The present study was aimed to examine the antibacterial potential of Brassica nigra essential oil (BNEO) against Ralstonia solanacearum, causal agent of bacterial wilt and Nitrosomonas sp., the nitrifying bacteria. In poisoned food assay, BNEO showed 100% growth inhibition of R. solancearum at ≥ 125 µg mL-1. Revalidation of findings by volatile assay employing inverted Petri plate technique exhibited 100% bacterial growth inhibition caused by vapors of BNEO, even at 50 µg mL-1 concentration. In the broth microdilution assay, the BNEO exhibited significant antibacterial activity only at higher concentrations (>500 µg mL-1). At 500 µg mL-1, BNEO showed 80% bacterial growth inhibition over control, which was at par with that of streptomycin (5 µg mL-1). In resazurin microtitre-plate assay, the maximum concentration of BNEO, at which color change occurred was 512 µg mL-1 (T9), and thus 512 µg mL-1 was concluded as the minimum inhibitory concentration (MIC). BNEO effectively inhibited the activity of Nitrosomonas spp. with 30-65% nitrification inhibition at the dose of 400 mkg-1 of Urea-N. Homology modeled protein targets assisted computational tool-based novel analysis helped to understand that the antibacterial potency of BNEO is due to preferable binding efficiency of allyl isothiocyanate (AITC), the major active ingredient of BNEO.

The cultivation of Panax notoginseng is often seriously hindered by root rot disease caused by the accumulation of soil-borne pathogens. Here, the inhibitory activity of Brassica juncea volatiles on P. notoginseng root rot pathogens was assessed and compounds in volatiles were identified. Furthermore, the antimicrobial activity and mechanism of allyl isothiocyanate (AITC) were deciphered by integrated transcriptome and metabolome analyses. The volatiles of B. juncea showed dose-dependent antimicrobial activity against root rot pathogens. AITC, identified as the main volatile compound, not only significantly inhibited pathogen growth in vitro but also suppressed root rot disease in the field. Integrated transcriptomic and metabolomics analysis revealed that AITC inhibited Fusarium solani by interfering with energy production and induced the accumulation of ROS by decreasing the content of glutathione (GSH). In summary, B. juncea releases AITC to inhibit soil-borne pathogens and could be used as a rotation crop or soil fumigant to alleviate root rot disease.

Doxorubicin (DOX) is an effective anthracycline chemotherapeutic drug. Nevertheless, the cardiotoxicity adverse effect restricts its clinical benefit. Allyl isothiocyanate (AITC) is a natural antioxidant and anti-inflammatory agent. In the present study, we investigated the effect of AITC on cardiotoxicity of DOX. Thirty-two adult male albino rats were divided into four groups; control, AITC, DOX, and AITC + DOX. AITC was administrated orally (25 mg/kg/day) for 7 days, and DOX was given as a single i.p. injection (15 mg/kg) on the third day. Mortality rate was observed during the experiment. Cardiac toxicity markers (lactate dehydrogenase (LDH), creatine kinase (CK-MB), and cardiac Troponin I (cTn-I)) were evaluated in serum samples obtained from all groups after 48 hours of DOX injection. DOX-treated group showed 40% mortality and a significant increase in cardiac enzymes. This increase was accompanied by degenerated cardiomyocytes, and inflammatory cells infiltrates. Interestingly, AITC administration alleviated myocardial oxidative stress induced by DOX as attenuated the increase in malondialdehyde (MDA), and nitric oxide (NO) while resulted in elevations of the antioxidant reduced glutathione (GSH) level as well as superoxide dismutase (SOD) activity. Furthermore, the inflammatory cytokine, TNF-α, was reduced upon administration of AITC with DOX. The cardio-protection of AITC is attributed to increase the expression of cytoprotective nuclear factor erythroid 2-related factor 2 (Nrf2). Subsequently, heme oxygenase 1 (HO-1) level was elevated by AITC to correct the oxidative stress induced by DOX in the heart. Accordingly, AITC ameliorated acute cardiotoxicity associated with DOX treatment via attenuation of oxidative stress and the induced-tissue inflammatory injury. Background Soil fumigants-the most effective agrochemicals for managing soil-borne diseases-have been extensively used. However, high volatility, moderate toxicity, and insufficient effective duration considerably limit their application. In the present study, a combination of modified biochar and polyurea microcapsules (MCs) via interface polymerization to co-encapsulate allyl isothiocyanate (AITC) was developed as a model fumigant for controlled release (AITC@BC-MCs). Results The physical characteristics of biochar modified by sand milling were significantly improved. In addition, chemical properties and morphological features of AITC@BC-MCs characterized by integrated methods revealed successful preparation of BC-MCs. Compared with monolayer MCs, BC-MCs could significantly delay AITC release due to the composite obstruction effect. Moreover, modified biochar endowed the cargo molecules with a pH-responsive release property. Additionally, this composite showed a longer persistent duration by prolonging AITC degradation half-life, which was 3.2-3.5 times that of the AITC technical concentrate under different soil conditions. Finally, the control efficacy of AITC@BC-MC against pathogens, including nematodes and fungi, as well as against weeds was significantly enhanced at the same dose, but it did not inhibit seed germination and growth after 10 days when fumigated soil was aerated. Conclusion Construction of a composite encapsulation system enhanced pesticide efficacy, reduced dose via controlled release, and delayed fumigant degradation in soil, indicating the great potential of this strategy for developing an effective and environmentally friendly fumigant formulation. The environmental impact of methyl bromide (MB) has resulted in its phase out as an insecticidal fumigant except for critical use exempted categories. Consequently, there is an urgent need to develop an environmentally sustainable MB alternative fumigant. trans-Cinnamaldehyde (TC), benzaldehyde, allyl isothiocyanate (AITC), hexanal, and ethyl formate (EF) are naturally occurring plant volatiles with insecticidal properties. This study assessed the toxicity of these plant volatiles to adult and egg stages of the spotted-wing drosophila (SWD) (Drosophila suzukii Matsumura). The plant volatile treatments had a significant effect on adult SWD mortality. The descending order of toxicity to adult SWD was benzaldehyde > AITC > TC > hexanal > EF at a headspace concentration of 0.50 μL/L air for 24 h. All the volatiles, at a concentration of 4.00 μL/L air, significantly inhibited larval emergence from SWD eggs in artificial diet compared to the control. At a 0.50 μL/L air level, among the volatiles tested, only AITC exhibited 100% inhibition against larval emergence from SWD eggs in blueberry fruits after 24 h exposure. In summary, this study shows that all volatiles tested elicited varying degrees of toxicity toward SWD adults and eggs. However, AITC was the most efficacious volatile and the one with the greatest promise as a post-harvest fumigant for both adult and egg stages of SWD.

