Honeycomb-like porous chitosan (CS) films are attractive tools for developing functional materials for filters, catalyses, adsorbents, biomaterials, etc. A simple method for fabricating honeycomb-like porous CS films without special reagents, facilities, and techniques would make them accessible. Here we introduce an easily available method for fabricating honeycomb-like CS films without a strong acid/base, toxic reagents, or special facilities/techniques. An aqueous solution containing CS and poly(N-isopropylacrylamide) (PNIPAm) was allowed to stand at 25 °C to evaporate water. After 3 days, CS–PNIPAm composite films with homogenously phase-separated PNIPAm particles were obtained. The PNIPAm particles were removed by immersion in methanol, and the resulting films dried under reduced pressure to become honeycomb-like porous CS films. The pore size could be varied in the range of 0.5–3.0 μm by altering the CS concentration and the molecular weight of CS where the pore size was reduced under conditions with stronger interaction between CS molecules. We reveal that the key to success with this system is the decrease of lower critical solution temperature (LCST) of PNIPAm with water evaporation. In addition, we confirmed the removed PNIPAm was recyclable in this system. Furthermore, we found this method was also applicable to alginate. The proposed facile method for fabricating honeycomb-like porous polymeric films could provide various functional porous materials.

In this study, the effects of chitosan and surface-deacetylated chitin nanofibrils (SDACNFs) on hair growth were evaluated. In human follicle dermal papilla cells in vitro, chitosan and SDACNFs were shown to increase cell growth on day 3 after the initiation of treatment, together with an increase in the production of fibroblast growth factor-7 (FGF-7) by these cells on day 3. Furthermore, in an in vivo study in mice, chitosan and SDACNF application promoted hair growth. The number of anagen follicles significantly increased compared with that in the control group, whereas the number of telogen follicles significantly decreased in the chitosan and SDACNF groups. In the chitosan and SDACNFs groups, moreover, the expression levels of FGF-7 and Sonic hedgehog were significantly upregulated in hair follicles. Overall, our results demonstrated that chitosan and SDACNFs promoted hair growth and therefore may have applications as novel therapeutic agents for the treatment of hair loss in patients.

Surface wrinkling to fabricate hierarchical surface topographies has attracted much attention because of the potential and multifunctional applications of hierarchical surface wrinkles beyond uniform wrinkles. Although many reports have described the preparation of hierarchical wrinkles induced by mechanical stress and heat, fabrication through drying-induced shrinkage has hardly been reported. Here we introduce hierarchical surface wrinkles and bumps generated on a chitosan film via the preparation of double-skin layers with κ- and ι-carrageenans, respectively, and subsequent drying. Double-skin layers are fabricated on a swollen chitosan film, called a chitosan hydrogel film, that is soaked first in κ- or ι-carrageenan solution and then in water to remove excess adsorbed κ- or ι-carrageenan. After the film is dried, hierarchical microscopic surface architectures are observed. In the case of the κ-carrageenan system, the wrinkles are hierarchical, consisting of wrinkles (6.2 ± 2.8 μm) that have smaller buckles (0.23 ± 0.09 μm). We reveal that the wrinkles or the smaller buckles are caused by plane inhomogeneous shrinkage between the κ-carrageenan layer and the chitosan film or by the aggregation of the κ-carrageenan layer upon drying, respectively. Interestingly, the ι-carrageenan system showed hierarchical bumps consisting of semispherical bumps (5.6 ± 2.1 μm) that have smaller bumps (0.78 ± 0.27 μm). We reveal that the larger bumps are generated during the immersion of the chitosan hydrogel film into ι-carrageenan solution. The smaller bumps are generated by the aggregation of the ι-carrageenan layer that occurs during drying; this process requires the plane compression strain caused by the shrinkage of the chitosan hydrogel film.

