Polysaccharides derived from seaweeds can be used as biostimulants to enhance plant resistance to different stressors. In this study, we investigated the effects of applying low molecular weight polysaccharides (LPU) derived from Ulva prolifera with 14.2 kDa on the responses of wheat (Triticum aestivum) to osmotic stress. The results showed that osmotic stress simulated using polyethylene glycol inhibited seedling growth, whereas we observed increases in the fresh weights and shoot lengths of seedlings treated with polysaccharide for 120 h. Furthermore, we observed enhanced activities of antioxidant enzymes, and significant reductions in malondialdehyde content of 23.13%, 19.82%, and 20.04% in response treatment for 120 h with 0.01%, 0.03%, and 0.05% LPU, respectively, relative to those in the group treated with polyethylene glycol alone. In all treatments, expression of the P5CS gene was upregulated to promote proline accumulation. Moreover, after 120 h, exogenously applied LPU induced the expression of stress-related genes, including SnRK2, Wabi5, Wrab18, and Wdhn13. Collectively, these findings indicate that LPU might have the effect of regulating the abscisic acid-dependent pathway in wheat, thereby increasing seedling antioxidant capacity and growth. Application of LPU may accordingly represent an effective approach for enhancing the resistance to osmotic stress in wheat.

The buildup of organic matter and organisms on surfaces exposed to marine environments, known as biofouling, is a disruptive and costly process affecting maritime operations. Previous research has identified some of the surface characteristics particularly suited to the creation of antifouling and fouling-release surfaces, but there remains room for improvement against both macrofouling and microfouling organisms. Characterization of their adhesives has shown that many rely on oxidative chemistries. In this work, we explore the incorporation of the stable radical 2,2,6,6-tetramethylpipiderin-1-oxyl (TEMPO) as a component in an amphiphilic block copolymer system to act as an inhibitor for marine cements, disrupting adhesion of macrofouling organisms. Using polystyrene-b-poly(dimethylsiloxane-r-vinylmethysiloxane) block copolymers, pendent vinyl groups were functionalized with TEMPO and poly(ethylene glycol) to construct an amphiphilic material with redox active character. The antifouling and fouling-release performance of these materials was investigated through settlement and removal assays of three model fouling organisms and correlated to surface structure and chemistry. Surfaces showed significant antifouling character and fouling-release performance was increased substantially toward barnacles by the incorporation of stable radicals, indicating their potential for marine antifouling applications.

Green tides have been increasing in frequency and severity in coastal areas in recent years, and thus, the adverse effects of green tides on the environment have attracted much research attention. Allelopathy is one of the most significant effects of green tide algae on the surrounding organisms. In this study, a series of experiments were developed to fully investigate the allelopathic effects of the green tide alga Ulva linza on two common coastal phytoplankton species and to isolate and identify the chemical compositions of the allelochemicals. Our results indicated that the fresh tissue, dry powder, and aqueous extracts all inhibited the population growth of the two experimental phytoplankton species, indicating the occurrence of allelopathy; further analysis of the allelochemicals identified multiple kinds of fatty acids as allelopathic compounds, in which the most abundant ones were hexadecanoic acid; 9Z, 12Z, 15Z-octadecatetrienoic acid; and 9E-octadecenoic acid. Our findings enrich the database for research on allelopathy between marine green tide algae and phytoplankton. Both algae and phytoplankton could increase their own competitive abilities during bloom formation, thereby changing the phytoplankton community structure.

In order to analyze whether the marine macroalga Ulva lactuca can absorb and metabolize anthracene (ANT), the alga was cultivated with 5 µM ANT for 0–72 h, and the level of ANT was detected in the culture medium, and in the alga. The level of ANT rapidly decreased in the culture medium reaching a minimal level at 6 h, and rapidly increased in the alga reaching a maximal level at 12 h and then decreased to reach a minimal level at 48 h of culture. In addition, ANT induced an increase in hydrogen peroxide that remained until 72 h and a higher increase in superoxide anions that reach a maximal level at 24 h and remained unchanged until 72 h, indicating that ANT induced an oxidative stress condition. ANT induced an increase in lipoperoxides that reached a maximal level at 24 h and decreased at 48 h indicating that oxidative stress caused membrane damage. The activity of antioxidant enzymes SOD, CAT, AP, GR and GP increased in the alga treated with ANT whereas DHAR remained unchanged. The level of transcripts encoding these antioxidant enzymes increased and those encoding DHAR did not change. Inhibitors of monooxygenases, dioxygenases, polyphenol oxidases, glutathione-S-transferases and sulfotransferases induced an increase in the level of ANT in the alga cultivated for 24 h. These results strongly suggest that ANT is rapidly absorbed and metabolized in U. lactuca and the latter involves Phase I and II metabolizing enzymes.

As a biocompatible polymer, ulvan has applications in countless fields. Therefore, the following study intends to extract ulvan from Ulva fasciata, emphasizing its use for biomedical and industrial applications in the manufacture of nanofibrous webs. The extracted ulvan was characterized using FT-IR, DSC, XRD, GPC, and NMR. The extracted ulvan's FT-IR spectra confirmed that it is a sulfated polysaccharide. The HPLC analysis showed that the extracted ulvan is composed of rhamnose, xylose, glucose and glucuronic acid. NMR showed that the proton chemical shifts at 1.3 are due to methyl protons of rhamnose 3-sulfate in the ulvan samples. The X-ray diffractograms suggested that the extracted ulvan is semi-crystalline polymer with major crystalline reflection at 2θ of 29.4°. Deionized water has been successfully used to produce ulvan/polyvinyl alcohol (ulvan/PVA) nanofibers as an eco-friendly solvent. It was found that the ulvan-to-PVA (1:2) ratio results in nanofiber that is well handled and smooth. In addition to pretreated ones, the ulvan extracted without organic solvent pretreatment showed bead free nanofibers. It is concluded that pretreatment with organic solvent in ulvan extraction, particularly in the manufacture of nanofibers, is not recommended. In addition, the resulting nanofibrous mat has sufficient mechanical properties for various applications to be incorporated.

In this work, surface-modifying amphiphilic additives (SMAAs) were synthesized via hydrosilylation using various polymethylhydrosiloxanes (PMHS) and allyl-terminated polyethylene glycol monomethyl ethers (APEG) of varying molecular weights. The additives synthesized were incorporated into a hydrophobic, self-stratifying siloxane-polyurethane (SiPU) coating system to produce an amphiphilic surface. Contact angle experiments and atomic force microscopy (AFM), in a dry and hydrated state, were performed to assess changes in surface wettability and morphology. The antifouling and fouling-release (AF/FR) performances were evaluated by performing laboratory biological assays using the marine bacterium Cellulophaga lytica, the microalga Navicula incerta, the macroalga Ulva linza, the barnacle Amphibalanus amphitrite, and the marine mussel, Geukensia demissa. Several of the formulations showed improved AF/FR performance vs the base SiPU and performed better than some of the commercial standard marine coatings. Formulations containing SMAAs with a low grafting density of relatively high molecular weight PEG chains showed the best performance overall.

