A method is designed to quickly form protein hydrogels, based on the self-assembly of highly concentrated lysozyme solutions in acidic conditions. Their properties can be easily modulated by selecting the curing temperature. Molecular insights on the gelation pathway, derived by in situ FTIR spectroscopy, are related to calorimetric and rheological results, providing a consistent picture on structure-property correlations. In these self-crowded samples, the thermal unfolding induces the rapid formation of amyloid aggregates, leading to temperature-dependent quasi-stationary levels of antiparallel cross β-sheet links, attributed to kinetically trapped oligomers. Upon subsequent cooling, thermoreversible hydrogels develop by the formation of interoligomer contacts. Through heating/cooling cycles, the starting solutions can be largely recovered back, due to oligomer-to-monomer dissociation and refolding. Overall, transparent protein hydrogels can be easily formed in self-crowding conditions and their properties explained, considering the formation of interconnected amyloid oligomers. This type of biomaterial might be relevant in different fields, along with analogous systems of a fibrillar nature more commonly considered.

Background The pathophysiology of heart failure with preserved ejection fraction (HFpEF) is a matter of investigation and its diagnosis remains challenging. Although the mechanisms that are responsible for the development of HFpEF are not fully understood, it is well known that nearly 80% of patients with HFpEF have concomitant hypertension.Methods We investigated whether early biochemical alterations were detectable during HFpEF progression in salt-induced hypertensive rats, using Fourier-transformed infrared (FTIR) and Raman spectroscopic techniques as a new diagnostic approach. Results Greater protein content and, specifically, greater collagen deposition were observed in the left atrium and right ventricle of hypertensive rats, together with altered metabolism of myocytes. Additionally, Raman spectra indicated a conformational change, or different degree of phosphorylation/methylation, in tyrosine-rich proteins. A correlation was found between tyrosine content and cardiac fibrosis of both right and left ventricles. Microcalcifications were detected in the left and right atria of control animals, with a progressive augmentation from six to 22 weeks. A further increase occurred in the left ventricle and right atrium of 22-week salt-fed animals, and a positive correlation was shown between the mineral deposits and the cardiac size of the left ventricle. Conclusions Overall, FTIR and Raman techniques proved to be sensitive to early biochemical changes in HFpEF and preceded clinical humoral and imaging markers.

Chronic high glucose (HG) exposure increases methylglyoxal (MG)-derived AGEs and is involved in the onset of pathological conditions, such as diabetes, atherosclerosis and chronic‐degenerative diseases. Under physiologic condition the harmful effects of MG are contrasted by glyoxalase system that is involved in the detoxification of Reactive Carbonyl Species (RCS) and maintain the homeostasis of the redox environment of the cell. Polyphenols are the most abundant antioxidants in the diet and present various health benefits. The study aimed at investigating the role of polyphenols extracted from an apple high in polyphenols (Calville White Winter), on glyco-oxidative stress induced by chronic HG-exposure. Intestinal Caco-2 cells were treated in physiological glucose condition (25mM) as a control and in HG condition (50mM) with or without apple extract for one week. Our data demonstrated that HG-treatment triggers glyco-oxidation stress with a significantly increase in ROS, lipid peroxidation, AGEs and Glyoxalase I (GlxI) activity with a significant decrease in total antioxidant intracellular defense. Treatment with polyphenols under HG condition restores to the control levels GlxI activity, decreases Glyoxalase II (GlxII) in relation to the control and induces a drop of glyco-oxidative damage. This paper seeks to highlight the roles of polyphenols in glyco-oxidative stress.

Brillouin–Raman microspectroscopy is presented as an innovative label-free all-optical investigation approachable to characterize the chemical composition and the mechanical properties of human tissues at micrometric resolution. Brillouin maps unveil mechanical heterogeneities in a human femoral diaphysis, showing a ubiquitous co-existence of hard and soft components, even in the most compact sections. The novel correlative analysis of Brillouin and Raman maps shows that the relative intensity of Brillouin peaks is a good proxy for the fraction of mineralized fibers and that the stiffness (longitudinal elastic modulus) of the hard component is linearly dependent on the hydroxyapatite concentration. For the soft component, a gradient of composition is found, ranging from an abundance of proteins in the more compact, external, bone to abundance of lipids, carotenoids, and heme groups approaching the trabecular, inner, part of the diaphysis. This work unveils the strong potential of correlative mechano-chemical characterization of human tissues at a micrometric resolution for both fundamental and translational research.

(1) Carthamus Tinctorius L. (safflower) is extensively used in traditional herbal medicine. (2) The aim of this study was to investigate the bioactive properties of polyphenol extracts from flowers of Carthamus Tinctorius (CT) cultivated in Italy. We also evaluated the properties of two bioactive water-soluble flavonoid compounds, hydroxy safflor yellow A (HSYA) and safflor yellow A (SYA), contained in Carthamus Tinctorius petals. (3) The total polyphenol content was 3.5 &plusmn, 0.2 g gallic acid equivalent (GAE)/100 g, flavonoids content was 330 &plusmn, 23 mg catechin equivalent (CE)/100 g in the flowers. The extract showed a high antioxidant activity evaluated by oxygen radical absorbance capacity (ORAC) and 2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assays. In addition, flower extract, SYA, and HSYA were able to reduce the susceptibility of low-density lipoprotein to copper-induced lipid peroxidation. In order to investigate the bioactive properties of flower extract, SYA, and HSYA we also studied their modulatory effect of oxidative stress on human dermal fibroblasts (HuDe) oxidized by tert-butyl hydroperoxide (t-BOOH). The CT extract at concentrations ranging from 0.01&ndash, 20 mg GAE/mL of polyphenols, exerted a protective effect against t-BOOH triggered oxidative stress. At higher concentration the extract exerted a pro-oxidant effect. Similar results have been obtained using HSYA and SYA. (4) These results demonstrate a biphasic effect exerted by HSYA, SYA, and flower extracts on oxidative stress.

