The primary source of soil pollution is a complex mixture of numerous inorganic and organic compounds (including chlorinated compounds, nutrients, and heavy metals, etc.). The presence of all of these compounds makes remediation and cleanup difficult. In this study, the phytoremediation ability of Jatropha curcas and Pongamia pinnata was tested to remove nickel (Ni) and Zinc (Zn) from paper mill and municipal landfill contaminated soils, to understand the uptake potential and to estimate the accumulation pattern of Ni and Zn in the vegetative parts of the plant. The experiments were carried out in pots (3 kg capacity) and the different combinations of soil were made by mixing the contaminated soil with a reference soil (forest soil) as T 0 , T 25 , T 50 , T 75 and T 100. The plant biomass, chlorophyll content, proline, nitrate reductase activity and metal removal efficiency (%)were determined after 120 DAS (i.e., the days after sowing). The results of the study showed that with increasing metal stress, there is a reduction in the above-ground biomass content in both the plant species with a slightly less impact on the root biomass. Over a period of 4 months, J. curcas and P. pinnata removed 82–86% and 93-90% Ni, respectively. The removal of Zn was significantly less as compared to Ni as most of the Zn remained in the belowground part (roots) and in the soil. Besides, the phytostabilization capacities of the plants were calculated on the basis of their tolerance index (TI), bioaccumulation factor (BAF) and translocation factor (TF). The low BAF and TF values with increasing heavy metals (HMs) content indicates its higher phytostabilization capacity in the root and rhizospheric region as compared to phytoaccumulation. A cornerstone of the management and conservation of archaeological sites is recording their physical characteristics. Documenting and describing the site is an essential step that allows for delineating the components of the site and for collecting and synthesizing information and documentation (Demas, 2012). The information produced by such work assists in the decision-making process for custodians, site managers, public officials, conservators, and other related experts. Rigorous documentation may also serve a broader purpose: over time, it becomes the primary archival and monitoring record. Both scholars and the public use this information and interpret the site, and they can serve as a posterity record in the event of catastrophic or gradual loss of the heritage asset. In May 2018 the Getty Conservation Institute and the Department of Antiquities of Cyprus collaborated with the Carleton Immersive Media Studio in undertaking the documentation of Nea Paphos, a World Heritage site with very important mosaic pavements in the eastern Mediterranean. This contribution outlines the critical components of the documentation project: field study, field measurements, data processing, validation, GIS, and integration of external data. The current applications in the energy sector are based largely on fossil fuels which release greenhouse gas emissions to the atmosphere. To face the issue of global warming, the energy sector has to transfer to and develop sustainable energy solutions that do not release carbon emissions. This is one of the primary motivators for the SDEWES conference as well as for this review, and previous ones, examining the most recent works based on sustainable and green energy production in such fields. The 17th Conference on the Sustainable Development of Energy, Water, and Environment Systems (SDEWES) was held on 6–10 November 2022 in Paphos, Cyprus. The SDEWES conference aims at solving complex and ongoing concerns that approach a long-term perspective and supporting innovative solutions and continuous monitoring and evaluation. This review paper aims at collecting the main presented papers focused on the following hot topics: low-carbon technologies based on renewable and clean-energy systems, including mainly biomass, solar, and wind energy applications; energy storage systems; hydrogen-based systems; energy-saving strategies in buildings; and the adoption of smart management strategies using renewable energy systems. These topics are investigated in order to propose solutions to address the issues of climate change, water scarcity, and energy saving. From the analyzed works, we note that some key issues for sustainable development remain to be further addressed: such as novel and advanced energy storage systems, green hydrogen production, novel low-temperature district heating and cooling networks, novel solar technologies for the simultaneous production of power and high temperature heat, solar desalination for hydrogen production systems, and agrivoltaic systems for the production of power and food. Lead telluride and germanium telluride are well-known IV-VI semiconductors, which is now the focus of research due to the perspective of application as thermoelectrics for midrange temperatures. Solid solutions and heterostructures on this basis, obtained by molecular beam epitaxy, are a promising direction for the development of these materials. In this paper, we have focused on the Raman spectra excited by the 514.5 nm laser line (out of resonance) of PbTe, GeTe, (Pb, Ge)Te, and (Pb, Ge, Eu)Te layers grown on BaF2 (111) monocrystalline substrates. The obtained phonon properties are related to the properties of the corresponding bulk materials or can be explained by a model that takes into account the difference in the masses of the constituent elements only, as is the case with the local mode of Ge in PbTe (registered at about 181 cm−1). Multiphonon processes registered for this phonon are a consequence of the change in the electronic structure of PbTe and electron-phonon interaction. This paper presents an overview for the Special Issue of Clean Technology and Environmental Policy journal (CTEP), and it includes accepted papers from: 5th South East European Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from May 22–26, 2022, in Vlore, Albania. 3rd Latin American Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from July 24–28, 2022, in Sao Paulo, Brazil. 17th Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from November 6–10, 2022, in Paphos, Cyprus. Considering CTEP's policy of high-quality research papers, guest editors have invited 35 research articles, presented at the SDEWES 2022 conference. After a vigorous review process, 13 papers have been accepted for publication in this special issue. All 13 accepted papers are briefly presented in this overview together with a wider view that presents research efforts within the SDEWES community published through previous SDEWES special issues. This paper presents the results of ceramic synthesis in the field of a powerful flux of high-energy electrons on powder mixtures. The synthesis is carried out via the direct exposure of the radiation flux to a mixture with high speed (up to 10 g/s) and efficiency without the use of any methods or means for stimulation. These synthesis qualities provide the opportunity to optimize compositions and conditions in a short time while maintaining the purity of the ceramics. The possibility of synthesizing ceramics from powders of metal oxides and fluorides (MgF2, BaF2, WO3, Ga2O3, Al2O3, Y2O3, ZrO2, MgO) and complex compounds from their stoichiometric mixtures (Y3Al3O12, Y3AlxGa(5−x) O12, MgAl2O4, ZnAl2O4, MgWO4, ZnWO4, BaxMg(2−x) F4), including activators, is demonstrated. The ceramics synthesized in the field of high-energy electron flux have a structure and luminescence properties similar to those obtained by other methods, such as thermal methods. The results of studying the processes of energy transfer of the electron beam mixture, quantitative assessments of the distribution of absorbed energy, and the dissipation of this energy are presented. The optimal conditions for beam treatment of the mixture during synthesis are determined. It is shown that the efficiency of radiation synthesis of ceramics depends on the particle dispersion of the initial