Biomaterials for effective hemorrhage control are urgently needed in clinics as uncontrolled bleeding is associated with high mortality. Herein, we developed an injectable and in situ photo-crosslinkable hybrid hemostatic hydrogel by combining pectin methacrylate (PECMA) and gelatin methacryloyl (GelMA). This modular material system combines ionic- and photo-crosslinking chemistries to design interpenetrating networks (IPN) exhibiting tunable rheology, highly porous structure, and controllable swelling and mechanical properties. By simply changing the calcium (0–15 mM) and polymer (1.5–7%) content used for the sequential crosslinking of hydrogels via calcium gelation and UV-photopolymerization, it was possible to precisely modulate the injectability, degradation, and swelling ratio. Moreover, it is demonstrated that PECMA/GelMA hydrogels present good cytocompatibility and uniquely synergize the hemostatic properties of calcium ions on PECMA, the amine residues on GelMA, and the highly porous network toward rapid blood absorption and fast coagulation effect. An in vitro porcine skin bleeding model confirmed that the hydrogel could be directly injected into the wound and rapidly photo-crosslinked, circumventing the bleeding and decreasing the coagulation time by 39%. Importantly, the crosslinked hydrogel could be easily removed to prevent secondary wound injury. Overall, this injectable hybrid PECMA/GelMA hydrogel stands as a promising hemostatic material.

A new POM-templated metal-organic framework [Ag8L4][PMo12O40]·OH {HL = 5-[4-(1,2,4-triazol-4-yl)phenyl]-1H-tetrazole} containing metal-organic nanocage and left- and right-handed helixes has been isolated under hydrothermal conditions. The Keggin POM plays a template role in the formation of macrocycle and metal-organic nanocage, as shown by X-ray diffraction analysis. The new compound represents a scarce example of POM-encapsulated metal-organic nanocage. Additionally, its photocatalytic degradation of RhB was also investigated.

The objective of this study was to evaluate the change of lipids in two whelk samples during cold storage. Results showed that the peroxide value (PV) and thiobarbituric acid reactive substance (TBARS) increased while the percentage of polyunsaturated fatty acid decreased, indicating that lipid oxidation occurred. The cold storage significantly reduced the levels of triacylglycerol (TAG), polar lipid (PoL), phosphatidylcholine (PC) and phosphatidylethanolamine (PE) but increased the levels of acid value (AV), free fatty acid (FFA) and monoacylglycerol, suggesting the hydrolysis of lipids. Moreover, the results showed that the lipoxygenase, acid lipase and phospholipase contributed to the hydrolysis and oxidation of lipids in the two whelks. Additionally, partial least squares discriminant analysis showed PC was positively correlated with PE, PoL and TAG, but negatively correlated with AV, FFA, PV and TBARS, indicating there is a close relationship between hydrolysis and oxidation of lipids.

Acute myeloid leukemia (AML) is a life-threatening hematological disease. Novel diagnostic and prognostic markers will be essential for new therapeutics and for significantly improving the disease prognosis. To characterize the metabolic features associated with AML and search for potential diagnostic and prognostic methods, here we analyzed the phenotypic characteristics of serum metabolite composition (metabonome) in a cohort of 183 patients with de novo acute myeloid leukemia together with 232 age- and gender-matched healthy controls using (1)H NMR spectroscopy in conjunction with multivariate data analysis. We observed significant serum metabonomic differences between AML patients and healthy controls and between AML patients with favorable and intermediate cytogenetic risks. Such differences were highlighted by systems differentiations in multiple metabolic pathways including glycolysis/gluconeogenesis, TCA cycle, biosynthesis of proteins and lipoproteins, and metabolism of fatty acids and cell membrane components, especially choline and its phosphorylated derivatives. This demonstrated the NMR-based metabonomics as a rapid and less invasive method for potential AML diagnosis and prognosis. The serum metabolic phenotypes observed here indicated that integration of metabonomics with other techniques will be useful for better understanding the biochemistry of pathogenesis and progression of leukemia.

