Due to the increase in consumption and the possibilities of exportation and industrialization, blueberry cultivation has been expanding in Brazil. Since Rabbiteye is one of the more cultivated groups of blueberries in Brazil, this study aimed to assess the productive characterization of the blueberry cultivar Bluegem, group Rabbiteye. Assessments were conducted with plants in full production during two harvest seasons. We collected data on the following variables: flower and vegetative bud distribution, open flowers and formed fruit in two different shoot lengths, and number of floral primordia. The results showed a greater number of vegetative buds and the intercalation between flower and vegetative buds in a proportion of 78% and 48% in long and short shoots, respectively. The number of total buds and the number of flower and vegetative buds were influenced by the harvest season and the shoot length. The number of floral primordia was influenced by the bud position on the shoot, but not by the harvest season. The number of flowers and fruits, in general, was greater in apical buds and in long shoots.

Spectral remote sensing and proximal sensors are important tools for managing the plant-pathogen relationship. The lack of experimental planning and the probability of error in agricultural studies may result in work repetition and, consequently, in financial expenses and costs with human resources. To reduce such problems, determining the optimum size of the experimental plot for treatments is one of the adopted methods. The objective of this study was to estimate the optimum plot size for reflectance in soybeans that were treated with different fungicide levels according to the methods of modified maximum curvature and maximum distance. Reflectance readings were carried out for the soybean crop with a radiometer GreenSeeker®, considering basic units of 0.45 m² in an area of ten rows, 10 m long, for each treatment. Treatments were applied to create a gradient of Asian soybean rust, varying the number of fungicide applications. Data were collected in two phenological stages (R5.5 and R6), obtaining 300 simulations of experimental area for each stage. Based on the results, the use of 5.40 m² plots with a group of three rows, 4 m long, is recommended. RESUMO O sensoriamento remoto espectral e o sensor proximal são ferramentas importantes para gerenciar a relação planta-patógeno. A falta de planejamento experimental e a probabilidade de erro em estudos agrícolas podem resultar em retrabalho e, consequentemente, despesas financeiras e de recursos humanos. Uma maneira de reduzir esse problema é determinar o tamanho ótimo de parcela experimental para realização dos tratamentos. O objetivo deste estudo foi estimar o tamanho ótimo de parcela para refletância em soja que foi tratada com diferentes doses de fungicida, usando os métodos de curvatura máxima modificada e distância máxima. As leituras de refletância foram realizadas na cultura de soja com o auxílio de um radiômetro GreenSeeker®, com unidades básicas de 0,45 m², em uma área de dez linhas, com 10 metros de comprimento, em cada tratamento. Os tratamentos foram aplicados para criar um gradiente da doença ferrugem asiática da soja, variando o número de aplicações de fungicidas. Os dados foram coletados em dois estágios fenológicos (R5.5 e R6), obtendo-se 300 simulações de áreas experimentais em cada estágio. Com base nos resultados, recomenda-se o uso de parcelas de 5,40 m², com um grupo de três linhas, com 4 m de comprimento.

The influence of NGF in male reproduction in some animal species and humans has already been assessed. Many of these effects are mediated by the distribution and abundance of tropomyosin receptor kinase A (TrKA) and p75 neurotrophin (p75NTR) receptors on sperm cells. The aim of this research was to investigate the role of NGF and its receptors, TrKA and p75NTR, in rabbit sperm outcomes during in vitro storage. Major semen traits (kinetic parameters, apoptotic, necrotic and live sperm) were recorded in rabbit semen samples from 0 to 12 h of storage (every 4 h). Three experimental hypotheses were formulated: i) sperm storage changes NGF receptor abundance in rabbit sperm; ii) TrKA and p75NTR differently modulate NGF signalling (assessed by the neutralisation of receptors); iii) NGF-receptor interactions show different responses during storage (evaluated by the addition of exogenous NGF). The results demonstrate that: (i) the receptor number changed in a time-dependent manner with a significant increase in p75NTR after 8–12 h of storage; ii) the neutralisation of NGF receptors largely affected VCL, apoptotic, necrotic and live cells during sperm storage, i.e. blockade of TrKA significantly increased speed, capacitation, necrosis and apoptosis, whereas blockade of p75NTR improved motility and live cells; iii) the addition of exogenous human NGF (100 ng/mL) at different time points of storage (0, 4, 8 h) differently influenced sperm traits i.e. NGF addition at time 0 positively affected all the pro-vital traits (kinetic, live cells) whereas, after 4–8 h, the effect of NGF was null or negative. In conclusion, NGF affects kinetic and other physiological traits (capacitation, apoptosis and necrosis) of rabbit sperm in a time-dependent manner. Most of these modifications are modulated by the receptors involved (TrKA or p75NTR), which changed considerably during sperm storage (increase of p75NTR).

The nerve growth factor (NGF), a member of the neurotrophins family, plays an important role in the nervous system but also in the reproductive apparatus and in sperm traits. The aim of this review is to summarize the effect of NGF on sperm functions of mammals. NGF was detected in seminal plasma of many animal species and its receptors, TrkA and p75NTR, are present in both epididymal and ejaculated sperm. The NGF, either endogenous or exogenous, may affects the kinetics and other sperm traits (e.g. capacitation, apoptosis, necrosis and acrosome reaction) and these effects appear modulated by the different receptors involved (TrkA or p75NTR). Although the animal species can affect the localization of receptors, TrkA seems mainly to be confined in the head and p75NTR in the midpiece and tail. Considering that some seminal disorders and sperm traits have been correlated with a lower NGF concentration, this review provides recent insights on the role of NGF on sperm cells and the possible mechanism implicated.

Summary Salmonella is the second main cause of foodborne illness in poultry production. It is one of the most problematic zoonoses in terms of public health worldwide because of the difficulty in controlling it and its significant morbidity and mortality rates. Recent surveys have shown that small flocks of laying hens have the same or higher prevalence of salmonellosis than larger flocks, mainly due to a lack of control actions, that is, the control of mice and wild animals, employees, and poor management practices. In this regard, different physical and chemical procedures have proven efficacious for reducing external and/or internal Salmonella contamination. This research evaluated the effect of ozone and UV-C rays on Salmonella growth and hen egg quality. Microbiological evaluation was performed on 120 eggs: negative control (C−), eggs not contaminated with Salmonella; positive control (C+), eggs contaminated; contaminated ozonate-treated (O, 600 mg/h for 2 h); and contaminated UV-C–irradiated (ʎ, 254 nm for 15 s) eggs. Moreover, 30 eggs were used (10/group) for the quality assessments of the C, O, and UV-C groups. A 2 log10 CFU/g reduction in Salmonella on contaminated eggs was found in the UV-C group compared with the C+ and O groups. Compared with UV-C treatment, ozonization reduced the amount of yolk tocols and carotenoids (by 2 times). The lipid oxidative status decreased (−1.5 times), similar to the cholesterol level (−28.5%), whereas the amount of cholesterol and its oxidized products increased (+82.1%) in the O group compared with the C group. UV-C irradiation is an effective strategy to reduce Salmonella contamination in eggs without negatively affecting the quality. Therefore, UV treatments remain among the more promising procedures.

Glyphosate is the most commonly used herbicide worldwide. Its improper use during recent decades has resulted in glyphosate contamination of soils and waters. Fungal bioremediation is an environmentally friendly, cost effective, and feasible solution to glyphosate contamination in soils. In this study, several saprotrophic fungi isolated from agricultural environments were screened for their ability to tolerate and utilise Roundup in different cultural conditions as a nutritional source. Purpureocillium lilacinum was further screened to evaluate the ability to break down and utilise glyphosate as a P source in a liquid medium. The dose–response effect for Roundup, and the difference in toxicity between pure glyphosate and Roundup were also studied. This study reports the ability of several strains to tolerate 1 mM and 10 mM Roundup and to utilise it as nutritional source. P. lilacinum was reported for the first time for its ability to degrade glyphosate to a considerable extent (80%) and to utilise it as a P source, without showing dose-dependent negative effects on growth. Pure glyphosate was found to be more toxic than Roundup for P. lilacinum. Our results showed that pure glyphosate toxicity can be only partially addressed by the pH decrease determined in the culture medium. In conclusion, our study emphasises the noteworthy potential of P. lilacinum in glyphosate degradation.

