ExbBD is part of a cytoplasmic membrane molecular motor driven by the proton-motive force. It belongs to the larger family of motors involved in nutriment import across the outer membrane of Gram-negative bacteria (ExbBD), flagellar rotation (MotAB) or late steps of cell division in Gram-negative bacteria (TolQR). ExbB and ExbD are integral membrane proteins with three (ExbB) or one (ExbD) transmembrane segment. Here we have solved by single-particle cryo-EM the structures of ExbB alone and of the ExbB-ExbD complex of the opportunistic pathogen Serratia marcescens. ExbBSm alone behaves as a stable pentamer, and the complex displays the ExbB5-ExbD2 stoichiometry. This is similar to what has been observed for ExbB-ExbD complexes from Escherichia coli and Pseudomonas savastanoi as well as MotAB complexes from various species. We identified residues located in the first TM of ExbBSm and ExbBEc that are likely involved in the interaction with TonB/HasB and that are essential for function. ExbBSm has a ca. 40 residues long periplasmic extension absent in E. coli. Such long ExbBs are found in some Gammaproteobacteria, and several genera of Alphaproteobacteria. We show that this extension interacts with HasB, a dedicated TonB paralog from the heme acquisition system (Has) from S. marcescens. We also show that it is involved in heme acquisition via the Has system from S. marcescens. ExbBSm represents thus a new class of ExbB protein and our results shed light on the specificity determinants between the ExbB-ExbD complex and their associated TonB partners.

The Shigella effector IpaA co-opts the focal adhesion protein vinculin to promote bacterial invasion. Here, we show that IpaA triggers an unreported mode of vinculin activation through the cooperative binding of its three vinculin-binding sites (VBSs) leading to vinculin oligomerization via its D1 and D2 head subdomains and highly stable adhesions resisting actin relaxing drugs. Using cross-linking mass spectrometry, we found that while IpaA VBSs1-2 bound to D1, IpaA VBS3 interacted with D2, a subdomain masked to other known VBSs. Structural modeling indicated that as opposed to canonical activation linked to interaction with D1, these combined VBSs interactions triggered major allosteric changes leading to D1D2 oligomerization. A cysteine-clamp preventing these changes and D1D2 oligomerization impaired growth of vinculin microclusters and cell adhesion. We propose that D1D2-mediated vinculin oligomerization occurs during the maturation of adhesion structures to enable the scaffolding of high-order vinculin complexes, and is triggered by Shigella IpaA to promote bacterial invasion in the absence of mechanotransduction.SummaryThe Shigella IpaA effector binds to cryptic vinculin sites leading to oligomerization via its head domain. Comment in Type VI secretion system baseplate. [Nat Microbiol. 2018]; International audience; To support their growth in a competitive environment and cause pathogenesis, bacteria have evolved a broad repertoire of macromolecular machineries to deliver specific effectors and toxins. Among these multiprotein complexes, the type VI secretion system (T6SS) is a contractile nanomachine that targets both prokaryotic and eukaryotic cells. The T6SS comprises two functional subcomplexes: a bacteriophage-related tail structure anchored to the cell envelope by a membrane complex. As in other contractile injection systems, the tail is composed of an inner tube wrapped by a sheath and built on the baseplate. In the T6SS, the baseplate is not only the tail assembly platform, but also docks the tail to the membrane complex and hence serves as an evolutionary adaptor. Here we define the biogenesis pathway and report the cryo-electron microscopy (cryo-EM) structure of the wedge protein complex of the T6SS from enteroaggregative Escherichia coli (EAEC). Using an integrative approach, we unveil the molecular architecture of the whole T6SS baseplate and its interaction with the tail sheath, offering detailed insights into its biogenesis and function. We discuss architectural and mechanistic similarities but also reveal key differences with the T4 phage and Mu phage baseplates.

