We created a volumetric template of the marmoset (Callithrix jacchus) brain, which enables localization of the cortical areas defined in the Paxinos et al. (The marmoset brain in stereotaxic coordinates. Elsevier Academic Press, Cambridge, 2012) marmoset brain atlas, as well as seven broader cortical regions (occipital, temporal, parietal, prefrontal, motor, limbic, insular), different brain compartments (white matter, gray matter, cerebro-spinal fluid including ventricular spaces), and various other structures (brain stem, cerebellum, olfactory bulb, hippocampus). The template was designed from T1-weighted MR images acquired using a 3 T MRI scanner. It was based on a single fully segmented marmoset brain image, which was transported onto the mean of 13 adult marmoset brain images using a diffeomorphic strategy that fully preserves the brain topology. In addition, we offer an automatic segmentation pipeline which fully exploits the proposed template. The segmentation pipeline was quantitatively assessed by comparing the results of manual and automated segmentations. An associated program, written in Python, can be used from a command-line interface, or used interactively as a module of the 3DSlicer software. This program can be applied to the analysis of multimodal images, to map specific cortical areas in lesions or to define the seeds for further tractography analyses.

It is well established that human life expectancy increases considerably with an ever-growing number of people suffering from age-related cognitive decline and degenerative brain diseases. This necessitates the development of animal models to counteract or stop the progression of the decline early enough. Presently, primate models are few, and many studies argue for the marmoset as an interesting primate model presenting a short life span and being easily available in research laboratories. In this article, we propose the marmoset as a valid model for cognitive decline. Using a computer touch screen, we trained 35 marmosets from 2 to 14 years of age to perform reversal learning and delayed-matching-to-position tasks. We found typical age-related cognitive deficits related to executive functions and spatial working memory. Applying a recursive algorithm, we detected 2 critical periods from which deficits appear. Mainly, response strategy deficits appear from age 4, whereas impairments in inhibitory control appear from age 7–8. Furthermore, the presence of outliers, sometimes at an early age, suggests pathological cognitive deficits that would require imaging exploration in parallel to behavior.

Our knowledge of human brain evolution primarily relies on the interpretation of palaeoneurological evidence. In this context, an endocast or replica of the inside of the bony braincase can be used to reconstruct a timeline of cerebral changes that occurred during human evolution, including changes in topographic extension and structural organisation of cortical areas. These changes can be tracked by identifying cerebral imprints, particularly cortical sulci. The description of these crucial landmarks in fossil endocasts is, however, challenging. High-resolution imaging techniques in palaeoneurology offer new opportunities for tracking detailed endocranial neural characteristics. In this study, we use high-resolution imaging techniques to document the variation in extant human endocranial sulcal patterns for subsequent use as a platform for comparison with the fossil record. We selected 20 extant human crania from the Pretoria Bone Collection (University of Pretoria, South Africa), which were detailed using X-ray microtomography at a spatial resolution ranging from 94 to 123 μm (isometric). We used Endex to extract, and Matlab to analyse the cortical imprints on the endocasts. We consistently identified superior, middle and inferior sulci on the frontal lobe; and superior and inferior sulci on the temporal lobe. We were able to label sulci bordering critical functional areas such as Broca's cap. Mapping the sulcal patterns on extant endocasts is a prerequisite for constructing an atlas which can be used for automatic sulci recognition.

The relation between normal and pathological aging and the cerebrovascular component is still unclear. In this context, the common marmoset, which has the advantage of enabling longitudinal studies over a reasonable timeframe, appears as a good pre-clinical model. However, there is still a lack of quantitative information on the macrovascular structure of the marmoset brain. In this paper, we investigate the potentiality of multi-angle TOF MR angiography using a 3T MRI scanner to perform morphometric analysis of the marmoset brain vasculature. Our image processing pipeline greatly relies on the use of multiscale vesselness enhancement filters to help extract the 3D macrovasculature and perform subsequent morphometric calculations. Although multi-angle acquisition does not improve morphometric analysis significantly as compared to single-angle acquisition, it improves the network extraction by increasing the robustness of image processing algorithms.

