Rapid diagnosis by using small, simple, and portable devices could represent one of the best strategies to limit the damage and contain the spread of viral, bacterial or protozoa diseases, principally when they can be transmitted by air and are highly contagious, as some respiratory viruses are. The presence of antibodies in blood or serum samples is not the best option for deciding when a person must be quarantined to stop transmission of disease, given that cured patients have antibodies, so the best diagnosis methods rely on the use of nucleic acid amplification procedures. Here we present a very simple device and detection principle, based on paper discs coupled to contactless conductivity (C4D) sensors, can provide fast and easy diagnostics that are needed when an epidemic outbreak develops. The paper device presented here solves one of the main drawbacks that nucleic acid amplification tests have when they are performed outside of central laboratories. As the device is sealed before amplification and integrally disposed in this way, amplimers release cannot occur, allowing repetitive testing in the physician’s practice, ambulances, or other places that are not prepared to avoid cross-contamination of new samples. The use of very low volume samples allows efficient reagent use and the development of low cost, simple, and disposable point-of-care diagnostic systems., Author summary In 2005, the World Health Organization (WHO) recognized Chagas Disease as a neglected tropical disease. Meanwhile the serological tests, recommended by WHO, can be performed for chronic disease diagnosis, the nucleic acid amplification tests must be performed for the detection of the acute phase of the disease. Although the existing laboratory diagnosis tests for Chagas Disease are sensitive and highly reproducible, they cannot be performed in rural, low infrastructure environments, where this disease prevails. In this sense, the use of simple and portable analytical devices may be able to offer an affordable solution to this problem, allowing fast sampling, diagnosis and treatment prescription in one simple and fast intervention, as the performed by short term medical missions. In this study we show for the first time a diagnosis test comprising low cost materials and employing a contactless and label-free conductivity detection system that is used to read the result of a nucleic acid amplification reaction. The test showed high sensitivity for Chagas Disease diagnosis showing the potential to be used in rural and low income places.

International audience; Mosquito control is implemented when arboviruses are detected in patients or in field-collected mosquitoes. However, mass screening of mosquitoes is usually laborious and expensive, requiring specialized expertise and equipment. Detection of virus in mosquito saliva using honey-impregnated filter papers seems to be a promising method as it is non-destructive and allows monitoring the viral excretion dynamics over time from the same mosquito. Here we test the use of filter papers to detect chikungunya virus in mosquito saliva in laboratory conditions, before proposing this method in large-scale mosquito surveillance programs. We found that 0.9 cm 2 cards impregnated with a 50% honey solution could replace the forced salivation technique as they offered a viral RNA detection until 7 days after oral infection of Aedes aegypti and Aedes albopictus mosquitoes with CHIKV.

Many microbial phenotypes are differentially or exclusively expressed on agar surfaces, including biofilms, motility, and sociality. However, agar-based assays are limited by their low throughput, which increases costs, lab waste, space requirements, and the time required to conduct experiments. Here, we demonstrate the use of wax-printed microfluidic paper-based analytical devices (μPADs) to measure linear growth rate of microbes on an agar growth media as a means of circumventing the aforementioned limitations. The main production materials of the proposed μPAD design are a wax printer, filter paper, and empty pipet boxes. A single wax-printed μPAD allowing 8 independent, agar-grown colonies costs $0.07, compared to $0.20 and $9.37 for the same number of replicates on traditional microtiter/spectrophotometry and Petri dish assays, respectively. We optimized the μPAD design for channel width (3 mm), agar volume (780 μL/channel), and microbe inoculation method (razor-blade). Comparative analyses of the traditional and proposed μPAD methods for measuring growth rate of nonmotile (Saccharomyces cerevisiae) and motile (flagellated Escherichia coli) microorganisms suggested the μPAD assays conferred a comparable degree of accuracy and reliability to growth rate measurements as their traditional counterparts. We substantiated this claim with strong, positive correlations between the traditional and μPAD assay, a significant nonzero slope in the model relating the two assays, a nonsignificant difference between the relative standard errors of the two techniques, and an analysis of inter-device reliability. Therefore, μPAD designs merit consideration for the development of enhanced-throughput, low-cost microbial growth and motility assays.

