BackgroundDuring the wars in Afghanistan and Iraq most injuries to service members involved the musculoskeletal system. These wounds often occurred around joints, and in some cases result in traumatic arthrotomy-a diagnosis that is not always clear, especially when there is no concomitant articular fracture. The aim of the present study is to evaluate the diagnosis and treatment of peri-articular blast injuries without fracture.MethodsThe study cohort included 12 consecutive patients (12 involved extremities) who sustained peri-articular blast wounds of the extremities without fractures. The diagnosis of penetrating articular injury was based on clinical examination, radiographic findings, or aspiration. A peri-articular wound was defined as any wound, or radio-opaque blast fragment, within 5 cm of a joint. The New Injury Severity Score (NISS) was calculated for each patient. Four patients had upper, and 8 patients had lower extremity injuries. Nine of 12 patients had joint capsular penetration and underwent joint irrigation and debridement.ResultsTwo patients had retained intra-articular metal fragments. One patient had soft tissue blast wounds within 5 cm of a joint but did not have joint capsule penetration. There were no significant differences (p = 0.23) between the distribution of wounds to upper versus lower extremities. However, there were a significantly greater number of blast injuries attributed to Improvised Explosive Devices (IEDs) than from other blast mechanisms (p = 0.01).ConclusionExtremity blast injuries in the vicinity of joints involving only soft tissues present a unique challenge in surgical management. A high index of suspicion should be maintained for joint capsular penetration so that intra-articular injuries may be appropriately treated.

Today, considering the increased cases of terroristic operations, structural design engineers are starting to acknowledge blast-induced loads alongside other design parameters. Since a blast-induced load exhibits a much shorter period than the natural period of the structure, it is unlikely to induce general damages or stimulate the entire structure immediately after the loading, but rather local damages in individual structural elements are the common form of damage. These local damages, however, can set the scene for the gradual failure of the structure. The present study on structures with moment-resisting frames, buckling-restrained braces (BRBs), and conventional steel braces was conducted through an alternative load path (ALP, also known as column elimination) and exact modeling of the blast on the steel frames was undertaken using LS-DYNA software. In this work, the behavior of three five-story steel frames under a blast-induced load of 100 kg TNT equivalent at a distance of 10 m from the structural columns for a period of 100 ms was investigated. Results of analyzing the blast-induced effect on the frames showed that the frame equipped with buckling-restrained braces (BRB) exhibited improved overall peak displacement across the entire frame, peak induced velocity, and peak induced acceleration by 30%, 30%, and 46%, respectively, as compared to other steel frames exposed to blast loads. This paper analyzed the technical principle and advantages of GB-SAR, and combined engineering practices to study the site selection, data acquisition scheme and data processing of GB-SAR station, and made a complete emergency monitoring scheme. Taking the construction blasting scene of an engineering construction as an example, the blasting point was located in the central position where the whole reinforced slope needs to be monitored. The emergency monitoring was carried out by using GB-SAR and the deformation information was obtained by using time-series data processing method, which verified the feasibility of the proposed emergency monitoring scheme and the reliability of data processing. The results show that, the blasting has little influence on the well-supported structure, and the shape variable is less than 1. 5 mm. After the explosion, the position shape variable near the center increases sharply, and the maximum value can reach 103 mm during the monitoring period. It is further confirmed that the GB-SAR can effectively monitor the impact of blasting on the surrounding construction scene, and can provide strong support for construction operations and project risk investigation. From terrorist attacks to industrial accidents, comprehending how structures respond to blast waves is critical for ensuring public safety and designing resilient structures. Studying these effects typically involves two main methods: free-field tests with live explosives and shock tube tests. Although shock tube testing offers certain advantages, both approaches are costly and demand significant space. This research aims to develop a cost-effective and straightforward technique for generating stress waves that closely replicate the progressive and spatial characteristics of free-field or shock tube blast waves. This method was designed to evaluate the dynamic response of laminated glass panels. The stress wave was generated by impacting a piston on the fluid inside a tube, which was connected to a fluid chamber. This setup produced impulsive loads that were distributed across a laminated glass test panel. Moreover, it was used to simulate the shock near filed explosions for a certain part of a structure. High-speed cameras were utilized to analyze the initial velocity of flying glass fragments. The apparatus successfully produced various blast waves and impulsive profiles for different drop weight heights. The initial velocities of randomly selected flying shards ranged from 3 m/s to 4 m/s. Abstract Interferon-gamma (IFN-γ) is a versatile pleiotropic cytokine essential for both innate and adaptive immune responses. It exhibits both pro-inflammatory and anti-inflammatory properties, making it a promising therapeutic candidate for treating various infectious diseases and cancers. We present IFNepitope2, a host-specific technique to annotate IFN-γ inducing peptides, it is an updated version of IFNepitope introduced by Dhanda et al. In this study, dataset used for developing prediction method contain experimentally validated 25,492 and 7983 IFN-γ inducing peptides in human and mouse host, respectively. In initial phase, machine learning techniques have been exploited to develop classification model using wide range of peptide features. Further, to improve machine learning based models or alignment free models, we explore potential of similarity-based technique BLAST. Finally, a hybrid model has been developed that combine best machine learning based model with BLAST. In most of the case, models based on extra tree perform better than other machine learning techniques. In case of peptide features, compositional feature particularly dipeptide composition performs better than one-hot encoding or binary profile. Our best machine learning based models achieved AUROC 0.89 and 0.83 for human and mouse host, respectively. The hybrid model achieved the AUROC 0.90 and 0.85 for human and mouse host, respectively. All models have been evaluated on an independent/validation dataset not used for training or testing these models. Newly developed method performs better than existing method on independent dataset. This paper summarizes the blast events and resulting damage found on more than 100 bridges in Iraq caused by terrorist attacks. Most structures were two to three lanes wide and composed of a superstructure with prestressed concrete beams that supported a concrete deck. In most cases, terrorists placed charges either on the deck or at the base of columns. Typical structural system failures were due to either intermediate column failure, girder end failure, or girder end failure in combination with midspan failure. It was found that a single blast load on the deck rarely caused collapse, while three or more blast loads distributed across the deck nearly always caused deck and/or span collapse. If at least two girders remained largely undamaged, one lane of traffic could often be directed safely over the structure, allowing a heavily damaged bridge to remain in temporary use. When columns were attacked, charges placed symmetrically caused the columns to collapse downward, whereas charges placed asymmetrically caused significant lateral movement. Although most bridges were simple span, substantial resiliency was observed, where the structures could support large amounts of load even when some critical components theoretically had near-zero capacity. It was further found that separating components, even slightly, could significantly reduce blast damage propagation. Brief recommendations are given to mitigate blast damage. [ABSTRACT FROM AUTHOR]

We present the characterization of the entomopathogenic nematode isolate LD_CU, and its associated symbiotic entomopathogenic bacteria. Morphological data of the present strain showed similitude with type population of Steinernema siamkayai, with small morphometric variations. BLASTn analysis of the ITS rRNA sequence of the nematode revealed 100% similarity with S. siamkayai, confirming conspecificity. The pairwise alignment showed no nucleotide differences with the type population of S. siamkayai. Phylogenetic analysis based on ITS rRNA gene sequences supported these findings, placing isolate LD_CU in a monophyletic clade with S. siamkayai, which forms a sister clade with S. huense, S. cumgarense, S. tami, and S. minutum. Biochemical tests identified the associated bacterial symbiont as Xenorhabdus stockiae. The isolate tested negative for oxidase, catalase, nitrate reductase, O-Nitrophenyl-β-D galactopyranoside (ONPG), methyl red, tryptophan deaminase, indole production, ornithine, lysine, citrate, malonate, hydrogen sulfide, acetoin, and phenylalanine deaminase, but positive for urea and esculin hydrolysis, motility, and acid production from glucose fermentation. The bacteria exhibited neutral red adsorption on MacConkey agar and formed brownish pigmented colonies on nutrient agar. Molecular characterization using 16S rRNA gene sequences revealed 100% similarity with X. stockiae and 90% similarity with X. innexi, indicating significant divergence from other Xenorhabdus species. Phylogenetic analysis showed a close relation to X. stockiae VP-2016b and distinct differences from X. innexi DSM 16336T. Pairwise alignment confirmed no nucleotide differences between the present bacterial strain and X. stockiae. This detailed and comprehensive profiling supports the accurate identification of Steinernema siamkayai and its bacterial symbiont, Xenorhabdus stockiae, and contributes valuable information to the taxonomy and phylogeny of these organisms.

