Behavioural attraction of the malaria vector Anopheles coluzzii to human host odour increases during adult maturation. We have previously demonstrated that the onset of host seeking in An. coluzzii coincides with an increased sensitivity of the CO2-sensitive neurons and abundance of chemosensory receptor gene transcripts in the maxillary palp. In this study, we extend our analysis to the antenna. Functional characterisation of the near-complete repertoire of odorant receptors (Ors) expressed in this tissue, to fractioned human odour, reveals a subset of salient human odorants to be detected by Ors at physiological relevant concentrations. When presented as a blend in their ratio of natural emission, these odorants elicit attraction by host-seeking mosquitoes, emphasising that Ors alone can mediate this behaviour. However, the same blend inhibits attraction in teneral mosquitoes. This switch in behavioural response indicates a change in valence during adult maturation. Quantitative analysis of Or transcript abundance and in vivo electrophysiological analysis reveal that the olfactory system of female An. coluzzii undergoes concerted changes that correlate with the onset of host seeking. We conclude that changes in Or abundance modulate peripheral olfactory coding, resulting in ecologically relevant behavioural effects.

Septoria tritici blotch (STB) caused by the fungal pathogen Zymoseptoria tritici and powdery mildew (PM) caused by Blumeria graminis f.sp tritici (Bgt) are among the forefront foliar diseases of wheat that lead to a significant loss of grain yield and quality. Resistance breeding aimed at developing varieties with inherent resistance to STB and PM diseases has been the most sustainable and environment-friendly approach. In this study, 175 winter wheat landraces and historical cultivars originated from the Nordic region were evaluated for adult-plant resistance (APR) to STB and PM in Denmark, Estonia, Lithuania, and Sweden. Genome-wide association study (GWAS) and genomic prediction (GP) were performed based on the adult-plant response to STB and PM in field conditions using 7,401 single-nucleotide polymorphism (SNP) markers generated by 20K SNP chip. Genotype-by-environment interaction was significant for both disease scores. GWAS detected stable and environment-specific quantitative trait locis (QTLs) on chromosomes 1A, 1B, 1D, 2B, 3B, 4A, 5A, 6A, and 6B for STB and 2A, 2D, 3A, 4B, 5A, 6B, 7A, and 7B for PM adult-plant disease resistance. GP accuracy was improved when assisted with QTL from GWAS as a fixed effect. The GWAS-assisted GP accuracy ranged within 0.53–0.75 and 0.36–0.83 for STB and PM, respectively, across the tested environments. This study highlights that landraces and historical cultivars are a valuable source of APR to STB and PM. Such germplasm could be used to identify and introgress novel resistance genes to modern breeding lines.

Over the past 70 years, the world has witnessed extraordinary growth in crop productivity, enabled by a suite of technological advances, including higher yielding crop varieties, improved farm management, synthetic agrochemicals, and agricultural mechanization. While this “Green Revolution” intensified crop production, and is credited with reducing famine and malnutrition, its benefits were accompanied by several undesirable collateral effects (Pingali, 2012). These include a narrowing of agricultural biodiversity, stemming from increased monoculture and greater reliance on a smaller number of crops and crop varieties for the majority of our calories. This reduction in diversity has created vulnerabilities to pest and disease epidemics, climate variation, and ultimately to human health (Harlan, 1972). The value of crop diversity has long been recognized (Vavilov, 1992). A global system of genebanks (e.g., www.genebanks.org/genebanks/) was established in the 1970s to conserve the abundant genetic variation found in traditional “landrace” varieties of crops and in crop wild relatives (Harlan, 1972). While preserving crop variation is a critical first step, the time has come to make use of this variation to breed more resilient crops. The DivSeek International Network (https://divseekintl.org/) is a scientific, not-for-profit organization that aims to accelerate such efforts.

Background and aims Hepatitis delta virus (HDV) infection is associated with fast progression to liver cirrhosis and liver complications. Previous studies have, however, been mainly from tertiary care centers, with risk for referral bias toward patients with worse outcomes. Furthermore, the impact of HDV viremia per se on liver-related outcomes is not really known outside the human immunodeficiency virus co-infection setting. We have therefore evaluated the long-term impact of HDV viremia on liver-related outcomes in a nationwide cohort of patients with hepatitis B and D co-infection, cared for at secondary care centers in Sweden. Approach and results In total, 337 patients with anti-HDV positivity, including 233 patients with HDV RNA viremia and 91 without HDV viremia at baseline, were retrospectively studied, with a mean follow-up of 6.5 years (range, 0.5-33.1). The long-term risks for liver-related events (i.e., hepatocellular carcinoma [HCC], hepatic decompensation, or liver-related death/transplantation) were assessed, using Cox regression analysis. The risk for liver-related events and HCC was 3.8-fold and 2.6-fold higher, respectively, in patients with HDV viremia compared with those without viremia, although the latter was not statistically significant. Among patients with HDV viremia with no baseline cirrhosis, the cumulative risk of being free of liver cirrhosis or liver-related events was 81.9% and 64.0% after 5 and 10 years of follow-up, respectively. This corresponds to an incidence rate of 0.04 cases per person-year. Conclusions HDV RNA viremia is associated with a 3.8-fold higher risk for liver-related outcomes. The prognosis was rather poor for patients with HDV viremia without cirrhosis at baseline, but it was nevertheless more benign than previous estimates from tertiary centers. Our findings may be of importance when making decisions about treatment and evaluating potential outcomes of upcoming antivirals against HDV.

