Escherichia coli O157:H7 is one of the major foodborne pathogenic bacterial that cause infectious diseases in humans. The previous found that a combination of kojic acid and tea polyphenols exhibited better activity against E. coli O157:H7 than using either alone. This study aimed to explore responses underlying the antibacterial mechanisms of kojic acid and tea polyphenols from the gene level. The functional enrichment analysis by comparing kojic acid and tea polyphenols individually or synergistically against E. coli O157:H7 found that acid resistance systems in kojic acid were activated, and the cell membrane and genomic DNA were destructed in the cells, resulting in “oxygen starvation”. The oxidative stress response triggered by tea polyphenols inhibited both sulfur uptake and the synthesis of ATP, which affected the bacteria's life metabolic process. Interestingly, we found that kojic acid combined with tea polyphenols hindered the uptake of iron that played an essential role in the synthesis of DNA, respiration, tricarboxylic acid cycle. The results suggested that the iron uptake pathways may represent a novel approach for kojic acid and tea polyphenols synergistically against E. coli O157:H7 and provided a theoretical basis for bacterial pathogen control in the food industry.

Abstract Background Immunotherapy brings new hope to patients with advanced gastric cancer. However, liver metastases can reduce the efficacy of immunotherapy in patients. Tumor-associated macrophages (TAMs) may be the cause of this reduction in efficacy. SPP1 + TAMs are considered to have immunosuppressive properties. We aimed to investigate the involvement of SPP1 + TAMs in the metastasis of gastric cancer. Methods The single-cell transcriptome was combined with batched BULK datasets for analysis. Animal models were used to verify the analysis results. Results We reveal the interaction of SPP1 + TAMs with CD8 + exhausted T cells in metastatic cancer. Among these interactions, GDF15-TGFBR2 may play a key immunosuppressive role. We constructed an LR score to quantify interactions based on ligands and receptors. The LR score is highly correlated with various immune features and clinical molecular subtypes. The LR score may also guide the prediction of the efficacy of immunotherapy and prognosis. Conclusions The crosstalk between SPP1 + TAMs and CD8 + exhausted T cells plays a key immunosuppressive role in the gastric metastatic cancer microenvironment. Graphical Abstract

The study aimed to investigate the inhibitory activity and mechanism of action of allyl isothiocyanate (AITC) against Clostridium perfringens (C. perfringens). Firstly, the inhibitory effect of AITC on C. perfringens was evaluated by determining the minimum inhibitory concentration (MIC) and plotting the growth curve. Secondly, the effect of AITC on C. perfringens cell membranes was assessed by scanning electron microscopy to observe cell morphology and by measuring cell membrane integrity with a propidium iodide staining assay. In addition, the impact of AITC on the metabolism of C. perfringens was investigated using SDS-PAGE profile and an ATPase activity assay. Finally, the inhibitory effect of AITC on C. perfringens in cooked ground pork was examined. The experimental results showed that AITC could effectively inhibit the growth of C. perfringens, and the MIC of AITC was determined to be 0.1 μL/mL. AITC was able to induce cell membrane deformations, such as rupture and depression, resulting in the loss of C. perfringens cell membrane integrity, and the degree of membrane damage increased with AITC concentration. Meanwhile, AITC could reduce the protein content and ATPase activity of C. perfringens, which had an impact on normal cellular metabolism. Abstract The RNA 5-methylcytosine (m5C) modification has been demonstrated to be an important epigenetic regulator and to impact colorectal cancer (CRC) progression. However, the potential roles of m5C modification in immune cell infiltration in the CRC tumor microenvironment (TME) remain unknown. The m5C modification phenotypes were comprehensively evaluated based on 14 m5C regulators in a meta-CRC cohort of 1792 patients and systematically correlated with the m5C modification phenotypes, immune cell infiltration characteristics and known biological processes. The m5Cscore model was constructed by principal component analysis (PCA) algorithms to quantify the m5C modification phenotypes of individual CRC samples and was used to predict the immunotherapy response. We identified three m5C modification phenotypes associated with distinct clinical outcomes and biological processes among the 1792 meta-CRC patients. Three phenotypes with a highly consistent TME landscape and characteristics were revealed: immune excluded, immune desert and immune inflammation. The meta-CRC patients were divided into high and low m5Cscore subgroups based on the m5Cscore. The m5Cscore was confirmed to have a negative correlation with infiltrating immune cells and PD-L1 expression and a positive correlation with tumor mutation burden (TMB), mutation rate and microsatellite instability (MSI) score. Moreover, patients in the low m5Cscore group had better immunotherapy responses and significant durable survival benefits in independent anti-PD-1/L1 immunotherapy cohorts for the immune checkpoint inhibitor (ICI) therapeutic strategy. This study revealed that m5C modification plays a crucial role in TME composition and complexity. Comprehensive evaluation of the m5C modification phenotypes of individual patients will enhance our understanding of TME characteristics and promote the application of more appropriate and personalized treatment strategies.

