There are many morphological and histological studies in the world to investigate the structure of the external and internal organs of insects. These studies not only serve to illuminate the biology of insect species, but also to reveal taxonomic relationships. For this purpose, the detailed structure of the testis and accessory glands that are the main organs of the male reproductive system of Bolua turkiyae Ünal, 1999 (Orthoptera: Tettigoniidae) was searched with a light microscope, scanning electron microscope, and transmission electron microscope techniques in 2019-2020 in Gazi University, Prof. Dr. Zekiye Suludere Electron Microscope Center in this study. B. turkiyae is an endemic species distributed around Bolu province in Turkey. The reproductive system of male B. turkiyae is comprised of testis, vas deferens, and accessory glands. When the cross-sections of the oval and yellow testis were examined, it was observed that they contain many follicles. There are cysts in these follicles that have the developmental stages of the sperm. From the distal ends of the follicles to the proximal ends, the germarium, growth zone, maturation zone, and transformation zone are located respectively. In these stages, spermatogonium, spermatocyte, spermatids, and finally spermatozoa are formed. Accessory glands are hollow tubular structures of two different sizes and diameters. Single-layered cubic or pyramidal cells surround the lumen in both groups of accessory glands. It is hoped that these new data will contribute to our understanding of the structure of the male reproductive system of B. turkiyae. [ABSTRACT FROM AUTHOR]

Histology has significant importance in the medical field and healthcare services in terms of microbiological studies. Automatic analysis of tissues and organs based on histological images is an open problem due to the shortcomings of necessary tools. Moreover, the accurate identification and analysis of tissues that is a combination of cells are essential to understanding the mechanisms of diseases and to making a diagnosis. The effective performance of machine learning (ML) and deep learning (DL) methods has provided the solution to several state-of-the-art medical problems. In this study, a novel histological dataset was created using the preparations prepared both for students in laboratory courses and obtained by ourselves in the Department of Histology and Embryology. The created dataset consists of blood, connective, epithelial, muscle, and nervous tissue. Blood, connective, epithelial, muscle, and nervous tissue preparations were obtained from human tissues or tissues from various human-like mammals at different times. Various ML techniques have been tested to provide a comprehensive analysis of performance in classification. In experimental studies, AdaBoost (AB), Artificial Neural Networks (ANN), Decision Tree (DT), Logistic Regression (LR), Naive Bayes (NB), Random Forest (RF), and Support Vector Machines (SVM) have been analyzed. The proposed artificial intelligence (AI) framework is useful as educational material for undergraduate and graduate students in medical faculties and health sciences, especially during pandemic and distance education periods. In addition, it can also be utilized as a computer-aided medical decision support system for medical experts to minimize spent-time and job performance losses. [ABSTRACT FROM AUTHOR]

CARCINOID , *APPENDIX (Anatomy) , *NEUROENDOCRINE tumors , *HISTOLOGY , *APPENDICITIS - Abstract
Goblet cell carcinoid (GCC) is an enigmatic and rare tumour involving the appendix almost exclusively. It is a rare, aggressive subtype of appendiceal tumours with neuroendocrine features, and controversy exists with regards to therapeutic strategy. We herein highlight a 56-year-old lady with GCC whom she presented initially with a typical presentation of acute appendicitis. The histologic hallmark of this entity is the presence of clusters of goblet cells in the lamina propria or submucosa stain for various neuroendocrine markers, though the intensity is often patchy. GCC of the appendix is rare and challenging. In advanced form, it can present with carcinoid symptoms but occasionally mimics classical presentations of acute appendicitis. A tumour measuring less than 1 cm and located at the tip of the appendix is sufficient for a simple appendicectomy which warrants a promising prognosis. [ABSTRACT FROM AUTHOR]

Aim: This study discusses the historical background of distance learning and its pros and cons from the perspective of students and teachers. This study aimed to conduct a comparison of the effectiveness of distance learning during the quarantine period at Kharkiv National Medical University (KNMU), and the national lockdown at the University of Cape Town (UCT). Material and Methods: A survey was used to investigate student’s perspectives about distance education. An online survey consisting of 19 questions was conducted on Google Forms platform among 395 students at KNMU and among 124 students of UCT obtained from Vula site statistics. Individuals’ learning progress during histological and anatomical classes using different methods such as virtual aggregators of slides and 3D programs were also observed. Results: At KNMU, most students (72.2%, n=285) agreed that the main advantages of distance education were the extra time they had to prepare for classes and revise study materials whereas the major challenge they faced while having distance education was the lack of clinical approach to patients (69.1%, n=273). At UCT, students found that the most helpful aspect of distance learning was pre-recorded lecture videos (66.9%, n=83). Conclusion: The pandemic has shown that distance learning is possible, but the question remains: has it proved to be efficient, and is it an inseparable part of the education system today? The role of distance education in the twenty-first century can be summarized as distance education programs catching huge popularity in the educational world with having and spreading discussable sides.

