The fate of the West Antarctic Ice Sheet (WAIS) 1 is the largest cause of uncertainty in long-term sea-level projections. In the last interglacial (LIG) around 125,000 years ago, data suggest that sea level was several metres higher than today 2-4 , and required a significant contribution from Antarctic ice loss, with WAIS usually implicated. Antarctica and the Southern Ocean were warmer than today 5-8 , by amounts comparable to those expected by 2100 under moderate to high future warming scenarios. However, direct evidence about the size of WAIS in the LIG is sparse. Here we use sea salt data from an ice core from Skytrain Ice Rise, adjacent to WAIS, to show that, during most of the LIG, the Ronne Ice Shelf was still in place, and close to its current extent. Water isotope data are consistent with a retreat of WAIS 9 , but seem inconsistent with more dramatic model realizations 10 in which both WAIS and the large Antarctic ice shelves were lost. This new constraint calls for a reappraisal of other elements of the LIG sea-level budget. Warm Airmass Intrusions (WAIs) from the mid‐latitudes significantly impact the Arctic water budget. Here, we combine water vapor isotope measurements from the MOSAiC expedition, with a Lagrangian‐based process attribution diagnostic to track moisture transformation in the central Arctic Ocean during two WAIs, under contrasting sea‐ice concentrations (SIC). During winter with high SIC, two moisture supplies are identified. The first is Arctic moisture, locally‐sourced over the sea ice, with isotopic composition influenced by kinetic fractionation during ice‐cloud formation and vapor deposition. This moisture is rapidly overprinted by low‐latitude moisture advected poleward during WAI. In summer under low SIC, moisture is supplied through evaporation from land and ocean, with moisture removal via liquid‐cloud and dew formation. The isotopic composition reflects the influence of higher relative humidity at the evaporation sites. Given the projected increase of frequency and duration of WAIs, our study contributes to assessing process changes in the Arctic water cycle. Plain Language Summary: The movement of warm and moist airmasses from lower latitudes has a big effect on the Arctic climate system. We used data from the MOSAiC drift expedition, where we measured the isotopic composition of water vapor. Water isotopes are powerful tracers of where moisture came from and how it changed during the transport. We focused on two specific warm air intrusions, occurring in February and September 2020 respectively, when the amount of sea ice was different. During the winter, the isotopic composition of the airmasses was primarily influenced by in‐Arctic moisture exchanges over sea ice. This local moisture was swiftly replaced by isotopically‐distinct warmer and moister airmasses coming from lower latitudes during the warm intrusion. In summer, when there was less sea ice, we found that water came mainly from ocean evaporation with additional land evaporation during the air intrusion. The isotopic composition of vapor was influenced by how humid the places it came from were. As warm air intrusions are expected to happen more often and last longer in the future, our study helps us understand how they affect the Arctic water cycle. Frequency and intensity of warm and moist air-mass intrusions into the Arctic have increased over the past decades and have been related to sea ice melt. During our year-long expedition in the remote central Arctic Ocean, a record-breaking increase in temperature, moisture and downwelling-longwave radiation was observed in mid-April 2020, during an air-mass intrusion carrying air pollutants from northern Eurasia. The two-day intrusion, caused drastic changes in the aerosol size distribution, chemical composition and particle hygroscopicity. Here we show how the intrusion transformed the Arctic from a remote low-particle environment to an area comparable to a central-European urban setting. Additionally, the intrusion resulted in an explosive increase in cloud condensation nuclei, which can have direct effects on Arctic clouds' radiation, their precipitation patterns, and their lifetime. The COVID-19 pandemic has focused public attention on occupational groups that ensure the maintenance of critical infrastructure, provision of medical care and supply of essential goods. This paper examines the working conditions in critical jobs based on representative data from the German BAuA Working Time Survey 2019. Our analyses reveal that essential workers are more likely to perform unskilled or semiskilled activities and work in cleaning, transport and logistics, health care occupations as well as IT and natural science services. Regarding the working conditions, essential workers are paid comparatively less and are more physically proximate to others at work than nonessential workers. They more often work atypical hours, such as day and night shifts and on weekends, and have less autonomy in their working time. Additionally, critical jobs are characterised by muscular and skeletal strain due to working positions and carrying heavy loads significantly more often. Observations from CMET (Controlled Meteorological) balloons are analyzed in combination with mesoscale model simulations to provide insights into tropospheric meteorological conditions (temperature, humidity, wind-speed) around Svalbard, European High Arctic. Five Controlled Meteorological (CMET) balloons were launched from Ny-Ålesund in Svalbard over 5-12 May 2011, and measured vertical atmospheric profiles above Spitsbergen Island and over coastal areas to both the east and west. One notable CMET flight achieved a suite of 18 continuous soundings that probed the Arctic marine boundary layer over a period of more than 10 h. The CMET profiles are compared to simulations using the Weather Research and Forecasting (WRF) model using nested grids and three different boundary layer schemes. Variability between the three model schemes was typically smaller than the discrepancies between the model runs and the observations. Over Spitsbergen, the CMET flights identified temperature inversions and low-level jets (LLJ) that were not captured by the model. Nevertheless, the model largely reproduced time-series obtained from the Ny-Ålesund meteorological station, with exception of surface winds during the LLJ. Over sea-ice east of Svalbard the model underestimated potential temperature and overestimated wind-speed compared to the CMET observations. This is most likely due to the full sea-ice coverage assumed by the model, and consequent underestimation of ocean-atmosphere exchange in the presence of leads or fractional coverage. The suite of continuous CMET soundings over a sea-ice free region to the northwest of Svalbard are analysed spatially and temporally, and compared to the model. The observed along-flight daytime increase in relative humidity is interpreted in terms of the diurnal cycle, and in the context of marine and terrestrial air-mass