Summary: Infections caused by non‐tuberculous mycobacteria (NTM) are reported as emerging disease in many countries worldwide. The occurrence of NTM in different hosts and their implication as obligate or opportunistic pathogen remain largely unclear. Lymph nodes and faecal samples of clinically healthy Swiss cattle at slaughter were analysed for the presence of NTM. Based on the examined lymph nodes, NTM were detected in 20% of 108 cattle originating from different premises. The 22 isolates belonged to five different species of Mycobacteria (M. avium subsp. hominissuis, M. kansasii, M. persicum, “M. lymphaticum” and M. europaeum). M. avium subsp. hominissuis (63%) and M. kansasii (18%) thereby predominated and were found in lymph nodes with and without macroscopic changes. Moreover, M. persicum found in two cattle has recently been described as a human pathogen and is closely related to M. kansasii. Amongst cattle with lymph nodes positive for mycobacteria, viable NTM were occasionally also detected in bovine faeces. However, the isolated NTM species from lymph nodes and respective faecal samples (M. hassiacum, M. phlei and M. vaccae) did not coincide. Moreover, NTM species identified amongst isolates from the slaughterhouse environment clearly differed from those from lymph nodes and faecal samples, excluding cross‐contamination of the tissue specimens through the environment or laboratory processing. Assuming that some NTM interfere with the detection of bovine tuberculosis (bTB), the present findings in healthy animals emphasize the need of more specific diagnostic tools for bTB eradication programs. The awareness of geohazards in the subaqueous environment has steadily increased in the past years and there is an increased need to assess these hazards in a quantitative sense. Prime examples are subaqueous landslides, which can be triggered by a number of processes including earthquakes or human activities, and which may impact offshore and onshore infrastructure and communities. In the literature, a plenitude of subaqueous landslide events are related to historical earthquakes, including cases from lakes in Switzerland. Here, we present an approach for a basin-wide earthquake-triggered subaquatic landslide hazard assessment for Lake Zurich, which is surrounded by a densely populated shoreline. Our analysis is based on high-resolution sediment-mechanical and geophysical input data. Slope stabilities are calculated with a grid-based limit equilibrium model on an infinite slope, which uses Monte Carlo sampled input data from a sediment-mechanical stratigraphy of the lateral slopes. Combined with probabilistic ground-shaking forecasts from a recent national seismic hazard analysis, subaquatic earthquake-triggered landslide hazard maps are constructed for different mean return periods, ranging from 475 to 9975 years. Our results provide a first quantitative landslide hazard estimation for the lateral slopes in Lake Zurich. Furthermore, a back-analysis of a case-study site indicates that pseudostatic accelerations in the range between 0.04 and 0.08 g were needed to trigger a well-investigated subaqueous landslide, dated to ~2210 cal. years B.P. Subaqueous landslides and their consequences, such as tsunamis, can cause serious damage to offshore infrastructure and coastal communities. Stability analyses of submerged slopes are therefore crucial, yet complex steps for hazard assessment, as many geotechnical and morphological factors need to be considered. Typically, deterministic models with data from a few sampling locations are used for the evaluation of slope stabilities, as high efforts are required to ensure high spatial data coverage. This study presents a simple but flexible approach for the probabilistic stability assessment of subaqueous slopes that takes into account the spatial variability of geotechnical data. The study area (~2 km) in Lake Zurich (northern Switzerland) shows three distinct subaquatic landslides with well-defined headscarps, translation areas (i.e. the zone where translational sliding occurred) and mass transport deposits. The ages of the landslides are known (~2,210 and ~640 cal. yr BP, and 1918 AD), and their triggers have been assigned to different mechanisms by previous studies. A combination of geophysical, geotechnical, and sedimentological methods served to analyse the subaquatic slope in great spatial detail: 3.5 kHz pinger seismic reflection data and a 300 kHz multibeam bathymetric dataset (1 m grid) were used for the detection of landslide features and for the layout of a coring and an in situ cone penetration testing campaign. The assignment of