Recent research in Technology-Enhanced Learning (TEL) demonstrated several important benefits that semantic technologies can bring to the TEL domain. An underlying assumption for most of these research efforts is the existence of a domain ontology. The second unspoken assumption follows that educators will build domain ontologies for their courses. However, ontologies are hard to build, especially for not-tech-savvy users. Tools for ontology extraction from text aim to overcome this problem. We have conducted an empirical study with educators, both from Information Technology (IT) and non-IT domains, where they used current ontology extraction tools to build domain ontologies for their courses from their existing course material. Based on the obtained study results we have drawn conclusions about the existing ontology extraction tools and provided recommendations for their future development so that they can be beneficial for the TEL domain. The article deals with design and design of a thin-walled heat sink for the robot control system, the measurement of the transverse roughness of the touch surfaces and the programming procedure in Autodesk Inventor 2019 software. In this software, a design of the 3D model of the heat sink generated based on power supply and NC code generation for the Fanuc control system. The production it-self carried out on the CNC Vertical Machining Center Pinnacle 2100 with this control system. The longitudinal surface roughness was measured with a Mi-tutoyo SJ 400 dredger and the measurement result was the maximum Ra = 0.69 μm and Rz = 4.1 μμm.

The paper is focused on the influence of the coolant pressure on the surface roughness of the workpiece when machining stainless steels. The components were machined on a STAR SR-32J dual spindle machining center and an external cooling unit HYTEK CHAV 160/150-AF-F-OL was used for cooling. Two stainless steel components were investigated, namely the gas control valve rod and the high-pressure control valve housing, which require low roughness Ra after machining (less than 0.375 and 0.25 micrometers respectively). The first component was tested at 8 different pressures in the range of 150 bar - 10 bar and the second component at 4 different pressures in the range of 120 bar - 10 bar. The roughness parameters were measured by the contact method using the MITUTOYO Surftest SJ-410 Roughness Tester and the Alicona InfiniteFocus optical microscope. Based on these sample input parameters, it was evaluated howmuch the pressure affects the surface quality or suggested its reduction due to the high cost of operation of the external high-pressure equipment.

This paper is focused on analysis of residual stress of functional surfaces and subsurface layers created by precision technologies of hard machining for progressive constructional materials of forging tools. Methods of experiments are oriented on monitoring of residual stress in surface which is created by hard turning (roughing and finishing operations). Subsequently these surfaces were etched in thin layers by electro-chemical polishing. The residual stress was monitored in each etched layer. The measuring was executed by portable X-ray diffractometer for detection of residual stress and structural phases. The results significantly indicate rise and distribution of residual stress in surface and subsurface layers and their impact on functional properties of surface integrity.

The incorporation of additive manufactured (AM) metal parts to real assemblies is a crucial issue for the increasing of their industrial utilization. The presented research is devoted to the electron beam welding (EBW) of dissimilar steel joints. Dissimilarity is defined by the various types of steel and manufacturing processes used for the creation of specimens. Conventional AISI 316 stainless steel, selective laser melted (SLM) SS 316L stainless steel, and SLM M300 maraging steel were welded at variable parameters in the form of a welding current and a welding velocity. EBW joints were evaluated considering the macroscopic and microscopic characteristics, as well as a reached microhardness. The obtained preliminary results represent important input data for the follow-up experiments focused on the setting of optimal EBW parameters of welding the dissimilar joints including SLM products, with the consideration of their basic macroscopical and microscopical characteristics, mechanical properties, and residual stresses.

We investigated the use of boron-doped diamond (BDD) with different surface morphologies for the enhanced detection of nine different peptides by matrix-assisted laser desorption/ionisation mass spectrometry (MALDI-MS). For the first time, we compared three different nanostructured BDD film morphologies (Continuous, Nanograss, and Nanotips) with differently terminated surfaces (-H, -O, and -F) to commercially available Ground Steel plates. All these surfaces were evaluated for their effectiveness in detecting the nine different peptides by MALDI-MS. Our results demonstrated that certain nanostructured BDD surfaces exhibited superior performance for the detection of especially hydrophobic peptides (e.g., bradykinin 1-7, substance P, and the renin substrate), with a limit of detection of down to 2.3 pM. Further investigation showed that hydrophobic peptides (e.g., bradykinin 1-7, substance P, and the renin substrate) were effectively detected on hydrogen-terminated BDD surfaces. On the other hand, the highly acidic negatively charged peptide adrenocorticotropic hormone fragment 18-39 was effectively identified on oxygen-/fluorine-terminated BDD surfaces. Furthermore, BDD surfaces reduced sodium adduct contamination significantly.

