COVID-19 is a global pandemic that has claimed millions of lives. This disease is caused by a coronavirus, SARS-CoV-2, which requires the binding of its spike protein to angiotensin-converting enzyme 2 (ACE2) for infection of the host cell. Morinda citrifolia (noni) fruit juice has antiviral activity that involves enhancement of immune system function. SARS-CoV-2 spike-ACE2 interaction experiments were carried out to further investigate the antiviral properties of noni juice and its major iridoids. Noni juice inhibited binding by approximately 69%. Scandoside was the most active of the three iridoids evaluated, reducing average spike protein-ACE2 interaction by 79.25%. The iridoids worked synergistically towards inhibiting spike protein binding when assayed together, improving activity by more than 22% above the expected level. But the modest activity of the most abundant iridoid, deacetylasperulosidic acid, indicates that other phytochemicals (i.e. scopoletin, quercetin, rutin and kaempferol) are also involved. Our results suggest that the presence of several biological active phytochemicals in noni juice enhances resistance to SARS-CoV-2 by interfering with its ability to bind ACE2. This is a new and significant anti-viral mechanism of noni juice that does not directly involve its immunomodulatory properties.

Objective: To investigate the cytotoxic activity and molecular mechanism(s) of two Thai noni juice (TNJ) products ethanolic extracts against cholangiocarcinoma (CCA) cell lines and non-cancerous cells, and to explore phenolic acid compositions of TNJ products. Methods: Phenolic acid profiles of TNJ Chiangrai (TNJ-Cr) and TNJ Buasri (TNJ-Bs) ethanolic extracts were determined by HPLC. The cytotoxicity of TNJ ethanolic extracts on cancer and non-cancerous cells was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide and trypan blue assays. Mechanism(s) underlying the anti-CCA activity of TNJ ethanolic extracts were determined by cell cycle, apoptosis, and reactive oxygen species (ROS) generation assays. The expression levels of proteins involved in apoptosis and ERK signaling were evaluated by Western blot analysis. Results: Phenolic acid profiles of both TNJ ethanolic extracts showed that the p-hydroxybenzoic, vanillic, and protocatechuic acids were the major phenolic acids in TNJ products. Cytotoxicity assays revealed that the TNJ-Cr and TNJ-Bs ethanolic extracts reduced viability of CCA cell lines through induction of apoptosis by up-regulation of p53 and Bax proapoptotic proteins. Both TNJ ethanolic extracts promoted ROS generation by activating the ERK1/2 signaling in well-differentiated CCA cells KKU-213B. Meanwhile, TNJ ethanolic extracts did not induce ROS production in poorly differentiated CCA cells KKU-100. Both TNJ ethanolic extracts showed no toxicity to human peripheral blood mononuclear cells. Conclusions: TNJ ethanolic extracts could inhibit CCA cell proliferation by inducing ROS generation and apoptosis and may be applicable for combination therapies in CCA treatment.

Noni juice can inhibit the growth of Escherichia coli bacteria. Noni juice extraction needs concentration to extract so that the preparation is more stable in the storage process. The purpose of this study was to determine the antibacterial activity and the amount of the minimum inhibitory concentration of noni juice extract from E. coli bacteria in vitro. This type of experimental research with a completely randomized one-way design. The extract was obtained by concentrating the Noni leaf extract. Extract quality control parameters include organoleptic, yield, drying shrinkage, and qualitative tests of flavonoid and anthraquinone compounds. Antibacterial activity test using the disc diffusion method with an extract concentration of 1.56%; 3.12%; 6.25%; 12.5%; and 25% with 3 replications. Morinda citrifolia (noni) fruit juice has exhibited a variety of biological activities in human clinical trials, indicating that it influences multiple systems of the body. Since the 1990s, the endocannabinoid system (ECS) has been found to modulate the activity of other organ systems. To investigate noni’s potential impact on the ECS, extracts from freeze-dried noni fruit were evaluated in fatty acid amide hydrolase (FAAH) and monoacylglycerol lipase (MAGL) inhibition assays. The ethyl acetate extract demonstrated the greatest activity against both enzymes. Lignans in this extract also inhibited enzyme activities, with americanin A being the most active in both assays. Americanoic acid and 3,3’-bisdemethylpinoresinol were the next most active compounds. These results suggest that lignans in noni fruit may influence endocannabinoid levels within the body via FAAH and MAGL inhibition. This reveals another set of probable mechanisms of action by which noni juice affects human health.

