BackgroundThe sinoatrial node (SN) generates the heart rate (HR). Its spontaneous activity is regulated by a complex interplay between the modulation by the autonomic nervous system (ANS) and intrinsic factors including ion channels in SN cells. However, the systemic and intrinsic regulatory mechanisms are still poorly understood. This study aimed to elucidate the sex-specific differences in heart morphology and SN function, particularly focusing on basal HR, expression and function of hyperpolarization-activated HCN4 and HCN1 channels and mRNA abundance of ion channels and mRNA abundance of ion channels contributing to diastolic depolarization (DD) and spontaneous action potentials (APs).MethodsBody weight, heart weight and tibia length of 2- to 3-month-old male and female mice were measured. Conscious in-vivo HR of male and female mice was recorded via electrocardiography (ECG). Unconscious ex-vivo HR, stroke volume (SV) and ejection fraction (EF) were recorded via echocardiography. Ex-vivo HR was measured via Langendorff apparatus. Volume of atria, ventricles and whole hearts were measured from the ex-vivo hearts by microcomputed tomography (micro-CT). Immunohistochemistry targeting HCN4 and HCN1 was conducted in the SN and RA tissues from both male and female hearts. The funny current (If) of SN cells in 1 nM and following wash-on of 1 μM isoproterenol (ISO) were recorded via whole cell patch clamp. The APs of SN tissue were recorded via sharp microelectrode and optical mapping of membrane voltage. The relative abundance of mRNAs was measured in male and female mice by qPCR.ResultsHeart weight to tibia length ratio and heart volume of females were significantly smaller than males. Unconscious in-vivo HR in male mice was higher than that in females. Conscious in-vivo HR, ex-vivo HR, SV, and EF showed no notable difference between male and female mice. Immunohistochemistry revealed HCN4, HCN1, and the sum of HCN4 and HCN1, expression in the SN was notably elevated compared with the RA in both male and females, but there was no sex difference in these channels expression. There were also no significant sex differences in the V0.5 of If in SN cells in the presence of 1 nM ISO, however wash-on 1 μM ISO in the same cells induced a significantly increased shift of V0.5 to more positive voltages in males than in females. The expression of mRNA coding for adrenergic receptor beta-1 (Adrb1) and cholinergic receptors muscarinic 2 (chrm2) in male mice was higher compared with that in female mice. Early diastolic depolarization (EDD) rate in APs from peripheral SN (pSN) from male mice were higher than these in female mice. Mice of both sexes showed equivalent frequency of SN APs and spatial localization of the leading site in control, and similar significant response to ISO 100 nM superfusion.ConclusionMales display faster in-vivo HR, but not ex-vivo HR, than females associated with increased expression of Adrb1 in male versus female. This suggests a possible difference in the β-adrenergic modulation in males and females, possibly related to the greater ISO response of If observed in cells from males. The role of hormonal influences or differential expression of other ion channels may explain these sex-specific variations in HR dynamics. Further investigations are necessary to pinpoint the precise molecular substrates responsible for these differences.

Research purpose: The sinoatrial node (SN) is the main pacemaker site, and it is located in the junctional area of the superior vena cava within the right atrium (RA). The precise micro-anatomy of the SN in males and females in ageing and obesity remains unclear. Basic procedures: Human SN/RA specimens were dissected from 25 post-mortem hearts (preserved in 4 % formaldehyde solution), under Polish local ethical rules. The SN/RA tissue blocks were embedded in paraffin. Masson's Trichrome staining and immunohistochemistry for CD31 (a marker of endothelial cells) and CD68 (a marker of macrophages) were performed. Images at different magnifications were taken and analysed. 12-lead ECGs from 24 patients under Polish local ethical rules were obtained. Heart rate and P wave morphologies from lead II, lead III and lead aVF were analysed. Statistical analysis was performed with the unpaired t-test. Principal results: Heart weight to body weight ratio was significantly higher in aged obese males vs. their female counterparts. In the RA samples, there was an increase in connective tissue and decreased myocyte content from aged obese females compared to aged obese males. Aged non-obese males had significantly increased cellular hypertrophy than the aged non-obese females. Both the aged obese and aged non-obese females showed more CD3 but less CD68 expressing cells than males. In the SN samples, CD31 and CD68 expressing cells were higher in both aged non-obese and aged obese males than their female counterparts. Major conclusions: Ageing and/or obesity are more likely to impact these cardiac tissues through increased inflammation, increased immune response and hypertrophy.