Microglia are cells that protect brain tissue from invading agents and toxic substances, first by releasing pro-inflammatory cytokines, and thereafter by clearing tissue by phagocytosis. Microglia express ferritin, a protein with ferroxidase activity capable of storing iron, a metal that accumulates in brain during aging. Increasing evidence suggests that ferritin plays an important role in inflammation. However, it is not known if ferritin/iron content can be related to the activation state of microglia. To this end, we aimed to delineate the role of ferritin in microglia activation in a non-human primate model. We analyzed brains of male marmosets and observed an increased density of ferritin+ microglia with an activated phenotype in hippocampus and cortex of old marmosets (mean age 11.25 ± 0.70 years) compared to younger subjects. This was accompanied by an increased number of dystrophic microglia in old marmosets. However, in aged subjects (mean age 16.83 ± 2.59 years) the number of ferritin+ microglia was decreased compared to old ones. Meanwhile, the content of iron in brain tissue and cells with oxidized RNA increased during aging in all hippocampal and cortical regions analyzed. Abundant amoeboid microglia were commonly observed surrounding neurons with oxidized RNA. Notably, amoeboid microglia were arginase1+ and IL-10+, indicative of a M2 phenotype. Some of those M2 cells also presented RNA oxidation and a dystrophic phenotype. Therefore, our data suggest that ferritin confers protection to microglia in adult and old marmosets, while in aged subjects the decline in ferritin and the increased amount of iron in brain tissue may be related to the increased number of cells with oxidized RNA, perhaps precluding the onset of neurodegeneration.

The number of studies that use the common marmoset (Callithrix jacchus) in various fields of neurosciences is increasing dramatically. In general, animals enter the study when their health status is considered satisfactory on the basis of classical clinical investigations. In behavioral studies, variations of score between individuals are frequently observed, some of them being considered as poor performers or outliers. Experimenters rarely consider the fact that it could be related to some brain anomaly. This raises the important issue of the reliability of such classical behavioral approaches without using complementary imaging, especially in animals lacking striking external clinical signs. Here we report the case of a young marmoset which presented a set of cognitive impairments in two different tasks compared to other age-matched animals. Brain imaging revealed a patent right lateral ventricular enlargement with a mild hippocampal atrophy. This abnormality could explain the cognitive impairments of this animal. Such a case points to the importance of complementing behavioral studies by imaging explorations to avoid experimental bias.

Tissue non-specific alkaline phosphatase (TNAP) may be involved in the synthesis of GABA and adenosine, which are the main inhibitory neurotransmitters in cortex. We explored this putative TNAP function through electrophysiological recording (local field potential ) in slices of mouse somatosensory cortex maintained in vitro . We used tetramisole , a well documented TNAP inhibitor, to block TNAP activity. We expected that inhibiting TNAP with tetramisole would lead to an increase of neuronal response amplitude, owing to a diminished availability of GABA and/or adenosine. Instead, we found that tetramisole reduced neuronal response amplitude in a dose-dependent manner. Tetramisole also decreased axonal conduction velocity. Levamisole had identical effects. Several control experiments demonstrated that these actions of tetramisole were independent from this compound acting on TNAP. In particular, tetramisole effects were not stereo-specific and they were not mimicked by another inhibitor of TNAP, MLS-0038949 . The decrease of axonal conduction velocity and preliminary intracellular data suggest that tetramisole blocks voltage-dependent sodium channels . Our results imply that levamisole or tetramisole should not be used with the sole purpose of inhibiting TNAP in living excitable cells as it will also block all processes that are activity-dependent. Our data and a review of the literature indicate that tetramisole may have at least four different targets in the nervous system. We discuss these results with respect to the neurological side effects that were observed when levamisole and tetramisole were used for medical purposes, and that may recur nowadays due to the recent use of levamisole and tetramisole as cocaine adulterants.

Despite the growing body of evidence pointing on the involvement of tissue non-specific alkaline phosphatase (TNAP) in brain function and diseases like epilepsy and Alzheimer’s disease, our understanding about the role of TNAP in the regulation of neurotransmission is severely limited. The aim of our study was to integrate the fragmented knowledge into a comprehensive view regarding neuronal functions of TNAP using objective tools. As a model we used the signal transduction molecular network of a pyramidal neuron after complementing with TNAP related data and performed the analysis using graph theoretic tools. The analyses show that TNAP is in the crossroad of numerous pathways and therefore is one of the key players of the neuronal signal transduction network. Through many of its connections, most notably with molecules of the purinergic system, TNAP serves as a controller by funnelling signal flow towards a subset of molecules. TNAP also appears as the source of signal to be spread via interactions with molecules involved among others in neurodegeneration. Cluster analyses identified TNAP as part of the second messenger signalling cascade. However, TNAP also forms connections with other functional groups involved in neuronal signal transduction. The results indicate the distinct ways of involvement of TNAP in multiple neuronal functions and diseases.

