Infrared spectroscopy is an effective technique extensively used in research and industry for the label-free and unambiguous identification of molecular species. However, the sensitivity of this technique is severely limited as a result of Beer's law and, the small infrared absorption cross-section that make prohibitively weak the absorption signals, of minute amounts of analyte as those present in monolayers. This limitation can be overcome by enhancing the infrared vibration of molecules through the enhancement of the electromagnetic (EM) field. Surface Enhanced InfraRed Absorption (SEIRA) using resonant metal Nano-scale Antennas (NAs) can provide huge electromagnetic fields on the nanometer scale featuring localized collective oscillations of electrons, an effect named Localized Surface Plasmonic Resonances (LSPRsWe here report on a series of 2D arrays of cross-shaped NAs having several mm(2) area coverage (metasurface) as SEIRA optimized antennas, which can be used in practical applications such as the vibrational sensing of chemical and biological analytes. Cross-shape designed NAs are insensitive to the polarization of the electromagnetic radiation impinging the active area. Due to the random orientation of the dipole moments of molecules they are particularly suitable for the construction of bio-molecular sensors. At the same time, the 2D-array configuration ensures a good near-field signal enhancement arising from the coupling between neighbour NAs Moreover, SEIRA NAs can be easily integrated with micrometre-sized channels and be suitable for the high sensitivity, real time analysis of IR emitting samples, in matrices where IR spectroscopy is severely limited due to absorption bands of liquid water. We present the design, fabrication and experimental characterization of large-area metasurfaces based on cross-shaped plasmonic NAs for the spectroscopic characterization of various types of compounds and for sensing applications in the mid-infrared range. The cross-shaped NAs we have designed exhibit SEIRA phenomena which are very sensitive to both refractive index changes in the surrounding medium and to the specific molecular vibration band emerging from surface adsorbed molecules. To test this effect on our device, we have used as model compounds small molecules (molecular weight (MW) < 500 g/mol) containing triple bond groups resonating at about 2100 cm(-1) and a large polymer (MW (similar to) 950,000 g/mol) containing carbonyl groups resonating at wavenumbers of about 1700 cm-1. We show a sensitivity of 600 nm/RIU at different wavelengths at a maximum amount of immobilized small molecule of 0.7 fmoles and a SEIRA enhancement factor of 48,000. We also show the device potential to reveal chemical reactions, occurring on the sensor surface at the same scale, where the nitrile group is converted to a triazole ring.

Fourier transform infrared micro-spectroscopy (&mu, FT-IR) is nowadays considered a valuable tool for investigating the changes occurring in human cells after exposure to ionizing radiation. Recently, considerable attention has been devoted to the use of this optical technique in the study of cells exposed to proton beams, that are being increasingly adopted in cancer therapy. Different experimental configurations are used for proton irradiation and subsequent spectra acquisition. To facilitate the use of &mu, FT-IR, it may be useful to investigate new experimental approaches capable of speeding up and simplifying the irradiation and measurements phases. Here, we propose the use of low-e-substrates slides for cell culture, allowing the irradiation and spectra acquisition in transflection mode in a fast and direct way. In recent years, there has been a wide debate about the validity of these supports, but many researchers agree that the artifacts due to the presence of the electromagnetic standing wave effects are negligible in many practical cases. We investigated human normal breast cells (MCF-10 cell line) fixed immediately after the irradiation with graded proton radiation doses (0, 0.5, 2, and 4 Gy). The spectra obtained in transflection geometry showed characteristics very similar to those present in the spectra acquired in transmission geometry and confirm the validity of the chosen approach. The analysis of spectra indicates the occurrence of significant changes in DNA and lipids components of cells. Modifications in protein secondary structure are also evidenced.