Abstract Non-alcoholic steatohepatitis (NASH) is a severe type of the non-alcoholic fatty liver disease (NAFLD). NASH is a growing global health concern due to its increasing morbidity, lack of well-defined biomarkers and lack of clinically effective treatments. Using metabolomic analysis, the most significantly changed active lipid sphingosine d18:1 [So(d18:1)] is selected from NASH patients. So(d18:1) inhibits macrophage HIF-2α as a direct inhibitor and promotes the inflammatory factors secretion. Male macrophage-specific HIF-2α knockout and overexpression mice verified the protective effect of HIF-2α on NASH progression. Importantly, the HIF-2α stabilizer FG-4592 alleviates liver inflammation and fibrosis in NASH, which indicated that macrophage HIF-2α is a potential drug target for NASH treatment. Overall, this study confirms that So(d18:1) promotes NASH and clarifies that So(d18:1) inhibits the transcriptional activity of HIF-2α in liver macrophages by suppressing the interaction of HIF-2α with ARNT, suggesting that macrophage HIF-2α may be a potential target for the treatment of NASH.

Abstract Background Polycystic ovary syndrome (PCOS) is a reproductive endocrine disorder with multiple metabolic abnormalities. Most PCOS patients have concomitant metabolic syndromes such as insulin resistance and obesity, which often lead to the development of type II diabetes and cardiovascular disease with serious consequences. Current treatment of PCOS with symptomatic treatments such as hormone replacement, which has many side effects. Research on its origin and pathogenesis is urgently needed. Although improving the metabolic status of the body can alleviate reproductive function in some patients, there is still a subset of patients with metabolically normal PCOS that lacks therapeutic tools to address ovarian etiology. Methods The effect of IL-22 on PCOS ovarian function was verified in a non-metabolic PCOS mouse model induced by dehydroepiandrosterone (DHEA) and rosiglitazone, as well as granulosa cell -specific STAT3 knockout (Fshr cre+ Stat3 f/f ) mice (10 groups totally and n = 5 per group). Mice were maintained under controlled temperature and lighting conditions with free access to food and water in a specific pathogen-free (SPF) facility. Secondary follicles separated from Fshr cre+ Stat3 f/f mice were cultured in vitro with DHEA to mimic the hyperandrogenic environment in PCOS ovaries (4 groups and n = 7 per group) and then were treated with IL-22 to investigate the specific role of IL-22 on ovarian function. Results We developed a non-metabolic mice model with rosiglitazone superimposed on DHEA. This model has normal metabolic function as evidenced by normal glucose tolerance without insulin resistance and PCOS-like ovarian function as evidenced by irregular estrous cycle, polycystic ovarian morphology (PCOM), abnormalities in sex hormone level. Supplementation with IL-22 improved these ovarian functions in non-metabolic PCOS mice. Application of DHEA in an in vitro follicular culture system to simulate PCOS follicular developmental block and ovulation impairment. Follicles from Fshr cre+ Stat3 f/f did not show improvement in POCS follicle development with the addition of IL-22. In DHEA-induced PCOS mice, selective ablation of STAT3 in granulosa cells significantly reversed the ameliorative effect of IL-22 on ovarian function. Conclusion IL-22 can improve non-metabolic PCOS mice ovarian function. Granulosa cells deficient in STAT3 reverses the role of IL-22 in alleviating ovary dysfunction in non-metabolic PCOS mice.

Abstract Soft bioelectronic devices exhibit motion-adaptive properties for neural interfaces to investigate complex neural circuits. Here, we develop a fabrication approach through the control of metamorphic polymers’ amorphous-crystalline transition to miniaturize and integrate multiple components into hydrogel bioelectronics. We attain an about 80% diameter reduction in chemically cross-linked polyvinyl alcohol hydrogel fibers in a fully hydrated state. This strategy allows regulation of hydrogel properties, including refractive index (1.37-1.40 at 480 nm), light transmission (>96%), stretchability (139-169%), bending stiffness (4.6 ± 1.4 N/m), and elastic modulus (2.8-9.3 MPa). To exploit the applications, we apply step-index hydrogel optical probes in the mouse ventral tegmental area, coupled with fiber photometry recordings and social behavioral assays. Additionally, we fabricate carbon nanotubes-PVA hydrogel microelectrodes by incorporating conductive nanomaterials in hydrogel for spontaneous neural activities recording. We enable simultaneous optogenetic stimulation and electrophysiological recordings of light-triggered neural activities in Channelrhodopsin-2 transgenic mice.

