Nanoparticles with multifunctionality and high colloidal stability are essential for biomedical applications. However, their use is often hindered by the formation of thick coating shells and/or nanoparticle agglomeration. Herein, we report a single nanoparticle coating strategy to form 1 nm polymeric shells with a variety of chemical functional groups and surface charges. Under exposure to alternating magnetic field, nanosecond thermal energy pulses trigger a polymerization in the region only a few nanometers from the magnetic nanoparticle (MNP) surface. Modular coatings containing functional groups, according to the respective choice of monomers, are possible. In addition, the surface charge can be tuned from negative through neutral to positive. We adopted a coating method for use in biomedical targeting studies where obtaining compact nanoparticles with the desired surface charge is critical. A single MNP with a zwitterionic charge can provide excellent colloidal stability and cell-specific targeting.

Contrast agents for magnetic resonance imaging (MRI) improve anatomical visualizations. However, owing to poor image resolution in whole-body MRI, resolving fine structures is challenging. Here, we report that a nanoparticle with a polysaccharide supramolecular core and a shell of amorphous-like hydrous ferric oxide generating strong T1 MRI contrast (with a relaxivity coefficient ratio of ~1.2) facilitates the imaging, at resolutions of the order of a few hundred micrometres, of cerebral, coronary and peripheral microvessels in rodents and of lower-extremity vessels in rabbits. The nanoparticle can be synthesized at room temperature in aqueous solution and in the absence of surfactants, has blood circulation and renal clearance profiles that prevent opsonization, and leads to better imaging performance than Dotarem (gadoterate meglumine), a clinically approved gadolinium-based MRI contrast agent. The nanoparticle's biocompatibility and imaging performance may prove advantageous in a broad range of preclinical and clinical applications of MRI.

Well-defined recyclable Pd- and Rh-bipyridyl group-impregnated SBA-15 catalysts were prepared for C-C bond coupling reaction and selective hydrogenation reactions, respectively. These SBA-15 derived ligands for the catalysts were prepared by direct and indirect co-condensation method using bipyridyl-linked methallylsilane. This indirect method, involving methoxysilane generated from methallylsilane shows higher loading efficiency of transition metal catalysts on SBA-15 than the direct use of methallylsilane.

The current gold standard for COVID-19 confirmation is RT-PCR, which possesses high sensitivity and specificity, but a long turnaround time has been a drawback. Here, we introduce a nanoPCR platform that can yield breakthroughs in RT-PCR speed and decentralize the diagnostic testing. A key innovation is to exploit magneto-plasmonic nanoparticles for dual purposes: fast thermocycling through plasmonic heating and enhanced signal detection via magnetic fluorescence switch. We further integrated these functionalities into a portable system to streamline point-of-care (POC) COVID-19 diagnostics with a “one-step-finish” approach from sampling to result. When applied with clinical COVID-19 samples, nanoPCR achieved an analytical performance of sensitivity and specificity comparable to that of a benchtop instrument but completed the entire detection within 18 minutes (vs. 2.5 hours for regular RT-PCR), which makes it ideal for rapid POC diagnostics. This platform technology is readily scalable for higher throughput and expandable to other disease targets, promising broader impacts in on-site molecular testing.

Nanoscale dynamic processes such as the diffusion of ions within solid-state structures are critical for understanding and tuning material properties in a wide range of areas, such as energy storage and conversion, catalysis, and optoelectronics. In the generation of new types of nanocrystals (NCs), diffusion-mediated ion exchange reactions have also been proposed as one of the most effective transformational strategies. However, retaining the original morphology and crystal structure of metal oxide NCs has been challenging because of Kirkendall void formation, and there has been no success, especially for anion exchange. Here we show that with the aid of an oxygen extracting reagent (OER), anion diffusion is dramatically accelerated and morphology-conserving anion exchange without Kirkendall void formation is possible. Adherens junctions (AJs) create spatially and mechanically discrete microdomains at the interfaces of cells. Using a mechanogenetic platform that generates artificial AJs with controlled protein localization, clustering, and mechanical loading, we report that AJs also organize proteolytic hotspots for γ-secretase with a spatially-regulated substrate selectivity that is critical in the processing of Notch and other transmembrane proteins. Membrane microdomains outside of AJs exclusively organize Notch ligand-receptor engagement (LRE-µdomain) to initialize receptor activation. Conversely, membrane microdomains within AJs exclusively serve to coordinate regulated intramembrane proteolysis (RIP-µdomain). They do so by concentrating γ-secretase and primed receptors while excluding full-length Notch. AJs induce these functionally distinct microdomains by means of cholesterol-dependent γ-secretase recruitment and size-dependent protein segregation. By excluding full-length Notch from RIP-µdomains, AJs prevents inappropriate enzyme-substrate interactions and suppresses spurious Notch activation. Ligand-induced ectodomain shedding eliminates size-dependent segregation, releasing Notch to translocate into AJs for processing by γ-secretase. This mechanism directs radial differentiative expansion of ventricular zone-neural progenitor cells in vivo and more broadly regulates the proteolysis of large cell-surface receptors like amyloid precursor protein. These findings suggest an unprecedented role of AJs in creating size-selective spatial switches that choreograph γ-secretase processing of multiple transmembrane proteins regulating development, homeostasis, and disease.

