Though with a less dramatic growth compared to photogrammetry, remote sensing from multispectral imagery taken by UAV (Unmanned Aerial Vehicles) platforms is applied to vegetation health monitoring, crop management, water quality assessments, geological inspections and much more, with a sizeable number of multispectral cameras now available on the market. As for satellite images, a key point in remote sensing is calibration, both geometric and radiometric, and the modelling of disturbances, to get well co-registered reflectance data. Leaving aside radiometric calibration, this paper focuses on how to best georeference the different bands one with respect to the other. This is normally achieved by the so-called band-to-band registration (BBR). Here, a straightforward approach is proposed, that exploits the multi-camera geometry and, unlike BBR, all the information contents of the bands, as inter-band matches are searched (and possibly found) for every pair of bands and not only between a reference and a slave band. Tests on images taken with the 9-band MAIA S2 camera are presented, discussing the pro and cons of pre-calibration and on-the-job calibration of the camera parameters of each sensor. The results found show that the proposed method is at least as good as the BBR ones.

Traditionally, data co-registration of survey epochs in photogrammetry relied on Ground Control Points (GCP) to keep the reference system unchanged. In the last years, Unmanned Aerial Systems (UAV) are increasingly used in photogrammetric environmental monitoring. The diffusion of affordable UAV platforms equipped with GNSS (Global Navigation Satellite System) centimetre-grade receivers might reduce, but not eliminate, the need for GCP. Conversely, if GNSS-assisted orientation cannot be used or if additional ground control and reliability checks are required, alternatives to repeated GCP survey have been proposed, taking advantage of Structure from Motion (SfM) photogrammetry. In particular, co-registering different epochs image blocks together, identifying corresponding features, has been demonstrated as a viable and efficient approach. In this paper four different strategies easily implementable in a generic commercial photogrammetric software are presented and compared considering three different test sites in Italy subject to different amounts of environmental changes. The influence of the amount and distribution of inter-epoch corresponding points on the accuracy of the reconstruction is investigated. The results show that some of the tested strategies obtains very good results and can be used (although not needed) also in RTK centimetre-grade UAV surveys, leveraging the additional information coming from previous epochs survey to actually increase the survey accuracy and reliability.

A dome-shape deformation has been found to affect the photogrammetric surface reconstruction in several real and simulated experiments. Its origin has been recognised in inaccurate estimation of the camera parameters and many papers already concentrated on conditions to avoid its development, especially as far as block design is concerned. This paper presents a Monte Carlo simulation to investigate surface reconstruction elevation errors in UAV (Unmanned Aerial Vehicle) photogrammetric blocks. The simulation tests are designed to find out the effect of block shape, camera axis inclination, side-lap, cross strips addition and block control by GCP or GNSS-assisted on the extent of the deformations. The main findings are: i) that GNSS-assisted blocks are generally more robust compared to GCP-controlled ones; ii) that, in GNSS-assisted blocks, unless a mix of nadiral and inclined strips is present, at least one fixed GCP must be provided; iii) that cross strip can conveniently be slimmed to save flight time and processing time; iv) that the effectiveness of GNSS deteriorate as the block shape slims out.

Unmanned Aerial Vehicles (UAV) are established platforms for photogrammetric surveys in remote areas. They are lightweight, easy to operate and can allow access to remote sites otherwise difficult (or impossible) to be surveyed with other techniques. Very good accuracy can be obtained also with low-cost UAV platforms as far as a reliable ground control is provided. However, placing ground control points (GCP) in these contexts is time consuming and requires accessibility that, in some cases, can be troublesome. RTK-capable UAV platforms are now available at reasonable costs and can overcome most of these problems, requiring just few (or none at all) GCP and still obtaining accurate results. The paper will present a set of experiments performed in cooperation with ARPA VdA (the Environmental Protection Agency of Valle d’Aosta region, Italy) on a test site in the Italian Alps using a Dji Phantom 4 RTK platform. Its goals are: a) compare accuracies obtainable with different calibration procedures (pre- or on-the-job/self-calibration); b) evaluate the accuracy improvements using different number of GCP when the site allows for it; and c) compare alternative positioning modes for camera projection centres determination, (Network RTK, RTK, Post Processing Kinematic and Single Point Positioning).

The research project ‘Moon Mapping’ has been established in 2014 between the Italian and Chinese Governments to promote cooperation and exchange between undergraduate students from both countries. The operational phase of the project started in early 2015, and will end in 2017, for a total length of three years. The main aim is to train new scholars to be able to work on different kinds of remotely-sensed data collected over the Moon surface by the Chinese space missions Chang’E-1/2. The project coordination has been assigned to the Italian Space Agency for the Italian side and to the Center of Space Exploration, China Ministry of Education, for the Chinese side. Several Chinese universities and Italian national research institutes and universities have been officially involved in this project. Six main research topics have been identified: (1) map of the solar wind ion; (2) geomorphological map of the Moon; (3) data preprocessing of Chang’E-1 mission; (4) map of element distribution; (5) establishment of 3D digital visualization system; and (6) compilation and publication of a tutorial on joint lunar mapping.

