Abstract

In this paper, we study a paratransit application in which children are transported every day from their homes to their special schools or medical-social establishments. To optimize this transportation system, the establishments collaborate to propose a joint transportation plan. We propose a new algorithm to jointly build vehicle routes that visit several establishments and simultaneously set the establishments’ opening hours. This algorithm combines a large neighborhood search, the resolution of a route-based model, and the progressive shrinkage of the planning window. It is applied to a real case from the area of Lyon in France, including 34 schools and 575 heterogeneous users served by a heterogeneous fleet of reconfigurable vehicles. On average, we show that in addition to the 10% of saving that can be expected by sharing vehicle routes between schools, 7% of additional savings can be achieved by school bell adjustment. This cost saving also decreases average user ride times and the number of vehicles required, creating longer routes that are more attractive for driver services. Abstract

In this paper, we study the Landau equation under the Navier-Stokes scaling in the torus for hard and moderately soft potentials. More precisely, we investigate the Cauchy theory in a perturbative framework and establish some new short time regularization estimates for our rescaled nonlinear Landau equation. These estimates are quantified in time and optimal, indeed, we obtain the instantaneous expected anisotropic gain of regularity (see [53] for the corresponding hypoelliptic estimates on the linearized Landau collision operator). Moreover, the estimates giving the gain of regularity in the velocity variable are uniform in the Knudsen number. Intertwining these new estimates on the Landau equation with estimates on the Navier-Stokes-Fourier system, we are then able to obtain a result of strong convergence towards this fluid system.

Abstract

This paper discusses interdisciplinary collaborative practices based on the AdA project (meaning “Affektrhetoriken des Audiovisuellen” https://projectada.github.io/) in which institutionally situated research groups from computational sciences and film studies developed approaches for the analysis of the temporal dynamics of audiovisual expressivity by bringing together a theoretically informed methodology of film analysis and computational video analysis and semantic video annotation. In an earlier publication, we already presented how co-building a Digital Humanities tool could be seen as instrumental genesis. In it we concentrated on the iterative development of a visualisation framework to highlight the need for a common language and for the consideration of time needed for collaboration.We would like to pursue the reflection on the instrumental genesis (Rabardel, 1995) of collaboration tools along the time dimension. We saw that every step of tool development for such qualitative and exploratory research requires time: may it be for tool sketching, development, testing, or documenting its usage for manual annotation.

Abstract

International audience; Instrumental genesis through interdisciplinary collaboration-reflections on the emergence of a visualisation framework for video annotation data XML This paper presents, discusses and reflects on the development of a visualization framework for the analysis of the temporal dynamics of audiovisual expressivity. The main focus lies on the instrumental genesis process (Rabardel 1995; Longchamp 2012)-a concept trying to express and analyze the co-evolution of instruments and the practices they make possible-underlying this development. It is described through the collaboration and communication processes between computer science scholars and humanities scholars in finding new ways of visualizing complex datasets for exploration and presentation in the realm of film-studies research. It draws on the outcome and concrete usage of the visualizations in publications and presentations of a research group, the AdAproject, that investigates the audiovisual rhetorics of affect in audiovisual media on the financial crisis (2007-). These film analyses are based on theoretical assumptions on the process of film-viewing, the relation of the viewer's perception and the temporally unfolding audiovisual images, and a methodical approach that draws on 'steps' in the research process such as segmentation, description and qualification, called eMAEX (Kappelhoff et al. 2011-2016) to reconstruct these experiential figurations (Bakels et al. 2020a, 2020b). The main focus of this paper is the process of iterative development of visualizations as interactive interfaces generated with the open-source software Advene, that were an integral part of the research process. In this regard, the timeline visualization is not only of interest for visual argumentation in (digital) humanities publications, but also for the creation of annotations as well as the exploration of this data. In the first part of the paper we describe this interdisciplinary collaboration as instrumental genesis on a general level-as an evolving and iterative process. In the second part we focus on the specific challenge of designing a visualization framework for the temporal dynamics of audiovisual aesthetics. Lastly we zoom out by reflecting on experiences and insights that might be of interest for the wider digital humanities community.

Abstract

Controversial reports have suggested that SARS-CoV E and 3a proteins may be viroporins that conduct currents through the plasma membrane of the infected cells. If true, these proteins would represent accessible targets for the development of new antiviral drugs by using high-throughput patch-clamp techniques. Here we aimed at better characterizing the cell responses induced by E or 3a protein with a particular focus on the ion conductances measured at the cell surface. First, we show that expression of SARS-CoV-2 E or 3a protein in CHO cells gives rise to cells with newly-acquired round shape, tending to detach from the Petri dish. This suggests that cell death is induced upon expression of E or 3a protein. We confirmed this hypothesis by using flow cytometry, in agreement with earlier reports on other cell types. In adhering cells expressing E or 3a protein, whole-cell currents were in fact not different from the control condition indicating that E and 3a proteins are not plasma membrane viroporins. In contrast, recording currents on detached cells uncovered outwardly-rectifying currents, much larger than those observed in control. The current characteristics are reminiscent of what was previously observed in cells expressing SARS-CoV-1 E or 3a proteins. Herein, we illustrate for the first time that carbenoxolone blocks these outward currents suggesting that they are conducted by pannexin channels, mostly likely activated by cell morphology change and/or cell death. Alongside we also demonstrate that truncation of the C-terminal PDZ binding motifs reduces the proportion of dying cells but does not prevent pannexin currents suggesting distinct pathways for cell death and pannexin currents induced by E and 3a proteins. We conclude that SARS-CoV-2 E and 3a proteins are not acting as viroporins expressed at the plasma membrane.Author SummaryA viroporin (or viral porin) is a class of proteins that is encoded by a virus genome. It is named porin because its biological role is to conduct ions through a pore that it created in a lipid membrane such as the one surrounding a human cell. if such viroporin is present at the external membrane of a human cell infected by a virus, it can be an easy target of an antiviral agent which thus does not have to enter the cell to be active. One example of viroporin is the flu M2 protein that is the target of amantadine, an antiviral agent used against flu. In previous studies, two proteins of SARS-CoV viruses, named E protein and 3a protein, have been suggested to be viroporins at the surface of infected human cells, potentially opening a new research avenue against SARS. Here we demonstrate that both proteins are not viroporins at the external membrane but they rather trigger changes in the cell shape and promote cell death. They only indirectly induce the activity of a porin that is encoded by the cell genome, named pannexin.

