The present study discussed the optimization of the ultrasonic‐assisted extraction of polysaccharides from daylily polysaccharides (DPs). The extracted crude polysaccharides were further separated and purified, and the antioxidant activities including 1,1‐diphenyl‐2‐111 picrylhydrazyl (DPPH) radical scavenging, 2,2'‐azinobis‐(3‐ethylbenzthiazoline‐6‐sulphonate) (ABTS) radical scavenging, hydroxyl radical scavenging, and ferric‐reducing antioxidant power (FRAP) activities of the obtained fractions were also evaluated. The results showed that the optimal ultrasonic‐assisted extraction parameters with DPs yield of 15.25 ± 1.13% were water to powder ratio of 25 ml/g, extraction power of 694 W, extraction temperature of 71°C, extraction time of 38 min, and three times extraction. By DEAE Sepharose Fast Flow column, four water‐soluble polysaccharide fractions (DP‐1, DP‐2, DP‐3, and DP‐4) were successfully obtained. Monosaccharide component analysis showed that the four obtained fractions were all hetero‐polysaccharides that mainly contained rhamnose, arabinose, fructose, galactose, glucose, galacturonic acid, and glucuronic acid in different molar ratios. All the four DP fractions did show obvious antioxidant activities in vitro, and the DP‐3 component had relatively high ABTS free radical scavenging activity. Overall, our research showed that DPs could provide cheap raw materials for the development of natural antioxidants in medicines, functional foods, and even cosmetics. The results showed that the optimal ultrasonic‐assisted extraction yield of DPs was 15.25 ± 1.13%. In the present study, purification and characterization of enzymatic hydrolysates of polysaccharide from Enteromorpha prolifera (HPE) are described. HPE was sequentially purified by DEAE Cellulose-52 chromatography and Sephadex G-100 chromatography to afford three fractions, namely, PHPE1, PHPE2, and PHPE3. Molecular weights of these three fractions were measured to be 103, 45.4, and 9.8 kDa, respectively, using high performance gel permeation chromatography (HPGPC). The three fractions were evaluated for their antioxidant activities by determining their ability to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, and superoxide anion radicals. The leaf of Magnolia officinalis is abundant in natural resources as a novel potential antioxidant and has great commercial value. In this study, the contents of total phenolics, total flavonoids, magnolol and honokiol in different solvent extracts (acetone, methanol and water) were measured. Meanwhile, the antioxidant activity of extracts from M. officinalis leaves were evaluated using 1,1-diphenyl-2-picrylhydrazyl free radical scavenging assay, 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt free radical scavenging assay, FRAP assays and β-carotene/linoleic acid system. In addition, the correlation between the antioxidant activity and the contents of the active compounds was analyzed. Experimental results indicated that three extracts exhibited antioxidant effects in a concentration-dependent manner, and acetone extracts had the highest antioxidant activity, next methanol and water. There was an intimate relativity between the contents of magnolol and honokiol in different solvent extracts and the antioxidant capability of them. So the magnolol and honokiol of M. officinalis leaves as a representative of the active compounds were extracted and purified by using the macroporous resin column. The purity of depurated sample could reach 82.6 %, which indicates the M. officinalis leaves have the potential to substitute the production of magnolol and honokiol in M. officinalis. The results of this study can provide theoretical and technical references for the reasonable use of the M. officinalis leaves and alleviate the shortage of M. officinalis resources. Abstract: The crude polysaccharides (GFP) were isolated from the fruiting bodies of Grifola frondosa and purified by DEAE cellulose-52 chromatography and Sephadex G-100 size-exclusion chromatography in that order. Three main fractions, GFP-1, GFP-2 and GFP-3, were obtained through the isolation and purification steps. Then the antioxidant activities of these three fractions were investigated in vitro. The results showed that GFP-1, GFP-2 and GFP-3 possessed significant inhibitory effects on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical and superoxide radical; their reducing power, ferrous ions chelating effect and the inhibition ability of the rat liver lipid oxidation where also strong. These results suggest that G. frondosa polysaccharides could be a suitable natural antioxidant and may be the functional foods for humans. [Copyright &y& Elsevier]

Abstract: A water-soluble polysaccharide (FUP-1) was obtained from Fritillaria ussuriensis Maxim through warm water extraction, ethanol precipitation, anion-exchange and gel-permeation chromatography. Structural analysis of this compound revealed that the average molecular weight of FUP-1 was 4.1×104 Da, and the major monosaccharide component was xylose, followed by glucose and galactose, at low levels. In vitro antioxidant assays showed that FUP-1 exhibited strong hydroxyl, superoxide anion and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activities. During in vivo antioxidant testing, the administration of FUP-1 could largely attenuate the increase in d-gal-induced malondialdehyde (MDA) and renew the activities of superoxide dismutase (SOD) in serums as well as glutathione peroxidase (GSH-Px) and the total antioxidant capacity (T-AOC) in the livers of d-gal-treated mice. Overall, the results of the present study