The current work investigates the potential of two low-cost off-the-shelf quadcopters for multi-view reconstruction of sub-vertical rock faces. The two platforms used are a DJI Phantom 1 equipped with a Gopro Hero 3+ Black and a DJI Phantom 3 Professional with integrated camera. The study area is a small sub-vertical rock face. Several flights were performed with both cameras set in time-lapse mode. Hence, images were taken automatically but the flights were performed manually as the investigated rock face is very irregular which required manual adjustment of the yaw and roll for optimal coverage. The digital images were processed with commercial SfM software packages. Several processing settings were investigated in order to find out the one providing the most accurate 3D reconstruction of the rock face. To this aim, all 3D models produced with both platforms are compared to a point cloud obtained with a terrestrial laser scanner. Firstly, the difference between the use of coded ground control targets and the use of natural features was studied. Coded targets generally provide the best accuracy, but they need to be placed on the surface, which is not always possible, as sub-vertical rock faces are not easily accessible. Nevertheless, natural features can provide a good alternative if wisely chosen as shown in this work. Secondly, the influence of using fixed interior orientation parameters or self-calibration was investigated. The results show that, in the case of the used sensors and camera networks, self-calibration provides better results. To support such empirical finding, a numerical investigation using a Monte Carlo simulation was performed.

A preliminary survey campaign is essential in projects of restoration, urban renewal, rebuilding or promotion of architectural heritage. Today several survey techniques allow full 3D object restitution and modelling that provides a richer description than simple 2D representations. However, the amount of data to collect increases dramatically and a trade-off between efficiency and productivity from one side and assuring accuracy and completeness of the results on the other must be found. Depending on the extent and the complexity of the task, a single technique or a combination of several ones might be employed. Especially when documentation at different scales and with different levels of detail are foreseen, the latter will likely be necessary. The paper describes two architectural surveys in Italy: the old village of Navelli (AQ), affected by the earthquake in 2009, and the two most relevant remains in Codiponte (MS), damaged by the earthquake in 2013, both in the context of a project of restoration and conservation. In both sites, a 3D survey was necessary to represent effectively the objects. An integrated survey campaign was performed in both cases, which consists of a GPS network as support for georeferencing, an aerial survey and a field survey made by laser scanner and close range photogrammetry. The two case studies, thanks to their peculiarities, can be taken as exemplar to wonder if the integration of different surveying techniques is today still mandatory or, considering the technical advances of each technology, it is in fact just optional.

The great amount of data that will be produced during the imaging of Mercury by the stereo camera (STC) of the BepiColombo mission needs a compromise with the restrictions imposed by the band downlink that could drastically reduce the duration and frequency of the observations. The implementation of an on-board real time data compression strategy preserving as much information as possible is therefore mandatory. The degradation that image compression might cause to the DTM accuracy is worth to be investigated. During the stereo-validation procedure of the innovative STC imaging system, several image pairs of an anorthosite sample and a modelled piece of concrete have been acquired under different illumination angles. This set of images has been used to test the effects of the compression algorithm (Langevin and Forni, 2000) on the accuracy of the DTM produced by dense image matching. Different configurations taking in account at the same time both the illumination of the surface and the compression ratio, have been considered. The accuracy of the DTMs is evaluated by comparison with a high resolution laser-scan acquisition of the same targets. The error assessment included also an analysis on the image plane indicating the influence of the compression procedure on the image measurements.

Photogrammetry has been used since long time to periodically control the evolution of landslides, either from aerial images as well as from ground. Landslides control and monitoring systems face a large variety of cases and situations: in hardly accessible environments, like glacial areas and high mountain locations, it is not simple finding a survey method and a measurement control system, which are capable to reliably assess, with low costs, the expected displacement and its accuracy. For this reason, the behaviour of these events presents the geologists and the surveyor each time with different challenges. The use of UAS (Unmanned Aerial System) represents, in this context, a recent and valid option to perform the data acquisition both in safety and quickly, avoiding hazards and risks for the operators while at the same time containing the costs. The paper presents an innovative monitoring system based on UAS-photogrammetry, GNSS survey and DSM change detection techniques to evaluate the Gran Sommetta rock glacier surface movements over the period 2012-2014. Since 2012, the surface movements of the glacier are monitored by ARPAVdA (a regional environmental protection agency) as a case study for the impact of climate change on high-mountain infrastructures. In such scenarios, in fact, a low-cost monitoring activity can provide important data to improve our knowledge about glacier dynamics connected to climate changes and to prevent risks in anthropic Alps areas. To evaluate the displacements of the rock glacier different techniques were proposed: the most reliable uses the orthophoto of the area and rely on a manual identification of corresponding features performed by a trained operator. To further limit the costs and improve the density of displacement information two automatic procedures were developed as well.