The outbreak of vibriosis from Vibrio parahaemolyticus (V. parahaemolyticus) is one of common pathogenic diseases found in the mariculture environment. In this study, the inhibitory effect of Ulva fasciata (U. fasciata) on the culturability, motility, and biofilm formation of V. parahaemolyticus ATCC17802 was examined by co-culturing system. Results showed that both of secretion and live tissue of U. fasciata could convert culturable V. parahaemolyticus ATCC17802 to non-culturable, both reaching more than 99% of inhibition rate after 3-day co-culture, and higher density (12 g L−1) of U. fasciata exhibited stronger inhibition. The twitching behavior of V. parahaemolyticus ATCC17802 was more easily affected by U. fasciata than the swimming behavior after 3-day co-culture, with the inhibitory rates varying at the ranges of 1.70–30.29% (twitching behavior) and 10.06–44.86% (swimming behavior) under the different environmental factors (salinity, NO3−-N and PO43−-P concentrations), but no significant correlation was found. The greatest inhibition effect on V. parahaemolyticus ATCC17802 biofilm formation occurred at 12 h, with inhibition rates at the range of 11.03–67.10 %, while there was still no significant correlation between inhibition rate and the three environmental factors. The different environmental factors might induce U. fasciata to excrete different levels of secondary metabolites, which caused the various inhibitory effect on the cultivability, motility, and biofilm formation of V. parahaemolyticus ATCC17802.

Polyelectrolyte multilayer (PEM) assembly is a versatile tool to construct low-fouling coatings. For application in the marine environment, their structure needs to be stabilized by covalent linkage. Here, we introduce an approach for spin coating of silane-based sol-gel chemistries using layer-by-layer assembly of polysaccharide-based hybrid polymer coatings (LBLHPs). The silane sol-gel chemistry allows the films to be cross-linked under water-based and mild reaction conditions. Two different silanes were used for this purpose, a conventional triethoxymethyl silane and a de novo synthesized zwitterionic silane. The polysaccharide-silane hybrid polymer coatings were thoroughly characterized with spectroscopic ellipsometry, water contact angle (WCA) goniometry, attenuated total reflection-Fourier transform infrared spectroscopy, and atomic force microscopy. The coatings showed good stability in seawater, smooth surfaces, a high degree of hydration, and WCAs below or close to the Berg limit. Amphiphilic surfaces, containing both hydrophilic and hydrophobic domains, offer desirable performance for many applications such as marine coatings or anti-icing purposes. This work explores the effect of the concentration of amphiphilic moieties on converting a polyurethane (PU) system to a coating having fouling-release properties. A novel amphiphilic compound is synthesized and added at increasing amounts to a PU system, where the amount of the additive is the only variable in the study. The additive-modified surfaces are characterized by a variety of techniques including ATR-FTIR, XPS, contact angle measurements, and AFM. Surface characterizations indicate the presence of amphiphilic domains on the surface due to the introduction of the self-stratifying amphiphilic additive. The fouling-release properties of the surfaces are assessed with three biological assays using Ulva linza, Cellulophaga lytica, and Navicula Incerta as the test organisms. A change in the fouling-release performance is observed and plateaued once a certain amount of amphiphilicity is attained in the coating system, which we call the critical amphiphilic concentration (CAC).

Green macroalgae comprise significant amount of structural carbohydrates for their conversion to liquid biofuels. However, it generally relies on species characteristics and the variability in seasonal profile to determine its route for bioprocessing. Hence, this study was conducted to analyze the indigenous marine macroalgal strain (Ulva prolifera) with respect to periodic trend and reducing sugar extraction. Consequently, in our investigation, the monthly variation in sugar profile and bioethanol yield was assessed between the monsoon and post-monsoon seasons, of which relatively high reducing sugar and fermentative bioethanol yield of about 0.152 ± 0.009 g/gdw and 6.275 ± 0.161 g/L was obtained for the October-month isolate (MITM10). Thereafter, the biochemical profile of this collected biomass (MITM10) revealed carbohydrate 34.98 ± 3.30%, protein 12.45 ± 0.49%, and lipid 1.93 ± 0.07%, respectively, on dry weight basis. Of these, the total carbohydrate fraction yielded the maximum reducing sugar of 0.156 ± 0.005 g/gdw under optimal conditions (11.07% (w/v) dosage, 0.9 M H2SO4, 121°C for 50 min) for thermal-acid hydrolysis. Furthermore, the elimination of polysaccharides was confirmed using the characterization techniques scanning electron microscopy (SEM) and Fourier transform infrared (FT-IR) spectroscopy. Therefore, the present thermochemical treatment method provides a species-specific novel strategy to breakdown the macroalgal cell wall polysaccharides that enhances sugar extraction for its utilization as an efficient bioenergy resource.

The hydrolytic activity of different types of polyphosphates, and the induction of soluble pyrophosphatase (sPPase, EC 3.6.1.1) activity have been assessed in cell extracts of nutrient limited green seaweed Ulva pseudorotundata Cormaci, Furnari &amp, Alongi subjected to different phosphorus regimes. Following a long period of nutrient limitation, the addition of different types of (poly)phosphates to artificial seawater enhanced growth rates on fresh weight and area, but not on dry weight bases. Chlorophyll and internal P content were affected by P supply. In contrast, internal soluble reactive P was kept low and was little affected by P additions. Soluble protein content increased in all treatments, as ammonium was added to prevent N limitation. The C:N:P atomic ratio revealed great changes depending on the nutrient regime along the experiment. Cell extracts of U. pseudorotundata were capable of hydrolyzing polyphosphates of different chain lengths (pyro, tripoly, trimeta, and polyphosphates) at high rates. The sPPase activity was kept very low in P limited plants. Following N and different kind of P additions, sPPase activity was kept low in the control, but slightly stimulated after 3 days when expressed on a protein basis. The highest activities were found at the end of the experiment under pyro and polyphosphate additions (7 days). The importance of alternative P sources to phosphate and the potential role of internal soluble pyrophosphatases in macroalgae are discussed.

Polysaccharide multilayers consisting of hyaluronic acid and chitosan were prepared by layer-by-layer assembly. To be used in seawater, the multilayers were crosslinked to a different degree using thermal or chemical methods. ATR-FTIR revealed different amide densities as a result of the crosslinking conditions. AFM showed that the crosslinking affected the roughness and swelling behavior of the coatings. The stability and degradability of the multilayers in aqueous environments were monitored with spectroscopic ellipsometry. The resistance of the coatings against non-specific protein adsorption was characterized by SPR spectroscopy. Settlement assays using Ulva linza zoospores and removal assays using the diatom Navicula incerta showed that the slowly degradable coatings were less prone to fouling than the strongly crosslinked ones. Thus, the coatings were a suitable model system to show that crosslinking the multilayers under mild conditions and equipping the coatings with controlled degradation rates enhances their antifouling and fouling-release properties against marine fouling organisms.

