The production of indigo, primarily used by the denim industry, increases year by year, and is mainly of synthetic origin. The textile industry, on which its production depends, is responsible for 10% of greenhouse gases and 20% of water pollution. However, the source of this pigment/colorant, mainly based on petrochemistry, remains a key issue today. Extracting indigo from plants is becoming a popular answer and requires an understanding and evaluation of the entire process, from raw material to pigment recovery. In this study, the indigotin precursor, indoxyl, derived from the hydrolysis of O-glycosides biomass extracted in water, was oxidized to obtain the desired pigment. This step is the most sensitive, as variations have been observed during this phase. Consequently, the standardization of the oxidation process was established to determine the extract capacity to consistently produce the blue dye pigment. Partial hydrolysis of the O-glycosides, the indoxyl precursors, was identified as a factor causing this yield variability in the obtained extracts. Once the precursors were fully chemically hydrolyzed, plants harvested during summer and during a freezing period showed a similar capacity to produce indigotin, with values of 412 ± 25 ppm and 379 ± 0 ppm, respectively. This result showed that in freezing conditions, the enzymatic material was not available, resulting in the lack of indigotin formation. To address the use of oxidation in an alkaline medium, a spontaneous oxidation method was proposed. Background and Aim: Rice straw, a widely available agricultural byproduct globally, has significant potential as a basal diet for livestock. The major challenge lies in obtaining high-protein foliage that can be easily extracted using natural water rather than chemical solvents. This study aimed to assess the ability of distilled water to extract protein concentrate from Indigofera leaves (Indigofera zollingeriana Miq.) and to evaluate its effectiveness in enhancing rumen feed fermentation and digestibility in low-quality rice straw basal diets. Materials and Methods: The study was conducted in two experimental series. Experiment 1 was designed to explore the ability of distilled water to extract protein concentrate from fresh and dry Indigofera leaves by comparing it with the 0.1 N NaOH standard solvent. Experiment 2 focused on the in vitro digestibility of protein concentrates extracted from fresh Indigofera leaves based on optimal findings from experiment 1. Five treatments consisting of 0.5% and 1.0% protein concentrate and two extractants (distilled water and 0.1 N NaOH) were used to extract protein from Indigofera leaves. These extracts were then added to rice straw-based diets. Rice straw without supplements was used as a control. The treatments were arranged using a randomized complete design with five replicates. Results: The results of experiment 1 showed that distilled water was superior to 0.1 N NaOH for extracting protein concentrate from fresh Indigofera leaves, as revealed by higher dry matter, protein yield, total amino acids (AA), and total essential AA (EAA) production. For in vitro experiment 2, supplementation with distilled water-extracted protein concentrates successfully increased rumen fermentation and digestibility in rice straw basal diets, as indicated by higher gas production, total volatile fatty acid, and microbial protein levels compared with 0.1 N NaOH. Conclusion: Findings from this study confirm that Indigofera leaf protein concentrate offers a new alternative for enhancing rumen feed fermentation and the digestibility of low-quality rice straw diets. This study implies that it is an easy, costeffective, and environmentally friendly approach, particularly beneficial for smallholders, to extract protein concentrate from fresh Indigofera leaves using distilled water and use it to enhance the quality of rice straw for ruminant feed. The limitation of this study is that the Indigofera supplement was established using in vitro digestibility under controlled laboratory conditions, which does not reflect real rumen conditions. This study investigates the effect of different cutting types and planting media on the growth of Indigofera zollingeriana, a valuable forage crop. The research used a factorial randomized block design (2 x 2) with 40 experimental units. The treatments included two cutting sources--stem cuttings (A1) and shoot cuttings (A2)--and two planting media--natural soil (B1) and natural soil supplemented with liquid organic fertilizer from rabbit urine (B2). Growth parameters such as the number of leaves, plant height, and stem diameter were measured. The results indicate that cutting type significantly affects leaf production, with shoot cuttings (A2) producing a higher number of leaves than stem cuttings (A1) (P<0.05). However, the planting media and its interaction with the cutting type had no significant impact on plant height and stem diameter (P>0.05). These findings suggest that using shoot cuttings could enhance the propagation and growth of Indigofera zollingeriana for sustainable forage production. Indigofera koreana is an endemic species to Korea, yet its phylogenetic position remains unresolved. In this study, we sequenced and analyzed plastid genomes (plastomes) to clarify the phylogenetic standing of I. koreana. We produced next-generation sequencing data for I. koreana and its close relative, Indigofera kirilowii. The plastomes of both species were successfully assembled, measuring 159,517 bp for I. kirilowii and 159,459 bp for I. koreana. Both species shared the same number of 128 intact genes and had identical 35.7% guanine–cytosine content. Phylogenetic analyses utilizing complete plastome and coding sequence–only data, along with 20 other Indigofera plastomes, were conducted to elucidate their evolutionary relationships. Our findings indicate that I. koreana forms a monophyletic group with Indigofera decora, Indigofera carlesii, and I. kirilowii, supported by maximum bootstrap values. Specifically, the phylogenetic tree derived from complete plastome data strongly suggests that I. koreana is most closely related to I. kirilowii. While these results are in line with previously suggested infra-generic classifications, they highlight the need for more comprehensive genomic studies to further unravel the evolutionary dynamics within the Indigofera genus, especially considering the role of polyploidization in species diversification.

Leaf extracts are known to be rich in phytoconstituent biomolecules, making them a valuable source of medicinal compounds. They also serve as both capping and reducing agents in nanoparticle fabrication. A reaction between CuSO4.5H2O aqueous solution and the Indigofera tinctoria leaf extract results in the formation of stable copper nanoparticles. Phytochemical screening of the Indigofera tinctoria leaf extract revealed the presence of various compounds including carbohydrates, terpenoids, phenol, tannins, flavonoids, saponins, and glycosides in the sample. The biosynthesized copper nanoparticles (CuNPs) were subsequently subjected to various forms of analysis. Techniques used included UV-visible (UV-vis) spectroscopy, Fourier transform infrared spectroscopy (FTIR), and scanning electron microscopy. An observable change in the color of solutions from pale to thick brown indicated the formation of CuNPs. Further confirmation came from UV-vis spectroscopy, which established the production of CuNPs at 500 nm. FTIR analysis revealed that the CuNPs were covered by organic residues. Background: Indigofera gerardiana (IG) is traditionally used for its medicinal properties, but scientific validation is limited. This study aimed to evaluate its pharmacological effects, specifically anti-inflammatory, analgesic, and antipyretic activities, using established animal models. Methods: This experimental study was conducted at the Pharmacology Department of Khyber Medical College and University, Peshawar between Feb 2022 and Aug 2023. The n-hexane fraction of IG was tested at concentrations of 100mg/kg, 150mg/kg, and 200mg/kg in in-vivo animal models. Anti-inflammatory effects were assessed with paw edema models in 30 rats, analgesic effects in 30 mice using writhing and hot plate tests, and antipyretic activity in 30 rabbits with yeast-induced fever. Data were analyzed using one-way ANOVA with Dunnett’s post hoc test. Results: The n-hexane fraction of IG showed a dose-dependent anti-inflammatory effect, reducing carrageenan-induced edema by 12.5% and histamine-induced edema by under 15% at 200 mg/kg i.p. after 3 hours. Analgesic activity was also dose-dependent, with pain inhibition in the acetic acid-induced writhing model of 16.9% at 100 mg/kg, 20.76% at 150 mg/kg, and 26.13% at 200 mg/kg. In the hot plate test, maximum pain inhibition occurred at 200 mg/kg after 90 minutes (p < 0.001). However, IG extract did not show significant antipyretic effects compared to paracetamol, which effectively reduced fever at all doses (p < 0.001). Conclusion: The study findings suggest that the n-hexane portion of IG exhibits significant anti-inflammatory and analgesic activities across various animal models while showing no substantial impact on yeast-induced pyrexia.

