Current cancer therapy has been restricted by the hypoxic microenvironment of tumors, especially for strongly oxygen-dependent photodynamic therapy. To defeat tumor hypoxia, an oxygen-irrelevant radical nanogenerator, PI/FBC, is developed by the co-assembly of iodized polymer PI and NIR photosensitizer FBC, and further evaluated as a remote controllable free radical generation platform for enhancing antitumor efficiency. The PI/FBC radical nanogenerator can be excited by NIR light to produce ROS through transfer of energy to oxygen and induce the formation of toxic iodine radicals via electron transfer to PI. Notably, unlike conventional tumor treatments, such a radical nanogenerator is controllable and insusceptible to oxygen concentration. Moreover, benefiting from the strong NIR emission of FBC, the distribution of the PI/FBC radical nanogenerator can be monitored without incorporating other imaging agents. This PI/FBC radical nanogenerator treatment will no doubt broaden the family of antitumor strategies by using non-oxygen radicals, which is significant for reference in the development of promising anticancer agents., An iodized polymer-based radical nanogenerator, PI/FBC, with strong NIR emission was prepared for NIR-mediated hypoxia-irrelevant tumor therapy.

Thermoresponsive polymers incorporated with photo-absorbing agents have been widely utilized for controlled drug delivery using light as an external stimulus. However, previously developed thermoresponsive drug carriers have disadvantages such as low biocompatibility and implantation failure. In the present study, gold nanorods (GNRs)-encapsulated poly(N-vinyl caprolactam) (PVCL) (GNR-PVCL) microparticles were synthesized by the stop-flow lithography (SFL) method. The SFL method enabled the fabrication of near-infrared (NIR) light-responsive GNR-PVCL microparticles of uniform size, which can allow localized injection. Doxorubicin (DOX) was encapsulated into GNR-PVCL microparticles to achieve NIR light-responsive anticancer therapy. DOX-loaded GNR-PVCL (DOX-GNR-PVCL) microparticles exhibited NIR light-triggered drug release due to the photothermal effect of GNRs, which increases the local temperature above the volume phase transition temperature of GNR-PVCL microparticles. In addition, DOX-GNR-PVCL exhibited controlled DOX release in response to the periodic irradiation of NIR light. Moreover, we demonstrated the efficient intracellular release of DOX upon NIR light exposure, and thus, NIR light-responsive anticancer activity. This study demonstrates that DOX-GNR-PVCL microparticles have significant potential as implantable drug carriers enabling NIR light-triggered drug release.

The solar spectral converters mainly involve the energy transfer between two codoped ions. Here, we report a series of Ce3+, Cr3+, Ln3+ (Ln = Yb, Nd, Er) tridoped Gd3Sc2Ga3O12 (GSGO) phosphors with improved absorption and increasing near infrared (NIR) emission. We observed the multiple energy transfer behaviors of Cr3+→Ln3+, Ce3+→Ln3+, Ce3+→Cr3+, and Ce3+→Cr3+→Ln3+ in GSGO matrix. When Ce3+ is introduced into the GSGO:Cr3+,Ln3+ phosphors, the energy transfer of Ce3+→Cr3+→Ln3+ has been realized by utilizing the energy transfer bridge of the Cr3+ ion. Consequently, GSGO:Ce3+,Cr3+,Ln3+ can absorb almost all ultraviolet and visible (UV–Vis) light and produce strong NIR light thanks to the synergistic effect of Ce3+→Cr3+→Ln3+, improving the photovoltaic conversion efficiency of c-Si solar cells. Our results show that the prepared GSGO:Ce3+,Cr3+,Ln3+ have the potential application in the solar spectral material for c-Si solar cells. Meanwhile, the strategy of multiple energy transfers gives a new way to design the spectral conversion materials with wider absorption for c-Si solar cells.

Various nanoplatforms have been developed to visualize intracellular microRNAs (miRNAs) because of their clinical significance in tumor progression and diagnosis. However, the diffusion-limited motion of the nanoplatforms penalizes the miRNA imaging efficiency in cells. Herein, we fabricated a near-infrared (NIR) light-propelled Janus-based nanoplatform to advance the imaging response. The Janus nanomotor covered with an Au half-shell was loaded by the endocytosis adjuvant of the MnO2 nanosheet for delivering a miRNA-responsive hQN (hairpin DNA quadrangular nanostructure) probe with a catalyzed hairpin assembly (CHA). Once the nanoplatform entered into cells, the MnO2 nanosheet was degraded to Mn2+ by endogenous fuels (such as glutathione) to release the hQN probe. The NIR light irradiation of the nanoplatform generated a heat gradient and thus propelled motion of the nanoplatform. This process accelerated the intracellular reaction of the hQN probe with miRNAs to trigger the cascade CHA amplification with an enhanced fluorescence readout.

Cancer has become a severe threat to human life due to its high mortality and metastatic rate. Effective inhibition and killing of cancer cells using chemotherapeutic drugs have been a promising means in clinical cancer therapy. However, the low selectivity, drug-resistance, uncontrollability and serious side effects of chemotherapy significantly limit its further development. There is an urgent need for new treatment strategies to compensate for deficiencies inherent in chemotherapy alone. A growing body of research shows that combined treatment strategies have the potential to overcome this dilemma by achieving significantly enhanced synergistic effects and reduced side effects. Emerging phase change materials (PCMs) create an ideal nanoplatform for cancer combination therapy due to their universal loading properties, stable and temperature-responsive phase transition capability, and excellent natural biocompatibility/biodegradability. The release of therapeutic agents can be precisely controlled through external, non-intrusive stimuli (such as NIR light and ultrasound), avoiding systemic toxicity associated with conventional chemotherapy. Herein, the construction methods and design principles of PCM-based nanoplatforms serving as strict gatekeeper and smart payload delivery systems are discussed in detail. Moreover, the advantages and disadvantages of these nanoplatforms are provided. A suitable discussion and perspective of the remaining challenges and future opportunities for PCM-based nanoplatforms in cancer treatment are also given in conclusion.