The growing use of UAS platform for aerial photogrammetry comes with a new family of Computer Vision highly automated processing software expressly built to manage the peculiar characteristics of these blocks of images. It is of interest to photogrammetrist and professionals, therefore, to find out whether the image orientation and DSM generation methods implemented in such software are reliable and the DSMs and orthophotos are accurate. On a more general basis, it is interesting to figure out whether it is still worth applying the standard rules of aerial photogrammetry to the case of drones, achieving the same inner strength and the same accuracies as well. With such goals in mind, a test area has been set up at the University Campus in Parma. A large number of ground points has been measured on natural as well as signalized points, to provide a comprehensive test field, to check the accuracy performance of different UAS systems. In the test area, points both at ground-level and features on the buildings roofs were measured, in order to obtain a distributed support also altimetrically. Control points were set on different types of surfaces (buildings, asphalt, target, fields of grass and bumps); break lines, were also employed. The paper presents the results of a comparison between two different surveys for DEM (Digital Elevation Model) generation, performed at 70 m and 140 m flying height, using a Falcon 8 UAS.

The paper describes the design and testing of a photogrammetric measurement protocol set up to determine the discrepancies between the planned and actual position of computer-guided template-based dental implants. Two moulds with the implants positioned in pre- and post- intervention are produced and separately imaged with a highly redundant block of convergent images; the model with the implants is positioned on a steel frame with control points and with suitable targets attached. The theoretical accuracy of the system is better than 20 micrometers and 0.3–0.4° respectively for positions of implants and directions of implant axes. In order to compare positions and angles between the planned and actual position of an implant, coordinates and axes directions are brought to a common reference system with a Helmert transformation. A procedure for comparison of positions and directions to identify out-of-tolerance discrepancies is presented; a numerical simulation study shows the effectiveness of the procedure in identifying the implants with significant discrepancies between pre- and post- intervention.

Photogrammetry has been used since long to periodically control the evolution of landslides; however, true monitoring is reserved to robotic total stations and ground based InSAR systems, capable of high frequency, high accurate 24h/day response. This paper presents the first results of a fixed terrestrial stereo photogrammetric system developed to monitor shape changes of the scene. The system is made of two reflex cameras, each contained in a sealed box with a control computer that periodically acquires an image and send it to a host computer; once an image pair is received from the two cameras, the DSM of the scene is generated by image correlation and made available for archiving or analysis. The system has been installed and is being tested on the Mont de la Saxe landslide, where several monitoring system are active. Some instability of the camera attitude has been noticed and is corrected with an automated procedure. First comparisons with InSAR data show a good agreement.

Nowadays, the scientific institutions involved in planetary mapping are working on new strategies to produce accurate high resolution DTMs from space images at planetary scale, usually dealing with extremely large data volumes. From a methodological point of view, despite the introduction of a series of new algorithms for image matching (e.g. the Semi Global Matching) that yield superior results (especially because they produce usually smooth and continuous surfaces) with lower processing times, the preference in this field still goes to well established area-based matching techniques. Many efforts are consequently directed to improve each phase of the photogrammetric process, from image pre-processing to DTM interpolation. In this context, the Dense Matcher software (DM) developed at the University of Parma has been recently optimized to cope with very high resolution images provided by the most recent missions (LROC NAC and HiRISE) focusing the efforts mainly to the improvement of the correlation phase and the process automation. Important changes have been made to the correlation algorithm, still maintaining its high performance in terms of precision and accuracy, by implementing an advanced version of the Least Squares Matching (LSM) algorithm. In particular, an iterative algorithm has been developed to adapt the geometric transformation in image resampling using different shape functions as originally proposed by other authors in different applications.

A large number of strategies and implementations of Structure from Motion (SFM) have been developed; some are being included in photogrammetric software packages. However, finding specific parameters highlighting the efficiency, the accuracy and the reliability of a SFM strategy it is difficult. While most strategies will almost always succeed in any given task (i.e. the orientation of a certain image sequence), judging how good is actually the result, is somehow still an open issue. In the first part of this paper some alternatives and parameters are presented and their pro and cons discussed, with the objective to find out whether there are alternatives to the well-established method of the check points. In the second part, the results of most of the image sequences made available by the workshop organizers, processed with the SFM approach developed by the authors, are presented; results for the sequences where external information (such as ground control points or a DTM) were available are presented in more detail.

Automatic orientation of image sequences in close range photogrammetry is becoming more and more important, not least to maintain a degree of competitiveness with other survey techniques, such as laser scanning. The objective of this paper is to compare two Structure from Motion (SFM) strategies. The previous strategy has been used at our Department for some years already in a wide range of projects and is based on the Harris operator and the fundamental matrix plus the trifocal tensor estimation to filter out the outliers. While it has in most cases performed satisfactorily, the percentage of accepted matches is generally smaller than expected; sometimes this leads to failure of the successful estimation of the trifocal tensor. The second one has only recently been implemented and is still under testing; it is based on the SURF operator and the 5-point relative orientation algorithm. The paper will show a comparison between the two strategies on a series of test cases.

In the generation of Digital Terrain Models (DTMs) from space images area-based image matching techniques are used in most cases; the image matching strategy, though, is to be tailored to the specific mission characteristics. In this paper, a series of tests have been performed with real and synthetic space images to study the performance of our implementation of the Least Squares Matching algorithm. To this aim, patches extracted from a pair of Hirise images were processed with our DTM generation software Dense Matcher (DM) as well as with Ames Stereo Pipeline (ASP) and the results compared to a reference DTM; moreover, synthetic images, generated from the reference DTM and overlaid with artificial texture ideal for image matching, were processed as well, to discriminate the influence of texture and surface topography on the DTM accuracy. With real images, both software show RMSE ranging from 15 to 38 cm; with synthetic images, processed only with DM, RMSE ranges from 12 to 18 cm.

