Saponins are amphiphilic molecules of pharmaceutical interest and most of their biological activities (i.e., cytotoxic, hemolytic, fungicide, etc.) are associated to their membranolytic properties. These molecules are secondary metabolites present in numerous plants and in some marine animals, such as sea cucumbers and starfishes. Structurally, all saponins correspond to the combination of a hydrophilic glycan, consisting of sugar chain(s), linked to a hydrophobic triterpenoidic or steroidic aglycone, named the sapogenin. Saponins present a high structural diversity and their structural characterization remains extremely challenging. Ideally, saponin structures are best established using nuclear magnetic resonance experiments conducted on isolated molecules. However, the extreme structural diversity of saponins makes them challenging from a structural analysis point of view since, most of the time, saponin extracts consist in a huge number of congeners presenting only subtle structural differences. In the present review, we wish to offer an overview of the literature related to the development of mass spectrometry for the study of saponins. This review will demonstrate that most of the past and current mass spectrometry methods, including electron, electrospray and matrix-assisted laser desorption/ionization ionizations, gas/liquid chromatography coupled to (tandem) mass spectrometry, collision-induced dissociation including MS3 experiments, multiple reaction monitoring based quantification, ion mobility experiments, and so forth, have been used for saponin investigations with great success on enriched extracts but also directly on tissues using imaging methods.

Modern mass spectrometry methods provide a huge benefit to saponin structural characterization, especially when combined with collision-induced dissociation experiments to obtain a partial description of the saponin (ion) structure. However, the complete description of the structures of these ubiquitous secondary metabolites remain challenging, especially since isomeric saponins presenting small differences are often present in a single extract. As a typical example, the horse chestnut triterpene glycosides, the so-called escins, comprise isomeric saponins containing subtle differences such as cis-trans ethylenic configuration (stereoisomers) of a side chain or distinct positions of an acetyl group (regioisomers) on the aglycone. In the present paper, the coupling of liquid chromatography and ion mobility mass spectrometry has been used to distinguish regioisomeric and stereoisomeric saponins. Ion mobility arrival time distributions (ATDs) were recorded for the stereoisomeric and regioisomeric saponin ions demonstrating that isomeric saponins can be partially separated using ion mobility on a commercially available traveling wave ion mobility (TWIMS) mass spectrometer. Small differences in the ATD can only be monitored when the isomeric saponins are separated with liquid chromatography prior to the IM-MS analysis. However, gas phase separation between stereoisomeric and regioisomeric saponin ions can be successfully realized, without any LC separation, on a cyclic ion mobility-enabled quadrupole time-of-flight (Q-cIM-oaToF) mass spectrometer. The main outcome of the present paper is that the structural analysis of regioisomeric and stereoisomeric natural compounds that represents a real challenge can take huge advantages of ion mobility experiments but only if increased ion mobility resolution is attainable.

Despite offering robust mechanical properties, polymer networks suffer from a lack of recyclability, reshaping, and healability. Designing stiff and remendable polymer networks that can repair under mild conditions remains a challenge to extend their field of applications. Herein, we describe a simple approach to design a nonisocyanate-based polyurethane network featuring multiresponsiveness (to humidity and temperature) and outstanding healing properties, as obtained by combining iminoboronate and boroxine chemistry. In spite of the presence of abundant dynamic bonds, the network has a high stiffness (Young's modulus of 551 MPa) and tensile strength (11 MPa). C?N iminoboronate and B-O boroxine exchange reactions at high temperature enable efficient network recycling over multiple cycles without compromising its properties. Collagen has a triple helix form, structured by a [-Gly-Xaa-Yaa-] repetition, where Xaa and Yaa are amino acids. This repeating unit can be post-translationally modified by enzymes, where proline is often hydroxylated into hydroxyproline (Hyp). Two Hyp isomers occur in collagen: 4-hydroxyproline (4Hyp, Gly-Xaa-Pro, substrate for 4-prolyl hydroxylase) and 3-hydroxyproline (3Hyp, Gly-Pro-4Hyp, substrate for 3-prolyl hydroxylase). If 4Hyp is lacking at the Yaa position, then Pro at the Xaa position should remain unmodified. Nevertheless, in literature 41 positions have been described where Hyp occurs at the Xaa position (?xHyp) lacking an adjacent 4Hyp. We report four additional positions in liver and colorectal liver metastasis tissue (CRLM). We studied the sequence commonalities between the 45 known positions of ?xHyp. Alanine and glutamine were frequently present adjacent to ?xHyp. We showed that proline, position 584 in COL1A2, had a lower rate of modification in CRLM than in healthy liver. The isomeric identity of ?xHyp, that is, 3- and/or 4Hyp, remains unknown. We present a proof of principle identification of ?xHyp. This identification is based on liquid chromatography retention time differences and mass spectrometry using ETD-HCD fragmentation, complemented by ab initio calculations. Both techniques identify ?xHyp at position 584 in COL1A2 as 4-hydroxyproline (4xHyp).

