Broiler chickens selected for rapid growth are highly susceptible to dilated cardiomyopathy (DCM). In order to elucidate the pathophysiology of DCM, the present study examines the fundamental features of pathological remodelling associated with DCM in broiler chickens using light microscopy, transmission electron microscopy (TEM), and synchrotron Fourier Transform Infrared (FTIR) micro-spectroscopy. The morphological features and FTIR spectra of the left ventricular myocardium were compared among broiler chickens affected by DCM with clinical signs of heart pump failure, apparently normal fast-growing broiler chickens showing signs of subclinical DCM (high risk of heart failure), slow-growing broiler chickens (low risk of heart failure) and Leghorn chickens (resistant to heart failure, used here as physiological reference). The findings indicate that DCM and heart pump failure in fast-growing broiler chickens are a result of a complex metabolic syndrome involving multiple catabolic pathways. Our data indicate that a good deal of DCM pathophysiology in chickens selected for rapid growth is associated with conformational changes of cardiac proteins, and pathological changes indicative of accumulation of misfolded and aggregated proteins in the affected cardiomyocytes. From TEM image analysis it is evident that the affected cardiomyocytes demonstrate significant difficulty in the disposal of damaged proteins and maintenance of proteostasis, which leads to pathological remodelling of the heart and contractile dysfunction. It appears that the underlying causes of accumulation of damaged proteins are associated with dysregulated auto phagosome and proteasome systems, which, in susceptible individuals, create a milieu conducive for the development of DCM and heart failure. RESEARCH HIGHLIGHTS The light and electron microscopy image analyses revealed degenerative changes and protein aggregates in the cardiomyocytes of chickens affected by DCM. The analyses of FTIR spectra of the myocardium revealed that DCM and heart pump failure in broiler chickens are associated with conformational changes of myocardial proteins. The morphological changes in cardiomyocytes and conformational changes in myocardial proteins architecture are integral constituents of pathophysiology of DCM in fast-growing broiler chickens.

Eimeria is an intracellular obligate apicomplexan parasite that parasitizes the intestinal epithelial cells of livestock and poultry, exhibiting strong host and tissue tropism. Parasite–host interactions involve complex networks and vary as the parasites develop in the host. However, understanding the underlying mechanisms remains a challenge. Acknowledging the lack of studies on Eimeria invasion mechanism, we described the possible invasion process through comparative analysis with other apicomplexan parasites and explored the fact that parasite–host interactions serve as a prerequisite for successful recognition, penetration of the intestinal mechanical barrier, and completion of the invasion. Although it is recognized that microbiota can enhance the host immune capacity to resist Eimeria invasion, changes in the microenvironment can, in turn, contribute to Eimeria invasion and may be associated with reduced immune capacity. We also discuss the immune evasion strategies of Eimeria, emphasizing that the host employs sophisticated immune regulatory mechanisms to suppress immune evasion by parasites, thereby sustaining a balanced immune response. This review aims to deepen our understanding of Eimeria–host interactions, providing a theoretical basis for the study of the pathogenicity of Eimeria and the development of novel anticoccidial drugs. Femoral bone degeneration has been recognized as an important cause of lameness in broiler chickens for many years, but the pathogenesis of this condition has not been completely elucidated. The current work presents comprehensive analyses of changes associated with femoral bone degeneration based on findings from gross pathology, histopathology, biochemistry, and synchrotron-based imaging techniques. Gross lesions were predominantly seen in epiphysis and metaphysis of the proximal femur, and infrequently in distal femur, but we did not observe gross lesions in the diaphysis. Bone fractures were observed occasionally, but the most common lesions involved separation of articular cartilage of the femoral bone head, with progressive erosions of the subchondral bone. In advanced cases, on histopathological examination, changes in femoral bone were indicative of chondronecrosis and osteonecrosis. Computed tomography revealed that the degenerative process involves loss of trabecular bone. The course of the lesion development in the mineralized matrix appears to be coupled with increased bone resorption associated with excessive proliferation of pathologically altered osteoclasts. Light microscopy, Fourier transform infrared spectroscopy, and biochemical analysis provided consistent evidence that lowered protein content of the bone organic matrix is an integral component of femoral bone pathology, but these changes do not appear to be associated with excessive activity of matrix metalloproteinases. Taken together, our findings indicate that femoral bone degeneration is associated with structural changes occurring in both inorganic and organic matrix of the bone, but insufficiency in protein metabolism is most probably a primary aetiological factor in the natural history of femoral bone degeneration. However, it is important to stress that our findings do not negate the importance of bacterial infection in the evolution of this condition. Pathogens play a critical role in the progressive pathogenesis of this condition, which ultimately is manifested, in most instances, as femoral head necrosis.

The present work describes specific congenital intestinal accidents found in commercial broiler chickens where the affected individuals have their intestinal loops translocated outside the abdomen. These anomalies have been observed in many embryos that failed to hatch, in newly hatched chicks, as well as in broilers at various stages of growth. It appears that these lesions stem from incomplete closure of the abdominal cavity during the last stages of embryonic development.

Thermal processing of meat products generates cardiotoxic compounds capable of inducing heart failure in both humans and laboratory animals. Such compounds may be present in broiler diets because supplements such as meat meal (MM), which are commonly used in broiler rations, are rendered at high temperature. Our objective was to evaluate whether putative cardiotoxic compounds in MM increase the risk of heart failure in broilers. The treatment and control diets were prepared by mixing the condensed MM extract (equivalent to dietary MM inclusion of 25%) or placebo (condensed extraction medium) with commercial broiler feed, and the respective diets were offered to commercial male broilers randomly allocated to either treatment or control groups. Broilers fed a diet spiked with MM extract showed a higher incidence (P<0.05) of chronic heart failure (65.5%) in comparison with the control group (55.4%). Postmortem examination upon termination of the experiment revealed that, in comparison with control broilers, broilers fed diet containing MM extract showed higher incidence of lesions indicative of subclinical heart disease evidenced grossly by ventricular dilation and pericardial effusions, microscopically by changes characteristic of cardiomyocyte degeneration, and ultrastructurally by changes in contractile elements and in mitochondria. Measurements of cardiac high-energy phosphates revealed that broilers fed the diet containing MM extract had lower (P<0.05) levels of cardiac energy reserve as compared with birds fed control diet. We conclude that cardiotoxic factors that can induce patho-physiological changes in the heart are present in MM.

Early-lactating dairy cows mobilize body protein to provide amino acids that are directed toward gluconeogenesis and milk protein synthesis. Propylene glycol (PG) is a precursor of ruminal propionate, and feeding PG has been reported to improve energy supply by increasing blood glucose. Our hypothesis was that feeding PG could spare body protein by providing an alternative source of carbon for gluconeogenesis. The major objectives of this study were 1) to delineate the effects of pre- and postpartum PG supplementation in transition dairy cows on whole-body nitrogen balance, urinary 3-methylhistidine (3-MH) excretion, body composition, and gene expression profiles for the major protein degradation pathways in skeletal muscle; and 2) to characterize the changes in body protein metabolism during the periparturient period. Sixteen pregnant cows (7 primiparous and 9 multiparous) were paired based on expected calving dates and then randomly assigned within each pair to either a basal diet (control) or basal diet plus 600 mL/d of PG. Diets were fed twice daily for ad libitum intake, and PG was fed in equal amounts as a top dress from d -7 to d 45. All measurements were conducted at 3 time intervals starting at d -14 +/- 5, d 15, and d 38 relative to calving. Propylene glycol had no effect on whole-body N balance, urinary 3-MH excretion, or body composition. However, N balance was lower at d 15 and 38, compared with d -14. Urinary excretion of 3-MH was lower at d -14 than at d 15 and 38. Supplemental PG had no effect on body weight (BW) and all components of empty BW. On average, cows fed both diets mobilized 19 kg of body fat and 14 kg of body protein between d -14 and d 38. Supplemental PG had no effect on mRNA abundance in skeletal muscle for m-calpain, and the 14-kDa ubiquitin-carrier protein E2 (14-kDa E2) and proteasome 26S subunit-ATPase components of the ubiquitin-mediated proteolytic pathway; however, PG supplementation downregulated mRNA expression for mu-calpain at d 15, and tended to downregulate mRNA expression for ubiquitin at d 15 and 38. Relative to calving, mRNA abundance for m- and mu-calpain, ubiquitin, and 14-kDa E2 were greater at d 15 compared with d -14 and d 38. In summary, these results indicate that transitional effects on whole-body metabolism and gene expression for the Ca(2+)-dependent and ubiquitin-mediated proteolytic pathways in skeletal muscle were more pronounced than those elicited by PG supplementation.