Dimethyl sulfoxide (DMSO) is widely used in a number of biological and biotechnological applications, mainly because of its effects on the cell plasma membrane, but the molecular origins of this action are yet to be fully clarified. In this work, we used two- and three-component synthetic membranes (liposomes) and the plasma membrane of human erythrocytes to investigate the effect of DMSO when added to the membrane-solvating environment. Fourier transform infrared spectroscopy and thermal fluctuation spectroscopy revealed significant differences in the response of the two types of liposome systems to DMSO in terms of the bilayer thermotropic behavior, available free volume of the bilayer, its excess surface area, and bending elasticity. DMSO also alters the mechanical properties of the erythrocyte membrane in a concentration-dependent manner and is capable of increasing membrane permeability to ATP at even relatively low concentrations (3% v/v and above). Taken in its entirety, these results show that DMSO is likely to have a differential effect on heterogeneous biological membranes, depending on their local composition and structure, and could affect membrane-hosted biological functions.

Glioblastoma multiforme (GBM) is the most frequent malignant brain tumor characterized by highly heterogeneous subpopulations. In order to reveal the heterogeneous cell response, single cell analysis is an essential requirement. In this study, optical microscopy and Raman microspectroscopy were used to follow the stress response of U251 single cells adherent on a silicon substrate. Cultured cells on silicon substrate were treated with hydrogen peroxide to promote apoptosis. Under these conditions expected changes occurred after a few hours and were revealed by the reduction of cytochrome c, lipid, nucleic acid and protein Raman signals: this ensured the possibility to analyse U251 cell line as grown on Si substrate, and to monitor the response of single cells to stress conditions. As a consequence, we used microRaman to monitor the effects induced by nutrient depletion: a fast change of Raman spectra showed two different sub-populations of sensible and resistant U251 cells. Furthermore, spectral variations after DMSO addition were associated to volume changes and confirmed by morphological analysis. Thus, our results highlight the sensitivity of Raman microspectroscopy to detect rapid variations of macromolecule concentration due to oxidative stress and/or cell volume changes at the single cell level.

SH-SY5Y cell line is a model for neural diseases originating from neuroblastoma, a paediatric embryonal malignancy that may differentiate and regress to a benign state. By monitoring the differentiation in single living cells, it could be possible to reach a better understanding of the process and, consequently, to develop new therapeutic approaches. In this work, we investigated the potential of Raman microspectroscopy to monitor biochemical changes in single living SH-SY5Y cells treated with a combination of retinoic acid (RA) and brain-derived neurotrophic factor to stimulate the differentiation toward a neuronal phenotype. The results show that the spectroscopic analysis is able to detect biochemical alterations consistent with the differentiation process. Moreover, we investigated also the influence of the cell culture substrate on the treatment outcome. Silicon and aluminium are 2 materials suitable for the spectroscopic analysis and of interest in neuroscience applications. Even though silicon has an ensured biocompatibility, it is subject to alterations after the treatment with RA. Aluminium revealed to be a cost-effective cell culture substrate for the RA and brain-derived neurotrophic factor treatment of SH-SY5Y cells and particularly suitable for Raman microspectroscopy. Water is a strong self-associated liquid with peculiar properties that crucially depend on H-bonding. As regards its molecular dynamics, only recently has water reorientation been successfully described based on a jump mechanism, which is responsible for the overall H-bonding exchange. Here, using high-resolution broad-band depolarized light scattering, we have investigated the reorientational dynamics of formamide (FA) as a function of concentration from the neat liquid to diluted aqueous solutions. Our main findings indicate that in the diluted regime the water rearrangement can trigger the motion of FA solute molecules, which are forced to reorient at the same rate as water. This highlights an exceptional behavior of FA, which perfectly substitutes water within its network. Besides other fundamental implications connected with the relevance of FA, its water-like behavior provides rare experimental evidence of a solute whose dynamics is completely slaved to the solvent. © 2017 American Chemical Society.

The lateral diffusion of lipids and of small molecules inside a membrane is strictly related to the arrangement of acyl chains and to their mobility. In this study, we use FTIR and time resolved 2D-IR spectroscopic techniques to characterize the structure and dynamics of the hydrophobic region of palmitoyl-oleylphosphatidylcholine/cholesterol vesicles dispersed in water/dimethylsulfoxide solutions. By means of a non-polar probe, hexacarbonyl tungsten, we monitor the distribution of free volumes inside the bilayer and the conformational dynamics of hydrophobic tails in relation to the different compositions of the membrane or the different compositions of the solvent. Despite the important structural changes induced by the presence of DMSO in the solvating medium, the picosecond dynamics of the membrane is preserved under the different conditions.

