Introduction: Alzheimer's disease is a neurodegenerative disease characterized by progressive deterioration of superior brain functions especially memory which generally affects older adults. Animal models of Alzheimer's are partially representative of the disease conditions under study since there is no model that fully incorporates all the characteristics that are present in humans, so similar conditions to AD in animal models should not be actually called Alzheimer's disease; however, these models are very useful for the study of several alterations involved in the pathophysiology of AD, including mainly the deterioration of memory capacity. The objective of this article is to determine the diversity of animal models of Alzheimer's disease, biological plausibility and the main affective disorders and behavior studied. Method: The keywords “animal models of Alzheimer's disease + mood/affective disorders” were placed in the Google scholar and NCBI databases, the first 5 pages of the search were considered. Results: 7 animal models were found that are used to model the disease and 5 for affective disorders. Conclusion: Among the animal models found, the rat model has been the most studied in terms of mood/affective disorders. We believe that more studies are needed where molecular changes are correlated with animal behavior of AD. The mood disorders studied are depression, anxiety, dysthymia, bipolar disorder, and drug-induced affective disorders. Resumen: Introducción: La enfermedad de Alzheimer es una enfermedad neurodegenerativa caracterizada por el deterioro progresivo de las funciones cerebrales superiores, especialmente la memoria, que generalmente afecta a los adultos mayores. Los modelos animales de la enfermedad de Alzheimer son parcialmente representativos de las condiciones de la enfermedad en estudio, ya que no existe un modelo que incorpore completamente todas las características que están presentes en los humanos, por lo que condiciones similares a la EA en modelos animales no deberían llamarse enfermedad de Alzheimer; sin embargo, estos modelos son de gran utilidad para el estudio de diversas alteraciones involucradas en la fisiopatología de la EA, incluyendo principalmente el deterioro de la capacidad de memoria. El objetivo de este artículo es determinar la diversidad de modelos animales de la enfermedad de Alzheimer, la plausibilidad biológica y los principales trastornos afectivos y de conducta estudiados. Método: Las palabras clave “modelos animales de la enfermedad de Alzheimer + trastornos del estado de ánimo/afectivo” se colocaron en las bases de datos de Google academic y NCBI, se consideraron las primeras 5 páginas de la búsqueda. Resultados: Se encontraron 7 modelos animales que sirven para modelar la enfermedad y 5 para los trastornos afectivos. Conclusión: Entre los modelos animales encontrados, el modelo de rata ha sido el más estudiado en términos de trastornos del estado de ánimo/afectivo. Creemos que se necesitan más estudios donde los cambios moleculares se correlacionen con el comportamiento animal de la EA. Los trastornos del estado de ánimo estudiados son la depresión, la ansiedad, la distimia, el trastorno bipolar y los trastornos afectivos inducidos por fármacos.

4. Distributary behavior of biphasic flows with wood load on an experimental alluvial fan

Alluvial fans are episodically affected by the distributary dynamics caused by extreme biphasic flow processes. The solid-fraction component of biphasic flows is generally represented by inorganic sediment, but these events may also carry significant amounts of large wood. The aim of the study was to assess on a physical alluvial fan model the randomness of morphodynamical processes and exposure associated to a set of specific loading conditions and at exploring how these patterns change if large wood is added to the biphasic mixture in a fixed proportion of the solid fraction. The experiments were conducted varying systematically the total volume, the sediment fraction of the biphasic mixture and the applied stream power. Two sets of experiments were run, either with and without a fixed proportion of the solid fraction constituted by large wood. Our results confirm that the exposure associated to specific loading conditions exhibits a remarkable randomness, that the applied stream power has a decisive effect on the variability of exposure and that the fixed portion of large wood strongly interfere with the distributary dynamics on alluvial fans.

RESUMEN Introducción y objetivos: Existe un creciente interés en el desarrollo de dispositivos para el tratamiento de la insuficiencia mitral (IM) de forma mínimamente invasiva. Para este propósito, disponer de un modelo experimental que reproduzca las condiciones de la enfermedad sería de gran utilidad. Nuestro objetivo fue validar un modelo experimental de IM en cerdos. Métodos: Para esta fase de creación del modelo se han utilizado 3 cerdos de raza large white, de 2 meses de edad y un peso de 25 ± 3 kg. Se provocó un infarto en el territorio de la arteria circunfleja que afectó la perfusión del músculo papilar posteromedial, y posteriormente se sometió al animal a una sobrecarga de volumen mediante creación de un shunt arteriovenoso, con la conexión de la aorta y la pulmonar mediante un tubo de dacrón. Se realizó análisis mediante ecocardiografía y resonancia magnética antes de la intervención y a las 8 semanas, y posteriormente el animal fue eutanasiado para realizar el estudio anatomopatológico. Resultados: De los 3 cerdos, 1 falleció durante la intervención por fibrilación ventricular y los otros 2 sobrevivieron al procedimiento y fueron eutanasiados como estaba previsto a las 8 semanas. Hoy en día, el estudio de la dinámica estructural es fundamentalmente teórico con acceso a ciertas simulaciones vía \emph{software}, sin embargo, este proyecto intenta que el estudiante pueda entender y observar de manera física las respuestas dinámicas de modelos experimentales. Estos modelos corresponden a pórticos planos de múltiples grados de libertad, que están sometidos a aceleración en la base, la cual es generada por una mesa vibratoria. Los resultados obtenidos se compararán con teóricos. Estos fueron obtenidos basándose en la descomposición modal y en el método de Newmark para el cálculo de la respuesta dinámica, considerando variación lineal en la aceleración de cada piso. La aplicación generada, ATH Dynamic Responses, proporcionó las respuestas teóricas, mediante una interfaz gráfica amigable para el usuario. Los modelos experimentales están constituidos por dos materiales: acero inoxidable (parantes) y láminas de acrílico (pisos), y fueron ensayados sobre una mesa vibratoria. La adquisición de datos se realizó mediante acelerómetros que se colocaron en cada piso y sobre la mesa vibratoria, fueron corregidos, tanto por línea base como con el filtro pasa bajo. Los resultados obtenidos muestran que la instrumentación con una mesa vibratoria y adquisición de datos con acelerómetros proporcionan valores muy similares a los teóricos en cuanto a respuestas dinámicas y propiedades modales.

Se describe un modelo experimental para evaluar la acción de drogas, formulaciones medicamentosas o extractos vegetales sobre el tránsito gastrointestinal. Se administran los compuestos problema a 4 ratones, 30 minutos antes de comenzar el bioensayo, para asegurar la absorción. Inmediatamente antes del ensayo se suministra una solución de sulfato de bario per os y se toman radiografías seriadas cada 30 minutos. Observando el progreso de la sustancia radiopaca en el tracto gastrointestinal puede evaluarse comparativamente la acción de distintos compuestos entre sí o respecto a un grupo control. Este método posee ventajas sobre otros ya descriptos al ser incruento, reproducible, confiable y económico. Estas características permiten su empleo en forma rutinaria para detectar compuestos que evidencien una actividad farmacológica promisoria.

The construction process of a viscometer, developed in collaboration with a final project student, is here presented. It is intended to be used by first year's students to know the viscosity as a fluid property, for both Newtonian and non-Newtonian flows. Viscosity determination is crucial for the fluids behaviour knowledge related to their reologic and physical properties. These have great implications in engineering aspects such as friction or lubrication. With the present experimental model device three different fluids are analyzed (water, kétchup and a mixture with cornstarch and water). Tangential stress is measured versus velocity in order to characterize all the fluids in different thermal conditions. A mathematical fit process is proposed to be done in order to adjust the results to expected analytical expressions, obtaining good results for these fittings, with R2 greater than 0.88 in any case.

Resumen El desarrollo de esta investigación culmina en un prototipo experimental realizado por la síntesis metodológica del diseño experimental, para lo cual se consideraron como referencia los sistemas plegables. Este concepto se llevó a cabo en el proceso del prototipo de paraguas plegable. Dicha modelación tuvo como referentes históricos el teatro ambulante del arquitecto Emilio Pérez Piñero, el cual se fundamenta en el principio de una estructura plegable, y Leonardo Da Vinci, quien creó un sistema de estructuras recíprocas que desarrolló por medio de esquemas constructivos y manifiestos. Siguiendo estos conceptos se creó el prototipo, con el manejo del software WinTess, donde se calculó la estructura portante y la membrana de dicho modelo propuesto. Lo más destacado fue el proceso de construcción de este prototipo, con lo cual se generó nuevo conocimiento técnico en cuanto a la simulación y fabricación de sistemas plegables y transformables, que tienen la capacidad de plegarse, moverse y erigirse en otro lugar.Palabras clave: arquitectura plegable; arquitectura transformable; detalles constructivos; modelo experimental; simulación estructural en 3D; tensoestructuras. This concept was developed through a folding umbrella prototype. Said modeling had two historical referents: the itinerant theater of architect Emilio Pérez Piñero, based on the principle of a folding structure, and Leonardo Da Vinci, who created a system of reciprocal structures using constructive and manifest schemes. Following these concepts, the prototype was developed using the Witness software, where the supporting structure and the membrane were calculated. 14. Efficacy Evaluation of a non-human experimental model for ultrasound-guided superficial venous puncture: clinical randomized assay.