influences. Analysis of the balloon trajectory during the CMET soundings identifies strong wind-shear, with a low-level channeled flow. The study highlights the challenges of modelling the Arctic atmosphere, especially in coastal zones with varying topography, sea-ice and surface conditions. In this context, CMET balloons provide a valuable technology for profiling the free atmosphere and boundary layer in remote regions where few other observations are available for model validation. Background: A key issue in the treatment of periprosthetic joint infection (PJI) is the correct diagnosis. The main problem is lack of diagnostic tools able to diagnose a PJI with high accuracy. Alpha-defensin has been proposed as a possible solution, but in the current literature, there is a lack of independent validation., Questions/purposes: We performed a prospective study to determine (1) what is the sensitivity, specificity, and positive and the negative predictive values of the alpha-defensin immunoassay test in diagnosing PJI; and (2) which clinical features may be responsible for false-positive and false-negative results?, Methods: Preoperative aspiration was performed in all patients presenting with a painful hip/knee arthroplasty, including both primary and revision implants. Metallosis, other inflammatory comorbidities, and previous/concomitant antibiotic therapy were not considered as exclusion criteria. An inadequate amount of synovial fluid for culture was an exclusion criterion. A total of 156 patients (65 knees, 91 hips) were included in this prospective study. At the time of revision, synovial fluid samples were taken to perform the alpha-defensin assay. During surgical débridement of tissue, samples for cultures and histologic evaluation were taken, and samples were cultured until positive or until negative at 14 days. A diagnosis of PJI was confirmed in 29 patients according to the International Consensus Group on PJI., Results: The sensitivity of the alpha-defensin immunoassay was 97% (95% confidence interval [CI], 92%-99%), the specificity was 97% (95% CI, 92%-99%), the positive predictive value was 88% (95% CI, 81%-92%), and the negative predictive value was 99% (95% CI, 96%-99%). Among four false-positive patients, two had metallosis and one had polyethylene wear. The false-negative case presented with a draining sinus, and intraoperative cultures were also negative., Conclusions: Alpha-defensin assay appears to be a reliable test, but followup evaluation is needed to estimate longer term performance of the test. The authors believe that alpha-defensin has demonstrated itself to be sufficiently robust that PJI diagnostic criteria now should include this test. Future studies are needed to compare the differences among the diagnostic capability of the available tests, in particular when metallosis is present, because metallosis may predispose the test to a false-positive result., Level of Evidence: Level I, diagnostic study.

Background: Charcot–Marie–Tooth (CMT) disease is an inherited peripheral neuropathy primarily involving motor and sensory neurons. Mutations in INF2, an actin assembly factor, cause two diseases: peripheral neuropathy CMT-DIE (MIM614455) and/or focal segmental glomerulosclerosis (FSGS). These two phenotypes arise from the progressive degeneration affecting podocytes and Schwann cells. In general, nerve enlargement has been reported in 25% of the demyelinating CMT subtype (CMT1), while little is known about the CMT-DIE caused by INF2 variants. Methods: To characterize the peripheral nerve phenotype of INF2-related CMT, we studied the clinical course, imaging, histology, and germline genetic variants in two unrelated CMT-DIE patients. Results: Patient 1 (INF2 p.Gly73Asp) and patient 2 (p.Val108Asp) first noticed walking difficulties at 10 to 12 years old. Both of them were electrophysiologically diagnosed with demyelinating neuropathy. In patient 2, the sural nerve biopsy revealed an onion bulb formation. Both patients developed nephrotic syndrome almost simultaneously with CMT and progressed into renal failure at the age of 16 to 17 years. Around the age of 30 years, both patients manifested multiple hypertrophy of the trunk, plexus, and root in the cervical, brachial, lumbosacral nerves, and cauda equina. The histology of the cervical mass in patient 2 revealed Schwannoma. Exome analysis showed that patient 2 harbors a germline LZTR1 p.Arg68Gly variant, while patient 1 has no schwannomatosis-related mutations. Conclusions: Peripheral neuropathy caused by INF2 variants may lead to the development of multifocal hypertrophy with age, likely due to the initial demyelination and subsequent Schwann cell proliferation. Background and Objectives: Myoclonus is already associated with a wide variety of drugs and systemic conditions. As new components are discovered, more drugs are suspected of causing this disabling abnormal involuntary movement. This systematic review aims to assess the medications associated with drug-induced myoclonus (DIM). Materials and Methods: Two reviewers assessed the PubMed database using the search term "myoclonus", without language restriction, for articles published between 1955 and 2024. The medications found were divided into classes and sub-classes, and the subclasses were graded according to their level of evidence. Results: From 12,097 results, 1115 were found to be DIM. The subclasses of medications with level A evidence were intravenous anesthetics (etomidate), cephalosporins (ceftazidime, cefepime), fluoroquinolones (ciprofloxacin), selective serotonin reuptake inhibitors (citalopram, escitalopram, paroxetine, sertraline), tricyclic antidepressant (amitriptyline), glutamate antagonist (amantadine), atypical antipsychotics (clozapine, quetiapine), antiseizure medications (carbamazepine, oxcarbazepine, phenytoin, gabapentin, pregabalin, valproate), pure opioid agonist (fentanyl, morphine), bismuth salts, and mood stabilizers (lithium). The single medication with the highest number of reports was etomidate. Drug-induced asterixis is associated with a specific list of medications. The neurotransmitters likely involved in DIM are serotonin, dopamine, gamma-aminobutyric acid (GABA), and glutamate. Conclusions: DIM may be reversible with management that can include drug discontinuation, dose adjustment, and the prescription of a medication used to treat idiopathic myoclonus. Based on the main clinical constellation of symptoms and pathophysiological mechanisms found in this study, DIM can be categorized into three types: type 1 (serotonin syndrome), type 2 (non-serotonin syndrome), and type 3 (unknown). The classical theory of homogeneous isotropic turbulence predicts that the ratio of transverse to longitudinal structure functions or power spectra is equal to 4/3 in the inertial subrange. For the typical turbulence cascade in the inertial subrange, it also predicts a power law scaling with an exponent of +2/3 and -5/3 for the structure functions and the power spectra, respectively. The goal of