An increasing number of studies take advantage of ion mobility spectrometry (IMS) coupled to mass spectrometry (IMS-MS) to investigate the spatial structure of gaseous ions. Synthetic polymers occupy a unique place in the field of IMS-MS. Indeed, due to their intrinsic dispersity, they offer a broad range of homologous ions with different lengths. To help rationalize experimental data, various theoretical approaches have been described. First, the study of trend lines is proposed to derive physicochemical and structural parameters. However, the evaluation of data fitting reflects the overall behavior of the ions without reflecting specific information on their conformation. Atomistic simulations constitute another approach that provide accurate information about the ion shape. The overall scope of this review is dedicated to the synergy between IMS-MS and theoretical approaches, including computational chemistry, demonstrating the essential role they play to fully understand/interpret IMS-MS data.

The research area of nonlinear topologies of conjugated polymers (CPs) became a topic of growing interest due to their unique optoelectronic properties compared to CPs with linear topologies. Nonetheless, the field of the third-order nonlinear optical response (TONO) is still largely unexplored, although, theoretically, nonlinear CPs should give a higher TONO response. Therefore, linear CPs and a trigonal nonlinear CP were synthesized via a combination of a controlled chain-growth polymerization, post-polymerization reactions and a click reaction. Afterward, the TONO response of the CPs was investigated with the highly sensitive third-harmonic scattering technique using solvatochromism experiments. In contrast to what was expected, trigonal nonlinear CPs do not possess a higher TONO response compared to their linear counterparts.

International audience; Research toward bio-based polymers is an expanding field due to environmental concerns. A library of new aliphatic diol-diamide monomers with different chain lengths between the two amido groups was synthesized from sustainable levulinic acid and various linear aliphatic diamines (C 2-C 10). The monomers were prepared by diacylation of the diamines followed by reduction of the ketones to alcohols. These secondary diols were successfully recognized by an enzyme and polymerised in solution through a lipase-catalysed polycondensation. Poly(ester-co-amide)s with number-average molecular weights (M n) in the range of 1300-7200 g mol −1 were obtained, with dispersities between 1.5 and 1.8. An improvement of the M n value was observed upon increasing the monomer chain length. The variation of the aliphatic diol allows modulating the thermal properties of the final polymers. The glass transition temperatures were found to be between −23°C and 0°C. The polymers containing a long aliphatic segment (C 8-C 10) were able to crystallize (melting temperature of 90-97°C). TGA analyses showed that the ester linkages degrade at lower temperatures than the amide bonds. The stability of the latter was found to be higher when the number of methylene units increased from 2 (355°C) to 10 (378°C). Synthetic polymers occupy a unique place in the field of ion mobility mass spectrometry (IMS-MS). Indeed, due to their intrinsic dispersity, they have the asset to offer a broad range of homologous ions with different lengths that can be detected in several charge states. In addition, the gas-phase structure of polymer ions mostly depends on their ability to screen the adducted charges. Several works dealing with linear, cyclic, and star-shaped polymers have already shown that the gas-phase structure of polymer ions heavily relies on the polymer architecture, i.e., the primary structure. In the present work, we move a step further by evaluating whether a relationship exists between the primary and secondary structures of synthetic homo and copolymers. The IMS-MS experiments will be further complemented by MD simulations. To highlight the effectiveness of IMS separation, we selected isomeric homo and copolymers made of lactide (LA) and propiolactone (PL) units. In this way, the mass analysis becomes useless since isomeric comonomer sequences can coexist for any given chain length. An UPLC method was implemented in the workflow to successfully separate all PL-LA comonomer sequences before infusion in the IMS-MS instrument. The analysis of doubly charged copolymers showed that the comonomer sequence has an impact on the IMS response. However, this only holds for copolymer ions with precise sizes and charge states, and this is therefore not a rule of thumb.

Saponins are plant secondary metabolites. There are associated with defensive roles due to their cytotoxicity and are active against microorganisms. Saponins are frequently targeted to develop efficient drugs. Plant biomass containing saponins deserves sustained interest to develop high-added value applications. A key issue when considering the use of saponins for human healthcare is their toxicity that must be modulated before envisaging any biomedical application. This can only go through understanding the saponin-membrane interactions. Quinoa is abundantly consumed worldwide, but the quinoa husk is discarded due to its astringent taste associated with its saponin content. Here, we focus on the saponins of the quinoa husk extract (QE). We qualitatively and quantitively characterized the QE saponins using mass spectrometry. They are bidesmosidic molecules, with two oligosaccharidic chains appended on the aglycone with two different linkages, a glycosidic bond and an ester function. The latter can be hydrolyzed to prepare monodesmosidic molecules. The microwave-assisted hydrolysis reaction was optimized to produce monodesmosidic saponins. The membranolytic activity of the saponins was assayed based on their hemolytic activity that was shown to be drastically increased upon hydrolysis. In silico investigations confirmed that the monodesmosidic saponins interact preferentially with a model phospholipid bilayer, explaining the measured increased hemolytic activity.

