Aerogel scaffolds are nanostructured materials with beneficial properties for tissue engineering applications. The tracing of the state of the aerogels after their implantation is challenging due to their variable biodegradation rate and the lack of suitable strategies capable of in vivo monitoring the scaffolds. Upconversion nanoparticles (UCNPs) have emerged as advanced tools for in vitro bioimaging because of their fluorescence properties. In this work, highly fluorescent UCNPs were loaded into aerogels to obtain theranostic implants for tissue engineering and bioimaging applications. 3D-printed alginate-hydroxyapatite aerogels labeled with UCNPs were manufactured by 3D-printing and supercritical CO2 drying to generate personalize-to-patient aerogels. The physicochemical performance of the resulting structures was evaluated by printing fidelity measurements, nitrogen adsorption-desorption analysis, and different microscopies (confocal, transmission and scanning electron microscopies). Stability of the aerogels in terms of physicochemical properties was also tested after 3 years of storage. Biocompatibility was evaluated in vitro by different cell and hemocompatibility assays, in ovo and in vivo by safety and bioimaging studies using different murine models. Cytokines profile, tissue index and histological evaluations of the main organs unveiled an in vivo downregulation of the inflammation after implantation of the scaffolds. UCNPs-decorated aerogels were first-time manufactured and long-term traceable by fluorescence-based bioimaging until 3 weeks post-implantation, thereby endorsing their suitability as tissue engineering and theranostic nanodevices (i.e. bifunctional implants).

Abstract Background Habitat transitions have considerable consequences in organism homeostasis, as they require the adjustment of several concurrent physiological compartments to maintain stability and adapt to a changing environment. Within the range of molecules with a crucial role in the regulation of different physiological processes, neuropeptides are key agents. Here, we examined the coding status of several neuropeptides and their receptors with pleiotropic activity in Cetacea. Results Analysis of 202 mammalian genomes, including 41 species of Cetacea, exposed an intricate mutational landscape compatible with gene sequence modification and loss. Specifically for Cetacea, in the 12 genes analysed we have determined patterns of loss ranging from species-specific disruptive mutations (e.g. neuropeptide FF-amide peptide precursor; NPFF) to complete erosion of the gene across the cetacean stem lineage (e.g. somatostatin receptor 4; SSTR4). Conclusions Impairment of some of these neuromodulators may have contributed to the unique energetic metabolism, circadian rhythmicity and diving response displayed by this group of iconic mammals.

Abstract We describe the geographical variation in tree species composition across Amazonian forests and show how environmental conditions are associated with species turnover. Our analyses are based on 2023 forest inventory plots (1 ha) that provide abundance data for a total of 5188 tree species. Within-plot species composition reflected both local environmental conditions (especially soil nutrients and hydrology) and geographical regions. A broader-scale view of species turnover was obtained by interpolating the relative tree species abundances over Amazonia into 47,441 0.1-degree grid cells. Two main dimensions of spatial change in tree species composition were identified. The first was a gradient between western Amazonia at the Andean forelands (with young geology and relatively nutrient-rich soils) and central–eastern Amazonia associated with the Guiana and Brazilian Shields (with more ancient geology and poor soils). The second gradient was between the wet forests of the northwest and the drier forests in southern Amazonia. Isolines linking cells of similar composition crossed major Amazonian rivers, suggesting that tree species distributions are not limited by rivers. Even though some areas of relatively sharp species turnover were identified, mostly the tree species composition changed gradually over large extents, which does not support delimiting clear discrete biogeographic regions within Amazonia.

Kelp forests are facing a global decline due to climate change and human-induced stressors, underlining the urgency for proactive interventions. Among the most used restoration methods, “green gravel” has emerged as a promising solution for the recovery of degraded kelp forests. While initial findings suggest its potential effectiveness, green gravel remains a novel approach that requires fine-tuned protocols and optimisation across all stages of the process. In this study, we assessed the performance of two different seeding densities for kelp growth on green gravel to optimise the use of seeding material. Our results show that, while the juveniles in the high-density treatment grew faster, they also had a higher mortality rate during green gravel production. This was a hypothesised outcome, as growing sporophytes reared under laboratory conditions allows for faster production of a large number of individuals, increasing competition for space, which may drive higher mortality rates. Comprehensive experimentation is essential to unlock the full potential of green gravel and ensure its efficiency in all process steps, to achieve successful kelp forest restoration. Well-defined and optimised protocols are indispensable for minimising production costs, simplifying logistics, and allowing future efforts to scale up.

Macroalgae are a natural oceanic resource of inexhaustible abundance for the biomass energy industry with growth rates that are three to four times greater than those of terrestrial plants. The objective of this study was to evaluate the sustainability of macroalgae as biomass for biorefining through two investigations. Firstly, the deposition of macroalgae was sampled through 28 collections on seven beaches in the city of Maceió, Brazil, over a two-year period using a zigzag sampling method, covering a deposition area of 135,000 m2. From this, it was estimated that daily collection would yield 5.03 t/ha of dry biomass. Secondly, the calorific values of macroalgal biomass energy and pellet compounds were calculated. The lower calorific value (8.82 MJ/kg) found from a compound of 13 species analyzed was similar to that of the main biomass used in Brazil to obtain energy, i.e., sugarcane bagasse, which has been evaluated as 8.91 MJ/kg. Macroalgal biomass in the form of condensed energy pellets was found to have a higher calorific value of 20.18 MJ/kg, i.e., 1.2% greater than the average for terrestrial biomass pellets. Based on the results obtained, it was observed that macroalgal biomass has the possibility of becoming a new renewable feedstock with potential for bioenergy. The estimates for the deposition of biomass show possibilities for producing biofuels from marine algal raw material, which provides scope for creating another sustainable alternative for global energy issues with a reduction in environmental problems.

The timely establishment of functional neo-vasculature is pivotal for successful tissue development and regeneration, remaining a central challenge in tissue engineering. In this study, we present a novel (micro)vascularization strategy that explores the use of specialized “vascular units” (VUs) as building blocks to initiate blood vessel formation and create perfusable, stroma-embedded 3D microvascular networks from the bottom-up. We demonstrate that VUs composed of endothelial progenitor cells and organ-specific fibroblasts exhibit high angiogenic potential when embedded in fibrin hydrogels. This leads to the formation of VUs-derived capillaries, which fuse with adjacent capillaries to form stable microvascular beds within a supportive, extracellular matrix-rich fibroblastic microenvironment. Using a custom-designed biomimetic fibrin-based vessel-on-chip (VoC), we show that VUs-derived capillaries can inosculate with endothelialized microfluidic channels in the VoC and become perfused. Moreover, VUs can establish capillary bridges between channels, extending the microvascular network throughout the entire device. When VUs and intestinal organoids (IOs) are combined within the VoC, the VUs-derived capillaries and the intestinal fibroblasts progressively reach and envelop the IOs. This promotes the formation of a supportive vascularized stroma around multiple IOs in a single device. These findings underscore the remarkable potential of VUs as building blocks for engineering microvascular networks, with versatile applications spanning from regenerative medicine to advanced in vitro models.