Background The importance of phase Ⅰ cardiac rehabilitation after percutaneous coronary intervention (PCI) has been confirmed, but there is suboptimal adherence among patients. Therefore, investigating the adherence of PCI patients to phaseⅠ cardiac rehabilitation and the influencing factors can provide a theoretical foundation for improving the adherence of patients. Objective To clarifythe influencing factors of the adherence of PCI patients to phaseⅠ cardiac rehabilitation and the effect pathways by the structural equation model construction, in order to provide the oretical support for improving the adherence of PCI patients to phase Ⅰ cardiac rehabilitation. Methods Patients with PCI enrolled in the Cardiac Rehabilitation Center of the Second Hospital of Harbin Medical University from August to December in 2021 were selected as the research objects by convenience sampling. The general demographic information questionnaire, therapy adherence questionnaire, health belief of coronary heart disease questionnaire, Patient Health Questionnaire-9 (PHQ-9), Generalized Anxiexy Disorde-7 (GAD-7), Family Adaptability and Cohesion Evaluation Scale (FACES), Chronic Illness Resource Survey (CIRS), Gensini score were used to investigate the patients by the end of phaseⅠ cardiac rehabilitation. Spearman rank correlation analysis was used to verify the correlations between health belief, depression, anxiety, family adaptability and cohesion, chronic illness resource utilization and cardiac rehabilitation adherence, respectively. Based on correlation analysis, the hypothetical model of the influencing factors of the adherence of PCI patients to phaseⅠ cardiac rehabilitation was constructed combined with Anderson's model of health service utilization. Maximum likelihood method was used to fit and modify the model constantly. Structural equation model was used to analyze the relationship among influencing factors. Results A total of 443 questionnaires were distributed and 430 valid questionnaires were returned, with a valid return rate of 97.06%. The results of multiple linear regression analysis showed that health belief (β=0.427), depression (β=-0.057), anxiety (β=-0.130), family adaptability and cohesion (β=0.242), chronic illness resource (β=0.140) were independent factors of the adherence to phaseⅠcardiac rehabilitation of PCI patients (P0.9. The results of the intermediate effects test showed that health belief, family adaptability and cohesion, chronic illness resource had positive direct effect on the adherence of PCI patients to phaseⅠ cardiac rehabilitation (β=0.395, 0.277, 0.152, P

Climate change has become one of the biggest challenges facing the world, which will affect the process of sustainable development in all aspects such as economy, society, environment, etc., and the integration of climate change into the sustainable development management strategy will help to realize the process of SDGs indicators. However, there is a lack of quantitative research on the correlation between climate variables and sustainable development, especially from the perspective of climate change. Therefore, the experiment systematically proposes a framework for ecosystem service-based sustainable development assessment in the context of climate change, and tries to explore the response relationship between climate variables and sustainable development goals. The experiment takes the central Yunnan urban agglomeration as the study area, and based on the climate data of CMIP6, adopts the PLUS model and the InVEST model, predicts the spatial distribution of ESs and calculates the indicators of future SDGs, and explores the response of the climate to the indicators of SDGs by using the Spearman's correlation analysis. The experimental results showed that both the overall ESs and SDGs metrics declined under the influence of future climate change. Through the response analysis, among the eight climate indicators, evapotranspiration, dryness, barometric pressure, and temperature showed negative impacts in the study go, while precipitation, relative humidity, wind speed, and solar radiation promoted sustainable progress in the study area. However, under the influence of future climate change, the positive indicators may translate into negative impacts. There is a strong correlation between climate change and sustainable development, and the results of this experiment can provide a scientific reference for the sustainable development of similar regions in order to minimize the negative impacts of climate change in the future.