In mining operations the evaluation of the stability condition of the excavated front are critic to ensure a safe and correct planning of the subsequent activities. The procedure currently used to this aim has some shortcomings: safety for the geologist, completeness of data collection and objective documentation of the results. In the last decade it has been shown that the geostructural parameters necessary to the stability analysis can be derived from high resolution digital surface models (DSM) of rock faces. With the objective to overcome the limitation of the traditional survey and to minimize data capture times, so reducing delays on mining site operations, a photogrammetric system to generate high resolution DSM of tunnels has been realized. A fast, effective and complete data capture method has been developed and the orientation and restitution phases have been largely automated. The survey operations take no more than required to the traditional ones; no additional topographic measurements other than those available are required. To make the data processing fast and economic our Structure from Motion procedure has been slightly modified to adapt to the peculiar block geometry while, the DSM of the tunnel is created using automatic image correlation techniques. The geomechanical data are sampled on the DSM, by using the acquired images in a GUI and a segmentation procedure to select discontinuity planes. To allow an easier and faster identification of relevant features of the surface of the tunnel, using again an automatic procedure, an orthophoto of the tunnel is produced. A case study where a tunnel section of ca. 130 m has been surveyed is presented.

Objectives Adult T-cell leukemia-lymphoma (ATL), is a highly malignant T-cell neoplasm caused by human T-cell leukemia virus type 1 (HTLV-1), chSaracterized by poor prognosis. Two viral proteins, Tax-1 and HTLV-1 basic-zipper factor (HBZ) play important roles in the pathogenesis of ATL. While Tax-1 can be found in both the cytoplasm and nucleus of HTLV-1 infected patients, HBZ is exclusively localized in the cytoplasm of HTLV-1 asymptomatic carriers and in patients with the chronic neurologic disease HTLV-I-associated myelopathy/tropical spastic paraparesis (HAM/TSP). HBZ is only localized in the nucleus of ATL cell lines, suggesting that the nuclear localization of HBZ can be a hallmark of neoplastic transformation. Methods In order to clarify this crucial point, we investigated in detail the pattern of HBZ expression in ATL patients. We made use of our monoclonal antibody 4D4-F3, that at present is the only reported reagent, among the few described, able to detect endogenous HBZ by immunofluorescence and confocal microscopy in cells from asymptomatic carriers, HAM/TSP and ATL patients. Results We found that HBZ is localized both in the cytoplasm and nucleus of cells of ATL patients irrespective of their clinical status, with a strong preference for the cytoplasmic localization. Also Tax-1 is localized in both compartments. Conclusions As HBZ is exclusively localized in the cytoplasm in asymptomatic carriers and in non-neoplastic pathologies, this finding shows that neoplastic transformation consequent to HTLV-1 infection is accompanied and associated with the capacity of HBZ to translocate to the nucleus, which suggests a role of cytoplasmic-to-nuclear translocation in HTLV-1-mediated oncogenesis. Objectives Adult T-cell leukemia/lymphoma (ATL) is a T-cell lymphoproliferative neoplasm caused by the human T-cell leukemia virus type 1 (HTLV-1). Two viral proteins, Tax-1 and HBZ play important roles in HTLV-1 infectivity and in HTLV-1-associated pathologies by altering key pathways of cell homeostasis. However, the molecular mechanisms through which the two viral proteins, particularly HBZ, induce and/or sustain the oncogenic process are still largely elusive. Previous results suggested that HBZ interaction with nuclear factors may alter cell cycle and cell proliferation. Methods We investigated the endogenous HBZ interactome in leukemic cells by immunoprecipitating the HBZ-interacting complexes of ATL-2 leukemic cells, followed by tandem mass spectrometry analyses. RNA seq analysis was performed to decipher the differential gene expression and splicing modifications related to HTLV-1. Here we compared ATL-2 with MOLT-4, a non HTLV-1 derived leukemic T cell line and further compared with HBZ-induced modifications in an isogenic system composed by Jurkat T cells and stably HBZ transfected Jurkat derivatives. Results The endogenous HBZ interactome of ATL-2 cells identified 249 interactors covering three main clusters corresponding to protein families mainly involved in mRNA splicing, nonsense-mediated RNA decay (NMD) and JAK-STAT signaling pathway. Here we analyzed in detail the cluster involved in RNA splicing. RNAseq analysis showed that HBZ specifically altered the transcription of many genes, including crucial oncogenes, by affecting different splicing events. Consistently, the two RNA helicases, DDX5 and its paralog DDX17, recently shown to be involved in alternative splicing of cellular genes after NF-κB activation by HTLV1 Tax-1, interacted and partially co-localized with HBZ. Conclusion For the first time, a complete picture of the endogenous HBZ interactome was elucidated. The wide interaction of HBZ with molecules involved in RNA splicing and the subsequent transcriptome alteration strongly suggests an unprecedented complex role of the viral oncogene in the establishment of the leukemic state. Background/objectives: New SARS-CoV-2 variants are continuously emerging, making it essential to assess the efficacy of vaccine-induced immune protection. Limited information is available regarding T cell responses to BA.2.86 and JN.1 variants, particularly in elderly individuals., Methods: We evaluated T cell and total IgG responses against the receptor-binding domain (RBD) of the ancestral SARS-CoV-2 strain, as well as BA.2.86 and JN.1 omicron subvariants, in two groups of subjects. One group consisted of SARS-CoV-2-exposed elderly individuals who were fully vaccinated with the BNT162B2 mRNA vaccine, with a booster dose of the updated 2023-2024 COVID-19 vaccine (XBB.1.5) at least 15 days after receiving a booster dose of the updated 2023-2024 COVID-19 vaccine. The second group consisted of healthcare workers who were unexposed to SARS-CoV-2 one month after the booster dose of the first-generation BNT162b2 mRNA vaccine. T cell activation-induced markers (AIM) and IFN-γ secretion were evaluated by flow cytometry and ELISpot assays, respectively., Results: Elderly subjects showed reduced IgG levels against JN.1 compared with the ancestral strain. BA.2.86 stimulation resulted in lower IFN-γ levels in the elderly versus the COVID-19-naïve group. AIM analysis showed that among T cells, CD4+ were the most responsive, with a reduced proportion of JN.1-reactive CD4+ T cells compared with the ancestral strain in the SARS-CoV-2-unexposed group. Despite receiving the updated booster, the elderly group showed reduced CD4+ T cell reactivity to BA.2.86., Conclusions: The XBB.1.5-containing vaccine induced lower CD4+ T cell responses against BA.2.86 in the elderly. CD4+ T cells from BNT16b2-vaccinated, COVID-19-naïve subjects recognized ancestral and BA.2.86 RBD strains while showing reduced responses to JN.1. These results emphasize the need for tailored vaccine strategies for emerging variants, particularly in vulnerable populations.

