Abstract

Rapid detection of a biological agent is essential to anticipate a threat to the protection of biodiversity and ecosystems. Our goal is to miniaturize a magnetic pathogen detection system in order to fabricate an efficient and portable system. The detection device is based on flat, multilayer coils associated with microfluidic structures to detect magnetic nanoparticles linked to pathogen agents. One type of immunological diagnosis is based on the measurement of the magnetic sensitivity of magnetic nanoparticles (MNPs), which are markers connected to pathogens. This method of analysis involves the coupling of antibodies or antigen proteins with MNPs. Among the available magnetic techniques, the frequency mixing method has a definite advantage by making it possible to quantify MNPs. An external magnetic field composed of a low- and a high-frequency field is applied to the sample reservoir. Then, the response signal is measured and analyzed. In this paper, magnetic microcoils are implemented on a multilayer Printed Circuit Board (PCB), and a microfluidics microstructure is designed in connection with the planar coils. Simulation software, COMSOL version 5.3, provides an analytical perspective to choose the number of turns in magnetic coils and to understand the effects of changing the shape and dimensions of the microfluidics microstructure.

3. Large reciprocal magneto-optical effect induced by all-dielectric resonant gratings based on a magnetic nanocomposite

Abstract

The beneficial combination of micro- and nano-patterned surfaces with magneto-optical materials was investigated over the recent years. Due to their resonant behavior, these structures are commonly used to enhance the non-reciprocal magneto-optical effects. In this paper, a novel kind of magneto-optical intensity effect is enhanced with an all-dielectric grating patterned on a magnetic nanocomposite layer. This nanocomposite is made of CoFe _2 O _4 nanoparticles (NPs) embedded in a silica matrix by sol–gel technique. The demonstrated magneto-optical intensity effect is reciprocal and it is observed with transverse magnetic field, for both polarization (TE and TM) and small angles of incidence. Such effect is not explained by the classical appearance of off-diagonal elements in the permittivity tensor of the magneto-optical material under magnetic field. However, it can be attributed to a magneto-induced reciprocal modification of the diagonal elements. Furthermore, this effect strongly depends on the NPs orientation inside the magneto-optical film and can originate from the magnetostrictive property of the magnetic CoFe _2 O _4 NPs.

4. High Temperature Continuous Flow Syntheses of Iron Oxide Nanoflowers Using the Polyol Route in a Multi-Parametric Millifluidic Device

Abstract

One of the most versatile routes for the elaboration of nanomaterials in materials science, including the synthesis of magnetic iron oxide nanoclusters, is the high-temperature polyol process. However, despite its versatility, this process still lacks reproducibility and scale-up, in addition to the low yield obtained in final materials. In this work, we demonstrate a home-made multiparametric continuous flow millifluidic system that can operate at high temperatures (up to 400 °C). After optimization, we validate its potential for the production of nanomaterials using the polyol route at 220 °C by elaborating ferrite iron oxide nanoclusters called nanoflowers (CoFe2O4, Fe3O4, MnFe2O4) with well-controlled nanostructure and composition, which are highly demanded due to their physical properties. Moreover, we demonstrate that by using such a continuous process, the chemical yield and reproducibility of the nanoflower synthesis are strongly improved as well as the possibility to produce these nanomaterials on a large scale with quantities up to 45 g per day.

Abstract

A magnetic frequency mixing technique with a set of miniaturized planar coils was investigated for use with a completely integrated Lab-on-Chip (LoC) pathogen sensing system. The system allows the detection and quantification of superparamagnetic beads. Additionally, in terms of magnetic nanoparticle characterization ability, the system can be used for immunoassays using the beads as markers. Analytical calculations and simulations for both excitation and pick-up coils are presented; the goal was to investigate the miniaturization of simple and cost-effective planar spiral coils. Following these calculations, a Printed Circuit Board (PCB) prototype was designed, manufactured, and tested for limit of detection, linear response, and validation of theoretical concepts. Using the magnetic frequency mixing technique, a limit of detection of 15 µg/mL of 20 nm core-sized nanoparticles was achieved without any shielding.

Abstract

The magnetic properties of cobalt ferrites nanoparticles prepared using three different processes (thermal decomposition, polyol and co-precipitation synthesis routes) are investigated by X-ray Absorption Spectroscopy and X-ray Magnetic Circular Dichroism. The repartition of cobalt and iron ions amongst the interstitial sites of the spinel structure is determined and correlated with their magnetic properties. The study reports on the influence of the synthesis method on the crystallographic order within a nanoparticle, and hence, on the magnetic anisotropy.