Thermostable cellulases have wide variety of applications and distinctive advantages, but their low titer becomes the hurdle in their commercialization. In the present work, an assessment of optimum levels of significant factors (temperature, moisture ratio, inoculum size, and ammonium sulfate) and the effect of their interactions on production of thermostable CMCase, FPase, and β-glucosidase by Thermoascus aurantiacus RCKK under solid-state fermentation (SSF) was carried out using central composite design (CCD) of response surface methodology (RSM). The study revealed 33, 13, and 8 % improvement in FPase, CMCase, and β-glucosidase production, respectively. Moreover, crude cellulase from T. aurantiacus RCKK efficiently hydrolyzed office waste paper, algal pulp (Gracillaria verulosa), and biologically treated wheat straw at 60 °C with sugar release of about 830 mg/ml, 285 mg/g, and 260 mg/g of the substrate, respectively. The thermostable enzyme from T. aurantiacus RCKK holds potential to be used in biofuel industry.

Background Reduced susceptibility of mosquito vectors to currently used insecticides hampers control interventions. Recently pyriproxyfen, an insect growth regulator has been demonstrated to effectively reduce the reproductive potential in vector mosquitoes. Methods Pyriproxyfen (PPF), in different concentrations (0.75%, 0.075%, 0.0075%, and 0.00075%) was applied on papers and Indian wild type Aedes aegypti female mosquitoes (N ≥ 20 for each treatment) were exposed onto it as per WHO guidelines, to study the reproductive disruption. PPF concentration on treated papers was quantitatively cross-determined using HPLC method. Reduction in fecundity, fertility and adult emergence in exposed female Ae. aegypti was determined. Abnormal development in ovary and eggs of exposed females was studied microscopically after different time intervals. Results Eggs laid, eggs hatched, pupae formed and adults emerged per female exposed in both before blood meal and after blood meal groups declined significantly from lowest to highest concentration of PPF (F ≥ 5.2; p < 0.02). Adult emergence inhibition in females exposed to PPF before and after blood meal groups ranged from 58.8% [OR = 0.18 (95% CI = 0.09–0.36)] to 79.2% [OR = 0.04 (95% CI = 0.02–0.10)] and 64.4% [OR = 0.12 (95% CI = 0.05–0.28)] to 77.1% [OR = 0.05 (95% CI = 0.02–0.14)] respectively in different concentrations. The probit model used suggested that FI50 (50% fertility inhibition) and EI50 (50% emergence inhibition) were 0.002% (p = 0.82) and 0.0001% (p = 0.99) for females exposed before blood meal, while 0.01% (p = 0.63) and, Author summary Development of resistance against insecticides has challenged mosquito control programmes globally and prompted the research of alternative options that can complement insecticides. An insect growth regulator, pyriproxyfen (PPF) usage against mosquitoes can effectively reduce the vector population. PPF mainly inhibits the metamorphosis of mosquito larvae into pupae and prevent the adult emergence, therefore it is generally applied in mosquito breeding habitats. PPF has been shown to exhibit delayed residual effect that may impair the reproductive capacity by affecting the survival, fecundity and fertility of adult mosquito exposed through tarsal contact. Presently, the effectiveness of different concentrations of PPF intended to be delivered through contact have been evaluated against dengue vector Ae. aegypti. Results suggested that very low PPF concentration treated surfaces drastically reduce the fecundity, fertility and adult emergence in mosquitoes. Study suggests that control interventions based on PPF-treated surfaces could provide an additional route to target mosquito vector control by overall population density reduction.

