Introduction: Sea cucumber, Apostichopus japonicus, is becoming popular around the world due to its nutritional and medicinal properties. There are still no detailed chemical studies of the lipid class, glycolipids compositions of sea cucumber. Methods: This study was conducted to determine the lipid class and glycolipid compositions of dried sea cucumber, A. japonicus, and analyze fatty acid compositions of Monogalactosyl Diglycerides (MGDG), Steryl Glycosides (SG) and Sulfoquinovosyl Diglycerides (SQDG). Total lipids of sea cucumber were extracted by Bligh and Dyer method and Sep-Pak Silica plus long cartridge, and Thin Layer Chromatography (TLC) silica gel G-60 F254 was used for the separation of different lipid classes and glycolipid compositions. The composition of fatty acids was analyzed by GC. Results & Conclusion: The level of total lipids in the dried sea cucumber, Apostichopus japonicus, was 4 ± 1% of dry weight (w/w) and the amount of neutral lipids, glycolipids and phospholipids was 31 ± 1%, 29 ± 1% and 40 ± 1% of the total lipids (w/w), respectively. MGDG, SG and SQDG were the major glycolipids, and the contents were 37.5 ± 0.3%, 33.8 ± 0.5% and 23.6 ± 0.7% of the total glycolipids (w/w), respectively and significantly higher than other glycolipids (p < 0.05). SQDG contained much higher Arachidonic Acid (AA), Eicosapentaenoic Acid (EPA) and MGDG contained higher Docosahexaenoic Acid (DHA) compared with SG (p < 0.05). Further investigation is required to understand the positional distribution of fatty acids and molecular species in MGDG, SG and SQDG in detail.

In a previous study, our research group demonstrated that sea cucumber (Apostichopus japonicus) extracts ameliorated allergic airway inflammation through CD4+CD25+Foxp3+ T (regulatory T; Treg) cell activation and recruitment to the lung. In this study, we aimed to determine which components of sea cucumber contribute to the amelioration of airway inflammation. We used n-hexane fractionation to separate sea cucumber into three phases (n-hexane, alcohol, and solid) and evaluated the ability of each phase to elevate Il10 expression in splenocytes and ameliorate symptoms in mice with ovalbumin (OVA)/alum-induced asthma. Splenocytes treated with the n-hexane phase showed a significant increase in Il10 expression. In the n-hexane phase, 47 fatty acids were identified. Individual fatty acids that comprised at least 5% of the total fatty acids were 16:0, 16:1n-7, 18:0, 18:1n-7, 20:4n-6, and 20:5n-3 (eicosapentaenoic acid). After administering the n-hexane phase to mice with OVA/alum-induced asthma, their asthma symptoms were ameliorated. Several immunomodulatory effects were observed in the n-hexane phase-pretreated group, compared with a vehicle control group. First, eosinophil infiltration and goblet cell hyperplasia were significantly reduced around the airways. Second, the concentrations of Th2-related cytokines (IL-4, IL-5, and IL-13) and Th17-related cytokines (IL-17) were significantly decreased in the spleen and bronchoalveolar lavage fluid (BALF). Finally, the concentrations of TGF-β and IL-10, which are associated with Treg cells, were significantly increased in the BALF and splenocyte culture medium. In conclusion, a fatty acid-rich fraction (n-hexane phase) of sea cucumber extract ameliorated allergic airway inflammation in a mouse model.

