Simple Summary: Since they have a unique phenotype, with an approximately 20% increase in muscle mass, we hypothesized that Belgian blue (BB) cows may have hematologic and biochemical characteristics that are significantly different from those of other cattle breeds. However, most available reference intervals in cattle are derived from dairy cows, mainly Holstein Friesian (HF) cows. Therefore, the aim of our study was to measure and compare biochemical and hematologic parameters in clinically healthy BB and HF cows. We studied 183 clinically healthy adult BB and HF cows. Our results confirmed that BB and HF represent different populations from a laboratory perspective. Therefore, we propose the first breed-specific reference intervals for BB, which are essential to improve the health management of this breed. Belgian blue (BB) cattle have an 11-bp deletion in myostatin that causes skeletal muscle hyperplasia and increased muscle mass, leading to a 'double-muscled' phenotype. Preliminary data suggest that this phenotype may be associated with breed-specific hematologic and biochemical values. Therefore, in this study, we sought to compare hematologic and serum biochemical parameters in healthy BB and Holstein Friesian (HF) cows and to propose breed-specific reference intervals for BB cows. Hematologic parameters, total protein, creatinine, creatine kinase (CK) and aspartate transaminase (AST) activities, albumin, and globulins were measured in 183 clinically healthy adult BB and HF cows. There were significant differences between BB and HF cows in 17 of 27 measured parameters. BB cows had significantly higher creatinine concentration and CK and AST activities (p < 0.001). RBCs, hemoglobin, hematocrit (p < 0.001), MCV and lymphocytes (p < 0.05) were also significantly higher in BB cows compared with HF cows. The average N/L ratio was greater than 1 in both breeds. These results suggest that BB and HF cows have significantly different clinically relevant hematologic and serum biochemical values, and, therefore, breed-specific reference intervals should be used. Eastern collared lizards (Crotaphytus collaris) are growing in popularity as exotic pets in the United States and worldwide. However, clinical biochemistry reference data to support the interpretation of health and disease for this species are lacking. This study evaluated 87 apparently healthy eastern collared lizards (C. collaris). Blood samples were collected from the ventral coccygeal vein and assayed via an Abaxis VetScan VS2 analyzer and avian/reptile profile plus rotor. Although lizard weight and snout–vent length (SVL) were normally distributed, most biochemical analytes, except albumin and total protein, were not. Many analytes were affected, albeit slightly, by lizard age, SVL, body condition score, gravidity and/or recent oviposition, sex, health status, and color and locality. However, except for calcium, phosphorous, and albumin for gravid or immediately postoviposition females, biochemical values remained within the generated reference interval. Limitations of this study included that the VetScan VS2 avian/reptile profile plus rotor was unable to successfully provide values for bile acids for most of these apparently healthy lizards. When compared to biochemistry analyte values of lizards from the suborder Iguania from the western hemisphere, eastern collared lizard biochemistries were similar for some analytes, but a relatively increased plasma glucose and uric acid occurs in this species, which could affect the diagnosis of clinical disease or other health abnormalities. Background: Wistar rats are extensively used as the model for assessing toxicity and efficacy in preclinical research. Hematological and biochemical laboratory data are essential for evaluating specific variations in the physiological and functional profile of a laboratory animal. Establishing hematological and biochemical reference values for Wistar (han) rats at various age intervals was the goal of this work. Male and female Wistar rats (n = 660) of ages 6–8 weeks, 10–14 weeks and > 6 months were used in the experiment. Blood and serum were collected from these rats under fasting conditions. Results: We observed that the majority of hematological and biochemical parameters were significantly influenced by sex and age. Hematological changes were significantly correlated to aging were increased red blood cells, hemoglobin, hematocrit, neutrophils, monocytes and eosinophils in both sexes, as well as decreased platelet, mean corpuscular volume, mean corpuscular hemoglobin and lymphocytes in both sexes. White blood cells of male rats were considerably higher than those of female rats in all age ranges. For biochemistry, increase in glucose, total protein and creatinine were seen in both sexes, along with increases in urea in females and alanine aminotransferase in males. Age was significantly associated with decreased alkaline phosphatase in both sexes. Conclusions: When using Wistar rats as a model, these reference values may be useful in evaluating the results. Intrahepatic cholestasis of pregnancy (ICP) is complicated by adverse fetal outcomes and even fetal death, the mechanism remains unclear. This study aims at evaluating the differential expression of mTORC2-AKT-IP3R signaling pathway, which accurately regulate Ca2+ transfer across mitochondria-associated membranes (MAMs) and determine the stress intensity experienced by endoplasmic reticulum (ER) and mitochondria, in patients diagnosed with ICP. We combined western blot analysis and placental immunofluorescence co-localization detection to assess the expression and co-localization of the mTORC2-AKT-IP3R signaling pathway in severe (maternal total bile acid (TBA) levels ≥40 μmol/L) and mild (maternal TBA 10–40 μmol/L) ICP. Compared with the control and mild ICP groups, phosphorylated protein kinase B (p-AKT) levels were significantly upregulated in the severe ICP group. Placental Rictor levels were lower in the mild ICP group than in the control group and were further downregulated in the severe ICP group. IP3R3 and p-IP3R3 levels were lower in placentas in the severe ICP group than in those in the mild ICP and control groups. Moreover, the co-localization of IP3R3 and p-AKT in patients in the mild and severe ICP groups was significantly elevated compared with that in patients in the control group. In patients with severe ICP, limited expression of Rictor and elevated p-AKT levels would suppress IP3R3/p-IP3R3 levels in MAMs. This inhibition might influence the transportation of Ca2+ from the ER to the mitochondria, thus weaken the stress adaptation associated with MAMs. Our results reveal the possible pathophysiological mechanism of adverse fetal outcomes in ICP. In the production of broiler poultry products, market operators use intensive technologies that involve the use of various, environmentally friendly nutraceuticals, in