Legume flours contain elevated levels of healthy functional components. The aim of the present paper was to evaluate the levels of polyphenols and flavonoids and total antioxidant capacity of pasta obtained using 100% legume flours (100% pea flour, 100% red lentil flour, and 60% grass pea flour and 40% chickpea flour). Moreover, the post-prandial glycaemic responses, the glycaemic index and glycaemic load were evaluated. The levels of the bioactive compounds (total polyphenols and flavonoids) and the antioxidant properties were higher in all products obtained with legume flours compared with durum wheat semolina (DWS) pasta. After cooking by boiling, a decrease in the total content of polyphenols of about 50%–60% was observed in all pasta samples. The comparison of the post-prandial increase of glucose after intake of pasta obtained with 100% legume flours demonstrated significant differences compared with DWS pasta. Glycaemic index (GI) and glycaemic load (GL) values of DWS pasta were 52.59 ± 8.41% and 26.14 ± 3.12, respectively. The GI values and GL values were 20.07 ± 7.67% and 6.88 ± 2.63 in the product containing grass pea and chickpea flour, 22.38 ± 6.92% and 8.30 ± 2.56 in the product containing red lentil flour, and 23.39 ± 6.72% and 8.65 ± 2.50 in the product containing pea flour. The results suggest that pasta formulated with legume flours is a low glycaemic food and can be considered a suitable source of polyphenols and flavonoids in human diet.

We report on a study of protein aggregation induced on different cell samples by dimethyl sulfoxide (DMSO) addition. DMSO is the most commonly used cryoprotectant because it is supposed to readily diffuse across lipid bilayers, thus reducing water activity within cells; despite its large use, the mechanism of penetration and even the main interaction features with cell components are far from being understood. In the present work, infrared absorption spectroscopy is successfully applied to real time detection of chemical and structural changes occurring in cells during dehydration from water and water/DMSO suspensions. As a most interesting result, DMSO is observed to favor protein aggregation both in cellular model systems, as cultured lymphocytes and fibroblasts, and in human samples for clinic use, as hematopoietic stem cells from cord blood. This effect is evidenced at low water content, analogously to what is observed for protein solutions. Such tendency is not specific of the type of protein and suggests one possible origin of DMSO toxicity.

In this paper we test the ability of Raman micro-spectroscopy and Raman mapping to investigate the status of cells grown in adhesion on different substrates. The spectra of immortalized SH-SY5Y cells, grown on silicon and on metallic substrates are compared with those obtained for the same type of cells adhering on organic polyaniline (PANI), a memristive substrate chosen to achieve a living bio-hybrid system. Raman spectra give information on the status of the single cell, its local biochemical composition, and on the modifications induced by the substrate interaction. The good agreement between Raman spectra collected from cells adhering on different substrates confirms that the PANI, besides allowing the cell growth, doesn't strongly affect the general biochemical properties of the cell. The investigation of the cellular state in a label free condition is challenging and the obtained results confirm the Raman ability to achieve this information.

In the present study we focus on the effect of dimethyl sulfoxide (DMSO) on sphingomyelin (SM) model membranes composed by a binary mixture of a natural SM extract from porcine brain (BSM), and cholesterol (Chol). We compare the BSM properties with those of another natural SM extract from egg yolk (ESM), mainly palmitoyl-SM, in order to follow the effects of lipid heterogeneity on thermotropic and packing properties of model membranes. With respect to ESM, BSM accounts for a higher contribution of gauche conformers and a higher disorder as a consequence of the significant variability of acyl chain length and unsaturation degree. The ability to form a packed and ordered bilayer is extremely important for the stability and the permeability of SM-Chol membranes. DMSO is extensively employed in a range of biological and pharmaceutical application that include drug delivery and cryopreservation. DMSO affects hydration, structure and other physical properties of the membrane and, at relatively high concentrations, is known to enhance their drug permeability. This work helps on increasing the basic knowledge of the effect of DMSO on the structural properties of lipid vesicles that mime natural membranes and can also provide useful information to their use as drug carriers.

The goal of the current study was to identify potential roles of paraoxonase-2 in bladder carcinogenesis. T24 bladder cancer cells were transfected with plasmids inducing paraoxonase-2 silencing or overexpression. Upon the selection of clones stably down- or upregulating paraoxonase-2, cell proliferation, migration, and the production of reactive oxygen species were evaluated, before and after treatment with cisplatin and gemcitabine, used alone or in combination. The activity levels of both caspase-3 and caspase-8 were also analyzed. shRNA-mediated gene silencing and the overexpression of paraoxonase-2 revealed that the enzyme was able to promote both the proliferation and migration of T24 cells. Moreover, the knockdown of paraoxonase-2 was significantly associated with a reduced cell viability of T24 cells treated with chemotherapeutic drugs and led to both an increase of reactive oxygen species production and caspase-3 and caspase-8 activation. Conversely, under treatment with anti-neoplastic compounds, a higher proliferative capacity was found in T24 cells overexpressing paraoxonase-2 compared with controls. In addition, upon enzyme upregulation, both the production of reactive oxygen species and activation of caspase-3 and caspase-8 were reduced. Although further analyses will be required to fully understand the involvement of paraoxonase-2 in bladder tumorigenesis and in mechanisms leading to the development of chemoresistance, the data reported in this study seem to demonstrate that the enzyme could exert a great impact on tumor progression and susceptibility to chemotherapy, thus suggesting paraoxonase-2 as a novel and interesting molecular target for effective bladder cancer treatment.

Human sample, from patients or healthy donors, are a valuable link between basic research and clinic. Especially in translational research, they play an essential role in understanding development and progression of diseases as well as in developing new diagnostic and therapeutic tools. Stored in biobanks, fast access to appropriate material becomes possible. However, biobanking in a clinical context faces several challenges. In practice, collecting samples during clinical routine does not allow to strictly adhere to protocols of sample collection in all aspects. This may influence sample quality to variable degrees. Time from sample draw to asservation is a variable factor, and influences of prolonged storage at ambient temperature of tissues are not well understood. We investigated whether delays between 5 minutes and 3 hours, and the use of RNAlater RNA-preserving reagent would lead to a relevant drop in sample quality, measured by quantitative mRNA expression analysis. Our findings suggest that even under ambient conditions, delays up to 3 hours do not have a major impact on sample quality as long as the tissue remains intact.