Chitin nanofibers were prepared from crab shell for the purpose of utilizing crab shells. After the series chemical treatment and wet pulverization treatment, a uniform fibrous substance having a width of about 10 nm was obtained. The reason why fine fibrous chitin can be obtained is the structure of the crab shell. A characteristic of chitin nanofibers is their high dispersibility in water. Therefore, processing ability is improved, and biological properties of the nanofibers can be evaluated. Chitin nanofibers whose surface is modified to chitosan can be obtained after treatment with a relatively medium concentration of sodium hydroxide. Since chitosan nanofibers have an amino group on the surface, they are positively charged in an acidic aqueous solution. Chitin and chitosan nanofibers have various physiological functions when taken or applied to the skin. Effects of oral ingestion of chitosan nanofibers on chronic kidney disease (CKD) model, non-alcoholic steatohepatitis model, and inflammatory bowel disease model were studied. Furthermore, the biological effects of chitin and chitosan nanofibers for the skin are also investigated.

Background/Aim: Chitosan-coated iron oxide nanoparticles (Chi-NP) have gained attention because of their biocompatibility, biodegradability, low toxicity and targetability under magnetic field. In this study, we investigated various biological properties of Chi-NP. Materials and Methods: Chi-NP was prepared by mixing magnetic NP with chitosan FL-80. Particle size was determined by scanning and transmission electron microscopes, cell viability by MTT assay, cell cycle distribution by cell sorter, synergism with anticancer drugs by combination index, PGE(2) production in human gingival fibroblast was assayed by ELISA. Results: The synthetic process of Chi-NP from FL-80 and magnetic NP increased the affinity to cells, up to the level attained by nanofibers. Upon contact with the culture medium, Chi-NP instantly formed aggregates and interfered with intracellular uptake. Aggregated Chi-NP did not show cytotoxicity, synergism with anticancer drugs, induce apoptosis (accumulation of subG1 cell population), protect the cells from X-ray-induced damage, nor affected both basal and IL-1β-induced PGE(2) production. Conclusion: Chi-NP is biologically inert and shows high affinity to cells, further confirming its superiority as a scaffold for drug delivery.

Sorbitol polyglycidyl ether (SPE), which is a bio-based water-soluble epoxy resin, was cured with lysine (Lys) and arginine (Arg) as basic amino acids and with cysteine (Cys) as an acidic amino acid. Furthermore, biocomposites of the Lys- and Arg-cured SPE (SPE-Lys and SPE-Arg) and Cys-cured SPE (SPE-Cys) with chitin nanofiber (ChNF) not higher than 10 wt% were successfully prepared by compression molding and casting methods, respectively. Also, SPE-Cys biocomposites with chitosan nanofiber (CsNF) not higher than 2 wt% were successfully prepared by the casting method. The FT-IR spectral analysis of the cured resins suggested that the e-amino and carboxy groups of Lys, α-amino and carboxy groups of Arg, thiol group of Cys mainly reacted with epoxy groups of SPE. The FT-IR spectral analysis of the ChNF biocomposites suggested that a side reaction of SPE with acetic acid which was contained in ChNF suspension occurred. The dynamic mechanical analysis revealed that a higher order of the loss modulus peak temperature (Tα) was SPE-Lys (39 °C) > SPE-Arg (33 °C) > SPE-Cys (21 °C). Although Tαs of ChNF biocomposites were lower than those of the corresponding cured resins, Tαs of SPE-Cys/CsNF biocomposites were higher than that of SPE-Cys by the action of CsNF as a basic catalyst. The storage moduli over the range of 50–100 °C for SPE-Lys/ChNF, SPE-Cys/ChNF and SPE-Cys/CsNF biocomposites were much higher than those for the corresponding cured resins. SPE-Lys/ChNF and SPE-Cys/CsNF biocomposites exhibited higher tensile strengths and moduli than SPE-Lys and SPE-Cys did, respectively.

