Deep Eutectic Solvents (DES) have emerged as an alternative for many applications in different chemical sectors (to be used during the upstream and downstream processing, or as performance additives). While traditionally coined as green solvents, the petrochemical and energy-demanding origin of some DES components, together with some reported toxicological data, have been often overlooked. This perspective discusses the possible fate of DES once they have been used as synthetic reaction media, particularly related to the downstream unit to recover the product from the reaction mixture, and to the final DES disposal. The Total Carbon Dioxide Release (TCR) (measured as kg CO2 per kg product) is used to compare different options. After a downstream processing to recover the product often involving an organic solvent which will be incinerated (producing CO2), the used DES media can either be incinerated or diluted to some degree to be divested to a Wastewater Treatment Plant (WWTP). A mild wastewater treatment - involving state-of-the-art microbial processing steps, appears more promising than the incineration option, both in terms of CO2 production, as well as to avoid the potential formation of halide compounds (e.g. from chloride) during the incineration. However, to reach the WWTP, a key factor is the dilution degree of the DES, and the biodegradability that DES (components) may display by wastewater microorganisms. At a range of 1 : 20 dilution, a production of ∼16 kg CO2 per kg product may be expected in the WWTP, for a synthesis in DES containing 100 g substrate loading per L. Research is urgently needed to assess whether the treatment of DES as diluted wastewater may be a sustainable (and economic) solution for the DES fate.

Biocatalysis holds immense potential for pharmaceutical development as it enables synthetic routes to various chiral building blocks with unparalleled selectivity. Therein, solvent and water use account for a large contribution to the environmental impact of the reactions. In the spirit of Green Chemistry, a transition from traditional highly diluted aqueous systems to intensified non-aqueous media to overcome limitations (e.g., water shortages, recalcitrant wastewater treatments, and low substrate loadings) has been observed. Benefiting from the spectacular advances in various enzyme stabilization techniques, a plethora of biotransformations in non-conventional media have been established. Deep eutectic solvents (DESs) emerge as a sort of (potentially) greener non-aqueous medium with increasing use in biocatalysis. This review discusses the state-of-the-art of biotransformations in DESs with a focus on biocatalytic pathways for the synthesis of active pharmaceutical ingredients (APIs). Representative examples of different enzyme classes are discussed, together with a critical vision of the limitations and discussing prospects of using DESs for biocatalysis.

The outstanding developments in molecular biology techniques allow nowadays the screening, large-scale production, and designing of biocatalysts, adapting them to desired reactions. Many enzymes can perform reactions both in aqueous and non-aqueous media, broadening even further the opportunities to integrate them in complex pharmaceutical multi-step syntheses. Areas covered This paper showcases several examples of biocatalysis in the pharmaceutical industry, covering examples of different enzymes, such as lipases, oxidoreductases, and transaminases, to deliver active drugs through complex synthetic routes. Examples are critically discussed in terms of reaction conditions, motivation for using an enzyme, and how biocatalysts can be integrated in multi-step syntheses. When possible, biocatalytic routes are benchmarked with chemical reactions. Expert Opinion The reported enzymatic examples are performed with high substrate loadings (>100 g L−1) and with excellent selectivity, making them inspiring strategies for present and future industrial applications. Sugar (fatty acid) esters are industrially relevant compounds, with a cumbersome production process due to the solubility issues of the substrates, which forces the use of environmentally unfriendly reaction media. Herein, deep eutectic solvents (DESs) are considered as a promising solution: several literature examples use glucose and different acyl donors to illustrate the efficient synthesis of sugar esters in classic DESs like choline chloride/urea (ChCl/U). However, this paper discloses that when sugars like lactose or other disaccharides are used, enzymes cannot efficiently perform (trans)esterifications in DESs, while the same reaction can proceed in mixtures like pyridine/tetrahydrofuran (Py/THF). This could be explained by computational solubility studies and molecular dynamics simulations of both reaction media, showing two effects: (i) on the one hand, large acyl donors (more than C10) display poor solubility in DESs and (ii) on the other hand, disaccharides interact with DES components. Thus, the DES affects the conformation of lactose (compared to the conformation observed in the Py/THF mixture), in such a way that the enzymatic reaction results impaired. Despite that classic DESs (e.g., ChCl/U) may not be useful for generalizing their use in saccharide ester syntheses, the achieved theoretical understanding of the reaction may enable the design of future DESs that can combine enzyme compatibility with eco-friendliness and efficiency in sugar chemistry.