Allyl isothiocyanate (AITC), as a fumigant, plays an important role in soil control of nematodes, soil-borne pathogens, and weeds, but its effects on soil microorganisms are unclear. In this study, the effects of AITC on microbial diversity and community composition of Capsicum annuum L. soil were investigated through Illumina high-throughput sequencing. The results showed that microbial diversity and community structure were significantly influenced by AITC. AITC reduced the diversity of soil bacteria, stimulated the diversity of the soil fungal community, and significantly changed the structure of fungal community. AITC decreased the relative abundance of dominant bacteria Planctomycetes, Acinetobacter, Pseudodeganella, and RB41, but increased that of Lysobacter, Sphingomonas, Pseudomonas, Luteimonas, Pseudoxanthomonas, and Bacillus at the genera level, while for fungi, Trichoderma, Neurospora, and Lasiodiplodia decreased significantly and Aspergillus, Cladosporium, Fusarium, Penicillium, and Saccharomyces were higher than the control. The correlation analysis suggested cellulase had a significant correlation with fungal operational taxonomic units (OTUs) and there was a significant correlation between cellulase and fungal diversity, while catalase, cellulose, sucrase, and urease were the major contributors in the shift of the community structure. Our results will provide useful information for the use of AITC in the assessment of environmental and ecological security.

Background Understanding the specific inhibitory effects of different Brassica seed meals (BSMs) on soilborne pathogens is important for their application as biocontrol agents for controlling plant disease. In this study, the seed meals of Brassica napus L. (BnSM), Brassica campestris L. (BcSM), and Brassica juncea L. (BjSM), and the combined seed meal of BcSM and BjSM (CSM, 1:1), were selected for investigation. The inhibitory effects of these seed meals on the plant pathogen Ralstonia solanacearum (Smith) and tomato bacterial wilt, were assessed and compared. Results All the BSMs significantly inhibited the growth of R. solanacearum in vitro. Furthermore, the BSMs could effectively suppress R. solanacearum virulence traits, including motility, exopolysaccharide production, dehydrogenase activity, virulence-related gene expression, and colonization in the soil. Among them, BjSM showed the best inhibiting effects, and CSM displayed synergic toxicity against R. solanacearum. In addition, the predominant antibacterial compounds in BcSM and BjSM were identified as the volatile compounds, 3-butenyl isothiocyanate and allyl isothiocyanate, respectively. Finally, pot experiment verified that the control effects of BjSM and CSM on tomato wilt reached more than 90%. Conclusion This is the first study to report on the ability of different kinds of BSMs to suppress the virulence of R. solanacearum and biocontrol efficiencies against bacterial wilt in tomato plants. Furtherly, the main antibacterial compounds in the BSMs were identified. The results demonstrated that CSM may possess potential for controlling bacterial wilt caused by R. solanacearum. The results provide a fresh perspective for comprehending the mechanism underlying BSM suppression of pathogens and plant disease. This article is protected by copyright. All rights reserved.

The cruciferous vegetables are richest sources of isothiocyanates and among them, allyl isothiocyanate (AITC) is one of the major constituents. It has been known to possess various activities of therapeutic interest. Keeping this in view, the present study was planned to investigate antioxidant capacity of AITC using Cupric Reducing Antioxidant Capacity (CUPRAC) Assay, 2, 2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay, β-Carotene bleaching (BCB) assay and superoxide anion scavenging assay. The antimutagenic potency of AITC was determined by modifying Vibrio harveyi bioluminescence assay against direct acting mutagen 4-nitro-o-phenylenediamine (NPD) and indirect acting mutagens 2-acetylaminofluorene (2-AAF), 7,12-Dimethylbenz[a]anthracene (DMBA) in presence and absence of S9. Furthermore, the antihemolytic activity was evaluated using hemolysis assay. Among the different activities carried out, it was observed that AITC exhibited efficient antimutagenic potential against direct acting mutagen NPD and indirect acting mutagens 2-AAF and DMBA in the presence as well as absence of S9 fraction in Vibrio harveyi bioluminescence assay. Overall, our results established the efficiency of AITC to act as strong antimutagenic agent, efficient anti-hemolytic agent and mild antioxidant agent thus, making AITC an interesting candidate to be further explored for drug discovery.

The aims of this work were to study growth, isothiocyanate (ITC), bioactive content, antioxidant activity and anticancer activity of mustard green (MG) microgreens grown from seeds treated with cold plasma at 21 and 23 kV for 5 min. Microgreens from plasma-treated seeds at 23 kV showed almost 2-fold increased ITC content (1.57 ± 0.05 mmol/100 g DW) compared to MG from seeds without plasma (control), showed the highest total phenolic content (TPC) (6.76 ± 0.14 mg GAE/g DW) and total flavonoid content (TFC) (0.16±0.01 mg RE/g DW). However, MG plasma-treated seeds at 21 kV showed the highest antioxidant activity from 2,2-Diphenyl-1-picrylhydrazyl (DPPH) assay (3.51 ± 0.38 mg TE/g DW). Allyl isothiocyanate and 3-butenyl isothiocyanate were the dominant ITCs in MG. The highest cytotoxicities using 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay against MCF-7 (IC50 of 32.44 ± 1.64 μg/mL) and HepG2 (IC50 of 28.58 ± 1.04 μg/mL) after 72 h exposure were found in MG from plasma-treated seeds at 23 kV and MG from control seeds, respectively. However, MG from plasma-treated seeds at 21 kV exhibited the highest antiproliferative effect against MCF-7 (IC50 of 23.23 ± 0.23 μg/mL) and HepG2 (IC50 of 20.44 ± 0.56 μg/mL) for 14 days and also the most potent antimigratory effect. MG from cold plasma inhibited MMP-9 protein expression in both cancers indicating antimigratory property. MG from cold plasma also significantly reduced MMP-9 mRNA expression in both cancers when compared to the control and untreated cells. In conclusion, cold plasma treatment on seeds seemed to be an innovative tool to enhance ITC, TPC, TFC and anticancer properties of MG microgreens for better health implications. © 2021 Friends Science Publishers

The effect of TRPA1-ion channel on thermoregulatory responses depending on the level of its activity was studied in Wistar rats. To activate the TRPA1 ion channel localized in the skin, its agonist allyl isothiocyanate (AITC) was used in different concentrations (0.04, 0.4, 1, and 2.5%). Low concentration of AITC (0.04%) enhanced and high concentrations (1 and 2.5%), on the contrary, inhibited cold-defense responses (decreased their magnitude and led to their later initiation due to an increase in temperature thresholds). With an increase in TRPA1 activation, the increase in temperature thresholds (afferent link) was ahead of the decrease in the magnitude of responses (efferent link), which can attest to different sensitivity of these processes to TRPA1 activation.

Background/purpose Cisplatin-resistant oral cancer is clinically difficult to manage and the dose-dependent toxicities of cisplatin has been widely concerned. Allyl isothiocyanate (AITC), known as mustard oil, is a plant-derived compound abundant in cruciferous vegetables. It is reported to have anti-cancer potential as a natural dietary chemopreventive compound against a variety of cancers, but the effect of AITC on cisplatin-resistant cancer cells is still little-known. Methods Human CAL27-cisplatin-resistant oral cancer cells (CAR cells) were examined to investigate the antitumor properties of AITC. 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide (MTT) assay, IncuCyte™ S3 cell proliferation assay, 4′,6-diamidino-2-phenylindole (DAPI) and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining as well as Western blot analysis were deployed. Results AITC decreased CAR cell viability, induced cell death of CAR cells and inhibited the confluences of cultured CAR cells. When CAR cells were treated with AITC, activation of caspase-3 and caspase-9 by AITC was observed and could be reversed by Z-VAD-fmk (pan-caspase inhibitor). Furthermore, the protein expressions of phosphorylated protein kinase B (p-AKT) and phosphorylated mammalian target of rapamycin (p-mTOR) were suppressed in AITC-treated CAR cells, whereas protein expressions of Bax, cytochrome c, Apaf-1, cleaved caspase-3, and cleaved caspase-9 were upregulated in AITC-treated CAR cells. Conclusion AITC can inhibit Akt/mTOR proliferation signaling and promote mitochondria-dependent apoptotic pathway through AITC-enhanced activities of caspase-3 and caspase-9 in CAR cells.