Abstract

It is very well known that the fraction of energy in a hadron collision going into electromagnetic particles (electrons and photons, including those from decays) has a large impact on the number of muons produced in air shower cascades. Recent measurements at the LHC confirm features that can be linked to a mixture of different underlying particle production mechanisms such as a collective statistical hadronization (core) in addition to the expected string fragmentation (corona). Since the two mechanisms have a different electromagnetic energy fraction, we present a possible connection between statistical hadronization in hadron collisions and muon production in air showers. Using a novel approach, we demonstrate that the core-corona effect as observed at the LHC could be part of the solution for the lack of muon production in simulations of high energy cosmic rays. To probe this hypothesis, we study hadronization in high energy hadron collisions using calorimetric information over a large range of pseudorapidity in combination with the multiplicity of central tracks. As an experimental observable, we propose the production of energy in electromagnetic particles versus hadrons, as a function of pseudorapidity and central charged particle multiplicity.

International audience; This paper considers the problem of maximizing an expectation function over a finite set, or finite-arm bandit problem. We first propose a naive stochastic bandit algorithm for obtaining a probably approximately correct (PAC) solution to this discrete optimization problem in relative precision, that is a solution which solves the optimization problem up to a relative error smaller than a prescribed tolerance, with high probability. We also propose an adaptive stochastic bandit algorithm which provides a PAC-solution with the same guarantees. The adaptive algorithm outperforms the mean complexity of the naive algorithm in terms of number of generated samples and is particularly well suited for applications with high sampling cost.

Abstract

International audience; Thymic epithelial tumors are rare neoplastic proliferations of thymic epithelial cells. The aggressiveness of these malignancies increases as higher is the histologic subtype, being thymic carcinoma the most aggressive subtype, with a greater tendency to metastatic spread. In metastatic setting, there is no standard treatment after progression on platinum-based chemotherapy. In this scenario, monotherapy treatment either with lenvatinib, a multi-tyrosine kinase inhibitor with antiangiogenic properties, or pembrolizumab, an immune-checkpoint inhibitor, has reported clinical activity. Potential combination of both agents may have synergistic activity as reported in other cancer types. PECATI trial is a single-arm, investigator-initiated phase II study aiming to assess the activity and safety of the combination of lenvatinib and pembrolizumab in 43 patients with advanced B3-thymoma or thymic carcinoma who progressed on or after at least one previous line of platinum-based chemotherapy. The primary endpoint of the trial is 5-month progression-free survival rate and the secondary endpoints include overall response rate, duration of response, and overall survival.

Abstract

In this paper, we aim to account for the distribution and interpretation of a novel class of free choice items in Romanian, which we refer to as additive free choice items (ADD-FCIs). We show that the internal composition of ADD-FCIs, as well as their distribution, differs from that attested for other free choice paradigms discussed in the literature. Morphologically, ADD-FCIs are a more complex variant of regular universal FCIs, by virtue of an additional morpheme. This morpheme plays an additive role when it functions as a stand-alone particle, and we propose that its role is similarly additive when it functions as an infix in ADD-FCIs. Couched in an exhaustification framework, we put forward a novel compositional account that can derive the interpretation of these ADD-FCIs. As for their distribution, these elements are only found in the presence of the conditional mood, as well as unconditional structures. We show how our analysis, coupled with the Viability Constraint used to explain the distribution of regular FCIs, can also account for their restricted distribution. In doing so, we further show how the pattern we investigate opens new perspectives regarding the licensing of free choice items in unconditionals.

Abstract

International audience; The article proposes an analysis of the implicit ideas that characterize secondary school geography, using the case of metropolization. The analysis is based on a corpus of students' notebooks, textbooks and the program for the sixth grade of secondary school. It revealed, on the one hand, implicit knowledge for the students concerning the division of knowledge and the ways of doing things in the discipline, and on the other hand, a tacit agreement between the students and the teachers concerning the point of view on the world and the position of knowledge constructed. The theme of metropolization makes it possible to uncover an implicit norm of national framing of disciplinary knowledge in high school, in contradiction with the multi-situated approaches of a scientific geography where the production of knowledge has become globalized.; L’article propose une analyse des implicites qui caractérisent la géographie du secondaire à partir du cas de la métropolisation. L'analyse s’appuie sur un corpus de cahiers d’élèves, de manuels et sur le programme de la classe de première. Elle amène à distinguer d’une part, des implicites pour les élèves concernant le découpage des savoirs et les façons de faire dans la discipline, et d’autre part, un accord tacite concernant les élèves et les enseignants à propos du point de vue sur le monde et de la position de savoir construite. Le thème de la métropolisation permet de mettre au jour une norme implicite de cadrage national du savoir disciplinaire en lycée, en porte-à-faux avec les approches multisituées d’une géographie scientifique où la production de savoir s’est mondialisée.

International audience; Intermittent computing is an emerging paradigm for systems without batteries and powered by intermittent energy sources. This paradigm promises a more energy-efficient design of computing systems. It seems particularly well suited to the field of connected sensors that form the first level of the Internet of Things. This application domain requires a reactive computing model. The definition of an intermittent and reactive model is a problem that has not yet been fully explored in the literature. In this paper, we focus on the modeling and analysis of intermittent reactive systems. We first introduce an extension of Time Petri Nets with cost to model the different dimensions of the system: concurrency, real time, energy consumption and reward representing the gains generated by the system when it has succeeded in carrying out certain actions. We then aim to synthesize optimal runs of the model that achieve the highest possible reward under a given cost (energy) constraint. We propose a symbolic algorithm for constrained-cost state space computation and prove its termination. We then present algorithms for the synthesis of the optimal traces from an exhaustive or partial state space exploration. We finally illustrate the cost-optimal traces synthesis on a case study and show how that can be used online for joint management of computing time and energy.

Abstract

In this work we continue to investigate well-posedness for stable driven McKean-Vlasov SDEs with distributional interaction kernel following the approach introduced in [8]. We specifically focus on the impact of the Besov smoothness of the initial condition and quantify how it affects the corresponding density estimates for the SDE. In particular, we manage to attain some critical thresholds allowing to revisit/address in a stable noise setting some concrete physical and biological models.