While neural cell transplantation represents a promising therapy for neurodegenerative diseases, the formation of functional networks of transplanted cells with host neurons constitutes one of the challenging steps. Here, we introduce a magnetic guidance methodology that controls neurite growth signaling via magnetic nanoparticles (MNPs) conjugated with antibodies targeting the deleted in colorectal cancer (DCC) receptor (DCC-MNPs). Activation of the DCC receptors by clusterization and subsequent axonal growth of the induced neuronal (iN) cells was performed in a spatially controlled manner. In addition to the directionality of the magnetically controlled axon projection, axonal growth of the iN cells assisted the formation of functional connections with pre-existing primary neurons. Our results suggest magnetic guidance as a strategy for improving neuronal connectivity by spatially guiding the axonal projections of transplanted neural cells for synaptic interactions with the host tissue.

Multifunctional magnetic nanoparticles have shown great promise as next-generation imaging and perturbation probes for deciphering molecular and cellular processes. As a consequence of multicomponent integration into a single nanosystem, pre-existing nanoprobes are typically large and show limited access to biological targets present in a crowded microenvironment. Here, we apply organic-phase surface PEGylation, click chemistry, and charge-based valency discrimination principles to develop compact, modular, and monovalent magnetofluorescent nanoparticles (MFNs). We show that MFNs exhibit highly efficient labeling to target receptors present in cells with a dense and thick glycocalyx layer. We use these MFNs to interrogate the E-cadherin-mediated adherens junction formation and F-actin polymerization in a three-dimensional space, demonstrating the utility as modular and versatile mechanogenetic probes in the most demanding single-cell perturbation applications.

The fine structure of the band edge exciton and the dark exciton photoluminescence (PL) are topics of significant interest in the research of semiconducting metal halide perovskite nanocrystals, with several conflicting reports on the level ordering of the bright and dark states and the accessibility of the emitting dark states. Recently, we observed the intense dark exciton PL in strongly confined CsPbBr3 nanocrystals at cryogenic temperatures, in contrast to weakly confined nanocrystals lacking dark exciton PL, which was explained by the confinement enhanced bright-dark exciton splitting. In this work, we investigated the size-dependence of the dark exciton photoluminescence properties in CsPbBr3 and CsPbI3 quantum dots in the strongly confined regime, showing the clear role of confinement in determining the bright-dark energy splitting (ΔEBD) and the dark exciton lifetime (τD). We observe the increase in both ΔEBD and τD with increasing quantum confinement in CsPbBr3 and CsPbI3 QDs, consistent with the earlier predictions on the size-dependence of ΔEBD and τD. Our results show that quantum confinement plays a crucial role in determining the accessibility to the dark exciton PL and its characteristics in metal halide perovskite nanocrystals.

Aberrant cleavage of Notch by γ-secretase is implicated in numerous diseases, but how cleavage is regulated in space and time is unclear. Here, we report that cadherin-based adherens junctions (cadAJs) are sites of high cell-surface γ-secretase activity, as well as sites of constrained physical space that excludes γ-secretase substrates having large extracellular domains (ECDs) like Notch. ECD shedding initiates drastic spatial relocalization of Notch to cadAJs, allowing enzyme-substrate interactions and downstream signaling. Spatial mutations by adjusting the ECD size or the physical constraint alter signaling. Dysregulation of this spatial switch promotes precocious differentiation of ventricular zone neural progenitor cells in vivo. We show the generality of this spatial switch for amyloid precursor protein proteolysis. Thus, cadAJs create spatially distinct biochemical compartments regulating cleavage events involving γ-secretase and preventing aberrant activation of receptors.