Extraction of ripened noni juice has been carried out using ethanol and water. The purpose of this study was to determine the total phenolic, total flavonoids, scopoletin, content and antioxidant activity of the ripened noni juice extract. The activity test was carried out on the ripened noni juice extract without and with the addition of ethanol and water solvent. The phytochemical assays of ethanol extract of the ripened noni juice showed that thetotal phenolic content was 3,94 mg gallic acid equivalent/g extract, the total flavonoids was 0,59 mg quercetin equivalent /g extract, the IC50 antioxidant activity was 24,92 mg/L, scopoletin content was 2,45 mg/gram.Keywords : Antioxidant, flavonoids, phenolic, ripened noni juice, scopoletin

This study investigated the effect of noni juice (NJ) addition on physico-chemical, sensory, and bioactive properties of yogurt. NJ addition increased the acidity, yogurt starter count, total phenolics, total flavonoids, antioxidant activity, and flavor of yogurt. However, it reduced the viscosity, syneresis, and color parameters. Furthermore, whether feeding yogurt with or without NJ (7%, v/v) (NY or Y) (20 g/kg of diet) could alter the response of mice to dextran sodium sulfate (DSS)-induced ulcerative colitis (UC) was investigated. Intriguingly, dietary NY significantly ameliorated body weight loss, colon length, and histopathological changes in the UC mice. This relief of symptoms might be associated with the decreased mRNA expression of pro-inflammatory cytokines including IL-6 and IFN-γ and the increased mRNA expression of anti-inflammatory cytokine, IL-10. These results suggest the feasibility of NJ addition into yogurt which could serve as a potential adjuvant dietary therapy for reducing inflammation in UC patients.

Ginseng extract (GE), ferulic acid (≥99%) (FA), and a fermented noni juice powder (FNJP), were investigated for their antioxidant and antimicrobial activities in vitro. Half inhibitory concentration (IC50) was 29.87, 0.45 and 3.82 mg/mL, for GE, FA, and FNJP, respectively. The capacity of the three extracts to inhibit polyphenol oxidase from three vegetable matrices ranged between no inhibition and 95.1 % (depending on the extract and PPO source). In the study of peroxidation prevention of three types fats, only ferulic acid delayed lipid peroxidation of olive oil when applied at 10 mg/mL. The extracts’ antimicrobial activity was studied on thirteen bacterial strains using the disk diffusion assay and the microdilution assay. Minimal inhibitory concentration (MIC) values were 5.5 mg/mL of GE for Listeria monocytogenes, 1.7 mg/mL of FA for Staphylococcus aureus, L. monocytogenes 1/2 and 4b, and 4.2 mg/mL of FNJP for Bacillus cereus. The increases in lag phase, and decreases in growth rate and in asymptotic value of the bacteria growing under different concentrations of the three compounds were described. Application of 5-fluorouracil (5-FU) in cholangiocarcinoma (CCA) is limited by adverse side effects and chemoresistance. Therefore, the combination therapy of 5-FU with other substances, especially natural products may provide a new strategy for CCA treatment. The aim of this study was to evaluate the combination effects of 5-FU and two ethanolic extracts of Thai noni juice (TNJ) products on CCA cell lines and nude mice xenografts. The results of antiproliferative assay showed the combination treatment of 5-FU and each TNJ ethanolic extract exerted more cytotoxicity on CCA cells than either single agent treatment. Synergistic effects of drug combinations can enable the dose reduction of 5-FU. The mechanism underlying a combination treatment was apoptosis induction through an activation of p53 and Bax proteins. In the nude mouse xenograft model, combination treatments of 5-FU with each TNJ ethanolic extract suppressed the growth of CCA cells implanted mice more than single agent treatments with no effects on mouse body weight, kidney, and spleen. Moreover, low doses of TNJ ethanolic extracts reduced the hepatotoxicity of 5-FU in nude mice. Taken together, these data suggested that the ethanolic extracts of TNJ products can enhance the anti-CCA effect and reduce toxicity of 5-FU.