Abstract Background Trauma can result in systemic inflammation that leads to organ dysfunction, but the impact on the brain, particularly following extracranial insults, has been largely overlooked. Methods Building upon our prior findings, we aimed to understand the impact of systemic inflammation on neuroinflammatory gene transcripts in eight brain regions in rats exposed to (1) blast overpressure exposure [BOP], (2) cutaneous thermal injury [BU], (3) complex extremity injury, 3 hours (h) of tourniquet-induced ischemia, and hind limb amputation [CEI+tI+HLA], (4) BOP+BU or (5) BOP+CEI and delayed HLA [BOP+CEI+dHLA] at 6, 24, and 168 h post-injury (hpi). Results Globally, the number and magnitude of differentially expressed genes (DEGs) correlated with injury severity, systemic inflammation markers, and end-organ damage, driven by several chemokines/cytokines (Csf3, Cxcr2, Il16, and Tgfb2), neurosteroids/prostaglandins (Cyp19a1, Ptger2, and Ptger3), and markers of neurodegeneration (Gfap, Grin2b, and Homer1). Regional neuroinflammatory activity was least impacted following BOP. Non-blast trauma (in the BU and CEI+tI+HLA groups) contributed to an earlier, robust and diverse neuroinflammatory response across brain regions (up to 2–50-fold greater than that in the BOP group), while combined trauma (in the BOP+CEI+dHLA group) significantly advanced neuroinflammation in all regions except for the cerebellum. In contrast, BOP+BU resulted in differential activity of several critical neuroinflammatory-neurodegenerative markers compared to BU. t-SNE plots of DEGs demonstrated that the onset, extent, and duration of the inflammatory response are brain region dependent. Regardless of injury type, the thalamus and hypothalamus, which are critical for maintaining homeostasis, had the most DEGs. Our results indicate that neuroinflammation in all groups progressively increased or remained at peak levels over the study duration, while markers of end-organ dysfunction decreased or otherwise resolved. Conclusions Collectively, these findings emphasize the brain's sensitivity to mediators of systemic inflammation and provide an example of immune-brain crosstalk. Follow-on molecular and behavioral investigations are warranted to understand the short- to long-term pathophysiological consequences on the brain, particularly the mechanism of blood–brain barrier breakdown, immune cell penetration–activation, and microglial activation.

The article explores the extent to which medieval polemical authors resorted to patristic originals and how much they adopted patristic argumentation. The authors used computational text reuse analysis using BLAST (Basic Local Alignment Search Tool) to compare 189 classical and medieval texts, mainly from open repositories of digitized texts, to find similarities. The corpus includes classical works, particularly Augustine’s anti-heretical treatises, canon law, inquisition manuals, exempla collections and florilegia, sermons, and theological commentaries. The lack of medieval texts after ca. 1200 in machine-readable format is the greatest hindrance to building a representative medieval corpus. The authors propose that although medieval polemicists saw Augustine and other Church fathers as models of Christian champions fighting heresy, intensive engagement with patristic theology took place in medieval works with limited circulation and influence. Abstract Background Precise evaluation of fungal conidia production may facilitate studies on resistance mechanisms and plant breeding for disease resistance. In this study, hyperspectral imaging (HSI) was used to quantify the sporulation of Magnaporthe oryzae on the leaves of rice cultivars grown under controlled conditions. Three rice genotypes (CO 39, Nipponbare, IR64) differing in susceptibility to blast were inoculated with M. oryzae isolates Guy 11 and Li1497. Spectral information (450–850 nm, 140 wavebands) of typical leaf blast symptoms was recorded before and after induction of sporulation of the pathogen. Results M. oryzae produced more conidia on the highly susceptible genotype than on the moderately susceptible genotype, whereas the resistant genotype resulted in no sporulation. Changes in reflectance spectra recorded before and after induction of sporulation were significantly higher in genotype CO 39 than in Nipponbare. The spectral angle mapper algorithm for supervised classification allowed for the classification of blast symptom subareas and the quantification of lesion areas with M. oryzae sporulation. The correlation between the area under the difference spectrum (viz. spectral difference without and with sporulation) and the number of conidia per lesion and the number of conidia per lesion area was positive and count-based differences in rice - M. oryzae interaction could be reproduced in the spectral data. Conclusions HSI provided a precise and objective method of assessing M. oryzae conidia production on infected rice plants, revealing differences that could not be detected visually.