The use of oxidoreductases in organic-aqueous biphasic systems is advantageous (effective solvation of reactants, minimization of substrate/product-induced inhibition, improved volumetric productivity, and straightforward downstream processing). This paper explores the effects of organic solvents on horse liver alcohol dehydrogenase (HLADH) by combining experimental and computational studies. Various organic solvents displaying a broad range of hydrophobicity and functionalities are used, namely, ethyl acetate, 2-methyltetrahydrofuran, methyl tert-butyl ether, cyclopentyl methyl ether, toluene, cyclohexane, heptane, and dodecane. The catalytic performance of model enzyme horse liver alcohol dehydrogenase concerning its activity, stability, and selectivity is experimentally evaluated. The results are interpreted with molecular dynamics simulations by assessing the (i) protein location in biphasic media, (ii) organic solvent distribution, and (iii) enzyme conformation. Herein, the stability states the robustness of the enzyme while storing it in biphasic media without catalysis taking place. Overall, different toxicities of the solvent to the enzyme can be pinpointed: "molecular toxicity", related to the solvent functional groups, and "interfacial toxicity", related to the position of the enzyme at the interface. Likewise, some solvents are more prone to be located close to the active site of the enzyme, triggering other effects on the enzymatic performance. Thus, methyl tert-butyl ether resulted as an optimal option for the enzyme, whereas other solvents like toluene and 2-methyltetrahydrofuran were detrimental. The combined forces of experiments and simulations have been shown to be useful tools to study the effects of reaction media, thus guiding solvent selection.

5-hydroxymethylfurfural (HMF) is produced upon dehydration of C6 sugars in biorefineries. As the product, it remains either in aqueous solutions, or is in situ extracted to an organic medium (biphasic system). For the subsequent oxidation of HMF to 2,5-furandicarboxylic acid (FDCA), ‘media-agnostic’ catalysts that can be efficiently used in different conditions, from aqueous to biphasic, and to organic (microaqueous) media, are of interest. Here, the concept of a one-pot biocatalytic cascade for production of FDCA from HMF was reported, using galactose oxidase (GalOx) for the formation of 2,5-diformylfuran (DFF), followed by the lipase-mediated peracid oxidation of DFF to FDCA. GalOx maintained its catalytic activity upon exposure to a range of organic solvents with only 1 % (v/v) of water. The oxidation of HMF to 2,5-diformylfuran (DFF) was successfully established in ethyl acetate-based biphasic or microaqueous systems. To validate the concept, the reaction was conducted at 5 % (v/v) water, and integrated in a cascade where DFF was subsequently oxidized to FDCA in a reaction catalyzed by Candida antarctica lipase B. © 2022 The Authors. For a knowledge-based design of enzyme catalysis in deep eutectic solvents (DESs), the influence of the DES properties (e.g., water activity and viscosity) and the impact of DESs and their individual components must be assessed. This paper investigates three different DESs: choline chloride-glycerol (ChCl-Gly), choline chloride-ethylene glycol (ChCl-EG), and ethyl ammonium chloride-glycerol (EACl-Gly). The specific activity and half-life time of horse liver alcohol dehydrogenase (HLADH) were experimentally determined in these DESs with water contents ranging from 0%-100%. HLADH showed limited activity in neat DESs, which was enhanced by adding water. Experiments with individual DES components of ChCl-Gly were carried out to clarify their individual influence. Glycerol acts as a strong stabilizer for the enzyme, whereas choline chloride's results are deleterious. To understand the experimental findings, molecular dynamics (MD) simulations were carried out to quantify the solvation layer and calculate the spatial distribution of the solvent molecules around HLADH. The experimental and the in silico approach suggests that designing novel DESs with higher glycerol loadings would result in improved media for biocatalysis. This is demonstrated by performing the reduction of cinnamaldehyde to cinnamyl alcohol-a relevant compound for the food industry and cosmetics-in ChCl-Gly (1 : 9 molar ratio), with 20 vol% water to decrease the viscosity. HLADH is highly active and stable under these new conditions, giving promising productivity (15.3 g L-1 d-1). This paper demonstrates that DESs can be designed to be both substrate-solubilizers and enzyme-compatible, opening new research lines for green chemistry and biocatalysis.