Morinda citrifolia L., known as noni, originated from Indonesia exhibits various pharmacological activities including anti-inflammatory properties. However, the validity of noni fruit juice as a treatment for rheumatoid arthritis (RA), an autoimmune disorder, has not been confirmed yet. Therefore, the main purpose of this research was to evaluate the efficacy of noni fruit juice (INFJ) made in Indonesia using SKG mice as an animal model of RA, which shows the resembling characteristics of human RA patients. Furthermore, the safety of INFJ was examined by repeated dose experiments in mice. INFJ was mixed with water at 50% and administered to SKG mice sensitized with mannan, free access for 4 weeks. Arthritis scores of fore- and hind-leg joints were measured and the joints were histopathologically examined. The sub-acute and sub-chronic toxicities of INFJ were evaluated using BALB/c mice. The arthritic scores were significantly lower from the 7 d after sensitization in the INFJ group than the control group. Histopathological examinations of the joints revealed inhibition of severity of RA. In both toxicity studies, INFJ did not show any toxicities. INFJ exhibited anti-arthritic activity in arthritic and histopathological examinations of the joints in SKG mice. Present study was the first report where noni juice may be effective against RA. The dose of noni juice showing efficacy against RA was confirmed safe from repeated dose studies in mice.

Morinda citrifolia (noni) fruit juice has been shown to have a wide variety of potential health benefits in human clinical trials. It may also influence the endocannabinoid system of the body. Since the main ingredient of the product studied in these clinical trials was juice made from noni fruit puree from French Polynesia, it was evaluated for its ability to inhibit the two major endocannabinoid degradation enzymes in vitro. Noni fruit juice inhibited both fatty acid amide hydrolase (FAAH) and monoacylglycerol lipase (MAGL) in a concentration-dependent manner, suggesting that it may help maintain anandamide and 2-arachidonoylglycerol levels. Samples of the puree were also analyzed for the presence of characteristic phytochemical markers of authentic noni fruit such as scopoletin, rutin, quercetin, deacetylasperulosidic acid and asperulosidic acid, all of which were present. Also present was scandoside, which is reported for the first time as being identified in noni fruit or its juice. Some of these compounds may contribute to the FAAH and MAGL inhibiting activity of noni juice. These results reveal another set of mechanisms by which noni juice possibly supports mental health, maintains joint health, relieves discomfort and modulates the immune system.

The change in phenols, polysaccharides and volatile profiles of noni juice from laboratory- and factory-scale fermentation was analyzed during a 63-day fermentation process. The phenol and polysaccharide contents and aroma characteristics clearly changed according to fermentation scale and time conditions. The flavonoid content in noni juice gradually increased with fermentation. Seventy-three volatile compounds were identified by solid-phase microextraction coupled with gas chromatography–mass spectrometry (SPME-GC-MS). Methyl hexanoate, 3-methyl-3-buten-1-ol, octanoic acid, hexanoic acid and 2-heptanone were found to be the main aroma components of fresh and fermented noni juice. A decrease in octanoic acid and hexanoic acid contents resulted in the less pungent aroma in noni juice from factory-scale fermentation. The results of principal component analysis of the electronic nose suggested that the difference in nitrogen oxide, alkanes, alcohols, and aromatic and sulfur compounds, contributed to the discrimination of noni juice from different fermentation times and scales.

Rubber plants (Hevea brasilensis) include sap plants, because this group has plant tissue that contains a lot of latex and the sap flows out when the plant tissue is injured (Budiman, 2012). The obstacle most often encountered in the process of making rubber processed materials is the large number of farmers who use coagulation materials that are not recommended by the government, thus decreasing the quality of the rubber produced. Therefore, it is necessary to use alternative coagulants that do not degrade and damage the quality of rubber, such as noni juice. The purpose of this study was to determine whether the production of noni juice coagulant as a coagulant can affect the ash content of the latex and to determine the correct dose of noni juice coagulant in reducing the ash content of the latex. This study used a randomized block design (RBD) with a single factor and the factors studied were the addition of coagulant doses of noni juice with 7 treatments (K0: natural latex (control), K1: 25 ml, K2: 50 ml, K3: 75 ml. , K4: 100 ml, K5: 125 ml of noni juice coagulant material and K6: 25 ml of salvage 90 and added 1 l of latex per experimental unit). The results of this study indicate that the volume of the noni juice coagulant has a very significant effect on the contact time, the gross weight of the bokar, and the ash content of the latex. The best dose of volume of noni juice coagulant to produce the lowest ash content of latex is the treatment of 75 ml of noni juice coagulant in 1 liter of liquid latex.