particular, probiotic preparations. In the technology of raising broiler chickens, probiotic preparations are necessary as a means for the prevention and treatment of gastrointestinal diseases, stimulating the growth and increasing the productivity of poultry. The purpose of the study is to investigate the effect of probiotic biologics on haematological parameters when it is fed to broiler chickens. The experiment used 20 broiler chickens, which were given a probiotic from 28 to 42 days in the following amounts: 0.5 g/10 dm3, 2.0 and 4.0 g/10 dm3 of water. It was found that the morpho-biochemical parameters of poultry blood corresponded to the physiological standards for broiler chickens of the established age and confirmed the absence of pathophysiological changes in their body. On days 35 and 42 of rearing, an increase in the number of leukocytes from 4.4 to 17.2% and the haemoglobin content by 3.9 and 6.2%, respectively, was observed in the blood of broiler chickens fed the probiotic at a dose of 4.0 g/10 dm3 compared to the control group. On day 35 of the study, a 1.2-fold increase in total serum protein was observed when drinking a probiotic in poultry, respectively, at doses of 2.0 and 4.0 g/10 dm3 of water, and on day 42 - 1.0 times for its drinking at a dose of 4.0 g/10 dm3 of water. It was found that the content of inorganic calcium and phosphorus in the poultry blood serum corresponded to physiological limits, which indicates a sufficient level of mineral nutrition in the body of broiler chickens. The absence of changes in the activity of alanine aminotransferase and aspartate aminotransferase, the content of total lipids, cholesterol, and creatinine in the blood serum of broiler chickens of the experimental groups indicates the hepato- and nephronethoxicity of the probiotic. Graves' disease (GD) is the most common cause of hyperthyroidism in children. A common GD symptom is a goiter. The usual biochemical profile in children with GD is a decreased thyroid hormone stimulating hormone (TSH) level and high free thyroxine (FT4) and free triiodothyronine (FT3) concentrations. The presence of thyroid receptor antibodies (TRAb) is the most important specific immunological sign for diagnosing GD. The treatment choices for pediatric GD are anti-thyroid drugs (ATDs), radioiodine, and thyroidectomy, but the risks and benefits of each modality are different. Management recommendations include the first-line use of a prolonged course of ATDs for at least 3 years and potentially 5 years or more. Rituximab and Teprotumumab are new novel alternative medications for the treatment of adult patients with GD and Graves' orbitopathy respectively, but evidence of the efficacy and safety of these drugs in pediatric patients with GD is lacking. Background: The elderly population in India is expected to increase to 319 million by 2050. Managing critically ill elderly patients in intensive care units (ICUs) is a difficult task. Proper planning and development of healthcare infrastructure are of prime importance to face this challenge. Objectives: To study the clinical profile and outcomes of elderly patients admitted to the medical ICUs. Materials and methods: A time-bound, prospective observational study on elderly patients admitted to medical ICUs for more than 48 hours was conducted from March 2019 to September 2020. The demographic, biochemical, hematologic, and microbiological data on antibiotic susceptibility patterns on various organisms and procalcitonin (PCT) reports were collected. Acute Physiology and Chronic Health Evaluation II (APACHE II) score was calculated. Various treatment modalities, such as mechanical ventilation, inotropes, hemodialysis, antibiotics, culture report in sepsis patients, and length of ICU stay were collected. Results: The age of the patients and the length of their ICU stay were not significantly associated with outcomes. Sepsis and APACHE II scores are significantly associated with outcomes. Receipt of mechanical ventilation, vasopressor support, and hemodialysis are significantly associated with mortality (p < 0.001). Conclusion: The patients' ages were not significantly associated with outcomes. The most common cause of death among elderly patients was found to be sepsis, followed by pneumonia. In elderly ICU patients, gram-negative organisms are the most common causative agents in bloodstream infections. Simple Summary: We explored the effect of age, gender, weight-for-height indices, sedation protocol, and housing conditions on the hematologic and serum biochemical values of captive rhesus and cynomolgus macaques. Several blood parameters demonstrated significant and clinically relevant changes in relation to the investigated variables. The results will provide veterinarians and researchers with important reference intervals for evaluating experimental results and health control from rhesus and cynomolgus macaques. Several physiological characteristics and housing conditions are known to affect hematologic and serum biochemical values in macaques. However, the studies that have been conducted either report values calculated based on a small number of animals, were designed specifically to document the effect of a particular condition on the normal range of hematologic and serum biochemical values, or used parametric assumptions to calculate hematologic and serum biochemical reference intervals. We conducted a retrospective longitudinal cohort study to estimate reference intervals for hematologic and serum biochemical values in clinically healthy macaques based on observed percentiles without parametric assumptions. Data were obtained as part of the Biomedical Primate Research Centre (Rijswijk, The Netherlands) health monitoring program between 2018 and 2021. In total, 4009 blood samples from 1475 macaques were analyzed with a maximum of one repeat per year per animal. Data were established by species, gender, age, weight-for-height indices, pregnancy, sedation protocol, and housing conditions. Most of the parameters profoundly affected just some hematologic and serum biochemical values. A significant glucose difference was observed between the ketamine and ketamine-medetomidine sedation protocols. The results emphasize the importance of establishing uniform experimental groups with validated animal husbandry and housing conditions to improve the reproducibility of the experiments. The increasing level of anthropogenic environmental pollution and effective means to reduce the negative impact of xenobiotics on animal and human health is an urgent problem today. Considering this, the purpose of the study is to examine the effect of heavy metals on accumulation processes under poisoning conditions, and biochemical parameters in the body of rats. Analogue groups were formed of rats of the same age, gender, and body weight to conduct the study. Rats were poisoned with solutions of copper sulfate, zinc sulfate, cadmium