Objective We investigated the anti-proliferative or cytotoxic activities of five nutraceutical compounds: allyl isothiocyanate, β-carotene, caffeine, capsaicin, and lupanine that we consume respectively, for example, from mustard seeds, carrot, coffee, pepper, and lupin seeds against cancer cell lines (human colon: HCT 116 p53 wild type, HCT 116 p53−/− and lymphoblastic: CEM/CCRF, CEM/ADR5000). Result Out of the five compounds tested in vitro, capsaicin and β-carotene were more cytotoxic than the other three compounds against the four cancer cell lines. The most potent nutraceutical compound was capsaicin and it exerted its highest cytotoxicity against HCT 116 p53−/− with IC50 value of 19.67 ± 0.06 µM. It is worth considering capsaicin for further development of anticancer drug against both colon and leukemia cancer types.

Isothiocyanates (ITCs), present as glucosinolate precursors in cruciferous vegetables, have shown anti-inflammatory, antioxidant and anticarcinogenic activities. Here, we compared the effects of three different ITCs on ROS production and on the expression of matrix metalloproteinase (MMP)-2 and -9, which represent important pathogenetic factors of various neurological diseases. Primary cultures of rat astrocytes were activated by LPS and simultaneously treated with different doses of Allyl isothiocyanate (AITC), 2-Phenethyl isothiocyanate (PEITC) and 2-Sulforaphane (SFN). Results showed that SFN and PEITC were able to counteract ROS production induced by H2O2. The zymographic analysis of cell culture supernatants evidenced that PEITC and SFN were the most effective inhibitors of MMP-9, whereas, only SFN significantly inhibited MMP-2 activity. PCR analysis showed that all the ITCs used significantly inhibited both MMP-2 and MMP-9 expression. The investigation on the mitogen-activated protein kinase (MAPK) signaling pathway demonstrated that ITCs modulate MMP transcription by inhibition of extracellular-regulated protein kinase (ERK) activity. Results of this study suggest that ITCs could be promising nutraceutical agents for the prevention and complementary treatment of neurological diseases associated with MMP involvement.

The present study aimed to examine the effect of allyl isothiocyanate (AITC) on chronic obstructive pulmonary disease and to investigate whether upregulation of multidrug resistance-associated protein 1 (MRP1) associated with the activation of the PARK7 (DJ-1)/nuclear factor erythroid 2-related factor 2 (Nrf2) axis. Lung function indexes and histopathological changes in mice were assessed by lung function detection and H&E staining. The expression levels of Nrf2, MRP1, heme oxygenase-1 (HO-1), and DJ-1 were determined by immunohistochemistry, Western blotting and reverse transcription-quantitative polymerase chain reaction. Next, the expression of DJ-1 in human bronchial epithelial (16HBE) cells was silenced by siRNA, and the effect of DJ-1 expression level on cigarette smoke extract (CSE)-stimulated protein degradation and AITC-induced protein expression was examined. The expression of DJ-1, Nrf2, HO-1, and MRP1 was significantly decreased in the wild type model group, while the expression of each protein was significantly increased after administration of AITC. Silencing the expression of DJ-1 in 16HBE cells accelerated CSE-induced protein degradation, and significantly attenuated the AITC-induced mRNA and protein expression of Nrf2 and MRP1. The present study describes a novel mechanism by which AITC induces MRP1 expression by protecting against CS/CSE-mediated DJ-1 protein degradation via activation of the DJ-1/Nrf2 axis.

Since lycopene Z-isomers exhibit greater bioavailability and biological activity than the naturally occurring all-E-isomer, efficient manufacturing methods for (Z)-lycopene-rich materials are urgently needed. Herein, a method was developed for Z-isomerization of (all-E)-lycopene in tomato oleoresin using heat treatment and a natural catalyst, viz. allyl isothiocyanate (AITC). For practical application of this isomerization technology, no organic solvents were used, and instead, oils and fats were used as the reaction medium. The Z-isomerization of (all-E)-lycopene was promoted by heating (>120 °C) even when oil and fat media were used. Allyl isothiocyanate enhanced thermal Z-isomerization and improved the (5Z)-lycopene content, which shows higher biological activity compared to the other Z-isomers. The thermal isomerization efficiency with AITC was further improved by using certain vegetable oils such as argan and olive oils. In addition, the storage stability of (Z)-lycopene-rich tomato concentrates dispersed in olive oil was evaluated. The total Z-isomer ratio and lycopene concentration decreased with longer storage periods, and it was revealed that (5Z)-lycopene showed excellent storage stability among the mono-Z-isomers.

BACKGROUND A combination of high-pressure processing (HPP) and antimicrobials is a well-known approach for enhancing the microbiological safety of foods. However, few studies have applied multiple antimicrobials simultaneously with HPP, which could be an additional hurdle for microbial inactivation. The present study applied a full factorial design to investigate the impact of HPP (225-325 MPa; 10-20 min), allyl isothiocyanate (AITC) (0.3-0.9 g kg-1 ) and trans-cinnamaldehyde (tCinn) (1.0-2.0 g kg-1 ) on the inactivation of Shiga toxin-producing Escherichia coli (STEC) O157:H7 and uropathogenic E. coli (UPEC) in ground chicken meat. RESULTS The regulatory requirement of 5-log reduction was achieved at 305 MPa, 18 min, 0.8 g kg-1 AITC and 1.7 g kg-1 tCinn for STEC O157:H7 and at 293 MPa, 16 min, 0.6 g kg-1 AITC and 1.6 g kg-1 tCinn for UPEC, as specified by response surface analysis and verified via experiments. The surviving population was eliminated by post-treatment storage of 9 days at 10 °C. The developed linear regression models showed r2 > 0.9 for the E. coli inactivation. The correlation of sensory and chemically evaluated pungency of mustard products was investigated via a time-intensity (TI) study and quantification of allyl isothiocyanate (AITC) contents using high-performance liquid chromatography (HPLC). Sweet, medium hot, hot, and extra hot commercial mustard products from different brands were examined. Notably, we found significant differences (p < 0.05) between the maximum perceived pungency intensity of various mustard products. The maximum perceived intensity (Imax ), the duration of the decreasing phase (DURDec ), and the area under the curve (AUC) values increased proportionally to the increase in the sample AITC content and were also higher in products classified as hot than in sweet mustards. The AITC concentration varied greatly between products from different brands and also between different sensory evaluated pungency levels. Furthermore, sensory evaluations and analytical results were correlated using regression analysis. The best correlation (correlation coefficient 0.891) was observed between the AITC concentration and AUC, when compared to that between the AITC concentration and DURDec (correlation coefficient 0.856) or the Imax value (correlation coefficient 0.803). The calculated regression model indicates that a higher AITC content induces an intensified trigeminal pungency sensation and that the sensory and chemical evaluations of mustard products were positively correlated. Therefore, by using this regression model, the sensory rating of mustard products may be predicted by chemical analysis of the AITC contents. PRACTICAL APPLICATION: This research paper provides a method to quantify the pungency inducing irritant allyl isothiocyanate in commercial mustard products and demonstrates a correlation between sensory and chemical data. Therefore, the amounts of sensory tests in product quality assurance can be reduced and replaced or at least supported by chemical quantification of pungent substances (especially AITC) in mustard products.