전 세계 해양에서 이루어지고 있는 어업에 의한 수산물 생산 량은 지난 20년 동안 연간 9,000만 톤 수준에서 정체되고 있지 만, 수산물에 대한 수요는 꾸준히 증가하여 현재 1억 5천만 톤 에 이르고 있기 때문에 그 부족분을 수산양식에 의한 생산으로 충족시키고 있다(FAO, 2010). 현재 수산양식에 의한 수산물 생 산방법은 육상의 노지양식과 연안의 가두리 양식에 의하여 이 루어지고 있다. 이들 두 양식양법은 기후변화 및 환경변화의 영 향으로 생산이 불안정하고 생태파괴, 질병전파, 적조발생과 같 은 연안환경 오염 문제를 유발하고 있다. 이러한 문제를 해결 하기 위하여 변온동물인 양식생물을 연중 사육할 수 있고, 적 조 및 태풍과 같은 자연재해를 극복할 수 있는 육상순환양식 시스템(Recirculating Aquaculture Systems, RAS)이 적극적으 로 도입되고 있다(Aruety et al., 2016). 그러나 대부분의 육상 RAS 양식장은 인공배합사료를 사용하여 고밀도로 양식생물을 생산하기 때문에 이들 생산결과에 의해서 생겨난 고형오물의 처리는 새로운 사회적 문제로 대두되고 있다(Mirzoyan et al., 2008). 양식장에서 배출된 고형오물은 어류가 섭식하지 못한 잔여사료와 소화되지 못한 영양소로 구성되어 유기물 함량이 50-92% 차지한다(Piedrahita, 2003; Gebauer, 2004; Gebauer and Eikebrokk, 2006). 어류 1 kg을 생산하기 위하여 인공배합 사료 1-2 kg을 급이해야 하고 그 중에서 약 36%는 고형오물 형 태로 배출된다(Naylor et al., 2000). 지금까지 육상어류양식장 에서 배출된 고형오물을 처리하기 위한 방안으로 유기질비료 로 사용(Danaher et al., 2009), 바이오가스로 활용(Luo et al., 2013), 식물환경복원(Ghaly et al., 2005) 등을 시도하였지만 아 직 실용화되지 못하고 있다. 또한 고염류와 고농도 유기물 때문 에 하수폐수처리장에서 처리가 곤란할 뿐만 아니라(Mirzoyan et al., 2008), 2016년부터 해양환경관리법에 의하여 해양투기 가 금지되기 때문에 이에 대한 대책이 시급하고도 절실하다. 해삼(Apostichopus japonicus)은 주로 한국 전 연안, 중국 발 해만, 일본 그리고 러시아의 태평양연안에서 한대 해역까지 북 동 태평양 전역에 이르고 있으며 한국과 중국 그리고 일본에 서 중요한 수산양식생물이다(Slater and Carton, 2007). 해삼 은 저서생물로서 바닥의 퇴적물 중에 섞여 있는 미세조류, 박 해삼사료원료로서 육상순환여과양식장 고형오물의 이화학적 특징과 해삼(Apostichopus japonicus)의 소화흡수율

We conducted a 90-day feeding experiment to evaluate the growth and energy budget of the sea cucumber Apostichopus japonicus. Sea cucumbers with a mean initial wet body weight of 3.03±0.06 g were fed one of the following three diets: an eel fecal solid diet, rainbow trout fecal solid diet, or a commercial diet at a water temperature of 17±1°C and salinity of 32±1 psu. The results suggested that the diets affected the final body weight, specific growth rate (SGR), food ingestion, feces production ratio, and hence the growth and energy budget of the sea cucumbers. Sea cucumbers fed the eel and rainbow trout fecal solid diets showed poorer energy absorption, assimilation, and growth than those fed the commercial diet. The sea cucumbers fed the commercial diet had a significantly higher SGR and proportion of energy used for growth than those fed the other two diets. Sea cucumbers fed the rainbow trout fecal solid diet showed a comparatively higher ingestion rate and feces production ratio than those fed the commercial diet. The eel and rainbow trout fecal solid diets, therefore, were not suitable for sea cucumbers in intensive cultivation. Our findings will facilitate further development of more appropriate diets for culture of sea cucumber.

조피볼락(Sebastes schlegeli)은 동북아 연안, 일본 홋가이도 이남 및 중국 연안에 분포하며, 수심 100 m 내외의 천해 암초나 인공어초 등에 서식하기 때문에 자원증강이나 레저용으로 중요 한 위치를 차지하고 있어 경제적으로나 사회적으로 가치가 큰 어종이다(Boehlert et al., 1986). 조피볼락은 알을 산란하는 일 반어류와는 달리 체내에서 수정하여 새끼를 산출하는 태생어 류이다(Corey et al., 1983). 태생어류인 볼락류에서 산출된 자 어들은 인공사료에 길들이기가 어려운 특징을 갖고 있어 인공 종묘생산에 있어서 장시간에 걸쳐서 살아있는 먹이생물을 공 급해야 한다는 문제점을 가지고 있다(Lee et al., 1993a; Lee et al., 1993b). 현재 인공종묘생산을 위하여 성립되어 있는 먹이생 물의 공급체계는 해수클로렐라-로티퍼(Rotifer)-알테미아(Artemia)-인공배합사료로의 공급체계가 구축되어 있다(Fujita, 1973; Kitajima, 1983; Hirayama, 1985; Yamasaki et al., 1987; Yoshimura et al., 1996). 현재, 산업적인 측면에서 볼락류의 인 공종묘생산에 있어서 가장 큰 문제점은 불확실성이 너무 높다 는 것이다. 이러한 이유는 여러 가지 가설이 있지만, 가장 유력 시 되고 있는 가설은 반세기 걸쳐서 그대로 유지해온 외줄타기 식의 먹이생물 공급체계에 두고 있다(Lee et al., 1993c; Lee et al., 2004). 현재 구축된 먹이생물 중에서 클로렐라와 로티퍼를 배양하여 먹이로 활용하는 데는 문제가 없으나, Artemia는 염 호라는 극한 상황의 환경에서 자란 갑각류의 난을 채집하여 건 조 통조림화한 것으로서 전량 수입해서 사용하고 있다(Kim et al., 1999). 또한 Artemia의 지질영양학적인 문제점에 대해서는 물벼룩(Moina macrocopa) 급여가 조피볼락(Sebastes schlegeli) 자어의 성장에 미치는 영향 정우철·Feng Jin·최종국·이정태·최병대·강석중