BACKGROUND AND PURPOSE Muscarinic and adrenergic G protein-coupled receptors (GPCRs) are the targets of rare peptide toxins isolated from snake or cone snail venoms. We used a screen to identify novel toxins from Dendroaspis angusticeps targeting aminergic GPCRs. These toxins may offer new candidates for the development of new tools and drugs. EXPERIMENTAL APPROACH In binding experiments with 3H-rauwolscine, we studied the interactions of green mamba venom fractions with α2-adrenoceptors from rat brain synaptosomes. We isolated, sequenced and chemically synthesized a novel peptide, ρ-Da1b. This peptide was pharmacologically characterized using binding experiments and functional tests on human α2-adrenoceptors expressed in mammalian cells. KEY RESULTS ρ-Da1b, a 66-amino acid peptide stabilized by four disulphide bridges, belongs to the three-finger-fold peptide family. Its synthetic homologue inhibited 80% of 3H-rauwolscine binding to the three α2-adrenoceptor subtypes, with an affinity between 14 and 73 nM and Hill slopes close to unity. Functional experiments on α2A-adrenoceptor demonstrated that ρ-Da1b is an antagonist, shifting adrenaline activation curves to the right. Schild regression revealed slopes of 0.97 and 0.67 and pA2 values of 5.93 and 5.32 for yohimbine and ρ-Da1b, respectively. CONCLUSIONS AND IMPLICATIONS ρ-Da1b is the first toxin identified to specifically interact with α2-adrenoceptors, extending the list of class A GPCRs sensitive to toxins. Additionally, its affinity and atypical mode of interaction open up the possibility of its use as a new pharmacological tool, in the study of the physiological roles of α2-adrenoceptor subtypes.

A spore-forming, rod-shaped Gram-strain-positive bacterium, strain 656.84T, was isolated from human faeces in 1984. It contained anteiso-C15 : 0 as the major cellular fatty acid, meso-diaminopimelic acid was found in the cell wall peptidoglycan, the polar lipid profile consisted of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol and aminophospholipids as the major components, and the predominant menaquinone was MK-7. The DNA G+C content was 52.9 mol%. The results of comparative 16S rRNA gene sequence studies placed strain 656.84T within the genus Paenibacillus. Its closest phylogenetic relatives were Paenibacillus barengoltzii and Paenibacillus timonensis. Levels of DNA–DNA relatedness between strain 656.84T and Paenibacillus timonensis CIP 108005T and Paenibacillus barengoltzii CIP 109354T were 17.3 % and 36.8 %, respectively, indicating that strain 656.84T represents a distinct species. On the basis of phenotypic and genotypic results, strain 656.84T is considered to represent a novel species within the genus Paenibacillus, for which the name Paenibacillus faecis sp. nov. is proposed; the type strain is 656.84T ( = DSM 23593T = CIP 101062T).

The success of S. pneumoniae as a major human pathogen is largely due to its remarkable genomic plasticity, allowing efficient escape from antimicrobials action and host immune response. Natural transformation, or the active uptake and chromosomal integration of exogenous DNA during the transitory differentiated state competence, is the main mechanism for horizontal gene transfer and genomic makeover in pneumococci. Although transforming DNA has been proposed to be captured by Type 4 pili (T4P) in Gram-negative bacteria, and a competence-inducible comG operon encoding proteins homologous to T4P-biogenesis components is present in transformable Gram-positive bacteria, a prevailing hypothesis has been that S. pneumoniae assembles only short pseudopili to destabilize the cell wall for DNA entry. We recently identified a micrometer-sized T4P-like pilus on competent pneumococci, which likely serves as initial DNA receptor. A subsequent study, however, visualized a different structure - short, ‘plaited’ polymers - released in the medium of competent S. pneumoniae. Biochemical observation of concurrent pilin secretion led the authors to propose that the ‘plaited’ structures correspond to transformation pili acting as peptidoglycan drills that leave DNA entry pores upon secretion. Here we show that the ‘plaited’ filaments are not related to natural transformation as they are released by non-competent pneumococci, as well as by cells with disrupted pilus biogenesis components. Combining electron microscopy visualization with structural, biochemical and proteomic analyses, we further identify the ‘plaited’ polymers as spirosomes: macromolecular assemblies of the fermentative acetaldehyde-alcohol dehydrogenase enzyme AdhE that is well conserved in a broad range of Gram-positive and Gram-negative bacteria., Author Summary Streptococcus pneumoniae often escapes prevention and treatment through rapid horizontal gene transfer via natural transformation. Uptake of exogenous DNA requires expression of a transformation pilus but two markedly different models for pilus assembly and function have been proposed. We previously reported a long, Type 4 pilus-like appendage on the surface of competent pneumococci that binds extracellular DNA as initial receptor, while a separate study proposed that secreted short, ‘plaited’ transformation pili act simply as peptidoglycan drills to open DNA gateways. Here we show that the ‘plaited’ structures are not competence-specific or related to transformation. We further demonstrate that these are macromolecular assemblies of the metabolic enzyme acetaldehyde-alcohol dehydrogenase—or spirosomes—broadly conserved across the bacterial kingdom.