Light-based theranostics have become indispensable tools in the field of cancer nanomedicine. Specifically, near infrared (NIR) light mediated imaging and therapy of deeply seated tumors using a single multi-functional nanoplatform have gained significant attention. To this end, several multi-functional nanomaterials have been utilized to tackle cancer and thereby achieve significant outcomes. The present review mainly focuses on the recent advances in the development of NIR light activatable multi-functional materials such as small molecules, quantum dots, and metallic nanostructures for the diagnosis and treatment of deeply seated tumors. The need for improved disease detection and enhanced treatment options, together with realistic considerations for clinically translatable nanomaterials will be the key driving factors for theranostic agent research in the near future. NIR-light mediated cancer imaging and therapeutic approaches offer several advantages in terms of minimal invasiveness, deeper tissue penetration, spatiotemporal resolution, and molecular specificities. Herein, we have reviewed the recent developments in NIR light responsive multi-functional nanostructures for phototheranostic applications in cancer therapy.

A non-toxic nano system using a cleverly designed dual light can be an important treatment strategy in cancer therapy. Here, we propose a unique concept of synergistic non-toxic nanoplatform, which operates with dual light. An apoferritin nano-platform whose surface was modified with Verteporfin (Visudyne (R)) and with ultra-small CuS nanoparticles in the center demonstrated the synergistic effect of photodynamic therapy (PDT) and photothermal therapy (PTT). The synergistic effect of PDT and PTT were achieved with 808 nm laser light and 690 nm LED light, respectively. These results not only give a new horizon to multifunctional nanostructures for biological applications but also show a new way to design the novel PDT and PTT agents.

Purpose Antibiotic resistance issues associated with microbial pathogenesis are considered to be one of the most serious current threats to health. Fortunately, TiO2, a photoactive semiconductor, was proven to have antibacterial activity and is being widely utilized. However, its use is limited to the short range of absorption wavelength. Methods In this work, heterostructured TiO2-FeS2 nanocomposites (NCs) were successfully prepared by a facile solution approach to enhance light-induced antibacterial activity over a broader absorption range. Results In TiO2-FeS2 NCs, FeS2 NPs, as light harvesters, can effectively increase light absorption from the visible (Vis) to near-infrared (NIR). Results of light-induced antibacterial activities indicated that TiO2-FeS2 NCs had better antibacterial activity than that of only TiO2 nanoparticles (NPs) or only FeS2 NPs. Reactive oxygen species (ROS) measurements also showed that TiO2-FeS2 NCs produced the highest relative ROS levels. Unlike TiO2 NPs, TiO2-FeS2 NCs, under light irradiation with a 515-nm filter, could absorb light wavelengths longer than 515 nm to generate ROS. In the mechanistic study, we found that TiO2 NPs in TiO2-FeS2 NCs could absorb ultraviolet (UV) light to generate photoinduced electrons and holes for ROS generation, including ⋅O2 - and ⋅OH; FeS2 NPs efficiently harvested Vis to NIR light to generate photoinduced electrons, which then were transferred to TiO2 NPs to facilitate ROS generation. Conclusion TiO2-FeS2 NCs with superior light-induced antibacterial activity could be a promising antibacterial agent against bacterial infections.

Ovarian cancer survival and recurrence rate are drastically affected by the amount of tumor that can be surgically removed prior to chemotherapy. Surgeons are currently limited to visual inspection, making smaller tumors difficult to remove surgically. Enhancing the surgeon’s ability to selectively remove cancerous tissue would have a positive effect on a patient’s prognosis. One approach to aid in surgical tumor removal involves using targeted fluorescent probes to selectively label cancerous tissue. To date, there has been a trade off in balancing two requirements for the surgeon: the ability to see maximal tumors and the ability to identify these tumors by eye while performing the surgery. The ability to see maximal tumors has been prioritized and this has led to the use of fluorophores activated by near-infrared (NIR) light as NIR penetrates most deeply in this surgical setting, but the light emitted by traditional NIR fluorophores is invisible to the naked eye. This has necessitated the use of specialty detectors and monitors that the surgeon must consult while performing the surgery. In this study, we develop nanoparticles that selectively label ovarian tumors and are activated by NIR light but emit visible light. This potentially allows for maximal tumor observation and real time detection by eye during surgery. We designed two generations of up-converting nanoparticles (UCNPs) that emit green light when illuminated with NIR light. These particles specifically label ovarian tumors most likely via tumor-associated macrophages (TAMs), which are prominent in the tumor microenvironment. Our results demonstrate that this approach is viable means of visualizing tumors during surgery without the need for complicated, expensive, and bulky detection equipment. Continued improvement and experimentation could expand our approach into a much needed surgical technique to aid ovarian tumor removal.

Bacterial infections are becoming a serious public health challenge because the abuse of antibiotics worldwide has caused bacterial resistance and the occurrence of superbugs. In this work, a new near-infrared (NIR) photoresponsive coating was constructed on Ti implant material, which was composed of MnO2, IR780 photosensitizer and polydopamine (PDA). Under the irradiation of 808 nm NIR light, the component of MnO2 in the coating exhibited high photothermal performance, and IR780 could be stimulated to generate ROS as a photosensitizer. The addition of PDA not only improved the cytocompatibility of the composite coating but also strengthened the light absorption ability of the system. Under the irradiation of 808 nm NIR light for 15 min, the composite coating killed 99.94% Staphylococcus aureus and 99.89% Escherichia coli on Ti plates. This rapid and highly effective bactericidal rate was ascribed to the synergistic effects of photothermal performance and photodynamic effect of the coatings excited by NIR light. Cell culture tests showed that PDA/IR780@MnO2–Ti had good cytocompatibility due to the excellent biocompatibility of PDA and nanostructured surface. Therefore, this surface system will be a very promising plateform to safely and effectively kill bacteria on biomedical devices and implants in a short time.