Optical techniques are increasingly employed for monitoring cell–matrix interactions in suitably prepared 3D scaffolds. The ability of designing and realizing synthetic extracellular matrix with well-controlled optical properties is a crucial need in this field. For this purpose a crosslinked hyaluronic acid (HA) scaffold is prepared. Fourier transform infrared and UV–vis spectroscopies enable to monitor the scaffold preparation process and to evidence scaffold high transparency and low fluorescence in the visible range. 3D optical characteristics of the HA scaffold are tested by two-photon microscopy (TPM) imaging of embedded fluorescent microbeads and alive keratinocytes labeled with vital PKH67 dye at different depths from the scaffold surface. Some useful indications about the potentiality of TPM measurements for the determination of attenuation coefficient of turbid media are also reported. Moreover, the use of the presented HA scaffold for preparing tissue phantoms for fluorescence imaging or diffuse imaging is proposed. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. Surface-enhanced Raman Spectroscopy (SERS) is a vibrational spectroscopy holding potentials for a rapid evaluation of quality and composition of food industry products without any need of sample preparation. Among many nanomaterials, gold nanoparticles (GNPs) and their colloidal dispersions have attracted great interest for SERS applications due to their unique properties of small size, large surface area to volume ratio, high reactivity to the living cells, stability over high temperatures. In this frame, a low-cost substrate, based on home-made 30-nm sized GNPs, has been designed and used for the investigation of commercial fruit juices and pulp. Thanks to the use of a wavelet denoising procedure and background subtraction spectra with clear features have been obtained. Their analysis has enabled to evidence the presence of components of great importance for the quality evaluation of the products, such as fructose and pectin. The overall inspection of the results has confirmed the potentialities of SERS in food industry.

Micro-Attenuated Total Reflection (ATR) Fourier Transform Infrared spectroscopy was used to investigate sol–gel layers for biosensing applications prior and after glucose oxidase (GOD) immobilization. The changes occurring in sol–gel infrared spectrum after GOD immobilization were clearly evidenced confirming the retaining of the enzyme activity. Moreover, micro-ATR experimental technique allowed us to investigate the spatial distribution of enzyme concentration. The non-destructive nature of our approach also enabled to monitor the time stability of sol–gel layers and of embedded GOD. The temporal evolution of some peaks in infrared spectra of these sol–gel layers was compared with absorption and steady-state fluorescence measurements. The results reported here confirm that micro-ATR infrared spectroscopy can be usefully employed for a non- or minimally invasive detailed characterization of supports for enzyme immobilization.

A monolithic silica gel matrix with entrapped glucose oxidase was constructed as a bioactive element in an optical biosensor for glucose determination. Physicochemical and biochemical characterizations of the catalytic matrix were performed, and the intrinsic fluorescence of immobilised glucose oxidase (GOD) was investigated in the UV and visible range by performing steady state and time course measurements. In all cases, the silica gel matrix proved to be a suitable support for optical biosensing owing to its superior optical properties (e.g., high transmittance and reliable fluorescence and GOD absorption spectra after immobilisation). From steady state measurements, calibration curves were obtained as a function of glucose concentration. When time course measurements were performed, the silica gel support displayed a larger linear calibration range and higher sensitivity than other immobilisation systems. In addition, a glucose optical biosensor was developed and characterised using as catalytic element GOD immobilised on a gel disk bound to a bundle of optical fibres.

A hollow-fiber enzyme reactor, operating under isothermal and nonisothermal conditions, was built employing a polypropylene hollow fiber onto which beta-galactosidase was immobilized. Hexamethylenediamine and glutaraldehyde were used as spacer and coupling agent, respectively. Glucose production was studied as a function of temperature, substrate concentration, and size of the transmembrane temperature gradient. The actual average temperature differences across the polypropylene fiber, to which reference was done to evaluate the effect of the nonisothermal conditions, were calculated by means of a mathematical approach, which made it possible to know, using computer simulation, the radial and axial temperature profiles inside the bioreactor and across the membrane. Percent activity increases, proportional to the size of the temperature gradients, were found when the enzyme activities under nonisothermal conditions were compared to those measured under comparable isothermal conditions. Percent reductions of the production times, proportional to the applied temperature gradients, were also calculated. The advantage of employing nonisothermal bioreactors in biotechnological industrial process was discussed.

