An integrated computer software system for macromolecular crystallography (MX) data collection at the BL02U1 and BL10U2 beamlines of the Shanghai Synchrotron Radiation Facility is described. The system, Finback, implements a set of features designed for the automated MX beamlines, and is marked with a user-friendly web-based graphical user interface (GUI) for interactive data collection. The Finback client GUI can run on modern browsers and has been developed using several modern web technologies including WebSocket, WebGL, WebWorker and WebAssembly. Finback supports multiple concurrent sessions, so on-site and remote users can access the beamline simultaneously. Finback also cooperates with the deployed experimental data and information management system, the relevant experimental parameters and results are automatically deposited to a database.

Abstract The persistent pandemic of coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and its variants accentuates the great demand for developing effective therapeutic agents. Here, we report the development of an orally bioavailable SARS-CoV-2 3C-like protease (3CLpro) inhibitor, namely simnotrelvir, and its preclinical evaluation, which lay the foundation for clinical trials studies as well as the conditional approval of simnotrelvir in combination with ritonavir for the treatment of COVID-19. The structure-based optimization of boceprevir, an approved HCV protease inhibitor, leads to identification of simnotrelvir that covalently inhibits SARS-CoV-2 3CLpro with an enthalpy-driven thermodynamic binding signature. Multiple enzymatic assays reveal that simnotrelvir is a potent pan-CoV 3CLpro inhibitor but has high selectivity. It effectively blocks replications of SARS-CoV-2 variants in cell-based assays and exhibits good pharmacokinetic and safety profiles in male and female rats and monkeys, leading to robust oral efficacy in a male mouse model of SARS-CoV-2 Delta infection in which it not only significantly reduces lung viral loads but also eliminates the virus from brains. The discovery of simnotrelvir thereby highlights the utility of structure-based development of marked protease inhibitors for providing a small molecule therapeutic effectively combatting human coronaviruses.

To resolve photons hungry for weak diffraction samples by the crystallographic method, a double-multilayer monochromator (DMM) was employed on an undulator beamline (BL17UM) at the Shanghai Synchrotron Radiation Facility (SSRF) to provide a focused sub-micron beam with high brightness for macromolecular crystallography experiments. High-quality crystallographic datasets from model protein crystal samples were collected and processed by an existing crystallographic program for structure solution and refinement. The data quality was compared with datasets from a normal silicon crystal monochromator to evaluate the bandwidth of the DMM effect on these crystallographic data. This experiment demonstrates that multilayer optics on an undulator beamline may play a valuable role in satisfying the demands of structure-related research, which requires novel methods.

Eleutherococcus nodiflorus (Dunn) S. Y. Hu is a momentous medicinal plant belonging to the Araliaceae family. In the current investigation, we determined the complete chloroplast genome of E. nodiflorus and analyzed the phylogenetic relationship among Eleutherococcus plants. The chloroplast genome of E. nodiflorus exhibited a typical quadripartite structure with a full length of 156,770 bp, including 133 genes, containing 88 protein-coding genes, 8 rRNA genes, 37 tRNA genes, and 1 presumed pseudogene (ycf1). The overall GC content observed was 37.95%, with the highest GC content of 43.02% found in the IR region. Comparative genome analysis revealed five highly variable regions among Eleutherococcus species, providing potential markers for further investigations on species identification and population genetics. A total of 44 small simple repeats were identified throughout the chloroplast genome of E. nodiflorus. The phylogenetic analysis indicated a sister relationship between E. nodiflorus and E. eleutheristylus, suggesting a close genetic relationship between the two Eleutherococcus plants. These results enhance the understanding of the plant evolution within Eleutherococcus plants and provide basic genetic resources for the development of species identification and investigation of population genetic diversity of the Eleutherococcus genus and Araliaceae.

SARS-CoV-2 3CL protease (3CLpro) is essential for coronavirus replication and of great interest as an antiviral drug target. Here, the authors show that the naturally occurring flavonoid myricetin is a non-peptidomimetic and covalent inhibitor of 3CLpro, and they solve crystal structures of 3CLpro with myricetin and derivatives, which reveal that the pyrogallol group covalently modifies the catalytic cysteine.

Selectively inhibiting N-Methyl-D-aspartate receptors (NMDARs) containing the GluN2C/2D subunits has been challenging. Here, using electrophysiology and X-ray crystallography, authors show that compounds UBP791 and UBP1700 show over 40- and 50-fold selectivity for GluN2C/2D compared to GluN2A.

MMACHC is an essential protein for the body to metabolise vitamin B12, and its deficiency will cause cblC-type methylmalonic aciduria and homocystinuria. MMACHC can interact with cyanocobalamin (a type of vitamin B12) cofactor and plays an important role in targeting cyanocobalamin to the enzyme of interest. In this paper, the GST-tag fusion-tagged MMACHC protein was successfully expressed by Escherichia coli (E. coli) low-temperature induction, and the high-purity MMACHC protein was successfully purified by affinity chromatography and gel filtration. Further, the crystal structure of MMACHC and cyanocobalamin complex was obtained with a resolution of 1.93 Å using X-ray diffraction. By analysing the complex structure of MMACHC and cyanocobalamin, we revealed the reasons for the diversity of MMACHC substrates and explained the reasons for the differences in disease conditions caused by different MMACHC site mutations. The acquisition of the complex structure of MMACHC and cyanocobalamin will play a significant role in promoting research on the metabolic pathway of vitamin B12.

Thaxtomins, a family of phytotoxins produced by Streptomyces spp., can cause dramatic plant cell hypertrophy and seedling stunting. Thaxtomin A is the dominant form from Streptomyces scabies and has demonstrated herbicidal action. TxtE, a cytochrome P450 enzyme from Streptomyces scabies 87.22, catalyzes direct nitration of the indolyl moiety of L-tryptophan to L-4-nitrotryptophan using nitric oxide, dioxygen and NADPH. The crystal structure of TxtE was determined at 2.1 Å resolution and described in this work. A clearly defined substrate access channel is observed and can be classified as channel 2a, which is common in bacteria cytochrome P450 enzymes. A continuous hydrogen bond chain from the active site to the external solvent is observed. Compared with other cytochrome P450 enzymes, TxtE shows a unique proton transfer pathway which crosses the helix I distortion. Polar contacts of Arg59, Tyr89, Asn293, Thr296, and Glu394 with L-tryptophan are seen using molecular docking analysis, which are potentially important for substrate recognition and binding. After mutating Arg59, Asn293, Thr296 or Glu394 to leucine, the substrate binding ability of TxtE was lost or decreased significantly. Based on the docking and mutation results, a possible mechanism for substrate recognition and binding is proposed.

