Portulaca oleracea (PO) is a commonly known medicinal crop that is an important ingredient for traditional Chinese medicine (TCM) due to its use as a vegetable in the diet. PO has been recorded to be frequently adulterated by other related species in the market of herbal plants, distorting the PO plant identity. Thus, identification of the botanical origin of PO is a crucial step before pharmaceutical or functional food application. In this research, a quick assay named “loop-mediated isothermal amplification (LAMP)” was built for the specific and sensitive authentication of PO DNA. On the basis of the divergences in the internal transcribed spacer 2 (ITS2) sequence between PO and its adulterant species, the LAMP primers were designed and verified their specificity, sensitivity, and application for the PO DNA authentication. The detection limit of the LAMP assay for PO DNA identification specifically was 100 fg under isothermal conditions at 63 °C for 30 min. In addition, different heat-processed PO samples can be applied for use in PO authentication in the LAMP assay. These samples of PO were more susceptible to the effect of steaming in authentication by PCR than boiling and drying treatment. Furthermore, commercial PO samples pursued from herbal markets were used to display their applicability of the developed LAMP analysis for PO postharvest authentication, and the investigation found that approximately 68.4% of PO specimens in the marketplace of herbal remedies were adulterated. In summary, the specific, sensitive, and rapid LAMP assay for PO authentication was first successfully developed herein, and its practical application for the inspection of adulteration in PO samples from the herbal market was shown. This LAMP assay created in this study will be useful to authenticate the botanical origin of PO and its commercial products.

Background Internal ribosome entry sites (IRESs) allow the translation of a transcript independent of its cap structure. They are distributed in some viruses and cellular RNA. The element is applied in dual gene expression in a single vector. Although it appears the lower efficiency of IRES-mediated translation than that of cap-dependent translation, it is with the crucial needs to know the precise differences in translational efficacy between upstream cistrons (cap-dependent) and downstream cistrons (IRES-mediate, cap-independent) before applying the bicistronic vector in biomedical applications. Material and methods This study aimed to provide real examples and showed the precise differences for translational efficiency dependent upon target gene locations. We generated various bicistronic constructs with quantifiable reporter genes as upstream and downstream cistrons of the encephalomyocarditis virus (EMCV) IRES to precisely evaluate the efficacy of IRES-mediated translation in mammalian cells. Results There was no significant difference in protein production when the reporter gene was cloned as an upstream cistron. However, lower levels of protein production were obtained when the reporter gene was located downstream of the IRES. Moreover, in the presence of an upstream cistron, a markedly reduced level of protein production was observed. Conclusion Our findings demonstrate the version of the EMCV IRES that is provided in many commercial vectors is relatively less efficient than cap-dependent translation and provide valuable information regarding the utilization of IRES to facilitate the expression of more than one protein from a transcript.

The aim of this study was to screen and isolate soil bacteria capable of conducting xylose and glucose aerobic biotransformation. Four bacterial strains were isolated from green onions field and identified as Arthrobacter species IL01, IL02, IL03 and IL04. The maximum specific growth rates of IL01, IL02, IL03 and IL04 during xylose fermentation are 0.076, 0.094, 0.128 and 0.088 hr–1, respectively. Those four strains also present the ability to simultaneously conduct glucose and xylose biotransformation. This study indicated that the aerobic bacterial strains isolated could provide a potential bioprocess for bioenergy production in the future.

Background: Cinnamomum osmophloeum (Co), a member of the Lauraceae, is an indigenous medicinal crop in Taiwan, and it contains higher cinnamaldehyde in essential oil than do other Cinnamomum species. Among these species, Cinnamomum burmannii (Cb) is frequently adulterated as Co because of their similar morphological characteristics or features. Objective: The purpose of this study was to develop a DNA-based molecular method for rapid authentication of the indigenous Co and prevention of its adulteration. Materials and Methods: The internal transcribed spacer (ITS) regions of nuclear ribosomal DNA from various Cinnamomum species were amplified by polymerase chain reaction (PCR), and these obtained sequences were used for sequence analysis. Based on the sequence variants among various Cinnamomum species, restriction fragment length polymorphism (RFLP) was used to differentiate these Cinnamomum plants. Results: Two restriction endonucleases, MylI and EcoRV, were specifically used to digest the PCR-amplified ITS DNA from seven Cinnamomum species. The PCR-RFLP results demonstrated that the different restriction patterns that were produced by these two restriction enzymes clearly distinguished Co from Cb and five other Cinnamomum species simultaneously. Conclusion: The PCR-RFLP analysis developed in this study provides an alternative method for rapidly identifying Cinnamomum plants at the species level using DNA polymorphisms.