Bacterial contamination is a severe issue that affects medical devices, hospital tools and surfaces. When microorganisms adhere to a surface (e.g., medical devices or implants) they can develop into a biofilm, thereby becoming more resistant to conventional biocides and disinfectants. Nanoparticles can be used as an antibacterial agent in medical instruments or as a protective coating in implantable devices. In particular, attention is being drawn to photothermally active nanoparticles that are capable of converting absorbed light into heat. These nanoparticles can efficiently eradicate bacteria and biofilms upon light activation (predominantly near the infrared to near-infrared spectral region) due a rapid and pronounced local temperature increase. By using this approach new, protective, antibacterial surfaces and materials can be developed that can be remotely activated on demand. In this review, we summarize the state-of-the art regarding the application of various photothermally active nanoparticles and their corresponding nanocomposites for the light-triggered eradication of bacteria and biofilms.

In the present study, chiral Cux Coy S nanoparticles (NPs) were developed to selectively induce apoptosis of senescent cells using both an alternating magnetic field (AMF) and near infrared (NIR) photon illumination. The chiral effects on living cells were investigated, and d-Cux Coy S NPs showed about 2.5 times higher of internalized ability than l-NPs. By modifying beta 2 macroglobulin (MG), senescent cells were effectively eliminated by d-Cux Coy S NPs without damaging the activities of normal cells under AMF and photon illumination. Compared to the individual application of NIR illumination and AMF, their synergistic effect induced the production of caspase-3 with a much shorter treatment time and higher efficiency due to the more serious photon-induced cellular redox and mechanical damage of cellular skeleton. Moreover, the developed strategy was successfully used to remove senescent cells in vivo. This study developed a controllable way of regulating cell activities using chiral NPs, which will provide a valuable way for treating diseases and promoting health.

The development of a highly effective photosensitizer (PS) that can be activated with a low-power single light is a pressing issue. Herein, we report a PS for synergistic photodynamic and photothermal therapy constructed through self-assembly of poly(selenoviologen) on the surface of core-shell NaYF4:Yb/Tm@NaYF4 upconversion nanoparticles. The hybrid UCNPs/PSeV PS showed strong ROS generation ability and high photothermal conversion efficiency (∼52.5%) under the mildest reported-to-date irradiation conditions (λ = 980 nm, 150 mW/cm2, 4 min), leading to a high efficiency in killing methicillin-resistant Staphylococcus aureus (MRSA) both in vitro and in vivo. Remarkably, after intravenous injection, the reported PS accumulated preferentially in deep MRSA-infected tissues and achieved an excellent therapeutic index. This PS design realizes a low-power single-NIR light-triggered synergistic phototherapy and provides a simple and versatile strategy to develop safe clinically translatable agents for efficient treatment of deep tissue bacterial inflammations.

Exported fresh intact pineapples must fulfill the minimum internal quality requirement of 12 degree brix. Even though near-infrared (NIR) spectroscopic approaches are promising to non-destructively and rapidly assess the internal quality of intact pineapples, these approaches involve expensive and complex NIR spectroscopic instrumentation. Thus, this research evaluates the performance of a proposed pre-dispersive NIR light sensing approach in non-destructively classifying the Brix of pineapples using K-fold cross-validation, holdout validation, and sensitive analysis. First, the proposed pre-dispersive NIR sensing device that consisted of a light sensing element and five NIR light emitting diodes with peak wavelengths of 780, 850, 870, 910, and 940 nm, respectively, was developed. After that, the diffuse reflectance NIR light of intact pineapples was non-destructively acquired using the developed NIR sensing device before their Brix values were conventionally measured using a digital refractometer. Next, an artificial neural network (ANN) was trained and optimized to classify the Brix values of pineapples using the acquired NIR light. The results of the sensitivity analysis showed that either one wavelength that was near to the water absorbance or chlorophyll band was redundant in the classification. The performance of the trained ANN was tested using new pineapples with the optimal classification accuracy of 80.56%. This indicates that the proposed pre-dispersive NIR light sensing approach coupled with the ANN is promising to be an alternative to non-destructively classifying the internal quality of fruits.

Light-mediated therapy has many unique merits but monotherapy strategies rarely completely inhibit tumor growth because resistance often develops. Combination therapy is a promising strategy in oncology and has demonstrated superior safety and efficacy over monotherapy. Here, we conjugated a scintillator complex and gold nanorod nanosensitizer for dual-modal image-guided photothermal and X-ray-induced photodynamic therapy (PDT). Lanthanide complexes were successfully conjugated and offer excellent X-ray-excited optical luminescence for PDT effects. The strong near-infrared (NIR) light and X-ray absorption abilities of gold nanorods make the nanosensitizer function as both a photothermal agent for photothermal therapy and a radiosensitizer for enhanced radiotherapy. The studies in vitro and in vivo demonstrated that the nanosensitizer offers good dual-modal imaging capability and significantly suppresses tumor progression under NIR light and X-ray irradiation. This work shows the great potential of conjugating scintillator lanthanide complexes and gold nanosensitizers for multimodal image-guided therapy of deep-seated tumors.

Chronic wounds present a high risk of infection due to delayed and incomplete healing, leading to increased health risks and financial burden to health-care systems. Numerous approaches to promote wound healing have been extensively explored, especially the development of effective wound dressing materials embedded with therapeutic drug molecules. Despite advances made in this area, a remaining challenge to be addressed is the controlled, on-demand release of therapeutic molecules using noncytotoxic stimulus, for example, near-infrared (NIR) excitation. Organic chromophores have been well developed for multimodality imaging-guided photothermal therapy (PTT) due to their outstanding optical properties and excellent designability. However, the theranostic efficiencies of most currently available organic chromophores are restricted intrinsically, owing to their poor photostability or complex synthesis procedures. These drawbacks not only increase their cost of synthesis, but also cause side effects in PTT. Method: We presented a facile strategy for constructing a near-infrared (NIR)-absorbing perylenediimide structured with pH-responsive piperazine ring at the bay region. The chromophore was conjugated with carboxyl-end-capped PEG as side chains that can self-assemble into nanoparticles (NPs) in aqueous solution. The NIR optical properties and photothermal conversation ability of PPDI-NPs were investigated. We then studied the imaging-guided PTT of PPDI-NPs under NIR light illumination in 4T1 cells and mice respectively. Results: The excellent photostable PPDI-NPs had near-infrared fluorescence (NIRF) emission and high photothermal conversion efficiency in acidic microenvironment. Importantly, PPDI-NPs can be utilized for the precise detection of tumors by NIRF/photoacoustic/thermal trimodality imaging. Efficient PTT of PPDI-NPs was applied in vitro and in vivo with high biosafety. Conclusion: In summary, we developed pH-responsive perylenediimide nanoparticles as multifunctional phototheranostic agent with high stability and simple synthesis procedures. This study offers a promising organic chromophore for developing phototheranostics in cancer therapy.