Introduction: The use of a venipuncture simulator facilitates technique learning and improves skills, which reduces the risk of venipuncture complications in humans. Objective: To evaluate the efficacy of a non-human experimental model for Ultrasound guided superficial venipuncture. Methods: We randomized 39 nurses in two groups: A and B. The training had three steps: 1 - theoretical class, 2 - practical class, with the ultrasound device and 3 - ultrasound-guided puncture training in the non-human model. The group A participated in steps 1, 2 and 3 and group B in steps 1 and 2. After training, both groups were released for ultrasound guided venipuncture. Results: The success in puncture in group A (n = 20) was 90% and in group B (n = 19) it was 68.42%. In the sum of the identification and the puncture times, the average of group A was 61.5 seconds (95% CI, 33.58; 106.95) and in group B was 148.0 seconds (95% CI, 114.54; 208.44), which was statistically significant (p = 0.007, without overlapping the interval estimates). 15. Avaliação da atividade biológica de preparações homeopáticas de antiadiponectina em camundongos submetidos a dieta hipercalórica.

Adiponectin was originally identified as a protein expressed and produced by the abdominal fat adipocytes of several animal species which function is to maintain the homeostasis of the adipose tissue vis-à-vis the body energy demands. Homeopathic preparations of endogenous substances are able to modulate several cell functions. In the present study we investigated possible effects of a homeopathic preparation of anti-adiponectin antibodies on the control of the abdominal fat by means of an experimental model. Forty Swiss male adult mice were fed a commercial dietary supplement for 1 month and 7 days and treated with anti-adiponectin 6cH per oral route. Parameters assessed were weight gain and macro- and microscopic characteristics of the subcutaneous adipose tissue and liver. The experimental group exhibited slightly less weight gain and statistically significant reduction of the adipocyte diameter. We did not detect significant inflammatory or degenerative changes in the liver. The mechanisms involved in the detected changes need to be investigated in future studies. [ABSTRACT FROM AUTHOR]

Abstract Helminth parasites have been studied as potential accumulators for different pollutants. Echinostoma paraensei is a foodborne trematode whose vertebrate host, the rodent Nectomys squamipes, is naturally exposed to environmental pesticides. However, little information exists regarding the pesticide’s effects on helminths. This study investigated the morphological effects on the trematode, E. paraensei, after experimental Roundup® herbicide exposure, in concentrations below those recommended for agricultural use. After two hours of exposure, scanning electron microscopy (SEM) showed changes to the tegument, such as furrowing, shrinkage, peeling, spines loss on the peristomic collar, and histopathological evidence of altered cells in the cecum and acinus vitelline glands with vacuoles and structural changes to the muscular layers. Glycidic content was decreased, primarily in the connective tissue. As E. paraensei is an intestinal parasite of the semi-aquatic wild rodent, N. squamipes, it is predisposed to pesticide exposure resulting from agricultural practices. Therefore, we emphasize the need to evaluate its impact on helminth parasites, due to their pivotal role in regulating host populations.

21. Experimenting on Contextualism: Between-Subjects vs. Within-Subjects.

According to contextualism, vast majority of natural-language expressions are context-sensitive. When testing whether this claim is reflected in Folk intuitions, some interesting methodological questions were raised such as: which experimental design is more appropriate for testing contextualism - the within- or the between-subject design? The main thesis of this paper is that the between-subject design should be preferred. The first experiment aims at assessing the difference between the results obtained for within-subjects measurements (where all participants assess all contexts) and between-subject measurements (where respondents evaluating different contexts are distinct groups). It is shown that the within-subject design provides data that seems to support contextualism. However, I present an alternative, invariantist interpretation of these results, therefore showing that the within-subject design does not allow to empirically distinguish between contextualism and invariantism. The second experiment further elaborates the issue of how perceiving the contrast between contexts can affect subjects' judgments - I show that certain kinds of contexts may elicit opposite intuitions when contrasted with different contexts. La prevalencia de infecciones por Toxoplasma gondii en el ser humano es de 5-90% según la zona geográfica; en Costa Rica por ejemplo, la seroprevalencia es de un 58%, por lo que es importante comprender algunos procesos inmunológicos, propios en estas afectaciones parasitarias. Con el objeto de determinar si el Trypanosoma musculi ejerce procesos de inmunosupresión sobre Toxoplasma gondii se realizó un experimento en el que se inocularon ratones Swiss con T. musculi cuatro, cinco, seis y siete días previos a la infección con T. gondii, ocurriendo la inmunosupresión cuando la inoculación con T. musculi fue hecha cuatro días antes. Además, la cantidad de tripomastigotos inoculados no influyó en el proceso. Se probaron tres cepas de T. gondii aisladas de las heces de un gato casero (TFC), de un Leopardus pardalis (TLP), de un Leopardus wiedii y de la carne de un Bos taurus (TBT). La cepa TLP resultó ser muy patógena, matando a los animales en un tiempo corto, independientemente de la inoculación con T. musculi; para las otras cepas se mantuvo el patrón de inmunosupresión en los ratones. Se reporta entonces un modelo experimental de inmunosupresión, aspecto muy en boga en este momento, por su relación con enfermedades que inducen esta condición en el ser humano, especialmente a enfermedades como el cáncer y el SIDA. Este modelo es más fácil de aplicar experimentalmente que el correspondiente con T. lewisi previamente descrito, el cual usa ratas blancas de más difícil manejo que los ratones usados en este estudio.

Se realizó un estudio experimental de casos y controles en el período comprendido entre noviembre de 2010 y mayo de 2011, con el objetivo de caracterizar el comportamiento del daño oxidativo en un modelo experimental de hiperglicemia e hiperlipidemia inducida por sacarosa en ratas wistar en la Unidad de Toxicología Experimental de la Universidad de Ciencias Médicas de Villa Clara. Para ello se utilizaron ratas machos como modelo biológico, con el fin de evaluar la actividad oxidativa inducida por sacarosa. El modelo fue establecido a partir de la administración de una dieta rica en sacarosa, para lo cual se determinaron los por cientos de hemólisis, fotohemólisis y la susceptibilidad a sustancias con efectos hemolíticos. Se encontró aumento en los valores de glucemia y triacilglicéridos de las ratas del grupo estudio, con diferencias significativas con respecto al grupo control. Asimismo, se obtuvieron diferencias significativas entre ambos grupos estudiados en los test de susceptibilidad a la hemólisis y la fotohemólisis. Se produjo incremento significativo en los niveles de glicemia y triacilglicéridos relacionados con la dieta rica en sacarosa. Se observaron incrementos significativos en los valores de los por cientos de hemólisis en el ensayo de susceptibilidad a la hemólisis y la fotohemólisis y en el primer caso se correspondió con el grupo sometido a la dieta rica en sacarosa. Se observó una fuerte correlación entre los niveles de triglicéridos del grupo estudio en el último mes de experimentación y los valores obtenidos en el test de fotohemólisis.We carried out an experimental research of cases and controls in the period from November 2010 to May 2011, with the objective of characterizing the behavior of the oxidative damage in an experimental model of saccharose-induced hyperglycemia and hyperlipidemia in Wistar rats in the Unit of Experimental Toxicology of the Medical Sciences University of Villa Clara. We used male rats as biological model, with the objective of evaluating the saccharose- induced oxidative activity. The model was set up on the basis of a diet rich in saccharose, and for that there were calculated the hemolysis, photohemolysis percentages and the susceptibility to substances with hemolytic effects. We found an increase in the glycemia and triacilglyceride values of the rats in the studied group, with significant differences in those of the control group. There were also found significant differences between the both studied groups in the tests of susceptibility to the hemolysis and photohemolysis. There it was a significant increase in the glycemia and triacilglycerides levels related with the saccharose-enriched diet. We observed significant increases in the percentages values of the hemolysis in the tests of susceptibility to the hemolysis and photohemolysis, and in the first case it corresponded with the group receiving a saccharose- enriched diet. There it was a strong correlation between the triglycerides level of the studied group in the last month of the research and the values obtained in the photohemolysis test.

La catalepsia fármaco-inducida en roedores es un modelo experimental muy utilizado para evaluar extrapiramidalismo. No existe una estandarización de la técnica, y en ocasiones las metodologías utilizadas son complicadas en su ejecución y en la evaluación de los resultados. También se han señalado diversos factores que pudieran llevar a una pseudocatalepsia o respuesta falsa positiva. El objetivo de este trabajo fue desarrollar una técnica sencilla y fiable donde no se observara pseudocatalepsia y verificar la viabilidad de su aplicación. Se describe un procedimiento utilizando ratones en un dispositivo artesanal y se muestran los resultados obtenidos luego de modificar las variables posición del animal, tiempo para realizar la observación y número de observaciones en un mismo animal. Se empleó haloperidol como droga de referencia. Se concluyó que la técnica propuesta es de fácil aplicación y consistente en sus resultados, sin que se observara pseudocatalepsia en la muestra utilizada.Drug-induced catalepsy in rodents is an experimental model commonly used to study extrapyramidalism. The technique has not been standardized, and the methodologies used are difficult to conduct and do not always facilitate the evaluation of results. Reference has also been made to various factors which might lead to pseudocatalepsy or to a false positive response. The objective of this study was to develop a simple, reliable technique in which pseudocatalepsy would not be observed, and verify the viability of its application. A description is presented of a procedure using mice in a handmade device and the results obtained after modifying the variables posture of the animal, time to make the observation, and number of observations for a given animal. Haloperidol was used as reference drug. It was concluded that the technique proposed is of easy application and yields consistent results, without any evidence of pseudocatalepsy in the sample used.