this study is to document the statistics of those ratios and exponents derived from aircraft observations, quantify their departures from theoretical predictions, and point out the differences among the aircraft. We estimate the transverse-to-longitudinal ratios and the scaling exponents from in situ high-rate turbulence measurements collected by three research aircraft during four field experiments in two regimes of the marine atmospheric boundary layer: shallow trade-wind convection and subtropical stratocumulus. The bulk values representing the inertial subrange were derived by fitting power law formulae to the structure functions and to the power spectra computed separately for the three components of the turbulent wind velocity measured in horizontal flight segments. The composite scale-by-scale transverse-to-longitudinal ratios were derived by averaging over the segments at common non-dimensional scales. The variability in the results can be attributed to how the wind velocity components are measured on each aircraft. The differences related to environmental conditions, e.g. between characteristic levels and regimes of the boundary layer, are of secondary importance. Experiment-averaged transverse-to-longitudinal ratios are 23 %–45 % smaller than predicted by the theory. The deviations of average scaling exponents with respect to the theoretical values range from - 34 % to + 47 % for structure functions and from - 24 % to + 22 % for power spectra, depending on experiment and velocity component. The composite scale-by-scale transverse-to-longitudinal ratios decrease and increasingly depart from 4/3 with decreasing scale, in contrast to previous experimental studies on local isotropy. Background: Resting heart rate (HR) and HR variability (HRV) are widely used parameters to assess cardiac autonomic nervous system function noninvasively. While resting assessments can be performed during sleep or after awakening, it would be relevant to know how interchangeable the results of these measurements are. This study aimed at examining the alignment between nocturnal and morning assessments during regular endurance training and in response to intensive training. A total of 24 recreational runners performed a 3-week baseline period (BL) and a 2-week overload (OL) period (Lucia's training impulse + 80%). Their running performance was assessed with a 3000-m running test after the BL and OL. The participants recorded daily their nocturnal HR and HRV (the natural logarithm of the root mean square of successive differences; LnRMSSD) with a photoplethysmography-based wrist device and performed an orthostatic test (2-min supine, 2-min standing) every morning with a chest-strap HR sensor. The HR and LnRMSSD segments that were analyzed from the nocturnal recordings included start value (SleepStart), end value (SleepEnd), first 4-h segment 30 min after detected sleep onset (Sleep4h), and full sleep time (SleepFull). The morning segments consisted of the last-minute average in both body positions. All segments were compared at BL and in response to the 3000-m test and OL. Results: All nocturnal HR and LnRMSSD segments correlated with supine and standing segments at BL (r = 0.42 to 0.91, p < 0.05). After the 3000-m test, the HR increased and LnRMSSD decreased only in the SleepStart, Sleep4h, and SleepFull segments (p < 0.05). In response to the OL, the standing HR decreased (p < 0.01), while the LnRMSSD increased (p < 0.05) in all segments except for SleepStart. The Pearson correlations between relative changes in nocturnal and morning segments were − 0.11 to 0.72 (3000-m) and − 0.25 to 0.79 (OL). The OL response in Sleep4h HR and LnRMSSD correlated with the relative change in 3000-m time (r = 0.63, p = 0.001 and r=-0.50, p = 0.013, respectively). Conclusions: Nocturnal and morning HR and LnRMSSD correlated moderately or highly in the majority of cases during the BL, but their responses to intensive training were not similarly aligned, especially in LnRMSSD. The nocturnal segments seemed to be sensitive to physical loading, and their responses were associated with the performance-related training responses. Key Points: • Moderate-to-high correlations were found between nocturnal and morning assessments of HR and HRV during regular endurance training in recreational runners. • In their responses to maximum exercise, nocturnal and morning HR and HRV were not necessarily aligned, as nocturnal segments seemed more responsive to such a stimulus. • While lower HR and higher HRV were associated with better endurance performance in all recording segments, changes in these parameters after the overload period correlated only in nocturnal segments. Paleoclimate information has played an instrumental role in showing how fast climate can vary and how large these changes can be. It provided the first vivid demonstration of the relationships between atmospheric greenhouse gas concentrations and surface air temperatures, as well as striking representations of climate change impacts and possible feedbacks within the climate system, such as those associated with vegetation or ice sheet changes. Here, a short review of recent advances in paleoclimate studies is provided, with the objective of showing what this information on past climates and environments can bring to research on current and possible future climates. We advocate that (1) paleoclimatic and paleoenvironmental information can be leveraged for narratives about climate change, in particular at the local and regional levels, (2) paleoclimate data is essential for out-of-range tests of climate models, since future climates are also out of the range of recent climate information used for calibrating climate models, (3) paleoclimate data, in particular for the last millennia, is essential for taking multi-centennial and multi-millennial variability into account when describing trends related to anthropogenic forcings and attributing climate change signals, in particular for extreme and rare events, and (4) paleoclimates also provide extremely valuable information for initializing the slow components of climate models. In addition, we show how paleoclimate studies can be beneficial to put recent and future climate change into context and improve our knowledge on key processes. Introduction: Orthostatic intolerance is highly prevalent in patients with myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) and is caused by an abnormal reduction in cerebral blood flow (CBF). In healthy controls (HCs), the regulation of CBF is complex and cardiac output (CO) is an important determinant of CBF: a review showed that a 30% reduction in CO results in a 10% reduction in CBF. In previous and separate ME/CFS studies, we showed that CO and CBF decreased to a similar extent during tilt testing. The aim of the study: to test the relationship between CBF and CO, which seems to be abnormal in ME/CFS patients and is different from that in HCs. Methods: In this retrospective study we analyzed this relationship in a large group of