We previously developed bio-based wrinkled surfaces induced by wood-mimetic skins upon drying in which microscopic wrinkles were fabricated on a chitosan (CS) film by immersing it in a phenolic acid solution, followed by horseradish peroxidase (HRP)-catalyzed surface reaction and drying. However, the detailed structure of the resulting wood-mimetic skins, including crosslinking mode and thickness, has not been clarified due to the difficulty of the analysis. Here, we prepare wrinkled films using ferulic acid (FE), vanillic acid (VA), and homovanillic acid (HO) and characterize their structures to clarify the unknown characteristics of wood-mimetic skin. Chemical and structural analyses of wood-mimetic skins prepared using VA and HO indicate that the crosslinking structure in the skin is composed of ionic bonds between CS and an oligophenolic residue generated by the HRP-catalyzed reaction on the CS surface. Moreover, the quantity of these ionic bonds is related to the skin hardness and wrinkle size. Finally, SEM and TOF-SIMS analyses indicate that the skin thickness is on the submicron order (

The multifunctional beneficial properties of chitosan in biomedical applications have been recognized in recent years. A major focus of these studies concerned the antioxidant properties of chitosan and its derivatives. In patients and in model rats with chronic renal failure (CRF), the application of chitosan demonstrated high antioxidant activity as well as renoprotective effects. Ingestion of surface-deacetylated chitin (SDAC) nanofibers by CRF rats resulted in a significant decrease in the levels of uremic toxins and oxidative stress, when compared with SDAC. The preparation of elastic gels SDACNFs/ sulfobutyl ether β-cyclodextrin (SBE-β-CD), starting with SDACNFs nanofibers with SBE-β-CD and containing prednisolone proved useful in the study of ulcerous colitis as an experimental model of another disease related to oxidative stress. This technique proved to be an efficient strategy for studies of the effects of chitosan and derivatives, such as SDACNF, in diseases linked to oxidative stress.

In order to synthesize a promising material for developing a novel peptide/protein delivery system, guanidinylation of chitooligosaccharides with 1-amidinopyrazole hydrochloride was investigated herein. The production of guanidinylated chitooligosaccharides was demonstrated by infrared spectroscopy (IR), nuclear magnetic resonance (NMR), and elemental analyses. Interestingly, we found that the reducing end in the guanidinylated chitooligosaccharides was converted to a cyclic guanidine structure (2-[(aminoiminomethyl)amino]-2-deoxy-d-glucose structure). This reaction was carefully proven by the guanidinylation of d-glucosamine. Although this is not the first report on the synthesis of the 2-[(aminoiminomethyl)amino]-2-deoxy-d-glucose, it has provided a rational synthetic route using the high reactivity of the reducing end. Furthermore, we found that the interaction between chitooligosaccharides and bovine serum albumin is weak when in a neutral pH environment; however, it is significantly improved by guanidinylation. The guanidinylated chitooligosaccharides are useful not only for the development of a novel drug delivery system but also as a chitinase/chitosanase inhibitor and an antibacterial agent.

Chitin, an N-acetylglucosamine polymer, is well-known to have unique biological functions, such as growth promotion and disease resistance induction in plants. Chitin has been expectedly used for improving crop yield using its functions, however, chitin derivatives, such as chitin oligosaccharide (CO) and chitosan, are widely used instead since chitin is difficult to handle because of its insolubility. Chitin nanofiber (CNF), produced from chitin through nanofibrillation, retains its polymeric structure and can be dispersed uniformly even in water. Here, the effects of CO and CNF on plant responses were directly compared in soybeans (Glycine max) to define the most effective method to produce chitin derivatives for plant response induction. The growth promotion of aerial parts was observed only in CNF-treated plants. The transcriptome analysis showed that the number of differentially expressed genes (DEGs) in CNF-treated soybeans was higher than in CO-treated soybeans. Notably, the expression patterns of DEGs were mostly similar but were strongly induced by CNF treatment as compared with the CO group. These results reveal that CNF can induce stronger plant response to chitin than CO in soybeans, suggesting nanofibrillation, rather than oligomerization, as a more effective method to produce chitin derivatives for plant response induction.