The detection of defects in aluminium alloys using eddy current testing (ECT) can be restricted by higher electrical conductivity. Considering the occurrence of discontinuities during the selective laser melting (SLM) process, checking the ability of the ECT method for the mentioned purpose could bring simple and fast material identification. The research described here is focused on the application of three ECT probes with different frequency ranges (0.3-100 kHz overall) for the identification of artificial defects in SLM aluminium alloy AlSi10Mg. Standard penetration depth for the mentioned frequency range and identification abilities of used probes expressed through lift-off diagrams precede the main part of the research. Experimental specimens were designed in four groups to check the signal sensitivity to variations in the size and depth of cavities. The signal behavior was evaluated according to notch-type and hole-type artificial defects' presence on the surface of the material and spherical cavities in subsurface layers, filled and unfilled by unmolten powder. The maximal penetration depth of the identified defect, the smallest detectable notch-type and hole-type artificial defect, the main characteristics of signal curves based on defect properties and circumstances for distinguishing between the application of measurement regime were stated. These conclusions represent baselines for the creation of ECT methodology for the defectoscopy of evaluated material.

The present study aimed to enhance the efficiency and efficacy of the Al/Cu joint production process implemented by the company VEMID Ltd., Jagodina, Serbia, by attaining sound joints within a very short welding time. For this purpose, the present study aimed at investigating the accuracy and the quality of the continuous drive friction welding (CDFW) process, as well as the optimum combination of CDFW parameters with highest joint efficiency in terms of investigated properties. The accuracy was estimated through an analysis of temperature-time curves recorded during CDFW using an infrared camera. The quality was evaluated through an investigation of the properties of Al/Cu joints produced using different friction (66.7, 88.9, and 133.3 MPa) and forging (88.9, 222.2, and 355.6 MPa) pressures and a constant total welding time (4 s) and rotational speed (2100 rpm). Thermal imaging with an infrared camera demonstrated that the actual total welding time was 15% longer compared to the nominal value. This was attributed to the slow pressure response of the pneumatic brake system. The relative changes in the maximum surface temperature ( T MS ) during the CDFW process corresponded to changes in welding pressures, indicating the potential of the thermal imaging method for monitoring and assessing this process. A preliminary investigation demonstrated that Al/Cu joints produced using welding pressures less than 88.9 MPa often displayed the presence of non-joined micro-regions at the Al/Cu interface and a significant thickness of interfacial Al 2 Cu (up to 1 µm). However, when friction pressure was set at 66.7 MPa, an increase in the forging pressure to 222.2 MPa eliminated the presence of non-joined micro-regions and reduced the thickness of Al 2 Cu to 0.5 µm on the average level. These Al/Cu joints achieved the highest joint efficiencies in terms of strength (100%) and ductility (61%). They exhibited an electrical conductivity higher than 92% of the theoretical value. A further increase in any welding pressure produced similar or deteriorated properties, accompanied by an increase in the consumption of raw materials and energy. Such turn of events was counterproductive to the original goal of increasing the efficiency and efficacy of the CDFW process.

The presented manuscript focuses on the influence of process parameters of abrasive water jet technology on the creation of non-transient erosive grooves. The processed stainless steel SS 316L is additively manufactured using the selective laser melting (SLM) method. Due to the distinct mechanical properties of this material resulting from the production process, the material was machined in two planes according to the direction of the printing layers. The experimental part employed a planned experiment utilizing the DoE (Design of Experiment) method. Experiments aimed at varying process parameters (traverse speed, standoff distance, abrasive mass flow) were conducted at a water pressure of 50 MPa, assessing the parameters' impact on the removed material and the properties of the resulting non-transient erosion groove. The properties of the erosion groove, such as shape and the material removal (area of erosion groove), were evaluated. The influences of process parameters on the observed parameters were assessed using the analysis of variance (ANOVA) method. Experiment preparation and setup were based on a thorough theoretical analysis of the machining process with the abrasive water jet (AWJ) method. The experiment also highlights the diverse properties of the SS 316L material prepared using the SLM method when machined with AWJ technology.

