Objective: To examine the potential effect of the probiotic strain Lactobacillus plantarum LP45 on osteoporosis and to explore the involved molecular mechanisms. Methods: A rat model of glucocorticoid-induced osteoporosis (GIO) was established, which was also orally administered with increasing doses of LP45 for 8 weeks. After the termination of the 8-week treatment, the tibia and femur bones of rats were analyzed for bone histomorphometry, bone mineral content (BMC), and bone mineral density (BMD). Femoral biomechanics were assessed. In addition, levels of osteocalcin, tartrate-resistant acid phosphatase 5 (TRAP5), osteoprotegerin (OPG), and receptor activator of nuclear factor kappa-B ligand (RANKL) in the serum and bone marrow were also measured using ELISA, Western blot, and real time-polymerase chain reaction. Results: GIO caused obvious defects in tibia and femur bone structures, in terms of tissue/bone volume, trabecular separation, trabecular thickness, and trabecular number, which could be rescued by LP45 dose dependently. The GIO-induced reductions in BMC, BMD, osteoblast surfaces per bone surface (BS), as well as elevated osteoclast surface per BS were largely restored by LP45 administration dose-dependently. LP45 also increased femoral biomechanics of GIO rats. Importantly, LP45 dose-dependently restored the changes of osteocalcin, TRAP5, OPG, and RANKL in the serum as well as bone marrow of GIO rats. Conclusion: Oral LP45 administration could significantly prevent bone defects in GIO rats, suggesting its potential as a dietary supplement with beneficial effects against osteoporosis, which might involve the RANKL/OPG signaling pathway.

Breast ultrasound (BUS) imaging is commonly used in the early detection of breast cancer as a portable, valuable, and widely available diagnosis tool. Automated BUS image classification and segmentation can assist radiologists in making accurate and fast decisions. Recent studies illustrate that tumor, peritumoral, and background regions of BUS images provide valuable information for BUS image segmentation or classification. However, few studies have investigated the influence of these three regions on multi-task learning. In this study, we propose an RMTL-Net to simultaneously segment tumor regions and classify tumors in BUS images into benign or malignant categories. To improve both segmentation and classification performance, we design a regional attention (RA) module that employs the predicted probability maps to automatically guide the classifier to learn important category-sensitive information in the tumor, peritumoral, and background regions and seamlessly fuse them to obtain a better feature representation. We conduct detailed ablation experiments of the proposed RA module and comparative experiments with four recent state-of-the-art peer multi-task learning methods, three single-task segmentation methods, and four single-task classification methods on two public BUS datasets. Experimental results show that the proposed RMTL-Net achieves the best overall segmentation and classification accuracy in terms of five segmentation metrics and six classification metrics.

Objective: This study was designed to compare the effects of hamstring muscle injection of botulinum toxin type A (BoNT/A) combined with periarticular injection to those of simple periarticular injection alone, in terms of early pain reduction and functional recovery following total knee arthroplasty (TKA). Methods: This study included 40 patients who underwent elective unilateral TKA at our hospital from February 2021 to December 2021. The patients were randomly assigned to either the experimental group (BoNT/A injection into the hamstring muscle + periarticular injection, n = 20 cases) or the control group (0.9% normal saline injection into the hamstring muscle + periarticular injection, n = 20 cases). The preoperative and postoperative 3-day HSS scores were compared between the two groups. Postoperative visual analog scale (VAS) pain scores were observed and compared between the two groups. The Lovett grading method was used to evaluate the postoperative muscle strength of both groups. Additionally, postoperative knee joint pain scores, maximum active flexion angle, and any postoperative adverse reactions were observed and compared between the two groups. Results: The experimental group showed significantly lower VAS pain scores than the control group at 24 h, 48 h, and 72 h after the operation, with a statistically significant difference (P < 0.05). Furthermore, compared to the control group, the patients in the experimental group exhibited larger knee joint active flexion angles at 1 day, 3 days, and 5 days post-operation, also demonstrating a statistically significant difference (P < 0.05). These findings hold scientific significance (P < 0.05). Additionally, the combined administration of BoNT/A hamstring muscle injection with periarticular injection exhibited a low incidence of adverse reactions during the perioperative period and had no significant effect on quadriceps muscle strength. Conclusion: The combination of BoNT/A hamstring muscle injection with periarticular injection effectively alleviates early postoperative pain following TKA and promotes early functional recovery of the knee joint.