(1) Background: Hyperglycemia leads to several biochemical and physiological consequences, such as the generation of advanced glycation end products (AGEs) and reactive oxygen species (ROS), which are involved in the development of several human diseases. Intestinal cells are continuously exposed to pro-oxidants and lipid peroxidation products from ingested foods, and also to glyco-oxidative damage. It has been reported that free radical generation may be linked to the development of inflammation-related gastrointestinal diseases. (2) Methods: The effects of high glucose (HG) treatment (50 mM) were assessed in terms of free radical production, lipid peroxidation, and AGEs formation. Furthermore, the expression and the antiapoptotic and antioxidant activity of the paraoxonase-2 (PON2) enzyme in intestinal cells has been investigated. (3) Results: Caco-2 cells treated with media supplied with high glucose (HG) (50 mM) showed, with respect to physiological glucose concentration (25 mM), an increase in ROS production, lipid peroxidation, and AGEs formation. Moreover, a lower PON2 expression and activity in HG-treated cells was related to activation of the apoptotic pathways. (4) Conclusions: Our results demonstrated that high glucose concentrations triggered glyco-oxidative stress in intestinal cells, the downregulation of PON2 could result in a higher oxidative stress and might contribute to intestinal dysfunction.

The properties of lipid membranes at low temperature are important for a number of biomedical and biotechnological applications, and the success of these applications depends on understanding the effects of temperature changes on intermolecular lipid-lipid and lipid-water interactions. Here, we use Fourier transform infrared spectroscopy to study lipid suspensions in water/dimethyl sulfoxide (DMSO) solutions in the -60 to 30 °C range. DMSO is a cryopreservative agent of cellular systems, and its action is largely related to its interaction with the lipid membrane, especially in the low-temperature regime. In the present work, we analyze the effects of solvent composition on the structural and thermotropic properties of cholesterol (chol)-loaded liposomes of palmitoyl-oleylphosphatidylcholine (POPC) because POPC/chol liposomes are suitable models of the plasmatic membrane. To this extent, we compare the properties of lipid vesicles suspended in water and water/DMSO solution at 0.10 DMSO mole fraction and we observe that the gel phase of the membrane has an increased thermal stability on DMSO addition. We estimate that the amount of unfrozen water at T = -60 °C is much reduced by the presence of DMSO, both in the gel- and the liquid-ordered phase of the membrane. Interestingly, we also evidence a reduced hydration of the lipid heads in the presence of DMSO when the vesicles are dispersed in a liquid solution, whereas the addition of DMSO does not alter the hydration state of phosphate and carbonyl groups in the frozen state of the membrane.

The thermal aggregation of lysozyme has been analyzed in water/ethanol solutions at low pH to induce the specific protein aggregation pathway which leads to fibrillar structures in a few hours. In this solvating medium, the protein undergoes a conformational rearrangement promoting the formation of fibrils that are structurally similar to amyloid ones. As the process evolves with different steps, a multitechnique approach has been used by means of analytical probes that can be selectively sensitive in the detection of the different stages of protein association. Fourier transform infrared spectroscopy, intrinsic fluorescence, stationary fluorescence anisotropy, transmission electron microscopy (TEM), and atomic force microscopy (AFM) measurements have been carried out at different times to access and characterize the whole aggregation pathway. The data recorded with different experimental setups revealed different sensitivity to different stages of protein assembling. The whole set of data together with the direct visualization of different aggregate structures by use of TEM and AFM imaging enable to discuss a possible mechanism of fibrillation.

In this study, Fourier transform infrared, Raman and Brillouin spectroscopy have been used to study lipid phase behavior of hydrated as well as dried multilamellar l-α-phosphatidylcholine assemblies, in order to compare limitations and potentials of the different techniques. Dried lipid samples have been studied in the presence and absence of trehalose, which is known to affect the phase behavior of these systems. The methylene C-H stretching (2800–3000 cm−1) region in infrared (IR) and Raman spectra provided mutually consistent information on the rearrangement of lipid acyl chains occurring at the lipid melting temperature. IR spectra have a higher signal-to-noise ratio, thus permitting a more precise evaluation of the melting temperature. In the hydrated lipid samples, the C-H stretching region in the Raman spectra is less affected by the contribution of water compared with that in the IR spectra. Raman spectra are particularly suitable to simultaneously study both lipid and water contributions allowing to distinguish ice from non-frozen water below 0 °C. Brillouin light scattering was used to probe the collective dynamics, i.e. the propagation velocity and the attenuation of longitudinal acoustic modes in the lipid samples. Lipid phase transitions are evident from a change in the temperature behavior of the acoustic velocity. Moreover, a strong relaxation process with a characteristic time of 14 ps was observed in the sample dried without trehalose with a maximum in acoustic attenuation at about 45 °C, which likely reflects the rearrangement of acyl chains. Dimethyl sulfoxide (DMSO) is a universal solvent widely used in many fields, from basic research to industrial applications. At low concentration, it is the most important cryoprotectant agent against cellular damage caused during a freeze-thaw cycle. Although the effects of this cosolvent on the physico-chemical properties of a lipid bilayer have been extensively studied with both in vitro and in vivo experiments, the molecular mechanism of cryopreservation is not completely understood. Cholesterol (Chol) represents one of the essential cell membrane component and is fundamental to maintain the integrity and fluidity of the membrane. Here we report a study on the effect of DMSO on the stability of Chol-containing model membranes. We investigated the effect of DMSO on thermal stability of model membranes formed by dipalmitoylphospatidylcholine (DPPC) and DPPC/Chol by means of Fourier Transform Infrared Spectroscopy (FT-IR) and Differential Scanning Calorimetry (DSC) measurements. It is well known that cholesterol reduces the thermal stability of DPPC vesicles and also the pre-transition is abolished. Our results show that DMSO induces a stabilization of the lipid bilayer of DPPC liposomes increasing both the pre- and main transition temperatures. In DPPC/Chol liposomes a similar thermal stabilization was observed for the main transition indicating that DMSO is capable to stabilize the lipid bilayer even in the presence of the sterol. Moreover, by direct inspection of the hydration degree of the lipid bilayers, we evidenced the role played by DMSO on the thermal stability of the membrane as connected to the hydration of the polar head groups.