Background: Glioblastoma (GBM) is the most frequent and aggressive brain tumor in adults with the lowest survival rates five years post-diagnosis. Oncolytic viruses (OVs) selectively target and damage cancer cells, and for this reason they are being investigated as new therapeutic tools also against GBM., Methods: An oncolytic herpes simplex virus type 1 (oHSV-1) with deletions in the γ34.5 neurovirulence gene and the US12 gene, expressing enhanced green fluorescent protein (EGFP-oHSV-1) as reporter gene was generated and tested for its capacity to infect and kill the murine GL261 glioblastoma (GBM) cell line. Syngeneic mice were orthotopically injected with GL261cells. Seven days post-implantation, EGFP-oHSV-1 was administered intratumorally. Twenty-one days after parental tumor challenge in the opposite brain hemisphere, mice were sacrified and their brains were analysed by immunohistochemistry to assess tumor presence and cell infiltrate., Results: oHSV-1 replicates and induces cell death of GL261 cells in vitro. A single intracranial injection of EGFP-oHSV-1 in established GL261 tumors significantly prolongs survival in all treated mice compared to placebo treatment. Notably, 45% of treated mice became long-term survivors, and rejected GL261 cells upon rechallenge in the contralateral brain hemisphere, indicating an anamnestic antitumoral immune response. Post-mortem analysis revealed a profound modification of the tumor microenvironment with increased infiltration of CD4 + and CD8 + T lymphocytes, intertumoral vascular collapse and activation and redistribution of macrophage, microglia, and astroglia in the tumor area, with the formation of intense fibrotic tissue suggestive of complete rejection in long-term survivor mice., Conclusions: EGFP-oHSV1 demonstrates potent antitumoral activity in an immunocompetent GBM model as a monotherapy, resulting from direct cell killing combined with the stimulation of a protective adaptive immune response. The different susceptibility to HIV-1 infection in U937 cells-permissive (Plus) or nonpermissive (Minus)-is linked to the expression in Minus cells of interferon (IFN)-γ inducible antiviral factors such as tripartite motif-containing protein 22 (TRIM22) and class II transactivator (CIITA). CIITA interacts with Cyclin T1, a key component of the Positive-Transcription Elongation Factor b (P-TEFb) complex needed for the efficient transcription of HIV-1 upon interaction with the viral transactivator Tat. TRIM22 interacts with CIITA, recruiting it into nuclear bodies together with Cyclin T1. A 50 kDa Cyclin T1 was found only in Minus cells, alongside the canonical 80 kDa protein. The expression of this truncated form remained unaffected by proteasome inhibitors but was reduced by IFNγ treatment. Unlike the nuclear full-length protein, truncated Cyclin T1 was also present in the cytoplasm, and this subcellular localization correlated with its capacity to inhibit Tat-mediated HIV-1 transcription. The 50 kDa Cyclin T1 in Minus cells likely contributes to their non-permissive phenotype by acting as a dominant negative factor, disrupting P-TEFb complex formation and function. Its reduction upon IFNγ treatment suggests a regulatory loop by which its inhibitory role on HIV-1 replication is then exerted by the IFNγ-induced CIITA, which binds to the canonical Cyclin T1, displacing it from the P-TEFb complex.