Gastric cancer is one of the most common malignant tumors in the world. Alpha fetoprotein (AFP)-positive gastric cancer (AFPP-GC) is considered a special entity among gastric cancers. There is still controversy regarding the clinicopathological characteristics and prognosis of AFPP-GC, and the potential mechanism underlying its high malignant potential is still unclear. A comprehensive description of AFPP-GC genomic characteristics and regulatory mechanisms is lacking. This study analyzed the pathological characteristics and prognosis of AFPP-GC by utilizing clinical samples. The results showed that AFPP-GC has a poor prognosis and a high of risk liver metastasis. Tissue transcriptome sequencing showed that genes with high expression in AFPP-GC were involved in the activation of various cancer pathways, and genes with low expression were involved in the immune response. Single-sample gene set enrichment analysis showed that overexpression of AFP in AFPP-GC significantly inhibited the infiltration of CD8+ T cells. To further explore the genomic characteristics of AFPP-GC, the signaling pathway by which AFP regulates the invasion and metastasis of AFPP-GC cells was discussed. The results showed that AFPP-GC may promote cell invasion by regulating the PTEN/AKT1/SOX5/CES1 signaling axis. This study reveals the molecular mechanism underlying the increased malignant potential of AFPP-GC vs. AFP-negative gastric cancer (AFPN-GC). This provides important information for individualized treatment of AFPP-GC.

Summary: The acidic microenvironment is considered an important factor in colorectal cancer (CRC) that contributes to malignant transformation. However, the underlying mechanism remains unclear. In a previous study, we confirmed that IDH1 K224 deacetylation promotes enzymatic activity and the production of α-KG. Here, we further investigate the effect of IDH1 hyperacetylation on the CRC acidic microenvironment. We demonstrate that increased α-KG affects hydroxylation of Ago2 and mediates miR-9-5p targeting NHE1 protein. Knockdown of NHE1 dramatically attenuates CRC cell proliferation and migration by restricting transport of intracellular H+ out of cells. Furthermore, we show that miR-9-5p is the microRNA with the most significant difference in the alteration of IDH1 K224 acetylation and can downregulate NHE1 mRNA. Our data also indicate that hydroxylation stabilizes Ago2, which in turn promotes miR-9-5p activity. Taken together, our results reveal a novel mechanism through which IDH1 deacetylation regulates the cellular acidic microenvironment and inhibits CRC metastasis.

Vegetation indexes have been widely used to qualitatively and quantitatively evaluate vegetation cover and its growth vigor. To further extend the study of vegetation indexes, this paper proposes to study the spatial and temporal distribution characteristics and specific driving mechanisms of vegetation indexes based on the example of Yunnan Province, China, and also adds the study of spatial and temporal prediction methods of vegetation indexes. This paper used data on this region’s normalized vegetation index (NDVI), three meteorological factors, and eight social factors from 1998 to 2019. The dynamic change in and driving mechanism of the NDVI were studied using mean value analysis, univariate linear trend regression analysis, and partial correlation analysis. In addition, the Fourier function model and the CA–Markov model were also used to predict the NDVI of Yunnan Province from 2020 to 2030 in time and space. The results show that: (1) The NDVI value in Yunnan Province is high, showing a significant growth trend. The increased vegetation coverage area has increased in the past 22 years without substantial vegetation degradation. (2) The positive promotion of meteorological factors is greater than the negative inhibition. The partial correlation of relative humidity among meteorological factors is the highest, which is the main driving factor. (3) The NDVI value is significantly positively correlated with population and economy and negatively correlated with pasture land and agricultural area. (4) The NDVI values are predicted well in time (R = 0.64) and space (Kappa = 0.8086 and 0.806), satisfying the accuracy requirements. This paper aims to enrich the theoretical and technical system of ecological environment research by studying the dynamic change, driving mechanism, and spatiotemporal prediction of the normalized vegetation index. Its results can provide the necessary theoretical basis for the simulation and prediction of vegetation indexes.