AIM: To evaluate three types of ventilation systems for operating rooms with respect to air cleanliness (in colony forming units, CFU/m(3)), energy consumption, and working environment comfort (noise and draught) as reported by surgical team members.METHODS: Two commonly used ventilation systems, vertical laminar airflow (LAF) and turbulent mixed airflow (TMA), were compared with a newly developed ventilation technique: temperature controlled airflow (TcAF). CFU concentrations were measured at three locations in an operating room during 45 orthopaedic surgeries: close to the wound (FINDINGS: We showed that LAF and TcAF, but not TMA, resulted in less than 10 CFU/m(3) at all measurement locations in the room during ongoing surgery. Median values of CFU/m(3) close to the wound (250 samples) were 0 for LAF, 1 for TcAF and 10 for TMA. Peripherally in the room, the CFU concentrations were lowest for TcAF. The CFU concentrations did not scale proportionally with airflow rates. Compared to LAF, TcAF's power consumption was 28% lower and there was significantly less disturbance from noise and draught.CONCLUSION: TcAF and LAF remove bacteria more efficiently from the air than TMA, especially close to the wound and instrument table. Like LAF, the new TcAF ventilation system maintained very low levels of CFU in the air, but TcAF used substantially less energy and provided a more comfortable working environment than LAF. This enables energy savings with preserved air quality.

BACKGROUND: Minimal hepatic encephalopathy (MHE) is clinically undetectable and the diagnosis requires psychometric tests. However, a lack of clarity exists as to whether the tests are in fact able to detect changes in cognition.AIM: To examine if the continuous reaction time test (CRT) can detect changes in cognition with anti-HE intervention in patients with cirrhosis and without clinically manifest hepatic encephalopathy (HE).METHODS: Firstly, we conducted a reproducibility analysis and secondly measured change in CRT induced by anti-HE treatment in a randomized controlled pilot study: We stratified 44 patients with liver cirrhosis and without clinically manifest HE according to a normal (n = 22) or abnormal (n = 22) CRT. Each stratum was then block randomized to receive multimodal anti-HE intervention (lactulose+branched-chain amino acids+rifaximin) or triple placebos for 3 months in a double-blinded fashion. The CRT is a simple PC-based test and the test result, the CRT index (normal threshold > 1.9), describes the patient's stability of alertness during the 10-minute test. Our study outcome was the change in CRT index in each group at study exit. The portosystemic encephalopathy (PSE) test, a paper-and-pencil test battery (normal threshold above -5), was used as a comparator test according to international guidelines.RESULTS: The patients with an abnormal CRT index who were randomized to receive the active intervention normalized or improved their CRT index (mean change 0.92 ± 0.29, p = 0.01). Additionally, their PSE improved (change 3.85 ± 1.83, p = 0.03). There was no such effect in any of the other study groups.CONCLUSION: In this cohort of patients with liver cirrhosis and no manifest HE, the CRT identified a group in whom cognition improved with intensive anti-HE intervention. This finding infers that the CRT can detect a response to treatment and might help in selecting patients for treatment.

Summary Barley (Hordeum vulgare L.) possesses a large and highly repetitive genome of 5.1 Gb that has hindered the development of a complete sequence. In 2012, the International Barley Sequencing Consortium released a resource integrating whole‐genome shotgun sequences with a physical and genetic framework. However, because only 6278 bacterial artificial chromosome (BACs) in the physical map were sequenced, fine structure was limited. To gain access to the gene‐containing portion of the barley genome at high resolution, we identified and sequenced 15 622 BACs representing the minimal tiling path of 72 052 physical‐mapped gene‐bearing BACs. This generated ~1.7 Gb of genomic sequence containing an estimated 2/3 of all Morex barley genes. Exploration of these sequenced BACs revealed that although distal ends of chromosomes contain most of the gene‐enriched BACs and are characterized by high recombination rates, there are also gene‐dense regions with suppressed recombination. We made use of published map‐anchored sequence data from Aegilops tauschii to develop a synteny viewer between barley and the ancestor of the wheat D‐genome. Except for some notable inversions, there is a high level of collinearity between the two species. The software HarvEST:Barley provides facile access to BAC sequences and their annotations, along with the barley–Ae. tauschii synteny viewer. These BAC sequences constitute a resource to improve the efficiency of marker development, map‐based cloning, and comparative genomics in barley and related crops. Additional knowledge about regions of the barley genome that are gene‐dense but low recombination is particularly relevant., Significance Statement We provide new genome sequence resources for barley and related species, to improve the efficiency of marker development, map‐based cloning, comparative genomics studies, and to provide insights into genome architecture. Notably, some of the gene‐rich islands in the barley genome deviate markedly from assumption that gene density and high recombination frequency are correlated.