In empirical art research, understanding how viewers judge visual artworks as beautiful is often explored through the study of attributes-specific inherent characteristics or artwork features such as color, complexity, and emotional expressiveness. These attributes form the basis for subjective evaluations, including the judgment of beauty. Building on this conceptual framework, our study examines the beauty judgments of 54 Western artworks made by native Japanese and German speakers, utilizing an extreme randomized trees model-a data-driven machine learning approach-to investigate cross-cultural differences in evaluation behavior. Our analysis of 17 attributes revealed that visual harmony, color variety, valence, and complexity significantly influenced beauty judgments across both cultural cohorts. Notably, preferences for complexity diverged significantly: while the native Japanese speakers found simpler artworks as more beautiful, the native German speakers evaluated more complex artworks as more beautiful. Further cultural distinctions were observed: for the native German speakers, emotional expressiveness was a significant factor, whereas for the native Japanese speakers, attributes such as brushwork, color world, and saturation were more impactful. Our findings illuminate the nuanced role that cultural context plays in shaping aesthetic judgments and demonstrate the utility of machine learning in unravelling these complex dynamics. Oral Squamous Cell Carcinoma (OSCC) is the most common malignant tumor of the oral cavity. Despite recent advances in the field of oral cancer therapy, including the introduction of immunotherapeutic approaches, the 5-year survival rate remains steadily assessed around 50%. Thus, there is an urgent need for new therapeutic strategies. After the characterization of the immune phenotype of three human OSCC cell lines (CAL-27, SCC-25, and SCC-4) and one mouse OSCC cell line (MOC2) showing their similarities to resected patient tumors, we explored for the first time an experimental preclinical model of therapeutic vaccination with mouse OSCC MOC2 cell line stably expressing MHC class II antigens after CIITA gene transfection (MOC2-CIITA). Mice injected with MOC2-CIITA reject or strongly retard tumor growth; more importantly, vaccinated animals that fully reject MOC2-CIITA tumors display anti-tumor immunological memory protective against challenge with parental MOC2 tumor cells. Further experiments of adoptive cell transfer or in vivo cell depletion show that both CD4 + and CD8 + T lymphocytes prove fundamental in tumor rejection. Metabolism and biological functions of the nitrogen-rich compound guanidine have long been neglected. The discovery of four classes of guanidine-sensing riboswitches and two pathways for guanidine degradation in bacteria hint at widespread sources of unconjugated guanidine in nature. So far, only three enzymes from a narrow range of bacteria and fungi have been shown to produce guanidine, with the ethylene-forming enzyme (EFE) as the most prominent example. Here, we show that a related class of Fe 2+ - and 2-oxoglutarate-dependent dioxygenases (2-ODD-C23) highly conserved among plants and algae catalyze the hydroxylation of homoarginine at the C6-position. Spontaneous decay of 6-hydroxyhomoarginine yields guanidine and 2-aminoadipate-6-semialdehyde. The latter can be reduced to pipecolate by pyrroline-5-carboxylate reductase but more likely is oxidized to aminoadipate by aldehyde dehydrogenase ALDH7B in vivo . Arabidopsis has three 2-ODD-C23 isoforms, among which Din11 is unusual because it also accepted arginine as substrate, which was not the case for the other 2-ODD-C23 isoforms from Arabidopsis or other plants. In contrast to EFE, none of the three Arabidopsis enzymes produced ethylene. Guanidine contents were typically between 10 and 20 nmol*(g fresh weight) -1 in Arabidopsis but increased to 100 or 300 nmol*(g fresh weight) -1 after homoarginine feeding or treatment with Din11-inducing methyljasmonate, respectively. In 2-ODD-C23 triple mutants, the guanidine content was strongly reduced, whereas it increased in overexpression plants. The incorporation of non-canonical amino acids (ncAAs) into proteins is a powerful technique used in various research fields. Genetic code expansion (GCE) is the most common way to achieve this: a specific codon is selected to be decoded by a dedicated tRNA orthogonal to the endogenous ones. In the past 30 years, great progress has been made to obtain novel tRNA synthetases (aaRSs) accepting a variety of ncAAs with distinct physicochemical properties, to develop robust in vitro assays or approaches for codon reassignment. This sparked the use of the technique, leading to the accumulation of publications, from which gathering all relevant information can appear daunting. Here we present iNClusive (https://non-canonical-aas.biologie.uni-freiburg.de/), a manually curated, extensive repository using standardized nomenclature that provides organized information on ncAAs successfully incorporated into target proteins as verified by mass spectrometry. Since we focused on tRNA synthetase-based tRNA loading, we provide the sequence of the tRNA and aaRS used for the incorporation. Derived from more than 687 peer-reviewed publications, it currently contains 2432 entries about 466 ncAAs, 569 protein targets, 500 aaRSs and 144 tRNAs. We investigated the basis for better performance of transgenic Nicotiana tabacum plants with G6PDH-isoenzyme replacement in the cytosol (Xanthi::cP2::cytRNAi, Scharte et al., 2009). After six generations of selfing, infiltration of Phytophthora nicotianae zoospores into source leaves confirmed that defence responses (ROS, callose) are accelerated, showing as fast cell death of the infected tissue. Yet, stress-related hormone profiles resembled susceptible Xanthi and not resistant cultivar SNN, hinting at mainly metabolic adjustments in the transgenic lines. Leaves of non-stressed plants contained twofold elevated fructose-2,6-bisphosphate (F2,6P 2 ) levels, leading to partial sugar retention (soluble sugars, starch) and elevated hexose-to-sucrose ratios, but also more lipids. Above-ground biomass lay in between susceptible Xanthi and resistant SNN, with photo-assimilates preferentially allocated to inflorescences. Seeds were heavier with higher lipid-to-carbohydrate ratios, resulting in increased harvest yields - also under water limitation. Abiotic stress tolerance (salt, drought) was improved during germination, and in floated leaf disks of non-stressed plants. In leaves of salt-watered plants, proline accumulated to higher levels during illumination, concomitant with efficient NADP(H) use and recycling. Non-stressed plants showed enhanced PSII-induction kinetics (upon dark-light transition) with little differences at the stationary phase. Leaf exudates contained 10% less sucrose, similar amino acids, but more fatty acids - especially in the light. Export of specific fatty acids via the phloem may contribute to both, earlier flowering and higher seed yields of the Xanthi-cP2 lines. Apparently, metabolic priming by F2,6P 2 -combined with sustained NADP(H) turnover-bypasses the genetically fixed growth-defence trade-off, rendering tobacco plants more stress-resilient and productive., (© 2023 The Authors. Human nucleotide-binding oligomerization domain (NOD)-like receptors (NLR) include a large family of proteins that have important functions in basic physio-pathological processes like inflammation, cell death and regulation of transcription of key molecules for the homeostasis of the immune system. They are all characterized by a common backbone structure (the STAND ATPase module consisting in a nucleotide-binding domain (NBD), an helical domain 1 (HD1) and a winged helix domain (WHD), used by both prokaryotes and eukaryotes as defense mechanism. In this review, we will focus on the MHC class II transactivator (CIITA), the master regulator of MHC class II (MHC-II) gene expression and the founding member of NLR. Although a consistent part of the described NLR family components is often recalled as innate or intrinsic immune sensors, CIITA in fact occupies a special place as a unique example of regulator of both intrinsic and adaptive immunity. Although the safety and efficacy of COVID-19 vaccines in older people are critical to their success, little is known about their immunogenicity among elderly residents of long-term care facilities (LTCFs). A single-center prospective cohort study was conducted: a total IgG antibody titer, neutralizing antibodies against Wild-type, Delta Plus, and Omicron BA.2 variants and T cell response, were measured eight months after the second dose of BNT162b2 vaccine (T0) and at least 15 days after the booster (T1). Forty-nine LTCF residents, with a median age of 84.8 ± 10.6 years, were enrolled. Previous COVID-19 infection was documented in 42.9% of the subjects one year before T0. At T1, the IgG titers increased up to 10-fold. This ratio was lower in the subjects with previous COVID-19 infection. At T1, IgG levels were similar in both groups. The neutralizing activity against Omicron BA.2 was significantly lower (65%) than that measured against Wild-type and Delta Plus (90%). A significant increase of T cell-specific immune response was observed after the booster. Frailty, older age, sex, cognitive impairment, and comorbidities did not affect antibody titers or T cell response. In the elderly sample analyzed, the BNT162b2 mRNA COVID-19 vaccine produced immunogenicity regardless of frailty.