유글레나(euglenoids)는 직경 15-500 μm 크기의 나사선 모양 으로 꼬인 길쭉한 방추형인데 매우 자유롭게 변형하며, 체내에 엽록체를 가지고 광합성을 하는 점으로 보아 식물에 속한다고 하지만, 세포벽이 없고 편모로 유영생활을 하므로 원생동물의 편모충류로 취급하기도 한다(Rodriguez-Zavala et al., 2010). 즉, 식물과 동물의 중간에 위치한다고 볼 수 있는데 엽록소를 가 지고 광합성을 하는 것은 식물적 특성이며, 입이나 수축포를 가 지고 자유롭게 움직이는 것은 동물적 특성에 속한다(Ruiz et al., 2004). 대부분 담수에서 살며 작은 연못이나 도랑에서 흔히 볼 수 있고 내만의 염수역에서도 볼 수 있다. 현재, 유글레나를 함 유하는 조류에 의한 유용물질 생산의 가능성에 관해서 각종 산 업계가 주목하고 있으며(Hayashi et al., 1994; Chisti and Yan, 2011), 유글레나 분말은 식품으로서의 실용화가 이미 수행되고 있으며(Choi et al., 2004), 고등식물에 비하여 석유화학 대체원 료, 혹은 연료의 원료 등 유용물질을 고수율로서 생산할 수 있 는 능력을 가지고 있다(Ishikawa et al., 2008; Lira-Silva et al., 2011). 또한 수많은 미세조류 중에서도 여러 분야에서 유용성 이 기대되고 있는 유글레나는 광합성을 통해 대기 중 이산화탄 소를 섭취해 고정하고 산소를 배출하므로 이를 이용한 하폐수 의 영양염류 제거 및 회수된 바이오매스를 Bio-fuel로 생산하 는 시스템 개발을 위한 연구가 진행되고 있다(Navarro et al., 1997; Ramalho et al., 1998; Tucci et al., 2006; Jasso-Cha vez et al., 2010). 현재 유글레나의 배양방법으로는 자가영양 배양방법과 타가영양 배양방법으로 이루어지고 있으나, 대부 배양방법에 따른 Euglena gracilis의 성장 및 지방산 조성 정우철·최종국·강창민·최병대·강석중*

유글레나는 엽록체를 가지고 광합성을 하는 점에서는 식물 로 취급하지만, 세포벽이 없고 편모로 유영생활을 하는 점에서 는 동물의 편모충류로 분류하기도 한다(Ruiz et al., 2004; Rodriguez-Zavala et al., 2010). 유글레나는 Vitamin E, Pramylon 와 같은 기능성 물질 뿐만 아니라 EPA와 DHA 같은 오메가-3 고도불포화지방산을 함유하고 있기 때문에 어류 자치어의 먹 이생물도 관심을 받고 있다(Harwood, 1988; Hayashi et al., 1993; James and Browse, 1999; Barsanti et al., 2000; Choi et al., 2013). 이들 기능성물질과 지방산의 함량은 자가영양배 양(autotrophic), 타가영양배양(heterotrophic) 및 혼합영양배 양(Mixotrophic) 배양조건에 따라서 달라진다(Wen and Chen, 2003; Wang et al., 2012). 즉 목적하는 기능성물질을 얻기 위해 서는 배양방법의 결정이 중요하다. 전보(Jeong et al., 2015)의 연구에 의하면 유글레나의 바이오매스와 오메가-3고도불포화 지방산의 함량 측면에서는 혼합배양방법이 최적배양방법인 것 으로 나타났다. 따라서 본 실험에서는 혼합영양배양 조건하에 서 배양온도와 pH와 같은 물리적 환경조건과 탄소원, 질소원 및 C/N비율과 같은 영양조건을 달리했을 때 따른 유글레나의 성장과 지질에 미치는 최적 배양조건을 구하고자 하였다.