Abstract The obese heart undergoes metabolic remodeling and exhibits impaired calcium (Ca2+) homeostasis, which are two critical assaults leading to cardiac dysfunction. The molecular mechanisms underlying these alterations in obese heart are not well understood. Here, we show that the Rab-GTPase activating protein AS160 is a lipid-responsive regulator of Ca2+ homeostasis through governing lysophosphatidylinositol metabolism and signaling. Palmitic acid/high fat diet inhibits AS160 activity through phosphorylation by NEK6, which consequently activates its downstream target Rab8a. Inactivation of AS160 in cardiomyocytes elevates cytosolic Ca2+ that subsequently impairs cardiac contractility. Mechanistically, Rab8a downstream of AS160 interacts with DDHD1 to increase lysophosphatidylinositol metabolism and signaling that leads to Ca2+ release from sarcoplasmic reticulum. Inactivation of NEK6 prevents inhibition of AS160 by palmitic acid/high fat diet, and alleviates cardiac dysfunction in high fat diet-fed mice. Together, our findings reveal a regulatory mechanism governing metabolic remodeling and Ca2+ homeostasis in obese heart, and have therapeutic implications to combat obesity cardiomyopathy.

Background: Myelodysplastic syndromes (MDS) are a class of myeloid neoplasms featuring inefficient maturation and differentiation of hematopoietic cells, blood cytopenia, and a high risk of leukemia onset. The diagnosis of MDS remains a challenging task owing to its complexity, heterogeneity, and the lack of specific characteristics. Methods: To look for an easy and inexpensive diagnostic method for MDS, we tried to establish an FCM scoring systems (FCSS) with a combination of antibodies for diagnosis and prognostic stratification of MDS. This FCSS adopted four parameters; i.e., the frequency of myeloblasts in nucleated cells, the ratio between pro-B cells and CD117+ cells, the ratio of CD45 mean fluorescence intensity between lymphocytes and myeloblasts, and the ratio of SSC peak values between mature granulocytes and lymphocytes. Results: We tested the correlation between the total FCSS score with conventional IPSS-R. Additionally, the correlation between the score of each FCSS parameter and IPSS-R was also evaluated. We found that total FCSS score had a positive correlation with IPSS-R, while FCSS parameter 1 and 4 were also correlated with IPSS-R. Furthermore, this FCSS had a sound sensitivity and specificity in the diagnosis of MDS. Conclusions: The FCSS represents a convenient and affordable approach for the diagnosis and prognostic stratification of MDS. [ABSTRACT FROM AUTHOR]

Background: Dancers represent the primary demographic affected by ankle joint injuries. In certain movements, some Latin dancers prefer landing on the Forefoot (FT), while others prefer landing on the Entire foot (ET). Different stance patterns can have varying impacts on dancers’ risk of ankle joint injuries. The purpose of this study is to investigate the differences in lower limb biomechanics between Forefoot (FT) dancers and Entire foot (ET) dancers.Method: A group of 21 FT dancers (mean age 23.50 (S.D. 1.12) years) was compared to a group of 21 ET dancers (mean age 23.33 (S.D. 0.94) years), performing the kicking movements of the Jive in response to the corresponding music. We import data collected from Vicon and force plates into OpenSim to establish musculoskeletal models for computing kinematics, dynamics, muscle forces, and muscle co-activation.Result: In the sagittal plane: ankle angle (0%–100%, p < 0.001), In the coronal plane: ankle angle (0%–9.83%, p = 0.001) (44.34%–79.52%, p = 0.003), (88.56%–100%, p = 0.037), ankle velocity (3.73%–11.65%, p = 0.017) (94.72–100%, p = 0.031); SPM analysis revealed that FT dancers exhibited significantly smaller muscle force than ET dancers around the ankle joint during the stance phase. Furthermore, FT dancers displayed reduced co-activation compared to ET dancers around the ankle joint during the descending phase, while demonstrating higher co-activation around the knee joint than ET dancers.Conclusion: This study biomechanically demonstrates that in various stance patterns within Latin dance, a reduction in lower limb stance area leads to weakened muscle strength and reduced co-activation around the ankle joint, and results in increased ankle inversion angles and velocities, thereby heightening the risk of ankle sprains. Nevertheless, the increased co-activation around the knee joint in FT dancers may be a compensatory response for reducing the lower limb stance area in order to maintain stability.

Previous research has primarily focused on evaluating the activity of individual muscles in dancers, often neglecting their synergistic interactions. Investigating the differences in lower limb muscle synergy during landing between dancers and healthy controls will contribute to a comprehensive understanding of their neuromuscular control patterns. This study enrolled 22 Latin dancers and 22 healthy participants, who performed a task involving landing from a 30 cm high platform. The data were collected using Vicon systems, force plates, and electromyography (EMG). The processed EMG data were subjected to non-negative matrix factorization (NNMF) for decomposition, followed by classification using K-means clustering algorithm and Pearson correlation coefficients. Three synergies were extracted for both Latin dancers and healthy participants. Synergy 1 showed increased contributions from the tibialis anterior (p < 0.001) and medial gastrocnemius (p = 0.024) in Latin dancers compared to healthy participants. Synergy 3 highlighted significantly greater contributions from the vastus lateralis in healthy participants compared to Latin dancers (p = 0.039). This study demonstrates that Latin dancers exhibit muscle synergies similar to those observed in healthy controls, revealing specific adjustments in the tibialis anterior and medial gastrocnemius muscles among dancers. This research illustrates how dancers optimize control strategies during landing tasks, offering a novel perspective for comprehensively understanding dancers’ neuromuscular control patterns.