The present study examined the possible role of reactive oxygen species in the pathogenesis of heart failure in broilers. Data were collected from three groups of birds at various risk of heart failure: Leghorn chickens (resistant to heart failure), slow-growing feed-restricted broilers (low risk of heart failure), fast-growing ad libitum fed broilers (high risk of heart failure), and broilers with congestive heart failure (CHF). In the first part of the study, basic clinical parameters and ultrastructural changes were examined in the context of lipid peroxidation of the ventricular myocardium. This was followed by the study of in vitro changes in the activity of selected cytosolic enzymes (creatine kinase and lactate dehydrogenase) and mitochondrial enzymes (pyruvate dehydrogenase and alpha-ketoglutarate dehydrogenase) in the presence of oxidants (hydrogen peroxide or tertiary butyl hydroperoxide). The distinctive clinical feature in the fast-growing broilers and in the broilers with CHF as compared with slow-growing broilers or Leghorn chickens was a significantly lower heart rate (P <0.05). Electron microscopy revealed marked morphological changes in myocardial mitochondria in these broilers (i.e. fast-growing broilers and broilers with CHF). The level of malondialdehyde equivalents, an indicator of lipid peroxidation subsequent to generated oxidative stress, was significantly higher (P <0.05) in ad libitum fed broilers and was highest (P <0.01) in broilers with CHF. In vitro, the presence of oxidants had a detrimental effect on creatine kinase and alpha-ketoglutarate dehydrogenase activity, while lactate dehydrogenase activity increased. The activity of pyruvate dehydrogenase was not altered by oxidants. Our results indicate that the deterioration of heart function in fast-growing commercial broilers in our experimental model is associated with oxidative stress leading to lipid peroxidation of cellular and mitochondrial membranes, and decreased activity of myocardial creatine kinase and alpha-ketoglutarate dehydrogenase enzymes critical for energy synthesis and transformation pathways.

Plant-based simple phenolics are natural antimicrobial and antioxidant compounds that may possess biological activity when included in animal diets. Previous work in our lab has shown that sinapic acid (SA) found in canola meal improves nutrient retention and alters gut microbiota when fed at low levels (0 to 0.10%) to broiler chickens. It was of interest to investigate the effect of dietary SA in laying hens that possess a more mature intestinal microbiota. Therefore, 2 experiments were conducted to determine the effects of dietary SA on performance, nutrient digestibility, and egg quality in laying hens. Sinapic acid (Sigma-Aldrich, 98% purity) was included in diets at 0, 0.025, 0.050, and 0.075% in experiment 1, and 0 and 0.5% in experiment 2. Both experiments were conducted for 21 d. Dietary treatment did not affect production traits and gastrointestinal tract volatile fatty acid production in either experiment. Apparent metabolizable energy and protein digestibility were also unaffected by treatment in experiment 1, but in experiment 2, feeding 0.5% SA increased both AME(n) and protein digestibility in comparison to the unsupplemented control. Egg weight, albumen weight, yolk weight, egg specific gravity, shell thickness, and Haugh units were not significantly affected by dietary treatments in either of the experiments. Sinapic acid was not detected in eggs from experiment 1 but in experiment 2, low levels of SA were detected in both the yolk and albumen of eggs from hens fed 0.5% SA. Further research is needed to more completely define the relationship between dietary SA level and nutrient utilization as well as the accumulation of SA in egg components.

Modern strains of fast-growing meat type poultry are highly susceptible to heart failure. Heart-related mortalities are observed predominantly in fast-growing broiler chickens, with ascites and sudden death syndrome being the most common heart-related conditions in modern broiler flocks. This paper examines the role of structural, molecular, and biochemical factors pertinent to the pathophysiology of heart failure in fast-growing broilers. Evidence explaining the pathogenesis of acute and chronic heart failure, in the context of the underlying molecular and biochemical changes in the cardiomyocytes, contractile apparatus, and extracellular matrix in the ventricular myocardium are critically evaluated and discussed with reference to the clinical signs associated with deterioration of heart pump function. The secondary pathophysiological effects on the cardiovascular system, resulting from hemodynamic changes associated with the failing heart pump, are also reviewed and critically discussed.

The search for alternative therapies to antimicrobial growth promoters (AGP) in poultry production has gained momentum in the past years because of consumer preference and government restrictions on the use of AGP in animal production. Flavonoids are plant-derived metabolites that have been studied for their health-promoting properties that could potentially be used as an alternative to AGP in poultry. In a previous study, we showed that the inclusion of a flavonoid-rich corn cultivar (PennHFD1) in the diet improved the health of broilers undergoing necrotic enteritis. However, the mechanisms of action by which the PennHFD1-based diet ameliorated necrotic enteritis are unknown. This study describes the microbial diversity and composition of the jejunum and ileum of chickens co-infected with Eimeria maxima and Clostridium perfringens and treated with a high-flavonoid corn-based diet. Luminal content and mucosal samples from the jejunum and ileum were collected for DNA extraction, 16S rRNA amplicon sequencing and data analyses. The infection model and the dietary treatments significantly changed the alfa diversity indices (Mucosal samples: ASVs, P = 0.04; Luminal content samples: ASVs, P = 0.03), and beta diversities (Mucosal samples: P < 0.01, Luminal content: P < 0.01) of the ileal samples but not those of the jejunal samples. The microbial composition revealed that birds fed the high-flavonoid corn diet had a lower relative abundance of C. perfringens compared to birds fed the commercial corn diet. The treatments also changed the relative abundance of other bacteria that are related to gut health, such as Lactobacillus. We concluded that both the infection model and the dietary high-flavonoid corn changed the broilers' gut microbial diversity and composition. In addition, the decrease in the relative abundance of C. perfringens corroborates with a decrease in mortality and intestinal lesions due to necrotic enteritis. Collecting different segments and sample types provided a broader understanding of the changes in the gut microbiota among treatments. [ABSTRACT FROM AUTHOR]

In ovo injection of camel lactoferrin (cLF36) as an antimicrobial peptide was applied in Ross 308 fertile eggs and tested in 320 post-hatched chickens challenged with Clostridium perfringens (Cp). In 8 treatments and five replicates of 8 birds each, performance, jejunum morphometry and ileal microbial counts of chickens were assayed. Feed intake and feed conversion ratio of the chickens affected by treatments. Together with the positive control group under the Cp (108 cfu/g) challenge and the negative control group under the antibiotic (AB) challenge, the highest villi length was observed. The highest crypt depth was related to the treatment with the Cp challenge and the lowest value was related to the in ovo injection of cLF36 group and combined Cp and AB challenges. The number of Clostridium spp. in the ileal contents increased in the chickens challenged with Cp (P < 0.05). The greatest change was observed in the treatment with injection of cLF36 during the embryonic period and challenge with Cp and the lowest value was related to negative control treatment. In addition, the difference between treatments with cLF36 in ovo injection during the embryonic period and challenge with or without Cp challenge was significantly increased. In the groups under the Cp challenge, the population of E. coli was numerically increased. Based on the obtained results, cLF36, derived from camel milk, could change some of the indices in performance. It caused morphological changes in the villi of ilium and caused a decrease the microbial counts of Clostridium spp., similar to the AB group in the chickens challenged with Cp. Our research attempts to create a new window for in ovo administration of cLF36, according to its beneficial effects in the present study, can be introduced as a candidate for growth-promoting antibiotics. Lectins are capable of altering intestinal morphology by binding to and disrupting the intestinal brush border membrane. They are also known to alter the weight of lymphoid organs. Therefore, we evaluated the effect of soybean lectin (SBL) on intestinal morphology and lymphoid organ weights of poults fed diets containing SBL. Dietary treatments evaluated in this study included a cornstarch and casein-based control (lectin-free) semipurified diet (PD) and semipurified diets containing 0.024 or 0.048% SBL (PDL and PDH, respectively). Experimental diets were fed from hatch to 14 d. Morphological evaluation of the intestine involved measurement of the villi height and perimeter, crypt depth, villus:crypt, and thickness of the muscle layer in the jejunum. Intestinal physical characteristics were also determined by measuring intestinal weight, length, and volume. Results indicated that 0.048% SBL in PDH increased villus:crypt and reduced total intestinal length in turkey poults. In addition, both the 0.024 and 0.048% dietary SBL levels reduced thymus weights. It was concluded that dietary SBL up to 0.048% enhanced intestinal development by increasing villus:crypt, but might alter the structural integrity of lymphoid organs.