Increased tolerance to pathogens is an important goal in conventional and biotechnology-assisted grapevine breeding programs worldwide. Fungal and viral pathogens cause direct losses in berry production, but also affect the quality of the final products. Precision breeding strategies allow the introduction of resistance characters in elite cultivars, although the factors determining the plant?s overall performance are not fully characterized. Grapevine plants expressing defense proteins, from fungal or plant origins, or of the coat protein gene of grapevine leafroll-associated virus 3 (GLRaV-3) were generated by Agrobacterium -mediated transformation of somatic embryos and shoot apical meristems. The responses of the transformed lines to pathogen challenges were investigated by biochemical, phytopathological and molecular methods. The expression of a Metarhizium anisopliae chitinase gene delayed pathogenesis and disease progression against the necrotrophic pathogen Botrytis cinerea. Modified lines expressing a Solanum nigrum osmotin-like protein also exhibited slower disease progression, but to a smaller extent. Grapevine lines carrying two hairpin- inducing constructs had lower GLRaV-3 titers when challenged by grafting, although disease symptoms and viral multiplication were detected. The levels of global genome methylation were determined for the genetically engineered lines, and correlation analyses demonstrated the association between higher levels of methylated DNA and larger portions of virus-derived sequences. Resistance expression was also negatively correlated with the contents of introduced viral sequences and genome methylation, indicating that the effectiveness of resistance strategies employing sequences of viral origin is subject to epigenetic regulation in grapevine. Cucumber (Cucumis sativus Linnaeus, 1753) stands out in the Brazilian agribusiness of vegetables as it can be consumed in salads (in natura or pickled) or even used for the production of cosmetics and drugs. During cucumber cultivation, several insectpests, mainly from the Orders Hemiptera, Thysanoptera, Lepidoptera, can cause significant yield losses. Therefore, this study aimed to evaluate the entomofaunistic diversity of arthropods in cucumber crop using Moericke and Pitfall traps. Cucumber seedlings were commercially obtained and grown up in the experimental area of the Federal University of Fronteira Sul – Campus Chapecó (SC). Three samplings were carried out between October and December, 2015. The sampled specimens were identified to the taxon of Order and placed in vials containing alcohol solution (70%). A total of 1.651 specimens were identified and classified into 11 different Orders. Pitfall traps sampled a greater number of orders (9) and specimens (1.309), with emphasis to the Order Hymenoptera (928). For the aerial traps (Moericke), 342 specimens, allocated in eight different Orders, were identified, with the Order Diptera as the most abundant one (153 individuals). Moericke and Pitfall traps showed a Shannon-Weaver diversity index of 1.53 and 1.02, respectively.

This study aimed to assess the effect of rearing system (intensive vs. free-range under vineyard) on the qualitative characteristics of goose meat. In particular, the integration of a goose raising system with an organic grape production was evaluated. Six hundred Romagnola geese of both sexes were divided into two groups at 21 d of age: Vineyard group (V) – 480 geese (120 geese/ha) pasturing in 4 ha of vineyard, and Control Group (C) – 120 geese of the same genotype in the experimental farm of Perugia, without any access to the pasture. Live weight, feed consumption and other performance were registered weekly on 30 selected and marked animals/group. At 160 days of age, 15 geese/group were slaughtered and characteristics of carcase, breast and drumstick (physical, chemical, oxidative status, fatty acids profile) were evaluated. The chemical analysis of diet and pasture and the ingestion of crude protein, digestible energy and bioactive compounds were also estimated. Vineyard geese, showed lower productive performance than the C ones (live and carcase weight), however, due to pasture availability, the intake of bioactive compounds (vitamin E, retinol, n-3 long-chain fatty acids) was higher and positively affected the antioxidant content of breast and drumstick. The higher kinetic activity of V geese reduced the fat amount of carcase and meat, whereas increased the development of drumstick muscle (higher meat/bone ratio) and worsened the oxidative status of meat. Concluding, the free-range vineyard geese resulted in a positive payoff on the geese meat quality viewpoint.Highlights Pasture enhance the amount of VIT E in goose muscle. Tenebrio molitor (T. molitor) (mealworm) larvae are one of the most promising insects for feed–food purposes. Mealworms are rich in several macro and micro nutritional elements and can be practically reared on side stream substrates. In this study, the effects of seven different cooking techniques were tested on the nutritional value of mealworms focusing the attention on protein digestibility, fatty acid (FA) profile, and oxidative status. Uncooked larvae (UC) were used as control and compared to two combinations of temperature/time in oven cooking (70°C for 30 min, OC70-30, 150°C for 10 min, OC150-10), two methods of frying (mealworms fried in sunflower oil as deep fry, DF, or pan fry, PF), microwaving (MW), boiling (in plastic bag under vacuum, BO), and steaming (ST). Proximate composition, in vitro digestibility (gastric and duodenal), FA profile, and oxidative status (tocopherol and tocotrienol, carbonyl, and lipid oxidation) were then tested. Cooking technique affected all the tested parameters. As expected, cooking affected proximate composition in relation to the method applied (dry matter increased after oven cooking and frying; lipids increased by frying). In vitro digestion revealed the highest value for the OC70-30 method, followed by UC and ST. Deep frying revealed the worst digestibility percentage. FA profile was deeply affected by the cooking technique, with general decrease in SFA and MUFA. The highest modifications in FA profile were revealed in ST larvae with an increased percentage of linoleic acid linked to the lowering of SFA and MUFA contents. Furthermore, deep frying larvae in sunflower oil increased the relative abundance of PUFAs. Tocols values were higher in DF and MW groups than PF (about 6-fold more) and all other groups (7-fold more). Carbonyls increased with oven cooking (OC150-10 and OC70-30), whereas the values were lower with frying and similar to ST and UC. Lipid oxidation was highest as well in OC150-10 but similar to frying methods (DF and PF). Based on the obtained results, it can be concluded that mealworm larvae surely meet human nutritional requirements, but the cooking method must be carefully chosen to maintain a high nutritional value.

A oliveira (Olea europaea L.) é composta por folhas que podem ser consumidas, mas costumam ser descartadas pela colheita mecanizada. Uma alternativa para viabilizar o descarte das folhas de oliveiras, é elaborar a farinha de folha de oliveira (FFO) realizando o aproveitamento. Com isso, o objetivo desta pesquisa foi elaborar um pão de queijo vegetariano adicionado de farinha da folha de oliveira a fim de viabilizar uma nova alternativa de aproveitamento tecnológico desse resíduo. Foram desenvolvidas cinco formulações de pães de queijo, alterando-se a quantidade de farinha das folhas de oliveira adicionada (0,95 – 2,85%) e submetidas a avaliação das características sensoriais pelo método de Análise Descritiva Quantitativa. Os resultados demonstraram que, dentre os quinze atributos avaliados, os sabores herbáceo, amargo e de queijo juntamente com o aroma herbáceo apresentaram diferença significativa. Entre os atributos analisados, a FFO contribuiu para o maior sabor amargo dos pães de queijo, seguido de sabor e aroma herbáceo. Conclui-se que foi possível elaborar um pão de queijo vegetariano adicionado de farinha de folha de oliveira como uma alternativa de aproveitamento tecnológico desse resíduo.

