In production, an intelligent analysis of the data provided along the production line bears huge potential for increasing process efficiency and reducing production costs. The (continuous) collection of relevant data is a crucial precondition for every Industry 4.0 or smart technology. While new machines have internal controllers and sensors to meet those requirements, this is usually not the case for machines being already in use. Especially in the textile industry, it is often standard to keep old machines since the manufacturing methods themselves haven’t changed significantly over the past decades. Hence, the successful exploitation of digitalization advantages requires these companies to first develop and implement a digitization strategy. In this paper we present a concept which allows to develop modular, scalable, flexible solutions considering the whole digitization process from data acquisition to storage. The concept is complemented by a guideline for its application in industry. Experience from a prototypical application in a textile company is described. The concept enables companies to determine the various conditions and requirements for digitization, to analyze different possibilities and to deduce scenarios that lead to a solution for the effective utilization of Industry 4.0 technologies.

The neural representation of information suffers from "noise"-the trial-to-trial variability in the response of neurons. The impact of correlated noise upon population coding has been debated, but a direct connection between theory and experiment remains tenuous. Here, we substantiate this connection and propose a refined theoretical picture. Using simultaneous recordings from a population of direction-selective retinal ganglion cells, we demonstrate that coding benefits from noise correlations. The effect is appreciable already in small populations, yet it is a collective phenomenon. Furthermore, the stimulus-dependent structure of correlation is key. We develop simple functional models that capture the stimulus-dependent statistics. We then use them to quantify the performance of population coding, which depends upon interplays of feature sensitivities and noise correlations in the population. Because favorable structures of correlation emerge robustly in circuits with noisy, nonlinear elements, they will arise and benefit coding beyond the confines of retina.

Reliable real-time low-latency spike sorting with large data throughput is essential for studies of neural network dynamics and for brain-machine interfaces (BMIs), in which the stimulation of neural networks is based on the networks' most recent activity. However, the majority of existing multi-electrode spike-sorting algorithms are unsuited for processing high quantities of simultaneously recorded data. Recording from large neuronal networks using large high-density electrode sets (thousands of electrodes) imposes high demands on the data-processing hardware regarding computational complexity and data transmission bandwidth; this, in turn, entails demanding requirements in terms of chip area, memory resources and processing latency. This paper presents computational complexity optimization techniques, which facilitate the use of spike-sorting algorithms in large multi-electrode-based recording systems. The techniques are then applied to a previously published algorithm, on its own, unsuited for large electrode set recordings. Further, a real-time low-latency high-performance VLSI hardware architecture of the modified algorithm is presented, featuring a folded structure capable of processing the activity of hundreds of neurons simultaneously. The hardware is reconfigurable “on-the-fly” and adaptable to the nonstationarities of neuronal recordings. By transmitting exclusively spike time stamps and/or spike waveforms, its real-time processing offers the possibility of data bandwidth and data storage reduction.

Spike sorting, i.e., the separation of the firing activity of different neurons from extracellular measurements, is a crucial but often error-prone step in the analysis of neuronal responses. Usually, three different problems have to be solved: the detection of spikes in the extracellular recordings, the estimation of the number of neurons and their prototypical (template) spike waveforms, and the assignment of individual spikes to those putative neurons. If the template spike waveforms are known, template matching can be used to solve the detection and classification problem. Here, we show that for the colored Gaussian noise case the optimal template matching is given by a form of linear filtering, which can be derived via linear discriminant analysis. This provides a Bayesian interpretation for the well-known matched filter output. Moreover, with this approach it is possible to compute a spike detection threshold analytically. The method can be implemented by a linear filter bank derived from the templates, and can be used for online spike sorting of multielectrode recordings. It may also be applicable to detection and classification problems of transient signals in general. Its application significantly decreases the error rate on two publicly available spike-sorting benchmark data sets in comparison to state-of-the-art template matching procedures. CMOS high-density transducer arrays enable fundamentally new neuroscientific insights through, e.g., facilitating investigation of axonal signaling characteristics, with the "axonal" side of neuronal activity being largely inaccessible to established methods. They also enable high-throughput monitoring of potentially all action potentials in a larger neuronal network (> 1000 neurons) over extended time to see developmental effects or effects of disturbances. Emerging multi-electrode-based brain-machine interfaces (BMIs) and large multi-electrode arrays used in in vitro experiments, enable recording of single neuron's activity on multiple electrodes and allow for an in-depth investigation of neural preparations, even at a sub-cellular level. However, the use of these devices entails stringent area and power consumption constraints for the signal-processing hardware units. In addition, the high autonomy of these units and an ability to automatically adapt to changes in the recorded neural preparations is required. Implementing spike detection in close proximity to recording electrodes offers the advantage of reducing the transmission data bandwidth. By eliminating the need of transmitting the full, redundant recordings of neural activity and by transmitting only the spike waveforms or spike times, significant power savings can be achieved in the majority of cases. Here, we present a low-complexity, unsupervised, adaptable, real-time spike-detection method targeting multi-electrode recording devices and compare this method to other spike-detection methods with regard to complexity and performance.