A wavelet multi-component decomposition algorithm has been used for data analysis of micro-Raman spectra of blood serum samples from patients affected by pemphigus vulgaris at different stages. Pemphigus is a chronic, autoimmune, blistering disease of the skin and mucous membranes with a potentially fatal outcome. Spectra were measured by means of a Raman confocal microspectrometer apparatus using the 632.8 nm line of a He-Ne laser source. A discrete wavelet transform decomposition method has been applied to the recorded Raman spectra in order to overcome problems related to low-level signals and the presence of noise and background components due to light scattering and fluorescence. This numerical data treatment can automatically extract quantitative information from the Raman spectra and makes more reliable the data comparison. Even if an exhaustive investigation has not been done in this work, the feasibility of the follow-up monitoring of pemphigus vulgaris pathology has been clearly proved with useful implications for the clinical applications.

Surface functionalization by layers of hydrophobins, amphiphilic proteins produced by fungi offers a promising and green strategy for fabrication of biomedical and bioanalytical devices. The layering process of the Vmh2 hydrophobin from Pleurotus ostreatus on Teflon membrane has been investigated by Attenuated Total Reflection Fourier Transform Infrared (ATR FT-IR) spectroscopy. In particular, protein layers obtained with hydrophobin purified with two different procedures and in various coating conditions have been examined. The layers have been characterized by quantifying the amide I and amide II band area together with the lipid/amide ratio and carbohydrate/amide ratio. This characterization can be very useful in evaluating the best purification strategy and coating conditions. Moreover the analysis of the secondary structure of the layered protein using the deconvolution procedure of amide I band indicate the prevalent contribution from β-sheet state. The results inferred by infrared spectroscopy have been also confirmed by scanning electron microscopy imaging.

Optical techniques are increasingly employed for monitoring cell-matrix interactions and the availability of 3D scaffolds with controlled optical properties can be extremely useful in this field. Hyaluronic acid (HA) is a glycosaminoglycan found in extracellular tissue in many parts of the body which is becoming more and more popular in biomaterials science as key material for the creation of new tissues using scaffolds. In this frame, a crosslinked hyaluronic acid-lys scaffold has been prepared and characterized. The peculiar optical properties of this scaffold make it a good candidate for exploiting optical techniques for monitoring cell-matrix interactions. Its optical and physico-chemical properties have been investigated by means of UV-VIS and FT-IR spectroscopy. In particular, transmittance and fluorescence measurements have shown low levels of background signals coming from the scaffold itself. In addition the optical quality of 3D scaffolds has been preliminarily tested by two-photon microscopy imaging of embedded fluorescent microbeads and labelled human keratinocites cells at different depths from the scaffold surface.

Summary Objective. This study aims to give an insight into the chemical and structural modifications occurring in laser treated dentine surface, as a tool to plan a new class of dental materials specific for the dental restoration when laser technologies are used. For this purpose the effect of Er:YAG (Erbium–Yttrium Aluminium Garnet) laser treatments on human dentine surface has been investigated. Methods. The dentine surface of periodontally compromised teeth cut along the main axis was treated in vitro with laser pulses having the same energy but time duration varying in the range of 100–1000 ms. The chemical characteristics and the morphology of the laser treated surface have been studied with micro-Raman spectroscopy (m-RS) and scansion electron microscopy (SEM). Results. The m-RS and SEM analysis of laser treated teeth have been compared with those of surfaces treated with conventional mechanical methodologies. The laser pulse time duration affects the surface morphology and the chemistry of dentine, mainly of organic components. The effects have been monitored by analysing the C–H group stretch vibration mode of the m-RS spectrum. Conclusion. The laser treatment can determine different chemical microenvironments on the treated dentine surface, depending on the time duration of laser pulse used. Treatments with very long pulse results in a the dentine surface very similar to that obtained with a conventional mechanical drill, while with very short laser pulses a strong modification of the dentine organic components has been observed. q 2004 Elsevier Ltd. The potential use of micro-Raman spectroscopy for estimating the microstructural changes occurring in periodontal ligaments after orthodontic treatment was explored. An in vitro investigation on bovine ligaments was carried out in order to find the appropriate conditions for studying human cases. The results were compared with preliminary analysis from human samples.