The identification of residual stresses (RS) in components made by selective laser melting (SLM) is necessary for subsequent technological optimization. The presented research is devoted to evaluating the influence of the combination of laser power (P), scanning velocity (v) and the rarely considered number of layers (n L ) on surface residual stresses in SLM stainless steel SS 316L. Experimental parameters were set based on the Design of Experiment (DoE) method, with follow-up X-ray diffraction (XRD) measurements and data processing using analysis of variance (ANOVA) and regression analysis. The obtained data are a valuable stepping-stone for the subsequent design of research focused on the application of sustainable eco-friendly Abrasive Water Jet (AWJ) peening for RS modification in the evaluated material.

Screen-printed sensors with chemically deposited boron-doped diamond electrodes (BDDE) were modified with different types of gold nanoparticles (AuNPs) according to a new original procedure. Physically and electrochemically deposited AuNPs had various sizes and also nanoporous character. They also differ in shape and density of surface coverage. The developed sensors were characterized using scanning electron microscopy and Raman spectroscopy. Their electrochemical properties were studied using cyclic voltammetry and electrochemical impedance spectrometry of selected outer sphere ([Ru(NH 3 ) 6 ]Cl 3 ) and inner sphere (K 3 [Fe(CN) 6 ], dopamine) redox markers. The application possibilities of such novel screen-printed sensors with BDDE modified by AuNPs were verified in the analysis of the neurotransmitter dopamine. The best analytical performance was achieved using printed sensors modified with the smallest AuNPs. The achieved limit of detection values in nanomolar concentrations (2.5 nmol L -1 ) are much lower than those of unmodified electrodes, which confirms the significant catalytic effects of gold nanoparticles on the surface of the working electrode. Residual stress occurs in the materials after different methods of processing due to the application of pressure and/or thermal gradient. The occurrence of residual stresses can be observed in both subtractive and additive-manufactured (AM) materials and objects. However, pressure residual stresses are considered, in some cases, to have a positive effect; there are applications where the neutral stress state is required. As there is a lack of standards describing the heat treatment of AM materials, there is a need for experimental research in this field. The objective of this article is to determine the heat treatment thermal regime to achieve close to zero stress state in the subsurface layer of additively manufactured AM316L stainless steel. The presented objective leads to the long-term goal of neutral etalons for eddy current residual stress testing preparation. A semi-product intended for the experiment was prepared using the Selective Laser Melting (SLM) process and subsequently cut, using Abrasive Water Jet (AWJ) technology, into experimental specimens, which were consequently heat-treated in combination with four temperatures and three holding times. Residual stresses were measured using X-ray diffraction (XRD), and microstructure variations were observed and examined. A combination of higher temperature and longer duration of heat treatment caused more significant stress relaxation, and the original stress state of the material influenced a degree of this relaxation. The microstructure formed of cellular grains changed slightly in the form of grain growth with randomly occurring unmolten powder particles, porosity, and inclusion precipitation.

Additive manufacturing has many positives, but its incorporation into functional parts production is restricted by the presence of defects. Eddy current testing provides solutions for their identification; however, some methodology and measurement standards for AM (additive manufacturing) products are still missing. The main purpose of the experiment described within this article was to check the ability of eddy current testing to identify AM stainless steel parts and to examine the data obtained by eddy currents variation under the influence of various types of designed artificial defects. Experimental samples were designed and prepared with SLM (selective laser melting) technology. Artificial defects, included in the samples, were detected using the eddy current testing device, taking the important circumstances of this non-destructive method into account. The presented research shows significant potential for eddy current testing to identify defects in AM products, with a resolution of various types and sizes of defects. The obtained data output shows the importance of choosing the right measurement regime, excitation frequency and secondary parameters setup. Besides the eddy current testing conditions, defect properties also play a significant role, such as their shape, size, if they are filled with unmolten powder or if they reach the surface.