powders. Powders with particle sizes of 1–10 µm, uniform for the synthesis of ceramics of complex compositions, are optimal. A hypothesis is put forward that ionization processes, resulting in the radiolysis of particles and the exchange of elements in the ion–electron plasma, dominate in the formation of new structural phases during radiation synthesis. Cancer development is a complex process involving both genetic and epigenetic changes. The SWI/SNF (switch/sucrose non‐fermentable) chromatin remodelling complex, one of the most studied ATP‐dependent complexes, plays an important role in coordinating chromatin structural stability, gene expression and post‐translational modifications. The SWI/SNF complex can be classified into BAF, PBAF and GBAF according to their constituent subunits. Cancer genome sequencing studies have shown a high incidence of mutations in genes encoding subunits of the SWI/SNF chromatin remodelling complex, with abnormalities in one or more of these genes present in nearly 25% of all cancers, which indicating that stabilizing normal expression of genes encoding subunits in the SWI/SNF complex may prevent tumorigenesis. In this paper, we will review the relationship between the SWI/SNF complex and some clinical tumours and its mechanism of action. The aim is to provide a theoretical basis to guide the diagnosis and treatment of tumours caused by mutations or inactivation of one or more genes encoding subunits of the SWI/SNF complex in the clinical setting. Fast and heavy inorganic scintillators with suitable radiation tolerance are required to face the challenges presented at future hadron colliders of high energy and intensity. Up to 5 GGy and 5 × 1018 neq/cm2 of one-MeV-equivalent neutron fluence is expected by the forward calorimeter at the Future Hadron Circular Collider. This paper reports the results of an investigation of proton- and neutron-induced radiation damage in various fast and heavy inorganic scintillators, such as LYSO:Ce crystals, LuAG:Ce ceramics, and BaF2 crystals. The experiments were carried out at the Blue Room with 800 MeV proton fluence up to 3.0 × 1015 p/cm2 and at the East Port with one MeV equivalent neutron fluence up to 9.2 × 1015 neq/cm2, respectively, at the Los Alamos Neutron Science Center. Experiments were also carried out at the CERN PS-IRRAD proton facility with 24 GeV proton fluence up to 8.2 × 1015 p/cm2. Research and development will continue to develop LuAG:Ce ceramics and BaF2:Y crystals with improved optical quality, F/T ratio, and radiation hardness. [ABSTRACT FROM AUTHOR]

In this paper, Er3+ doped TeO 2 -ZnF 2 -BaF 2 -KF-Ta 2 O 5 tellurite glasses with low hydroxyl content (∼0.03 × 10−19 cm−3) were investigated employing both glass composition and glass melting process optimization. The Raman spectra and physical properties were characterized to analyze the structure of the glasses. Under the pumping of 980 nm LD laser, intense up-conversion fluorescence at 1.5 and 2.7 μm of samples and their lifetimes were detected and analyzed, and the related transition mechanisms with gradient-varying Er3+ doping concentrations were discussed. The maximum absorption and emission cross section at 2.7 μm was calculated to be 6.4 × 10−21 cm2 and 6.8 × 10−21 cm2, correspondingly, which were higher than those of traditional tellurite glasses. Using the calculated and measured spectroscopic parameters of bulk tellurite glass, a dual-wavelength pumping model was established to verify the feasibility of mid-infrared laser output in similar tellurite glass fiber. Experimental results support the assertion that the Er3+ doped tellurite glasses hold promise as a candidate laser gain medium for mid-infrared fiber laser systems. [ABSTRACT FROM AUTHOR]

Tuberculosis is a widespread disease all over the world. Early diagnosis plays a crucial role in the treatment of patients and can reduce the mortality rate. A new biosensor based on a binary photonic crystal is designed for the detection of mycobacterium tuberculosis bacteria in the blood. The structure air/(A/B)N/(Defect)/(A/B)N/air is employed, where A, B and N are the first and second layers and the number of periods, respectively. Layers A and B are chosen as GaAs and BaF2, respectively. The defect layer is the blood sample. The transfer matrix method (TMM) is utilized to simulate the structure. Some parameters, such as central wavelength, defect layer thickness and incident angle are optimized to maximize the sensitivity. The optimum sensitivity is found 3197.18 nm/RIU, which is extremely high when compared to the most recent papers published in this field. In addition, our sensor has a simple design, tunable fabrication and low cost and can be useful for the detection of many organisms that can exist in the blood. [ABSTRACT FROM AUTHOR]

The aim of the paper is to extend the standard set of ceramic vessels used for thermal processing of food with two forms: jugs and colanders. The paper presents the functional analysis of ceramic jugs and colanders made of cooking ware found in early Roman layers of the residential quarter of ancient Nea Paphos, excavated by the Polish Archaeological Mission of Warsaw University at Kato Paphos. The examination is based on the shape of the vessels, the fabric, as well as on specific traces of use visible on the pots, such as mineral deposit on the inside or soot on the outside surface. The thin-walled cooking pottery of the early to late Roman period originating from the area of Morphou Bay in the northern part of Cyprus, excavated by the Paphos Agora Project of the Jagiellonian University in Kraków, is studied in this paper in order to shed new light on the nature of the supply patterns of this extraordinary class of pottery. The collected data--on the macroscopic characteristic of the ware and shape--and a chronological analysis as well as quantitative study of the assemblage have indicated a continuous presence of this group in Nea Paphos, reflecting perhaps some sort of special social requirements combined with an economic background. In 2006 UNESCO report has identified soil loss as one of the main threats of climate change with possible impact to natural and cultural heritage. The study illustrated in this paper shows the results from geomatic perspective, applying an interdisciplinary approach undertaken in order to identify major natural hazards affecting cultural landscapes and archaeological heritage in rural areas in Cyprus. In particular, Earth Observation (EO) and ground-based methods were identified and applied for mapping, monitoring and estimation of the possible soil loss caused by soil erosion. Special attention was given to the land use/land cover factor (C) and its impact on the overall estimation of the soil-loss. Cover factor represents the effect of soil-disturbing activities, plants, crop sequence and productivity level, soil cover and subsurface bio-mass on soil erosion. Urban areas have a definite role in retarding the recharge process, leading to increased runoff and soil loss in the broader area. On the other hand, natural vegetation plays a predominant role in reducing water erosion. The land use change was estimated based on the difference of the NDVI value between Landsat 5 TM and Sentinel-2 data for the period between 1980s' until today. Cover factor was then estimated for both periods and significant land use changes were further examined in areas of significant cultural and natural landscape value. The results were then compared in order to study the impact of land use change on the soil erosion and hence on the soil loss rate in the selected areas. [ABSTRACT FROM AUTHOR]