Aim: The research paper presents the characteristic of cytoarchitectonics of the thymus of intact white mature male laboratory rats. Topicality of the study is due to the need to clarify the data on the contribution of each type of thymus cells in the formation of its structure. The aim of the research was to determine the specifics of localization and ultramicroscopic structure of thymus cells in male mature Wistar laboratory rats. Material and Methods: The study was conducted using histological and ultramicroscopic methods on 10 mature male laboratory rats, weighing 130-150 g. Semi-thin (0.5-1 μm) and ultrathin (0.05-0.2 μm) sections were made on a microtome UMTP-4 (Ukraine), which were stained with 1% methylene blue solution with the addition of 1% sodium tetraborate solution. Histological analysis and photographic recording were performed using Olympus light microscope (Japan) and DSM 510 camcorder with magnification in 1000 times. Results: With a detailed study of the semi-thin and ultrathin sections in the thymus lobules the specifics of localization and ultramicroscopic structure of thymus cells were clearly identified. The features of localization and ultramicroscopic structure of epithelial, mesenchymal, vascular and hematopoietic thymus cells were determined from the point of view of their functional loads and interactions. Conclusion: The described structural peculiarities of the components of the thymus and their relative location in different zones reflect significant organ polymorphism, which must be taken into account in order to achieve the required level of objectivity in the result evaluation of simulated biomedical experiments.

Aim: To prospectively compare the sensitivity of Wang 22-gauge needle aspiration and 19-gauge needle aspiration with bronchial biopsy and determine whether there was difference between 22-gauge and 19-gauge needle aspirations in the diagnosis of endobronchial malignancies. Materials and methods: All patients (63 patients) in the study underwent fiberoptic bronchoscopy that included 22-gauge needle aspiration and 19-gauge needle aspiration. In 50 patients bronchial biopsy was done. The sensitivities of the individual techniques were compared. Results: Cancer was diagnosed in 48 patients by 22-gauge needle aspiration and 19-gauge needle aspiration, and in 43 patients by bronchial biopsy. The sensitivity of bronchial biopsy was 0.86. The addition of 22-gauge and 19-gauge needle aspirations to bronchial biopsy increased the sensitivity to 0.90 and 0.88, respectively, but the difference was not statistically significant (P = 1.0). The maximal diagnostic yield was obtained with the combination of needle aspirations and forceps biopsy in the detection of submucosal or peribronchial bronchogenic carcinoma (100%) (P < 0.05). Conclusion: The combination of forceps biopsy and transbronchial needle aspirations should be used for the highest rate of bronchoscopic diagnostic yield. However, there was no difference for increasing diagnostic yield in patients with visible endobronchial lesions between 22-gauge and 19- gauge needles. [ABSTRACT FROM AUTHOR]

Human growth hormone (GH) is crucial modulator of cellular metabolisms, including cell proliferation and organ development, by stimulating insulin-like growth factor-1 (IGF-1), which has various functions such as cell proliferation, tissue growth, survival, or neuroprotection. Therefore, GH is implicated as a critical player in the cell and can enhance neurogenesis and provide neuroprotection during the treatment of neurological diseases such as Parkinson’s disease (PD). In this study, the neuroprotective role of GH was investigated in rotenone-induced PD models for the first time. Both SH-SY5Y and SK-N-AS neuroblastoma cells were exposed to rotenone to mimic PD pathogenesis as stated in previous studies. Our data demonstrated that overexpression of GH led to the resistance of the SH-SY5Y and SK-N-AS cell lines to rotenone treatment. The levels of ER stress markers, CHOP, PERK, XBP-1, and ATF6, were higher in wt cells than GH+ SH-SY5Y cells. However, the level of autophagy markers LC3 increased and the levels of reactive oxygen species (ROS) decreased with the overexpression of GH. Furthermore, while rotenone significantly increased the SubG1 population in the cell cycle of SH-SY5Y wt cells, there was a minor alteration in GH+ cell population. Concomitantly, the levels of the proapoptotic marker, cleaved-PARP, and positive staining of Annexin V in SH-SY5Y wt cells were higher after rotenone treatment. Together, these results revealed that overexpression of GH enhanced the autophagy response by triggering the ER stress of SH-SY5Y cells to rotenone exposure and showed a neuroprotective effect in vitro PD models.