La columna vertebral del perro ha sido utilizada como modelo experimental en medicina humana, y es uno de los animales domésticos más estudiados en medicina veterinaria. s in embargo, existen limitados datos sobre la amplitud del disco intervertebral (DIV) y el foramen intervertebral (FIV). e l propósito de este trabajo es medir cuantitativamente la amplitud de los DIV y FIV , e igualmente, la distancia entre la emergencia de los nervios espinales lumbares y el borde anterior del cuerpo vertebral caudal adyacente o altura del triángulo de seguridad de Kambin (ATK). s e realizó un estudio descriptivo en cinco cadáveres de perros. s e midieron: los diametros discales ventrales, la distancia entre la emergencia de los nervios espinales y el borde anterior del cuerpo vertebral caudal adyacente (CVCA), y los diámetros de los F iv de los niveles toracolumbares T12 – L6. La amplitud del div en el borde ventral fue en promedio de 7,34 mm ( SD ± 2,209), y la distancia entre los nervios espinales y el borde posterior del DIV, de 3,56 mm ( sd 1,366). Los diámetros promedio del F iv fueron en su aspecto craneocaudal de 5,24 mm ( sd 1,838) y dorsoventral de 4,73 mm ( sd 1,098). s e evidenció para esta muestra una correlación gráfica entre la a TK y la ADIV, y una longitud máxima de todas las estructuras medidas en el segmento L4 – L5. a demás, se plantea la posibilidad del desarrollo de instrumentales intradiscales y transforaminales con un diámetro de 3 mm, hechos a medida para caninos a partir de los 10 kg de peso.

RACIONAL: O refluxo duodenogástrico tem sido implicado como potencial carcinógeno para o estômago e esôfago e é um dos fatores que podem explicar o desenvolvimento de câncer no coto gástrico. Modelos experimentais de carcinogênese no estômago ressecado ou nas gastrojejunoanastomoses estão bem definidos. OBJETIVOS: Desenvolver um modelo experimental de carcinogênese gástrica através de piloroplastia à Finney, avaliar a influência da ingestão de nitrito de sódio nesse modelo, analisar as concentrações de ácidos biliares e o valor do pH gástrico. MÉTODOS: Foram operados 110 ratos Wistar divididos em quatro grupos: Grupo I (15 ratos) submetidos à laparotomia (grupo Sham); Grupo II (15 ratos) submetidos à laparotomia (Sham) e à ingestão de nitrito de sódio na água de beber; Grupo III (40 ratos) submetidos à piloroplastia à Finney; Grupo IV (40 ratos) submetidos à piloroplastia à Finney e à ingestão de nitrito de sódio na água de beber. Após 50 semanas da operação, os ratos foram sacrificados, coletadas amostras de suco gástrico para análise do pH, dosagem dos ácidos biliares, e realizada análise histológica. RESULTADOS: A mortalidade pós-operatória imediata foi de 9% e, ao longo do experimento, 10 ratos morreram. O grupo controle (I) não apresentou lesões gástricas; o grupo controle com nitrito de sódio (II) desenvolveu papilomas no pré-estômago em 16.6%; os grupos operados com piloroplastia apresentaram adenomas em 10,3% no Grupo III e 14,2% no Grupo IV, e adenocarcinoma em 55,1%, no grupo III e 14,2% no Grupo IV. A implantação de glândulas para dentro da submucosa e muscular, na zona de anastomose (implantação mucosa), não foi critério suficiente para decidir sobre a malignidade das lesões, sendo necessária a presença simultânea de atipias celulares. A concentração de ácidos biliares do suco gástrico foi maior nos Grupos III e IV. A medida do pH gástrico não foi diferente nos grupos estudados. CONCLUSÃO: 1) A piloroplastia à Finney é modelo experimental adequado de carcinogênese gástrica; 2) ela induziu refluxo duodenogástrico; 3) o refluxo duodenogástrico atuou como carcinógeno para o estômago; 4) não houve relação entre o pH gástrico e o desenvolvimento de carcinoma; 5) o nitrito de sódio não atuou como carcinógeno para o estômago dos ratos.BACKGROUND: The duodenogastric reflux has been implicated as a potential carcinogen for the stomach and esophagus and is one of the factors that may explain the development of gastric stump cancer. Experimental models of carcinogenesis in the stomach stump or in the duodenogastric anastomosis are well defined. AIM: To develop an experimental model of gastric carcinogenesis through the Finney pyloroplasty, evaluate the influence of ingestion of sodium nitrite in this model, analyze the concentrations of bile acids and the pH of the stomach. METHODS: A hundred and ten Wistar rats were operated and divided into four groups: Group I (15 rats) underwent laparotomy (Sham group); Group II (15 rats) underwent laparotomy (Sham) and ingestion of sodium nitrite in drinking water; Group III (40 rats) submitted to the Finney pyloroplasty and Group IV (40 rats) submitted to the Finney pyloroplasty and ingestion of sodium nitrite in drinking water. After 50 weeks of surgery, the rats were sacrificed and samples collected for analysis of gastric pH, dosing of bile acids and histological analysis. RESULTS: The immediate postoperative mortality was 9%, and during the experiment, 10 rats died. The control group (I) did not show gastric lesions; the control group with sodium nitrite (II) developed papillomas in the pre-stomach in 16.6%; the operated groups with pyloroplasty had adenomas in 10.3% in Group III and 14.2 % in Group IV, and adenocarcinoma in 55.1% in group III and 14.2% in Group IV. The implementation of glands into the submucosa and muscle in the area of anastomosis (mucosa deployment) was not sufficient criterion for deciding on the malignancy of the lesions, requiring the simultaneous presence of atypical cells. The concentration of bile acids in gastric juice was higher in Groups III and IV. The measurement of gastric pH was not different in both groups. CONCLUSION: 1) The Finney pyloroplasty is suitable experimental model of gastric carcinogenesis; 2) it induced duodenogastric reflux; 3) the duodenogastric reflux served as a carcinogen for the stomach; 4) there was no relationship between pH and the development of gastric carcinoma; 5 ) sodium nitrite did not act as a carcinogen for the stomach of rats.

27. Elastase-induced pulmonary emphysema: insights from experimental models

Several distinct stimuli can be used to reproduce histological and functional features of human emphysema, a leading cause of disability and death. Since cigarette smoke is the main cause of emphysema in humans, experimental researches have attempted to reproduce this situation. However, this is an expensive and cumbersome method of emphysema induction, and simpler, more efficacious alternatives have been sought. Among these approaches, elastolytic enzymes have been widely used to reproduce some characteristics of human cigarette smoke-induced disease, such as: augmentation of airspaces, inflammatory cell influx into the lungs, and systemic inflammation. Nevertheless, the use of elastase-induced emphysema models is still controversial, since the disease pathways involved in elastase induction may differ from those occurring in smoke-induced emphysema. This indicates that the choice of an emphysema model may impact the results of new therapies or drugs being tested. The aim of this review is to compare the mechanisms of disease induction in smoke and elastase emphysema models, to describe the differences among various elastase models, and to establish the advantages and disadvantages of elastase-induced emphysema models. More studies are required to shed light on the mechanisms of elastase-induced emphysema.Diversos estímulos podem ser utilizados para reproduzir características histológicas e funcionais do enfisema humano, uma das principais causas de incapacidade e morte. Uma vez que a fumaça de cigarro é a principal causa de enfisema em humanos, estudos experimentais têm tentado reproduzir esta situação. No entanto, esse é um método dispendioso e complicado para a indução do enfisema e, alternativas mais simples e eficazes, têm sido pesquisadas. Entre essas abordagens, enzimas elastolíticas vêm sendo amplamente utilizadas para reproduzir algumas das características do enfisema humano, tais como: aumento dos espaços aéreos, influxo de células inflamatórias nos pulmões e inflamação sistêmica. Entretanto, o uso de modelos de enfisema induzido por elastase permanece controverso, uma vez que as vias de ação da doença envolvidas na indução com elastase podem diferir das que ocorrem no enfisema induzido pelo fumo. Isso indica que a escolha de um modelo de enfisema pode influenciar os resultados de novas terapias ou drogas a serem testadas. O objetivo desta revisão é comparar os mecanismos da indução da doença em modelos de enfisema por fumaça e elastase, descrever as diferenças entre os vários modelos de elastase e, estabelecer as vantagens e desvantagens dos modelos de enfisema por elastase. Mais estudos são necessários para elucidar os mecanismos relacionados ao enfisema induzido por elastase.