Through the incorporation of chiral and achiral poly(3-alkylthiophenes) (P3ATs) into a star-shaped system, a well-defined supramolecular organization is obtained lacking the unfavorable linear lamellar structure typically obtained for P3AT. Through the combination of a controlled chain-growth polymerization and efficient postpolymerization and click reactions, well-defined star-shaped P3ATs with a low dispersity of 1.1 were obtained. The combination of UV–vis, circular dichroism (CD), atomic force microscopy (AFM), and transmission electron microscopy (TEM) measurements showed the formation of a strong (chiral) supramolecular organization into fibers, different and stronger than those obtained with the linear P3ATs. The fact that the width of the fibers is in good agreement with the width of a single star-shaped P3AT excludes the formation of a linear lamellar structure. In this study, poly((S)-3-(3′,7′-dimethyloctyl)thiophene) is synthesized via KCTCP, using a catechol-based external Ni-initiator. After characterization, the polymer's catechol functionality is deprotected and coupled with magnetite nanoparticles and the supramolecular organization is studied using UV/Vis, CD, AFM and Faraday measurements. We conclude that no long range supramolecular ordering is present in the hybrid material and that only interchain interactions between polymer chains attached on different nanoparticles are present. Also the effect of the new initiator and the branched monomer side chain on the controlled character of the KCTCP is investigated. It is found that a methyl group on the 3-position of the monomer sidechain already affects the controlled character of the polymerization. Also, a mechanism for the observed effects of the branched monomer and the new catechol-based initiator on the controlled character of the polymerization is proposed.

Well-defined poly(lactide)-based block copolymers were synthesized by a heterogeneous solvent and metal-free green approach by using organocatalysts in supercritical carbon dioxide (scCO2). We first report on the homopolymerization of both l - and d , l -lactide by organocatalyzed Ring-Opening Polymerization (o-ROP) by using a bicomponent organocatalyst composed of a thiourea derivative and various tertiary amines as cocatalysts. Control over the molar mass and dispersity is achieved until high monomer conversion although the polylactides are insoluble in the polymerization medium. The precision synthesis of PLA-based block copolymers from various CO2-phobic hydroxyl end-capped macroinitiators such as polyethylene glycol, polycaprolactone, polybutylene succinate and polyphosphoester was then reported. Merging scCO2 with this organocatalytic system provides therefore a unique tool for the design under solvent-free conditions of poly(lactide)-based block copolymers that are insoluble in scCO2.

Recent advances in molecular dynamics (MD) simulations have made it possible to examine the behavior of large charged droplets that contain analytes such as proteins or polymers, thereby providing insights into electrospray ionization (ESI) mechanisms. In the present study, we use this approach to investigate the release of polylactide (PLA) ions from water/acetonitrile ESI droplets. We found that cationized gaseous PLA ions can be formed via various competing pathways. Some MD runs showed extrusion and subsequent separation of polymer chains from the droplet, as envisioned by the chain ejection model (CEM). On other occasions the PLA chains remained inside the droplets and were released after solvent evaporation to dryness, consistent with the charge residue model (CRM). Following their release from ESI droplets, the nascent gaseous PLA ions were subjected to structural relaxation for several μs in vacuo. The MD conformations generated in this way for various PLA charge states compared favorably to experimental results obtained by ion mobility spectrometry-mass spectrometry (IMS-MS). The structures of all PLA ions evolved during relaxation in the gas phase. However, some macroion species retained features that resembled their nascent structures. For this subset of ions, the IMS-MS response appears to be strongly correlated with the ESI release mechanism (CEM vs. CRM). The former favored extended structures, whereas the latter preferentially generated compact conformers.

A poly(3-hexylthiophene)-block-perfluoropolyether-block-poly(3-hexylthiophene) (P3HT-b-PFPE-b-P3HT) triblock copolymer was synthesized by copper(I)-catalyzed alkyne–azide cycloaddition (CuAAC) in 95% yield. The copolymer was unequivocally characterized by size exclusion chromatography reaching 27 000 g mol−1 and nuclear magnetic resonance (NMR). As expected, diffusion-ordered spectroscopy NMR experiment revealed a lower diffusion coefficient (2.56 × 10-10 m2 s−1) for the P3HT-b-PFPE-b-P3HT triblock copolymer than the ethynyl end-functionalized P3HT (2.83 × 10-10 m2 s−1). Such a triblock copolymer exhibits i) a good thermal stability with no significant weight loss until 320 °C under air, ii) a glass transition of -50 °C due to the soft PFPE segment and iii) a melting peak arising from P3HT at 236 °C. The UV–vis absorption spectrum exhibits a similar absorption profile to that of pure P3HT with a vibronic structure. Analysis of terpolymer thin films by atomic force microscopy imaging and grazing incidence wide-angle X-ray scattering reveals the formation of P3HT crystallites and an “edge-on” orientation of the polymer chains towards the substrate. Preliminary photovoltaic studies on blends of this triblock copolymer combined with a fullerene derivative indicate that the devices using P3HT-b-PFPE-b-P3HT show an almost similar power conversion efficiency to commercial P3HT.

Herein, we developed a straightforward aqueous synthesis of unprecedented alternating peptide-peptoid copolymers via a Ugi four-component reaction applied to dipeptides. Functional peptide-peptoid copolymers as well as block copolymers were produced in a single step in the presence of acrylic acid and carboxylic acid-terminated poly(ethylene glycol). The thermoresponsiveness of these polymers is highlighted.

Cyclodextrin is commonly used in the formation of supramolecular complexes with an array of targets ranging from metal ion sensing to modulation of peptide properties. Many of the applications of these supramolecular complexes are measured by monitoring the modulation of the optical properties of a chromophore upon formation or breaking of the complex. The transposition of these complexes to the gas phase allows their study in a size selected manner, based on the isolation of specific stoichiometries to be analyzed independently. Forster Resonance Energy Transfer (FRET) is used to study the structure of these supramolecular complexes in solution phase. This motivates the use of action-FRET – a recently developed gas-phase extension of the FRET technique – to study the structure of gaseous supramolecular complexes. Here, the feasibility of performing action-FRET on a supramolecular assembly constituted by a chromophore tagged cyclodextrin receptor and a donor chromophore tagged amyloid beta peptide fragment is considered.