In very high gravity (VHG) fermentation, yeast cells are subjected to a multitude of challenging conditions, including the osmotic pressure exerted by the high sugar content of the wort and the stress factors associated with the high ethanol concentrations present at the end of the fermentation cycle. The response of this biological system to abiotic stresses may be enhanced through biochemical and physiological routes. Silica may play a significant role in regulating the cellular homeostasis of yeast. Alternatively, it is expected that this outcome may be achieved through biochemical responses from the effects of vitamins on yeast cells and the physiological yeast route changing by the culture medium aeration. The objective of this study was to investigate the effects of adding 500 mg L−1 of silica on corn ethanol wort medium and the possibility of supplementing the same wort with vitamins alongside aeration (0.2 v v−1 min−1) as an alternative resource to sustain the fermentation yield rather than adding silica in a fed-batch fermentation cycle with yeast recycling. Upon completion of the five fermentation cycles, yeast samples subjected to the treatment with the addition of silica exhibited a 3.1% higher fermentation yield in comparison to the results observed in the vitamins plus aeration medium bath. Even though greater biomass production (19.1 g L−1) was observed through aerobic yeast behavior in vitaminized supplemented corn medium, the provided silica had a more beneficial effect on yeast stress relief for very high gravity fermentation in a corn hydrolyzed wort with cell recycling.

Spatial epidemiology tools play a critical role in effectively allocating resources to curb the spread of animal diseases. This study focuses on classical scrapie (CS), an animal prion disease identified in Portugal, which infects small ruminant flocks and has been shown to be experimentally transmissible to wild cervids. Utilising remote sensing technologies and semi-automatic classification models, we aimed to evaluate the risk of interspecies prion transmission from domestic small ruminants to wild cervids (hosts). To achieve this, we gathered data related to hosts and infected small ruminant flocks. Furthermore, we collected and processed freely available, medium-resolution satellite imagery to derive vegetative and biophysical spectral indices capable of representing the primary habitat features. By employing a pixel-based species distribution model, we integrated the compiled geographical distribution data and spectral data with five supervised classification algorithms (random forest, classification tree analysis, artificial neural network, generalised linear model, and generalised additive model). The consensus map allowed accurate predictions of spatialised regions exhibiting spectral characteristics similar to where CS and its hosts were initially identified. By overlapping suitable territories for disease and host occurrence, we created a spatially explicit tool that assesses the risk of prion spill-over from domestic small ruminants to wild cervids. The described methodology is highly replicable and freely accessible, thus emphasising its practical utility. This study underscores the substantial contribution of model-based spatial analysis to disease monitoring and lays the groundwork for defining populations at risk and implementing targeted control and prevention strategies, thus safeguarding both animal and public health.

This study presents the first record of T. lindbergi from the southwestern Atlantic Ocean, based on specimens collected from the smalleye croaker Nebris microps Cuvier (Sciaenidae), off the coast of Maranhão Island, State of Maranhão, Brazil. Our approach included a morphological analysis complemented by DNA sequencing (28S, ITS2 rDNA, and cox1 mtDNA). Our phylogenetic analysis revealed the affinity of T. lindbergi to its congener T. laticaudi Parukhin, 1969, a digenean parasite commonly found in hydrophiine snakes inhabiting the Pacific Ocean. The interspecific divergence between T. lindbergi and T. laticaudi measures 3.80% for 28S, 7.49–7.64% for ITS2, and 16.29–16.70% for cox1. Our findings expand the documented geographic range of T. lindbergi into the southwestern Atlantic Ocean, report a novel host record, and increase the number of hemiurids known from Brazil to 30 species. Additionally, this study represents the initial documentation of a marine digenean fish within the North Brazil Shelf.

Yeast is one of the co-products of ethanol plants, which can be used as a nutritional supplement in animal feed due to its high protein content. Given the importance of yeast contribution to the nutritional properties of DDG (dried distillers’ grains), the aim of this study was to assess how different levels of aeration affect the biomass production and the quality of yeast providing new insights into yeast production, offering an alternative source of income for the corn ethanol industry. For this purpose, yeasts were grown in a fed-batch process, and different concentrations of aeration in the medium were tested, namely 0.5, 1.0, and 1.5 volume of air per volume of wort per minute (v v−1 min−1). At the end of the cellular biomass production process, yeasts grown with 0.5 (v v−1 min−1) aeration in the reactor showed higher biomass formation (19.86 g L−1), cellular yield (g g−1), and a lower formation of succinic acid (0.70 g L−1) and acetic acid (0.11 g L−1). Aeration influenced an increase of 1.0% in the protein content in yeast. In conclusion, lower levels of aeration in the yeast production process enables more efficient sugar utilization for biomass formation and is a potential strategy to increase the protein content and the commercial value of DDG.

Leukodystrophies represent a large and complex group of inherited disorders affecting the white matter of the central nervous system. Adult-onset leukoencephalopathy with axonal spheroids and pigmented glia (ALSP) is a rare leukodystrophy which still needs the proper identification of diagnostic, prognostic, and monitoring biomarkers. The aim of this study was to determine the diagnostic and prognostic value of chitinases and neurofilament light chain as biomarkers for ALSP. A cross-sectional study was performed to analyze cerebrospinal fluid levels of chitinases (chitotriosidase and chitinase 3-like 2) and neurofilament light chain in five different groups: (i) normal health individuals; (ii) patients with definitive diagnosis of ALSP and genetic confirmation; (iii) asymptomatic patients with CSF1R variants; (iv) patients with other adult-onset leukodystrophies; and (v) patients with amyotrophic lateral sclerosis (external control group). Chitinase levels showed a statistical correlation with clinical assessment parameters in ALSP patients. Chitinase levels were also distinct between ALSP and the other leukodystrophies. Significant differences were noted in the levels of chitinases and neurofilament light chain comparing symptomatic (ALSP) and asymptomatic individuals with CSF1R variants. This study is the first to establish chitinases as a potential biomarker for ALSP and confirms neurofilament light chain as a good biomarker for primary microgliopathies.

Protein substitutes (PS) without tyrosine (Tyr) and phenylalanine (Phe), are an essential source of synthetic protein in the treatment of tyrosinemia (HT). In the UK, the only available protein substitutes for HT are Tyr/ Phe free amino acid liquid or powders or formulations based on glycomacropeptide (CGMP). A tablet Tyr/ Phe free amino acid supplement (AAT) has now been introduced. The aim of this two-part prospective, longitudinal intervention study was to assess the efficacy, acceptability, and tolerance of AAT in children aged >8 years with HTI. Part 1: was a 28-day acceptability/ tolerance study, part 2, was a 12-month extension study examining efficacy of AAT. Anthropometry and blood Tyr/ Phe were assessed. All subjects were taking NTBC [2-(2-nitro-4-triflourothybenzoyl) cyclohexane-1, 3-dione] with a Tyr restricted diet. Eight subjects with HTI were recruited (4 boys, and 4 girls) with a median age of 14.3y (range 10.4–17.3); 3 were Caucasian and 5 of Pakistani origin. The median (range) protein equivalent from PS was 60 g/d (50–60), natural protein 20 g/d (15–30), and NTBC 30 mg/d (25–80). No subjects were taking Phe supplements. Five (63%) subjects completed part 1, with 4 taking all their PS requirements as AAT. Subjects reported AAT were tasteless and had no odour. No adverse gastrointestinal symptoms were recorded, with two reporting improvements in abdominal discomfort. At 12 months, 4 subjects had a non-significant decrease in blood Tyr/ Phe compared to the 12 months pre-treatment. Median blood Tyr (μmol/ L) pre-intervention was 500 (320–590); and at 12 months, 450 (290–530). Median blood Phe (μmol/L) pre-intervention was 40 (30–40); and at 12 months 30 (30–50). Median height z scores remained unchanged, but there was a small decrease in weight z score (pre-study weight − 0.1 (−1.4 to1.1), 12 m − 0.3 (−1.4 to 1.3) and BMI (pre- study BMI 0.2 (−2 to 1.4), and 12 m, −0.1 (−2.5 to 1.5)). Conclusion: AAT were useful for some adolescents with HTI who struggled with the taste and volume of conventional powdered and liquid PS.