Phosphorene, a monolayer of black phosphorus (BP), is an elemental two-dimensional material with interesting physical properties, such as high charge carrier mobility and exotic anisotropic in-plane properties. To fundamentally understand these various physical properties, it is critically important to conduct an atomic-scale structural investigation of phosphorene, particularly regarding various defects and preferred edge configurations. However, it has been challenging to investigate mono- and few-layer phosphorene because of technical difficulties arising in the preparation of a high-quality sample and damages induced during the characterization process. Here, we successfully fabricate high-quality monolayer phosphorene using a controlled thinning process with transmission electron microscopy, and subsequently perform atomic-resolution imaging. Graphene protection suppresses the e-beam-induced damage to multi-layer BP and one-side graphene protection facilitates the layer-by-layer thinning of the samples, rendering high-quality monolayer and bilayer regions. We also observe the formation of atomic-scale crystalline edges predominantly aligned along the zigzag and (101) terminations, which is originated from edge kinetics under e-beam-induced sputtering process. Recent suggestions of nanoscale heat confinement on the surface of synthetic and biogenic magnetic nanoparticles during heating by radio frequency-alternating magnetic fields have generated intense interest because of the potential utility of this phenomenon for noninvasive control of biomolecular and cellular function. However, such confinement would represent a significant departure from the classical heat transfer theory. Here, we report an experimental investigation of nanoscale heat confinement on the surface of several types of iron oxide nanoparticles commonly used in biological research, using an all-optical method devoid of the potential artifacts present in previous studies. By simultaneously measuring the fluorescence of distinct thermochromic dyes attached to the particle surface or dissolved in the surrounding fluid during radio frequency magnetic stimulation, we found no measurable difference between the nanoparticle surface temperature and that of the surrounding fluid for three distinct nanoparticle types. Furthermore, the metalloprotein ferritin produced no temperature increase on the protein surface nor in the surrounding fluid. Experiments mimicking the designs of previous studies revealed potential sources of the artifacts. These findings inform the use of magnetic nanoparticle hyperthermia in engineered cellular and molecular systems.

Multidrug resistance (MDR) is a leading cause of failure in current chemotherapy treatment and constitutes a formidable challenge in therapeutics. Here, we demonstrate that a nanoscale magnetic tandem apoptosis trigger (m-TAT), which consists of a magnetic nanoparticle and chemodrug (e.g., doxorubicin), can completely remove MDR cancer cells in both in vitro and in vivo systems. m-TAT simultaneously activates extrinsic and intrinsic apoptosis signals in a synergistic fashion and downregulates the drug efflux pump (e.g., P-glycoprotein) which is one of the main causes of MDR. The tandem apoptosis strategy uses low level of chemodrug (in the nanomolar (nM) range) to eliminate MDR cancer cells. We further demonstrate that apoptosis of MDR cancer cells can be achieved in a spatially selective manner with single-cell level precision. Our study indicates that nanoscale tandem activation of convergent signaling pathways is a new platform concept to overcome MDR with high efficacy and specificity.

Achieving a molecular level understanding of chemical reactions on the surface of solid-state nanomaterials is important, but challenging. For example, the fully saturated basal plane is believed to be practically inert and its surface chemistry has been poorly explored, while two-dimensional (2D) layered transition-metal chalcogenides (TMCs) display unique reactivities due to their unusual anisotropic nature, where the edges consisting of unsaturated metals and chalcogens are sites for key chemical reactions. Herein, we report the use of Lewis acids/bases to elucidate the chemical reactivity of the basal plane in 2D layered TMCs. Electrophilic addition by Lewis acids (i.e., AlCl3) selectively onto sulfides in the basal plane followed by transmetalation and subsequent etching affords nanopores where such chemical activations are initiated and propagated from the localized positions of the basal plane. Cell-based therapies are attractive for treating various degenerative disorders and cancer but delivering functional cells to the region of interest in vivo remains difficult. The problem is exacerbated in dense biological matrices such as solid tissues because these environments impose significant steric hindrances for cell movement. Here, we show that neural stem cells transfected with zinc-doped ferrite magnetic nanoparticles (ZnMNPs) can be pulled by an external magnet to migrate to the desired location in the brain. These magnetically labeled cells (Mag-Cells) can migrate because ZnMNPs generate sufficiently strong mechanical forces to overcome steric hindrances in the brain tissues. Once at the site of lesion, Mag-Cells show enhanced neuronal differentiation and greater secretion of neurotrophic factors than unlabeled control stem cells. Our study shows that ZnMNPs activate zinc-mediated Wnt signaling to facilitate neuronal differentiation. When implemented in a rodent brain stroke model, Mag-Cells led to significant recovery of locomotor performance in the impaired limbs of the animals. Our findings provide a simple magnetic method for controlling migration of stem cells with high therapeutic functions, offering a valuable tool for other cell-based therapies.