The present study examined several basic attributes of first-line defence mechanisms in the skin as potential factors that may explain the susceptibility of broiler chickens to cellulitis. The variables including structural characteristics of the skin, physicochemical properties and cellular responses to the challenge with pathogens were compared between two categories of chickens, a strain of fast-growing commercial broiler chickens (susceptible to cellulitis) and leghorn chickens (resistant to cellulitis). There were substantial differences between leghorns and broilers with regard to physiological characteristics of the skin. Broiler skin was more amenable to injury and the wound-healing process was slow. Compared with leghorns, the lesions resulting from sub-dermal challenge in broilers were more severe and disseminated over a larger area. Mobilization of phagocytic cells (heterophils and macrophages) in leghorns was brisk even in the areas distant from the site of infection, whereas only few heterophils were recruited in the skin of broilers. The functional competence of heterophils in broilers was inferior when compared with leghorns. Based on the present finding, the predisposition of broilers to cellulitis appears to be primarily associated with the inferior first line of defence of their skin. Broilers in commercial situations may be at higher risk to succumb to even minor infection and eventually develop cellulitis because: (1) structural weaknesses of the skin may predispose broilers to skin injury and thus the risk of skin infection by pathogens is increased; (2) broiler skin surface is more likely to provide a conducive environment for colonization of Escherichia coli; (3) in the event of infection, poor recruitment of phagocytic cells to the site of infection may readily lead to widespread colonization of the tissue by pathogens causing cellulitis and (4) poor functional quality of the phagocytic cells that are mobilized compromise the ability of the host to contain the spread of infection.

This work examines the effects of lupine-based diets on the status of the host's riboflavin and zinc. Test diets contained 35% soybean meal (control) or raw (40%) or dehulled (35%) lupine seed meal and were isocaloric (13.4 MJ of AME/kg) and isonitrogenous (23% crude protein). Each diet was offered ad libitum to a group of 16 male commercial broiler chicks for 21 d, starting at 1 d of age. Broilers fed lupine diets had lower feed intakes and growth rates. All sections of the intestinal tract were significantly enlarged (P < 0.01) in all groups fed lupine-based diets in comparison with broilers fed the soybean meal diet, but there were no differences in the morphologies of the mucosa, submucosa, muscularis, or serosa. Increased size of the duodenum, jejunum, and ileum was predominantly attributed to the increase in length rather than intestinal tissue mass. Hence, the enlargement of the intestinal tract was consistent with physiological hyperplasia and not pathological remodeling and hypertrophy. Overall, broilers fed lupine-based diets had more viscous digesta than those fed the soybean meal diet, but the differences were statistically not significant. Blood plasma Zn concentration did not differ between broilers fed lupine-based diets and those fed soybean-meal-based diets, and all broilers fed lupine-based diets had significantly higher (P < 0.001) riboflavin concentrations. In this context, it is apparent that the bioavailability of these micronutrients from lupine diets is not compromised. Intestinal tissue hyperplasia may be interpreted as physiological adaptation to increase absorptive capacity and thus maximize absorption of essential nutrients in the face of antinutritional factors in the diet.

This study reports novel insight into the aetiology of pulmonary hypertension and ascites in broiler chickens. The scope of measurements was focused on anatomical and functional parameters, and blood flow patterns in leghorns (resistant to ascites), fast-growing broilers (susceptible to ascites), broilers developing ascites, and ascitic broilers evaluated in vivo using echocardiography, and further examined in the context of postmortem findings. Both, in vivo observed features and postmortem findings, showed clear differences between broilers and leghorns, and between normal and ascitic broilers. Abnormalities in the heart chamber geometry and blood flow patterns were detected upon echocardiographic examination in all ascitic broilers. Right and left atrio-ventricular (AV) valve regurgitation were common findings in ascitic broilers and some apparently normal broilers, with left AV valve insufficiency being a predominant feature with respect to degree and frequency of occurrence. Blood flow disturbances were not detected in leghorns. Left ventricular fractional shortening (functional parameter) was considerably reduced (P < 0.01) in ascitic birds (mean: 21.7 +/- 2.0 SE) in comparison with normal broilers (mean: 39.1 +/- 3.6 SE), or leghorns (mean: 43.3 +/- 2.4 SE). The presented findings indicate that pathological and functional changes in the left ventricle and atrium play a significant role in the pathogenesis of ascites in broilers. Severe dilation of the left atrium and pulmonary veins seen on postmortem examination, as well as regurgitant blood flow in the left atrium, demonstrated by Doppler study in ascitic birds, provide evidence that chronically elevated pressure in the left atrium is involved in the aetiology of pulmonary hypertension and ascites in fast-growing broilers.

This study describes the adverse effects of dietary lupines in broiler chickens for which lupine seeds (Lupinus angustifolius) in raw, dehulled, or autoclaved forms were used as a replacement for soybean meal (SBM) in practical diets. Test diets contained 35% SBM (control) or raw (40%), autoclaved (40%), or dehulled (35%) lupine seed meal. All diets were isocaloric (3,230 kcal/kg AME) and isonitrogenous (23% crude protein). Each diet was offered ad libitum to a group of 16 (four replicates with four birds per replicate) day-old male broiler chicks for 21 d. Chemical analysis of lupine seeds showed no detectable levels of mycotoxins, and total alkaloid contents were below 0.01%. Decreased food intake and growth rate were the main signs observed in all birds fed lupine-based diets. These adverse effects were observed during the first week and persisted throughout the trial. Acute signs of toxicity were observed in four chicks fed the diet containing raw lupine seed during the first week of exposure. Initial clinical signs included leg weakness, lack of coordination, and torticollis. In later stages, during Weeks 2 and 3, some birds fed lupine-based diets showed signs of muscle paralysis and skeletal deformity. Postmortem examination did not show gross pathological changes associated with the dietary treatments. Liver microsomal cytochrome P-50 content was higher (P < 0.05) in birds fed the raw lupine-based diet (mean 0.56 pmol/mg protein) in comparison with controls (mean 0.25 pmol/mg protein), which indicated a systemic effect. Based on the present results, it can be stated that high levels of some varieties of sweet lupines in broiler diets may cause significant adverse effects manifested as 1) decreased feed intake and growth rate in most of the birds, and 2) specific signs of acute and chronic toxicity in some individuals.

The present study examines ultrastructural and molecular changes in ventricular myocardium associated with ascites cases in fast-growing broilers raised at low altitude. Extensive ultrastructural lesions were seen in the left and right ventricular myocardium of broilers with fulminant heart failure and ascites. Significant changes included lesions in the myofibril contractile apparatus, altered mitochondria, marked reduction in the myofibril component, and changes in the extracellular matrix (ECM) architecture. No lesions were observed in hearts of slow growing broilers, but mild to moderate changes (predominantly in the left ventriculum) were apparent in the hearts from some clinically normal, fast-growing broilers. SDS-PAGE profiles of washed myofibrils showed several distinctly different bands in preparations from left ventricular myocardium of ascitic birds. Western blot analysis of these samples revealed several fragments of myosin heavy chain, M-protein, and titin. Based on gelatinolytic activity, matrix metalloproteinases (MMP) in the cytosolic fraction of ventricular myocardium homogenates were identified as MMP-2. The relative activity of this enzyme appears to be considerably higher in preparations from broilers, particularly in the preparations from the left ventriculum of fast-growing broilers, in comparison to leghorns or slow growing broilers. The nature and distribution of the changes in the heart indicate that chronic cardiomyopathic process in the left ventricular myocardium occurs during the development of ascites. It is postulated that progressive deterioration of the left heart pump function caused by initial lesions in the left ventricular myocardium is a significant factor in the development of pulmonary hypertension and the pathogenesis of ascites in broilers raised at low altitude.

The present study examines the differences in blood gas parameters, cardiac output, cardiac index, oxygen delivery and tissue oxygen extraction in slow growing chickens (leghorn and feed restricted broilers), fast growing chickens (broilers fed ad libitum) and chickens with fulminant heart failure and ascites. In comparison to leghorns, broiler chickens had lower pO2 and O2 saturation levels in venous blood (P < 0.001). At the age of 35 days, broilers had arterial and venous pO2 significantly lower than 7-day-old broilers (P < 0.05). Overall, blood pO2 and O2 saturation tended to decline, and CO2 content tended to increase with age. Chickens developing ascites had lower blood pO2 and O2 saturation levels, and higher blood CO2 content in comparison to normal chickens (P < 0.05). In comparison to other chickens, ascitic chickens had the lowest pO2 and O2 saturation, and highest CO2 content in both venous and arterial blood (all P < 0.001). Broilers at 35 days of age had higher arterial O2 content than leghorn chicks, and there were only minor differences between normal and ascitic chickens. However, ascitic chickens had the lowest venous O2 content (P < 0.001), but the highest tissue O2 extraction index (P < 0.001). Cardiac index was higher in leghorn chicks than in broilers (P 0.001). Ascitic birds had the lowest cardiac index (P < 0.001). Oxygen delivery was higher in leghorns than in broilers (P < 0.001). Ascitic birds had the lowest oxygen delivery index. The present study has identified significant differences in previously unexamined performance indicators of the cardiovascular system between slow growing chickens, fast growing chickens and chickens with heart failure. Low cardiac index in broiler chickens appears to be the key haemodynamic problem leading to hypoxaemia and ultimately cardiovascular failure in fast growing broilers.