patients. To compare the patient data with those of HCs, we focused on patients with a normal heart rate (HR) and blood pressure (BP) response to upright tilt. Also, the influence of clinical data was analyzed. A total of 534 ME/CFS patients and 49 HCs underwent tilt testing with measurements of HR, BP, CBF, CO, and end-tidal PCO2. To measure CBF, extracranial Doppler flow velocity and vessel diameters were obtained using a GE echo system. The same device was used to measure suprasternal aortic flow velocities. End-tidal PCO2 was recorded using a Nonin Lifesense device. Results: In 46 (9%) patients, CO and CBF changes were in the normal range for HCs, and in 488 (91%) an abnormal CO and CBF reduction was found. In patients with abnormal CO and CBF reductions, the slope of the regression line of CO versus CBF reduction was almost 1. The multiple regression analysis of the latter group showed that the CO reduction for the most part predicted the CBF reduction, with a limited role for the PETCO2 reduction. Conclusions: Two different patient groups with a normal HR and BP response during the tilt were identified: those with a CO and CBF in the normal range for HCs and those with an abnormal CO and CBF reduction during the tilt (91% of patients). In the latter group of patients, an almost 1:1 relationship between the CO and CBF reduction suggests the absence of compensatory vasodilation in the cerebral vasculature. Land surface temperature (LST) has gained increased attention in cryospheric research. While various global satellite LST products are available, none of them is specially designed for the pan-Arctic region. Based on the recently published EUMETSAT Advanced Very High Resolution Radiometer (AVHRR) fundamental data record (FDR), a new LST product (1981–2021) with daily resolution is developed for the pan-Arctic region. Validation shows good accuracy with an average mean absolute error (MAE) of 1.71 K and a MAE range of 0.62–3.07 K against in situ LST data from the Surface Radiation Budget (SURFRAD) network and Karlsruhe Institute of Technology (KIT) sites. Long-term stability, a strong requirement for trend analysis, is assessed by comparing LST with air temperatures from ERA5-Land (T2M) and air temperature data from the EUSTACE (https://www.eustaceproject.org , last access: 17 December 2024) global station dataset. Long-term stability might not be fulfilled mainly due to the orbit drift of the NOAA satellites. Therefore, the analysis is split into two periods: the arctic winter months, which are unaffected by solar illumination and, therefore, orbital drift, and the summer months. The analysis for the winter months results in correlation values (r) of 0.44–0.83, whereas for the summer months (r) values range between 0.37–0.84. Analysis of anomaly differences revealed instabilities for the summer months at a few stations. The same stability analysis for the winter months revealed only one station with instabilities in comparison to station air temperature. Discrepancies between the temperature anomalies recorded at the stations and ERA5-Land T2M were also found. This highlights the limited influence of orbital drift on the LST product, with the winter months presenting good stability across all stations, which makes these data a valuable source for studying LST changes in the pan-Arctic region over the last 40 years. Tropical free‐tropospheric humidity plays a crucial role for the Earth's radiative balance and climate sensitivity. In addition to atmospheric humidity, stable water isotopes can provide important information about the hydrological cycle. We use the isotope‐ and water tagging‐enabled version of the COSMOiso model to determine isotopic fingerprints of diagnosed moisture pathways over the western tropical Atlantic (WTA). A convection‐permitting, high‐resolution (5 km) nudged simulation is performed for January–February 2020. During this period, the target region is characterized by alternating large‐scale circulation regimes with different humidity and isotope signatures. Moist conditions in the middle troposphere (300–650 hPa) are associated with moisture transport from the south, east, southeast, as well as evaporation from the North Atlantic, while dry conditions correspond to extratropical transport from the north and west. To predict the contribution of different moisture sources, we used a statistical model based on the local specific humidity and temperature as predictors and obtained an R‐squared (R2) of 0.52. Adding water isotopes improved the prediction (R2 = 0.73), showing that isotopes provide unique information on moisture sources and transport patterns beyond conventional local observations. High-resolution three-dimensional (3D) wind velocity measurements are of major importance for the characterization of atmospheric turbulence. The use of a multi-beam wind lidar focusing on a measurement volume from different directions is a promising approach for obtaining such wind data. This paper provides a detailed study of the propagation of measurement uncertainty of a three-beam wind lidar designed for mounting on airborne platforms with geometrical constraints that lead to increased measurement uncertainties of the wind components transverse to the main axis of the system. The uncertainty analysis is based on synthetic wind data generated by an Ornstein–Uhlenbeck process as well as on experimental wind data from airborne and ground-based 3D ultrasonic anemometers. For typical atmospheric conditions, we show that the measurement uncertainty of the transverse components can be reduced by about 30 %–50 % by applying an appropriate post-processing algorithm. Optimized post-processing parameters can be determined in an actual experiment by characterizing measured data in terms of variance and correlation time of wind fluctuations, allowing for the optimized design of a multi-beam wind lidar with strong geometrical limitations. We introduce a novel method to retrieve the cloud fraction and the optical thickness of liquid clouds over a water surface based on polarimetry. The approach is well suited for satellite observations providing multi-angle polarization measurements, in particular those of the Hyper-Angular Rainbow Polarimeter #2 (HARP2). Unlike commonly used methods to derive cloud fractions, our method does not depend on the spatial resolution of observations, and it does not require any threshold values for cloud detection. Based on radiative transfer simulations, we show that the cloud fraction and the cloud optical thickness can be derived from measurements at two viewing angles: one within the cloudbow and one in the sun glint region. In the cloudbow, the degree of polarization mainly depends on the cloud optical thickness. Conversely, for a viewing direction in the sun glint region, the degree of polarization depends on the clear fraction of the pixel, because here the radiation scattered by cloud droplets is almost unpolarized, whereas radiation reflected by the surface is highly polarized. Utilizing these