The present paper deals with a comprehensive enumeration of leguminous taxa found in Birbhum District of West Bengal, India. We recorded 140 species, one subspecies, and two varieties, representing 82 genera from 24 tribes under four subfamilies, viz., Cercidoideae (6 species in 2 genera belonging to 1 tribe), Detarioideae (3 species in 3 genera belonging to 3 tribes), Caesalpinioideae (43 species in 24 genera belonging to 5 tribes), and Papilionoideae (91 taxa including 88 species, 1 subspecies and 2 varieties in 53 genera belonging to 15 tribes). Herbs (59 taxa) were the largest group, followed by trees (39), climbers (23), and shrubs (22). Crotalaria and Indigofera (7 species each) emerged as the dominant genera, followed by Senna (6) and Bauhinia and Senegalia with five species each. In this district, the legumes are represented under 24 tribes, of which the Phaseoleae is the largest comprising 24 taxa, followed by Desmodieae (16) and Cassieae (12). Regarding endemics, Indigofera prostrata exclusive to West Bengal is recorded from Birbhum District, while Grona brachystachya, Hardwickia binata, Pterocarpus marsupium, and P. santalinus are endemic to India. This study was conducted to evaluate the acute dermal and ocular irritant potential of three natural products obtained from medicinal plants Murraya paniculata (PNI), Eucalyptus sp. (PNII), and Indigofera suffruticosa Mill. (PNIII) for topical use in veterinary medicine. For the assessment of acute dermal and ocular irritability, methodologies from the Organization for Economic Cooperation and Development (OECD) were employed through guidelines 404 and 405, respectively, using New Zealand albino rabbits. The evaluated products showed no signs or symptoms of irritation/corrosion on the skin and eyes, exhibiting dermal and ocular irritation indices within the range of classification of non-irritating substances, according to the OECD Harmonized Integrated Classification System. The bioactive extracts of traditional medicinal plants are rich in polyphenols and help to rejuvenate skin. The study was designed to assess the skin rejuvenating effects of a stable cream enriched with 4% I. argentea (IaMe) extract. The quantity of polyphenols by spectrophotometric methods was TPC, 101.55 ± 0.03 mg GAE/g and total flavonoid content; 77.14 ± 0.13 mg QE/g, while HPLC-PDA revealed gallic acid; 4.91, chlorogenic acid 48.12, p-coumaric acid 0.43, and rutin 14.23 µg/g. The significant results of biological activities were observed as DPPH; 81.81% ± 0.05%, tyrosinase; 72% ± 0.23% compared to ascorbic acid (92.43% ± 0.03%), and kojic acid (78.80% ± 0.19%) respectively. Moreover, the promising sun protection effects Sun protection factor of extract (20.53) and formulation (10.59) were observed. The active cream formulation (w/o emulsion) was developed with liquid paraffin, beeswax, IaMe extract, and ABIL EM 90, which was stable for 90 days as shown by various stability parameters. The rheological results demonstrated the active formulation's non-Newtonian and pseudoplastic characteristics and nearly spherical globules by SEM. The IaMe loaded cream was further investigated on human trial subjects for skin rejuvenating effects and visualized in 3D skin images. Herein, the results were significant compared to placebo. IaMe formulation causes a substantial drop in skin melanin from -1.70% (2 weeks) to -10.8% (12 weeks). Furthermore, it showed a significant increase in skin moisture and elasticity index from 7.7% to 39.15% and 2%-30%, respectively. According to the findings, Indigofera argentea extract has promising bioactivities and skin rejuvenating properties, rationalizing the traditional use and encouraging its exploitation for effective and economical cosmeceuticals. Background: Leishmaniasis is highly prevalent worldwide, and while conventional medicine offers numerous treatment methods for cutaneous leishmaniasis, Iranian traditional medicine suggests various remedies. We aimed to evaluate the efficacy of an herbal combination containing Indigofera argentea leaves, Pistacia atlantica resin, and Salvia hispanica seeds in patients with zoonotic cutaneous leishmaniasis. Methods: This study was conducted at the Leishmaniasis Department of Chabahar Health Center in southeastern Iran in 2021. It was a double-blinded randomized clinical trial involving 68 patients enrolled after clinical diagnosis, examination of smear samples using Giemsa staining, and PCR confirmation. The volunteers were randomly divided into intervention and control groups. Both groups received 'Glucantime ® as the primary medication weekly until complete healing or up to 12 weeks. Glucantime ® was administered intralesionally at a dosage of 0.1 cc on the wound's margin, with repeat injections at 1 cm intervals along the wound edge when necessary. In addition to the main drug, the intervention group received the herbal product topically twice daily until wound healing or for up to 4 weeks, while the control group received a placebo in the same manner. Results: 82.35% of patients in the intervention group and 20.58% in the control group achieved complete healing within four weeks. There was a significant difference between the two groups (P < 0.001). Conclusion: The herbal product demonstrated effectiveness in treating patients with zoonotic cutaneous leishmaniasis. Indigo quality is determined by its indigotin content. Another quality indicator is colour. For an evaluation of species, indigo samples from Indigofera tinctoria, Indigofera suffruticosa, Indigofera arrecta, Persicaria tinctoria, Strobilanthes cusia and Wrightia laevis cultivated in Austria and China were visually classified and analysed spectrophotometrically and using a L*a*b* measuring device. In addition to a standardised hot-extraction method without lime, some samples were extracted simulating traditional methods at ambient temperatures using lime. The highest indigotin contents were achieved with Indigofera arrecta (55%, Austria) and Strobilanthes cusia (56%, China). There were no statistically significant differences between the indigo extraction yields of the species cultivated in Austria, but Indigofera arrecta and Persicaria tinctoria had statistically significantly higher indigotin extraction yields than Indigofera tinctoria and Indigofera suffruticosa. From the species extracted in China, Strobilanthes cusia showed higher values in all parameters than Indigofera tinctoria, Indigofera suffruticosa and Wrightia laevis. Compared with the standardised method, the method simulating local practice yielded more indigo but had a lower indigotin content; the indigotin extraction yields did not differ greatly. L*a*b* values enabled precise estimations of the indigotin content, making it an interesting option for quality control, as inexpensive, easy-to-handle L*a*b* measuring instruments have become available. Indigofera jintongpenensis, a new species of the subfamily Papilionoideae of Fabaceae, is described and illustrated from Yunnan, southwest China. The new species is characterised by having a prostrate habit, flexible stems and branches, as well as spreading, sub-basifixed, asymmetrically 2-branched trichomes covering the entire plant, discoid calyx, and racemose inflorescences 6–8-flowered, short, 1–2 (–3.5) cm in length, apparently shorter than the leaf. A distribution map and comparison of morphological diagnostic characters with its morphologically similar species are provided. This research was done as a factorial experiment with 5 replications was carried out. The seeds were primed using putrescine at concentrations of 0 (hydropriming), 0.5, and 1 mM, and control were used as control. Salinity treatment was applied under normal, low, moderate and severe (0, 50, 100, and 150 mM NaCl, respectively) salinity stress. According to the findings, increasing salinity levels reduced germination percentage, (GP), germination rate, (GR), coefficient of velocity of germination, (CVG), seed weight vigour index, (SWVI), by 29, 66, 53 and 25%, respectively and seedling fresh weight, seedling dry weight, roots length and shoot length by 37, 14, 72 and 61%, respectively, while increasing mean germination time (MGT), percentage of dry matter, total soluble sugar (TSS), reactive oxygen species (ROS) and malondialdehyde (MDA) by 112, 53, 57,16 and 182%, respectively. When seeds were primed using putrescine reduced the effects of oxidative stress by reducing ROS and MDA levels by 11 and 47%, respectively compared to control and improved the GP, GR, CVG, SWVI, root length and shoot length by 31, 81, 67, 36, 27, 19%, and decreased MGT by 40%. Although seed priming using 1 mM putrescine was effective in some parameters, there was no significant difference when compared to 0.5 mM putrescine. As a result, seed priming indigo using 0.5 mM putrescine with increasing dry matter by 73% appears to be more practical and economical in terms of mitigating the negative effects of salinity stress. Program Peningkatan Kapasitas Organisasi Mahasiswa (PPK Ormawa) merupakan salah satu kegiatan yang difokuskan pada pengabdian dan pengembangan potensi masyarakat di daerah yang dijadikan sebagai mitra pengabdian selama jangka waktu tertentu secara berkelanjutan. Desa Sokawera, Kecamatan Cilongok, Kabupaten Banyumas, Provinsi Jawa Tengah dipilih menjadi desa sasaran karena memiliki potensi sumberdaya alam dan manusia yang melimpah. Namun potensi tersebut masih kurang dimanfaatkan karena rendahnya minat kaum muda dalam bidang peternakan serta rendahnya pengetahuan peternak dalam pemanfaatan teknologi pada bidang peternakan. PPK Ormawa membentuk Sanggar Ternak Muda “Berdikari” sebagai salah satu upaya untuk mengatasi permasalahan yang ada yakni menjadi tempat atau wadah bagi pemuda dan masyarakat Desa Sokawera untuk belajar mengenai dunia peternakan sehingga terjadi peningkatan sumber daya manusia di Desa Sokawera. Metode yang dilakukan dengan melakukan diskusi, pelatihan dengan sistem ceramah dan praktik, serta melakukan pre-test dan post-test sebagai parameter keberhasilan kuantitatif. Sebanyak 20 orang peserta yang mengikuti pelatihan dijadikan sampel dalam data dengan hasil yang berbeda. Pengujian dilakukan dengan melakukan uji banding ganda atau two sample t-test menghasilkan temuan bahwa analisis perbandingan antara rata-rata data pre-test dan post-test yang mewakilkan pemahaman pengurus sanggar terhadap materi pelatihan adalah berbeda secara signifikan. Kemajuan ini mampu meningkatkan kesejahteraan masyarakat Desa Sokawera.