The presence of alkaline phosphatase (AP) activity in the neural tissue has been described decades ago. However, only recent studies clarified the isotype, regional distribution and subcellular localization of the AP expressed in the cerebral cortex of diverse mammalian species including the human. In the primate brain the discovery that the bone AP isotype (TNAP) is expressed provided the opportunity of a deeper understanding of the role of this enzyme in neuronal functions based on the knowledge acquired by studying the role of the enzyme in hypophosphatasia, mostly in bone mineralization. TNAP exhibits widespread substrate specificity and, in the brain, it is potentially involved in the regulation of molecules which play fundamental roles in signal transmission and development. In light of these observations, the localization of TNAP in the human cerebral cortex is of high significance when considering that epilepsy is often diagnosed in hypophosphatasia. Here we overview our results on the identification of TNAP in the primate cerebral cortex: TNAP exhibits a noticeably high activity in the synapses and nodes of Ranvier, is specifically present in layer 4 of the sensory cortices and additionally in layer 5 of prefrontal, temporal and other associational areas in human. Our studies also indicate that bone AP activity depends on the level of sensory input and that its developmental time-course exhibits characteristic regional differences. The relevance of our findings regarding human cortical physiology and brain disorders are discussed.

The ubiquitous enzyme TNAP (tissue non-specific alkaline phosphatase) is found in numerous tissues such as liver, kidney and bone, but little attention has been paid to its expression and role in the brain. Observations in TNAP-KO mice, which analyzed the role of this enzyme in osteogenesis, had suggested that TNAP might be involved in GABA neurotransmission. Apart from its presence in endothelial cells, here we show a specific and strong alkaline phosphatase (AP) activity in the neuropile, matching the pattern of thalamo-cortical innervation in layer 4 of the primate sensory cortices (visual, auditory and somatosensory). Such a pattern is also evident in rodents and carnivores, making AP a powerful marker of primary sensory areas. Remarkably, AP activity is regulated by sensory experience as demonstrated by monocular deprivation paradigms in monkeys. The areal and laminar distribution of AP activity matches that of the GAD(65), the GABA synthesizing enzyme found in presynatic terminals. As our electron microscopic investigations indicate that AP is found at the neuronal membranes and in synaptic contacts, it is proposed that the neuronal AP isoform (NAP), may be a key enzyme in regulating neurotransmission and could therefore play an important role in developmental plasticity and activity-dependent cortical functions.

Cerebral aging has been linked to structural and functional changes in the brain throughout life. Here, we study the marmoset, a small non-human primate, in order to get insights into the mechanisms of brain aging in normal and pathological conditions. Imaging the brain of small animals with techniques such as MRI, quickly becomes a challenging task when compared with human brain imaging. Very often, a simple pre-processing step such as brain extraction cannot be achieved with classical tools. In this paper, we propose a diffeomorphic registration algorithm, which makes use of learned constraints to propagate the manual segmentation of a marmoset brain template to other MR images of marmoset brains. The main methological contribution of our paper is to explore a new strategy to automatically tune the spatial regularization of the deformations. Results show that we obtain a robust segmentation of the brain, even for images with a low contrast.

Evidence is emerging with regard to the role of tissue non-specific alkaline phosphatase (TNAP) in neural functions. As an ectophosphatase, this enzyme might influence neural activity and synaptic transmission in diverse ways. The localization of the enzyme in known neural circuits, such as the retina, might significantly advance an understanding of its role in normal and pathological functioning. However, the presence of TNAP in the retina is scarcely investigated. Our multispecies comparative study (zebrafish, cichlid, frog, chicken, mouse, rat, golden hamster, guinea pig, rabbit, sheep, cat, dog, ferret, squirrel monkey, human) using enzyme histochemistry and Western blots has shown the presence of TNAP activity in the retina of several mammalian species, including humans. Although the TNAP activity pattern varies across species, we have observed the following trends: (1) in all investigated species (except golden hamster), retinal vessels display TNAP activity; (2) TNAP activity consistently occurs in the photoreceptor layer; (3) in majority of the investigated species, marked TNAP activity is present in the outer and inner plexiform layers. In zebrafish, frog, chicken, guinea pig, and rat, TNAP histochemistry has revealed several sublayers of the inner plexiform layer. Frog, golden hamster, guinea pig, mouse, and human retinas possess a subpopulation of amacrine cells positively staining for TNAP activity. The expression of TNAP in critical sites of retinal signal transmission across a wide range of species suggests its fundamental, evolutionally conserved role in vision.