Bulk magnetite (Fe3O4), the loadstone used in magnetic compasses, has been known to exhibit magnetoelectric (ME) properties below ~10 K; however, corresponding ME effects in Fe3O4 nanoparticles have been enigmatic. We investigate quantitatively the ME coupling of spherical Fe3O4 nanoparticles with uniform diameters (d) from 3 to 15 nm embedded in an insulating host, using a sensitive ME susceptometer. The intrinsic ME susceptibility (MES) of the Fe3O4 nanoparticles is measured, exhibiting a maximum value of ~0.6 ps/m at 5 K for d=15 nm. We found that the MES is reduced with reduced d but remains finite until d=~5 nm, which is close to the critical thickness for observing the Verwey transition. Moreover, with reduced diameter, the critical temperature below which the MES becomes conspicuous increased systematically from 9.8 K in the bulk to 19.7 K in the nanoparticles with d=7 nm, reflecting the core-shell effect on the ME properties. Spatiotemporal interrogation of signal transduction at the single-cell level is necessary to answer a host of important biological questions. This protocol describes a nanotechnology-based single-cell and single-molecule perturbation tool, termed mechanogenetics, that enables precise spatial and mechanical control over genetically encoded cell-surface receptors in live cells. The key components of this tool are a magnetoplasmonic nanoparticle (MPN) actuator that delivers defined spatial and mechanical cues to receptors through target-specific one-to-one engagement and a micromagnetic tweezers (μMT) that remotely controls the magnitude of force exerted on a single MPN. In our approach, a SNAP-tagged cell-surface receptor of interest is conjugated with a single-stranded DNA oligonucleotide, which hybridizes to its complementary oligonucleotide on the MPN. This protocol consists of four major stages: (i) chemical synthesis of MPNs, (ii) conjugation with DNA and purification of monovalent MPNs, (iii) modular targeting of MPNs to cell-surface receptors, and (iv) control of spatial and mechanical properties of targeted mechanosensitive receptors in live cells by adjusting the μMT-to-MPN distance. Using benzylguanine (BG)-functionalized MPNs and model cell lines expressing either SNAP-tagged Notch or vascular endothelial cadherin (VE-cadherin), we provide stepwise instructions for mechanogenetic control of receptor clustering and for mechanical receptor activation. The ability of this method to differentially control spatial and mechanical inputs to targeted receptors makes it particularly useful for interrogating the differential contributions of each individual cue to cell signaling. The entire procedure takes up to 1 week.

Nanoparticle clusters (NPCs) have attracted significant interest owing to their unique characteristics arising from their collective individual properties. Nonetheless, the construction of NPCs in a structurally well-defined and size-controllable manner remains a challenge. Here we demonstrate a strategy to construct size-controlled NPCs using the DNA-binding zinc finger (ZnF) protein. Biotinylated ZnF was conjugated to DNA templates with different lengths, followed by incubation with neutravidin-conjugated nanoparticles. The sequence specificity of ZnF and programmable DNA templates enabled a size-controlled construction of NPCs, resulting in a homogeneous size distribution. We demonstrated the utility of magnetic NPCs by showing a three-fold increase in the spin-spin relaxivity in MRI compared with Feridex. Furthermore, folate-conjugated magnetic NPCs exhibited a specific targeting ability for HeLa cells. The present approach can be applicable to other nanoparticles, finding wide applications in many areas such as disease diagnosis, imaging, and delivery of drugs and genes.