The purpose of this study is to evaluate the efficiency of municipal wastewater treatment by a subsurface cobble-bed biofilm system (SCBS) in Taipei, Taiwan. In contrast to traditional wastewater treatment facilities, SCBS uses cobbles as the contact media in the biofilm treatment unit. In this study, the SCBS consists of a series of underground treatment units, including a sedimentation tank, a grit chamber, two bar screens, a pumping station, a distribution tank, a collection tank and an effluent tank. At the flowrate of 4000 m3/day, the average influent concentrations for biochemical oxygen demand, suspended solid, ammonium nitrogen, and total phosphorus were 66.99 mg/L, 26.14 mg/L, 17.33 mg/L, and 1.96 mg/L, respectively. After 39 months of operation, the measured influent and effluent results show that the treatment efficiencies obtained from the SCBS for biochemical oxygen demand, suspended solid, ammonium nitrogen, and total phosphorus are 91.3%, 84.0%, 84.0%, and 26.0%, respectively. The result of a first-order kinetic analysis shows that the NH3-N degradation constant is greater than the BOD degradation constant in this cobble-bed biofilm unit. Probability analysis revealed that the SCBS may be an attractive alternative from the perspectives of treatment efficiency for municipal wastewater treatment. Klebsiella spp. were found to be the predominant species in the biofilm system in the SCBS.

The perfect ginseng radix is collected when the ginseng root reaches a cultivation age of six years; this ensures the best mass quality and consistency of the plant's essential bioactive components. Since traditional means of authentication via physical appearance or smell are hardly reliable, an efficient analytical method that can determine the real cultivation age of dried ginseng radix in commercial products, especially ginseng products of various dosage forms, is urgently required. In the present study, chemical fingerprint by 1H-NMR spectroscopy was used on dried ginseng radix samples with cultivation ages ranging from 1–6 years. The resulting dataset was then analyzed by using principle component analysis and cluster analysis to build up a distributive model that allows the identification of the real cultivation age of the ginseng radix based on a plant metabolomic strategy. This quality surveillance method was able to clearly discriminate the 6 years old ginseng radix from the other ages, and could be applied on the evaluation of the real cultivation age for the various dried white ginseng radix samples and commercial products accurately.

Chicken anemia virus (CAV) is an anemia agent of breeder and young chicks. This virus is the cause of economic losses across the chicken industry worldwide as a consequence of severe anemia and immunodeficiency among the birds. Two genes of CAV encoding the VP1 and VP2 proteins were cloned and expressed in Escherichia coli BL21 (DE3). A Western blot assay using His-tag antiserum was used to assess the expression level of the CAV viral proteins in E. coli. The results demonstrated that only full-length VP2 can be successfully expressed in E. coli, but not full-length VP1. A serial of N-terminus deletions of the VP1 protein, VP1Nd30, VP1Nd60 and VP1 Nd129, were created using PCR in order to improve VP1 expression. The results demonstrated that all three of these recombinant VP1 mutant proteins can be expressed in E. coli. VP1Nd129 protein demonstrates the highest expression level compared to the other two proteins. The specificity of Nd129-VP1 and VP2 protein were confirmed by mass spectrometry. By comparing the expression level of VP1Nd129 and VP2 protein after the addition of IPTG, the results indicated that the VP1Nd129 protein gave a higher level of protein expression than VP2. The highest yields of VP1Nd129 and VP2 were 26.2 and 15.5 mg/L, respectively, after IPTG induction with 0.1 mM IPTG for 6 h, respectively. The identification of the optimized conditions for production of the CAV viral proteins VP1 and VP2 will allow them to be used in the future as an antigen for the development of vaccines and diagnostic tests.

This work evaluates the addition of solid phase oxygen, a magnesium peroxide (MgO(2)) formulation manufactured by Regenesis (oxygen-releasing compounds, ORC), to inhibit the production of hydrogen sulfide (H(2)S) in an SRB-enriched environment. The initial rate of release of oxygen by the ORC was determined over a short period by adding sodium sulfite (Na(2)SO(3)), which was a novel approach developed for this study. The ability of ORCs to control H(2)S by releasing oxygen was evaluated in a bench-scale column containing cultured sulfate reducing bacteria (SRB). After a series of batch tests, 0.4% ORC was found to be able to inhibit the formation of H(2)S for more than 40 days. In comparison, the concentration of H(2)S dropped from 20 mg S/L to 0.05 mg S/L immediately after 0.1% hydrogen peroxide (H(2)O(2)) was added, but began to recover just four days later. Thus, H(2)O(2) does not seem to be able to inhibit the production of sulfide for an extended period of time. By providing long-term inhibition of the SRB population, ORC provides a good alternative means of controlling the production of H(2)S in water.