Hydrophilic and hydrophobic interactions strongly affect the solvation dynamics of biomolecules. To understand their role, small model systems are generally employed to simplify the investigations. In this study the amphiphile trimethylamine N-oxide (TMAO) is chosen as an exemplar, and studied by means of extended frequency range depolarized light scattering (EDLS) experiments as a function of solute concentration. This technique proves to be a suitable tool for investigating different aspects of aqueous solvation, being able at the same time to provide information about relaxation processes and vibrational modes of solvent and solute. In the case study of TMAO, we find that the relaxation dynamics of hydration water is moderately retarded compared to the bulk, and the perturbation induced by the solute on surrounding water is confined to the first hydration shell. The results highlight the hydrophobic character of TMAO in its interaction with water. The number of molecules taking part in the solvation process decreases as the solute concentration increases, following a trend consistent with the hydration water-sharing model, and suggesting that aggregation between solute molecules is negligible. Finally, the analysis of the resonant modes in the THz region and the comparison with the corresponding results obtained for the isosteric molecule tert-butyl alcohol (TBA) allow us to provide new insights into the different solvating properties of these two biologically relevant molecules.

The effect of the rf power on the structural and electrical properties of nanostructured silicon carbon films deposited by Plasma Enhanced Chemical Vapour Deposition system, using silane and methane gas mixture highly diluted in hydrogen, has been investigated. The structural and electrical properties are found to depend strongly on rf power. The increase of the rf power decreases the size of the silicon crystallites as well as the crystalline fraction and increases the carbon content in the films. The study not only indicates the correlation between crystalline fraction and the electrical conductivity but also reveals the presence of nanocrystallites in the films deposited at high rf power.

The aim of this work is to investigate the stability to simulated solar radiation of some paintings samples through a new methodological approach adopting non-invasive spectroscopic techniques. In particular, commercial watercolours and iron oxide based pigments were used, these last ones being prepared for the experimental by gum Arabic in order to propose a possible substitute for traditional reintegration materials. Reflectance spectrophotometry in the visible range and Hyperspectral Imaging in the short wave infrared were chosen as non-invasive techniques for evaluation the stability to irradiation of the chosen pigments. These were studied before and after artificial ageing procedure performed in Solar Box chamber under controlled conditions. Data were treated and elaborated in order to evaluate the sensitivity of the chosen techniques in identifying the variations on paint layers, induced by photo-degradation, before they could be observed by eye. Furthermore a supervised classification method for monitoring the painted surface changes adopting a multivariate approach was successfully applied.

We use extended depolarized light scattering spectroscopy to study the dynamics of water in a lysozyme-trehalose aqueous solution over a broad time scale, from hundreds to fractions of picoseconds. We provide experimental evidence that the sugar, present in the ternary solution in quantity relevant for biopreservation, strongly modifies the solvation properties of the protein. By comparing aqueous solutions of lysozyme with and without trehalose, we show that the combined action of sugar and protein produces an exceptional dynamic slowdown of a fraction of water molecules around the protein, which become more than twice slower than in the absence of trehalose. We speculate that this ultraslow water may be caged between the sugar and protein surface, consistently with a water entrapment scenario. We also demonstrate that the dynamics of these water molecules gets slower and slower upon cooling. On the basis of these findings, we believe such ultraslow water close to the lysozyme is likely to be involved in the mechanism of bioprotection.

A Brillouin scattering investigation has been carried out on trehalose-water solutions in a wide range of concentrations (0 < phi < 0.74) in the ultraviolet regime. A complete set of data as a function of temperature (-10 degrees C

Biological interfaces characterized by a complex mixture of hydrophobic, hydrophilic, or charged moieties interfere with the cooperative rearrangement of the hydrogen-bond network of water. In the present study, this solute-induced dynamical perturbation is investigated by extended frequency range depolarized light scattering experiments on an aqueous solution of a variety of systems of different nature and complexity such as small hydrophobic and hydrophilic molecules, amino acids, dipeptides, and proteins. Our results suggest that a reductionist approach is not adequate to describe the rearrangement of hydration water because a significant increase of the dynamical retardation and extension of the perturbation occurs when increasing the chemical complexity of the solute.