Tuberoinfundibular dopamine (TIDA) neurons have cell bodies located in the arcuate nucleus of the mediobasal hypothalamus. They project to the external zone of the median eminence, and the dopamine (DA) released there is carried by the hypophysial portal vasculature to the anterior pituitary. The DA then activates D2 receptors to inhibit prolactin (PRL) secretion from lactotrophs. The TIDA neuronal population is the principal regulatory factor controlling PRL secretion. The neuroendocrine role subserved by TIDA neurons sets them apart from other dopaminergic populations like the nigrostriatal and mesolimbic DA neurons. TIDA neurons exhibit intrinsic oscillatory fluctuations in their membrane potential that give rise to phasic firing and bursting activity. TIDA neuronal activity is sexually differentiated and modulated by gonadal hormones and PRL, as well as an array of small molecule and peptide neurotransmitters. This review covers these characteristics.

This study aimed to report on a method for injecting botulinum toxin type A into the hamstring muscles combined with periarticular injection in total knee arthroplasty (TKA) patients. We enrolled patients who underwent elective unilateral TKA at our hospital from February 2021 to December 2021 and administered botulinum toxin type A hamstring muscle injection combined with periarticular injection. We established and reported a detailed method for this combined approach, which could provide an alternative analgesic regimen after TKA in clinical practice.

Abstract One of the primary challenges of visual tracking is the variable appearance of the target object. As tracking proceeds, the target object can change its appearance due to illumination changes, rotations, deformations etc. Modern trackers incorporate online updating to learn how the target changes over time. However, they do not use the history of target appearance. To address this shortcoming, we uniquely use domain adaptation with the target appearance history to efficiently learn a temporally matching filter (TMF) during online updating. This TMF emphasizes the persistent features found in different appearances of the target object. It also improves the classification accuracy of the convolutional neural network by assisting the training of the classification layers without incurring the runtime overhead of updating the convolutional layers. Extensive experimental results demonstrate that the proposed TMF‐based tracker, which incorporates domain adaptation with the target appearance history, improves tracking performance on three benchmark video databases (OTB‐50, OTB‐100 and VOT2016) over other online learning trackers. Specifically, it improves the overlap success of VITAL and MDNet by 0.44 % and 1.03 % on the OTB‐100 dataset and improves the accuracy of VITAL and MDNet by 0.55 % and 0.06 % on the VOT2016 dataset, respectively.

Traffic sign detection is considered as one of the active research topics in transportation and computer vision. The previous works mainly focus on detecting traffic signs in images or in mobile light detection and ranging (LiDAR) data. In this paper, we propose a novel deep learning method to accurately detect traffic signs by fusing the complementary features from registered airborne geo-referenced color images and noisy airborne LiDAR data. Specifically, we first segment the airborne color images to road and non-road segments by integrating various local features in an inequality constraint quadratic optimization model. Next, we find the corresponding road regions in LiDAR data and extract high elevated objects above the road. We then segment the extracted objects to different regions corresponding to traffic sign candidates using Euclidean distance-based clustering. Finally, we find the corresponding traffic sign candidates in color images, extract their deep features, and represent them in a convex optimization model to classify the candidates. A set of extensive experiments have been carried out on the airborne geo-referenced color images and noisy airborne LiDAR data captured by Utah State University from I-15 highway. The results show the effectiveness of the proposed method in detecting traffic signs.

This study proposes an illumination‐invariant face‐recognition method called adaptive homomorphic eight local directional pattern (AH‐ELDP). AH‐ELDP first uses adaptive homomorphic filtering to reduce the influence of illumination from an input face image. It then applies an interpolative enhancement function to stretch the filtered image. Finally, it produces eight directional edge images using Kirsch compass masks and uses all the directional information to create an illumination‐insensitive representation. The author's extensive experiments show that the AH‐ELDP technique achieves the best face recognition accuracy of 99.45% for CMU‐PIE face images, 96.67% for Yale B face images and 84.42% for Extended Yale B face images using one image per subject for training when compared to seven representative state‐of‐the‐art techniques.

Abstract All-perovskite tandem solar cells have shown great promise in breaking the Shockley–Queisser limit of single-junction solar cells. However, the efficiency improvement of all-perovskite tandem solar cells is largely hindered by the surface defects induced non-radiative recombination loss in Sn–Pb mixed narrow bandgap perovskite films. Here, we report a surface reconstruction strategy utilizing a surface polishing agent, 1,4-butanediamine, together with a surface passivator, ethylenediammonium diiodide, to eliminate Sn-related defects and passivate organic cation and halide vacancy defects on the surface of Sn–Pb mixed perovskite films. Our strategy not only delivers high-quality Sn–Pb mixed perovskite films with a close-to-ideal stoichiometric ratio surface but also minimizes the non-radiative energy loss at the perovskite/electron transport layer interface. As a result, our Sn–Pb mixed perovskite solar cells with bandgaps of 1.32 and 1.25 eV realize power conversion efficiencies of 22.65% and 23.32%, respectively. Additionally, we further obtain a certified power conversion efficiency of 28.49% of two-junction all-perovskite tandem solar cells.