The present work evaluates trends in the incidence of cellulitis during the last decade using Canadian National Poultry Condemnation Records. In 1986, only 0.048% of the total slaughter broilers were condemned as a result of cellulitis lesions. Over the next 10 yr, steady increments in cellulitis condemnations were observed, and between 1986 and 1996, the percentage of cellulitis condemnation increased 11.8-fold. In 1996, more than 2.6 million broilers (0.568% of total slaughter) were condemned due to cellulitis; this constituted 30.1% of total condemnations, making it the number one condemnation category in 1996. In the context of dynamic increase in cellulitis, the problems concerning meat hygiene and possible health risk to the consumer are deliberated.

The results presented here are based on a collection of heart necropsy data obtained from a large population of ascitic and normal broiler chickens raised at low altitude. The hearts were subjected to gross postmortem evaluation. Generally, gross changes in the cardiac muscle predominantly consisted of various degrees of right and left ventricular enlargement and atrial enlargement, and were observed in all hearts from birds that developed ascites and in some apparently normal chickens. These hearts appeared flaccid, had a thin ventricular muscle wall, and lacked the normal tone of healthy heart muscle. Ventricular enlargement (mostly dilation) and AV valve degeneration (endocardiosis) were observed as early as 7 to 10 days of age and throughout the remaining growth period in broilers with or without ascites. The hearts from 264 ascitic and 255 apparently normal commercial broilers were used for quantitative analysis, with emphasis on dilatory changes of the ventriculi and detailed evaluation of the atrio-ventricular (AV) valves. The lesions of the AV apparatus and the degree of dilation of the ventriculi were graded according to a priori established standards, and categorized as mild, moderate or severe. Severe or moderate dilation of both the right and left ventricle were observed more frequently (P < 0.001) in ascitic birds than in normal birds. Of hearts from apparently normal birds, 34 and 61% had grossly normal right and left ventricle, respectively. Overall, 30% of ascitic birds had a grossly normal left ventricle, but some degree of right ventricular dilation was found in all ascitic birds. Prominent endocardiosis was a common feature in a majority of hearts from ascitic birds and in some apparently normal birds. Endocardiosis of the left AV valves was observed considerably more frequently (P < 0.001) than of right AV valves. Most severe lesions were found on left AV valves, and consisted of nodules or coalescing formations associated with annulus, valve cusps, chordae tendineae, and sometimes spread to the mural endocardium in the vicinity of the valve apparatus. Frequently the cusps appeared thickened and rigid, and had rolled edges. Chordae tendineae were frequently thickened, and sometimes ruptured. These severe left AV valve lesions were found most frequently (P < 0.001) in the hearts from ascitic birds (52%) in comparison to apparently normal birds (18%). The right AV valve lesions were mainly in the form of small nodules found around the orifice, and less frequently seen on the muscular leaflets of the right valve. Only in a few instances the nodules were larger, ranging from 1 to 3 mm in diameter. The severity of gross lesions on the left AV valve were indicative of stenosis and/or insufficiency of the valve apparatus. A hypothesis that left atrio-ventricular valve degeneration and left ventricular dilation should be considered as a possible aetiologic association with pulmonary hypertension, right ventricular failure, and ascites in broiler chickens is presented and discussed.

Brain stem auditory-evoked response (BAER) is a noninvasive technique used for detecting neurophysiological abnormalities of the brain stem along the auditory pathway. Brain stem auditory-evoked response recordings were obtained from subcutaneous skin electrodes from two control sheep and 22 other sheep fed high sulfur (S) diets with low or high concentration of thiamine (B1), copper (Cu), and molybdenum (Mo). At least four peaks (I,II,III,IV) of varied amplitude were observed in all animals. Neurophysiological abnormalities due to decreased conductivity and/or excitability of nerve fibers along the auditory pathway were found on the BAER recordings of sheep fed high S diet. Abnormalities of peaks and interpeak latencies within BAER recordings were related to histopathological observations of brain stem lesions. Lesions in the areas of the cochlear nuclei and lateral lemniscus were seen in conjunction with altered BAER components. However, abnormalities in BAER recordings and lesions in the brain stem also occurred in the absence of overt clinical signs. Analysis of interpeak latencies between peaks I and IV revealed significant differences among dietary groups. Sheep given diets low in Cu, Mo, and B1 were affected most. Factorial analysis indicated B1 and interactions among Cu, Mo, and B1 as significant factors influencing interpeak latencies.

Background: The purpose of this study was to explore the defensive effects of Vitamin E (Vit. E) and Urtica dioica (UD) in the mitigation of developmental anomalies induced by thiamethoxam (TMX) in chicken embryos. Methods: For this purpose, a total of 140 fertile eggs were equally divided into seven experimental groups (AG); Groups A and B were kept as negative and sham control, respectively. Group C was exposed to TMX while groups D and E were supplemented with Vit. E and U. dioica, respectively along with TMX. Group F received Vit. E and group G U. dioica, only. The eggs were examined on days 10th and 20th of incubation for the assessment of developmental flaws and musculoskeletal anomalies. Results: The mortality rate was highest (40%) in group C (TMX) followed by groups D and E (20 and 15%), respectively. Developing embryos were exposed to TMX for retarded growth weight and crown-rump length (CRL) were significantly decreased as compared to the control group. The highest survival rate was recorded in negative control group A. The teratogenic defects recorded in this study include growth retardation, decreased crown-rump length, shortened beak, exencephaly, feather scantiness, and limb deformities. Morphometric analysis revealed improved growth by all parameters in Vit. E and U. dioica supplemented groups. Conclusion: It was concluded that developmental defects are due to induced TMX, can be counteracted with Vit. E and U. dioica and have no phytochemicals negative effects. [ABSTRACT FROM AUTHOR]