The present study evaluated the effects of feed supplementation with olive oil by-products on the microbial quality of rabbit meat. Thirty-three New Zealand White rabbits were randomly assigned to three experimental grower–finisher diets. Each dietary group consisted of three experimental treatments: (1) a basal control diet (C), (2) a C diet supplemented with a low dosage of polyphenol (150 mg/kg, L), and (3) a C diet supplemented with a high dose of polyphenols (280 mg/kg, H). Polyphenol analyses in feed and meat were performed using the liquid-chromatography coupled to tandem mass spectrometry technique (LC-MS/MS). Higher amounts of sulphate metabolites were detected in the H group. Microbiological quality was evaluated on Longissimus lumborum muscles stored under aerobic conditions at 4 °C. The H diet exerted an inhibitory effect on microbial growth (p &lt, 0.001), notably for Pseudomonas spp., when compared to C and L diets, differences among the groups were observed starting from 6 days of storage. In the H group, the Pseudomonas spp. population showed an increase in the latency phase and a decrease in the maximum growth rate of the fitted curves in comparison with the C and L groups. The use of dietary polyphenols could be a strategy to reduce spoilage during meat storage.

This study aimed to develop an adaptability score (AS) for chicken strains, which includes behavioral, plumage conditions, and body lesion indicators through a multifactorial approach. A total of 600 male chickens from 6 poultry genotypes—Ranger Classic (R1), Ranger Gold (R2), Rowan Ranger (R3), Hubbard Red JA (A), CY Gen 5 × JA87 (CY), and M22 × JA87 (M)—were reared under organic conditions, fed ad libitum, and individually weighed weekly to calculate the daily weight gain (DWG). The behavioral observations consisted of the explorative attitude (EA), recorded at 21 days, and the behavioral patterns (BPs) recorded the week before the slaughter. The AS was established by a principal component analysis, and the AS of these genotypes was compared. Moreover, the effect of DWG and genotype on the AS was evaluated by univariable and multivariable regression models. Although the DWG and genotype were strictly dependent, genotype was the most important factor affecting the AS. In fact, its effect was significant both in univariable (p &lt, 0.001) and multivariable models (p &lt, 0.001). Conversely, the DWG was significant only in the univariable and lost significance when the effect of genotype was introduced in the model.

The aim of this study was to analyze the relationship between the kinetic behavior, carcass characteristics, oxidative status (blood and meat), and meat fatty acid profiles of 6 organically reared slower growing chicken genotypes (SrG). One hundred male chickens of 6 SrG were used: Ranger Classic (RC), Ranger Gold (RG), Rowan Ranger (RR), RedJA (RJ), CY Gen 5 JA87 (CY), and M22 × JA87 (M). Twenty chickens/genotype were selected to analyze behavior, while, 15 individuals were slaughtered and different traits were analyzed in the blood and drumstick meat. The variables were grouped into different principal components: kinetic activity (PC1, with explorative attitude as the highest score), productive performance (PC2, carcass and head/feet yields), blood (PC3, carbonyls, and TBARS) and meat (PC6, thiols, and TBARS) markers, technological traits (PC4, pH, and color), proximate meat composition (PCA5, moisture, lipids, protein, and ash), fatty acid profile, and nutritional indexes (PC7, IP, and PUFAn-3). Uni- and bivariate analyses showed a strong positive association between kinetic behaviors and blood and meat oxidation and a medium positive association with fatty acid profile and nutritional indexes, whereas a negative association was found between productive performance loads and the technological traits of meat. Generalized linear models showed that all PCs were influenced by genotype. In particular, CY and M resulted as less active genotypes; conversely, RR showed more kinetic activity, whereas RJ, RG, and RC exhibited intermediate levels of activity. Cluster analysis of kinetic behavior and blood or meat oxidative status highlighted 2 groups: nonwalking (NW: CY and M) and walking (W: RC, RG, RR, and RJ) animals. However, in the W group, another was visualized, constituted by genotypes with high kinetic activity resulting in the worst oxidative balance (Walking not trained-genotypes, Wnt: RR and RJ). The present results confirmed that the kinetic behavior of SrG genotypes is negatively correlated with productive performance. Furthermore, a significant association between kinetic behavior and blood (positively correlated) or meat (negatively correlated) oxidative status was noted. Such differences are mainly due to the intrinsic response of the genotypes used (i.e., training-walking capacity).

The transport of auxin controls the rate, direction and localization of plant growth and development. The course of auxin transport is defined by the polar subcellular localization of the PIN proteins, a family of auxin efflux transporters. However, little is known about the composition and regulation of the PIN protein complex. Here, using blue‐native PAGE and quantitative mass spectrometry, we identify native PIN core transport units as homo‐ and heteromers assembled from PIN1, PIN2, PIN3, PIN4 and PIN7 subunits only. Furthermore, we show that endogenous flavonols stabilize PIN dimers to regulate auxin efflux in the same way as does the auxin transport inhibitor 1‐naphthylphthalamic acid (NPA). This inhibitory mechanism is counteracted both by the natural auxin indole‐3‐acetic acid and by phosphomimetic amino acids introduced into the PIN1 cytoplasmic domain. Our results lend mechanistic insights into an endogenous control mechanism which regulates PIN function and opens the way for a deeper understanding of the protein environment and regulation of the polar auxin transport complex., Identification of native PIN‐FORMED (PIN) auxin transporter complexes in Arabidopsis thaliana shows formation of dimers that are stabilized by the auxin transport inhibitor NPA and natural flavonols.

Simple Summary Agroforestry is a practice, which consists of having orchard, crops and animals in the same land. This system shows many sustainability advantages like reductions of land use, permitting to obtain two productions (vegetal and animal) in the same area. Moreover, if the animals are well managed, they exert a double action by fertilizing and weeding the soil. The agroforestry system here studied consists of an organic vineyard where geese at two densities (High Geese Density-HGD and Low Geese Density-LGD) were reared. In the organic vineyard, only Copper (Cu) treatment is allowed, like antifungal. The aim was to investigate the chemical and biochemical properties of the soil with geese and the impact of Cu on the soil and animal tissues. The main results showed that the presence of animals improves the efficiency of the microbial biomass mainly in the upper soil horizons. Moreover, the grazing activity of geese removes Cu from the soil with the grass intake and showed a moderate accumulation in the liver. However, no significant difference was present in the edible tissues (breast and drumstick) of the vineyard geese in respect to the control ones. Abstract Agroforestry systems aim at increasing the productivity and the environmental sustainability of both crop and animal productions. The integration of small animals such as geese in the vineyard could represent an opportunity to improve farm income and reduce land use for grazing. The main objective of this work was to study the impact of geese rearing in an organic vineyard on the chemical and biochemical properties of the soil and the effect of Copper (Cu) supplied with the fungicide treatments. Furthermore, the amount of Cu in the animal tissues was also investigated. Three experimental areas within the vineyard were selected: High Geese Density (HGD-240 geese ha−1), Low Geese Density (LGD-120 geese ha−1) and Without Geese used as control soil (WG). The results indicated that both HGD and LGD did not affect the main chemical properties of the vineyard soils. LGD increased the amount and the efficiency of the microbial biomass in the upper soil horizons. Moreover, geese through the grazing activity reduced the Cu content in the vineyard soils, accumulating this element in their liver. However, the content of Cu in the breast and drumstick of vineyard geese did not show any significant difference in respect the meat of the control ones.

Sitophilus zeamais is one of the more important and severe pests of stored grains. Its control is carried out predominantly by synthetic insecticides, a practice that must be rethought assuming the increasing demands for pesticide-free foods. This work aimed at evaluating the bioactivity of Eucalyptus cv. “Urocam” essential oil on 10- to 30-day-old S. zeamais . Essential oil from the leaves of Eucalyptus cv. “Urocam” was sprayed on corn kernels, infected by S. zeamais, at the rate dosage of 0.0; 1.0; 2.0; 3.0 and 5.0 L t -1 . A completely randomized experimental design with five replicates was used to conduct the experiment. All data were analyzed by the F test and whenever a significant interaction was found, analysis of variance and regression tests were performed (Assistat 7.7 Beta software). The terpene with the highest concentration in the Eucalyptus cv. “Urocam” essential oil was Eucalyptol (74%). Based on our results, it can be inferred that Eucalyptus cv. “Urocam” essential oil presents a good potential of use in the dosages of 3.0 and 5.0 L t -1 of corn kernels due to the high mortality of S. zeamais within the first hours after application, reaching 100% by the end of the first 24 hours.