A solution-phase synthetic protocol to form two-dimensional (2D) single-layer transition-metal chalcogenides (TMCs) has long been sought; however, such efforts have been plagued with the spontaneous formation of multilayer sheets. In this study, we discovered a solution-phase synthetic protocol, called "diluted chalcogen continuous influx (DCCI)", where controlling the chalcogen source influx (e.g., H2S) during its reaction with the transition-metal halide precursor is the critical parameter for the formation of single-layer sheets as examined for the cases of group IV TMCs. The continuous influx of dilute H2S throughout the entire growth period is necessary for large sheet formation through the exclusive a- and b-axial growth processes. By contrast, the burst influx of highly concentrated H2S in the early stages of the growth process forms multilayer TMC nanodiscs. Our DCCI protocol is a new synthetic concept for single-layer TMCs and, in principle, can be operative for wide range of TMC nanosheets.

Capturing tumor cells has attracted the attention because such cells can be used for diagnosing the occurrence and progression of cancers. Here we present a simple and efficient capturing of tumor cells using anti-EGFR antibody-conjugated magnetic nanoparticles. EGFR is a well-established epithelial cancer-specific marker that is over-expressed in many types of tumor cells. The conjugation of an anti-EGFR antibody with magnetic nanoparticles through a PEG-linker was shown to be highly efficient for preventing the non-specific binding of cells and improving the stability of the resulting nanoparticles even at a high salt concentration. The use of EGFR-targetable nanoparticles resulted in about 57% capture efficiency in 20 min for two cancer cell lines, PC9 (lung cancer) and HeLa (cervical cancer). The survival rate of the captured cells was approximately 85%, and most of the cells were retrievable, providing options for a further analysis of captured cells such as a mutational investigation of EGFR-related cancers. The present approach can be used for capturing circulating tumor cells (CTCs) expressing EGFR and the subsequent diagnostic analysis of the captured cells.

Overcoming resistance: Heat-treated cancer cells possess a protective mechanism for resistance and survival. Resistance-free apoptosis-inducing magnetic nanoparticles (RAINs) successfully promote hyperthermic apoptosis, obstructing cell survival by triggering two functional units of heat generation and the release of geldanamycin (GM) for heat shock protein (Hsp) inhibition under an alternating magnetic field (AMF).

The intrinsic nature of small-molecule chemotherapeutics, including i) limited aqueous solubility, ii) systemic toxicity due to non-specific whole-body distribution, and iii) potential development of drug resistance after initial administration, compromises their treatment efficacy.[1] Recently, nanoparticle (NP)-based drug delivery systems have been considered as promising alternatives to overcome some of these limitations and begin to resolve obstacles in the disease management in clinical oncology.[2] The intraparticular space of a NP vector can be employed to package drug payloads without constrain associated with their solubility. Further, NP vectors exhibit enhanced permeability and retention (EPR) effects[3] that facilitate the differential uptake, leading to preferential spatio-distribution in tumor.[4] However, conventional NP drug delivery systems tend to passively release drug payloads, limiting the ability to release an effective drug concentration at a desired time window. Therefore, there is a need to develop next-generation NP drug delivery system such as a stimuli-responsive drug release system with a goal of achieving spatio-temporal control, by which an acute level of drug concentration can be delivered at the time point the NP vectors reach maximum tumor accumulation.[5] By doing so, it is expected to dramatically improve therapeutic effects in tumor and effectively reduce systematic toxicity at a minute drug dosage.[6]

Two-dimensional (2D) layered nanostructures are emerging fast due to their exceptional materials properties. While the importance of physical approaches (e.g., guest intercalation and exfoliation) of 2D layered nanomaterials has been recognized, an understanding of basic chemical reactions of these materials, especially in nanoscale regime, is obscure. Here, we show how chemical stimuli can influence the fate of reaction pathways of 2D layered nanocrystals. Depending on the chemical characteristics (Lewis acid ((1)O2) or base (H2O)) of external stimuli, TiS2 nanocrystal is respectively transformed to either a TiO2 nanodisc through a "compositional metathesis" or a TiO2 toroid through multistage "edge-selective structural transformation" processes. These chemical reactions can serve as the new design concept for functional 2D layered nanostructures. For example, TiS2(disc)-TiO2(shell) nanocrystal constitutes a high performance type II heterojunction which not only a wide range solar energy coverage (~80%) with near-infrared absorption edge, but also possesses enhanced electron transfer property.