Plastination is a laboratory process to obtain permanent dry tissue and organ sample. That can be used in the future for investigation and educational purposes. This methodology is based on dehydration and penetration of synthetic substances such as silicon into tissue. In this study, it was aimed to deplastinate previously plastinated kidneys in order to examine them under the light microscope. In this study, 14 sheep kidneys were used, seven samples of control and seven samples of plastination-deplastination (p / d) group. Kidneys in both control and p/d groups were fixed in 10% formalin. The samples in the control group were embedded in paraffin following routine tissue processing protocol. However, the samples in the p/d group were deplastinated in alcohol and methylbenzene and embedded into paraffin. 5 μm thick sections obtained from paraffin blocks were stained with hematoxylin and eosin (H&E), periodic acid-shiff (PAS) and then examined under the light microscope. Typical histological structures were observed in the control group. Small fragments were obtained as it was challenging to obtain sections from the P/d group blocks. Morphological structures were visible with some pseudo degenerations and wrong staining. This study is the first study that demonstrates alcohol and methylbenzene deplastination can be partially successful for evaluating plastinated kidney samples under a light microscope. However, we believe that the kidney may have limitations due to its wide parenchyma compared with literature conclusions. Nevertheless, more studies are required to develop the optimum protocols., Plastinasyon, gelecekte araştırma ve eğitim amaçlı kullanılabilecek kalıcı kuru doku ve organ örnekleri elde etmeye yönelik bir laboratuvar işlemidir.

Neoadjuvant chemotherapy (NACT) in gastroesophageal junction (GEJ) and gastric cancer (GC) was shown to improve survival in recent studies. We aimed to share our real-life experience of patients who received NACT to compare the efficacy and toxicity profile of different chemotherapy regimens in our country. This retrospective multicentre study included locally advanced GC and GEJ cancer patients who received NACT between 2007 and 2021. Relation between CT regimens and pathological evaluation were analysed. A total of 794 patients from 45 oncology centers in Turkey were included. Median age at the time of diagnosis was 60 (range: 18-86). Most frequent NACT regimens used were FLOT (65.4\%), DCF (17.4\%) and ECF (8.1\%), respectively. In the total study group, pathological complete remission (pCR) rate was 7.2\%, R0 resection rate 86.4\%, and D2 dissection rate was 66.8\%. Rate of pCR and near-CR (24\%), and R0 resection (84\%) were numerically higher in FLOT arm (p > 0.05). Patients who received FLOT had also higher chemotherapy-related toxicity rate compared to patients who received other regimens (p > 0.05). Median follow-up time was 16 months (range: 1-154 months). Estimated median overall survival (OS) was 58.4months (95\% CI: 35.2-85.7) and disease-free survival (DFS) was 50.7 months (95\% CI: 25.4-75.9). The highest 3-year estimated OS rate was also shown in FLOT arm (68\%). We still do not know which NACT regimen is the best choice for daily practice. Clinicians should tailor treatment regimens according to patients' multifactorial status and comorbidities for to obtain best outcomes. Longer follow-up period needs to validate our results.