In pathogenic bacteria post-translationally modified proteins have been found to promote bacterial survival, replication and evasion from the host immune system. In the human pathogen Neisseria meningitidis, the protein PilE (15–18 kDa) is the major building block of type IV pili, extracellular filamentous organelles that play a major role in mediating pathogenesis. Previous reports have shown that PilE can be expressed as a number of different proteoforms, each harbouring its own set of post-translational modifications (PTMs) and that specific proteoforms are key in promoting bacterial virulence. Efficient tools that allow complete PTM mapping of proteins involved in bacterial infection are therefore strongly needed. As we show in this study, a simple combination of mass profiling and bottom-up proteomics is fundamentally unable to achieve this goal when more than two proteoforms are present simultaneously. In a N. meningitidis strain isolated from a patient with meningitis, mass profiling revealed the presence of four major proteoforms of PilE, in a 1:1:1:1 ratio. Due to the complexity of the sample, a top-down approach was required to achieve complete PTM mapping for all four proteoforms, highlighting an unprecedented extent of glycosylation. Top-down mass spectrometry therefore appears to be a promising tool for the analysis of highly post-translationally modified proteins involved in bacterial virulence.

Microdochium nivale , a wheat pathogenic filamentous fungus, appeared to be very sensitive in in vivo laboratory assays to fenpropimorph and tridemorph and to a lesser extent to fenpropidin, piperalin, and spiroxamine. It accumulated Δ 8 -sterols when grown in the presence of one of these sterol biosynthesis inhibitors. Thus, in M. nivale , these fungicides seemed to be very good inhibitors of the sterol Δ 8 → Δ 7 -isomerase. This hypothesis was then confirmed by sterol Δ 8 → Δ 7 -isomerase inhibition assays in cell-free enzyme systems.

Natural genetic transformation is widely distributed in bacteria and generally occurs during a genetically programmed differentiated state called competence. This process promotes genome plasticity and adaptability in Gram-negative and Gram-positive bacteria. Transformation requires the binding and internalization of exogenous DNA, the mechanisms of which are unclear. Here, we report the discovery of a transformation pilus at the surface of competent Streptococcus pneumoniae cells. This Type IV-like pilus, which is primarily composed of the ComGC pilin, is required for transformation. We provide evidence that it directly binds DNA and propose that the transformation pilus is the primary DNA receptor on the bacterial cell during transformation in S. pneumoniae. Being a central component of the transformation apparatus, the transformation pilus enables S. pneumoniae, a major Gram-positive human pathogen, to acquire resistance to antibiotics and to escape vaccines through the binding and incorporation of new genetic material., Author Summary Natural genetic transformation, first discovered in Streptococcus pneumoniae by Griffith in 1928, is observed in many Gram-negative and Gram-positive bacteria. This process promotes genome plasticity and adaptability. In particular, it enables many human pathogens such as Streptococcus pneumoniae, Staphylococcus aureus or Neisseria gonorrhoeae to acquire resistance to antibiotics and/or to escape vaccines through the binding and incorporation of new genetic material. While it is well established that this process requires the binding and internalization of external DNA, the molecular details of these steps are unknown. In this study, we discovered a new appendage at the surface of S. pneumoniae cells. We show that this appendage is similar in morphology and composition to appendages called Type IV pili commonly found in Gram-negative bacteria. We demonstrate that this new pneumococcal pilus is essential for transformation and that it directly binds DNA. We propose that the transformation pilus is an essential piece of the transformation apparatus by capturing exogenous DNA at the bacterial cell surface.

