Abstract

International audience; Mycobacterium avium subsp. paratuberculosis is responsible for paratuberculosis in animals. This disease, leading to an inflammation of the gastrointestinal tract, has a high impact on animal health and an important economic burden. The environmental life cycle of M. avium subsp. paratuberculosis is poorly understood and several studies suggest that free-living amoebae (FLA) might be a potential environmental host. FLA are protozoa found in water and soil that are described as reservoirs of pathogenic and nonpathogenic bacteria in the environment. Indeed, bacteria able to survive within these amoebae would survive phagocytosis from immune cells. In this study, we assessed the in vitro interactions between several strains of M. avium subsp. paratuberculosis and Acanthamoeba castellanii. The results indicate that the bacteria were able to grow within the amoeba and that they can survive for several days within their host. To explore the presence of M. avium subsp. paratuberculosis in environmental amoebae, we sampled water from farms positive for paratuberculosis. A M. avium subsp. paratuberculosis strain was detected within an environmental amoeba identified as related to the poorly described Rosculus genus. The bacterial strain was genotyped, showing that it was similar to previous infectious strains isolated from cattle. In conclusion, we described that various M. avium subsp. paratuberculosis strains were able to grow within amoebae and that these bacteria could be found on farm within amoebae isolated from the cattle environment. It validates that infected amoebae might be a reservoir and vector for the transmission of M. avium subsp. paratuberculosis.

Abstract

National audience; Mycobacterium bovis est une bactérie zoonotique du complexe de M. tuberculosis (MTBC), endémique dans de nombreux pays, principal responsable de la tuberculose chez les bovins (bTB). A l’inverse, M. tuberculosis est à l’origine de tuberculose humaine mais rarement bovine. Un programme national d’assainissement puis d’éradication (fin des années 50) a permis à la France d’être reconnue officiellement indemne de bTB en 2000 (prévalence cheptel annuelle < 0,1%). Cependant depuis 5 ans, on observe une recrudescence de la bTB qui met en péril le statut de la France : la perte du statut indemne mènerait à d’importantes pertes économiques pour les éleveurs et l’industrie agroalimentaire, notamment par l’impossibilité des échanges commerciaux. Pour mieux comprendre la dynamique de la maladie, et afin de réaliser un suivi épidémiologique plus précis des foyers et des liens entre foyers dans l’espace et dans le temps, nous avons réalisé le génotypage de la souchothèque de M. bovis du Laboratoire National de Référence-Anses (2500 isolats, 1978-2011) par deux méthodes : le spoligotypage et le MLVA. Ce couplage permet une identification précise de souches pour comprendre les schémas d’évolution et de dissémination de la bactérie. Dans cette étude nous avons sélectionné les types de souches françaises de M. bovis les plus représentatives dans l’histoire de la bTB (souches anciennes disparues, endémiques depuis plusieurs années, et d’émergence locale). Nous réaliserons une caractérisation de SNPs sur de putatifs gènes de virulence qui pourraient être à l’origine de différents profils de pathogénicité-transmissibilité. Ces données devraient nous permettre de comprendre si l’émergence actuelle de la maladie résulte d’une augmentation de la virulence ou de facteurs humains (techniques/ type d’élevage).

National audience; De nombreux éleveurs de chèvres utilisent un traitement d'induction et de synchronisation de l’œstrus. Cette étude se propose d'évaluer son influence sur la concentration cellulaire du lait de chèvre en relation avec la production laitière, le statut infectieux des demi-mamelles, le taux butyreux et le taux protéique. Les résultats montrent une augmentation significative de la concentration cellulaire en cellules somatiques durant le cycle œstral induit, avec un effet plus marqué pour les demi-mamelles infectées que pour les saines. Ce phénomène est vraisemblablement lié en partie à une diminution de la production laitière. Après la pose des éponges à J13 et J20, le taux butyreux est significativement diminué et le taux protéique augmente. Ces résultats mériteraient d'être validés dans d'autres troupeaux, mais on peut d' ores et déjà considérer que l'effet de la synchronisation sur les concentrations cellulaires du lait de tank va dépendre de la prévalence des infections mammaires dans le troupeau considéré. La prise en compte d'une moyenne géométrique «glissante" permettrait d'en atténuer les effets négatifs pour le paiement du lait aux éleveurs.