Background and objectives: Prevalence estimates for classical homocystinuria (HCU) are variable and likely underestimated due to underdiagnosis. Claims data represent a strong but seldom used resource to analyze prevalence of HCU. The aim of this study was to estimate a prevalence range of HCU in the US utilizing a combination of diagnosis codes, total homocysteine levels, and clinical presentations indicative of HCU. Methods: This was a non-interventional retrospective cohort study, using Optum's de-identified Market Clarity Data, with a patient identification period from January 01, 2016, through September 30, 2021. An algorithm was developed to identify 2 cohorts of patients using broad and strict definitions of HCU. The index date was the date within the identification period on which the first criterion was met for the inclusion criteria. Baseline demographics, clinical characteristics, and complications were assessed and summarized using descriptive statistics. Crude and standardized prevalence estimates were calculated. Results: There were 3880 and 633 patients that met the relevant inclusion criteria for the broad and strict cohorts, respectively. The projected US prevalence of HCU was calculated to be 17,631 and 3466 based on the broad and strict definitions, respectively. The average annual standardized prevalence across 2016–2020 was 5.29 and 1.04 per 100,000 people for the broad and strict cohorts, respectively. Conclusions: Prevalence estimates of HCU vary depending on databases or datasets used and identification criteria. Many patients with clinical presentations suggesting a diagnosis of HCU did not have an associated diagnosis, potentially indicating underdiagnosis or underreporting. Future research should study alternative methods, such as the identification algorithm in our analysis, to better diagnose and understand the true prevalence of HCU.

Polycystic Ovary Syndrome (PCOS) is the most common endocrine disorder that affects women of reproductive age, representing the primary cause of anovulatory infertility. The nonapeptide oxytocin (OT) plays an important role in cognitive, emotional, and reproductive functions in human beings. Oxytocin receptors are expressed in several body parts, including the ovaries. Despite this, the possible role played by oxytocin in symptoms of PCOS is not clear. The present systematic review aimed at understanding the presence of possible oxytocin level alterations in PCOS, the connection between alterations of OT levels and the symptoms of PCOS, and the effect of oxytocin administration in PCOS. After a systematic search in the principal databases, eight studies, five human and three animal, were included. Four human studies and one animal study highlighted the role played by oxytocin in fertility issues related to PCOS. Three human and two animal studies investigated the role of body weight and OT levels. Studies that analyzed oxytocin basal levels in women agreed that PCOS is associated with a reduction in the serum level of oxytocin. Two human studies and one animal study agreed about lower levels of oxytocin, confirming a possible implication of the dysfunction of OT in the pathogenesis of PCOS.

The InBIO Barcoding Initiative (IBI) Orthoptera dataset contains records of 420 specimens covering all the eleven Orthoptera families occurring in Portugal. Specimens were collected in continental Portugal from 2005 to 2021 and were morphologically identified to species level by taxonomists. A total of 119 species were identified corresponding to about 77% of all the orthopteran species known from continental Portugal.DNA barcodes of 54 taxa were made public for the first time at the Barcode of Life Data System (BOLD). Furthermore, the submitted sequences were found to cluster in 129 BINs (Barcode Index Numbers), 35 of which were new additions to the Barcode of Life Data System (BOLD). All specimens have their DNA barcodes publicly accessible through BOLD online database. Stenobothrus lineatus is recorded for the first time for continental Portugal. This dataset greatly increases the knowledge on the DNA barcodes and distribution of Orthoptera from Portugal. All DNA extractions and most specimens are deposited in the IBI collection at CIBIO, Research Center in Biodiversity and Genetic Resources.

The InBIO Barcoding Initiative (IBI) Dataset - DS-IBILP08 contains records of 2350 specimens of moths (Lepidoptera species that do not belong to the superfamily Papilionoidea). All specimens have been morphologically identified to species or subspecies level and represent 1158 species in total. The species of this dataset correspond to about 42% of mainland Portuguese Lepidoptera species. All specimens were collected in mainland Portugal between 2001 and 2022. All DNA extracts and over 96% of the specimens are deposited in the IBI collection at CIBIO, Research Center in Biodiversity and Genetic Resources.The authors enabled "The InBIO Barcoding Initiative Database: DNA barcodes of Portuguese moths" in order to release the majority of data of DNA barcodes of Portuguese moths within the InBIO Barcoding Initiative. This dataset increases the knowledge on the DNA barcodes of 1158 species from Portugal belonging to 51 families. There is an increase in DNA barcodes of 205% in Portuguese specimens publicly available. The dataset includes 61 new Barcode Index Numbers. All specimens have their DNA barcodes publicly accessible through BOLD online database and the distribution data can be accessed through the Global Biodiversity Information Facility (GBIF).

Abstract Background The reconstruction of the evolutionary history of organisms has been greatly influenced by the advent of molecular techniques, leading to a significant increase in studies utilizing genomic data from different species. However, the lack of standardization in gene nomenclature poses a challenge in database searches and evolutionary analyses, impacting the accuracy of results obtained. Results To address this issue, a Python class for standardizing gene nomenclatures, SynGenes, has been developed. It automatically recognizes and converts different nomenclature variations into a standardized form, facilitating comprehensive and accurate searches. Additionally, SynGenes offers a web form for individual searches using different names associated with the same gene. The SynGenes database contains a total of 545 gene name variations for mitochondrial and 2485 for chloroplasts genes, providing a valuable resource for researchers. Conclusions The SynGenes platform offers a solution for standardizing gene nomenclatures of mitochondrial and chloroplast genes and providing a standardized search solution for specific markers in GenBank. Evaluation of SynGenes effectiveness through research conducted on GenBank and PubMedCentral demonstrated its ability to yield a greater number of outcomes compared to conventional searches, ensuring more comprehensive and accurate results. This tool is crucial for accurate database searches, and consequently, evolutionary analyses, addressing the challenges posed by non-standardized gene nomenclature. Graphical abstract

ABSTRACTAmong the transmissible spongiform encephalopathies (TSEs), chronic wasting disease (CWD) in cervids is now a rising concern in wildlife within Europe, after the detection of the first case in Norway in 2016, in a wild reindeer and until June 2022 a total of 34 cases were described in Norway, Sweden and Finland. The definite diagnosis is post-mortem, performed in target areas of the brain and lymph nodes. Samples are first screened using a rapid test and, if positive, confirmed by immunohistochemistry and Western immunoblotting. The study of the genetics of the prion protein gene, PRNP, has been proved to be a valuable tool for determining the relative susceptibility to TSEs. In the present study, the exon 3 of PRNP gene of 143 samples from red deer (Cervus elaphus) and fallow deer (Dama dama) of Portugal was analysed. Three single nucleotide polymorphisms (SNPs) were found in red deer – codon A136A, codon T98A, codon Q226E – and no sequence variation was detected in fallow deer. The low genetic diversity found in our samples is compatible with previous studies in Europe. The comparison with results from North America suggests that the free-ranging deer from our study may present susceptibility to CWD, although lack of experimental data and the necessity of continuous survey are necessary to evaluate these populations.