This study investigates conformational aspects of ulvans (F2) and their polycarboxyl derivatives obtained through periodate–chlorite oxidation (C3) followed by DEAE-Sephacel fractioning (C3b and C3c). Size exclusion chromatography coupled with laser light scattering and viscometric detection, in addition to circular dichroism (CD) and molecular modeling analyses, suggested that F2 had a compact sphere conformation with a helical motif as secondary structure. In contrast, all the polycarboxyl ulvans showed a random coil conformation, although C3c (NaSO3− 21.0%; COO− 1.81 mmol·g−1; Mw 18 kg·mol−1) had a more rigid and constrained backbone than C3 (NaSO3− 21.0%; COO− 1.81 mmol·g−1; Mw 49 kg·mol−1), largely due to its higher sulfate and carboxyl content. Despite the higher ionic character of C3c, its anticoagulant activity (ACA), determined by activated partial thromboplastin time (APTT) assay, was not improved compared to that of C3. Moreover, C3b (NaSO3− 14.1%; COO− 1.23 mmol·g−1; Mw 8.1 kg·mol−1) had higher activity than F2 (NaSO3− 20.6.%; COO− 0.36 mmol·g−1; Mw 123 kg·mol−1), even with its lower sulfate content and molar mass. These results suggest that the ACA of polycarboxyl ulvans relies on carboxyl and sulfate content and may depend, in addition, on a proper flexible conformation.

Cardiovascular diseases are key complications primarily associated with hyperthyroidism disorders. The present study sought to ameliorate hyperthyroidism-mediated cardiovascular inflammations and related oxidative stress paradigms in experimental rats using the broadly distributed green seaweed Ulva fasciata. Forty-eight adult male albino rats were recruited and randomly classified into six groups. Hyperthyroidism was stimulated using L-thyroxine sodium at a dose of 100 μg/kg i.p. for 3 weeks daily. Further, 200 mg/kg b.wt. concentration of the U. fasciata methanolic (U. fasciata-MeOH) extract was the recommended dose and administrated orally to the hyperthyroid rats. The standard commercial drug "propranolol hydrochloride" was also tested at a dose of 10 mg/kg i.p. to compare the findings obtained from the seaweed extract. A combined treatment with the U. fasciata-MeOH extract and propranolol hydrochloride was also assessed. Our results implied that the treatment of hyperthyroid rats with the U. fasciata-MeOH extract significantly reduced serum levels of the thyroid hormones T3 and T4, proinflammatory cytokines (TNF-α, MPO, and CRP), triglycerides and total cholesterol, as well as the cardiac biomarkers CK-MB, LDH, and troponin to thresholds close to those of the standard drug. In addition, levels of high-density lipoprotein cholesterol (HDL-C) and interleukin 10 (IL-10) were significantly upregulated. Hyperthyroid rats only treated with propranolol hydrochloride, or with a combination of the drug and the seaweed extract, conferred the same observations. Histopathological architecture boosted our interesting findings where the myocardium tissues in hyperthyroid rats, administrated the U. fasciata-MeOH extract or/and propranolol hydrochloride, exhibited more or less a normal structure as the control, reflecting the potential cardiovascular recovery exerted by this seaweed extract. In vitro DPPH, ABTS, and FRAP antioxidant assays of the U. fasciata-MeOH extract showed an outstanding ROS-scavenging potential. HPLC analysis of the U. fasciata-MeOH extract unraveled an inestimable valuable array of phenolics (mainly p-coumaric, gallic, ferulic, chlorogenic, and syringic acids) and flavonoids (hesperidin, kaempferol, catechin, quercetin, and rutin). Conclusively, the seaweed U. fasciata is a profitable source of antioxidant polyphenolics characterized by having a pharmaceutical potential against hyperthyroidism-linked cardiovascular inflammations and oxidative stress patterns due to their substantial free radical quenching properties, and also via regulating the signalling pathways of the proinflammatory, lipid profile, and cardiac biomarkers.

Marine green algae are valuable sources of diverse health-promoting bioactive components. Ulvan is suitable for biological applications due to its unique structure and numerous bioactivities. Here, the complex structure of ulvan from Ulva pertusa was analyzed using specific ulvan lyase degradation, MS, and NMR detection. Its structure mainly consists of →4)-β-d-GlcA-(1 → 4)-α-l-Rha3S-(1 → and →4)-β-d-Xyl-(1 → 4)-α-l-Rha3S-(1 → repeating units. Small amounts of →4)-α-l-IdoA-(1 → 4)-α-l-Rha3S-(1 → unit also exist. In addition, a minor number of branches, a single GlcA, and a long branch containing GlcA-Glc were linked to Rha3S. The antiviral activity of the ulvan and its degraded fragments were further investigated. Ulvan (1068.2 kDa) and ulvan-F1 (38.5 kDa) with relatively high molecular weight showed potency of inhibiting the infection and replication of vesicular stomatitis virus (VSV) at 100 μg/mL, the inhibition rate of VSV replication was 40.75% and 40.13%, respectively. These results indicated that ulvan has potential as a functional agent.

Polyacrylamide (PAM) has been used as a coagulant aid in water treatment process for past decades, but it has caused great damages to human nervous system. Developing new coagulant aid with high biological safety is urgently demanded. This study provides a natural biomacromolecule coagulant aid with good biosecurity-Enteromorpha prolifera polysaccharide (Ep). Its coagulant aid efficiency and mechanism were investigated in terms of organics removal, floc properties and membrane fouling degree. In addition, contrast experiments were conducted with PAM to evaluate its potential of industrial applications. Results showed that organics removal could be increased by 23% when 0.3 mg/L Ep was used, which exhibited comparable aid effects to PAM. Due to the bridging-sweep aid role of Ep, flocs sizes, growth rate and recovery factor reached 470 μm, 62.6 μm/min and 0.492, respectively, while only 170 μm, 14.0 μm/min and 0.326 were obtained by PAM. Additionally, flocs exhibited more porous and multi-branched structures when Ep was applied, which caused less ultrafiltration membrane fouling (eventual J/J0 value = 0.52). As a result, Ep could be considered as a potential substitute of PAM, since better biosecurity, higher organics removal and lower membrane fouling could be obtained simultaneously by Ep addition.

Smart films were produced with ulvan extracted from the green seaweed Ulva. lactuca at different extraction conditions, by varying the glycerol or sorbitol in specific plasticizer concentration. Chemical composition and molecular weight characteristics of ulvans were carried out by GLC and HPSEC analysis, respectively. Regardless of the extraction procedure, the investigation revealed that ulvans were selected as reproducible and suitable materials of stimuli-responsive due to its peculiar chemical composition and self-aggregation molecular behavior. When using the terminology of stimuli-responsive system, we declare that the physico-chemical, barrier, optical, and mechanical characteristics of ulvan films system are strongly responsive to extraction conditions and to plasticizer type and concentration. As plasticizer concentration increased, thickness, moisture content (MC), water solubility, water vapor permeability (WVP), transparency, elongation at break (EAB), usually UV and visible light protection of ulvan films increased, while, their tensile strength (TS) decreased significantly. The films prepared with glycerol exhibited better solubility, transparency and mechanical properties, than those with sorbitol. However, the films formed with sorbitol had the greatest moisture resistance and more effective vapor, visible and UV light barrier as well as the lowest solubility. The FTIR spectra evidenced the expected outcome of the chemical interaction and miscibility kind between polysaccharides and plasticizers.