sulfate, and lead nitrate for 14 days. Using the method of infrared spectroscopy, substantial differences in the spatial structure of protein components in intact and poisoned animals were established. The difference between the spectral characteristics of the examined tissues is clearly demonstrated by the statistical indicators of skewness and kurtosis. It was determined that poisoning of rats with copper, zinc, cadmium, and lead ions affects the course of glycolysis reactions and the tricarboxylic acid cycle, which leads to a likely increase in serum concentrations of lactate and pyruvate, oxaloacetate and α-ketoglutarate and a decrease in Malate content compared to intact rats. It was established that under the conditions of poisoning, there is also a substantial increase (P < 0.05) in the content of the examined heavy metals in the blood, liver, and kidneys. In animals poisoned with heavy metals, a decrease in the pool of free amino acids in the kidneys is observed. In particular, the content of aspartic acid, valine, glycine, tyrosine, and cystine (more than 1.5 times) in the kidneys of such rats decreases; alanine, leucine, serine, taurine, threonine, phenylalanine (more than 2.0 times), lysine -- 3.4-4.9 times. Therewith, an increase in the level of isoleucine and methionine by 1.3-1.5 times, ornithine -- by 1.8-2.1 times, and glutamic acid -- by 4.4-5.3 times in rats of the experimental group compared to intact ones was identified. Background: SLC10A1 codes for the sodium taurocholate cotransporting polypeptide (NTCP). The SLC10A1S267F mutation is associated with loss of function of bile acid (BA) uptake and defined as a new type of hypercholanemia. This kind of hypercholanemia is characterized by high levels of serum BA. However, limited studies have been conducted on this topic. Objectives: This study aimed to describe the biochemical and bioinformatic characterization of patients with an SLC10A1S267F mutation, as well as to dissect pathogenesis in hypercholanemia. Methods: In this study, a total of 12 individuals (including 5 homozygous, 3 heterozygous, and 4 wild-type individuals) were recruited. Whole-genome sequencing (WGS) and Sanger sequencing were used to confirm the genotype. Tests of liver function, renal function, and serum lipid level, in addition to routine blood tests, were performed to evaluate the clinical consequences of patients with an SLC10A1S267F mutation. The ClinVar website and protein prediction tools were used to analyze other cholesterol and BAs related gene mutations in SLC10A1S267F patients, as well as to evaluate their possible effects on serum BA levels of patients. Results: All SLC10A1S267F homozygous patients displayed high levels of BAs. Liver and renal functions were generally normal. According to previous reports, homozygous patients are prone to vitamin D deficiency and deviated blood lipids. However, all homozygous individuals had normal levels of blood lipids, thyroid hormones, and vitamin D (25(OH)D). Moreover, except for the SLC10A1S267F mutation, according to the WGS results, multiple gene mutations were found in 5 homozygous and might affect the level of BAs, but the SLC10A1S267F mutation still is the most important reason resulting in a high level of BAs. Conclusions: This study provided a more detailed description of the SLC10A1S267F mutation-induced hypercholanemia, delivering a new idea that there might be some mutations in SLC10A1S267F homozygotes, probably influencing BA metabolism. Glucose meters have been developed for determining the hyperglycemic and hypoglycemic disorders as well as checking daily glucose levels for diabetic patients. Although multiple commercial glucometers have been presented, the attendance of interferences in the measuring of glucose is still a challenge. This problem can be managed using the proper design of potentiostat as the main part of glucometers. Here, we introduce a novel electrochemical circuit design comprising a microcontroller to specify the optimum zero point for the glucose measuring. The proper configuration of the potentiostat circuit leads to the correct detection of zero point for glucose. For this, the counter electrode (CE) is placed in high impedance mode, as well as the working electrode (WE) and reference electrode (RE) in the feedback path of the transimpedance amplifier circuit. The linear range of the proposed sensor was detected in the concentration ranges of 57–600 mg/dL with a linear equation of ${y}={0.0018} {x}-{0.0693}$ and a sensitivity of 0.0018. A fully integrated biochemical sensing platform is developed for blood diagnostics. The biochemical analyses of blood include the enzymatic reaction of the sample with a reagent that produces a colorimetric variation of solutions. The developed platform focuses on providing software solutions to the hardware dependencies in the existing analyzers by replacing the physical optical filter wheel assembly with software-based wavelength filtering. The experiments are performed to sense these variations with the developed image acquisition platform and the software-based wavelength filtering unit. The machine learning approach is used to train the system to predict unknown concentrations using the experimentally generated dataset. To verify the usability of the developed platform, the sensing of glucose, albumin, and cholesterol was carried out in the linear range from 10 to 400 mg/dL, from 1 to 8 g/dL, and from 10 to 800 mg/dL with a limit of detection (LOD) of 2.8 mg/dL, 0.64 g/dL, and 3.14 mg/dL, respectively. The results obtained are matching with the commercial biochemistry analyzer. The system is tested thoroughly, and the results show that the developed blood sensing platform is reliable, portable, and accurate due to the minimization of hardware moving parts, and offers extreme flexibility of operation due to software solutions, facilitating numerous blood analysis all-in-one instruments. Background: Diabetic nephropathy (DN) is one of the most important complications of type 2 diabetes (T2DM). Oxidative stress and inflammatory cytokines play an essential role in the development and progression of DN. Despite adopting appropriate therapies, many patients with DN progress to end-stage renal disease (ESRD). Therefore, exploring innovative strategies for better management of DN is crucial. Crocin, a natural compound found in saffron, has profound antioxidant, antifibrotic and anti-inflammatory properties. This study aimed to evaluate the therapeutic effects of crocin in attenuation of the progression of DN. Methods: In this randomized, triple-blind, placebo-controlled clinical trial, 44 patients with T2DM and microalbuminuria were randomly assigned to receive either crocin (15 mg/day) or a placebo for 90 days. Eventually, 40 patients completed the study: 21 patients in the crocin group and 19 in the placebo group. The primary outcome was a change in urine