Reciprocal crosstalk between endocrine and immune systems has been well-documented both in physiological and pathological conditions, although the connection between the immune system and thyroid hormones (THs) remains largely unclear. Inflammation and infection are two important processes modulated by the immune system, which have profound effects on both central and peripheral THs metabolism. Conversely, optimal levels of THs are necessary for the maintenance of immune function and response. Although some effects of THs are mediated by their binding to cell membrane integrin receptors, triggering a non-genomic response, most of the actions of these hormones involve their binding to specific nuclear thyroid receptors (TRs), which generate a genomic response by modulating the activity of a great variety of transcription factors. In this special review on THs role in health and disease, we highlight the relevance of these hormones in the molecular mechanisms linked to inflammation upon their binding to specific nuclear receptors. In particular, we focus on THs effects on different signaling pathways involved in the inflammation associated with various infectious and/or pathological processes, emphasizing those mediated by NF-kB, p38MAPK and JAK/STAT. The findings showed in this review suggest new opportunities to improve current therapeutic strategies for the treatment of inflammation associated with several infections and/or diseases, such as cancer, sepsis or Covid-19 infection.

Excitotoxicity due to excessive glutamate release causes oxidative stress and neuronal death, and is a feature of many brain diseases. Here the authors show that protein kinase D1 is inactivated by excitotoxicity in a model of stroke and that its activation can be neuroprotective.

Polyethylene (PE) is the most-produced polyolefin, and consequently, it is the most widely found plastic waste worldwide. PE biodegradation is under study by applying different (micro)organisms in order to understand the biodegradative mechanism in the majority of microbes. This study aims to identify novel bacterial species with compelling metabolic potential and strategic genetic repertoires for PE biodegradation. Pseudomonas citronellolis E5 is newly isolated from solid organic waste contaminated with plastic debris, and Rhodococcus erythropolis D4 was selected for its promising potential in biodegradable plastic determined by its genetic repertoire. P. citronellolis E5 was selected for its ability to grow on PE as the only carbon and energy source. Meaningful extracellular secreted laccase activity was also characterized for D4 during growth on PE (E5 and D4 strains have a laccase activity of (2 ± 1)×10–3 U mg−1 and (3 ± 1)×10–3 U mg−1, respectively). Despite the highest level of cell numbers recorded at 7 days of growth on PE for both strains, the patterns of the metabolic products obtained and degraded during 60 days on PE were dissimilar in the two bacteria at different sampling times. However, they mainly produced metabolites belonging to carboxylic acids and alkanes with varying numbers of carbons in the aliphatic chains. Whole-genome sequence analyses of P. citronellolis E5 compared to R. erythropolis D4 and genetic determinant prediction (by gene annotation and multiple sequence alignment with reference gene products) have been performed, providing a list of 16 and 42 gene products putatively related to different metabolic steps of PE biodegradation. Altogether, these results support insights into PE biodegradation by bacteria of the Pseudomonas and Rhodococcus genera from metabolic and genetic perspectives as a base to build up novel biotechnological platforms.

Oxidative stress caused by reactive oxygen species (ROS) affects the aging process and increases the likelihood of several diseases. A new frontier in its prevention includes bioactive foods and natural extracts that can be introduced by the diet in combination with specific probiotics. Among the natural compounds that we can introduce by the diet, Panax ginseng extract is one of the most utilized since it contains a vast number of bioactive molecules such as phenolic acids, flavonoids, and polysaccharides that have been shown to possess antioxidant, anti-ageing, anti-cancer, and immunomodulatory activity. In this work, the ability of a P. ginseng extract in combination with a probiotic formulation was taken into consideration to evaluate its effects on the modulation of in vitro reconstructed human gut microbiota (HGM). After evaluating the growth of the individual strains on the ginseng extract, we tested the in vitro reconstructed HGM setup (probiotics, minimal core, and whole community) using 2% w/v ginseng as the only carbon and energy source. The probiotic strains reached the highest growth, while the minimal core and the whole community showed almost the same growth. Specifically, the presence of the ginseng extract favors L. plantarum and B. animalis subsp. lactis among the probiotics, while B. cellulosilyticus prevails over the other strains in the minimal core condition. In the presence of both probiotics and minimal core strains, L. plantarum, B. animalis subsp. lactis, and B. cellulosilyticus reach the highest growth values. The bacterial metabolites produced during ginseng extract fermentation in the three conditions were administered to human intestinal epithelial cells (HT-29) to investigate a potential antioxidant effect. Remarkably, our results highlighted a significant reduction in the total ROS and a slightly reduction in the cytosolic superoxide anion content in HT-29 cells treated with bacterial metabolites deriving from ginseng extract fermentation by the whole community.

