1. Lung Injury Induces Alveolar Type 2 Cell Hypertrophy and Polyploidy with Implications for Repair and Regeneration

Abstract: Epithelial polyploidization post-injury is a conserved phenomenon, recently shown to improve barrier restoration during wound healing. Whether lung injury can induce alveolar epithelial polyploidy is not known. We show that bleomycin injury induces AT2 cell hypertrophy and polyploidy. AT2 polyploidization is also seen in short term ex vivo cultures, where AT2-to-AT1 trans-differentiation is associated with substantial binucleation due to failed cytokinesis. Both hypertrophic and polyploid features of AT2 cells can be attenuated by inhibiting the integrated stress response (ISR) using the small molecule ISRIB. These data suggest that AT2 hypertrophic growth and polyploidization may be a feature of alveolar epithelial injury. As AT2 cells serve as facultative progenitors for the distal lung epithelium, a propensity for injury-induced binucleation has implications for AT2 self-renewal and regenerative potential upon reinjury, which may benefit from targeting the ISR.

Abstract: Central micturition control and urine storage involve a multisynaptic neuronal circuit for the efferent control of the urinary bladder. Electrical stimulation of the lateral preoptic area (LPA) at the level of the decussation of the anterior commissure in cats evokes relaxation of the bladder, whereas ventral stimulation of LPA evokes vigorous contraction. Endogenous Angiotensin-(1–7) [(Ang-(1–7)] synthesis depends on ACE-2, and its actions on binding to Mas receptors, which were found in LPA neurons. We aimed to investigate the Ang-(1–7) actions into the LPA on intravesical pressure (IP) and cardiovascular parameters. The gene and protein expressions of Mas receptors and ACE-2 were also evaluated in the LPA. Angiotensin-(1–7) (5 nmol/μL) or A-779 (Mas receptor antagonist, 50 nmol/μL) was injected into the LPA in anesthetized female Wistar rats; and the IP, mean arterial pressure (MAP), heart rate (HR), and renal conductance (RC) were recorded for 30 min. Unilateral injection of Ang-(1–7) into the LPA increased IP (187.46 ± 37.23%) with peak response at ∼23–25-min post-injection and yielded no changes in MAP, HR, and RC. Unilateral or bilateral injections of A-779 into the LPA decreased IP (−15.88 ± 2.76 and −27.30 ± 3.40%, respectively) and elicited no changes in MAP, HR, and RC. The genes and the protein expression of Mas receptors and ACE-2 were found in the LPA. Therefore, the LPA is an important part of the circuit involved in the urinary bladder control, in which the Ang-(1–7) synthetized into the LPA activates Mas receptors for increasing the IP independent on changes in RC and cardiovascular parameters.

Abstract: INTRODUCTION Anismus is a pathology that is responsible for up to 50% of constipation cases, there are mainly 2 treatments: biofeedback and the use of botulinum toxin, with similar results. Does the application of botulinum toxin have a better cost-effectiveness index compared to biofeedback as a treatment in patients with anismus? The objective is to establish whether the application of botulinum toxin as a treatment in patients with anismus represents a better cost-effectiveness index compared to biofeedback. MATERIAL AND METHODS A randomized prospective cost-effectiveness trial was conducted evaluating the effectiveness of biofeedback treatment compared to the application of botulinum toxin in patients with anismus. Patients were followed up at month 1, 3, and 6 after management. RESULTS A total of 9 patients were included, 4 in the botulinum toxin group and 5 in the biofeedback group. The efficacy of botulinum toxin was 75% at month 1 and 3 and 50% at month 6. The efficacy of biofeedback remained at 80% at 6 months. The expense for biofeedback treatment was $ 17,458.32 MXN compared to $ 7,846.08 MXN for botulinum toxin. CONCLUSION A decrease in the average cost of $ 9,612.24 MXN was obtained in favor of the use of botulinum toxin for the treatment of anismus.