Abstract

This paper is aimed at investigating the process of photocrosslinking under Deep-UV irradiation of nanocomposite thin films doped with cobalt ferrite magnetic nanoparticles (MNPs). This material is composed of a hybrid sol–gel matrix in which MNP can be introduced with high concentrations up to 20 vol%. Deep-UV (193 nm) is not only interesting for high-resolution patterning but we also show an efficient photopolymerization pathway even in the presence of high concentration of MNPs. In this study, we demonstrate that the photocrosslinking is based on the free radical polymerization of the methacrylate functions of the hybrid precursor. This process is initiated by Titanium-oxo clusters. The impact of the nanoparticles on the photopolymerization kinetic and photopatterning is investigated. We finally show that the photosensitive nanocomposite is suitable to obtain micropatterns with sub-micron resolution, with a simple and versatile process, which opens many opportunities for fabrication of miniaturized magneto-optical devices for photonic applications.

Abstract

Highlights • An algorithm for automatic Covid-19 quantification based on 2D & 3D deep convolutional neural networks is presented. • A Covid-19-specific holistic, highly compact multi-omics signature integrating imaging/clinical/ biological data and associated comorbidities for automatic patient staging is presented and evaluated. • Short and Long-term prognosis for clinical resources optimization offering alternative/complementary means to facilitate triage for Covid-19 • Clinically-relevant quantification and staging tool validated by comparison with clinical experts is reported., Graphical abstract, Coronavirus disease 2019 (COVID-19) emerged in 2019 and disseminated around the world rapidly. Computed tomography (CT) imaging has been proven to be an important tool for screening, disease quantification and staging. The latter is of extreme importance for organizational anticipation (availability of intensive care unit beds, patient management planning) as well as to accelerate drug development through rapid, reproducible and quantified assessment of treatment response. Even if currently there are no specific guidelines for the staging of the patients, CT together with some clinical and biological biomarkers are used. In this study, we collected a multi-center cohort and we investigated the use of medical imaging and artificial intelligence for disease quantification, staging and outcome prediction. Our approach relies on automatic deep learning-based disease quantification using an ensemble of architectures, and a data-driven consensus for the staging and outcome prediction of the patients fusing imaging biomarkers with clinical and biological attributes. Highly promising results on multiple external/independent evaluation cohorts as well as comparisons with expert human readers demonstrate the potentials of our approach.

Purpose The purpose of this study was to determine the prevalence and imaging characteristics of incidentally diagnosed COVID-19 pneumonia on computed tomography (CT). Materials and methods This retrospective study was conducted between March 20th and March 31st, 2020 at Cochin hospital, Paris France. Thoracic CT examinations of all patients referred for another reason than a suspicion of SARS-CoV-2 infection were reviewed. CT images were analyzed by a chest radiologist to confirm the presence of findings consistent with COVID-19 pneumonia and quantify disease extent. Clinical and biological data (C-reactive protein serum level [CRP] and white blood cell count) of patients with CT findings suggestive for COVID-19 pneumonia were retrieved from the electronic medical chart. Results During the study period, among 205 diagnostic CT examinations, six examinations (6/205, 3%) in 6 different patients (4 men, 2 women; median age, 57 years) revealed images highly suggestive of COVID-19 pneumonia. The final diagnosis was confirmed by RT-PCR. Three inpatients were suspected of extra thoracic infection whereas three outpatients were either fully asymptomatic or presented with fatigue only. All had increased CRP serum level and lymphopenia. Disease extent on CT was mild to moderate in 5/6 patients (83%) and severe in 1/6 patient (17%). Conclusion Cumulative incidence of fortuitous diagnosis if COVID-19 pneumonia did not exceed 3% during the highest pandemic phase and was predominantly associated with limited lung involvement.

Abstract

International audience; Fluids responding to magnetic fields (ferrofluids) offer a scene with no equivalent in nature to explore long-range magnetic dipole interactions. Here, we studied the very original class of binary ferrofluids, embedding soft and hard ferrimagnetic nanoparticles. We used a combination of x-ray magnetic spectroscopy measurements supported by multi-scale experimental techniques and Monte-Carlo simulations to unveil the origin of the emergent macroscopic magnetic properties of the binary mixture. We found that the association of soft and hard magnetic nanoparticles in the fluid has a considerable influence on their inherent magnetic properties. While the ferrofluid remains in a single phase, magnetic interactions at the nanoscale between both types of particles induce a modification of their respective coercive fields. By connecting the microscopic properties of binary ferrofluids containing small particles, our findings lay the groundwork for the manipulation of magnetic interactions between particles at the nanometer scale in magnetic liquids.