Abstract Breast cancer (BRCA) has become the most common type of cancer in women. Improving the therapeutic response remains a challenge. Phosphatase and tensin homologue deleted on chromosome 10 (PTEN) is a classic tumour suppressor with emerging new functions discovered in recent years, and myeloid PTEN loss has been reported to impair antitumour immunity. In this study, we revealed a novel mechanism by which myeloid PTEN potentially affects antitumour immunity in BRCA. We detected accelerated stress granule (SG) assembly under oxidative stress in PTEN-deficient bone marrow-derived macrophages (BMDMs) through the EGR1-promoted upregulation of TIAL1 transcription. PI3K/AKT/mTOR (PAM) pathway activation also promoted SG formation. ATP consumption during SG assembly in BMDMs impaired the phagocytic ability of 4T1 cells, potentially contributing to the disruption of antitumour immunity. In a BRCA neoadjuvant cohort, we observed a poorer response in myeloid PTENlow patients with G3BP1 aggregating as SGs in CD68+ cells, a finding that was consistent with the observation in our study that PTEN-deficient macrophages tended to more readily assemble SGs with impaired phagocytosis. Our results revealed the unconventional impact of SGs on BMDMs and might provide new perspectives on drug resistance and therapeutic strategies for the treatment of BRCA patients.

Abstract Background Protein phosphatase class 2 C (PP2C) is the largest protein phosphatase family in plants. Members of the PP2C gene family are involved in a variety of physiological pathways in plants, including the abscisic acid signalling pathway, the regulation of plant growth and development, etc., and are capable of responding to a wide range of biotic and abiotic stresses, and play an important role in plant growth, development, and response to stress. Apocynum is a perennial persistent herb, divided into Apocynum venetum and Apocynum hendersonii. It mainly grows in saline soil, deserts and other harsh environments, and is widely used in saline soil improvement, ecological restoration, textiles and medicine. A. hendersonii was found to be more tolerant to adverse conditions. The main purpose of this study was to investigate the PP2C gene family and its expression pattern under salt stress and to identify important candidate genes related to salt tolerance. Results In this study, 68 AvPP2C genes and 68 AhPP2C genes were identified from the genomes of A. venetum and A. hendersonii, respectively. They were classified into 13 subgroups based on their phylogenetic relationships and were further analyzed for their subcellular locations, gene structures, conserved structural domains, and cis-acting elements. The results of qRT-PCR analyses of seven AvPP2C genes and seven AhPP2C genes proved that they differed significantly in gene expression under salt stress. It has been observed that the PP2C genes in A. venetum and A. hendersonii exhibit different expression patterns. Specifically, AvPP2C2, 6, 24, 27, 41 and AhPP2C2, 6, 24, 27, 42 have shown significant differences in expression under salt stress. This indicates that these genes may play a crucial role in the salt tolerance mechanism of A. venetum and A. hendersonii. Conclusions In this study, we conducted a genome-wide analysis of the AvPP2C and AhPP2C gene families in Apocynum, which provided a reference for further understanding the functional characteristics of these genes.

Attentional control, guided by top-down processes, enables selective focus on pertinent information, while habituation, influenced by bottom-up factors and prior experiences, shapes cognitive responses by emphasizing stimulus relevance. These two fundamental processes collaborate to regulate cognitive behavior, with the prefrontal cortex and its subregions playing a pivotal role. Nevertheless, the intricate neural mechanisms underlying the interaction between attentional control and habituation are still a subject of ongoing exploration. To our knowledge, there is a dearth of comprehensive studies on the functional connectivity between subsystems within the prefrontal cortex during attentional control processes in both primates and humans. Utilizing stereo-electroencephalogram (SEEG) recordings during the Stroop task, we observed top-down dominance effects and corresponding connectivity patterns among the orbitofrontal cortex (OFC), the middle frontal gyrus (MFG), and the inferior frontal gyrus (IFG) during heightened attentional control. These findings highlighting the involvement of OFC in habituation through top-down attention. Our study unveils unique connectivity profiles, shedding light on the neural interplay between top-down and bottom-up attentional control processes, shaping goal-directed attention.