Abstract: During the coronavirus disease 2019 (COVID-19) pandemic, many countries opted for strict public health measures, including closing schools. After some time, they have started relaxing some of those restrictions. To avoid overwhelming health systems, predictions for the number of new COVID-19 cases need to be considered when choosing a school reopening strategy. Using a computer simulation based on a stochastic compartmental model that includes a heterogeneous and dynamic network, we analyse different strategies to reopen schools in the São Paulo Metropolitan Area, including one similar to the official reopening plan. Our model allows us to describe different types of relations between people, each type with a different infectiousness. Based on our simulations and model assumptions, our results indicate that reopening schools with all students at once has a big impact on the number of new COVID-19 cases, which could cause a collapse of the health system. On the other hand, our results also show that a controlled school reopening could possibly avoid the collapse of the health system, depending on how people follow sanitary measures. We estimate that postponing the schools' return date for after a vaccine becomes available may save tens of thousands of lives just in the São Paulo Metropolitan Area compared to a controlled reopening considering a worst-case scenario. We also discuss our model constraints and the uncertainty of its parameters.

Abstract: The objective of this study was to characterize the vaginal bacterial flora and their antibiotic susceptibility in vitro after, application, and the reuse of an intravaginal progesterone device (CIDR) in controlling the estrous cycle in sheep. Thirty Texel ewe lambs were used, after carefully assessed on their health and body condition. Experimental groups were formed by new CIDR (CIDRn, n = 15) and CIDR reused (CIDRu, n = 15). Treatment with progesterone lasted 6 days. Sterile swabs were used for vaginal sample collection, a procedure performed previously of CIDR insertion and on the moment of CIDR removing. The predominant flora prior to insertion of intravaginal devices proved to be 93.3% (14/15) and 80.0% (12/15) of gram-positive agents, especially of the genera Bacillus sp. and Staphylococcus sp. in CIDRn CIDRu groups, respectively. Upon removal of the devices, however, it was observed that the emergence of gram-negative agents in 100% of the samples CIDRn, most of them belonging to the Enterobacteriaceae genus. In CIDRu group, there was a balance between negative- and gram-positive agents on the removal of the devices. Regarding to susceptibility, our findings revelated some degree of resistance in all samples. Therefore, our conclusions pointed out that intravaginal devices (CIDR), when employed in the first use, or reuse, for a period of 6 days, promote a change in vaginal flora of sheep, highlighting the predominance of gram-negative agents during its removal.

Abstract: Os Papilomavírus humanos são os principais agentes causadores de câneeres genitais. Epidemins de HPV têm se tomado cada vez mais freqUentes no mundo inteiro, sendo a infecção pelo HPV hoje estimada como a mais comum das Doenças Sexualmente Transmissíveis. O nosso estudo teve como objetivo pesquisar a presença do HPV em biópsins de lesões do trato genital mnsculino, compatíveis com infecção por este vírus, pelo método de hibridização in sim. Foram examinadns 75 biópsias de lesões classificadns à peniscopia em: condilomas, lesões papulares e lesões acetobrancas . A prevalência da infecção foi de 78,7% (59/75), com predominância dos tipos 6 e 11, de baixo risco, detectados em 59.3% (35/59) dos casos positivos e a presença de HPVs de alto risco em 37,2% (22/59) das amostrns. A importância da detecção destes HPVs de alto risco está no fato dos pacientes do sexo masculino serem a principal fonte de infecção do trato genital feminino, onde os HPVs exercem com maior eficiência seu potencial de transformação maligna.

Abstract: A two-dimensional class of mean-field models serving as a minimal frame to study long-range interaction in two space dimensions is considered. In the case of an anisotropic mixed attractive-repulsive interaction, an initially spatially homogeneous cold fluid is dynamically unstable and evolves towards a quasi-stationary state in which the less energetic particles get trapped into clusters forming a Bravais-like lattice, mimicking a crystalline state. Superimposed to this, one observes in symplectic numerical simulations a flux of slightly more energetic particles channeling through this crystalline background. The resultant system combines the rigidity features of a solid, as particles from a displaced core are shown to snap back into place after a transient, and the dynamical diffusive features of a liquid for the fraction of channeling and free particles. The combination of solid and liquid properties is numerically observed here within the classical context. The quantum transposition of the model may be experimentally reached using the latest ultracold atoms techniques to generate long-range interactions.