Molybdenum disulfide (MoS 2 ) screen-printed working electrodes were developed for dopamine (DA) electrochemical sensing. MoS 2 working electrodes were prepared from high viscosity screen-printable inks containing various concentrations and sizes of MoS 2 particles and ethylcellulose binder. Rheological properties of MoS 2 inks and their suitability for screen-printing were analyzed by viscosity curve, screen-printing simulation and oscillatory modulus. MoS 2 inks were screen-printed onto conductive FTO (Fluorine-doped Tin Oxide) substrates. Optical microscopy and scanning electron microscopy with energy-dispersive X-ray spectroscopy (SEM/EDX) analysis were used to characterize the homogeneity, topography and thickness of the screen-printed MoS 2 electrodes. The electrochemical performance was assessed through differential pulse voltammetry. Results showed an extensive linear detection of dopamine from 1 µM to 300 µM (R 2  = 0.996, sensitivity of 5.00 × 10 -8 A μM -1 ), with the best limit of detection being 246 nM. This work demonstrated the possibility of simple, low-cost and rapid preparation of high viscosity MoS 2 ink and their use to produce screen-printed FTO/MoS 2 electrodes for dopamine detection., (© 2022. The development of cost-effective co-catalysts of high photocatalytic activity and recyclability is still a challenge in the energy transformation domain. In this study, 0D/2D Schottky heterojunctions, consisting of 0D ZnO and 2D Ti 3 C 2 , were successfully synthesized by the electrostatic self-assembling of ZnO nanoparticles on Ti 3 C 2 nanosheets. In constructing these heterojunctions, Ti 3 C 2 nanosheets acted as a co-catalyst for enhancing the transfer of excitons and their separation to support the photocatalytic response of ZnO. The as-prepared ZnO/Ti 3 C 2 composites demonstrate an abbreviated charge transit channel, a huge interfacial contact area and the interfacial electrons' transport potential. The extended optical response and large reactive area of the ZnO/Ti 3 C 2 composite promoted the formation of excitons and reactive sites on the photocatalyst's surface. The ZnO/Ti 3 C 2 Schottky heterojunction showed significantly high photocatalytic activity for hydrogen production from a water-ethanol solution under the light illumination in the visible region. The hydrogen evolution overoptimized the ZnO/Ti 3 C 2 composition with 30 wt.% of Ti 3 C 2 , which was eight times higher than the pristine ZnO. These findings can be helpful in developing 0D/2D heterojunction systems for photocatalytic applications by utilizing Ti 3 C 2 as a low-cost co-catalyst.

Modern manufacturing engineering requires quick and reasonable solutions during the production planning stage, ensuring production efficiency and cost reduction. This research aims to create a scientific approach to the rational choice of a locating chart for complexly shaped parts. It is an important stage during the manufacturing technology and fixture design process. The systematization of the designed and technological features of complexly shaped parts and the definition of the features that impact a locating chart create the fundamentals for justification. A scientific approach has been developed using the complex combination of the part's features and a decision-making approach using the example of bracket-type parts. The matrix of design and technological features of parts was developed including steel AISI 3135 and cast iron DIN 1691. The classification of locating charts for bracket-type parts was defined. A mathematical model of the rational choice of the locating chart according to the structural code of the workpiece was verified in case studies from the practice. As a result, a decision-making approach was applied to the rational choice of the locating chart for any bracket-type part. The proposed solutions improve the production planning stage for machine building, automotive, and other industries.

New screen-printed sensor with a boron-doped diamond working electrode (SP/BDDE) was fabricated using a large-area linear antenna microwave chemical deposition vapor system (LA-MWCVD) with a novel precursor composition. It combines the advantages of disposable printed sensors, such as tailored design, low cost, and easy mass production, with excellent electrochemical properties of BDDE, including a wide available potential window, low background currents, chemical resistance, and resistance to passivation. The newly prepared SP/BDDEs were characterized by scanning electron microscopy (SEM) and Raman spectroscopy. Their electrochemical properties were investigated by cyclic voltammetry and electrochemical impedance spectroscopy using inner sphere ([Fe(CN) 6 ] 4-/3- ) and outer sphere ([Ru(NH 3 ) 6 ] 2+/3+ ) redox probes. Moreover, the applicability of these new sensors was verified by analysis of the anti-inflammatory drug lornoxicam in model and pharmaceutical samples. Using optimized differential pulse voltammetry in Britton-Robinson buffer of pH 3, detection limits for lornoxicam were 9 × 10 -8 mol L -1 . The oxidation mechanism of lornoxicam was investigated using bulk electrolysis and online electrochemical cell with mass spectrometry; nine distinct reaction steps and corresponding products and intermediates were identified.