Ripening of barrel-salted herring ( Clupea harengus ) is evaluated by the use of fluorescence spectroscopy and protein determinations. During ripening, protein degradation takes place in the herring and protein is extracted into the brine. The present study aims at identifying parameters which are correlated to the ripening characteristics of barrel-salted herring and which can provide a better understanding of the ripening process. Front face fluorescence landscapes were obtained by measuring directly on the brine from barrel-salted herring. These data were analyzed by parallel factor analysis (PARAFAC), which revealed four fluorophores, tryptophan (two states), vitamin B6 and riboflavin. All four parameters showed an increase in concentration during the storage period corresponding to an increase in protein content that varied from 3 g/100 g at day 60 to 5 g/100 g after 277 days of storage. It was not possible to see a difference in the development of the four fluorophores during the ripening period. The protein content was predicted from the fluorescence landscapes by partial least squares (PLS). The use of unfolded fluorescence spectra gave an RMSECV of 0.26 g/100 g and a correlation between the measured protein content and the predicted values of 0.86.

As the name dehydrins implies, these proteins are typically expressed in response to dehydration which can be caused by drought, osmotic stress or freezing temperatures 1. In general, dehydrins occur in plants as multi-gene families. Four Arabidopsis dehydrins (LTI29, ERD14, COR47 and RAB18) have been tested for protection of thylakoid membranes during freeze thaw cycle in vitro. 2,3 Our firstly reported results show that dehydrins LTI29, ERD14, COR47 have cryoprotective activity while RAB18 did not protect the thylakoid membranes at low temperatures. The cryoprotective activity reached a maximum of 50% to 60% at protein concentration of 140-250 ?g/ml in the assay. A contribution of dehydrins to freezing tolerance in vivo is supported by the observation of Nylander et al. (2001)4 that LTI29 and COR47 are cold induced at mRNA and protein expression level.

TiO2 dye sensitized solar cells (DSSCs) are emerging as an alternative technology to photovoltaic junction devices. DSSCs conventionally use liquid-type electrolytes but solid and gel polymer electrolytes are more promising due to many advantages. However, their conductivity needs to be enhanced in order to make them useful in practical device applications. In this work, an iodide ion conducting polymer electrolyte was prepared using polyacrylonitrile (PEO), tetrahexylammonium iodide salt with ethylene carbonate and propylene carbonate plasticisers. The salt composition was varied to find the optimum conductivity. The sample containing 120% salt with respect to PEO weight showed the highest conductivity. This optimum electrolyte showed a glass transition at -102.3°C and an ambient temperature conductivity of 2.1×10-3 S cm−1 at 30°C. The optimized gel electrolyte was used in a quasi solid-state DSSC and a ISC of 9.37 mA cm-2, a VOC of 696 mV and an efficiency of 2.8% were achieved under irradiation of 1000 W m−2. In addition, a solid polymer electrolyte was fabricated using the same salt with polyethylene oxide host polymer and ethylene carbonate. The salt composition was varied to obtain the highest conductivity and the best performing sample with composition 65% salt with respect to PEO weight showed a conductivity of 4.7×10-4 S cm-2 at 30°C. This electrolyte was used in a DSSC and it showed a ISC of 1.26 mA cm-2, a VOC of 745 mV and efficiency of 0.6%.