We describe herein a method of recharging used commercial spin columns or assembling homemade spin columns using filter paper as binding material for cost-effective, low throughput nucleic acid purification. The efficiency of filter paper-based spin columns was evaluated for purification of nucleic acids from various sources. Following protocols of commercial kits, we found filter paper to be a useful binding material for purification of nucleic acids, including plant genomic DNA, plant total RNA, PCR products, and DNA from agarose gels. However, filter paper has a weak binding affinity to plasmid DNA in tested miniprep protocols. Protocols for the use of filter paper recharged spin columns or homemade spin columns for low throughput purification of plant genomic DNA and total RNA with unused commercial kit buffers or less expensive homemade buffers are presented.

Ricin is a highly-toxic compound derived from castor plant beans. Several incidents involving contamination of residences and buildings due to ricin production or dissemination have occurred in recent years. The goal of this study was to determine whether ricin bioactivity could be attenuated in reasonable time via simple modifications of the indoor environment. Attenuation was assessed on six different materials as a function of temperature, relative humidity (RH), and contact time, using both a pure and crude preparation of the toxin. Ricin bioactivity was quantified via a cytotoxicity assay, and attenuation determined as the difference in ricin recovered from test and positive controls. The results showed that pure ricin could be attenuated successfully, while the crude ricin was generally more persistent and results more variable. We found no significant attenuation in crude ricin after two weeks at typical indoor environmental conditions, except on steel. Attenuation mostly improved with increasing temperature, but the effect of RH varied. For pure ricin, heat treatments at 40°C for 5 days or 50°C for 2–3 days achieved greater than 96% attenuation on steel. In contrast, appreciable recovery of the crude ricin preparation still occurred at 40°C after two weeks.

This study characterized the microfauna community during treatment of pulp and paper wastewater in a UNOX system aerated with pure oxygen, and with a high organic loading rate (0.4±0.06kg BOD/kg MLSSday), low sludge retention time (3.73±0.33 day), and high oxygen concentration (≤20mg O2/L) in comparison to municipal treatment systems. In the aeration tank, temperatures were high, averaging 35.7°C (March-May), then 38.9°C (June-August). Effluent quality was acceptable: 180±22mg COD/L, 7.2±2.1mg BOD5/L, and 33±5mg TSS/L. Background/Aims: The nucleo-cytoplasmic large DNA viruses (NCLDV) constitute an apparently monophyletic group that consists of 6 families of viruses infecting a broad variety of eukaryotes. A comprehensive genome comparison and maximum-likelihood reconstruction of NCLDV evolution reveal a set of approximately 50 conserved genes that can be tentatively mapped to the genome of the common ancestor of this class of eukaryotic viruses. We address the origins and evolution of NCLDV. Results: Phylogenetic analysis indicates that some of the major clades of NCLDV infect diverse animals and protists, suggestive of early radiation of the NCLDV, possibly concomitant with eukaryogenesis. The core NCLDV genes seem to have originated from different sources including homologous genes of bacteriophages, bacteria and eukaryotes. These observations are compatible with a scenario of the origin of the NCLDV at an early stage of the evolution of eukaryotes through extensive mixing of genes from widely different genomes. Conclusions: The common ancestor of the NCLDV probably evolved from a bacteriophage as a result of recruitment of numerous eukaryotic and some bacterial genes, and concomitant loss of the majority of phage genes except for a small core of genes coding for proteins essential for virus genome replication and virion formation.

13C NMR analyses of hydrophobic dissolved organic matter (DOM) fractions isolated from a landfill leachate contaminated groundwater near Norman, OK; the Colorado River aqueduct near Los Angeles, CA; Anaheim Lake, an infiltration basin for the Santa Ana River in Orange County, CA; and groundwater from the Tomago Sand Beds, near Sydney, Australia, found branched methyl groups and quaternary aliphatic carbon structures that are indicative of terpenoid hydrocarbon precursors. Significant amounts of lignin precursors, commonly postulated to be the major source of DOM, were found only in trace quantities by thermochemolysis/gas chromatography/mass spectrometry of the Norman Landfill and Tomago Sand Bed hydrophobic DOM fractions. Electrospray/tandem mass spectrometry of the Tomago Sand Bed hydrophobic acid DOM found an ion series differing by 14 daltons, which is indicative of aliphatic and aryl-aliphatic polycarboxylic acids. The product obtained from ozonation of the resin acid, abietic acid, gave a similar ion series. Terpenoid precursors of DOM are postulated to be derived from resin acid paper sizing agents in the Norman Landfill, algal and bacterial terpenoids in the Colorado River and Anaheim Lake, and terrestrial plant terpenoids in the Tomago Sand Beds.