Essential oils are widely recognized as an efficient and safe alternative for controlling pests in foods. However, a few studies have determined the persistence of these compounds in stored grains. The present study optimized and validated a fast and effective method for extraction and quantification of allyl isothiocyanate (AITC—the main component of mustard essential oil) residue in cowpea beans. It also investigated the persistence of this substance in the grains. The proposed method employs headspace solid-phase microextraction (HS-SPME) and gas chromatography with a flame ionization detector (GC/FID). For optimizing it, a central composite design (CCD) was used, where the best conditions for the extraction of the AITC were achieved using 15 min fiber exposure at 30 °C. The performance of the method was assessed by studying selectivity, linearity, limits of detection (LOD) and quantification (LOQ), precision, and accuracy. The LOD and LOQ for AITC were 0.11 and 0.33 μg kg–1, respectively. The determination coefficient (R2) was above 0.99. The relative recovery rate ranged from 108.2 to 114.8%, with an interday coefficient of variation below 9%. After 36 h, no residue was detected in the samples, demonstrating that the AITC has low persistence and can be safely used as a bioinsecticide for grains.

Horseradish (Armoracia rusticana) leaves pomace, which contains high-value bioactive compounds, is the product resulting from pressing horseradish leaves for juice production. The aim of the current research was to investigate the effect of convective, microwave-vacuum and freeze-drying on the content of bioactive compounds in horseradish leaves pomace. Convective hot air-drying was performed at 40, 60 and 80 °C. The total phenolic content (TPC), total flavonoid content (TFC), total flavan-3-ol content, total phenolic acid content, total flavonol content, chlorophylls and total carotenoids, and antioxidant activity were determined by spectrophotometric methods. Individual profiles of phenols and organic acids are estimated by high-performance liquid chromatography (HPLC), but volatile compounds are estimated by gas chromatography (GC). Totally, 14 individual phenolic compounds, 8 organic acids, and 49 volatile compounds were analysed in the studied samples. The main phenolic compound identified in horseradish leaves pomace was rutin (3231 mg/100 g DW), among organic acids—quinic and malic acids, and volatile compounds—allyl isothiocyanate, 3-butenenitrile and benzyl alcohol. In the drying process, the content of some (total flavan-3-ols, total carotenoids content) compounds increased, but others (TPC, total organic acids content) decreased, and it was drying method-dependent. Freeze-drying caused the reduction of TPC by 29%, whereas convective drying by 53–59%. Fresh pomace contains such isothiocyanates as allyl isothiocyanate and butyl isothiocyanate, which were completely lost in the drying process. Freeze-drying allowed the best retention of various phenolic and volatile compounds in horseradish leaves pomace.

Background and purpose Develop a translational assay of Transient Receptor Potential Ankyrin 1 (TRPA1) activity for use as a preclinical and clinical biomarker. Experimental approach Allyl isothiocyanate (AITC), capsaicin or citric acid were applied to ears of wildtype and Trpa1-knock out (Trpa1 KO) rats, and changes in dermal blood flow (DBF) were measured by laser speckle contrast imaging. In humans, the DBF, pain and itch responses to 5-20% AITC applied to the forearm were measured and safety was evaluated. Reproducibility of the DBF, pain and itch responses to topically applied 10% and 15% AITC were assessed at two visits separated by 13-15 days. DBF changes were summarized at 5-minute intervals as areas under the curve (AUC) and maxima. Intraclass correlation coefficient (ICC) was calculated to assess arm-arm and period-period reproducibility. Key results AITC- and citric acid-induced DBF were significantly reduced in Trpa1 KO rats compared to wildtype (90 ± 2% and 65 ± 11% reduction, respectively), whereas capsaicin response did not differ. In humans, each AITC concentration significantly increased DBF compared to vehicle with the maximal increase occurring 5 minutes post application. Ten percent and 15% AITC were selected as safe and effective stimuli. AUC from 0 to 5 minutes was the most reproducible metric of AITC-induced DBF across arms (ICC = 0.92) and periods (ICC = 0.85). Subject-reported pain was more reproducible than itch across visits (ICC = 0.76 vs 0.17, respectively). Conclusion and implications AITC-induced DBF is a suitable target engagement biomarker of TRPA1 activity for preclinical and clinical studies of TRPA1 antagonists.

Steam has long been used to disinfest greenhouse soils. However, there is increasing interest in expanding the use of steam for in-field soil disinfestation as an alternative to chemical fumigants. Previous studies demonstrated that allyl-isothiocyanate (AITC) reduced viability of weed seeds and plant pathogen propagules, but AITC has a low vapor pressure and is relatively immobile in soil. Heat has been used in the past to enhance the mobility of soil fumigants such as methyl bromide (i.e., “hot gassing”). The effect of steam heat on the mobility of AITC is unknown. The objective of this study was to investigate the potential synergistic effect of steam plus ATIC against weed seeds and a plant pathogen. AITC alone did not reduce the viability of the four weed species and the number of Verticillium dahliae microsclerotia. The steam + AITC treatment reduced the viability of V. dahliae at 12.5 and 18 cm distances by 82% and 88%, respectively, and knotweed and nettle seeds at 70 cm from injection point by 75% and 86%, respectively, from the center of microplots compared with steam alone. The results suggest that AITC and steam have a complementary effect on soilborne pests because steam increases the mobility of AITC.

The human gut microbiota plays a critical role in the regulation of adiposity, obesity and metabolic and cardiovascular disease. Wasabi is a pungent spice and its active component, allyl isothiocyanate, improves plasma triacylglycerol, cholesterol and high blood pressure in rodents, but it is unclear if this occurs through alterations to the composition of the microbiota. The aim of this study was to determine the effectiveness of Wasabi japonica stem and rhizome blend on ameliorating cardiovascular disease parameters including plasma sodium concentration, systolic blood pressure (SBP), plasma endothelin-1 and angiotensin II concentrations by altering the gut microbiota in a Wistar rat model of obesity and metabolic syndrome. Rats were randomized to receive a corn starch or high-carbohydrate/high-fat diet for 8 weeks before being allocated to supplementation with wasabi powder (5% (w/w) in food) or not for an additional 8 weeks. At the end of the trial, rats were grouped according to blood pressure status. Wasabi supplementation prevented the development of hypertension and was also associated with significantly increased abundance of Allobaculum, Sutterella, Uncl. S247, Uncl. Coriobacteriaceae and Bifidobacterium. Hypertension was positively correlated with higher abundance of Oscillospira, Uncl. Lachnospiraceae and Uncl. Clostridiales, Uncl. Bacteroidales and Butyricimonas. Oscillospira and Butyricimonas abundances were specifically positively correlated with systolic blood pressure. Overall, the improved host cardiovascular health in diet-induced obese rats supplemented with wasabi powder may involve changes to the gut microbiota composition.