The purpose of the study was to determine the effect of three different levels of dietary linoleic acid (LA) intake on the growth of MDA-MB-435 human breast cancer cells in the mammary fat pads of nude mice, and their metastasis to the lungs. These diets were isocaloric, and contained different mixtures of safflower (LA-rich) and coconut (saturated fatty acid-rich) oils to provide 23% (w/w) total fat, with 2, 8, and 12% (w/w) LA. A fourth group was fed a low-fat, 5% (w/w) corn oil diet. There were 25 mice in each dietary group. A necropsy, 12 weeks after the tumor cell injections, the primary tumor weights in the 12% LA (4.1 +/- 2.7 g)- and 8% LA (3.5 +/- 1.7 g)-fed groups were significantly greater (P < 0.05) than those fed the 2% LA diet (2.5 +/- 1.5 g); they did not differ significantly from the weights of mammary fat pad tumors in the 5% corn oil-fed mice. The incidence of grossly visible pulmonary metastatic nodules was not significantly different between the 8 and 12% LA-fed mice, but was higher for both groups compared with the 2% LA-fed group (P < 0.05), with a similar trend in comparison with the 5% corn oil group. The mean total calculated volumes of the macroscopic metastases per tumor-bearing mouse were significantly greater in the 8 and 12% LA (157 +/- 250.7 and 99.1 +/- 140.0 mm3, respectively), compared with the 2% LA (23.3 +/- 51.8 mm3)- and 5% corn oil (24.5 +/- 35.1 mm3)-fed mice; all P < 0.05. Micrometastases were observed most frequently in the 5% corn oil and 2% LA dietary groups, but none of the differences were statistically significant. No differences were detected in the concentrations of prostaglandin E, leukotriene B4, or 5-hydroxyeicosatetraenoic acid in tumors from mice fed the four different diets.

Glandular kallikrein (a trypsin-like serine protease) is an estrogen-induced and dopamine-repressed protein in the rat anterior pituitary which appears to be associated with lactotrophs. This study examined glandular kallikrein levels in diethylstilbestrol (DES)-induced pituitary tumors in F344 rats and compared it to plasma and pituitary prolactin, and pituitary wet weight. Ovariectomized F344 rats were implanted with Silastic tubes containing 0 or 5 mg DES for 1, 3, 5, 7, or 9 weeks. Glandular kallikrein was measured by microenzymatic assay using D-valylleucylarginyl-p-nitroanilide following trypsin treatment of extracts to activate latent forms of glandular kallikrein. Prolactin was measured by radioimmunoassay. DES induced steady time-dependent increases in pituitary wet weight with 7- and 16-fold increases observed by 5 and 9 weeks, respectively. Growth rates averaged 11.4 mg/week during the first 5 weeks of DES exposure, and then increased to 23.2 mg/week between weeks 5 and 9. Glandular kallikrein total activity (nmol/min/pituitary) increased 130- and 240-fold after 3 and 5 weeks of DES exposure, respectively, and then abruptly plateaued. The specific activity (nmol/min/mg protein) of glandular kallikrein peaked at 3-5 weeks (36-fold increase compared to controls) and then declined as pituitary protein but not glandular kallikrein continued to increase. Total pituitary prolactin constantly rose during DES exposure with 12- and 26-fold increases after 5 and 9 weeks, respectively. Plasma prolactin levels also continuously rose during exposure to DES with 130- and 290-fold increases after 5 and 9 weeks, respectively. No major strain differences were found with regard to sensitivity to the acute effects of estrogen or dopaminergic stimulation on glandular kallikrein induction. DES-induced pituitary tumors in F344 rats are well known to arise via lactotroph proliferation, and the striking elevation in glandular kallikrein and prolactin during the early phases of tumor growth provide further support for a localization of glandular kallikrein in lactotrophs. However, the abrupt stabilization in glandular kallikrein levels by week 5 was unexpected and may signal a biochemical transformation of the tissue during tumor progression.