geotechnical data to lithological units enabled the construction of a sediment-mechanical stratigraphy that consists of four units, each with characteristic profiles of bulk density and shear strength. The thickness of each mechanical unit can be flexibly adapted to the local lithological unit thicknesses identified from sediment cores and seismic reflection profiles correlated to sediment cores. The sediment-mechanical stratigraphy was used as input for a Monte Carlo simulated limit-equilibrium model on an infinite slope for the assessment of the present slope stability and for a back analysis of past landslides in the study area, both for static and earthquake-triggered scenarios. The results show that the location of failure initiation in the model is consistent with stratigraphic analysis and failure-plane identification from sediment cores. Furthermore, today's sediment-charged slopes are failure-prone, even for a static case. This approach of including an adaptable sediment-mechanical stratigraphy into a limit-equilibrium slope stability analysis may be applied as well to the marine realm. [ABSTRACT FROM AUTHOR]

Background: The assessment of a liver biopsy remains the gold standard for diagnosing and staging fatty liver in dairy cows, which is often necessary for diagnostic and research purposes. Accuracy of the diagnosis relays on the quality of the biopsy, the assumed representativeness of a small tissue sample for a disease process throughout the liver and accurate human evaluation of histologic specimens. The objective of the present study was to assess the distribution of triacylglycerol (TAG) infiltration throughout the parenchyma of livers with different degrees of fatty liver in dairy cows. In addition, histopathological scores from the corresponding specimens were compared to a quantitative measurement of TAG, as well as the agreement between two observers. Methods: Thirty livers with different degrees of lipid infiltration were selected and 10 different locations throughout the liver were assessed. The TAG content was measured enzymatically, calculated in % or mg/g wet weight, and assigned to a scoring system. Corresponding tissue specimens were stained with hematoxylin-eosin (H&E) and Oil red O (ORO) for histopathological evaluation, using a scoring system. Results: The difference in TAG content between any locations was less than 2%. Based on the scoring system the TAG concentration was even distributed in 79.3% of the livers. Based on kappa statistics the agreement between two pathologists and staining technique in scoring histological specimens was moderate to fair. Conclusions: Overall the distribution of TAG throughout the liver and the accuracy of human evaluation of liver biopsies may lead to acceptable diagnoses for clinical purposes. Within the liver lobules a common pattern of lipid distribution depending on severity could be observed. Intramyocardial abscesses are rare in cattle and may lead to unspecific clinical signs. This case report describes the clinical and echocardiographic findings in an 8 year old Brown Swiss cow presented with an intramural myocardial abscess. The main clinical findings were anorexia, drop in milk yield, fever, tachycardia, and hyperfibrinogenemia. Neither heart murmurs nor cardiac arrhythmias were present on auscultation. Transthoracic echocardiographic examination revealed a prominent intramural mass embedded in the left ventricular free wall and bulging into the lumen of the left ventricle. Background Feline pancreas-specific lipase (Spec fPL) is considered a useful test for the antemortem diagnosis of pancreatitis in cats. A recent study found good agreement between the results of the Spec fPL and catalytic 1,2- o-dilauryl-rac-glycero-3-glutaric acid-(6′-methylresorufin) ester ( DGGR) lipase assay. Prospective studies evaluating their sensitivity and specificity are lacking. Objectives To compare the results of the Spec fPL and the DGGR assays with a standardized histologic assessment of the pancreas. Animals Sixty client-owned cats presented for necropsy. Methods Prospective study: Spec fPL concentrations and serum DGGR lipase activity were measured from the same blood sample. The pancreas was removed within 3 hours after euthanasia; serial transverse sections were made every 0.5 cm throughout the entire pancreas and reviewed using a histologic grading scheme. Sensitivity and specificity for the Spec fPL and DGGR assay results were determined. Results The sensitivity and specificity for the Spec fPL assay (cutoff value ≥5.4 μg/L) was 42.1 [95% confidence interval (95% CI), 29.4-55.9%] and 100% (95% CI, 31.0-100.0%). The