International audience; Nicotinic acetylcholine receptors (nAChRs) are one of the most important families in the ligand-gated ion channel superfamily due to their involvement in primordial brain functions and in several neurodegenerative pathologies. The discovery of new ligands which can bind with high affinity and selectivity to nAChR subtypes is of prime interest in order to study these receptors and to potentially discover new drugs for treating various pathologies. Predatory cone snails of the genus Conus hunt their prey using venoms containing a large number of small, highly structured peptides called conotoxins. Conotoxins are classified in different structural families and target a large panel of receptors and ion channels. Interestingly, nAChRs represent the only subgroup for which Conus has developed seven distinct families of conotoxins. Conus venoms have thus received much attention as they could represent a potential source of selective ligands of nAChR subtypes. We describe the mass spectrometric-based approaches which led to the discovery of a novel α-conotoxin targeting muscular nAChR from the venom of Conus ermineus. The presence of several posttranslational modifications complicated the N-terminal sequencing. To discriminate between the different possible sequences, analogs with variable N-terminus were synthesized and fragmented by MS/MS. Understanding the fragmentation pathways in the low m/z range appeared crucial to determine the right sequence. The biological activity of this novel α-conotoxin (α-EIIA) that belongs to the unusual α4/4 subfamily was determined by binding experiments. The results revealed not only its selectivity for the muscular nAChR, but also a clear discrimination between the two binding sites described for this receptor.

International audience; The Gram-negative bacterium Neisseria meningitidis asymptomatically colonizes the throat of 10 to 30% of the human population, but throat colonization can also act as the port of entry to the blood (septicemia) and then the brain (meningitis). Colonization is mediated by filamentous organelles referred to as type IV pili, which allow the formation of bacterial aggregates associated with host cells. We found that proliferation of N. meningitidis in contact with host cells increased the transcription of a bacterial gene encoding a transferase that adds phosphoglycerol onto type IV pili. This unusual posttranslational modification specifically released type IV pili-dependent contacts between bacteria. In turn, this regulated detachment process allowed propagation of the bacterium to new colonization sites and also migration across the epithelium, a prerequisite for dissemination and invasive disease.

International audience; PURPOSE: To determine the human centrin 2 (Hscen 2) protein response to oxidising radicals in vitro and to evaluate the consequences on its biological functions. MATERIALS AND METHODS: Hscen 2 was submitted to hydroxyl and azide radicals produced by radiolysis in the absence of oxygen. The resulting products were characterised by biochemical, spectroscopic and mass spectrometry techniques. Their thermodynamics parameters of complexation with C-terminal fragment of Xeroderma pigmentosum C protein (C-XPC), one of the Hscen 2 cellular partners, were quantified by isothermal titration calorimetry (ITC). RESULTS: Both hydroxyl and azide radicals induce centrin 2 polymerisation as we characterised several intermolecular cross-links generating dimers, trimers, tetramers and higher molecular mass species. These cross-links result from the formation of a covalent bond between the only tyrosine residue (Tyr 172) located in the C-terminal region of each monomer. Remarkably, dimerisation occurs for doses as low as a few grays. Moreover, this Hscen2 dimer has a lower affinity and stoechiometry binding to C-XPC. CONCLUSIONS: These results show that as oxidative radicals induce high proportions of irreversible damages (polymerisation) centrin 2 is highly sensitive to ionising radiation. This could have important consequences on its biological functions.