Ulvan, a sulfated polysaccharide extracted from the green seaweed genus Ulva, has bioactive properties including an immunomodulating capacity. The immunomodulatory capacity of ulvan from Ulva ohnoi, however, has not been assessed in detail. We depolymerised purified ulvan from U. ohnoi to obtain a range of molecular weight fractions (Mw 7, 9, 13, 21, 209 kDa), which were characterised by constituent sugar analysis, SEC-MALLS, and NMR. Ulvan fractions contained 48.8–54.7 mol% rhamnose, 32.5–35.9 mol% glucuronic acid, 4.5–7.3 mol% iduronic acid, and 3.3–5.6 mol% xylose. 1H and 13C NMR was consistent with hydrolysis occurring at the anomeric centre without further modification to the oligosaccharide structure. The in vitro immunomodulatory effect of ulvan fractions was quantified by measuring levels of inflammatory-mediating signalling molecules released from LPS-stimulated RAW264.7 murine macrophages. All ulvan fractions showed no toxicity on RAW264.7 cells at concentrations below 100 μg mL−1 over 48 h. Secreted interleukin-10 and prostaglandin E2 demonstrated an anti-inflammatory effect by higher molecular weight ulvan fractions at 100 μg mL−1. To a lesser extent, these fractions also enhanced the LPS-induced inflammation through minor increases of IL-1β and IL-6. This study confirms that ulvan from U. ohnoi has a mild in vitro immunomodulatory effect.

The present study investigated dietary Enteromorpha prolifera polysaccharide (EPP) on the growth performance, body composition and non-specific immunity of crucian carp Carassius auratus. Crucian carps with body weight of 51.24 ± 4.08 g were randomly divided into five groups: one group was fed with basic diet, while the other four groups were fed with diets containing 20, 40, 60 and 80 g/kg EPP. After 60 days of feeding, dietary administration of 40 g/kg EPP increased the body weight gain rate, feed conversion ratio, specific growth rate, body crude protein content, intestine digestive enzyme activities, serum lysozyme activity, acid phosphatase activity, alkaline phosphatase activity, complement 3 level, superoxide dismutase activity and catalase activity and decreased body fat content and serum total cholesterol, triacylglycerol, alanine aminotransferase activity and glutamic oxaloacetic transaminase activity compared with those of the control group. All levels of EPP improved the resistance to Aeromonas hydrophilia compared with the control group. The results indicated that EPP could promote the growth of crucian carp and improve their disease resistance and may be used as a dietary supplement for crucian carp.

In this study, purification of polysaccharide ulvan by anion exchange chromatography was prepared, and the major polysaccharide fraction (FU) was collected at 1.0 M NaCl elute by anion exchange chromatography, then high sulfate content purified ulvan (HFU) was prepared with sulfur trioxide/N,N-dimethylformamide (SO3-DMF) in formamide. The antioxidant activity and the antihyperlipidemic activity of HFU in mice were determined. The results showed that treatment with HFU could improve the antioxidant and antihyperlipidemic activity. Compared with the hyperlipidemic group, the antihyperlipidemic activity of HFU (125 mg/kg) was the strongest, TC concentrations were significantly decreased by 26.7% (P

This study investigated the effects of dietary Enteromorpha powder supplementation on the productive performance, egg quality, and antioxidant performance of Zi geese during the late laying period. Three hundred twelve Zi geese (1 yr old) were randomly allocated into 2 cohorts to form a control group and an experimental group (with each cohort including 6 replicates and 21 female geese and 5 male geese in each replicate). The control group was fed a basal diet, and the experimental group was fed a diet containing 3% Enteromorpha powder. The data showed that Enteromorpha powder supplementation significantly improved egg production, laying rate, average daily egg weight (P < 0.01), and egg yolk color (P < 0.05). Supplementation decreased the ADFI and feed conversion rate (P < 0.01). Compared with the control group, glutathione peroxidase (GSH-Px) activity was significantly higher in serum and ovary tissue (P < 0.05), but GSH-Px activity was lower in liver tissue (P < 0.01). Malondialdehyde was reduced in liver and ovary tissue (P < 0.05) in the Enteromorpha powder supplementation group. Meanwhile, the expression of the CAT gene was significantly upregulated in the liver (P < 0.01) in the Enteromorpha group. These results indicate that dietary Enteromorpha powder supplementation improved productive performance and reduced the level of lipid peroxidation in Zi geese during the late laying period.

Ulvan was the polysaccharide (U) from a large edible green algae Ulva pertusa. In this study, phosphorylated ulvan (PU) was prepared by the sodium trimetaphosphate -sodium tripolyphosphate method. Antioxidant and antihyperlipidemic effects of U and PU were investigated employing in vivo systems. The PU was confirmed by IR, 31P NMR and 13C NMR spectra. And in addition, we found that the PU3 group at the dose of 500 mg/kg showed stronger antioxidant activity. Compared with hyperlipidemia group, it significantly increased GSH-Px (34.29%; P

A useful growth process model for Ulva prolifera with light limitation considering the nutrient effect was proposed to better understand the development of macroalgae bloom. The interrelationship between light and nutrient limitation was demonstrated to obtain the mechanism. As a case study, thresholds of light and nutrients for different stages of Ulva prolifera growth were estimated using the proposed model. Limits of light intensity, nitrate, and phosphate concentration on the initial growth were found to be 40.0 W m−2, 6.5 μmol L−1, and 0.27 μmol L−1, respectively. The minimal light intensity for initial growth was found to increase monotonously with decrease in the nutrient concentration. It was also observed that the minimal light intensity for growth increases as the density of Ulva prolifera increases and the nutrient concentration decreases. Similarly, the minimal nutrient concentration for initial growth increases monotonously with decrease in the light intensity. In addition, the minimal nutrient concentration for growth increases with increase in the density of Ulva prolifera and decrease in the light intensity. It was demonstrated that the phosphate limitation on the initial growth of Ulva prolifera seedling can occur in most coastal waters of the southern Yellow Sea and a tendency of approaching the phosphate limitation on the growth of the floating thalli of Ulva prolifera exists. Evidence was provided to support the argument that the macroalgae thalli from aquaculture rafts, rather than that from seedlings or spores, can contribute to the original biomass of the floating green Ulva prolifera in the southern Yellow Sea. The model presented in this study can provide new insights into the interrelationship between the light and nutrient limitation, as well as into the growth mechanism of floating seaweeds. It can also provide a more accurate prediction of seaweed growth in light- and nutrient-limited environments.