Producing of extra virgin olive oils (EVOOs) containing pleasant sensory notes depends on fruits quality and production process and is strongly associated with their classification that is based on aroma and sensory taste. Consolidated as an efficient method, the direct headspace solid phase microextraction technique (HS-SPME) was utilized to characterize the volatile organic compounds (VOCs) profile, which contributes to the aroma of olive oils from southwestern (Serra da Mantiqueira region) and southern (Campanha Gaucha region) Brazil. In this work, the relationship between the VOCs and sensory characteristics has been established; 19 EVOO samples (12 from Campanha Gaucha and 7 from Serra da Mantiqueira) were studied. Indeed, the main volatile compounds were analyzed and grouped by their classification as well stood up with the trained sensorial panel perceptions. Relevant correlation between artichoke notes and ripe EVOO and between herbaceous notes and green EVOO was found. Additional correlations were observed for C5 and C6 VOCs with green and fruit/floral notes. The results denote the high quality among the samples and imply that besides the genetic factor, ripe or green classification influenced the volatile composition. PRACTICAL APPLICATION: As the Brazilian olive oil production is increasing, knowing about different sensory characteristics and its correlation with the volatile compounds of extra virgin olive oil represents a good tool to improve the quality. Moreover, the application of direct SPME method was possible evidence in the differentiation of ripe and green olive oils, beyond the production region and in consonance with its sensory notes and characteristics.

In alterative rearing systems, the use of outdoor space has a crucial role. It is well known that only some commercial poultry genotypes are suitable to be reared in these systems. It is necessary to find a balance between productive performance and adaptability. The aim of this study was to evaluate the productive performance, behavior, and welfare status of six poultry genotypes reared in an organic system. One hundred males/genotype (Hubbard RedJA (A), CY5XJA87 (CY), M22XJA87 (M), Ranger Classic (R1), Ranger Gold (R2), and Rowan Ranger (R3)) were reared from 1 to 81 days of age. The number of culled birds was recorded daily, whereas live weight and feed consumption were recorded weekly. Behavior evaluation was undertaken through a computerized system one week before slaughtering, the breast yield and muscle/bone ratio of the drumstick was also evaluated in refrigerated carcasses. The results showed that A and R3 had good adaptability, showing active behaviors and satisfactory productive performance 3083.6 g and 3022.1 g, respectively. Although CY and M achieved the best productive performance, they did not appear adapted to the organic system due to a higher frequency of static behaviors (rest and roost), mortality, footpad dermatitis, breast blisters, and poor feather condition.

Selenium (Se) is an important micronutrient for living organisms, since it is involved in several physiological and metabolic processes. Se intake in humans is often low and very seldom excessive, and its bioavailability depends also on its chemical form, with organic Se as the most available after ingestion. The main dietary source of Se for humans is represented by plants, since many species are able to metabolize and accumulate organic Se in edible parts to be consumed directly (leaves, flowers, fruits, seeds, and sprouts) or after processing (oil, wine, etc.). Countless studies have recently investigated the Se biofortification of plants to produce Se-enriched foods and elicit the production of secondary metabolites, which may benefit human health when incorporated into the diet. Moreover, feeding animals Se-rich diets may provide Se-enriched meat. This work reviews the most recent literature on the nutraceutical profile of Se-enriched foods from plant and animal sources. Composting is a technique used to obtain a nutrient-rich, plant-like organic compost. Considering that there are few studies that shows the efficiency of these composts in the cultivation of vegetables, the objective of this study was to evaluate, in the cultivation of lettuce, the quality of the final compost obtained from tree pruning, dairy sludge and coffee grounds—compost 1; grass, dairy sludge and coffee grounds—compost 2; grass, dairy sludge, coffee grounds and boiler ash—compost 3. Lettuce (Lactuca Sativa L.) were planted for the study with the three composts and a control object-without compost, and a portion with NPK fertilizer. The evaluated parameters were plant growth, head diameter, leaf number, dry mass and fresh mass. The results shows statistical similarities between the organic fertilizers and NPK fertilizer in the evaluated parameters and the control plot presented inferior results to the other plots, emphasizing the need for fertilization at each planting.

Summary The genetic transformation of plant cells is critically dependent on the availability of efficient selectable marker gene. Sulfonamides are herbicides that, by inhibiting the folic acid biosynthetic pathway, suppress the growth of untransformed cells. Sulfonamide resistance genes that were previously developed as selectable markers for plant transformation were based on the assumption that, in plants, the folic acid biosynthetic pathway resides in the chloroplast compartment. Consequently, the Sul resistance protein, a herbicide‐insensitive dihydropteroate synthase, was targeted to the chloroplast. Although these vectors produce transgenic plants, the transformation efficiencies are low compared to other markers. Here, we show that this inefficiency is due to the erroneous assumption that the folic acid pathway is located in chloroplasts. When the RbcS transit peptide was replaced by a transit peptide for protein import into mitochondria, the compartment where folic acid biosynthesis takes place in yeast, much higher resistance to sulfonamide and much higher transformation efficiencies are obtained, suggesting that current sul vectors are likely to function due to low‐level mistargeting of the resistance protein to mitochondria. We constructed a series of optimized transformation vectors and demonstrate that they produce transgenic events at very high frequency in both the seed plant tobacco and the green alga Chlamydomonas reinhardtii. Co‐transformation experiments in tobacco revealed that sul is even superior to nptII, the currently most efficient selectable marker gene, and thus provides an attractive marker for the high‐throughput genetic transformation of plants and algae.

The aim of the present study was to investigate the effect of transport on welfare traits, several haematological parameters and carcase hygiene in two different chicken genotypes (fast- and slow-growing strains) reared under free-range conditions. For this aim, two hundred male chicks, 100 from fast-growing (Ross 308, R) and 100 from slow-growing (Naked Neck, NN) strain were farmed. At the end of the rearing period, at 81 days of age, 56 birds/strain were randomly selected for slaughtering and submitted to two different pre-slaughter conditions: no transport (0h) or 4 hours of transport (4h). Tonic immobility (TI), blood parameters and carcase hygiene traits were determined. Strain and transport significantly affected TI of birds. Both experimental factors and their interaction significantly affected plasma creatine kinase, alkaline phosphatase, alanine aminotransferase and aspartate aminotransferase. Cholesterol and triglycerides were not different between the experimental groups, whereas glucose decreased after 4 hours of transport in both strains. A significant difference between groups for the heterophils/lymphocytes ratio after transport was also observed, with NN being higher than Ross. Concerning the oxidative stress, we observed a higher ROS production in NN chickens. The carcase microbial characteristics showed a higher level of contamination after the transport (total viable counts), but not concerning the Enterobacteriaceae counts.

The capability of some insects to be reared on by-products, waste and former foodstuff make them a very promising response to the increasing request of animals’ products. Mealworm larvae (Tenebrio molitor) are widely studied for feed and food purposes. In this study the effects of five diets were tested on the fatty acids (FA) profile, the lipid metabolism and the antioxidant molecules of the larvae. Three diets were composed by single ingredients as cookies, bread leftovers and brewery spent grains. Cookies were also mixed with bread or spent grains to increase the dietary lipids contents. Frass was studied to determine microbial contamination and FA/active biological molecules metabolisms. All the tested parameters were affected by the diets. Interestingly, beside the differences related to the diets, larvae showed a common trend in FA profile, with five FA representing between 87.25% and 94.59% of the total FAs (C18:1, C16:0, C18:2, C14:0 and C18:0). Lipid metabolism showed to be affected by FA profile of the diets, as well as retinol and tocopherol isoforms. Zeaxanthin, present into the diets, was not detected into the larvae but in the frass. Results point out a high nutritional value of mealworm larvae fed former foodstuff.