Close., Recent advances in high-throughput genotyping have made it easier to combine information from different mapping populations into consensus genetic maps, which provide increased marker density and genome coverage compared to individual maps. Previously, a single nucleotide polymorphism (SNP)-based genotyping platform was developed and used to genotype 373 individuals in four barley (Hordeum vulgare L.) mapping populations. This led to a 2943 SNP consensus genetic map with 975 unique positions. In this work, we add data from six additional populations and more individuals from one of the original populations to develop an improved consensus map from 1133 individuals. A stringent and systematic analysis of each of the 10 populations was performed to achieve uniformity. This involved reexamination of the four populations included in the previous map. As a consequence, we present a robust consensus genetic map that contains 2994 SNP loci mapped to 1163 unique positions. The map spans 1137.3 cM with an average density of one marker bin per 0.99 cM. A novel application of the genotyping platform for gene detection allowed the assignment of 2930 genes to flow-sorted chromosomes or arms, confirmed the position of 2545 SNP-mapped loci, added chromosome or arm allocations to an additional 370 SNP loci, and delineated pericentromeric regions for chromosomes 2H to 7H. Marker order has been improved and map resolution has been increased by almost 20%. These increased precision outcomes enable more optimized SNP selection for markerassisted breeding and support association genetic analysis and map-based cloning. Charged particles and photons have been measured in central Pb + Pb collisions at 158 A GeV in a common (eta-phi)-phase space region in the WA98 experiment at the CERN SPS. The measured distributions have been analyzed to quantify the frequency with which phase space regions of varying sizes have either small or large neutral pion fraction. The measured results are compared with VENUS model simulated events and with mixed events. Events with both large and small charged-neutral fluctuations are observed to occur more frequently than expected statistically, as deduced from mixed events, or as predicted by model simulations, with the difference becoming more prominent with decreasing size of the A Delta eta-Delta phi region. Forty-five cases of retinoblastoma in Sweden treated from 1959–1965 are reviewed. A variety of conservative methods were used in 23 eyes of which 13 were cured by radiotherapy. Local application of 60Co was found to be the most favourable method.

An unstable [t1/2 at 37 degrees = 32 +/- 2 (SD) sec] intermediate, thromboxane A2, was detected in the conversion of prostaglandin G2 into 8-(1-hydroxy-3-oxopropyl)-9,12L-dihydroxy-5,10-heptadecadienoic acid (thromboxane B2) in platelets. The intermediate was trapped by addition of methanol, ethanol, or sodium azide to suspensions of washed human platelets incubated for 30 sec with arachidonic acid or prostaglandin G2. The structures of the resulting derivatives demonstrated that the intermediate possessed an oxane ring as in thromboxane B2 but lacked its hemiacetal hydroxyl group. Additional experiments using 18O2 or [2H8]arachidonic acid in the formation of thromboxane B2 and CH3O2H for the trapping of thromboxane A2, together with information on the t1/2 of the intermediate, indicated the presence of an oxetane structure in thromboxane A2. Incubation of arachidonic acid or prostaglandin G2 with washed platelets led to formation of an unstable factor that induced irreversible platelet aggregation and caused release of [14C]serotonin from platelets that had been incubated with [14C]serotonin. The properties and the mode of formation of this factor indicated that it was identical with thromboxane A2. Furthermore, evidence is presented that the more unstable and major component of rabbit aorta contracting substance (RCS) formed in platelets and guinea pig lung is also thromboxane A2.

Low temperature and drought have major influences on plant growth and productivity. To identify barley genes involved in responses to these stresses and to specifically test the hypothesis that the dehydrin (Dhn) multigene family can serve as an indicator of the entire transcriptome response, we investigated the response of barley cv. Morex to: (1) gradual drought over 21 days and (2) low temperature including chilling, freeze-thaw cycles, and deacclimation over 33 days. We found 4,153 genes that responded to at least one component of these two stress regimes, about one fourth of all genes called "present" under any condition. About 44% (1,822 of 4,153) responded specifically to drought, whereas only 3.8% (158 of 4,153) were chilling specific and 2.8% (119 of 4,153) freeze-thaw specific, with 34.1% responsive to freeze-thaw and drought. The intersection between chilling and drought (31.9%) was somewhat smaller than the intersection between freeze-thaw and drought, implying an element of osmotic stress response to freeze-thaw. About 82.4% of the responsive genes were similar to Arabidopsis genes. The expression of 13 barley Dhn genes mirrored the global clustering of all transcripts, with specific combinations of Dhn genes providing an excellent indicator of each stress response. Data from these studies provide a robust reference data set for abiotic stress.

We report mapping of translocation breakpoints using a microarray. We used complex RNA to compare normal hexaploid wheat (17,000 Mb genome) to a ditelosomic stock missing the short arm of chromosome 1B (1BS) and wheat-rye translocations that replace portions of 1BS with rye 1RS. Transcripts detected by a probe set can come from all three Triticeae genomes in ABD hexaploid wheat, and sequences of homoeologous genes on 1AS, 1BS and 1DS often differ from each other. Absence or replacement of 1BS therefore must sometimes result in patterns within a probe set that deviate from hexaploid wheat. We termed these 'high variance probe sets' (HVPs) and examined the extent to which HVPs associated with 1BS aneuploidy are related to rice genes on syntenic rice chromosome 5 short arm (5S). We observed an enrichment of such probe sets to 15-20% of all HVPs, while 1BS represents approximately 2% of the total genome. In total 257 HVPs constitute wheat 1BS markers. Two wheat-rye translocations subdivided 1BS HVPs into three groups, allocating translocation breakpoints to narrow intervals defined by rice 5S coordinates. This approach could be extended to the entire wheat genome or any organism with suitable aneuploid or translocation stocks.

In the latter, we have implemented a synteny viewer between barley andAegilops tauschiito aid in comparative genome analysis.

