Deletion and site-directed point mutants of the polycistronic P/C mRNA of Sendai virus revealed that one of the nonstructural proteins of this virus, the C' protein, initiates from an ACG codon. This ACG codon occurs in an optimum sequence context and precedes the first AUG of the P/C mRNA. The results presented in this communication are consistent with the concept that the ribosomes scan the P/C mRNA to initiate several proteins from its different initiator codons. The arrangement of several weak initiator codons in tandem in an mRNA, i.e. non-AUG in optimum sequence context and AUG in suboptimum sequence context, may represent an alternate means to regulate gene expression in eukaryotes and their viruses.

Betaine, Arsonium Ylides A systematic study on the nature of decomposition of betaine formed by nucleophilic attack of semistabilized arsonium ylides on thiocarbonyl substrates has been carried out. A series of substituted benzylidenetriphenylarsenanes have been generated from their corresponding salts and condensed with few cyclic thioketones to yield exocyclic olefins as exclusive products. The non-availability of thioepoxides indicates that the mode of decomposition of such type of betaines is quite analogous to the betaine formed by phosphonium ylides. The structure of the products have been assigned by IR and NMR spectral studies.

Capped and uncapped methylated oligonucleotides, five and six nucleotides in length, were synthesized in vitro in spring viremia of carp virus transcription reaction mixtures containing ATP + CTP, ATP + CTP + GTP, or ATP + CTP + UTP, but not in any other combinations of two or three ribonucleoside triphosphates. The oligonucleotides that have been characterized are consistent with the structures: m7GpppAmpNpCpNpN, GpppAmpNpCpNpN, pppAmpNpCpNpN, and ppAmpNpCpNpN--i.e., similar to those of the termini of transcripts made in complete reaction mixtures. Because both capped and uncapped methylated oligonucleotides were synthesized, it can be concluded that methylation of the penultimate nucleotide can precede capping and methylation of the capping nucleotide. Our results also indicate that capping and methylation are processes that can take place prior to mRNA chain completion.

A water-soluble tuberculin-active peptidoglycan (TAPG) with a molecular weight of ca. 28,000 to 30,000 was isolated from the culture filtrate of Mycobacterium tuberculosis. TAPG was approximately four to five times more potent than tuberculin purified protein derivative S in guinea pigs sensitized with M. tuberculosis or M. bovis (freeze-dried BCG). It showed little or no cross-reactivity at a dose of 0.1 to 0.4 microgram in guinea pigs sensitized with M. kansasii, M. scrofulaceum, M. intracellulare, or M. avium. TAPG did not show any adjuvant activity when injected in guinea pigs in a water-in-oil emulsion containing ovalbumin. TAPG, in Freund incomplete adjuvant, proved to be an effective immunogen for inducing delayed hypersensitivity in guinea pigs. Chemical analysis of TAPG showed that it contains proline, glutamic acid, alanine, diaminopimelic acid, tyrosine, threonine, glucosamine, and the reducing sugars, arabinose and galactose. In immunoelectrophoretic studies with reference M. tuberculosis H37Rv antiserum, TAPG did not show any precipitin bands.

The Sendai virus polycistronic P/C mRNA encodes the P and C proteins from alternate overlapping reading frames. To determine the functions of these proteins in virus replication, hammerhead ribozymes were targeted to cleave the 5'-untranslated region of the P/C mRNA. Both cell-free and intracellular assays were employed to determine ribozyme efficacy. To appropriately compare activities between cell-free and intracellular assays, identical ribozymes were synthesized in vitro as well as expressed in cells. Ribozyme parameters, namely hybridization arm length (HAL) and nonhybridizing extraneous sequences (NES), were found to have rate-determining properties. In cell-free reactions, ribozymes with 13-mer HAL were up to 10-fold more efficient than those with 9-mer HAL. Ribozymes with 9-mer HAL were relatively ineffective in transfected cells. Minimizing the number of NES increased ribozyme efficiency in vitro. However, ribozymes with minimal NES were essentially inert intracellularly. The NES at the termini of the most effective intracellular ribozyme, Rz13st ( approximately 95% inhibition of the p gene expression), were predicted to fold into stem-loop structures. These structures most likely increase ribozyme stability as evidenced by the 8-fold higher resistance to ribonuclease T2 digestion of Rz13st compared with Rz13B. Our results suggest that when designing effective intracellular ribozymes, parameters that enhance formation of productive ribozyme:substrate duplexes and that increase RNA stability should be optimized.