Biodegradable polymer nanoparticles have recently received considerable attention for drug delivery. One of the most investigated system is represented by cisplatin-loaded poly-(D, L-lactic-co-glycolic)acid (PLGA) nanoparticles. FT-IR microspectroscopy has been used for characterizing these particles. In addition an univariate analysis procedure has been adopted for interpreting experimental results when the spectrum modifications are relatively weak.

An optical model for Raman response of thin metal films has been developed, taking into account attenuation effects and multi-reflections occurring at film edges. Film thickness and surface morphology of nanometer thin NbN films have been inferred in the framework of this model using micro-Raman measurements. Results have been compared with those obtained by means of spectroscopic ellipsometry. The absolute value of the thickness is determined with a precision better than 20% on nanometric scale allowing us to control surface morphology with high accuracy also on large areas.

To study immobilized protein interactions with dissolved substrates is a very important topic both from a fundamental and technological standpoint. In the present report we illustrate the preliminary results obtained on sol-gel immobilized glucose oxidase (GOD) using a standard de-scanned two-photon microscope based on a modified confocal scanhead with internal detectors and a Ti:sapphire laser as a source. Data acquisition conditions were preliminary defined using functionalized beads of different dimensions. Various sol-gel supports were then investigated by monitoring endogeneous fluorescence due to the flavoadenine (FAD) molecules, present in GOD. Linear absorption and fluorescence spectroscopy along with Fourier Transform Infrared microscopy were employed for a full-optical characterization of the samples. The results show that GOD immobilization processes can be successfully monitored in some cases and also the interaction with glucose could be studied by this approach. This assessment holds potentials to better understand the characteristic of immobilized enzymes biocatalysis and to develop new biosensing schemes.

We report an experimental study of two-photon absorption in GaAs/AlGaAs quantum wires. Strongly anisotropic selection rules are observed depending on the polarization direction of the exciting field with respect to the carrier confinement direction in the wires.

The investigation of materials suitable for enzyme immobilization in biosensing applications has a widespread interest. There are many studies on physico-chemical properties of these materials at macroscopic level but few studies have been devoted to examine and correlate these properties at microscopic level. FT-IR spectroscopy with Micro-Attenuated Total Reflection (Micro-ATR) approach can be extremely useful for understanding a variety of aspects of materials which can be used for optimising immobilization procedures. Moreover, this experimental approach is particularly simple to use (no sample preparation is required) and minimally invasive. Using a Perkin Elmer Spectrum One FT-IR spectrometer equipped with a mercury-cadmium-telluride detector and a micro-ATR element we investigated different materials used for immobilization procedures with various enzymes widely used for biosensing in environmental and clinical applications. In particular, composite membranes constituted by a chemically modified poly-acrylonitrile (PAN) membrane plus layers of tethered chitosan of different molecular weight have been examined. Also silica gel matrices without and with glucose oxidase have been investigated. Spectra have been analysed and the contribution of principal functional groups has been evidenced.

An "in vitro" study of Raman spectra from oral human tissues is reported in order to the develop a diagnostic method suitable for "in vivo" oral pathology follow-up. The investigated pathology is Pemphigus Vulgaris (PV) for which new techniques for guiding and monitoring therapy would be particularly useful. Raman spectra were obtained in the wavenumber regions from 1000 to 1800 cm -1 and 2700 to 3200 cm -1 from tissues from patients at different stages of pathology (active PV, under therapy and in PV remission stage) as confirmed by histopathological and immunofluorescence analysis. Differences in the spectra depending on tissue illness stage arise in 1150-1250 cm -1 (amide III) and 1420-1450 cm -1 (CH 3 deformation) regions and around 1650 cm -1 (amide I) and 2930 cm -1 (CH 3 symmetric stretch). A wavelet deconvolution procedure was applied to the spectra for better discriminating among the three different stages of illness and a linear regression analysis was used to fully exploit the content of information of Raman spectra.