Different Light-Absorbing Snow Impurities (LASI) can deposit on snow- and ice-covered surfaces. These particles are able to decrease snow and ice albedo and trigger positive albedo feedback. The aim of this work was to develop a new method to quantify the carbonaceous fractions that are present in snow and ice samples that contribute significantly to their darkening. Currently, in the literature, there is an absence of a unified and accepted method to perform these studies. To set up the method proposed here, snow samples were collected at two Italian locations, Claviere and Val di Pejo (Northern Italy). The samples were analyzed using two main techniques, Total Organic Carbon analysis (TOC analysis) and Thermal Optical analysis in Transmittance mode (TOT), which enabled the speciation of the carbonaceous fraction into organic (OC), inorganic (IC), and elemental carbon (EC), and further into the soluble and insoluble parts. The results highlighted a correlation between the nature of the sample (i.e., location, age, and exposure of the snow) and the experimental results, giving validity to the method. For example, the abundant presence of terrigenous constituents was reflected in high amounts of insoluble IC. Moreover, due to the trend between insoluble IC and Elemental Carbon (EC), the role of IC in TOT analysis was investigated. Indeed, IC turned out to be an interfering agent, suggesting that the two techniques (TOC analysis and TOT) are complementary and therefore need to be used in parallel when performing these studies. Finally, the results obtained indicate that the newly proposed method is suitable for studying the carbonaceous fractions in snow samples.

Due to the Covid-19 pandemic, the use of disposable face masks has been adopted worldwide as a precautionary measure to slow down the transmission of the virus. This has determined an unprecedented rise in the production of these protective equipments, and unfortunately to a new form of environmental contamination due to the improper disposal. To provide a preliminary estimation of the release of microfibers by a surgical mask dumped in the marine environment, we carried out artificial weathering experiments. Results indicated that a single surgical mask submitted to 180 hours UV-light irradiation and vigorous stirring in artificial seawater may release up to 173,000 fibers/ day. Moreover, SEM and micro-FTIR analysis carried out onto surgical masks collected from Italian beaches highlighted the same morphological and chemical degradation signature observed in the masks subjected to the artificially weathering experiments, confirming the risks of a similar microfiber release into the marine environment.

The octocoral family Alcyoniidae represents a rich source of bioactive substances with intriguing and unique structural features. This review aims to provide an updated overview of the compounds isolated from Alcyoniidae and displaying potential cytotoxic activity. In order to allow a better comparison among the bioactive compounds, we focused on molecules evaluated in vitro by using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, by far the most widely used method to analyze cell proliferation and viability. Specifically, we surveyed the last thirty years of research, finding 153 papers reporting on 344 compounds with proven cytotoxicity. The data were organized in tables to provide a ranking of the most active compounds, to be exploited for the selection of the most promising candidates for further screening and pre-clinical evaluation as anti-cancer agents. Specifically, we found that (22S,24S)-24-methyl-22,25-epoxyfurost-5-ene-3β,20β-diol (16), 3β,11-dihydroxy-24-methylene-9,11-secocholestan-5-en-9-one (23), (24S)-ergostane-3β,5α,6β,25 tetraol (146), sinulerectadione (227), sinulerectol C (229), and cladieunicellin I (277) exhibited stronger cytotoxicity than their respective positive control and that their mechanism of action has not yet been further investigated.

Maitake (Grifola frondosa) is a medicinal mushroom known for its peculiar biological activities due to the presence of functional components, including dietary fibers and glucans, that can improve human health through the modulation of the gut microbiota. In this paper, a Maitake ethanol/water extract was prepared and characterized through enzymatic and chemical assays. The prebiotic potential of the extract was evaluated by the growth of some probiotic strains and of a selected probiotic consortium. The results revealed the prebiotic properties due to the stimulation of the growth of the probiotic strains, also in consortium, leading to the production of SCFAs, including lactic, succinic, and valeric acid analyzed via GC-MSD. Then, their beneficials effect were employed in evaluating the vitality of three different healthy and tumoral colorectal cell lines (CCD841, CACO-2, and HT-29) and the viability rescue after co-exposure to different stressor agents and the probiotic consortium secondary metabolites. These metabolites exerted positive effects on colorectal cell lines, in particular in protection from reactive oxygen species.