Blue denim, a billion-dollar industry, is currently dyed with indigo in an unsustainable process requiring harsh reducing and alkaline chemicals. Forming indigo directly in the yarn through indican (indoxyl-β-glucoside) is a promising alternative route with mild conditions. Indican eliminates the requirement for reducing agent while still ending as indigo, the only known molecule yielding the unique hue of blue denim. However, a bulk source of indican is missing. Here, we employ enzyme and process engineering guided by techno-economic analyses to develop an economically viable drop-in indican synthesis technology. Rational engineering of PtUGT1, a glycosyltransferase from the indigo plant, alleviated the severe substrate inactivation observed with the wildtype enzyme at the titers needed for bulk production. We further describe a mild, light-driven dyeing process. Finally, we conduct techno-economic, social sustainability, and comparative life-cycle assessments. These indicate that the presented technologies have the potential to significantly reduce environmental impacts from blue denim dyeing with only a modest cost increase. Conventional blue denim dyeing has both environmental and health-related consequences. Climate change and other extinction facilitators have caused significant shifts in the distribution patterns of many species during the past few decades. Restoring and protecting lesser-known species may be more challenging without adequate biogeographical information. To address this knowledge gap, the current study set out to determine the global spatial distribution patterns of Indigofera oblongifolia (Forssk) a relatively lesser-known leguminous species. This was accomplished by utilizing three distinct bioclimatic temporal frames (current, 2050, and 2070) and four greenhouse gas scenarios (RCPs 2.6, 4.5, 6.0, and 8.5), in addition to non-climatic predictors such as global livestock population, human modification of terrestrial ecosystems, and global fertilizers application (nitrogen and phosphorus). Furthermore, we evaluate the degree of indigenousness using the geographical area, habitat suitability categories, and number of polygons. This research reveals that climatic predictors outperform non-climatic predictors in terms of improving model quality. Precipitation Seasonality is one of the most important factors influencing this species' optimum habitat suitability up to 150 mm for the current, 2050 RCP 8.5 and 2070-RCPs 2.6, 4.5, and 8.5. Our ellipsoid niche modelling extends the range of precipitation during the wettest quarter and maximum temperature during the warmest month to 637 mm and 26.5–31.80 degrees Celsius, respectively. India has a higher indigenous score in the optimal class than the African region. This findings suggest that the species in question tends to occupy contiguous regions in Africa, while in India, it is dispersed into several smaller meta-populations. [ABSTRACT FROM AUTHOR]

Background: Multidrug-resistant bacterial strains cause several serious infections that can be fatal, such as Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumonia, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacteriaceae (often referred to as ESKAPE pathogens). Since ancient times, several indigenous medical systems in India have utilized diverse medicinal plants (approximately 80,000 species) as conventional treatments for a variety of illnesses. A member of the Fabaceae family, also referred to as "Himalayan indigo," Indigofera heterantha Wall, is well known for its therapeutic properties. Methods: The present study investigated the antibacterial, antifungal and antihelmintic properties of the roots, bark, leaves, and flowers of I. heterantha from the Kashmir Himalayas. The effectiveness of the extracts against bacteria, fungi, and earthworms. Three of the tested organisms for bacteria were ESKAPE pathogens, as they are responsible for creating fatal bacterial infections. The antifungal potency of I. heterantha aqueous and methanolic extracts was evaluated using the Agar Well Diffusion Assay. The antihelmintic activity was carried out on an adult Pheretima posthuma Indian earth worm, which shares physiological and anatomical similarities with human intestinal roundworm parasites. Results: The methanolic extracts of root and bark have shown prominent activity against all bacterial strains, whereas aqueous extracts of flower, root, and leaves have shown promising activity against Staphylococcus aureus. The aqueous extract demonstrated good activity against S. cerevisiae at a concentration of 200 mg/ml with a zone of inhibition of 16 mm, while the methanolic extract displayed comparable activity against the fungal strains. The remaining two strains, P. crysogenum and A. fumigatus, were only moderately active in response to the extracts. All the extracts have shown anthelmintic activity except aqueous flower. Conclusion: These results will pave the way for the bioassay-guided isolation of bioactive constituents that may act as hits for further development as potential antibacterial agents against drug-resistant microbial and helminthic infections. The leaf extract of Indigofera linnaei Ali, an Indian medicinal plant, was utilized in the synthesis of copper oxide nanoparticles (CuO-NPs). Green chemistry is a safe and cost-effective method for the synthesis of nanoparticles using plant extracts. The synthesis of CuO NPs was confirmed using ultraviolet–visible (UV-visible) spectrum λ-max data with two peaks at 269 and 337 nm. Different functional groups were identified using Fourier-transform infrared spectroscopy (FT-IR). X-ray diffraction (XRD) was used to confirm the crystalline structure of the CuO-nanoparticles. Scanning electron microscopy (SEM) and energy dispersive X-ray (EDX) analyses were performed to examine the surface morphology and elemental composition of the biosynthesized CuO-NPs. Furthermore, the synthesized CuO-NPs exhibited antibacterial activity against Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, and Enterococcus faecalis. Additionally, they exhibited a good insecticidal effect on Culex quinquefasciatus larvae, with low LC50 55.716 µg/mL and LC90 123.657 µg/mL values. The CuO-NPs inhibited human breast cancer cells in a concentration-dependent manner, with an IC50 value of 63.13 µg/mL. [ABSTRACT FROM AUTHOR]

The waste from the use of synthetic dye in the textile industry leads to environmental concerns, causing serious human and animal health issues. Research on waste recycling efforts has been made. However, this process is still challenging and expensive. In this context, the development of natural dye and optimization of its extraction process have attracted attention recent attention. Indigofera plants are found in several genera and are excellent sources of dye, known as indigo dye. There are three main genera of indigo, Isatis spp., Persicaria tinctoria (Ait) Spach (Polygonum tinctorium Ait), and Indigofera spp. that are used to produce industrial Indigo. These can be used as a dye by clothing manufacturers. In addition to being a potential commercial crop in the near future, indigo is a legume and can be used to further diversify the existing cropping systems in the U.S. In the U.S. Southern Great Plains, this crop could be an alternative crop to diversify and sustain the cotton-based cropping system that is predominant in the region. However, little research has been conducted regarding the agronomics and genetics of this crop. The aim of the present study is to investigate the ameliorative potential of the aqueous extract of Indigofera tinctoria (IT) in aging-induced inflammation and its associated cardiac hypertrophy and fibrosis. Young (3-month-old) and aged (24–26-month-old) male Wistar albino rats were grouped into young control, aged control, aged + IT, and young + IT. The animals in the supplementary groups received 200 mg/kg BWT of aqueous