Objective: The objective was to evaluate the effects of UV-B irradiated vitamin D-enriched yeast supplementation on milk yield, milk composition, vitamin D in milk, milk fatty acids, blood chemistry, and 25(OH)D status in dairy cows. Methods: Six Thai Friesian cows (milk production, 11.2±2.0 kg/d; body weight, 415.0±20.0 kg; and days in milk, 90.0±6.0) were allocated to each treatment in a 3×3 Latin square design, with three treatments and three periods. Each period of the Latin square lasted 49 days consisting of 14 days for diet adaptation and 35 days for sample collection. Dairy cows were randomly assigned to one of three treatments: i) feeding a basal diet without yeast (CON); ii) basal diet + 5 g of live yeast (75 IU/head/d of vitamin D2; LY); and iii) basal diet + 5 g of UV-B irradiated vitamin D enriched yeast (150,000 IU/head/d of vitamin D2; VDY). Feed intake and milk production were recorded daily, milk sample collection occurred on days 14 and 35 of each collection period, and blood plasma was collected on days 0, 7, 14, 21, 28, and 35 of each collection period. Results: The results show that after a trial period of 14 and 35 days, the VDY group had significantly higher vitamin D content in milk than the LY and CON groups (376.41 vs 305.15, 302.14 ng/L and 413.46 vs 306.76, 301.12 ng/L, respectively). At days 7, 14, 21, 28, and 35 of the experiment, cows fed the VDY group had significantly higher 25(OH)D2 status in blood than the CON and LY groups (51.07 vs 47.16, 48.05 ng/mL; 54.96 vs 45.43, 46.91 ng/mL; 56.16 vs 46.87, 47.16 ng/mL; 60.67 vs 44.39, 46.17 ng/mL and 63.91 vs 45.88, 46.88 ng/mL), respectively. Conclusion: In conclusion, UV-B irradiated vitamin D-enriched yeast supplementation could improve vitamin D content in the milk and 25(OH)D status in dairy cows during the lactation period. Background: Excessive reactive oxygen species (ROS) and subsequent mitochondrial dysfunction are pivotal in initiating cardiac hypertrophy. To explore nano-selenium's (SeNP's) preventive potential against this condition, the authors evaluated chemically synthesized chitosan-SeNPs and biosynthesized Bacillus cereus YC-3-SeNPs in an angiotensin II (Ang II)-induced cardiac hypertrophy model. Methods: This investigation encompassed ROS measurement, mitochondrial membrane potential analysis, transmission electron microscopy, gene and protein expression analyses, protein carbonylation assays, serum antioxidant quantification and histological staining. Results: SeNPs effectively countered Ang II-induced cardiac hypertrophy by reducing ROS, restoring mitochondrial and protein kinase 2α (CK2-α) function, activating antioxidant pathways and enhancing serum antioxidant levels. Conclusion: This finding underscores SeNPs' role in attenuating Ang II-induced myocardial hypertrophy both in vitro and in vivo. Enlargement of the heart is called cardiac hypertrophy; this is caused by too many reactive oxygen species, which are compounds that damage the mitochondria of cells. The mitochondria provide energy to cells and their disruption can cause a significantly negative effect on cells and the tissues and organs cells make up. Selenium is a type of metal that must be consumed in small amounts to stay healthy; it has antioxidant effects, meaning it can stop reactive oxygen species and potentially prevent cardiac hypertrophy. Nano-selenium (SeNP), consisting of tiny, spherical particles containing selenium, may be a more effective way of delivering selenium as an antioxidant to prevent cardiac hypertrophy. SeNPs were made synthetically and from a type of bacterium called Bacillus cereus; both SeNPs demonstrated antioxidant effects in heart cells taken from chicken embryos and live chickens. These results suggest that SeNPs could be developed into medication to combat cardiac hypertrophy. Chicken domestication began at least 3,500 years ago for purposes of divination, cockfighting, and food. Prior to industrial scale chicken production, domestication selected larger birds with increased egg production. In the mid-20th century companies began intensive selection with the broiler (meat) industry focusing on improved feed conversion, rapid growth, and breast muscle yield. Here we present proteomic analysis comparing the modern broiler line, Ross 708, with the UIUC legacy line which is not selected for growth traits. Breast muscle proteome analysis identifies cellular processes that have responded to human directed artificial selection. Mass spectrometry was used to identify protein level differences in the breast muscle of 6-day old chicks from Modern and Legacy lines. Our results indicate elevated levels of stress proteins, ribosomal proteins and proteins that participate in the innate immune pathway in the Modern chickens. Furthermore, the comparative analyses indicated expression differences for proteins involved in multiple biochemical pathways. In particular, the Modern line had elevated levels of proteins affecting the pentose phosphate pathway, TCA cycle and fatty acid oxidation while proteins involved in the first phase of glycolysis were reduced compared to the Legacy line. These analyses provide hypotheses linking the morphometric changes driven by human directed selection to biochemical pathways. Following development of polioencephalomalacia in one of 105 cattle in a farm in southeastern Saskatchewan, a study was initiated to monitor thiamine (B(1)) and copper (Cu) status, and to evaluate interactive nutritional factors which may have been responsible for this occurrence. It was evident that a combination of high sulfur (S) and low Cu intake was responsible for the depletion of blood B(1) and plasma Cu. Supplementation with trace minerals alone resulted in a significant (p<0.05) improvement in both B(1) and Cu status of the herd. We recommend that herds exposed to high intakes of S be supplemented with Cu up to 50 mg/kg feed dry matter to alleviate potential deficiencies of B(1) and Cu.

The effect of high dietary sulfur (S) supplementation on blood thiamine (B1) concentration, biochemical indices of liver, muscle and kidney damage and selected plasma electrolytes was studied in six sheep. Three of these sheep received an additional 230 mg thiamine/kg diet (Group 2). After approximately 2.5-3 weeks on this diet, all three sheep in the non-B1-supplemented group (Group 1) showed loss of appetite and developed mild neurological signs: depression, intermittent signs of excitation and head pressing. Increases in blood B1 concentration and plasma creatine kinase (CK) and aspartate aminotransferase (AST) were observed during this time in all affected animals. Clinical signs lasted only for two to five days. Sheep in group 2 were clinically normal throughout the experiment, but all of these animals also had elevated blood B1 concentrations and plasma CK activity at the 3 wk sampling. Plasma magnesium concentrations of group 1 sheep were elevated at the 2.5-3 wk and 6 wk samplings but they declined significantly (p less than 0.05) to low normal levels thereafter. Magnesium concentrations of group 2 sheep were low at the beginning but progressively increased during the course of the experiment. At necropsy, brain lesions suggestive of polioencephalomalacia (PEM) were observed in all sheep but were most marked in group 1. It is speculated that PEM may be caused by a direct toxic effect of S, S metabolites or B1 antimetabolites in the brain rather than by an in vivo B1 deficiency per se.