Aromatic amino acid homeostasis was investigated in cell suspension cultures of Nicotiana plumbaginifolia and was related to the activity of the first enzyme in aromatic biosynthesis, 3-deoxy-D- arabino -heptulosonate-7-phosphate (DAHP) synthase. An inverse relationship was found between the intracellular content of free phenylalanine, tyrosine and tryptophan and enzyme specific activity levels, suggesting the occurrence of end-product control mechanisms. Two DAHP synthase isogenes are present in wild tobacco that showed a different expression pattern during the culture growth cycle. Intracellular levels of aromatic amino acids were increased or decreased by adding the culture medium with phenylalanine, tyrosine and tryptophan, or with sublethal doses of the shikimate pathway inhibitor glyphosate, respectively. As a consequence, enzyme levels varied in the opposite direction. The concomitant exogenous supply of glutamine further reduced enzyme activity in mid-log cells, suggesting induction by both aromatic amino acid depletion and nitrogen starvation.

Sucrose is essential for plants for several reasons: It is a source of energy, a signaling molecule, and a source of carbon skeletons. Sucrose phosphate synthase (SPS) catalyzes the conversion of uridine diphosphate glucose and fructose-6-phosphate to sucrose-6-phosphate, which is rapidly dephosphorylated by sucrose phosphatase. SPS is critical in the accumulation of sucrose because it catalyzes an irreversible reaction. In Arabidopsis thaliana , SPSs form a gene family of four members, whose specific functions are not clear yet. In the present work, the role of SPSA2 was investigated in Arabidopsis under both control and drought stress conditions. In seeds and seedlings, major phenotypic traits were not different in wild-type compared with spsa2 knockout plants. By contrast, 35-day-old plants showed some differences in metabolites and enzyme activities even under control conditions. In response to drought, SPSA2 was transcriptionally activated, and the divergences between the two genotypes were higher, with spsa2 showing reduced proline accumulation and increased lipid peroxidation. Total soluble sugars and fructose concentrations were about halved compared with wild-type plants, and the plastid component of the oxidative pentose phosphate pathway was activated. Unlike previous reports, our results support the involvement of SPSA2 in both carbon partitioning and drought response.