In the mycelium of all the Fusarium species studied, the major sterol was ergosterol in the absence of fenpropimorph. In the presence of this fungicide fenpropimorph-sensitive strains, with the exception of F. nivale , accumulated mainly Δ 8,14 -sterols and tolerant ones accumulated either both Δ 8,14 - and Δ 8 -sterols or only Δ 8 -sterols. This seemed to indicate that fenpropimorph toxicity was related to sterol Δ 14 -reductase sensitivity. In F. nivale , which was highly sensitive to fenpropimorph, the accumulation of only Δ 8 -sterols indicated that the main target enzyme was sterol Δ 8 →Δ 7 -isomerase. In addition to Δ 8,14 -and Δ 8 -sterols, squalene was accumulated in very high amounts in several strains; the level of squalene accumulation did not seem to be correlated with fenpropimorph sensitivity.

The importance of protein glycosylation in the interaction of pathogenic bacteria with their host is becoming increasingly clear. Neisseria meningitidis , the etiological agent of cerebrospinal meningitis, crosses cellular barriers after adhering to host cells through type IV pili. Pilin glycosylation genes ( pgl ) are responsible for the glycosylation of PilE, the major subunit of type IV pili, with the 2,4-diacetamido-2,4,6-trideoxyhexose residue. Nearly half of the clinical isolates, however, display an insertion in the pglBCD operon, which is anticipated to lead to a different, unidentified glycosylation. Here the structure of pilin glycosylation was determined in such a strain by “top-down” MS approaches. MALDI-TOF, nanoelectrospray ionization Fourier transform ion cyclotron resonance, and nanoelectrospray ionization quadrupole TOF MS analysis of purified pili preparations originating from N. meningitidis strains, either wild type or deficient for pilin glycosylation, revealed a glycan mass inconsistent with 2,4-diacetamido-2,4,6-trideoxyhexose or any sugar in the databases. This unusual modification was determined by in-source dissociation of the sugar from the protein followed by tandem MS analysis with collision-induced fragmentation to be a hexose modified with a glyceramido and an acetamido group. We further show genetically that the nature of the sugar present on the pilin is determined by the carboxyl-terminal region of the pglB gene modified by the insertion in the pglBCD locus. We thus report a previously undiscovered monosaccharide involved in posttranslational modification of type IV pilin subunits by a MS-based approach and determine the molecular basis of its biosynthesis.

International audience; Recognition of the mother is of major importance for the survival of mammalian neonates. This recognition is based, immediately after birth, on the detection of odours that have been learned by the fetus in utero. If the ethological basis of a transnatal olfactory continuity is well established, little is known on the nature of its olfactory cues, and nothing about the presence of potential carrier proteins in the maternal fluids such as amniotic fluid, colostrum and milk. We have identified the components of the pig putative maternal pheromone in these fluids of the sow. We also used a ligand-oriented approach to functionally characterize carrier proteins for these compounds in the maternal fluids. Six proteins were identified, using binding assay, immunodetection and peptide mapping by mass spectrometry. These proteins are known to transport hydrophobic ligands in biological fluids. Among them, alpha-1 acid glycoprotein (AGP) and odorant-binding protein (OBP) have been described in the oral sphere of piglets as being involved in the detection of pig putative maternal pheromone components. These are the first chemical and biochemical data supporting a transnatal olfactory continuity between the fetal and the postnatal environments.

International audience; This study targets to express the piglet odorant-binding protein (p1OBP) and compare the engineered product to the corresponding native protein forms, i.e. plOBP and adult porcine OBP (pOBP). Using the natural signal peptide from the cDNA sequence, up to 40 mg 1-1 of secreted recombinant piglet OBP (rOBP) has been produced in a minimal culture medium. No significant difference in molecular mass between rOBP and native plOBP could be observed by mass spectrometry following or not trypsin digestion. rOBP and pOBP shared similar immunoreactivity towards polyclonal anti-pOBP antibodies, suggesting a proper processing and folding of the recombinant product. Both plOBP and rOBP displayed comparable binding properties towards fatty acids present in the putative maternal pheromone and a steroid, component of the boar sex pheromone. Furthermore, the rOBP product was found to bind to an olfactory receptor, for which pOBP binding was previously characterized. Taken together, these findings suggest that rOBP, produced in Pichia pastoris, exhibits structural and functional properties comparable to those of the native lipocalins from both young or adult animal.