Due to the high energy absorption capability and excellent bending strength of the sandwich panels, they are mostly preferred as protective structures against explosive blast attacks. The evaluation of their blast-mitigation characteristics through experiments is highly dangerous, costly, time-consuming, and polluting for the environment. Therefore, in this presented work, a series of numerical analyses were performed to evaluate the blast mitigation of the outstanding sandwich panels with honeycomb, corrugated, auxetic, and foam cores. The masses of sandwich panels were kept constant, and their areal densities were maintained constant throughout the study to effectively compare their performance under identical air blast loading conditions. To apply the air blast loads to the designed sandwiches, 1–3kg of spherical-shaped trinitrotoluene charges were used for a 100mm stand-off distance. The sandwiches are made of high-strength AL-6XN steel and crushable aluminum foam. The rate-dependent Johnson–Cook constitutive model of plasticity and the crushable foam model with volumetric hardening were used for the evaluation of sandwich panels’ plastic deformations. The findings of the work depicted that the honeycomb core is more efficient than the other core structures of the same masses because the sandwich panels with honeycomb core provide smaller back skin deflections, globalized core crushing, and higher core energy absorption than the other sandwich panels for extreme conditions of air blast loading. Blast-related mild traumatic brain injury (blast-mTBI) can result in a spectrum of persistent symptoms leading to substantial functional impairment and reduced quality of life. Clinical evaluation and discernment from other conditions common to military service can be challenging and subject to patient recall bias and the limitations of available assessment measures. The need for objective biomarkers to facilitate accurate diagnosis, not just for symptom management and rehabilitation but for prognostication and disability compensation purposes is clear. Toward this end, we compared regional brain [18F]fluorodeoxyglucose–positron emission tomography ([18F]FDG-PET) intensity-scaled uptake measurements and motor, neuropsychological, and behavioral assessments in 79 combat Veterans with retrospectively recalled blast-mTBI with 41 control participants having no lifetime history of TBI. Using an agnostic and unbiased approach, we found significantly increased left pallidum [18F]FDG-uptake in Veterans with blast-mTBI versus control participants, p < 0.0001; q = 3.29 × 10−9 [Cohen's d, 1.38, 95% confidence interval (0.96, 1.79)]. The degree of left pallidum [18F]FDG-uptake correlated with the number of self-reported blast-mTBIs, r2 = 0.22; p < 0.0001. Greater [18F]FDG-uptake in the left pallidum provided excellent discrimination between Veterans with blast-mTBI and controls, with a receiver operator characteristic area under the curve of 0.859 (p < 0.0001) and likelihood ratio of 21.19 (threshold:SUVR ≥ 0.895). Deficits in executive function assessed using the Behavior Rating Inventory of Executive Function–Adult Global Executive Composite T-score were identified in Veterans with blast-mTBI compared with controls, p < 0.0001. Regression-based mediation analyses determined that in Veterans with blast-mTBI, increased [18F]FDG-uptake in the left pallidum-mediated executive function impairments, adjusted causal mediation estimate p = 0.021; total effect estimate, p = 0.039. Measures of working and prospective memory (Auditory Consonant Trigrams test and Memory for Intentions Test, respectively) were negatively correlated with left pallidum [18F]FDG-uptake, p < 0.0001, with mTBI as a covariate. Increased left pallidum [18F]FDG-uptake in Veterans with blast-mTBI compared with controls did not covary with dominant handedness or with motor activity assessed using the Unified Parkinson's Disease Rating Scale. Localized increased [18F]FDG-uptake in the left pallidum may reflect a compensatory response to functional deficits following blast-mTBI. Limited imaging resolution does not allow us to distinguish subregions of the pallidum; however, the significant correlation of our data with behavioral but not motor outcomes suggests involvement of the ventral pallidum, which is known to regulate motivation, behavior, and emotions through basal ganglia–thalamo–cortical circuits. Increased [18F]FDG-uptake in the left pallidum in blast-mTBI versus control participants was consistently identified using two different PET scanners, supporting the generalizability of this finding. Although confirmation of our results by single-subject-to-cohort analyses will be required before clinical deployment, this study provides proof of concept that [18F]FDG-PET bears promise as a readily available noninvasive biomarker for blast-mTBI. Blast is the most common injury mechanism in conflicts of this century due to the widespread use of explosives, confirmed by recent conflicts such as in Ukraine. Data from conflicts in the last century such as Northern Ireland, the Falklands, and Vietnam up to the present day show that between 16% and 21% of personnel suffered a traumatic brain injury. Typical features of fatal brain injury to those outside of a vehicle (hereafter referred to as dismounted) due to blast include the presence of hemorrhagic brain injury alongside skull fractures rather than isolated penetrating injuries more typical of traditional ballistic head injuries. The heterogeneity of dismounted blast has meant that analysis from databases is limited and therefore a detailed look at the radiological aspects of injury is needed to understand the mechanism and pathology of dismounted blast brain injury. The aim of this study was to identify the head and spinal injuries in fatalities due to dismounted blast. All UK military fatalities from dismounted blast who suffered a head injury from 2007–2013 in the Iraq and Afghanistan conflicts were identified retrospectively. Postmortem computerized tomography images (CTPMs) were interrogated for injuries to the head, neck, and spine. All injuries were documented and classified using a radiology brain injury classification (BIC) tool. Chi-squared (χ2) and Fisher's exact tests were used to investigate correlations between injuries, along with odds ratios for determining the direction of correlation. The correlations were clustered. There were 71 fatalities from dismounted blast with an associated head injury with a CTPM or initial CT available for analysis. The results showed the heterogeneity of injury from dismounted blast but also some potential identifiable injury constellations. These were: intracranial haemorrhage, intracranial deep haemorrhage, spinal injury, and facial injury. These identified injury patterns can now be investigated to consider injury mechanisms and so develop mitigation strategies or clinical treatments. Level of Evidence: Observational. Rice, a critical staple on a global scale, faces escalating challenges in yield preservation due to the rising prevalence of abiotic and biotic stressors, exacerbated by frequent climatic fluctuations in recent years. Moreover, the scorching climate prevalent in the rice-growing regions of South China poses obstacles to the cultivation of good-quality, heavy-grain varieties. Addressing this dilemma requires the development of resilient varieties capable of withstanding multiple stress factors. To achieve this objective, our study employed the broadspectrum blast-resistant line Digu, the brown planthopper (BPH)-resistant line ASD7, and the heavy-grain backbone restorer lines Fuhui838 (FH838) and Shuhui527 (SH527) as parental materials for hybridization and multiple crossings. The incorporation of molecular markers facilitated the rapid pyramiding of six target genes (Pi5, Pita, Pid2, Pid3, Bph2, and