We previously showed that immunoreactivity to N-Methyl-D-aspartate (NMDA) receptors in primary visual cortex of Callithrix jacchus is regulated by visual activity during the second and third postnatal months (Fonta et al., 1997). The purpose of the present study was to show that the columnar pattern of high and low NMDAR1 immunoreactivity observed in monocularly deprived animals corresponds to ocular dominance columns linked to the nondeprived and deprived eye, respectively. We compared cortical distribution of NMDAR1 receptors and the projection zones of thalamic afferents, revealed by transneuronal transport of tritiated proline, in 2-month-old, either monocularly deprived or control, marmosets. The data show that ocular dominance columns exist in 2-month-old marmosets and that a 2-week monocular deprivation by means of eyelid suture leads to a modification of the thalamo-cortical afferents organization. Experiments of neuronal tracing and immunohistochemistry performed on the same animals demonstrated that cortical domains with decreased NMDAR1 level correspond to the deprived eye columns. These investigations, coupled to the previous results, strongly suggest that the NMDA receptors, regulated by visual activity, are involved in the refining of ocular dominance columns in the primary visual cortex of juvenile marmoset.

During the early postnatal phase of high neuronal plasticity, an altered visual input leads to great modifications of visual cortex organization [Y. Fregnac, M. Imbert, Development of neuronal selectivity in primary visual cortex of cat, Physiol. Rev., 64 (1984) 375–434; D.H. Hubel, T.N. Wiesel, S. LeVay, Plasticity of ocular dominance columns in monkey striate cortex, Philos. Trans. R. Soc. London, Ser. B, 278 (1977) 377–409.]. We used refined differential screening of an organized cDNA library to identify the genes that may participate in this plasticity. We isolated a candidate plasticity gene encoding for a 163 aa protein that is closely related to the human and yeast Skp1p, a key factor in cell cycle progression [C. Bai, K. Hofman, L. Ma, M. Goebl, J.W. Harper, S.J. Elledge, SKP1 connects cell cycle regulators to the ubiquitin proteolysis machinery through a novel motif, the F-box, Cell, 86 (1996) 263–274; C. Connelly, P. Hieter, Budding yeast SKP1 encodes an evolutionary conserved kinetochore protein required for cell cycle progression, Cell, 86 (1996) 275–285; H. Zhang, R. Kobayashi, K. Galaktionov, D. Beach, p19Skp1 and p45Skp2 are essential elements of the cyclin A-CDK2 S phase kinase, Cell, 82 (1995) 915–925.]. Northern blot analysis showed that the expression of SKP1 (Skp1p gene) dramatically decreased after 2 h of light stimulation in the visual cortex of young dark-reared rats. This down regulation lasted at least 72 h. It was specific for the critical period as we did not observe any significant regulation of SKP1 mRNA by light in adult dark-reared rat brain. The down regulation was observed in the superior colliculus but also in the frontal cortex and in the hippocampus. The fact that this down regulation was not restricted to the visual system, suggested that it could be produced by dark rearing-induced hormonal changes. The significance of SKP1 expression in the brain and its regulation are discussed.

It has been demonstrated that the primary visual cortex is highly sensitive to manipulations of the visual environment during a specific, early, postdevelopmental period: the critical period. Pharmacological studies have shown that N-methyl-D-aspartate (NMDA) receptors are involved in the plasticity of the visual cortex just as they are involved in the induction of long-term potentiation (LTP), another activity-dependent form of plasticity. The setting up of synaptic connectivity in the neocortex may rely on LTP-like mechanisms. By using immunohistochemistry techniques, we tested the hypothesis of the role of subunit R1 of NMDA (NMDAR1) receptors in the thalamocortical afferent segregation into ocular-dominance columns in the New World monkey,Callithrix jacchus.We employed early and short (2 weeks) monocular-deprivation periods at different ages of postnatal development (17, 46, 67, 107, and 188 postnatal days). We observed heterogeneous distribution of NMDAR1 in the layer IVC receiving the thalamic inputs if the deprivation was realized between the ages of 46 and 107 days. Layers IVCa and IVCb were involved differently as a function of the deprivation age. The striped pattern lost its differential intensity with the postnatal age. These results are compared with the ocular-dominance pattern evolution described in other works on this primate. They provide evidence of the NMDAR1 role in the modular organization, within time limits, during the postnatal development of the primary visual cortex. J. Comp. Neurol.