The aim of this study was to investigate the acute toxicity of pulp-paper industry wastewater using traditional and enrichment toxicity tests and to emphasize the importance of toxicity tests in wastewater discharge regulations. Enrichment toxicity tests are novel applications and give an idea of whether there is potential toxicity or growth-limiting and -stimulating conditions. Different organisms were used such as bacteria (floc and coliform bacteria), algae (Chlorella sp.), protozoa (Vorticella sp.), and fish (Lepistes sp.) to represent four trophic levels. Furthermore, chemical oxygen demand (COD) fractionation results were compared with these tests to assess the effect of COD subcategories on the determination of possible toxicity. The pulp-paper industry results revealed acute toxicity to at least two organisms in 6 of 20 effluent samples. The toxicity test results were assessed with chemical analyses such as COD, biochemical oxygen demand (BOD), color, absorbable organic halogen (AOXs), and phenol. It was observed that the toxicity of the effluents could not be explained by using physicochemical analyses in four cases for the pulp-paper industry. In this study, the ability of algae to treat a wood-based pulp and paper industry wastewater was investigated. Tests were performed in batch reactors seeded with a mixed culture of algae. Under different lighting and initial wastewater strength conditions, changes in COD, AOX and color contents of reactors were followed with time. Algae were found to remove up to 58% of COD, 84% of color and 80% of AOX from pulp and paper industry wastewaters. No remarkable differences were observed in COD and color when light intensity and wastewater strength were changed, while AOX removals were strongly affected. Algal species identification studies revealed that some green algae (Chlorella) and diatom species were dominant in the treatment. The study also showed that algae grew mixotrophically, while the main mechanism of color and organics removal from pulping effluents was partly metabolism and partly metabolic conversion of colored and chlorinated molecules to non-colored and non-chlorinated molecules. Adsorption onto algal biomass was not so effective.

Although coastal eutrophication is generally recognised as a recent phenomenon related to the well-documented increase in riverine nutrient delivery during the last 30 or 40 years, a few historical records paradoxically show that, in some places like the Southern Bight of the North Sea, or the Northern Adriatic, algal proliferation as intense as presently observed was already regularly occurring at the end of the 19th century. Estimated riverine nutrient loads from diffuse sources or from domestic point sources of waste water at that time are too low to account for these observations. We attempted a retrospective evaluation of the possible contribution of industrial activity to nutrient river loading. The figures indicate that, by the end of the last century, large scale use of traditional processes in textile and paper industries, in tanneries, candles factories and others was responsible for a dominant part of the nutrient load carried by rivers in Western Europe and could have caused nutrient inputs to coastal zones similar to the present ones.