Abstract

Poster - Session : Public Health and Mycobacterium avium subsp. paratuberculosis in the environment; International audience; Background - In the subspecies M. avium ssp. paratuberculosis (Map) two groups, known as Cattle (C) and Sheep (S), have been defined by genotyping. Recent studies show that Map C and S have different phenotypes with respect to infection of macrophages and iron metabolism. Map is adapted to the gastrointestinal tract of ruminant, but the mechanism of entry is currently unknown. In this study, we investigated the phenotype of the Map-host interaction, involving the virulence factor heparin-binding hemagglutinin (HBHA), for both groups of Map. HBHA is described in M. tuberculosis as a major adhesin required for extra pulmonary dissemination of the tubercle bacillus. Method - A large collection of Map isolates (types C & S) were genotyped by MIRU-VNTR and RFLP-IS900. The polymorphism of the hbha gene was investigated by fragment analysis using GcneN:apper technology. Structure-functions properties of recombinant HBHA (types C & S) were analyzed by Heparin Sepharose chromatography and SPR analysis based on Biacore technology. Results - In silico analyses of both types of Map have revealed two forms of hbha. This observation, showing that hbha is distinct according to the group, was confirmed using GeneMapper on 83 Map strains (65 Map C & 18 Map S) with various genotypes. We found that Map type C produces HBHA with a short C-terminal domain, while that of typeS presents a long (-terminal domain, similar to that of HBHA produced by M. tuberculosis. The purification of HBHA from Map type C and S by Heparin-sepharose chromatography highlighted a correlation between their affinities to heparin and the length of their C-terminal domain confirmed by Biacore analysis. Conclusion - We show for the first time that the types C and S of Map may be distinguished by the type of HBHA they produce, which differs in size and adherence properties. Thus, HBHA participates in the genotypic and phenotypic differences observed between the C and S types of Map.

Abstract

Members of the Mycobacterium avium complex (MAC) are ubiquitous bacteria that can be found in water, food, and other environmental samples and are considered opportunistic pathogens for numerous animal species, mainly birds and pigs, as well as for humans. We have recently demonstrated the usefulness of a PCR-based mycobacterial interspersed repetitive-unit-variable-number tandem-repeat (MIRU-VNTR) typing for the molecular characterization of M. avium subsp. paratuberculosis and M. avium strains exclusively isolated from AIDS patients. In the present study we extended our analysis, based on eight MIRU-VNTR markers, to a strain collection comprehensively comprising the other M. avium subspecies, including M. avium subsp. avium, M. avium subsp. hominissuis, and M. avium subsp. silvaticum, isolated from numerous animal species, HIV-positive and HIV-negative humans, and environmental sources. All strains were fully typeable, with the discriminatory index being 0.885, which is almost equal to that obtained by IS1311 restriction fragment length polymorphism (RFLP) typing as a reference. In contrast to IS1311 RFLP typing, MIRU-VNTR typing was able to further discriminate M. avium subsp. avium strains. MIRU-VNTR alleles strongly associated with or specific for M. avium subspecies were detected in several markers. Moreover, the MIRU-VNTR typing-based results were consistent with a scenario of the independent evolution of M. avium subsp. avium/M. avium subsp. silvaticum and M. avium subsp. paratuberculosis from M. avium subsp. hominissuis, previously proposed on the basis of multilocus sequence analysis. MIRU-VNTR typing therefore appears to be a convenient typing method capable of distinguishing the three main subspecies and strains of the complex and providing new epidemiological knowledge on MAC.