The sequestration of elevated atmospheric CO2 levels in seawater results in increasing acidification of oceans and it is unclear what the consequences of this will be on seaweed ecophysiology and ecological services they provide in the coastal ecosystem. In the present study, we examined the physiological and biochemical response of intertidal green seaweed Ulva compressa to elevated pCO2 induced acidification. The green seaweed was exposed to control (pH 8.1) and acidified (pH 7.7) conditions for 2 weeks following which net primary productivity, pigment content, oxidative status and antioxidant enzymes, primary and secondary metabolites, and mineral content were assessed. We observed an increase in primary productivity of the acidified samples, which was associated with increased levels of photosynthetic pigments. Consequently, primary metabolites levels were increased in the thalli grown under lowered pH conditions. There was also richness in various minerals and polyunsaturated fatty acids, indicating that the low pH elevated the nutritional quality of U. compressa. We found that low pH reduced malondialdehyde (MDA) content, suggesting reduced oxidative stress. Consistently we found reduced total antioxidant capacity and a general reduction in the majority of enzymatic and non-enzymatic antioxidants in the thalli grown under acidified conditions. Our results indicate that U. compressa will benefit from seawater acidification by improving productivity. Emerging knowledge shows the importance of preweaning nutrition on programming the gastrointestinal microbiome and development of the gut barrier function. The aim of this study was to assess the effects of supplementing cow milk with either intact dried Ulva sp., Ascophyllum nodosum, or Saccharina latissima on growth performance and several gut health parameters of preweaning dairy calves. Forty male Holstein calves were selected based on birth weight (41 ± 4 kg) and plasma Brix percentage (≥8.7%) at d 2 of life. From d 2 to d 42 of life, the control calves (n = 10) were fed with cow milk (8 L/d) without seaweed supplementation, and the experimental calves were fed with cow milk (8 L/d) supplemented with either Ulva sp. (n = 10), A. nodosum (n = 10), or S. latissima (n = 10) at a concentration of 50 g/8 L of cow milk per day (i.e., 5% on a dry matter basis). Calves were weighed every week, and body weight gain and calf starter intake were monitored weekly. At d 42 ± 3 of life, calves were slaughtered. The organ weights and digesta pH from the reticulorumen, mid- and end small intestine, and mid-colon were recorded. A tissue sample (5 cm) collected from the mid-small intestine was analyzed for histomorphology. Digesta from the mid-small intestine and mid-colon were analyzed for lactobacilli, Escherichia coli, and Enterobacteriaceae, and short-chain fatty acid profile. Weight gain of the calves was not affected by seaweed supplementation. Proportional organ weights were not affected by seaweed supplementation except for reticulorumen weight, which was higher in calves fed Ulva sp. Both the mid-small intestinal and mid-colonic digesta populations of lactobacilli, Enterobacteriaceae, and E. coli, as well as the mid-small intestinal histomorphology in seaweed-supplemented calves were not different from control calves. However, acetic acid proportion in mid-colonic digesta was increased in calves fed Ulva sp. and A. nodosum, whereas butyric acid proportion was decreased compared with the control calves. Digesta pH in mid- and end small intestine and mid-colon were not affected, whereas ruminal pH was increased in calves fed Ulva sp. compared with the control calves. In conclusion, intact dried seaweed supplementation did not improve the growth or selected gut health parameters (i.e., histomorphology, digesta pH, bacteria, and short-chain fatty acids) in preweaning Holstein calves.

Reactive oxygen species (ROS) are the key factors that cause many diseases in the human body. Polysaccharides from seaweed have been shown to have significant antioxidant activity both in vivo and in vitro. The ameliorative effect of Ulva lactuca polysaccharide extract (UPE) on renal injury induced by oxidative stress was analyzed. As shown by hematoxylin–eosin staining results, UPE can significantly improve the kidney injury induced by D-galactose (D-gal). Additionally, the protective mechanism of UPE on the kidney was explored. The results showed that UPE could decrease the levels of serum creatinine (Scr), blood urea nitrogen (BUN), serum cystatin C (Cys-C), lipid peroxidation, protein carbonylation, and DNA oxidative damage (8-OHdG) and improve kidney glutathione content. Moreover, UPE significantly increased the activities of superoxide dismutase and glutathione peroxidase and total antioxidant activity in mice. UPE also decreased the levels of inflammatory cytokines TNF-α and IL-6. Further investigation into the expression of apoptotic protein caspase-3 showed that UPE decreased the expression of apoptotic protein caspase-3. These results indicate that UPE has a potential therapeutic effect on renal injury caused by oxidative stress, providing a new theoretical basis for the treatment of oxidative damage diseases in the future.

Enhancing the cholesterol turnover in the brain via activation of liver x receptors can restore memory in a mouse model for Alzheimer's disease. The edible Asian brown alga Sargassum fusiforme (Hijiki) contains high amounts of oxysterols such as (313, 244)-stigmasta-5, 28-dien-3, 24-diol (24[R, S]-saringosterol) that are a potent liver x receptor agonists. We aimed to find native European seaweed species with contents of 24(R, S)-saringosterol that are comparable to those found in Sargassum fusiforme. Additionally, we hypothesize that seasonal variations modify the amount of 24(R, S)-saringosterol in seaweeds. Sterols and oxysterols were extracted with chloroform/ methanol from various seaweed species harvested in the Eastern Scheldt in different seasons between October 2016 and September 2017. Identification and quantification of the lipids was performed by gas chromatographymass spectrometry and gas chromatography- flame ionization detection. We confirmed that brown algae Undaria pinnatifida harvested in February and Sargassum muticum harvested in October contained the highest amounts of 24(R, S)-saringosterol (32.4 +/- 15.25 mu g/g, mean +/- S.D. and 32.95 +/- 2.91 mu g/g, respectively) and its precursor fucosterol (1.48 +/- 0.11 mg/g), higher than Sargassum fusiforme (20.94 +/- 3.00 mu g/g, mean +/- S.D.), while Ascophyllum nodosum and Fucus vesiculosus and Fucus serratus contained amounts of 24(R, S)-saringosterol (22.09 +/- 3.45 mu g/g, 18.04 +/- 0.52 mu g/g and 19.47 +/- 9.01 mu g/g, mean +/- S.D., respectively) comparable to Sargassum fusiforme. In other algae only minor amounts of these sterols were observed. The green algae Ulva lactuca contained only 0.29 mg/g fucosterol and 10.3 mu g/g 24 (R, S)-saringosterol, while all investigated red algae did not contain any 24(R, S)-saringosterol or fucosterol. In the Eastern Scheldt algae harvested in September/October delivered the highest yield for 24(R, S)-saringosterol, with the exception of Undaria pinnatifida that showed the highest levels in February. We showed that exposure of lipid extracts of Ulva lactuca to sunlight at room temperature or in the presence of oxygen to UV-C light lead to the quantitative conversion of fucosterol into 24(R, S)saringosterol. Exposing pure fucosterol to UV-light did not convert any fucosterol into 24(R, S)-saringosterol underscoring the requirement of seaweed constituents in the conversion of fucosterol into 24(R, S)-saringosterol. In conclusion, we showed that brown seaweeds harvested from the Eastern Scheldt contain amounts of 24(R, S)saringosterol comparable to Sargassum fusiforme, varying per season and showing the highest amounts in spring. In accordance with these observations the amount of 24(R, S)-saringosterol in the brown seaweeds can be modulated by light.