extract of IT orally once a day for 21 days. Aged animals showed prolonged QT interval and increased weight and volume of the heart with a thickening ventricular wall. Infiltration of leukocytes and increased cardiomyocyte diameter and decreased numerical density along with cardiomyocyte apoptosis and increased collagen accumulation were also seen in aged myocardium when compared to the young. The expression profile of various pro-inflammatory cytokines such as IL-6, IL-1β, TNF-α, NFκB, and iNOS was increased with a concomitant reduction in IL-10 expression in the aged compared to the young. In addition, a marked increase in ROS generation, TGF-β, and α-SMA levels is evident in the aged myocardium. These pathological changes were greatly reversed in aged animals supplemented with IT. Furthermore, the aged + IT group showed repression of pro-inflammatory markers with a subsequent increase in IL-10 expression. Contrarily, no marked changes were observed between young and young + IT groups. Taken together, it is concluded that the aqueous extract of Indigofera tinctoria suppresses cardiac fibrosis and hypertrophy by repressing the inflammation and its associated activation of TGFβ and myofibroblast conversion. BackgroundIndigo naturalis (IN) is an herbal medicine that has been used for ulcerative colitis with an unclear mechanism of action. Indigo and indirubin, its main constituents, are ligands of the aryl hydrocarbon receptor (AhR). We assessed the safety, efficacy, and colon AhR activity of IN given orally to patients with treatment-refractory ulcerative colitis. The role of AhR in IN benefit was further evaluated with an AhR antagonist in a murine colitis model.MethodsThis open-label, dose-escalation study sequentially treated 11 patients with ulcerative colitis with either IN 500 mg/day or 1.5 g/day for 8 weeks, followed by a 4-week non-treatment period. The primary efficacy endpoint was clinical response at week 8, assessed by total Mayo score. Secondary endpoints included clinical remission, Ulcerative Colitis Endoscopic Index of Severity, quality of life, and colon AhR activity measured by cytochrome P450 1A1 (CYP1A1) RNA expression.ResultsTen of 11 (91%) patients, including 8/9 (89%) with moderate-to-severe disease, achieved a clinical response. Among these 10 patients, all had failed treatment with 5-aminosalicylic acid, 8 patients with a tumour necrosis factor (TNF)-alpha inhibitor, and 6 patients with TNF-alpha inhibitor and vedolizumab. Five patients were corticosteroid dependent. Clinical response was observed in all five patients who had been recommended for colectomy. Three patients achieved clinical remission. All patients experienced improved endoscopic severity and quality of life. Four weeks after treatment completion, six patients had worsened partial Mayo scores. Four patients progressed to colectomy after study completion. Colon CYP1A1 RNA expression increased 12 557-fold at week 8 among six patients evaluated. No patient discontinued IN due to an adverse event. Concomitant administration of 3-methoxy-4-nitroflavone, an AhR antagonist, in a murine colitis model abrogated the benefit of IN.ConclusionIN is a potentially effective therapy for patients with treatment-refractory ulcerative colitis. Proper feed optimization is vital for the health and growth of goats, with fodder quality being a central factor. This study evaluated the impact of feeding goats guinea grass mixed with various tree legumes on their growth and feed consumption. Nine upgraded goats were used for this research, divided equally among three treatment groups. Each treatment was repeated three times, with one goat per repetition. The experimental treatments included guinea grass paired with Gliricidia (T1), Indigofera (T2), or Ipil-ipil (T3). The results showed that the T2 (Guinea grass + Indigofera tinctoria) combination resulted in the highest feed intake at 29.94kg over five weeks, a finding significant with a p-value of 0.04. Nutrient analysis demonstrated a significant intake of crude protein for T2 with a p-value of 0.038. Moreover, T2 exhibited the most substantial total weight gain (3016.67g) and an average daily gain of 86.19g/day, with p-values of <0.001 and 0.013 respectively. In conclusion, the mix of Guinea grass and Indigofera tinctoria offers the best results in terms of feed intake and goat growth performance. [ABSTRACT FROM AUTHOR]

A proper planting system and efficient management are needed to cultivate forage effectively. Therefore, this research aimed to evaluate forage production and nutrient content of different elephant grass varieties grown with Indigofera in the intercropping system and were harvested at different intervals of defoliation. The research was conducted in the dry and rainy seasons using a randomized block design with 3 factors (2x2x2) and 4 replications. The first factor was two different elephant grass varieties consisting of cv. Pakchong and Taiwan, the second factor was the planting pattern, including intercropping and monoculture, while the third factor was harvesting age, comprising 50 days and 60 days. The results showed an interaction effect between grass varieties, planting patterns, and harvesting ages. Intercropping the Pakchong varieties with Indigofera and harvesting at 60 days produced the highest forage fresh weight and DM production, ADF as well as NDF contents and yields, CP yield, and the highest carrying capacity (p<0.05) according to DM intake. When harvested at 50 days, Indigofera increased the protein content of forage, but this effect did not occur when it was harvested at 60 days. Furthermore, nutrient yields were not affected by interactions of three factors in the rainy season (p>0.05), but in the dry season, the highest ash and CP yields were produced by the Pakchong variety intercropped with Indigofera and harvesting at 60 days. The highest ADF and NDF yields were obtained from the Taiwan varieties grown in monoculture and harvested at 60 days. Based on the results, it was concluded that the two varieties of elephant grass could be cultivated with Indigofera in the intercropping system and harvesting time of 60 days will produce the highest nutrient yield and carrying capacity. Faktor H merupakan komposisi substrat (substitusi DI dengan SDP) yaitu, H1= SDP 0% + DI 30%; H2= SDP 2% + DI 28%; H3= SDP 4% + DI 26%; H4 = SDP 6% + DI 24%; faktor N adalah jenis kemasan, N0 = tanpa dikemas; N1= plastik; N2= kertas; N3= karung goni. The aims of this study to determine the quality of physical (aroma, color, texture) and the properties of physical (compactness, density, moisture content) of wafers with substrate composition (substitution of indigofera leaves/IL with papaya leaf silage/PLS) and different packaging. The study used a factored completely randomized design (CRD) (4x4) with 3 replications. The H factor is the composition of the substrate (substitution of IL with PLS), namely, H1= PLS 0% + IL 30% H2= PLS 2% + IL 28%; H3= PLS 4% + IL 26%; H4 = PLS 6% + IL 24%; factor N is the type of packaging, N0 = no packaging; N1= plastic; N2= paper; N3 = gunny sack. Parameters measured were quality of physical (color, texture, aroma) and properties of physical (specific gravity, density and moisture content) of wafers. The results showed that the interaction between the type of packaging and the composition of the substrate (P0.01) affected the texture and aroma of wafers. Composition of substrate significantly (P0.01) affected the density and aroma of wafers. The type of packaging significantly (P0.01) affected the physical quality (aroma, color, texture) and physical properties (moisture content). Substitution of 28% IL with 2% PLS on wafers with gunny sack resulted in the best texture of wafer (coarse, dense and not slimy texture, score 3.44) and substitution of 24% DI with 6% SDP with gunny sack packaging with the best aroma of wafer (typical wafer aroma, score 3.46).