Background Morinda citrifolia (Noni) is an herbal remedy with multiple therapeutic properties such as antioxidant, anticancer, anti-inflammatory, antihypertensive effects due to rich in phytochemical constituents. Purpose To evaluate chemoprotective effect of noni in dipping cisplatin (CP)-induced cytotoxicity in normal (healthy) cells through an antioxidant activity using in-vitro and in-vivo. Methods Chemoprotective study of fresh noni juice (NJ) and divine noni gold (DNG) at the doses (0.3125–10 mg/ml) were performed against CP (3.125–100 µM) induced cytotoxicity in normal cells (BEAS-2B) by Sulforhodamine B assay at 24 h and 48 h and the percentage of growth inhibition was calculated. Similarly, the protective effect of noni (360 mg/b.wt.) were investigated against CP (5 mg/ml) induced cardiotoxicity in healthy mice through the investigation of serum biochemical markers, endogenous antioxidant enzymes were interpreted. Finally, the chemoprotective mechanism of DNG was accomplished Nrf2 expression following CP challenged NIH-3T3 cells was performed by the western blot method. Results Treatment with noni at tested dose exhibited the least toxicity in normal cells (BEAS-2B). CP at the following concentrations induced cell death in time and dose-dependent manner. Whereas, noni revealed a significant cytoprotective effect against CP-induced cytotoxicity in the normal cells. Further in in-vivo study, treatment with noni at 360 mg/b.wt in CP injected animals attenuated cardiac injury as indicated by quenching the activities of serum biochemical markers, alleviated enzymatic and non-enzymatic antioxidant enzymes. Furthermore, noni attenuated CP induced toxicity by indirect antioxidant activity were proved by Nrf2 expression. Conclusion Results suggest that DNG is more potent than NJ. Findings confirmed that DNG protects against CP-induced toxicity by activating the Nrf2 expression. More studies are needed to comprehend the mechanisms of DNG cytoprotection.

NaClO is not suitable as a root canal irrigant because of its cytotoxicity. Good biocompatibility irrigants are required to have antibacterial activity. Many herbal products like Bee glue, Noni juice and Azadirachta indica have such properties. This study aims to investigate the possible effects of propolis, MCJ and Neem on bacterial infections and cytotoxicity in primary plaque colonizers. Direct contact and agar diffusion tests evaluated the antibacterial activity of herbal products against Fusobacterium, Candida albicans and Prevotella. The CCK 8 test determined the influence of these herbal products on the proliferation of human apical papilla stem cells (hSCAPs) and human periodontal fibroblasts (hPDLFs). A migration assay test was performed in addition to quantitative real-time PCR which measured osteogenic differentiation in hSCAPs. All herbal extracts tested in this study exhibited antibacterial activity comparable to NaClO against bacterial infections, while the strongest bacteriostatic effect was shown in the herbal treated group. These extracts had much weaker effects on the proliferation and migration of hSCAPs and hPDLFs as calculated by the CCK-8 assay against NaClO. Bee glue treatment had the most potent effect on osteogenic differentiation, followed by treatment with Noni juice and Azadirachta indica (Neem), while NaClO showed the lowest effect. For primary plaque colonizers of immature or advanced permanent teeth, Bee glue, Noni juice, and Azadirachta indica can be promising irrigants with good biocompatibility. Microencapsulation of fermented noni juice (FNJ) into powder format could protect bioactive compounds, reduce the unpleasant odour and improve the acceptability for consumers. Blends of maltodextrin (MD) and gum acacia (GA) were used to achieve spray-drying microencapsulation of noni juice at different blending ratios. The physicochemical properties including microstructure, moisture content, water activity, particle size, bulk/tapped density, dissolution rate, ATR-FTIR and the bioaccessibility of bioactive compounds in powders during in vitro digestion were examined. Results showed that blends produced with more GA produced microcapsules with lower moisture content, water activity and bulk/tapped density, but slower powder dissolution. The ATR-FTIR results suggested that there were no significant chemical interactions between the core material and carrier or between the MD and GA in the blend powders. The spray-dried noni juice powder produced using the blends with higher ratio of GA to MD showed a better protection on the bioactive compounds, resulting in a higher bioaccessibility of powders during in vitro digestion. This study provides insights into microencapsulation of noni juice using blends of MD and GA and examines the physicochemical properties and bioaccessibilities of spray-dried powders as affected by the selected carriers.