dependencies, we developed a retrieval using a simple lookup table approach. Based on sensitivity studies, we show that prior information about wind speed and aerosol optical thickness improves the accuracy of the cloud fraction retrieval. Prior information about the cloud droplet size distribution can reduce the uncertainty of the cloud optical thickness retrieval. The prior information should be obtained by combining our method with already existing aerosol and cloud retrieval algorithms. We performed 3D radiative transfer simulations and found that the cloud optical thickness is generally underestimated due to neglect of 3D scattering effects. The cloud fraction is overestimated in cloud shadows and underestimated in in-scattering regions. As a demonstration, we apply the methodology to airborne observations from polarization cameras of the Munich Aerosol Cloud Scanner (specMACS) instrument. The high-spatial-resolution data (10–20 m) have been averaged to a spatial resolution of approximately 2.5 km to mimic satellite observations. From the average linear polarization at scattering angles of 140 and 110°, we derive continuous cloud fraction values and the corresponding cloud optical thicknesses. Comparison for cases including low, medium, and high cloud fractions shows that the retrieval, using only reflected polarized radiances at two scattering angles, provides accurate estimates of the cloud fraction for observations with coarse spatial resolution. Cloud geometry and in particular cloud top heights can be derived from 2-D camera measurements by applying a stereographic method to data from an overflight over a scene of clouds see, e.g.,. Although airplane overpasses are relatively fast, cloud motion with the wind is important and can result in errors in the cloud localization. Here, the impact of the wind is investigated using the method from for measurements of the airborne hyperspectral imaging system spectrometer of the Munich Aerosol Cloud Scanner (specMACS). Further, a method for cloud motion correction using model winds from the European Centre for Medium-Range Weather Forecasts (ECMWF) is presented. It is shown that the update is important as the original algorithm without the cloud motion correction can over- or underestimate the cloud top heights by several hundred meters, depending on the wind speed and the relative wind direction. This is validated using data from the Elucidating the role of clouds–circulation coupling in climate (EUREC 4 A) campaign and realistic 3-D radiative transfer simulations. From the comparison of the derived cloud top heights with the expected ones from the model input, an average accuracy of the cloud top heights of less than (20±140) m (mean deviation and 1 standard deviation) is estimated for the updated method. In this study, we present and analyze the first continuous time series of relevant aerosol precursor vapors from the central Arctic (north of 80° N) during the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition. These precursor vapors include sulfuric acid (SA), methanesulfonic acid (MSA), and iodic acid (IA). We use FLEXPART simulations, inverse modeling, sulfur dioxide (SO2) mixing ratios, and chlorophyll a (chl a) observations to interpret the seasonal variability in the vapor concentrations and identify dominant sources. Our results show that both natural and anthropogenic sources are relevant for the concentrations of SA in the Arctic, but anthropogenic sources associated with Arctic haze are the most prevalent. MSA concentrations are an order of magnitude higher during polar day than during polar night due to seasonal changes in biological activity. Peak MSA concentrations were observed in May, which corresponds with the timing of the annual peak in chl a concentrations north of 75° N. IA concentrations exhibit two distinct peaks during the year, namely a dominant peak in spring and a secondary peak in autumn, suggesting that seasonal IA concentrations depend on both solar radiation and sea ice conditions. In general, the seasonal cycles of SA, MSA, and IA in the central Arctic Ocean are related to sea ice conditions, and we expect that changes in the Arctic environment will affect the concentrations of these vapors in the future. The magnitude of these changes and the subsequent influence on aerosol processes remains uncertain, highlighting the need for continued observations of these precursor vapors in the Arctic. Numerical methods and simulation codes are essential for the advancement of our understanding of complex atmospheric processes. As technology and computer hardware continue to evolve, the development of sophisticated code is vital for accurate and efficient simulations. In this paper, we present the recent advancements made in the FLEXible PARTicle dispersion model (FLEXPART), a Lagrangian particle dispersion model, which has been used in a wide range of atmospheric transport studies over the past 3 decades, extending from tracing radionuclides from the Fukushima nuclear disaster, to inverse modelling of greenhouse gases, and to the study of atmospheric moisture cycles. This version of FLEXPART includes notable improvements in accuracy and computational efficiency. (1) By leveraging the native vertical coordinates of European Centre for Medium Range Weather Forecasts (ECMWF) Integrated Forecasting System (IFS) instead of interpolating to terrain-following coordinates, we achieved an improvement in trajectory accuracy, leading to a ∼8 %–10 % reduction in conservation errors for quasi-conservative quantities like potential vorticity. (2) The shape of aerosol particles is now accounted for in the gravitational settling and dry-deposition calculation, increasing the simulation accuracy for non-spherical aerosol particles such as microplastic fibres. (3) Wet deposition has been improved by the introduction of a new below-cloud scheme, by a new cloud identification scheme, and by improving the interpolation of precipitation. (4) Functionality from a separate version of FLEXPART, the FLEXPART CTM (chemical transport model), is implemented, which includes linear chemical reactions. Additionally, the incorporation of Open Multi-Processing parallelisation makes the model better suited for handling large input data. Furthermore, we introduced novel methods for the input and output of particle properties and distributions. Users now have the option to run FLEXPART with more flexible particle input data, providing greater adaptability for specific research scenarios (e.g. effective backward simulations corresponding to satellite retrievals). Finally, a new user manual (https://flexpart.img.univie.ac.at/docs/ , last access: 11 September 2024) and restructuring of the source code into modules will serve as a basis for further development. In the central Arctic, high-quality water vapour observations are sparse due to the low density of meteorological stations and uncertainties in satellite remote sensing. Different reanalyses also disagree on the amount of water vapour in