Both of carbon dioxide (CO2) and near-infrared (NIR) light as triggers for non-invasive remotely control are attracting wide attentions due to their good biocompatibility and easy operation. Here, CO2/NIR light dual controlled nanoparticles are proposed to remotely regulate the unzipping of dsDNA by using imidazole functionalized conjugated polymer nanoparticles (imidazole-CPNs). The dsDNA successfully coats on the shell of imidazole-CPNs to form imidazole-CPNs/dsDNA assembly due to intensively electrostatic interaction triggered by CO2. Furthermore, the unzipping process of dsDNA is remotely controlled by NIR light based on the photothermal effect, and it can be readily monitored by the fluorescence intensity of ethidium bromide (EB) and CD spectra of dsDNA. Thus, dual stimulation responsive imidazole-CPNs effectively control dsDNA unzipping under CO2 stimulus and NIR light, promising a new direction in the biological applications of DNA, such as the treatments of diseases caused by gene duplication abnormality.

Recently, the rapid development of biomaterials has induced great interest in the precisely targeted treatment of bone-related diseases, including bone cancers, infections, and inflammation. Realizing noninvasive therapeutic effects, as well as improving bone tissue regeneration, is essential for the success of bone‑related disease therapies. In recent years, researchers have focused on the development of stimuli-responsive strategies to treat bone-related diseases and to realize bone regeneration. Among the various external stimuli for targeted therapy, near infrared (NIR) light has attracted considerable interests due to its high tissue penetration capacity, minimal damage toward normal tissues, and easy remote control properties. The main objective of this systematic review was to reveal the current applications of NIR light-assisted phototherapy for bone-related disease treatment and bone tissue regeneration. Database collection was completed by June 1, 2020, and a total of 81 relevant studies were finally included. We outlined the various therapeutic applications of photothermal, photodynamic and photobiomodulation effects under NIR light irradiation for bone‑related disease treatment and bone regeneration, based on the retrieved literatures. In addition, the advantages and promising applications of NIR light-responsive drug delivery systems for spatiotemporal-controlled therapy were summarized. These findings have revealed that NIR light-assisted phototherapy plays an important role in bone-related disease treatment and bone tissue regeneration, with significant promise for further biomedical and clinical applications.

The overprescription and improper use of antibiotics have contributed to the evolution of bacterial resistance, making it urgent to develop alternative therapies and agents with better efficacy as well as less toxicity to combat bacterial infections and keep new resistance from developing. In this work, a novel light-activable nano-antibiotic platform (TC-PCM@GNC-PND) was constructed by the incorporation of gold nanocages (GNC) and two thermosensitive gatekeepers, phase-change materials (PCM) and thermosensitive polymer poly(N-isopropylacrylamide-co-diethylaminoethyl methacrylate) (PND), to realize precisely the synergy of photothermal and antimicrobial drugs. GNC exhibits an excellent photothermal effect owing to its strong absorbance in the near-infrared (NIR) region, and hollow interiors make it a favorable vehicle for loading various antibiotics such as tetracycline (TC). The release of the encapsulated drugs could be precisely controlled by NIR light through the dual thermosensitive interaction of liquid-solid transition of PCM and coil-granule transition of PND, improving efficacy and alleviating side effects with on-demand drug release. The thermosensitive hydrogel was formed in situ upon application with body temperature, enhancing retention of the antimicrobial agent in local infectious sites. Highly effective ablation of bacteria is achieved both in vitro and in periodontitis models with little toxicity owing to the synergy of photothermal effects and chemotherapeutic drug release induced by NIR. This study could provide guidance for the design of antibacterial materials and shed substantial light on synergistic treatment.

Owing to the structural controllability, pore modification and unique semiconductor property of metal-organic frameworks (MOFs), the investigation on MOFs as efficient photocatalysts for hydrogen production from water splitting under UV, visible or even NIR light irradiation have made great progress. In this review, we systematically summarized the recent advances of MOFs-based photocatalysts for hydrogen production from water, and divided them into three categories: MOFs, MOFs composites and MOFs-derived photocatalysts. The high hydrogen production efficiency and possible hydrogen production mechanism of MOFs-based photocatalysts were analyzed in detail. A brief perspective was given to achieve high effective and stable MOFs-based materials for photolysis of water, providing suggestions to explore new and efficient MOFs-based photocatalysts.

A novel light-responsive nanovalve is facilely prepared on mesoporous silica (mSiO2) coated upconversion nanoparticles (UCNP) through modification of photo-cleavagable pyrenemethyl ester (Py) molecules on its surface. These Py molecules can associate with β-cyclodextrin (β-CD) molecules as gatekeepers to block the mSiO2 pore channels and entrap the loaded drugs. Upon NIR light irradiation, NIR light is upconverted to ultraviolet (UV) and visible (Vis) light by UCNP, and the modified Py molecules are cleaved by the upconverted UV light leading to the detachment of β-CD gatekeepers to trigger drug release. For the fabrication of a monitoring system, β-CD is covalently conjugated with the fluorescein isothiocyanate (FITC) as drug release indicator (FITC-β-CD). The luminescence resonance energy transfer (LRET) from UCNP to Py/FITC-β-CD before drug release results in the quenching of Vis emission of UCNP while the detachment of Py/FITC-β-CD nanovalves after drug release diminishes the LRET between UCNP and FITC-β-CD which recovers the luminescence of UCNP, the change in luminescence provides a potential capability for one to quantitatively monitor the drug release process. in vitro study shows a remarkable killing ability on HeLa cancer cell of this system.