Molecular dynamics and structural properties of water-tert-butyl alcohol (TBA) mixtures are studied as a function of concentration by extended depolarized light scattering (EDLS) experiments. The wide frequency range, going from fraction to several thousand GHz, explored by EDLS allows distinguishing TBA rotational dynamics from structural relaxation of water and intermolecular vibrational and librational modes of the solution. Contributions to the water relaxation originating from two distinct populations, i.e. hydration and bulk water, are clearly identified. The dynamic retardation factor of hydration water with respect to the bulk, ? ? 4, almost concentration independent, is one of the smallest found by EDLS among a variety of systems of different nature and complexity. This result, together with the small number of water molecules perturbed by the presence of TBA, supports the idea that hydrophobic simple molecules are less effective than hydrophilic and more complex molecules in perturbing the H-bond network of liquid water. At increasing TBA concentrations the average number of perturbed water molecules shows a pronounced decrease and the characteristic frequency of librational motions reduces significantly, both of which are results consistent with the occurrence of self-aggregation of TBA molecules. The effects of the silane concentration f on the structural, optical and electrical properties of undoped hydrogenated silicon films prepared in a plasma-enhanced chemical vapour deposition system have been studied. The electrical conductivity and Hall mobility appear to be controlled by microstructures induced by silane concentration and a clear electrical transport transition from crystalline to amorphous phase has been found when 3%

Molecular dynamics of acetone-alkali metal halide (LiBr, LiI) solutions were investigated using depolarized Rayleigh scattering (DRS) and low-frequency Raman spectroscopy in the frequency range from ~0.5 to 200 cm(-1) (~20 GHz to 6 THz). These experiments probe fast dynamical fluctuations of the polarizability anisotropy at picosecond and sub-picosecond time scales that are mainly driven by acetone orientational dynamics. Two distinct contributions were revealed: a fast process (units of picosecond, ps) related to the essentially unperturbed bulk solvent and a slow one (tens of ps) assigned to acetone molecules forming Li(+) solvation shells, decelerated by the motional constraint imposed by the cation. The increase of LiBr and LiI concentration significantly slows down the overall solvent relaxation as a consequence of the increased fraction of acetone molecules involved in the ion solvation shells. The global retardation is larger in LiI than LiBr solutions consistently with viscosity trends. This is explained in terms of ion association (at least ion pairing) more favorably promoted by Br(-) than I(-), with reduced Li(+)-acetone interactions in LiBr than LiI solutions. Anion-induced modulation of the Li(+)···O═C contacts, largely responsible for electrostriction phenomena, also affects the reduced THz-Raman spectral density, ascribed to ultrafast librational motions of acetone molecules. Overall, these findings enlighten the interplay between ion-dipole and ion-ion interactions on the fast solvation dynamics in electrolyte solutions of a typical polar aprotic solvent.

When a solute is dissolved in water, their mutual interactions determine the molecular properties of the solute on one hand, and the structure and dynamics of the surrounding water particles (the so-called hydration water) on the other. The very existence of soft matter and its peculiar properties are largely due to the wide variety of possible water-solute interactions. In this context, water is not an inert medium but rather an active component, and hydration water plays a crucial role in determining the structure, stability, dynamics, and function of matter. This review focuses on the collective dynamics of hydration water in terms of retardation with respect to the bulk, and of the number of molecules whose dynamics is perturbed. Since water environments are in a dynamic equilibrium, with molecules continuously exchanging from around the solute towards the bulk and vice versa, we examine the ability of different techniques to measure the water dynamics on the basis of the explored time scales and exchange rates. Special emphasis is given to the collective dynamics probed by extended depolarized light scattering and we discuss whether and to what extent the results obtained in aqueous solutions of small molecules can be extrapolated to the case of large biomacromolecules. In fact, recent experiments performed on solutions of increasing complexity clearly indicate that a reductionist approach is not adequate to describe their collective dynamics. We conclude this review by presenting current ideas that are being developed to describe the dynamics of water interacting with macromolecules.

A living bio-hybrid system has been successfully implemented. It is constituted by neuroblastic cells, the SH-SY5Y human neuroblastoma cells, adhering to a poly-anyline (PANI) a semiconductor polymer with memristive properties. By a multidisciplinary approach, the biocompatibility of the substrate has been analyzed and the functionality of the adhering cells has been investigated. We found that the PANI films can support the cell adhesion. Moreover, the SH-SY5Y cells were successfully differentiated into neuron-like cells for in vitro applications demonstrating that PANI can also promote cell differentiation. In order to deeply characterize the modifications of the bio-functionality induced by the cell-substrate interaction, the functional properties of the cells have been characterized by electrophysiology and Raman spectroscopy. Our results confirm that the PANI films do not strongly affect the general properties of the cells, ensuring their viability without toxic effects on their physiology. Ascribed to the adhesion process, however, a slight increase of the markers of the cell suffering has been evidenced by Raman spectroscopy and accordingly the electrophysiology shows a reduction at positive stimulations in the cells excitability.

In the present study, FTIR spectroscopy was used to monitor the freeze-thaw cycle of two cellular lines (HuDe and Jurkat) suspended in three different media: phosphate buffer solution (PBS); dimethylsulfoxide (DMSO)/PBS solution at 0.1 DMSO molar fraction; and CryoSure (0.1 DMSO molar fraction PBS solution+dextran 5% w/v) solution. The Trypan Blue test was also applied before freezing and after thawing each cell sample to estimate the recovery of membrane integrity after thermal treatment, and correlate this datum with spectroscopic results. By following the temperature evolution of two different spectral components (the libration and bending combination mode ?c(H2O) at 2000-2500cm-1, and the methylene symmetric stretching vibration ?sym(CH2) at about 2850cm-1) in the -120÷28°C range, we evidenced the main transition of lipid membrane in connection with cell dehydration, as induced by ice formation in the extracellular medium. In particular, in DMSO/PBS and CryoSure samples we observed a transition to a more rigid state of the lipid membrane together with an increased amount of non-freezable water in the extracellular medium; these results are connected to the role of DMSO as a cryoprotective agent irrespective of the nature of cell type.