Wxb ). Through a comprehensive evaluation encompassing blast resistance, BPH resistance, cold tolerance, grain appearance, and quality, alongside agronomic trait selection, a promising restorer line, Guihui5501 (GH5501), was successfully developed. It demonstrated broad-spectrum resistance to blast, exhibiting a resistance frequency of 77.33% against 75 artificially inoculated isolates, moderate resistance to BPH (3.78 grade), strong cold tolerance during the seedling stage (1.80 grade), and characteristics of heavy grains (1,000-grain weight reaching 35.64 g) with good grain quality. The primary rice quality parameters for GH5501, with the exception of alkali spreading value, either met or exceeded the second-grade national standard for premium edible rice varieties, signifying a significant advancement in the production of good-quality heavy-grain varieties in the southern rice-growing regions. Utilizing GH5501, a hybrid combination named Nayou5501, characterized by high yield, good quality, and resistance to multiple stresses, was bred and received approval as a rice variety in Guangxi in 2021. Furthermore, genomic analysis with gene chips revealed that GH5501 possessed an additional 20 exceptional alleles, such as NRT1.1B for efficient nitrogen utilization, SKC1 for salt tolerance, and STV11 for resistance to rice stripe virus. Consequently, the restorer line GH5501 could serve as a valuable resource for the subsequent breeding of high-yielding, good-quality, and stress-tolerant hybrid rice varieties. Background: Precise evaluation of fungal conidia production may facilitate studies on resistance mechanisms and plant breeding for disease resistance. In this study, hyperspectral imaging (HSI) was used to quantify the sporulation of Magnaporthe oryzae on the leaves of rice cultivars grown under controlled conditions. Three rice genotypes (CO 39, Nipponbare, IR64) differing in susceptibility to blast were inoculated with M. oryzae isolates Guy 11 and Li1497. Spectral information (450–850 nm, 140 wavebands) of typical leaf blast symptoms was recorded before and after induction of sporulation of the pathogen. Results: M. oryzae produced more conidia on the highly susceptible genotype than on the moderately susceptible genotype, whereas the resistant genotype resulted in no sporulation. Changes in reflectance spectra recorded before and after induction of sporulation were significantly higher in genotype CO 39 than in Nipponbare. The spectral angle mapper algorithm for supervised classification allowed for the classification of blast symptom subareas and the quantification of lesion areas with M. oryzae sporulation. The correlation between the area under the difference spectrum (viz. spectral difference without and with sporulation) and the number of conidia per lesion and the number of conidia per lesion area was positive and count-based differences in rice - M. oryzae interaction could be reproduced in the spectral data. Conclusions: HSI provided a precise and objective method of assessing M. oryzae conidia production on infected rice plants, revealing differences that could not be detected visually. 2023 was an important year for research in traumatic brain injury (TBI), particularly as it concerned interests in neuropathology. After reviewing the literature, we present the advancements that we felt were of particular importance to the neuropathology community. Highlighted are articles that report upon: (1) the first large-cohort assessment for the neuropathology of intimate partner violence, (2) the assessment of chronic traumatic encephalopathy (CTE) in young athletes, (3) the observation of cortical sulcal depth vascular changes in CTE, (4) a proposal for a tau immunohistochemical panel to evaluate complex cases of CTE in the context of multiple tauopathies, (5) the relationship of TBI and/or CTE with TDP-43 pathology, (6) repetitive TBI inducing pathology in C9orf72-transgenic mice, (7) radiologic patterns of head and neck injury following vehicular underbody blast exposure, (8) chronic alterations in brain metal content following repetitive impact TBI, (9) neurovascular unit injury following low-level blast exposure, and finally (10) an assessment of Muhammad Ali’s clinical history leading to the conclusion that he suffered from young-onset, idiopathic Parkinson Disease. We close our writing with in memoriam to Dr. Byron A. Kakulas, a renowned figure in the neuropathology of spinal cord injury who we lost in 2023.

Abstract Large-scale flank collapses are one of the main hazards associated with the evolution of volcanic islands. Precisely dating such events is critical to evaluate the frequency of destabilization episodes and further assess the triggering mechanism(s) associated with internal and/or external factors, such as volcano dynamics, regional tectonics, and global paleoclimatic changes. Here, we constrain the age of a pumice-rich pyroclastic deposit exposed on the eastern flank of Flores Island (Azores), which we interpret as a co-blast deposit generated by a major flank collapse that destroyed the whole western flank of the former volcanic edifice. Twelve single-grain 40Ar/39Ar analyses, performed on 250–500 µm anorthoclase feldspars (mean K/Ca close to 5) with our high-sensitivity multi-collector NGX mass spectrometer, provide a robust weighted mean age of 1.32 ± 0.01 Ma for this eruption. This new age is consistent with previous K/Ar data bracketing the flank collapse between 1.30 ± 0.04 and 1.18 ± 0.09 Ma, and indicates that this event occurred at the end of the main construction phase of the volcano. The explosion produced pumice-rich layers preceded by a lahar as attested by a polygenetic mudflow deposit underlying the dated deposit. From the geochemistry of lavas erupted just before and after the collapse, we speculate upon the possible role of magmatic processes on flank destabilization. We propose a first hypothesis where differentiation in a shallow magma reservoir could have favored edifice inflation, ground shaking, and flank failure, triggering a decompression-induced violent eruption. Overall, our study shows that high-sensitivity mass spectrometers have now reached analytical performances allowing to measure precisely and accurately ages on relatively small and moderately K-rich single feldspars, which is of the utmost importance for dating heterogeneous blasts and tephra deposits that may have been induced by large-scale flank collapses during the late Quaternary.

Rice as a staple food for very large population suffers from various biotic and abiotic stresses. Among the biotic stresses, Blast, Bacterial leaf blight and Brown plant hopper (BPH) are considered to most potential threats that significantly affect the rice productivity. The use of chemical usage for controlling these diseases and pest attacks is not environmentally friendly and is expensive. Using Molecular Marker assisted backcross breeding program we have improved one of our hybrid parental line with 5 genes (xa13, Xa21, Pi54, Bph20 and Bph21). We have developed 15 near isogenic lines having similar agronomical characters as of recipient parent, they are promising for their direct induction in breeding program. Our results indicated that out of fifteen lines, Six lines i.e. GK 101-12, GK 101-15, GK 101-9, GK 101-5 and GK 101-2 out-performed in all the three locations tested, they had not only have very high level of resistance to BLB, Blast and BPH but also showed significantly higher yield compared to susceptible check as well as recipient parent. These lines may be used for improvement of our existing hybrid and also for developing of new hybrids with other CMS lines. These lines are also useful for developing new set of restorer lines through (R x R) and also useful for introgression of wide spectrum of resistance to the varieties/hybrid.