Abstract

International audience; The role of extracellular vesicles (EVs) from faeces (fEVs) and small circulating EVs (cEVs) in liver diseases such as non-alcoholic fatty diseases (NAFLD) and nonalcoholic steatohepatitis (NASH) has not been demonstrated. fEVs and cEVs of healthy donors, NAFLD and NASH patients were isolated and characterized. The effects of EVs were evaluated in intestinal, endothelial, Kupffer and stellate cells. Nonmuscular myosin light chain kinase (nmMLCK) deficient mice were used in vivo. Bacterial origins of fEVs were analysed by 16s rDNA gene sequencing. fEVs and small cEVs were composed of prokaryotic and eukaryotic origins. Only NASH-fEVs exerted deleterious effects. NASH-fEVs increased intestinal permeability and reduced expression of tight junction proteins that were prevented by nmMLCK inhibition, increased endothelial cell permeability and inflammatory cytokines and chemokines requiring TLR4/lipopolysaccharide pathway. NASH-fEVs and NASH-cEVs activated profibrotic and proinflammatory proteins of hepatic stellate cells. Treatment with NASH-fEVs evoked an increase in intestinal permeability in wild type but not in nmMLCK deficient mice. Bacterial origins of fEVs were different between NAFLD and NASH patients and 16 amplicon sequence variants were differentially abundant. We demonstrate that fEVs actively participate in barrier dysfunctions leading to liver injuries underscoring the role of nmMLCK and lipopolysaccharide carried by fEVs.

Contains fulltext : 290818.pdf (Publisher’s version ) (Open Access) BACKGROUND: Flow cytometry (FCM) aids the diagnosis and prognostic stratification of patients with suspected or confirmed myelodysplastic syndrome (MDS). Over the past few years, significant progress has been made in the FCM field concerning technical issues (including software and hardware) and pre-analytical procedures. METHODS: Recommendations are made based on the data and expert discussions generated from 13 yearly meetings of the European LeukemiaNet international MDS Flow working group. RESULTS: We report here on the experiences and recommendations concerning (1) the optimal methods of sample processing and handling, (2) antibody panels and fluorochromes, and (3) current hardware technologies. CONCLUSIONS: These recommendations will support and facilitate the appropriate application of FCM assays in the diagnostic workup of MDS patients. Further standardization and harmonization will be required to integrate FCM in MDS diagnostic evaluations in daily practice. 01 januari 2023

Abstract

Non-cyanobacteria diazotrophs (NCDs) were shown to dominate in surface waters shifting the long-held paradigm of cyanobacteria dominance and raising fundamental questions on how these putative heterotrophic bacteria thrive in sunlit oceans. Here, we report an unprecedented finding in the widely used model diatom Phaeodactylum tricornutum (Pt) of NCDs sustaining diatom cells in the absence of bioavailable nitrogen. We identified PtNCDs using metagenomics sequencing and detected nitrogenase gene in silico and/or by PCR. We demonstrated nitrogen fixation in PtNCDs and their close genetic affiliation with NCDs from the environment. We showed the wide occurrence of this type of symbiosis with the isolation of NCDs from other microalgae, their identification in the environment, and predicted their associations with photosynthetic microalgae. Overall, this study provides evidence for a previously overlooked symbiosis using a multidisciplinary model-based approach, which will help understand the different players driving global marine nitrogen fixation.

Abstract

International audience; This paper presents a variant of the vehicle routing problem regarding the delivery of products to customers in cities with a combination of walking and driving. The objective is first to offer a better modeling of delivery problems in congested cities and also to evaluate potential savings in traveled distances and parking times. We introduce the Park-and-Loop Routing Problem with Parking Selection (PLRP-PS) in which a parking space or loading zone has to be found for the driver and his vehicle before he walks to deliver to one or several customers. In this paper, we will focus on cases where parking locations should be selected among a large set of parking areas. To solve this problem, we developed a variant of the large neighborhood search metaheuristic called Small and Large Neighborhood Search (SLNS). We will focus on designing and comparing simple and efficient techniques to select parking spots for vehicles before goods are delivered by walking trips. The efficiency of the approach is demonstrated in the park-andloop routing problem, with eleven new best solutions found on an existing benchmark. Some realistic instances were generated based on open data from the city of Nantes, France. In these instances, we find that combining walking and driving to deliver to the center of a city can save 19% of working time on average compared to the classical vehicle routing approach.

Abstract

International audience; Computer musicians refer to mesostructures as the intermediate levels of articulation between the microstructure of waveshapes and the macrostructure of musical forms. Examples of mesostructures include melody, arpeggios, syncopation, polyphonic grouping, and textural contrast. Despite their central role in musical expression, they have received limited attention in recent applications of deep learning to the analysis and synthesis of musical audio. Currently, autoencoders and neural audio synthesizers are only trained and evaluated at the scale of microstructure: i.e., local amplitude variations up to 100 milliseconds or so. In this paper, we formulate and address the problem of mesostructural audio modeling via a composition of a differentiable arpeggiator and time-frequency scattering. We empirically demonstrate that time-frequency scattering serves as a differentiable model of similarity between synthesis parameters that govern mesostructure. By exposing the sensitivity of short-time spectral distances to time alignment, we motivate the need for a time-invariant and multiscale differentiable time-frequency model of similarity at the level of both local spectra and spectrotemporal modulations.