The polycistronic P/C mRNA of Sendai virus is translated under cell-free conditions into five proteins (P, C', C, Y1, and Y2) from overlapping reading frames. In this study, we showed that in addition to the P, C', and C proteins, Y1 and Y2 were expressed by six different Sendai virus strains in infected cells. The Y proteins exhibited strain-specific variation in their gel mobility which corresponds to the variation seen in the cognate C proteins. While the relative levels of the P, C', and C proteins were consistent among various cell lines, the levels of Y1 and Y2 proteins varied among the cell lines used for viral infection.

To determine the translation initiation potential of the 5‘ proximal four AUGs of the polycistronic P/C mRNA of Sendai virus, we have developed an efficient system for oligonucleotide-directed site-specific mutagenesis utilizing a plasmid vector which contains the replication origin of the single-stranded DNA phage f1 and the phage SP6 promoter. Utilizing uridine containing single-stranded DNA templates of the plasmid vector carrying the P/C gene we introduced point mutations in all four of the putative initiator codons (AUGs) of the P/C gene. Based on the analysis of in vitro translation products from the SP6 polymerase-directed transcripts of the mutants, the first and second AUG codons were assigned to the P and C proteins, respectively. Proteins detected in vitro, Y1 and Y2, initiated from the third and fourth AUG codons, respectively, and originated within the C reading frame. Since the C’ protein is encoded in the same reading frame as the C protein but did not initiate from any of the 5‘ proximal AUGs, the synthesis of the C’ protein may start from a non-AUG initiator codon in the P/C gene.

The assembly of nucleocapsids is an essential step in the replicative cycle of vesicular stomatitis virus (VSV). In this study, we have examined the early events of vesicular stomatitis virus nucleocapsid assembly in BHK-21 cells. Nuclease-resistant intracellular nucleocapsids were isolated at various stages of assembly and analyzed for RNA and protein contents. The smallest ribonucleoprotein complex formed during nucleocapsid assembly contains the 5'-terminal 65 nucleotides of nascent viral RNA complexed with the viral proteins N and NS. Elongation of the assembling nucleocapsids proceeds unidirectionally towards the 3' terminus by the sequential addition of viral proteins which incrementally protect short stretches of the growing RNA chain. Pulse-chase studies show that the assembling nucleocapsids can be chased into full-length nucleocapsids which are incorporated into mature virions. Our results also suggest an involvement of the cytoskeletal framework during nucleocapsid assembly.

Direct sequencing of nine Sendai virus defective interfering RNA species revealed two kinds of 3'-terminal sequences. Six RNA species had 3' termini identical to the virus genome (negative strand), confirming that internal deletions are a frequent cause of Sendai virus defectiveness. The other three RNA species had 3'-terminal sequences identical to that described as the complement of the 5' terminus of the virus genome (R. A. Lazzarini, J. D. Keene, and M. Schubert, Cell 26:145-154, 1981), indicating that they are of the copy-back type. Extensive homology between these two types of 3' sequences evidently accounts for the ability of the copy-back sequence to function as an initiation signal for viral RNA replication. There may not be a selective advantage of one type of terminus over the other, since one defective interfering strain possessed two RNA species, one of which had the genomic 3' terminus and the other copy-back type.

Translation of Sendai virus nucleocapsid protein (NP) and phosphoprotein (P/C) mRNAs in rabbit reticulocyte lysates in the presence of antisense oligodeoxynucleotides (15-20-mers) showed that oligonucleotides having complementarity within the 5′ noncoding region of the mRNAs blocked translation, oligonucleotides having complementarity within 20 nucleotides upstream from the initiator codon blocked translation only partially, and oligonucleotides complementary to the coding region of mRNA had no effect on translation. The results suggest the possibility that the 80 S initiation complex may form about 20 bases upstream from the initiator codon. Alternatively, the 40 S preinitiation complex may recognize an initiator codon at least 20 nucleotides upstream from the codon, activating a helix-destabilizing process.