Chitosan produced by the deacetylation of chitin is a cationic polymer with antimicrobial properties. In this study, we demonstrate the improvement of chitosan properties by nanofibrillation. Nanofiber sheets were prepared from nanofibrillated chitosan under neutral conditions. The Young’s modulus and tensile strength of the chitosan NF sheets were higher than those of the chitosan sheets prepared from dissolving chitosan in acetic acid. The chitosan NF sheets showed strong mycelial growth inhibition against dermatophytes Microsporum and Trichophyton. Moreover, the chitosan NF sheets exhibited resistance to degradation by the fungi, suggesting potentials long-lasting usage. In addition, surface-deacetylated chitin nanofiber (SDCNF) sheets were prepared. The SDCNF sheet had a high Young’s modulus and tensile strength and showed antifungal activity to dermatophytes. These data indicate that nanofibrillation improved the properties of chitosan. Thus, chitosan NF and SDCNF sheets are useful candidates for antimicrobial materials.

A facile method of preparing cyclodextrin-grafted chitosan (CyD-g-CS) by means of the carboxymethlation of cyclodextrins and the dehydration-condensation reaction to chitosan was developed. In this method, crude product containing ~10% of the carboxymethylated-cyclodextrin was subjected to the grafting reaction without further purification. The product (CyD-g-CS) was finally purified by dialysis. Furthermore, doxorubicin-capturing nanoparticles composed of CyD-g-CS and sodium triphosphate were prepared.

This study evaluated the effects of oral administration of surface-deacetylated chitin nanofibers (SDACNFs) on hypercholesterolemia using an experimental model. All rats were fed a high cholesterol diet with 1% w/w cholesterol and 0.5% w/w cholic acid for 28 days. Rats were divided equally into four groups: the control group was administered 0.05% acetic acid dissolved in tap water, and the SDACNF, chitosan (CS), and cellulose nanofiber (CLNF) groups were administered 0.1% CNF, CS, or CLNF dissolved in the tap water, respectively, during the experimental period. Changes in body weight, intake of food and water, and organ weight were measured. Serum blood chemistry and histopathological examination of the liver were performed. Administration of SDACNF did not affect body weight change, food and water intake, or organ weights. Administration of SDACNF and CS decreased the diet-induced increase in serum total cholesterol, chylomicron, very-low-density lipoprotein, and phospholipid levels on day 14. Moreover, oral administration of SDACNFs suppressed the increase of alanine transaminase levels on day 29 and suppressed vacuolar degeneration and accumulation of lipid droplets in liver tissue. These data indicate that SDACNF has potential as a functional food for patients with hypercholesterolemia.

Chitin (β-(1-4)-poly-N-acetyl-D-glucosamine) is widely distributed in nature and is the second most abundant polysaccharide after cellulose. It is often converted to its more deacetylated derivative, chitosan. Previously, many reports have indicated the accelerating effects of chitin, chitosan, and its derivatives on wound healing. More recently, chemically modified or nano-fibrous chitin and chitosan have been developed, and their effects on wound healing have been evaluated. In this review, the studies on the wound-healing effects of chitin, chitosan, and its derivatives are summarized. Moreover, the development of adhesive-based chitin and chitosan are also described. The evidence indicates that chitin, chitosan, and its derivatives are beneficial for the wound healing process. More recently, it is also indicate that some nano-based materials from chitin and chitosan are beneficial than chitin and chitosan for wound healing. Clinical applications of nano-based chitin and chitosan are also expected.

Chitosan nanofibers were easily prepared from fully deacetylated chitosan dry powder using a high-pressure waterjet system. From SEM observation, after 10 cycles of treatment, most of the chitosan had been reduced to homogeneous nanofibers measuring tens of nanometers. On the other hand, further mechanical treatment did not show a significant change. Relative crystallinity of chitosan nanofibers gradually decreased as the number of passes increased since high-pressure waterjet treatment damaged the crystalline region of chitosan nanofibers. The transmittance of the chitosan nanofiber slurry increased steeply, as the number of passes increased, indicating that the chitosan fibers were disintegrated effectively. Viscosity of chitosan nanofiber slurry also showed that the chitosan disintegrated well into nanofibers up to 10 passes. Above 10 passes, disintegration efficiency was saturated. The molecular weights of the nanofibers steeply decreased due to the depolymerization of chitosan by mechanical disintegration. The Young's modulus and tensile strength of chitosan nanofiber sheets were improved as the number of treatments increased, but further treatments deteriorated the tensile strength.