Morinda citrifolia (Rubiaceae), also known as noni, is a plant found in the Hawaiian and Tahitian islands. It is considered as one of the most important plants brought to Hawaii by the first Polynesians. The yellow fruits have a distinctive ”grenade-like” shape and can grow up to a size of 12 cm. It has a foul taste and a soapy smell when ripe. It was proposed that the fruits of noni might suppress the growth of tumors by stimulating the immune system. In recent years, noni juice has been sold in the US market as a nutraceutical supplement. Here we addressed the volatile compounds in the fresh and ripe noni fruits. Carboxylic acids, aldehydes, ketones, alcohols, esters, terpenes and six organosulfur compounds were isolated by SDE and SPME, and further identified by GC/AED and GC/MS. The sulfur compounds identified may make a major contribution to the characteristic flavor of noni fruit juice. SPME is useful to recover low boiling point and low molecular weigh compounds; in contrast, SDE owns higher extraction capacity and higher recovery for polar compounds. SPME and SDE can provide complementary information; when use together, a deep understanding of aroma profile of food systems can be expected.

Morinda citrifolia L., also known as noni, has a long history of traditional use in the Hawaiian and Taihitian islands as cureall medicine. Recently, commercial noni juice products have been imported from different countries and delivered directly to consumers with claims of multiple health benefits, including anti-cancer and immuno-stimulating activities. In this study, phenolic compounds content, free radical and superoxide anion radical scavenging activities were determined in two noni juices with different extent of maturity from traditional fermentation process. Ripe noni fruits gave higher juice yield with a lighter color. In general, juice from ripe fruits contained higher total quantities of phenolic compounds, condensed tannins, flavonoids and scopoletin, and thus exhibiting better scavenging capacity for free radical, superoxide anion radicals and H2O2 and higher ACE inhibitory activity. Fermentation processing gave better juice recovery, whereas prolonged fermentation time reduced the reductive phytochemical content and some related bioactivities.

Objective The current treatment of leishmaniasis induces strong side effects and increasing numbers of cases of resistance to reference drugs have been reported. The discovery of the therapeutic properties of active substances in plant extracts represents an interesting field of research into a more efficient treatment against leishmaniasis. Morinda citrifolia, commonly known as noni, has demonstrated promising results as antileishmanial and immunomodulator. Thus, the aim of this work was to evaluate activity against axenic amastigote and hydrogen peroxide induction capacity by M. citrifolia fruit juice. Results Phytochemical screening identified anthraquinones, flavonoids, alkaloids, terpenoids, steroids, saponins, coumarins, phenolic compounds, tannins, anthocyanidins and chalcones. Noni juice exhibited dose-dependent activity and an IC50 of 240.1 µg/mL for axenic amastigotes. An absence of endotoxins was observed at the concentrations analyzed, while no cytotoxic effects were identified. Noni juice induced hydrogen peroxide production in BALB/c peritoneal macrophages but not in macrophages infected with Leishmania amazonensis. M. citrifolia fruit juice exhibited antileishmanial activity against L. amazonensis axenic amastigotes and activated macrophages by hydrogen peroxide induction, asserting its potential for further research into new forms of leishmaniasis treatment.

The fruit of Morinda citrifolia, commonly known as noni, has an extensive history of use as a food and medicine throughout the tropics. Among its many uses, noni was believed to promote skin health. In vivo wound healing studies reveal that noni fruit juice ingestion increases collagen production and deposition via increased expression of extracellular matrix protein genes. Noni juice also appears to promote fibroblast proliferation. In vitro studies indicate that noni juice also protects fibroblasts via activation of the transcription factor Nrf2 and protects extracellular matrix collagen by inhibiting matrix metalloproteinases. In vitro and in vivo study findings are corroborated by a clinical trial where hospitalization times were reduced, and wound granulation quality was improved in pediatric burn patients. The findings of other human studies also indicate that noni juice has the potential to protect skin collagen. These include reduced plasma reactive oxygen species in cigarette smokers, lowered skin glycation levels in overweight and obese adults, as well as reduced skin glycation levels among noni juice consumers within the general population. These in vitro, in vivo and human studies reveal that there are multiple mechanisms of action through which noni juice promotes collagen synthesis and inhibits collagen degradation. The procollagen properties of noni juice aid wound healing, increase skin elasticity, and improve the overall appearance of the skin.