Abstract

It has been proposed that magnetic flux expulsion due to outer core fluid upwellings may affect the geomagnetic secular variation on the core-mantle boundary (Bloxham, 1986). In this process intense horizontal field lines are concentrated below the outer boundary, introducing small radial length scales and consequently strong radial diffusion. We explore such magnetic boundary layers in numerical dynamo simulations with heterogeneous outer boundary heat flux inferred from a tomographic model of lower mantle seismic shear waves velocity anomalies. Our scheme associates magnetic boundary layers to peak horizontal magnetic fields at the top of the shell. In our models mean magnetic boundary layer thickness ranges ≈200-400 km and decreases with increasing magnetic Reynolds number. Extrapolation or interpolation to Earth's core conditions based on total core flow amplitude or its poloidal part gives mean magnetic boundary layer thickness of ≈220 and ≈260-330 km, respectively. We find magnetic boundary layers associated with the azimuthal field at the equatorial region, whereas magnetic boundary layers associated with the meridional field are found at mid latitudes. Negative outer boundary heat flux anomalies yield preferred locations of expulsion of azimuthal field below Africa and the Pacific (at low latitudes of the Northern Hemisphere.), while positive outer boundary heat flux anomalies yield preferred locations of expulsion of meridional field below the Americas and East Asia. Abstract

This article reports on the inclusive production cross section of several quarkonium states, $\mathrm{J}/\psi$, $\psi {\rm (2S)}$, $\Upsilon\rm(1S)$, $\Upsilon\rm(2S)$, and $\Upsilon\rm(3S)$, measured with the ALICE detector at the LHC, in pp collisions at $\sqrt{s} = 5.02$ TeV. The analysis is performed in the dimuon decay channel at forward rapidity ($2.5 < y < 4$). The integrated cross sections and transverse-momentum ($p_{\rm T}$) and rapidity ($y$) differential cross sections for $\mathrm{J}/\psi$, $\psi {\rm (2S)}$, $\Upsilon\rm(1S)$, and the $\psi {\rm (2S)}$-to-$\mathrm{J}/\psi$ cross section ratios are presented. Abstract

International audience; Aggressive B-cell malignancies, such as mantle cell lymphoma (MCL), are microenvironment-dependent tumors and a better understanding of the dialogs occurring in lymphoma protective ecosystems will provide new perspectives to increase treatment efficiency. To identify novel molecular regulations, we performed a transcriptomic analysis based on the comparison of circulating (n=77) versus MCL lymph nodes (n=107) together with RNA sequencing of malignant (n=8) versus normal B-cell (n=6) samples. This integrated analysis led to the discovery of microenvironment-dependent and tumor-specific secretion of the interleukin-32 beta (IL32β), whose expression was confirmed in situ within MCL lymph nodes by multiplex immunohistochemistry. Using ex vivo models of primary MCL cells (n=23), we demonstrated that, through the secretion of IL32β, the tumor was able to polarize monocytes into specific MCL-associated macrophages, which in turn favor tumor survival. We highlighted that while IL32β-stimulated macrophages secreted several protumoral factors, they supported tumor survival through a soluble dialog, mostly driven by BAFF. Finally, we demonstrated the efficacy of selective NIK/alternative-NFNB inhibition to counteract microenvironment-dependent induction of IL32β and BAFF-dependent survival of MCL cells. This data uncovered the IL32β/BAFF axis as a previously undescribed pathway involved in lymphoma-associated macrophages polarization and tumor survival, which could be counteracted through selective NIK inhibition.

Abstract

γδ T cells stimulated by phosphoantigens (pAg) are potent effectors that secrete Th1 cytokines and kill tumor cells. Consequently, they are considered candidates for use in cancer immunotherapy. However, they have proven only moderately effective in several clinical trials. We studied the consequences of pAg-stimulated γδ T-cell interactions with natural killer (NK) cells and CD8+ T cells, major innate and adaptive effectors, respectively. We found that pAg-stimulated γδ T cells suppressed NK-cell responses to “missing-self” but had no effect on antigen-specific CD8+ T-cell responses. Extensive analysis of the secreted cytokines showed that pAg-stimulated γδ T cells had a proinflammatory profile. CMV-pp65–specific CD8+ T cells primed with pAg-stimulated γδ T cells showed little effect on responses to pp65-loaded target cells. By contrast, NK cells primed similarly with γδ T cells had impaired capacity to degranulate and produce IFNγ in response to HLA class I–deficient targets. This effect depended on BTN3A1 and required direct contact between NK cells and γδ T cells. γδ T-cell priming of NK cells also led to a downregulation of NKG2D and NKp44 on NK cells. Every NK-cell subset was affected by γδ T cell–mediated immunosuppression, but the strongest effect was on KIR+NKG2A– NK cells. We therefore report a previously unknown function for γδ T cells, as brakes of NK-cell responses to “missing-self.” This provides a new perspective for optimizing the use of γδ T cells in cancer immunotherapy and for assessing their role in immune responses to pAg-producing pathogens.See related Spotlight by Kabelitz, p. 543.

Abstract

One-dimensional bin-packing is a well-known combinatorial optimization problem which is strongly NP-hard. It consists of allocating a given set of items of different sizes into bins of the same capacity to minimize the number of bins used. The capacity of each bin cannot be exceeded. This paper deals with some variants of this problem to take into account the cases when there are items with uncertain sizes. The goal is to obtain robust solutions taking into account possible variations of item sizes around their nominal values. First, two robust approaches are considered which are based on a stability radius calculation, to ensure that the stability radius, measured either with the Manhattan or Chebyshev norm, is not below a given threshold. Then, a complementary robust approach is applied which is based on a relative resiliency calculation. To solve to optimality these robust variants of the bin-packing problem, a compact 0-1 linear programming formulation, which is also valid for the standard bin-packing problem, is introduced. Then, a Dantzig-Wolfe decomposition is suggested in order to provide a set-cover reformulation with a stronger linear relaxation, but an exponential number of columns. Finally, to obtain integer optimal solutions, a branch-and-price algorithm is developed, whose linear relaxation of the set-cover formulation is solved by a dynamic column generation. Numerical experiments are conducted on adapted benchmark sets from the literature. The performance of the branch-and-price algorithm allows us to investigate what protection against uncertainty is offered by each approach, and at which cost of robustness.

Abstract

International audience; The replication crisis in psychology has led to question popular phenomena such as ego depletion, which has been criticized after studies failed to replicate. Here, we describe limitations in the literature that contributed to these failures and suggest how they may be addressed. At the theoretical level, the literature focuses on two out of at least eight identified auxiliary hypotheses. Thus, the majority of the hypotheses related to the three core assumptions of the ego-depletion theory have been overlooked, thereby preventing the rejection of the theory as a whole. At the experimental level, we argue that the low replicability of ego-depletion studies could be explained by the absence of a comprehensive, integrative, and falsifiable definition of self-control, which is central to the concept of ego depletion; by an unclear or absent distinction between ego depletion and mental fatigue, two phenomena that rely on different processes; and by the low validity of the tasks used to induce ego depletion. Finally, we make conceptual and methodological suggestions for a more rigorous investigation of ego depletion, discuss the necessity to take into account its dynamic and multicomponent nature, and suggest using the term self-control fatigue instead.