28. Polioencefalomalacia experimental em bovinos induzida por toxicose por enxofre

O presente trabalho teve como objetivos avaliar os sinais clínicos, as concentrações do sulfeto de hidrogênio ruminal e as alterações anatomopatológicas associadas à intoxicação experimental por enxofre em bovinos. Foram utilizados dez bezerros mestiços leiteiros, sendo que quatro bovinos ingeriram ração sem sulfato de sódio (G1) e seis consumiram ração com sulfato de sódio (G2). Exames clínicos (temperatura retal, frequência cardíaca e respiratória e motricidade ruminal) e laboratoriais (hemograma, fibrinogênio, proteína plasmática, pH do fluido ruminal, concentração do sulfeto de hidrogênio ruminal, líquido cerebrospinal e histopatológico) foram realizados. A temperatura retal, frequência cardíaca, hemograma, fibrinogênio, proteína plasmática, pH do fluido ruminal e os valores do líquido cerebrospinal estavam dentro dos valores de referência para a espécie. Taquipnéia, hipomotricidade ruminal e elevados valores de sulfeto de hidrogênio ruminal foram observados nos bezerros do grupo G2. Um bezerro do grupo G2 apresentou sinais neurológicos e lesões histopatológicas de PEM. Dois animais de cada grupo foram eutanasiados. Lesões microscópicas foram observadas nos bezerros do G2. Histologicamente as alterações observadas foram necrose neuronal cortical e lesões hemorrágicas nos núcleos basais, tálamo, mesencéfalo, ponte e bulbo. O protocolo experimental constituído por uma dieta rica em carboidrato de alta fermentação, baixa quantidade de fibra efetiva e altos níveis de enxofre (0,52%) ocasionou alterações clinicas e histológicas e elevadas concentrações de sulfeto de hidrogênio ruminal compatíveis com quadro de intoxicação por enxofre.

29. Desenvolvimento de um modelo experimental de hemodiálise em cães Development of a dog model of hemodialysis

Com o objetivo de desenvolver um modelo de hemodiálise (HD) em cães, foram estudados 18 animais, sem raça definida, machos, clinicamente sadios, com peso corporal variando entre sete e 14 kg. O acesso vascular foi obtido através de implantação do cateter de duplo lúmen em veia jugular externa. As sessões de HD, em número de cinco por animal, com até três horas de duração, foram realizadas em hemodialisadora de sistema proporcional com ultrafiltração (UF) controlada, com solução dialisante padrão e tampão bicarbonato. A UF foi ajustada para HD isovolêmica, utilizou-se perfil de sódio, e para anticoagulação heparina sódica. Os animais foram mantidos anestesiados com cloridrato de levomepromazina e propofol. Foram avaliados dados hematológicos, bioquímicos, hemogasometria, pressão arterial sistêmica e tempo de coagulação ativado. Foi observada diminuição do número global de hemácias, volume globular, hemoglobina e leucócitos. Em relação aos exames bioquímicos, houve manutenção nos níveis de sódio sérico, e quanto à hemogasometria, a manutenção da SO2. A pressão arterial sistêmica manteve-se constante. Os resultados obtidos no presente trabalho permitiram concluir que foi possível o desenvolvimento do modelo proposto e mostrou que a HD em cães é um método viável e seguro, que poderá contribuir para o tratamento clínico da insuficiência renal nesta espécie.To develop a model of hemodialysis (HD), 18 healthy dogs, without a defined breed, males, weighing 7-14 kg, were studied. A double lumen catheter was inserted into the jugular vein for vascular access. HD sessions, totalizing 5 for each dog, were performed with a proportional HD machine, controlled isovolemic ultrafiltration, standard dialysis solution and bicarbonate buffer. Sodium profile and sodium heparin were used. During HD sessions the dogs were kept anesthetized (levomepromazine and propofol). Hematological and biochemistry data, blood gas analysis, systemic arterial blood pressure, and activated clotting time were evaluated. Serum biochemistry e blood gas analysis showed, respectively, sodium and SO2 maintenance. Systemic blood pressure kept sustained during HD sessions. It could be concluded that it was possible to develop a HD dog model that is a safe and viable technique to be used in chronic renal failure patients.

Para estabelecer um modelo experimental para o estudo da etiologia, patologia e patogênese da polioencefalomalacia (PEM) em bovinos, a condição foi induzida em quatro novilhos pela administração oral de amprólio nas doses diárias de 500 e 350mg/kg de peso vivo, respectivamente por 22 e 26-28 dias. Todos os bovinos morreram espontaneamente ou foram eutanasiados in extremis após um curso clínico de 4-7 dias. Três bovinos que receberam 1.000mg/kg de amprólio e dois que receberam 500mg/kg morreram espontaneamente com quadro clínico agudo a subagudo sem desenvolverem sinais e lesões de PEM. Nos novilhos que PEM foi reproduzida, os sinais neurológicos incluíram marcada apatia, incoordenação, posição de cavalete, quedas ocasionais, hiperexcitabilidade, tremores musculares, cegueira, bruxismo, estrabismo, nistagmo, midríase, opistótono, decúbito lateral e movimentos de pedalagem. Os principais achados de necropsia eram restritos ao encéfalo e consistiam de tumefação, achatamento, amolecimento e amarelamento das circunvoluções cerebrais. Histologicamente, havia necrose neuronal segmentar e laminar (neurônios vermelhos) associada a edema, tumefação endotelial, separação das lâminas de neurônios do córtex telencefálico ou entre as substâncias cinzenta e branca e infiltração moderada a acentuada de macrófagos espumosos. Essas alterações eram mais acentuadas nos lobos telencefálicos frontal, parietal e occipital. Adicionalmente, lesões similares e moderadas foram detectadas no mesencéfalo e hipocampo. A necrose neuronal e o edema afetaram uniformemente as camadas de neurônios da substância cinzenta dos lobos telencefálicos frontal, parietal e occipital. Esse modelo experimental de PEM com administração oral de amprólio parece ser útil para o estudo da doença em bovinos, conforme observado anteriormente em ovinos.In order to establish an experimental model for the study of the etiology, pathology, and pathogenesis of polioencephalomalacia (PEM) in cattle, the condition was induced into four steers by oral administration of amprolium at daily doses of 500 and 350mg per kg of body weight respectively for 22 and 26-28 days. All steers died spontaneously or were euthanized in extremis after being sick for 4-7 days. Three steers that received the drug at 1,000mg/kg and two that received 500mg/kg died spontaneously with acute or subacute clinical signs and without lesions and signs of PEM. In those steers in which PEM was reproduced, the neurological signs included marked apathy, incoordination, sawhorse stance, occasional falls, hyperexcitability, muscle tremors, blindness, grinding of teeth, strabismus, nystagmus, mydriasis, opisthotonus, and lateral recumbency with paddling movements. Main gross lesions were restricted to the brain and included swelling, flattening, softening and yellow discoloration of the cerebral circumvolutions. Histologically, there was segmental laminar neuronal necrosis (red neurons) associated with edema, swelling of endothelial cells, cleavage of laminar neuronal layers or between gray and white matter and moderate to severe infiltration by foamy macrophages (gitter cells). These changes were more marked in the frontal, parietal and occipital telencephalic lobes. Additionally, similar and moderate lesions were detected in the midbrain and hippocampus. Neuronal necrosis and edema affected uniformly the neurons layers of the grey matter of the frontal, parietal and occipital lobes. This experimental model of PEM with oral administration of amprolium may be useful for the study in cattle, as previously observed in sheep.

Foram avaliados dois protocolos de administração, em ratos sadios, de uma solução de fatores hepatotróficos (FH), composta por aminoácidos, vitaminas, sais minerais, glicose, insulina, glucagon e triiodotironina (T3). A solução foi administrada durante 10 dias, 40mg/kg/dia, i.p., em duas, grupo 2xFH (n=15), ou três doses, grupo 3xFH (n=15), diárias. Foram observados os efeitos na proliferação celular dos hepatócitos, na angiogênese e na matriz extracelular hepática, assim como as possíveis reações adversas. Os animais dos grupos 2xFH e 3xFH apresentaram aumento da massa hepática de 30,1% e 22,5%, respectivamente, em relação ao grupo-controle (CT; n=15). O índice de proliferação hepatocelular foi maior nos grupos 2xFH (1,4%) e 3xFH (1,2%) em relação ao grupo CT (0,53%), e a densitometria relativa do fator de crescimento do endotélio vascular pelo imunoblot não revelou diferença estatística entre os três grupos. Nos grupos 2xFH e 3xFH, houve redução do colágeno intersticial em relação ao grupo CT. A solução de FH estimulou o crescimento hepático e reduziu o volume de colágeno perissinusoidal. A administração em três doses diárias resultou em mortalidade de 26,7%, possivelmente pelo excessivo estresse da manipulação e pela menor adaptação fisiológica dos ratos, o que não ocorreu nos grupos 2xFH e CT. Para esse tipo de abordagem em ratos, o procedimento experimental mais apropriado, seguro, com melhor chance de adaptação dos animais e com resultados significativos é a aplicação dos FH em duas doses diárias.