Chicken vision is sensitive to ultraviolet (UV) light containing the UVA spectrum, while UVB plays a key role in the endogenous production of vitamin D3. However, commercially available light sources are typically deficient in the UV spectrum and thus may not adequately fulfill the lighting requirements of indoor-housed laying hens. We hypothesized that supplementary UVB light may improve egg production and egg quality, and bone health during early lay relative to UVA supplementation or standard control lighting. To investigate the effects of UV light supplementation, an experiment was conducted on 252 ISA Brown hens during 16 to 27 weeks of age. Birds were housed in eighteen pens (14 hens/pen) under three different light treatment groups each with six replications: (i) UVO: standard control lighting with LED white light, (ii) UVA: control lighting plus supplemental daylight with an avian bulb, and (iii) UVA/B: control lighting plus a supplemental full spectrum reptile bulb containing both UVA and UVB wavelengths. Hen-day egg production and egg quality, blood parameters including plasma Ca and P, and serum 25(OH)D3, and hen body weight and external health scoring were measured at different age points; while bone quality was assessed at the end of the experiment at 27 weeks. Data were analyzed in JMP® 16.0 using general linear mixed models with α level set at 0.05. Results showed that UVA and UVA/B supplemented birds reached sexual maturity (50% production) 3 and 1 day earlier, respectively, than control birds. There was a trend for UV lights to increase hen-day egg production (P = 0.06). Among egg quality traits, only eggshell reflectivity and yolk index were affected by UV lights (P = 0.02 and 0.01, respectively); however, most of the egg quality traits changed over age (all P < 0.01). Post-hoc tests showed higher serum 25(OH)D3 in the UVA/B group relative to control hens (P < 0.05); but there was no treatment effect on plasma Ca and P or on bone quality parameters (all P > 0.05). A significant interaction was observed between light treatment and age for the number of comb wounds (P = 0.0004), with the UV supplemented hens showing more comb wounds after 24 weeks. These results demonstrated that supplemental UVA/B light had minimal effects on egg production and egg quality, whereas, UVA/B exposure may increase vitamin D3 synthesis during the early laying period. Background: Even with the technological advances in management, health and genetics applied to poultry farming worldwide, there is still a high rate of carcasses condemnation at slaughterhouses, which result in losses for the poultry production chain. Thus, this work aimed to evaluate the condemnation occurrence index (COI) and adjusted seasonal index (ASI) of poultry (turkey, griller, and heavy chicken) between 2009 and 2019, in a slaughterhouse enable to export in southeastern Brazil. Data were obtained from official spreadsheets from the Brazilian Federal Inspection Service (FIS) and used to calculate the COI, correlation analysis between the main causes of condemnation, and ASI assessments throughout the year. Results: Seven percent (55,594,318) of the poultry carcasses were condemned (partial or total), and the most frequent causes, contamination, and contusion/traumatic injury, amounted to 63.5% of the total condemnation. There was a trend of increasing condemnation throughout the time series evaluated, with COI varying between 45,282–149,809 condemnations per 1,000,000 poultry slaughtered. Considering the ASI, it was identified that for ascitic syndrome, July has a higher index value (1.63) than the months between January–June (P < 0.05). Conclusions: The main causes of condemnation were contamination and contusion/traumatic injury, both technological causes. ASI showed that in July there is a greater carcasses condemnation due to ascitic syndrome than in the months between January and June. The variations observed in the ASIs can provide subsidies for preventive measures and optimization of human and financial resources, generating positive impacts on food safety, productivity, and profitability of the sector. The aim of this study was to evaluate the effects of dietary zinc (Zn) supplementation on performance, immune responses and gastrointestinal tract features of broilers fed wheat-soybean meal diet. In addition, optimised dietary Zn level was estimated based on dose-response data. A total of 500 one-day-old male Ross-308 broiler chicks were randomly assigned to 10 dietary treatments with five replicates and ten birds per replicate in a 5 × 2 factorial arrangement of five graded (+40 mg/kg) dietary Zn levels with or without phytase (500 FTU/kg of diet) supplementation. By increasing dietary Zn concentration, final live body weight (LBW), daily weight gain (WG), feed intake (FI), and European Production Efficiency Factor (EPEF), antibody titres against the sheep red blood cell inoculation, cutaneous basophils hypersensitivity to PHA-P injection, jejunum villus height, and apparent dry matter digestibility (ADMD) increased with a significant linear trend. Phytase supplementation improved WG and ADMD and decreased pancreas and intestine relative weights. The interaction effects of dietary Zn concentration and phytase supplementation on the whole of evaluated traits were not significant. Optimum dietary Zn levels were estimated by the regression models at 70, 82, 98, and 110 mg/kg of diet for optimised EPEF and immunity, LBW and WG, appetites, and ADMD, respectively. It is concluded, in the broiler chickens fed wheat-soybean meal diet, Zn and phytase supplementation can improve growth performance, immune response, digestibility, and intestinal features. The Zn requirement is higher than the NRC (1994) recommendation. The efficacy of dietary Zn was not affected by dietary phytase supplementation. Arginine (Arg), lysine (Lys), and methionine (Met) can be used to support the health status of turkeys. The present study investigated selected performance, gut integrity, and immunological parameters in turkeys reared in optimal or challenge conditions. The experiment lasted for 28 days, and it had a completely randomized 2 × 3 factorial design with two levels of dietary Arg, Lys and Met (high or low) and challenge with Clostridium perfringens (C. perfringens), Escherichia coli lipopolysaccharide (LPS) or no challenge (placebo). Increased dietary levels of Arg, Lys and Met had a beneficial effect on turkey performance and immunological parameters, and it improved selected indicators responsible for maintaining gut integrity in different challenge conditions. Under optimal conditions (with no challenge), high ArgLysMet diets did not compromise bird performance and they improved selected performance parameters in challenged birds. The immune system of turkeys was not excessively stimulated by high ArgLysMet diets, which did not disrupt the redox balance and had no negative effect on gut integrity. High ArgLysMet diets increased the expression levels of selected genes encoding nutrient transporters and tight junction proteins. However, the influence exerted by different dietary inclusion levels of Arg, Lys and Met on gut integrity was largely determined by the stressor (C. perfringens vs. LPS). Further studies are required to investigate the role of Arg, Lys and Met levels in the diet on the immune response, gut function and performance of turkeys in different challenge conditions. No study has demonstrated the use of dietary Cannabis-derived cannabidiol (CBD) to alter the stress response in chickens or examined its effects on meat volatile compounds (VOCs). Here, we subjected chickens to dysbiosis via C. perfringens infection or Escherichia coli lipopolysaccharide (LPS) treatment and investigated the potential link between meat VOCs and cecal bacterial activity and the ameliorative effect of CBD. The cecal bacterial production of short-chain fatty acids (SCFAs) was closely correlated with meat VOCs. CBD supplementation reduced the formation of breast meat spoilage VOCs, including alcohols, trimethylamine and pentanoic acid, in the challenged birds, partly by decreasing cecal putrefactive SCFA production. Meat VOC/cecal SCFA relationships differed according to the challenge, and CBD attenuated the effects of C. perfringens infection better than the effects of LPS challenge on meat VOCs. Transgenerational effects of certain nutrients such as essential fatty acids are gaining increased attention in the field of human medicine and animal sciences as a new tool to improve health and animal performance during perinatal life. Omega-3 (n-3) and omega-6 (n-6) fatty acids are denoted by the position of the first double bond from methyl end of the hydrocarbon chain. Alpha-linolenic acid (18:3 n-3) and linoleic acid (18:2 n-6) are essential n-3 and n-6 fatty acids and cannot be synthesized by the vertebrates including chickens. Alpha-linolenic acid and linoleic acid are the parent fatty acids of long chain (> 20–22C) n-3 and n-6 polyunsaturated fatty acids (PUFA) such as eicosapentaenoic acid (20:5 n-3, EPA), docosapentaenoic acid (22:5 n-3/or 22:5 n-6, DPA), docosahexaenoic acid (22:6 n-3, DHA) and arachidonic acid (20:4 n-6). As components of cell membrane phospholipids, PUFA serves as precursors of eicosanoids, act as ligands for membrane receptors and transcription factors that regulate gene expression and are pivotal for normal chick growth and development. Considering the role of egg lipids as the sole source of essential fatty acids to the hatchling, dietary deficiencies or inadequate in ovo supply may have repercussions in tissue PUFA incorporation, lipid metabolism, chick growth and development during pre and early post-hatch period. This review focus on studies showing how maternal dietary n-3 or n-6 fatty acids can lead to remodeling of long chain n-3 and n-6 PUFA in the hatching egg and progeny chick tissue phospholipid molecular species and its impact on chick growth and PUFA metabolism during early life. Background: Two experiments were conducted to establish an optimal NE challenge model and evaluate the efficacy of stimbiotic (STB) supplementation in necrotic enteritis (NE) challenged broilers. In Exp. 1, a total of 120 Arbor Acres (AA) broilers (45.0 ± 0.21 g) were randomly assigned to 6 treatments in a 3 × 2 factorial arrangement. Vaccine treatments included non-challenge (0), × 10 the recommended dose (× 10) or × 20 the recommended dose (× 20) by the manufacturer. Clostridium perfringens (CP) treatments were non-challenge (No) or 3 mL of 2.2 × 107 CFU CP challenge (Yes). In Exp. 2, a total of 72 AA broilers (40.17 ± 0.27 g) were randomly assigned to 6 treatments in a 3 × 2 factorial arrangement. Dietary treatments included non-additive (CON), 100 mg/kg STB (STB) and 100 mg/kg STB on top of a typical commercial blend including an essential oil, probiotics, and enzyme (CB). Challenge treatments included non-NE challenge (No) and NE challenge (Yes) as established in Exp. 1. Results: In Exp. 1, CP and vaccine challenge decreased (P < 0.05) body weight (BW), body weight gain (BWG) and feed intake (FI), and increased (P < 0.05) the number of broilers with diarrhea and intestinal lesions. The oral administration of × 20 recommended dose of vaccines coupled with 3 mL of 2.2 × 107 CFU CP resulted in (P < 0.01) a significantly increased incidence of wet litter and intestinal lesions. Thus, this treatment was chosen as the challenge model for the successful inducement of NE in Exp. 2. In Exp. 2, the NE challenge negatively affected (P < 0.01) growth performance, ileal morphology, immunoglobulin contents in blood, caecal microbiota