Abstract Background Circular RNAs (circRNAs) have emerged as vital regulators of the initiation and progression of diverse kinds of human cancers. In this study, we explored the role of hsa_circ_0000231 and its downstream pathway in CRC. Methods The expression profile of circRNAs in 5 pairs of CRC tissues and adjacent normal tissues were analyzed by Microarray. Quantitative real-time PCR and in situ hybridization and Base Scope Assay were used to determine the level and prognostic values of hsa_circ_0000231. Then, functional experiments in vitro and in vivo were performed to investigate the effects of hsa_circ_0000231 on cell proliferation. Mechanistically, fluorescent in situ hybridization, dual luciferase reporter assay, RNA pull-down and RNA immunoprecipitation experiments were performed to confirm the interaction between hsa_circ_0000231 and IGF2BP3 or has_miR-375. Results We acquired data through circRNA microarray profiles, showing that the expression of hsa_circ_0000231 was upregulated in CRC primary tissues compared to adjacent normal tissues, which was indicated poor prognosis of patients with CRC. Functional analysis indicated that inhibition of hsa_circ_0000231 in CRC cell lines could suppress CRC cell proliferation as well as tumorigenesis in vitro and in vivo. The mechanistic analysis showed that hsa_circ_0000231 might, on the one hand, act as a competing endogenous RNA of miR-375 to promote cyclin D2 (CCND2) and, on the other hand, bind to the IGF2BP3 protein to prevent CCND2 degradation. Conclusions The findings suggested that hsa_circ_0000231 facilitated CRC progression by sponging miR-375 or binding to IGF2BP3 to modulate CCND2, implying that hsa_circ_0000231 might be a potential new diagnostic and therapeutic biomarker of CRC.

Abstract Background Growing evidence has shown that the prognosis for colon cancer depends on changes in microenvironment. The purpose of this study was to elucidate the prognostic value of long noncoding RNAs (lncRNAs) related to immune microenvironment (IM) in colon cancer. Methods Single sample gene set enrichment analysis (ssGSEA) was used to identify the subtypes of colon cancer based on the immune genomes of 29 immune signatures. Cox regression analysis identified a lncRNA signatures associated with immune infiltration. The Tumor Immune Estimation Resource database was used to analyze immune cell content. Results Colon cancer samples were divided into three subtypes by unsupervised cluster analysis. Cox regression analysis identified an immune infiltration-related 5-lncRNA signature. This signature combined with clinical factors can effectively improve the predictive ability for the overall survival (OS) of colon cancer. At the same time, we found that the expression of H19 affects the content of B cells and macrophages in the microenvironment of colon cancer and affects the prognosis of colon cancer. Finally, we constructed the H19 regulatory network and further analyzed the possible mechanisms. We found that knocking down the expression of H19 can significantly inhibit the expression of CCND1 and VEGFA. At the same time, the immunohistochemical assay found that the expression of CCND1 and VEGFA protein was significantly positively correlated with the infiltration of M2 type macrophages. Conclusion The findings may help to formulate clinical strategies and understand the underlying mechanisms of H19 regulation. H19 may be a biomarker for targeted treatment of colon cancer.

In a chalcopyrite and pyrite flotation system, the effect and mechanism of organic depressant sodium dimethyl dithiocarbamate on the hydrophobicity of chalcopyrite and pyrite were studied. The micro-flotation test results revealed that the pyrite recovery efficiency decreased from 80.31% to 23.68% under weakly alkaline environment (pH 8.5) after adding SDD. The zeta potential results indicate that the potential on the pyrite surface decreased after the SDD was added. The coordination group –CSS– in SDD can trap cations and generate hydrophilic dithiocarbamate on the pyrite surface. The results of the scanning electron microscopy and energy dispersive spectrometry show that after the addition of SDD, complexes were formed on the surfaces of the pyrite, while the surface morphology of the chalcopyrite hardly changed. The X-ray photoelectron spectroscopy results showed that the SDD was adsorbed on the pyrite surface and reduced the hydrophobicity of the pyrite. The Time-of-flight secondary ion mass spectrometry analysis results revealed that the normalized peak value (0.001) of CSN− on the chalcopyrite surface was considerably lower than that on the pyrite surface (0.0113) after the addition of SDD. All these results have established that SDD influenced the hydrophobicity of pyrite and had little influence on chalcopyrite hydrophobicity.