The use of oxidoreductases (EC1) in non-conventional reaction media has been increasingly explored. In particular, deep eutectic solvents (DESs) have emerged as a novel class of solvents. Herein, an in-depth study of bioreduction with an alcohol dehydrogenase (ADH) in the DES glyceline is presented. The activity and stability of ADH in mixtures of glyceline/water with varying water contents were measured. Furthermore, the thermodynamic water activity and viscosity of mixtures of glyceline/water have been determined. For a better understanding of the observations, molecular dynamics simulations were performed to quantify the molecular flexibility, hydration layer, and intraprotein hydrogen bonds of ADH. The behavior of the enzyme in DESs follows the classic dependence of water activity (aW) in non-conventional media. At low aW values (<0.2), ADH does not show any activity; at higher aW values, the activity was still lower than that in pure water due to the high viscosities of the DES. These findings could be further explained by increased enzyme flexibility with increasing water content.

The broad interdisciplinary nature of biocatalysis fosters innovation, as different technical fields are interconnected and synergized. A way to depict that innovation is by conducting a survey on patent activities. This paper analyses the intellectual property activities of the last five years (2014–2019) with a specific focus on biocatalysis applied to asymmetric synthesis. Furthermore, to reflect the inventive and innovative steps, only patents that were granted during that period are considered. Patent searches using several keywords (e.g., enzyme names) have been conducted by using several patent engine servers (e.g., Espacenet, SciFinder, Google Patents), with focus on granted patents during the period 2014–2019. Around 200 granted patents have been identified, covering all enzyme types. The inventive pattern focuses on the protection of novel protein sequences, as well as on new substrates. In some other cases, combined processes, multi-step enzymatic reactions, as well as process conditions are the innovative basis. Both industries and academic groups are active in patenting. As a conclusion of this survey, we can assert that biocatalysis is increasingly recognized as a useful tool for asymmetric synthesis and being considered as an innovative option to build IP and protect synthetic routes.

The quest for sustainable solvents is currently a matter of intense research and development, as solvents significantly contribute heavily to the waste generated by chemical industries. Cyclopentyl methyl ether (CPME) is a promising eco-friendly solvent with valuable properties such as low peroxide formation rate, stability under basic and acidic conditions, and relatively high boiling point. This Review discusses the potential use of CPME for applications in biotechnology (e.g., biotransformations, as solvent or cosolvent), biorefineries, and bioeconomy (e.g., for furan synthesis or as an extractive agent in liquid–liquid separations), as well as for other purposes, such as chromatography or peptide synthesis. Although CPME is currently produced by petrochemical means with a remarkably high atom economy, its biogenic production can be envisaged from substrates such as cyclopentanol or cyclopentanone, which can be derived from furfural or from (bio-based) adipic acid, respectively. The combination of the promising properties of CPME as a (co)solvent with a future (economic) biogenic origin would be advantageous for setting strategies aligned with the sustainable chemistry principles.

Amino acids are of paramount importance as chiral building blocks of life, for drug development in modern medicinal chemistry, and for the manufacture of industrial products. In this work, the stereoselective synthesis of (S)- and (R)-2-amino-4-hydroxybutanoic acid was accomplished using a systems biocatalysis approach comprising a biocatalytic one-pot cyclic cascade by coupling of an aldol reaction with an ensuing stereoselective transamination. A class II pyruvate aldolase from E. coli, expressed as a soluble fusion protein, in tandem with either an S- or R-selective, pyridoxal phosphate dependent transaminase was used as a catalyst to realize the conversion, with formaldehyde and alanine being the sole starting materials. Interestingly, the class II pyruvate aldolase was found to tolerate formaldehyde concentrations of up to 1.4 M. The cascade system was found to reach product concentrations for (S)- or (R)-2-amino-4-hydroxybutanoic acid of at least 0.4 M, rendering yields between 86% and >95%, respectively, productivities of >80 g L–1 d–1, and ee values of >99%.

The present invention refers to an enzyme consisting of a fusion protein particularly useful as shown through-out the present invention for carrying out the carbon-carbon bond-forming reaction known as the aldol Reaction, preferably for carrying out an aldol reaction by using aldehydes as substrates and preferably pyruvate or a salt thereof, for producing hydroxyketoacids. Said enzyme is made by binding an aldolase to a maltose binding protein. The enzymes display full activity under "highly denaturing" substrate loadings (aldehydes, > 1 M).