[Objective] In order to improve the quality of urban space, it is necessary to optimize the design of rest facilities in the external space of urban rail transit stations. [Method] The external space of Wushan Station on Guangzhou Metro Line 3 with very high level of public activities is taken as the research area and investigated based on the PSPL (public space-public life) research method. The crowd flow change in the area at different time periods is analyzed through on-site counting. The evaluation and demand of the main users of the space is explored and analyzed through interviews. The route of the crowd flow in the area at peak and off-peak periods is counted and analyzed through behavioral note. The reasons for the spatial and temporal differences in the activities of the crowds and their activity demands are further explored through on-site investigation. Based on the characteristics of the temporal and spatial differences of crowd activities in this area, an optimal design strategy of foldable and modular rest facilities is proposed. [Result & Conclusion] Folding seats can not only meet the basic needs of pedestrians for a short rest, waiting or staying, but also flexibly use the space. When not in use, the seats can be folded back to the original facilities, greatly saving the public space. The modular design uses cuboid block modules instead of decorative facilities such as flower boxes, because the free combination of modules can make use of the space to guide crowd flow reasonably, which can not only meet the different needs of different people at different time periods, and but also improve the interaction between people and space.

IntroductionThe sinus node (SN) is the main pacemaker site of the heart, located in the upper right atrium at the junction of the superior vena cava and right atrium. The precise morphology of the SN in the human heart remains relatively unclear especially the SN microscopical anatomy in the hearts of aged and obese individuals. In this study, the histology of the SN with surrounding right atrial (RA) muscle was analyzed from young non-obese, aged non-obese, aged obese and young obese individuals. The impacts of aging and obesity on fibrosis, apoptosis and cellular hypertrophy were investigated in the SN and RA. Moreover, the impact of obesity on P wave morphology in ECG was also analyzed to determine the speed and conduction of the impulse generated by the SN.MethodsHuman SN/RA specimens were dissected from 23 post-mortem hearts (preserved in 4% formaldehyde solution), under Polish local ethical rules. The SN/RA tissue blocks were embedded in paraffin and histologically stained with Masson’s Trichrome. High and low-magnification images were taken, and analysis was done for appropriate statistical tests on Prism (GraphPad, USA). 12-lead ECGs from 14 patients under Polish local ethical rules were obtained. The P wave morphologies from lead II, lead III and lead aVF were analyzed.ResultsCompared to the surrounding RA, the SN in all four groups has significantly more connective tissue (P ≤ 0.05) (young non-obese individuals, aged non-obese individuals, aged obese individuals and young obese individuals) and significantly smaller nodal cells (P ≤ 0.05) (young non-obese individuals, aged non-obese individuals, aged obese individuals, young obese individuals). In aging, overall, there was a significant increase in fibrosis, apoptosis, and cellular hypertrophy in the SN (P ≤ 0.05) and RA (P ≤ 0.05). Obesity did not further exacerbate fibrosis but caused a further increase in cellular hypertrophy (SN P ≤ 0.05, RA P ≤ 0.05), especially in young obese individuals. However, there was more infiltrating fat within the SN and RA bundles in obesity. Compared to the young non-obese individuals, the young obese individuals showed decreased P wave amplitude and P wave slope in aVF lead.DiscussionAging and obesity are two risk factors for extensive fibrosis and cellular hypertrophy in SN and RA. Obesity exacerbates the morphological alterations, especially hypertrophy of nodal and atrial myocytes. These morphological alterations might lead to functional alterations and eventually cause cardiovascular diseases, such as SN dysfunction, atrial fibrillation, bradycardia, and heart failure.