Abstract Increasing evidence has clarified that the tumor microenvironment (TME) is closely related to the prognosis and therapeutic efficacy of cancer. However, there is no reliable TME evaluation system used to accurately predict the prognosis of and therapeutic efficacy in gastric cancer. We evaluated the immune microenvironment score (IMS) of 1422 gastric cancer samples based on 51 immune cell signatures. We explored the relationship between the IMS and prognosis, immune cell infiltration, cancer subtype, and potential immune escape mechanisms. The results show that activation of the stroma and decreased levels of immune infiltration were associated with a low IMS. A high IMS was characterized by Epstein–Barr virus infection, increased mutation load, microsatellite instability, and immune cell infiltration. A high IMS was also related to high expression of immune checkpoint molecules (PD-1/PD-L1). Finally, patients with a high IMS had a better response to PD-1/PD-L1 inhibitors and may be more suitable for immune checkpoint inhibitors (area under the curve = 0.81). In addition, a low IMS may be converted into the immune-infiltrating subtype after romidepsin treatment. Stratification based on the IMS may enable gastric cancer patients to benefit more from immunotherapy and help identify new cancer treatment strategies.

Faced with the high heterogeneity and poor prognosis of colorectal cancer (CRC), this study sought to find new predictive prognostic strategies to improve the situation. Cuproptosis is a novel cell death mechanism that relies on copper regulation. However, the role of cuproptosis-related gene (CRG) in CRC remains to be elucidated. In this study, we comprehensively assessed the CRG landscape in CRC based on The Cancer Genome Atlas (TCGA). We identified differential expression and genetic alterations of CRG in CRC. CRG is highly correlated with initiation, progression, prognosis, and immune infiltration of CRC. We construct a risk score signature containing 3 CRGs based on LASSO. We explored the correlation of CRG-Score with clinicopathological features of CRC. Age, stage, and CRG-Score were integrated to construct a nomogram. The nomogram has robust predictive performance. We also understand the correlation of CRG-Score with CRC immune landscape. CRG-Score can effectively predict the immune landscape of CRC patients. Low-risk CRC patients have greater immunogenicity and higher immune checkpoint expression. Low-risk CRC patients may be better candidates for immunotherapy. At the same time, we also predicted more sensitive drugs in the high-risk CRC patients. In conclusion, the CRG risk score signature is a strong prognostic marker and may help provide new insights into the treatment of individuals with CRC.

Abstract Organic cathode materials have potential applications in rechargeable batteries due to their several advantages such as high specific capacity, flexible designability, plentiful raw materials, environmental friendliness, and renewability. However, their high solubility in organic electrolytes strongly impedes the further research progress. Thus, it is highly desirable to develop some new strategies to address this issue. Herein, we report one method to address this dissolution issue by increasing molecular weight without reducing theoretical capacity, where a novel Calix[8]quinone (C8Q) with 8 p‐benzoquinone units connected by methylene groups was designed and prepared in a good yield (total: 23%). C8Q exhibits higher cycle stability (268 mAh g−1) in lithium‐ion batteries (LIBs) compared to the same series of substances Calix[4]quinone (C4Q, 30 mAh g−1) and Calix[6]quinone (C6Q, 207 mAh g−1) after 100 cycles at 0.2 C. Moreover, C8Q shows better electrochemical performance in 4.2 M LiTFSI‐AN highly‐concentrated electrolyte with special aggregate structure configured with high‐dissociation salt LiTFSI and low‐viscosity solvent acetonitrile (AN), namely, C8Q possesses high capacity (340 mAh g−1 after 100 cycles at 0.2 C), superior rate ability (440 mAh g−1 at 0.1 C and 167 mAh g−1 at 5 C), and ultra‐long cycle life (220 mAh g−1 after 1000 cycles at 0.5 C). This work could provide a promising strategy to address the dissolution issue of organic electrode materials in organic electrolyte.