sensitivity and specificity for the DGGR assay (cutoff value >26 U/L) was 36.8 (95% CI, 24.7-50.7%) and 100% (95% CI, 31.0-100.0%). When lymphocytic inflammation up to 10% of a section was considered normal, the sensitivity and specificity for Spec fPL assay (cutoff value ≥5.4 μg/L) was 61.1 (95% CI, 36.1-81.7%) and 69.0% (95% CI, 52.8-81.9%) and the sensitivity and specificity for the DGGR assay (cutoff value >26 U/L) was 66.7 (95% CI, 41.2-85.6%) and 78.6% (95% CI, 62.8-89.2%). In Switzerland, dicrocoeliasis is regarded as the most significant parasitic infection of llamas and alpacas. Fasciola hepatica infestation is also a problem but less common. The aim of the present retrospective study was to evaluate the lungs of New World camelids (NWCs) for evidence of arterial hypertension in association with liver changes due to liver fluke infestation. The lungs of 20 llamas and 20 alpacas with liver fluke infestation were histologically evaluated. The hematoxylin and eosin and van Gieson (VG)–elastica stains as well as immunohistology for the expression of α–smooth muscle actin (α-SMA) were used to visualize the structures of arterial walls. Parasitology of fecal matter (11 llamas and 17 alpacas) confirmed that most of these animals were infested with both Dicrocoelium dendriticum and other gastrointestinal parasites. In most cases (10/12 llamas, 4/6 alpacas), liver enzyme activity in serum was elevated. Histologically, arteries in the lungs of 9 of 20 llamas (45%) and 3 of 20 alpacas (15%) showed severe intimal and adventitial and slight to moderate medial thickening, which was confirmed with α-SMA and VG-elastica staining. All animals exhibited typical liver changes, such as fibrosis and biliary hyperplasia, in association with the presence of liver flukes. This study shows that liver flukes can induce proliferative changes in lung arteries in NWCs that resemble those seen with pulmonary arterial hypertension due to liver parasites in humans. However, the degree of liver fluke infestation was not correlated with the extent of liver damage, or with the amount of thoracic or abdominal effusion or pulmonary arterial changes. Summary: Two horses were presented with non-specific clinical signs of several weeks' duration and were humanely destroyed due to a poor prognosis. At necropsy examination, both horses had multiple small, white nodules replacing pancreatic tissue and involving the serosal surface of the abdominal cavity, the liver and the lung. Microscopically, neoplastic cells were organized in acini and contained abundant (case 1) or sparse (horse 2) intracytoplasmic zymogen granules. Immunohistochemically, both tumours expressed amylase and pan-cytokeratin, but not insulin or neuron-specific enolase. In case 2, a low percentage of neoplastic cells expressed glucagon and synaptophysin. The presence of zymogen granules was confirmed in both cases by electron microscopy and occasional fibrillary or glucagon granules were observed in cases 1 and 2, respectively. Cattle persistently infected with a noncytopathic Bovine viral diarrhea virus (BVDV) are at risk of developing fatal “mucosal disease” (MD). The authors investigated the role of various apoptosis pathways in the pathogenesis of lesions in animals suffering from MD. Therefore, they compared the expression of caspase-3, caspase-8, caspase-9, and Bcl-2L1 (Bcl-x) in tissues of 6 BVDV-free control animals, 7 persistently infected (PI) animals that showed no signs of MD (non-MD PI animals), and 11 animals with MD and correlated the staining with the localization of mucosal lesions. Caspase-3 and -9 staining were markedly stronger in MD cases and were associated with mucosal lesions, even though non-MD PI animals and negative controls also expressed caspase-9. Conversely, caspase-8 was not elevated in any of the animals analyzed. Interestingly, Bcl-x also colocalized with mucosal lesions in the MD cases. However, Bcl-x was similarly expressed in tissues from all 3 groups, and thus, its role in apoptosis needs to be clarified. This study clearly illustrates ex vivo that the activation of the intrinsic, but not the extrinsic, apoptosis pathway is a key element in the pathogenesis of MD lesions observed in cattle persistently infected with BVDV. However, whether direct induction of apoptosis in infected cells or indirect effects induced by the virus are responsible for the lesions observed remains to be established. Gastrointestinal stromal tumors (GISTs) are defined as specific CD117-(Kit, stem cell factor receptor) expressing tumors of the gastrointestinal (GI) tract. They are believed to originate from the interstitial