The Ebola virus is a deadly pathogen that has been threatening public health for decades. Recent studies have revealed alternative viral invasion routes where Ebola virus approaches cells via interactions among phosphatidylserine (PS), PS binding ligands such as Gas6, and TAM family receptors such as Axl. In this study, we investigate the interactions among phosphatidylserine on the Ebola viral-like particle (VLP) membrane, human Gas6, and human Axl using atomic force microscope-based single molecule force spectroscopy to compare their binding strength and affinity from a biomechanical perspective. The impact of calcium ions on their interactions is also studied and quantified to provide more details on the calcium-dependent phosphatidylserine-Gas6 binding mechanism. Our results indicate that, in the presence of calcium ions, the binding strengths of VLP-Gas6 and VLP-Gas6-Axl increase but are still weaker than that of Gas6-Axl, and the binding affinity of VLP-Gas6 and VLP-Gas6-Axl is largely improved. The binding strength and affinity of Gas6-Axl basically remain the same, indicating no impact in the presence of calcium ions. Together, our study suggests that, under physiological conditions with calcium present, the Ebola virus can utilize its membrane phosphatidylserine to dock on cell surface via Gas6-Axl bound complex.

Abstract The ICT service industry has become a burgeoning industry at a high and stable speed. Their equitable distribution can improve national and global positive peace. This paper aimed to verify the characteristics of spatio-temporal evolution and its influencing factors in the ICT service industry. Based on the data from 31 Provinces in China from 2015 to 2019, this paper uses location quotient, spatial autocorrelation methods and spatial econometric analysis to explore the development characteristics, evolution and influencing factors of the ICT service industry, respectively. The main results are shown as follows: (1) China's ICT service industry is mainly concentrated in Beijing, Shanghai, Zhejiang, Tibet, and Guangdong, with a trend of specialisation development. They are not only distributed in cities with relatively superior overall development but also those with superior industrial and development carrier elements. Technological relevance, aggregation, and political difference might have an impact on promoting the emergence and development of these industries. (2) ICT service industry is characterised by stable and highly concentrated development. Numbers between three to five significant provinces and types with high-high (HH) and high-low (HL) clusters of local spatio-temporal association kept stable in the period. The HH was in eastern coastal areas, including Zhejiang, Shanghai, Jiangsu, and Shandong, and the HL was in Guangdong in 2015. There is a definite spatial correlation in spatial distribution with constant strengthening. (3) TUR, NDN, MIAT and the area were shown to have a significant role in promoting the ICT service industry, while NW, GDP and ICT Employment were shown to have a significant negative impact on this industry. Correspondingly, two strategies were put forward here: (1) accelerating the inter-provincial networking development of the ICT service industry, and (2) strengthening government policy guidance for the ICT service industry. These outcomes can not only provide a scientific basis and theoretical support for the distribution of strategies and resources for these industries at the theoretical level but also improve resource integration from the national perspective and the efficiency of resource use at the practical level.

In the honey bee, the queen’s death severely threatens the survival of the colony. In an emergency, new queens are reared from young worker larvae, where nepotism is thought to influence the choice of queen candidates by the workers. This article simulates the emergency queen-rearing process in a colony under natural conditions and records the results of colony selection (without nepotism). In queenless colonies, worker larvae aged three days or younger were preferred for queen rearing, and 1-day-old larvae were the first to be selected for the queen-cell cups. In the capping stage, the number of capped queen cells selected from the 1-day-old larvae was much higher than the 3-day-old larvae. On the first day, the number of emerging queens reared from 1-day-old larvae was significantly higher than the queens reared from 2-day-old and 3-day-old larvae. However, there was no significant difference in the birth weights of queens reared from 1-day-old, 2-day-old, or 3-day-old larvae. When the newly emerged queens were introduced into the original queenless colony, 1-day-old larval queens triggered more worker followers than 2-day-old larval queens. The expression of ovarian development-related genes (vg, hex110, and Jh) was higher in queens reared from 1-day-old larvae than those reared from 2-day-old and 3-day-old larvae, indicating that the quality of the queens reared from 1-day-old larvae is superior. This study shows that in the absence of nepotism, the colony selection of queen candidates at the larval stage, capping stage, and emerging stage is not final, but is gradually optimized to maximize colony development through a “quality control” process.