Holothuria (Platyperona) sanctori is a common sea cucumber of the Mediterranean Sea. Although its ecology and molecular phylogeny have been investigated in previous works, nothing is known about its saponin content, contrarily to other Mediterranean holothuroids. Saponins are bioactive triterpene glycosides that present interesting pharmacological/biological properties, and which can be used as taxonomic markers. This work highlights the different saponins produced by H. sanctori . Mass spectrometry ( MALDI-MS and MS/MS ) analyses highlighted a total of 18 different saponins, including eight new congeners. Presumptive molecular structures are proposed for all these molecules. A higher diversity of saponins is found in the body wall (12) than in the Cuvierian tubules (8). In addition to this qualitative difference, saponins were also 2–3 fold more concentrated in the Cuvierian tubules than in the integument. Our results demonstrate that H . sanctori , in accordance with its phylogenetic position, is the second species of Holothuria described to date possessing exclusively non-sulfated saponins. The species presents its own mix of saponins, acting as a true chemical signature.

This work deals with the synthesis and characterization of new diblock copolymers based on π-conjugated and depolymerizable units. These diblock copolymers are based on a regioregular poly(3-hexylthiophene) sequence associated with a sacrificial block, namely polyphthalaldehyde. The conjugated polymer was obtained by Grignard metathesis polymerization and end-capped by an alkynyl group while the depolymerizable segment was synthesized by an anionic cyclopolymerization from an azide moiety. Diblock copolymers with different molecular weights were then successfully synthesized via an alkyne–azide coupling reaction. Under specific conditions, these copolymers self-assemble into fibrillar nanostructures in thin films. The elimination of polyphthalaldehyde was carried out by thermal treatment, generating nanoporous poly(3-hexylthiophene) films. Fully bio- and CO2-sourced non-isocyanate polyurethanes (NIPUs) were synthesized by reaction of carbonated soybean oil (CSBO) either with biobased short diamines or amino-telechelic oligoamides derived from fatty acids to achieve respectively thermoset or thermoplastic NIPUs. Biobased carbonated vegetable oils were first obtained by metal-free coupling reactions of CO2 with epoxidized soybean oils under supercritical conditions (120 °C, 100 bar) before complete characterization by FTIR, 1H NMR, and electrospray ionization mass spectroscopy (ESI-MS). In a second step, biobased NIPUs were produced by melt-blending of the so-produced cyclocarbonated oil with the biobased aminated derivatives. The thermal and mechanical properties of resulting polymers were found to be depending on the cyclocarbonated vegetable oil/amine ratio. Algae biomass is nowadays considered as a unique reservoir of a huge variety of different molecules, such as lipids, carbohydrates, proteins and secondary metabolites, awaiting development in various fields. In particular, the valorization of the algae polysaccharide content is still in its infancy and intense research activity is starting to appear. Oligosaccharides with defined molecular weight and structures are extremely interesting for different applications such as plant biostimulation or biopesticide. Oligosaccharide are often prepared in medium to large scale upon chemical-based hydrolysis reactions from the extracted polysaccharides in harsh conditions (temperature and pH), leading to the loss of the fragile groups such as the sulfate functions in carrageenans. In the present work, microwave-based hydrolysis, as a green chemistry method, is optimized to produce bioactive oligosaccharides from carrageenans. Microwave hydrolysis process is first optimized on a commercially available carrageenan and further applied on carrageenans extracted from two algae, namely Kappaphycus alvarezii or Eucheuma spinosum that are produced in large quantities in the world and have a high content of carrageenans. We also develop a complete set of characterization methods, i.e. size exclusion chromatography, reducing sugar and residual sulfate determination and mass spectrometry, to perform the follow-up of the hydrolysis reaction. We here demonstrate that fast (5 min) and controlled hydrolysis of carrageenans can be achieved in pure water under microwave activation, while preserving the nature of the active sulfate groups (>95%).

End-functionalized poly(3-hexylthiophene) (P3HT) and poly(2,2,6,6-tetramethylpiperidinyloxy-4-yl methacrylate) (PTMA) were synthesized using controlled polymerization techniques and linked together to lead to a diblock copolymer structure via click cycloaddition. These diblock copolymers were further blended with carbon nanotubes (CNTs) to be used as cathodes in organic radical batteries. The performances of these batteries were studied during charge/discharge cycling at constant and variable current densities (C rate). In order to shed light on the importance of the diblock copolymer architecture, the results were compared to the capacities measured on electrodes made of blends of P3HT and PTMA homopolymers. Theoretical simulations were finally used to corroborate the good electrochemical properties of the synthesized diblock copolymers.

International audience; Environmental regulation and fossil resources depletion boost the search for new polymeric materials produced from biomass. Here, we report the synthesis of a novel diester bearing a pendant lactam unit from methyl levulinate and aspartic acid. The palladiumcatalyzed reductive amination/cyclization sequence was carefully optimized to afford the diacid with high yield (>95 %). In a second step, the compound was esterified to give the corresponding diester. The latter monomer was copolymerized with α-ω linear diols yielding polyesters with molecular weights up to 20.5 kg.mol-1 .