Previously, we have shown that barley (Hordeum vulgare) plants carrying a mutation preventing chloroplast development are completely frost susceptible as well as impaired in the expression of several cold-regulated genes. Here we investigated the transcriptome of barley albina and xantha mutants and the corresponding wild type to assess the effect of the chloroplast on expression of cold-regulated genes. First, by comparing control wild type against cold-hardened wild-type plants 2,735 probe sets with statistically significant changes (P = 0.05; ≥2-fold change) were identified. Expression of these wild-type cold-regulated genes was then analyzed in control and cold-hardened mutants. Only about 11% of the genes cold regulated in wild type were regulated to a similar extent in all genotypes (chloroplast-independent cold-regulated genes); this class includes many genes known to be under C-repeat binding factor control. C-repeat binding factor genes were also equally induced in mutants and wild-type plants. About 67% of wild-type cold-regulated genes were not regulated by cold in any mutant (chloroplast-dependent cold-regulated genes). We found that the lack of cold regulation in the mutants is due to the presence of signaling pathway(s) normally cold activated in wild type but constitutively active in the mutants, as well as to the disruption of low-temperature signaling pathway(s) due to the absence of active chloroplasts. We also found that photooxidative stress signaling pathway is constitutively active in the mutants. These results demonstrate the major role of the chloroplast in the control of the molecular adaptation to cold.

Motivation: Genomic DNA was hybridized to oligonucleotide microarrays to identify single-feature polymorphisms (SFP) for Arabidopsis, which has a genome size of ∼130 Mb. However, that method does not work well for organisms such as barley, with a much larger 5200 Mb genome. In the present study, we demonstrate SFP detection using a small number of replicate datasets and complex RNA as a surrogate for barley DNA. To identify single probes defining SFPs in the data, we developed a method using robustified projection pursuit (RPP). This method first evaluates, for each probe set, the overall differentiation of signal intensities between two genotypes and then measures the contribution of the individual probes within the probe set to the overall differentiation. Results: RNA from whole seedlings with and without dehydration stress provided 'present' calls for ∼75% of probe sets. Using triplicated data, among the 5% of 'present' probe sets identified as most likely to contain at least one SFP probe, at least 80% are correctly predicted. This was determined by direct sequencing of PCR amplicons derived from barley genomic DNA. Using a 5 percentile cutoff, we defined 2007 SFP probes contained in 1684 probe sets by combining three parental genotype comparisons: Steptoe versus Morex, Morex versus Barke and Oregon Wolfe Barley Dominant versus Recessive. Near-infrared reflectance (NIR) spectroscopy in the spectral range of 1000–2500 nm, was measured directly on brine from barrel salted herring, to investigate the potential of NIR as a fast method to determine the protein content. A principal component analysis performed on the NIR spectra shows two groups, separating the first 100 days of storage from the storage time exceeding 100 days. A partial least-squares regression model between selected regions of the NIR spectra and the protein content yields a correlation coefficient of 0.93 and a prediction error (RMSECV) of 0.25 g/100 g. The results clearly indicate that NIR spectroscopy has a potential as a fast and noninvasive method for assessing the protein content in brine from barrel salted herring, which again may be used as an indicator for the ripening quality of barrel salted herring.

Temperature-programmed desorption (TPD) of self-assembled monolayers (SAMs) on gold is investigated by using in parallel mass spectrometry (MS) and infrared reflection−absorption spectroscopy (IRAS). Monolayers formed by HS(CH2)n−OH (n = 18, 22) and HS(CH2)15−CONH−(CH2CH2O)−H (EG1) are compared to reveal the influence of specifically introduced hydrogen-bonding groups on their thermal stability. The overall desorption process of the above molecules is found to occur in two main steps; a disordering of the alkyl chains followed by a complex series of decomposition/desorption reactions. The final step of the process involves desorption of sulfur from different chemisorption states. The amide-group-containing SAM, which is stabilized by lateral hydrogen bonds, displays a substantial delay of the alkyl chain disordering by about 50 K, as compared to the linear chain alcohols HS(CH2)n−OH. Highlight text A thorough study of starch biosynthesis and deposition in a non-domesticated wild grass was performed using Brachypodium distachyon as a model., Brachypodium distachyon is a non-domesticated cereal. Nonetheless, Brachypodium was recently introduced as a model plant for temperate cereals. This study compares grain starch metabolism in Brachypodium and barley (Hordeum vulgare). In Brachypodium, we identified and annotated 28 genes involved in starch metabolism and identified important motifs including transit peptides and putative carbohydrate-binding modules (CBMs) of the families CBM20, CBM45, CBM48, and CBM53. Starch content was markedly lower in Brachypodium grains (12%) compared to barley grains (47%). Brachypodium starch granules were doughnut shaped and bimodally distributed into distinct small B-type (2.5–10 µm) and very small C-type (0.5–2.5 µm) granules. Large A-type granules, typical of cereals, were absent. Starch-bound phosphate, important for starch degradation, was 2-fold lower in Brachypodium compared with barley indicating different requirements for starch mobilization. The amylopectin branch profiles were similar and the amylose content was only slightly higher compared with barley cv. Golden Promise. The crystallinity of Brachypodium starch granules was low (10%) compared to barley (20%) as determined by wide-angle X-ray scattering (WAXS) and molecular disorder was confirmed by differential scanning calorimetry (DSC). The expression profiles in grain for most genes were distinctly different for Brachypodium compared to barley, typically showing earlier decline during the course of development, which can explain the low starch content and differences in starch molecular structure and granule characteristics. High transitory starch levels were observed in leaves of Brachypodium (2.8% after 14h of light) compared to barley (1.9% after 14h of light). The data suggest important pre-domesticated features of cereals.