Rice blast, caused by the fungus Magnaporthe oryzae, and brown planthopper (BPH) infestation are two of the most destructive problems of rice production in China. The development of multi-resistant varieties is widely recognized as the most efficient and environmentally friendly approach to controlling crop diseases and pests. Functional molecular markers (FMMs) have been developed from functional variants in the genic region associated with trait variation, greatly enhancing the efficiency of identifying and pyramiding valuable genes in crop breeding. In this study, two FMMs and a multiplex PCR amplification system were developed for two major broad-spectrum BPH resistance genes, Bph6 and Bph9. With the assistance of FMMs in the multi-resistant rice restorer-line development pipeline, two lines (Huahui7713 and Huahui3006) with blast and BPH resistance were developed by pyramiding three resistance genes Pigm, Bph6 and Bph9. Two new hybrid rice varieties, Weiliangyou7713 and Xuanliangyou3006, derived from Huahui7713 and Huahui3006, have been developed and commercialized in China. Weiliangyou7713 and Xuanliangyou3006 exhibit enhanced resistance to both blast and BPH, while maintaining optimal yield and grain quality. The adoption of Weiliangyou7713 continues to expand, now being cultivated on a large scale, which is promising for its future role in reducing the dependence on chemical fungicides and pesticides in rice production. This suggests that the implementation of Huahui7713 and Huahui3006 in targeted breeding programs could be highly beneficial for developing rice varieties with strong resistance to blast and BPH. Background: The family Geminiviridae contains plant viruses commonly referred to as geminiviruses, which cause diseases in numerous important plants. Geminiviridae is one of the most important family of plant-infecting viruses that has been divided into fourteen recognized genera. Amongst these, the genus Begomovirus is the largest and economically most important one, viruses of the genus are further categorized into bipartite (two genomic components called DNA-A and DNA-B) and monopartite (single genomic component or DNA-A) begomoviruses. Method: The genomic DNA was extracted from symptomatic plant samples and both the DNA-(B and A) components of a bipartite begomovirus were amplified via rolling circular amplification (RCA) and PCR. The amplified DNA-A and DNA-B were cloned and sequenced by Primer walking, Sanger sequencing, and Illumina sequencing methods. The phylogenetic and biogeographic analyses of both components were performed by various bioinformatics tools. Results: DNA-A (accession number MW722701) and DNA-B (accession number MW722782) of begomoviruses that were sequenced shared >95% sequence similarity with the bipartite begomovirus known as Tomato Leaf Curl New Delhi virus (TLCuNDV). Phylogenetic and biogeographic analyses proposed several possible ancestors, and multiple dispersal and vicariance events may be involved in the evolution of this virus. Conclusions: This is the first time a bipartite begomovirus has been found to infect a plant in the study area (District Swat, Pakistan). The increased use of chemicals in rice farming poses significant issues regarding the emergence of pesticide/fungicide resistance and environmental sustainability concerns. This study was aimed at the genetic improvement of blast, bacterial leaf blight (BB) and gall midge resistance in a popular rice variety CO 51 which already harbours a blast resistance gene Pi54. Efforts were made to pyramid an additional blast resistance gene Pi9 along with two BB resistance genes (xa13 and Xa21) and two gall midge resistance genes (Gm1 and Gm4) into an elite rice variety CO 51 to enhance the resistance level to biotic stresses. The superior lines were selected using functional markers conferring resistance to blast (NBS4 and Pi54MAS linked to Pi9 and Pi54 genes, respectively) and BB [(xa13Prom (xa13) and pTA248 (Xa21)] and SSR markers linked to Gm1 (RM1328) and Gm4 (RM22550) for phenotypic screening and agronomic evaluation. The genotyping and phenotyping of F6 and BC2F6 progenies of CO 51 X 562-4, for agronomic traits and resistance to BB and blast, identified ten superior progenies in F6 and five superior progenies in BC2F6. The breeding lines harbouring both xa13+Xa21 exhibited high levels of resistance to BB (score ≤ 1 cm) and Pi9+Pi54 exhibited strong resistance to blast (score ≤ 2). Introduction: Although traumatic brain injury (TBI) has been linked with adverse long-term health, less research has examined whether TBI is linked with nonclinical outcomes including involuntary job loss. Symptoms associated with TBI may influence one’s ability to maintain gainful employment including employment in the U.S. military. That influence may impact military service members with exposure to repetitive low-level blast (LLB). Understanding the association between TBI and involuntary job loss outcomes among military populations is particularly important as it may be associated with differences in eligibility for post-service benefits. The purpose of the present research was to determine whether (1) TBI and related conditions are associated with involuntary job loss (i.e., medical and administrative separations from service) among military personnel, and (2) occupational risk of LLB is associated with involuntary job loss in both the presence and absence of clinical diagnoses of TBI and related conditions. Method: This research leveraged population-level data from the Career History Archival Medical and Personnel System for enlisted personnel who served on active duty between 2005–2015. Risk of LLB exposure was categorized using military occupational specialty as a proxy. Medical diagnoses were identified using ICD-9 codes. Separations for medical and administrative reasons were identified. Results: Risk for administrative separation differed across medical diagnoses of interest, but those who worked in high-risk occupations were more likely to be administratively separated than those working in low-risk occupations. Risk for medical separation was associated with occupational risk of LLB and each of the diagnoses of interest, though significant interactions suggested that the effects of certain diagnoses of interest (e.g., concussion, cognitive problems, postconcussive syndrome, migraines) on medical separations was greater among those working in high-risk occupations. Discussion: Taken together, the present research suggests that TBI and associated medical conditions, as well as occupational risk of LLB, are associated with long-term involuntary job loss for medical reasons. This study is the first to demonstrate involuntary military job loss outcomes associated with TBI, mental health conditions, and conditions associated with blast exposure using both inpatient and outpatient population-level data and may have important implications for civilian employment and post-service benefits. The glycocalyx is a proteoglycan–glycoprotein structure lining the luminal surface of the vascular endothelium and is susceptible to damage due to blast overpressure (BOP) exposure. The glycocalyx is essential in maintaining the structural and functional integrity of the vasculature and regulation of cerebral blood flow (CBF). Assessment of alterations in the density of the glycocalyx; its components (heparan sulphate proteoglycan (HSPG/syndecan-2), heparan sulphate (HS), and chondroitin sulphate (CS)); CBF; and the effect of hypercapnia on CBF was conducted at 2–3 h, 1, 3, 14, and 28 days after a high-intensity (18.9 PSI/131 kPa peak pressure, 10.95 ms duration, and 70.26 PSI·ms/484.42 kPa·ms impulse) BOP exposure in rats. A significant reduction in the density of the glycocalyx was observed 2–3 h, 1-, and 3 days after the blast exposure. The glycocalyx recovered by 28 days after exposure and was associated with an increase in HS (14 and 28 days) and in HSPG/syndecan-2 and CS (28 days) in the frontal cortex. In separate experiments, we observed significant decreases in CBF and a diminished response to hypercapnia at all time points with some recovery at 3 days. Given the role of the glycocalyx in regulating physiological function of the cerebral vasculature, damage to the glycocalyx after BOP exposure may result in the onset of pathogenesis and progression of cerebrovascular dysfunction leading to neuropathology. Injuries and subclinical effects from exposure to blasts are of significant concern in military operational settings, including tactical training, and are associated with self-reported concussion-like symptomology and physiological changes such as increased intestinal permeability (IP), which was investigated in this study. Time-series gene expression and IP biomarker data were generated from "breachers" exposed to controlled, low-level explosive blast during training. Samples from 30 male participants at pre-, post-, and follow-up blast exposure the next day were assayed via RNA-seq and ELISA. A battery of symptom data was also collected at each of these time points that acutely showed elevated