pacemaker cells of Cajal (ICC) or their progenitor cells. In horses only a few cases of GISTs are described in the literature. The innovative BoneWelding(®) technology, where ultrasound energy bonds bioresorbable implants to bone, was tested for its feasibility in spine surgery and its local thermal effects. The three tested concepts consisted of implementation of a resorbable plating system, two converging polymer pins and suture anchors to the cervical vertebral bodies. Bioresorbable polylactide implants (PLDLLA 70/30) were inserted ventrally into the third and fourth vertebral body of seven sheep, of which six were sacrificed at 2 months and one sheep immediately after temperature measurements during implant insertion. Polymer screws were used as controls. Qualitative, semi-quantitative histological, and quantitative histomorphometrical evaluation showed excellent anchorage of the implants, new mineralized bone at the implant-bone interface, no inflammatory cell reaction or thermal damage to the adjacent bone in response to the novel insertion technology. The application of two converging pins, parallel inserted polymer pins, or fusion of the implant to the polymer plates did not affect the overall excellent tissue tolerance of the technology. The article examines the distribution of borna disease virus (BDV) antigen in tissues of naturally infected bicolored white-toothed shrews, Crocidura leucodon. The authors mention that immunohistochemistry and TaqMan real-time reverse transcription polymerase chain reaction (RT-PCR) were used in the assessment. They report that the study indicate that BDV antigen and nucleic acid were identified in several organs like the nerve tissue and neurons and in parenchymal cells.

A 13-year-old neutered male European short-hair cat was presented because of blindness and behavioural abnormalities. On physical examination, abnormal behaviour, compulsive walking, circling, continuous vocalization and blindness were the main neurological signs. In addition, abdominal alopecia, thin and inelastic skin, weight loss despite polyphagia, polyuria and polydipsia were present. Laboratory investigation revealed diabetes mellitus and pituitary-dependent hypercortisolism. Diagnostic imaging showed bilaterally enlarged adrenals and a large pituitary mass. Summary: A bull aged 16 months with bilateral testicular hypoplasia and azoospermia was persistently infected with bovine viral diarrhoea virus (BVDV). Viral antigen was detected in serum and semen by ELISA, but the animal was serologically negative. After slaughter, the genital tract was examined histopathologically and by immunohistochemistry, including double immunolabelling with BVDV antibody and either S-100 antibody (for Sertoli cells) or ferritin antibody (for Leydig cells). The seminiferous tubules of both testes were lined by a single layer of Sertoli cells and the germinal epithelium was completely absent except for a few remaining spermatogonia. BVDV antigen was demonstrated (1) in the media of arterial vessel walls of the testis, epididymis, urethra, prostate, and vesicular and bulbourethral glands, (2) in epithelial cells of the ductus epididymidis, the accessory glands and the urethra, and (3) in the testis, mainly in Sertoli cells and to a lesser extent in the spermatogonia that remained, but not in Leydig cells. In a retrospective study, covering the period from 1986 to 1995, tissues of aborted fetuses were re-examined. A total of 347 cases were tested immunohistochemically, among them samples of 223 brains were examined for Neospora caninum, Toxoplasma gondii and Bovine Virus Diarrhoea Virus (BVDV), and 249 placentae for Chlamydiaceae. Two real-time PCR assays, one for N. caninum, and one for T. gondii, were developed. These potential abortion-inducing agents were detected – and confirmed by PCR, except for BVDV – in 16.1% ( N. caninum), 0% ( T. gondii), 9.9% (BVDV) and 0.8% ( Chlamydiales) of the cases examined. Immunohistochemistry proved to be inadequate for the detection of the protozoal epitopes, whereas it was confirmed as a very useful tool for the detection of BVDV. In abortion material, PCR is considered to be more suitable for the detection of protozoa and Chlamydophila abortus, an adequate sampling presupposed. [ABSTRACT FROM AUTHOR]