The thermal denaturation of hen egg white lysozyme (HEWL) in D(2)O was followed by IR absorption after addition of dimethyl sulfoxide (DMSO) at different molar fractions. Amide I intensity and position revealed that DMSO reduces the thermal stability of the native protein and favors the formation of ordered aggregates. The comparison with ethanol/water solutions evidenced that ethanol (partially deuterated ethanol EtOD) has a stronger effect on the thermal stability of HEWL: the same down-shift of melting temperature was measured at 0.18 and 0.30 molar fraction of ethanol and DMSO, respectively. This is probably due to lower polarity of EtOD/D(2)O with respect to DMSO/D(2)O solutions. A kinetic study of protein assembling at 0.30 DMSO molar fraction, was also performed at different temperatures. The high viscosity of the solvent was observed to cause a sensitive slowing down of aggregation rate in comparison to that of water/alcohol solutions. The evidence of a retarded self-assembling put forward a possible explanation for the use of dimethyl sulfoxide as a protectant of protein structure. In fact, for both organic solvents a nonspecific interaction with the protein and a water-mediated action is deduced, but the addition of DMSO reduces the irreversible denaturation due to kinetic effects and this can be exploited for lessening one of the main degradation routes of globular proteins during freezing-thawing cycles.

Different spectroscopic techniques were applied for studying the structural properties of lysozyme in salt-free aqueous solutions. The results of vibrational and Brillouin scattering measurements were compared to obtain both single-molecule and collective properties of the solutions. The characterization of the protein system, from the conformation of the polypeptide chain to the exposure of side chains to the solvent and the arrangement of the solution network, was then achieved in the range 25–85 °C. Through the analysis of the indole breathing mode, a different environment for the six tryptophan residues of an unfolded lysozyme could be evidenced. Short and long exposures to high temperatures were used to modulate the competition between the thermally induced reversible and irreversible denaturation processes. These different thermal treatments were applied to distinguish between the effects of global unfolding of the single molecule from those of self-aggregation and gel formation. It has been observed that clusterization occurs at melting temperatures with slow kinetics; also, aggregates evolve from the completely unfolded state of the protein and lead to a sensitive increase in viscosity. This effect probably hinders any further conformational rearrangement of the molecules in the aggregate; thus as a consequence, the disordered structure of clusters does not change to give the β-sheet organization, characteristic of filaments or fibrils. International audience; We describe the fabrication of Ge nanowires during a single co-deposition step of Ge and Mn at high temperature. In these experimental conditions, a phase separation occurs and two different phases Ge and Ge1 − xMnx are formed with Ge1 − xMnx in the shape of small clusters distributed randomly in the Ge matrix. Because of the high deposition temperature, a new Ge1 − xMnx phase with low eutectic point is stabilized; this phase is different from the one (commonly Ge3Mn5) stabilized at lower temperature. During the growth process at 350 °C, the crystalline clusters remain solid but they are highly mobile and can float at the surface, serving as seeds to direct the growth of crystalline Ge nanowires from the vapor. The sketch steps of NWs formation are first the phase separation with formation of specific Ge1 − xMnx critical nuclei with low eutectic point and second the growth of Ge NWs directed by the Ge1 − xMnx solid cluster seeds. Ge NWs growth is forced along particular crystalline axis by the cluster seeds that lower the interfacial energy Ge/Ge1 − xMnx and the energy formation of the germanium crystal stabilizes the cluster position at the tip of the NWs. The density of NWs can be tuned by varying the nominal Mn concentration since this density is related to the number of clusters with the specific Ge1 − xMnx phase (with low eutectic point). The single step MBE process presented here has the main advantage to fully avoid any incorporation of unintentional impurity into Ge nanowires (apart from Mn atoms) and could be applied to several other systems. This work also provides new insights into the vapor–solid–solid growth mechanisms of Ge NWs.

The collective properties of liquid tert-butyl alcohol (TBA) were analysed by low-wavenumber Raman (LWR) scattering spectroscopy. Vibrational and relaxation phenomena of this H-bonding system were assessed in pure liquid phase at different temperatures in the 15–70 °C range, and in solution with 2,2′-dimethyl butane (2,2′-DMB) and water as a function of composition in the 0.7⩽xTBA⩽0.9 range at constant temperature (T = 25 °C). The LWR spectrum of pure TBA (below 150 cm−1) was expressed by the dynamical (or Raman) susceptibility ???? and reproduced by curve fitting using three functional forms. The high-wavenumber band, whose intensity is poorly dependent on the temperature, was assigned to the ultrafast librational mode; the remaining components with an intensity that increased with temperature were attributed to relaxation dynamics in the range of picosecond (‘slow’ 3 ps) and sub-picosecond (‘fast’, 0.4 ps) timescales. Adding 2,2′-DMB to TBA had no significant effect on the intermolecular interactions in alcohol-rich solutions, with almost unchanged LWR scattering features. On the other hand, water added to TBA determined an increase in interactions, similar to the effect of a temperature decrease in pure liquid alcohol; this was clearly depicted by the LWR profiles. Moreover, through the analysis of the OH stretching bands of water in solution, the confinement of aqueous pools in the hydrophilic spaces of alcohol-rich solutions was confirmed. Light scattering spectra on aqueous solutions of trehalose were recorded in a wide frequency range combining the use of a double monochromator and a multipass Fabry-Perot interferometer. Experimental results indicate the presence of a slow relaxation mode related to the solute dynamics, which is clearly separated from the solvent one. The spectral analysis reveals the existence of two separate solvent relaxation processes assigned to hydrating and bulk water molecules. The picosecond dynamics of water molecules directly interacting with the solute (proximal water) is consistently delayed with the corresponding relaxation time increase is about 5-6 times compared to the bulk. The slowing down induced by the sugar on the water dynamics mainly involves a restricted hydration layer constituted of 16-18 water molecules. These results improve our knowledge about the influence of carbohydrates on the fast rearrangement dynamics of water and may serve as a model to gain important insight on basic solvation properties of other biorelevant systems in aqueous media.