The biotransformation of octylphenol polyethoxylate surfactant (Triton X-100) and bacterial communities in soil microcosms was investigated. The soil microcosms were designed to simulate real sites of bioremediation. The soil used in this study was contaminated with pesticides and alkylphenol polyethoxylate surfactants over a long period of time. The nitrogen source, (NH4)2SO4 and the mineral salt basal solution were added to the microcosm and the water content was adjusted to 50% with distilled water. The microcosms were aerated using an aeration system with an air flow rate of 0.3 l min(-1). The exogenous bacterial strain Pseudomonas sp. SH4 was added to the microcosms to increase the rate of biotransformation of the Triton X-100. The number of microorganisms and the theoretical extent of formation of carbon dioxide were estimated to evaluate the biotransformation of Triton X-100 in the microcosms. The analytical results revealed that the exogenous bacteria could increase the rate of transformation of Triton X-100 by approximately 50%. Aeration of the microcosm increased the biotransformation of Triton X-100 by 45%. Bacterial count of 6.8x10(10) MPN g(-1) of soil was achieved in the M5 microcosm. Analysis of the bacterial community by 16S rDNA sequences revealed that Pseudomonas sp. SH4 could dominate all the microcosms to which it was added as an exogenous bacteria. The quantity of the indigenous bacterial strains Stenotrophomonas maltophilia, Stenotrophomonas sp. and clone 4-70 were also enhanced in the microcosms by the aeration, and the addition of carbon and nitrogen source.

Background Chicken anemia virus (CAV), the causative agent chicken anemia, is the only member of the genus Gyrovirus of the Circoviridae family. CAV is an immune suppressive virus and causes anemia, lymph organ atrophy and immunodeficiency. The production and biochemical characterization of VP1 protein and its use in a subunit vaccine or as part of a diagnostic kit would be useful to CAV infection prevention. Results Significantly increased expression of the recombinant full-length VP1 capsid protein from chicken anemia virus was demonstrated using an E. coli expression system. The VP1 gene was cloned into various different expression vectors and then these were expressed in a number of different E. coli strains. The expression of CAV VP1 in E. coli was significantly increased when VP1 was fused with GST protein rather than a His-tag. By optimizing the various rare amino acid codons within the N-terminus of the VP1 protein, the expression level of the VP1 protein in E. coli BL21(DE3)-pLysS was further increased significantly. The highest protein expression level obtained was 17.5 g/L per liter of bacterial culture after induction with 0.1 mM IPTG for 2 h. After purification by GST affinity chromatography, the purified full-length VP1 protein produced in this way was demonstrated to have good antigenicity and was able to be recognized by CAV-positive chicken serum in an ELISA assay. Conclusions Purified recombinant VP1 protein with the gene's codons optimized in the N-terminal region has potential as chimeric protein that, when expressed in E. coli, may be useful in the future for the development of subunit vaccines and diagnostic tests.

Chicken anemia virus (CAV) is an important viral pathogen that causes anemia and severe immunodeficiency syndrome in chickens worldwide. In this study, a potential diagnostic monoclonal antibody against the CAV VP1 protein was developed which can precisely recognize the CAV antigen for diagnostic and virus recovery purposes. The VP1 gene of CAV encoding the N-terminus-deleted VP1 protein, VP1Nd129, was cloned into an Escherichia (E.) coli expression vector. After isopropyl-β-D-thiogalactopyronoside induction, VP1Nd129 protein was shown to be successfully expressed in the E. coli. By performing an enzyme-linked immunoabsorbent assay using two coating antigens, purified VP1Nd129 and CAV-infected liver tissue lysate, E3 monoclonal antibody (mAb) was found to have higher reactivity against VP1 protein than the other positive clones according to the result of limiting dilution method from 64 clones. Using immunohistochemistry, the presence of the VP1-specific mAb, E3, was confirmed using CAV-infected liver and thymus tissues as positive-infected samples. Additionally, CAV particle purification was also performed using an immunoaffinity column containing E3 mAb. The monoclonal E3 mAb developed in this study will not only be very useful for detecting CAV infection and performing histopathology studies of infected chickens, but may also be used to purify CAV particles in the future.