Subsidence, the downward vertical land motion (VLM), plays a pivotal role in contributing to the risk of coastal flooding. Accurately estimating VLM and identifying its potential features related to subsidence can provide crucial information for stakeholders to make better-informed decisions. This study aimed to estimate large-scale subsidence at the Texas Gulf Coast and identify potential subsidence features using explainable models. Nine potential features were considered for modeling the VLM, ranging from natural terrain variations to anthropogenic activities. These features were used to train a random forest (RF) machine learning model. Explainable artificial intelligence (XAI) techniques including SHapley Additive exPlanations (SHAP) and impurity- and permutation-based feature importance were used to identify the contributions to subsidence. The results demonstrated favorable performance of the RF model, achieving an $R^{2}$ value of 0.56 during validation. XAI results underscored the significance of the digital elevation model in explaining subsidence at the Texas Coast. Additionally, XAI analysis highlighted the overall contribution of subsidence from anthropogenic activities, such as hydrocarbon extraction and groundwater withdrawal. Furthermore, the sample-level SHAP map provided detailed and reasonable subsidence-attribution results across the study area, showing potential for automatic and data-driven explanations of the VLM.

Environmentally friendly technologies for the prevention and control of crop diseases and insect pests are important to reduce the use of chemical pesticides, improve the quality of agricultural products, protect the environment, and promote sustainable development of crop production. On the basis of Internet of Things (IoT) technology, we developed a prevention and control system for crop diseases and insect pests with two main components: a plant protection device (the hardware) and an information management system (the software). To be suitable for both facility- and field-based production scenarios, we incorporated two types of plant protection devices, utilizing ozone sterilization and light-trap technologies. The devices were equipped with various sensors to realize real-time collection and monitoring of data on the crop production environment. The information management system has an IoT-based architecture and includes a mobile device app to enable remote control of the plant protection devices for intelligent management of plant protection data. The system can achieve efficient management of large-scale equipment applications and multi-device collaborative work to prevent and control pests and diseases. The developed system has operated successfully for several years in China and has been applied to cucumber, tomato, rice, and other crops. We demonstrate the effectiveness and practicality of the system in a greenhouse facility and in the field.

The study of the properties of ferroelectric materials against irradiation has a long history. However, anti−irradiation research on the ferroelectric domain has not been carried out. In this paper, the irradiation of switched domain structure is innovatively proposed. The switched domain of 700 nm lithium niobate (LiNbO3, LN) thin film remains stable after gamma irradiation from 1 krad to 10 Mrad, which was prepared by piezoresponse force microscopy (PFM). In addition, the changing law of domain wall resistivity is explored through different sample voltages, and it is verified that the irradiated domain wall conductivity is still larger than the domain. This domain wall current (DWC) property can be applied to storage, logic, sensing, and other devices. Based on these, a ferroelectric domain irradiation resistance model is established, which explains the reason at an atomic level. The results open a possibility for exploiting ferroelectric materials as the foundation in the application of space and nuclear fields.

Mapping large-scale coastal subsidence is significant in providing valuable support to decision-making stakeholders to recognize impacts of potential natural disasters. However, this task presents significant challenges due to its highly complex nature of the spatial–temporal variability. Recent advances in the observation capability of synthetic aperture radar (SAR) missions and the processing algorithms of interferometric SAR (InSAR) techniques have made it possible to efficiently map large-scale subsidence from space. This study utilized the persistent scatterer (PS) InSAR method to process four swathes of Sentinel-1 SAR images captured between 2016 and 2022 to map large-scale subsidence along the Texas coastline. The subsidence map, encompassing over 1.7 million PS points, was calibrated/validated with observations of 115 continuously operating global navigation satellite system (cGNSS) stations. Nineteen subsiding hotspots were identified and potential subsidence drivers were analyzed, including hydrocarbon extraction (HE), groundwater withdrawal (GW), and salt domes. The results suggest that HE activities were the primary driver of observed subsidence in the Coastal Bend and South Texas, as well as in the Southeast Texas regions. In the Houston-Galveston Area, subsidence appears to be influenced by a combination of GW, HE, and salt dome movements. Within 50 km of the Texas shorelines, subsidence rates were approximately −1.0 mm/yr within the coastal vicinity, while inland areas experience a gradually increasing trend. Specific coastal areas, such as Corpus Christi, Freeport, Seabrook, San Leon, and others, may face elevated flooding risks during high tide and storm events with the sea-level rise trend up to 24.0 mm/yr in relation to coastal subsiding land.