Emerging studies have focused on ways to treat cancers by modulating T cell activation. However, whether B cell receptor signaling in the tumor microenvironment (TME) can be harnessed for immunotherapy is unclear. Here, we report that an Asia-specific variant of human IgG1 containing a Gly396 to Arg396 substitution (hIgG1-G396R) conferred improved survival of patients with colorectal cancer (CRC). Mice with knockin of the murine functional homolog mIgG2c-G400R recapitulated the alleviated tumorigenesis and progression in murine colon carcinoma models. Immune profiling of the TME revealed broad mobilizations of IgG1+ plasma cells, CD8+ T cells, CD103+ DCs, and active tertiary lymphoid structure formation, suggesting an effective antitumor microenvironment in hIgG1-G396R CRC patients. Mechanistically, this variant potentiated tumor-associated antigen–specific (TAA-specific) plasma cell differentiation and thus antibody production. These elevated TAA-specific IgG2c antibodies in turn efficiently boosted the antibody-dependent tumor cell phagocytosis and TAA presentation to effector CD8+ T cells. Notably, adoptive transfer of TAA-specific class-switched memory B cells harboring this variant exhibited therapeutic efficacy in murine tumor models, indicating their clinical potential. All these results prompted a prospective investigation of hIgG1-G396R in patients with CRC as a biomarker for clinical prognosis and demonstrated that manipulating the functionality of IgG1+ memory B cells in tumors could improve immunotherapy outcomes.

Terrestrial vegetation, a critical component of the Earth’s land surface, directly impacts the planet’s material and energy balance. This study investigated the dynamics of terrestrial vegetation in China from 2000 to 2019 using three remote sensing products (NDVI, EVI, and SIF) and explored the driving mechanisms behind these changes. We considered three meteorological factors, nine land use types, and two socio-economic factors while employing mathematical models to analyze the data. Additionally, we used the CA–Markov model to predict the spatial distribution of vegetation remote sensing products for 2020–2025. Our findings indicate the following: (1) Throughout the study period, the vegetation indices, NDVI, EVI, and SIF, all exhibited increasing trends. The SIF showed a more direct response to vegetation cover changes and was less influenced by other driving factors. The SIF outperforms the NDVI and EVI in detecting vegetation trend changes, particularly regarding sensitivity. (2) Vegetation cover changes are driven by multiple meteorological factors, such as temperature, precipitation, and relative humidity. These factors exhibit a strong spatial correlation with the distribution of vegetation remote sensing products. Among these factors, the SIF shows a higher sensitivity to temperature compared to the NDVI and EVI, while the NDVI and EVI display greater sensitivity to precipitation and relative humidity. (3) Within the study area, land use types reveal a gradient from northwest to southeast, which is consistent with the spatial distribution of the vegetation remote sensing products. For green vegetation types, the three remote sensing products exhibit varying sensitivity levels, with the SIF demonstrating the highest sensitivity to green vegetation types. (4) Overall, the future vegetation outlook in China is promising, especially in the southeastern regions where significant vegetation improvement trends are evident. However, the vegetation conditions in some northwestern areas remain less favorable, necessitating the reinforcement of ecological construction and improvement measures. Additionally, a significant positive correlation exists between population size, GDP, and vegetation remote sensing products. This study highlights the variability in the dynamics and driving mechanisms of terrestrial vegetation remote sensing products in China and employs the CA–Markov model for predicting future vegetation patterns. Our research contributes to the theoretical and technical understanding of remote sensing for terrestrial vegetation in the Chinese context.

A landslide is one of the most destructive natural disasters in the world. The accurate modeling and prediction of landslide hazards have been used as some of the vital tools for landslide disaster prevention and control. The purpose of this study was to explore the application of coupling models in landslide susceptibility evaluation. This paper used Weixin County as the research object. First, according to the landslide catalog database constructed, there were 345 landslides in the study area. Twelve environmental factors were selected, including terrain (elevation, slope, slope direction, plane curvature, and profile curvature), geological structure (stratigraphic lithology and distance from fault zone), meteorological hydrology (average annual rainfall and distance to rivers), and land cover (NDVI, land use, and distance to roads). Then, a single model (logistic regression, support vector machine, and random forest) and a coupled model (IV–LR, IV–SVM, IV–RF, FR–LR, FR–SVM, and FR–RF) based on information volume and frequency ratio were constructed, and the accuracy and reliability of the models were compared and analyzed. Finally, the influence of environmental factors on landslide susceptibility under the optimal model was discussed. The results showed that the prediction accuracy of the nine models ranged from 75.2% (LR model) to 94.9% (FR–RF model), and the coupling accuracy was generally higher than that of the single model. Therefore, the coupling model could improve the prediction accuracy of the model to a certain extent. The FR–RF coupling model had the highest accuracy. Under the optimal model FR–RF, distance from the road, NDVI, and land use were the three most important environmental factors, ac-counting for 20.15%, 13.37%, and 9.69%, respectively. Therefore, it was necessary for Weixin County to strengthen the monitoring of mountains near roads and areas with sparse vegetation to prevent landslides caused by human activities and rainfall.