This study presents a taxonomic revision of the genus Indigofera L. (tribe Indigofereae, subfamily Faboideae, Fabaceae) in Egypt. The revision is based on 58 specimens representing 13 taxa of Indigofera collected from different locations and habitats. The morphological characteristics of the vegetative and reproductive parts, such as indumentum; leaf shape, and size; stipule shape and length; calyx shape and length; corolla color, shape, and size; pod and seeds (color, shape, and size) were proved to have high taxonomic significance in distinguishing the species belonging to the genus Indigofera. Generally, all parts of the plant are covered with dense white two-armed hairs, which give the plant a silvery appearance. In Indigofera cordifolia B. Heyne ex Roth, the leaves are unifoliolate, but they are compound among the other investigated species. An artificial key to the studied taxa, full species description and illustrations, local and global distribution, habitat, and conservation status are provided to facilitate the identification and recognition of these species. Introduction: Indigofera L. is the third largest genus in Fabaceae and includes economically important species that are used for indigo dye-producing, medicinal, ornamental, and soil and water conservation. The genus is taxonomically difficult due to the high level of overlap in morphological characters of interspecies, fewer reliability states for classification, and extensive adaptive evolution. Previous characteristic-based taxonomy and nuclear ITS-based phylogenies have contributed to our understanding of Indigofera taxonomy and evolution. However, the lack of chloroplast genomic resources limits our comprehensive understanding of the phylogenetic relationships and evolutionary processes of Indigofera. Methods: Here, we newly assembled 18 chloroplast genomes of Indigofera. We performed a series of analyses of genome structure, nucleotide diversity, phylogenetic analysis, species pairwise Ka/Ks ratios, and positive selection analysis by combining with allied species in Papilionoideae. Results and discussion: The chloroplast genomes of Indigofera exhibited highly conserved structures and ranged in size from 157,918 to 160,040 bp, containing 83 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Thirteen highly variable regions were identified, of which trnK-rbcL, ndhF-trnL, and ycf1 were considered as candidate DNA barcodes for species identification of Indigofera. Phylogenetic analysis using maximum likelihood (ML) and Bayesian inference (BI) methods based on complete chloroplast genome and protein-coding genes (PCGs) generated a well-resolved phylogeny of Indigofera and allied species. Indigofera monophyly was strongly supported, and four monophyletic lineages (i.e., the Pantropical, East Asian, Tethyan, and Palaeotropical clades) were resolved within the genus. The species pairwise Ka/Ks ratios showed values lower than 1, and 13 genes with significant posterior probabilities for codon sites were identified in the positive selection analysis using the branch-site model, eight of which were associated with photosynthesis. Positive selection of accD suggested that Indigofera species have experienced adaptive evolution to selection pressures imposed by their herbivores and pathogens. Our study provided insight into the structural variation of chloroplast genomes, phylogenetic relationships, and adaptive evolution in Indigofera. These results will facilitate future studies on species identification, interspecific and intraspecific delimitation, adaptive evolution, and the phylogenetic relationships of the genus Indigofera. Background: Textile dyes which are eliminated as unfixed dyes through the effluent from textile industry cause deleterious effect on the fresh water fish. Thus, toxicity tests were conducted using indigo dye on Oreochromis niloticus juveniles (mean weight 30.00 ± 0.73 g) as a test fish under bioassay system. Varying concentration of 0.00 (control), 0.5, 1.0, 1.5, 2.0, and 2.5 mg/l containing graded levels of Indigofera were applied in experimental tanks to determine the lethal concentration. The LC50 of dye at 96 h was 1.3 mg/l of water. The histopathology (heart, gills, kidney and spleen) and water quality parameters (Dissolved oxygen, pH and temperature) were determined using standard methods and behavioural responses were observed. Results: Histopathology of heart, gills, kidney and spleen revealed degeneration of cells, space formation, slight cellular changes and vacuolation among the treatments especially in the higher concentration of 1.5 mg/l, 2.0 mg/l, and 2.5 mg/l of the dye solution used except in the control. The pH and DO of control were significantly different from the experimental units while there was no significant variation in the temperature of the control and all the experimental units. Effects of indigo dye on water parameters were significant (P < 0.05) throughout the experiment. Behavioural responses exhibited by the experimental fish include irregular swimming, hyperventilation, rapid opercula movement, and restlessness. The dye concentration and exposure period both boosted these behaviours. Conclusions: The findings of this study indicated that indigo dye solution is toxic to fish, and that fish opercula movement and mortality were influenced by the dosage of each concentration and the duration of exposure. Objective. Indigofera bungeana is a shrub with high quality protein that has been widely utilized for forage grass in the semi-arid regions of China. This study aimed to enrich the currently available knowledge and clarify the detailed drought stress regulatory mechanisms in I. bungeana, and provide a theoretical foundation for the cultivation and resistance breeding of forage crops. Methods. This study evaluates the response mechanism to drought stress by exploiting multiple parameters and transcriptomic analyses of a 1-year-old seedlings of I. bungeana in a pot experiment. Results. Drought stress significantly caused physiological changes in I. bungeana. The antioxidant enzyme activities and osmoregulation substance content of I. bungeana showed an increase under drought. Moreover, 3,978 and 6,923 differentially expressed genes were approved by transcriptome in leaves and roots. The transcription factors, hormone signal transduction, carbohydrate metabolism of regulatory network were observed to have increased. In both tissues, genes related to plant hormone signaling transduction pathway might play a more pivotal role in drought tolerance. Transcription factors families like basic helix-loop-helix (bHLH), vian myeloblastosis viral oncogene homolog (MYB), basic leucine zipper (bZIP) and the metabolic pathway related-genes like serine/threonine-phosphatase 2C (PP2C), SNF1-related protein kinase 2 (SnRK2), indole-3-acetic acid (IAA), auxin (AUX28), small auxin up-regulated rna (SAUR), sucrose synthase (SUS), sucrosecarriers (SUC) were highlighted for future research about drought stress resistance in Indigofera bungeana. Conclusion. Our study posited I. bungeana mainly participate in various physiological and metabolic activities to response severe drought stress, by regulating the expression of the related genes in hormone signal transduction. With the aim to investigate the active chemical constituents for the roots of Indigofera stachyodes, the petroleum ether part of this plant was separated and purified using silica gel column chromatography, Sephadex LH-20 column chromatography, semi-preparative high performance liquid chromatography, and recrystallization methods. The structures of obtained monomeric compounds were identified through modern spectral techniques by detail analysis of their electrospray ionization mass spectrometry, hydrogen and carbon nuclear magnetic resonance spectra data, as well as compared with the reported references. The results were as follows: twenty-two natural compounds were identified as β-sitostenone (1), stigmasta-3,6-di-one (2), 6β-hydroxystigmast-4-en-3-one (3), (22E)-5α,8α-epidioxyergosta-6,22-dien-3β-ol (4), medicarpin (5), sativan (6), 2′,4′-dihydroxychaicone (7), 6,7-dimethoxy-4-hydroxy-1-naphthoic acid (8), p-hydroxybenzoic acid ethyl ester (9), 2,4-dihydroxybenzoic acid ethyl ester (10), (9E,11E)-13-oxo-9,11-ocatadecadienoic acid (11), (9E,11E)-13-oxo-9,11-octadecadienoic acid methyl ester (12), 9-oxo-10E,12E-octadecadienoic acid methyl ester (13), 9-hydroxy-10E,12Z-octadecadienoic acid methyl ester (14), pinellic acid (15) and 9-oxo-10E,12E-octadecadienoic acid-(2S)-2,3-dihydroxypropyl ester (16), β-sitosterol (17), betulinic acid (18), 3β-hydroxyolean-12-en-11-one (19), maackiain (20), phellopterin (21), palmitic acid (22). Of these compounds, ten compounds (compounds 1-3, 7, 11-16) were isolated from the Fabaceae for the first time; sixteen compounds (compounds 1-16) were obtained from Indigofera plant for the first time; at the same time, compounds 1-16, 18, 19, 21 were obtained from the roots of Indigofera stachyodes for the first time. Thus, the results of this research enrich the chemical structure types of roots of I. stachyodes, and afford a scientific foundation for its further rational use.