Seaweed is a promising sustainable source of vegan protein as its farming does not require arable land, pesticides/insecticides, nor freshwater supply. However, to be explored as a novel protein source the content and nutritional quality of protein in seaweed need to be improved. We assessed the influence of pH-shift processing on protein degree of hydrolysis (%DH), protein/peptide size distribution, accessibility, and cell bioavailability of Ulva fenestrata proteins after in vitro gastrointestinal digestion. pH-shift processing of Ulva, which concentrated its proteins 3.5-times, significantly improved the %DH from 27.7±2.6% to 35.7±2.1% and the amino acid accessibility from 56.9±4.1% to 72.7±0.6%. Due to the higher amino acid accessibility, the amount of most amino acids transported across the cell monolayers was higher in the protein extracts. Regarding bioavailability, both Ulva and protein extracts were as bioavailable as casein. The protein/peptide molecular size distribution after digestion did not disclose a clear association with bioavailability.

The present study tested the outdoor cultivation of Haloferax mediterranei for PHA production from green macroalgae Ulva sp. in pneumatically agitated bioreactors and applied ultrasonic separation for enhanced settling of archaeal cells. Scaled-up cultivation (40 L) yielded maximum biomass productivity of 50.1 ± 0.11 mg·L-1·h−1 with a PHA productivity of 27 ± 0.01 mg·L-1·h−1 and conversion yield of 0.107 g PHA per gram UlvaDW. The maximum mass fraction of PHA achieved in biomass was calculated to be 56% w/w. Ultrasonic harvesting of Hfx. mediterranei cells approached 30% removal at energy inputs around 7.8 kWh·m−3, and indicated no significant aggregation enhancement by Ca2+ addition. Molecular weight analysis showed an increase in Polydispersity Index (PDI) when the corresponding air velocities were increased suggesting that the polymer was more homogeneous at lower mixing velocities. The current study demonstrated scalable processes for PHA production using Ulva sp. feedstock providing new technologies for halophilic biorefinery.

Ulva sp. is known to be a source of bioactive compounds such as ulvans, but to date, their biological activity on skin commensal and/or opportunistic pathogen bacteria has not been re­ported. In this study, the effects of poly- and oligosaccharide fractions produced by enzyme-assisted extraction and depolymerization were investigated, for the first time in vitro, on cutaneous bacteria: Staphylococcus aureus, Staphylococcus epidermidis, and Cutibacterium acnes. At 1000 μg/mL, poly- and oligosaccharide fractions did not affect the growth of the bacteria regarding their generation time. Polysaccharide Ulva sp. fractions at 1000 μg/mL did not alter the bacterial biofilm formation, while oligosaccharide fractions modified S. epidermidis and C. acnes biofilm structures. None of the fractions at 1000 μg/mL significantly modified the cytotoxic potential of S. epidermidis and S. aureus towards keratinocytes. However, poly- and oligosaccharide fractions at 1000 μg/mL induced a decrease in the inflammatory potential of both acneic and non-acneic C. acnes strains on keratinocytes of up to 39.8%, the strongest and most significant effect occurred when the bacteria were grown in the presence of polysaccharide fractions. Our research shows that poly- and oligosaccharide Ulva sp. fractions present notable biological activities on cutaneous bacteria, especially towards C. acnes acneic and non-acneic strains, which supports their potential use for dermo-cosmetic applications.

Multi-scale macroalgae growth models are required for the efficient design of sustainable, economically viable, and environmentally safe farms. Here, we develop a multi-scale model for Ulva sp. macroalgae growth and nitrogen sequestration in an intensive cultivation farm, regulated by temperature, light, and nutrients. The model incorporates a range of scales by incorporating spatial effects in two steps: light extinction at the reactor scale (1 m) and nutrient absorption at the farm scale (1 km). The model was validated on real data from an experimental reactor installed in the sea. Biomass production rates, chemical compositions, and nitrogen removal were simulated under different seasons, levels of dilution in the environment and water-exchange rate in the reactor. This multi-scale model provides an important tool for environmental authorities and seaweed farmers who desire to upscale to large bioremediation and/or macroalgae biomass production farms, thus promoting the marine sustainable development and the macroalgae-based bioeconomy., Zollmann et al. develop a multi-scale model for Ulva sp. macroalgae growth, biochemical composition, and nitrogen sequestration, regulated by temperature, light and nutrients. Their results demonstrate that this model can be used to design environmentally friendly and economically sustainable seaweed farms.

In order to understand how darkness/irradiance and low nighttime temperature might alter physiology of Ulva prolifera under lower salinity conditions, we analyzed the growth rates, water content, superoxide dismutase (SOD) activity, total soluble proteins (SPs) and carbohydrates content at the end of dark and light period under three temperature levels (25–25 °C treatment: 25 °C for day and night; 15–15 °C treatment: 15 °C for day and night; 25–15 °C treatment: 25 °C for day with 15 °C for night) and two salinity conditions (15, 25), meanwhile, the pigment content (chlorophyll a and b), chlorophyll fluorescence and photosynthetic oxygen evolution also were determined during light phase. We found that the U. prolifera showed higher growth rate and SOD activity during dark phase at 25 °C, but this dark-induced increase could not be observed at 15 °C. The reasons for this increase varied, however, maybe not included water content and SPs for no significant difference in water content observed under all the treatments, as well as lower SPs content for dark period aside that at 15 °C and salinity 15. Compared to other two temperature treatments, the thalli grown at 25–15 °C showed higher growth rate and the photosynthetic oxygen evolution rate in light phase under salinity 15 conditions, although the maximum relative electron transport rate (rETRmax) showed higher value under 25 °C treatment. These results indicate that the darkness and the lower nighttime temperature maybe responsible reason for the rapid growth of these green tide algae.