The adaptation of chickens to free-range rearing systems mainly involves the locomotory behavior, which is very different in fast-growing (FG) and slow-growing (SG) strains. This study aimed to compare the effect of moderate locomotory activity (induced and prolonged) on the blood oxidative status in a slow-growing chicken strain with that in a fast-growing one. Thirty FG (Ross 308) birds and 30 SG (Hubbard) birds were divided into 2 groups for each strain and subjected to different treatments: no exercise (the control group [C]) and 1 h of walking at 4 km/h (the exercise group [E]). Daily exercise was promoted by operators, who walked behind the animals around a paddock. Blood samples were obtained weekly from both groups. For the E group, samples were obtained before exercise (E1) and after exercise (E2). Oxidative markers (creatine kinase [CK], reactive oxygen molecular substance [ROMS], thiobarbituric acid reactive substances [TBARS]), and antioxidant compounds (α-tocopherol [α-T], γ-tocopherol [γ-T], δ-tocopherol [δ-T], α-tocotrienol [α-T3], γ-tocotrienol [γ-T3], retinol, and carotenoids) were evaluated. In both strains, the CK level was higher in chickens subjected to exercise; however, its increase was greater in the FG group than in the SG one (1.56-fold vs. 1.08-fold). The antioxidant status was worse in FG strain birds subjected to exercise, whereas the status remained nearly the same in the SG strain birds. The α-T and retinol concentrations were significantly reduced by exercise, primarily in the FG group, whereas the other antioxidant compounds (α-T3, γ-T3, γ-T, δ-T, lutein, and zeaxanthin) were unaffected by strain or treatment. The FG and SG strains had different responses to exercise, and only the SG showed a progressive reduction in TBARS and ROMS values during the 28-day experiment. Accordingly, moderate exercise may be beneficial only when the birds have suitable behavioral characteristics (e.g., higher kinetic activity, rusticity, and explorative nature) or physical characteristics (e.g., low body weight); otherwise, exercise is highly stressful and affects physiology and well-being.

The stomach content and samples of Longissimus dorsi muscle of 32 feral wild boars were collected in two different feeding areas (forest and farmland) of Umbria region (Italy). The animals from each feeding area were divided into two age classes: class 1 (12–24 months of age; 48 kg average weight) and class 2 (animals older than 2 years of age; 84 kg average weight). The major food categories consumed were hard mast and crops (89.02–75.98%). The L*(lightness) and a*(red to green colour) values of the meat were affected by the feeding area as well as the b*(yellow colour) value; the age significantly affected only the a* and the b* value of the meat. The α-tocopherol was the most abundant vitamin E homologue, ranged between 520.63 and 1881.33 ng/g and was higher in farmland areas. The index of lipid oxidation (TBARS) ranged from 0.093 and 0.140 mg MDA/kg and was higher in wild boars from farmland. The monounsaturated fatty acids (MUFA) ranged between 38.36 and 46.75% and were higher in wild boar of class 2. The total polyunsaturated fatty acids (PUFA) as well as PUFAn-6 were affected by age, while PUFAn-3 was only affected by feeding area and ranged from 0.91 and 1.99 in farmland and forest, respectively. The feeding area affects the intramuscular fat contents in terms of nutritional characteristics of the meat: the n-6/n-3 ratio that was lower in meat from animals hunted in the forest area (p ≤ .001), as well as the ARA/(EPA + DHA) ratio (p ≤ .01).

The nutritional profile and the consumer preference of fish burgers made from carp, goldfish, perch, and tench were investigated. Perch and goldfish burgers, which have the highest proportion of eicosapentaenoic acid (6.63% and 4.90%, respectively) and docosahexaenoic acid (20.28% and 11.99%, respectively), showed lower absolute amounts of these fatty acids (17.03 and 10.10 mg/100 g food, respectively) than tench and carp (31.16 and 49.63 mg/100 g food, respectively). The consumer evaluation established that differences in the perception of the fish burgers exist. The overall liking and taste were the main attributes that influenced both consumer’s acceptance and purchase intent.

The phenolic diterpene carnosic acid (CA, C20H28O4) exerts antioxidant, anti-inflammatory, anti-apoptotic, and anti-cancer effects in mammalian cells. CA activates the nuclear factor erythroid 2-related factor 2 (Nrf2), among other signaling pathways, and restores cell viability in several in vitro and in vivo experimental models. We have previously reported that CA affords mitochondrial protection against various chemical challenges. However, it was not clear yet whether CA would prevent chemically induced impairment of the tricarboxylic acid cycle (TCA) function in mammalian cells. In the present work, we found that a pretreatment of human neuroblastoma SH-SY5Y cells with CA at 1 μM for 12 h prevented the hydrogen peroxide (H2O2)-induced impairment of the TCA enzymes (aconitase, α-ketoglutarate dehydrogenase (α-KGDH), succinate dehydrogenase (SDH)) and abolished the inhibition of the complexes I and V and restored the levels of ATP by a mechanism associated with Nrf2. CA also exhibited antioxidant abilities by enhancing the levels of reduced glutathione (GSH) and decreasing the content oxidative stress markers (cellular 8-oxo-2'-deoxyguanosine (8-oxo-dG), and mitochondrial malondialdehyde (MDA), protein carbonyl, and 3-nitrotyrosine). Silencing of Nrf2 by small interfering RNA (siRNA) abrogated the protective effects elicited by CA in mitochondria of SH-SY5Y cells. Therefore, CA prevented the H2O2-triggered mitochondrial impairment by an Nrf2-dependent mechanism. The specific role of Nrf2 in ameliorating the function of TCA enzymes function needs further research.

Background The nerve growth factor (NGF), a member of the neurotrophins family, plays an important role not only in the nervous but also in other non-nervous systems such as the reproductive system. The aim of the paper is to study the in vitro effect of NGF on rabbit sperm functions. Methods Ten adult rabbit bucks were collected five times, and pooled semen samples have been analysed. NGF was quantified in seminal plasma, and the distribution of NGF receptors (TrKA and p75NTR) in sperm was established. Moreover, the dose-effect of NGF on motility rate and track speed was evaluated. Successively, the effect of the neutralisation of NGF receptors was assessed to verify the specific role of each receptor. Untreated sperm were used as control. Results Our study identified several interesting results: i) We detected NGF in seminal plasma and TrKA and p75NTR in sperm surface. In particular, TrKA is localised in the head and p75NTR in the midpiece and tail of rabbit sperm. ii) Once the optimal dose of NGF (100 ng/mL) was established, its addition affected both kinetics and other physiological traits (capacitation, apoptosis and necrosis) of rabbit sperm. (iii) The neutralisation of TrKA and p75NTR receptors affected sperm traits differently. In particular, sperm speed, apoptosis and capacitation seemed mainly modulated via p75NTR receptor, whereas motile, live cells, necrosis and acrosome reaction were modulated via TrKA. Conclusion For the first time, we showed the presence of p75NTR in rabbit sperm. NGF affects kinetic and other physiological traits of rabbit sperm. Most of these changes are modulated by the receptors involved (TrKA or p75NTR). Considering that some seminal disorders in human have been correlated with a lower NGF concentration and no studies have been done on the possible involvement of NGF receptors, these findings also provide new insights on human fertility.

A Política Nacional de Mobilidade Urbana (NPUM) surgiu com o objetivo de guiar as cidades em decisões que garantam mais acessibilidade e sustentabilidade com prioridades para transporte coletivo (ônibus veículos sobre trilhos) e não motorizado. O presente trabalho propõe uma sistemática de avaliação da efetividade prática da PNMU no que tange o transporte público e uso de bicicleta, através de alguns indicadores do Índice de Mobilidade Urbana Sustentável (IMUS), criado por Rodrigues da Silva et al. (2008), com modificação do método original. Foram comparadas quatro capitais brasileiras: Curitiba, São Paulo, Rio de Janeiro e Belo Horizonte. Os indicadores foram analisados temporalmente, de 2010 até 2015, para traçar uma evolução das cidades frente às premissas da Lei de Mobilidade de 2012 e compará-las utilizando o teste estatístico T-Student. Observou-se que as evoluções não têm distinção significativa, porém as médias de pontuação sim. Os resultados são inconclusivos para a comparação, devido à dificuldade de se encontrar dados abertos necessários para melhor avaliar os indicadores. Ainda assim, o IMUS se apresenta como um critério promissor de avaliação da NPUM.