The disease is characterized histologically by intradermal blisters and immunopathologically by the finding of tissue bound and circulating immunoglobulin G (IgG) antibody directed against the cell surface of keratinocytes. More than 150 spectra were measured by means of a Raman confocal microspectrometer apparatus using the 632.8 nm line of a He-Ne laser source. A discrete wavelet transform decomposition method has been applied to the recorded Raman spectra in order to overcome related to low-level signals and the presence of noise and background components due to light scattering and fluorescence. The results indicate that appropriate data processing can contribute to enlarge the medical applications of micro-Raman spectroscopy.

The two-photon absorption spectra of GaAs/AlGaAs multiple quantum well and superlattice structures have been experimentally investigated by means of the nonlinear luminescence technique in different polarization configurations. A strong excitonic effect overlapping the interband two-photon spectrum has been found and the selection rules for the excitonic transitions have shown to greatly change for different polarizations. The comparison of linear and nonlinear absorption measurements provides important information on the excited states of excitons in multiple quantum wells.

The formation of a hybrid layer at the dentin/resin interface is a critical point in the curative processes. New restorative materials are currently under investigation in order to improve the characteristics of this interface layer. In this paper micro-Raman spectroscopy and Environmental Scanning Electron Microscope (E-SEM) analysis have been performed to investigate the interface properties of three different restorative materials. The experimental investigation has been performed on tooth cavities prepared both by Er:YAG laser ablation and by conventional diamond bur. Laser prepared cavities were realized using 100 ms pulsed light beam with energy of 350 mJ, at a frequency of 20 Hz. The cavities were filled with three different composites: True-Vitality (Den-Mat), Enamel Plus (Micerium), Supreme (3M). The treated samples were sectioned perpendicularly to the exposed dentin surface and the morphological characterization of the samples was performed by means of E-SEM operating in wet mode. This equipment permits to obtain a high-resolution image of surface without conductive coating process. The same samples were then examined by micro-Raman spectroscopy that has already shown its validity in the study of dentin/resin interfaces.

Micro-Raman Spectroscopy (μ-RS) provides a unique tool in medicine for a not invasive and real time analysis of biological tissue for biopsy and "in vivo" investigation. Based on the evaluation of molecular vibration frequencies, the μ-RS is able to detect the main molecular bonds of protein constituents, as the C-H and C-C ones. Changes in frequency or in the relative intensity of the vibration modes revealed by μ-RS can be related to changes of chemical bond and of protein structure induced by pathology. The μ-RS has been performed on samples of oral tissue from informed patients, affected by pemphigus vulgaris (an oral pathology) in an advanced regression state. The biopsies were thin slices (about 1mm thick) with 6mm diameter. The sample was measured through a 170 μm thick cover-glass. The experimental set-up was mainly composed by a He-Ne laser and a monochromator equipped with a Peltier cell and with a grating of 1800 grooves/mm. The laser light was focused on the sample surface by means of a long focal length 50X optical objective. The main protein bonds are clearly detectable in the considered samples and this give important information on the integrity and on the state of tissue components (lipids and proteins), and consequently on the occurrence of pathology. The potential application of this method for in vivo analysis is an invaluable alternative to biopsy and pathological examinations for many medical application as screening diagnostic, therapy progress examination, and surgical support.

A wavelet multi-component decomposition algorithm is proposed for processing data from micro-Raman spectroscopy (μ-RS) of biological tissue. The μ-RS has been recently recognized as a promising tool for the biopsy test and in vivo diagnosis of degenerative human tissue pathologies, due to the high chemical and structural information contents of this spectroscopic technique. However, measurements of biological tissues are usually hampered by typically low-level signals and by the presence of noise and background components caused by light diffusion or fluorescence processes. In order to overcome these problems, a numerical method based on discrete wavelet transform is used for the analysis of data from μ-RS measurements performed in vitro on animal (pig and chicken) tissue samples and, in a preliminary form, on human skin and oral tissue biopsy from normal subjects. Visible light μ-RS was performed using a He–Ne laser and a monochromator with a liquid nitrogen cooled charge coupled device equipped with a grating of 1800 grooves mm−1. The validity of the proposed data procedure has been tested on the well-characterized Raman spectra of reference acetylsalicylic acid samples.