the central Arctic. The Multidisciplinary drifting Observatory for the Study of the Arctic Climate (MOSAiC) expedition provides comprehensive observations that are suitable for evaluating satellite products and reanalyses. Radiosonde observations provide high-quality water vapour estimates with a high vertical but a low temporal resolution. Observations from the microwave radiometers (MWRs) on board the research vessel Polarstern complement these observations through high temporal resolution. In this study, we demonstrate the high accuracy of the combination of the two MWRs HATPRO (Humidity and Temperature Profiler) and MiRAC-P (Microwave Radiometer for Arctic Clouds – Passive). For this purpose, we developed new retrievals of integrated water vapour (IWV) and profiles of specific humidity and temperature using a neural network approach, including observations from both HATPRO and MiRAC-P to utilize their different water vapour sensitivity. The retrievals were trained with the European Centre for Medium-Range Weather Forecasts (ECMWF) reanalysis version 5 (ERA5) and synthetic MWR observations simulated with the Passive and Active Microwave radiative TRAnsfer tool (PAMTRA). We applied the retrievals to synthetic and real observations and evaluated them with ERA5 and radiosondes launched during MOSAiC, respectively. To assess the benefit of the combination of HATPRO and MiRAC-P compared to single MWR retrievals, we compared the errors with respect to MOSAiC radiosondes and computed the vertical information content of the specific humidity profiles. The root mean squared error (RMSE) of IWV was reduced by up to 15 %. Specific humidity biases and RMSE were reduced by up to 75 % and 50 %, respectively. The vertical information content of specific humidity could be increased from 1.7 to 2.4 degrees of freedom. We also computed relative humidity from the retrieved temperature and specific humidity profiles and found that RMSE was reduced from 45 % to 15 %. Finally, we show a case study demonstrating the enhanced humidity profiling capabilities compared to the standard HATPRO-based retrievals. The vertical resolution of the retrieved specific humidity profiles is still low compared to radiosondes, but the case study revealed the potential to resolve major humidity inversions. Background: Stroke survivors often experience autonomic nervous system (ANS) dysfunction. While Transcranial Direct Current Stimulation (tDCS) has been shown to modulate the ANS when applied to the left hemisphere, its effects on the right hemisphere remain unexplored. Objective: We aimed to compare the effects of tDCS applied to both the injured and the contralateral hemispheres on heart rate variability (HRV) and functional capacity in individuals post-stroke. Methods: Twenty individuals with cerebral hemisphere lesions (ten with right-hemisphere lesions and ten with left-hemisphere lesions) were randomized into four groups: anodal and sham tDCS on the left temporal cortex (T3) and anodal and sham tDCS on the right temporal cortex (T4). HRV was assessed before the intervention, after the six-minute walk test (6MWT), and following tDCS. HRV data were categorized into frequency ranges: low frequency (LF), high frequency (HF), and sympathovagal balance. The 6MWT (meters) was conducted both pre- and post-tDCS. Results: In individuals with right-hemisphere lesions, a higher global LF value was observed (right side: 71.4 ± 16.8 nu vs. left side: 65.7 ± 17.3 nu; p = 0.008), as well as lower values of the HF component (right side: 29.5 ± 18.9 nu vs. left side: 34.0 ± 17.4 nu; p = 0.047), consequently exhibiting higher global values of the low/high-frequency ratio (right side: 3.9 ± 2.8 vs. left side: 2.9 ± 2.4). Regarding the stimulation site, tDCS over T3 led to a lower overall value of the low/high-frequency ratio (left hemisphere: 3.0 ± 2.2 vs. right hemisphere: 3.7 ± 2.9; p = 0.040) regardless of the lesion location. A significant increase in the distance covered in the 6MWT was observed for individuals with lesions in both hemispheres after tDCS at T3. Conclusions: Participants with right-hemisphere lesions exhibited superior global sympathetic autonomic nervous system activity. When the tDCS was applied on the left hemisphere, it maintained lower sympathovagal balance values and improved functional capacity regardless of the hemisphere affected by the stroke. Wind-blown snow is a frequent phenomenon in high-elevation and polar regions which impacts the surface energy and mass balance of these areas. Loose surface snow gets eroded and transported by wind, which influences the snow particles' physical properties (size, shape, optical properties) that determine the characteristics of the emerging wind-impacted snowpack layer. During airborne snow transport, the governing processes happen on the micro-scale while the particles are transported over long distances. The unfolding processes and the evolution of the particles' physical properties are thus difficult to observe in situ. Here, we used cold-laboratory ring wind tunnel experiments as an interim solution to study the governing processes during airborne snow transport with stable water isotopes as tracers for these micro-scale processes. Repeated analysis of airborne-sampled snow by micro-computed tomography (µ CT) documented a growing and rounding of snow particles with transport time, with a concurrent decrease in specific surface area. Stable water isotope analysis of airborne snow and water vapour allowed us to attribute this evolution to the process of airborne snow metamorphism. The changes observed in the snow isotopic composition showed a clear isotopic signature of metamorphic deposition, which requires particle–air temperature gradients. These results question the validity of the thermal-equilibrium assumption between particles and air inside the saltation layer of wind-blown snow events, where the conditions are similar to the ones found in the wind tunnel. Our results thus refine the understanding of the governing processes in the saltation layer and suggest that the snow's isotopic composition can inform on local wind-blown snow events as the original snow isotope signal gets overprinted by airborne snow metamorphism. Within transport times of 3 h, we observed changes in the isotope signal of airborne snow of up to +1.47 ‰ in δ18 O, ±5.7 ‰ in δ D, and -6.1 ‰ in d-excess. The impact of a slightly modified broadband snow surface albedo parameterization, which explicitly considers the cloud dependence of the snow albedo, is evaluated in simulations of a coupled regional climate model of the Arctic. The cloud dependence of the snow albedo leads to a more realistic simulation of the variability of the surface albedo during the snowmelt period in late May and June. In particular, the reproduction of lower albedo values under cloud-free/broken-cloud conditions during the snowmelt period represents an improvement and results in an earlier disappearance of the snow cover and an earlier onset of sea-ice melt. In this