Abstract: A new analytical method for mercury speciation in offshore petroleum produced water samples (PW) was developed using the combination of extraction (assisted by ultrasound in the presence of surfactant) and distillation before propylation and determination by gas chromatography cold vapor atomic fluorescence spectrometry (GC-CV-AFS). Ultrasonic (US) treatment (40 min) and the use of Triton X-114 (at 0.5% w/v at pH 3) improved recoveries during the distillation used to separate mercury species from the interfering PW matrix (rich in dispersed oil and salt). Recoveries as high as 92% (Hg2+), 87% (CH3Hg) and 86% (CH3CH2Hg) were achieved. Conditions were adjusted to minimize artifact formation during the samples treatment process in systems containing the major PW matrix components. The method detection limits were 5.0, 8.0 and 11.0 pg L- 1 respectively for Hg2+, CH3Hg and CH3CH2Hg. The use of matrix matched standards (containing mineral oil at 40 mg L−1 and NaCl at 30 g L−1) improved accuracy of the method that was successfully applied for the analysis of four different PW samples, obtained from off-shore operations, with Hg2+ and CH3Hg detected at the ng L−1 level.

Abstract: Continuing search for anticancer compounds from the marine environment, we have studied microorganisms that inhabit intertidal sediments of the northeastern Brazilian coast. Of the 32 strains isolated, 13 were selected for biological evaluation of their crude extracts. The acetate extract obtained from a Gram-negative bacterium was strongly active against cancer cell lines with IC(50) values that ranged from 0.04 (HL60 leukemia cells) to 0.26 μg/ml (MDA MB-435 melanoma cells). The bacterium was identified as a Pseudoalteromonas sp. based on 16S rRNA gene sequencing. A bioassay-guided fractionation of the active extract led to the isolation of prodigiosin, a well-known tripyrrole red pigment with immunosuppressive and anticancer activities. Further experiments with ErbB-2 overexpressing cell lines, including HB4a-C3.6 (moderate overexpression), HB4a-C5.2 (high overexpression), and the parental HB4a cell line, were performed. Prodigiosin was moderately active toward HB4a cells with an IC(50) of 4.6 μg/ml, while it was 115 and 18 times more active toward HB4a-C3.6 cells (IC(50) of 0.04 μg/ml) and HB4a-C5.2 (IC(50) of 0.26 μg/ml) cells, respectively. These data suggest that, in spite of its previously described apoptosis-inducing properties, prodigiosin can selectively recognize cells overexpressing ErbB-2, which could be highly appealing in human breast cancer therapy.

Abstract: Jasmonates are signaling molecules that play key roles in wound response and regulate the biosynthesis of many defensive proteins, including proteases. In this study, we investigate the effects of wounding and methyl jasmonate (MJ) application on the protein expression pattern of Ricinus communis L. leaves and on proteolytic activity. Gelatin zymography demonstrated that both MJ and mechanical wounding induce alterations in the proteolytic pattern of castor bean leaves (R. communis L.). Expression of two cysteine proteases (38 and 29 kDa) was induced by the treatments employed; however, MJ induced a higher protease level than mechanical wounding during the stress period (24, 48, and 72 h). The increase in protease activity mirrors the decline in soluble protein content and rubisco degradation that may indicate initiation of senescence in castor plants. The 29 kDa protease has an acidic optimal pH; whereas the 38 kDa protease has a neutral optimum activity. Both proteases were almost completely inhibited by E-64 and cystatin. The significant induction of these proteins by MJ suggests a possible role of cysteine proteases in leaf senescence as well as their involvement in regulating both the wound response and MJ in castor bean plants.

Abstract: Significant effort has been made world-wide to boost biofuels with the expectation of a positive contribution to renewable fuel and greenhouse gas reduction. Jatropha curcas L. has proved to be an opportunistic crop in tropical areas, particularly in unfavorable environments. For this reason, analyses of toxicity and allergy caused by its seeds and pollen are necessary. A 12 kDa, allergenic 2S albumin, denoted Jat c 1, was isolated from Physic nut (J. curcas) seeds. Jat c 1 binds IgE attached to rat mast cells, inducing histamine release. It also showed strong cross-reactivity with the major allergens from castor bean, Ric c 1 and Ric c 3.