Surface-deacetylated chitin nanofiber reinforced chitosan films were prepared. The nano-composite films were highly transparent of approximately 84% at 600 nm due to the nanometer-sized fillers and chitosan matrix, which were embedded in the cavities and on the rough surface of the nanofiber networks. Due to the extended crystalline structure, the nanofibers worked effectively as reinforcement filler to improve the Young's modulus and the tensile strength of the chitosan film. After 10% blending of nanofiber, these properties were increased by 65% and 94%, respectively. Moreover, thermal expansion was also significantly decreased from 35.3 to 26.1 ppm K(-1) after 10% addition of nanofibers. Surface-deacetylated chitin nanofiber and the nano-composite films showed antifungal activity against A. alternata.

Chitosan nanofibers were easily prepared from dry chitosan powder using the Star Burst system, which employs a high-pressure water jet system. Although the chitosan nanofibers became thinner as the number of Star Burst passes increased, the fiber thickness did not change significantly above 10 passes. Crystallinity and the chitosan nanofiber length decreased after extensive treatment due to the strong collision forces breaking the fibers. The mechanical properties and thermal expansion of the chitosan nanofiber sheets were improved by increasing the number of passes up to 10, but further treatment resulted in a deterioration of these properties.

We characterized the antifungal activity of the Bacillus circulans subclass III MH-K1 chitosanase (MH-K1 chitosanase), which is one of the most intensively studied glycoside hydrolases (GHs) that belong to GH family 46. MH-K1 chitosanase inhibited the growth of zygomycetes fungi, Rhizopus and Mucor, even at 10 pmol (0.3 μg)/ml culture probably via its fungistatic effect. The amino acid substitution E37Q abolished the antifungal activity of MH-K1 chitosanase, but retained binding to chitotriose. The E37Q mutant was fused with green fluorescent protein (GFP) at its N-terminus and proved to act as a chitosan probe in combination with wheat-germ agglutinin (WGA), which is a chitin-specific binding lectin. The GFP-fused MH-K1 chitosanase mutant E37Q (GFP-E37Q) bound clearly to the hyphae of the Rhizopus and Mucor strains, indicating the presence of chitosan. In contrast, Cy5-labelled WGA (Cy5-WGA), but not GFP-E37Q, stained the hyphae of non-zygomycetes species, i.e. Fusarium oxysporum, Penicillium expansum, and Aspergillus awamori. When the mycelia of Rhizopus oryzae were treated with wild type MH-K1 chitosanase, they could not bind to GFP-E37Q but were stained instead by Cy5-WGA. We conclude that chitin is covered by chitosan in the cell walls of R. oryzae.

We previously developed biobased wrinkled surfaces based on wood mimetic skins in which microscopic wrinkles were fabricated on a chitosan film by immersion in a phenolic acid solution, horseradish peroxidase-catalyzed surface reaction, and drying. Here, we prepared a diverse range of wrinkled films by immersion treatment at 30, 40, 50, and 60 °C in p-coumaric acid and then investigated the correlation between wrinkle morphology and mechanical properties. Wrinkle wavelengths gradually decreased as the immersion temperature increased as well as the previous report. In order to clarify the mechanisms responsible for the different wrinkle morphologies, the films were subjected to elastic moduli measurement and GPC analysis after immersion treatment. These experiments provided evidence that the chitosan around the film surface decomposed along with the immersion process. The decomposition was accelerated by higher immersion temperature, suggesting that higher temperatures led to the formation of softer skins, inducing smaller wrinkles. In fact, wrinkle morphologies with this system were predominately determined by the hardness of the wood mimetic skins. This phenomenon is consistent with the fundamentals of surface wrinkling in nature. This study is the first to demonstrate that artificial wrinkling triggered by water evaporation can be controlled by precise control of the surface hardness of soft material.