Albumin-to-Creatinine Ratio (uACR) from baseline to the end of the treatment period. We also evaluated metabolic, anthropometric, and biochemical parameters as the secondary outcomes. Results: The results of the present study showed that uACR increased in both groups, but the increment was not significantly higher in the crocin group compared with the placebo. Serum levels of transforming growth factor-β (TGF-β) decreased in the crocin group and increased in the placebo group, but none of these changes was significant. Crocin significantly reduced triglyceride (TG) as an important metabolic parameter (P-Value = 0.03). Conclusion: This study has shown that crocin may be a safe and potential adjunct to conventional therapies for DN patients but because of our limitations such as short duration of the treatment period, and prescribing low doses of crocin, we could not achieve the significant level. Growth hormone (GH) is secreted by cells in the anterior pituitary on two time scales: discrete pulses over minutes that occur within a 24-hr pattern. Secretion reflects the balance of stimulatory and inhibitory inputs from the hypothalamus and is influenced by gonadal steroids, stress, nutrition, and sleep/wake states. We propose a novel approach for the analysis of GH data and use this approach to quantify (i) the timing, amplitude and the number of GH pulses and (ii) GH infusion, clearance and basal secretion (i.e., time invariant) rates, using serum GH sampled every 10 minutes during an 8-hour sleep study in 18 adolescents. In our method, we approximate hormonal secretory events by deconvolving GH data via a two-step coordinate descent approach. The first step utilizes a sparse-recovery approach to estimate the timing and amplitude of GH secretory events. The second step estimates physiological parameters. Our method identifies the timing and amplitude of GH pulses and system parameters from experimental and simulated data, with a median ${R^2}$ R 2 of 0.93, among experimental data. Recovering GH pulses and model parameters using this approach may improve the quantification of GH parameters under different physiological and pathological conditions and the design and monitoring of interventions. Continuous glucose monitoring is of great significance in the diagnosis and treatment of diabetes. In this paper, a miniaturized and automatic optofluidic system was designed for continuous glucose monitoring. Interstitial fluid (ISF) was first obtained by an ISF transdermal extracting chip. Then, the glucose concentration in the ISF was detected by enzyme colorimetry. For accurate detection of the glucose concentration, the volume of transdermally extracted ISF was measured, and the volumes of ISF and colorimetric reagent in the mixed sample were controlled and mixed uniformly. To test the precision and stability of the system, different concentrations of glucose solutions (30–200 mg/dL) were used to simulate ISF extracted from skin. The results revealed that the measured glucose concentration correlated well with the reference concentration (R2 = 0.9976). In addition, the system achieved high-precision measurement of a low glucose concentration (30 mg/dL) even as the ISF volume fluctuated widely (0.5– $2.5 ~\mu \text{L}$), and the average absolute error was only 1.27%. Therefore, the system realized high precision for automatic and continuous monitoring of glucose in microvolume ISF samples, which would be a promising tool for the continuous monitoring of blood glucose concentration. Eimeria infection is one of the most common protozoal disease affecting sheep worldwide. This study was carried out to investigate the effect of Eimeria species (spp.) infection in native sheep breed at Sohag governorate. Total number of investigated sheep was 101. All sheep were examined clinically; blood and fecal samples were collected from each animal. Coprological examination was performed using the standard flotation technique. Eimeria oocysts were counted microscopically using the high power. Oocysts numbers were used to classify the degree of infection into mild and severe. Complete blood picture was performed using automatic blood cell counter. Levels of total protein, albumin, and globulin were determined in sera of infected sheep. In addition to the activities of liver enzymes (ALT, AST, ALP) and serum calcium level were measured using the spectrophotometer. Results of hematology and blood biochemistry were analyzed statistically to compare between mild and heavy infections. The total number of sheep infected with Eimeria spp. was 67/101 (66.4%). Infected sheep manifested variable clinical signs such as bloody and/or mucoid diarrhea, dehydration, lethargy, emaciation, fever and loss of wool. In this study, total erythrocytic count and hemoglobin concentration were significantly decreased in heavy Eimeria spp. infection. Total leucocytes count and granulocytes percentage were significantly elevated in mildly infected sheep. While, lymphocyte percentage was increased in case of heavy infection. Total proteins were significantly decreased in case of heavy infection. Albumin/globulin ratio was considerably altered. The liver enzymes, ALT and ALP were elevated in heavily infected sheep. AST was significantly higher when compared with mild cases. This study highlighted the risk of heavy Eimeriosis that can result in severe anemia and altered liver functions. Background: Rehmanniae Radix (RR), an herb with numerous pharmacological effects, is widely used in traditional Chinese medicine for the treatment of blood deficiency syndrome, either alone or in combination with other herbs. However, the mechanism by which processed Rehmanniae Radix (PRR) improves blood enrichment efficacy has not been clearly defined. Methods: Ultra-performance liquid chromatography coupled to quadrupole time-of-flight mass (UPLC-Q-TOF/MS) and biochemical methods were combined to explore the hematopoietic functional mechanisms of PRR on blood deficiency in a rat model, as well as the potential active ingredient for blood enrichment efficacy. The pharmacological effects of PRR were evaluated on a rat blood deficiency model induced by cyclophosphamide in combination with 1-acetyl-2-phenylhydrazine. The blood routine index, including white blood cell (WBC), red blood cell (RBC), and platelet (PLT) counts, as well as hemoglobin (HGB) level, and the changing metabolite profile based on urine and serum were assessed. Nontargeted metabolomic studies, combined with biochemical analyses, were employed to clarify pharmacological mechanisms. Results: PRR significantly increased the blood routine index levels and reversed the levels of SOD, GSH, and ATP. The PRR group was similar to the control group, as determined from the metabolic profile. All of the 60 biomarkers, representing the typical metabolic characteristics of the blood-deficient rat model, mainly involved energy metabolism dysfunction, the