Abstract: To determine the effect of gonadotropins on insulin- and insulin-like growth factor (IGF-I)-induced bovine granulosa cell functions, granulosa cells from bovine ovarian follicles were cultured for 2 days in the presence of 10% fetal calf serum (FCS), and then cultured for an additional 2 days in serum-free medium with added hormones. In the presence of 0 or 1 ng/mL of insulin or IGF-I, FSH had little or no effect (P0.05) on estradiol production by granulosa cells from both small (1-5mm) and large (or = 8mm) follicles. However, in the presence ofor = 3 ng/mL of insulin, FSH increased (P0.05) estradiol production by granulosa cells from small and large follicles such that the estimated dose (ED(50)) of insulin necessary to stimulate 50% of the maximum estradiol production was decreased by 2- to 3-fold from 22 to 28 ng/mL in the absence of FSH to 7-14 ng/mL in the presence of FSH. Similarly, in the presence ofor = 3 ng/mL of IGF-I, FSH increased (P0.05) estradiol production by granulosa cells from small and large follicles such that the ED(50) of IGF-I for estradiol production was decreased by 4- to 5-fold from 25 to 36 ng/mL in the absence of FSH to 5-6 ng/mL in the presence of FSH. In the presence of FSH, the maximal effect of insulin on estradiol production was much greater than that of IGF-I (137- versus 12-fold increase) and were not additive; when combined, 100 ng/mL of IGF-I completely blocked the stimulatory effect of 100 ng/mL of insulin. In the absence of FSH, the maximal effect of insulin and IGF-I on estradiol production was similar. Concomitant treatment with 30 ng/mL of LH reduced (P0.05) insulin-stimulated estradiol production by 52% on day 1 and 19% on day 2 of treatment. Insulin, IGF-I and FSH also increased (P0.05) granulosa cell numbers and progesterone production but their maximal effects were less (i.e.,4-fold increase) than their effects on estradiol production. In conclusion, insulin and IGF-I synergize with FSH to directly regulate ovarian follicular function in cattle, particularly granulosa cell aromatase activity.

Abstract: Objective Although improving mothers' knowledge about breastfeeding can increase rates and duration of breastfeeding, little is known about the influence of fathers' knowledge. The purpose of this study was to assess the knowledge of mothers and fathers about breastfeeding before and after receiving postpartum advice and its relationship to the frequency of breastfeeding. Methods A clinical trial was performed with mothers and fathers of normal children born at the Hospital de Clinicas de Porto Alegre, Brazil, between July 1994 and March 1995. The study intervention consisted of postpartum advice supplied by means of a video film discussing basic topics of breastfeeding, an explanatory leaflet, and open discussion after viewing the video. The first 208 couples comprised the control group, the next 197 comprised experimental group 1, and the remaining 196 comprised experimental group 2. Immediately after delivery, mothers and fathers in the three groups answered a test on breastfeeding knowledge; they completed the same test at the end of the first month. All families received home visits at the end of the first, second, fourth, and sixth months, or until breastfeeding ceased. Logistic regression was used to evaluate the association between the mothers' and fathers' knowledge and frequency of breastfeeding. Results Postpartum advice increased the breastfeeding knowledge of mothers and fathers. The mothers with the highest level of knowledge had a 6.5 times higher chance of exclusively breastfeeding at the end of the third month, and 1.97 times higher chance of continuing breastfeeding to the end of the sixth month compared with other mothers. The fathers' knowledge also significantly influenced breastfeeding rates. The children whose fathers knew more had a 1.76 higher chance of being exclusively breastfed at the end of the first month, and 1.91 higher chance of receiving maternal milk at the end of the third month. Conclusion A simple, inexpensive strategy can increase the level of breastfeeding knowledge of mothers and fathers and, consequently, have a positive impact on the frequency of breastfeeding.

Abstract: The objectives of this study were to research the effects of herbage allowance (HA) on dry matter (DM) intake, milk yield, grazing efficiency and grazing behaviour of dairy cows grazing alfalfa (Medicago sativa L.) pastures, and to compare the estimates of herbage DM intake using either an external intake marker developed from a purified enriched lignin (LIPE®) or the animal performance method, the latter based on calculations of energy requirements.Twenty six spring calving crossbred Holstein-Jersey dairy cows (84 days in milk) were balanced and assigned to either the low HA (Lo-HA; 10.0 kg DM/cow/day) or the high HA treatment (Hi-HA; 14.5 kg DM/cow/day). Cows were offered alfalfa (Medicago sativa L.) pastures at grazing, Sorghum (Sorghum bicolor) silage (3.7 kg DM/cow/day) and concentrates (5.6 kg DM/cow/day). Herbage DM intake was estimated using the sward cutting method, the animal performance method and an indigestible intake marker (LIPE®). Abstract: The relative susceptibility of 4 tropical maize pests, Spodoptera frugiperda (J. E. Smith),Diatraea grandiosella Dyar, D. saccharalis (F.), and Helicoverpa zea (Boddie), to the lepidopteran-specific CryI-type proteins produced by B. thuringiensis is presented. The toxin with the highest potency against H. zea larvae was the CryIAc toxin. S. frugiperda larvae were susceptible to CryID and CryIF toxins. The CryIB toxin showed to be highly toxic against D. grandiosella and D. saccharalis. This information will establish a basis for selecting B. Thuringiensis strains producing the appropriate CryI proteins to be used for the biological control of these tropical pests.