Although extensive research is in progress on the use of chitin and chitosan, poor solubility has been one of the main obstacles to their effective utilization. Introduction of carbohydrate branches into chitin and chitosans gave branched polysaccharide analogs, which showed good water solubility and induced new chemical and biological functions. In this study, hyperbranched chitosan derivatives were synthesized by introducing chitosan branches to the amino group of chitosan by the reductive N -alkylation method. Compared with the linear chitosan, the hyperbranched derivatives exhibited improved water solubility in the physiological conditions (pH 6.8). Although the viscosity of starting chitosans for main chain increased with the increase in their weight average molecular weights (80–190 kDa), the corresponding hyperbranched chitosan derivatives showed relatively low viscosities.

Microscopic surface architectures that can be easily manufactured have been in demand as mechano-structural cues for tissue engineering. Microscopic surface reliefs synthesized by wrinkling were expected as cell culture scaffolds for cell proliferation, control of cellular alignment and differentiation, and spheroid generation. We previously developed bio-based wrinkled films prepared via lignification-mimetic reactions and drying. Although these films are expected as a candidate for cell culture scaffolds, stability and morphology of the wrinkled surfaces in aqueous buffer solutions were not explored. Here, we investigate the surface morphologies of the wrinkled films in phosphate-buffered saline, and their application to 3T3 cell culture. The wrinkled film prepared with the immersion treatment at 40 °C maintained its wrinkled structure in phosphate-buffered saline even after five days, although the wrinkles were broadened by hydration of the skin layer. Interestingly, higher cell numbers were observed in the 3T3 cell culture using the wrinkled film than using flat film with the same surface composition. In addition, the high biocompatibility of the wrinkled film was confirmed by in vivo experiments. These results strongly encourage application of the wrinkled film as a mechano-structural cue. Studies of the advanced applications for the wrinkled films are now in progress.

Chitin (β-(1-4)-poly-N-acetyl-D-glucosamine) is widely distributed in nature and is the second most abundant polysaccharide after cellulose. Chitin occurs in nature as ordered macrofibrils. It is the major structural component in the exoskeleton of crab and shrimp shells and the cell wall of fungi and yeast. As chitin is not readily dissolved in common solvents, it is often converted to its more deacetylated derivative, chitosan. Chitin, chitosan, and its derivatives are widely used in tissue engineering, wound healing, and as functional foods. Recently, easy methods for the preparation of chitin and chitosan nanofibers have been developed, and studies on biomedical applications of chitin and chitosan nanofibers are ongoing. Chitin and chitosan nanofibers are considered to have great potential for various biomedical applications, because they have several useful properties such as high specific surface area and high porosity. This review summarizes methods for the preparation of chitin and chitosan nanofibers. Further, biomedical applications of chitin and chitosan nanofibers in (i) tissue engineering, (ii) wound dressing, (iii) cosmetic and skin health, (iv) stem cell technology, (v) anti-cancer treatments and drug delivery, (vi) anti-inflammatory treatments, and (vii) obesity treatment are summarized. Many studies indicate that chitin and chitosan nanofibers are suitable materials for various biomedical applications.