Abstract Cutaneous leishmaniasis (CL) is considered a public health problem. Current treatments have disadvantages because they are invasive and have serious side effects, and thus there is a need for research into new, more effective pharmacological alternatives. Plants are promising sources of bioactive substances, and new analogues can be obtained through chemical reactions. The present study aimed to evaluate the antileishmanial effects of the analog dillapiole n-butyl ether (DBE) extracted from Piper aduncum leaves. The cytotoxic potential of DBE was evaluated at concentrations of 15.62 to 500 µM in peritoneal macrophages for 48 h, and in RAW 264.7 macrophages for 72 h using a dose-response method. The antileishmanial activity in L. amazonensis promastigotes used concentrations of 0.2 to 4.5 µM for 24, 48 and 72 h and the quantification of the cellular infection rate used a concentration of 4.5 µM of DBE against the amastigote forms internalized in macrophages for 24 h and 48 h. Nitric oxide was quantified from macrophages previously treated with DBE for 24 h and 48 h. The dosage of reactive oxygen species used a concentration of 4.5 µM of DBE incubated together with dichlorofluorescein acetate for 1, 3, 6, and 24 h. For the molecular modeling of DBE, the Leishmania protein, available in the “Protein Data Bank” database, was used. The studied molecule was not toxic to cells and presented a CC50 of 413 µM in peritoneal macrophages and 373.5 µM in RAW 264.7. The analogue inhibited promastigote forms of L. amazonensis with an IC50 of 1.6 µM for 72 h. DBE presented an infection rate of 17% and 12%, dillapiole of 24% and 14% and Pentacarinat® of 10% and 9% over 48 h. DBE demonstrated a binding energy of -7.8 for the U53 enzyme. It is concluded that the analogue showed promising antileishmanial activity for future in vivo tests.

Group B Streptococcus (GBS) is a major human and animal pathogen that threatens public health and food security. Spill-over and spill-back between host species is possible due to adaptation and amplification of GBS in new niches but the evolutionary and functional mechanisms underpinning those phenomena are poorly known. Based on analysis of 1,254 curated genomes from all major GBS host species and six continents, we found that the global GBS population comprises host-generalist, host-adapted and host-restricted sublineages, which are found across host groups, preferentially within one host group, or exclusively within one host group, respectively, and show distinct levels of recombination. Strikingly, the association of GBS genomes with the three major host groups (humans, cattle, fish) is driven by a single accessory gene cluster per host, regardless of sublineage or the breadth of host spectrum. Moreover, those gene clusters are shared with other streptococcal species occupying the same niche and are functionally relevant for host tropism. Our findings demonstrate (1) the heterogeneity of genome plasticity within a bacterial species of public health importance, enabling the identification of high-risk clones; (2) the contribution of inter-species gene transmission to the evolution of GBS; and (3) the importance of considering the role of animal hosts, and the accessory gene pool associated with their microbiota, in the evolution of multi-host bacterial pathogens. Collectively, these phenomena may explain the adaptation and clonal expansion of GBS in animal reservoirs and the risk of spill-over and spill-back between animals and humans.

Abstract Renealmia comprises approximately 85 herbaceous and rhizomatous plants in Africa and America known for their metabolite diversity. This study characterized the anatomy and histochemistry of the vegetative organs of three native Brazilian Renealmia species, R. breviscapa and R. nicolaioides, collected from the Amazon Forest in Acre and R. chrysotricha from the Atlantic Forest in Rio de Janeiro. Samples were fixed and processed using conventional plant anatomy and micromorphology techniques, including histochemical tests to identify classes of compounds in vegetative organs. Characteristics including oil idioblasts, dorsiventral leaf structure, tetracytic stomata, unicellular trichomes, and prismatic crystals were common among the studied species of Renealmia, and thus significant for the family. Distinctive characteristics were identified in the vegetative organs of the three species, including the cellular distribution of root phloem, covering of rhizome tissue, different types and distributions of idioblasts and phenolic trichomes in rhizomes and leaves, presence or absence of petiole, and subepidermal layer in the intercostal region of leaves. Mineral content importance as a diagnostic characteristic of the genus is also emphasized. The results of this study advance the understanding of the distinctive anatomical characteristics of the three species of Renealmia in different environments, two in the Atlantic Forest and one in the Amazon Forest.

The worldwide dispersal of the ectoparasitic mite Varroa destructor from its Asian origins has fundamentally transformed the relationship of the honey bee (Apis mellifera) with several of its viruses, via changes in transmission and/or host immunosuppression. The extent to which honey bee-virus relationships change after Varroa invasion is poorly understood for most viruses, in part because there are few places in the world with several geographically close but completely isolated honey bee populations that either have, or have not, been exposed long-term to Varroa, allowing for separate ecological, epidemiological, and adaptive relationships to develop between honey bees and their viruses, in relation to the mite's presence or absence. The Azores is one such place, as it contains islands with and without the mite. Here, we combined qPCR with meta-amplicon deep sequencing to uncover the relationship between Varroa presence, and the prevalence, load, diversity, and phylogeographic structure of eight honey bee viruses screened across the archipelago. Four viruses were not detected on any island (ABPV-Acute bee paralysis virus, KBV-Kashmir bee virus, IAPV-Israeli acute bee paralysis virus, BeeMLV-Bee macula-like virus); one (SBV-Sacbrood virus) was detected only on mite-infested islands; one (CBPV-Chronic bee paralysis virus) occurred on some islands, and two (BQCV-Black queen cell virus, LSV-Lake Sinai virus,) were present on every single island. This multi-virus screening builds upon a parallel survey of Deformed wing virus (DWV) strains that uncovered a remarkably heterogeneous viral landscape featuring Varroa-infested islands dominated by DWV-A and -B, Varroa-free islands naïve to DWV, and a refuge of the rare DWV-C dominating the easternmost Varroa-free islands. While all four detected viruses investigated here were affected by Varroa for one or two parameters (usually prevalence and/or the Richness component of ASV diversity), the strongest effect was observed for the multi-strain LSV. Varroa unambiguously led to elevated prevalence, load, and diversity (Richness and Shannon Index) of LSV, with these results largely shaped by LSV-2, a major LSV strain. Unprecedented insights into the mite-virus relationship were further gained from implementing a phylogeographic approach. In addition to enabling the identification of a novel LSV strain that dominated the unique viral landscape of the easternmost islands, this approach, in combination with the recovered diversity patterns, strongly suggests that Varroa is driving the evolutionary change of LSV in the Azores. This study greatly advances the current understanding of the effect of Varroa on the epidemiology and adaptive evolution of these less-studied viruses, whose relationship with Varroa has thus far been poorly defined.

A educação moral é de grande relevância para o desenvolvimento do ser humano, as abordagens morais, quando inseridas nos currículos, contribuem na qualificação docente e melhoria do processo de ensino-aprendizagem. Considerando essas questões, esta pesquisa centralizou-se na educação em valores morais nos espaços escolares, destacando a sua importância no ensino de Ciências. O objetivo foi traçar um perfil sobre formação moral apresentado por professores de Ciências. O referencial teórico foi baseado nas cinco linhas paradigmáticas de Puig. É uma pesquisa qualitativa, em que a obtenção dos dados foi realizada por meio da escala Likert, um questionário aplicado para dez professores. Os resultados indicaram que a maioria dos professores apresentou um perfil paradigmático sobre formação moral polissêmico e conhecimento superficial.