This paper presents a research project which integrates technological tools for developing a complete system for monitoring and determining irrigation demand on a systematic basis in Cyprus. Such tools are multi-spectral remotely sensed data dynamic water budget simulation and optimization, crop evapotranspiration (ETc) models and microsensor technology. The main aim is to estimate ETc in Cyprus and, furthermore, to undertake the required measures for an effective irrigation water management in the future. Evapotranspiration is difficult to determine since it combines various meteorological and field parameters while in literature quite many different models for estimating ETc are put forward. The proposed wireless sensor network acts as a monitoring tool for providing measurements of the necessary parameters: meteorological, climatic data and other auxiliary parameters required by the irrigation model in order to determine the irrigation demand. Reflectance is determined directly from satellite images. Finally, using the WaterWare irrigation software, irrigation scheduling is planned for the area of interest in Paphos, Cyprus. This area is located at almost sea level and is characterized by mild micro-climate. The results of the paper refer to year 2009 and show the daily water requirements of the specific crop in study. A novel double‐cladding Ho3+/Tm3+ co‐doped Bi2O3–GeO2–Ga2O3–BaF2 glass fiber, which can be applied to a 2.0‐μm infrared laser, was fabricated by a rod‐tube drawing method. The thermal properties of the glass were studied by differential scanning calorimetry. It showed good thermal stability and matching thermal expansion coefficient for fiber drawing when Tx−Tg > 193°C and the maximum difference of the thermal expansion coefficient is 3.55 × 10−6/°C or less. The 2.0‐μm luminescence characteristics were studied using the central wavelength of 808 nm pump light excitation. The results show that when the concentration ratio of Ho3+/Tm3+ reaches 0.5 mol%:1.0 mol%, the maximum fluorescence intensity was obtained in the core glass, the emission cross section reached 10.09 × 10−21 cm2, and the maximum phonon energy was 751 cm−1. In this paper, a continuous laser output with a maximum power of 0.986 W and a wavelength of 2030 nm was obtained using an erbium‐doped fiber laser as a pump source in a 0.5 m long Ho3+/Tm3+ co‐doped glass fiber. In short, the results show that Ho3+/Tm3+ co‐doped 36Bi2O3–30GeO2–15Ga2O3–10BaF2–9Na2O glass fiber has excellent laser properties, and it is an ideal mid‐infrared fiber material for a 2.0‐μm fiber laser with excellent characteristics [ABSTRACT FROM AUTHOR]

Brazil's Atlantic Forest (BAF) is a highly fragmented, strategic environmental and socio-economic region that represents the fourth biodiversity hotspot while also producing many commodities that are exported globally. Human disturbance plays a pivotal role as a driver of BAF's soil dynamics and behaviors. The soils under Late Primary and Secondary Semideciduous Seasonal Forests (LPSF and LSSF) were characterized by high to moderate resilience, with improved chemical properties as human disturbance decreased. The Transitional Forest to Cerrado (TFC) had the worst soil conditions. Disturbed Primary and Secondary Semideciduous Seasonal Forests (DPSF and DSSF) represent a transitional stage between LPSF/LSSF and TFC. Accordingly, SOCs stocks increased from TFC << DPSF, DSSF < LPSF, LSSF. In BAF soils, to avoid unreliable data, SOCs measurements should be (i) conducted to at least 1 m soil depth and (ii) quantified with a CHN analyzer. Human disturbance strongly affected the positive feedback between vegetation succession, SOCs, and soil nutrition. Soil development decreased as human disturbance increased, thus negatively affecting SOCs. Soils in the BAF require a long time to recover after the end of human disturbance, thus suggesting that preservation strategies should be prioritized in remnant BAF fragments. We present sub-50 fs soliton pulse generation from a diode-pumped Kerr-lens mode-locked laser based on an Yb3+-doped BaF2 crystal. Utilizing a spatially single-mode, fiber-coupled InGaAs laser diode at 976 nm as a pump source, the Yb:BaF2 laser generates pulses as short as 46 fs at 1060.1 nm with an average output power of 45 mW at a pulse repetition rate of ~65.6 MHz via soft-aperture Kerr-lens mode locking. To the best of our knowledge, this represents the first demonstration of Kerr-lens mode-locked operation of the Yb:BaF2 crystal, as well as the shortest pulse duration ever achieved from any diode-pumped mode-locked laser based on an Yb3+-doped alkaline-earth fluoride crystal. Prior to the next generation sequencing and characterization of the tumor genome landscape, mutations in the SWI/SNF chromatin remodeling complex and the KEAP1-NRF2 signaling pathway were underappreciated. While these two classes of mutations appeared to independently contribute to tumor development, recent reports have demonstrated a mechanistic link between these two regulatory mechanisms in specific cancer types and cell models. In this work, we expand upon these data by exploring the relationship between mutations in BAF and PBAF subunits of the SWI/SNF complex and activation of NRF2 signal transduction across many cancer types. ARID1A/B mutations were strongly associated with NRF2 transcriptional activity in head and neck squamous carcinomas (HNSC). Many additional tumor types showed significant association between NRF2 signaling and mutation of specific components of the SWI/SNF complex. Different effects of BAF and PBAF mutations on the polarity of NRF2 signaling were observed. Overall, our results support a context-dependent functional link between SWI/SNF and NRF2 mutations across human cancers and implicate ARID1A inactivation in HPV-negative HNSC in promoting tumor progression and survival through activation of the KEAP1-NRF2 signaling pathway. Purpose: Acute lung injury (ALI) is a severe pulmonary disease characterized by damage to the alveoli and pulmonary blood vessels, leading to severe impairment of lung function. Studies on the effect of capsaicin (8-methyl-N-geranyl-6-nonamide, CAP) on lipopolysaccharide (LPS)-induced ALI in bronchial epithelial cells transformed with Ad12-SV40 2B (BEAS-2B) are still limited. This study aimed to investigate the effect and specific mechanism by which CAP improves LPS-induced ALI.Methods: The present study investigated the effect of CAP and the potential underlying mechanisms in LPS-induced ALI in vitro and vivo via RNA sequencing, Western blotting (WB), quantitative real-time reverse transcription PCR (qRT‒PCR), enzyme-linked immunosorbent assay (ELISA), and transmission electron microscopy (TEM). The TRPV1 inhibitor AMG9810 and the AKT agonist SC79 were used to confirm the protective effect of the TRPV1/AKT axis against ALI. The autophagy agonist rapamycin (Rapa) and the autophagy inhibitors 3-methyladenine (3-MA) and bafilomycin A1 (Baf-A1) were used to clarify the characteristics of LPS-induced autophagy.Results: Our findings demonstrated that CAP effectively suppressed inflammation and autophagy in LPS-induced ALI, both in vivo and in vitro. This mechanism involves regulation by the TRPV1/AKT signaling pathway. By activating TRPV1, CAP reduces the expression of P-AKT, thereby exerting its anti-inflammatory and inhibitory effects on pro-death autophagy. Furthermore, prior administration of CAP provided substantial protection to mice against ALI induced by LPS, reduced the lung wet/dry ratio, decreased proinflammatory cytokine expression, and downregulated LC3 expression.Conclusion: Taken together, our results indicate that CAP protects against LPS-induced ALI by inhibiting inflammatory responses and autophagic death through the TRPV1/AKT signaling pathway, presenting a novel strategy for ALI therapy. ATP-dependent chromatin remodeling SWI/SNF complexes exist in three subcomplexes: canonical BAF (cBAF), polybromo BAF (PBAF), and a newly described non-canonical BAF (ncBAF). While cBAF and PBAF regulate fates of multiple cell types, roles for ncBAF in hematopoietic stem cells (HSCs) have not been investigated. Motivated by recent discovery of disrupted expression of BRD9, an essential component of ncBAF, in multiple cancers, including clonal hematopoietic disorders, we evaluate here the role of BRD9 in normal and malignant HSCs. BRD9 loss enhances chromatin accessibility, promoting myeloid lineage skewing while impairing B cell development. BRD9 significantly colocalizes with