Abstract: Background The plasma concentration ratio of triglyceride (TG)/high-density lipoprotein cholesterol (HDL-C) can identify cardiometabolic risk and cardiovascular disease. The visceral adiposity index is a sex-specific index, in which measurements of body mass index and waist circumference are combined with TG and HDL-C concentrations. The current analysis was initiated to see if the visceral adiposity index would improve the ability of the TG/HDL-C to identify increased cardiometabolic risk and outcome. Methods Cardiometabolic data were obtained in 2003 from 926 apparently healthy individuals, 796 of whom were evaluated in 2012 for evidence of incident cardiovascular disease. The relationship between TG/HDL-C and values for visceral adiposity index was evaluated by Pearson's correlation coefficient. The relative risks for first cardiovascular event between individuals above and below the TG/HDL-C sex-specific cut points, and in the top quartile of visceral adiposity index versus the remaining 3 quartiles, were estimated using Cox proportional hazard models. Results TG/HDL-C concentration and visceral adiposity index were highly correlated ( r = 0.99) in both men and women. Although more men (133 vs121) and women (73 vs 59) were identified as being at "high risk" by an elevated TG/HDL-C ratio, the individual cardiometabolic risk factors were essentially identical with either index used. However, the hazard ratio of developing cardiovascular disease was significantly increased in individuals with an elevated TG/HDL-C, whereas it was not the case when the visceral adiposity index was used to define "high risk." Conclusion The visceral adiposity index does not identify individuals with an adverse cardiometabolic profile any better than the TG/HDL-C.

29. Influence of perioestrous suprabasal progesterone levels on cycle length, oestrous behaviour and ovulation in heifers

Abstract: In order to induce suprabasal plasma concentrations of progesterone after luteolysis and to determine their effect on oestrous behaviour and ovulation, heifers subcutaneously received silicone implants containing 2.5 (n = 4), 5 (n = 4), 6 (n = 3), 7.5 (n = 3) or 10 (n = 4) g of progesterone, or an empty implant (controls, n = 5) between days 8 and 25 of the cycle (ovulation designated Day 0). Growth of dominant follicles and time of ovulation were determined by ultrasound, and signs of oestrus were recorded and scored. Blood was collected at 2–4 h intervals from Days 15 to 27 and assayed for progesterone concentration. In all heifers, plasma concentrations of progesterone sharply decreased during Days 16–18. Control heifers had their lowest progesterone levels on Days 20.5 and 21, standing oestrus on Day 19.5 ± 0.4 (mean ± SEM), and ovulated on Day 20.7 ± 0.4. A similar pattern was observed in heifers treated with 2.5 and 5 g progesterone. Heifers treated with 6, 7.5 and 10 g of progesterone showed an extended (P < 0.05) interovulatory interval. Onset of prooestrus and time of maximum expression of signs of oestrus were not significantly different from those in controls. However, there was an absence of standing oestrus in most of the cases, signs of oestrus lasted longer (P < 0.05) and were weaker in intensity when doses increased. In these groups, the lowest progesterone concentrations were attained shortly after implant removal. Some heifers treated with 6 and 7.5 g of progesterone had standing oestrus and post oestrous bleeding as seen in the controls but ovulation occurred from Days 24.5 to 27. When plasma progesterone concentrations were over 1 nmol 1−1, disturbed oestrus and delayed ovulation occurred. The extended period of prooestrus and oestrus and delayed ovulation were similar to that described in cases of repeat breeding. It is suggested that suprabasal plasma concentrations of progesterone, after luteolysis, may lead to asynchrony between onset of oestrus and ovulation and consequently be a cause of repeat breeding in cattle.