Purpose. The functional status of the ankle joint is critical during dynamic movements in high-intensity sports like basketball and volleyball, particularly when performing actions such as stopping jumps. Limited ankle dorsiflexion is associated with increased injury risk and biomechanical changes during stop-jump tasks. Therefore, this study aims to investigate how restricting ankle dorsiflexion affects lower extremity biomechanics during the stop-jump phase, with a focus on the adaptive changes that occur in response to this restriction. Initially, 18 participants during stop-jumping with no wedge plate (NW), 10° wedge plate (10 W), and 20° wedge plate (20 W) using dominant leg data were collected to explore the relationship between limiting ankle mobility and lower extremity biomechanics. Following this, a musculoskeletal model was developed to simulate and calculate biomechanical data. Finally, one-dimensional parametric statistical mapping (SPM1D) was utilized to evaluate between-group variation in outcome variables using a one-way repeated measures analysis of variance (ANOVA). ObjectiveThis study aims to establish hypertension risk nomograms for Chinese male and female adults, respectively.MethodA series of questionnaire surveys, physical assessments, and biochemical indicator tests were performed on 18,367 adult participants in China. The optimization of variable selection was conducted by running cyclic coordinate descent with 10-fold cross-validation through the least absolute shrinkage and selection operator (LASSO) regression. The nomograms were built by including the predictors selected through multivariable logistic regression. Calibration plots, receiver operating characteristic curves (ROC), decision curve analysis (DCA), clinical impact curves (CIC), and net reduction curve plots (NRC) were used to validate the models.ResultsOut of a total of 18 variables, 5 predictors—namely age, body mass index, waistline, hipline, and resting heart rate—were identified for the hypertension risk predictive model for men with an area under the ROC of 0.693 in the training set and 0.707 in the validation set. Seven predictors—namely age, body mass index, body weight, cardiovascular disease history, waistline, resting heart rate, and daily activity level—were identified for the hypertension risk predictive model for women with an area under the ROC of 0.720 in the training set and 0.748 in the validation set. The nomograms for both men and women were externally well-validated.ConclusionGender differences may induce heterogeneity in hypertension risk prediction between men and women. Besides basic demographic and anthropometric parameters, information related to the functional status of the cardiovascular system and physical activity appears to be necessary.

BackgroundNeutrophil extracellular traps (NETs) have been shown to play a pivotal role in promoting metastasis and immune escape in hepatocellular carcinoma (HCC). Therefore, noninvasive tests to detect the formation of NETs in tumors can have significant implications for the treatment and prognoses of patients. Here, we sought to develop and validate a computed tomography (CT)-based radiomics model to predict the gene expression profiles that regulate the formation of NETs in HCC.MethodsThis study included 1133 HCC patients from five retrospective cohorts. Based on the mRNA expression levels of 69 biomarkers correlated with NET formation, a 6-gene score (NETs score, NETS) was constructed in cohort 1 from TCIA database (n=52) and validated in cohort 2 (n=232) from ICGC database and cohort 3 (n=365) from TCGA database. And then based on the radiomics features of CT images, a radiomics signature (RNETS) was developed in cohort 1 to predict NETS status (high- or low-NETS). We further employed two cohorts from Nanfang Hospital (Guangzhou, China) to evaluate the predictive power of RNETS in predicting prognosis in cohort 4 (n=347) and the responses to PD-1 inhibitor of HCC patients in cohort 5 (n=137).ResultsFor NETS, in cohort 1, the area under the curve (AUC) values predicting 1, 2, and 3-year overall survival (OS) were 0.836, 0.879, and 0.902, respectively. The low-NETS was associated with better survival and higher levels of immune cell infiltration. The RNETS yielded an AUC value of 0.853 in distinguishing between high-NETS or low-NETS and patients with low-RNETS were associated with significantly longer survival time in cohort 1 (P

Abstract The purpose of this study was to compare swing time and golf club angle parameters during golf swings using three, two dimensional (2D) low cost, Augmented-Video-based-Portable-Systems (AVPS) (Kinovea, SiliconCoach Pro, SiliconCoach Live). Twelve right-handed golfers performed three golf swings whilst being recorded by a high-speed 2D video camera. Footage was then analysed using AVPS-software and the results compared using both descriptive and inferential statistics. There were no significant differences for swing time and the golf phase measurements between the 2D and 3D software comparisons. In general, the results showed a high Intra class Correlation Coefficient (ICC > 0.929) and Cronbach’s Coefficient Alpha (CCA > 0.924) reliability for both the kinematic and temporal parameters. The inter-rater reliability test for the swing time and kinematic golf phase measurements on average were strong. Irrespective of the AVPS software investigated, the cost effective AVPS can produce reliable output measures that benefit golf analyses.

Insulin promotes hepatic lipogenesis, though underlying regulation remains unclear. Here the authors show that insulin translocates TRIM24 from the nucleus into cytosolic P-bodies to stabilise hepatic Pparγ mRNA, and that inactivation of TRIM24 promotes Pparγ degradation and alleviates hepatosteatosis.

Abstract Insulin signals through its receptor to recruit insulin receptor substrates (IRS) and phosphatidylinositol 3-kinase (PI3K) to the plasma membrane for production of phosphatidylinositol-3,4,5-trisphosphate (PIP3) from phosphatidylinositol-4,5-bisphosphate [PI(4,5)P2], which consequently activates protein kinase B (PKB). How insulin signals transduce from the plasma membrane into the cytoplasm is not clearly understood. Here we show that liquid–liquid phase separation (LLPS) plays a critical role in spatiotemporal control of insulin signaling through regulating multiple components including IRS1. Both protein concentration and insulin stimulation can drive the formation of intracellular IRS1 condensates through LLPS. Components including PI(4,5)P2, p85-PI3K and PDK1 are constitutively present in IRS1 condensates whereas production of PIP3 and recruitment of PKB in them are induced by insulin. Thus, IRS1 condensates function as intracellular signal hubs to mediate insulin signaling, whose formation is impaired in insulin resistant cells. Collectively, these data reveal an important function of LLPS in spatiotemporal control of insulin signaling.

Introduction: Cross-court and the long-line topspin forehand is the common and basic stroke skill in table tennis. The purpose of this study was to investigate the differences in lumbar and pelvis movements between cross-court and long-line topspin forehand strokes in table tennis based on musculoskeletal demands using OpenSim.Materials and Methods: The eight-camera Vicon system and Kistler force platform were used to measure kinematics and kinetics in the lumbar and pelvis movement of sixteen participants (Weight: 69.89 ± 1.58 kg; Height: 1.73 ± 0.03 m; Age: 22.89 ± 2.03 years; BMI: 23.45 ± 0.69 kg/m2; Experience: 8.33 ± 0.71 years) during cross-court and long-line topspin forehand play. The data was imputed into OpenSim providing the establishment of the Giat2392 musculoskeletal model for simulation. One-dimensional statistical parametric mapping and independent samples t-test was performed in MATLAB and SPSS to analyze the kinematics and kinetics.Results: The results show that the range of motion, peak moment, and maximum angle of the lumbar and pelvis movement in cross-court play were significantly higher than in the long-line stroke play. The moment of long-line in the sagittal and frontal plane was significantly higher than cross-court play in the early stroke phase.Conclusion: The lumbar and pelvis embody greater weight transfer and greater energy production mechanisms when players performed cross-court compared to long-line topspin forehand. Beginners could enhance their motor control strategies in forehand topspin skills and master this skill more easily based on the results of this study.