First, new global robust fixed-time stability results are proposed for scalar systems by using constant and variable exponent coefficients. Then, they are applied to global robust fixed-time stabilization of a class of uncertain nonlinear second order systems by using sliding mode control. All the results are illustrated in simulation.

Abstract

Geological disasters such as slope failure and landslides can cause loss of life and property. Therefore, reproducing their evolution process is of great importance for risk assessment and mitigation. The recently developed SIMSAND critical state sand model combined with the smoothed particle hydrodynamics (SPH) method is adopted in this work to study slope failure under large deformations. To illustrate the efficiency and accuracy of the SIMSAND-SPH approach, a series of slope collapse studies using the discrete element method (DEM) considering various particle shapes (i.e. spherical, tetrahedral and elongated) is adopted as benchmarks. The parameters of the SIMSAND model are calibrated using DEM triaxial tests. In comparison to the DEM simulations, the runout distance and final slope height are well characterized with the SIMSAND-SPH approach with less computational cost. All comparisons show that the SIMSAND-SPH approach is highly efficient and accurate, which can be an alternative numerical tool to simulate real scale granular flow.

Abstract

This paper addresses the complexity reduction of stochastic homogenization of a class of random materials for a stationary diffusion equation. A cost-efficient approximation of the correctors is obtained using a method designed to exploit quasi-periodicity. Accuracy and cost reduction are investigated for local perturbations or small transformations of periodic materials as well as for materials with no periodicity but a mesoscopic structure, for which the limitations of the method are shown. Finally, for materials outside the scope of this method, we propose to use the approximation of homogenized quantities as control variates for variance reduction of a more accurate and costly Monte Carlo estimator (using a multifidelity Monte Carlo method). The resulting cost reduction is illustrated in a numerical experiment and compared with a control variate method from weakly stochastic homogenization. The limits of this variance reduction technique are tested on materials without periodicity or mesoscopic structure.

Abstract

Background Muscular dystrophies (MDs) are inherited diseases in which a dysregulation of the immune response exacerbates disease severity and are characterized by infiltration of various immune cell types leading to muscle inflammation, fiber necrosis and fibrosis. Immunosuppressive properties have been attributed to mesenchymal stem cells (MSCs) that regulate the phenotype and function of different immune cells. However, such properties were poorly considered until now for adult stem cells with myogenic potential and advanced as possible therapeutic candidates for MDs. In the present study, we investigated the immunoregulatory potential of human MuStem (hMuStem) cells, for which we previously demonstrated that they can survive in injured muscle and robustly counteract adverse tissue remodeling. Methods The impact of hMuStem cells or their secretome on the proliferative and phenotypic properties of T-cells was explored by co-culture experiments with either peripheral blood mononucleated cells or CD3-sorted T-cells. A comparative study was produced with the bone marrow (BM)-MSCs. The expression profile of immune cell-related markers on hMuStem cells was determined by flow cytometry while their secretory profile was examined by ELISA assays. Finally, the paracrine and cell contact-dependent effects of hMuStem cells on the T-cell-mediated cytotoxic response were analyzed through IFN-γ expression and lysis activity. Results Here, we show that hMuStem cells have an immunosuppressive phenotype and can inhibit the proliferation and the cytotoxic response of T-cells as well as promote the generation of regulatory T-cells through direct contact and via soluble factors. These effects are associated, in part, with the production of mediators including heme-oxygenase-1, leukemia inhibitory factor and intracellular cell adhesion molecule-1, all of which are produced at significantly higher levels by hMuStem cells than BM-MSCs. While the production of prostaglandin E2 is involved in the suppression of T-cell proliferation by both hMuStem cells and BM-MSCs, the participation of inducible nitric oxide synthase activity appears to be specific to hMuStem cell-mediated one. Conclusions Together, our findings demonstrate that hMuStem cells are potent immunoregulatory cells. Combined with their myogenic potential, the attribution of these properties reinforces the positioning of hMuStem cells as candidate therapeutic agents for the treatment of MDs.

Highlights • Radiotherapy can trigger cytokine release syndrome and tumor lysis syndrome in association with chemotherapy drugs. • Extreme caution must be given when irradiating large tumor volumes at high risk of tumor lysis. • Evidence of cytokine release following radiotherapy unveils the systemic interactions of radiotherapy with the immune system., We present the case of a 53-year-old woman treated with analgesic radiotherapy for a multiple myeloma bone lesion of the forearm. After a first fraction of 5 Gray (Gy), she presented with an acute respiratory syndrome with fever a few hours after the treatment. The same symptoms occurred after the second fraction 3 days later. The patient recovered quickly thanks to intravenous hydration and suspension of the radiotherapy. Biological tests revealed a tumor lysis syndrome. We concluded that the clinical symptoms could be defined as cytokine release syndrome. This is the second time in the literature that cytokine release syndrome has been described following radiotherapy. First, we synthesize TLS and radiotherapy to determine how radiotherapy could be a trigger associated with other well-known factors. Furthermore, we discuss radiotherapy and cytokine release syndrome. Summary We present the case of a woman treated with analgesic radiotherapy for a multiple myeloma bone lesion. Following the first and the second treatment fraction, the patient presented with an acute respiratory syndrome with fever and biological tests revealed a tumor lysis syndrome. We concluded that the clinical symptoms could be defined as cytokine release syndrome. Abstract