Background: Abnormal expression of lncRNA is closely related to the occurrence and metastasis of osteosarcoma. The tumor immune microenvironment (TIM) is considered to be an important factor affecting the prognosis and treatment of osteosarcoma. This study aims to explore the effect of immune-related lncRNAs (IRLs) on the prognosis of osteosarcoma and its relationship with the TIM.Methods: Ninety-five osteosarcoma samples from the TARGET database were included. Iterative LASSO regression and multivariate Cox regression analysis were used to screen the IRLs signature with the optimal AUC. The predict function was used to calculate the risk score and divide osteosarcoma into a high-risk group and low-risk group based on the optimal cut-off value of the risk score. The lncRNAs in IRLs signature that affect metastasis were screened for in vitro validation. Single sample gene set enrichment analysis (ssGSEA) and ESTIMATE algorithms were used to evaluate the role of TIM in the influence of IRLs on osteosarcoma prognosis.Results: Ten IRLs constituted the IRLs signature, with an AUC of 0.96. The recurrence and metastasis rates of osteosarcoma in the high-risk group were higher than those in the low-risk group. In vitro experiments showed that knockdown of lncRNA (AC006033.2) could increase the proliferation, migration, and invasion of osteosarcoma. ssGSEA and ESTIMATE results showed that the immune cell content and immune score in the low-risk group were generally higher than those in the high-risk group. In addition, the expression levels of immune escape-related genes were higher in the high-risk group.Conclusion: The IRLs signature is a reliable biomarker for the prognosis of osteosarcoma, and they alter the prognosis of osteosarcoma. In addition, IRLs signature and patient prognosis may be related to TIM in osteosarcoma. The higher the content of immune cells in the TIM of osteosarcoma, the lower the risk score of patients and the better the prognosis. The higher the expression of immune escape-related genes, the lower the risk score of patients and the better the prognosis.

Polyphenols are found in free and bound forms in foods, both of which exert a wide range of pharmacological and therapeutic effects. Here, 3T3-L1 pre-adipocytes and high-fat diet (HFD)-induced mice were used to investigate the potential anti-obesity effect of bound phenolics extracted from lotus seeds (LBP). Phenolic compounds including protocatechuic, caffeic and ellagic acids were identified in LBP. In vitro results showed LBP significantly decreased intracellular lipid accumulation; while oral administration of LBP was found to retard body weight gain and improve plasma lipid profile in HFD-induced mice. Moreover, LBP increased phosphorylation of adenosine monophosphate-activated protein kinase (AMPK) accompanied with down-regulation of adipogenic/lipogenic genes (Pparγ, C/ebpα, aP2, Srebp-1c, Acc1, Fas, Lpl) expression; and up-regulation of lipolytic genes (Hsl, Pgc-1α, Sirt1, Cpt1α) and adipokine adiponectin expression. Taken together, these observations suggested lotus seeds bound phenolics may alleviate obesity, in part by modulating the AMPK signaling pathway.

Growing evidence suggests that immune-related genes (IRGs) and long non-coding RNAs (lncRNAs) can serve as prognostic markers of overall survival (OS) in patients with colon cancer. This study aimed to identify an immune-related lncRNA signature for the prospective assessment of prognosis in these patients. Gene expression and clinical data of colon cancer patients were downloaded from The Cancer Genome Atlas (TCGA). Immune-related lncRNAs were identified by a correlation analysis between IRGs and lncRNAs. In total, 447 samples were divided into a training cohort (224 samples) and a testing cohort (223 samples). Univariate, lasso and multivariate Cox regression analyses identified an immune-related nine-lncRNA signature closely related to OS in colon cancer patients in the training dataset. A risk score formula involving nine immune-related lncRNAs was developed to evaluate the prognostic value of the lncRNA signature in the training dataset. Colon cancer patients with a high risk score had poorer OS than those with a low risk score. A multivariate Cox regression analysis confirmed that the immune-related nine-lncRNA signature could be an independent prognostic factor in colon cancer patients. The results were further confirmed in the testing cohort and the entire TCGA cohort. Furthermore, a gene set enrichment analysis revealed several pathways with significant enrichment in the high- and low-risk groups that may be helpful in formulating clinical strategies and understanding the underlying mechanisms. Finally, a quantitative real-time polymerase chain reaction assay found that the nine lncRNAs were significantly differentially expressed in colon cancer cell lines. The results of this study indicate that this signature has important clinical implications for improving predictive outcomes and guiding individualized treatment in colon cancer patients. These lncRNAs could be potential biomarkers affecting the prognosis of colon cancer.