in the caecum, footpad dermatitis, intestinal lesion scores, tumour necrosis factor (TNF-α) and endotoxin in the serum compared with the non-NE challenged birds. The supplementation of STB and CB in diets enhanced (P < 0.05) growth performance, intestinal microbiota, and blood profiles by stimulating ileal morphology (VH and VH:CD) and propionate production in the cecum, and there were no differences in measured variables between STB and CB supplemented birds. Conclusion: Overall, these results indicate that STB supplementation was able to reduce the inflammatory response and improve the performance of NE challenged birds, and the supplementation of STB alone was as effective as a typical commercial blend containing a number of other additives. Background: Air pollution has been associated with neurodevelopmental disorders in epidemiological studies. In our studies in mice, developmental exposures to ambient ultrafine particulate (UFP) matter either postnatally or gestationally results in neurotoxic consequences that include brain metal dyshomeostasis, including significant increases in brain Fe. Since Fe is redox active and neurotoxic to brain in excess, this study examined the extent to which postnatal Fe inhalation exposure, might contribute to the observed neurotoxicity of UFPs. Mice were exposed to 1 µg/m3 Fe oxide nanoparticles alone, or in conjunction with sulfur dioxide (Fe (1 µg/m3) + SO2 (SO2 at 1.31 mg/m3, 500 ppb) from postnatal days 4–7 and 10–13 for 4 h/day. Results: Overarching results included the observations that Fe + SO2 produced greater neurotoxicity than did Fe alone, that females appeared to show greater vulnerability to these exposures than did males, and that profiles of effects differed by sex. Consistent with metal dyshomeostasis, both Fe only and Fe + SO2 exposures altered correlations of Fe and of sulfur (S) with other metals in a sex and tissue-specific manner. Specifically, altered metal levels in lung, but particularly in frontal cortex were found, with reductions produced by Fe in females, but increases produced by Fe + SO2 in males. At PND14, marked changes in brain frontal cortex and striatal neurotransmitter systems were observed, particularly in response to combined Fe + SO2 as compared to Fe only, in glutamatergic and dopaminergic functions that were of opposite directions by sex. Changes in markers of trans-sulfuration in frontal cortex likewise differed in females as compared to males. Residual neurotransmitter changes were limited at PND60. Increases in serum glutathione and Il-1a were female-specific effects of combined Fe + SO2. Conclusions: Collectively, these findings suggest a role for the Fe contamination in air pollution in the observed neurotoxicity of ambient UFPs and that such involvement may be different by chemical mixture. Tissue resident immune system cells in the chicken intestine play a significant role in the protection against pathogens. However, very little is known about these cells. The current study was conducted to further characterize chicken intestinal immune system cells. Furthermore, this study aimed to assess the immune modulatory action of a highly virulent Clostridium perfringens , a commonly found chicken intestinal microbe, in comparison with a non-commensal, Lactococcus lactis , on intestine-derived immune system cells. The results demonstrated varying distribution of innate and adaptive immune cells along the avian gut-associated lymphoid tissue (GALT) in the duodenum, jejunum, ileum, and cecal tonsils. In addition, steady-state and tissue-specific presence of CD25+ cells among αβ and γδ T-cell subsets was assessed along the intestine. Ex vivo stimulation with C. perfringens or L. lactis resulted in a significant increase in the frequency of CD25+ T cells (γδ and αβ T cells). In addition, significantly more cell death was observed in ex vivo stimulation with C. perfringens , which was indirectly correlated with a decrease in macrophage activation based on nitric oxide (NO) production with no effect on lymphoid cell responsiveness as per intracellular interferon (IFN)-gamma (γ) staining. Ex vivo stimulation with L. lactis activated γδ T cells and αβ T cells, based on intracellular IFN-γ staining, while it had limited effect on macrophages. However, the ability of γδ and αβ T cells to produce IFN-γ and the ability of macrophages production of NO was rescued in the presence of L. lactis. Background: The goal of this study was to evaluate marker-assisted selection (MAS) in broiler chickens using previously mapped gene regions associated with ascites syndrome incidence. The second-generation MAS products were assessed for impact on ascites phenotype and whether there were associated changes in important production traits. Previously, we used whole genome resequencing (WGR) to fine-map 28 chromosomal regions as associated with ascites phenotype in our experimental ascites broiler line (Relaxed, REL) based on a hypobaric chamber challenge. Genotypes for single nucleotide polymorphisms (SNPs) in mapped regions on chromosomes 2 and 22, were used for MAS in our REL line. After two generations, birds homozygous for the genotypes associated with resistance for both chromosomal regions were established. The MAS F2 generation was then compared to the REL line for ascites susceptibility and 25 production traits. Results: Selection based on SNPs in the carboxypeptidase Q (CPQ, Gga2) and leucine rich repeat transmembrane neuronal 4 (LRRTM4, Gga22) gene regions resulted in a sex- and simulated altitude- dependent reduction of ascites incidence in two F2 cohorts of the MAS line. Comparisons of the F2 MAS and REL lines for production traits when reared at ambient pressure found no significant negative impacts for feed intake (FI), feed conversion ratio (FCR), or deboned part yields for either sex for two F2 cohorts. There were, however, improvements in the MAS for full-trial body weight gain (BWG), FCR, absolute and relative tender weights, and relative drumstick weight. Conclusions: These results validate the mapping of the 28 chromosomal regions and demonstrate that fine mapping by WGR is an effective strategy for addressing a complex trait; it also stands as the first successful SNP-based selection program against a complex disease trait, such as ascites. The MAS line is comparable and, in some instances, superior, in growth performance to the REL control while being more resistant to ascites. The objective of this study was isolating and characterising Clostridium perfringens from chickens in Vietnam and identifying virulence factors involved with enteritis. Five hundred thirty-one faecal and sixty-eight intestinal samples were collected from healthy and diseased chickens for the C. perfringens isolation. The presence of virulence factors was determined by multiplex PCR. The netB gene of the selected isolates was sequenced and checked for its expression by SDS-PAGE. Two hundred seventy-two C. perfringens isolates were collected. All of them were shown to be positive for the cpa gene. The netB gene was detected in 26.56% of the C. perfringens isolates from the healthy chickens, while 43.45% of the isolates from the faeces and 45% of the isolates from the intestinal samples were positive for this gene in the diseased birds. All eight isolates positive to netB from the diseased chickens showed 100% identity in the netB sequence and produced the NetB toxin in vitro, whereas only two out of eight healthy chicken-derived isolates produced this toxin. Nine out of ten chickens experimentally infected with the C. perfringens netB-positive isolate showed typical signs of enteritis. The cpa gene was the most prevalent virulence factor identified in the bacteria C. perfringens, but the netB gene could be a major player responsible for necrotic enteritis progression in chickens. Locomotor problems are among one of the main concerns in the current poultry industry, causing major economic losses and affecting animal welfare. The most common bone anomalies in the femur are dyschondroplasia, femoral head separation (FHS), and bacterial chondronecrosis with osteomyelitis (BCO), also known as femoral head necrosis (FHN). The present study aimed to identify differentially expressed (DE) genes in the articular cartilage (AC) of normal and FHS-affected broilers by RNA-Seq analysis. In the transcriptome analysis, 12,169 genes were expressed in the femur AC. Of those, 107 genes were DE (FDR < 0.05) between normal and affected chickens, of which 9 were downregulated and 98 were upregulated in the affected broilers. In the gene-set enrichment analysis using the DE genes, 79 biological processes (BP) were identified and were grouped into 12 superclusters. The main BP found were involved in the response to biotic stimulus, gas transport, cellular activation, carbohydrate-derived catabolism, multi-organism regulation, immune system, muscle contraction, multi-organism process, cytolysis, leukocytes and cell adhesion. In this study, the first transcriptome analysis of the broilers femur articular cartilage was performed, and a set of candidate genes (AvBD1, AvBD2, ANK1, EPX, ADA, RHAG) that could trigger changes in the broiler´s femoral growth plate was identified. Epithelial damage and loss of barrier integrity occur following intestinal infections in humans and animals. Gut health was evaluated by electron microscopy in an avian model that exposed birds to subclinical necrotic enteritis (NE) and fed them a diet supplemented with the probiotic Bacillus amyloliquefaciens strain H57 (H57). Scanning electron microscopy of ileal mucosa revealed significant villus damage, including focal erosions of epithelial cells and villous atrophy, while transmission electron microscopy demonstrated severe enterocyte damage and loss of cellular integrity in NE-exposed birds. In particular, mitochondria were morphologically altered, appearing irregular in shape or swollen, and containing electron-lucent regions of matrix and damaged cristae. Apical junctional complexes between adjacent enterocytes were significantly shorter, and the adherens junction was saccular, suggesting loss of epithelial integrity in NE birds. Segmented filamentous bacteria attached to villi, which play an important role in intestinal immunity, were more numerous in birds exposed to NE. The results suggest that mitochondrial damage may be an important initiator of NE pathogenesis, while H57 maintains epithelium and improves the integrity of intestinal mucosa. Increased blood lactic acid is likely to be involved in the incidence of sudden death syndrome (SDS) in broiler chicken. Guanidinoacetic acid (GAA) with direct or indirect influence on the cardiovascular system may provide, to some degree, protection against lactic acidemia in birds. A total number of 144 male broiler chicks (Ross 308) were used in a complete randomised block design to investigate the effects of dietary supplementation of GAA at 0.6, 1.2, 1.8, 2.4 and 3 g/kg on the physiological response of broiler chicks to an acute lactic acidosis in an 8-days experiment. Birds receiving 1.8 and 3 g/kg GAA exhibited lesser mortality percentage compared with the control birds and those received dietary GAA at 0.6 and 1.2 g/kg (p <.03). Dietary inclusion of 1.2 g/kg GAA to broiler chicks increased serum concentration of uric acid compared to the control birds and those received 2.4 g/kg (p <.02). The mean serum concentration of creatinine was greater in the birds receiving different GAA levels compared with the control birds (p <.0001). The serum concentration of nitric oxide decreased in the birds receiving 1.2 g/kg GAA by 21.05 and 15.57% compared