A strategy for the synthesis of a series of closely related oligo(ethylene glycol)-terminated alkanethiol amides (principally HS(CH(2))(m)CONH(CH(2)CH(2)O)(n)H; m = 2, 5, 11, 15, n = 1, 2, 4, 6, 8, 10, 12) and analogous esters has been developed. These compounds were made to study the structure and stability of self-assembled monolayers (SAMs) on gold in the prospect of designing new biosensing interfaces. For this purpose, monodisperse heterofunctional oligo(ethylene glycols) with up to 12 units were prepared. Selective monoacylation of the symmetrical tetra- and hexa(ethylene glycol) diols as their mesylates with the use of silver(I) oxide was performed. The synthetic approach was based on carbodiimide couplings of various oligo(ethylene glycol) derivatives to omega-(acetylthio) carboxylic acids via a terminal amino or hydroxyl function. SAM structures on gold were studied with respect to thickness, wettability (water contact angles approximately 30 degrees ), and conformation. A good fit was obtained for the relation between monolayer thickness (d) and the number of units in the oligo(ethylene glycol) chain (n): d = 2.8n + 21.8 (A). Interestingly, the corresponding infrared spectroscopy analysis showed a dramatic change in conformation of the oligomeric chains from all-trans (n = 4) to helical (nor = 6) conformation. A crystalline helical structure was observed in the SAMs for n6.

A comparative study of the self-assembly and phase behavior of seven different oligo(ethylene glycol) (OEG)-terminated alkanethiols on polycrystalline gold surfaces is presented. The general structure of the compounds is HS(CH2)m-X-EGn, where m = 11, 15; n = 2, 4, 6, and the linkages X are amide (−CONH−), ester (−COO−), or ether (−O−) groups. The amide and ester groups give rise to the intermolecular hydrogen bonding and dipole−dipole interactions, respectively, whereas the ether lacks specific interactions. The results from contact angle goniometry, null ellipsometry, and infrared reflection−absorption spectroscopy (IRAS) indicate that the intermolecular interactions can be partly used to control the conformation and order of the OEG portion of the self-assembled monolayers (SAMs). A measurement of direct photon production in {sup 208}Pb+ {sup 208}Pb collisions at 158A GeV has been carried out in the CERN WA98 experiment. The invariant yield of direct photons in central collisions is extracted as a function of transverse momentum in the interval 0.5 1.5 GeV/c . The result constitutes the first observation of direct photons in ultrarelativistic heavy-ion collisions. It could be significant for diagnosis of quark-gluon-plasma formation.

Three-particle correlations have been measured for identified {pi}{sup -} from central 158A GeV Pb+Pb collisions by the WA98 experiment at CERN. A substantial contribution of the genuine three-body correlation has been found as expected for a mainly chaotic and symmetric source.

This letter explores the phase behavior of oligo(ethylene glycol) self-assembled monolayers using temperature-programmed infrared reflection absorption spectroscopy. The monolayers are formed by self-assembly of hexa(ethylene glycol) (EG6) and tetra(ethylene glycol) (EG4)-terminated and amide group containing alkanethiols on polycrystalline gold. The ethylene glycol portions of the two monolayers are known to exist in two different conformations at room temperature: EG6 in helical and EG4 in all-trans (zigzag). The helical phase of the EG6 gradually diminishes upon increasing the temperature and a pronounced conformational transition occurs around 60 °C, leading to a rapidly increasing population of all-trans conformers along the EG6 chain. The EG4 SAM exhibits a much simpler phase behavior. The oligomer conformation is marginally affected upon increasing the temperature to 75 °C, displaying the dominating all-trans phase, which possibly coexists with a small fraction of gauche-rich (disordered) regions....