VqmA is a highly conserved transcriptional regulator of the quorum-sensing system of Vibrio cholerae, a major human pathogen that continues to imperil human health. VqmA represses biofilm formation and plays an important role in V. cholerae pathogenicity in the human host. Although VqmA's biological function is well understood, the molecular mechanisms by which its specific ligand (and effector), 3,5-dimethylpyrazine- 2-ol (DPO), controls transcription of the target gene, vqmR, remain obscure. To elucidate the molecular mechanism of DPO binding, we used structural analyses and biochemical assays to study the V. cholerae VqmA--DPO--DNA complex. These analyses revealed that VqmA contains an N-terminal homodimer domain (PAS) and a C-terminal DNA-binding domain (DBD).Weobserved thatVqmAdirectly binds to aDPO molecule via a compact hydrophobic pocket, consisting of a six-stranded antiparallel β-sheet and several α-helices. We also found that the VqmA dimer interacts with the quasi-palindromic sequence of the vqmR promoter through its DBD. The results of the biochemical studies indicated that a water atom and VqmA residues Phe-67 and Lys-101 play a key role in effector recognition, which is also assisted by Tyr-36 and Phe-99. This is the first molecular level view of the VqmA dimer bound to DPO and DNA. The structure--function analyses presented here improve our understanding of the complex mechanisms in the transcriptional regulation of VqmA in Vibrio spp. and may inform the design of drugs to manage V. cholerae infections. The running biomechanics of running shoes have been extensively investigated. However, there is limited knowledge about the use of bionic shoes compared to neutral shoes, along with the velocities involved in their use. The aim of this study was to examine the biomechanical alterations associated with various running velocities of bionic shoes. By removing different thicknesses of the forefoot section, bionic shoes created a more natural shape—close to that of a human foot. The study included 16 heel strike runners running at 10 km/h, 12 km/h and 14 km/h in bionic shoes and neutral shoes, respectively. A two-way ANOVA and SPM1d were employed for examining kinematic and kinetic differences. Regarding the results for the shoes, increased ROM was observed for the bionic shoes for the hip (p < 0.001) and ankle joints (p < 0.001). Ankle positive work (p < 0.001) and negative work (p = 0.042) also showed significant differences. Regarding the velocity results, hip ROM (p < 0.001) increased and peak knee angular velocity (p = 0.018) increased, while knee ROM (p = 0.023) decreased. The interaction effects only existed in hip (p = 0.031) and ankle (p = 0.008) ROM. The results of this study suggested that the impact of running propulsion in the bionic shoes was minimal. However, with increased velocities, the bionic shoes demonstrated the ability to absorb more force, created a more stable training environment, and contributed to injury prevention for the hip and ankle joints.

Introduction: The purpose of this study was to evaluate the effect of running-induced fatigue on the characteristic asymmetry of running gait and to identify non-linear differences in bilateral lower limbs and fatigued gait by building a machine learning model.Methods: Data on bilateral lower limb three-dimensional ground reaction forces were collected from 14 male amateur runners before and after a running-induced fatigue experiment. The symmetry function (SF) was used to assess the degree of symmetry of running gait. Statistical parameter mapping (Paired sample T-test) algorithm was used to examine bilateral lower limb differences and asymmetry changes pre- and post-fatigue of time series data. The support vector ma-chine (SVM) algorithm was used to recognize the gait characteristics of both lower limbs before and after fatigue and to build the optimal algorithm model by setting different kernel functions.Results: The results showed that the ground reaction forces were asymmetrical (SF > 0.5) both pre-and post-fatigue and mainly concentrated in the medial-lateral direction. The asymmetry of the medial-lateral direction increased significantly after fatigue (p < 0.05). In addition, we concluded that the polynomial kernel function could make the SVM model the most accurate in classifying left and right gait features (accuracy of 85.3%, 82.4%, and 82.4% in medial-lateral, anterior-posterior and vertical directions, respectively). Gaussian radial basis kernel function was the optimal kernel function of the SVM algorithm model for fatigue gait recognition in the medial-lateral and vertical directions (accuracy of 54.2% and 62.5%, respectively). Moreover, polynomial was the optimal kernel function of the anterior-posterior di-rection (accuracy = 54.2%).Discussion: We proved in this study that the SVM algorithm model depicted good performance in identifying asymmetric and fatigue gaits. These findings can provide implications for running injury prevention, movement monitoring, and gait assessment.

BackgroundThe most commonly acknowledged non-scarring alopecia are androgenetic alopecia (AGA) and alopecia areata (AA). Previous studies have revealed various risk factors associated with alopecia. However, the relationship between leukocyte telomere length (LTL) and non-scarring alopecia remains unclear.MethodsA two-sample Mendelian randomization (MR) analysis was performed to evaluate the causality between genetically predicted LTL and the risk of non-scarring alopecia. MR analyses were performed using the inverse variance-weighted (IVW) method and complemented with other MR methods.ResultsThe summary statistics of the genome-wide association studies (GWAS) for AGA and AA were obtained from the FinnGen biobank, which included 119,185 and 211,428 individuals, respectively. A total of 126 single nucleotide polymorphisms (SNPs) with genome-wide significance were selected as the instrumental variables for LTL. The MR analyses suggested a causal relationship between LTL and AGA, and the risk of AGA increased by 3.19 times as the genetically predicted LTL was shortened by one standard deviation in log transformed form under the IVW method (OR = 4.19, 95% CI = 1.20–14.61, p = 0.024). The other MR methods also demonstrated a similar trend of the effect of LTL on AGA. There was no causal relationship between LTL and AA (p > 0.05). Sensitivity analyses further demonstrated that the current results were less likely to be affected by confounders and bias.ConclusionOur results suggested a potential causal relationship between LTL and AGA, and shortened LTL was associated with an increased risk of AGA.