Light scattering processes in unconfined nematic liquid crystals have been investigated by means of time-resolved transmittance and static light scattering techniques. In time-resolved experiments, sample temperature has been modulated and different modalities of propagation have been probed. The results here obtained show that a nematic polydomain liquid crystal exhibits scattering properties strongly different from a nematic monodomain and, in a sense, it behaves as an optically thin sample. Such a behaviour may be ascribed to the strong backscattering exhibited by the polydomain sample, as indicated by static light scattering experiments, where the angular dependence of the scattered intensity was measured, revealing a preferential backward direction. The two experimental techniques here adopted have shown their complementary role to understand the actual scattering regime. The diffusion constant D has been determined by means of time-resolved measurements and a comparison with the results obtained in monodomain nematic liquid crystals has been attempted. Raman spectroscopy of oral tissues is a promising tool for in vivo diagnosis of oral pathologies, due to the high chemical and structural information content of Raman spectra. However, measurements on biological tissues are usually hindered by low level signals and by the presence of interfering noise and background components due to light diffusion or fluorescence processes. Numerical methods can be used in data analysis, in order to overcome these problems. In this work the wavelet multicomponent decomposition approach has been tested in a series of micro-Raman measurements performed on “in vitro” animal tissue samples. The experimental set-up was mainly composed by a He-Ne laser and a monochromator equipped with a liquid nitrogen cooled CCD equipped with a grating of 1800 grooves/mm. The laser light was focused on the sample surface by means of a 50 X optical objective. The resulting spectra were analysed using a wavelet software package and the contribution of different vibration modes have been singled out. In particular, the C=C stretching mode, and the CH2 bending mode of amide I and amide III and tyrosine contributions were present. The validity of wavelet approach in the data treatment has been also successfully tested on aspirin.

The role of a spatially varying absorptive inhomogeneity located at different depths within a turbid material has been investigated. This inhomogeneity has been characterized by a spatially dependent Gaussian distribution of its absorption coefficient. The present study has been performed calculating the time-resolved contrast function in the framework of the first-order perturbative approach to the diffusion equation for a slab geometry and a coaxial measurement scheme. The model has allowed us to take into account different locations of the inclusion along the source-detector axis. The accuracy of time-resolved contrast predictions has been analyzed through comparisons with results of the finite element method that has been used to numerically solve the diffusion equation. Recovery of the absorption perturbation parameter of the inhomogeneity for different axial positions has also been investigated.

Results on image quality parameters of a time-resolved imaging system are reported. Spatial resolution has been investigated by using the edge response function method (ERF) whereas a small absorbing inclusion hidden inside a turbid slab has been adopted to analyse the contrast and noise of images obtained by different reconstruction algorithms.

Cavities in human teeth were fabricated by means of Er:YAG laser and compared with samples processed by using conventional drill. The cavities of lased and unlased samples were cured with two different composite resins ( Bis-GMA vs PEX) to investigate differences in their adhesion properties. Morphological characterization of the sample surfaces by SEM analysis was performed. The same samples were then examined by micro-Raman spectroscopy by using He-Ne laser visible light excitation, in order to characterize the dentin/resin interfaces. The Raman spectra exhibited typical peaks of dentine. Resin contributions to the Raman spectra were also detected and have been used to monitor the interface properties of the boundary region. The dependence of the Raman spectrum on spatial position has been monitored. In both lased and unlased samples the PEX diffusion in dentine is limited in a region of order of 10 μm depth. A larger interdiffusion region is observed at the boundary of dentine and Bis-GMA resin.

Well-characterized phantom objects are necessary for investigating the performances of optical imaging systems based on time-resolved transmittance and fluorescence. For this purposes we have prepared inhomogeneous phantoms made of gelatinous objects placed in aqueous solutions of 10% Intralipid with different concentrations. The gelatinous objects have been prepared using a mixture of 10% Intralipid with agar at which absorbing ink have been added for transmittance based optical imaging system. For fluorescence measurements proper fluorescent dyes (rhodamine6G and IR125) have been added. Conventional optical characterization by spectrophotometric and spectrofluorimetric measurements have been performed. In addition, time-resolved transmittance and fluorescence measurements have been carried out. In particular, time-correlated single photon counting system has been used for time-resolved transmittance measurements. For time-resolved fluorescence measurements an optical imaging system based on a Ti:Sa laser and streak camera has been employed.