The applicability of the Whole Effluent Assessment concept for the proof of compliance with the “best available techniques” has been analysed with paper mill wastewater from Germany by considering its persistency (P), potentially bio-accumulative substances (B) and toxicity (T). Twenty wastewater samples from 13 paper mills using different types of cellulose fibres as raw materials have been tested in DIN or ISO standardised bioassays: the algae, daphnia, luminescent bacteria, duckweed (Lemna), fish-egg and umu tests with lowest ineffective dilution (LID) as test result. The potentially bio-accumulative substances (PBS) were determined by solid-phase microextraction and referred to the reference compound 2,3-dimethylnaphthalene. Usually, a primary chemical–physical treatment of the wastewater was followed by a single or multi-stage biological treatment. One indirectly discharged wastewater sample was pre-treated biologically in the Zahn–Wellens test before determining its ecotoxicity. No toxicity or genotoxicity at all was detected in the acute daphnia and fish egg as well as the umu assay. In the luminescent bacteria test, moderate toxicity (up to LIDlb = 6) was observed. Wastewater of four paper mills demonstrated elevated or high algae toxicity (up to LIDA = 128), which was in line with the results of the Lemna test, which mostly was less sensitive than the algae test (up to LIDDW = 8). One indirectly discharged wastewater sample was biodegraded in the Zahn–Wellens test by 96% and was not toxic after this treatment. Low levels of PBS have been detected (median 3.27 mmol L−1). The colouration of the wastewater samples in the visible band did not correlate with algae toxicity and thus is not considered as its primary origin. Further analysis with a partial wastewater stream from thermomechanically produced groundwood pulp (TMP) revealed no algae or luminescent bacteria toxicity after pre-treatment of the sample in the Zahn–Wellens test (chemical oxygen demand elimination 85% in 7 days). Thus, the algae toxicity of the respective paper mill cannot be explained with the TMP partial stream; presumably other raw materials such as biocides might be the source of algae toxicity. Comparative data from wastewater surveillance of authorities confirmed the range of ecotoxicity observed in the study. Wastewater from paper mills generally has no or a moderate ecotoxicity (median LID 1 and 2) while the maximum LID values, especially for the algae and daphnia tests, are considerably elevated (LIDA up to 128, LIDD up to 48). Wastewater from paper mills generally is low to moderately ecotoxic to aquatic organisms in acute toxicity tests. Some samples show effects in the chronic algae growth inhibition test which cannot be explained exclusively with colouration of the samples. The origin of elevated algae ecotoxicity could not be determined. In the algae test, often flat dose–response relationships and growth promotion at higher dilution factors have been observed, indicating that several effects are overlapping. At least one bioassay should be included in routine wastewater control of paper mills because the paper manufacturing industry is among the most water consuming. Although the algae test was the most sensitive test, it might not be the most appropriate test because of the complex relationship of colouration and inhibition and the smooth dose–effect relationship or even promotion of algae growth often observed. The Lemna test would be a suitable method which also detects inhibitors of photosynthesis and is not disturbed by wastewater colouration.

Huge efforts have been made both in adopting more environmental-friendly bleaching processes, and in developing advanced oxidation processes and more effective biological treatments for the reduction of deleterious impacts of paper mill effluents. Even so, the success of such treatments is frequently reported in terms of chemical parameters without a proper evaluation of the effluent's toxicity mitigation. This is the first study reporting an exhaustive evaluation of the toxicity of a secondary bleached kraft pulp mill effluent, after either tertiary treatment with the soft-rot fungi Rhizopus oryzae or with a photo-Fenton oxidation, using a battery of freshwater species. As it has been reported the photo-Fenton/UV treatment has proved to be the most effective in reducing the colour and the COD (chemical oxygen demand) of the effluent. Nevertheless, extremely low EC 50 values were reported for almost all species, after this tertiary treatment. The treatment with R. oryzae was less effective in terms of colour removal and COD reduction, but proved to be the most promising in reducing toxicity.

The pulp and paper industry has invested heavily over recent years in cleaner processing, to reduce losses and minimise its impact on the environment. Over the past fifteen years, a New Zealand integrated bleached kraft mill has undergone a comprehensive programme of upgrades to increase production, reduce water consumption and streamline its biological treatment process. Whilst the overall discharge of contaminants from the site decreased, the treatment system performance did not show a concurrent improvement as may have been expected. Reduced BOD removal, low dissolved oxygen levels, and poor solids settlability were symptomatic of phosphorus limitation in the aerated lagoon treatment system. The wastewater entering the system was found to be phosphorus limited at a BOD:P ratio of 100:0.2. Mono-ammonium-phosphate was supplemented, at approximately 30 kg P/d, to raise the phosphorus levels to a BOD:P ratio of 100:0.3. Treatment efficiencies improved very quickly after phosphorus dosage, with a 50% reduction in BOD and TSS discharge, a significant increase in dissolved oxygen levels, and improved BOD removal (85% to 93%). This case study demonstrates that whilst more closed operation can result in reduced discharge of organic loads, there may be negative impacts on the availability of nutrients for balanced biological growth.