Diplotaxis erucoides is a wild edible plant (WEP) of the Brassicaceae family, growing as a weed in Mediterranean countries. It contains glucosinolates, which are hydrolyzed by myrosinase rendering isothiocyanates; these compounds are responsible for its characteristic aroma and flavor, similar to that of mustard or wasabi. The aim of this study was to characterize the Diplotaxis erucoides plant by analyzing its volatile, sugar, organic acid, and sensory profiles. This information will be essential to decide whether it is possible to develop sustainable and healthy alternatives to commercial mustard and wasabi using this WEP. Two Diplotaxis-based products were prepared one cream and one mayonnaise. Allyl isothiocyanate represented more than 80% of total content of volatile compounds in the leaves, pods and stems. The sugar and organic acid content of Diplotaxis and its based products were very low. The Diplotaxis products were characterized by having intense Diplotaxis odor/aroma, herbaceous flavor and long aftertaste, but lacked some pungency. Diplotaxis erucoides is a sustainable and healthy option to produce herbaceous and pungent sauces.

Papillomatous digital dermatitis (PDD) is a foot disease causing lameness in dairy cattle. It is regarded as a polymicrobial infection, although its etiology is not fully understood. PDD is treated by the topical or systemic administration of antibiotics such as lincomycin (LCM); however, the milk of the cows cannot be marketed during the treatment and withdrawal period due to the residual antibiotics in milk. Allyl isothiocyanate (AITC), an extract of Wasabia japonica (known as wasabi or Japanese horseradish) widely employed as a food additive, can be used as an alternative antimicrobial agent that overcomes this problem. We previously showed that AITC is as effective as LCM in PDD treatment. Here, using the samples obtained in the previous clinical study, we analyzed changes in the bacterial population in the PDD-associated microbiota after AITC treatment and compared those with that following LCM treatment by 16S ribosomal RNA (rRNA)-based amplicon analysis. Both treatments induced major changes in the bacterial population, and Treponema species, which have been regarded as the major causative agents of PDD, were efficiently eliminated by both agents. However, the AITC-treated samples exhibited higher diversity compared with pretreatment samples, but this trend was not observed for LCM treatment, probably reflecting different antibacterial activities of the two agents. Importantly, this analysis detected population changes before morphological changes in PDD lesions (clinical signs of healing) became evident, indicating that 16S rRNA-based amplicon analysis represents an efficient strategy for analyzing and monitoring the treatment efficiency of PDD as well as other polymicrobial diseases.

Red imported fire ants, Solenopsis invicta Buren (Hymenoptera: Formicidae), which are among the most harmful alien ants, were first detected in Japan in a shipping container from China in May 2017, and continue to enter Japan via this trade pathway. Fumigation is one of the most effective ways to eliminate S. invicta from goods transported in containers. The present study is the first to evaluate the effect of fumigation with allyl isothiocyanate (AITC) against S. invicta. Fumigation using microencapsulated AITC pellets in a gas-barrier bag for 24 h completely killed S. invicta. Furthermore, our fumigation system was able to maintain a high concentration of AITC vapor for at least 13 days. AITC, a naturally occurring compound in wasabi [Eutrema japonicum (Miq.) Koidz], has been demonstrated to be safe for humans and the environment, but its use as a fumigant is limited by its strong volatility and pungency. However, its encapsulation solves this problem by allowing controlled vapor release. Thus, the use of microencapsulated AITC in combination with a gas-barrier bag has considerable potential to provide an effective and easy-to-use protocol for fumigation of S. invicta.

Allyl isothiocyanate (AITC) is a glucosinolate produced in cruciferous plant species. AITC is known to act as a pesticide on microorganisms, insects, and weeds. Synthetic AITC is registered as a biopesticide for agricultural soil treatment use in the United States and elsewhere in the world. Although a potent pesticide, reports on the weed and pathogen control efficacy of synthetic AITC applied as soil disinfectant are highly variable. Due to the low vapor pressure of AITC, questions remain as to whether pest and weed control efficacy can be improved by combining it with other chemicals. The objective of this study was to assess the control efficacy of AITC stand-alone applications vs. applications, in which AITC was combined with the standard-fumigants chloropicrin, 1,3-dichloropicrin, and methyl isothiocyanate. Two shank-applied on-farm field trials were conducted in cut flower [delphinium (Delphinium elatum), ranunculus (Ranunculus asiaticus)] fields, and two drip tape applied field trials in strawberry (Fragaria ×ananassa) fields in California. Weed pressure, weed seed viability, nematode survival, and pathogen survival of Pythium ultimum, fusarium wilt (Fusarium oxysporum), and verticillium wilt (Verticillium dahliae) were assessed. Cumulative yearly yield of marketable fruit was assessed in the strawberry field trials. The results of this study show that the use of AITC as a stand-alone treatment provided no consistent weed or pathogen control efficacy. However, our results also indicate that shank and drip applied multitactic fumigation approaches with AITC can efficiently control soil-borne diseases and weeds. These findings have potential implications, especially in those areas where certain fumigants are restricted due to regulations and/or availability.

The glucosinolate–myrosinase system is a well-known defense system that has been shown to induce stomatal closure in Brassicales. Isothiocyanates are highly reactive hydrolysates of glucosinolates, and an isothiocyanate, allyl isothiocyanate (AITC), induces stomatal closure accompanied by elevation of free cytosolic Ca2+ concentration ([Ca2+]cyt) in Arabidopsis. It remains unknown whether AITC inhibits light-induced stomatal opening. This study investigated the role of Ca2+ in AITC-induced stomatal closure and inhibition of light-induced stomatal opening. AITC induced stomatal closure and inhibited light-induced stomatal opening in a dose-dependent manner. A Ca2+ channel inhibitor, La3+, a Ca2+chelator, EGTA, and an inhibitor of Ca2+ release from internal stores, nicotinamide, inhibited AITC-induced [Ca2+]cyt elevation and stomatal closure, but did not affect inhibition of light-induced stomatal opening. AITC activated non-selective Ca2+-permeable cation channels and inhibited inward-rectifying K+ (K+in) channels in a Ca2+-independent manner. AITC also inhibited stomatal opening induced by fusicoccin, a plasma membrane H+-ATPase activator, but had no significant effect on fusicoccin-induced phosphorylation of the penultimate threonine of H+-ATPase. Taken together, these results suggest that AITC induces Ca2+ influx and Ca2+ release to elevate [Ca2+]cyt, which is essential for AITC-induced stomatal closure but not for inhibition of K+in channels and light-induced stomatal opening., Allyl isothiocyanate (AITC) induces Ca2+ influx and Ca2+ release to elevate [Ca2+]cyt, which is essential for AITC-induced stomatal closure but not for inhibition of K+in channels or light-induced stomatal opening.

