Species of the Microcystis genus are the most common bloom-forming toxic cyanobacteria worldwide. They belong to a clade of unicellular cyanobacteria whose ability to reach high biomasses during blooms is linked to the formation of colonies. Colonial lifestyle provides several advantages under stressing conditions of light intensity, ultraviolet light, toxic substances and grazing. The progression from a single-celled organism to multicellularity in Microcystis has usually been interpreted as individual phenotypic responses of the cyanobacterial cells to the environment. Here, we synthesize current knowledge about Microcystis colonial lifestyle and its role in the organism ecology. We then briefly review the available information on Microcystis microbiome and propose that changes leading from single cells to colonies are the consequence of specific and tightly regulated signals between the cyanobacterium and its microbiome through a biofilm-like mechanism. Prevailing social, economic and political ideas and paradigms constitute the lens through which scientists observe, assess, and understand the world. This affects how we interpret and understand the mechanisms governing the interaction between organisms and has led, in most cases, to dominant explanations and paradigms that are difficult to overthrow. This is the case of ecological theory, whose perspectives have followed the rationale of societal changes. From the industrial revolution to very recently, species competition for resources was regarded as one of the main drivers of species interactions. Nowadays, a new and rapidly growing way of thinking emerged, fueled by the high sequencing capacities, ultra-resolution microscopy and the slowly growing number of different social and gender perspectives participating in ecological studies: that living beings are not just single organisms interacting with other single organisms, but complex communities of macro- and microorganisms living and evolving together. The information emerging from this field is bringing new light to previously disregarded aspects of the ecological interactions that, in our opinion, will change the main paradigms in ecology. As members of a South American scientific network of Aquatic Microbial Ecology (MicroSudAqua), here we propose to explore alternative explanations for ecological observations, searching for new traits accounting for cooperation between microorganisms as a fundamental evolutionary and ecological strategy. Fildes Peninsula, on King George Island, has been greatly influenced by recent rapid climate warming. Lakes are pervasive features of Fildes Peninsula landscapes, some of which are used as water sources for Antarctic stations. We studied seven Fildes Peninsula lakes to explore differences among lakes and between seasons in phytoplankton and bacterioplankton communities. We measured environmental variables, analysed pigments using high-performance liquid chromatography and examined bacterial DNA through high-throughput sequencing of the 16S rRNA gene. The main driver structuring microbial communities was the season (i.e. spring vs autumn). Chlorophyceae were the dominant phytoplankton group in all lakes and both seasons. Indicator bacteria for each season were identified, including Flavobacterium , Polaromonas and Oxalobacteraceae as indicators of spring conditions under thick ice, whereas Frankiales and Verrucomicrobia were indicator species of autumn following the ice-free summer. The indicator species for spring are generally observed in oligotrophic conditions, whereas many of the autumn indicators are commonly found in soils. There were lesser between-lake differences in microbial communities in autumn, at the end of the open-water period, than in spring at the end of the ice-covered period. The seasonal differences in community structure and activity of the planktonic bacterial assemblage were investigated in the freshwater and brackish-water zones of a shallow coastal lagoon of the southwestern Atlantic Ocean. It was found that the pelagic zone of the studied lagoon harbored repeated short-term blooms of single bacterial species.

Raphidiopsis raciborskii is a toxic, invasive bacteria with a defined biogeographic pattern attributed to the generation of ecotypes subjected to local environmental filters and to phenotypic plasticity. The interactions taking place between the cyanobacterium and the other bacteria inhabiting the external polysaccharide-rich matrix surrounding the cells, or phycosphere, may be ecotype-specific and would have different influence on the carbon and nutrient cycling in the ecosystem. Here, we describe the bacterial community or microbiome (assessed by 16S rRNA metagenomics) associated to two R. raciborskii strains that have been described as different ecotypes: the saxitoxin-producer MVCC19 and the non-toxic LB2897. Our results showed that both ecotypes share 50% of their microbiomes and differ in their dominant taxa. The taxon having the highest abundance in the microbiome of MVCC19 was Neorhizobium (22.5% relative abundance), while the dominant taxon in LB2897 was the Planctomycetes SM1A02 (26.2% relative abundance). These groups exhibit different metabolic capabilities regarding nitrogen acquisition (symbiotic nitrogen-fixing in Neorhizobium vs. anammox in SM1A02), suggesting the existence of ecotype-specific microbiomes that play a relevant role in cyanobacterial nicheadaptation. In addition, as saxitoxin and analogues are nitrogen-rich (7 atoms per molecule), we hypothesise that saxitoxin-producing R. raciborskii benefits from external sources of nitrogen provided by the microbiome bacteria. Understanding the mechanisms underlying community assembly helps to define success and susceptibility to biological invasions. Here, we explored phytoplankton community assembly following niche and neutral paradigms and using a trait‐based approach. Under the hypothesis that the morphology‐based functional groups (MBFG) clusters