Glioblastoma is the most malignant tumor of the central nervous system. Current treatments based on surgery, chemotherapy, and radiotherapy, and more recently on selected immunological approaches, unfortunately produce dismal outcomes, and less than 2% of patients survive after 5 years. Thus, there is an urgent need for new therapeutic strategies. Here, we report unprecedented positive results in terms of protection from glioblastoma growth in an animal experimental system after vaccination with glioblastoma GL261 cells stably expressing the MHC class II transactivator CIITA. Mice injected with GL261-CIITA express de novo MHC class II molecules and reject or strongly retard tumor growth as a consequence of rapid infiltration with CD4+ and CD8+ T cells. Importantly, mice vaccinated with GL261-CIITA cells by injection in the right brain hemisphere strongly reject parental GL261 tumors injected in the opposite brain hemisphere, indicating not only the acquisition of anti-tumor immune memory but also the capacity of immune T cells to migrate within the brain, overcoming the blood-brain barrier. GL261-CIITA cells are a potent anti-glioblastoma vaccine, stimulating a protective adaptive anti-tumor immune response in vivo as a consequence of CIITA-driven MHC class II expression and consequent acquisition of surrogate antigen-presenting function toward tumor-specific CD4+ Th cells. Background: To date, only a few studies reported data regarding the development of mucosal immune response after the BNT162b2-booster vaccination., Methods: Samples of both serum and saliva of 50 healthcare workers were collected at the day of the booster dose (T3) and after two weeks (T4). Anti-S1-protein IgG and IgA antibody titres and the neutralizing antibodies against the Wuhan wild-type Receptor-Binding Domain in both serum and saliva were measured by quantitative and competitive ELISA, respectively. Data were compared with those recorded after the primary vaccination cycle (T2). Neutralizing antibodies against the variants of concern were measured in those individuals with anti-Wuhan neutralizing antibodies in their saliva., Findings: After eight months from the second dose, IgG decreased in both serum (T2 GMC : 23,838.5 ng/ml; T3 GMC : 1473.8 ng/ml) and saliva (T2 GMC : 12.9 ng/ml; T3 GMC : 0.3 ng/ml). Consistently, serum IgA decreased (T2 GMC : 48.6 ng/ml; T3 GMC : 6.4 ng/ml); however, salivary IgA showed a different behaviour and increased (T2 GMC : 0.06 ng/ml; T3 GMC : 0.41 ng/ml), indicating a delayed activation of mucosal immunity. The booster elicited higher titres of both IgG and IgA when compared with the primary cycle, in both serum (IgG T4 GMC : 98,493.9 ng/ml; IgA T4 GMC : 187.5 ng/ml) and saliva (IgG T4 GMC : 21.9 ng/ml; IgA T4 GMC : 0.65 ng/ml). Moreover, the booster re-established the neutralizing activity in the serum of all individuals, not only against the Wuhan wild-type antigen (N = 50; INH: 91.6%) but also against the variants (Delta INH: 91.3%; Delta Plus INH: 89.8%; Omicron BA.1 INH: 85.1%). By contrast, the salivary neutralizing activity was high against the Wuhan antigen in 72% of individuals (N = 36, INH: 62.2%), but decreased against the variants, especially against the Omicron BA.1 variant (Delta N = 27, INH: 43.1%; Delta Plus N = 24, INH: 35.2%; Omicron BA.1 N = 4; INH: 4.7%). The enzyme that, in Saccharomyces cerevisiae, catalyzes the last step in both proline synthesis and arginine catabolism, δ 1 -pyrroline-5-carboxylate (P5C) reductase, was purified to near homogeneity and characterized thoroughly. Retention patterns upon gel permeation chromatography were consistent with a homodecameric composition of the holomer. High lability of the purified preparations and stabilization by reducing compounds suggested susceptibility to reactive-oxygen-species-mediated damage. Both NADH and NADPH were used as the electron donor, the latter resulting in a 3-fold higher V max . However, a higher affinity toward NADH was evident, and the NADPH-dependent activity was inhibited by NAD + , proline, arginine, and a variety of anions. With proline and arginine, the inhibition was of the competitive type with respect to the specific substrate, and of the uncompetitive- or mixed-type with respect to NADPH, respectively. The results suggest that, contrary to the enzyme from higher plants, yeast P5C reductase may preferentially use NADH in vivo. An in silico analysis was also performed to investigate the structural basis of such enzyme features. Superposition of the protein model with the experimental structure of P5C reductase from Medicago truncatula allowed us to hypothesize on the possible allosteric sites for arginine and anion binding, and the cysteine pairs that may be involved in disulfide formation.