On-fiber micro-structures are favorable platforms for sensing in aqueous environments. Durable in-liquid operation with NIR light transmitted through fused optical microknot fibers defined on adiabatic tapers is presented in straight versus folded configurations. We suggest and demonstrate two schemes for OMF deployment: folded configuration which is intended for remote sensing, and straight configuration on either hydrophilic or hydrophobic substrate, the latter of which is useful in packaged geometries. We study experimentally the OMF preparation methods in folded, straight on glass surface and straight on PDMS surfaces. The persistent detection of NIR light transmission resonances in the presence of volatile liquid capabilities are experimentally demonstrated in the various configurations.

Nanodevices have potential as intelligent sensing systems for detection of microRNAs (miRNAs) in living cells. However, the resolution offered by "always active" nanodevices is often insufficient to manipulate miRNA sensing with high spatiotemporal control. In this work, using DNA nanotechnology we constructed an activatable DNA nanodevice programmed to detect miRNAs in vitro and in vivo with the high spatial and temporal precision of NIR light. Our nanodevice is functionalized on the surface of upconversion nanoparticles (UCNPs) with a rationally designed DNA beacon that displays UV light-activatable miRNA sensing activity. The UCNPs absorb deep-tissue-penetrable NIR light and emit high-energy UV light locally, which serve as transducers to operate the nanodevice in the NIR window. The nanodevice can naturally enter cells and enable remote regulation of its fluorescent imaging activity for miRNAs in living cells by NIR light illumination in a chosen place and time. Furthermore, we demonstrate that the nanodevice can be expanded to activatable imaging of intratumoral miRNAs in living mice. This work illustrates the potential of DNA nanodevices for miRNA detection with high spatiotemporal resolution, which could expand the toolbox of technologies for precise biological and medical analysis.

Remote, rapid, and ink-free printing/erasure on fabrics has great potential to revolutionize specialized clothing in numerous applications including fashion/aesthetic and security fields, but the construction of such smart fabrics has not been realized due to underlying obstacles in obtaining suitable photoreversible color-switching systems (PCSS). To address this problem, we have prepared TiO2- x nanorods as photocatalytic and photothermal component. With redox dyes as reversible color indicators and hydroxyethyl cellulose (HEC) as polymer matrix, TiO2- x/dye/HEC-based PCSS is coated on poly(dimethylsiloxane)-treated cotton fabric. Under 365 nm light irradiation, discoloration occurs in 180 s, resulting from the efficient photocatalytic reduction of the dye. On the contrary, when the colorless fabric is irradiated by 808 nm light, recoloration occurs in a very short time (∼100 s), far lower than the traditional heating mode (30-8 min at 90-150 °C). This rapid recoloration should be attributed to the localized high temperature (164.3-184.5 °C) induced by photothermal effect of TiO2- x. Particularly, when TiO2- x/dye/HEC-based PCSS is extended to coat commercial clothes (such as T-shirts), red/green/blue figures/letters can be rapidly and remotely printed by UV-light pen and then erased by near-infrared light, with high cycle stability. Therefore, such rewritable smart fabric represents an attractive alternative to regular clothes in meeting the increasing aesthetic or camouflage needs.

The use of cheap and safe near-infrared (NIR) light is still the subject of intense research efforts but remains a huge challenge due to the associated low photon energy (wavelength from 0.78 to 2.5 µm). In this study, a series of 17 NIR dyes mainly based on a well-established cyanine scaffold is proposed. Remarkably, 11 of them were never synthesized before. Markedly, noncharged structures, negatively charged cyanine bearing Na+ as counter cation, and positively charged cyanines bearing (B(Ph)4−) or (I−) as counter anions were examined as promising NIR light photoinitiating systems. Excellent photoinitiating abilities were found for some reported dyes when used in combination with iodonium salt and amine. Markedly, photothermal effects with a huge heater behavior were also observed for different NIR dye structures. Interestingly, the synthesis of interpenetrating polymer networks (IPNs, e.g., for the polymerization of acrylate/epoxy monomer blends) can also be carried out upon NIR light with the proposed systems.

Climatic gradients frequently predict large-scale ecogeographical patterns in animal coloration, but the underlying causes are often difficult to disentangle. We examined ecogeographical patterns of reflectance among 343 European butterfly species and isolated the role of selection for thermal benefits by comparing animal-visible and near-infrared (NIR) wavebands. NIR light accounts for ~50% of solar energy but cannot be seen by animals so functions primarily in thermal control. We found that reflectance of both dorsal and ventral surfaces shows thermally adaptive correlations with climatic factors including temperature and precipitation. This adaptive variation was more prominent in NIR than animal-visible wavebands and for body regions (thorax-abdomen and basal wings) that are most important for thermoregulation. Thermal environments also predicted the reflectance difference between dorsal and ventral surfaces, which may be due to modulation between requirements for heating and cooling. These results highlight the importance of climatic gradients in shaping the reflectance properties of butterflies at a continent-wide scale.