species with similar niche, we analysed how trait‐related differences in fitness influence dominance of an invasive species. This was based on literature review, field data and model simulations. We predict that invading species can be dominant if: 1) do not belong to the local MBFG but use unexploited areas of the niche, or 2) belong to the resident MBFG but exhibit a higher fitness due to a particular combination of traits. The invasive dinoflagellate Ceratium furcoides was used as the model species to evaluate these hypotheses, its morphological (e.g. volume) and physiological (e.g. growth rates) traits were compared with species from the same (V: photosynthetic flagellates) and different (VII: colonial cyanobacteria) MBFG. Fitness was estimated using models parametrized with MBFG rates (R*, ability to draw down phosphate) under different environmental conditions (i.e. flushing). Results contributed to support both hypotheses. First, the alternation of C. furcoides and cyanobacteria dominance was explained by the use of different niches. Secondly, species from MBFG V were dominant under similar environments. Within this group V C. furcoides showed higher fitness under low flushing and high predation, advantage provided by a distinctive combination of traits. The application of trait‐based approaches to represent the niche and estimate fitness along environmental gradients was useful to evaluate community assembly and can be used to predict the dominance of microbial species invasions. Ceratium furcoides is an invasive freshwater dinoflagellate that in the last three decades has expanded its geographic distribution in South America, being recently found in Paraná River floodplain (Argentina). Despite growing concern about the presence and impacts of this invader, information regarding genetic diversity in the Southern Hemisphere is missing. This work constitutes the first phylogenetic characterization of Ceratium populations of South America, particularly, from the Paraná system. After taxonomic identification as C. furcoides based on morphological traits, two sequencing‐based approaches were applied using the ribosomal 18S gene: Sanger sequencing to isolated individuals and high‐throughput amplicon sequencing (HTS) to environmental DNA. The sequence of C. furcoides obtained shared 100% identity to Asian sequences, and formed a highly supported clade in the constructed reference phylogenetic tree. HTS helped to recover low‐frequency genetic variants suggesting the presence of different population of C. furcoides, and to alert potential invasion in its early stages. [ABSTRACT FROM AUTHOR]

Glyphosate-based herbicides are widely used for several crops, such as transgenic soybean and forestry. The aim of this study was to determine the effect of glyphosate on the community structure of riverine bacterioplankton and to evaluate the potential of bacterioplankton to degrade the herbicide. River water to which 13C-labelled glyphosate (10, 100 µg L–1) was added or not (control) was incubated for 6 days at the temperature measured in situ (20°C). Significant differences in bacterioplankton community composition, as assessed by microfluidics-based automated ribosomal intergenic spacer analysis, were found among treatments, with differences in the presence of 100 µg L–1 of glyphosate being more pronounced, namely significant decreases in bacterial richness and diversity. The glyphosate degradation product aminomethylphosphonic acid (AMPA) was detected, accounting for 1.2% of glyphosate conversion in water with 100 µg L–1 of 13C-labelled glyphosate, together with a significant enrichment of 13C in the bacterial biomass. These findings suggest that glyphosate had a direct detrimental effect on most bacterioplankton taxa, but enriched those that were able to degrade the herbicide. Together, the results indicate that glyphosate degradation in the river assessed would be a slow process (months–years), taking place through the AMPA degradation pathway and meaning glyphosate accumulate in the ecosystem. After incubating river water with carbon-13-labelled glyphosate for 6 days, we found significant shifts in bacterioplankton community composition. Bacterial richness and diversity were decreased in the 100 µg L–1 glyphosate treatment, where 1.2% of glyphosate was converted to AMPA and a significant enrichment of carbon-13 in the bacterial biomass was detected. The aim of this study was to determine the origin (human, bovine or porcine) and the concentration of the fecal sources of contamination in waters from Santa Lucía basin and Uruguay River in Uruguay by using host-specific viral markers (adenoviruses and polyomaviruses) as microbial source tracking (MST). Between June 2015 and May 2016, monthly collections of surface water samples were performed in six sites in Santa Lucía basin and four sites in Uruguay River (n = 120 samples). Viral concentration was carried out using an absorption-elution method. Detection and quantification of human and porcine adenovirus (HAdV and PAdV, respectively) and human and bovine polyomavirus (HPyV and BoPyV, respectively) were performed by quantitative PCR (qPCR). To evaluate the infectivity of circulating HAdV, an integrated cell culture-qPCR (ICC-qPCR) was used. A logistic regression analysis was carried out to estimate the influence of environmental variables on the virus presence in surface waters. Overall, HAdV was the prevalent (18%; 21/120) followed by BoPyV (11%; 13/120) and HPyV (3%; 3/120), whereas PAdV was not detected in this study. The mean concentration ranged from 1.5 × 104 genomic copies/L (gc/L) for HAdV to 1.8 × 102 gc/L for HPyV. Infective HAdVs were observed in two out of ten analyzed samples. A significant effect of environmental temperature (p = 0.001) and river (p = 0.012) on the presence of human viruses was found. These results suggest that fecal contamination could affect the water quality of these rivers, showing deficiencies in the procedure