A total of 100 currency notes of various denominations in circulation were randomly studied for bacterial, fungal and protozoal contamination. All except four notes yielded one or more bacteria. Bacterial culture yielded single isolate in 33 notes, two in 44 notes, three in 12 notes and four in 7 notes. The predominant bacterial isolate was Bacillus sps followed by Coagulase negative Staphylococci and Micrococcus sps. Other bacteria that are either potential or confirmed pathogens included K. pneumoniae, E. coli, S. aureus, Pseudomonas sps and S. typhi. Only two notes were positive for Acid fast bacilli. 28 samples did not yield any fungal growth. Overall 118 fungal isolates were isolated, of which 34 could not be identified. All the fungi isolated were saprophytes. Saline and Iodine wet mount did not reveal any parasitic forms. We recommend that currency notes must be handled with caution.

Freshwater algal biomass and orange and lemon peels were assessed as tissue paper pulp supplements. Cellulose and hemicellulose contents of algal biomass were 7.1% and 16.3%, respectively, whereas for citrus peels cellulose content ranged from 12.7% to 13.6% and hemicellulose from 5.3% to 6.1%. For all materials, lignin and ash content was 2% or lower, rendering them suitable for use as paper pulp supplements. The addition of algal biomass to paper pulp increased its mechanical strength significantly. However, brightness was adversely affected by chlorophyll. The addition of citrus peels in paper pulp had no effect on breaking length, increased bursting strength and decreased tearing resistance. Brightness was negatively affected at proportions of 10%, because citrus peel particles behave as coloured pigments. The cost of both materials is about 45% lower than that of conventional pulp, resulting in a 0.9-4.5% reduction in final paper price upon their addition to the pulp.

Spirotox is a short-term acute toxicity test with a large ciliated protozoan Spirostomum ambiguum. The test was created in the Department of Environmental Health Sciences, Medical University of Warsaw, Poland, during the early 1990s. It was presented for the first time during the 6th International Symposium on Toxicity Assessment, in Berlin in 1993. S. ambiguum was very resistant to a wide range of environmental conditions, especially to pH and dissolved oxygen. Over the last 10 years the sensitivity of S. ambiguum to many classes of toxicants was evaluated. Spirotox was found to be very sensitive to heavy metals, fungicides, and pharmaceuticals used to cure diseases of the human nervous system. On the other hand, it was generally less sensitive than standard bioassays to simple organics. Spirotox was also used for analysis of cyanobacterial blooms. Though it was moderately sensitive to hepatotoxins, the test seems to be a good tool for evaluation of the entire toxicity of blooms. The last but not the least of the applications of the Spirotox test was evaluation of the toxicity of extracts from medical devices. Protozoa, which are the simplest eukaryotes, seem to be good screening bioassays for monitoring the technology of medical device production.

Most studies focus on the ecotoxicity of pulp and paper mill effluents, rather than on how they affect the physicochemical and biological structure and the intrinsic ecological capabilities of the receiving watercourses. We investigated the impact of such effluents on the water quality, microplankton system and microbial self-purification capacity (degradation of polymeric organic compounds via extracellular enzymes) of the Biobio River in Chile. The physicochemical impact on the water quality was indicated by raised conductivity, by the pollution of the water body with nitrate, nitrite and soluble reactive phosphorus, by the appearance of tannin and lignin, and by the steady accumulation of inorganic and organic suspended matter (SPM) along the river. From the biological structure of the microplankton system, very low and declining concentrations of chlorophyll a and heterotrophic flagellate densities were determined. The pulp and paper mill effluents introduced high bacterial abundances and biomass concentrations into the river water. This reflects the effective use made of the abundantly available inorganic and organic nutrients within this industrial and municipal process water by bacteria adapted to these extreme environments, additionally supported by concomitant low grazing pressure derivable from low heterotrophic flagellate abundances. Indeed, in one section of the river affected by a pulp mill, the plant was found to significantly contribute to the self-cleaning capacity of the river. However, this elevated degradation capacity was not enough to compensate for the additionally discharged organic material which, together with the toxic effects of the paper plant effluents, significantly interferes with the ecological status of the Biobio River.