Summary: Three dogs (two Rottweilers and a Flat-coated retriever) showed various neurological signs, including apathy, depression, circling, a partial decrease in functions associated with cranial nerves, seizures, hyperaesthesia, proprioceptive deficits, and increased spinal reflexes. In all three cases, necropsy revealed a solid, distinct, white mass in the brain and multiple, poorly demarcated, firm nodular proliferations in the lung; in one case the liver was also affected. Histopathological examination showed loosely aggregated, pleomorphic cells, with abundant eosinophilic cytoplasm. The neoplastic cells sometimes contained vacuoles or phagocytized cells. Binucleated and multinucleated giant cells, and mitotic figures, were common. Immunohistochemically, the tumour cells reacted strongly for lysozyme and vimentin, but there was no reaction for S-100 protein, cytokeratin, CD3 or CD79a. The histological and immunohistochemical examinations indicated a histiocytic origin of the tumour cells and malignant histiocytosis was therefore diagnosed. [Copyright &y& Elsevier]

Summary: Equine and feline dysautonomias are characterized histopathologically by degenerating neurons with chromatolysis, pyknotic and sometimes eccentric nuclei, and loss of Nissl substance in the peripheral autonomic ganglia. Because it may be difficult to distinguish pathological from post-mortem changes in affected ganglia by histopathological examination, synaptophysin was evaluated as an immunohistochemical marker. Degenerating neurons showed strong intracytoplasmic labelling indicating abnormal accumulation of synaptophysin. It was concluded that synaptophysin immunohistochemistry is a helpful tool for detecting degenerating neurons in equine (grass sickness) and feline (Key-Gaskell syndrome) dysautonomias. This study involved 13 calves, one to 50 days of age, born to first calf heifers that had been pastured on one of seven alpine communal pastures in the canton of St. Gallen during the summer of 1995. Of a total of 993 cattle pastured, 61 were pregnant heifers that were negative for bovine virus diarrhoea (BVD) antigen and for BVD antibodies at the start of pasturing. Seroconversion occurred in 26 of these pregnant heifers during the pasture period. Blood samples and skin biopsy specimens of calves born to 13 of these were examined for BVD antigen by antigen-ELISA and by an immunohistochemical technique, respectively. Blood samples were positive for BVD antigen in four calves, questionable in one calf, negative in seven and missing in one prematurely born calf. The four calves that were positive for BVD antigen in the blood were also positive in skin biopsies. Of the seven calves with a negative or missing blood test, six had positive and two had negative skin samples. Based on the combined results of blood and skin testing, 11 of 13 calves were positive for BVD antigen. Of the 11 infected calves, six were normal at birth, four were smaller than normal and one was premature and weak and was euthanized on humane grounds. Of the four small calves, two developed diarrhoea and died within the first month of life. The two calves that were negative for BVD antigen were clinically normal. The results of this study not only demonstrate the occurrence of in-utero infection with BVD virus, but also stress the importance of alpine communal pasturing in the spread of BVD virus. Because the prevention of infection with BVD virus on communal pastures does not seem feasible, it is recommended that all calves born to cows from such pastures be tested for BVD antigen. This report describes a buck with cerebral gliomatosis. The animal was severely apathetic to somnolent. Neurological examination revealed generalised ataxia and hypermetria of the fore limbs. There was bilateral mydriasis and severely decreased menace and pupillary light reflexes. Sensitivity to pricking with a needle was markedly reduced over the entire body. There was a delayed response to adduction, abduction and crossing of the limbs and rocking of the animal. Examination of cerebrospinal fluid indicated mild mixed-cell inflammation. Based on all of the findings, an abscess or tumour of the central nervous system with localisation in the cerebrum was suspected. Because of the grave prognosis, the goat was euthanased and a post mortem examination performed. No macroscopic abnormalities were seen in any of the organs including the brain. Histologically, there was extensive diffuse glial cell hyperplasia in the white matter of the cerebral hemispheres and in the brain stem. [ABSTRACT FROM AUTHOR]

The article discusses the study which shows the evidence of BTV-8 field strains to cross the placental barrier in sheep under experimental conditions. The study used three Dorset ewes in which each respondent is inoculated and administered intradermal injections. In addition, the study found low presence of antibody titres of the lamb after experimental BTV-8 infection in sheep with strain-dependent feature.