This experiment aimed to evaluate chemical composition, in vitro rumen fermentation, digestibility, and methane emissions of forages including bede grass, gamal (Gliricidia sepium), Indigofera, lamtoro (Leucaena leucocephala), elephant grass, mini elephant grass and Thai elephant grass. Forage samples were dried at 60oC for 24 hours, then ground to 1 mm sieve. The ground samples were used subsequently to determine the chemical composition and in vitro rumen fermentation test. Incubation was carried out in a water bath at 39oC for 48 h without replication. The results showed that all forages contained various crude protein at a range of 9-20% DM. The proportion of neutral detergent insoluble CP (NDICP) and acid detergent insoluble CP (ADICP) in forage was generally low, but the highest was found in bede grass. The highest NH3 concentration was found in Indigofera incubation, and lowest was in bede grass. All forage samples, except for mini elephant grass and Indigofera, had IVDMD and IVOMD below 70%. We identified low methane emissions from forage at 48 hours of incubation. It was concluded that legumes had a higher crude protein than in grass species and had low methane emissions. However, the relatively low digestibility of legume may limit its utilization.

Penggunaan pewarnaan tekstil yang tepat agar hasilnya maksimal. Teknik celup pada kain denim dan teknik semprot pada pewarnaan alam jarang ditemui, sehingga menjadi inovasi yang perlu dikembangkan. Lingkungan penulis memiliki potensi bahan baku yang dapat dibudidayakan untuk peluang usaha di bidang pertanian. Permasalahan yang dihadapi yaitu bagaimana proses pembuatan pewarna alam sebagai pewarna tekstil, bagaimana hasil pewarnaan alam pada pembuatan ready to wear. Tujuannya untuk mengetahui proses pembuatan pewarna alam sebagai pewarna tekstil, untuk mengetahui hasil pewarna alam pada pembuatan ready to wear. Metodologi penilitian yang digunakan yaitu RD dan analisis data diskriptif. Hasil dari penerapan teknik pewarnaan tie dye, celup dan semprot menggunakan indigofera dan jelawe pada kain denim dan katun dengan fiksasi menghasilkan warna yang berbeda. Bahan jelawe menghasilkan warna kuning kecoklatan dan indigofera menghasilkan warna biru. Kain denim memiliki hasil yang bagus menggunakan warna jelawe dan teknik celup. Kain katun memiliki hasil yang bagus pada kedua warna menggunakan teknik celup dan semprot. Warna indigofera tidak dapat menggunakan teknik semprot karena memiliki tekstur pasta yang sulit menempel. Fiksasi tunjung menghasilkan warna gelap, kapur menjadikan warna sedikit cerah dan tawas menjadikan warna cerah. Saran yang dari penelitian ini yaitu cara pewarnaan menggunakan warna dasar yang sama agar warna rata.

In this study, Indigofera tinctoria extract (ITLE) along with maltodextrin in different concentrations was encapsulated using a freeze dryer, and some physicochemical properties were measured. Powder containing 30% maltodextrin was selected as the optimal powder for use in ice‐cream production in four levels (0%–1.8%), and some quality parameters were examined. The results showed that with increasing the carrier concentration, moisture content, aw, solubility, a*, and b* of the powders decreased; bulk density, tapped density, and L* did not change significantly; total phenolic content and antioxidant activity of powders also increased significantly (p <.05). Addition of encapsulated ITLE to the ice cream caused a significant decrease in L*, b*, and melting rate, a significant increase in a*, overrun, and hardness of the samples and no change in the viscosity of the ice‐cream mix (p <.05). Ice cream containing 1.2% encapsulated ITLE had higher sensory acceptance than other levels following control ice cream. The results of this study showed that ITLE can be used as a desirable additive in the production as a natural antioxidant and color agent. [ABSTRACT FROM AUTHOR]

Summary: Microfibres (diameter <5 mm) and textile dyes released from textile industries are ubiquitous, cause environmental pollution, and harm aquatic flora, fauna, animals and human life. Therefore, enzymatic abatement of microfibre pollution and textile dye detoxification is essential. Microbial enzymes for such application present major challenges of scale and affordability to clean up large scale pollution. Therefore, enzymes required for the biodegradation of microfibres and indigo dye were expressed in transplastomic tobacco plants through chloroplast genetic engineering. Integration of laccase and lignin peroxidase genes into the tobacco chloroplast genomes and homoplasmy was confirmed by Southern blots. Decolorization (up to 86%) of samples containing indigo dye (100 mg/L) was obtained using cp‐laccase (0.5% plant enzyme powder). Significant (8‐fold) reduction in commercial microbial cellulase cocktail was achieved in pretreated cotton fibre hydrolysis by supplementing cost effective cellulases (endoglucanases, ß‐glucosidases) and accessory enzymes (swollenin, xylanase, lipase) and ligninases (laccase lignin peroxidase) expressed in chloroplasts. Microfibre hydrolysis using cocktail of Cp‐cellulases and Cp‐accessory enzymes along with minimal dose (0.25% and 0.5%) of commercial cellulase blend (Ctec2) showed 88%–89% of sugar release from pretreated cotton and microfibres. Cp‐ligninases, Cp‐cellulases and Cp‐accessory enzymes were stable in freeze dried leaves up to 15 and 36 months respectively at room temperature, when protected from light. Use of plant powder for decolorization or hydrolysis eliminated the need for preservatives, purification or concentration or cold chain. The typification of the names of three non-endemic species of Indigofera in Australia is revisited. Firstly, the uncertain typification of an introduced species, I. hochstetteri Baker, is considered; a previous, imprecise lectotypification is recognised and a second step lectotype is designated. Secondly, an earlier error in the citation of the type of another introduced species, I. sessiliflora DC., is corrected and specific details of the holotype are provided. Simple Summary: Indigo waste is a by-product of the processing of natural indigo dye. Indigo waste could be utilized as a protein source in ruminant rations, which would reduce the cost of feed. We evaluated the effects of the inclusion of indigo waste in concentrate diets on the feed utilization, rumen fermentation, hematology, immune function and growth performance of growing beef cattle. The present findings suggest that the inclusion of indigo waste at low levels in concentrate diets maintains feed intake, digestibility, rumen fermentation or growth performance in growing beef cattle without affecting hematology or immune function. This experiment was conducted to assess the effect of indigo waste on the feed intake, digestibility, rumen fermentation, hematology, immune response and growth performance in growing beef cattle. Twenty crossbred beef cattle with an initial body weight (BW) of 145 ± 11 kg were fed four levels of indigo waste for 90 days in a trial. Additions of indigo waste at 0%, 10%, 20% and 30% in a concentrate diet using a completely randomized design (CRD). Cattle were fed concentrate at 1.8% BW, with rice straw fed ad libitum. The concentrate intake decreased linearly (p = 0.01) with the addition of indigo waste. The supplementation with indigo waste reduced dry matter (DM) and organic matter (OM) digestibility cubically (p = 0.03 and p = 0.02, respectively), while increasing neutral detergent fiber (NDF) digestibility cubically (p = 0.02). The final BW of beef cattle decreased linearly (p = 0.03) with the addition of indigo waste. The inclusion of indigo waste decreased the average daily gain (ADG) and gain-to-feed ratio (G:F) linearly (p < 0.01) from 0 to 90 days. The nutrient digestibility, ADG and G:F of beef cattle fed 10% indigo waste in the diet was similar when compared with the control (0% indigo waste). The ruminal pH, ammonia-nitrogen (NH3-N) and total volatile fatty acid (VFA) concentrations were similar among treatments (p > 0.05). The proportion of acetate increased linearly (p < 0.01) but propionate decreased linearly (p < 0.01), resulting in an increase in the acetate to propionate ratio (p < 0.01) when