Aim: COVID-19 pandemic had a significant impact on the way medical education is delivered. In most countries education was provided remotely. In this study, we aimed to study whether social and online habits, computer readiness, online communication skills, and Internet Use Disorder (IUD) were significant characteristics affecting the preferences of medical students for online or traditional classroom teaching in histology and embryology. Material and Method: In 2019-2020, the fall semester for Turkish universities was face-to-face. Due to the COVID-19 pandemic, the spring semester of that academic year was online. We asked 1,144 public and private university students in Istanbul about demographic characteristics and their preferences in learning and general living, using two subscales of the Online Learning Readiness Scale (OLRS) and the Young Internet Addiction Scale. We compared online and face-to-face success levels using quiz results. Results: Students generally preferred traditional lectures. However, those with higher OLRS scores preferred online lectures and learning materials and were more adapted to online living. Internet use disorder did not correlate with online lecture preferences. The results from private and public universities were not considerably different. The success levels after online lectures were better than after face-to-face lectures, which suggests that the preferences and satisfaction of students are different from their learning achievement. Conclusion: Preferences of students should be the priority in planning medical education. They want online lectures available as complementary to face-to-face lectures. Therefore, instructors should improve their online communication and teaching skills to enhance students’ online lecture satisfaction. The IUD does not possess a risk in this condition. Contrarily, regularly planned complementary online courses will enhance students online learning habits, online readiness and achievement and enables a more effective continuation of education in cases of natural disasters and pandemics. Objectives: The aim of this study was to examine levator palpebralis superior muscle histologically in patients with congenital blepharoptosis and to investigate the relationship between these findings and age, sex and degree of blefaroptosis in this patient group. Materials and methods: Levator muscle of 13 patients with congenital ptosis, who had applied to Dicle University Medical Faculty Ophthalmology Clinic and had undergone levator palpebralis superior muscle resection between january 2009-january 2010, has been examined histopathologically in Histology and Embriology Deparment. During preoperative period, ptosis amount, levator function (LF), tear functions, Bell\'s phenomenon and jaw-winking phenomenon were evaluated. All patients underwent resection of levator palpebralis superior muscle. Received postoperative levator muscle was examined by light microscopy. Results: The average age of 9 (69.2%) male and 4 (30.8%) female cases were 10.61 ± 4.77 (4- 19) years. In histological examination, the quality and quantity of the levator muscle fibrils have been assessed. There was no relationship detected between histological features of levator palpebralis superior muscle and patient\'s age and gender (p>0.05). Patients with weak levator palpebralis superior muscle were detected to have fatty degeneration histologically. The higher the levator palpebralis superior muscle function revealed decreased fatty degeneration and increased skeletal muscle fibrils. Conclusion: More ultrastructural studies in larger populations are needed to support the relationship between structure and function of levator palpebralis superior muscle in patients with congenital blepharoptosis., Amaç: Bu çalışmanın amacı konjenital blefaroptozisli olgularda levator palpebra superior kasının histolojik olarak incelenmesi ve elde edilen bulguların hastaların yaş, cinsiyet ve blefaroptozis derecesi ile olan ilişkisinin araştırılmasıdır.

Objective: Experimental studies regarding nerve regeneration use a number of methods to compose a nerve graft group for making comparisons among various peripheral nerve repair techniques. It is unclear that whether these methods used interchangeably yield similar functional, physiological and morphological results or not. The aim of this study is to demonstrate functional and morphological results of these diverse nerve grafting techniques, thereby a possible necessity of standardization. Material and Methods: Thirty-two adult male Sprague-Dawley rats were divided into four groups and used. A nerve defect measuring 1.5 cm was created in the sciatic nerve of the rat, and in group one, split ends of the defect were buried into neighbouring muscles (nerve defect group). In groups two and three, resected nerve segment was sutured to its own place in its original direction (orthotopic graft group) or after reversing it 180 degrees (inverted graft group). In group four, a sural nerve graft taken from the same leg was used to bridge the defect (distant graft group). Functional, electrophysiological and histometric results obtained at 21st week were compared among the groups in order to see whether they were similar or not. Results. There were significant differences among the groups. Regarding functional results, orthotopic and inverted graft groups gave superior results compared to nerve defect and distant graft groups. Most importantly, it is clear that different nerve grafting techniques can not be used interchangeably. Conclusion: The results of the present study suggest that the use of a constant method in order to compose a nerve graft group in experimental studies may be convenient to avoid contradictory results among the researches. In other words, standardization of nerve grafting methods in experimental studies is mandatory.