with the control birds and those received 1.8 g/kg of GAA, respectively (p <.04). A lesser frequency for liver colour score 5 was observed in the birds fed on the diet with 0.6 g/k GAA (p <.0001). It was concluded that dietary administration of GAA deteriorates the adaptive physiological responses to an acute lactic acidosis in broiler chicken possibly through liver and kidney dysfunction. Dietary guanidinoacetic acid decreased mortality but showed an adverse effect on growth performance in chickens exposed to acute lactic acidosis. The adverse effects of guanidinoacetic acid may happen through liver and kidney dysfunction. Valeriana carnosa and V. clarionifolia stand out as principal elements in the indigenous pharmacopeias of Patagonia; however, their phytochemical characterization is unknown. This study constitutes the starting point of a general project that aims to characterize secondary metabolites in these species. The variability of phenolic compounds in root ethanolic extracts was analyzed and compared for thirteen populations of V. carnosa and two of V. clarionifolia from the south of Argentinean Patagonia. Phenolic content was quantified by the Folin-Ciocalteu method and the putative phenolic compound profiles were investigated using HPLC-UV-MS. Antioxidant activity was evaluated through 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assays. Total phenolic content values ranged from 5.6 to 16.6 mg GAE/g in V. carnosa and 7.3 to 9.7 mg GAE/g in V. clarionifolia. Antioxidant evaluation results evidenced that the percentage of neutralized DPPH varied between 26% and 85% in V. carnosa and 39% and 58% in V. clarionifolia. A positive correlation between total phenolic content and antioxidant activity (r = 0.90) was observed. In V. carnosa total phenolic content was not correlated with altitude or latitude (p > 0.05), and chemical variability seems to be associated with genetic variability and/or different growing habitats (microclimatic conditions). However, the presence of some specific phenolic compounds was associated with latitude. In V. carnosa and V. clarionifolia 15 and 10 phenolic compounds were tentatively identified, respectively, and several of these are reported to have beneficial attributes from a phytomedical viewpoint. The aim of the present study was to evaluate the effects of dietary total sulphur amino acids (TSAA) levels and guanidinoacetic acid (GAA) supplementation on performance, carcase yield and energetic molecular metabolites in heart and breast muscle of broiler chickens fed wheat-soy diet. A total of 450, male one-day-old Ross-308 broiler chicks were assigned in a 3 × 3 factorial arrangement of nine treatments, five replicates and 10 birds each. Dietary treatments included three graded levels of supplementary TSAA (+0.4 g/kg per level) resulting in a deficient, adequate and plus level of recommended by strain, with three levels of GAA (0, 0.6 and 1.2 g/kg) and were fed for 35 days. Dietary TSAA levels only enhanced performance during the starter period (p <.05). At adequate TSAA level, weight gain (WG) and feed conversion ratio (FCR) in the grower phase were positively affected by supplementing GAA at 1.2 g/kg (p <.05). The FCR during the overall experimental period was significantly lower when feeding GAA at 0.6 and 1.2 g/kg compared to non-supplemented (p <.05). Heart and breast muscle ATP/AMP ratio and breast muscle ADP and PCr concentration and PCr/CrN ratio in the birds fed 1.2 g/kg GAA diet were significantly higher than non-supplemented (p <.05). It is concluded, effects of dietary GAA supplementation were influenced by dietary TSAA level, this indicates to need for proper dietary sulphur amino acid formulation by GAA supplementation. Dietary GAA supplementation could improve broiler chicken's growth performance via regulating some aspects of energy metabolism. Guanidinoacetic acid plays a critical role in energy homeostasis. Matrix metalloproteinases (MMPs) play an important role in intestinal extracellular matrix homeostasis. An overexpression of MMPs results in tissue destruction and local inflammation and has been associated with multiple inflammatory diseases. These host proteases might also be important in tissue damage caused by infectious agents, such as in intestinal damage in Clostridium perfringens-induced avian necrotic enteritis (NE). The aim of the present study was to elucidate the effect of a C. perfringens infection on the MMP activity in the small intestine of birds with a pre-disposing coccidial infection to obtain a more thorough understanding of the pathogenesis of NE. For this purpose, the gelatinolytic activity present in jejunal tissue of Eimeria infected birds which were challenged with either a pathogenic C. perfringens type G strain or a commensal C. perfringens type A strain was analyzed using substrate zymography. The results show that infection of broilers with Eimeria and different C. perfringens strains, independent of their pathogenicity, decreases the expression of a 40–45 kDa host collagenase in the jejunum, as compared to the expression in Eimeria-infected control birds. It was also shown that the expression of 2 MMPs with molecular weights of approximately 50–60 and 60–70 kDa was significantly lower in necrotic tissue as compared to the activity in macroscopically healthy tissue adjacent to the lesion. These results indicate that host collagenases are not elicited by the C. perfringens infection for permeabilizing the host mucosa to allow penetration of the NetB toxin in Eimeria infected broilers. Background: Intensive selection for growth rate (GR) in broiler chickens carries negative after-effects, such as aberrations in skeletal development and the immune system, heart failure, and deterioration of meat quality. In Poland, fast-growing chicken populations are highly non-uniform in term of growth rate, which is highly unprofitable for poultry producers. Therefore, the identification of genetic markers for boiler GR that could support the selection process is needed. The hypothalamus is strongly associated with growth regulation by inducing important pituitary hormones. Therefore, the present study used this tissue to pinpoint genes involved in chicken growth control. Results: The experiment included male broilers of Ross 308 strain in two developmental stages, after 3rd and 6th week of age, which were maintained in the same housing and feeding conditions. The obtained results show for the overexpression of genes related to orexigenic molecules, such as neuropeptide Y (NPY), aldehyde dehydrogenase 1 family, member A1 (ALDH1A1), galanin (GAL), and pro-melanin concentrating hormone (PMCH) in low GR cockerels. Conclusion: The results reveal strong associations between satiety centre and the growth process. Riboflavin (vitamin B2) is essential for monogastric animals. It is mainly produced by recombinant microorganisms (Candida famata, Bacillus subtilis and Ashbya gossypii). The availability of genetically modified organism (GMO)-free riboflavin, obligatory in European organic agriculture, is a major issue. Besides, requirements for organic livestock might differ from conventional production because other genotypes and feed formulations are used. The effects of a fermentation suspension with a high native content of riboflavin produced with unmodified A. gossypii by fermentation were investigated at graded dosages as an alternative to conventional (GMO-based) riboflavin in slow-growing broilers on performance traits and health and welfare indicators. In 2 runs with 800 animals each, Ranger Gold™ broilers were fed with 4 dietary treatments. For starter diets (day 1 to 18), treatments included a basal diet (1) without any riboflavin supplementation (negative control, N-C), (2) with conventional riboflavin supplementation (Cuxavit B2 80% riboflavin) at 9.6 mg/kg (positive control, P-C), (3) with riboflavin supplementation from the alternative source at 3.5 mg/kg (A-low) and (4) with riboflavin supplementation from the alternative source at 9.6 mg/kg (A-high). For the finisher diet (day 29 until slaughtering), P-C and A-high were supplemented with 8.0 mg/kg and A-low with 3.5 mg/kg. Diets were formulated according to organic regulations. Animals were kept in floor pens with 20 chickens per pen. Weekly, BW, feed and water consumption were recorded. Every second week, animal-based health and welfare indicators (feather score and footpad dermatitis) were scored. Slaughter traits were assessed for five males and females per pen at 62/63 days of age. Final body weight of A-high differed from N-C and A-low, but not from P-C. From week 2 until six years of age, A-high had a higher daily weight gain when compared to all other groups. With 74.4%, dressing percentage was higher in A-high compared with all other groups (73.3%). Breast percentage of A-low was lower than that of both control groups but did not differ from A-high. The highest frequency of liver scores indicating fatty liver syndrome was found in P-C, followed by N-C and A-low. Feather scores did not respond to treatment; the highest frequency of mild footpad dermatitis was observed in A-high, however at a low prevalence. In conclusion, the tested fermentation suspension with a high native content of riboflavin derived from fermentation of A. gossypii can be used at levels of commercial recommendations as alternative to riboflavin produced from GMO in broiler feeding. The brain produces steroids de novo from cholesterol, so‐called "neurosteroids." The Purkinje cell, a cerebellar neuron, was discovered as a major site of the biosynthesis of neurosteroids including sex steroids, such as progesterone, from cholesterol in the brain. Allopregnanolone, a progesterone metabolite, is also synthesized in the cerebellum and acts on the Purkinje cell to prevent cell death of this neuron. Recently, the pineal gland was discovered as an important site of the biosynthesis of neurosteroids. Allopregnanolone, a major pineal neurosteroid, acts on the Purkinje cell for the survival of this neuron by suppressing the expression of caspase‐3, a crucial mediator of apoptosis. Background: Among the most prominent health problems marring the global poultry industry for several decades are skeletal abnormalities. The aim of this study was to investigate a recent emergence of a novel form of skeletal deformity affecting cervical spine in broiler chickens. This work presents the natural history of this newly emerging skeletal anomaly along with long term observations of epidemiological trends in commercial broiler flocks, and clinical and pathological features. Results: In distinction from other forms of skeletal deformities commonly reported in broiler chickens, this new form of cervical spine anomaly have been observed in newly hatched chicks and in fully developed embryos that died in the shell. On clinical and post mortem examination this condition presents characteristic features consistent with congenital cervical scoliosis and torticollis (CCST). The pathogenesis of CCST appears to be linked to pathological remodeling of the cervical vertebrae bone associated with excessive activity of osteoclasts. Long term observations indicate that the incidence of CCST showed increasing epidemiological trends over time. More recently CCST has been observed in newly hatched chicks with incidence ranging from 0.1 to > 1%, and in fully developed embryos that failed to hatch about 4 to 5%. Conclusions: The increasing trends in incidence of CCST in commercial broiler flocks are of concern from an economic perspective, and also represent a very specific and important aspect of animal welfare.