IntroductionRespiratory diseases such as chronic obstructive pulmonary disease, obstructive sleep apnea syndrome, and COVID-19 may cause a decrease in arterial oxygen saturation (SaO2). The continuous monitoring of oxygen levels may be beneficial for the early detection of hypoxemia and timely intervention. Wearable non-invasive pulse oximetry devices measuring peripheral oxygen saturation (SpO2) have been garnering increasing popularity. However, there is still a strong need for extended and robust clinical validation of such devices, especially to address topical concerns about disparities in performances across racial groups. This prospective clinical validation aimed to assess the accuracy of the reflective pulse oximeter function of the EmbracePlus wristband during a controlled hypoxia study in accordance with the ISO 80601-2-61:2017 standard and the Food & Drug Administration (FDA) guidance.MethodsHealthy adult participants were recruited in a controlled desaturation protocol to reproduce mild, moderate, and severe hypoxic conditions with SaO2 ranging from 100% to 70% (ClinicalTrials.gov registration #NCT04964609). The SpO2 level was estimated with an EmbracePlus device placed on the participant's wrist and the reference SaO2 was obtained from blood samples analyzed with a multiwavelength co-oximeter.ResultsThe controlled hypoxia study yielded 373 conclusive measurements on 15 subjects, including 30% of participants with dark skin pigmentation (V–VI on the Fitzpatrick scale). The accuracy root mean square (Arms) error was found to be 2.4%, within the 3.5% limit recommended by the FDA. With the development of inventory planning and the contradiction between land supply and demand, urban renewal development has been gradually replacing reconstruction in China's community redevelopment projects. Such projects need multiple stakeholders' engagement. However, China's patterns of public engagement with top-down governance are different from those in developed countries with bottom-up initiatives. This fact also indicates that such developed patterns are not suitable for the context of China. Meanwhile, research on micro rehabilitation is relatively new and requires further analytical work on development pattern analysis. Therefore, the protagonist status of different stakeholders and allowing them to participate in redevelopment projects are social issues that need to be solved urgently. This paper explores patterns of public engagement in community micro-rehabilitation projects in China. Eleven communities in Guangzhou are taken as cases through participatory observation, document analysis, and interviews. This paper analyses opinions and comments from different stakeholders and summarises their information delivery paths. The findings indicate four present patterns: single-threaded, representative feedback, property involvement, and external party service patterns. Through comparative analysis, the study highlights that the participating stakeholders in the four patterns involved the projects to different degrees. However, they are all still in the ''Tokenism'' degree, which is in the middle category of the ladder of engagement. Furthermore, an appropriate and sustainable pattern is put forward to provide a reference and research basis for improving public engagement in community micro-rehabilitation projects in China.

Cities face considerable fundamental sustainability challenges, and scholars have developed many sustainability assessment tools (SATs) to assess and address these problems. As an important pillar of the tools, the institutional dimension, though added to one of the main dimensions, needs to be stressed more in the existing studies, especially in the field of political geography. Territory, as one of the core concepts of political geography, is considered an essential practical perspective of institutional issues. This paper aims to clarify the limitations of the institutional dimension in SATs from the perspective of territory. Nineteen SAT tools are filtered and reviewed after refining the concept of institutional sustainability. Their categories and indicators are divided into four themes in order to clarify the intent of the institutional dimensions. After documentary analysis, we argue that the main shortcomings of the existing research are the need for more balanced integrality and spatial embeddedness of institutional indicators in the tools. The institutional dimension should be first considered before defining other dimensions through more detailed explanations than the existing one and should lay the roots in the specific institutional arrangements. This paper suggests that the preferential consideration for institutional dimension and the appropriate increase of its specific gravity can be considered in future SAT optimization and development. Some sociological approaches, like grounded theory, can be regarded as an introduction to form institutional indicators based on specific institutional arrangements. This study can provide an opportunity to improve the existing sustainability assessment tools or develop new tools to reflect more holistic understandings of institutional sustainability.

Methionine is an essential amino acid, involved in the promotion of growth, immunity, and regulation of energy metabolism. Over the decades, research has long focused on the beneficial effects of methionine supplementation, while data on positive effects of methionine restriction (MR) were first published in 1993. MR is a low-methionine dietary intervention that has been reported to ameliorate aging and aging-related health concomitants and diseases, such as obesity, type 2 diabetes, and cognitive disorders. In addition, MR seems to be an approach to prolong lifespan which has been validated extensively in various animal models, such as Caenorhabditis elegans, Drosophila, yeast, and murine models. MR appears to be associated with a reduction in oxidative stress via so far mainly undiscovered mechanisms, and these changes in redox status appear to be one of the underlying mechanisms for lifespan extension and beneficial health effects. In the present review, the association of methionine metabolism pathways with redox homeostasis is described. In addition, the effects of MR on lifespan, age-related implications, comorbidities, and diseases are discussed.