Summary Background The use of cyclophosphamide in patients with acute exacerbation of idiopathic pulmonary fibrosis (IPF) is unknown. Our study was designed to evaluate the efficacy and safety of four cyclophosphamide pulses in addition to high-dose methylprednisolone in this population. Methods In this double-blind, placebo-controlled trial done in 35 departments across 31 hospitals in France, adult patients (≥18 years) with acute exacerbation of IPF and those with suspected acute exacerbation of IPF were randomly assigned in a 1:1 ratio using a web-based system to receive either intravenous pulses of cyclophosphamide (600 mg/m2) plus uromitexan as haemorrhagic cystitis prophylaxis (200 mg/m2) at the time of cyclophosphamide administration and then again, 4 h later, or placebo at days 0, 15, 30, and 60. Random assignment was stratified according to the severity of IPF and was block-balanced with variable block sizes of four or six patients. Patients receiving mechanical ventilation, with active infection, with active cancer, or who were registered on the lung transplant waiting list were excluded. All patients received standardised high-dose glucocorticoids. The investigators, patients, and the sponsor were masked to the treatment assignments. The primary endpoint was 3-month all-cause mortality, analysed by a χ2 test adhering to an intention-to-treat principle. The trial is now complete and registered with ClinicalTrials.gov , NCT02460588 . Findings Between Jan 22, 2016, and July 19, 2018, 183 patients were assessed for eligibility, of whom 120 patients were randomly assigned and 119 patients (62 [52%] with severe IPF) received at least one dose of cyclophosphamide (n=60) or placebo (n=59), all of whom were included in the intention-to-treat analysis. The 3-month all-cause mortality was 45% (27/60) in patients given cyclophosphamide compared with 31% (18/59) in the placebo group (difference 14·5% [95% CI −3·1 to 31·6]; p=0·10). Similar results were found after adjustment by IPF severity (odds ratio [OR] 1·89 [95% CI 0·89–4·04]). The risk of death at 3 months, independent of the treatment received, was higher with severe than non-severe IPF (OR 2·62 [1·12–6·12]) and was lower with the use of antifibrotic therapy (OR 0·33 [0·13–0·82]). Adverse events were similar between groups by 6 months (25 [42%] in the cyclophosphamide group vs 30 [51%] in the placebo group) and their proportion, including infections, did not differ. Overall infection was the main adverse event and occurred in 20 (33%) of 60 patients in the cyclophosphamide group versus 21 (36%) of 59 patients in the placebo group. Interpretation In patients with acute exacerbation of IPF, adding intravenous cyclophosphamide pulses to glucocorticoids increased 3-month mortality. These findings provide evidence against the use of intravenous cyclophosphamide in such patients. Abstract

International audience; Amorphous calcium carbonate nanoparticles (ACC NPs) are promising multifunctional materials for healthcare applications. Due to their instability in aqueous media, pure ACC NPs for the biomedical field are increasingly synthesised in absolute ethanol, using the ammonia diffusion method (ADM). Although this method presents the advantage of providing stable ACC NPs without additives, it requires the use of pure ethanol as solvent. New insights into the formation mechanisms of ACC NPs in ethanol using gas diffusion are presented in this article. The optimisation of the process according to these findings can increase the mass concentration of ACC NPs by a factor of 3.5. As a result, the amount of ethanol required to produce a target mass of particles is significantly decreased, reducing the ecological impact of the process. The stabilisation of the resulting ACC NPs in aqueous media is achieved by a short-time process using phospholipids based on the ethanol injection method. By using the natural electrostatic affinity of negatively charged materials for the positive surface of ACC NPs in ethanol, we reduced the process time from 24 h to 2 minutes, compared with the closest state of the art, decreasing the operating time and corresponding energy consumption. The process does not require the use of synthetic PEGylated lipids for steric stabilisation. In addition, a natural egg-sourced phospholipid was identified as an efficient stabiliser for the first time. The upscaling of our process was successfully demonstrated using a 50 L reactor for bulk synthesis, as well as a continuous flow reactor for industrial continuous flow production.

Abstract

International audience; Assessment of visual encryption of video and image content requires a reliable estimation of content recognizability and low quality. As pointed out in the literature, current methods are insufficient and research into this topic, as well as into the relation between low quality and recognizability, is still lacking. This lack of research is primarily due to a lack of data. To improve on the status-quo we have taken a recognizability database and performed a subjective quality evaluation on a subset of the images. This gives us a new database with both subjective recognizability and quality information and allows to delve into the relation between low quality and recognizability. We analyze the relationship between quality and recognizability as well as the predictive quality of state of the art visual quality indices. We show that the visual quality indices are poor indicators for the estimation of recognizability. Furthermore, we show that they must be a poor fit because of the disparity between two distinct perceptual tasks: quality and recognizability.

Abstract

International audience; Traditional manufacturing systems face a major transition challenge toward intelligent and sustainable manufacturing systems. Energy-efficient manufacturing has become one of the main focuses of this transition because of the high and critical consumption of energy for the production of many industrial sectors. To this aim, modeling Energy Consumption (EC) behavior is a primary step to monitor and reduce consumption. However, it requires a deep understanding of the system’s kinematic and dynamic behaviors. Hence, creating a model from scratch can be challenging, which may result in a model that does not correctly represent the real system. With the advancement of digital technologies, it is now possible to collect and analyze data from manufacturing systems in real-time. This opens the door to the possibility of modeling the energy consumption behavior of different machine states based on a data-driven approach and keeping the current energy consumption under control and monitored using real-time data from the equipment. Prior research has been conducted in the literature incorporating EC modeling into a Digital Twin (DT). However, the addressed issue remains an open challenge due to its complexity. The proposed methodology solves the lack of literature by proposing a methodology that makes the EC modeling within the reach of any researcher or practitioner in the field. The current paper proposes a data-driven methodology for integrating the EC model into Digital Twins. The methodology is based on measurements to identify different segments and sub-states of EC of production equipment, using techniques such as segmentation and regression. It relies on power absorption measurement of industrial equipment to generate EC related-parameters to be fed into the DT model to monitor the current operating condition of the physical system. This work contributes to the DT-based sustainable transition by allowing to monitor and quantitatively measure those parameters which could be controlled to reduce EC. A case study on an industrial robot is used to validate and assess the performance of the approach in a laboratory environment.

Abstract

This article seeks to determine the acoustic correlates of gemination in Standard Somali (Afroasiatic, Cushitic), in particular whether closure duration is the primary acoustic correlate distinguishing singleton and geminate stops, with immediate consequences for the analysis of word-initial strengthening. We provide an acoustic analysis of word-initial and word-internal voiced singletons as well as of their geminate counterparts on the basis of a production experiment conducted with four native speakers. Three temporal and four non-temporal acoustic properties of /b d ɡ/ and /bb dd ɡɡ/ are examined and systematically compared (closure duration, release burst duration, vowel duration; and closure amplitude, release amplitude, presence of a release burst, (de)voicing). We argue that the opposition between singleton and geminate voiced stops is primarily realized as the manner contrast approximant [β̞ ð̞ ɣ̞] vs. stop [b d ɡ]. Word-initially, Somali exhibits various peculiarities that are reminiscent of the cross-linguistically attested phenomenon of domain-initial strengthening. This article provides the first study of this phenomenon in Somali. We establish that word-initial /b d ɡ/ and word-medial /bb dd ɡɡ/ share the same closure duration, release burst duration, and vowel duration within the Prosodic Word. They also have a similar closure amplitude, and voicing properties. Moreover, the acoustic properties of word-initial /b d ɡ/ remain constant, and do not depend on their position in the prosodic hierarchy. On the basis of these results, the article also aims at providing new insights in the phonological representation of Somali geminates and word boundaries, and thus contributes to the understanding of word-initial strengthening in Somali.