CTCF, whose chromatin recruitment is augmented by BRD9 loss, leading to altered chromatin state and expression of myeloid-related genes within intact topologically associating domains. These data uncover ncBAF as critical for cell fate specification in HSCs via three-dimensional regulation of gene expression and illuminate roles for ncBAF in normal and malignant hematopoiesis. BRD9 is a core non-canonical BAF component. Here the authors show that BRD9 plays a pivotal role in regulating the disease-related cell fate of hematopoietic stem cells. Malignant rhabdoid tumor (MRT) is a highly malignant and often lethal childhood cancer. MRTs are genetically defined by bi-allelic inactivating mutations in SMARCB1, a member of the BRG1/BRM-associated factors (BAF) chromatin remodeling complex. Mutations in BAF complex members are common in human cancer, yet their contribution to tumorigenesis remains in many cases poorly understood. Here, we study derailed regulatory landscapes as a consequence of SMARCB1 loss in the context of MRT. Our multi-omics approach on patient-derived MRT organoids reveals a dramatic reshaping of the regulatory landscape upon SMARCB1 reconstitution. Chromosome conformation capture experiments subsequently reveal patient-specific looping of distal enhancer regions with the promoter of the MYC oncogene. This intertumoral heterogeneity in MYC enhancer utilization is also present in patient MRT tissues as shown by combined single-cell RNA-seq and ATAC-seq. We show that loss of SMARCB1 activates patient-specific epigenetic reprogramming underlying MRT tumorigenesis. The regulatory landscape of malignant rhabdoid tumor (MRT) due to SMARCB1 loss remains to be explored. We are living in the 'plasticene' era. Marine plastic pollution is a global problem, and increasing effort and funds are being invested towards identifying and implementing solutions. It is well accepted that the implemented solutions should take into account local specificities and culture, which requires the involvement and engagement of local decision-makers and key actors. This paper presents how the DeCyDe-4 method, which is based on a participatory decision-support concept, was implemented to develop community-based policy tools and action plans to combat marine litter. Through a presentation of the step-by-step implementation of the method at a 3-day workshop in the Paphos District of Cyprus, the DeCyDe-4 method is unfolded so as to showcase how a multi-disciplinary group of local stakeholders can co-create a Policy Tool, and identify and prioritize measures and solutions to marine litter. The case study shows that DeCyDe-4 can support a varied group of stakeholders to negotiate a complex environmental problem such as marine litter, and to make informed, unbiased decisions, through a process that encourages active participation and dialogue, so that opposite opinions are heard, consensus is reached and ownership of the solutions is achieved. The subject of this paper is a reconstruction of the architectural decoration of a façade of the House of Aion in Nea Paphos. During an excavation carried out in 1997 several pieces of decorated architectural elements were uncovered in room 19, among others the fragments of an arch, a lintel, an engaged column, an impost and two consoles. Those blocks served as a base for the reconstruction of the architectural frame of the main gateway. It took the form of a cantilevered, blind arcade of five spans erected above the main gate. Each span consisted of two engaged columns supporting an arch with a shallow niche underneath it, probably for a kind of decorative element. The reconstruction was based on similar architectural details known from the main room of the House of Aion, the Porta Aurea at the palace of Diocletian in Split, and the western façade of the Felix Romuliana Palace in Gamzigrad. During the 2014 season a late Hellenistic well was explored as part of the excavations on the Nea Paphos Agora. Many special finds, including ceramic vessels, iron and bronze items, and lamps were found. Moreover, on the bottom of the well a piece of textile attached to metal application was discovered. Oil lamps as archaeological finds and in museum collections provide a wealth of information. Various types of studies may be applied to investigate their meaning in ancient times. When several methods are used simultaneously, the objects may be interpreted according to distinct aspects, enabling us to study this group of ceramic objects from a multidisciplinary, comprehensive perspective. TeO2-BaF2-Er2O3-Dy2O3 laser glasses were prepared using the melt-quenching method. The bound water that can capture the excited state energy was reduced by physical and chemical methods. We did not observe a significant Er3+ emission peak at 2.7 μm in fluorescence spectra, which may be due to the efficient energy transfer process (ET2). Meanwhile, we found a broadband gain span of approximately 400 nm in fluorescence spectra at the 2.85 μm band, attributed to the 'vector summation' of the energy level radiation transition and the change of the glass network. Subsequently, we explored the structural properties of the glass. The results indicated that the Gaussian peak located at 250 cm−1 drifts toward 370 cm−1, which may be caused by the fracture or recombination of Te-O-Te and a decrease in the bridge oxygen content with the increasing concentration of Er2O3. The topology cage structure around the luminescence center of rare earth ions is changed and the stability of the optically active center is enhanced, finally contributing to the enhancement of luminescence. Meanwhile, the maximum σemi and gain coefficient of Dy3+ reach up to 7.22 × 10−21 cm2 and 7.37 cm−1, respectively. The comprehensive results show that the fluorotellurite glass designed in this study is expected to be a gain medium for mid-infrared lasers in remote sensing monitoring, military, and other fields. Group 3 medulloblastoma is one of the most aggressive types of childhood brain tumors. Roughly 30% of cases carry genetic alterations in MYC, SMARCA4, or both genes combined. While overexpression of MYC has previously been shown to drive medulloblastoma formation in mice, the functional significance of SMARCA4 mutations and their suitability as a therapeutic target remain largely unclear. To address this issue, we combined overexpression of MYC with a loss of SMARCA4 in granule cell precursors. Both alterations did not increase proliferation of granule cell precursors in vitro. However, combined MYC overexpression and SMARCA4 loss successfully induced tumor formation in vivo after orthotopic transplantation in recipient mice. Resulting tumors displayed anaplastic histology and exclusively consisted of SMARCA4-negative cells although a mixture of recombined and non-recombined cells was injected. These observations provide first evidence for a tumor-promoting role of a SMARCA4 deficiency in the development of medulloblastoma. In comparing the transcriptome of tumors to the cells of origin and an established Sonic Hedgehog medulloblastoma model, we gathered first hints on deregulated gene expression that could be specifically involved in SMARCA4/MYC driven tumorigenesis. Finally, an integration of RNA sequencing and DNA methylation data of murine tumors with human samples revealed a high resemblance to human Group 3 medulloblastoma on the molecular level. Altogether, the development of SMARCA4-deficient medulloblastomas in mice paves the way to deciphering the role of frequently occurring SMARCA4 alterations in Group 3 medulloblastoma with the perspective to explore targeted therapeutic options. A series of CoCrFeNiMn high-entropy alloy matrix self-lubricating composites were prepared by spark plasma sintering. The composites are composed of an FCC phase, Cr7C3, Ag, and eutectic fluoride BaF2/CaF2 phases. The microstructure of the composites is uniform. The additional phases distribute along the boundary of equiaxed grains of the FCC phase. The compressive yield strength and fracture toughness decrease with the increase of eutectic fluoride BaF2/CaF2. The composites are susceptible to brittle cleavage fracture. [ABSTRACT FROM AUTHOR]