Effects of natural catalysts, isothiocyanates and polysulfides, on Z-isomerization and decomposition of (all-E)-carotenoids (lycopene, β-carotene, and astaxanthin) after heat treatment were investigated. When isothiocyanates were added to (all-E)-carotenoid solutions and heated, Z-isomerization and decomposition of carotenoids were enhanced and the degree differed depending on the isothiocyanate type. Interestingly, when polysulfides were applied in the same manner, in addition to promoting the Z-isomerization reaction, they markedly improved the thermal stability of carotenoids. Successively, we investigated the reaction characteristics of allyl isothiocyanate (AITC) and diallyl disulfide (DADS) using (all-E)-lycopene; that is, effects of the amount added, solvent used, and reaction temperature and time, as well as the combination use on Z-isomerization and decomposition of lycopene, were investigated. With increases in the amount added and reaction temperature and time, Z-isomerization of lycopene was promoted for both catalysts. The high-temperature treatment tests clearly showed that AITC induced thermal decomposition of lycopene, whereas DADS improved the lycopene stability. Moreover, the simultaneous use of AITC and DADS resulted in a synergetic effect on the Z-isomerization efficiency.

Due to its biological composition, poultry meat is perishable by nature and susceptible to quality deterioration such as microbial spoilage and oxidation changes. Meat and meat products with healthy, extended shelf-life and good organoleptic properties are highly demanded by consumers. In current study, the antimicrobial and antioxidant effects of allyl-isothiocyanate (AITC) as a natural additive on raw chicken meat during chilling storage were determined. For this purpose, samples of minced were treated with different concentrations of AITC (100, 200, 300 and 500 ppm) and control no AITC added, the samples packaged and stored for 8 days at 4 °C. Fluctuations with no significant effect were noticed in TBARS values during the storage period. Simultaneously, AITC with higher concentration showed lower mesophilic aerobic counts compared to control and meat containing a low concentration of AITC. Moreover, compared to the beginning of storage, AITC decreased water holding capacity (WHC) of meat this can affect other physicochemical properties of meat. Further study needed to determine the effect of AITC on the physicochemical properties of meat and food products.

Fusarium wilt (Fusarium oxysporum f. sp. lycopersici), root-knot nematodes (Meloidogyne spp.), and purple nutsedge (Cyperus rotundus L.) are among the most damaging soilborne pests for tomato (Lycopersicon esculentum Mill.) production in the southeastern United States. Allyl isothiocyanate (allyl ITC) was evaluated as a potential fumigant alternative for control of soilborne pathogens, nematodes, and weeds. Shank- or drip-injected allyl ITC at rates ranging from 221 to 367 kg ha−1 exhibited excellent performance, reducing the recovery of total F. oxysporum from treated soils. Shank- or drip-injected allyl ITC at 367 kg ha−1 provided equivalent control of C. rotundus compared with 1,3-dichloropropene + chloropicrin and metam potassium, respectively. Totally impermeable film (TIF) did not further reduce the recovery of F. oxysporum and various nematodes from soil treated with allyl ITC compared with virtually impermeable film (VIF). However, TIF mulch significantly improved C. rotundus control versus shank- or drip-injected allyl ITC treatments under VIF mulch. Overall, allyl ITC is an effective methyl bromide alternative against F. oxysporum, C. rotundus, and plant-parasitic nematodes Criconemella spp. and Hoplolaimus spp. in plasticulture tomato production.

Angiogenesis, invasion, and metastasis are the main events of cancer cells. JAK1/STAT3 is a key intracellular signaling transduction path, which controls the growth, differentiation, apoptosis, invasion, and angiogenesis in various cancer cells. The existing study explored the impact of AITC on the JAK-1/STAT-3 pathway in DMBAinjected rat mammary tumorigenesis. The mammary tumor was initiated through a single dose of 25 mg DMBA/rat through a subcutaneous injection administered near the mammary gland. We observed decreased body weight and augmented the total number of tumors, tumor incidence, tumor volume, well-developed tumor, and histopathological abnormalities in DMBA-injected rats and that were modulated after being treated with AITC. Staining of mammary tissues showed a high accumulation of collagen in DMBAtreated rats and it was normalized by the AITC treatment. Moreover, DMBA-induced mammary tissues showed up-regulated expressions of EGFR, pJAK-1, pSTAT-3, the nuclear fraction of STAT-3, and its associated products like VEGF, VEGFR2, HIF-1α, MMP-2, and MMP-9 and the down-regulated expressions of cytosolic STAT-3 and TIMP-2. Oral administration of AITC on DMBAtreated rats inhibits angiogenesis and invasion through modified these angiogenic and invasive markers. The finding of the current study is further confirmed by molecular docking analysis that shows a strong binding interaction between AITC with STAT-3 and cocrystal structure of STAT3 glide energy of -18.123 and − 72.246 (kcal/mole), respectively. Overall, the results suggested that AITC inhibits activation of the JAK-1/STAT-3 path, which subsequently prevents angiogenesis and invasion. It was recommended that AITC might develop a beneficial effect against breast cancer.

Producing methane from anaerobic co-digestion of kitchen waste and waste activated sludge has been widely implemented in real-world situations. However, the fate and impact of allyl isothiocyanate (AITC), a main active component in cruciferous vegetables, in the anaerobic co-digestion has never been documented. This study therefore aims to provide such supports. Experimental results exhibited that AITC was degraded completely by microorganisms and served as a substrate to produce methane. As AITC increased from 0 to 60 mg/L, the maximum methane production decreased from 285.1 to 35.8 mL/g VS, and the optimum digestion time was also prolonged. The mechanism study demonstrated that AITC induced cell apoptosis by modifying the physicochemical properties of cell membrane, which resulted in inhibitions to the procedure of anaerobic co-digestion. The high-throughput sequencing showed that AITC enriched the microorganism for degradation of complex organic compounds such as Bacillus, but lessened anaerobes involved in hydrolysis, acidogenesis, and methanogenesis.

Poultry is the major sources of foodborne salmonellosis. Antibiotic resistance and a surge in zoonotic diseases warrant the use of natural alternatives. Glucosinolates (GLs) are naturally occurring antimicrobial compounds in rapeseed and canola. This study investigated the effect of feeding rapeseed, canola meal, and allyl isothiocyanate (AITC; Brassica secondary metabolites) on growth performance (GP), gut health, and the potential antimicrobial activity against nalidixic acid-resistant Salmonella Typhimurium (STNR) in chickens. A total of 640 one-day-old male Cobb 500 broilers were randomly allocated to 8 treatments with 8 replicated cages and 10 birds per cage. Dietary treatments were nonchallenge control (NC, corn-SBM based), challenge (Salmonella) control (CC), 10% rapeseed (10RS), 30% rapeseed (30RS), 20% canola meal (20CLM), 40% canola meal (40CLM), 500 ppm AITC (500AITC), and 1,500 ppm AITC (1500AITC). On d 1, all the birds except NC were orally challenged with STNR (7 log CFU/bird). The chickens were reared for 21 d, and their FI and BW were recorded weekly. Salmonella cecal colonization and fecal shedding were quantified, whereas organ translocation (OT) of STNR to the spleen, liver, and kidney was tested on 0, 3, 6, 13, and 20-d postchallenge (dpc). Data were subjected to one-way ANOVA, and the means were separated by Duncan's test, except mortality and OT data analyzed after transformation by square root of (n +1) (P < 0.05). Overall, feeding 30RS resulted in reduced BW (P = 0.003), BWG (P = 0.003), and FI (P = 0.001) compared to CC, 500AITC, and 1500AITC. Similarly, feeding 20CLM resulted in lower BW and BWG compared to CC (P < 0.05) and increased FCR compared to 1500AITC (P = 0.03). Feeding CC resulted in higher mortality compared to NC and 30RS (P = 0.03). Cecal colonization of STNR was reduced (P < 0.0001) for 30RS on 6 dpc and 500AITC on 6 and 13 dpc (P < 0.0001). Although no difference in gut permeability was observed 6 dpc (P > 0.05), OT of STNR population was the highest for CC in the spleen (P = 0.05). In the liver, 10RS showed reduced OT compared to 20CLM on 13 dpc (P = 0.03), whereas 30RS showed the lowest OT on 6 dpc in the kidney. Fecal shedding was lowest for 30RS on 6 dpc (P = 0.004). Histomorphology showed 30RS had the highest duodenum (P = 0.01) and jejunum (P = 0.02) villus height (VH) and VH to crypt depth (CD) ratio compared to the other treatments, whereas 1500AITC showed similar results to 30RS. Both 30RS and 1500AITC contained comparatively higher functional GL metabolites and were able to maintain gut health. Including higher levels of rapeseed or AITC in poultry feed can reduce Salmonella colonization in the feces and their translocation to other organs.