Temperature control is an important method of self-defense in biological systems. For example, one response mounted by humans in an effort to fight injury, including viral and bacterial infections, involves an increase in body temperature, thus producing the well-recognized symptoms of fever.[1] Today, the idea of using artificial temperature control for disease removal is being realized with the aid of various techniques, such as ultrasound, near-infrared light, and magnetic field by increasing localized temperature in a targeted region.[2] Magnetic nanoparticles have attracted considerable attention for hyperthermia applications owing to their ability to generate heat effectively when exposed to an alternating magnetic field without a penetration depth limit.[3] Hyperthermia, the artificially induced heat treatment of a disease, uses temperatures ranging between 42 °C and 47°C. Generally, a temperature below 45 °C induces apoptotic cell death.[4] As compared to necrosis, apoptosis is a more benign form of “programmed” cell death.[5] Nonliving cells produced as the result of apoptotic process are cleaned by phagocytosis without affecting neighboring normal cells. In contrast, necrosis, typically generated by harsh and high-temperature hyperthermia, is considered relatively harmful because it can be correlated with inflammatory disease and metastasis.[6] However, achieving effective apoptotic hyperthermia is often difficult, as cells typically acquire resistance to induced thermal stress.[7] Repeated exposures to high temperatures with high concentration of magnetic nanoparticles are usually necessary to achieve a useful level of therapeutic efficacy even though the conditions could favor necrotic cell death rather than apoptosis. Because cancer cells are susceptible to heat at about 43 °C, while most normal tissues remain undamaged,[8] hyperthermia using this temperature defines a recognized but unmet goal.

While interesting and unprecedented material characteristics of two dimensionality (2-D) layered nanomaterials are emerging, their reliable synthetic methodologies are not well developed. In this study we demonstrate general applicability of synthetic protocols to a wide range of colloidal 2-D layered transition-metal chalcogenide (TMC) nanocrystals. As distinctly different from other nanocrystals, we discovered that 2-D layered TMC nanocrystals are unstable in the presence of reactive radicals from elemental chalcogen during the crystal formation. We first introduce the synthesis of titanium sulfide and selenide where well-defined single crystallinity and lateral size controllability are verified, and then such synthetic protocols are extended to all of group IV and V transition-metal sulfide (TiS(2), ZrS(2), HfS(2), VS(2), NbS(2), and TaS(2)) and selenide (TiSe(2), ZrSe(3), HfSe(3), VSe(2), NbSe(2), and TaSe(2)) nanocrystals. The use of appropriate chalcogen source is found to be critical for the successful synthesis of 2-D layered TMC nanocrystals. CS(2) is an efficient chalcogen precursor for metal sulfide nanocrystals, whereas elemental Se is appropriate for metal selenide nanocrystals. We briefly discuss the effects of reactive radical characteristics of elemental S and Se on the formation of 2-D layered TMC nanocrystals.

The regulation of cellular activities in a controlled manner is one of the most challenging issues in fields ranging from cell biology to biomedicine. Nanoparticles have the potential of becoming useful tools for controlling cell signalling pathways in a space and time selective fashion. Here, we have developed magnetic nanoparticles that turn on apoptosis cell signalling by using a magnetic field in a remote and non-invasive manner. The magnetic switch consists of zinc-doped iron oxide magnetic nanoparticles (Zn(0.4)Fe(2.6)O(4)), conjugated with a targeting antibody for death receptor 4 (DR4) of DLD-1 colon cancer cells. The magnetic switch, in its On mode when a magnetic field is applied to aggregate magnetic nanoparticle-bound DR4s, promotes apoptosis signalling pathways. We have also demonstrated that the magnetic switch is operable at the micrometre scale and that it can be applied in an in vivo system where apoptotic morphological changes of zebrafish are successfully induced.

Regioselective chemical reactions and structural transformations of two-dimensional (2D) layered transition-metal chalcogenide (TMC) nanocrystals are described. Upon exposure of 2D TiS(2) nanodiscs to a chemical stimulus, such as Cu ion, selective chemical reaction begins to occur at the peripheral edges. This edge reaction is followed by ion diffusion, which is facilitated by interlayer nanochannels and leads to the formation of a heteroepitaxial TiS(2)-Cu(2)S intermediate. These processes eventually result in the generation of a single-crystalline, double-convex toroidal Cu(2)S nanostructure. Such 2D regioselective chemical reactions also take place when other ionic reactants are used. The observations made and chemical principles uncovered in this effort indicate that a general approach exists for building various toroidal nanocrystals of substances such as Ag(2)S, MnS, and CdS.