International audience; Mutations in the tissue-nonspecific alkaline phosphatase (TNAP) gene can result in skeletal and dental hypomineralization and severe neurological symptoms. TNAP is expressed in the synaptic cleft and the node of Ranvier in normal adults. Using TNAP knockout (KO) mice (Akp2(-/-)), we studied synaptogenesis and myelination with light- and electron microscopy during the early postnatal days. Ablation of TNAP function resulted in a significant decrease of the white matter of the spinal cord accompanied by ultrastructural evidence of cellular degradation around the paranodal regions and a decreased ratio and diameter of the myelinated axons. In the cerebral cortex, myelinated axons, while present in wild-type, were absent in the Akp2( -/- ) mice and these animals also displayed a significantly increased proportion of immature cortical synapses. The results suggest that TNAP deficiency could contribute to neurological symptoms related to myelin abnormalities and synaptic dysfunction, among which epilepsy, consistently present in the Akp2(-/-) mice and observed in severe cases of hypophosphatasia.

International audience; Vascular domains have been described as being coupled to neuronal functional units enabling dynamic blood supply to the cerebral cyto-architecture. Recent experiments have shown that penetrating arterioles of the grey matter are the building blocks for such units. Nevertheless, vascular territories are still poorly known, as the collection and analysis of large three-dimensional micro-vascular networks are difficult. By using an exhaustive reconstruction of the micro-vascular network in an 18mm(3) volume of marmoset cerebral cortex, we numerically computed the blood flow in each blood vessel. We thus defined arterial and venular territories and examined their overlap. A large part of the intracortical vascular network was found to be supplied by several arteries and drained by several venules. We quantified this multiple potential to compensate for deficiencies by introducing a new robustness parameter. Robustness proved to be positively correlated with cortical depth and a systematic investigation of coupling maps indicated local patterns of overlap between neighbouring arteries and neighbouring venules. However, arterio-venular coupling did not have a spatial pattern of overlap but showed locally preferential functional coupling, especially of one artery with two venules, supporting the notion of vascular units. We concluded that intra-cortical perfusion in the primate was characterised by both very narrow functional beds and a large capacity for compensatory redistribution, far beyond the nearest neighbour collaterals.

International audience; The enzyme tissue non-specific alkaline phosphatase (TNAP) belongs to the ectophosphatase family. It is present in large amounts in bone in which it plays a role in mineralization but little is known about its function in other tissues. Arguments are accumulating for its involvement in the brain, in particular in view of the neurological symptoms accompanying human TNAP deficiencies. We have previously shown, by histochemistry, alkaline phosphatase (AP) activity in monkey brain vessels and parenchyma in which AP exhibits specific patterns. Here, we clearly attribute this activity to TNAP expression rather than to other APs in primates (human and marmoset) and in rodents (rat and mouse). We have not found any brain-specific transcripts but our data demonstrate that neuronal and endothelial cells exclusively express the bone TNAP transcript in all species tested, except in mouse neurons in which liver TNAP transcripts have also been detected. Moreover, we highlight the developmental regulation of TNAP expression; this also acts during neuronal differentiation. Our study should help to characterize the regulation of the expression of this ectophosphatase in various cell types of the central nervous system.

International audience; The ectoenzyme tissue non-specific alkaline phosphatase (TNAP) is mostly known for its role in bone mineralization. However, in the severe form of hypophosphatasia, TNAP deficiency also results in epileptic seizures, suggesting a role of this enzyme in brain functions. Accordingly, TNAP activity was shown in the neuropil of the cerebral cortex in diverse mammalian species. However in spite of its clinical significance, the neuronal localization of TNAP has not been investigated in the human brain. By using enzyme histochemistry, we found an unprecedented pattern of TNAP activity appearing as an uninterrupted layer across diverse occipital-, frontal- and temporal lobe areas of the human cerebral cortex. This marked TNAP-active band was localized infragranulary in layer 5 as defined by quantitative comparisons on parallel sections stained by various techniques to reveal the laminar pattern. On the contrary, TNAP activity was localized in layer 4 of the primary visual and somatosensory cortices, which is consistent with earlier observations on other species. This result suggests that the expression of TNAP in the thalamo-recipient granular layer is an evolutionary conserved feature of the sensory cortex. The observations of the present study also suggest that diverse neurocognitive functions share a common cerebral cortical mechanism depending on TNAP activity in layer 5. In summary, the present data point on the distinctive role of layer 5 in cortical computation and neurological disorders caused by TNAP dysfunctions in the human brain.