symptom reporting related to headache, concentration, dizziness, and taking longer to think, dissipating ~16 h following blast exposure. Evidence for bacterial translocation into circulation following blast exposure was detected by significant stepwise increase in microbial diversity (measured via alpha-diversity p = 0.049). Alterations in levels of IP protein biomarkers (i.e., Zonulin, LBP, Claudin-3, I-FABP) assessed in a subset of these participants (n = 23) further evidenced blast exposure associates with IP. The observed symptom profile was consistent with mild traumatic brain injury and was further associated with changes in bacterial translocation and intestinal permeability, suggesting that IP may be linked to a decrease in cognitive functioning. Rice blast and bacterial leaf blight, are major disease, significantly threatens rice yield in all rice growing regions under favorable conditions and identification of resistance genes and their superior haplotypes is a potential strategy for effectively managing and controlling this devastating disease. In this study, we conducted a genome-wide association study (GWAS) using a diverse set of 147 rice accessions for blast and bacterial blight diseases in replications. Results revealed 23 (9 for blast and 14 for BLB) significant marker-trait associations (MTAs) that corresponded to 107 and 210 candidate genes for blast and BLB, respectively. The haplo-pheno analysis of the candidate genes led to the identification of eight superior haplotypes for blast,with an average SES score ranging from 0.00 to 1.33, and five superior haplotypes for BLB, with scores ranging from 1.52cm to 4.86cm superior haplotypes. Among these, superior haplotypes LOC_OS12G39700-H4 and LOC_Os06g30440-H33 were identified with the lowest average blast scores of 0.00-0.67, and superior haplotype LOC_Os02g12660-H39 exhibited the lowest average lesion length (1.88 - 2.06cm) for BLB. A total of ten accessions for blast and eight accessions for BLB were identified carrying superior haplotypes were identified. These haplotypes belong to aus and indx subpopulations of five countries (Bangladesh, Brazil, India, Myanmar, and Pakistan). For BLB resistance, eight accessions from six countries (Bangladesh, China, India, Myanmar, Pakistan, and Sri Lanka) and four subpopulations (aus, ind1A, ind2, and ind3) were identified carrying superior haplotypes. Interestingly, four candidate genes, LOC_Os06g21040, LOC_Os04g23960, LOC_Os12g39700, and LOC_Os01g24640 encoding transposon and retrotransposon proteins were among those with superior haplotypes known to play a crucial role in plant defense responses. These identified superior haplotypes have the potential to be combined into a single genetic background through haplotype-based breeding for a broader resistance spectrum against blast and bacterial blight diseases. Many military veterans who experienced blast-related traumatic brain injuries in the conflicts in Iraq and Afghanistan currently suffer from chronic cognitive and mental health problems that include depression and post-traumatic stress disorder (PTSD). Male rats exposed to repetitive low-level blast develop cognitive and PTSD-related behavioral traits that are present for more than 1 year after exposure. We previously reported that a group II metabotropic receptor (mGluR2/3) antagonist reversed blast-induced behavioral traits. In this report, we explored mGluR2/3 expression following blast exposure in male rats. Western blotting revealed that mGluR2 protein (but not mGluR3) was increased in all brain regions studied (anterior cortex, hippocampus, and amygdala) at 43 or 52 weeks after blast exposure but not at 2 weeks or 6 weeks. mGluR2 RNA was elevated at 52 weeks while mGluR3 was not. Immunohistochemical staining revealed no changes in the principally presynaptic localization of mGluR2 by blast exposure. Administering the mGluR2/3 antagonist LY341495 after behavioral traits had emerged rapidly reversed blast-induced effects on novel object recognition and cued fear responses 10 months following blast exposure. These studies support alterations in mGluR2 receptors as a key pathophysiological event following blast exposure and provide further support for group II metabotropic receptors as therapeutic targets in the neurobehavioral effects that follow blast injury. The use of chemical usage for controlling these diseases and pest attacks is not environmentally friendly and is expensive. Using Molecular Marker assisted backcross breeding program we have improved one of our hybrid parental line with 5 genes (xa13, Xa21, Pi54, Bph20 and Bph21). We have developed 15 near isogenic lines having similar agronomical characters as of recipient parent, they are promising for their direct induction in breeding program. Our results indicated that out of fifteen lines, Six lines i.e. GK 101-12, GK 101-15, GK 101-9, GK 101-5 and GK 101-2 out-performed in all the three locations tested, they had not only have very high level of resistance to BLB, Blast and BPH but also showed significantly higher yield compared to susceptible check as well as recipient parent. These lines may be used for improvement of our existing hybrid and also for developing of new hybrids with other CMS lines. These lines are also useful for developing new set of restorer lines through (R x R) and also useful for introgression of wide spectrum of resistance to the varieties/hybrid. [ABSTRACT FROM AUTHOR]

Background: Acute myeloid leukemia (AML) is a hematological malignancy generally marked by the unregulated proliferation of myeloid series blast cells. The condition of hematologic malignancy is often associated with increased anti-apoptotic activity. One of the Bcl-2 protein families, Bcl-xL, has an important role in controlling apoptosis / programmed cell death in hematologic malignancies. This study, determined the correlations between anti-apoptotic activity from Bcl-xL expression analysis with the number of bone marrow blasts, CD34 activity as a marker of blast cells, and CD7 as an aberration marker are often found in AML patients. Aim: Analysis of the correlation between blast number, and expression of Bcl-xL, CD34, and CD7 in adult Acute Myeloid Leukemia patients. Method: An observational cross-sectional study was performed on 30 adult patients who have recently been diagnosed with Acute Myeloid Leukemia using bone marrow aspiration for examination of the number of blasts by a microscope. Examination of the expression of Bcl-xL, CD34, and CD7 was performed by BD FACSCalibur based on the measured Mean Fluorescence Intensity (MFI). Results: A total of 30 AML patients had a range of blast count 20 - 82%, Bcl-xL expression with MFI 93.06 - 441.09, CD34 expression with MFI 1.06 - 1,452.48, CD7 expression with MFI 9.31 - 90.58. In this study, there was no significant correlation between Bcl-xL expression as an indicator of anti-apoptotic properties with blast count r = 0.118 (p = 0.534), CD34 expression r 0.225 (p = 0.231) and CD7 expression r = 0.148 (p = 0.435). Conclusion: Bcl-xL expression as an indicator of anti-apoptotic properties in adult AML patients had no correlations with the proliferation of blast cells in AML. Our objective was to evaluate the combined effect of blast freezing vacuum packaged USDA Low Choice Longissimus lumborum beef steaks followed by consumer freezing of retail overwrapped steaks upon objective and subjective measures of beef palatability. The experimental design utilized a randomized complete block with a 3 × 3 treatment structure of targeted freezing treatments initially in vacuum and secondly in overwrap packaging (unfrozen = NOT; blast frozen at −34.4°C = BF; consumer-frozen at −17.8°C = CF) to accomplish 9 treatment combinations. Descriptive sensory attributes were evaluated by trained panelists on a 100-point line scale. Slice shear force and expressible moisture were assessed. Data were analyzed via PROC GLIMMIX using a randomized complete block design with a 3 × 3 treatment structure. Of descriptive panel attributes, overall juiciness was the only interaction observed (P = 0.006). Though similar to steaks initially CF in the vacuum package and followed by a second freeze (CF/BF or CF/CF) in the overwrap package, steaks singly frozen to simulate a CF (CF/NOT and NOT/CF) resulted in the overall driest ratings by panelists (P = 0.006). Only bloody/serumy differed (P = 0.002) within the initial freeze, where steaks not frozen (NOT) in the vacuum package were rated higher than those that were frozen (BF or CF). During the second freeze in the overwrap package after retail display, steaks BF rated higher for oxidized (P = 0.051) off-flavor than steaks CF and higher than both CF and NOT steaks for refrigerator-stale (P = 0.006) off-flavor; all other attributes did not differ (P ≥ 0.155). Although some freezing combinations that included CF were generally lower for overall juiciness, BF vacuum packaged steaks had no effect on palatability when compared to NOT steaks. The beef industry and consumers should feel confident using freezing as a means to extend shelf-life of beef steaks.