Emerging complementary metal oxide semiconductor (CMOS)-based, high-density microelectrode array (HD-MEA) devices provide high spatial resolution at subcellular level and a large number of readout channels. These devices allow for simultaneous recording of extracellular activity of a large number of neurons with every neuron being detected by multiple electrodes. To analyze the recorded signals, spiking events have to be assigned to individual neurons, a process referred to as “spike sorting.” For a set of observed signals, which constitute a linear mixture of a set of source signals, independent component (IC) analysis (ICA) can be used to demix blindly the data and extract the individual source signals. This technique offers great potential to alleviate the problem of spike sorting in HD-MEA recordings, as it represents an unsupervised method to separate the neuronal sources. The separated sources or ICs then constitute estimates of single-neuron signals, and threshold detection on the ICs yields the sorted spike times. However, it is unknown to what extent extracellular neuronal recordings meet the requirements of ICA. In this paper, we evaluate the applicability of ICA to spike sorting of HD-MEA recordings. The analysis of extracellular neuronal signals, recorded at high spatiotemporal resolution, reveals that the recorded data cannot be modeled as a purely linear mixture. As a consequence, ICA fails to separate completely the neuronal signals and cannot be used as a stand-alone method for spike sorting in HD-MEA recordings. We assessed the demixing performance of ICA using simulated data sets and found that the performance strongly depends on neuronal density and spike amplitude. Furthermore, we show how postprocessing techniques can be used to overcome the most severe limitations of ICA. In combination with these postprocessing techniques, ICA represents a viable method to facilitate rapid spike sorting of multidimensional neuronal recordings.

In the case of extracellular recordings, spike detection algorithms are necessary in order to retrieve information about neuronal activity form the data. We present a new spike detection algorithm which is based on methods from the field of blind equalization and beamforming. In contrast to existing approaches, our method estimates several waveforms directly from the data and corresponding linear filters are constructed. The estimation is done in an unsupervised manner, and the few parameters of the algorithm are intuitive to set. The algorithm allows for superior detection performance, even when multiple neurons with various waveforms are present in the data. We compare our method with current state-of-the-art spike detection algorithms, and show that the proposed method achieves favorable results.

We present a method which allows for the blind source separation of sparse overcomplete mixtures. In this method, linear filters are used to find a new representation of the data and to enhance the signal-to-noise ratio. Further, "Deconfusion", a method similar to the independent component analysis, decorrelates the filter outputs. In particular, the method was developed to extract neural activity signals from extracellular recordings. In this sense, the method can be viewed as a combined spike detection and classification algorithm. We compare the performance of our method to those of existing spike sorting algorithms, and also apply it to recordings from real experiments with macaque monkeys.

The brain decodes the visual scene from the action potentials of ∼20 retinal ganglion cell types. Among the retinal ganglion cells, direction-selective ganglion cells (DSGCs) encode motion direction. Several studies have focused on the encoding or decoding of motion direction by recording multiunit activity, mainly in the visual cortex. In this study, we simultaneously recorded from all four types of ON-OFF DSGCs of the rabbit retina using a microelectronics-based high-density microelectrode array (HDMEA) and decoded their concerted activity using probabilistic and linear decoders. Furthermore, we investigated how the modification of stimulus parameters (velocity, size, angle of moving object) and the use of different tuning curve fits influenced decoding precision. Finally, we simulated ON-OFF DSGC activity, based on real data, in order to understand how tuning curve widths and the angular distribution of the cells' preferred directions influence decoding performance. We found that probabilistic decoding strategies outperformed, on average, linear methods and that decoding precision was robust to changes in stimulus parameters such as velocity. The removal of noise correlations among cells, by random shuffling trials, caused a drop in decoding precision. Moreover, we found that tuning curves are broad in order to minimize large errors at the expense of a higher average error, and that the retinal direction-selective system would not substantially benefit, on average, from having more than four types of ON-OFF DSGCs or from a perfect alignment of the cells' preferred directions.

Extracellular recordings are a key tool to record the activity of neurons in vivo. Especially in the case of experiments with behaving animals, however, the tedious procedure of electrode placement can take a considerable amount of expensive and restricted experimental time. Furthermore, due to tissue drifts and other sources of variability in the recording setup, the position of the electrodes with respect to the recorded neurons can change causing low recording quality. The contributions of this work are threefold. We introduce a quality measure for the recording position of the electrode which should be maximized during recordings and is especially suitable for the use of multi-electrodes. An automated positioning system based on this quality measure is proposed. The system is able to find favorable recording positions and adapts the electrode position smoothly to changes of the neuron positions. Finally, we evaluate the system using a new simulator for extracellular recordings based on realistically reconstructed 3D neurons.

Multi-channel electrical recordings of neural activity in the brain is an increasingly powerful method revealing new aspects of neural communication, computation, and prosthetics. However, while planar silicon-based CMOS devices in conventional electronics scale rapidly, neural interface devices have not kept pace. Here, we present a new strategy to interface silicon-based chips with three-dimensional microwire arrays, providing the link between rapidly-developing electronics and high density neural interfaces. The system consists of a bundle of microwires mated to large-scale microelectrode arrays, such as camera chips. This system has excellent recording performance, demonstrated via single unit and local-field potential recordings in isolated retina and in the motor cortex or striatum of awake moving mice. The modular design enables a variety of microwire types and sizes to be integrated with different types of pixel arrays, connecting the rapid progress of commercial multiplexing, digitisation and data acquisition hardware together with a three-dimensional neural interface., Science Advances, 6 (12), ISSN:2375-2548