- Abstract
The study was conducted on 64 white laboratory rats (8 – intact, 56 – experimental groups). In 56 animals, stress was modeled with a sharp sound stimulus, and two populations were distinguished: "stress-tolerant" (24) and "stress-intolerant" (32). After stress, animals of the control group were fed with balanced (ordinary) food for 40 days, and those of the main group - with lowprotein food. 12 stress-tolerant and 16 stress-intolerant rats were included in each group. An "agranular" structural type of the orbital shell was detected in an intact white laboratory rat. In animals tolerant to audiogenic stress, most of the changes in the orbital shell are reversible ("reactive"), some are "limited" and only a few are irreversible. Morphologically, the sequence of "early period alteration - postalterational stabilization and initiation of recovery - complete recovery or incomplete recovery with the formation of residual symptoms" was detected in the orbital cortex in stress-resistant white laboratory rats. A high-intensity and statistically reliable direct proportional relationship between low-protein food ration and the frequency of residual morphological signs in the orbital shell under conditions of stress intolerance was determined. THREE-dimensional imaging , *CAPILLARIES , *CORD blood , *TOLUIDINE blue , *PLACENTA - Abstract
Purpose: Visualization of the three-dimensional structures is performed by examining two-dimensional sections in histology. It is not always possible to comprehend structure-function integrity. Therefore, it was planned to obtain three-dimensional (3D) models of the capillaries in the placenta terminal villus by using the program developed in the Başkent University's laboratory and used in studies in different disciplines. The program's suitability for histology studies will also be assessed. Materials and Methods: A term placenta tissue sample was obtained from a 33-year-old healthy woman. Following the plastic embedding procedure, semi-thin serial sections were taken from these blocks. The sections were stained using toluidine blue and examined and photographed by using a light microscope. The 3D modeling procedure was conducted on the light micrographs of serial sections in order to reconstruct villous capillaries. Results: 3D images were obtained from two-dimensional light micrographs by using the same program. It was determined that the vessels running parallel to each other branched harmoniously, forming a capillary network in the terminal villus. Additionally, challenges encountered during the utilization of this program in histological studies are also presented in this study. Conclusion: Fetal blood reaches every part of the terminal villus through the branching capillary network. This feature would increase the surface area of vasculo-syncytial membrane and facilitate feto-maternal exchange. This can be clearly visualized by using 3D models. Three-dimensional modeling helps to understand the structure of all organs and tissues. Given that the current program is freely accessible, it will not incur additional costs for researchers and is open to further improvement by the programmer (erhankiziltan@gmail.com). Objective: This study aimed to compare clinicopatolojic risk factors and postoperative morbidity of primary cytoreductive surgery and neoadjuvant chemotherapy followed by interval cytoreductive surgery. Study Design: Between January 2019 and January 2023,103 cases whohadova-rian cancer with patients evaluated retrospectively in two groups as primary cytoreductive surgery and neoadjuvant chemotherapy followed by interval cytoreductive surgery. The cases in two groups were compared for comorbidities, CA-125 levels, hemoglobin levels, types of surgery histopathological results, length of hospital stay and postoperative complications. Results: Of the 108 patients, 49 (47.6.5%) underwent primary cytoreductive surgery, and 54 (52.4%) had neoadjuvant chemotherapy followed by interval cytoreductive surgery. Compared with primary cytoreductive group, neoadjuvant group was associated with significantly more serous histology (p=0.04), advanced stage (p=0.002), lymph node dissection operation (p=0.002), and increased CA-125 (< 0.001). 98.1% of the patients underwent maximal cytoreduction and 1.9% suboptimal surgery. There was no statistically significant difference between the neoadjuvant group and the primary cytoreduction group in terms of operation type, length of hospital stay, comorbidity and wound complications. Conclusion: The rates of maximal cytoreduction were high in ovarian cancer patients in the geriatric age group. There was no difference between neoadjuvant chemotherapy followed by interval surgery group and received primary cytoreductive surgery groups in terms of maximal cytoreduction and postoperative complications. In order to minimize surgical outcomes and postoperative morbidity, it may be important to examine and treat patients in centers specialized in gynecology oncology in tertiary centers. POLYMERASE chain reaction , *CYTOMEGALOVIRUSES , *ULCERATIVE colitis - Abstract
Objective: To show the importance of tissue Cytomegalovirus (CMV) PCR to diagnose CMV infection in patients with treatment-resistant ulcerative colitis. Methods: Fifteen patients who had previously been diagnosed as ulcerative colitis with clinical, endoscopic, histological, and radiological criteria and who had referred to our clinic with acute exacerbation who were diagnosed with CMV colitis and received CMV treatment, were evaluated retrospectively. After the demographic and clinical characteristics of the patients were noted, Mayo scores were evaluated to determine ulcerative colitis activity. Patients' age, gender, laboratory values, type of colitis, and Mayo scores were recorded. Biopsy results, CMV IgM, serum, and tissue CMV DNA levels were noted. Descriptive statistical parameters of the demographical data were calculated. Results: Eleven of the patients were male, and four were female. The mean age of the patients was 42±11,3. The mean duration of the disease was 28±46 months. Twelve patients had pancolitis, and three patients had distal colitis. All patients' Mayo score was 12 points. All patients were steroid-resistant, and none had previously received biological agent therapy. 7 of 15 patients had serum CMV DNA levels over 1000 copies/mL. Tissue CMV DNA levels of 8 patients were found higher than 250 copies/mg, although serum CMV DNA levels were below 1000 copies/mL. Pathology samples of 11 patients were evaluated, and CMV inclusion bodies were not detected. All patients received ganciclovir for CMV treatment. After treatment, CMV DNA of all patients was negative, and diarrhea and inflammation markers were reduced. Conclusion: For the diagnosis of CMV, CMV-PCR in colon tissue specimens should also be considered in addition to endoscopic appearance and serum CMV DNA levels. [ABSTRACT FROM AUTHOR]