Monitoring vertical land motion (VLM) along coastlines, which influences the dynamics of sea level changes in relation to the land, is a challenging task due to its inherent high spatiotemporal variability and limited availability of observations. This study aimed to investigate the rates, patterns, and drivers of land subsidence near the coastal town of San Leon, TX, United States, since the 1990s, utilizing a range of space and terrestrial geodetic techniques. These techniques included interferometric synthetic aperture radar (InSAR), global navigation satellite systems (GNSS), tide gauge (TG), and satellite radar altimetry (SRA). The small baseline subset (SBAS) InSAR method was adopted to process 254 images from three synthetic aperture radar (SAR) sensors, i.e., ERS-2 between 1995 and 1999, ALOS-1 PALSAR between 2006 and 2011, and Sentinel-1 between 2016 and 2020. The results from InSAR subsidence maps were verified by comparing with high-accuracy vertical positioning observations at ten continuously operating GNSS (cGNSS) stations. Within the study area, a special attention was given to the Eagle Point TG station where sea level has been significantly rising relative to the sinking land. Long-term time series of land subsidence at Eagle Point obtained from sea-level difference between TG and SRA observations were confirmed and compared against InSAR and the cGNSS station observations recorded in close proximity. Gaussian Process regression (GPR) was then employed to model the VLM processes at Eagle Point using: (1) the combined results of InSAR and sea-level difference (i.e., GPR 1), and (2) the InSAR results alone (i.e., GPR 2). A 0.9 mm/yr divergence was found between GPR 1 and GPR 2 models, indicating the potential to accurately estimate long-term VLM with InSAR standalone measurements even if multi-year observation gaps intermittently occur, especially for inland areas where measurement data from other geodetic techniques, such as GNSS, TG, and SRA, are not available. Further investigations suggest that land subsidence around Eagle Point since 1998 was related to anthropogenic activities such as hydrocarbon pumping from oil and gas wells that were situated in close proximity to the TG station.

Introduction: Outer membrane proteins are crucial in maintaining the structural stability and permeability of the outer membrane. Outer membrane proteins exhibit several functions such as antigenicity and strong immunogenicity, which have potential applications in clinical diagnosis and disease prevention. However, wet experiments for studying OMPs are time and capital-intensive, thereby necessitating the use of computational methods for their identification.Methods: In this study, we developed a computational model to predict outer membrane proteins. The non-redundant dataset consists of a positive set of 208 outer membrane proteins and a negative set of 876 non-outer membrane proteins. In this study, we employed the pseudo amino acid composition method to extract feature vectors and subsequently utilized the support vector machine for prediction.Results and Discussion: In the Jackknife cross-validation, the overall accuracy and the area under receiver operating characteristic curve were observed to be 93.19% and 0.966, respectively. These results demonstrate that our model can produce accurate predictions, and could serve as a valuable guide for experimental research on outer membrane proteins.

Summary: Apocynum species have great application prospects in textile and phytoremediation of saline soil, are rich in flavonoids, and possess medicinal significance. Here, we report the draft genomes of Apocynum venetum and Apocynum hendersonii, and elucidate their evolutionary relationship. The high synteny and collinearity between the two suggested that they have experienced the same WGD event. Comparative analysis revealed that flavone 3-hydroxylase (ApF3H) and differentially evolved flavonoid 3-O-glucosyl transferase (ApUFGT) genes are critical for determining natural variation in flavonoid biosynthesis between the species. Overexpression of ApF3H-1 enhanced the total flavonoid content and promoted the antioxidant capacity of transformed plants compared to the wild-type. ApUFGT5 and 6 explained the diversification of flavonoids or their derivatives. These data provide biochemical insight and knowledge on the genetic regulation of flavonoid biosynthesis, supporting the adoption of these genes in breeding programs aimed at the multipurpose utilization of the plants.