Sea fog poses a considerable challenge to port operations, impacting maritime safety and efficiency. During the past five years, the average annual downtime of the navigation dispatch department in Ningbo Zhoushan Port due to weather was 800–1000 h, of which approximately 300 h can be attributed to sea fog. This study addresses the issue by developing a comprehensive sea fog monitoring system for Ningbo Zhoushan Port. The system utilizes automatic weather stations (AWS) and visibility laser imaging, detection, and ranging (LIDAR) to assess sea fog severity and improve monitoring accuracy. By increasing monitoring frequency and adopting corresponding warning measures, the system aims to enhance maritime safety and efficiency in Ningbo Zhoushan Port. The results showed that the implemented system successfully determines sea fog severity, enables real-time monitoring, and provides precise visibility assessments. Joint assessments revealed a substantial increase in the annual operating time and revenue of the port. These findings underscore the importance of advanced monitoring techniques in optimizing port operations, reducing collision risks, and mitigating economic losses caused by sea fog.

Designing child-friendly streetscapes is a pragmatic and effective approach to addressing the limited outdoor play spaces and social exclusion experienced by migrant children living in vulnerable residential areas. However, the existing research and guidelines on streetscape design lack specificity for migrant workers’ communities and fail to provide full-cycle design process guidance for real-world practices. By taking a social–ecological perspective and synthesizing the existing literature, this paper develops an integrated design framework with seven indicators to guide three stages of child-friendly streetscape regeneration in migrant workers’ communities. The effectiveness of this framework was validated by application in a case project in a typical migrant workers’ community in Ningbo, China. The results show that the social–ecological design framework can help maximize opportunities for various children’s activities on the street with limited spatial resources. It also contributes to creating inclusive streetscapes to promote social cohesion by integrating social and cultural elements, children’s participation, and place-making activities. However, institutional barriers, cultural norms, and limited resources impede children’s participation and sustainable operations, which calls for more attention to be paid to “software” building. The new framework and research findings serve as a valuable guide and reference for practitioners in the field of child-friendly environmental design, especially in the context of marginalized communities in developing countries.

Research purpose: The cardiac conduction system (CCS) regulates electrical impulses across the heart and cardiac arrhythmias cause structural remodelling of the CCS. Since its discovery over a century ago, the precise anatomy and differences between the human CCS in healthy, aged, and obese human hearts has remained relatively unknown. Using iodine potassium-iodide (I2KI) and graphene oxide (GO) as contrast agents for high-resolution micro-computed tomography (micro-CT), we explored and identified the anatomies of whole healthy, aged, and obese hearts, including their CCS. Basic procedures: Human specimens were obtained from 5 post-mortem hearts, under local ethical rules and stored in the Dobrzynski laboratory, under the Human Tissue Act 2004. Specimens were stained with I2KI or GO contrast agents before scanning with micro-CT. Data obtained from micro-CT was uploaded onto Amira v6.5 software for analysis, 3-dimensional reconstructions, and segmentation of relevant structures. Following micro-CT analyses, tissue blocks were cryosectioned and stained for histological assessments. Main findings: There are obvious anatomical structural differences between the healthy, aged and obese hearts. Compared to the healthy heart, the aged heart and obese heart had larger chambers; thicker myocardial walls; thicker blood vessels; more extensive nodal regions and connective tissue; more epicardial fat; and fewer Purkinje fibres.Our use of I2KI and GO as contrast agents for high-resolution micro-CT scanning contribute to - and expands - the current understanding of CCS structural variations between healthy, aged and obese human hearts.These current and novel techniques can have key impacts on our anatomical understandings for current treatments for cardiovascular disease and the development of mathematical models of aged and diseased hearts. Thus ultimately aiding in the reduction of cardiac morbidities and reduction of patient death rate.

A submetric spatial resolution Raman optical time-domain reflectometry (ROTDR) temperature sensor assisted by the Wiener deconvolution postprocessing algorithm has been proposed and experimentally demonstrated. Without modifying the typical configuration of the ROTDR sensor and the adopted pump pulse width, the Wiener demodulation algorithm is able to recover temperature perturbations of a smaller spatial scale by deconvoluting the acquired Stokes and anti-Stokes signals. Numerical simulations have been conducted to analyze the spatial resolution achieved by the algorithm. Assisted by the algorithm, a typical ROTDR sensor adopting pump pulses of 20 ns width can realize the distributed temperature sensing with a spatial resolution of 0.5 m and temperature accuracy of 1.99 °C over a 2.1-km sensing fiber.