Tenebrio molitor (mealworm) larvae represent one of the most interesting edible insects and could be reared on alternative feeds, such as former foodstuff products (FFPs). In the present work, five different FFPs (brewery spent grains, bread and cookie leftovers, and mixes of brewer&rsquo, s spent grain or bread with cookies) were employed as feeding substrates. Larvae&rsquo, s growth performances, chemical composition, microbial loads, and antioxidant status were determined. Chemical compositions of the substrates affected all the tested parameters. Brewery spent grains-fed larvae showed a faster growth period and higher crude protein and carbohydrate contents. The use of cookies as a single substrate or their addition to spent grains or bread increased the lipids contents, while growth was delayed. Microbial loads were partially affected by the fed diet. The antioxidant status of larvae showed different concentrations of tocopherols isoforms (&delta, &gamma, &alpha, ) in relation to the diet, however, no differences were detected in relation to the global antioxidant capacity (2,2-azinobis-(3 ethylbenzothiazoline-6-sulfonic acid), ABTS reducing activity, 1,1-diphenyl-2-pircydrazyl, DPPH radical scavenging activity, ferric reducing ability, FRAP). Results point out a high plasticity of mealworm larvae and the potential to tailor the final outcomes in relation to the substrate employed. Mealworms could be practically reared on FFPs to produce food-feed with high nutrient values.

In the last two decades, the human sperm count linearly decreased in Western countries. Health problems, lifestyle, pollutants, and dietary behaviours are considered as the main risk factors, and the unbalance of dietaryn‐6/n‐3fatty acids is one of the most relevant. The aim of the present research is to study the effect of different dietary sources ofn‐3polyunsaturated fatty acids (PUFA) on reproductive traits using rabbit buck as the animal model. Fifteen rabbit bucks were assigned to three experimental groups: the control group, the FLAX group fed 10% extruded flaxseed, and the FISH group fed 3.5% fish oil for 110 days (50-day adaptation and 60-day experimental periods). Semen samples were collected weekly, whereas blood was collected every two weeks for the analytical determination of semen traits, oxidative status, fatty acid profiles, isoprostanes, neuroprostanes, and the immunocytochemistry of docosahexaenoic acid (DHA) and eicosapentaenoic (EPA) acid. At the end of the trial, the rabbits were killed and the testes were removed and stored for the analysis of fatty acid profile and immunocytochemistry. Results showed that dietary administration ofn‐3PUFA improved the track speed of the sperm and increased then‐3long-chain PUFA mainly confined in the sperm tail. Seminal plasma increased the thiobarbituric reactive substances (TBARs) by three times in the groups fed supplementaln‐3, whereas the F2-isoprotanes (F2-IsoPs) and F4-neuroprostanes (F4-NeuroPs) were lower and higher, respectively, in both supplemented groups than in the control. The testes and sperm showed a higher DHA and EPA distribution in rabbits from then‐3supplemented groups compared with the control. In conclusion, supplemental dietaryn‐3PUFA improved sperm motion traits and resulted in an enrichment of membrane fatty acid in the sperm and testes of the rabbits. However, such an increased amount of PUFA negatively affected the sperm oxidative status, which was mainly correlated with the generation of F4-NeuroPs with respect to F2-IsoPs. Accordingly, the latter cannot be considered a good marker of oxidation when diets rich inn‐3PUFA are provided.

The present research studied the effect of liquorice extract (in feed and/or directly in burgers) on the shelf-life of rabbit meat. Before weaning, 28 individually caged rabbit does with their litters were divided in two dietary groups: Control, receiving a commercial diet, and Liquorice, receiving the Control diet supplemented with 6 g liquorice extract/kg (L). At 12 weeks of age, 15 fattened rabbits/treatment (one rabbit/cage) were slaughtered and their hindlegs dissected. Hindleg meat was trimmed, individually minced and divided into two parts: one of them was mixed with 0.25% (w/w) L. Storage time significantly reduced the amount of polyunsaturated fatty acids in the burger, to a different magnitude depending on the unsaturation level, experimental group and storage time. The α-tocopherol content showed higher levels during storage in the burgers from the Liquorice group. Also TBARs values showed a significant positive effect of dietary liquorice and a progressive increase at days 3 and 6 of storage.

We evaluated the effects of two types of colony cages, in which rabbit does were always in a group (C1), and where they were in combi cages furnished with removable internal walls to allow both individual and grouphousing (C2), in addition to the control group (C: conventional individual cage), on welfare, reproductive performance, and global efficiency. Forty-eight New Zealand White nulliparous rabbit does underwent artificially insemination (AI) and were divided into three groups, and reared in the different systems for about 1 year. The reproductive rhythm provides AIs at weaning (30d). In the C1 system, does were continuously grouped, while in C2, walls were inserted four days before kindling and removed 1week after it (60% of the timesheet in group). Reproductive traits and behaviour were evaluated during the entire year. The behavioural observations were performed around days 7, 36, and 44, corresponding to the inclusion of the does in the maternal cages, the insertion of walls four days before kindling, and the removal of the walls 1week after parturition in the C2 group, respectively. The percentages of does with severe skin injuries and the distribution of the injuries on different parts of body were also registered. Does reared in conventional cages showed the greatest presence of stereotype behaviours, while the C1 group showed the highest (p &lt, 0.05) incidence of aggressiveness after regrouping (attack, dominance features, and lower allo-grooming) in comparison to the C2 group (17% and 22%, in C2 and C1 does, respectively).Individually caged does achieved the best productive performance (sexual receptivity, fertility, kindling rate, and number of kits born alive and at weaning). The C1 group showed the lowest performance (p &lt, 0.05), whereas C2 showed an intermediate one. Does housed in the combi cage (C2) had higher (p &lt, 0.05) receptivity and fertility rates and higher numbers of kits born alive and at weaning (79.2% and 76.2%, 7.95 and 7.20, respectively) than the C1 group, but lower values (p &lt, 0.05) than does that were individually housed.

Standard feeds are imbalanced in term of n-6/n-3 polyunsaturated fatty acids (PUFA) ratio, with a low proportion of the latter. The reproductive system appears to be strongly affected by administration of n-3 PUFA, and ingredients rich in α-linolenic acid (ALA; i.e. vegetable sources) or EPA and DHA acids (i.e. fish oil) can be included in animal diets to balance PUFA intake. The aim of this study was to evaluate the effect of dietary supplementation with flaxseed (ALA) or fish oil (EPA and DHA) on PUFA metabolism in rabbit does. A total of 60 New Zealand White female rabbits were assigned to three experimental groups: control group, FLAX group fed 10% extruded flaxseed and FISH group fed 3% fish oil. Blood, milk, liver and ovaries were collected from the does to assess the lipid composition; furthermore, FADS2 gene expression was assessed in liver and ovary tissues. Reproductive performance of does was also recorded. The fertility rate and number of weaned rabbits improved with n-3 dietary supplementation: does at first parity showed the lowest reproductive results, but the administration of n-3 reduced the gap between primiparous and multiparous does. Feed consumption and milk production were not affected by the feeding regime. The fatty acid composition of milk, plasma, liver and ovaries were widely influenced by diet, showing higher concentrations of n-3 long-chain PUFA (LCP) in does fed with n-3 enriched diets. FISH diet resulted in the highest n-3 LCP enrichment, whereas in the FLAX group, this increase was lower. Blood and milk showed low levels of LCP, whereas liver and ovaries were the main sites of n-3 LCP synthesis and accumulation. Accordingly, although the liver is the main metabolic centre for LCP synthesis, ovaries also have a prominent role in LCP generation. FADS2 expression in liver and ovary tissue was downregulated by FISH administration. In conclusion, the enrichment of diets with n-3 PUFA could be an effective strategy for improving the reproductive performance of does.