Nowadays, attempts to use nanoparticles for medical applications are on the rise. However, the effect of nanoparticles in the body has not been clarified. In this study, we aimed to examine the pharmacokinetics of magnetized nanoparticles. FITC conjugated SA-MnMEIO nanoparticles were prepared for the purpose of tracing. Nanoparticles (15 nm diameter) were injected into the tail vein of BALB/c mice at a dose of 20 mg (Mn+Fe)/kg. A mouse was housed in a metabolic cage and sacrificed serially up to 14 days for the sampling of blood and tissues from various organs (i.e., heart, kidney, liver, and spleen). The concentration of nanoparticles was measured by detecting FITC using spectrofluorophotometer. Nanoparticles showed a half-life of 8.20 h. The fluorescence intensities of nanoparticles in tissues showed different time-dependent patterns among different organs. In the heart and the liver, the fluorescence intensities increased up to 2 weeks of observation, while those in the kidney and the spleen decreased. The results indicate that nanoparticles have unique pharmacokinetic characteristics and suggest that pharmacokinetic study is essential for the development of new nanoparticles for medical use.

Two-dimensional (2D) layered nanostructures have received increasing interest due to their unique nanoscale phenomena and their potential applications ranging from electronics and energy to catalysis. Recent investigation has revealed that laterally confined layered nanocrystals (LCLN) have remarkably enhanced electrochemical properties compared to their bulk counterparts owing to nanoscale characteristics that include large surface areas, finite lateral sizes, and enhanced open-edge morphologies. Among the variety of layered materials that have been described, tin sulfides are of particular interest because of their unique structural properties. SnS2 has a layered CdI2-type structure, composed of tin atoms sandwiched between two layers of hexagonally disposed closepacked sulfur atoms. The 2D layered characteristics of this substance are revealed in alkali metal intercalation phenomena, and by investigating anisotropy of properties such as electric and photoelectric conductivity. Owing to their large theoretical capacities for battery applications, bulk or micron sized tin-based materials have been extensively studied as possible alternatives for commercially available carbon electrodes. However, the main drawback of this system has been stemming from the large volume changes and accompanying sharp decrease in capacity that occur during electrochemical cycles. Aifantis et al. have reported that active sites with spherical and smaller volume fraction could improve electrochemical properties of anode materials from the view point of fracture mechanics. Other researchers also have reported that nanoscale tin sulfide-based materials would lead to an improvement in the cycling stability of these systems. Nanoscale distance-dependent phenomena, such as Forster resonance energy transfer, are important interactions for use in sensing and imaging, but their versatility for bioimaging can be limited by undesirable photon interactions with the surrounding biological matrix, especially in in vivo systems. Here, we report a new type of magnetism-based nanoscale distance-dependent phenomenon that can quantitatively and reversibly sense and image intra-/intermolecular interactions of biologically important targets. We introduce distance-dependent magnetic resonance tuning (MRET), which occurs between a paramagnetic 'enhancer' and a superparamagnetic 'quencher', where the T1 magnetic resonance imaging (MRI) signal is tuned ON or OFF depending on the separation distance between the quencher and the enhancer. With MRET, we demonstrate the principle of an MRI-based ruler for nanometre-scale distance measurement and the successful detection of both molecular interactions (for example, cleavage, binding, folding and unfolding) and biological targets in in vitro and in vivo systems. MRET can serve as a novel sensing principle to augment the exploration of a wide range of biological systems.