cattle were fed with indigo waste supplementation. Increasing indigo waste levels did not influence blood urea nitrogen (BUN) levels, hematological parameters or immune responses (IgA, IgM and IgG) (p > 0.05). With the aim to investigate the active chemical constituents for the roots of Indigofera stachyodes, the petroleum ether part of this plant was separated and purified using silica gel column chromatography, Sephadex LH-20 column chromatography, semi-preparative high performance liquid chromatography, and recrystallization methods. The structures of obtained monomeric compounds were identified through modern spectral techniques by detail analysis of their electrospray ionization mass spectrometry, hydrogen and carbon nuclear magnetic resonance spectra data, as well as compared with the reported references. This study aimed to evaluate the effect of mixed feed sorghum-Indigofera on Garut ewes reproductive performance stimulated with Pregnant Mare Serum Gonadotropin (PMSG) hormone. Twenty-eight multiparous Garut ewes were grouped based on feed type (local forage (LF) vs. mixed feed sorghum-Indigofera (SI)), for a 30-day acclimatization period with a completely randomized design. Following acclimatization, all animals were synchronized with PGF2a (i.m.) twice, 11 days apart. Half the population of each group was stimulated with 500 IU PMSG after the second injection. Estrous characteristics were observed for seven days, followed by natural mating. Pregnancy status and the number of fetuses were detected 30 days after mating using ultrasound (USG), and reproductive efficiency was evaluated after parturition. Data during acclimatization were analyzed using a two-sample t-test, while the reproductive performances used a General Linear Model (GLM). Estrous characteristics showed no interaction (p>0.05) between feed types and PMSG stimulation. The onset of estrous in the SI treatment occurred 9.33 hours faster (p<0.05) than in the LF treatment. Meanwhile, the duration of estrous in the group stimulated with PMSG was 18.67 hours longer (p<0.05) than without PMSG. The vulva temperature at standing heat was 38.16±0.69 °C (p>0.05), with the pregnancy rate ranging 42.85%-100%. Interestingly, the SI group treated with or without PMSG had a larger litter size (1.43-4.50) compared to the LF group (1.25-1.33). It is concluded that feeding with SI had improved reproductive success, with the highest number of ewes giving birth. Additionally, the ewes fed SI with PMSG had the largest litter size. The use of natural dyes as textile dyes is increasing due to environmentally friendly standards and does not require the use of synthetic dyes. By banning the use of synthetic dyes containing azo groups, this is the right moment to reintroduce natural dyes that have been abandoned for a long time. This study aims to identify the active compound of tantrum leaves (Indigofera tinctoria) as a natural textile dye by maceration extraction method with variations in maceration time and material treatment on wet and dry tantrum leaves. This experimental research method was carried out using several stages of analysis, namely the phytochemical test, FTIR (Fourier Transform Infrared Spectroscopy), and UV-Vis (Ultraviolet-Visible Light) spectrophotometer. The results of the phytochemical tests showed that the compounds contained in the Indigofera tinctoria plant were indigo, flavonoids, steroids, and alkaloids. The FTIR test indicated that the dye content in the extract was from the indigo group. The results of the UV-Vis spectrum showed that there was maximum absorption in wet tantrum leaf extract, dry tantrum leaf extract, and alkaline extract at wavelengths of 406.50 nm, 365.50 nm, and 664.50 nm, respectively. ABSTRACT. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian pelet mengandung tepung daun Indigofera terhadap produktivitas kambing Boerka periode bunting dan laktasi. Ternak yang digunakan adalah kambing Boerka bunting bulan keempat sebanyak 60 ekor dengan variasi paritas ke 2 dan 3 dan rataan bobot badan saat dikawinkan adalah 32,63±5,29 kg. Penelitian menggunakan Rancangan Acak Kelompok (RAK) dengan 3 perlakuan pakan dengan 20 kali ulangan: rumput lapang + Pelet komersil (R1); rumput lapang + pelet Indigofera 10% (R2); rumput lapang + pelet Indigofera 20% (R3). Parameter yang diukur adalah bobot badan anak lahir dan bobot badan anak prasapih (umur 3 bulan). Data dianalisis menggunakan Analysis of Variance (ANOVA ) satu arah dan dilanjutkan dengan Uji Jarak Berganda Duncan. Hasil analisis statistik menunjukkan bahwa pemberian tepung daun Indigofera 10% memberikan pengaruh nyata terhadap bobot lahir anak kambing betina dan tidak berbeda nyata terhadap parameter lainnya namun cenderung menurun pada pemberian tepung daun Indigofera 20%. Dapat disimpulkan bahwa penggunaan tepung daun Indigofera dalam pelet sampai 20% memberikan pengaruh yang tidak nyata terhadap produktivitas kambing Boerka periode bunting dan laktasi. A total of 60 heads of Boerka goat who were pregnant at 4th, parity of 2nd and 3rd and average of body weight when mated were 32,63±5,29 kg. Research used randomized block design with 3 feed treatments and 20 replicated: native grass + commercial pellet (R1); native grass + Indigofera’s pellet 10% (R2); native grass + Indigofera’s pellet 20%. The parameter measured were body weight of lamb born and body weight of lamb at pre-weaning (3 month aged). Data were analyzed used Analysis of Variance (ANOVA) one way dan continued with Multiple’s Range Test of Duncan. The result of statistical analysis showed that dietary Indigofera leaf meal 10% gave significant effect on birth weight of female lamb and non significant effect on other parameters but tends to decreased in dietary Indigofera leaf meal 20%. It could be conclude that utilization of Indigofera leaf meal until 20% in pellet gave not significant effect on productivity of Boerka goat at pregnancy and lactating period.

The study aimed to analyze the pharmacological properties of medicinal plant Indigofera hochstetteri Baker extracts. Preliminary phytochemical analysis revealed a diverse range of secondary metabolites present in it. TLC analysis detected numerous phytochemicals with varying Rf values, aiding in different solvent systems. GC-MS analysis revealed the presence of 29 bioactive compounds with diverse pharmacological activities, including anti-inflammatory, antioxidant, analgesic and antimicrobial properties. Antimicrobial effect of I. hochstetteri Baker methanolic extract showed significant inhibitory effects against E. coli , E. aerogenes , S. flexneri , P. aeruginosa , S. aureus , E. faecalis , B. cereus , and fungal strain C. albicans. The methanol extract also showed significant antifungal activity by inhibiting the growth of Sclerotium rolfsii in food poisoning method. MTT assays revealed significant cytotoxic activity of methanolic extract against human leukemia HL-60 cancer cells with IC 50 of 116.01 μg/mL. In apoptotic study, I. hochstetteri Baker methanolic extract showed 28.84% viable cells, 30.2% early apoptosis, 35.54% late apoptosis, and 5.86% necrosis comparatively similar with standard used. The extract showed significant anti-inflammatory effect on HRBC stabilization, and protein denaturation of BSA and egg albumin denaturation with IC 50 of 193.62 μg/mL, 113.94 μg/mL respectively. In anti-diabetic assays like α-amylase, α-glucosidase, and Glucose uptake assay, I. hochstetteri extract showed good anti-diabetic effect with IC 50 of 60.64 μg/mL, 169.34 μg/mL, and 205.63 μg/mL respectively. A new species, Indigofera jaisalmerica C.S. Purohit et R.N. Kulloli. sp nov. from Indian desert (Jaisalmer district, Rajasthan, India) is described and illustrated. The new species is closely related with Indigofera cordifolia Roth, but differs in having a leaf more than 2 cm long with scattered hairs on both surfaces, trichomes antler shape on leaf surface, inflorescence axillary spicate with 14–17 flowers, calyx slightly longer than flower, fruit strictly one-seeded and enclosed by calyx lobes. Detailed taxonomic description along with colored photo plate, distribution map, SEM study of leaf and seeds, and associates are provided here for its easy identification.