Environmental light pollution due to artificial light may increase the rate and severity of retinal diseases, and plant-based nutritional interventions with antioxidant properties have the potential to reverse this phenomenon. We aimed to investigate the potential effects of allyl isothiocyanate (AITC) against white light-emitting diode (LED)-induced retinal degeneration (RD) in the rats.Twenty-eight male rats were allocated as: (i) Control, (ii) LED, (iii) LED + AITC (10 mg/kg BW), (iv) LED + AITC (20 mg/kg BW). Rats were administered with AITC for 28 days, followed by two days of intense environmental LED light (750AITC lowered the serum and retina malondialdehyde (MDA) levels while significantly (In conclusion, four weeks of AITC administration to the rats showed specific protective effects against two days of intense LED light-induced retinal damage; through antiinflammatory, antioxidant, anti-apoptotic, and modulating mitochondrial metabolic pathways.

Glucosinolates are secondary plant metabolites occurring in Brassicaceae plants. Upon tissue disruption these compounds can be enzymatically hydrolyzed into isothiocyanates. The latter are very reactive and can react with nucleophiles during food processing such as cooking. Here, a novel type of glucosinolate degradation product was identified resulting from the reaction of the isothiocyanates sulforaphane and allyl isothiocyanate with thioglucose during aqueous heat treatment. The cyclic compounds were isolated and their structure elucidated by NMR spectroscopy and high resolution mass spectrometry as 4-hydroxy-3-(4-(methylsulfinyl)butyl)thiazolidine-2-thione and 3-allyl-4-hydroxythiazolidine-2-thione. Based on experiments with isotope-labeled reagents, the determination of the diastereomeric ratio and further reactions, a reaction mechanism was proposed. Finally, the formation of the two 3-alk(en)yl-4-hydroxythiazolidine-2-thiones was quantified in boiled cabbage samples with contents of 92 pmol/g respectively 19 pmol/g fresh weight using standard addition method.

The effects of yellow mustard (Brassica juncea) and its characteristic component allyl isothiocyanate (AITC) on the formation of 2-amino-y1-methyl-6-phenylimidazo[4,5-b] pyridine (PhIP) in roast beef patties and PhIP-producing model systems were investigated. The probable inhibitory pathways of AITC on PhIP formation were also investigated in the model systems. The results revealed that yellow mustard and AITC can reduce PhIP in roast beef patties up to 41.7% and 60.2%, respectively. The rate of inhibition of PhIP also reached 64.8% in the PhIP-producing model systems. Furthermore, AITC could react with creatinine and phenylalanine in the model system (reducing each by 15.0%%–23.7% and 31.4%–55.8%, respectively). AITC showed the great scavenging ability of free radical scavenging (up to 64.2%). AITC also reacted with the intermediate phenylacetaldehyde (16.9%–30.8%) and the final product PhIP (7.0%–24.6%). It is speculated that AITC can inhibit PhIP through competitive inhibition of precursors, blocking intermediate, free radical scavenging, and direct elimination of PhIP.

Environmental concerns related to synthetic pesticides and the emphasis on the adoption of an integrated pest management concept as a cardinal principle have strengthened the focus of global research and development on botanical pesticides. A scientific understanding of the mode of action of biomolecules over a range of pests is key to the successful development of biopesticides. The present investigation focuses on the in silico protein-ligand interactions of allyl isothiocyanate (AITC), a major constituent of black mustard (Brassica nigra) essential oil (MEO) against two pests, namely, Meloidogyne incognita (Mi) and Fusarium oxysporum f. sp. lycopersici (Fol), that cause severe yield losses in agricultural crops, especially in vegetables. The in vitro bioassay results of MEO against Mi exhibited an exposure time dependent on the lethal concentration causing 50% mortality (LC50) values of 47.7, 30.3, and 20.4 μg ml−1 at 24, 48, and 72 h of exposure, respectively. The study revealed short-term nematostatic activity at lower concentrations, with nematicidal activity at higher concentrations upon prolonged exposure. Black mustard essential oil displayed excellent in vitro Fol mycelial growth inhibition, with an effective concentration to cause 50% inhibition (EC50) value of 6.42 μg ml−1. In order to decipher the mechanism of action of MEO, its major component, AITC (87.6%), which was identified by gas chromatography–mass spectrometry (GC-MS), was subjected to in silico docking and simulation studies against seven and eight putative target proteins of Mi and Fol, respectively. Allyl isothiocyanate exhibited the highest binding affinity with the binding sites of acetyl cholinesterase (AChE), followed by odorant response gene-1 (ODR1) and neuropeptide G-protein coupled receptor (nGPCR) in Mi, suggesting the possible suppression of neurotransmission and chemosensing functions. Among the target proteins of Fol, AITC was the most effective protein in blocking chitin synthase (CS), followed by 2,3-dihydroxy benzoic acid decarboxylase (6m53) and trypsinase (1try), thus inferring these as the principal molecular targets of fungal growth. Taken together, the study establishes the potential of MEO as a novel biopesticide lead, which will be utilized further to manage the Mi–Fol disease complex.

The effects of some sulfur-containing compounds on the isomerization and degradation of lycopene, phytofluene, and phytoene under different thermal treatment conditions were studied in detail. Isothiocyanates such as allyl isothiocyanate (AITC) and polysulfides like dimethyl trisulfide (DMTS) had the effect on the configuration of PTF (phytofluene), PT (phytoene), and lycopene. The proportion of their naturally occurring Z-isomers (Z1,2-PTF and 15-Z-PT) decreased and transformed into other isomers including all-trans configuration, while Z-lycopene increased significantly after thermal treatment, especially for 5-Z-lycopene. The results showed that increase in heating temperature, time, and the concentration of DMTS and AITC could promote the isomerization reaction effectively to some extent. In addition, 15-Z-PT and the newly formed Z4-PTF were the predominant isomers in tomato at the equilibrium. Unlike the lycopene, which degraded significantly during heat treatment, the isomers of PTF and PT were stable enough to resist decomposition. Moreover, the isomerization of three carotenoids was enhanced, and the bioaccessibility of lycopene increased significantly with the addition of shii-take mushroom containing sulfur compounds, while there was no positive effect observed in that of PTF and PT.