peripheral circulation system, and oxidative damage in the body. This improvement may be attributed to changes in polysaccharide and sixteen non-polysaccharide compounds in PRR, which were caused by processing RR with rice wine. There is an increasing significance for the breed, age, and sex-related variations in hematological and biochemical parameters in veterinary medicine. The primary objective of this study was to identify possible breed-related differences and to determine the effect of age and sex on hematologic and serum biochemical parameters in the Bosnian and Herzegovinian - Croatian Shepherd dog Tornjak raised in Bosnia and Herzegovina (B&H). Data from 19 hematological and 17 serum biochemical parameters were analyzed in the group of 100 dogs (47 females and 53 males), sexually intact and aged between one month and twelve years. Breed affected TP (total protein) and ALB (albumin) value. The median TP value was slightly above the reference interval and the ALB median value was in the upper limit. Variations in hematological and serum biochemical parameters were 6 sex-related and 23 age-related. Blood biochemistry of indigenous sheep breeds in Bauchi, Nigeria was assessed. Blood samples were collected from one hundred and twenty sheep comprising Yankasa, Balami, Uda (32 per breed) and West Africa Dwarf (24) at the Abubakar Tafawa Balewa University Teaching and Research Farm, Muda Lawal, Durum and Gwallaga markets. Audio comb serum auto-analyser was used for biochemical analyses. The results indicated that Uda had the highest values of sodium (136.34±0.59 mmol/L), urea (6.10±0.30 mmol/L), total protein (7.55±0.14 mmol/L), total cholesterol (4.12±0.25 mmol/L) and alkaline phosphate (109.97±3.01 mmol/L). Aspartate amino transferase (13.68±0.38 Iµ/L) was higher in female sheep than in their male counterparts while the males had higher glucose value (5.07±0.16 mmol/L). Sodium (135±0.85 mm/L), TP (7.08±0.13 g/dl) and AP (106.81±2.16 g/dl) were higher in the dry season while those of CRT (106.21±1.93 mmol/L) and ALB (4.52±0.34 g/dl) were higher in wet season. However, the values of Na+ (135.00±0.59 mmol/L) and GLC (5.17±0.20 mmol/L) were higher in adult than in young. The present research demonstrates a novel 1D photonic crystal (PhC) based reconfigurable biosensor pertaining to label-free detection of different concentrations of progesterone and estradiol, which play a vital role in developing reproductive hormones in women. The proposed sensor is designed by an alternative arrangement of Na3 AlF6 and CeO2, with a central defect layer. A thin layer of novel phase change chalcogenide material (Ge2 Sb2 Te5) is deposited along the two sides of the defect layer to improve the sensing performance. Numerical simulation of transmission spectrum for TE mode is carried out by using the transfer matrix method (TMM). The mainstay of this research is centered on the assay of shift in the defect mode position and intensity with respect to different concentrations of analyte, by changing the phase of the GST material from amorphous to crystalline. Interestingly, we observed a high tunability in defect mode wavelength, when the phase is changed from amorphous to crystalline, which leads to accomplishment of a high sensitivity of 1.75 nm/nmol/L for progesterone and 20.5 nm/nmol/L for estradiol. Aside from sensitivity, other significant parameters like figure of merit and detection limit are computed, which give a deep insight into the sensing performance. These encouraging sensing performances pave the path for efficient detection of different concentrations of progesterone and estradiol to monitor various gynecological problems in women. The morphological and biochemical characters and effect of a highly protein diet on Japanese quail were investigated. Total participants in this study were twenty adult male and eighteen female Japanese quail, they were at three different old-stages; 30-days, 90-day and 180-day old. They were offered highly-protein food and water for two weeks. After two weeks, the serum has been centrifugated, hence different indicators including the total protein were assessed. Kidneys were examined with a light microscopy, their masses were estimated. Data analysis was performed using SPSS-22. we observed a decrease in width of middle lobe of kidneys in 90-days old quail birds (22%) and 180-days old quail birds (18%). The perimeter of anterior lobe has been increasing as the age increases, whereas it was a decrease in the middle lobe (35%) and posterior lobe (31%) in 90-days old quail birds. As compared between sex-based groups, it has been shown increase in absolute value of kidney weight among 90-days old (23%) and in 90-days old (7%) female quail birds. Absolute mass has been found more in left kidney at 30-days old (16%) and lower in the right kidney in all age groups female quail birds than that of males. A decrease in length of three lobes of the kidneys was observed in 180-days old females of quail birds (20%), whereas it has been found significantly increase in the width of front lobe (11%) in females quail birds. There was an increase in the amount of general protein and level of albumin in females and males in all age groups. The globin was higher in males than was in females. When comparing the ratio of phosphorus and calcium, it was observed that both of them were higher in females than males. Alkaline phosphate and hemoglobin were elevated in males in 180-days old. The current study was planned to know the effect of breeding season on blood biochemicals of dromedary male camel in Pakistan. The clinically healthy male Marecha camels (n=5) from five to ten years old were investigated in non-breeding "August" and breeding "February" months. They were housed in half-opened pens, TMR, 6-8 hours browsing and twice-watering was performed. Blood biochemicals were studied by using standard kits through biochemistry analyzer. The glucose, cholesterol and triglyceride values (P<0.05) were found to be 136.68±5.46, 57.12±4.44, 35.18±3.26 and 142.62±4.38, 59.32±4.26, 36.97±3.63 mg/dl respectively in August and February, being elevated in rut-camels. The total protein, urea and creatinine concentrations varied non-significantly among animals. While the calcium and phosphorus concentration (P<0.05) was found to be 9.88±1.16, 4.77±0.8 and 9.06±1.18, 4.06±0.9 mg/dl respectively in August and February, lower in rut-camels. Such biochemical investigations are evident of the importance of serum bio-gram which has pronounced effect on sexual behavior and may be used for detection of rutting and non-rutting males. Objectives: Placental expression of neuropilin-1 (NRP1), a proangiogenic member of the vascular endothelial growth factor receptor family involved in sprouting angiogenesis, was recently discovered to be downregulated in pregnancies with fetal growth restriction (FGR) and abnormal umbilical artery (UA) Doppler. Soluble