The Delta(++)-resonance production in central 158 A GeV Pb-208 + Pb-208 collisions at the CERN SPS has been studied. The Delta(++) production was estimated from the invariant mass spectrum of p pi(+)-pairs by subtracting a mixed event background. The measured Delta(++) abundance is compared with the results from other experiments at lower energies, and with a model calculation assuming thermal and chemical equilibrium. An event-by-event analysis of the azimuthal angular correlation with respect to the reaction plane has been carried out for K+ and pi(+) emission near mid-rapidity in 158 A . GeV Pb + Pb collisions. In semi-central collisions, K+ mesons are found to be preferentially emitted out of the reaction plane, while pi(+) mesons are emitted in the reaction plane. The results suggest that the kaon emission is influenced by in-medium potential effects in addition to collective flow effects. (C) 1999 Published by Elsevier Science B.V. All rights reserved.

Neutral pion production in central 158A GeV [sup 208]Pb+ [sup 208] Pb collisions has been studied in the WA98 experiment at the CERN Super Proton Synchrotron. The [pi][sup 0] transverse mass spectrum has been analyzed in terms of a thermal model with hydrodynamic expansion. The high accuracy and large kinematic coverage of the measurement allow one to limit previously noted ambiguities in the extracted freeze-out parameters. The experimental data on the interactions of 10.6A GeV gold nuclei in nuclear emulsions are analyzed with particular emphasis of target separation interactions and study of critical exponents. Charged fragment moments, conditional moments as well as two and three – body asymmetries of the fast moving projectile particles are determined in terms of the total charge remaining bound in the multiply charged projectile fragments. Some differences in the average yields of helium nuclei and heavier fragments are observed, which may be attributed to a target effect. However, two and three-body asymmetries and conditional moments indicate that the breakup mechanism of the projectile seems to be independent of target mass. We looked for evidence of critical point observable in finite nuclei by study the resulting charged fragments distributions. We have obtained the values for the critical exponents γ, β and τ and compare our results with those at lower energy experiment (1.0A GeV data). The values suggest that a phase transition like behavior, is observed.

The production of neutral pions in 158A GeV {sup 208}Pb+ {sup 208}Pb collisions has been studied in the WA98 experiment at the CERN Super Proton Synchrotron (SPS). Transverse momentum spectra are studied for the range 0.3{le}m{sub T}{minus}m{sub 0}{le}4.0 GeV /c . The results for central collisions are compared to various models. The centrality dependence of the neutral pion spectral shape and yield is investigated. An invariance of the spectral shape and a simple scaling of the yield with the number of participating nucleons is observed for centralities with greater than about 30thinspthinspparticipating nucleons. The restoration of chiral symmetry and its subsequent breaking through a phase transition has been predicted to create regions of Disoriented Chiral Condensates (DCC). This phenomenon has been predicted to cause anomalous fluctuations in the relative production of charged and neutral pions in high-energy hadronic and nuclear collisions. The WA98 experiment has been used to measure charged and photon multiplicities in the central region of 158 AGeV Pb+Pb collisions at the CERN SPS. In a sample of 212646 events, no clear DCC signal can be distinguished. Feeding clofibrate to rats and mice results in a strong induction of acyl-CoA thioesterase activity in the liver that is mainly due to increases in the enzyme activities in mitochondria and cytosol. The cytosolic acyl-CoA thioesterase protein of about 40 kDa, referred to as CTE-I, is strongly induced by the treatment. We report here the molecular cloning of the cDNA corresponding to the rat and mouse enzymes, and the further characterization of the mouse CTE-I by recombinant expression in bacteria and regulation of expression of the enzyme. The cDNAs corresponding to the rat and mouse enzymes contained open reading frames encoding proteins of 419 amino acids with calculated molecular masses of 45938 Da and 46135 Da, respectively. Sequence analysis revealed an active site serine consensus sequence commonly found in lipases and carboxylesterases. Recombinant expression of the mouse CTE-I cDNA in Escherichia coli resulted in production of immunoreactive protein that was mainly active with long-chain acyl-CoAs. Northern blot analysis showed that the full-length CTE-I cDNA probe hybridized to two major transcripts corresponding to CTE-I and MTE-I (mitochondrial acyl-CoA thioesterase I), respectively. The expression of both mRNA species was found to be highly regulated. As expected, both CTE-I and MTE-I were strongly upregulated (> 50-fold) by clofibrate treatment. Interestingly, fasting for 48 h resulted in a similar magnitude of induction as two days of clofibrate feeding. In addition, feeding a fat-free diet resulted in down-regulation of CTE-I mRNA. CTE-I mRNA was strongly expressed in kidney and brown adipose tissue and MTE-I mRNA was expressed mainly in brown adipose tissue and heart but was also expressed in kidney and white adipose tissue. Dietary regulation and tissue-specific expression suggest that CTE-I and MTE-I play important roles in lipid metabolism.