The new conjugated poly(4,4-bis(2-ethylhexyl)cyclopentadithiophene-alt-p-phenylene) (PCPDTPP) has been synthesized in a controlled way via catalyst transfer condensative polymerization (CTCP) of the corresponding biaryl monomer. Therefore, a series of Ni catalysts as well as (new) Pd-PEPPSI catalysts were employed in different Kumada CTCPs (KCTCPs) and Negishi CTCPs (NCTCPs). A series of tests confirmed the absence of undesired transfer and termination reactions and, hence, the controlled character of the chain-growth polymerization with Ni(dppe)Cl2 as catalyst. This resulted in polymers with low dispersities (D = 1.2), Mn of 33 kDa, and relatively good end-group control. Furthermore, formation of PCPDTPP-b-poly(3-octylthiophene) (P3OT) block copolymers was possible. Saponins are natural compounds presenting a high structural diversity whose structural characterization remains extremely challenging. Ideally, saponin structures are best established using nuclear magnetic resonance experiments conducted on isolated molecules. However, saponins are also increasingly characterized using tandem mass spectrometry (MS/MS) coupled with liquid chromatography, even if collision-induced dissociation (CID) experiments are often quite limited in accurately determining the saponin structure.We consider here ion mobility mass spectrometry (IMMS) as an orthogonal tool for the structural characterization of saponin isomers by comparing the experimental collisional cross sections (CCSs) of saponin ions with theoretical CCSs for candidate ion structures. Indeed, state-of-the-art theoretical calculations perfectly complement the experimental results, allowing the ion structures to be deciphered at the molecular level.We demonstrate that ion mobility can contribute to the structural characterization of saponins because different saponin ions present significantly distinct CCSs. Depending on the nature of the cation (in the positive ion mode), the differences in CCSs can also be exacerbated, optimizing the gas-phase separation. When associated with molecular dynamics simulations, the CCS data can be used to describe the interactions between the cations, i.e. HOur work contributes to resolve the relationship between the primary and secondary structures of saponin ions. However, it is obvious that the structural diversity and complexity of the saponins cannot be definitively unraveled by measuring a single numerical value, here the CCS. Consequently, the structural characterization of unknown saponins will be difficult to achieve based on IMMS alone. Nevertheless, we demonstrated that monodesmosidic and bidesmosidic saponins can be distinguished via IMMS.

To date, the controlled preparation of cyclic polymers is mainly achieved using click ligation strategies. The light-induced Diels-Alder click coupling has recently been shown to function as an attractive catalyst-free alternative to the copper-catalyzed alkyne-azide cycloaddition. However, the need for a light emitting system limits its applicability. To overcome this drawback, a sunlight-induced Diels-Alder reaction as an efficient route for the preparation of cyclic aliphatic polyesters in high yields and purity based on the same chemistry is demonstrated in the present contribution. The versatility and robustness of the proposed strategy against divergent atmospheric conditions is demonstrated. In light of the simplicity of the strategy presented here, it is suggested that the sunlight-induced Diels-Alder click-cyclization is a world-wide transposable process. A sunlight-induced Diels-Alder reaction is demonstrated for the preparation of cyclic polyesters. Under highly divergent atmospheric conditions, cyclic polyesters of high purity are obtained by taking advantage of the natural UV irradiation from the sun. We describe a simple and facile method for quantitatively converting bromine end-groups of well-defined polystyrene (PS, Mn,SEC = 4000 Da, Đ = 1.08) prepared by activators regenerated by electron transfer (ARGET) atom transfer radical polymerization (ATRP) into terminal alkenes by heating at 100 °C in dimethylformamide (DMF) without additional reagents. Subsequently, a facile quantitative post-functionalization of the terminal double bonds to various end functional polymers was performed via light-induced thiol–ene reactions. The quantitative end-group modifications as well as their thermal stability were assessed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-ToF-MS), nuclear magnetic resonance (1H NMR) spectroscopy and size-exclusion chromatography (SEC), evidencing the generated functional polystyrenes to be highly stable up to 200 °C for extended periods of time (24 h).