Social insects often show an extreme reproductive skew. In queenless ants, colonies consist of morphologically identical workers that can all potentially reproduce sexually. Similarly to that in social vertebrates, aggression in these ants functions to select the reproductive(s). We investigated the mechanisms underlying reproductive skew in the monogynous queenless ant Streblognathus peetersi. Behavioural observations of disturbed hierarchies were integrated with physiological measures of fertility (vitellogenin titre in the haemolymph) and chemical analysis of cuticular hydrocarbons, which are putative fertility pheromones. This multifaceted approach revealed that the colony reproductive is determined as a result of aggression between high-ranking workers, but once an alpha is established, chemical signalling is enough to maintain reproductive skew. As already reported in several species of ants and also in a social wasp, egg layers have distinct profiles of cuticular hydrocarbons compared with infertile workers. Importantly, ‘high rankers’ who are unable to lay eggs also have a specific cuticular profile; this is consistent with their intermediate state of fertility indicated by vitellogenin levels. Rather than just ovarian activity, the cuticular hydrocarbon profile thus reveals the individual hormonal state that underlies reproductive activity. We compare the fertility signal in queenless ants with the ‘badge of status’ reported in various birds. We discuss the evolutionary stability of this communication system and give special emphasis to ecological constraints and the high degree of intracolonial relatedness typical of social insects.

International audience; Improvement of the catalytic properties of fungal laccases is a current challenge for the efficient bioremediation of natural media polluted by xenobiotics. We developed the heterologous expression of a laccase from the white-rot fungus Trametes versicolor in the yeast Yarrowia lipolytica as a first step for enzyme evolution. The full-length cDNA consisted of a 1,561-bp open reading frame encoding lacIIIb, a 499-amino-acid protein and a 21-amino-acid signal peptide. Native and yeast secretion signals were used to direct the secretion of the enzyme, with the native signal yielding higher enzyme activity in the culture medium. The level of laccase activity secreted by the transformed yeast was similar to that observed for the non-induced wild-type strain of T. versicolor. The identity of the recombinant enzyme was checked by Western blot and matrix-assisted laser desorption/ionization time-of-flight analysis. Electrophoresis separation in native conditions indicated a molecular mass of the recombinant protein slightly higher (5 kDa) than that of the mature T. versicolor laccase IIIb, suggesting a limited excess of glycosylation. The laccase production level reached 2.5 mg/l (0.23 units/ml), which is suitable for engineering purpose.

International audience; The biotransformation of 16 polycyclic aromatic hydrocarbons (PAHs) in mixture was investigated in reactors in the presence of purified laccases of the fungus Pycnoporus cinnabarinus, ABTS as a redox mediator, 25% acetonitrile, and Tween 20. Several hydrocarbons from a synthetic mixture, such as anthracene and benzo[ a ]pyrene, were converted up to 80% into quinones, whereas others also belonging to three- and five-ring chemicals were less transformed. Chrysene and benzo[ k ]fluoranthene were not oxidized by the laccase mediator system. Moreover, hydrocarbons extracted from an industrial soil were all recalcitrant to enzymatic attack. This lack of reactivity of the laccases toward the hydrocarbons could be due to the presence of interfering compounds coextracted from the soil, such as metals.

Bovine odorant-binding protein (bOBP) is a dimeric lipocalin present in large amounts in the respiratory and olfactory nasal mucosa. The structure of bOBP refined at 2.0-A resolution revealed an elongated volume of electron density inside each buried cavity, indicating the presence of one (or several) naturally occurring copurified ligand(s) (Tegoni et al. (1996) Nat. Struct. Biol. 3, 863-867; Bianchet et al. (1996) Nat. Struct. Biol. 3, 934-939). In the present work, by combining mass spectrometry, x-ray crystallography (1.8-A resolution), and fluorescence, it has been unambiguously established that natural bOBP contains the racemic form of 1-octen-3-ol. This volatile substance is a typical component of bovine breath and in general of odorous body emanations of humans and animals. The compound 1-octen-3-ol is also an extremely potent olfactory attractant for many insect species, including some parasite vectors like Anopheles (Plasmodium) or Glossina (Trypanosoma). For the first time, a function can be assigned to an OBP, with a possible role of bOBP in the ecological relationships between bovine and insect species.