Lipoxygenase metabolites of arachidonic acid have been implicated in myocardial injury induced by coronary artery occlusion and reperfusion, as dual inhibitors of the lipoxygenase and cyclooxygenase enzymes, but not selective cyclooxygenase inhibitors, reduce infarct size. However, interpretation of these studies has been clouded by the lack of specificity of the drugs previously used. A specific 5-lipoxygenase inhibitor, REV-5901, has recently been developed. This drug inhibits A23187-induced immunoreactive leukotriene B4 generation by canine neutrophils (IC50 approximately 2.5 microM), and when given intravenously, attenuates the formation of a leukotriene D4-like material in blood ex vivo. The release of bioassayable leukotriene-like material from rabbit hearts infarcted in vivo and subsequently perfused in vitro is prevented by REV-5901. Moreover, the inhibitor also acts as an end-organ antagonist to prevent the spasmogenic effects of the peptide-containing leukotrienes in vitro with an IC50 approximately 0.1 microM. REV-5901 (10 + 2 mg/kg i.v.) reduces infarct size produced by coronary artery occlusion and reperfusion in the anesthetized dog from 56.6 +/- 2 to 28.6 +/- 3.7% of the hypoperfused zone. Salvage of the ischemic myocardium occurs independently of any apparent hemodynamic effect of the drug, but is accompanied by a diminution in neutrophil accumulation in the ischemic heart. It is proposed that inhibition of leukotriene B4 formation by REV-5901 suppresses the accumulation of neutrophils, thereby attenuating neutrophil-mediated cardiac damage.

Glandular kallikrein is a major estrogen-induced and dopamine-repressed protein of the rat anterior pituitary that appears to originate from lactotrophs. This study examined the development of glandular kallikrein levels in the anterior pituitary in both female and male rats and compared it to anterior pituitary prolactin. In addition, the development of glandular kallikrein levels in the neurointermediate lobe of the pituitary and the kidney were also examined. During puberty, a dramatic surge in glandular kallikrein occurred in female anterior pituitaries (16- to 20-fold increase) and levels remained elevated thereafter. The dynamics of the increase were biphasic--glandular kallikrein increased between Day 30 and 45, plateaued between Days 45 and 55, and then increased again between Days 55 and 65. Female anterior pituitary prolactin increased 7- to 8-fold during puberty. The rise during puberty was biphasic and was generally synchronized with increases in glandular kallikrein. However, the initial rise was proportionately less than that of glandular kallikrein, and the secondary surge was more dramatic. In contrast to females, anterior pituitary glandular kallikrein remained at low levels in male rats; prolactin levels also remained unchanged through puberty and increased moderately thereafter. Glandular kallikrein in the female neurointermediate lobe remained unchanged through Day 55, almost doubled on Day 60, and returned to prepubertal levels by Day 65; males did not exhibit the transient surge in neurointermediate lobe levels. Starting at age 60 days, renal glandular kallikrein was found to be slightly higher (15-20%) in females than in males.(ABSTRACT TRUNCATED AT 250 WORDS)

Recently, the metabolism of arachidonic acid to two unidentified products (Peak 1 and Peak 2) by a cytochrome P-450 dependent mixed function oxidase has been described in canine polymorphonuclear leukocytes (PMNs). This study assessed the biologic activity of one of these metabolites, Peak 2, on PMN function. Peak 2 was formed biologically following addition of exogenous arachidonic acid to canine PMNs pretreated with BW755c to inhibit lipoxygenase and cyclooxygenase enzymes, and purified by high performance liquid chromatography following separation by column chromatography. Peak 2 (20-200 ng/ml) inhibited calcium ionophore A23187-induced aggregation and the second phase of LTB4-induced aggregation. Additionally, Peak 2 inhibited A23187-induced PMN adhesion to columns of Sephadex G-25. BW755c (94 microM), which increased the formation of Peaks 1 and 2 by almost 300%, also inhibited A23187-induced PMN adhesion. In contrast, Peak 2 did not inhibit the release of superoxide anions or immunoreactive LTB4, after stimulation of the PMNs with A23187. Thus, Peak 2 may modulate some activities of canine PMNs. Because the biologic activity of Peak 2 is opposite to that of LTB4, which promotes PMN aggregation and adhesion, and because LTB4 may be metabolized by a cytochrome P-450-dependent mixed function oxidase to less active metabolites, this enzyme system may play a central role in the control of PMN function.