Linear polystyrenes carrying a mid-chain triazole, esters as well as terminal secondary bromines functionalities were synthesized via activators regenerated by electron transfer (ARGET) atom transfer radical polymerization (ATRP) using a bifunctional triazole containing initiator (3.8 kDa ≤ Mn,SEC ≤ 125 kDa, 1.08 ≤ ≤ 1.19) with the aim of understanding their behavior under thermal and thermomechanical stress. As reference materials-isolating the influence of individual functional groups-three polystyrene homopolymers carrying an ω-bromine chain-end functionality, α,ω-ester-bromine functionalities as well as α,ω-dibromine/mid-chain ester functionalities (2 kDa ≤ Mn,SEC ≤ 39 kDa, 1.06 ≤ ≤ 1.08) were prepared via ARGET ATRP. Furthermore, a well-defined triazole mid-chain functionalized block homopolymer, i.e. polystyrene-b-polystyrene (PS-b-PS, Mn,SEC = 4.4 kDa,= 1.08), was synthesized via a combination of ARGET ATRP and copper(i)-catalyzed azide-alkyne cycloaddition (CuAAC) as a reference material. Reference polymers without bromine and with ester/triazole functionalities were additionally investigated. Thermomechanical stress was applied to the polymers via small scale extrusion as well as a rheological assessment (G′(t), G′′(t)) under processing conditions. The thermally challenged polymers were analyzed by size-exclusion chromatography (SEC), matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-ToF-MS), proton nuclear magnetic resonance (1H NMR) and X-ray photoelectron spectroscopy (XPS) to arrive at a detailed image of the degradation susceptibility of individual functional groups, especially esters, bromines and triazole functions. The findings indicate an enhanced degradation of ATRP polymers via an accelerated ester cleavage due to HBr release at high temperatures accompanied by a concomitant molecular weight increase due to the formation of triazolium salts via the reaction of triazole units with bromine terminal chain ends. International audience; One of the main issues when using traveling wave ion mobility spectrometry (TWIMS) for the determination of collisional cross-section (CCS) concerns the need for a robust calibration procedure built from referent ions of known CCS. Here, we implement synthetic polymer ions as CCS calibrants in positive ion mode. Based on their intrinsic polydispersities, polymers offer in a single sample the opportunity to generate, upon electrospray ionization, numerous ions covering a broad mass range and a large CCS window for different charge states at a time. In addition, the key advantage of polymer ions as CCS calibrants lies in the robustness of their gas-phase structure with respect to the instrumental conditions, making them less prone to collisional-induced unfolding (CIU) than protein ions. In this paper, we present a CCS calibration procedure using sodium cationized polylactide and polyethylene glycol, PLA and PEG, as calibrants with reference CCS determined on a home-made drift tube. Our calibration procedure is further validated by testing the polymer calibration to determine CCS of numerous different ions for which CCS are reported in the literature.

Shape-memory polymers (SMPs) are a class of stimuli-responsive materials that have attracted tremendous attention in various applications, especially in the medical field. While most SMPs are thermally actuated, relating to a change of thermal transition (e.g., melting temperature), SMPs that can be actuated upon exposure to light are emerging. Recently, there has been new interest into multiple stimuli-responsive SMPs in order to cover the range of applications for these smart materials. Several families of polymers possessing various end-groups are characterized by ion mobility mass spectrometry (IMMS). A significant contribution of the end-groups to the ion collision cross section (CCS) is observed, although their role is neglected in current fitting models described in literature. Comparing polymers prepared from different synthetic procedures might thus, be misleading with the current theoretical treatments. We show that this issue is alleviated by comparing the CCS of various polymer ions (polyesters and polyethers) as a function of the number of atoms in the macroion instead of the usual representation involving the degree of polymerization. Finally, we extract the atom number density from the spectra which gives us the possibility to evaluate the compaction of polymer ions, and by extension to discern isomeric polymers.

Poly(3-hexylthiophene)s were synthesized with phosphonic ester, pyridine, thiol, and phenol end-groups using functionalized air-stable Ni initiators. The protected thiol- and phenol-functionalized P3HTs were converted into thiol and phenol P3HTs by quantitative postpolymerization reactions. 1H NMR and MALDI-ToF analysis showed very high degrees of functionalization and strong control over the polymerization except for the pyridine functionalized P3HT. The preparation of cyclic macromolecules has always represented a challenging task for polymer science, mainly because of difficulties in connecting chain extremities together. Initiated by the pioneering studies of Jacobson and Stockmayer, preparative pathways to cyclic polymers have been considerably improved within the last two decades thanks to the advent of both controlled polymerizations and efficient coupling reactions in organic chemistry. This Review aims to provide a critical up-to-date overview and illustrate the considerable efforts that have been made in the past few years to improve the availability of macrocycles for industrial and academic investigations through the use of the ring-closure approach. Particular attention is paid to methods for the preparation of monocycles over more complex architectures, since the latter are usually derived from the former.

Saponin analysis by mass spectrometry methods is nowadays progressively supplementing other analytical methods such as nuclear magnetic resonance (NMR). Indeed, saponin extracts from plant or marine animals are often constituted by a complex mixture of (slightly) different saponin molecules that requires extensive purification and separation steps to meet the requirement for NMR spectroscopy measurements. Based on its intrinsic features, mass spectrometry represents an inescapable tool to access the structures of saponins within extracts by using LC-MS, MALDI-MS, and tandem mass spectrometry experiments. The combination of different MS methods nowadays allows for a nice description of saponin structures, without extensive purification. However, the structural characterization process is based on low kinetic energy CID which cannot afford a total structure elucidation as far as stereochemistry is concerned. Moreover, the structural difference between saponins in a same extract is often so small that coelution upon LC-MS analysis is unavoidable, rendering the isomeric distinction and characterization by CID challenging or impossible. In the present paper, we introduce ion mobility in combination with liquid chromatography to better tackle the structural complexity of saponin congeners. When analyzing saponin extracts with MS-based methods, handling the data remains problematic for the comprehensive report of the results, but also for their efficient comparison. We here introduce an original schematic representation using sector diagrams that are constructed from mass spectrometry data. We strongly believe that the proposed data integration could be useful for data interpretation since it allows for a direct and fast comparison, both in terms of composition and relative proportion of the saponin contents in different extracts. Holothuroids produce triterpenoid saponins that act as chemical defenses against predators and parasites. These saponins interact with sterols of the plasma membranes, inducing the formation of pores and then cell lysis. To avoid such harms from their own saponins, holothuroids present specific sterols in their tissues. Despite the noxious cytotoxic effect of their chemical defenses, holothuroids host various associates that display specific adaptations to resist to saponin toxicity. Among them, symbiotic carapid fishes (i.e., pearlfishes) are resistant to ichthyotoxic saponins as they display no stress response and a survival time 45 times longer than free-living fishes without any specific gill adaptation. The present study aims at discovering the resistance mechanism(s) developed by carapids by addressing 3 hypotheses: carapids have (1) a mechanical barrier against the toxin constituted by a larger secretion of mucus than other fishes, (2) a bioactive barrier against the toxins constituted by a mucus effective on saponins and (3) a Δ7sterol tissue composition mimicking holothuroids that enable them to resist to saponins. First experiments showed that the mucus has no effective impact on saponin chemical structures. Mass spectrometry analyses showed that carapids, similarly to non-symbiotic fishes but contrary to their hosts, present a Δ5sterol tissue composition. However, two different procedures have shown that carapids produce six to ten times more mucus than control fishes, suggesting that a great quantity of mucus can protect carapids from their host’s saponins and acts as a mechanical barrier against toxins. Therefore, these results provide a new understanding of the carapids–holothuroids relationship.