Abstract Background During the wars in Afghanistan and Iraq most injuries to service members involved the musculoskeletal system. These wounds often occurred around joints, and in some cases result in traumatic arthrotomy—a diagnosis that is not always clear, especially when there is no concomitant articular fracture. The aim of the present study is to evaluate the diagnosis and treatment of peri-articular blast injuries without fracture. Methods The study cohort included 12 consecutive patients (12 involved extremities) who sustained peri-articular blast wounds of the extremities without fractures. The diagnosis of penetrating articular injury was based on clinical examination, radiographic findings, or aspiration. A peri-articular wound was defined as any wound, or radio-opaque blast fragment, within 5 cm of a joint. The New Injury Severity Score (NISS) was calculated for each patient. Four patients had upper, and 8 patients had lower extremity injuries. Nine of 12 patients had joint capsular penetration and underwent joint irrigation and debridement. Results Two patients had retained intra-articular metal fragments. One patient had soft tissue blast wounds within 5 cm of a joint but did not have joint capsule penetration. There were no significant differences (p = 0.23) between the distribution of wounds to upper versus lower extremities. However, there were a significantly greater number of blast injuries attributed to Improvised Explosive Devices (IEDs) than from other blast mechanisms (p = 0.01). Conclusion Extremity blast injuries in the vicinity of joints involving only soft tissues present a unique challenge in surgical management. A high index of suspicion should be maintained for joint capsular penetration so that intra-articular injuries may be appropriately treated.

Due to the growing need for sustainable and ultra-high-strength construction materials, scientists have created an innovative ultra-high-performance concrete called Geopolymer based ultra-high-performance concrete (GUHPC). Besides, in the last few decades, there have been a lot of explosions and ballistic attacks around the world, which have killed many civilians and fighters in border areas. In this context, this article reviews the fresh state and mechanical properties of GUHPC. Firstly, the ingredients of GUHPC and fresh properties such as setting time and flowability are briefly covered. Secondly, the review of compressive strength, flexure strength, tensile strength and modulus of elasticity of fibrous GUHPC. Thirdly, the blast and projectile impact resistance performance was reviewed. Finally, the microstructural characteristics were reviewed using the scanning electron microscope and X-ray Powder Diffraction. The review outcome reveals that the mechanical properties were increased when 30% silica fume was added to a higher dose of steel fibre to improve the microstructure of GUHPC. It is hypothesized that the brittleness of GUHPC was mitigated by adding 1.5% steel fibre reinforcement, which played a role in the decrease of contact explosion cratering and spalling. Removing the need for cement in GUHPC was a key factor in the review, indicating a promising potential for lowering carbon emissions. However, GUHPC research is still in its early stages, so more study is required before its full potential can be utilized.

The experiments were conducted to investigate the resistance levels among forty‑five lowland rice accessions at the Teaching and Research Farm, Federal College of Agriculture, Ishiagu (Latitude 5°56″N and Longitude 7°41″E) Ebonyi State, Nigeria. Forty‑five rice accessions were screened for their resistance to the blast disease in row planting during the 2017 and 2019 cropping seasons using Randomised Complete Block Design (RCBD) and were replicated thrice. The data on disease intensity and agronomic performance were subjected to Analysis of Variance and the significant means were separated with the Tukey HSD test at p < 0.05. Assessment of blast disease showed that the incidence, severity, and resistance levels varied significantly among rice tested. Accession NG / SA / JAN / 023 had the highest disease incidence (59.06 %) while accessions NG / SA / DEC / 07 / 0300, TOG 5453, TOG 6804, FARO‑22 and FARO‑57 had no blast symptoms for the two cropping seasons. The disease severity ranged from 0.00 to 3.43 for the two cropping seasons. Five accessions (NG / SA / DEC / 07 / 0300, TOG‑5453, TOG‑6804, FARO‑22, and FARO‑57) were highly resistant, 10 were resistant, 14 moderately resistant, and 16 were moderately susceptible to the disease. In the 2017 cropping season, the result of the grain yield showed that accession Local Foreign and FARO 26 had the value of 46.77 g apiece while NG / SA / DEC / 07 / 0278 had the least (11.00 g). However, in the 2019 cropping season, the results of the grain yield showed that accession Local Foreign produced the highest yield (56.00 g), FARO 26 had 40.31 g grain yield while NG / SA / DEC / 07 / 0285 (22.0 g) had the lowest grain yield. The study concluded that using the blast‑resistant cultivars is the best option in managing blast disease, and the five highly resistant accessions could be planted out in other agroecological zones for further studies on managing rice blast disease in the field.

This study examines the impact of lifetime blast exposure on white matter integrity in service members and veterans (SMVs). Participants were 227 SMVs, including those with a history of mild traumatic brain injury (mTBI; n?=?124), orthopedic injury controls (n?=?58), and non-injured controls (n?=?45), prospectively enrolled in a Defense and Veterans Brain Injury Center (DVBIC)/Traumatic Brain Injury Center of Excellence (TBICoE) study. Participants were divided into three groups based on number of self-reported lifetime blast exposures: none (n?=?53); low (i.e., 1?9 blasts; n?=?81); and high (i.e., ?10 blasts; n?=?93). All participants underwent diffusion tensor imaging (DTI) at least 11 months post-injury. Tract-of-interest (TOI) analysis was applied to investigate fractional anisotropy and mean, radial, and axial diffusivity (AD) in left and right total cerebral white matter as well as 24 tracts. Benjamini-Hochberg false discovery rate (FDR) correction was used. Regressions investigating blast exposure and mTBI on white matter integrity, controlling for age, revealed that the presence of mTBI history was associated with lower AD in the bilateral superior longitudinal fasciculus and arcuate fasciculus and left cingulum (?s?=??0.255 to ?0.174; ps?0.01); however, when non-injured controls were removed from the sample (but orthopedic injury controls remained), these relationships were attenuated and did not survive FDR correction. Regression models were rerun with modified post-traumatic stress disorder (PTSD) diagnosis added as a predictor. After FDR correction, PTSD was not significantly associated with white matter integrity in any of the models. Overall, there was no relationship between white matter integrity and self-reported lifetime blast exposure or PTSD.