A WUSCHEL-related homeobox (WOX) gene family has been implicated in promoting vegetative organs to embryonic transition and maintaining plant embryonic stem cell identity. Using genome-wide analysis, we identified 17 candidates, WOX genes in ramie (Boehmeria nivea). The genes (BnWOX) showed highly conserved homeodomain regions typical of WOX. Based on phylogenetic analysis, they were classified into three distinct groups: modern, intermediate, and ancient clades. The genes displayed 65% and 35% collinearities with their Arabidopsis thaliana and Oryza sativa ortholog, respectively, and exhibited similar motifs, suggesting similar functions. Furthermore, four segmental duplications (BnWOX10/14, BnWOX13A/13B, BnWOX9A/9B, and BnWOX6A/Maker00021031) and a tandem-duplicated pair (BnWOX5/7) among the putative ramie WOX genes were obtained, suggesting that whole-genome duplication (WGD) played a role in WOX gene expansion. Expression profiling analysis of the genes in the bud, leaf, stem, and root of the stem cuttings revealed higher expression levels of BnWOX10 and BnWOX14 in the stem and root and lower in the leaf consistent with the qRT-PCR analysis, suggesting their direct roles in ramie root formation. Analysis of the rooting characteristics and expression in the stem cuttings of sixty-seven different ramie genetic resources showed a possible involvement of BnWOX14 in the adventitious rooting of ramie. Thus, this study provides valuable information on ramie WOX genes and lays the foundation for further research.

Head and neck squamous cell carcinoma (HNSCC) is one of the most common human cancers; however, its outcome of pharmacotherapy is always very limited. Herein, we performed a batch query in the connectivity map (cMap) based on bioinformatics, queried out 35 compounds with therapeutic potential, and screened out parbendazole as a most promising compound, which had an excellent inhibitory effect on the proliferation of HNSCC cell lines. In addition, tubulin was identified as a primary target of parbendazole, and the direct binding between them was further verified. Parbendazole was further proved as an effective tubulin polymerization inhibitor, which can block the cell cycle, cause apoptosis and prevent cell migration, and it exhibited reasonable therapeutic effect and low toxicity in the in vivo and in vitro anti-tumor evaluation. Our study repositioned an anthelmintic parbendazole to treat HNSCC, which revealed a therapeutic utility and provided a new treatment option for human cancers.

It is crucial nowadays to accurately monitor vertical land motion (VLM) near coasts and understand its spatial and temporal variability to quantify its impact to water-land interactions. This study developed and employed a double difference (DD) network adjustment method to reconstruct daily VLM time series and estimate their trends using sea-level observations recorded at 24 tide gauge (TG) stations located on the Texas coastline between 1993 and 2020. Based on the first difference between TG and the satellite radar altimetry (SRA) observations, the method calculated DD between correlated TG pairs to mitigate sea-level variations. Then all potential connections of TG pairs across the 24 stations were combined to form a TG network regarding the DD values. On a daily basis, the DD values reflected relative elevation difference for a specific connection and the network could be solved via weighted least squares to reconstruct elevations (i.e., VLM time series) at different TG stations. Results indicated that at most TG stations, clear VLM patterns were restored and found to be largely consistent with time series derived from continuously-operating global navigation satellite system (cGNSS). In addition, greater than 80% of the TG stations used in the work showed a linear VLM trend with an uncertainty of less than or equal to 1.0 mm/yr. The proposed DD network adjustment method provides long and continuous VLM time-series results, and has a potential to complement other geodetic techniques, particularly along the coastal areas where TG stations were densely installed.

Stretchable, skin-conformal, and self-powered wearable pressure sensors have garnered significant attention for use in human joint bending motion monitoring. Here, a piezo-triboelectric pressure sensor (P-TPS) based on triboelectric nanogenerator and piezoelectric nanogenerator is demonstrated. The P-TPS can generate an enhanced electrical output by coupling the dual-mode triboelectrification and piezoelectric effect. The P-TPS shows high sensitivity (voltage = 0.3 V/kPa; current = 4.3 nA/kPa; pressure range = 0–200 kPa), high linearity, and good stability. Furthermore, it demonstrates a wide measurement range (0–800 kPa), table frequency response, and fast response time. Additionally, all components of the P-TPS are fabricated using flexible and stretchable materials, affording satisfactory stretchability and excellent skin conformality. Owing to their ability to self-power, they can be attached to the outside of joints to monitor human joint bending movements in real time. Hence, this study provides a novel method of using a stretchable and skin-conformal piezo-triboelectric nanogenerator with high electrical performance as a self-powered pressure sensor, which offers significant potential in personalized recognition, medical research, and human machine interface.