Alternative techniques have been investigated for effectiveness in caries removal because conventional metallic dental burs can lead to an excessive loss of sound tissue. The aim of the present study is to realize a preliminary approach in obtaining effective polymer mixtures for polymeric bur development, capable of removing primary dental caries using combinations of polymers to ensure the requirements for such instruments, but also a greater compatibility with the teeth structure. This study assessed the main mechanical properties, water sorption, solubility and microscopic structure of four new polymer mixture recipes to provide essential features in obtaining experimental self-limited dental burs. Two mixtures have in their composition polymer mixtures of Bis-phenol A diglycidyl ether dimethacrylate/Triethylene glycol dimethacrylate/Urethane dimethacrylates (R1, R2), and two other mixtures have Bis-phenol A diglycidyl ether dimethacrylate/Polymethyl methacrylate/Methyl methacrylates (R3, R4). The incorporation of nanoparticles into the polymer matrix has become essential due to the need of polymer biocompatibility increasing along with teeth surface remineralization, so that the powder charge was added to four recipes, such as 5% glass with BaF2 and 0.5% graphene with silver particles. All data sets were analyzed using the One-Way ANOVA test. R3, R4 showed higher compressive strength and diametrical compression values; these values increased when glass and graphene were added. Moreover, the addition of glass particles lead to an increase in flexural strength. Regarding the sorption, sample R3 had the most significant differences between day 69 and the rest of the investigation days, while the solubility varied at different intervals. From the mechanical evaluation, we could conclude that the Bis-GMA/PMMA/MMA mixtures fit the mechanical characteristics supported by polymer burs, following future studies regarding their use on the affected dentin. Q960 high-strength steel is widely used in pressure vessels, bridges, offshore platforms and other important steel structural components because of its high strength and good plastic toughness, but alloy elements added to this kind of steel have strong hardenability, especially after welding, so the strength and toughness cannot meet the requirements, which limits its application in a wider range. In this paper, from the point of view of the metallurgical treatment of the weld, the goal is to improve the strength and toughness of the Q960 high strength steel weld metal In order to analyze the influence of Zr on the welding process of Q960 steel and the strengthening and toughening effect of weld metal, this paper takes Fe-Mn-Mo-Cr-Ni as the main alloy system, BaF2-CaF2-Al-Mg as the basic slag system, and adopts the method of melting consumable electrode self-shielded for welding, and analyzes the welding process, microstructure, tensile property and impact toughness of the welded joint. The experimental results show that when the weld metal contains 0.0061% Zr, the minimum spatter rate is only 7%, the maximum slag removal rate is 95%, the maximum hardness is 357HV, the maximum elongation is 34%, and the impact toughness is the highest. At this time, the acicular ferrite content in the weld microstructure is the highest, and there is a certain amount of equiaxed fine-grained ferrite, and the content of proeutectoid ferrite is the least, which effectively improves the strength and toughness of the weld metal. The Australian archaeological mission to Nea Paphos in Cyprus has begun exploring the remains of a structure to the rear of the cavea of the ancient theatre on the southeastern edge of the hill known as Fabrika. The preliminary results of this excavation suggest a significantly large post-medieval building constructed using stone taken from the former theatre. The building appears to have been used in some sort of industrial production, the precise nature of which is not yet determined, and is perhaps the source of the etymology of the name of the hill. This paper discusses a need for a reassessment of this area of the city of Nea Paphos in its more recent history, and discusses the needs for a better understanding of the urban relationship between Fabrika and the theatrical precinct during the Hellenistic and Roman periods. The aim of image-based localization (IBL) is to localize the real location of query image by matching reference image in database with GNSS-tags. Popular methods related to IBL commonly use street-level images, which have high value in practical application. Using street-level image to tackle IBL task has the primary challenges: existing works have not made targeted optimization for urban IBL tasks. Besides, the matching result is over-reliant on the quality of image features. Methods should address their practicality and robustness in engineering application, under metropolitan-scale. In response to these, this paper made following contributions: firstly, given the critical of buildings in distinguishing urban scenes, we contribute a feature called Building-Aware Feature (BAF). Secondly, in view of negative influence of complex urban scenes in retrieval process, we propose a retrieval method called Patch-Region Retrieval (PRR). To prove the effectiveness of BAF and PRR, we established an image-based localization experimental framework. Experiments prove that BAF can retain the feature points that fall on the building, and selectively lessen the feature points that fall on other things. While this effectively compresses the storage amount of feature index, we can also improve recall of localization results; implemented in the stage of geometric verification, PRR compares matching results of regional features and selects the best ranking as final result. PRR can enhance effectiveness of patch-regional feature. In addition, we fully confirmed the superiority of our proposed methods through a metropolitan-scale street-level image dataset. Background: Disrupted intestinal epithelial barrier is one of the major causes of Crohn's disease (CD). Novel molecular targets for intestinal epithelial barrier are essential to treatment of CD. Transmembrane and immunoglobulin domain-containing protein 1 (TMIGD1) is an adhesion molecule that regulates cell adhesion, migration, and enterocyte differentiation. However, the function and mechanism of TMIGD1 in CD and intestinal epithelial barrier has rarely been studied. Furthermore, the association between TMIGD1 and the clinical features of CD remains unclear. Methods: Transcriptome analysis on colonic mucosa from CD patients and healthy individuals were performed to identify dysregulated genes. Multi-omics integration of the 1000IBD cohort including genomics, transcriptomics of intestinal biopsies, and serum proteomics identified the association between genes and characteristics of CD. Inflammation was assessed by cytokine production in cell lines, organoids and intestinal-specific Tmigd1 knockout (Tmigd1INT-KO) mice. Epithelial barrier integrity was evaluated by trans-epithelium electrical resistance (TEER), paracellular permeability, and apical junction complex (AJC) expression. Co-immunoprecipitation, GST pull-down assays, mass spectrometry, proteomics, and transcriptome analysis were used to explore downstream mechanisms. Results: Multi-omics integration suggested that TMIGD1 was negatively associated with inflammatory characteristics of CD. TMIGD1 was downregulated in inflamed intestinal mucosa of patients with CD and mice colitis models. Tmigd1INT-KO mice were more susceptible to chemically induced colitis. In epithelial cell lines and colonic organoids, TMIGD1 knockdown caused impaired intestinal barrier integrity evidenced by increased paracellular permeability and reduced TEER and AJC expression. TMIGD1 knockdown in intestinal epithelial cells also induced pro-inflammatory cytokine production. Mechanistically, TMIGD1 directly interacted with cytoplasmic BAF nuclear assembly factor 1 (BANF1) to inhibit NF-κB activation. Exogenous expression of TMIGD1 and BANF1 restored intestinal barrier function and inhibited inflammation in vitro and in vivo. TMIGD1 expression predicted response to anti-TNF treatment in patients with CD. Conclusions: Our study demonstrated that TMIGD1 maintained intestinal barrier integrity and inactivated inflammation, and was therefore a potential therapeutic target for CD. [ABSTRACT FROM AUTHOR]