Abstract Background Pain control after hepatectomy is usually achieved by opioids. There are significant individual differences in the amount of opioids used after hepatectomy, and the metabolism of opioids is liver-dependent. The purpose of our study was to explore the possible risk factors for opioid consumption during the first 48 h after surgery. Methods In a retrospective study design involving 562 patients undergoing open or laparoscopic hepatectomy, all patients were treated with intravenous patient-controlled analgesia (IV-PCA) along with continuous and bolus doses of sufentanil for a duration of 48 h after surgery during the time period of August 2015 and February 2019. The primary endpoint was high sufentanil consumption 48 h after hepatectomy, and patients were divided into two groups: those with or without a high PCA sufentanil dosage depending on the third quartile (Q3). The secondary endpoint was the effect of a high PCA sufentanil dosage on various possible clinical risk factors. The relevant parameters were collected, and correlation and multivariate regression analyses were performed. Results The median operation time was 185 min (range, 115–250 min), and the median consumption of sufentanil 48 h after the operation was 91 μg (IQR, 64.00, 133.00). Factors related to the consumption of sufentanil at 48 h after hepatectomy included age, operation time, blood loss, intraoperative infusion (red blood cells and fresh-frozen plasma), pain during movement after surgery (day 1 and day 2), preoperative albumin, and postoperative blood urea nitrogen. Age (≤ 60 and > 60 years), extent of resection (minor hepatic resection and major hepatic resection), surgical approach (laparoscope and open) and operation time (min) were independent risk factors for sufentanil consumption at 48 h postoperatively. Conclusion Age younger than 60 years, major hepatic resection, an open approach and a longer operation are factors more likely to cause patients to require higher doses of sufentanil after hepatectomy, and the early identification of such patients can increase the efficacy of perioperative pain management.

Features extracted by deep networks have been popular in many visual search tasks. This article studies deep network structures and training schemes for mobile visual search. The goal is to learn an effective yet portable feature representation that is suitable for bridging the domain gap between mobile user photos and (mostly) professionally taken product images while keeping the computational cost acceptable for mobile based applications. The technical contributions are twofold. First,we propose an alternative of the contrastive loss popularly used for training deep Siamese networks, namely robust contrastive loss, where we relax the penalty on some positive and negative pairs to alleviate over fitting. Second, a simple multitask fine-tuning scheme is leveraged to train the network, which not only utilizes knowledge from the provided training photo pairs but also harnesses additional information from the large Image Net data set to regularize the fine tuning process. Extensive experiments on challenging real-world data sets demonstrate that both the robust contrastive loss and the multitask fine-tuning scheme are effective, leading to very promising results with a time cost suitable for mobile product search scenarios. Patellofemoral disorders are more common in female runners compared to their male counterparts. Differences in biomechanical characteristics between groups of runners could provide insight into the causes of higher rates of injury in female versus male runners, which would be useful to physical therapists and athletic trainers in development of individualized injury prevention programs. This review compares the differences in biomechanical characteristics between female and male runners. Electronic databases, including PubMed, Scopus, Web of Science, and Embase were searched in December 2021 for studies evaluating sex-specific differences in lower limb mechanics of healthy participants during running. Two independent reviewers determined the inclusion and quality of each research paper. Meta-analyses were used where possible. A total of 13 studies were selected. Means and standard deviations of reported data were retrieved from each selected paper for comparison of results. Three biomechanical variables, including dynamics, muscle activation, and kinematics, were compared between female and male runners. However, no differences were found in kinetic variables or muscle activation between groups due to insufficient data available from the selected studies. Meta-analyses of kinematic variables revealed that female runners exhibited significantly greater hip flexion angle, hip adduction angle, and hip internal rotation angle, but smaller knee flexion angle compared to male runners during running. We found significant differences in kinematic variables between female and male runners, which could influence the training advice of physical therapists and athletic trainers who work with runners, and inform the development of injury prevention programs.

The objective of this study was to examine the effects of running fatigue on the symmetry of lower limb biomechanical parameters in eighteen male amateur runners. The marker trajectories and ground reaction forces were collected before and after the running-induced fatigue protocol. Symmetry angles (SA) were used to quantify the symmetry of each parameter. Normality tests and Paired sample T-tests were carried out to detect bilateral lower limb differences and SA of parameters between pre- and post-fatigue. One-dimensional statistical parameter mapping (SPM_1d) was used to compare parameters with the characteristic of one-dimensional time series data of lower limbs. After fatigue, the SA of knee extension angles, knee abduction moment, and hip joint flexion moment increased by 17%, 10%, and 11% respectively. In comparison, the flexion angle of the knee joint decreased by 5%. The symmetry of internal rotation of ankle, knee and hip joints increased after fatigued, while the SA of external rotation of the three joints decreased significantly. These findings provide preliminary evidence that fatigue changes lower limb symmetry in running gait and may have implications for understanding running-related injuries and performance.

Background: Despite the growing interest in golf, the impact of motion control strategy on golf performance may be uncertain. A network meta-analysis was conducted to investigate the relationships between ground reaction force (GRF), foot positions, and club types in golf and determine whether different clubs have a different impact when swinging. Methods: Three databases were searched from the time of inception to March 2023. Eligibility criteria included studies that provided data using a driver to swing and compared outcomes to control data. Results: Searches yielded 6527 studies. Seven studies met the selection criteria (n = 422 participants). The results showed that an iron 6 is the best when considering the trail foot vertical ground reaction force (TVGRF), trail foot anteroposterior ground reaction force (TAGRF), and lead foot vertical ground reaction force (LVGRF). The pitching wedge was the best in the lead foot mediolateral ground reaction force (LMGRF) and lead foot anteroposterior ground reaction force (LAGRF). Iron 7 was the best in the trail foot mediolateral ground reaction force (TMGRF), and the lead foot was larger than the trail foot to the vertical GRF. Discussion: The study found that clubs may influence a player’s posture and swing power because golf clubs are available in various lengths and shapes. The lead foot generates a larger GRF than the trail foot; three-dimensional GRFs differ among golf clubs. When a golfer aims to maximize the distance of their drives, they must generate relatively more resultant horizontal reaction force (RFH). Golfers often use different clubs to achieve optimal performance on the course by controlling their motion. However, there needs to be a focus on the quality of the included studies because the sample size was too small, increasing the risk of bias associated with the results.