Nanoparticles have the potential to revolutionize current bio-medical diagnostic and therapeutic methods. For example, magnetic nanoparticles with unique superparamagnetism are emerging as next-generation probes for high performance magnetic resonance (MR) imaging. Their enhanced properties and nanoscale controllability in terms of size, composition, surface states, and magnetic spin structure have allowed for the highly sensitive and target-specific MR imaging of various biological systems including cancer detection, cell trafficking, and angiogenesis. On the other hand, viruses can be utilized as excellent delivery vehicles due to their facile cellular transfection and gene expression efficacies within their target cells. Such excellent properties of viruses offer promising prospects for gene therapy of genetic diseases and cancers, as well as for the genetic engineering of cells. Despite these prospects, a lack of understanding of their biological behaviors including in vivo migration, molecular recognition, gene delivery, and ultimate fate following their desired biofunctional applications limits their further development. There have been previous studies in the development of nanoparticle probing systems for viruses including virus-gold and virus-quantum dot nanoparticle systems. However, their utilization in the probing of viral gene delivery has not been investigated so far. Recently, Gd-based MR contrast agent-coated viruses were developed, but their functional behaviors including targetrecognition, cellular transfection, and gene delivery capabilities have not been demonstrated partly due to the intrinsically low sensitivity of Gd-based MR contrast agents. Our strategy is to hybridize the virus with magnetic nanoparticles into a single nanoparticle system with the dual-functional capabilities of target-specific MR imaging and gene delivery. Specifically, we fabricate hybrid nanoparticles of “enhanced green fluorescent protein (eGFP) promoter genecontaining adenovirus” and “manganese-doped magnetism engineered iron oxide (abbreviated as MnMEIO) nanoparticle”. As the viral gene delivery vector, we selected adenoviruses. Adenoviruses are known to be highly effective for transferring double-stranded DNAs to various cell types. Along with a gene delivery capability, adenoviruses possess targetspecificity to the cells with overexpression of Coxsackievirus B adenovirus receptor (CAR) which is known to facilitate the binding and intrusion of adenoviruses to the host cells. As the magnetic nanoparticle component we selected manganese-doped magnetism-engineered iron oxide (MnFe2O4, MnMEIO) nanoparticles, since it is known that MnMEIO exhibits exceptional MR contrast effects (R2 (=1/T2) value of 358 sec mM) and therefore is advantageous as probes for ultra-sensitive MR imaging. Briefly, the thermal reaction of manganese chloride (MnCl2) and iron tris(2,4-pentadionate) in hot organic solvents containing oleic acid and oleylamine capping molecules yielded hydrophobically capped MnMEIO nanoparticles. Their water-solubility and biocompatibility were attained by introducing 2,3-dimercaptosuccinic acids to the nanoparticle surface. Nanoparticles obtained were 12 nm with a high sizemonodispersity (r < 7%) and possessed single crystallinity, and a saturation magnetization value of 110 emu g (Mn+Fe). Hybridization of adenoviruses with MnMEIO nanoparticles was performed through a slight modification of a literature method, as follows. First, the capsid lysine residues of the adenoviruses were converted to maleimide groups by reacting them with sulfo-succinimidyl(4-N-maleimidomethyl)cyclohexane-1-carboxylate (sulfo-SMCC) cross-linkers. These groups were allowed to react with a large stoichiometric amount of MnMEIO nanoparticles, which resulted in the formation of adenovirus-MnMEIO hybrid nanoparticles by way of the nucleophilic addition of a surface thiol group of MnMEIO to the C O M M U N IC A IO N

We demonstrate a novel magnetophoretic immunoassay of allergen-specific immunoglobulin E (IgE) based on the magnetophoretic deflection velocity of a microbead that is proportional to the associated magnetic nanoparticles under enhanced magnetic field gradient in a microchannel. In this detection scheme, two types of house dust mites, Dermatophagoides farinae (D. farinae) and Dermatophagoides pteronyssinus (D. pteronyssinus), were used as the model allergens. Polystyrene microbeads were conjugated with each of the mite extracts followed by incubation with serum samples. The resulting mixture was then reacted with magnetic nanoparticle-conjugated anti-human IgE for detection of allergen-specific IgE by using sandwich immuno-reactions. A ferromagnetic microstructure combined with a permanent magnet was employed to increase the magnetic field gradient ( approximately 10(4) T/m) in a microfluidic device. The magnetophoretic velocities of microbeads were measured in a microchannel under applied magnetic field, and the averaged velocity was well correlated with the concentration of allergen-specific IgE in serum. From the analysis of pooled sera obtained from 44 patients, the detection limits of the allergen-specific human IgEs for D. farinae and D. pteronyssinus were determined to be 565 (0.045 IU/mL) and 268 fM (0.021 IU/mL), respectively. These values are 1 order of magnitude lower than those by a conventional CAP system. For evaluation of reproducibility and accuracy, unknown sera were subjected to a blind test by using the developed assay system, and they were compared with the CAP system. As a result, coefficient of variance was less than 10%, and the developed method enabled a fast assay with a tiny amount of serum ( approximately 10 microL).