Simple Summary: The generation of an artificial intestine could minimize the use of in vivo trials, which are presently necessary for testing aquafeeds. As well as being lengthy and expensive, they also require the use of many animals. Furthermore, although in vivo screening allows for a precise evaluation of the final effect of each feed, it does not improve our basic knowledge of the cellular and molecular mechanisms that determine the end results. This, in turn, severely limits our ability to understand and predict the biological value of ingredients, both individually and in differing combinations thereof. To circumvent these constraints, we previously derived and characterized two novel cell lines from the proximal (RTpi-MI) and distal intestine (RTdi-MI) of rainbow trout (Oncorhynchus mykiss), demonstrating that the two cell lines retain the main properties of the intestinal tracts from which they originate. In the present work, we used these cells to set up a functional intestinal barrier on a permeable membrane support to investigate the digestion and absorption of dietary amino acids in fish feed. The results demonstrate that the procedure we developed enables the quantification of individual amino acids that cross the intestinal wall. This new platform will now be used to screen and select new feed ingredients/formulations. The ever-increasing number and variation of raw materials utilized to provide alternative feed formulations continues to allow for a more sustainable and flexible approach. Testing all these options in vivo is still the most robust and reliable manner to pick the best raw material candidates, but it requires the use of large numbers of animals and is time-consuming and expensive. Therefore, we are developing an in vitro platform that can provide a reliable evaluation of new ingredients. The main aim of this work was to combine an in vitro digestion protocol of extruded, commercially relevant aquafeeds with the exposure of intestinal epithelial cells to the extracted bio-available fraction (BAF). The results show that 250,000 cells/cm2 represents the optimal seeding density and that up to 50% BAF concentration for up to 24 h had no negative effects on the epithelial barrier morphology and function. It is possible to determine amino acid digestibility and bioavailability in all the experimental conditions (with and without BSA, at 25% and 50% dilution) and at all time points (0, 6, and 24 h). However, BAF concentration, the medium used for its dilution, and the length of exposure to the different epithelial cell lines can all influence the results and, therefore, must be selected according to the final aim of the experiment. Heavy metals (HMs) have the characteristics of high toxicity, long residual time, and difficult degradation, which pose a potential threat to aquatic ecosystems. The distribution and migration of HMs in different media can reflect their potential impact on aquatic ecosystem health. In this study, we analyzed the distribution characteristics of seven HMs (Zn, Cu, Cr, Ni, Hg, As, and Pb) in the water and sediment and five groups of organisms (fish, Crustacean, cephalopod, bivalvia, and gastropoda) in the sea ranching area of Haizhou Bay and explored the regularity of HM accumulation from water and sediments to organisms. The results showed that in the water, Zn and Hg had the smallest concentrations in the nearshore area, Cr had the lowest concentrations in the offshore area, and As, Cu, Pb, and Ni had the lowest concentrations in the central area. In the sediment, Hg had the smallest concentrations in the nearshore area and As, Pb, Cr, Cu, Ni, and Zn had the lowest concentrations in the offshore area; the average contents of seven heavy metals all meet the Class I standard of sediments in China. The concentrations of Cu and Zn in crustaceans were significantly higher than those in fishes (p < 0.05), and As showed a higher enrichment effect in cephalopods. Under the influence of feeding habits and habitat environment, the BAF value of benthic crustaceans and bivalvia accumulated HMs from sediments is high, and the BAF value of fish accumulated HMs from water and sediments is low. Overall, the concentration of HMs in water is high, and HMs in sediments are in good condition, crustaceans and bivalviaa have a strong ability to accumulate HMs from water and sediments, while fish are weak. Selenium (Se) is an essential microelement for human or animal health. At high concentrations, it can cause Se poisoning. Human activities (such as coal burning and mining) threaten soil biota by mobilizing high levels of Se. We used the earthworm Eisenia fetida as a bio-indicator of environmental pollutants to investigate Se acute toxicity, enrichment, and distribution through exposure tests using filter paper, artificial soil and cow manure. The 24 h- and 48 h-LC 50 for the filter paper contact test were 2.7 and 1.52 μg/cm2. In artificial soil test, the 14 d-LC 50 and 14 d-biomass inhibition concentration (IC 20) were 63.86 and 59.81 mg/kg, respectively. The cow manure resulted in a 2.2- and 2.6-fold higher LC 50 and IC 20 than artificial soil results, respectively. Earthworms accumulated the largest Se load (89.47 mg/kg) in artificial soil containing 80 mg Se/kg and only accumulated 90.3 mg/kg in cow manure containing 160 mg Se/kg; 46.6–60.59% of the total Se was distributed in the tail of E. fetida. The Se enrichment rate (SER Se) and bioaccumulation factor (BAF Se) scored higher in artificial soil than in cow manure with the same Se concentration exposure, and the highest SER Se was 6.21 and 6.31 mg Se/kg earthworm/d, respectively. The highest BAF Se was 1.49 in artificial soil and 0.75 in cow manure. Our results demonstrate that selenite is more toxic to earthworms living in artificial soil than in cow manure. E. fetida possesses certain Se detoxification mechanisms by distributing Se in the tail. Abstract In this paper, BaAl 11 O 16 N (BAON) based oxynitride phosphors have been synthesized via the employment of the flux method and Eu2+/Mn2+ co-doped technique. The effects of different fluxes on phase formation of the phosphor have been investigated. It has been found that BaF 2 is the best flux to prepare BAON as it can decrease the pure phase-forming temperature (∼1550 °C) than the normal solid state synthesis (∼1700 °C). It also gives the best luminescence quantum efficiency. The crystallinity, morphology and grain size of the phosphor can be improved when more BaF 2 flux has been added. The photoluminescence (PL) spectra and decay curves for Eu2+/Mn2+co-doped BAON phosphors reveal that an efficient energy transfer (ET) from Eu2+ to Mn2+ takes place in the BAON host. The corresponding ET efficiencies and mechanisms are evaluated and the critical distance (R c) is calculated in terms of the experimental data and analysis. The emission colors of the co-doped phosphors can be tuned from blue to green by simply varying the doping concentrations of Mn2+. White light-emitting diodes (WLEDs) are fabricated by coating the as-prepared samples on a near ultraviolet (n- UV) LED chip to explore the real application potentials. The excellent performance of this WLEDs verifies that the co-doped phosphor can be used as an efficient green component. Abstract In this paper, we studied the gamma photon attenuation properties for different germanate glasses systems with composition Sb 2 O 3 –GeO 2 –SiO 2 –Al 2 O 3 , TeO 2 –WO 3 –GeO 2 , GeO 2 –TeO 2 /SiO 2 –PbO–CaO–Tm 2 O 3 and B 2 O 3 –GeO 2 –Gd 2 O 3 –Eu 2 O 3 –GdF 3 –BaF 2. The mass attenuation coefficients (μ/ρ) for the selected