way, the consideration of the cloud dependence of the snow albedo results in an amplification of the two-stage snow/ice albedo feedback in the model. This finds expression in considerably increased sea-ice melt during the summer months and ends up in a new quasi-stationary equilibrium in sea ice with statistically significant lower sea-ice volume and statistically significant lower summer sea-ice area. A crucial factor influencing the mass balance of the West Antarctic Ice Sheet is the Amundsen Sea Low (ASL), a climatological low‐pressure region situated off the West Antarctic coast. However, albeit the deepening of the ASL since the 1950s has been attributed to anthropogenic forcing, the multi‐decadal variability of the ASL remains poorly understood, because of a lack of long observations. Here, we apply a newly developed data assimilation method to reconstruct the ASL over 1870–2000. We study the forced and internal variability of the ASL using our new reconstruction in concert with existing large ensembles of climate model simulations. Our findings robustly demonstrate that an atmospheric teleconnection originating from the tropical Indo‐Pacific is the main driver of ASL variability at the multi‐decadal time scale, with resemblance to the Interdecadal Pacific Oscillation. Since the mid‐20th century, anthropogenic forcing has emerged as a dominant contributor to the strengthening of the ASL. Plain Language Summary: Changes in the West Antarctic Ice Sheet mass balance (i.e., the difference between the gain and loss of ice mass) are partly influenced by large‐scale winds, and in particular, a climatological low‐pressure feature located off the West Antarctic coast called the Amundsen Sea Low (ASL). Yet, although the long‐term strengthening of the ASL since the mid‐20th century has been demonstrated to be related to anthropogenic forcing, our understanding of the variability of the ASL on time‐scales of decades is poorly known. In this paper, we therefore investigate the origins of this variability since 1870, and quantify the relative contributions of human‐caused climate changes and natural variability of the climate system. For this purpose, we use several ensembles of model simulations as well as new climate reconstructions that combine paleoclimate records with model simulations using a statistical method. Our results indicate that the multi‐decadal variability of the ASL is strongly driven by tropical variability in the Indo‐Pacific through atmospheric connections between this region and the Amundsen Sea. Our reconstruction, when compared with a large ensemble of model simulations, indicates that since 1950, human‐induced climate forcing has become a dominant driver of long‐term ASL variability, contributing equally to tropical variability. Large-scale meteorological events (e.g. cyclones), referred to as synoptic events, strongly influence weather predictability but still cannot be fully characterised in the Arctic region because of the sparse coverage of measurements. Due to the fact that atmospheric dynamics in the lower stratosphere and troposphere influence the ozone field, one approach to analyse these events further is the use of space-borne measurements of ozone vertical distributions and total columns in addition to conventional parameters such as pressure or wind speed. In this study we investigate the link between cyclones and changes in stratospheric ozone by using a combination of unique measurements during the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) ship expedition, ozone profile and total column observations by satellite instruments (OMPS-LP, TROPOMI), and ERA5 reanalysis data. The final goal of the study is to assess whether the satellite ozone data can be used to obtain information about cyclones and provide herewith an additional value in the assimilation by numerical weather prediction models. Three special cases during the MOSAiC expedition were selected and classified for the analysis. They are one "normal" cyclone, where a low surface pressure coincides with a minimum in tropopause height, and two "untypical" cyclones, where this is not observed. The influence of cyclone events on ozone in the upper-troposphere lower-stratosphere (UTLS) region was investigated, using the fact that both are correlated with tropopause height changes. The negative correlation between tropopause height from ERA5 and ozone columns was investigated in the Arctic region for the 3-month period from June to August 2020. This was done using total ozone columns and sub-columns from TROPOMI, OMPS-LP, and MOSAiC ozonesonde data. The greatest influence of tropopause height changes on ozone contour levels occurs at an altitude between 10 and 20 km. Moreover, the lowering of the 250 ppb ozonopause (at about 11 km altitude) below 9 km was used to detect cyclones using OMPS-LP ozone observations. The potential of this approach was demonstrated in two case studies where the boundaries of cyclones could be determined using ozone observations. The results of this study can help improve our understanding of the relationship between cyclones, tropopause height, and ozone in the Arctic and demonstrate the usability of satellite ozone data in addition to the conventional parameters for investigating cyclones in the Arctic. Global warming is amplified in the Arctic. However, numerical models struggle to represent key processes that determine Arctic weather and climate. To collect data that help to constrain the models, the HALO–(AC) 3 aircraft campaign was conducted over the Norwegian and Greenland seas, the Fram Strait, and the central Arctic Ocean in March and April 2022. The campaign focused on one specific challenge posed by the models, namely the reasonable representation of transformations of air masses during their meridional transport into and out of the Arctic via northward moist- and warm-air intrusions (WAIs) and southward marine cold-air outbreaks (CAOs). Observations were made over areas of open ocean, the marginal sea ice zone, and the central Arctic sea ice. Two low-flying and one long-range, high-altitude research aircraft were flown in colocated formation whenever possible. To follow the air mass transformations, a quasi-Lagrangian flight strategy using trajectory calculations was realized, enabling us to sample the same moving-air parcels twice along their trajectories. Seven distinct WAI and 12 CAO cases were probed. From the quasi-Lagrangian measurements, we have quantified the diabatic heating/cooling and moistening/drying of the transported air masses. During CAOs, maximum values of 3 K h -1 warming and 0.3 g kg -1 h -1 moistening were obtained below 1 km altitude. From the observations of WAIs, diabatic cooling rates of up to 0.4 K h -1 and a moisture loss of up to 0.1 g kg -1 h -1 from the ground to about 5.5 km altitude were derived. Furthermore, the development of cloud macrophysical (cloud-top height and horizontal cloud cover) and microphysical (liquid water path, precipitation, and ice index) properties along