Sorbitol polyglycidyl ether (SPE) which is a bio-based water-soluble epoxy resin was cured with chitosan (CS) and/or a commercial water-soluble polyamidoamine- or polyetheramine-type epoxy hardener (PAA or PEA). Furthermore, biocomposites of the CS-cured SPE (CS-SPE) and CS/PAA- or CS/PEA-cured SPE (SPE-CA or SPE-CE) biocomposites with chitin nanofiber (CNF) were prepared by casting and compression molding methods, respectively. The curing reaction of epoxy and amino groups of the reactants was confirmed by the FT-IR spectral analysis. SPE-CS and SPE-CA were almost transparent films, while SPE-CE was opaque. Transparency of SPE-CS/CNF and SPE-CA/CNF became a little worse with increasing CNF content. The tanδ peak temperature of SPE-CS was higher than those of SPE-PAA and SPE-PEA. SPE-CA or SPE-CE exhibited two tanδ peak temperatures related to glass transitions of the CS-rich and PAA-rich or PEA-rich moieties. The tanδ peak temperatures related to the CS-rich and PAA-rich moieties increased with increasing CNF content. A higher order of tensile strengths and moduli of the cured resins was SPE-CS≫SPE-CA>SPE-CE. The tensile strength and modulus of each sample were much improved by the addition of 3wt% CNF, while further addition of CNF caused a lowering of the strength and modulus.

Previous reports indicate that the beneficial effect of chitin nanofibrils (CNFs), and chitosan nanofibrils (CSNFs) for wound healing. In this study, the wound healing effects of superficially deacetylated chitin nanofibrils (SDACNFs) were evaluated using an experimental model. In the experiments using circular excision wound model, SDACNFs induced re-epithelium and proliferation of the fibroblasts and collagen tissue. In the chitin, CNFs, and CSNFs, on the other hand, the e-epithelium and proliferation of the fibroblasts and collagen tissue were not induced perfectly compared with the SDACNFs group. In particular, re-epithelization was observed on day 4 in the only SDACNF group. Moreover, SDACNFs did not induce severe systemic inflammation in the linear incision wound model. The data indicated that SDACNFs effectively induced the proliferation and re-modeling phases compared with chitin, CNFs, and CSNFs in the wound. These data indicate that SDACNFs can be beneficial as a new biomaterial for wound healing.

Surface-deacetylated chitin nanofiber films plasticized with glycerol were prepared to control mechanical properties. Nanofiber networks were able to retain excessive glycerol content up to 70% to obtain self-standing film. All films were flexible and highly transparent independent of glycerol content. Glycerol significantly decreased the Young's moduli and tensile strengths, and increased the fracture strain due to its plasticizing effect. At the same time, glycerol did not change the high transparency or the low thermal expansion of the nanofiber film.

This study investigated the prophylactic effects of orally administered surface-deacetylated chitin nanofibers (SDACNFs) and chitosan against 5-fluorouracil (5-FU)-induced intestinal mucositis, which is a common side effect of 5-FU chemotherapy. SDACNFs and chitosan abolished histological abnormalities associated with intestinal mucositis and suppressed hypoproliferation and apoptosis of intestinal crypt cells. These results indicate that SDACNF and chitosan are useful agents for preventing mucositis induced by anti-cancer drugs.

Sustainable alternatives for high-performance and functional materials based on renewable resources are intensely needed as future alternatives for present-day, fossil-based materials. Nanochitin represents an emerging class of highly crystalline bionanoparticles with high intrinsic mechanical properties and the ability for conjugation into functional materials owing to reactive amine and hydroxyl groups. Herein we demonstrate that hydrogels containing surface-deacetylated chitin nanofibrils of micrometer length and average diameters of 9 nm, as imaged by cryogenic transmission electron microscopy, can be wet-spun into macrofibers via extrusion in a coagulation bath, a simple low energy and large-scale processing route. The resulting biofibers display attractive mechanical properties with a large plastic region of about 12% in strain, in which frictional sliding of nanofibrils allows dissipation of fracture energy and enables a high work-of-fracture of near 10 MJ/m3. We further show how to add functionality to these macrofibers by exploiting the amine functions of the surface chitosan groups to host catalytically active noble metal nanoparticles, furnishing biobased, renewable catalytic hybrids. These inorganic/organic macrofibers can be used repeatedly for fast catalytic reductions of model compounds without loss of activity, rendering the concept of hybridized chitin materials interesting as novel bioderived supports for nanoparticle catalysts.