The mechanisms of resistance to terbinafine, a squalene epoxidase inhibitor, and to fenpropimorph, a sterol Δ 14 -reductase and/or Δ 8 → Δ 7 -isomerase inhibitor, were investigated in laboratory mutants of the phytopathogenic fungus Nectria haematococca . Neither modified fungicide uptake nor fungicide metabolism could explain resistance in the mutants studied. The terbinafine-resistant mutants contained at least 10 times more squalene than the wild-type strain, and when cultivated in presence of terbinafine, they required a 30-times higher fungicide concentration to obtain a very high level of accumulated squalene similar to that of the wild-type strain. Thus, a reduced affinity of the squalene epoxidase toward both the substrate and the fungicide could explain terbinafine resistance in these mutants. In some fenpropimorph-resistant mutants, the effects of fenpropimorph on sterol biosynthesis were similar to those in the wild-type strain, suggesting that resistance could be due to tolerance to sterol Δ 8,14 -sterol accumulation and to Δ 5,7 -sterol decrease. In one of them, the major sterol in absence of fungicide was not ergosterol, but ergosta-5,7,22,24(24 1 )-tetraenol, indicating reduced Δ 24 (24 1 )-reductase activity. The potential role of a modified Δ 24 (24 1 ) reductase in fenpropimorph resistance has yet to be elucidated. In the remaining fenpropimorph-resistant mutants, much higher fenpropimorph concentrations were required to cause ergosterol decreases similar to that in the wild-type strain, suggesting a Δ 14 -reductase modification. Among them, some mutants accumulated Δ 8 -sterols as well as Δ 8,14 -sterols, indicating that the mechanism of resistance may be due to reduced affinity of the Δ 14 -reductase toward fenpropimorph, whereas the other mutants accumulated only Δ 8,14 -sterols. Those last mutants contained much larger amounts of 4,4-dimethyl and 4α-methyl sterols than the wild-type strain, in the absence of fungicide. The most abundant methylated sterol was 4,4-dimethylfecosterol, the presumed product of the Δ 14 -reductase, suggesting that resistance may be due to overproduction of the Δ 14 -reductase in these mutants.

Mycelium of Eutypa lata grown in solid and liquid cultures contained C 28 -sterols, mainly ergosterol, accompanied by much smaller amounts of episterol, ergostatetraenol, ergosta-7,22-dien-3β-ol and minor 4α-methyl and 4,4-dimethylsterols.

Ergosta-5,24(241)-dien-3β-ol was the major sterol of conidia of Uncinula necator, an obligate pathogenic fungus, which is the causal agent of vine powdery mildew. Other Δ5-sterols, such as cholesterol, ergost-5-en-3β-ol, stigmasta-5,22-dien-3β-ol and stigmast-5-en-3β-ol found in U. ecator host plant (Vitis vinifera) were detected as minor sterols in conidia of U. necator. The triadimenol-sensitive and resistant strains contained similar sterol compositions. Triadimenol treatment led to accumulation of 14α-methylsterols, such as eburicol and obtusifoliol. This suggests that sterol C-14 demethylase is the target for triadimenol in sterol biosynthesis in U. necator.