Solenostemma oleifolium is a tropical plant growing in the Algerian desert that is traditionally used to treat several illnesses, including infection. We investigate essential oil components from leaves of Solenostemma oleifolium (EOSO) and its antibacterial activity. Using Gas Chromatography and Mass Spectrometry (GC-MS), twenty compounds were identified in EOSO, including linalool (57.10%), terpineol (12.95%), trans-geraniol (12.65%), and nerol (4.67%). Nuclear magnetic resonance (NMR) analysis allowed us to confirm linalool as the main component of EOSO. Antibacterial activity was tested by agar diffusion and microdilution methods for minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC). For EOSO, the inhibition diameters ranged from 16.79 to 39.84 mm, the MIC ranged from 1.066 to 8.54 mg mL-1 and the MBC ranged from 20 to 100 mg mL-1. For linanool, the inhibition diameters ranged from 11.1 to 31.87 mm, the MIC ranged from 2.68 to 14.3 mg mL-1 and, the MBC ranged from 40 to 100 mg mL-1.EOSO and linalool exhibited significant antibacterial activity against all the tested bacteria. This study confirmed the antibacterial activity of the S. oleifolium essential oil and that linalool is the principal constituent of the EOSO.

YbbN/CnoX are proteins that display a Thioredoxin (Trx) domain linked to a tetratricopeptide domain. YbbN from Escherichia coli (EcYbbN) displays a co-chaperone (holdase) activity that is induced by HOCl. Here, we compared EcYbbN with YbbN proteins from Xylella fastidiosa (XfYbbN) and from Pseudomonas aeruginosa (PaYbbN). EcYbbN presents a redox active Cys residue at Trx domain (Cys63), 24 residues away from SQHC motif (SQHC[N24]C) that can form mixed disulfides with target proteins. In contrast, XfYbbN and PaYbbN present two Cys residues in the CXXC (CAPC) motif, while only PaYbbN shows the Cys residue equivalent to Cys63 of EcYbbN. Our phylogenetic analysis revealed that most of the YbbN proteins are in the bacteria domain of life and that their members can be divided into four groups according to the conserved Cys residues. EcYbbN (SQHC[N24]C), XfYbbN (CAPC[N24]V) and PaYbbN (CAPC[N24]C) are representatives of three sub-families. In contrast to EcYbbN, both XfYbbN and PaYbbN: (1) reduced an artificial disulfide (DTNB) and (2) supported the peroxidase activity of Peroxiredoxin Q from X. fastidiosa, suggesting that these proteins might function similarly to the canonical Trx enzymes. Indeed, XfYbbN was reduced by XfTrx reductase with a high catalytic efficiency (kcat/Km = 1.27 x 107 M−1 s−1), similar to the canonical XfTrx (XfTsnC). Furthermore, EcYbbN and XfYbbN, but not PaYbbN displayed HOCl-induced holdase activity. Remarkably, EcYbbN gained disulfide reductase activity while lost the HOCl-activated chaperone function, when the SQHC was replaced by CQHC. In contrast, the XfYbbN CAPA mutant lost the disulfide reductase activity, while kept its HOCl-induced chaperone function. In all cases, the induction of the holdase activity was accompanied by YbbN oligomerization. Finally, we showed that deletion of ybbN gene did not render in P. aeruginosa more sensitive stressful treatments. Therefore, YbbN/CnoX proteins display distinct properties, depending on the presence of the three conserved Cys residues.

Micro- and nanoplastics (MNPs) are an important atmospheric aerosol constituent. However, there still needs to be a standard procedure for their sampling and size fractionation, which is an obstacle to the aggregation and critical analysis of results obtained by different research groups. This review focuses on the sampling and fractionation methodologies used for MNPs. Moreover, a streamlined, simplified methodology for sampling and fractionation is proposed.

Yeast recycling, which is a common practice in sugarcane ethanol plants, could be expanded if it could be successfully implemented in corn-based ethanol production. However, the process of recycling the yeast remaining after fermentation is hampered by contaminating microorganisms that reduce the fermentation efficiency and compete with the yeast for the fermentable sugars. Currently, antibiotics are used to control microbial contamination. This study proposes chlorine dioxide and electron beam irradiation as alternative control methods for completely inactivating contaminants and minimizing their effect on recycled yeast. For that, wort sterilization using electron radiation (20 kGy) and treatment with a chemical biocide, namely chlorine dioxide (125 mg kg−1), were compared with non-treated wort. Five fermentation cycles were performed using fed-batch systems with 300 g L−1 of fermentable sugars. The results obtained in this study showed the inactivation of contaminants under the effect of electron beam irradiation, which led to an increase in the productivity, yield, and efficiency of fermentation by 0.21 g L−1h−1, 2.6%, and 4.7%, respectively. However, ClO2 did not show promising results in reducing contamination or improving fermentative parameters. Thus, electron beam irradiation of contaminated wort may be a suitable alternative to chemical biocides and would allow the use of recycled yeast in corn-based ethanol plants.

The Neotropical deer genus Mazama is characterized by homoplastic morphological characters, a high karyotypic diversity, and a polyphyletic condition. The species of the genus have been recovered into two multigeneric lineages, the subtribes Odocoileina and Blastocerina, of the tribe Odocoileini (New World deer) in the family Cervidae. Within the Blastocerina, gray brockets include two non-sister species, Subulo gouazoubira, occurring south of the Amazon region, and Passalites nemorivagus, occurring in the Guianas and in the Amazon region. We clarify the taxonomic status and phylogenetic position of Mazama americana citus Osgood, 1912 (referred to as either S. gouazoubira or P. nemorivagus by other authors). We collected a topotype of M. a. citus from the eastern shore of Lake Maracaibo, Venezuela, characterize it morphologically and cytogenetically (conventional banding and fluorescence in situ hybridization), and carry out a phylogenetic analysis of its whole mitogenome and Cytb alongside two additional specimens of M. a. citus from northwestern Venezuela. Our analyses reveal the topotype to be a large gray brocket with a cinnamon band above the eyes and 2n = 61 and FN = 70 karyotype. Using cattle whole chromosome painting and bacterial artificial chromosome X probes, we determined its karyotype to differ in at least 10 rearrangements from that of S. gouazoubira. Bayesian inference recovers M. a. citus within the Blastocerina subtribe, separated phylogenetically from other gray brockets (100% branch value), revealing the Osgood’s gray brocket to be a valid species that should be assigned to a new genus. We propose the generic name Bisbalus, with Bisbalus citus (Osgood, 1912) as the type species.

A population of burrowing owls (Athene cunicularia) under professional care at Zoomarine Portugal presented with sudden respiratory clinical signs. Clinical management included a thorough diagnosis plan, including in-house fecal analysis that revealed the presence of ovoid unioperculate eggs. In the postmortem examination of one hyperacute dyspneic specimen, adult nematode parasites were collected and identified based on their morphology as Cyathostoma (Hovorkonema) americana. Even after a broad-spectrum deworming protocol as part of the treatment and metaphylaxis approach, the incidence of parasitic reinfection was high. The complete clinical resolution was only accomplished after the identification and management of the possible focus of infection, a wild population of cattle egrets (Bubulcus ibis) that frequently congregated above the owls’ habitat. To the authors’ best knowledge, this is the first report of infection by Cyathostoma (Hovorkonema) americana in burrowing owls. Although nematodes of the family Syngamidae are not commonly included in the differential diagnosis of infectious respiratory agents of birds of the order Strigiformes, this report highlights the possibility of opportunistic parasitism in a zoological context, especially where there is a continued proximity to free-ranging avifauna.