of sewage discharge from regional cities, livestock and dairy farms. Among the bacterivorous protists, heterotrophic nanoflagellates (HNFs) are considered to be the main grazers of bacteria in freshwaters due to their size-selective grazing. In this work, we assessed the change of a riverine bacterial community in controlled incubations, where HNFs’ predation pressure was initially released through filtration. Filtration did not prevent the passage of cysts, which grew in the enrichments afterwards. Data on the composition of the bacterial community were gathered by Catalyzed Reporter Deposition FluorescentIn situHybridization (CARD-FISH) using 16S probes targeting phylogenetic groups. Bacterial cell size was also examined using image analysis. Overall, the initial filtration directly (through release of predation pressure) or indirectly (through competition among bacterial groups) affected the bacterial community composition. When nanoflagellate abundance rose, a reduction of bacterial abundance and changes in cell size distribution were observed.Gamma-ProteobacteriaandActinobacteriawere the groups showing the greatest reduction in abundance.Beta-Proteobacteriashowed a reduction of cell size and were found in aggregates.Alpha-ProteobacteriaandActinobacteriadeveloped two distinct filamentous morphotypes: short, segmented rods and long chains of rods. Our results showed that the release of the predation pressure and the successive rise of the nanoflagellates changed the bacterial community in terms of composition at large phylogenetic scale. HNF grazing is highly group-specific and seems to reconstruct the community based on cell size, and thus, not only drastically changing the bacterial community composition, but also increasing its functional diversity. We combined the use of polyclonal antibodies against saxitoxin with catalyzed reporter deposition to detect production of saxitoxin by the cyanobacterium Cylindrospermopsis raciborskii . The procedure is simple, allows detection of intracellular saxitoxin in cyanobacteria filaments by confocal laser microscopy and is a promising tool to study toxin production and metabolism. Ancient DNA ( aDNA) analysis of lake sediments is a promising tool for detecting shifts in past microbial assemblages in response to changing environmental conditions. We examined sediment core samples from subtropical, freshwater Laguna Blanca (Uruguay), which has been severely affected by cultural eutrophication since 1960 and where cyanobacterial blooms, particularly those of the saxitoxin-producer Cylindrospermopsis raciborskii, have been reported since the 1990s. Samples corresponding to ~1846, 1852, 2000 and 2007 AD were selected to perform denaturing gradient gel electrophoresis ( DGGE) analysis of the 16S-23S rRNA intergenic transcribed spacer (ribosomal ITS) to compare their prokaryotic assemblage composition. Each stratum showed different ITS patterns, but the composition of 21st century samples was clearly different than those of mid-19th century. This compositional change was correlated with shifts in sediment organic matter and chlorophyll a content, which were significantly higher in recent samples. The presence of saxitoxin-producing cyanobacteria was addressed by quantitative real-time PCR of the sxtU gene involved in toxin biosynthesis. This gene was present only in recent samples, for which clone libraries and ITS sequencing indicated the presence of Cyanobacteria. Phylogenetic analyses identified C. raciborskii only in the 2000 sample, shortly after several years when blooms were recorded in the lake. These data suggest the utility of aDNA for the reconstruction of microbial assemblage shifts in subtropical lakes, at least on centennial scales. The application of aDNA analysis to genes involved in cyanotoxin synthesis extends the applicability of molecular techniques in palaeolimnological studies to include key microbial community characteristics of great scientific and social interest. Proteus mirabilis is an opportunistic pathogen that frequently causes complicated urinary tract infections. Among a wide spectrum of potential virulence factors, outer membrane proteins ( OMPs) are critical for bacterial interactions and survival in different environments. In this work, we used a proteomic approach to assess P. mirabilis in vivo OMPs expression compared to in vitro, including iron replete and iron-restricted conditions. Three putative iron receptors, IreA, PMI0842, and PMI2596, were detected both in bacterium grown in vivo and in vitro under iron-restricted conditions. A prophage gene product, PMI1721, was detected only on in vivo growing bacterium, suggesting a potential role yet to be disclosed on the surface of P. mirabilis. Plasminogen, a host protein, was co-purified with OMPs of in vivo grown bacteria, which is in accordance with previous observations and suggests that plasminogen bound to P. mirabilis surface may be associated to virulence as seen in other bacterial pathogens. Western blots using sera of experimentally challenged mice showed that iron-regulated proteins are expressed and highly immunogenic during infection. This work confirms observations made by others for P. mirabilis and reveals details not yet described, suggesting new aspects of the bacterium pathogenesis that remain unknown. American foulbrood is a severe bacterial disease affecting larvae of the honeybee Apis mellifera and it is caused by Paenibacillus larvae larvae. The disease is present worldwide and cases have been reported in almost all the beekeeping regions of the five continents. During 2001 and 2002 we carried out a nationwide study to assess the presence and amount of P. l. larvae spores in honey samples from Uruguay, combining classic bacteriological, and molecular approaches. The distribution of P. l. larvae spores in honey of the whole country showed a clear pattern and may provide useful data for a control and prevention strategy of American foulbrood.