Onchocerciasis is a major public health problem caused by Onchocerca volvulus parasite and transmitted to humans via black flies (simulium) bites. The control of onchocerciasis relies much on the use of the chemical drug ivermectin, which is only effective against microfilariae and has led to drug resistance. This study was carried out to assess the in vitro antifilarial activity of methanolic extract of Indigofera tinctoria and its most active fractions on adult male O. ochengi worm, the closest model to O. volvulus, after 48 h and 72 h of treatment. Worms' viability was determined biochemically by MTT/formazan colorimetry assay. The promising plant extract's acute and subacute oral toxicity were evaluated on both mice and rats. The result revealed a highest antifilarial activity of the methanolic extract (L C 50 = 12.28 μ g / mL) compared to ivermectin (L C 50 = 26.50 μ g / mL) after 72 h of treatment. Out of the eight (08), chromatographic fractions screened, only three (03) fractions (C, F, and G) revealed the highest anti-Onchocerca activity after 72 h of treatment. An oral administration of the plant extract at a single dose of 2000 mg/kg did not produce any toxicity in mice. After repeated daily administration of methanolic extract of I. tinctoria (250 mg/kg, 500 mg/kg, and 1000 mg/kg) for 28 days, no significant changes in body weight, biochemical, and haematological parameters was observed. Histopathological examination of organs did not reveal any sign of alteration. The phytochemical analysis of the methanolic extract of I. tinctoria revealed the presence of various phenolic compounds. Therefore, this study demonstrated the potential antifilarial activity of Indigofera tinctoria and offered an alternative to treating onchocerciasis. Moreover, further studies could be developed in promising new antifilarial sources of the isolated compound and in vivo antifilarial activity of Indigofera tinctoria in the animal model needs to be studied. Background: Indigofera tinctoria plays the role of a natural dye source that produces indigo color and contains the secondary metabolite indican which is highly responsive to light intensity. This study aims to examine the critical period for the formation of the secondary metabolite indican on light intensity. Methods: The method used was a completely randomized block design with a split-plot design. The study consisted of 2 treatment factors, namely: length of shade (main plot) and light intensity (subplot). The length of shade included 5 levels, namely early growth phase (up to 1 month after planting), mid-growth phase (up to 2 months after planting), maximum growth phase (up to 3 months after planting), 1 month before harvest and 2 months before harvest. Light intensity had 3 levels, namely 50% light intensity (38,464.3 lux), 25% (19,232.15 lux) and 10% (7,692.86 lux). Result: The combination of duration of shade and light intensity affected the growth, yield and content of secondary metabolites (indican) in Indigofera tinctoria. The highest number of leaves, plant fresh weight and biomass was found in the combination of shade in the early growth phase (up to 1 month after planting) with 50% light intensity. The highest indican production was found in the midgrowth shading (up to 2 months after planting) with 10% light intensity, which was 843.33 ppm. Background: Proanthocyanidins (PAs) have always been considered as important medicinal value component. In order to gain insights into the PA biosynthesis regulatory network in I. stachyodes roots, we analyzed the transcriptome of the I. stachyodes in Leaf, Stem, RootI (one-year-old root), and RootII (two-year-old root). Results: In this study, a total of 110,779 non-redundant unigenes were obtained, of which 63,863 could be functionally annotated. Simultaneously, 75 structural genes that regulate PA biosynthesis were identified, of these 6 structural genes (IsF3′H1, IsANR2, IsLAR2, IsUGT72L1-3, IsMATE2, IsMATE3) may play an important role in the synthesis of PAs in I. stachyodes roots. Furthermore, co-expression network analysis revealed that 34 IsMYBs, 18 IsbHLHs, 15 IsWRKYs, 9 IsMADSs, and 3 IsWIPs hub TFs are potential regulators for PA accumulation. Among them, IsMYB24 and IsMYB79 may be closely involved in the PA biosynthesis in I. stachyodes roots. Conclusions: The biosynthesis of PAs in I. stachyodes roots is mainly produced by the subsequent pathway of cyanidin. Our work provides new insights into the molecular pathways underlying PA accumulation and enhances our global understanding of transcriptome dynamics throughout different tissues. Silver nanoparticles (AgNP's) have been successfully fabricated via bio-reduction of Indigofera tinctoria plant extract as the reducing and capping agent. The effects of pH and temperature on the formation of the AgNP's have been studied. The synthesized AgNP's have been characterized using UV-Visible spectroscopy, Fourier Transform Infrared (FTIR) spectroscopy, Zeta potential analysis, Scanning Electron Microscope (SEM) and Atomic Force Microscopy (AFM). Antimicrobial activities of the synthesized AgNP's have been tested against both bacterial and fungal strains by agar well diffusion method. The biomass-capped AgNP's imparted antimicrobial activity by inhibiting the growth of Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Aspergillus niger and Penicillium chrysogenum. The antioxidant activities of the synthesized AgNP's exhibit low IC50 value of ~55.72 µg/mL. Studies on drug loading and kinetics of drug release reveal that I. tinctoria AgNP's follow the zero-order kinetics at pH 4.6 and pH 7.4. The gradient value of 0.568 (pH 4.6) and 0.6 (pH 7.4) falls between 0.42 < n < 0.85 when fitted into Peppa's plot indicating that the drug release follow an anomalous transport or non-Fickian diffusion transport, indicating that the diffusion is time dependent. Objective: The study aimed to determine the effect of using a combination of cassava tuber (Manihot esculenta) and Indigofera zollingeriana leaf flour as an energy source supplemented with citric acid in the ration on performance, carcass quality, digesta pH, viscosity, and the number of villi in the intestines of broilers. Materials and Methods: The research design was completely randomized with five treatments and five replications, each containing four broilers. The treatment was a substitution of corn in the ration with a combination of cassava tuber and I. zollingeriana leaf (CIF): without CIF, 5% CIF substitute for corn + 0.2% citric acid, 10% CIF substitute for corn + 0.2% citric acid, 15% CIF corn substitute + 0.2% citric acid, and CIF corn substitute + 0.2% citric acid. Each treatment ration was supplemented with 0.2% citric acid. The variables that were looked at were ration consumption, body weight gain, feed conversion, live weight, percentage of the carcass, percentage of abdominal fat, pH, viscosity, and the number of broiler villi. Results: This study showed that the combination of CIF flour supplemented with 0.2% citric acid had a significant effect (p < 0.05) on body weight gain, ration conversion, live weight, carcass percentage, and the number of villi in broiler intestines. But it did not have a significant effect (p > 0.05) on the amount of feed eaten, the amount of fat in the abdomen, the pH of the digesta, or the thickness of the broilers' blood. Conclusions: The combination of CIF flour at a level of 10% supplemented with 0.2% citric acid can be used as an energy source to replace corn without having a bad effect on production performance, carcass quality, and small intestine characteristics of broilers.