Simple Summary The weaning of young rabbits is a critical period that is often accompanied by digestive troubles. Innovations in feeding strategy are urgently needed to preserve rabbit health and to reduce the use of antibiotics. We show here that providing dehydrated alfalfa during weaning is a promising solution to manage health status by favoring the establishment of a proper digestive microbiota. Abstract The improvement of rabbit gut microbiota by modifying nutritional components of the feed or favoring its early intake of feed has been previously investigated. The early administration of dehydrated alfalfa (A) or fresh grass (G) for rabbits, during the peri-weaning period (30 and 45 days of age), and their effect on performance and caecal microbiota compared to a standard diet (C) were evaluated. Until 15 days of age, nine litters/group were housed in the maternal cage and milked once per day. From 15 to 30 days, the young rabbits could consume both milk and solid feed (pelleted for C or supplemental feed for A and G). At 30 days of age, the rabbits were weaned and, until 45 days, were kept in single cages following the same dietary protocol. No significant changes were found in the milk intake or the individual weight of young rabbits at 30 and 45 days. The caecal Firmicutes/Bacteroidetes (bacterial phyla ratio) increased with age (from 2.43 to 6.05 on average, at 30 and 45 days). The Ruminococcaceae/Lachinospiraceae (bacterial family ratio) was highest in the A group at both ages, followed by G then C. The early administration of dehydrated alfalfa is a promising solution to improve health status by favoring an appropriate digestive microbiota.

Olive leaves (OL) are by-products of olive cultivation that are usually discarded, but due to the presence of bioactive compounds beneficial to health, they could be used. Thus, the objective of this paper is to develop a cracker from an olive leaf flour (OLF) and to verify the sensory effect of its use. Crackers were made from 5 formulations: F0 (without OLF), F3 (1,7 g/100 g), F5 (2,8 g/100 g), F7 (4 g/100 g), and F9 (5,1 g/100 g). In addition to OLF, the crackers had refined and whole wheat flour and almond flour. Oleuropein quantification, macronutrients, and sensory analysis were performed using Quantitative Descriptive Analysis (QDA), as well as consumer acceptance and preference, in addition to purchase intent. Pinocembrin (PB; 5,7-dihydroxyflavanone; C15H12O4) is a flavonoid found in propolis and exerts antioxidant, anti-inflammatory, and antimicrobial effects. Furthermore, PB has been studied as a neuroprotective agent. However, it remains to be understood whether and how PB would induce mitochondrial protection in mammalian cells. Therefore, we investigated here the mechanism involved in the protective effects elicited by PB in paraquat (PQ; 100 μM)-treated SH-SY5Y neuroblastoma cells. PB (25 μM) pretreatment (for 4 h) downregulated the levels of Bcl-2-associated X protein (Bax), blocked the release of cytochrome c to the cytosol, and inhibited the PQ-induced activation of caspase-9 and caspase-3. Besides, PB prevented mitochondrial dysfunction by suppressing the PQ-elicited inhibition of complexes I and V. Moreover, PB abrogated the loss of mitochondrial membrane potential (MMP) and the decline in ATP levels in the cells exposed to PQ. PB exerted antioxidant effects on mitochondria by decreasing the levels of redox impairment markers in mitochondrial membranes. Importantly, PB enhanced the levels of mitochondrial reduced glutathione (GSH). Upregulation of enzymes involved in the synthesis of GSH was seen in the cells exposed to PB. PB afforded mitochondrial protection by activating the extracellular signal-regulated kinase/nuclear factor erythroid 2-related factor 2 (Erk1/2-Nrf2) axis, since inhibition of Erk1/2 or silencing of Nrf2 abrogated these effects. Therefore, PB exerted mitochondrial and cellular protection by an Erk1/2-Nrf2-dependent mechanism.

Single-wall carbon nanotubes functionalized with polyethylene glycol (SWCNT-PEG) are promising materials for biomedical applications such as diagnostic devices and controlled drug-release systems. However, several questions about their toxicological profile remain unanswered. Thus, the aim of this study was to investigate the action of SWCNT-PEG in Danio rerio zebrafish embryos at the molecular, physiological and morphological levels. The SWCNT used in this study were synthesized by the high-pressure carbon monoxide process, purified and then functionalized with distearoyl phosphatidylethanolamine block copolymer-PEG (molecular weight 2 kDa). The characterization process was carried out with low-resolution transmission electron microscopy, thermogravimetric analysis and Raman spectroscopy. Individual zebrafish embryos were exposed to the SWCNT-PEG. Toxic effects occurred only at the highest concentration tested (1 ppm) and included high mortality rates, delayed hatching and decreased total larval length. For all the concentrations tested, the alkaline comet assay revealed no genotoxicity, and Raman spectroscopy measurements on the histological slices revealed no intracellular nanotubes. The results shown here demonstrate that SWCNT-PEG has low toxicity in zebrafish embryos, but more studies are needed to understand what mechanisms are involved. However, the presence of residual metals is possibly among the primary mechanisms responsible for the toxic effects observed, because the purification process was not able to remove all metal contamination, as demonstrated by the thermogravimetric analysis. Carnosic acid (CA; C20H28O4) is a phenolic diterpene found in rosemary (Rosmarinus officinalis L.) and exhibits protective properties, e.g., antioxidant, anti-inflammatory, antitumor, and antimicrobial activities. In this context, CA has been viewed as a neuroprotective agent due to its ability in rescuing neuronal cells from pro-oxidant and pro-apoptotic challenges. In the present work, we found that CA pretreatment at 1 µM for 12 h suppressed the mitochondria-related pro-oxidant and mitochondria-dependent pro-apoptotic effects of chlorpyrifos (CPF) in human neuroblastoma SH-SY5Y cells. CA prevented mitochondrial membrane potential disruption and decreased the levels of oxidative stress markers in mitochondrial membranes obtained from cells exposed to CPF. CA also inhibited cytochrome c release and activation of the caspases-9 and -3, as well as decreased DNA fragmentation, in CPF-treated cells. CA upregulated the content of glutathione (GSH) in mitochondria by a mechanism involving the activation of the phosphoinositide-3-kinase (PI3K)/Akt/nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway, since inhibition of PI3K/Akt or silencing of Nrf2 using siRNA strategy abolished the protection exerted by CA in SH-SY5Y cells. Therefore, CA protected mitochondria of SH-SY5Y cells through the activation of the PI3K/Akt/Nrf2 axis, causing upregulation of the mitochondrial GSH content and consequent antioxidant and anti-apoptotic effects.

The aim of the study was to evaluate the effect of transport length on in vivo oxidative status and breast meat characteristics in two chicken genotypes reared under free range conditions. A total of 200 male chicks, 100 from fast-growing (Ross 308) and 100 from medium-growing (Naked Neck) strain were used. Fifty-six of these, 28 for genotype, before slaughtering, were randomly allocated to 2 pre-slaughter conditions: absence (0 h) or 4 h of transport. The transport length significantly affected the in vivo oxidative status of broiler greatly reducing the α and δ-tocopherol, retinol and lutein + zeaxanthin content of plasma, and increased the oxidative stress (thiobarbituric acid reactive substances, TBARS) in both strains. Concerning meat quality, the pH (0, 2 and 24 h post-mortem) of breast muscles of chickens transported for 4 h, showed higher values, and respect to strains, Naked Neck had lower values. The pH values were negatively correlated with the lightness (2–24 h) and the shear force of meat. The transport length significantly affected the fatty acid profile of breast muscle, with a decrease in polyunsaturated fatty acids and an increase in TBARS value. Even the antioxidants content of breast was reduced by chicken transport (α-tocotrienol, α-, δ-tocopherol and lutein + zeaxanthin), especially in Naked Neck birds. In conclusion, the results indicate that transport for 4 h prior to slaughter, negatively affect the meat quality of poultry. Slow-growing chickens seem more sensible to stress transport due to the higher kinetic behaviour of these strains.