Microfiltered noni (Morinda citrifolia) juice was given by cannulation to 6 Wistar rats for 13 days; the rats were also fed on a standard food formulation containing whole soy bean flour. Twenty-four hour urine was collected before and after the nutritional intervention and was analyzed by UPLC-DAD/ESI+-QTof/MS to perform an untargeted metabolomics study. A supervised multivariate OSC-PLS-DA analysis was performed to discriminate between urine samples. Among the top 10 most discriminant ions, 8 compounds were tentatively identified. Six were significantly increased after noni juice ingestion: the glucuronidate form of S-(-) equol, daidzein, 2 isomers of dihydroxyquinoline, naphthyl and hippuric acid. On the contrary, the urinary excretion of the oligosaccharides stachyose and raffinose, was significantly reduced. Still highly statistically significant, glucuronated genistein and glycetein were also elevated in urine after noni juice ingestion. Noni juice appeared to considerably enhance the bioavailability of soy isoflavone and modify the metabolism of soy oligosaccharides. Noni (Morinda citrifolia L.) is native to the Polynesian and recognized in the tropical and subtropical countries as a sustainable crop with feasible commercial applications. It has been reported that the interest in developing noni plant as a novel source of bioactive compounds are increasing by the day. This review describes the safety, nutritional values, and the properties of noni seed oil (NSO) with potential industrial uses. In particular, the bioactive compounds, anti-nutrients, antioxidant activity, and IC50 values of noni seed and the chemical composition of NSO are also described. NSO has high contents of polyunsaturated fatty acids, total phytosterols and tocopherols that could be better choices for patients with high cholesterol and cardiovascular diseases. Extracts of noni seed have been shown to possess bioactive compounds that exhibit antioxidant, anti-mutagenic, anti-tumor, anti-inflammatory, anti-allergic, anti-viral, anti-fungal, anti-microbial, and anti-carcinogenic properties. Bioactive compound-rich noni fruit seed could be a potential source of functional foods. Moreover, noni seeds could be a valuable new source of vegetable oil because of its nutritional properties and non-toxic nature along with the increasing supply of seeds as by-products from noni juice industry. Comprehensive studies are needed on NSO to explore more potential product development. Moreover, further study is needed on the development of nutraceutical food products from noni seed by-products.

Background: Excessive reactive oxygen species (ROS) can result in inflammation and cytokine secretion in the liver, and then activate hepatic stellate cells that cause the accumulation of extracellular matrix proteins, especially collagen, in liver tissue. Naturally fermented noni juice (NJ; Morinda citrifolia) has been used for decades as a nutraceutical in humans. In this study, we intended to examine if NJ can ameliorate ROS-induced liver fibrosis via a thioacetamide (TAA)-induced rat model. The present study assesses the feasibility of noni and mulberry as a raw substrate for the production of probiotic noni and mulberry juice by lactic acid bacteria (Lactobacillus plantarum SK-3 and Pediococcus acidilactici M-3). Changes in pH, titratable acidity (lactic acid), cell survival, and antioxidant properties were examined during fermentation. Both the strains grew well in noni juice and mulberry juice after 48 hour fermentation. P. acidilactici M-3 produced less lactic acid than L. plantarum SK-3. After 28 days of cold storage, both tested strains survived the low-pH conditions in fermented noni juice and mulberry juice. Both the juices fermented with L. plantarum SK-3 had a high antioxidant capacity. The noni and mulberry juice fermented with L. plantarum SK-3 had shown the cholesterol-lowering ability better than the juices fermented with P. acidilactici M-3. Finally, L. plantarum SK-3 and P. acidilactici M-3 were found as optimal probiotics for fermentation with noni juice as well as mulberry juice. In this investigation, the results could be an indicator of the development of health-promoting food juices.