NRP1 (sNRP1) is an antagonist to NRP1; however, little is known about its role in normal and FGR pregnancies. This study tested the hypotheses that, first, sNRP1 would be detectable in maternal circulation and, second, its concentration would be upregulated in FGR pregnancies compared to those with normal fetal growth and this would correlate with the severity of the disease as assessed by UA Doppler.Methods: This was a prospective case-control pilot study of 40 singleton pregnancies (20 FGR cases and 20 uncomplicated controls) between 24 + 0 and 40 + 0 weeks' gestation followed in an academic perinatal center from January 2015 to May 2017. FGR was defined as an ultrasound-estimated fetal weight < 10th percentile for gestational age. The control group was matched to the FGR group for maternal age and gestational age at assessment. Fetal ultrasound biometry and UA Doppler were performed using standard protocols. Maternal plasma sNRP1 measurements were performed using a commercially available ELISA.Results: Contrary to the study hypothesis, maternal plasma sNRP1 levels were significantly decreased in FGR pregnancies as compared to those with normal fetal growth (137.4 ± 44.8 pg/mL vs 166.7 ± 36.9 pg/mL; P = 0.03). However, there was no significant difference in sNRP1 concentration between the control group and FGR pregnancies that had normal UA Doppler. Plasma sNRP1 was downregulated in FGR pregnancies with elevated UA systolic/diastolic ratio (P = 0.023) and those with UA absent or reversed end-diastolic flow (P = 0.005) in comparison to FGR pregnancies with normal UA Doppler. This suggests that biometrically small fetuses without hemodynamic compromise are small-for-gestational age rather than FGR.Conclusions: This study demonstrated a significant decrease in maternal plasma sNRP1 concentration in growth-restricted pregnancies with fetoplacental circulatory compromise. Hematological and biochemical blood parameters indicate both the degree of adaptation of the body of athletes to physical activity and the imbalance between body's adaptation capabilities and the loads imposed. This study investigates the balance of hematological and biochemical blood indicators in professional wrestlers (sambo, freestyle wrestling, and Greco-Roman wrestling). Sambo wrestlers were characterized by the highest counts of peripheral blood leukocytes. All wrestlers had higher counts of monocytes and granulocytes compared to the control group. Athletes specializing in freestyle and Greco-Roman wrestling had the highest blood hemoglobin values due to the increased average hemoglobin content in erythrocyte. Subjects of the control group had the smallest platelet count. The groups also exhibited differences in the shape and size of blood cells. The groups did not differ in the blood contents of transferases (ALT and ACT), while all athletes had higher levels of alkaline phosphatase. Wrestlers were characterized by a higher content of blood triglycerides, while cholesterol levels did not differ significantly between the groups. Sambo wrestlers had the highest level of creatinine. Testosterone content in athletes was lower than in the control group; this decrease was the least in freestyle wrestlers. The contents of cortisol, Т4, and TSH in athletes were higher than in the control group. At the same time, wrestlers had reduced levels of Т3, testosterone, and vitamin D. The present experiment was carried out to investigate the effect of different percentages (50 and 100%) of dried whey powder and sun flower meal treated with blood or formaldehyde on Biochemical blood parameters: Blood Glucose (BG), Blood Urea Nitrogen (BUN), Blood Cholesterol (BCH), Blood Total Protein (BTP), Blood Albumin (ALB) and Blood Globulin (GLO) in lambs fattening diets. Clostridium chauvoei, causing myonecrosis in livestock animals, lives in the feces, surface water and soil. The blackleg due to C. chauvoei is very common in dairy animals. Still, no report is available about the pathophysiology of disease in Cholistani cattle kept under tropical and desert conditions of Cholistan, Pakistan. Therefore, in this study, we report for the first time the pathophysiology of a visceral form of blackleg infection in indigenous cholistani cattle breed reared in desert conditions of Cholistan. Clinically morbid animals exhibited different signs of infection including fever, crepitation sounds, gaseous swelling and edematous lesions. Blood was collected for hematological and serum biochemical investigation. Hematological examination indicated a significant increased erythrocyte sedimentation rate, lower red blood cells, hematocrit, and total white blood cell count. Results on serum biochemistry showed significantly (p<0.02) increased creatine kinase levels, creatinine, urea, alanine aminotransferase, aspartate aminotransferase, alkaline phosphates, creatinine phosphokinase and lactate dehydrogenase in infected cattle. At necropsy level, light pink color fluid under swelling areas of skin, gas bubbles, dark to black in color of affected muscles and crepitation sounds at palpation were observed. Necropsy showed marked myocarditis, petechial hemorrhages, consolidation and severe pulmonary edema. Spleen showed petechial hemorrhages and congestion. Histological analysis of muscular tissues indicated severe inflammatory reaction comprising of cellular infiltration, marked edema, necrosis and disruption of myofibrils. PCR confirmed the presence of C. chauvoei in muscles, heart and exudates of the lungs. This is the first report on molecular detection of Clostridium chauvoei from a visceral form of Blackleg disease in cholistani breed of cattle naturally infected in the Cholistan desert of Punjab, Pakistan. Background: Comprehensive hematology and biochemistry RIs are currently lacking in the literature for young dairy calves based on sample sizes more than 120. Young dairy calves are at a relatively high risk of poor health and welfare outcomes. They have a high risk of morbidity and mortality in the first 2 weeks of life, and many are transported and fasted during this time. For example, non‐replacement calves in Australia and New Zealand are usually 5‐12 days old when transported to abattoirs, meaning that calves of this age group are potentially at risk of both health and welfare compromise. Given these factors, sound, comprehensive, age‐specific biochemical and hematologic RIs are needed for both veterinary clinical practice and to inform research on calf health and welfare. Objectives: The aim of this study was to generate age‐specific hematology and biochemistry RIs for dairy calves aged 5‐12 days. Methods: We collected blood samples from 141 fasted, healthy dairy calves on 10 Australian farms. Reference Value Advisor software was used to calculate nonparametric RIs for multiple biochemistry and hematology variables. Results: RIs for a panel of hematology and biochemistry variables