Carbon dioxide embolism is a rare but severe complication of laparoscopic hepatectomy. We reported a case of massive carbon dioxide embolism that developed into a rare paradoxical gas embolism during laparoscopic hepatectomy and resulted in reduced muscular power of the left upper extremity, severe pleural effusion and hypoalbuminemia after surgery. Fortunately, the patient fully recovered with positive prevention and postoperative treatment. This case report highlights that the surgeon and anaesthesiologist must be aware of the risks and the importance for prompt treatment when massive carbon dioxide embolism occurs.

BackgroundThe dynamic balance between oxidation and anti-oxidation in the body’s internal environment has a significant meaning for human health. Physical exercise and antioxidative supplementation could affect the balance of oxidation and anti-oxidation systems. The evidence on the effects of physical exercise and antioxidative supplementation is mixed.AimsTo identify the effects of physical exercise, antioxidative supplementation, and their combination on the dynamic balance between oxidation and anti-oxidation in different subgroups of healthy adults.MethodsAll studies which reported randomized controlled trials with healthy participants were screened and included from the databases of PubMed, Medline, Embase, and Ovid. All participants were reclassified according to their different daily life activities. All physical exercise interventions were reclassified according to the intensity. The effect size would be calculated in percent or factor units from the mean level change with its associated random-effect variance.ResultThere were 27 studies included in this review. The agreement between authors by using The Cochrane Collaboration Risk of Bias Assessment Tool reached a kappa-value of 0.72. Maintaining a regular physical exercise routine in an appropriate intensity would be beneficial to the body’s anti-oxidative potential. Anti-oxidative supplementation could have some positive but limited effects on the body’s anti-oxidative status and complex interaction with physical exercise.ConclusionKeeping a regular physical exercise routine and gradually increasing its intensity according to the individual’s daily life activity might be a better choice to maintain and enhancing the body’s antioxidation potential, only using anti-oxidative supplementation is not recommended. Objective. The purpose of this research was to use a new method the human and bar combination barycenter to exposit the differences between successful and failed characteristics of snatch attempts in competition. Try to establish an effective biomechanical method that can uncover the main factors for the failed snatch. The obtained results will provide valuable information for weightlifters to improve the success rate in snatch by altering their technical issues accordingly. Methods. A 3-D video analysis method was used to compare the characteristics of the heaviest successful and failed attempts of ten elite weightlifters in the men’s 73 kg category. The video was captured under competitive conditions at the 2019 World Weightlifting Championships, the 2019 Asian Weightlifting Championships, and the 2020 China Olympic Trial. The video data were digitized using the SIMI°Motion7.50 3-D system (Germany). Results. Significant difference (P>0.05) was not found between the successful and failed attempts in the parameters, such as the maximal vertical rising velocity, the maximal vertical height, and the vertical displacement of the barbell. The maximal descending acceleration of the human body, the time duration, the angles of the hip, and knee joints were no significant difference. However, significant differences were found in the variation of the human and bar combination barycenter on the X-axis in the inertial ascent stage and the squat support stage (t=2.862,P

TleD is a SAM (S-adenosyl-L-methionine)-dependent methyltransferase and acts as one of the key enzymes in the teleocidin B biosynthesis pathway. Besides methyl transferring, TleD also rearranges the geranyl and indole moieties of the precursor to form a six-membered ring. Moreover, it does not show homologies with any known terpenoid cyclases. In order to elucidate how such a remarkable reaction could be achieved, we determined the complex crystal structures of TleD and the cofactor analogue S-adenosyl-L-homocysteine with or without the substrate teleocidin A1. A domain-swapped pattern via an additional N-terminal α-helix is observed in TleD hexamers. Structural comparison and alignment shows that this additional N-terminal α-helix is the common feature of SAM methyltransferase-like cyclases TleD and SpnF. The residue Tyr21 anchors the additional N-terminal α-helix to a 'core SAM-MT fold' and is a key residue for catalytic activity. Molecular dynamics simulation results suggest that the dihedral angle C23-C24-C25-C26 of teleocidin A1 is preferred to 60-90± in the TleD and substrate complex structure, which tend to adopt a Re-face stereocenter at C25 position after reaction and is according to in vitro enzyme reaction experiments. Our results also demonstrate that methyl transfer can be a new chemical strategy for carbocation formation in the terpene cyclization, which is the key initial step. [ABSTRACT FROM AUTHOR]

BackgroundHepatocellular carcinoma (HCC) is one of the leading malignant tumors worldwide. Prognosis and long-term survival of HCC remain unsatisfactory, even after radical resection, and many non-invasive predictors have been explored for post-operative patients. Most prognostic prediction models were based on preoperative clinical characteristics and pathological findings. This study aimed to investigate the prognostic value of a newly constructed nomogram, which incorporated post-operative aspartate aminotransferase to lymphocyte ratio index (ALRI).MethodsA total of 771 HCC patients underwent radical resection from three medical centers were enrolled and grouped into the training cohort (n = 416) and validation cohort (n = 355). Prognostic prediction potential of ALRI was assessed by receiver operating curve (ROC) analysis. The Cox regression model was used to identify independent prognostic factors. Nomograms for overall survival (OS) and disease-free survival (DFS) were constructed and further validated externally.ResultsThe ROC analysis ranked ALRI as the most effective prediction marker for resected HCC patients, with the cut-off value determined at 22.6. Higher ALRI level positively correlated with larger tumor size, higher tumor node metastasis (TNM) stage, and inversely with lower albumin level and shorter OS and DFS. Nomograms for OS and DFS were capable of discriminating HCC patients into different risk-groups.ConclusionsPost-operative ALRI was of prediction value for HCC prognosis. This novel nomogram may categorize HCC patients into different risk groups, and offer individualized surveillance reference for post-operative patients.

The effect of heel elevation on the barbell squat remains controversial, and further exploration of muscle activity might help find additional evidence. Therefore, 20 healthy adult participants (10 males and 10 females) were recruited for this study to analyze the effects of heel height on lower extremity kinematics, kinetics, and muscle activity using the OpenSim individualized musculoskeletal model. One-way repeated measures ANOVA was used for statistical analysis. The results showed that when the heel was raised, the participant’s ankle dorsiflexion angle significantly decreased, and the percentage of ankle work was increased (p < 0.05). In addition, there was a significant increase in activation of the vastus lateralis, biceps femoris, and gastrocnemius muscles and a decrease in muscle activation of the anterior tibialis muscle (p < 0.05). An increase in knee moments and work done and a reduction in hip work were observed in male subjects (p < 0.05). In conclusion, heel raises affect lower extremity kinematics and kinetics during the barbell squat and alter the distribution of muscle activation and biomechanical loading of the joints in the lower extremity of participants to some extent, and there were gender differences in the results.

