Ischemia leading to heart attacks and strokes is the major cause of deaths in the world. Whether an occlusion occurs or not, depends on the ability of a growing thrombus to resist forces exerted on its structure. This manuscript provides the first known in vivo measurement of the stresses that clots can withstand, before yielding to the surrounding blood flow. Namely, Lattice-Boltzmann Method flow simulations are performed based on 3D clot geometries. The latter are estimated from intravital microscopy images of laser-induced injuries in cremaster microvasculature of live mice. In addition to reporting the blood clot yield stresses, we also show that the thrombus 'core' does not experience significant deformation, while its 'shell' does. This indicates that the latter is more prone to embolization. Hence, drugs should be designed to target the shell selectively, while leaving the core intact (to minimize excessive bleeding). Finally, we laid down a foundation for a nondimensionalization procedure, which unraveled a relationship between clot mechanics and biology. Hence, the proposed framework could ultimately lead to a unified theory of thrombogenesis, capable of explaining all clotting events. Thus, the findings presented herein will be beneficial to the understanding and treatment of heart attacks, strokes and hemophilia.

Flow-induced shear stresses have been found to be a stimulatory factor in pre-osteoblastic cells seeded in 3D porous scaffolds and cultured under continuous flow perfusion. However, due to the complex internal structure of the scaffolds, whole scaffold calculations of the local shear forces are computationally-intensive. Instead, representative volume elements (RVEs), which are obtained by extracting smaller portions of the scaffold, are commonly used in literature without a numerical accuracy standard. Hence, the goal of this study is to examine how closely the whole scaffold simulations are approximated by the two types of boundary conditions used to enable the RVEs: "wall boundary condition" (WBC) and "periodic boundary condition" (PBC). To that end, Lattice-Boltzmann Method fluid dynamics simulations were used to model the surface shear stresses in 3D scaffold reconstructions, obtained from high resolution microcomputed tomography images. It was found that despite the RVEs being sufficiently larger than 6 times the scaffold pore size (which is the only accuracy guideline found in literature), the stresses were still significantly under-predicted by both types of boundary conditions: between 20 and 80% average error, depending on the scaffold's porosity. Moreover, it was found that the error grew with higher porosity. This is likely due to the small pores dominating the flow field, and thereby negating the effects of the unrealistic boundary conditions, when the scaffold porosity is small. Finally, it was found that the PBC was always more accurate and computationally efficient than the WBC. Therefore, it is the recommended type of RVE. Overall, this work provides a previously-unavailable guidance to researchers regarding the best choice of boundary conditions for RVE simulations.

A custom-built mask aligner (CBMA), which fundamentally covers all the key features of a commercial mask aligner, while being low cost, light weight, and having low power consumption and high accuracy is constructed. The CBMA is comprised of a custom high fidelity LED light source, vacuum chuck and mask holder, high-precision translation and rotation stages, and high resolution digital microscopes. The total cost of the system is under $7,500, which is over ten times cheaper than a comparable commercial system. It produces a collimated ultraviolet illumination of 1.8-2.0 mW cm-2 over an area of a standard 4-inch wafer, at the plane of the photoresist exposure; and the alignment accuracy is characterized to be < 3 microns, which is sufficient for most microfluidic applications. Moreover, this manuscript provides detailed descriptions of the procedures needed to fabricate multilayered master molds using our CBMA. Finally, the capabilities of the CBMA are demonstrated by fabricating two and three-layer masters for micro-scale devices, commonly encountered in biomicrofluidics applications. The former is a flow-free chemical gradient generator and the latter is an addressable microfluidic stencil. Scanning electron microscopy is used to confirm that the master molds contain the intended features of different heights.

Quantitative Biology - Cell Behavior - Abstract
Directed fibroblast migration is central to highly proliferative processes in regenerative medicine and developmental biology, such as wound healing and embryogenesis. However, the mechanisms by which single fibroblasts affect each other's directional decisions, while chemotaxing in microscopic tissue pores, are not well understood. Therefore, we explored the effects of two types of relevant social interactions on fibroblast PDGF-BB-induced migration in microfluidic tissue-mimicking mazes: cell sequence and mitosis. Surprisingly, it was found that in both cases, the cells display behavior that is contradictory to the chemoattractant gradient established in the maze. In case of the sequence, the cells do not like to take the same path through the maze as their predecessor, when faced with a bifurcation. To the contrary, they tend to alternate - if a leading cell takes the shorter (steeper gradient) path, the cell following it chooses the longer (weaker gradient) path, and vice versa. Additionally, we found that when a mother cell divides, its two daughters go in opposite directions (even if it means migrating against the chemoattractant gradient and overcoming on-going cell traffic). Therefore, it is apparent that fibroblasts modify each other's directional decisions in a manner that is counter-intuitive to what is expected from classical chemotaxis theory. Consequently, accounting for these effects could lead to a better understanding of tissue generation in vivo, and result in more advanced engineered tissue products in vitro., Comment: see last page for supplemental materials