O objetivo deste estudo foi avaliar um novo modelo de lesão no ligamento suspensório (LS). Sob anestesia geral, um punch para biópsia cutânea de 0,6cm de diâmetro foi utilizado para criar uma lesão no centro do LS de ambos os membros torácicos e pélvicos, em seis equinos. Todos os animais se recuperam da cirurgia sem nenhuma complicação importante. Realizaram-se avaliações clínicas e ultrassonográficas no período pós-operatório. Durante as primeiras duas semanas, no local da lesão, houve redução no edema de leve para discreto e da dor de discreta para ausente. As lesões foram facilmente observadas ao exame ultrassonográfico 72 horas após a cirurgia como áreas anecoicas homogêneas, representando uma média (± erro padrão) de 33,5±5% da área do LS. Não houve alteração significativa na área de lesão durante as primeiras duas semanas (P=0,77). Concluiu-se que o modelo proposto foi eficiente para promover lesões controladas e homogêneas simultaneamente nos quatro LS, sem causar desconforto importante aos equinos. Esses achados, associados à possibilidade de se avaliar previamente o LS por meio de biópsia, demonstram que esse modelo de indução de lesões ligamentosas se apresenta com um método útil, principalmente se aplicado ao estudo de terapias destinadas a melhorar o processo de reparo do LS.The aim was to study a novel model of suspensory ligament (SL) lesion. Under general anesthesia, a punch for skin biopsy (0.6cm of diameter) was used to create a circular lesion in the middle of SL simultaneously in the four members of six horses. All horses recovered without any important complication. Clinical and ultrasonographic evaluations were made during the post operative period. During the first two weeks, edema changed from mild to discreet, and pain changed from discreet to none around the surgical site. The lesions were easily observed by ultrasonography 48h after surgery as a homogenous anechoic area representing a mean (± SEM) 33.5±5% of the SL area, and did not change size during the first two weeks (P=0.77). In conclusion, the proposed model was efficient to promote controlled and homogeneous lesions in the four SL simultaneously without causing important discomfort to the horses. Taking in account these findings together with the possibility of evaluation of the ligament biopsy previously to any treatment, this method may be useful especially in studies testing therapies for tissue repair.

OBJETIVO: Padronizar um modelo experimental de lesão de medula espinal em ratos Wistar, utilizaram-se um equipamento computadorizado para impacto por queda de peso e os parâmetros determinados pelo Multicenter Animal Spinal Cord Injury Study - MASCIS. MÉTODOS: Avaliaram-se 30 ratos, com idade variando entre 20 e 25 semanas de vida. O peso variou de 200 a 300 g, para as fêmeas, e de 232 a 430 g para os machos. Realizaram-se impactos com pesos de 10 g de 12,5; 25 e 50 mm de altura, controlando-se a velocidade de impacto e o coeficiente de compressão. O impacto ocorreu sobre a superfície da medula espinal na altura da décima vértebra torácica, após laminectomia. Monitoraram-se os sinais vitais e realizaram-se gasometrias previamente e posteriormente à lesão da medula. O volume de lesão foi avaliado pela análise quantitativa dos íons de sódio e potássio. RESULTADOS: Verificaram-se correlações estatisticamente significantes entre o volume de lesão e os parâmetros mecânicos. O volume de lesão provocado por queda de 50 mm de altura foi superior aos de 12,5 e 25 mm, que não diferiram entre si. CONCLUSÃO: O modelo demonstrou-se eficaz e capaz de gerar lesões medulares padronizadas em ratos Wistar.OBJECTIVE: To standardize an experimental model of spinal cord injury in Wistar rats, computerized weight fall impact equipment were used and the parameters were used determined by the multicenter animal spinal cord injury study - MASCIS. METHODS: Thirty rats were used, with age varying between 20 and 25 weeks, and weight ranging from 200 to 300g for females, and from 232 to 430g for males. The impacts were done with weights of 10g starting from 12.5, 25 and 50 mm of height, and the impact speed and compression coefficient were obtained. The impact occurred on the surface of the spinal cord at the level of the tenth thoracic vertebra after laminectomy. Vital signs were monitored and gas analysis was made before and after the spinal cord injury. The lesion volume was evaluated by the quantitative analysis of sodium and potassium ions. RESULTS: Statistically significant correlations were verified among the lesion volume and the mechanical parameters. The lesion volume caused by the fall from 50mm height was superior to that of the 12.5 and 25mm, which didn't differ from each other. CONCLUSION: The model demonstrated itself to be effective and capable of generating standard spinal cord injuries on Wistar rats.

Todos os ovinos morreram ou foram eutanasiados in extremis após um curso clínico de 3-7 dias. Os sinais clínicos incluíam depressão, incoordenação, midríase, bruxismo, cegueira e decúbito com opistótono e movimentos de pedalagem. Salivação excessiva e posição de cavalete foi observada em um ovino e mioclonias em um outro. Os principais achados de necropsia restringiam-se ao sistema nervoso central e incluíam tumefação do encéfalo com achatamento dos giros telencefálicos e hemorragias nos lobos parietal e occipital do telencéfalo; as hemorragias ocorriam também nas áreas submeníngeas da medula espinhal e do mesencéfalo. Histologicamente, havia necrose segmentar laminar de neurônios (neurônios vermelhos) associada a edema, tumefação de células endoteliais, hemorragias e infiltração por macrófagos espumosos (células gitter). Essas alterações eram mais marcadas nos lobos frontal, parietal e occipital do telencéfalo e havia uma demarcação abrupta entre as lesões e o neurópilo normal adjacente. Adicionalmente, lesões semelhantes, mas menos acentuadas, eram observadas no mesencéfalo, tálamo e hipocampo. Histologically, there was segmental laminar neuronal necrosis (red neurons) associated with edema, swelling of endothelial cells, hemorrhages and infiltration by foamy macrophages (gitter cells). These changes were more marked in the frontal, parietal and occipital telecephalic lobes and there was sharp demarcation between the lesions and the adjacent normal neuropile. Additionally, similar, but less marked lesions were seen in the mesencephalon, thalamus and hippocampus. Considering the consistent reproducible aspects of polioencephalomalacia in sheep using amprolium, this may be an useful model for the study of the disease.

No presente trabalho foi elaborada uma técnica para protocolo de colheita de medula óssea (MO) (10ml. kg-1), do osso femoral, para isolamento, quantificação e viabilidade da fração total de células mononucleares (CM). Para tanto, 40 cães machos ou fêmeas, sem raça definida, com idade aproximada de dois anos, pesando em torno de 10kg, foram submetidos a procedimento asséptico em ambiente cirúrgico para colheita de MO. Para a obtenção de uma quantidade suficiente de CM, durante o procedimento foi utilizada a agulha tipo Steis anatômica, que favoreceu a colheita de volume sangüíneo em menor espaço de tempo e não danificou a viabilidade celular. Também foi utilizado o Kit Bone Marrow collection, que teve a finalidade de filtrar as espículas ósseas, mantendo a integridade das CM colhidas durante o período decorrido para o acondiconamento do sangue. Durante o período da colheita de MO, os animais foram submetidos à collheita de sangue periférico (pré, trans e pós-operatório) para avaliações hematológicas e sofreram autotransfusão sangüínea para suprir a queda acentuada de hemoglobina ocorrida nos primeiros momentos da coletaheita. O total de MO colhida e filtrada foi colocado lentamente sob gradiente de densidade Histopaque (1.077g ml-1). O material foi centrifugado a 440 x g por 30 minutos e o anel de células foi colhido, lavado e centrifugado três vezes em meio contendo solução salina 0,9%, DMEM e soro sangüíneo autólogo estéril. Foi realizada a contagem do anel celular em câmara de Neubauer e foi verificada sua viabilidade utilizando corante vital. Neste estudo foi verificado que no volume de MO colhido foi possível obter a média de 2,57 x 10(6) (± 1,56) CM kg-1 e a viabilidade celular foi superior a 90% (96,72 ± 2,9%). Conclui-se que a técnica de colheita de MO com agulha Steis com lavagem celular no meio contendo soro autólogo e Kit Bone Marrow e agulha Steis com lavagem celular no meio contendo soro autólogo permite obter um número ideal e indicado de CM viáveis. Esse número pode ser aplicado diretamente em lesões teciduais com objetivo de potencializar o processo de regeneração.In the present research a new protocol to harvest 10ml kg-1 femoral bone marrow (BM) was developed to allow isolation, quantification and to test the mononuclear cell (MC) fraction viability. Forty male or female stray dogs, aging and weighting around two years old and 10kg respectively, were submitted to aseptic bone marrow harvest in a surgical environment. To achieve an ideal cell count of MC, an anatomical Steis needle was used during the procedure, which favored the indicated volume harvest in a shorter period of time without interfering cellular viability. A bone marrow collection kit was also used to filter bone fragments while maintaining harvested MC integrity during blood packaging. Meanwhile BM harvesting was conducted, animals peripheral blood collection was performed (pre, trans and post-operatory) to hematological evaluations and autologous blood transfusion was made to overcome the increased hemoglobin fall that takes place in the initial harvesting moments. The harvested and filtered BM was slowly placed over a Histopaque density gradient (1.077g ml-1). The material was centrifuged at 440g x for 30 minutes. The cellular ring was harvested, washed and three times centrifuged in saline 0.9%, DMEM and autologous sterile serum. Cellular ring count was conducted in neubauer chamber and its viability was performed with vital dye. In this study was possible to notice that with the harvested BM volume an average of 2.57 x 10(6) (± 1.56) MC kg-1 was obtained and the cell viability was over 90% (96.72 ± 2.9%). It was concluded that the bone marrow kit and Steis needle with autologous serum cellular wash BM harvesting technique allow an ideal MC number isolation which can be administered in tissue lesions to enhance the regeneration process.