Soluble silicates, commercially known as waterglass, are among the largest volume synthetic chemicals in the world. Silicon from waterglass is rapidly transformed to the biologically active orthosilicic acid (referred to as dissolved silicate). We used mesocosms to examine the impact of different concentrations of pulp mill effluent (PME) on structural and functional endpoints of a benthic assemblage in the Saint John River (NB, Canada) during 1999 and 2000. Previous studies on this effluent's effects produced conflicting results, with field surveys suggesting a pattern of mild nutrient enrichment, while laboratory toxicity tests linked effluent exposure to moderate contaminant effects. Experimental treatments included three concentrations of sulfite pulp mill effluent (0, 5, 10% v/v PME). Endpoints for the assessment included algal biomass and taxonomic composition, benthic invertebrate abundance and composition, and insect emergence. Low concentrations of PME increased periphyton biomass and caused changes in community structure within the diatom-dominated community. Pulp mill effluent addition had little effect on several structural endpoints measured for benthic invertebrates, including abundance and taxonomic richness, but significantly changed community composition. For both periphyton and benthic invertebrates, community composition endpoints were more sensitive indicators of PME exposure. Insect emergence was a highly relevant functional endpoint. When benthic and emerged insects were combined, total abundance increased with PME addition. Results from two trophic levels, which provided multiple lines of evidence, indicated that the main impact of these PME concentrations is nutrient enrichment rather than effluent toxicity. Our findings also suggest that benthic invertebrate and periphyton assemblages, algal biomass production, and insect emergence are sensitive response measures. Future studies may confirm this observation. The consideration of both functional and structural endpoints at different trophic levels can greatly improve our understanding the effects of discharges to rivers. Such an understanding could not have been obtained using standard assessment techniques and illustrates the value of mesocosms and the benthic community assemblage approach in environmental assessment.

The effects of temperature variations on aerobic biological wastewater treatment were evaluated with respect to treatment efficiency, solids discharges, sludge physicochemical properties and microbiology. The effects of controlled temperature shifts (from 35 degrees to 45 degrees C; from 45 degrees to 35 degrees C) and periodic temperature oscillations (from 31.5 degrees C to 40 degrees C, 6-day period, for 30 days) were assessed in 4 parallel, lab-scale sequencing batch reactors (SBRs) that treated pulp and paper mill effluent. Overall, the temperature shifts caused higher effluent suspended solids (ESS) levels (25-100 mg/L) and a decrease (up to 20%) in the removal efficiencies of soluble chemical oxygen demand (SCOD). Lower ESS levels were triggered by a slow (2 degrees C/day) versus a fast (10 degrees C/12h) temperature shift from 35 degrees to 45 degrees C, but the SCOD removal efficiencies decreased similarly in both cases (from 66+/-3% and 65+/-2% to 49+/-3% and 51+/-3%). Temperature oscillations caused an increased deterioration of the sludge settleability [high sludge volume indices (SVI); low zone settling velocities (ZSV)], high ESS levels and lower SCOD removals. The temperature transients were associated with poor sludge settleability (SVI100 mL/g MLSS, ZSV1 cm/min), more negatively charged sludge (up to -0.35+/-0.03 meq/g MLSS), increased filament abundance (approximately 4 to 4.5, subjective scale equivalent to very common), and decreased concentrations of protozoa and metazoa (25,000-50,000 microorganisms/mL sludge). The controlled, periodic temperature oscillations had a slight impact on SCOD removal efficiency (5% decrease), and did not seem to select for robust microorganisms that withstood the temperature shift. Sludge deflocculation and filament proliferation caused by these temperature transients may explain the higher ESS levels.