Abstract

Improving screening, discovering therapies, developing a vaccine and performing staging and prognosis are decisive steps in addressing the COVID-19 pandemic. Staging and prognosis are especially crucial for organizational anticipation (intensive-care bed availability, patient management planning) and accelerating drug development; through rapid, reproducible and quantified response-to-treatment assessment. In this letter, we report on an artificial intelligence solution for performing automatic staging and prognosis based on imaging, clinical, comorbidities and biological data. This approach relies on automatic computed tomography (CT)-based disease quantification using deep learning, robust data-driven identification of physiologically-inspired COVID-19 holistic patient profiling, and strong, reproducible staging/outcome prediction with good generalization properties using an ensemble of consensus methods. Highly promising results on multiple independent external evaluation cohorts along with comparisons with expert human readers demonstrate the potentials of our approach. The developed solution offers perspectives for optimal patient management, given the shortage of intensive care beds and ventilators1, 2, along with means to assess patient response to treatment.

Abstract

A significant enhancement of the longitudinal magneto-optical effect is demonstrated numerically and experimentally in transmission, and for small angles of incidence, through a subwavelength resonant structure consisting of a dielectric grating on top of a magneto-optical waveguide. The enhanced polarization rotation is associated with a high transmittance. These low footprint devices may thus be suitable for applications like magnetic field sensors or in non-destructive testing.

Abstract

The enhancement of magneto-optical (MO) effects in planar devices is commonly obtained through the combination of a plasmonic resonance and a MO material, or through the microstructuration of such materials. However, the devices often suffer from weak optical signal, or require fastidious lithography processes. We present a much simpler device made of a photoresist 1D grating processed on a MO composite planar waveguide. This latter is formed by magnetic nanoparticles embedded in a silica matrix. Such all-dielectric device produces much higher Q-factor resonances, which finally result in giant MO intensity effects. Their magnitude combined with the ability of the device to be processed on large scale and various substrates, are very promising for a wide range of sensing applications.

Abstract

We report on the design, fabrication, and characterization of an all-dielectric one-dimensional (1D) resonant device formed by a silicon nitride grating impregnated by a low-index magneto-optical silica-type matrix. This impregnation is realized through the dipping of the 966 nm periodic template in a sol–gel solution previously doped with CoFe2O4 nanoparticles, and able to fill the grating slits. By a proper adjustment of the geometrical parameters of such a photonic crystal membrane, simultaneous excitation of transverse electric (TE) and transverse magnetic (TM) polarization resonances is nearly achieved at 1570 nm. This TE/TM phase-matching situation leads to a fivefold enhancement of the Faraday effect in the resonance area with an increased merit factor of 0.32°. Moreover, the device demonstrates its ability to enhance longitudinal and transverse Kerr effects for the other directions of the applied magnetic field. Taking benefits from the ability of the nanocomposite material to be processed on photonic platforms, and despite its quite low magneto-optical activity compared to classical magnetic materials, this work proves that an all-dielectric 1D device can produce a high magneto-optical sensitivity to every magnetic field directions.

Abstract

Introduction Covid-19 pneumonia CT extent correlates well with outcome including mortality. However, CT is not widely available in many countries. This study aimed to explore the relationship between Covid-19 pneumonia CT extent and blood tests variations. The objective was to determine for the biological variables correlating with disease severity the cut-off values showing the best performance to predict the parenchymal extent of the pneumonia. Methods Bivariate correlations were calculated between biological variables and grade of disease extent on CT. Receiving Operating Characteristic curve analysis determined the best cutoffs for the strongest correlated biological variables. The performance of these variables to predict mild (50% of parenchyma involved) was evaluated. Results Correlations between biological variables and disease extent was evaluated in 168 patients included in this study. LDH, lymphocyte count and CRP showed the strongest correlations (with 0.67, −0.41 and 0.52 correlation coefficient, respectively). Patients were split into a training and a validation cohort according to their centers. If one variable was above/below the following cut-offs, LDH>380, CRP>80 or lymphocyte count, Highlights • Patients with mild respiratory symptoms are not necessarily referred for CT when they are diagnosed with SARS-CoV2 infection. • Previous studies demonstrated than extent of Covid-19 pneumonia on CT is related to patients' outcome. • LDH combined with CRP and lymphocyte count, help predicting parenchymal extent of the pneumonia when CT-scan is not available.