Vários aspectos da biologia do herpesvírus bovino tipo 5 (BoHV-5) têm sido estudados em coelhos, que desenvolvem infecção aguda e doença neurológica após inoculação experimental. A infecção aguda é seguida pelo estabelecimento de infecção latente, que pode ser reativada natural ou artificialmente. Os primeiros experimentos nesta espécie estabeleceram um protocolo de inoculação e monitoramento da infecção, e caracterizaram os principais aspectos virológicos, clínicos e patológicos da infecção aguda. A patogenia da infecção aguda, desde a replicação viral nos sítios de inoculação, vias e cinética de transporte viral até o encéfalo, distribuição e replicação viral no sistema nervoso central (SNC), tropismo celular e tecidual, manifestações clínicas e patologia no SNC foram detalhadamente estudados nestes animais. Posteriormente, vários aspectos biológicos e moleculares da infecção latente também foram elucidados a partir de inoculações de coelhos. Os coelhos também têm sido utilizados para estudar o fenótipo (neuroinvasividade, neurovirulência) de isolados de campo e de cepas vacinais recombinantes, proteção por imunidade passiva, proteção vacinal, eficácia de drogas anti-virais e terapêuticas de suporte da infecção neurológica. Este modelo experimental também foi utilizado para o estudo da origem e distribuição dos estímulos elétricos produzidos durante as convulsões - uma característica da infecção neurológica pelo BoHV-5 -, e para testes de medicamentos anti-convulsivantes. Ressalvadas as diferenças que certamente existem entre bovinos - os hospedeiros naturais - e coelhos, as observações oriundas deste modelo experimental tem contribuído sobremaneira para o conhecimento da biologia do BoHV-5. O presente trabalho apresenta uma coletânea de resultados e observações, publicadas ou não pelo grupo, ao longo de mais de uma década, envolvendo inoculações de coelhos para estudar diversos aspectos da infecção pelo BoHV-5.Several aspects of the biology of bovine herpesvirus 5 (BoHV-5) have been studied in rabbits, which develop acute infection and neurological disease upon experimental inoculation. The acute infection is followed by the establishment of latent infection, which can be naturally or artificially reactivated. The first experiments in rabbits established a protocol for virus inoculation and monitoring the infection, and characterized the main virological, clinical and pathological aspects of the acute infection. The pathogenesis of acute infection, from the initial viral replication at site of inoculation, pathways and kinetics of viral transport to the brain, distribution and virus replication in the central nervous system (CNS), cellular and tissue tropism, clinical signs and CNS pathology have been extensively studied using this animal model. Subsequently, several biological and molecular aspects of latent BoHV-5 infection have also been elucidated upon inoculation of rabbits. Rabbits have also been used to investigate the phenotype (neuroinvasiveness, neurogrowth) of field isolates and recombinant vaccine candidates, protection by passive immunity, vaccine protection, the efficacy of anti-viral drugs and support therapies for neurological disease. This animal model was also used to investigate the origin and distribution of electric impulses involved in seizures - a hallmark of BoHV-5 induced neurological infection - and also to test the efficacy of anti-convulsivants. In spite of the possible differences between rabbits and cattle - the natural host of the virus - the observations taken from this experimental model have greatly contributed to the knowledge of the biology of BoHV-5 infection. The present article presents a review of the main published and unpublished results and observations by our group, comprising more than a decade of studies on the pathogenesis of BoHV-5 infection in the rabbit model.

37. Efeito da prednisona em lesão medular aguda experimental em ratos Effect of prednisone on acute experimental spinal cord injury in rats

Foram utilizados 25 ratos (Rattus novergicus) submetidos a trauma experimental da medula espinhal, empregando-se aparelho estereotáxico com um peso de 50,5g comprimindo a duramáter durante cinco minutos. Após o trauma, os animais foram divididos em cinco grupos de cinco. O grupo A (controle) recebeu placebo oito horas após o trauma; os grupos B, C, D e E receberam prednisona oito, 24, 48 e 120 horas após o trauma, respectivamente. A prednisona foi administrada na dose inicial de 2mg/kg, durante cinco dias, com diminuição progressiva até o 26º dia. Os animais foram avaliados conforme a capacidade motora, posicionamento proprioceptivo, reflexo de localização, plano inclinado e sensibilidade dolorosa. Após 33 dias da cirurgia, foram sacrificados para avaliação histológica das medulas espinhais. Observaram-se degeneração das raízes nervosas, necrose medular, inflamação local e reação glial, sem diferenças entre os grupos. Não houve correlação entre os testes neurológicos. A utilização da prednisona na lesão medular aguda não teve efeito diferenciado com o tempo e não foi prejudicial na recuperação neurológica, mesmo quando utilizada tardiamente.Twent-five rats (Rattus novergicus) were submitted to an experimental spinal cord injury by using a 50.5g stereotaxic equipament to apply pressure on the duramater during five minutes. After the spinal cord injury, the animals were randomly distributed into five groups of five animals each: one group received placebo 8 hours after injury and the remaining four groups received prednisone at 8, 24, 48, and 120 hours after injury. Prednisone was applied at 2mg/kg dosage during five days and progressively reduced until the 26th day. Animals were evaluated by motor capacity, proprioceptive positioning, locating reflex, inclined plan, and sensibility to pain. The animals were sacrificed 33 days after surgery for histological study of the spinal cords that revealed degeneration of the rootlets, necrosis, inflammatory foci, and reactive gliosis, with no significant differences among the treated groups. The administration of high dosages of prednisone to animals with acute spinal cord injury had no deleterious effects on neurological recovery, even when used late.

Malignant brain tumor experimental models tend to employ cells that are immunologically compatible with the receptor animal. In this study, we have proposed an experimental model of encephalic tumor development by injecting C6 cells into athymic Rowett rats, aiming at reaching a model which more closely resembles to the human glioma tumor. In our model, we observed micro-infiltration of tumor cell clusters in the vicinity of the main tumor mass, and of more distal isolated tumor cells immersed in normal encephalic parenchyma. This degree of infiltration is superior to that usually observed in other C6 models.Modelos experimentais de tumores cerebrais malignos geralmente utilizam células imunologicamente compatíveis com o animal receptor. Neste estudo apresentamos um modelo experimental baseado na inoculação de células C6 em ratos atímicos Rowett, visando obter um tumor que se assemelhe mais àqueles observados nos seres humanos. Neste modelo observamos microinfiltração de ilhotas de células na periferia da massa tumoral principal e nas áreas mais distantes, células tumorais isoladas no tecido cerebral normal. Este grau de infiltração é superior àquele observado em outros modelos utilizando as células C6.

Rabbits have been used as an experimental model in many diseases and for the study of toxicology, pharmacology and surgery in many universities. However, some aspects of their macro anatomy need a more detailed description, especially the abdominal and pelvic arterial vascular system, which has a huge variability in distribution and trajectory. Thirty cadaveric adult New Zealand rabbits, 13 male and 17 female, with an average weight and rostrum-sacral length of 2.5 kg and 40cm, respectively, were used. The thoracic aorta was cannulated and the vascular system was filled with stained latex S-65. The celiac artery and its proximal branches were dissected and lengthened in order to evidence origin and proximal ramifications. The celiac artery emerged between the 12th and 13th thoracic vertebra in 11 (36.7%) rabbits; at the level of the 13th thoracic vertebra in 6 (20%) rabbits; between the 13th thoracic vertebra and the 1st lumbar vertebra in 12 (40%) rabbits; and at the level of the 1st lumbar vertebra in only one (3.3%) rabbit. The mean length of the celiac artery was 0.5cm. The celiac artery first branch was the lienal artery, the second branch was the left gastric artery and the hepatic artery arose from the left gastric artery in all the dissected rabbits. No relation was observed between the celiac artery length and the rostrum-sacral length in rabbits. The number of left gastric and lienal artery branches and the distribution of celiac artery origin are not gender dependent.Os coelhos têm sido usados como modelo experimental em diferentes patologias e para estudos de toxicologia, farmacologia e cirurgia em várias universidades. Entretanto apesar de sua grande utilização, muitos aspectos de sua macroanatomia, em especial os que se referem ao sistema vascular arterial que irrigam as viscerais abdomino-pélvicas ainda carecem de uma descrição mais detalhada, pois os vasos arteriais apresentam grande variabilidade na sua distribuição e trajeto. Foram utilizados 30 coelhos, 13 machos e 17 fêmeas, pesando em media 2,5 kg e apresentando comprimento rostro-sacral em torno de 40cm. A artéria aorta torácica foi canulada e através da mesma foi feita à fixação com solução de formaldeído a 10% e repleções vasculares com solução de Petrolátex S65 corado. A artéria celíaca e suas ramificações proximais foram dissecadas ao longo do seu percurso, registrando com auxílio de um paquímetro seu comprimento e sua esqueletopia. A artéria celíaca teve sua emergência de forma única diretamente da artéria aorta abdominal em todos os animais dissecados. Emitiu inicialmente a artéria lienal e a seguir a artéria gástrica esquerda que se continuou como hepática em todos os 30 animais. A artéria celíaca teve sua origem entre a 12ªe 13ª vértebra torácica em 11 animais (36,7 %), na 13ª vértebra torácica em 6 (20 %), entre a 13ª vértebra torácica e a 1ª vértebra lombar em 12 (40 %) e na 1ª vértebra lombar em apenas 1 animal (3,3%). O comprimento médio da artéria celíaca foi de 0,5cm. Não foi observada relação entre o comprimento da artéria celíaca e o comprimento rostro-sacral dos coelhos. O número de artérias gástricas esquerdas, ramificações principais da artéria lienal, bem como a origem da artéria celíaca independeram do sexo do animal.