Flavonoids are ubiquitous molecules in nature and are found in almost all plants, including fruits and vegetables. Although flavonoids are structurally similar, subtle differences in their structures lead to important changes in their biological activities. Over the years, mass spectrometry has become an ideal tool for the characterization of those important molecules. In particular, to overcome the challenge of structure assignment, tandem mass spectrometry has been used in numerous studies. In the present study, we submitted selected flavonoid 7- O-diglycosides to electrospray ionization to prepare different types of flavonoid ions, i.e. protonated, sodium-cationized and copper-cationized molecules. Most of the investigated reactions are already described in the literature in several papers and the aim of the present study is to present concise and coherent relations between collision-induced dissociation reactions, cationizing agents and flavonoid 7- O-diglycoside structures. Some insights into the reaction mechanisms and the role of the cationizing particles will also be attempted.

Tandem mass spectrometry is increasingly applied to polymer ions in the context of in-depth structural characterization. In particular, the well-known collision-induced dissociation (CID) methodology is nowadays advantageously used to characterize end-groups and to distinguish isomeric polymer ions. When applied to sodium-cationized polyesters, CID was demonstrated to mainly involve charge remote fragmentation processes and consecutive as well as competitive dissociation pathways. In the present report, an experimental and theoretical study of collision induced dissociation of isomeric sodium cationized polylactide was investigated. The investigated polymers were designed to obtain oligomers only differentiated on the basis of regioisomeric end-groups, using ortho-, meta-, and para-methylbenzyl alcohols as initiators. The CID spectra revealed the presence of competitive charge driven and charge remote fragmentations. In particular, the charge induced fragmentations allowed the minute distinction between the three isomeric end-groups. The intermediacy of an ion/neutral complex was proposed to account for the observed decomposition and some theoretical calculations were performed to support the proposal.

Plasma polymer films (PPFs) of allylamine are prepared varying the mean power injected in the plasma ( P mean ). The PPF behaviour in ethanol is studied through the analysis of the PPF physico-chemistry and the composition of the resulting ethanol solutions. By Visible light Spectroscopic Ellipsometry (VISSE), it is observed that the well-known decrease of the PPF thickness (Δ d ) upon immersion decreases as a function of P mean . This behaviour is attributed to an increase of the PPF cross-linking density ( χ ). By means of time-of-flight secondary ion mass spectrometry (ToF-SIMS) and further statistical processing of the data by principal component analysis (PCA), we propose a method to discriminate the samples according to χ . A significant increase in the PPF oxygen content is also observed after immersion which is ascribed to reaction between ethanol molecules and trapped radicals of the PPF network. On the other hand, taking into account the specificities of the gas chromatography–mass spectrometry (GC–MS) method, the resulting ethanol solutions are demonstrated to be free of chemical species containing a number of carbon atoms ranging from 7 to 40. However, Δ d (accounting for PPF restructuring) might come with the dissolution of PPF materials having less than 7 carbon atoms.

To avoid predation, holothuroids produce feeding-deterrent molecules in their body wall and viscera, the so-called saponins. Five tropical sea cucumber species of the family Holothuriidae were investigated in order to study their saponin content in two different organs, the body wall and the Cuvierian tubules. Mass spectrometry techniques (MALDI- and ESI-MS) were used to detect and analyze saponins. The smallest number of saponins was observed in Holothuria atra, which contained a total of four congeners, followed by Holothuria leucospilota, Pearsonothuria graeffei and Actinopyga echinites with six, eight and ten congeners, respectively. Bohadschia subrubra revealed the highest saponin diversity (19 congeners). Saponin mixtures also varied between the two body compartments within a given animal. A semi-quantitative approach completed these results and showed that a high diversity of saponins is not particularly correlated to a high saponin concentration. Although the complexity of the saponin mixtures described makes the elucidation of their respective biological roles difficult, the comparisons between species and between body compartments give some clues about how these molecules may act as predator repellents.