In micron or nano smart sensing systems, piezoelectric cantilever beams are distributed as major components in microsensors, actuators, and energy harvesters. This paper investigates the performance of four cantilever beam devices with “electric-force” conversion based on the inverse piezoelectric effect of lithium niobate (LiNbO3, LN) single-crystal materials. A new compact piezoelectric smart device model is proposed, designed as a single mass block connected by four beams, where devices exhibit smaller lateral errors (0.39–0.41%). The relationship between the displacement characteristics of cantilever beams and driving voltage was researched by applying excitation signals. The results show that the device has the maximum displacement at a first-order intrinsic frequency (fosc = 11.338 kHz), while the displacement shows a good linear relationship (R2 = 0.998) with driving voltage. The square wave signals of the same amplitude have greater “electrical-force” conversion efficiency. The output displacement can reach 12 nm, which is much higher than the output displacement with sinusoidal excitation. In addition, the relative displacement deviation of devices can be maintained within ±1% under multiple cycles of electrical signal loading. The small size, high reliability, and ultra-stability of Si–LN ferroelectric single-crystal cantilever beam devices with lower vibration amplitudes are promising for nanopositioning techniques in microscopy, diagnostics, and high-precision manufacturing applications.

The protein phosphatase 2C (PP2C), a key regulator of the ABA signaling pathway, plays important roles in plant growth and development, hormone signaling, and abiotic stress response. Although the PP2C gene family has been identified in many species, systematic analysis was still relatively lacking in ramie (Boehmeria nivea L.). In the present study, we identified 63 BnPP2C genes from the ramie genome, using bioinformatics analysis, and classified them into 12 subfamilies, and this classification was consistently supported by their gene structures and conserved motifs. In addition, we observed that the functional differentiation of the BnPP2C family of genes was restricted and that fragment replication played a major role in the amplification of the BnPP2C gene family. The promoter cis-regulatory elements of BnPP2C genes were mainly involved in light response regulation, phytohormone synthesis, transport and signaling, environmental stress response and plant growth and development regulation. We identified BnPP2C genes with tissue specificity, using ramie transcriptome data from different tissues, in rhizome leaves and bast fibers. The qRT-PCR results showed that the BnPP2C1, BnPP2C26 and BnPP2C27 genes had a strong response to drought, high salt and ABA, and there were a large number of stress-responsive elements in the promoter region of BnPP2C1 and BnPP2C26. The results suggested that BnPP2C1 and BnPP2C26 could be used as the candidate genes for drought and salt tolerance in ramie. These results provide a reference for further studies on the function of the PP2C gene and advance the development of the mechanism of ramie stress response, with a view to providing candidate genes for the molecular breeding of ramie for drought and salt tolerance.

Abstract Abscisic acid (ABA) is known as an important hormone regulating plant stress resistance, such as salt, drought and heavy metal resistance. However, the relationship between ABA and cadmium (Cd) enrichment in ramie (Boehmeria nivea L.) is still unclear to date. This study aimed to reveal the effect of ABA on Cd enrichment in ramie, and we received the following results: (1) Under Cd treatment, the Cd uptake of ramie increased with the increase of Cd concentration, but the chlorophyll content decreased. Under Cd treatment, the ABA content was highest in roots of ramie, followed by that in old leaves, and lowest in new leaves. Long-time treatment of high Cd concentration reduced the ability of endogenous ABA biosynthesis. (2) Spraying ABA on ramie plants (SORP) and adding ABA directly to the culture solution (ADCS) with low concentration can promote the growth of ramie and increase the amount of Cd uptake, and the effect of SORP is better. (3) The molecular reason for the decrease of chlorophyll content due to Cd stress, may be resulted from the down-regulated expression of the chlorophyll synthesis genes (BnPAO and BnNYC1) and the up-regulated expression of the chlorophyll degradation genes (BnCHLH, BnCHLG, BnHAP3A and BnPPR1). The elevated ABA content in ramie plants may due to the up-regulated expression of the ABA synthesis related genes (BnABA1, BnNCED3, and BnNCED5) and the genes (BnABCG40, BnNFXL2, BnPYL9, BnGCR2, BnGTG1, BnBGLU1, BnUTG1, BnVHAG1 and BnABI5) that encoding ABA transport and response proteins, which was consistent with the enhance the Cd uptake in ramie. Our study revealed the relationship between ABA and Cd uptake in ramie, which provided a reference for improving the enrichment of Cd in ramie.