Intelligent responsive microcarriers have emerged as a promising class of biomaterials for therapeutic delivery and tissue regeneration, since they can respond to external stimuli and release the loaded drugs in an active manner. Among various available stimuli, near‐infrared (NIR) light is particularly attractive because it can penetrate biotic tissues with sufficient intensity and minimal damage. In this work, a kind of photoresponsive delivery microcarriers (PDMs) is developed using microfluidics. The microcarriers consist of NIR‐absorbing graphene oxide, thermosensitive poly(N‐isopropylacrylamide), and biocompatible gelatin methacrylate. Under NIR light, the PDMs exhibit an evident volume shrinkage and effectively trigger the drug release. After the NIR light is switched off, the shrunken microcarriers return to their original size. This reversible process can be stably repeated for many cycles. An in vitro experiment demonstrates that the NIR‐radiated PDMs can actively release vascular endothelial growth factors and improve the tube formation of human umbilical vein endothelial cells. The results from the in vivo experiment also show an obvious photothermal effect and superior therapeutic efficacy of these PDMs in a rat model of tissue defects. These features make the PDMs an excellent drug delivery system and represent a great potential for clinical applications in tissue repair., Photoresponsive delivery microcarriers are developed using microfluidics, which consist of graphene oxide, poly(N‐isopropylacrylamide), and gelatin methacrylate. These microcarriers can actively respond to near‐infrared light and release the encapsulated drugs according to the irradiation frequency. After being implanted into a rat model of abdominal wall defects, they also exhibit potential value in tissue repair.

An on-demand drug delivery nanoplatform based on mesoporous silica (mSiO2) coated upconversion nanoparticles (UCNP@mSiO2) with a novel near-infrared (NIR) light-triggered hydrophobic-to-hydrophilic switch nanovalve was fabricated. The surface of UCNP@mSiO2 was first immobilized with hydrophobic 2-diazo-1,2-naphthoquinones (DNQ) guest molecules. After doxorubicin hydrochloride (DOX, a universal anticancer drug) was loaded into channels of mSiO2 shell, β-cyclodextrin (β-CD) host molecules with a hydrophobic cavity were added as gatekeepers to cap DNQ stalk molecules via hydrophobic affinity, which may play a role in the OFF state of the nanovalve to prevent the drug from being released. Upon 980 nm light irradiation, a NIR light-triggered hydrophobic-to-hydrophilic switch, that transformed the hydrophobic guest DNQ into hydrophilic guest 3-indenecarboxylic acid (ICA), took place so that the capped β-CD gatekeepers dissociated due to repulsion between β-CD host (hydrophobic) and ICA guest (hydrophilic), activating the ON state of the nanovalves to release drug. The in vitro studies prove that the nanoplatform enables on-demand drug release to efficiently kill HeLa cell upon NIR light regulation. The in vivo experiment results further confirm that the nanoplatform with such fabricated nanovalves is able to inhibit tumor growth in mice. The designed nanovalves based on the novel NIR light-triggered hydrophobic-to-hydrophilic switch strategy therefore may shed new light on future development of on-demand cancer therapy.

Early diagnosis and efficient therapy are very important for cancer management. Among the various intelligent delivery systems, near-infrared (NIR) light-triggered polymeric nanomicelles (PNMs) have emerged as a promising approach for imaging and photoactive delivery. This platform can be used for spatially and temporally controlled remote release of therapeutic payload molecules under NIR light irradiation. This review discusses the design and characteristics of PNMs as well as their recent application in imaging, drug delivery, and theranostics in photothermal and photodynamic therapy. The challenges and potential of PNMs in biomedical applications are also highlighted.

ZnIn2S4 is known to be a visible light-active photocatalyst. In this work, it is shown that by substituting part of the In atoms with vanadium, the visible light range of photocatalytic activity of such material can be extended, using the so-called in-gap band scheme that has been shown to enhance photovoltaic characteristics. Characterization of this material using several techniques, complemented by DFT calculations, will support this statement. Theranostic nanoplatforms integrating simultaneously photodynamic therapy (PDT) and photothermal therapy (PTT) exhibit intrinsic advantages in tumor therapy due to distinct mechanisms of action. However, it is challenging to achieve PDT and PTT under single near-infrared (NIR) laser irradiation with a nanoplatform utilizing conventional organic photodynamic agent and inorganic photothermal agent owing to the difference in inherent excitation wavelengths. Particularly, the single NIR light (660 nm)-triggered PTT and PDT nanoplatform, constructed from chlorin e6 (Ce6) and copper sulfide (CuS) nanoparticles (NPs), has never been reported. Herein, we, for the first time, designed and established a dual-modal phototherapeutic nanoplatform that achieved both PTT and PDT under single NIR laser (660 nm) irradiation for Ce6 and CuS NPs with the strategy of core & ndash;shell structured CuS@Carbon inte-grated with Ce6. Introducing of carbon shell not only endows small CuS NPs with excellent tumor accu-mulation, but also significantly strengthens the photothermal performance of CuS NPs, realizing efficient photothermal performance under 660 nm laser irradiation. Moreover, Ce6 in carbon shell endowed the nanoplatform with photodynamic effect under 660 nm laser irradiation. The as-prepared Ce6/ CuS@Carbon nanoplatform thus achieved dual-modal phototherapy under single NIR laser irradiation, significantly inhibiting tumor growth with minimal adverse effects and superior biosafety. In this study, Li-based blue- and green-emitting core@shell (C@S) upconversion nanophosphors (UCNPs) and NaGdF4-based red-emitting C@S UCNPs were synthesized, and IR-808 dyes were conjugated with the C@S UCNPs to enhance upconversion (UC) luminescence. The surface of the as-synthesized C@S UCNPs, which was originally capped with oleic acid, was modified with BF4&minus, to conjugate the IR-808 dye having a carboxyl functional group to the surface of the UCNPs. After the conjugation with IR-808 dyes, absorbance of the UCNPs was significantly increased. As a result, dye-sensitized blue (B)-, green (G)-, and red (R)-emitting UCNPs exhibited 87-fold, 10.8-fold, and 110-fold enhanced UC luminescence compared with B-, G-, and R-emitting Nd3+-doped C@S UCNPs under 800 nm near-infrared (NIR) light excitation, respectively. Consequently, dye-sensitized UCNPs exhibiting strong UC luminescence under 800 nm NIR light excitation have high applicability in a variety of biological applications.