Background: This paper presents research on the comparison and evaluation of lower limb power, movement, position on the court, and effectiveness in the players of men’s volleyball. The study involved professional players in the first division of a men’s volleyball team. The main aim of this paper was to show the correlation between lower limb power, movement, position on the pitch, and volleyball efficiency. Methods: The authors presented basic somatic characteristics of anthropology metrics, advanced research results obtained with the Optojump measuring system, players’ results statistics obtained with the Data Volley software (v.4.2024.01), and by applying scientific statistical methods and graphics interpretation. Results: The analyses with the Kruskal-Wallis test showed statistically significant differences in all the variables studied. Flight time (TF, H = 122.967; p < 0.001.) was the shortest for the players in the libero position, then the setter, attacker, middle and longest flight time was shown by the receiving players. The height of the jumps (JH, H = 123.018; p < 0.001) was the highest for receiving and mid players, and then attackers and setters, and the lowest jumps were recorded by libero players. The highest power of lower limbs (P, H = 20.392; p < 0.001) was characteristic for receivers, setters, mid-players and, curiously, libero achieved a better result than attackers. Conclusions: The analysis shows that the power of the lower extremities in volleyball players is associated with the function they perform on the court.

The global distribution of Aedes aegypti mosquitoes, particularly in tropical regions, poses a significant public health risk due to their apparent ability to transmit arboviruses such as West Nile virus (WNV). This study aimed to evaluate the vector competence of Ae. aegypti from São Tomé and Príncipe (STP) for the transmission of the WNV PT6.39 strain, considering its potential role as a bridge vector in a region where Culex quinquefasciatus would be the main vector. Aedes aegypti mosquitoes were collected, reared, and experimentally infected with WNV, with viral dissemination and transmission potential assessed 7, 14, and 21 days post infection (dpi). The results showed an increasing trend in infection rates, from 5% at 7 dpi to 35% at 21 dpi, with corresponding dissemination rates of 0%, 100%, and 43%. The transmission rates also increased from 0% at 7 dpi to 67% at 21 dpi, with a maximum transmission efficiency of 10% observed at the final time point. Although Ae. aegypti from STP demonstrated the potential to transmit WNV, the overall transmission efficiency remained relatively low. These findings provide necessary insights into the vector competence of Ae. aegypti in this region, highlighting the importance of continued monitoring and targeted vector control measures to mitigate the risk of potential WNV outbreaks.

Cracked and deteriorated asphalt are common problems on our roads, leading to safety concerns and requiring significant resources for rehabilitation and reconstruction. This study investigates bio-fog seals, a promising eco-friendly solution utilizing bio-based rejuvenators. These treatments penetrate aged asphalt, restoring its flexibility and resistance to cracking. We assessed the effectiveness of two bio-fog seal formulations—one containing sub-epoxidized soybean oil (SESO) and the other combining SESO with a biopolymer (BioMag). Applied to real pavement sections, the research evaluated how these bio-seals impacted key performance factors, such as stiffness, permeability, and drying time, and safety factors, including skid resistance and pavement marking visibility. The results indicate the bio-seals did not compromise skid resistance and the reflectivity of the markings, eliminating the need for repainting stripes. Additionally, they successfully reduced pavement stiffness, making the asphalt more flexible and crack-resistant. Remarkably, with rapid setting times, under 30 min, these treatments minimize traffic disruption and do not require a blotter material. Overall, this research demonstrates the potential of bio-fog seals as a sustainable solution for extending pavement lifespan and lowering long-term maintenance costs.

This paper presents a novel parallel manipulator capable of generating two translations (2T), inside a vertical plane, and two rotations (2R), about horizontal axes, which are required in aerospace, manufacturing and rehabilitation fields. These four degrees of freedom are reached by means of a unique RRU and three RSS kinematic chains connected to a rhomboid-shaped mobile platform. The kinematic analysis of the new manipulator is provided, which includes the resolution of the inverse position problem and the velocity equations relating to input and output variables. Additionally, a methodology is proposed for obtaining the workspace free of singularities, collisions and kinematic joint range limitation. This systematic methodology allows designers to identify the critical factors affecting the workspace and, thus, to rearrange the mechanical design accordingly for optimum path planning. We represent the workspace using its two-dimensional subspaces (i.e., translational and rotational workspace). The results are analyzed for different working modes of the manipulator to see its potential use in applications wherein 2T2R motion is necessary.

The competitiveness of tourist destinations is influenced by their relative attractiveness, which will play an essential role in their global success; gastronomy is one of the main motivations that lead tourists to visit a given destination. This research aims to investigate gastronomy’s role in the destination’s competitiveness and image through the analysis of online reputation, both in terms of ratings and sentiments provided by the experience, through the creation of an index of the online reputation of gastronomic image. Online restaurant reviews retrieved from TripAdvisor, from restaurants belonging to eight tourism destination regions, considered the competitive set to the Algarve, are analysed by applying sentiment analysis algorithms. With regard to the Algarve region, it was concluded that the Costa del Sol and the Tropical coast were the most competitive regions in terms of gastronomic image, where the inclusion of seafood products in meals was one of the strategic aspects used to increase the competitiveness of this region. These results can help restaurant managers and destination management organisations to better understand the different customer needs and how to increase their competitiveness.

Introduction: Metastatic prostate cancer (PCa) presents a significant challenge in oncology due to its high mortality rate and the absence of effective biomarkers for predicting patient outcomes. Building on previous research that highlighted the critical role of the long noncoding RNA (lncRNA) H19 and cell adhesion molecules in promoting tumor progression under hypoxia and estrogen stimulation, this study aimed to assess the potential of these components as prognostic biomarkers for PCa at the biopsy stage. Methods: This research utilized immunohistochemistry and droplet digital PCR to analyze formalin-fixed paraffin-embedded (FFPE) biopsies, focusing on specific markers within the H19/cell adhesion molecules pathway. Results: A novel multivariate analysis led to a “BioScore”, a composite biomarker score to predict disease progression. This score is based on evaluating five key markers: the expression levels of Hypoxia-Inducible Factor 2 Alpha (HIF-2α), endothelial Nitric Oxide Synthase (eNOS), β4 integrin, E-cadherin transcript (CDH1), and lncRNA H19. The criteria for the “BioScore” involve identifying three out of these five markers, combining elevated levels of HIF-2α, eNOS, β4 integrin, and CDH1 with reduced H19 expression. Conclusions: This finding suggests the possibility of identifying, at the time of biopsy, PCa patients at higher risk of metastasis based on dysregulation in the H19/cell adhesion molecules circuitry. This study provides a valuable opportunity for early intervention in managing PCa, potentially contributing to personalized treatment strategies.

In this paper, we present an analysis of the effectiveness of various machine learning algorithms in classifying astronomical objects using data from the third release (DR3) of the Gaia space mission. The dataset used includes spectral information from the satellite’s red and blue spectrophotometers. The primary goal is to achieve reliable classification with high confidence for symbiotic stars, planetary nebulae, and red giants. Symbiotic stars are binary systems formed by a high-temperature star (a white dwarf in most cases) and an evolved star (Mira type or red giant star); their spectra varies between the typical for these objects (depending on the orbital phase of the object) and present emission lines similar to those observed in PN spectra, which is the reason for this first selection. Several classification algorithms are evaluated, including Random Forest (RF), Support Vector Machine (SVM), Artificial Neural Networks (ANN), Gradient Boosting (GB), and Naive Bayes classifier. The evaluation is based on different metrics such as Precision, Recall, F1-Score, and the Kappa index. The study confirms the effectiveness of classifying the mentioned stars using only their spectral information. The models trained with Artificial Neural Networks and Random Forest demonstrated superior performance, surpassing an accuracy rate of 94.67%.