Regioseletive copolymerization of N-isopropylacrylamide (NIPAM) onto chitosan was achieved by atom transfer radical polymerization (ATRP) by using a regioselective 3,6-di-O-bromoisobutyryl-2-N-phthaloyl chitosan as a macroinitiator. The degree of polymerization (DP) of polyNIPAM onto the chitosan derivative changed by varying the ratio between NIPAM monomer, macroinitiator, ligand, and transition metal. ATRP successfully proceeded and a DP of polyNIPAM up to 110.5 was obtained. The thermal decomposition temperature of the 3,6-di-O-bromoisobutyryl-2-N-phthaloyl chitosan was significantly improved by increasing the DP of the NIPAM component. The polyNIPAM-g-chitosan derivative showed a thermoresponsive property. Although it formed a stable suspension in water at room temperature, it caused a hydrophilic-to-hydrophobic transition at around 32 °C, resulting in precipitate formation.

Synthesis of a highly regioselective amphiprotic chitosan derivative was achieved by click chemistry. The Huisgen cycloaddition between 6-azido-6-deoxy- N -pthaloyl-chitosan and methyl propiolate was successfully carried out in the presence of Cu(I) catalyst. After the reaction, both the phthaloyl protecting group and methyl group were completely removed by hydrazine. FT-IR and NMR spectroscopy as well as elemental analysis strongly support the structural uniformity of the desired amphiprotic chitosan derivative, which has both a carboxylic group with a 1,4-triazole linker at the C-6 position and an amino group at the C-2 position per repeating unit. The amphiprotic chitosan derivative was soluble under both acidic and basic aqueous conditions. In contrast, it formed nanoparticle under neutral condition due to the interaction between the positive ( NH 3 + ) and negative ( COO − ) ions on the chitosan derivative.

A short synthesis of chemoselective chitosan derivatives was achieved by copper-catalyzed Huisgen cycloaddition, which is an ideal reaction for click chemistry, by using N-(4-azidophthaloyl)-chitosan. N-(4-azidophthaloyl)-chitosan was prepared through chemoselective N-bromophthaloylation of chitosan in acidic water and subsequent azidation. The obtained N-(4-bromopthaloyl)-chitosan had higher solubility in common solvents than conventional phthaloyl chitosan. N-(4-azidophthaloyl)-chitosan was successfully converted with ethynyl derivatives having functional groups (hydroxymethyl, phenyl, and methyl ester) in the presence of copper(II) sulfate, sodium ascorbate and/or trimethylamine. FT-IR spectra, elemental analyses, and 1H and 13C NMR spectra supported that the desired chitosan derivatives were chemoselectively transferred by these groups with a 1,4-triazole linker.

A one-pot synthesis was used to produce chitosan derivatives with polyphenolic side chains via a regioselective phenolic coupling reaction. Under Mannich reaction conditions, treatment of chitosan with formaldehyde and methyl 2,4-dihydroxybenzoate gave N-(2,6-dihydroxy-3-methoxycarbonylphenyl)methylated chitosan in good yield (87%). Formation of a CC bond occurred regioselectively at the C(3) position of methyl 2,4-dihydroxybenzoate. Chitosan derivatives having various phenolic compounds as a side chain were easily synthesized in a similar manner. The chitosan derivatives showed good biodegradability and improved their solubility in methanol (9.8 mg mL−1) and 2-methoxyethanol (> 10 mg mL−1). The UV protection provided by the derivatives with phenolic benzophenone side chain was evaluated using UV spectra of polyethylene terephthalate and poly(vinyl butyral-co-vinyl alcohol-co-vinyl acetate) films coated with the derivatives and the derivatives absorbed effectively in the UV-A region (

Highly chemoselective N-phthaloylations of chitosan were achieved by reaction in aqueous acetic acid media in concentrations from 0 to 10.0%. The degree of substitution (DS) values of phthaloyl groups determined by elemental analysis and 1H NMR were approximately 1. The 1H and 13C NMR spectra strongly support the structural uniformity of N-phthaloyl chitosan.