Photodynamic therapy (PDT), a clinically approved cancer treatment strategy, features non-invasiveness, few side-effects, high spatial resolution, etc. The advancement of PDT has been significantly restricted by the penetration depth of the excitation light. Herein, an effective fluorogen, TBD, with aggregation-induced emission characteristics (AIEgen) and high reactive-oxygen-species (ROS) generation efficiency was reported and integrated with a near infrared (NIR) light excitable upconversion nanoparticle (UCNP) to construct NIR light excitable UCNP@TBD nanocomposites. The formed nanocomposite has excellent photostability, good biocompatibility, and efficient ROS generation under NIR light excitation via Forster resonance energy transfer (FRET), enabling NIR light excited PDT. Moreover, the proposed NIR light excited PDT can break the impasse between the penetration depth and excitation volume in conventional PDT, effectively improving the anticancer therapeutic outcome. In vitro cancer cell ablation and in vivo tumor growth inhibition validated that the proposed UCNP@TBD nanocomposite is a promising NIR light excitable PDT agent with great potential for future translational research.

Porphyrin‐based architectures have emerged as excellent candidates for artificial light‐harvesting antennae. Although tetrabenzoporphyrins (TBPs), the π‐extended “bigger brothers” of porphyrins, benefit from increased absorptivity in the near‐infrared (NIR), their utilization in light‐harvesting systems is still in its infancy. Within this work, we prepared two donor‐acceptor hetero dyads consisting of hexa‐peri‐hexabenzocoronenes (HBCs) to harvest high energetic near‐UV light, and a TBP core, which serves as visible to NIR light antenna and as subsequent energy sink of the conjugate. The HBCs are either attached to the meso‐position or via a maleimide‐bridge on the periphery of the macrocycle. Upon excitation of the HBC moieties, both dyads exhibit an efficient energy transfer to the TBP core. Furthermore, the TBP‐HBC dyads show superior light‐harvesting properties compared to the respective HBC‐porphyrin reference and the naturally occurring chlorophylls a/b.

Nanomaterials-based prodrug activation systems have been widely explored in cancer therapy, aiming at overcoming limited dosage formulation, systemic toxicity, and insufficient pharmacokinetic performance of parent drugs. For better delivery control, various stimuli systems, especially nanomaterials-based ones, have come to the forefront. Among them, near-infrared (NIR) light takes advantage of on-demand/site-specific regulation and non-invasiveness. In this review, we will address the developments of nanomaterials-based prodrug over the last decade, the activation mechanisms, and bioapplications under NIR light triggering. The advantages and limitations of NIR-triggered prodrug activation strategies and the perspectives of the next-generation prodrug nanomedicine will also be summarized. Here, a novel triple-responsive graphene oxide hybrid supramolecular hydrogel based on the electrostatic self-assembly between graphene oxide and a quaternized polymer and the host-guest inclusion between α-cyclodextrins and poly(ethylene glycol) monomethyl ether (mPEG) was constructed. The quaternized polymer was synthesized by quaternization between pH-sensitive poly( N, N-dimethylaminoethyl methacrylate) and bromine end-capped poly(ethylene glycol) monomethyl ether. The supramolecular hydrogels prepared from the host-guest inclusion of poly(ethylene glycol) monomethyl ether and α-cyclodextrins would turn into a mobile sol phase when the temperature was increased above a certain temperature (Tgel-sol). Graphene oxide sheets not only acted as a core material to provide additional cross-linking but also absorbed NIR light and converted NIR light into heat to trigger the gel-sol transition. The constructed graphene oxide hybrid cyclodextrin-based supramolecular hydrogels could respond to NIR light, temperature, and pH, which could be beneficial for controlled release of cargoes and would hold great promise in the field of delivery systems.

Metal organic frameworks (MOFs) have attracted great interest as a smart drug delivery platform in the battle against cancer for their intriguing properties, such as high drug loading efficiency and microenvironment responsive degradation. Hence, the development of multifunctional MOFs integrating several theranostic elements is of crucial importance. In this work, we constructed a theranostic nanoplatform (DOX/Pd@ZIF-8) based on metal-organic frameworks encapsulating 2D Pd nanosheets and DOX using an all-in-one strategy, taking the advantages of the smart drug delivery function of MOFs and the excellent optical properties of Pd nanosheets. Polydopamine (PDA) was then introduced (DOX/Pd@ZIF-8@PDA) to improve the biocompatibility. DOX/Pd@ZIF-8@PDA exhibited excellent photothermal conversion effects and optoacoustic effects due to the strong NIR light absorption of Pd nanosheets. DOX was released in a pH-dependent manner and the release rates were accelerated in acidic solutions. However burst drug release in a relatively short period could be achieved upon 808 nm laser irradiation. Moreover, the potential of DOX/Pd@ZIF-8@PDA implemented as photoacoustic imaging contrast agents in vitro and in vivo was fully exploited and dual factor activated synergetic photo-chemo cancer therapy was successfully carried out.

The design of light-responsive peptide hydrogels with controllable drug release characteristics is still a challenge. Here, we fabricated a NIR light-responsive short peptide hydrogel by using a thermo-responsive short peptide hydrogel and PEGylated NbSe2 nanosheets, namely, the NbSe2–PEG@YD hydrogel. Under NIR light irradiation, the hydrogel began to collapse owing to the local heating, leading to the release of the cargos. The hydrogel exhibited recoverability and the release rate of the cargos could be accurately modulated by the NIR light irradiation according to the requirement. The excellent biocompatibility of this hydrogel ensured that it could be further developed as a drug carrier for clinical application.

Stimuli-responsive systems, which can be used for temporally and spatially controllable therapeutic platforms, have been widely investigated in cancer therapy. Among a wide range of stimuli-responsive nanomaterials, transition metal dichalcogenides (TMDCs) have recently attracted great attention due to their large surface-to-volume ratio, atomic thickness, and other unique physicochemical properties. Thus, TMDCs are able to be responsive to various endogenous (e.g. acidic pH and overexpressed enzymes) or exogenous stimuli (e.g. light and magnetic). The majority of TMDC-based therapeutic platforms are triggered by near-infrared (NIR) light. However, due to the limited penetration of NIR light, novel strategies that are able to ablate deep-seated tumor tissues have emerged in recent years and have been applied to design multi-stimuli-responsive nano-systems. A comprehensive overview of the development of stimuli-responsive TMDC-based nanoplatforms for "smart" cancer therapy is presented to demonstrate a more intelligent and better controllable therapeutic strategy. Furthermore, the versatile properties of TMDCs and the typical responsive principles of certain stimuli-responsive platforms are discussed for a better understanding of selected examples in this review.