Abstract: The aim of the present study was to determine the degree of development and structural status of perfusion-fixed day-7 corpora lutea (CL) (using qualitative and quantitative histology at light and electron microscopy levels) in FSH-induced superovulated (SO) and untreated heifers. Blood samples were collected daily to monitor the plasma progesterone levels during the FSH treatment up to slaughter 7 days after oestrus. Meanwhile the ovarian activity was followed by ultrasonography and rectal palpation. At slaughter, the ovaries were fixed by vascular perfusion to avoid distortion of the structures and conventionally processed for electron microscopy. The volume density of the luteal tissue was calculated on the examined sections by point-counting. The histology of the corpora lutea, exception made of a higher incidence of degenerated luteal and endothelial cells in the superovulated animals, did not differ from that of the untreated animals, confirming their normal development. The mean weight and volume of the CL in SO animals was 50% smaller than that of the untreated heifers. The progesterone concentration (nmol/l) at day 7 was significantly positively correlated with the number of CLs/heifer (r = 0.93, P less than 0.01), with the weight of Cls/heifer (r = 0.97, P less than 0.005) and with the volume of CLs/heifer (r = 0.97, P less than 0.005). The results indicated that there were no morphological differences, in terms of histological structure and volume density, between the luteal tissue of SO heifers and the luteal tissue of non-SO heifers on day 7 of the oestrous cycle which could interfere with its endocrine function.

Abstract: Evidence based on immunological cross-reactivity and anti-diabetic properties has suggested the presence of insulin-like peptides in plants. The objective of the present study was to investigate the presence of insulin-like proteins in the leaves of Bauhinia variegata ("pata-de-vaca", "mororo"), a plant widely utilized in popular medicine as an anti-diabetic agent. We show that an insulin-like protein was present in the leaves of this plant. A chloroplast protein with a molecular mass similar to that of bovine insulin was extracted from 2-mm thick 15% SDS-PAGE gels and fractionated with a 2 x 24 cm Sephadex G-50 column. The activity of this insulin-like protein (0.48 mg/mL) on serum glucose levels of four-week-old Swiss albino (CF1) diabetic mice was similar to that of commercial swine insulin used as control. Further characterization of this molecule by reverse-phase hydrophobic HPLC chromatographic analysis as well as its antidiabetic activity on alloxan-induced mice showed that it has insulin-like properties. Immunolocalization of the insulin-like protein in the leaves of B. variegata was performed by transmission electron microscopy using a polyclonal anti-insulin human antibody. Localization in the leaf blades revealed that the insulin-like protein is present mainly in chloroplasts where it is also found associated with crystals which may be calcium oxalate. The presence of an insulin-like protein in chloroplasts may indicate its involvement in carbohydrate metabolism. This finding has strengthened our previous results and suggests that insulin-signaling pathways have been conserved through evolution.

34. Superovulatory response in lactating cows with different follicular dynamics

Abstract: To determine the influence of a dominant follicle on the superovulatory response in midlate lactating cows, the dynamics of follicular growth and the development of corpora lutea and embryos were studied. Real-time B-mode ultrasonography with a 7.5 MHz rectal linear-array transducer was used to scan the ovaries daily, from 8-12 days before superovulation until day 7 post-estrus, when embryos were recovered. On the first day of superovulation cows were classified as belonging to either a 'dominant' group (n = 12) having a 'dominant follicle', i.e.9 mm in diameter in growing phase or stable for4 days, or a 'non-dominant' group (n = 11) those carrying a 'non-dominant follicle', i.e. a follicle in a regressing phase or with the same diameter for4 days. Cows in the 'dominant' group had significantly higher numbers of 4-6 and 7-10 mm follicles than cows in the 'non-dominant' group on the third and the fifth day of superovulation. The total number of follicles (4 mm) preovulation and the number of CL day 7 postovulation was higher in the 'dominant' group than in the 'non-dominant' group. There were no differences between groups regarding the total number of embryos or transferable embryos recovered. The results indicate that superovulation in mid-late lactation is not negatively affected by the presence of a 'dominant follicle'. The criteria to define 'dominant' and 'non-dominant' follicles might be redefined when applied to lactating cows, whose response to superovulation seems to be under the influence of many other factors.