The first evidence of directed flow at 158 A GeV Pb + Pb collisions is observed using the Plastic Ball detector in WA98 experiment. The azimuthal angle of the total transverse energy, Phi(o), is determined for each event using the observed protons in the Plastic Ball. A clear azimuthal asymmetry of fragments is observed with respect to Phi(o). The degree of azimuthal asymmetry is found to be largest in mid-central collisions, which is consistent with the picture of the collective flow.The p(t) spectra of particles measured in the mid-rapidity spectrometer has been studied in conjunction with the Plastic Ball, and different p(t) slopes with respect to Phi(o) for proton and pi(+) are observed.

The problem of fatigue life predictions for variable amplitude loading is considered. A prediction model is proposed that is based on a level crossing count of the effective load, where the effective load is defined according to the crack closure concept. The proposed model is applied to block load sequences presented in the literature and compared to experimental data and other prediction methods. It can be concluded that, provided that the crack opening level can be determined, the model gives reasonable predictions.

Existing design rules for tank vehicles have proved insufficient, because vibrations often cause fatigue cracks. Measurements have been performed on the tank to provide a picture of the influence of different road types and filling ratios. Shock response spectrum analysis (SRS) was used to obtain a measure of single-dynamic events. To get a basis for dimensioning against fatigue a calculation of fatigue-damage response spectra (FDRS) has been performed. This relates the vibrations in the tank to the risk of fatigue damage. In lieu of cycle counting by rain-flow count techniques, which has certain disadvantages, a new model, the so called HdM model, for fatigue-life assessment based on level crossing has been used. Further development of the results of the analysis can be used to improve design criteria in transport tank regulations, as well as to determine optimum inspection intervals for, in particular, tanks for hazardous materials.

In this letter the distribution of slow target associated particles emitted in Au + Emulsion interactions at 11.6 A GeV/c is studied, The three models RQMD, FRITIOF and VENUS are used for comparisons and especially their treatment of rescattering is investigated.

Charged particle pseudorapidity density distributions in Au induced reactions in nuclear emulsion at 10.7 A GeV have been measured. In peripheral events the shower particle distribution exhibits a two peak structure while in central collisions it has a Gaussian shape. The spectator protons appear in the second peak in the pseudorapidity spectra and exhibits a broader momentum distribution than expected from Fermi motion. The produced particle yield in central collisions is not substantially different from phenomenological descriptions based on data from O and S induced reactions.

We have previously reported that long chain acyl-CoA thioesterase activity was induced about 10-fold in rat liver mitochondria, when treating rats with the peroxisome proliferator di(2-ethylhexyl)phthalate (Wilcke M., and Alexson S. E. H (1994) Eur. J. Biochem. 222, 803-811). Here we have characterized two enzymes which are responsible for the majority of long chain acyl-CoA thioesterase activity in mitochondria from animals treated with peroxisome proliferators. A 40-kDa enzyme was purified and characterized as a very long chain acyl-CoA thioesterase (MTE-I). The second enzyme was partially purified and characterized as a long chain acyl-CoA thioesterase (MTE-II). MTE-I was inhibited by p-chloromercuribenzoic acid, which implicates the importance of a cysteine residue in, or close, to the active site. Antibodies against MTE-I demonstrated the presence of immunologically related acyl-CoA thioesterases in peroxisomes and cytosol. High expression of MTE-I was found in liver from peroxisome proliferator treated rats and in heart and brown fat from control and induced rats. Comparison of physical and catalytical characteristics of the enzymes studied here and previously purified acyl-CoA thioesterases suggest that MTE-I and MTE-II are novel enzymes.

The synthesis of four bifunctionalized orthogonally N-protected oligosaccharides derived from lactose and mannose, intended as cross-linking derivatives, is described. The amino sugar at the non-reducing end is derivatized with an N-Boc-protected glycine moiety, and further connected to either a mannose (1→6) disaccharide or (1→3) lactose units (one, two or three) resulting in tri-, penta-, or heptasaccharides. All of the synthesized oligosaccharides have an Nbenzyloxycarbonyl-aminoethyl residue at the reducing end. The fully orthogonal N-Boc/N-Cbz protection group pattern enables further conjugation/derivatization and results in a hydrophilic cross-linking molecule. It was found that the order of the final synthetic steps were crucial to avoid acyl migration. A suitable amide coupling protocol has been applied to introduce the NBoc-protected glycine moiety in alcoholic solvent. The synthesized oligosaccharides will provide a model system to investigate the influence of length, structure and flexibility. The function of the cross-linked substituents thereby provide valuable insights into the role as a spacer molecule.

— A computer program that creates time histories for use in fatigue tests in servo-hydraulic machines is presented. The time histories can be tailored from certain user specifications, namely the irregularity factor and the level-crossing spectrum. The program is based on the theory of stochastic time series, and the formulae needed for the program are derived in the paper. Problems of optimality are treated both with respect to fulfilment of the specifications and with respect to machine performance. Examples from two fatigue-testing projects, where the program has been applied, are presented.