Intracerebral hemorrhage (ICH) is the most lethal subtype of stroke, but effective treatments are lacking, and neuroinflammation plays a key role in the pathogenesis. In the innate immune response to cerebral hemorrhage, microglia first appear around the injured tissue and are involved in the inflammatory cascade response. Microglia respond to acute brain injury by being activated and polarized to either a typical M1-like (pro-inflammatory) or an alternative M2-like (anti-inflammatory) phenotype. These two polarization states produce pro-inflammatory or anti-inflammatory. With the discovery of the molecular mechanisms and key signaling molecules related to the polarization of microglia in the brain, some targets that regulate the polarization of microglia to reduce the inflammatory response are considered a treatment for secondary brain tissue after ICH damage effective strategies. Therefore, how to promote the polarization of microglia to the M2 phenotype after ICH has become the focus of attention in recent years. This article reviews the mechanism of action of microglia’s M1 and M2 phenotypes in secondary brain injury after ICH. Moreover, it discusses compounds and natural pharmaceutical ingredients that can polarize the M1 to the M2 phenotype.

Polyphenols from peanut skin have been reported to possess many beneficial functions for human health, including anti-oxidative, antibacterial, anticancer, and other activities. To date, however, its anti-inflammatory effect and the underlying mechanism remain unclear. In this study, the anti-inflammatory effect of peanut skin procyanidins extract (PSPE) and peanut skin procyanidins (PSPc) were investigated by a dextran sodium sulfate (DSS)-induced colitis mouse model. The results showed that both PSPE and PSPc supplementation reversed the DSS-induced body weight loss and reduced disease activity index (DAI) values, accompanied by enhanced goblet cell numbers and tight junction protein claudin-1 expression in the colon. PSPE and PSPc treatment also suppressed the inflammatory responses and oxidative stress in the colon by down-regulating IL-1β, TNF-α, and MDA expressions. Meanwhile, PSPE and PSPc significantly altered the gut microbiota composition by increasing the relative abundance of Clostridium XlVb and Anaerotruncus, and inhibiting the relative abundance of Alistipes at the genus level. PSPE and PSPc also significantly elevated the production of short-chain fatty acids (SCFAs) in mice with colitis. The correlation analysis suggested that the protective effects of PSPE and PSPc on colitis might be related to the alteration of gut microbiota composition and the formation of SCFAs. In conclusion, the current research indicates that supplementation of PSPE and PSPc could be a promising nutritional strategy for colitis prevention and treatment.

Urban regeneration has become one of the most effective ways to develop urban areas that have declined. Compared with other types of urban regeneration, community micro-regeneration is characterised by scattered stakeholders. Existing studies on public participation in community micro-regeneration mainly focus on revealing the interaction between different stakeholders with less attention to the main users’ social profiles in their participation process. This paper explores residents’ social profiles influencing their participation in community micro-regeneration projects in China. An evaluation framework for residents’ participation in community micro-regeneration projects is designed through literature research, the Delphi technique, and an analytic hierarchy process (AHP) based on the case study of Yongtai community, Guangzhou. Relative residents’ social profiles with the data from the questionnaire and literature research are further verified by stepwise linear regression. The results show that residents’ participation is still in the stage of tokenism with high-level passive information receipt but low-level enthusiasm for interactive activities. The older (i.e., 65-years of age and above) and middle-income groups (i.e., between 3000 and 14,999 CNY/Month) are positively associated with their participation in the Yongtai community micro-regeneration project. These findings can provide references for managers to distribute social resources.

Understanding the functions of protein glycosylation critically depends on methods to efficiently enrich glycoproteins from complex samples. Here, the authors develop a strategy using dendrimer-conjugated benzoboroxole to enhance glycopeptide enrichment, providing the basis for more comprehensive glycoprotein analyses.