The regeneration of bone defects in osteoporotic patients presents a big challenge. Icariin (Ica), which is the main active component of traditional Chinese medicine - herba epimedii, has been widely loaded to scaffolds for the treatment of bone fracture and osteoporosis. To increase drug loading capacity, and meanwhile achieve a stable and safe drug delivery, in present study, a novel icariin derivative - thiolated icariin (Ica-SH) molecule with good cytocompatibility and bioactivity was incorporated into biphasic calcium phosphate (BCP) ceramic scaffold through Ica-SH intermolecular disulfide bonds (abbr., Ica-SH/BCP) to form film coating. Compared with Ica-loaded BCP scaffolds via physical adsorption (abbr., Ica/BCP), the formation of Ica-SH intermolecular disulfide bonds in Ica-SH/BCP scaffolds led to a decreased real-time drug release concentration, and a reduced drug release rate. The released drug from Ica-SH/BCP scaffolds could promote the proliferation, migration, and osteoblastic differentiation of ovariectomized rat bone mesenchymal stem cells (OVX-rBMSCs), and up-regulate the angiogenic gene expression in human umbilical vein endothelial cells (HUVECs) in vitro. The OVX-rat femoral condyle defect model also demonstrated that Ica-SH/BCP scaffolds promoted a superior bone regeneration in osteoporotic animals than BCP ones. These findings suggest that Ica-SH is a promising drug, which can be facilely incorporated into a variety of scaffolds for repairing bone defects of patients, in particular the ones with osteoporosis.

Objective: The lunge squat is one of the exercises to strengthen the lower limbs, however, there is little evidence of the effects of different equipment. The purpose of this study was to investigate the biomechanical effects of different types of equipment and loads on the lunge squat’s effect on the lower limbs. Methods: Fourteen male fitness novices participated in the experiment. Kinematics and kinetics in the sagittal plane using dumbbells, barbells, and weighted vests were measured using OpenSim. Two-way repeated measures ANOVA and one-dimensional statistical parametric mapping were used in the statistical analysis (SPM1D). Results: Range of motion (ROM) change in the knee joint was more obvious when using a barbell, whereas ROM when using a dumbbell was minimal. Compared to other joints, the joint moment at the hip joint was the largest and changed more significantly with increasing weight-bearing intensity, and the change was more pronounced with the dumbbell. For the center of pressure (COP) overall displacement, the dumbbell produced a smaller range of displacement. Conclusions: Dumbbells are suggested for male beginners to improve stability, barbells for the more experienced, and a low-weighted vest may be more appropriate for those with knee pain.

The purpose of this study was to investigate the variations of arch index from static standing to dynamic walking and running; furthermore, the interlimb symmetry was checked in the two populations. A total of eighty male participants were recruited for this study, with forty habitually barefoot and forty habitually shod males, respectively. Arch index (AI) was calculated following the previously established “gold standard” measurement via contact areas recorded from EMED. Repeated measure analysis of variance (ANOVA) was employed to compare the difference between static and dynamic walking and running arch index. Paired-samples t-test and symmetry index (SI) were used to investigate the symmetry of the left foot arch index and right foot arch index. It was found that the dynamic arch index was significantly higher than the static arch index in barefoot and shod males, showing an increase from static weight-bearing standing to dynamic walking and running. However, interlimb (right-left) symmetry in the foot arch index was observed in the two populations. Dynamic changes of the arch index may provide implications that need to be considered while designing shoe lasts or insoles. Knowledge of the healthy arch index range reported from this study could also be used as a standard baseline to probe into foot and arch disorders.

Standing long jump (SLJ) is complicated by the challenge of motor coordination in both the upper and lower segments. This movement is also considered to be a fundamental skill in a variety of sports. In particular, SLJ is an important test index for middle school students for assessing their physical fitness levels. This assessment takes the form of a physical fitness test high school entrance examination in some countries such as China. This minireview summarizes recent studies that have investigated how to improve the standing long jump performance from different aspects which include arm motion, takeoff angle, standing posture, warming-up exercise, and handheld weight. The common study limitations, controversial knowledge, and future research direction are also discussed in detail.

Abstract Background KIF18A is associated with a variety of tumours; however, the specific mechanism of action of KIF18A in hepatocellular carcinoma (HCC) remains unclear. In this study, in vitro and in vivo experiments were performed with the aim of exploring the potential function and molecular mechanism of kinesin KIF18A in the occurrence and development of HCC. Methods We detected the expression of KIF18A in tumour and adjacent tissues as well as cell proliferation, cell invasion and migration in hepatoma cells after silencing KIF18A. KIF18A-silenced hepatoma cells were subcutaneously injected into nude mice to verify the tumorigenicity of KIF18A. We also detected the expression of signal pathway-related proteins in hepatoma cells after KIF18A knockdown with the aim of exploring the association between KIF18A and related signalling pathways. Results The level of KIF18A protein was higher in liver cancer tissues than adjacent tissues. After silencing KIF18A in SMMC-7721 and HepG2 cells, cell growth was obviously inhibited; the migration and invasion abilities were significantly decreased and the in vivo tumour weight was decreased compared to the control group (0.201 ± 0.088 g vs 0.476 ± 0.126 g, p = 0.009). The expression of cell cycle-related protein (cyclin B1), invasion and metastasis-related proteins (MMP-7 and MMP-9) and Akt-related proteins in hepatoma cells was also decreased after knocking down KIF18A. Conclusions KIF18A may promote proliferation, invasion and metastasis of HCC cells by promoting the cell cycle signalling pathway as well as the Akt and MMP-7/MMP-9-related signalling pathways and may serve as a new target for the diagnosis and treatment of HCC.