Abstract

International audience; Magnetic anisotropies and crystallographic structures of iron-cobalt nanospinels obtained with the co-precipitation synthesis process are revealed using X-ray Absorption Spectroscopy and X-ray Magnetic Circular Dichroism. The chemical process allows to obtain nanoparticles of various sizes and chemical composition, but it is the site symmetry environment of Co2+ that is found to be the crucial parameter that governs the magnetic anisotropies of the nanospinels. The distribution of Co2+ among the crystallographic sites of the structure is directly linked to the temperature of the synthesis process. In parallel, the results also revealed that a superficial rich shell of iron is formed for the iron-cobalt nanospinels stable in acidic medium leading to chemically inhomogeneous nanoparticles.

Abstract

Magnetic hyperthermia (MHT) and photothermal therapy (PTT) are emergent state‐of‐the‐art modalities for thermal treatment of cancer. While their mechanisms of action have distinct physical bases, both approaches rely on nanoparticle‐mediated remote onset of thermotherapy. Yet, are the two heating techniques interchangeable? Here, the heating obtained either with MHT or with PTT is compared. The heating is assessed in distinct environments and involves a set of nanomaterials differing in shape (spheres, cubes, stars, shells, and rods) as well as in composition (maghemite, magnetite, cobalt ferrite, and gold). The nanoparticle's heating efficacy in an aqueous medium is first evaluated. Subsequently, the heating efficiency within the cellular environment, where intracellular processing markedly decreases MHT, is compared. Conversely, endosomal sequestration could have a positive effect on PTT. Finally, iron oxide nanocubes and gold nanostars are compared in MHT and PTT in vivo within the heterogeneous intratumoral environment. Overall, two distinct therapeutic approaches, related to high dosage allowing MHT and low dosage associated with PTT, are identified. It is also demonstrated that PTT mediated by magnetic nanoparticles has an efficacy that is comparable to that of plasmonic nanoparticles, but only at significant nanoparticle dosages. At low concentrations, only plasmonic nanoparticles can deliver a therapeutic heating.

Abstract

International audience; Multifunctional nanoparticles such as magneto‐plasmonic nanohybrids are rising theranostic agents. However, little is yet known of their fate within the cellular environment. In order to reach an understanding of their biotransformations, reliable metrics for tracking and quantification of such materials properties during their intracellular journey are needed. In this study, their long‐term (one month) intracellular fate is followed within stem‐cell spheroids used as tissue replicas. A set of magnetic (magnetization) and thermal (magnetic hyperthermia, photothermia) metrics is implemented to provide reliable insightsinto the intracellular status. It shows that biodegradation is modulated by the morphology and thickness of the gold shell. First a massive dissolution of the iron oxide core (nanoflower‐like) is observed, starting with dissociation of the multigrain structure. Second, it is demonstrated that an uninterrupted gold shell can preserve the magnetic core and properties (particularly magnetic hyperthermia). In addition to the magnetic and thermal metrics, intracellular high‐resolution chemical nanocartography evidences the gradual degradation of the magnetic cores. It also shows different transformation scenarios, from the release of small gold seeds when the magnetic core is dissolved (interesting for long‐term elimination) to the protection of the magnetic core (interesting for long‐term therapeutic applicability).

Abstract

Metallic nanoparticles have been increasingly suggested as prospective therapeutic nanoplatforms, yet their long-term fate and cellular processing in the body is poorly understood. Here we examined the role of an endogenous iron storage protein – namely the ferritin – in the remediation of biodegradable cobalt ferrite magnetic nanoparticles. Structural and elemental analysis of ferritins close to exogenous nanoparticles within spleens and livers of mice injected in vivo with cobalt ferrite nanoparticles, suggests the intracellular transfer of degradation-derived cobalt and iron, entrapped within endogenous protein cages. In addition, the capacity of ferritin cages to accommodate and store the degradation products of cobalt ferrite nanoparticles was investigated in vitro in the acidic environment mimicking the physiological conditions that are present within the lysosomes. The magnetic, colloidal and structural follow-up of nanoparticles and proteins in the lysosome-like medium confirmed the efficient remediation of nanoparticle-released cobalt and iron ions by ferritins in solution. Metal transfer into ferritins could represent a quintessential process in which biomolecules and homeostasis regulate the local degradation of nanoparticles and recycle their by-products.