Quantitative detection of external stimuli, such as light, heat, and force, is an important challenge for sensing materials. Near-infrared (NIR) light is used in a variety of fields, such as biological, physical, medical, and engineering applications. Visualization and quantitative detection of invisible NIR are significant for its safe use. Here we fabricate a paper-based device coated with polypyrrole (PPy) and layered polydiacetylene (PDA). As PPy induces evolution of heat with irradiation of NIR, the effective conjugation length of the layered PDA is changed by the dynamic structure changed through the heat conduction. The visible color change from blue to red proceeds on the paper device. The red-color intensity has a relationship with the irradiation power of NIR. These facts suggest that the irradiation time and power of NIR are quantified by the color. Moreover, the results indicate that layered PDA can be applied to the quantification of external stimuli through conversion from the desired stimulus to the detectable one.

The application of optical technologies in treating pathologies and monitoring disease states requires the development of soft, minimal invasive and implantable devices to deliver light to tissues inside the body. Here, we present soft and degradable optical waveguides from poly(d,l-lactide) and derived copolymers fabricated by extrusion printing in the desired dimensions and shapes. The obtained optical waveguides propagate VIS to NIR light in air and in tissue at penetration depths of tens of centimeters. Besides, the printed waveguides have elastomeric properties at body temperature and show softness and flexibility in the range relevant for implantable devices in soft organs. Printed waveguides were able to guide light across 8 cm tissue and activate photocleavage chemical reactions in a photoresponsive hydrogel (in vitro). The simplicity and flexibility of the fiber processing method and the optical and mechanical performance of the obtained waveguides exemplify how rational study of medically approved biomaterials can lead to useful inks for printing cost-effective and flexible optical components for potential use in medical contexts.

The development of light-controlled nitric oxide (NO)-releasing nanoplatforms that are capable of specifically targeting liver cancer cell lines and delivering an optimal amount of NO can significantly affect liver cancer therapy. Cancer cell-targeted imaging and efficient therapy are vital for tumor diagnosis and treatments. However, the development of multifunctional plasmonic nanoparticles with high-performance SERS-imaging and NIR light-triggered plasmonic photothermal therapy (PPTT) of cancer cells in both the first (NIR-I) and second (NIR-II) biological windows is still a big challenge. In the present work, gold nanostars which possess a broad NIR absorption band covering the NIR-I and NIR-II windows with good NIR SERS activity and photothermal effects were synthesized by a seed-mediated growth method, using gold chloride (HAuCl4), ascorbic acid (AA) and (1-hexadecyl) trimethylammonium chloride (CTAC) as growth solutions. The gold nanostars were further designed to be multifunctional nanoagents by labeling Raman molecules and then conjugating arginine–glycine–aspartic acid (RGD), which can serve as cancer cell-targeted SERS-imaging tags and photothermal nanoagents in both the NIR-I and NIR-II windows. The investigation of in vitro SERS-mapping and PPTT of the A549 human lung adenocarcinoma cells indicates that the proposed multifunctional gold nanostars have great potential for a wide spectrum of light-mediated applications, such as optical imaging and image-guided phototherapy in both the NIR-I and NIR-II biological windows.

Near-infrared (NIR) light-induced photothermal agent-based stimuli-responsive materials have attracted great interest from researchers. However, the highly smart release with precise control by NIR light is not yet well established because of the lack or inadequacy of intelligent release systems, such as premature release of drug and/or photothermal agent. Herein, we put forward a novel and convenient strategy to synthesize cyanine dye-functionalized polymeric materials, where cyanine dye was schemed to attach to polymeric materials by copolymerization, endowing the polymeric materials with NIR light-responsive photothermal property and fluorescent nature for real-time imaging of endocytosis and intracellular trafficking of nanovehicles. Meanwhile, the chemotherapy drug DOX was introduced into the cyanine-containing polymeric materials via formation of dynamic covalent hydrazone bond to circumvent the blood circulation barrier. The nanovehicles displayed fine pH/NIR light-controlled drug release and excellent tumor intracellular drug transposition, which were ulteriorly combined with photo-triggered hyperthermia for enhanced antitumor effect. Therefore, this multipronged design of theranostic nanovehicles with endogenous and exogenous stimuli-responsiveness provides a novel strategy to attain highly smart drug delivery for precise cancer therapy.

Light‐based technique, including optical imaging and photoregulation, has become one of the most important tools for both fundamental research and clinical practice, such as cell signal sensing, cancer diagnosis, tissue engineering, drug delivery, visual regulation, neuromodulation, and disease treatment. In particular, low energy near‐infrared (NIR, 700–1700 nm) light possesses lower phototoxicity and higher tissue penetration depth in living systems as compared with ultraviolet/visible light, making it a promising tool for in vivo applications. Currently, the NIR light‐based imaging and photoregulation strategies have offered a possibility to real‐time sense and/or modulate specific cellular events in deep tissues with subcellular accuracy. Herein, the recent progress with respect to NIR light for monitoring and modulating the spatiotemporal dynamics of cell functions in living systems are summarized. In particular, the applications of NIR light‐based techniques in cancer theranostics, regenerative medicine, and neuroscience research are systematically introduced and discussed. In addition, the challenges and prospects for NIR light‐based cell sensing and regulating techniques are comprehensively discussed., NIR light‐based technique, including optical imaging and photoreglation, has become one of the most important tools for both fundamental research and clinical practice. Herein, the recent progress of NIR light‐based cell function sensing and modulation in cancer theranostics, regenerative medicine, and neuroscience research is summarized.