Phyllanthus emblica L. fruits were extracted by a hot water assistant with ultrasonication to obtain aqueous Phyllanthus emblica L. extract (APE). The ameliorating functional dyspepsia (FD) effect of a low dose (150 mg/kg) and a high dose (300 mg/kg) of APE was exhibited by determining the gastrointestinal motility, gastrointestinal hormones, and gut microbiome shifts in reserpine induced FD male balb/c mice. APE increased the gastrointestinal motility including the gastric emptying (GE) rate and small intestinal transit (SIT) rate. The level of serum gastrointestinal hormones such as motilin (MTL) and gastrin (GAS) increased, and the vasoactive intestinal peptide (VIP) level decreased after the administration of APE. Furthermore, the gut microbiome analysis demonstrated that APE could regulate the microbiome structure and restore homeostasis by elevating useful bacterial abundance, while simultaneously decreasing harmful bacterial abundance. This study demonstrated the ameliorating FD effect of APE and its potential efficacy in curing functional gastrointestinal disorders and maintaining a healthy digestive tract.

BackgroundThe UHRF1 gene is an epigenetic modification factor that mediates tumor suppressor gene silencing in a variety of cancers. Related studies have reported that UHRF1 can inhibit the expression of the KISS1 gene. However, the regulatory mechanism underlying UHRF1 expression in colorectal cancer (CRC) is still unclear. The aim of this study was to gain a better understanding of the regulation of UHRF1 expression in CRC and to determine whether it regulates the mechanism by which KISS1 promotes CRC metastasis.MethodsIn the present study, the levels of miR-506, UHRF1 and KISS1 expression in CRC tissues and in human CRC cell lines were studied using quantitative real-time PCR (qRT-PCR) and Western blotting. Cell proliferation, migration, and invasion assays are used to detect cell proliferation, migration, and invasion. A dual-luciferase reporter system was used to confirm the target gene of miR-506.ResultsThis study found that UHRF1 protein is highly expressed in CRC tissues and negatively correlated with KISS1 protein expression. UHRF1 overexpression activates the PI3K/NF-κB signaling pathway by inhibiting the mRNA expression levels of pathway mediators. Bioinformatics analysis and luciferase reporter gene assays confirmed that miR-506 targets UHRF1.ConclusionThis study identified the regulation of UHRF1 expression in CRC and the mechanism of CRC metastasis. UHRF1 may be a new potential target molecule for future CRC metastasis treatment.

Fresh-cut fruits and vegetables are the main sources of foodborne illness outbreaks with implicated pathogens such as Escherichia coli O157:H7, Salmonella, and Listeria monocytogenes. This study aimed at investigating the influence of two key parameters (concentration of curcumin and illumination time) on the effects of curcumin-based photodynamic sterilization on the preservation of fresh-cut Hami melons. The results indicated that illumination with 50 μmol/L curcumin for 60 min using a blue LED lamp reduced the total aerobic microorganism count by ~1.8 log CFU/g in fresh-cut Hami melons. Besides this, the effects of photodynamic sterilization on the soluble solids content, color, water content, firmness, and sensory indices of the fresh-cut Hami melons were also evaluated. Compared to the control group, photodynamic sterilization can effectively delay the browning rate and maintain the luminosity, firmness, water content, and soluble solids content of fresh-cut Hami melon. The sensory quality was indeed preserved well after 9 days of storage in a fridge. These results showed that photodynamic sterilization is an effective and promising technology to prolong the shelf life of fresh-cut Hami melons.