IntraceLlular responses to olfactory signals (three components and the combination of mixtures which they can provide) of 90 morphological identified local and output interneurones of the bee antennal lobe (the so-called Homo LIN, Hetero LIN, Pluri ON and Uni ON) were analysed and classified into eight main categories of response profiles. The different morphological types exhibit differences m (i) background activity; (ii) response patterns; and (iii) selectivity and responsiveness to pure odours and mixtures. The results demonstrate that the responses to mixtures are unpredictable from the responses to the single components. Olfactory integration includes inhibitory processes. Results of the present work considered together with those related to the morphological and spatial aspects demonstrate that the antennal lobe layer of the bee olfactory neuronal network is characterized by a complex architecture and connectivity underlying highly complex and somewhat unpredictable intracellular responses.

Wholemount analysis with optic and confocal microscopy of iontophoretic ally stained (cobalt or Lucifer Yellow) bee antennal lobe interneurones led us to identify and classify four main types of olfactory responsive neurones, differing in their morphology and spatial distribution within the glomerular space and amongst the glomeruli in the whole antennal lobe. Two types of local interneurones (LIN), restricted to the antennal lobe were: (i) Homo LIN presenting an apparently similar density of arbors among all the glomeruli where they branch, the fine neurites being limited to the core of the glomeruli; (ii) Hetero LIN, also pluriglomerular, differ from the Homo LIN by a high dense neurite arborization in one particular glomerulus with fine branches invading the core and reaching the outermost part of this glomerulus. Two types of output interneurones (ON) having dendrites in the antennal lobe and axons projecting in other brain parts were: (i) Uni ON with dendrites invading the core and periphery of only one glomerulus; (ii) Pluri ON, pluriglomerular, with dendrites limited to the glomerular cores. The Uni ON axons follow the median tract and project to the mushroom bodies and lateral lobe of the protocerebrum. On the contrary, axons of Pluri ON follow several subtracts of the antenno-glomerular tract and project to various protocerebal structures or to the contralateral antennal lobe. Local interneurones and output interneurones both invade many glomeruli (1/3 to 2/3 and >50% of the total number, respectively), most being distributed in the different glomerular populations. The high complexity demonstrated in the architecture of the bee antennal lobe neurones might support the high functional complexity described elsewhere (Fonta a al., 1991; Sun et al., submitted).

International audience; This paper describes the use of a new 3D high-resolution imaging technique dedicated to functional vessels for a systematic quantitative study of angiogenesis in the primate cortex. We present a new method which permits, using synchrotron X-ray micro-tomography imaging, the identification of micro-vascular components as well as their automatic numerical digitalization and extraction from very large 3D image analysis and post-treatments. This method is used to analyze various levels of micro-vascular organization and their postnatal modifications. Comparing newborn- and adult marmosets, we found an increase in vascular volume (270%), exchange surface (260%) and vessel length (290%) associated to a decrease in distances between vessel and tissue (32%). The increase in relative vascular volumes between the two ages, examined through the whole cortical depth, has been found to be mainly sustained by events occurring at the capillary level, and only marginally at the perforating vessel level. This work shows that the postnatal cortical maturation classically described in terms of synaptogenesis, gliogenesis and connectivity plasticity is accompanied by an intensive remodeling of micro-vascular patterns.