Abstract

International audience; ABSTRACT Purpose PIK3CA pathogenic variants in the PIK3CA-related overgrowth spectrum (PROS) activate phosphoinositide 3-kinase signaling, providing a rationale for targeted therapy, but no drug has proven efficacy and safety in this population. Our aim was to establish the six-month tolerability and efficacy of low-dose taselisib, a selective class I PI3K inhibitor, in PROS patients. Methods Patients over 16 years with PROS and PIK3CA pathogenic variants were included in a phase IB/IIA multicenter, open-label single-arm trial (six patients at 1 mg/day of taselisib, then 24 at 2 mg/day). The primary outcome was the occurrence of dose limiting toxicity (DLT). Efficacy outcomes were the relative changes after treatment of (1) tissue volume at affected and unaffected sites, both clinically and on imaging; (2) cutaneous vascular outcomes when relevant; (3) biologic parameters; (4) quality of life; and (5) patient-reported outcomes. Results Among 19 enrolled patients, 2 experienced a DLT (enteritis and pachymeningitis) leading to early trial termination (17 treated, 10 completed the study). No serious adverse reaction occurred in the 1 mg cohort ( n = 6). No significant reduction in affected tissue volume was observed (mean −4.2%; p = 0.81; SD 14.01). Thirteen (76.4%) participants reported clinical improvement (pain reduction, chronic bleeding resolution, functional improvement). Conclusion Despite functional improvement, the safety profile of low-dose taselisib precludes its long-term use.

Abstract

International audience; A series of automatic time-lapse cameras installed along the southwestern side of Tasiapik Valley, near the village of Umiujaq, Nunavik (northern Québec) documented several departure modes and types of snow involved in snow avalanches during winter 2017-2018. These included cornice-avalanche dynamics, slab and loose snow avalanches, and clean and dirty snow avalanches. At the top of the selected slope, a camera monitored the development of a snow cornice beginning in November 2017, detecting multiple cornice failures over the winter and spring. The track and deposition area of the runout paths were monitored from two cameras downslope, revealing the concomitance of snow-cornice fall and snow avalanche triggering. Snow avalanche activity remained relatively infrequent until the end of May 2018. Spring snow avalanche activity is characterized by wet and dirty snow avalanches carrying debris to the foot of the slope and by runout zones located near the road along the slope.

The authors wish to thank Dr. Dominic Edelmann (Division of Biostatistics, DKFZ) for helpful advice about data analysis., There is overwhelming epidemiologic evidence that the risk of multiple myeloma (MM) has a solid genetic background. Genome-wide association studies (GWAS) have identified 23 risk loci that contribute to the genetic susceptibility of MM, but have low individual penetrance. Combining the SNPs in a polygenic risk score (PRS) is a possible approach to improve their usefulness. Using 2361 MM cases and 1415 controls from the International Multiple Myeloma rESEarch (IMMEnSE) consortium, we computed a weighted and an unweighted PRS. We observed associations with MM risk with OR = 3.44, 95% CI 2.53-4.69, p = 3.55 x 10(-15) for the highest vs. lowest quintile of the weighted score, and OR = 3.18, 95% CI 2.1 = 34-4.33, p = 1.62 x 10(-13) for the highest vs. lowest quintile of the unweighted score. We found a convincing association of a PRS generated with 23 SNPs and risk of MM. Abstract

Aims and context: Dark diversity is an emerging and promising concept proposed to estimate the recruitment potential of natural communities and guide conservation and restoration policies. It represents all the species that could be present in a community due to favourable environmental conditions, but are currently lacking. To date, experimental approaches only measured taxonomic dark diversity, mainly based on species coexistence, which relies partly on neutral processes. Thus, these approaches may fail to identify the biodiversity which is lacking for deterministic reasons, and can hence hardly bring out suitable restoration methods. Methods: Here, we propose a novel method to estimate dark diversity, which is based on more deterministic coexistence: the coexistence of functional features. We adapted the Beals co-occurrence index using functional groups, and we estimated functional dark diversity based on coexistence of functional groups. We then made use of functional dark diversity to address a persistent issue of restoration ecology: how does passive rewilding impact the ecological integrity of recovered communities? We compared spontaneous, secondary woodlands with ancient forests, in terms of taxonomic and functional dark diversity of vascular plants and spiders. Results: Our results indicated that functional dark diversity does not equate to taxonomic dark diversity. Considering plants, recent woodlands surprisingly harboured less functional dark diversity than ancient forests, while they had a very similar amount of taxonomic dark diversity. Concerning spiders, recent woodlands harboured a similar amount of functional dark diversity as ancient forests, but more taxonomic dark diversity. Also, the composition of functional dark diversity differed between forest types, shedding light on their past assembly processes and unveiling their potential for conservation and effective restoration. Synthesis: Functional dark diversity brings novel perspectives for ecological diagnostic and restoration. Combined to taxonomic dark diversity, it enables to identify easily the deterministic constrains which limit the re-assembly of ecological communities after land-use changes and to predict the realistic, possible establishments of functional features. Here, we showed that spontaneous woodlands can have very similar, sometimes even higher, ecological integrity than that of ancient forests, and hence may be valuable habitats to be conserved from an ecological perspective.