Com o objetivo de avaliar as características de distribuição de agentes hiperbáricos no líquido cérebro-espinhal (LCE) eqüino, é apresentado um modelo in vitro do espaço subaracnóide confeccionado em policloreto de vinila (PVC) transparente, no qual se adaptou um cateter para a infusão contínua de glicose a 10%, morfina hiperbárica, buprenorfina hiperbárica e metadona hiperbárica marcadas com azul de metileno para permitir a visualização da distribuição destas substâncias no LCE. Avaliaram-se a distância alcançada pelos diferentes agentes minuto a minuto e a forma de distribuição dos mesmos. Todas as substâncias testadas mostraram comportamento de distribuição vertical a partir da extremidade do cateter, quando se deu o início da migração cranial no LCE, sendo que o avanço máximo registrado foi aos 15 minutos após o término da infusão, com migração de 15cm para glicose a 10%, 13cm para a morfina hiperbárica, 18cm para a buprenorfina hiperbárica e 17cm para a metadona hiperbárica. O modelo proposto mostrou-se eficiente para a avaliação do comportamento físico dos agentes hiperbáricos no interior do espaço subaracnóide, oferecendo a possibilidade de teste de substâncias a serem utilizadas pela via subaracnóide de cavalos.The aim of this study was to evaluate the distribution characteristics of hyperbaric solutions in equine cerebrospinal fluid (CSF), in an in vitro model of the subarachnoid space. The model was made with transparent PVC in which a catheter was adapted to infuse 10% glucose, 10% hyperbaric morphine, hyperbaric buprenorphine, and hyperbaric methadone, dyed with methilene blue in order to visualize the solutions distribution in the CSF. The distance in cm reached by the different agents every minute as well as the pattern of distribution were evaluated. All tested solutions distributed itself in a forward and vertically pattern from the tip of the catheter. Maximum distance reached after 15 minutes were 15cm for 10% glucose, 13cm for hyperbaric morphine, 18cm for hyperbaric buprenorphine, and 17cm for hyperbaric methadone. The proposed model was efficient to evaluate the distribution pattern of hyperbaric solutions in equine CSF, offering a new possibility to physically test the administration of solutions in the equine subarachnoid space.

O presente trabalho relata a avaliação de cobaias como modelo para testes de imunogenicidade de vacinas inativadas contra o herpesvírus bovino tipo 1 (BoHV-1) e o vírus da diarréia viral bovina (BVDV). Para isso, cobaias (n=60) e bovinos (n=10) foram imunizados duas vezes, com intervalo de 28 dias, com uma vacina experimental contendo antígenos dos dois vírus, e testados para anticorpos neutralizantes 28 dias após a segunda dose. Os bovinos foram vacinados com a dose recomendada para a espécie (5mL); as cobaias foram distribuídas em seis grupos e imunizadas com doses fracionadas (0,005mL a 1,6mL). Os grupos de cobaias imunizadas com doses equivalentes a 1/16 (0,320mL) e 1/8 (0,640mL) da dose bovina desenvolveram títulos médios geométricos (GMTs) de 6,46 e 7,56, respectivamente, estatisticamente semelhantes aos dos bovinos (GMT=8) (P>0,05). Uma alta correlação dose-resposta (R²=0,95) foi observada entre as doses vacinais e os títulos de anticorpos neutralizantes anti-BoHV-1 nos grupos de cobaias. Por outro lado, não foi possível o estabelecimento de uma dose vacinal que induzisse em cobaias uma resposta neutralizante anti-BVDV em níveis semelhantes à induzida em bovinos. Apenas as cobaias imunizadas com as doses maiores (0,640 e 1,6mL) desenvolveram títulos neutralizantes de magnitude moderada (GMTs de 8 e 9, respectivamente), porém estatisticamente inferiores ao GMT dos bovinos (GMT=34,9) (PThe present study reports the use of guinea pigs as a model to study the immunogenicity of bovine herpesvirus type 1 (BoHV-1) and bovine viral diarrhea virus (BVDV) inactivated vaccines. To this purpose, guinea pigs (60) and calves (10) were immunized twice with a 28 day interval with an experimental vaccine containing antigens of both viruses and tested for virus neutralizing (VN) antibodies 28 days after the second dose. Calves were immunized with the recommended dose (5mL), while the guinea pigs were distributed in six groups and immunized with fractionated doses (0.005 to 1.6mL). Guinea pigs immunized with 1/16 (0.320mL) and 1/8 (0.640mL) of the bovine dose developed VN titers (GMTs) of 6.46 and 7.56, respectively, which were equivalent to the titers developed by calves (GMT=8) (P>0.05). A high correlation (R²=0.95) was observed between the antigen dose and the VN titer developed by all guinea pig groups. PURPOSE: To elaborate an experimental model of pulmonary carcinogenesis in Wistar rats. METHODS: Male Rattus norvegicus albinus, Wistar lineage was carried through an intra-pulmonary instillation of the Benzo[a]pyrene (B[a]P) dilution in alcohol 70%, a polycyclic aromatic hydrocarbon widely known by its power of tumoral induction. Three experimental groups had been formed with 08 animals each: Control Group (Alcohol 70%); B[a]P Group 10 mg/kg; e B[a]P Group 20mg/kg, submitted to euthanasia 08, 10, 12 and 14 weeks after the experimental procedure. The pulmonary sections had been colored by hematoxilin-eosin (HE) and submitted to the morphometrical analysis to describe the tissue alterations. RESULTS: The presence of diffuse inflammatory alterations was observed in all groups, however, at the analysis of the pulmonary tissue of the experimental groups, it had been observed hyperplasic alterations (BALT hyperplasia), and in one of the animals of the experimental group 20mg/kg (12 weeks), it was noticed the presence of cellular epithelial tracheal pleomorphism, suggesting the adenocarcinoma formation in situ. CONCLUSION: The main secondary alterations to the intra-pulmonary instillation of B[a]P in Wistar rats were: cellular proliferation, inflammatory alterations of several degrees and nodular lymphoid hyperplasias. The association of an activator agent of the pulmonary metabolic reply is necessary to establish the ideal reply-dose to the development of the lung cancer.OBJETIVO: Elaborar um modelo experimental de carcinogênese pulmonar em ratos wistar. MÉTODOS: Rattus norvegicus albinus, linhagem Wistar foram submetidos a instilação intra-pulmonar da diluição em álcool 70% de Benzo[a]pireno (B[a]P), um hidrocarboneto aromático policíclico amplamente conhecido por seu poder de indução tumoral. Foram formados três grupos experimentais com 08 animais cada: Grupo Controle (álcool 70%); Grupo B[a]P 10 mg/kg; e Grupo B[a]P 20mg/kg, submetidos a eutanásia 08, 10, 12 e 14 semanas após o procedimento experimental. As secções pulmonares foram coradas por HE e submetidas a análise morfométrica para descrição das alterações teciduais. RESULTADOS: em todos os grupos observou-se a presença de alterações inflamatórias difusas, porém na análise do tecido pulmonar dos grupos experimentais, observou-se alterações hiperplásicas (hiperplasia de BALT), e em um dos animais do grupo experimental 20mg/kg (12 semanas) notou-se a presença de pleomorfismo celular epitelial traqueal, sugerindo a formação de adenocarcinoma in situ. CONCLUSÃO: as principais alterações secundárias à instilação intra-pulmonar de B[a]P em ratos Wistar foram: proliferação celular, alterações inflamatórias de diversos graus e hiperplasias nodulares linfóides. A associação de um agente ativador da resposta metabólica pulmonar pode ser necessária para estabelecimento da dose-resposta ideal ao desenvolvimento do câncer de pulmão.