the southward pathways of the air masses were documented during CAOs, and the moisture budget during a specific WAI event was estimated. In addition, we discuss the statistical frequency of occurrence of the different thermodynamic phases of Arctic low-level clouds, the interaction of Arctic cirrus clouds with sea ice and water vapor, and the characteristics of microphysical and chemical properties of Arctic aerosol particles. This study emulates dropsondes to elucidate the extent to which sporadic airborne sondes adequately represent divergence of moisture transport in Arctic atmospheric rivers (ARs). The convergence of vertically integrated moisture transport (IVT) plays a crucial role as it favours precipitation that significantly affects Arctic sea ice properties. Long-range research aircraft can transect ARs and drop sondes to determine their IVT divergence. In order to assess the representativeness of future sonde-based IVT divergence in Arctic ARs, we disentangle the sonde-based deviations from an ideal instantaneous IVT divergence, which result from undersampling by a limited number of sondes and from the flight duration. Our synthetic study uses C3S Arctic Regional Reanalysis (CARRA) reanalyses to set up an idealised scenario for airborne AR observations. For nine Arctic spring ARs, we mimic flights transecting each AR in CARRA and emulate sonde-based IVT representation by picking single vertical profiles. The emulation quantifies IVT divergence observability by two approaches. First, sonde-based IVT and its divergence are compared to the continuous IVT interpolated onto the flight cross-section. The comparison specifies uncertainties of discrete sonde-based IVT variability and divergence. Second, we determine how temporal AR evolution affects IVT divergence values by contrasting time-propagating sonde-based values with the divergence based on instantaneous snapshots. For our Arctic AR cross-sections, we find that coherent wind and moisture variabilities contribute less than 10 % to the total transport. Both quantities are uncorrelated to a great extent. Moisture turns out to be the more variable quantity. We show that sounding spacing greater than 100 km results in errors greater than 10 % of the total IVT along AR cross-sections. For IVT divergence, the Arctic ARs exhibit similar differences in moisture advection and mass convergence across the embedded front as mid-latitude ARs, but we identify moisture advection as being dominant. Overall, we confirm the observability of IVT divergence with an uncertainty of around 25 %–50 % using a sequence of at least seven sondes per cross-section. Rather than sonde undersampling, it is the temporal AR evolution over the flight duration that leads to high deviations in divergence components. In order to realise the estimation of IVT divergence from dropsondes, flight planning should consider not only the sonde positioning, but also the minimisation of the flight duration. Anderson–Fabry disease (AFD) is a genetic lysosomal storage disorder caused by mutations in the α-galactosidase A gene, leading to impaired lysosomal function and resulting in both macrovascular and microvascular alterations. AFD patients often exhibit increased intima-media thickness (IMT) and reduced flow-mediated dilation (FMD), indicating non-atherosclerotic arterial thickening and the potential for cardiovascular events. Nailfold capillaroscopy, a non-invasive diagnostic tool, has shown potential in diagnosing and monitoring microcirculatory disorders in AFD, despite limited research. This study evaluates nailfold capillaroscopy findings in AFD patients, exploring correlations with GLA gene variant subgroups (associated with classical or late-onset phenotypes and variants of uncertain significance (VUSs)), and assessing morpho-functional differences between sexes. It aims to determine whether capillaroscopy can assist in the early identification of individuals with multiorgan vascular involvement. A retrospective observational study was conducted with 25 AFD patients from AOUP "G. Rodolico-San Marco" in Catania (2020–2023). Patients underwent genetic testing, enzyme activity evaluation, and nailfold capillaroscopy using Horus basic HS 200 videodermatoscopy. Parameters like angiotectonic disorder, vascular areas, capillary density, and intimal thickening were assessed. The study identified significant differences in capillaroscopy findings among patients with different GLA gene variant subgroups. Classic AFD variant patients showed reduced capillary length and signs of erythrocyte aggregation and dilated subpapillary plexus. No correlation was found between enzymatic activity and capillaroscopy parameters. However, Lyso-Gb3 levels were positively correlated with average capillary length (ῤ = 0.453; p = 0.059). Sex-specific differences in capillaroscopy findings were observed in neoangiogenesis and average capillary length, with distinct implications for men and women. This study highlights the potential of nailfold capillaroscopy in the diagnostic process and clinical management of AFD, particularly in relation to specific GLA gene mutations, as a valuable tool for the early diagnosis and monitoring of AFD. We document the isotopic evolution of near-surface snow at the East Greenland Ice Core Project (EastGRIP) ice core site in northeast Greenland using a time-resolved array of 1 m deep isotope (δ18O , δD) profiles. The snow profiles were taken from May–August during the 2017–2019 summer seasons. An age–depth model was developed and applied to each profile, mitigating the impacts of stratigraphic noise on isotope signals. Significant changes in deuterium excess (d) are observed in surface snow and near-surface snow as the snow ages. Decreases in d of up to 5 ‰ occur during summer seasons after deposition during two of the three summer seasons observed. The d always experiences a 3 ‰–5 ‰ increase after aging 1 year in the snow due to a broadening of the autumn d maximum. Models of idealized scenarios coupled with prior work indicate that the summertime post-depositional changes in d (Δd) can be explained by a combination of surface sublimation, forced ventilation of the near-surface snow down to 20–30 cm , and isotope-gradient-driven diffusion throughout the column. The interannual Δd is also partly explained with isotope-gradient-driven diffusion, but other mechanisms are at work that leave a bias in the d record. Thus, d does not just carry information about source-region conditions and transport history as is commonly assumed, but also integrates local conditions into summer snow layers as the snow ages through metamorphic processes. Finally, we observe a dramatic increase in the seasonal isotope-to-temperature sensitivity, which can be explained solely by isotope-gradient-driven diffusion. Our results are dependent on the site characteristics (e.g., wind, temperature, accumulation rate, snow properties) but indicate that more process-based research is necessary to understand water isotopes as climate proxies.