International audience; Cellular prion protein (PrP(c)) is a cell surface glycoprotein highly expressed in neurons, and a protease-resistant conformer of the protein accumulates in the brain parenchyma in prion diseases. In human prion diseases, visual cortex and visual function can be affected. We examined both the levels and the localization of PrP(c) in developing visual cortex of the common marmoset. Western blot analysis showed that PrP(c) increased from the day of birth through adulthood, and this increase correlated with the progression of synapse formation. Immunohistochemistry showed that PrP(c) was present in fiber tracts of the neonate, and this immunoreactivity was lost with maturation. Within the neuropil, the laminar distribution of PrP(c) changed with age. In the neonate, PrP(c) immunoreactivity was strongest in layer 1, where the earliest synapses form. At the end of the first postnatal week, layer 4C, as identified by its strong cytochrome oxidase activity, was noticeably lighter in terms of PrP(c) immunoreactivity than the adjacent layers. The contrast between the strong immunoreactivity in both supragranular and infragranular layers and weak immunoreactivity in layer 4C increased with age. Layers 2/3 and 5 contained more intense PrP(c) immunoreactivity; these layers receive thalamic input from the koniocellular division of the LGN, and these layers of the LGN also had strong PrP(c) immunoreactivity. Together, these results provide evidence for PrP(c) localization in an identified functional pathway and may shed some light on prion disease pathogenesis. J. Comp. Neurol. 504:646-658, 2007. A key molecular event in prion diseases is the conversion of cellular prion protein (PrP(c)) into an abnormal misfolded conformer (PrP(sc)). The PrP(c) N-terminal domain plays a central role in PrP(c) functions and in prion propagation. Because mammalian PrP(c) is found as a full-length and N-terminally truncated form, we examined the presence and amount of PrP(c) C-terminal fragment in the brain of different species. We found important variations between primates and rodents. In addition, our data show that the PrP(c) fragment is present in detergent-resistant raft domains, a membrane domain of critical importance for PrP(c) functions and its conversion into PrP(sc).

International audience; We have shown previously that the tissue nonspecific alkaline phosphatase (TNAP) is selectively expressed in the synaptic cleft of sensory cortical areas in adult mammals and, by using sensory deprivation, that TNAP activity depends on thalamocortical activity. We further analyzed this structural functional relationship by comparing the developmental pattern of TNAP activity to the maturation of the thalamocortical afferents in the primate brain (Callithrix jacchus). Cortical expression of alkaline phosphatase (AP) activity reflects the sequential maturation of the modality-specific sensory areas. Within the visual cortex, the regional and laminar distribution of AP correlates with the differential maturation of the magno- and parvocellular streams. AP activity, which is transiently expressed in the white matter, exhibits a complementary distributional pattern with myelin staining. Ultrastructural analysis revealed that AP activity is localized exclusively to the myelin-free axonal segments, including the node of Ranvier. It was also found that AP activity is gradually expressed in parallel with the maturation of synaptic contacts in the neuropile. These data suggest the involvement of AP, in addition to neurotransmitter synthesis previously suggested in the adult, in synaptic stabilization and in myelin pattern formation and put forward a role of AP in cortical plasticity and brain disorders.

We studied the relationship between vascularization and neuronal activity in the visual cortex during postnatal development in the primate. Analyses were focused on layer IVC that displays a sequential pattern of maturation for the magno- and parvocellular systems in separate sublayers, respectively IVC alpha and IVC beta. Cytochrome oxidase and endogenous alkaline phosphatase histochemistry was used to analyse, on the same sections, the laminar patterns of cortical activity and vessel density in the primary visual cortex of the marmoset (Callithrix jacchus). Experiments were carried out in five young and two adult animals. We showed that the temporal pattern of angiogenesis differs in layer IVC alpha and IVC beta. During the first postnatal month, vessel density is higher in IVC alpha than in IVC beta and runs parallel to cytochrome oxidase intensity. In 2-month-old animals, both vessel densities and cytochrome oxidase activity are similar in IVC alpha and IVC beta. In adults, the vessel densities in IVC alpha and IVC beta are the reverse of those observed during the first postnatal month. Vessel diameter does not account for this evolution in vascular patterns. In the discussion, we suggest that such a developmental time-course of angiogenesis might be linked to the synaptogenesis requirements that proceed differently for the magno- and parvocellular systems in the primate striate cortex.