The aim of the study was to define poultry adaptability to organic system, through the assessment of several endpoints. Eight hundred male birds of slow-growing birds (Ancona: A, Leghorn: L, crossbreed Cornish × Leghorn: CL), medium-growing (Gaina: G, Robusta Maculata: RM, Kabir: K, Naked Neck: NN) and fast-growing strains (Ross: R) were organically reared. A and L genotypes displayed a quicker reaction time when submitted to tonic immobility test, and a great variety of behaviour and exploiting all the pasture area. Concerning feather conditions L, A, CL G and RM showed the best values for all considered body regions, as well as the absolute absence of foot pad and breast blister lesions. Static behaviour of R and G chickens did not produce a significant oxidative burst whereas, the active behaviour of A, slow-growing birds, increased the oxygen demand. Plasma α-tocopherol followed the trend of kinetic and foraging activity being higher in slow-, intermediate in medium- and lower in fast-growing birds. The adaptability index showed the best result of slow-growing strains with intermediate results in medium-growing and the worst in fast-growing ones. There is a negative linear correlation between adaptation and daily weight gain. However, within the same sub-group (slow, medium and fast), there is no correlation between daily weight gain and adaptation to an organic system. Growing rabbits’ (42 rabbits/group, 3 rabbits/cage, 14 cages/treatment) diet was supplemented with 5% Spirulina (Arthrospira platensis) and 3% thyme (Thymus vulgaris L.) powder single (S or T) and in combination (ST) between 35 and 77 days of age. On day 0 (weaning at 35 days of age) 14 rabbits were vaccinated with 100 μg/animal ovalbumin to provoke immune response. Blood samples were taken on days 0, 14, 28 and 42 of the experimental period. Sampling dates significantly influenced total protein, albumin, glucose, cholesterol, urea, creatinine concentration and enzyme (AST, ALT, GGT) activities, with a significant age × diet interaction in the case of TP and CREA. There was a significant increase in ALT (+45 and 74%) and GGT (+87 and 102%) activity after immunisation. While Spirulina and thyme significantly ameliorated the rise in AST activity, their effect was inefficient in the case of GGT. Spirulina, both single and in combination showed a tendency in higher IgG level as compared to control (P0.05). Higher MDA concentration was measured in the red blood cells of S, T and ST animals, while no other significant diet effect on the antioxidant parameters was detected, however, significant sampling date × diet interaction was found in the case of GPx activity. Bitter vetch (Vicia ervilia (L.) Willd.), a grain legume crop well adapted in marginal soils, has mainly been used for animal feed. Nowadays, bitter vetch seeds in feed formulations are replaced by other protein sources such as soybean meal. However, in the context of sustainable economic development, it may be beneficial to enhance the cultivation of bitter vetch landraces in marginal areas. Fifty-six bitter vetch accessions of different provenance were preliminarily characterised by microsatellite DNA analysis to discriminate landraces suitable for specific and restricted environments. Twenty-two landraces of two genetically different groups were then selected for further characterisation by agro-morphological analyses. Being late-flowering with a seed yield of up to 3–4 t ha–1 in experimental field conditions, with neither chemical nor water input, these plants will be valuable material for long-term study to develop new cultivars adapted for seed production under organic agricultural systems in Southern Europe. The seeds of these 22 landraces were also evaluated, with positive results, as partial replacement of soybean in rabbit diets.

The present study tested the effect of a liquorice root extract (L) administration in the diet of growing rabbits and/or in their minced meat. A total of fifty-six rabbit does and their kits (3 weeks of age) were divided into 4 groups receiving a commercial diet (C) or the C diet supplemented with increasing levels of L: 2 g/kg (L2), 4 g/kg (L4), 6 g/kg (L6). At weaning (5 weeks of age), sixty rabbits/group were randomly selected, caged by 3, and fed ad libitum with the experimental diets until slaughter (12 weeks of age). At 6 weeks of age, 6 rabbits/diet were selected for gastrointestinal tract pH and caecal microbial count analyses. Live performance and health status were recorded weekly and daily, respectively. Carcass traits were measured after slaughter and hind leg (HL) and Longissimus lumborum (LL) meat were dissected for pHu and L*a*b* colour measurements. Minced meat from HL belonging to C and L6 dietary treatments was each split in two parts to incorporate liquorice extract at 0.25% (w/w), obtaining four groups of meat burgers: CC; CL; L6C and L6L. The pH, L*a*b* colour and microbial count (TVC, Coliforms, and Pseudomonas) were measured at days 0, 3 and 6 of refrigerated storage. A sensory analysis was conducted on LL meat of C and L6 dietary treatments, and tested attributes were odour, taste, flavour, texture and off-flavours. Adulteration of Mozzarella di Bufala Campana with cow milk is a common fraud because of the high price and limited seasonal availability of water buffalo milk. To identify such adulteration, this work proposes a novel approach based on the use of species-specific, low-molecular-weight biomarkers (LMWBs). Liquid chromatography–tandem mass spectrometry screening analyses identified β-carotene, lutein, and β-cryptoxanthin as LMWBs of cow milk, while ergocalciferol was found only in water buffalo milk. Adulterated mozzarellas were prepared in the laboratory and analyzed for the four biomarkers. Combined quantification of β-carotene and ergocalciferol enabled the detection of cow milk with a sensitivity threshold of 5% (w/w). The method was further tested by analyzing a certificated water buffalo mozzarella and several commercial products. This approach is alternative to conventional proteomic and genomic methods and is advantageous for routine operations as a result of its simplicity, speed, and low cost.

This experiment compared the productive performance and location of growing rabbits in pens without and with two-level platforms (wire-mesh or plastic-mesh). A total of 174 rabbits of both sexes weaned at 5 weeks of age were randomly divided into three groups (n = 58 rabbits/group, 2 pens/treatment, 29 rabbits/pen). The floor area of pens was 1.0 × 1.83 m, and the floor was made of wire-mesh. Two pens were equipped with wire-mesh (WP) and two pens with plastic-mesh elevated platforms (PP) on two levels, and two pens were without platforms (NoP). The objective of this study was to evaluate the effect of Curcuma longa powder and ascorbic acid on some quality traits of rabbit burgers. The burgers (burgers control with no additives; burgers with 3.5 g of turmeric powder/100g meat; burgers with 0.1g of ascorbic acid/100g meat) were analyzed at Days 0 and 7 for pH, color, drip loss, cooking loss, fatty acid profile, TBARS, antioxidant capacity (ABTS, DPPH and FRAP) and microbial growth. The addition of turmeric powder modified the meat color, produced an antioxidant capacity similar to ascorbic acid and determined a lower cooking loss than other formulations. Turmeric powder might be considered as a useful natural antioxidant, increasing the quality and extending the shelf life of rabbit burgers.

The aim of the current study was to investigate the association between the InDel polymorphism in the angiotensin I-converting enzyme gene (ACE) and the rs699 polymorphism in the angiotensinogen gene (AGT) and diabetes mellitus type 2 (DM2) in a sample population from Southern Brazil. A case-control study was conducted with 228 patients with DM2 and 183 controls without DM2. The ACE InDel polymorphism was genotyped by polymerase chain reaction (PCR) with specific primers, followed by electrophoresis on 1.5% agarose gel. The AGT rs699 polymorphism was genotyped using a real-time PCR assay. No significant association between the ACE InDel polymorphism and DM2 was detected (P = 0.97). However, regarding the AGT rs699 polymorphism, DM2 patients had a significantly higher frequency of the AG genotype and lower frequency of the GG genotype when compared to the controls (P = 0.03). Our results suggest that there is an association between the AGT rs699 polymorphism and DM2 in a Brazilian sample.