BACKGROUND: Edible seaweeds are one of the most important resources that currently exist for their multiple uses and applications. Like other food types, during their storage there are significant changes in sensory characteristics associated with loss of freshness and with production and/or degradation of their volatile compounds. In this study, multiple headspace sorptive extraction (MHSSE) coupled with gas chromatography/mass spectrometry (GC/MS) detection has been applied to an edible green seaweed (Ulva rigida) in order to determine the evolution of volatile compounds during storage for 12 days at 4 or 16 °C. RESULTS: The existing analytical methodology has been updated and validated for 18 additional compounds. At the end, 41 volatile compounds from different chemical families were quantified. Aldehydes, carboxylic acids and esters were the main classes found in U. rigida. Analytical and sensory data were submitted for statistical analysis. During storage, most volatile compounds were only affected by the number of storage days (P < 0.05), with significant decreases as storage time increased. Sensory analysis revealed significant changes in the samples stored for longer times (8, 10 and 12 days), with a clear influence of the boiled vegetable descriptor. CONCLUSION: In order to get a successful preservation of seaweeds and to maintain their marine character during storage, both temperature and time should be carefully controlled. © 2019 Society of Chemical Industry

In the present work, Ulva lactuca, a marine macroalgae was used for the production of biodiesel. The ultrasound assisted extraction of oil from autoclaved algal biomass was found effective with maximum yield. The maximum oil was extracted at optimal conditions of 5% moisture content of algal biomass, 0.15 mm size of biomass, 6:1 solvent: solid ratio, at 55 °C in 140 min. The n-hexane with co-solvent methyl tertbutyl ether has shown higher oil when compared to other co-solvents. The extracted oil was transesterified into biodiesel using silica doped with zinc oxide as novel heterogeneous nanocatalyst. The maximum biodiesel yield of 97.43% was obtained at optimized conditions of 800 °C calcination temperature, 8% catalyst concentration, 9:1 methanol to oil ratio, 55 °C reaction temperature and 50 min reaction time. The kinetics of the transesterification reaction was also studied. The Ulva lactuca was found as a potential source for biodiesel production.

To study the combined effects of multiple nitrogen (N) sources and salinity on the growth and physiology on macroalgae, we cultured Ulva prolifera under three N levels (N0, 0.1235 mg L−1; N1, 0.6 mg L−1; and N2, 4.4 mg L−1; the ratios were 18:74:8 for NH4–N, NO3–N, and NO2–N, respectively) and three salinity conditions (15, 25, and 35). Then, the growth, pigment content, photosynthetic performance, superoxide dismutase (SOD) activity, and contents of soluble protein and carbohydrates were measured. The results showed the following: (1) Compared to that grown at salinity 25, the growth of U. prolifera decreased under salinity 35, especially under the N0 and N2 levels, but there were no significant effects of salinity 15 under any of the N levels. (2) There were no significant effects of salinity on the chlorophyll a (Chla) content, but compared to the content at salinity 25, the chlorophyll b (Chlb) content was enhanced by salinity 15 and 35; lower ratio values between Chla and carotenoids (Car) occurred under the salinity 25 treatment. Under each salinity condition, the pigments were enhanced by a high N level. (3) A relatively higher salinity level decreased the photosynthetic oxygen evolution rate, while a higher N level increased this value. Compared to the rate at salinity 25, the dark respiration rate (Rd) significantly increased at salinity 15 under the N0 condition. (4) SOD activity was enhanced by a high N level, but no significant effects of salinity were observed. (5) The carbohydrate content was enhanced at salinity 35 under the N0 and N1 levels, and under salinity 15, this value increased with increasing N levels. In conclusion, although the growth of U. prolifera decreased at high N levels under high salinity conditions, a high N level induced an increase in photosynthesis, while no significant decrease in growth occurred. These findings indicate that low salinity and high N levels may be nonnegligible reasons why this species thrives, and low salinity was the better choice when this species was used for wastewater treatment.

Copper and zinc accumulation in macroalgae is a complex issue. While these metals exist as micronutrients and can serve to add nutritional value to the macroalgae when consumed by both plants and animals, elevated levels of the metals can reduce growth or even kill the algae. Many water parameters can influence the toxicity of the metals, though past studies have rarely isolated individual water parameters. This study aimed to independently determine the effects that salinity and alkalinity have on the growth and accumulation of these two metals on the macroalgae Ulva fasciata, distinguishing the effects of salinity and alkalinity as whole parameters from the collective effects of the water different constituents. The effect of salinity was determined using sodium chloride additions rather than seawater dilution, as performed in past studies, while alkalinity was tested using sodium bicarbonate additions to artificial seawater. The results of the study reinforce previous findings that copper is very toxic to macroalgae, even at low concentrations (50 µg L−1) though the effects of zinc remain inconclusive at 50 µg L−1, since the experiment was conducted over only a two-week trial period. The research suggests that salinity and alkalinity have no significant effect on the toxicity of copper to the growth of the macroalgae, but alkalinity significantly reduced copper and increased zinc accumulation in U. fasciata. The results of this study warrant further research in the field to investigate which other components of seawater and macroalgae reduce metal toxicity in the macroalgae. Additionally, these findings suggest the need for further refinement of toxicity models when adapted to macroalgae.

Numerous studies on poly γ-d-glutamicacid (γ-PGA) production have investigated terrestrial renewable sources for reducing production costs, but there are no studies using waste marine resources so far. We aimed to develop a cost-effective production method of γ-d-PGA by Bacillus sp. SJ-10 using green macroalgae (Ulva sp.) as a major substrate without hydrolysis pretreatment. The SJ-10 was shown to not only cause immediate tissue degradation of the Ulva membrane but also grew well as a sole substrate. The γ-d-PGA yield was 6.29 ± 0.34 g/L under optimized conditions via the response surface method, and the produced γ-d-PGA had a thermal decomposition temperature of 310°C and molecular weight of 250-1780 kDa. The calculated cost efficiency for the final yield was 32% when compared with complex media. Therefore, the present study provided a strategy for promoting an ecofriendly and cost-effective means to produce γ-d-PGA via a marine renewable resource.

In this study, iron oxide nanoparticles (IO-NPs) with a mean diameter of 102.85 nm were firstly synthesized via a facile green route using Ulva spp. aqueous extract as a bioreductant agent. Then, IO-NPs were loaded into carbonated hydroxyapatite (c-Hap) and the final product was named as the iron oxide nanoparticles loaded carbonated hydroxyapatite (IO-NPs-Lc-Hap). Subsequently, IO-NPs-Lc-Hap was characterized by FT-IR, SEM, XRD and EDX analysis methods. MG colour removal efficiencies of Ulva spp., Hap, IO-NPs and IO-NPs-Lc-Hap materials were also evaluated by adsorption and/or Fenton-like reaction methods. IO-NPs-Lc-Hap with the highest decolourization capacity was chosen as a heterogeneous Fenton-like catalyst for Malachite Green (MG). For Fenton-like decolourization of MG, the optimum H2O2 concentration, initial dye concentration and catalyst concentration were determined to be 30 mM, 100 mg/L and 1.0 g/L, respectively. At these optimum conditions, 100% decolourization efficiency and 33.3% COD removal were obtained. On the other hand, 94% decolourization efficiency and 42% COD removal were achieved for the real textile wastewater at the obtained optimum conditions. The experimental decolourization reaction rate for MG was determined as -rd = 0.0779 [(mg dye0.3) (g cat-0.3) (min-1)] × qt0.7. Also, the catalyst had high decolourization efficiencies at the end of six sequence usages.