indicate that FUP-1 exerts antioxidant activity not only through its own radical-scavenging activity but also by boosting the enzymatic and nonenzymatic antioxidant defense system of the host. Abstract: A water-soluble polysaccharide obtained from Acanthopanax senticosus leaves (ASL), was fractionated by DEAE–Sepharose fast-flow column chromatography, and purified by Sephadex G-75 gel-permeation column chromatography. The characteristics of ASP-2-1 were determined by chemical analysis, high-performance capillary electrophoresis (HPCE), high-performance gel-permeation chromatography (HPGPC). The results show that ASP-2-1 contained 89.47% carbohydrate, 7.45% uronic acid, 2.16% protein and seven kinds of monosaccharides including rhamnose, xylose, glucose, mannose, arabinose, galactose and glucuronic acid in a molar ratio of 7.45:18.63:25.15:0.93:8.35:2.79:5.69, with an average molecular weight of about 14,573Da. Furthermore, the immunobiological and antioxidant activities, in vitro, of ASP-2-1 were evaluated by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay and ferric-reducing antioxidant power assay (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH ), superoxide radical and hydroxyl radical ( OH) free radical-scavenging assay, respectively. The results showed that ASP-2-1 exhibited significantly higher immunomodulatory activities against the lymphocyte proliferation in vitro, pronounced reductive power (FRAP value: 785.1μM at 0.2mg/ml), strong hydroxyl radical (89.56% at 1mg/ml) scavenging activity, moderate superoxide radicals (65.32% at 1mg/ml) and DPPH radicals (68.9% at 1mg/ml) scavenging activities. Abstract: Four water-soluble polysaccharides fractions (ZSP1b, ZSP2, ZSP3c and ZSP4b) were isolated and purified from the fruiting bodies of Zizyphus Jujuba cv. Jinsixiaozao by DEAE-SepharoseCL-6B and SepharoseCL-6B column chromatography. Their chemical and physical characteristics were determined by chemical methods, gas chromatography (GC) and high-performance size-exclusion chromatography (HPSEC). Antioxidant activities of crude polysaccharide from the fruiting bodies of Zizyphus Jujuba cv. Jinsixiaozao (CZSP) and its purified fractions were investigated including superoxide anion, 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl radical-scavenging activity and reducing power. Antioxidant results showed that ZSP3c and ZSP4b containing more uronic acid had the stronger free radical scavenging activities than ZSP1b containing no uronic acid. Abstract: A new antioxidation protein named G4b was purified from Ginkgo biloba seed albumin. The results of chemical colorimetry and chemiluminescence methods showed that the hemi-inhibitory concentration (IC50) of G4b on the hydroxyl radical were 78.1μg/mL and 115.1μg/mL, and the IC50 on DPPH was 100.7μg/mL, while the IC50 on the superoxide anion and DNA injury were 23μg/mL and 27μg/mL, respectively, and the abundance of sulfur amino acids and aromatic amino acids contained in G4b might contribute to its strong antioxidation properties. G4b is a kind of homogeneous and novel protein with a molecular mass of 29,247Da, consisting of two peptides with similar molecular weights linked by a disulfide bond. Abstract: Three polysaccharide fractions (DDP1-1, DDP2-1 and DDP3-1) were successfully purified from the crude polysaccharide of Dendrobium denneanum by DEAE-Cellulose and Sephadex G-200 column chromatography. The average molecular weights (Mws) of these fractions were 51.5, 26.1 and 6.95kDa, respectively. Monosaccharide components analysis indicated that DDP1-1 and DDP2-1 were composed of arabinose, xylose, mannose, glucose and galactose in a molar ratio of 1.00:2.82:57.11:140.82:7.76 and 1.00:1.62:1.18:77.5:7.79. DDP3-1 was composed of arabinose, mannose, glucose and galactose in a molar ratio of 1.00:1.03:8.84:2.00. Abstract: Water-soluble salvianolic acid A (Sal A) and salvianolic acid B (Sal B) were successfully isolated and purified from the crude extract of Salvia miltiorrhiza by high-speed counter-current chromatography (HSCCC). The solvent system was n-hexane–ethyl acetate–methanol–water (3:6:6:10, v/v/v/v). 4.27mg of Sal A and 32.09mg of Sal B were obtained from 260mg of the crude sample. The purities of Sal A and Sal B were 96.67% and 97.43%, respectively. Their structures were identified by 1H NMR and 13C NMR. Antioxidant activities of Sal A and Sal B were also evaluated and compared by the methods of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS +) radical cation decolourisation assay. Both Sal A and Sal B showed high radical scavenging activities with their EC50 values being 1.43±0.09 and 1.81±0.01μg/ml in DPPH radical method. The ABTS results showed that Sal A and Sal B exhibited high total antioxidant activities, their EC50 values were 1.35±0.00 and 1.43±0.01μg/ml, respectively. [Copyright &y& Elsevier]

The water-soluble crude polysaccharide GM, obtained from the Gynostemma pentaphyllum Makino by boiling-water extraction and ethanol precipitation, was fractionated by DEAE–Sepharose CL-6B column chromatography, and purified by Sephadex G-100 column chromatography, giving three polysaccharide fractions termed GMA, GMB and GMC. The monosaccharide components of them were studied by PC and GC. On the basis of superoxide radical assay, hydroxyl radical assay and self-oxidation of 1,2,3-phentriol assay, the antioxidant activities of GM, GMA, GMB and GMC were investigated. Among these contents, GMC had the higher scavenging effects on superoxide radicals and inhibitory effects on self-oxidation of 1,2,3-phentriol, and so should be explored as a novel potential antioxidant.