Background: The family with sequence similarity 83 member D (FAM83D) protein is known to play a significant role in many human diseases. However, its role in cancer remains ambiguous. This study aimed to investigate the function of FAM83D in a pan-cancer analysis, with a special focus on breast cancer.Methods: Samples were collected from The Cancer Genome Atlas (TCGA) and used for bioinformatic analysis. Datasets from the Gene Expression Omnibus (GEO) and Genotype-Tissue Expression (GTEx) databases were also analyzed for verification. The potential value of FAM83D as a prognostic and diagnostic biomarker was visualized through R software. The “survival” and “GSVA” package were used for univariate, multivariate and pathway enrichment analyseis. We further analyzed the CancerSEA databases and TISIDB websites for single-cell and immune-related profiling. Lastly, we validated those data in vitro using quantitative reverse transcriptase-polymerase chain reaction (RT‒qPCR), cell counting kit-8 (CCK-8), transwell, flow cytometry, and tumorigenicity assays in a murine cell line model.Results: The expression of FAM83D in tumor samples was significantly higher than in normal tissues for most cancer types in the datasets. We confirmed this finding using RT‒qPCR in a breast cancer cell line. Analysis of multiple datasets suggests that overall survival (OS) was extremely poor for breast cancer patients with high FAM83D expression. The CCK-8 assay demonstrated that MCF-7 cell proliferation was inhibited after genetic silencing of FAM83D. Transwell assay showed that knockdown of FAM83D significantly inhibited the invasion and migration ability of MCF-7 cells compared to the control. The results of flow cytometry showed that silencing FAM83D could block the G1 phase of MCF-7 cells compared with negative groups. The tumorigenicity assay in nude mice indicated that the tumorigenic ability to silence FAM83D decreased compared.Conclusion: Results suggest that FAM83D expression can serve as a valuable biomarker and core gene across cancer types. Furthermore, FAM83D expression is significantly associated with MCF-7 cell proliferation and thus may be a prospective prognostic biomarker especially for breast cancer.

Chemotherapy, as one main strategy to relieve tumor progression, has a weak effect on triple-negative breast cancer (TNBC) chest wall metastasis. The development of near-infrared (NIR) light-responsive nanomaterials for chemodynamic therapy (CDT) and photothermal therapy (PTT) is a promising platform but still challenging in biomedicine. This study reports a peroxidase mimicking nanozyme (Fe-N-C SAzyme) against TNBC by CDT and PTT. Fe-N-C SAzyme generated reactive oxygen species (ROS) by decomposing H2O2 into hydroxyl radicals (•OH) and also induced light-to-heat conversion under the exposure of 808 nm laser irradiation. With these biological characteristics, the obtained Fe-N-C SAzymes displayed enhanced cell cytotoxicity and inhibition of cancer cell proliferation both in vitro and in vivo at a low dose of nanoagent and a moderate NIR laser power density. Besides, Fe-N-C nanoagent with its excellent ROS generation brought metabolic reprogramming of elevated glycolysis in tumor cells. In vivo experiments, when combined with PTT, the enhanced antitumor effect was found by the elimination of M-MDSC in tumor microenvironment. Fe-N-C SAzymes can serve as a new synergistic CDT and PTT nanoagent to simultaneously reprogram tumor metabolism and tumor microenvironment. It will provide prospects for chemodynamic/photothermal combined cancer therapy for TNBC chest wall metastasis based on the use of a single nanosystem.

Abstract Radish (Raphanus sativus) is an important vegetable worldwide that exhibits different flesh and skin colors. The anthocyanins responsible for the red and purple coloring in radishes possess nutritional value and pharmaceutical potential. To explore the structural and regulatory networks related to anthocyanin biosynthesis and identify key genes, we performed comparative transcriptome analyses of the skin and flesh of six colored radish accessions. The transcript profiles showed that each accession had a species-specific transcript profile. For radish pigmentation accumulation, the expression levels of anthocyanin biosynthetic genes (RsTT4, RsC4H, RsTT7, RsCCOAMT, RsDFR, and RsLDOX) were significantly upregulated in the red- and purple-colored accessions, but were downregulated or absent in the white and black accessions. The correlation test, combined with metabolome (PCC > 0.95), revealed five structural genes (RsTT4, RsDFR, RsCCOAMT, RsF3H, and RsBG8L) and three transcription factors (RsTT8-1, RsTT8-2, and RsPAR1) to be significantly correlated with flavonoids in the skin of the taproot. Four structural genes (RsBG8L, RsDFR, RsCCOAMT, and RsLDOX) and nine transcription factors (RsTT8-1, RsTT8-2, RsMYB24L, RsbHLH57, RsPAR2L, RsbHLH113L, RsOGR3L, RsMYB24, and RsMYB34L) were found to be significantly correlated with metabolites in the flesh of the taproot. This study provides a foundation for future studies on the gene functions and genetic diversity of radish pigmentation and should aid in the cultivation of new valuable radish varieties.