In the marmoset Callithrix jacchus, ocular dominance columns (ODC) have been reported to be present in young animals, but absent in adults (Spatz, 1989). We have studied in juvenile and adult animals the postnatal organization of the retino-geniculo-cortical afferents by means of transneuronal labeling. We show in the present work that ODC are present in the primary visual cortex of Callithrix jacchus, both in the adult and in the juvenile animal. The present work confirms the presence of ODC in the visual cortex of juvenile marmoset before the end of the first postnatal month. In 2-month-old animals, ODC are well demarcated in IVcα and IVcβ. In the adult marmosets, the present data clearly show that the primary visual cortex is also organized with ODC. In horizontal sections, they form a mosaic through the ventral and dorsal calcarine cortex and through the dorso-lateral occipital part of the striate cortex. In frontal sections, their presence is manifest in IVcβ within the calcarine cortex and they only faintly appear in IVcα. These new findings are important since they underline the usefulness of the adult New World Monkeys as a model in visual research.

The effects of gentamicin administrated to gravid mice on the vestibular hair cells of their offspring were investigated. Scanning electron microscopy showed the effect to be dose-dependent and age-dependent, and there was a different sensitivity to toxicity for each of the three types of vestibular receptors. The destructive action of gentamicin on the vestibule before birth provides a mechanism to study the influence of the vestibular system on the central nervous system throughout its development.

This study was undertaken to examine the expression of the protooncogene c-fos in an invertebrate, the honeybee. We investigated first the expression of Fos-like proteins, testing different methodologies (immunohistochemistry, immunocytochemistry, immunoblotting). These studies were carried out at the embryonic, nymphal and adult stages. We found immunoreactive bands at approximately 30 kD and 57 kD. The first ones are revealed at the embryonic, nymphal and adult stages. The approximately 57 kD proteins are present at the adult stage. Beside this qualitative variation, intensive modification occurs with a burst of protein expression a few days after the insect emergence. The results suggest that Fos-like proteins are found in the developing and maturing nervous system. Basal level of expression is found in the olfactory pathway, convincing to explore the protooncogene c-fos expression in the cellular and molecular analyses of olfactory plasticity.

Nous avons entrepris des etudes multidisciplinaires destinees a elucider les mecanismes neuronaux et comportementaux responsables des capacites d'adaptation de l'abeille aux modifications des conditions environnementales, en nous basant sur l'etude des reponses de cet insecte a des signaux chimiques complexes (fig 1). Des etudes comportementales de la discrimination olfactive ont ete entreprises en utilisant pour stimulus des emissions volatiles florales. L'etude du comportement de butinage, conduite en conditions naturelles et au laboratoire, sur des plantes cultivees telles que le tournesol, associee a l'identification chimique des emissions florales, a montre que:―les preferences inter-genotypes, liees a la qualite du nectar, etaient etablies sur la base d'un nombre limite de constituants discriminatifs au sein des aromes floraux

Volatile chemicals are known to be essential factors that trigger or release different kinds of bumble- bees (Bombus sp.) behaviour, in intraspecific and interspecific relationships. Chemical analyses of the in- volved semio-chemicals are now in progress, and in parallel we have studied the nervous bases of identified odour recognition, for both males and females (queens and workers) of B. hypnorum and B. terrestns. The insects' antennae are provided with the sensory equipment for the detection of volatile compounds; the distribu- tion of placodea-type olfactory sensilla was studied by scanning electron microscopy. Responses of antennal sensory cells to floral and pheromonal components were recorded (EAG). Queens present the most numerous sensillar population and the highest amplitudes in EAG responses. These results might be linked to the polymor- phism of the body size and therefore to the antennal size of the three kinds of insects, the queens being the biggest in this insect population. Odour sensitivity depends on the molecular weight and the chemical func- tion of the tested molecules and appears to be higher for some floral compounds such as vanillin and pheromonal derivatives such as farnesol.

Electrophysiological data (EAG) were recorded on adult bumblebees stimulated with floral and/or pheromonal pure odorants at different concentrations. The responses of queen, worker, and male bees are compared and the sensitivities of these insects to the pure odorants tested are discussed.

Cypermethrin is highly toxic to the honeybee Apis mellifera ligustica. The action of the chemical is rapid (within 2 days) and it has no long-term effects. The sensitivity of the bees increases with decrease in breeding temperature and with increasing age of the insect. The commercial formulation ‘QCymbush’ is repellent to bees: the effect appears to be due to the formulation ingredients because cypermethrin itself is not repellent. A floral odour lost its natural attractiveness in the presence of ‘Cymbush’. The repellency appears to persist for some 2 days after treatment, during which time the bees learnt to avoid the crop. No residues of cypermethrin were found in the hive products (pollen, wax or honey), nor in the oilseed rape at harvest.