Pier columns, as primary bridge supports, are vulnerable to extreme loading demands resulting from fire, vehicle collisions, and blasts. The combined effects of these demands may cause significant strength reduction, extensive damage, and potentially collapse. Although the response of bridge columns subjected to independent or combined vehicle impact and air blast has been widely investigated, the cumulative effects of fire in conjunction with these dynamic loads have received limited attention. Consequently, this study presents a unique and advanced multistep finite-element modeling approach using LS-DYNA to simulate the response of isolated round reinforced concrete bridge columns subjected to fire prior to and after a vehicle collision and subsequent air blast. The developed modeling approach incorporates uncoupled implicit heat transfer analyses and explicit structural analyses and was validated against published fire, impact, and blast test results. A parametric study that investigated the effects of various column diameters and fire exposure scenarios on performance was completed, considering fire occurring before or after impact and blast. Column performance was assessed based on damage levels, residual load capacities, and lateral displacements. Study findings enhance the understanding of bridge column performance under this combination of hazards, provide insight into improving bridge resiliency, and could potentially have practical implications for the design and maintenance of bridge columns. Specifically, the study reveals that fire prior to impact and blast was a more critical load sequence for larger column diameters that could potentially remain in service in their final damage states while being repaired. Moreover, the study indicated that for the same fire duration and column diameters, residual capacities and displacements corresponding to half-surface-area exposure were slightly lower than those exposed to fire on the full surface area, a finding that could be beneficial in many practical applications where protective techniques and tools could be implemented. Rice is one of the most important crops in the world and a staple food for more than half of the world's population. At present, the blast disease caused by the fungus Magnaporthe oryzae poses a severe threat to food security through reduction of rice yields worldwide. High phosphate fertilization has previously been shown to increase blast susceptibility. At present, however, our knowledge on the mechanisms underpinning phosphate-induced susceptibility to M. oryzae infection in rice is limited. In this work, we conducted live cell imaging on rice sheaths inoculated with a M. oryzae strain expressing two fluorescently-tagged M. oryzae effectors. We show that growing rice under high phosphate fertilization, and subsequent accumulation of phosphate in leaf sheaths, promotes invasive growth of M. oryzae. Consistent with this, stronger expression of M. oryzae effectors and Pathogenicity Mitogen-activated Protein Kinase (PMK1) occurs in leaf sheaths of rice plants grown under high a phosphate regime. Down-regulation of fungal genes encoding suppressors of plant cell death and up-regulation of plant cell death-inducing effectors also occurs in sheaths of phosphate over-accumulating rice plants. Treatment with high Pi causes alterations in the expression of fungal phosphate transporter genes potentially contributing to pathogen virulence. From the perspective of the plant, Pi accumulation in leaf sheaths prevents H2O2 accumulation early during M. oryzae infection which was associated to a weaker activation of Respiratory Burst Oxidase Homologs (RBOHs) genes involved in reactive oxygen species (ROS) production. Further, a weaker activation of defense-related genes occurs during infection in rice plants over-accumulating phosphate. From these results, it can be concluded that phosphate fertilization has an effect on the two interacting partners, pathogen and host. Phosphate-mediated stimulation of fungal effector genes (e.g., potentiation of fungal pathogenicity) in combination with repression of pathogen-inducible immune responses (e.g., ROS accumulation, defense gene expression) explains higher colonization by M. oryzae in rice tissues accumulating phosphate. Phosphate content can therefore be considered as an important factor in determining the outcome of the rice/M. oryzae interaction. As fertilizers and pesticides are commonly used in rice cultivation to maintain optimal yield and to prevent losses caused by pathogens, a better understanding of how phosphate impacts blast susceptibility is crucial for developing strategies to rationally optimize fertilizer and pesticide use in rice production. Blast-induced neurotrauma has received much attention over the past decade. Vascular injury occurs early following blast exposure. Indeed, in animal models that approximate human mild traumatic brain injury or subclinical blast exposure, vascular pathology can occur in the presence of a normal neuropil, suggesting that the vasculature is particularly vulnerable. Brain endothelial cells and their supporting glial and neuronal elements constitute a neurovascular unit (NVU). Blast injury disrupts gliovascular and neurovascular connections in addition to damaging endothelial cells, basal laminae, smooth muscle cells, and pericytes as well as causing extracellular matrix reorganization. Perivascular pathology becomes associated with phospho-tau accumulation and chronic perivascular inflammation. Disruption of the NVU should impact activity-dependent regulation of cerebral blood flow, blood–brain barrier permeability, and glymphatic flow. Here, we review work in an animal model of low-level blast injury that we have been studying for over a decade. We review work supporting the NVU as a locus of low-level blast injury. We integrate our findings with those from other laboratories studying similar models that collectively suggest that damage to astrocytes and other perivascular cells as well as chronic immune activation play a role in the persistent neurobehavioral changes that follow blast injury. Brain health is largely dependent on the metabolic regulation of amino acids. Brain injuries, diseases, and disorders can be detected through alterations in free amino acid (FAA) concentrations; and thus, mapping the changes has high diagnostic potential. Common methods focus on optimizing neurotransmitter quantification; however, recent focus has expanded to investigate the roles of molecular precursors in brain metabolism. An isocratic method using high performance liquid chromatography with electrochemical cell detection was developed to quantify a wide range of molecular precursors and neurotransmitters: alanine, arginine, aspartate, serine, taurine, threonine, tyrosine, glycine, glutamate, glutamine, and γ-Aminobutyric acid (GABA) following traumatic brain injury. First, baseline concentrations were determined in the serum, cerebrospinal fluid, hippocampus, cortex, and cerebellum of naïve male Sprague Dawley rats. A subsequent study was performed investigating acute changes in FAA concentrations following blast-induced traumatic brain injury (bTBI). Molecular precursor associated FAAs decreased in concentration at 4 h after injury in both the cortex and hippocampus while those serving as neurotransmitters remained unchanged. In particular, the influence of oxidative stress on the observed changes within alanine and arginine pathways following bTBI should be further investigated to elucidate the full therapeutic potential of these molecular precursors at acute time points. • Molecular precursors decrease at 4 h following bTBI, linked to oxidative stress. • Alanine and arginine pathways showed potential for bTBI therapeutic intervention. • An isocratic HPLC-ECD method quantified neurometabolites with high sensitivity. • Eleven amino acid concentrations are validated in rat CSF, serum, and brain tissue.