Marine macroalgae are a promising source of diverse bioactive compounds with applications in the biocontrol of harmful cyanobacteria blooms (cyanoHABs). In this work, we evaluated the potential algicidal activities of 14 species of seaweed collected from the coast of Souiria Laqdima, Morocco. Methanol extracts were screened in solid and liquid medium against the growth of the toxic cyanobacteria Microcystis aeruginosa and the microalgae Chlorella sp. used as food supplement. The results in solid medium revealed that the algicidal activity was limited to M. aeruginosa with the extract of Bornetia secundiflora showing the highest growth inhibition activity against Microcystis (27.33 ± 0.33 mm), whereas the extracts of Laminaria digitata, Halopytis incurvus, Ulva lactuca, and Sargasum muticum showed no inhibition. In liquid medium, the results indicated that all methanolic extracts of different macroalgae tested have a significant inhibitory effect on M. aeruginosa compared with that of the negative control. The maximum inhibition rates of M. aeruginosa were produced by the extracts of Bifurcaria tuberculata, Codium elongatum, and B. secundiflora. Moreover, the extracts of B. secundiflora recorded the maximum inhibition rate of Chlorella sp. Overall, the results highlight the potential of the extracts from macroalgae to control toxic cyanobacteria species. An up-flow fixed column study was conducted to remediate Remazol Brilliant Orange 3R (RBO3R) from contaminated solutions using biochar derived from Ulva lactuca biomass. The influences of column parameters on dye sorption were studied in detail, which include initial RBO3R concentration, bed depth, and flow rate. Optimization experiments indicated that maximum RBO3R column uptake of 0.114 mmol/g was observed at 0.25 mmol/L (initial RBO3R concentration), 0.3 L/hr (flow rate), and 25 cm (U. lactuca bed depth). Modeling of column sorption data was performed using the Yoon-Nelson, modified dose-response and Thomas models. The spent biochar was desorbed and rejuvenated using 0.01 M NaOH. The elutant (0.01 M NaOH) exhibited 99.7% efficiency, and the process was completed in 115 min with high overall concentration factor of 8.4. PRACTITIONER POINTS: This study explores the impact of column parameters on the dye removal potential of U. lactuca-derived biochar. At optimized condition, the biochar bed exhibited highest Remazol Brilliant Orange 3R uptake capacity of 0.114 mmol/g. The regeneration and desorption of U. lactuca-derived biochar bed was possible with NaOH (0.01 M) as elutant.

Intertidal macroalgae suffer different environmental conditions and mat densities during growing period. In the present study, Ulva lactuca Linnaeus were collected from high, intermediate, and low tidal zones at Nan'ao Island, China. These algal photosynthetic pigments and photosynthesis behaviors with different mat densities were measured. The aim is to examine how the physiological responses and acclimation match the representative tidal distribution and algal mat density. The photosynthetic pigment (chlorophyll a and carotenoid) contents and irradiance-saturated maximum photosynthetic rates (Pmax) were greater in low zone-grown U. lactuca compared with the algae grown at high and intermediate zones. Under low algal mat density, the Pmax, apparent photosynthetic efficiency (α), and dark respiration rate (Rd) of U. lactuca grown at low zone were increased, whereas the irradiance saturation points (Ik) were decreased, compared with the algae grown at higher zone. However, the Pmax of high and intermediate zone-grown U. lactuca at high algal mat density were greater than at low density. Moreover, the pH compensation point of low zone-grown thalli (9.98) was lower than the higher zone-grown thalli (more than 10.15); however, the chlorophyll fluorescence parameters (reflect photosynthetic system activity) of the thalli collected from the three different zones were similar. Therefore, we proposed that the effects of varied densities on the photosynthetic rates of these three tidal zone-grown U. lactuca thalli were different, which might be related with different capacity of HCO3- utilization of macroalgae at their zonations.

Extraction of protein from macroalgae, currently defined as “novel food”, is challenging and limited information about the health impacts of these proteins is available. Here, we report on a non-thermal, chemical-free green macroalgae Ulva sp. protein extraction by osmotic shock combined with pulsed electric fields (PEF) followed by hydraulic pressure. The extracted proteins were identified and annotated to allergens using sequence similarity. The allergenicity potential of PEF extracted proteins was compared to osmotic shock extracts and complete Ulva sp. proteome, extracted with the thermochemical method. The PEF extracts contained ‘superoxide dismutase’ (SOD), a known food allergen, osmotic shock extract contained ‘troponin C’, and thermochemical extract contained two additional potential food allergens ‘aldolase A’ and ‘thioredoxin h’. This study shows an importance and the need for deep investigation of algal proteins and protein extraction technology health impacts prior to large-scale release to the market of “novel food” derived proteins.

Petroleum is one of the most important pollutants in the marine ecosystem, and oil spills have a long-term effect on the marine environment. After an oil spill, petroleum continues to diffuse, flowing to the intertidal zone where it accumulates. Ulva pertusa is the main macroalgae species in the intertidal zone of the ocean, and petroleum is toxic to marine macroalgae. Fatty acids, which are involved in various physiological activities, including metabolism, are indispensable substances in organisms. Thus, an organism's fatty acid composition reflects its physiological state. Because fatty acids are carbon-rich compounds, their stable carbon isotope composition can be measured to assess the state of the organism. In this study, the effects of the water accommodated fraction (WAF) of different concentrations of 180# fuel oil and 0# diesel oil on the chlorophyll a and fatty acid carbon stable isotope composition (δ13CFAs) of U. pertusa were assessed. The fatty acid carbon stable isotope value varied among the different petroleum treatments, and high WAF concentrations significantly inhibited the synthesis of unsaturated fatty acids. Thus, exposure to petroleum products affected the synthesis of fatty acids in U. pertusa and also caused carbon stable isotope fractionation during the fatty acid synthesis process. The results indicate that the saturated fatty acid δ13C16:0 and the unsaturated fatty acid δ13C18:1 can be used as biomarkers to evaluate the degree of petroleum-induced stress in U. pertusa and that carbon stable isotope analysis of U. pertusa can be used as an environmental risk assessment tool in the intertidal zone.

Non-conventional carbon sources, such as macroalgae, are sustainable alternatives for large-scale production of biopolymers. The present study examined macroalgae-derived carbohydrates, as carbon sources for the production of polyhydroxyalkanoates (PHAs) by Haloferax mediterranei. Simulants of the hydrolysates of seven different macroalgal biomasses were prepared and the PHA production was studied. A maximum biomass concentration with maximum PHA content was detected in medium prepared from green macroalgae. The highest cell dry weight and PHA concentrations were 3.8 ± 0.2 g·L−1 and 2.2 ± 0.12 g·L−1 respectively when Haloferax mediterranei was grown in 25% (w/w) of Ulva sp. hydrolysate, at 42 °C temperature and initial pH of 7.2. Poly(3-hydroxy-butyrate-co-3-hydroxyvalerate was the major PHA constituent. The present study demonstrated that Ulva sp. is a promising feedstock for PHA production.