Catabolism of juvenile hormone was studied in vivo in the African locust by injection of the labelled natural enantiomer (10R) (12-3H) JH-III. Due to the poor solubility of JH in aqueous solution, it was injected in a water-miscible solvent. Ethanol was chosen for its apparently low toxicity towards the locust. In these experimental conditions, reverse phase liquid chromatography procedure (RP-HPLC) coupled with on line radiodetection, revealed an apolar metabolite of JH-III. This compound was found both in adult females and in fifth stadium larvae. We demonstrate that this metabolite resulted from substitution of the carboxyl methyl group of JH-III by some hydrophobic moiety. This compound co-migrates in our RP-HPLC system with the JH analog epoxy-ethyl farnesoate (JH-III ethyl ester) obtained by KCN-catalysed transesterification of JH-III in ethanol. Both JH-III ethyl ester of chemical origin and biological compounds extracted from locusts give the same spectra when analyzed by gas chromatography-mass spectroscopy (GC-MS). Transesterification of JH-III was not observed with locust tissues incubated in vitro but occurred in vivo even if JH was injected in other alcoholic solvents such as propanol. Our data suggest that transesterification of JH-III occurred in vivo and underline the role of injecting solvent in in vivo studies.

International audience; The trans- andcis-(Z)-α-bisabolene epoxides (trans- andcis-(Z)-αBE) are the main components of the male sex pheromone in Nezara viridula. The role of thecis isomer and the importance of thecis/trans proportion for the activity and the specificity of the pheromone are not clearly elucidated and were studied here. Interindividual variation of thecis/ trans proportion produced by males was studied by individual hexanic extracts in two strains originating from the south of France (SF) and French West Indies (FWI). The trans isomer composed 42–82% of bisabolene epoxides in SF males and 74–94% of bisabolene epoxides in FWI males. Means (± SD) significantly differ between SF (62.8%±8.4) and FWI (82.4%±5.9) males in spite of this interindividual variation. Different isomers of bisabolene epoxide were synthesized and their EAG activity on female antennae was compared. Racemictrans- andcis-(Z)-αBE elicited low EAGs, not different from the nonnaturaltrans andcis (E)-αBE that were inactive on behavior. Behavioral tests revealed that racemictrans- andcis-(Z)-αBE attracted 45% (P0.05) of the females, respectively. Binary blends containing 75/25, 50/50, and 25/75 proportions ofcis/ trans isomers were more attractive than trans-(Z)-agrBE alone and response of females to the 25%cis/75% trans blend was significantly more important than the response to trans-isomer alone (P

The leaf-cutting ants Acromyrmex octospinosus live in symbiosis with a fungus they cultivate on leaf fragments. The symbiote forms characteristic hyphal apical swellings called “gongylidia,” which are consumed in preference to the mycelium by the ants. However, the ants also ingest fluids derived from the plant leaves they handle to make the culture substrate. The 4-desmethylsterols of the ants were essentially (83%–94% of total 4-desmethylsterols) Δ5,7-sterols which were identified as ergosta-5,7,24(28)-trien-3β-ol, ergosta-5,7-dien-3β-ol, and ergosterol. These Δ5,7-sterols were all found in the gongylidia, whereas ergosterol mainly was found in the mycelium. Some Δ5-sterols were detected in the whole body, as well as in the expressed rectal fluid of the ants which partly contains the nonabsorbed material. However, the rectal fluid, as compared to the whole body, contained much higher amounts of Δ5-sterols originating mainly from the plant leaves. Lytic transglycosylases (LT) are redundant enzymes that play a critical role in peptidoglycan (PG) recycling and metabolism. LT(s) role in cell wall-modifying complexes and usefulness as antimicrobial drug targets remain elusive. We determined at high-resolution a structure of the membrane-bound homolog of the soluble LT fromNeisseriaspecies with a disordered active site helix (alpha helix 30). Alpha helix 30 is crucial for binding PG during catalysis1. Here we show using an alpha helix 30 deletion strain that LT (LtgA) determines the integrity of the cell wall, participates in cell division and separation, and can be manipulated to impair the fitness of the human pathogenNeisseria meningitidisduring infection. Characterization ofltgAhelix deleted strain interactome identified glycan chain remodeling enzymes whose function appear to be modulated by LTs. Targeting LTs can disrupt the PG machinery, which is fatal for the bacterium, a new approach for antibiotic development.