Lignans constitute a large group of phenolic plant secondary metabolites possessing high bioactivity. Their accurate determination in plant extracts with a complex chemical composition is challenging and requires advanced separation techniques. In the present study, a new approach to the determination of lignans in coniferous knotwood extracts as the promising industrial-scale source of such compounds based on comprehensive two-dimensional liquid chromatography separation and UV spectrophotometric detection is proposed. First and second-dimension column screening showed that the best results can be obtained using a combination of non-polar and polar hydroxy group embedded octadecyl stationary phases with moderate (~40%) "orthogonality". The optimization of LC × LC separation conditions allowed for the development of a new method for the quantification of the five lignans (secoisolariciresinol, matairesinol, pinoresinol, 7-hydroxymatairesinol, and nortrachelogenin) in knotwood extracts with limits of quantification in the range of 0.27–0.95 mg L−1 and a linear concentration range covering at least two orders of magnitude. Testing the developed method on coniferous (larch, fir, spruce, and pine) knotwood extracts demonstrated the high selectivity of the analysis and the advantages of LC × LC in the separation and accurate quantification of the compounds co-eluting in one-dimensional HPLC. - Abstract
The energy spectra of galactic cosmic rays carry fundamental information regarding their origin and propagation, but, near Earth, cosmic rays are significantly affected by the solar magnetic field which changes over time. The time dependence of proton and electron spectra were measured from July 2006 to December 2009 by PAMELA experiment, that is a ballooon-borne experiment collecting data since 15 June 2006. These studies allowed to obtain a more complete description of the cosmic radiation, providing fundamental information about the transport and modulation of cosmic rays inside the heliosphere. In this talk the study of the time dependence of the cosmic-ray protons and helium nuclei from the unusual 23rd solar minimum through the following period of solar maximum activity is presented., QC 20181221

The present research assesses the competitive status of PJSC “Gazprom” following the results of the first half of 2015. The authors have shown that a lack of relevance, and the imperfection of applied methods of ranking make it impossible use the existing ratings to assess the current competitiveness of economic entities. Therefore, a conclusion is made that the assessment of the impact of recession trends in the economy on the performance of major domestic companies may be made only by applying specific estimation methods of competitiveness of enterprises. The main methods for assessing the competitiveness of enterprises are analyzed; their strengths and weaknesses are identified. The authors have proved that for rapid assessment of the competitive status of many business entities the dynamic evaluation method of enterprise competitiveness proves most appropriated. Improvements are proposed to the mathematical models of the dynamic approach to the assessment of the competitiveness of enterprises, the mathematical apparatus and algorithm analysis; the theoretical and methodological principles underlying the dynamic approach are described. The methodological and methodical advantages of the dynamic approach are compared to traditional methods of assessing the competitiveness of enterprises. Practical aspects of the assessment and analysis of corporate competitiveness on the basis of a dynamic method are discussed. Calculations for assessing the competitiveness of PJSC “Gazprom” following the results of the first half of 2015 revealed the competitive edge and weaknesses of the company. The analysis of competitiveness is done for the industry. It is shown that unlike traditional ratings, which are usually based on a single indicator (as a rule the revenue or capitalization of the entity) taken over one accounting period, the ranking of companies with the use of the dynamic approach is a more accurate tool for assessing their competitiveness. In conclusion, the authors determine directions of further research on the subject.

- Abstract
- Abstract
A discussion is given of the continential drift theory in its various hypothetical stages. Through the analysis of the existing geological data, it is concluded that two supercontinents, Lavrasia and Gondvana, have existed prior to the middle of the Mesozoic Era. The massifs of the continental crust of Lavrasia consist of old platforms, such as North America, Greenland, Barents Sea, Asia, Russia, Siberia and Kolyma. As a result of the earth's rotation, convectional forces of the earth's crust and cosmogonic causes, the individual massifs become detached from Lavrasian super-continent and began to drift, forming ridges and deep faults. The geological structure of these formations can be associated with the land masses that had drifted away and constitute now either continents, islands, submarine ridges or trenches. - Abstract
The UP-1 digital-analog converter is intended for coordinating the operation of the M-20 digital computer with various analog computers. Substitution of the Dnepr-1 computer for the M-20 involved solution of the following problems: (1) as the Dnepr-1 delivers negative 7-v pulses while the Up-1 requires positive 40-50-v pulses, some circuits and assemblies in the Up-1 were altered; (2) special instructions were worked out for feeding digital information into the UP-1 and for taking it out; a simultaneous exchange of information would require 45 binary digits; only 26 digits were available in the Dnepr-1; 3) definitely space pulses are required for access to the UP-1; hence, such pulses had to be designated from those available in the Dnepr-1. Changes in schematics are shown., Edited machine trans. of Seminar Upravlyayushchie Mashiny i Sistemy. Trudy (USSR) n1 p29-37 1968, by Charles T. Ostertag.

- Abstract
Human ribosomal intergenic spacer (rIGS) contains in its central part two highly homologous 2 kb repeats, LR1 and LR2. In this paper, we investigate heterogeneity of the variable LR2 segment (LR2var) in the human rIGS. More than 500 LR2var copies from ten unrelated human genomes have been cloned and sequenced. Prolonged (G)n (AG)m compound microsatellite clusters with 'n' and 'm' notions fluctuating in random manner span central parts of almost all LR2var variants. Nucleotide sequences flanking the central microsatellite clusters are represented by more than 30 structural groups, with the two major (A and B) and six minor (C-H) ones. The analysis of sequencing data let us propose that the LR2var variability can be derived by various ways, including microsatellite DNA slip-strand mispairing during replication, non-equal crossover and segmental DNA exchange between LR1var and LR2var through the mechanism of gene conversion.