Abstract

The nanoscale distribution of magnetic anisotropies was measured in core@shell MnFe$_2$O$_4$@CoFe$_2$O$_4$ 7.0 nm particles using a combination of element selective magnetic spectroscopies with different probing depths. As this picture is not accessible by any other technique, emergent magnetic properties were revealed. The coercive field is not constant in a whole nanospinel. The very thin (0.5 nm) CoFe$_2$O$_4$ hard shell imposes a strong magnetic anisotropy to the otherwise very soft MnFe$_2$O$_4$ core: a large gradient in coercivity was measured inside the MnFe$_2$O$_4$ core with lower values close to the interface region, while the inner core presents a substantial coercive field (0.54 T) and a very high remnant magnetization (90% of the magnetization at saturation)., Comment: 27 pages (including supporting information)

Abstract

International audience; This paper describes the possibility to achieve a TE-TM mode conversion in a magneto-optical hybrid waveguide operating at k¼1550 nm. This hybrid device is made by coating a SiO2/ZrO2 layer doped with magnetic nanoparticles on an ion-exchanged glass waveguide. Soft annealing (90 C) and UV treatment, both compatible with the ion exchange process, have been implemented to finalize the magneto-optical film. Optical characterizations that have been carried out demonstrated the efficiency of these hybrid structures in terms of lateral confinement and mode conversion. Indeed, TE to TM mode conversion has been observed when a longitudinal magnetic field is applied to the device. The amount of this conversion is discussed taking into account the distribution of light between the layer and the guide, and the modal birefringence of the structure.

Abstract

International audience; Fluorescent organic nanoparticles (FON) made of small molecules recently arouse considerable interest in biomedical imaging and opto-electronics as potential microlasers or emitting displays.2 Their collective emission largely enhanced with respect to dilute media and their reduced sensitivity to external bleaching and quenching effects make them very attractive when powerful excitation is required as in multiphoton fluorescence spectroscopy. Fluorescence in the solid state is however facing two deleterious effects: competitive non-radiative quenching processes caused by strong intermolecular aggregation, and emission reabsorption owing to the inherent high chromophore density. The main strategies consist in introducing bulky substituents to prevent the formation of non-emissive π−π stacked assemblies as well as elaborating materials with a large optical Stokes shift. Hydrophobic materials are often obtained and considered of little use in bioimaging. We want to report herein the synthesis and the properties of a new family of push-pull amorphous compounds which form strong one-and two-photon emitting nanoparticles that can efficiently be utilized to image lipophilic cell membranes despite their water insolubility.The present series of fluorescent compounds consists in polar triphenylamino derivatives substituted by bulky tert-butylphenyl groups following a Suzuki aromatic cross coupling reaction as a final step. Both the twisted star-shaped triphenylamino-core and peripheral hindered substituents prevent deleterious π−π stacking and ensure high solubility in polar and apolar organic solvents.

The formation of two-dimensional self-assemblies of ordered, packed gold nanoparticles is described. The nanoparticles were coated by a diamide thiol ligand (DASH) that ensured the connection of the nanoparticles through hydrogen bonds. The stability of the colloidal solutions is discussed with respect to the stability of the normal dodecanethiol-coated particles.

Abstract

Ionic magnetic fluid (ferrofluid) is a stable suspension of magnetic nanoparticles in water. Cobalt ferrite nanoparticles are interesting in view of high-density recording storage. The size of the magnetic particles strongly influences the physical properties of the ferrofluids. In this study, we describe the synthesis of ionic magnetic fluid in the presence of tartrate ions. By varying the amount of organic ligands, nanoparticles in a large range of size are obtained: the mean diameter varies from 3 to 10 nm. The effect of tartrate ions on the stability of the ionic magnetic fluid is also studied in relation with the size of the magnetic particles and the amount of adsorbed ligand.

Abstract

An experimental and theoretical study on the Photonic Hall Effect (PHE) in liquid and gelled samples of ferrofluids is presented. The ferrofluids are aqueous colloidal suspensions of Fe(_{2})CoO(_{4}) particles, which can be considered as anisotropic and absorbing Rayleigh scatterers. The PHE is found to be produced by the orientation of the magnetic moments of the particles, as is also the case for the Faraday effect. The dependence of the PHE with respect to the concentration of the scatterers, the magnetic field and the polarization of the incident light is measured in liquid and in gelled samples and is compared to a simple model based on the use of a scattering matrix and the single scattering approximation., Comment: 20 pages, 11 figures, submitted

Abstract

High quality CoFe2O4 nanoparticles were synthesized only in 16 min, continuously in two coupled microreactors. The first microreactor induced the fast homogenization of the reagents mixture at ambient temperature so Fe3+ and Co2+ hydroxides precipitate, while a second microreactor heated at 98 °C allowed the fast aging and the evolution of amorphous hydroxides into faceted and crystalline CoFe2O4.