Sea cucumbers contain triterpene glycosides called saponins. We investigated the complex saponin mixture extracted from the common Mediterranean species Holothuria forskali. Two different body components were analyzed separately: the body wall (which protects the animal and is moreover the most important organ in terms of surface and weight) and the Cuvierian tubules (a defensive organ that can be expelled on predators in response to an attack). MALDI/MS and MALDI/MS/MS were used to detect saponins and describe their molecular structures. As isomers have been found in the Cuvierian tubules, LC/MS and LC/MS/MS were performed to identify each saponin separately. Twelve saponins have been detected in the body wall and 26 in the Cuvierian tubules. All the saponins from the body wall are also present in the Cuvierian tubules but the latter also contain 14 specific saponins. The presence of isomeric saponins complicated structure elucidation for the whole set but 16 saponins have been described tentatively. Among these, 3 had already been reported in the literature as holothurinosides A and C, and desholothurin A. Molecular structures have been proposed for the 13 others which, in the present work, have been provisionally named holothurinosides E, F, G, H, I, A(1), C(1), E(1), F(1), G(1), H(1) and I(1) and desholothurin A(1). The diversity and organ specificity of the saponins described here are much higher than what had been reported to date in any sea cucumber species.

Chiral recognition of enantiomers by host compounds is one of the most challenging topics in modern host-guest chemistry. Amongst the well-established methods, mass spectrometry (MS) is increasingly used nowadays, due to its low detection limit, short analysis time, and suitability for analyzing mixtures and for studying chiral effects in the gas phase. The development of electrospray-ionization (ESI) techniques provides an invaluable tool to study, in the gas phase, diastereoisomeric complex ions prepared from enantiomer ions and a chiral selector. This paper reports on an ESIMS and ESIMSMS study of the molecular mechanisms that intervene in the chiral-recognition phenomena observed between amino acids and a chiral crown ether. The modified crown ether, namely (+)-([18]crown-6)-2,3,11,12-tetracarboxylic acid, is used as the chiral selector when covalently bound on a stationary phase in liquid chromatography. This study was stimulated by the fact that, except with threonine and proline, consistent elution orders were observed, which indicates that the D enantiomers interact more strongly with the chiral selector than the L enantiomers. For proline, the lack of a primary amino group is likely to be responsible for the nonresolution of the two forms, whereas the second stereogenic center on threonine could explain the reversed elution order. In light of those observations, we performed mass spectrometry experiments to understand more deeply the enantiomeric recognition phenomena, both in solution by the enantiomer-labeled guest method and in the gas phase by gas-phase ligand-exchange ion/molecule reactions. The results have been further supported by quantum chemical calculations. One of the most interesting features of this work is the identification of a nonspecific interaction between proline and the crown ether upon ESIMS analysis.

This article deals with the Kumada Catalyst Transfer Polycondensation (KCTP) of 4,7-dioctylbenzo[2,1-b:3,4-b']dithiophene (BDP-Oct) using Ni(II) catalyst or In/cat combination. A combination of MALDI MS, GPC, and 31P NMR spectroscopy is used to reveal the failure of the KCTP of this particular monomer. Intermolecular transfer reactions to monomer appeared to prevent the formation of polymer. This result is remarkable, since isomeric benzo[1,2-b:4,5-b']dithiophene polymerizes in a controlled way. Saponins are secondary metabolites that are abundant and diversified in echinoderms. Mass spectrometry is increasingly used not only to identify saponin congeners within animal extracts but also to decipher the structure/biological activity relationships of these molecules by determining their inter-organ and inter-individual variability. The usual method requires extensive purification procedures to prepare saponin extracts compatible with mass spectrometry analysis. Here, we selected the sea star Asterias rubens as a model animal to prove that direct analysis of saponins can be performed on tissue sections. We also demonstrated that carboxymethyl cellulose can be used as an embedding medium to facilitate the cryosectioning procedure. Matrix-assisted laser desorption/ionization (MALDI) imaging was also revealed to afford interesting data on the distribution of saponin molecules within the tissues. We indeed highlight that saponins are located not only inside the body wall of the animals but also within the mucus layer that probably protects the animal against external aggressions. Graphical Abstract Saponins are the most abundant secondary metabolites in sea stars. They should therefore participate in important biological activities. Here, MALDI imaging is presented as a powerful method to determine the spatial distribution of saponins within the animal tissues. Ruthenium(ii) polyazaaromatic complexes have gained interest in recent decades as biomolecular tools, especially in the development of new phototherapeutic agents. These light emissive Ru complexes based on π-deficient ligands were first designed to allow a photo-induced electron transfer (PET) with the guanine base in DNA since their 3MLCT state is highly photo-oxidizing. Later the field of research was extended to proteins with the highlighting of a PET process with the tryptophan residue. This paper reports the kinetics of the luminescence quenching of [Ru(TAP)2phen]2+ by several selected peptide sequences containing at least one tryptophan residue. By using a peptide library we highlight the important parameters influencing the kinetics of the photo-electron transfer process, such as the net electrostatic charge and the number of tryptophan residues. The best peptide candidates were selected to study the formation of photo-products by MALDI-ToF mass spectrometry. A high photoreactivity of the [Ru(TAP)2phen]2+ complex was observed and multiple photoadducts were characterized, among them inter-peptidic adducts as well as intra-peptidic adducts.

Novel organocobalt complexes featuring weak C-CoL2 bonds (L = acetylacetonate) are prepared and used as sources of halomethyl radicals. They permit the precision synthesis of α-halide functionalized and telechelic polymers in organic media or in water. Substitution of halide by azide allows derivatization of polymers using the CuAAC click reaction.