Abstract

Despite interesting properties, the use of 67Cu, 186Re and 47Sc theranostic radionuclides in preclinical studies and clinical trials is curtailed by their limited availability due to a lack of widely established production methods. An IAEA Coordinated Research Project (CRP) was initiated to identify important technical issues related to the production and quality control of these emerging radionuclides and related radiopharmaceuticals, based on the request from IAEA Member States. The international team worked on targetry, separation, quality control and radiopharmaceutical aspects of the radionuclides obtained from research reactors and cyclotrons leading to preparation of a standard recommendations for all Member States. The CRP was initiated in 2016 with fourteen participants from thirteen Member States from four continents. Extraordinary results on the production, quality control and preclinical evaluation of selected radionuclides were reported in this project that was finalized in 2020. The outcomes, outputs and results of this project achieved by participating Member States are described in this minireview.

Abstract

International audience; Enceladus is the first planetary object for which direct sampling of a subsurface water reservoir, likely habitable, has been performed. Over a decade of flybys and seven flythroughs of its watery plume, the Cassini spacecraft determined that Enceladus possesses all the ingredients for life. The existence of active eruptions blasting fresh water into space, makes Enceladus the easiest target in the search for life elsewhere in the Solar System. Flying again through the plume with more advanced instruments, landing at the surface near active sources and collecting a sample for return to Earth are the natural next steps for assessing whether life emerges in this active world. Characterizing this habitable world also requires detailed mapping and monitoring of its tidally-induced activity, from the orbit as well as from the surface using complementary platforms. Such ambitious goals may be achieved in the future in the framework of ESA large or medium-class missions in partnership with other international agencies, in the same spirit of the successful Cassini-Huygens mission. For all these reasons, exploring habitable ocean worlds, with Enceladus as a primary target, should be a priority topic of the ESA Voyage 2050 programme.

Abstract

International audience; Recent development in smart devices has lead us to an explosion in data generation and heterogeneity, which requires new network solutions for better analysing and understanding traffic. These solutions should be intelligent and scalable in order to handle the huge amount of data automatically. With the progress of high-performance computing (HPC), it becomes feasible easily to deploy machine learning (ML) to solve complex problems and its efficiency has been validated in several domains (e.g., healthcare or computer vision). At the same time, network slicing (NS) has drawn significant attention from both industry and academia as it is essential to address the diversity of service requirements. Therefore, the adoption of ML within NS management is an interesting issue. In this paper, we have focused on analyzing network data with the objective of defining network slices according to traffic flow behaviors. For dimensionality reduction, the feature selection has been applied to select the most relevant features (15 out of 87 features) from a real dataset of more than 3 million instances. Then, a K-Means clustering is applied to better understand and distinguish behaviors of traffic. The results demonstrated a good correlation among instances in the same cluster generated by the unsupervised learning. This solution can be further integrated in a real environment using network function virtualization.

Abstract

Purpose: DYRK1A syndrome is among the most frequent monogenic forms of intellectual disability (ID). We refined the molecular and clinical description of this disorder and developed tools to improve interpretation of missense variants, which remains a major challenge in human genetics. Methods: We reported clinical and molecular data for 50 individuals with ID harboring DYRK1A variants and developed (1) a specific DYRK1A clinical score; (2) amino acid conservation data generated from 100 DYRK1A sequences across different taxa; (3) in vitro overexpression assays to study level, cellular localization, and kinase activity of DYRK1A mutant proteins; and (4) a specific blood DNA methylation signature. Results: This integrative approach was successful to reclassify several variants as pathogenic. However, we questioned the involvement of some others, such as p.Thr588Asn, still reported as likely pathogenic, and showed it does not cause an obvious phenotype in mice. Conclusion: Our study demonstrated the need for caution when interpreting variants in DYRK1A, even those occurring de novo. Abstract

International audience; While early maps are known from all over the world, the key questions always involve: what exactly do they show? And what spatial extent do they cover? In this context, we recently used 3D-modelling to re-examine a carved stone slab datable to the Early Bronze Age (c.2150–1600 BC) that was found at Saint-Bélec in Brittany. We show that the surface of the slab had been shaped in three dimensions to represent the relief of the surrounding landscape in which it was found, while several engraved motifs on it evoke contemporary structures known archaeologically. We argue that the Saint-Bélec slab represents an area of c.545 km2 corresponding to the extent of a prehistoric political entity. The carving and subsequent burying of the slab can be linked to the postulated rise and fall of hierarchical societies and raises many wider questions about socio-economic structures in temperate Europe at that time.

Abstract

International audience; This paper proposes a longitudinal control framework for platooning in an urban environment. The targetted application is to redistribute vehicles involved in car-sharing systems, where only the leader vehicle is human-driven. We propose a platoon model and control law considering actuator dynamics. This control relies on a hybrid Information Flow Topology (IFT), where the leader broadcasts its state but each follower only measures the position of its predecessor. A consensusbased control law incorporates the effect of the network/sensor time delay and the variable velocity of the leader. Conditions for the platoon internal and string stability are given. Experiments demonstrate the efficiency of the framework in simulation and real experiments with three commercial cars.

Abstract

Although splenectomy is still considered the most effective curative treatment for immune thrombocytopenia (ITP), its use has significantly declined in the last decade, especially since the approval of thrombopoietin receptor agonists (TPO-RAs). The main objective of the study was to determine whether splenectomy was still as effective nowadays, particularly for patients with failure to respond to TPO-RAs. Our secondary objective was to assess, among patients who relapsed after splenectomy, the pattern of response to treatments used before splenectomy. This multicentre retrospective study involved adults who underwent splenectomy for ITP in France from 2011 to 2020. Response status was defined according to international criteria. We included 185 patients, 100 (54.1%) and 135 (73.0%) patients had received TPO-RAs and/or rituximab before the splenectomy. The median follow-up after splenectomy was 39.2 months [16.5-63.0]. Overall, 144 (77.8%) patients had an initial response and 23 (12.4%) experienced relapse during follow-up, for an overall sustained response of 65.4%, similar to that observed in the pre-TPO-RA era. Among patients who received at least one TPO-RA or rituximab before splenectomy, 92/151 (60.9%) had a sustained response. Six of 13 (46%) patients with previous lack of response to both TPO-RAs and rituximab had a sustained response to splenectomy. Among patients with relapse after splenectomy, 13/21 (61.2%) patients responded to one TPO-Ras that failed before splenectomy. In conclusion, splenectomy is still a relevant option for treating adult primary ITP not responding to TPO-RAs and rituximab. Patients with lack of response or with relapse after splenectomy should be re-challenged with TPO-RAs. This article is protected by copyright. All rights reserved.