The sinus node (SN) is the heart’s primary pacemaker and has a unique expression of pacemaking ion channels and immune cell markers. The role of microribonucleic acids (miRNAs) in control of ion channels and immune function of the sinus node is not well understood. We have recently shown that hsa-miR-486-3p downregulates the main pacemaking channel HCN4 in the SN. In addition, we recently demonstrated that immune cells are significantly more abundant in the SN compared to the right atrium. The aim of this study was to validate the previously predicted interactions between miRNAs and mRNAs of key Ca2+ ion channels (involved in peacemaking) and mRNA of TPSAB1—(a mast cells marker) using luciferase assay. We now show that miR-486 significantly downregulates Cav1.3, Cav3.1, and TPSAB1-mediated luciferase activity, while miR-938 significantly downregulates only TPSAB1-mediated luciferase activity. This makes miR-486-3p a potential therapeutic target in the treatment of SN dysfunctions.

Polycystic ovary syndrome (PCOS) is one of the most common endocrine diseases among women of reproductive age and is associated with many metabolic manifestations, such as obesity, insulin resistance (IR) and hyperandrogenism. The underlying pathogenesis of these metabolic symptoms has not yet been fully elucidated. With the application of metabolomics techniques, a variety of metabolite changes have been observed in the serum and follicular fluid (FF) of PCOS patients and animal models. Changes in metabolites result from the daily diet and occur during uncommon physiological routines. However, some of these metabolite changes may provide evidence to explain possible mechanisms and new approaches for prevention and therapy. This article reviews the pathogenesis of PCOS metabolic symptoms and the relationship between metabolites and the pathophysiology of PCOS. Furthermore, the potential clinical application of some specific metabolites will be discussed.

Abstract High-throughput technologies have identified significant changes in patterns of mRNA expression over cancer development but the functional significance of these changes often rests upon the assumption that observed changes in levels of mRNA accurately reflect changes in levels of their encoded proteins. We systematically compared the expression of 4436 genes on the RNA and protein levels between discrete tumor samples collected from the ovary and from the omentum of the same OC patient. The overall correlation between global changes in levels of mRNA and their encoding proteins is low (r = 0.38). The majority of differences are on the protein level with no corresponding change on the mRNA level. Indirect and direct evidence indicates that a significant fraction of the differences may be mediated by microRNAs.

Thiols are important molecules in the environment and in biological processes. Cysteine (Cys), homocysteine (Hcy), glutathione (GSH) and hydrogen sulfide (H2S) play critical roles in a variety of physiological and pathological processes. The selective detection of thiols using reaction-based probes and sensors is very important in basic research and in disease diagnosis. This review focuses on the design of fluorescent and colorimetric probes and sensors for thiol detection. Thiol detection methods include probes and labeling agents based on nucleophilic addition and substitution, Michael addition, disulfide bond or Se-N bond cleavage, metal-sulfur interactions and more. Probes for H2S are based on nucleophilic cyclization, reduction and metal sulfide formation. Thiol probe and chemosensor design strategies and mechanism of action are discussed in this review.

The grassland areas in eastern Inner Mongolia and northern Xinjiang are rich in coal and groundwater resources. The mining fissures formed by high-intensity coal mining are easy to lead to the strong water-rich aquifer, and it is very easy to cause the destruction of the groundwater system in the overlying strata, the accident of water inrush and sand inrush is easy to happen, especially the height of coal caving is difficult to control in fully mechanized top coal caving mining under aquifer. In this paper, the system protection of overlying aquifer in extra-thick coal seam is taken as the research object, the protection of aquifer and the safety control of water-sand disaster are the research objectives, and the key technology of intelligent caving for overlying aquifer protection in extra-thick coal seam is developed, based on the technology of preventing and controlling water inrush and sand inrush in coal mine, the fundamental way of protecting the groundwater system above the extra-thick coal seam is obtained. An intelligent top-coal caving technology is developed and applied, including top-coal movement tracking system, multi-round memory caving method and system. Based on the precise exploration of hydrogeological conditions, the actual measurement of overburden failure height and the determination of top coal caving height, the intelligent top coal caving technology of multi-round coal caving is applied to Control the height of mining fracture zone. Taking the applied working face as an example, the hydrogeological fine exploration and drilling verification, the acquisition of overburden failure height and the intelligent top-coal caving technology are applied, the safety and precise control of water-sand disaster in overlying aquifer is realized in the process of mining, and the overlying strong water-rich aquifer is effectively protected.