Background: Pickleball is an exponentially growing sport with a lack of notation-based studies. Consequently, this research aimed to conduct a match analysis in men’s singles to enhance the understanding of the game and optimize training practices. Methods: Using observational methodology, a total of 1145 points were analyzed from the semifinal and final rounds of five Professional Pickleball Association Tour tournaments. Data were recorded with LINCE PLUS V.2.1.0 software using the OI-PICKLEBALL-S23 observational instrument. Descriptive statistical analyses were conducted with IBM-SPSS version 25.0, and gameplay patterns were detected using Theme 6.0 Edu. Statistical significance was set at p < 0.05. Results: The data indicated that service faults at the start of the game were minimal (2.4%). The server won fewer points than the returner in the overall set of analyzed points (46.6%). Most points were concluded in short rallies (1–4 shots; 43%) or medium-length rallies (5–8 shots; 44%), with the final shot predominantly occurring from striking zone 2, the area closest to the non-volley line (50.7%). Ground strokes (55.1%) and volleys (38.4%) were the most common final shots. Conclusions: The insights gained from this study can benefit high-performance players and coaches and provide a foundation for future notation-based research in pickleball.

Endophytic fungi are promising producers of phenolic compounds; however, the process of extracting these molecules from the culture medium is a crucial step for obtaining these metabolites. In this context, the best conditions for extracting phenolic compounds produced by Cophinforma mamane CF2-13, isolated from an Amazonian plant, were evaluated. The fungus was cultivated in a liquid medium to obtain its metabolites. The effects of the solvent, time, and temperature were investigated, as well as the use of ultrasound. The total phenolics and the antioxidant activity were assessed to define the best extraction conditions. The optimized extract was chemically characterized and used to analyze its potential in protecting olive oil from lipid oxidation. The best results were found using ethyl acetate at 20 °C for 2 h. These conditions led to 61.61 mg GAEq/g of extract of total phenolics, and an antioxidant capacity of 0.47, 0.45, and 0.29 mmol TEq/g of extract using the DPPH, ABTS, and FRAP methods, respectively. In addition, the optimized extract presented caffeic acid and was able to delay the lipid oxidation of olive oil. The potential of endophytic fungi to produce phenolic compounds is promising for the food industry since natural bioactive compounds have many benefits for human health.

Marine protected areas (MPAs) are widely promoted as effective tools for conserving biodiversity and safeguarding ecosystem services. However, MPA success can be hindered by a lack of legitimacy and low social support because of perceived negative effects on ecosystem services and human well-being. Despite these social challenges, the social dimensions of marine conservation, such as the effects of MPAs on coastal livelihoods and local communities’ perceptions of these effects, are often overlooked in conservation initiatives. In this study, we use a mixed methods approach, combining qualitative thematic analysis and quantitative network analysis derived from interviews and focus group discussions, to examine the perceptions of key stakeholder groups about the Litoral Norte MPA in Portugal. Our findings reveal that most stakeholder groups hold more negative than positive views about the governance and management of the MPA. Key concerns include unsatisfactory participation in MPA decisions and perceptions that the MPA fails to deliver positive social and ecological outcomes, such as increased community involvement, fair income distribution, and enhanced fish abundance. Policy makers and managers need to address these negative perceptions to improve conservation governance and management. By considering the stakeholder feedback presented in this study, such as fostering better engagement with the local community and transforming conflicts into opportunities for co-developing new conservation actions with local resource users, policy makers and managers can increase support for Litoral Norte and enhance the social and ecological outcomes of the MPA.

Light quality is a key factor affecting algal growth and biomass composition, particularly pigments such as carotenoids, known for their antioxidant properties. Light-emitting diodes (LEDs) are becoming a cost-effective solution for indoor seaweed production when compared to fluorescent bulbs, allowing full control of the light spectra. However, knowledge of its effects on Ulva biomass production is still scarce. In this study, we investigated the effects of LEDs on the phenotype of an Ulva lacinulata strain, collected on the Northern Portuguese coast. Effects of white (W), green (G), red (R), and blue (B) LEDs were evaluated for growth (fresh weight and area), photosynthetic activity, sporulation, and content of pigments and antioxidant compounds. The results showed that there were no significant differences in terms of fresh weight accumulation and reduced sporulation among the tested LEDs, while W light induced the highest expansion rate. Under G, U. lacinulata attained a quicker photoacclimation, and the highest content of pigments and total antioxidant activity; but with R and W, antioxidant compounds against the specific radicals O2•− and •NO were produced in a higher content when compared to other LEDs. Altogether, this study demonstrated that it is possible to modulate the bioactive properties of U. lacinulata by using W, R, and G light, opening the path to the production of biomass tailored for specific nutraceutical applications.

The current scenario of antifouling (AF) strategies to prevent the natural process of marine biofouling is based in the use of antifouling paints containing different active ingredients, believed to be harmful to the marine environment. Compounds called booster biocides are being used with copper as an alternative to the traditionally used tributyltin (TBT); however, some of them were recently found to accumulate in coastal waters at levels that are deleterious for marine organisms. More ecological alternatives were pursued, some of them based on the marine organism mechanisms’ production of specialized metabolites with AF activity. However, despite the investment in research on AF natural products and their synthetic analogues, many studies showed that natural AF alternatives do not perform as well as the traditional metal-based ones. In the search for AF agents with better performance and to understand which molecular motifs were responsible for the AF activity of natural compounds, synthetic analogues were produced and investigated for structure–AF activity relationship studies. This review is a comprehensive compilation of AF compounds synthesized in the last two decades with highlights on the data concerning their structure–activity relationship, providing a chemical toolbox for researchers to develop efficient nature-inspired AF agents.

During the desalting of salted cod, significant textural, histological, and biochemical changes occur. Understanding these changes is crucial for enhancing the preservation and extending the shelf life of desalted cod. This study aimed to investigate the physicochemical quality parameters and enzymatic activities during the desalting process of cod (16 h at 4 and 20 °C) and to extend the shelf life of desalted cod through high-pressure processing (HPP) at 400 and 550 MPa for 5 min. During desalting, a correlation was noted between the pH and trimethylamine content in samples desalted at 20 °C, with both parameters increasing in the initial 4 h and stabilizing thereafter. The soluble protein in cod muscle decreased over desalting time, as it dissolved into the desalting water. Enzymatic activity showed a decline in cathepsins (B and D) and acid phosphatase throughout desalting, whereas lipase activity increased, particularly at 20 °C. HPP effectively extended the shelf life of desalted cod by controlling endogenous microbial growth, enabling an extension to 14–21 days compared to the 7 days observed in untreated control samples. This study highlights quality changes during desalting, with lesser effects at lower temperatures. Subsequent HPP improved the microbiological quality of desalted cod during refrigerated storage.