Antiviral treatments inhibiting Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) replication may represent a strategy complementary to vaccination to fight the ongoing Coronavirus disease 19 (COVID-19) pandemic. Molecules or extracts inhibiting the SARS-CoV-2 chymotripsin-like protease (3CLPro) could contribute to reducing or suppressing SARS-CoV-2 replication. Using a targeted approach, we identified 17 plant products that are included in current and traditional cuisines as promising inhibitors of SARS-CoV-2 3CLPro activity. Methanolic extracts were evaluated in vitro for inhibition of SARS-CoV-2 3CLPro activity using a quenched fluorescence resonance energy transfer (FRET) assay. Extracts from turmeric (Curcuma longa) rhizomes, mustard (Brassica nigra) seeds, and wall rocket (Diplotaxis erucoides subsp. erucoides) at 500 µg mL−1 displayed significant inhibition of the 3CLPro activity, resulting in residual protease activities of 0.0%, 9.4%, and 14.9%, respectively. Using different extract concentrations, an IC50 value of 15.74 µg mL−1 was calculated for turmeric extract. Commercial curcumin inhibited the 3CLPro activity, but did not fully account for the inhibitory effect of turmeric rhizomes extracts, suggesting that other components of the turmeric extract must also play a main role in inhibiting the 3CLPro activity. Sinigrin, a major glucosinolate present in mustard seeds and wall rocket, did not have relevant 3CLPro inhibitory activity; however, its hydrolysis product allyl isothiocyanate had an IC50 value of 41.43 µg mL−1. The current study identifies plant extracts and molecules that can be of interest in the search for treatments against COVID-19, acting as a basis for future chemical, in vivo, and clinical trials.

Natural isothiocyanates (ITCs) are phytochemicals abundant in cruciferous vegetables with the general structure, R–N C S. They are bioactive organosulfur compounds derived from the hydrolysis of glucosinolates by myrosinase. A significant number of isothiocyanates have been isolated from different plant sources that include broccoli, Brussels sprouts, cabbage, cauliflower, kale, mustard, wasabi, and watercress. Several ITCs have been demonstrated to possess significant pharmacological properties including: antioxidant, anti-inflammatory, anti-cancer and antimicrobial activities. Due to their chemopreventive effects on many types of cancer, ITCs have been regarded as a promising anti-cancer therapeutic agent without major toxicity concerns. However, their clinical application has been hindered by several factors including their low aqueous solubility, low bioavailability, instability as well as their hormetic effect. Moreover, the typical dietary uptake of ITCs consumed for promotion of good health may be far from their bioactive (or cytotoxic) dose necessary for cancer prevention and/or treatment. Nanotechnology is one of best options to attain enhanced efficacy and minimize hormetic effect for ITCs. Nanoformulation of ITCs leads to enhance stability of ITCs in plasma and emphasize on their chemopreventive effects. This review provides a summary of the potential bioactivities of ITCs, their mechanisms of action for the prevention and treatment of cancer, as well as the recent research progress in their nanodelivery strategies to enhance solubility, bioavailability, and anti-cancer efficacy.

Brassica juncea L. (Brassicaceae) is widely used in making mustard oil. The petroleum ether extract of the seeds of this plant found to contain appreciable levels of secondary metabolites including phenolics, flavonoids, tannins and alkaloids that might be attributed to the good antioxidant activity of this extract. The extract exhibited broad antimicrobial activity against the tested pathogenic strains including Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, Klebsiella pneumonia, Candida albicans, Escherichia coli and Proteus vulgaris. 10 compounds were identified in the petroleum ether extract of Brassica juncea (L.) seeds using gas chromatography- electron ionization mass spectrometry (GC-EIMS) analysis. The identified constituents were 9,19-Cyclolanost-24-en-3-ol, acetate, (3.beta.)- Cycloartenyl acetate (34.77%), 3-Methylpentane (25.26%), Methyl cyclopentane (15.44%), octadec-9-enoic acid (oleic acid) (9.02%), Allyl isothiocyanate (3.43%), 3-Butenyl isothiocyanate (3.38%), hexanal (Caproaldehyde) (3.14%) and nonacosane (0.95%). In conclusion, Brassica juncea seeds could be used as valuable source of active constituents that possess remarkable biological activity and could be used in the field of medicine.

BACKGROUND Allyl isothiocyanate (AITC), a classic anti-inflammatory and antitumorigenic agent, was recently identified as a potential treatment for obesity and insulin resistance. However, little is known about its direct impact on the liver.

Background Transient receptor potential ankyrin 1 (TRPA1) channels may have a role in migraine as some substances known to cause headache activate the channel. In the craniovascular system such activation causes a calcitonin gene-related peptide (CGRP)-dependent increase in meningeal blood flow. TRPA1 channels in the endothelium of cerebral arteries cause vasodilation when activated. The headache preventive substance feverfew inhibits activation of TRPA1 channels. In this study we aim to compare and characterize the effect of the TRPA1 agonist allyl isothiocyanate (AITC) on the diameter of rat dural and pial arteries in vivo. Methods The genuine closed-cranial window technique in rats was used to examine changes in dural and pial artery diameter and mean arterial blood pressure (MABP) after intracarotid infusion of AITC. Blockade experiments were performed by intravenous infusion of olcegepant, HC-030031, sumatriptan or capsazepine immediately after infusion of AITC, in four different groups of rats. Results AITC caused a significant dilation of dural arteries, which was inhibited by HC-030031, olcegepant and sumatriptan, but not by capsazepine. In pial arteries AITC caused a significant dilation, which was not inhibited by any of the pre-treatments, suggesting a poor penetration of the blood-brain barrier or autoregulation due to dimethyl sulfoxide (DMSO) mediated decrease in MABP during HC-030031 infusion. AITC did not cause a significant change in MABP. Conclusion AITC causes dilation of dural arteries via a mechanism dependent on CGRP and TRPA1 that is sensitive to sumatriptan. AITC causes a small but significant dilation of pial arteries.

In vitro experiments showed that six isolates, belonging to different biovars, were equally susceptible to mustard essential oil (MEO) vapor. The proportion of cell mortality and colony growth inhibition were directly related to the dose of MEO. A 24 h exposure to 1 or 2 μL of AITC per liter of box volume resulted in mortality of 89% and 92% of cells, respectively, and exposure to 10 μL L−1 resulted in 100% cell mortality. The growth of developing colonies was reduced by 89% at 2 or 4 μL L−1 and by 96% at 6 or 10 μL L−1. The leakage of intracellular materials increased with increasing doses of MEO vapor. The decline in soil inoculum density by fumigation was directly related to the MEO dose. The initial inoculum density of approximately 3 × 105 colony-forming units per gram of soil declined by 67, 91 and 96% at fumigation doses of 25, 50 and 75 μL of allyl isothiocyanate (AITC) per liter of soil, respectively. The pathogen was not detected at a fumigation dose of 100 μL L−1. Disease control was also related to the fumigation dose. After 48 days, the bacterial wilt symptoms did not develop on the tomatoes when the soil was fumigated at 100 μL L−1 or higher.