germanate glasses were calculated at different energies (between 0.015 MeV and 15 MeV) using the MCNPX code. The calculated MCNPX μ/ρ values then compared with the WinXCom data to check the accuracy in the results. The results showed that the Sb 2 O 3 replacement by SiO 2 causes a decrease in μ/ρ for Sb 2 O 3 –GeO 2 –SiO 2 –Al 2 O 3 glass system. Also, it is found that for TeO 2 –WO 3 –GeO 2 glass system, μ/ρ decreases as GeO2 content increases from 1 to 20 mol%. While for GeO 2 –TeO 2 /SiO 2 –PbO–CaO–Tm 2 O 3 glass system, the results revealed that the glass samples contain TeO 2 possess higher μ/ρ values than the samples contain SiO 2. For the B 2 O 3 –GeO 2 –Gd 2 O 3 –Eu 2 O 3 –GdF 3 –BaF 2 glasses, we found that the μ/ρ values decrease as BaF 2 is increased from 0 to 15 mol%. Moreover, it is found that 50Sb-0Ge for series 1, TWG0 for series 2, 2G4T for series 3 and X = 0 for series 4 glass samples among the studied glasses have the highest effective atomic number values. On the basis of the results achieved, 50Sb-0Ge, TWG0, 2G4T and X = 0 samples have the best radiation shielding characteristics among the selected glasses. Growing concerns over environmental issues and sustainable living have resulted in increased interest in renewable energy and energy efficiency. The real estate market is no exception, with homeowners increasingly considering the market value of green and sustainable buildings, which can offer both energy efficiency and potential health benefits. This study investigates the level of interest among homeowners in investing in renewable energy sources and energy efficiency measures for their homes and how it relates to their perception of the market value of green or sustainable buildings in the real estate market. A survey was conducted in the Paphos urban complex in Cyprus, with 180 participants over the age of 18. The participants were selected through a random sampling method and were representative of the general population in terms of gender, age, and income. Data were collected on their attitudes towards renewable energy sources and energy efficiency, as well as their perceptions of the market value of green buildings. The data collected were analyzed using various statistical methods, including Cronbach's α coefficient, the non-parametric Friedman test, descriptive statistics, and factor analysis, with the Statistical Package for the Social Sciences (SPSS) being used for coding and analysis. Results indicate that 64% of the homeowners surveyed were interested in investing in renewable energy sources, and 72% were interested in energy efficiency measures. Additionally, findings suggest a moderate level of interest (58%) among homeowners in investing in renewable energy sources and that this is positively associated with their perception of the market value of green buildings. Furthermore, homeowners with higher income and education levels tend to be more interested in investing in renewable energy sources and energy efficiency measures and perceive green buildings as having higher market value. This study provides insights into the factors that drive homeowners' investment in renewable energy sources and energy efficiency measures, shedding light on the relationship between homeowners' perceptions of the market value of green buildings and their interest in such investments. Diffuse midline gliomas (DMGs) including diffuse intrinsic pontine gliomas (DIPGs) bearing lysine-to-methionine mutations in histone H3 at lysine 27 (H3K27M) are lethal childhood brain cancers. These tumors harbor a global reduction in the transcriptional repressive mark H3K27me3 accompanied by an increase in the transcriptional activation mark H3K27ac. We postulated that H3K27M mutations, in addition to altering H3K27 modifications, reprogram the master chromatin remodeling switch/sucrose nonfermentable (SWI/SNF) complex. The SWI/SNF complex can exist in two main forms termed BAF and PBAF that play central roles in neurodevelopment and cancer. Moreover, BAF antagonizes PRC2, the main enzyme catalyzing H3K27me3. We demonstrate that H3K27M gliomas show increased protein levels of the SWI/SNF complex ATPase subunits SMARCA4 and SMARCA2, and the PBAF component PBRM1. Additionally, knockdown of mutant H3K27M lowered SMARCA4 protein levels. The altered lysosomal function can induce drug redistribution which leads to drug resistance and poor prognosis for cancer patients. V-ATPase, an ATP-driven proton pump positioned at lysosomal surfaces, is responsible for maintaining the stability of lysosome. Herein, we reported that the potassium voltage-gated channel subfamily J member 15 (KCNJ15) protein, which may bind to V-ATPase, can regulate the function of lysosome. The deficiency of KCNJ15 protein in breast cancer cells led to drug aggregation as well as reduction of drug efficacy. The application of the V-ATPase inhibitor could inhibit the binding between KCNJ15 and V-ATPase, contributing to the amelioration of drug resistance. Clinical data analysis revealed that KCNJ15 deficiency was associated with higher histological grading, advanced stages, more metastases of lymph nodes, and shorter disease free survival of patients with breast cancer. KCNJ15 expression level is positively correlated with a high response rate after receiving neoadjuvant chemotherapy. Moreover, we revealed that the small molecule drug CMA/BAF can reverse drug resistance by disrupting the interaction between KCNJ15 and lysosomes. This work aimed to detect changes in trace element chemical speciation in sewage sludge (SS) after the ozonation process. The modified Community Bureau of Reference (BCR) sequential extraction procedure was performed to determine the chemical speciation of trace elements in SS after the ozonation process. To assess potential soil contamination with trace elements from sewage sludge after the ozonation process, the risk assessment code (RAC) coefficient was used. The bioaccumulation factor (BAF) and translocation factor (TF) values were also calculated to characterize the efficiency of trace element accumulation in the studied plant species from soil fertilized with sludges after the ozonation process. Generally, the mean concentration of total trace elements in the SS after the ozonation process was higher, but the differences were statistically significant only in the case of Mn, Cu, Pb, and Cd. The dominant fraction of Fe, Cr, Pb, and Cd was the residual fraction F4, while the extractable/exchangeable fraction F1 was present in the smallest amount. Therefore, in the case of Mn, Zn, and Ni, the ozonation process had a significant impact on the increase in the content of these elements in the F1 fraction. The application of the SS stabilized by ozonation process for maize and wheat fertilization did not significantly affect the bioaccumulation of most of the analyzed metals in aboveground biomass. Higher values of BAF coefficients after the application of ozonated SS were found only in the case of Cu and Ni. In turn, the determined TF coefficients were lower than 1 in most cases. The obtained results showed that the slight change in the concentration of Zn, Mn, and Ni in fraction F1 causes a specific risk of their mobility in the soil environment. It should be noted that due to the variable composition of sewage sludge, an analysis of the content of individual trace elements in chemical fractions should be carried out to assess its actual impact on the environment. Gammaherpesviruses, including Kaposi sarcoma-associated herpesvirus (KSHV) and Epstein-Barr virus (EBV), are DNA viruses that are globally associated with human cancers and establish lifelong latency in the human population. Detection of gammaherpesviral infection by the cGAS-STING innate immune DNA-sensing pathway is critical for suppressing viral reactivation from latency, a process that promotes viral pathogenesis and transmission. We report that barrier-to-autointegration factor 1 (BAF)-mediated suppression of the cGAS-STING signaling pathway is necessary for reactivation of KSHV and EBV. We demonstrate a role for BAF in