PURPOSE: To test the possibility of obtaining a practical and stable model of hyperinsulinemia and hyperglycemia in hamsters, substituting the drinking water by 10% or 20% fructose solutions for a period of 2, 4, or 6 months. METHODS: Male hamsters were divided into 3 main groups, further divided in 3 subgroups: Two months: Group Ia control (n = 51) received filtered water, Group Ib (n = 49) received 10% fructose solution instead of water, Group Ic (n=8) received 20% fructose solution instead of water. Four months: Group IIa control (n=8), Group IIb 10% fructose (n = 7), Group IIc 20% fructose (FIIc, n = 7). Six months: Group IIIa control (n = 6), Group IIIb 10% Fructose (n = 6), Group IIIc 20% Fructose (n = 5). All groups were fed with the same laboratory diet. The animals were weighed every 2 weeks during the study period. On the final day of each experiment (61st, 121st, and 181st day after the beginning of the study, respectively), the animals were weighed and anesthetized for blood collection to determine plasma glucose and insulin after at least a 12-h fast. Ten animals of group Ia and 10 of group Ib were evaluated to determine changes in macromolecular permeability induced by ischemia/reperfusion as measured in the cheek pouch microcirculation. RESULTS: Compared to controls, the animals that drank the 10% or 20% fructose solution had significantly greater weight gain (P < .001), fasting plasma glucose (P < .001) Reperfusion, after 30 min ischemia, resulted in an immediate but reversible increase in postcapillary leakage (L) of 89.0 ± 2.0 L/cm² (group Ia - controls), and 116.5 ± 4.8 L/cm² (group Ib 10% fructose), P < .001.These results suggest that chronic administration of either 10% or 20% fructose solutions could be used to experimentally induce a stable hamster model of hyperinsulinemia and hyperglycemia. CONCLUSION: The model might facilitate the study of basic mechanisms of hyperglycemia and hyperinsulinemia affecting the microvasculature as demonstrated by the findings regarding ischemia/reperfusion after only 2 months of treatment.OBJETIVO: Testar a possibilidade de obtenção de um modelo prático e estável de hiperinsulinemia e hiperglicemia em hamsters substituindo a água de beber por soluções de frutose a 10% ou 20% por um período de dois, quatro ou seis meses. ABSTRACT: Neosporosis is a parasitic disease caused by the protozoan Neospora caninum which results in major economic losses for cattle breeding due to abortion and other reproductive disorders. Gerbils (Meriones unguiculatus) are commonly used as experimental models for neosporosis due to their high susceptibility to N. caninum infection, both by oocysts ingestion as by tachyzoites/bradyzoites parenteral inoculation. However, the risk of transmission by tachyzoites ingestion is not fully elucidated. In this study, infection of neonate gerbils by N. caninum (NC-1 strain) tachyzoites inoculated by the oral route and the parasite distribution in gerbils' tissues were evaluated by protozoan DNA detection. Seventeen neonate gerbils, aged 4-5 days, were inoculated with 4x105 tachyzoites by the oral route and one gerbil was kept as uninfected control. N. caninum DNA was detected in 100% of the inoculated gerbils, showing that the oral route is effective as a potential route of infection of neonates by N. caninum tachyzoites. N. caninum DNA was reported in all organs evaluated (heart, lungs, kidneys, liver, spleen and brain), with different frequencies. These results showed systemically distributed infection of neonate gerbils after oral inoculation of tachyzoites.

Pain is a complex phenomenon defined as an aversive sensory or emotional experience, where there is awareness of tissue damage or tissue integrity threat, with physiological and behavioral changes that aim to reduce or avoid occurrence of such damage. This study aimed to develop and evaluate an alternative low cost assessment method of the cutaneous nociceptive threshold in horses. The device used to assess the nociceptive threshold was constructed using an infrared 250W, 220V light bulb equipped with a mechanism to standardize the distance between the light bulb and the cutaneous surface of the animals. Six adult horses, males and females, were submitted to three evaluations. The results were submitted to analysis of variance (ANOVA) with Tukey's Multiple Comparison Test as post-test. No differences were observed among the evaluations (p>0,05), indicating that this method fulfills the aim to emit a standard nociceptive stimulus able to trigger a regular and qualitatively similar response between the tested subjects. We concluded that the studied equipment can be constructed at an accessible cost and it has potential to be applied in models of experimental pain, although some improvements are needed. Introduction: the use of plants as a therapeutic resource is a very ancient practice, and has since been the subject of studies. In this context there is jambolan (Syzygiumcumini) whichis a plant belonging to the family Mirtaceae. The leave shave substance swith anti diabetic action, acting hypoglycemic function, mimicking the actions of insulin, regulating blood glucose levels. Objective: the aim of this study was to evaluate the antioxidant activityof the leaves of Syzygium cumini and the effects of this infusion on a diet enriched experimental model with high glucose concentrations in Drosophila melanogaster. Methods: to verify jambolan the antioxidant used the DPPH (2,2-diphenyl-1picrilhidrazila) second Brand - Williams et al (1995).. The Drosophila were separated by sex, weighed and treated with infusion jambolan leaves except for the control, for three days and on the fourth day were sacrificed and weighed to biochemistry Glucose PAP Liquiform enzymatic triglycerides (Labtest). Results and discussion: regarding males, jambolan effect was highly significant as a protective factor in the diet with 20% sucroseandalso in the diet with 30% sucrose. Conclusion: this study demonstrated that exposure to a high-carbohydrate diet was harmful to D. melanogaster and the Syzygiumtea cumini (jambolan) had a positive effecton glucose and triglycerides parameters, validating this invertebrate model as a tool for the investigation of Diabetes mellitus. The aim of this research was to study the development of the lung in Galea spixii, by gross anatomy, histological analysis and immunohistochemical techniques. Totally, 8 fetuses were used and allocated into three groups of age: Group I (33-35 days), Group II (38-40 days), and Group III (43-45 days) with given crown-rump - CR lenghts. According to the gross morphology, there were no differences in relation to the lung morphology among groups. In relation to lung maturation, the Group I showed early formation of the bronchi and bronchioles, which were richly surrounded by mesenchyme and small blood vessels, typical features of the pseudoglandular stage. Individuals from Group II showed higher amounts of tubular formations in the lung parenchyma and reduced mesenchyme, reaching the canalicular stage. The lung from individuals of the Group III was completely formed, reaching the alveolar phase. In the immunohistochemical analysis, the lung of individuals from both Groups I and II were positive for Pcna, Oct-4, and VEGF. In contrast, there was not labbeling on samples from Group III. Thus, the G. spixii lung anatomy resembles the agouti (Dasyprocta sp.) lung, in relation to the number of lobes and fissures. In regard to the lung development, G. spixii showed more similarity with the human fetal lung, since both are born in the stage of the alveolar phase, different to other rodent species. - Abstract
Prosocial behavior refers to voluntary actions intended to help or benefit other individuals or group of individuals. In this article we revise experimental research on prosocial behavior in childhood. The first manifestations of prosocial behavior begin to appear after the first year of life, in simple acts such as helping an adult to reach a pencil that fell on the floor, or pointing to an object the adult is looking for. Later on they become more complex and diverse: children begin to share when another individual is at a resource disadvantage, or to cooperate with others to achieve a common goal. These experimental studies in early and middle childhood, allow to have an overview of prosocial behavior during this stage of life. The most difficult behaviors for children to achieve are those where they have to provide emotional support to someone who is sad or frustrated, or those altruistic acts that would involve a sacrifice for the child such as giving up an object of his own to improve the welfare of the other. Neosporosis is a parasitic disease caused by the protozoan Neospora caninum which results in major economic losses for cattle breeding due to abortion and other reproductive disorders. Gerbils (Meriones unguiculatus) are commonly used as experimental models for neosporosis due to their high susceptibility to N. caninum infection, both by oocysts ingestion as by tachyzoites/bradyzoites parenteral inoculation. However, the risk of transmission by tachyzoites ingestion is not fully elucidated. In this study, infection of neonate gerbils by N. caninum (NC-1 strain) tachyzoites inoculated by the oral route and the parasite distribution in gerbils' tissues were evaluated by protozoan DNA detection. Seventeen neonate gerbils, aged 4-5 days, were inoculated with 4x1 05 tachyzoites by the oral route and one gerbil was kept as uninfected control. N. caninum DNA was detected in 100% of the inoculated gerbils, showing that the oral route is effective as a potential route of infection of neonates by N. caninum tachyzoites. N. caninum DNA was reported in all organs evaluated (heart, lungs, kidneys, liver, spleen and brain), with different frequencies. - Abstract
Introduction and objectives: Animal models are a useful tool for the evaluation of disease mechanisms and also for technologies for diagnosis and treatment. In this study we performed a descriptive analysis of the functional and structural cardiac changes occurred as a result of acute coronary occlusion in pigs and its evolution during 5 weeks. Methods: 19-Large White pigs, weighing 20 kg, randomized into 3-experimental series were used. After sternotomy, anterior descending coronary artery was occluded. Duration of occlusion: Series 1 (n = 6) 60 min; series 2 (n = 8) 90 min; series 3 (n = 5) 60 min followed for 5 weeks. The following parameters where then analyzed: global cardiac function (ECG, left ventricular and atrium pressures, aortic flow and cardiac echocardiography), regional contractility, troponin T and CK-MB levels, macroscopic and histological analyzes. Results: Coronary occlusion transiently altered the global cardiac function and produced increased cell damage markers, impaired regional contractility and produced histological changes. The increment of ischemic time (60 vs. 90 min) increased infarct size (13.4 ± 5.4% vs. 22.9 ± 7.8 S1 S2%; P=.04). After 5 weeks, morphological remodelling changes were evident. In 79% of cases ischemia triggered ventricular fibrillation. Conclusion: The porcine open chest model of acute myocardial infarction and reperfusion is valid for studying the pathophysiology of coronary ischemia, allows direct analysis of regional myocardial function and is easily retrievable in the event of serious arrhythmias.