In this paper, three Renaissance tapestries depicting scenes painted by Raffaello Sanzio, conserved at the Vatican Museum, were investigated using in-situ UV–Visible fluorimetric measurements. The results show that this technique is suitable for the detection of natural organic colorants used for dyeing the threads woven in these tapestries. The emission signals detected on red–purple colours were assigned to the colorant orcein and those on different nuances of blue and green colours to indigo by comparison with data from reference laboratory samples. The assignments were supported by chromatographic experiments carried out on threads taken from the back side of the tapestry in the same points analysed by spectrofluorimentry.

The structural properties of Ge:Mn samples grown by MBE in two different ways, namely coevaporated alloys and alloys fabricated from an alternate deposition of Mn and Ge layers, i.e., digital layer alloys (DLA), are compared. The study is based on the X-ray absorption spectroscopy (XAS) in the near edge range (XANES) and in the extended energy range (EXAFS). In particular, an EXAFS analysis shows how much the DLA technique helps to hamper the atomic diffusion of Mn from tetrahedral interstitial sites generally occupied during co-evaporation, and to what extent it is able to reduce the formation of an Mn5Ge3 alloy.

The low-wavenumber Raman (LWR) scattering from aqueous solutions of glucose, fructose, trehalose, and dextran have been investigated in the 10–60 °C range at three different sugar concentrations. The spectra below 250 cm−1 have been analyzed in terms of the imaginary part of the dynamical susceptibility , and described by the sum of quasi-elastic (QE) and inelastic (IE) contributions. The subtraction of the QE scattering shows the presence of two bands representing bending-like and stretching-like modes of tetrahedral H-bonded structures of liquid water. The differences of IE profiles in solutions and in the pure solvent have been discussed in terms of perturbations operated by sugar on the tetrahedral arrangement of H-bonded network in liquid water. We report depolarized light scattering measurements of water-glucose solutions in a wide spectral range from 0.01 to 1000 cm(-1), performed by the combined use of a double monocromator and a multipass Fabry-Perot interferometer. For the first time evidence is given of a low frequency relaxation process related to the glucose dynamics well separated from that of water. This low frequency process has a different origin with respect to the broad relaxation feature detected by dielectric spectroscopy and is interpreted in terms of Stokes-Einstein-Debye rotational diffusion of single glucose molecules. Protein low-frequency vibrational modes are an important portion of a proteins' dynamical repertoire. Yet, it is notoriously difficult to isolate specific vibrational features in the spectra of proteins. Given an appropriately chosen model peptide, and using different experimental conditions, we can simplify the system and gain useful insights into the protein vibrational properties. Combining neutron scattering, depolarized light scattering, and molecular dynamics simulations, we analyse the low frequency vibrations of biological molecules, comparing the results from a small globular protein, lysozyme, and an amphiphilic peptide, NALMA, both in solution and in powder states. Lysozyme and NALMA present similar spectral features in the frequency range between 1 and 10 THz. With the aid of MD simulations, we assign the spectral features to methyl groups' librations (1-5 THz) and hindered torsions (5-10 THz) in NALMA. Our data also show that, while proteins display boson peak vibrations in both powder and solution forms, NALMA exhibits boson peak vibrations in powder form only. This provides insight into the nature of this feature, suggesting a connection of BP collective motions to a characteristic length scale of heterogeneities present in the system. These results provide context for the use of model peptide systems to study protein dynamics; demonstrating both their utility, and the great care that has to be used in extrapolating results observed in powder to solutions.

Liquid 1-octanol and 2-octanol have been investigated by infrared (IR), Raman, and Brillouin experiments in the 10-90 degrees C temperature range. Self-association properties of the neat liquids are described in terms of a three-state model in which OH oscillators differently implicated in the formation of H-bonds are considered. The results are in quantitative agreement with recent computational studies for 1-octanol. The H-bond probability is obtained by Raman data, and a stochastic model of H-bonded chains gives a consistent picture of the self-association characteristics. Average values of hydrogen bond enthalpy and entropy are evaluated. The H-bond formation enthalpy is ca. -22 kJ/mol and is slightly dependent on the structural isomerism. The different degree of self-association for the two octanols is attributed to entropic factors. The more shielded 2-isomer forms larger fractions of smaller, less cooperative, and more ordered clusters, likely corresponding to cyclic structures. Signatures of a different cluster organization are also evidenced by comparing the H-bond energy dispersion (HBED) of OH stretching IR bands. A limiting cooperative H-bond enthalpy value of 27 kJ/mol is found. It is also proposed that the different H-bonding capabilities may modulate the extent of interaggregate hydrocarbon interactions, which is important in explaining the differences in molar volume, compressibility, and vaporization enthalpy for the two isomers.