in dairy calves aged 5‐12 days old were derived. The determination of hematological values is used to obtain knowledge about the health conditions of animal species. The bigheaded Amazon River turtles, (Peltocephalus dumerilianus) are considered one of the least known testudine species concerning their biology and health status. Herein, we determined the hematological and plasma biochemical parameters of 17 (eight males and nine females) adult P. dumerilianus to provide reference interval values for clinically healthy individuals. We collected the blood samples by puncturing the femoral vein using long heparinized hypodermic syringes. Sexual dimorphism for individuals was determined by external observation of the shape of the plastron. The average values obtained for the ten hematological and biochemical parameters analyzed were red blood cell count = 0.32 million µL-1; hematocrit = 20.6 %; hemoglobin = 8.5 g dL-1; mean corpuscular volume = 681.6 fL; mean corpuscular hemoglobin = 267.8 pg; mean corpuscular hemoglobin concentration = 41.9 g dL-1; glucose = 80.6 mg dL-1, total protein = 4.1 g dL-1, triglycerides = 388.9 mg dL-1, and total cholesterol = 79.3 mg dL-1. Despite the sexual dimorphism evidenced for the species, there was no significant statistical difference between males and females for both hematological and biochemical parameters analyzed herein. Based on these results, the population is considered healthy, with parameter values coinciding with previously reported reference ranges for testudines species in the region. The results obtained in this study can be used for assessing the health status of other Amazonian turtle populations, especially in actions aimed at cultivation strategies, management, and species conservation. The excellent characteristics of nanocellulose fiber-based papers have made them one of the most attractive and innovative materials for the development of electrochemical paper-based analytical devices (ePADs). Here, the authors describe a new ePAD based on cellulose nanofibers (CNs) for the determination of glucose concentration from whole blood samples. Cellulose acetate (CA) nanofibers were prepared by the electrospinning method. Then, the paper layer regenerated to cellulose by deacetylation in alkaline solution. To obtain a smooth and continuous CNs layer, it was treated with trimethyl chitosan (TMC). The formation of CA layer, its alkaline regeneration and TMC modification was confirmed by atomic force microscopy, scanning electron microscopy, Fourier-transform infrared spectroscopy, and energy dispersiveX-ray. Screen-printed three electrodes (SPEs) were fabricated by sputtering a thick layer of gold (Au) on the TMC/CNs substrate. Subsequently, reduced graphene oxide (rGO) was used to modify the surface of the working electrode. The fabrication steps of rGO-Au-SPE were characterized using cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and scanning electron microscop. The results showed that design, construction and modification process was well achieved. Next, glucose oxidase is successfully immobilized onto the regenerated CNs layer which has been previously located in the sensing area of the rGO-Au-SPEs. The ePAD glucose assay exhibited high sensitivity of $9.9\times 10^{-4}~\text{K}\Omega ^{-1} \cdot \text{mM} ^{-1}$ to glucose in the range 3.3 – 27.7 mM ($\text{R}^{2} =0.99$) with a detection limit of 0.1 mM, high reproducibility (RSD = 0.57-1.59%) and outstanding selectivity as well as stability. Background and Aim: Salmonella typhi infection occurs in most endemic areas. Patients suspected of typhoid fever located in the Badda and Shantinagar localities of Dhaka were the studied population. A total of 107 blood specimens were collected. Objective: This study attempted to determine the antibiotic susceptibility pattern of Salmonella typhi among different age and gender groups having clinical symptoms of typhoid fever. Methods: The microorganisms obtained from blood specimens were identified by phenotypical and biochemical tests following standard protocols. Antibiotic susceptibility test was done by the Agar-disc-diffusion method to understand antibiotic susceptibility of the isolates. All the Salmonella typhi isolates were serologically confirmed using a commercial antisera kit. Results and Conclusion: Out of 107, 74 Salmonella typhi isolates showed multidrug-resistant properties. Nalidixic acid, ampicillin, azithromycin, and ciprofloxacin showed high antibiotic resistance percentage. On the other hand meropenem, amikacin, gentamycin showed low antibiotic resistance. Age and sex groups also revealed significant attributes to the multidrug-resistant isolates. Multidrug-resistant pattern of the clinical isolates of Salmonella typhi of the current study has presented a matter of great public health concern in Dhaka city, Bangladesh. The first line of antibiotics can be prescribed, taking into consideration of age and gender. The sources of Salmonella typhi infection and mode of transmission must be identified. Resistin, a novel adipokine may play an important role in bone metabolism. The study is designed to discover the association of bone mineral density (BMD) with serum resistin levels, anthropometric measures and to elucidate serum resistin as a predictor of BMD in postmenopausal women. Postmenopausal women (n = 160) were recruited and divided into two groups, non-osteoporotic (n = 70) and osteoporotic (n = 90). BMD was evaluated by DXA scan. High serum resistin levels and low weight are independent contributors to low BMD and can influence BMD at lumbar spine, right femoral neck, right hip, left femoral neck, and left hip in postmenopausal women. Background: Premature ovarian insufficiency (POI) is a delicate medical problem in young women. This condition is not unchangeable and permanent but is associated with intermittent and unpredictable ovarian activity, resulting in low conception rate. Over the period of 8 years, the evaluation of secondary amenorrhea was conducted in 90 patients below the age of 40 who wished to restore fertility and is characterized by irregular or absent periods and reduced fertility. Symptoms include those associated with the natural menopause (night sweats and vaginal dryness), and with the long-term adverse effects of estrogen deficiency (osteoporosis and cardiovascular disease): The latter is believed to explain the shorter life expectancy associated with POI. Aim: To illustrate the predictive value of progesterone receptors (PRs), estrogen receptors (ERs), progesterone (PRG), estradiol (E2), Lulilizing hormones (LH), Follicle stimulating hormones (FSH) and Anti mullerian hormones (AMH) serum levels as diagnostic approach for POI. Method: Sixty (60) women with idiopathic POF with thirty (30) women as control groups were included in the study. Baseline investigation in all subjects included serum progesterone receptors(PRs), estrogen