The original idea for bionic shoes (BSs) involves combining the function of unstable foot conditions and the structure of the human plantar. The purpose of this study was to investigate the differences between the normal shoes (NS) and the BS during the stance phases of walking and running. A total of 15 Chinese males from Ningbo University were recruited for this study (age: 24.3 ± 2.01 years; height: 176.25 ± 7.11 cm, body weight (BW): 75.75 ± 8.35 kg). The participants were asked to perform a walking and running task. Statistical parametric mapping (SPM) analysis was used to investigate any differences between NSs and BSs during the walking and running stance phases. The results demonstrated that there were significant differences found (21.23–28.24%, p = 0.040; 84.47–100%, p = 0.017) in hip extension and flexion between the NS and the BS during the walking stance phase. There were no significant differences found in ankle and moment during the running stance phase. Significant differences were found in the rectus femoris (5.29–6.21%; p = 0.047), tibialis anterior (14.37–16.40%; p = 0.038), and medial gastrocnemius (25.55–46.86%; p < 0.001) between the NS and the BS during the walking stance phase. Significant differences were found in rectus femoris (12.83–13.10%, p = 0.049; 15.89–80.19%, p < 0.001), tibialis anterior (15.85–18.31%, p = 0.039; 21.14–24.71%, p = 0.030), medial gastrocnemius (80.70–90.44%; p = 0.007), and lateral gastrocnemius (11.16–27.93%, p < 0.001; 62.20–65.63%, p = 0.032; 77.56–93.45%, p < 0.001) between the NS and the BS during the running stance phase. These findings indicate that BSs are more efficient for muscle control than unstable shoes and maybe suitable for rehabilitation training.

Abstract Clinical outcomes of patients with hepatocellular carcinoma (HCC) are highly variable. This study aims to identify and validate a simple, readily available, and objective prognostic index for the management of HCC. Data from 724 HCC patients undergoing curative resection were evaluated and randomly divided into two cohorts for building and validating the prognostic index. A best model, NγLR = (neutrophil count [109/L] × γ-glutamyl transpeptidase [U/L]) /(lymphocyte count [109/L] × U/L), was selected. An optimal cut-off value of 103.6 for NγLR stratified patients into high NγLR (>103.6) and low NγLR (≤103.6) groups. NγLR > 103.6 was closely associated with HCC malignant characteristics. Elevated NγLR predicted a worse overall survival (OS) and progression-free survival (PFS) for HCC patients and remained an independent predictor for both types of survival. A series of poly(ethylene oxide) (PEO)/fullerene(C60) complexes are prepared by lyophilization. The intermolecular interaction and molecular motion in the complex are investigated by solid-state 13C NMR spectroscopy. An intense C60 signal due to the intermolecular cross-polarization is observed in the 13C CP/MAS spectra of the complex samples, indicating a high degree of dispersion of C60s in the complexes. By measuring the 13C spin-lattice relaxation times and 1H transverse relaxation times of the complex sample and by comparing the static 13C spectrum of the pure C60 sample with that of the complex sample, it is demonstrated that there exist n–π interactions between the n-orbitals of the PEO ether oxygen and the π-system of C60. The C60 molecules act as physical cross-links in the amorphous region of PEO, which greatly inhibit the mobility of the surrounding PEO chains, while the rapid isotropic rotation of C60 molecules is also reduced to some extent due to the interactions with the polymer chains. Prediction of prognosis of hepatocellular carcinoma (HCC) has shown an important role in improving treatment outcomes and preventing disease progression, however, the prognostic indicator of HCC is still lacking. The purpose of this study is to investigate the predictive value of GLR (gamma-glutamyl transpeptidase to lymphocyte count ratio) in single HCC with a tumor size (TS) ≤ 5 cm. A retrospective analysis was performed on 272 patients with TS ≤ 5 cm who underwent radical resection. The Pearson χ2 test was applied to discuss the relationship between HCC and GLR, alpha-fetoprotein (AFP). Then univariate and multivariate analysis was utilized to predict the risk factors for survival prognosis in patients. In this study, GLR showed a positive relation with tumor size, tumor-node-metastasis (TNM) stage, microvascular invasion, early recurrence, and serum aspartate aminotransferase (AST) level, while the AFP value only correlated with drinking. Elevated GLR value had poor overall survival (OS) and progression-free survival (PFS) of TS ≤ 5 cm HCC patients, GLR level and tumor size were closely related to the prognosis of small HCC patients compared with AFP. GLR may serve as a prognostic marker for dynamic monitoring of HCC patients with single TS ≤ 5 cm after radical resection.

This study was aimed to analyze the foot posture index and plantar pressure characteristics of fifteen badminton players and fifteen controls. The hypothesis was that people with the habit of playing badminton would be significantly different with nonplaying people in foot posture index, 3D foot surface data, and plantar pressure distribution. Nine regions of plantar pressure were measured by using the EMED force platform, and badminton players showed significantly higher peak pressure in the hallux (p=0.003), medial heel (p=0.016), and lateral heel (p=0.021) and force-time integral in the hallux (p=0.002), medial heel (p=0.026), and lateral heel (p=0.015). There is no asymmetrical plantar pressure distribution between the left foot and the right foot of players. The mean foot posture index values of male and female badminton players are 5.2 ± 1.95 and 5.7 ± 1.15, respectively, and comparatively, those values of male and female controls are 1.5 ± 1.73 and 1.7 ± 4.16, respectively. This study shows that significant differences in morphology between people with the habit of playing badminton and people without that habit could be taken as a factor for a future study in locomotion biomechanics characteristics and foot shape of badminton players and in a footwear design in order to reduce injury risks.

This study was to investigate the feasibility and safety of patient-controlled intravenous analgesia for opiate titration in patients with severe cancer. Eligible patients with cancer pain were randomly divided into two groups, which were titrated with a subcutaneous injection of morphine and patient-controlled intravenous analgesia, respectively. The time required for patients to begin titration to reach visual analog scale score ⩽ 3, the adverse reactions during titration, the time spent by nurses in titrating each patient, and the time to complete the titration with a delayed-release dosage form analgesic effect and adverse reaction. Barefoot and shod running has received increased attention in recent years, however, the influence of morphology-related foot function has not been explored. This study aimed to investigate morphology-related jumping and running biomechanical functions in habitually barefoot and shod males. A total of 90 barefoot males (Indians) and 130 shod males (Chinese), with significant forefoot and toe morphology differences, participated in a vertical jump and running test to enable the collection of kinematic and kinetic data. The difference of pressure distribution in the hallux and forefoot was shown while jumping and running. The unrestricted forefoot and toes of the barefoot group presented flexible movement and leverage functions to expand the forefoot loading area during performance of the two tasks. Findings related to morphology functions, especially in the forefoot and toe may provide useful information for footwear design.