The rate of technical development in bleaching of chemical pulp and the upgrading of process control and wastewater treatment systems in the pulp industry have been extremely rapid over the past few years. When assessing the environmental impacts of bleached kraft pulp mill effluents (BKMEs), it is therefore more important than ever to carefully characterize the bleaching process, the composition of the treated effluent, and the degree of exposure of sensitive target organisms in the receiving water body. These requirements have not always been fulfilled in previous reports on biological effects of BKMEs in Scandinavia. This work presents the results of a comprehensive field study of the impacts of a 350,000-tonne kraft mill, bleaching softwood and hardwood pulp in campaigns according to the sequence O(D25,C70+D5)(EOP)D(EP)D. The effluent is treated in an aerated lagoon with a mean retention time of 8-9 days, practically eliminating chlorate and resin acids, and reducing the discharge of adsorbable organic halogen (AOX) to an average of 1 t/day (1.3 and 0.4 kg/ADt for softwood and hardwood, respectively). The treated effluent is discharged through a 1.3-km-long diffuser, at a water depth of 9-12 m, into a well ventilated coastal area, giving a 1000-fold dilution within 3-4 km from the diffuser. The actual exposure of the coastal ecosystem to BKME components was determined by analysis of extractable organic chlorine in suspended solids and of conjugates of chlorophenolics in the bile of feral perch. Despite a major damage to the benthic communities that occurred about 10 years ago, and was due to large chlorate discharges at the time, no direct detrimental effects on benthic flora and fauna could be ascribed to the present BKME discharge. Instead, a clear recovery of the Fucus community, although not yet completed, could be demonstrated. Studies of the composition, abundance and biomass of the fish community, the recruitment and survival of fish fry, and the physiological status of perch, using a set of biomarkers, revealed that even in the most BKME-exposed area, only minor effects were detected. These effects were related to eutrophication/enrichment rather than to the action of toxic substances. The general effect picture, thus, was essentially of a different type than the one recorded in previous studies of mills, which used older technology and less effective process and effluent treatment control, and which were discharging into enclosed, shallow bays of the Baltic Sea.

The effect of 22 technological waste water samples and of some standards was tested on bacteria, fungi, chlorococcal algae, flagellata, plant cells, cells of Tubifex tubifex, hamster cells V79 and the fish Lebistes reticulatus. Of these 22 samples, some inhibition of cell life processes was displayed by the black liquor formed in the production of paper pulp and viscose pulp, by the waste solution produced during the preparation of bleaching agents for paper pulp and viscose pulp, and by the residual liquor after hypochlorite treatment of paper pulp.

Different process wastewaters from a sulfite pulp mill with bleaching were characterized by chemical analysis and toxicity tests. The amount of adsorbable organically bound halogen (AOX) from the bleachery was 3.6 kg per ton pulp. The extractable organically bound chlorine was 15% of AOX. Some identified organochlorine compounds in the effluent could be traced in the receiving water. Effluents from the chlorination and alkaline extraction stages and the condensate were the main contributors to the effluent toxicity. The effluents were particularly toxic to the alga Skeletonema costatum. The EC50 value for growth of the alga was 24–29 ml/liter of the total effluent. The toxicity cannot be ascribed to single chemical components in the effluents. Degradation of toxic components occurs after dilution of the effluents in the receiving water. The toxicity may be reduced to 30–50% of the initial toxicity within 1 week. Predictions of toxic effects in the receiving water, based on results of toxicity tests and estimated dilution, indicate that large areas are affected by the discharges. Some observations of the distribution of organisms in the receiving water indicate that predictions from the toxicity tests may be valid.

Cysts of Polytomella parva and Polytomella caeca were recovered after 7 days storage at cryogenic temperatures following drying on shredded filter paper, silica gel or without added substrate. Accelerated storage testing, by exposing dried material to elevated temperatures, indicated that shredded filter paper was the best of the substrates tested. Polytomella parva was recovered after 5 years storage at -30 degrees C when dried on filter paper but not when dried on silic agel. Determinations of the number of cysts recovered indicated that viable cysts survived all conditions of storage tested. However, excystment following storage was delayed, the extent depending on storage conditions and the substrate used for drying. Most rapid recovery occurred when cysts were rehydrated immediately after drying, and after storage on filter paper at below -70 degrees C.