destabilizing cGAS expression and show that inhibiting BAF expression in latently infected, reactivating, or uninfected cells leads to increased type I interferon-mediated antiviral responses and decreased viral replication. Furthermore, BAF overexpression resulted in decreased cGAS expression at the protein level. These results establish BAF as a key regulator of the lifecycle of gammaherpesviruses and a potential target for treating viral infections and malignancies. Although China has been the main manufacturer and consumer of phthalate esters (PAEs), human health ambient water quality criteria (AWQCs) have not been proposed for these chemicals. In this study, the distribution and bioaccumulation of six PAEs (dimethyl phthalate (DMP), diethyl phthalate (DEP), di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), bis (2-ethylhexyl) phthalate (DEHP), and di-n-octyl phthalate (DnOP)) were investigated in 11 edible fish species collected from Poyang Lake, China. The results showed that the total concentrations of the six PAEs in the fish ranged between 118.63 and 819.84 μg/kg wet weight (mean of 327.50 ± 190.44 μg/kg). DMP, DEP, DBP, and DEHP were detected in all samples, of which DEHP and DBP were two of the most predominant phthalates, accounting for more than 90% of the total PAEs. The DEHP concentrations in fish with different habitat preferences were different, demersal species were significantly higher than pelagic species (p < 0.05). The mean natural logarithmic bioaccumulation factors (log BAFs) of PAEs increased with increasing lipophilicity of the substances, which yielded the following regression equation: log BAF (L/kg) = 0.103 log Kow + 2.158 (r2 = 0.940, p < 0.05, n = 4). Using this quantitative structure–activity relationship to calculate BAFs for the remaining undetected substances (BBP and DnOP) to derive AWQCs. According to the natural parameters, the human health AWQCs relating to PAE concentrations for water and fish consumption were derived as 9.4 × 103 (DMP), 5.0 × 102 (DEP), 4.2 × 101 (DBP), 1.1 (BBP), 8.6 × 10− 2 (DEHP), and 2.0 (DnOP) μg/L. Human health risk assessment indicated that the dietary intake of DEHP may exert a carcinogenic effect on residents of the Poyang Lake region. Mutations in DNAJC5/CSPα are associated with adult neuronal ceroid lipofuscinosis (ANCL), a dominant-inherited neurodegenerative disease featuring lysosome-derived autofluorescent storage materials (AFSMs) termed lipofuscin. Functionally, DNAJC5 has been implicated in chaperoning synaptic proteins and in misfolding-associated protein secretion (MAPS), but how DNAJC5 dysfunction causes lipofuscinosis and neurodegeneration is unclear. Here we report two functionally distinct but coupled chaperoning activities of DNAJC5, which jointly regulate lysosomal homeostasis: While endolysosome-associated DNAJC5 promotes ESCRT-dependent microautophagy, a fraction of perinuclear and non-lysosomal DNAJC5 mediates MAPS. Functional proteomics identifies a previously unknown DNAJC5 interactor SLC3A2/CD98hc that is essential for the perinuclear DNAJC5 localization and MAPS but dispensable for microautophagy. Importantly, uncoupling these two processes, as seen in cells lacking SLC3A2 or expressing ANCL-associated DNAJC5 mutants, generates DNAJC5-containing AFSMs resembling NCL patient-derived lipofuscin and induces neurodegeneration in a Drosophila ANCL model. These findings suggest that MAPS safeguards microautophagy to avoid DNAJC5-associated lipofuscinosis and neurodegeneration. BAF and PBAF are mammalian SWI/SNF family chromatin remodeling complexes that possess multiple histone/DNA-binding subunits and create nucleosome-depleted/free regions for transcription activation. Despite previous structural studies and recent advance of SWI/SNF family complexes, it remains incompletely understood how PBAF-nucleosome complex is organized. Here we determined structure of 13-subunit human PBAF in complex with acetylated nucleosome in ADP-BeF3-bound state. Four PBAF-specific subunits work together with nine BAF/PBAF-shared subunits to generate PBAF-specific modular organization, distinct from that of BAF at various regions. PBAF-nucleosome structure reveals six histone-binding domains and four DNA-binding domains/modules, the majority of which directly bind histone/DNA. This multivalent nucleosome-binding pattern, not observed in previous studies, suggests that PBAF may integrate comprehensive chromatin information to target genomic loci for function. Our study reveals molecular organization of subunits and histone/DNA-binding domains/modules in PBAF-nucleosome complex and provides structural insights into PBAF-mediated nucleosome association complimentary to the recently reported PBAF-nucleosome structure. Haemoglobinopathies, including thalassaemias and sickle-cell syndromes, are demanding, lifelong conditions that pose a significant burden to patients, families, and healthcare systems. Despite the therapeutic advances and the resulting improvements in prognosis accomplished in past decades, these patients still face important challenges, including suboptimal access to quality care in areas with developing economies, changing epidemiology due to massive migration flows, an evolving clinical spectrum due to ageing in well-treated patients, and limited access to novel high-cost therapies. We herein describe the organization of healthcare services for haemoglobinopathies in Cyprus--with particular focus on beta-thalassaemia, the most prevalent condition in this region--along with selected patient outcomes. This report aims at underscoring the fact that nationally funded and well-coordinated prevention and care programmes for chronic and complex conditions, such as haemoglobinopathies, with active involvement from patient organizations lead to effective disease control and excellent outcomes in survival, quality of life, social adaptation, and public health savings, and allow timely and effective responses to emerging crises, such as the COVID-19 pandemic. The Cyprus paradigm could therefore serve as a blueprint for the organization or adaptation of haemoglobinopathy programs in other countries since these disorders are still widely occurring. Double-PHD fingers 3 (DPF3) is a BAF-associated human epigenetic regulator, which is increasingly recognised as a major contributor to various pathological contexts, such as cardiac defects, cancer, and neurodegenerative diseases. Recently, we unveiled that its two isoforms (DPF3b and DPF3a) are amyloidogenic intrinsically disordered proteins. DPF3 isoforms differ from their C-terminal region (C-TERb and C-TERa), containing zinc fingers and disordered domains. Herein, we investigated the disorder aggregation properties of C-TER isoforms. In agreement with the predictions, spectroscopy highlighted a lack of a highly ordered structure, especially for C-TERa. Over a few days, both C-TERs were shown to spontaneously assemble into similar antiparallel and parallel β-sheet-rich fibrils. Altered metal homeostasis being a neurodegeneration hallmark, we also assessed the influence of divalent metal cations, namely Cu2+, Mg2+, Ni2+, and Zn2+, on the C-TER aggregation pathway. Circular dichroism revealed that metal binding does not impair the formation of β-sheets, though metal-specific tertiary structure modifications were observed. Through intrinsic and extrinsic fluorescence, we found that metal cations differently affect C-TERb and C-TERa. Cu2+ and Ni2+ have a strong inhibitory effect on the aggregation of both isoforms, whereas Mg2+ impedes C-TERb fibrillation and, on the contrary, enhances that of C-TERa. Upon Zn2+ binding, C-TERb aggregation is also hindered, and the amyloid autofluorescence of C-TERa is remarkably red-shifted. Using electron microscopy, we confirmed that the metal-induced spectral changes are related to the morphological diversity of the aggregates. While metal-treated C-TERb formed breakable and fragmented filaments, C-TERa fibrils retained their flexibility and packing properties in the presence of Mg2+ and Zn2+ cations. [ABSTRACT FROM AUTHOR]