Cardiac ablation (CA) is an interventional electrophysiological procedure used to disrupt arrhythmic substrates in the myocardium by inducing localized scarring. Current CA training relies on the master–apprentice model. In different fields of medicine including CA, virtual and physical simulators have proven to enhance, and even outperform, conventional training modalities while providing a risk-free learning environment. Despite the benefits, high costs and operational difficulties limit the widespread use of interventional simulators. Our previous research introduced a low-cost CA simulator using a 3D-printed biatrial cardiac model, successfully recording ten ablation lesions on the phantom myocardium. In this work, we present and evaluate an enhanced version: compared to the previous version, the cardiac phantom’s electrical behavior and ablation settings were optimized to produce consistent lesions, while 3D-printed components improved the haptic and radiographic properties of the simulator. Seven cardiologists compared the experimental simulator’s performance to the leading commercial system from Heartroid in a 24-question survey on a 5-point Likert scale. The four following areas of fidelity were considered: catheter entry, anatomical correctness, radiographic appearance, and mapping and ablation. The experimental simulator significantly outperformed the commercial system (p < 0.01), particularly in radiographic appearance (p < 0.01). The results show the potential for the experimental simulator in routine CA training.

Metallic foams exhibit unique properties that make them suitable for diverse engineering applications. Accurate mechanical characterization is essential for assessing their performance under both monotonic and cyclic loading conditions. However, despite the advancements, the understanding of cyclic load responses in metallic foams has been limited. This study aims to propose a mesostructural model to assess the fatigue behavior of open-cell metal foams subjected to cyclic loading conditions. The proposed model considers the previous load history and is based on the analogy of progressive collapse, integrating a finite element model, a fatigue analysis model, an equivalent number of cycles model, and a failure criterion model. Validation against experimental data shows that the proposed model can reliably predict the fatigue life of the metallic foams for specific strain amplitudes.

Inula viscosa is a widely used plant in traditional Mediterranean and Middle Eastern medicine for various illnesses. I. viscosa has been shown to have anticancer effects against various cancers, but its effects against lung cancer have been under limited investigation. At the same time, I. viscosa is rich in terpenoids whose anti-lung cancer effects have been poorly investigated. This study aimed to examine the potential anticancer properties of methanolic and aqueous extracts of stems and leaves of I. viscosa and its terpenoid-rich fraction against human lung cancer A549 cells. Results showed that the methanolic extracts of I. viscosa had significantly higher polyphenol and flavonoid content and radical scavenging capacity than the aqueous extracts. In addition, leaves methanolic extracts (IVLM) caused the highest reduction in viability of A549 cells among all the extracts. IVLM also reduced the viability of human ovarian SK-OV-3, breast MCF-7, liver HepG2, and colorectal HCT116 cancer cells. A terpenoid-rich I. viscosa fraction (IVL DCM), prepared by liquid-liquid separation of IVLM in dichloromethane (DCM), displayed a substantial reduction in the viability of A549 cells (IC50 = 27.8 ± 1.5 µg/mL at 48 h) and the panel of tested cancerous cell lines but was not cytotoxic to normal human embryonic fibroblasts (HDFn). The assessment of IVL DCM phytochemical constituents using GC-MS analysis revealed 21 metabolites, highlighting an enrichment in terpenoids, such as lupeol and its derivatives, caryophyllene oxide, betulin, and isopulegol, known to exhibit proapoptotic and antimetastatic functions. IVL DCM also showed robust antioxidant capacity and decent polyphenol and flavonoid contents. Furthermore, Western blotting analysis indicated that IVL DCM reduced proliferation (reduction of proliferation marker Ki67 and induction of proliferation inhibitor proteins P21 and P27), contaminant with P38 MAP kinase activation, and induced the intrinsic apoptotic pathway (P53/BCL2/BAX/Caspase3/PARP) in A549 cells. IVL DCM also reduced the migration of A549 cells, potentially by reducing FAK activation. Future identification of anticancer metabolites of IVL DCM, especially terpenoids, is recommended. These data place I. viscosa as a new resource of herbal anticancer agents.

Background: Traumatic brain injury (TBI) is a major cause of death and disability, with a rising incidence in recent years. Factors such as age, sex, hypotension, low score on the Glasgow Coma Scale, use of invasive mechanical ventilation and vasopressors, etc., have been associated with mortality caused by TBI. The aim of this study was to identify the clinical and sociodemographic characteristics that influence the mortality or survival of patients with TBI in a tertiary care hospital in Mexico. Methods: A sample of 94 patients aged 18 years or older, from both sexes, with an admitting diagnosis of mild-to-severe head trauma, with initial prehospital treatment, was taken. Data were extracted from the Single Registry of Patients with TBI at the Ixtapaluca Regional High Specialty Hospital (HRAEI). Normality tests were used to decide on the corresponding statistical analysis. Results: No factors associated with mortality were found; however, survival analysis showed that the presence of seizures, aggregate limb trauma, and subjects with diabetes mellitus, heart disease or patients with four concomitant comorbidities had 100% mortality. In addition, having seizures in the prehospital setting increased the risk of mortality four times. Although they did not have a direct association with mortality, they significantly decreased survival. A larger sample size is probably required to obtain an association with mortality. Conclusions: These results reflect the severity of the clinical situation in this population and, although no risk factors were identified, they enlighten us about the conditions presented by patients who died.

Increasing economic and environmental concerns arising from the extensive exploration and dependence on fossil fuel-based materials have encouraged the search for eco-friendly alternatives. Fibers based on biomass-derived materials have been attracting growing interest. Among other features, the mechanical performance of bio-based fibers needs to be improved to effectively compete with their counterparts and emerge as viable substitutes. This review presents scientific advancements in the development of naturally derived fibers, and strategies for their production with tailored mechanical properties. The potential of natural precursor-based fibers for their conversion into high-performance carbon fibers is also emphasized. Studies reporting the mechanical properties of bio-based fibers developed by wet spinning are identified, analyzed, and discussed. These studies show that cellulose is the most studied material, while Ioncell technology is identified as the most suitable method for producing cellulose-based fibers with the highest tensile strength. Studies have also demonstrated that silk fibroin exhibits tensile strength and elongation at break ranging from 300 to 600 MPa and 30 to 50%. Although several novel processes have been explored, there are still challenges that need to be addressed for bio-based fibers to become feasible options, and to boost their usage across industries.

We developed a protein to rapidly and accurately diagnose Chagas disease, a life-threatening illness identified by the WHO as a critical worldwide public health risk. Limitations in present day serological tests are complicating the current health situation and contributing to most infected persons being unaware of their condition and therefore untreated. To improve diagnostic testing, we developed an immunological mimic of the etiological agent, Trypanosoma cruzi, by combining ten pathogen-specific epitopes within the beta-barrel protein structure of Thermal Green Protein. The resulting multi-epitope protein, DxCruziV3, displayed high specificity and sensitivity as the antibody capture reagent in an ELISA platform with an analytical sensitivity that exceeds WHO recommendations. Within an immunochromatographic platform, DxCruziV3 showed excellent performance for the point of application diagnosis in a region endemic for multiple diseases, the municipality of Barcelos in the state of Amazonas, Brazil. In total, 167 individuals were rapidly tested using whole blood from a finger stick. As recommended by the Brazilian Ministry of Health, venous blood samples were laboratory tested by conventional assays for comparison. Test results suggest utilizing DxCruziV3 in different assay platforms can confidently diagnose chronic infections by T. cruzi. Rapid and more accurate results will benefit everyone but will have the most noticeable impact in resource-limited rural areas where the disease is endemic.