Biodiesel from microalgae provides a promising alternative for biofuel production. Microalgal biodiesel can be produced under three major cultivation modes, namely, photoautotrophic, heterotrophic, and mixotrophic cultivation. Studies of biodiesel production from microalgae have been reported mostly focusing on photoautotrophic cultivation, mixotrophic cultivation has rarely been researched. This paper compared the biomass productivity, lipid content, and lipid productivity ofChlorella sorokinianaunder photoautotrophic, heterotrophic, and mixotrophic cultivation. Glucose was adopted as organic carbon source at five concentrations (0.1, 0.5, 1.0, 2.0, 5.0% glucose w/v). Results displayed that microalgal growth was significantly improved in glucose supplied cultures. Synergetic effect of photoautotrophy and heterotrophy existed in mixotrophic cultivation except for 5.0% culture. Highest biomass productivity of 1.178 g·L-1·d-1and highest lipid productivity of 582 mg·L-1·d-1was observed under mixotrophic cultivation with 2.0% (w/v) glucose addition. Lipid content ofC. sorokinianawas mostly higher in stationary phase than in exponential phase. Highest lipid content of 49.37% was observed in 2.0% mixotrophic culture, followed by 47.09% in 2.0% heterotrophic culture.

Reported studies of microalgal biodiesel production are mainly based on photoautotrophic cultivation, mixotrophic cultivation has rarely been researched. This paper compared the biomass productivity, lipid content, and lipid productivity of Scenedesmus sp. under photoautotrophic, heterotrophic, and mixotrophic cultivation. Glucose was added as organic carbon source at five concentrations (0.1, 0.5, 1.0, 2.0, 5.0% glucose w/v). Results displayed that microalgal growth was significantly improved in glucose supplied cultures. Synergetic effect of photoautotrophy and heterotrophy existed in all mixotrophic cultures. Highest biomass productivity of 1.307 g·L-1·d-1 and highest lipid productivity of 316 mg·L-1·d-1 was respectively observed under mixotrophic cultivation with 5.0% and 1.0% (w/v) glucose addition. Lipid content of Scenedesmus sp. under mixotrophic cultivation was mostly higher in stationary phase than in exponential phase. Highest lipid content of 27.73% was observed in 1.0% mixotrophic culture, followed by 24.66% in 1.0% heterotrophic culture.

It is well understood that the heat of micellization for surfactants is monotonically decreased along with an increase in temperature. However, this behavior for polymeric surfactants has never been carefully examined. In this study, the heat of micellization of poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) (PEO-PPO-PEO) triblock copolymers (Pluronics F68 and F88) in water as a function of temperature is carefully examined by using a high-sensitivity differential scanning calorimeter (HSDSC). The critical micelle temperature (CMT) decreases along with an increase in the concentration of Pluronic F68 (or F88). The heat of micellization decreases along with an increase in the temperature, as expected, when the CMT is higher than 55 and 42 degrees C for Pluronics F68 and F88, respectively. It is interesting to observe that the heat of micellization increases along with the temperature while the temperature is below 55 and 42 degrees C for Pluronics F68 and F88, respectively. The enthalpy-entropy compensation phenomenon for the micellization of Pluronics F68 and F88 in connection with the hydrophobicity is discussed.

Acute myeloid leukemia (AML) is a group of hematological malignancies with high heterogeneity. There is an increasing need to improve the risk stratification of AML patients, including those with normal cytogenetics, using molecular biomarkers. Here, we report a metabolomics study that identified a distinct glucose metabolism signature with 400 AML patients and 446 healthy controls. The glucose metabolism signature comprises a panel of 6 serum metabolite markers, which demonstrated prognostic value in cytogenetically normal AML patients. We generated a prognosis risk score (PRS) with 6 metabolite markers for each patient using principal component analysis. A low PRS was able to predict patients with poor survival independently of well-established markers. We further compared the gene expression patterns of AML blast cells between low and high PRS groups, which correlated well to the metabolic pathways involving the 6 metabolite markers, with enhanced glycolysis and tricarboxylic [corrected] acid cycle at gene expression level in low PRS group. In vitro results demonstrated enhanced glycolysis contributed to decreased sensitivity to antileukemic agent arabinofuranosyl cytidine (Ara-C), whereas inhibition of glycolysis suppressed AML cell proliferation and potentiated cytotoxicity of Ara-C. Our study provides strong evidence for the use of serum metabolites and metabolic pathways as novel prognostic markers and potential therapeutic targets for AML.