A detailed investigation on the use of time-resolved transmittance for the optical characterization of scattering media in which well-controlled quantities of absorbing ink were added has been performed. Particular attention has been devoted to analytical solutions to the diffusion equation and to other technical details used for fitting procedures in order to recover expected values for scattering and absorption coefficients.

We report two-photon absorption photoluminescence excitation (TPA-PLE) measurements in ZnSe/ZnSSe strained-layer superlattices (SLS). Polarization-dependent selection rules are observed in TPA spectra depending on the direction of the exciting field with respect to the growth axis of the SLS. Experimental exciton transition energies have found to be in good agreement with theoretical predictions considering strain and confinement effects. Furthermore by means of comparative non-linear technique the TPA coefficient has been measured at different energies and compared with theoretical predictions including quasi-bidimensional excitonic effects. A satisfactory agreement has been shown for the overall spectral behaviour and in some cases also for the TPA coefficient values.

A detailed investigation of the use of time-resolved trasmittance for the optical characterization of scattering media by use of different analytical solutions to the diffusion equation has been performed. A femtosecond Ti:sapphire laser working at 800 nm and a streak camera with a time resolution of a few picoseconds were employed. Different latex and Intralipid solutions as well as biological samples were investigated. Reduced scattering coefficients were evaluated, and good agreement with the Mie predictions was found. An estimation of the order of magnitude of the absorption coefficient was obtained for the low-absorbance samples examined. These studies confirm experimentally that time-resolved trasmittance can be employed usefully for evaluating μs′ values of thick scattering samples when a proper theoretical description that takes into account realistic boundary conditions is used.

In order to study the thermal behaviour of sintered carbide inserts during interrupted cutting, thermal cycles have been carried out by using a CO2 high-power laser with different conditions to simulate the actual cutting stress. It has been pointed out that the use of a laser is adequate for this purpose and that, even should the thermal fractures themselves not cause the collapse of the inserts, the thermal actions alone will be responsible for the growth of microfractures which weight heavily in predicting tool life.

An evaluation of coating performances in feedback controlled surface temperature laser heat treating has been made in terms of a simple energy balance model, by using a 500 W c.w. CO2 laser system able to perform such a kind of heat treating. Three different types of coating (manganese phosphate, titanium dioxide and colloidal graphite) have been investigated. The experimental results show that very different maximum processing speeds are ensured by each coating, in spite of their quite similar percentage of absorbed laser power. This more complete characterisation can be used to improve the performances of the aforesaid technique, avoiding any failure in controlled surface temperature laser heat treating.

Some predictions on the hardness and hardening depths on laser heat treatment of steels can be obtained when specific characteristics of both laser processes (heating and cooling rates) and laser heat treated steels (microhardness profiles) are taken into account. Some controlled surface temperature laser heat treatments have been carried out with a medium power c.w. CO 2 laser on a medium carbon steel (AISI/SAE1045), allowing these predictions to be tested. In particular, knowing the surface temperature has enabled an analytical algorithm to be used to describe thermal processes and a simple exponential expression to be employed to carefully predict the hardened case depth.

A method based on the control of surface temperature has been successfully applied to laser hardening of steels. In this way, a very simple approximation is used to predict the hardened depth and deeper treatments are obtained. In addition, this apparatus has allowed a complete characterization of the coatings generally used, thus making the heat treating process independent of their properties. Hence, performing heat treatments at a high automation level is possible.

A quantitative theory giving a unified view of certain laser applications was developed. The analysis and experimental procedure of a controlled temperature laser heat treating process are reported. Key features are the ease of predicting working results and the capability of getting on-line process diagnostics for use in a highly automated industrial environment.