receptors(ERs), progesterone (PRG), estradiol (E2), Lulilizing hormones (LH), Follicle stimulating hormones (FSH) and Anti mullerian hormones (AMH) levels were estimated using appropriate assays in biochemistry laboratory. Results: There was statistically significant differences in the serum progesterone receptors, estrogen receptors, progesterone, estradiol, LH, FSH and AMH in POI cases when compared with healthy control (P=<0.05). Conclusion: These results indicate that serum progesterone and estrogen receptors levels, is high in POI cases as a result of lack progesterone and estradiol hormones as a result of loss negative feedback inhibition, this result may be used to primary diagnosis of premature ovarian failure. Context: Metabolic differences between ectopic fat depots may provide novel insights to obesity-related diseases.Objective: To investigate the plasma metabolomic profiles in relation to visceral adipose tissue (VAT) volume and liver and pancreas fat percentages.Design: Cross-sectional.Setting: Multicenter at academic research laboratories.Patients: Magnetic resonance imaging (MRI) was used to assess VAT volume, the percentage of fat in the liver and pancreas (proton density fat fraction [PDFF]) at baseline in 310 individuals with a body mass index ≥ 25 kg/m2 and with serum triglycerides ≥ 1.7 mmol/l and/or type 2 diabetes screened for inclusion in the 2 effect of omega-3 carboxylic acid on liver fat content studies.Intervention: None.Main Outcome Measure: Metabolomic profiling with mass spectroscopy enabled the determination of 1063 plasma metabolites.Results: Thirty metabolites were associated with VAT volume, 31 with liver PDFF, and 2 with pancreas PDFF when adjusting for age, sex, total body fat mass, and fasting glucose. Liver PDFF and VAT shared 4 metabolites, while the 2 metabolites related to pancreas PDFF were unique. The top metabolites associated with liver PDFF were palmitoyl-palmitoleoyl-GPC (16:0/16:1), dihydrosphingomyelin (d18:0/22:0), and betaine. The addition of these metabolites to the Liver Fat Score improved C-statistics significantly (from 0.776 to 0.861, P = 0.0004), regarding discrimination of liver steatosis.Conclusion: Liver PDFF and VAT adipose tissue shared several metabolic associations, while those were not shared with pancreatic PDFF, indicating partly distinct metabolic profiles associated with different ectopic fat depots. Tissue plasminogen activator (tPA) a thrombolytic agent is commonly used for digesting the blood clot. tPA half‐life is low (4–6 min) and its administration needs a prolonged continuous infusion. Improving tPA half‐life could reduce enzyme dosage and enhance patient compliance. Nano‐carries could be used as delivery systems for the protection of enzymes physically, enhancing half‐life and increasing the stability of them. In this study, chitosan (CS) and polyethylene glycol (PEG) were used for the preparation of CS‐g‐PEG/tPA nanoparticles (NPs) via the ion gelation method. Particles' size and loading capacity were optimised by central composite design. Then, NPs cytotoxicity, release profile, enzyme activity and in vivo half‐life and coagulation time were investigated. The results showed that NPs does not have significant cytotoxicity. Release study revealed that a burst effect happened in the first 5 min and resulted in releasing 30% of tPA. Loading tPA in NPs could decrease 25% of its activity but the half‐life of it increases in comparison to free tPA in vivo. Also, blood coagulation time has significantly affected (p ‐value = 0.041) by encapsulated tPA in comparison to free tPA. So, CS‐g‐PEG/tPA could increase enzyme half‐life during the time and could be used as a non‐toxic candidate delivery system for tPA. [ABSTRACT FROM AUTHOR]

The therapeutics for type 2 diabetes mellitus has emerged in the current century towards nanomedicine incorporated with plant active compounds. In this study, Tinospora cordifolia loaded poly (D, L‐lactide) (PLA) nanoparticles (NPs) were evaluated in vivo for their anti‐hyperglycemic potency towards streptozotocin‐induced type 2 diabetic rats. T. cordifolia loaded PLA NPs were synthesised by the double solvent evaporation method using PLA polymer. The NPs were then characterised and administrated orally for 28 successive days to streptozotocin‐induced diabetic rats. The PLA NPs had significant anti‐diabetic effects which were equal to the existing anti‐diabetic drug glibenclamide. Sulphonylureas are extensively used in the treatment of type II diabetes; however, these drugs have complications of hypoglycaemia and weight gain. The current study aims at developing a potent antidiabetic drug that has lesser side effects and better management of its associated conditions. Zinc oxide nanoparticles (ZnO NPs) were synthesised using Syzygium cumini seed extract with an average size of 18.92 nm. In vitro studies on rat insulinoma (RIN‐5F) cells revealed that cells treated with synthesised ZnO NPs showed a dose‐dependent increase in insulin secretion. Streptozotocin‐fructose‐induced type II diabetic rats treated with ZnO NPS exhibited a significant reduction (p < 0.01) in the blood glucose levels, total cholesterol, triglycerides, and low‐density lipoprotein levels and increase (p < 0.01) in serum insulin and liver antioxidant enzyme levels proclaiming its role as a hypoglycaemic and hypolipidaemic drug. Treatment of ZnO NPs in diabetic rats exhibited an increased number of beta cells which was responsible for its increased insulin levels and reduced glucose levels. From the overall observations, biosynthesised ZnO NPs exhibited an efficacious hypoglycaemic effect in diabetic rats, so it can be suggested as a potent antidiabetic drug. [ABSTRACT FROM AUTHOR]

This study aimed to determine the influence of body condition score (BCS) on the metabolic and reproductive status of adult female Kacang goats (Capra hircus). Twenty-three local doe goats aged 3-6 years, which were not pregnant and had BCS ranging from 1-3 on a scale of 1-5, were studied. Blood samples were collected before slaughtering for biochemical analyses. After the goats were killed, the ovaries were collected for follicle and corpus luteum (CL) examination. Ovarian follicular fluid was aspirated for biochemical analyses. Most blood biochemistry parameters of the goats with BCS 1 -3 did not show significant differences (P>0.05). The ovaries with BCS 1 were acyclic; whereas the ovaries with BCSs 2 and 3 were cyclic. The ovarian follicular fluid concentrations of glucose, Ca, and Mg in the goats with BCS 1 were significantly lower than those of the goats with BCSs 2 and 3. The levels of TP, ALB, and cholesterol in the ovarian follicular fluid were significantly correlated with the blood results (P<0.05). In conclusion, Kacang goats with poor body conditions showed ovarian dysfunction. The levels of TP, ALB, and cholesterol in follicular fluid were related to the blood metabolite levels.