Abstract: Inthelastdecadesseveralinvestigatorshavesuggestedtheassociationofandrogenswithhypertension. Recently, some studies have shown that the incidence of cardiovascular diseases is increased in men with low testosterone levels, suggesting a protective role of androgens. Hypertension is one of the mostcommonproblemsinpregnancythatcomplicates5-10 %ofpregnancies.Anincreaseofmorbidity wasobservedinbabiesfrompregnantwomenwithhypertension.However,thepathogenesisremains unclearandthislimitstheabilitytopreventandtreatthispathology.Thebeneficialeffectsofandrogens for vascular system are associated with their ability to cause vasorelaxation. Inhuman vessels, this non genomiceffectofandrogensappeartobeduetoactivationoflargeconductancecalcium-activatedpotassiumchannels(BKCa)andvoltagegatedpotassiumchannels(KV)whichisinducedbycGMPincrease andaconsequentactivationofcGMP-dependentproteinkinase(PKG).Ontheotherhand,thegenomic effects of androgens concerning ionic channels are almost unknown. Abstract: A total of 99 paediatric patients (57 male, 42 female) aged 1 to 12 years, weighing 10 to 40kg and with acute pharyngo-amygdalitis were enrolled in a single-blind study to assess the efficacy and tolerability of nimesulide in comparison with naproxen when both drugs were administered over an 8-day treatment period. Among the 2 treatment groups comprising 99 evaluable patients, demographic analysis of age, weight and height did not reveal statistically significant differences. Evaluation of fever, pain, inflammation and nasal obstruction over the 8-day treatment period showed a significant improvement in these parameters for those patients treated with nimesulide when compared with naproxen from day 1, with remission of symptoms starting from day 3. These findings were complemented by a superior tolerability profile reported for nimesulide-treated patients. In conclusion, nimesulide appears to be a safe and effective treatment for paediatric patients with pharyngo-amygdalitis and it has shown superior efficacy and tolerability when compared with naproxen.

Abstract: Superovulation (SO) with FSH was induced in 23 dairy cows and 12 dairy heifers which were treated on the day of embryo collection (Day 7) with a single luteolytic injection of either 500 mg or 1000 mg of Cloprostenol (Clp). The animals were clinically examined and their plasma progesterone (P4) levels were monitored for 30 days after Clp injection. Estrus signs were checked twice daily. The ovaries were scanned by ultrasonography to follow corpora luteal (CL) regression, follicular, development and ovulation. The CLs regressed in all 34 animals responding to SO. Estrus signs occurred within 16 days after Clp injection in 91.2% of the population (19 cows and 12 heifers); ovulation occurred in 55.9% of the 34 animals responding to SO (12 cows and 7 heifers ). Of the animals showing estrus, the 7 cows and 5 heifers (38.7%) that did not ovulate either became cystic (4 cows and 3 heifers) or had follicular growth waves which never reached ovulation(3 cows and 2 heifers). Furthermore, 3 cows (8.8%) did not show estrus signs during this period. In the second cycle following Clp administration, the interestrus interval was within normal ranges (17 to 24 days) and estrus was followed by ovulation even in most of those animals with previously non-ovulated follicular waves. The cystic cases (20.6%) and 1 anestrus case, however, remained. Heifers had a significantly shorter interval from embryo collection to first estrus than cows. No significant differences in the effect of the 2 Clp dosages assayed on the interval from embryo collection to the first ovulatory estrus were found in those animals having a normal ovulatory estrus. The P4 plasma profiles confirmed both luteolysis after Clp injection and the occurrence of ovulation. In heifers, the concentration of P4 on the day of embryo collection and 24 h thereafter was positively correlated with the interval from collection to first estrus. Besides the satisfactory luteolytic effect of Clp, the resumption of cyclicity seemed to be more influenced by the disturbed ovarian dynamics at the time when Clp was injected than by its dosage.

Abstract: Abstract A two-dimensional class of mean-field models serving as a minimal frame to study long-range interaction in two space dimensions is considered. In the case of an anisotropic mixed attractive-repulsive interaction, an initially spatially homogeneous cold fluid is dynamically unstable and evolves towards a quasi-stationary state in which the less energetic particles get trapped into clusters forming a Bravais-like lattice, mimicking a crystalline state. Superimposed to this, one observes in symplectic numerical simulations a flux of slightly more energetic particles channeling through this crystalline background. The resultant system combines the rigidity features of a solid, as particles from a displaced core are shown to snap back into place after a transient, and the dynamical diffusive features of a liquid for the fraction of channeling and free particles. The combination of solid and liquid properties is numerically observed here within the classical context. The quantum transposition of the model may be experimentally reached using the latest ultracold atoms techniques to generate long-range interactions.
