Background The use of artificial intelligence (AI) assisted white light imaging (WLI) detection systems for superficial esophageal squamous cell carcinoma (SESCC) is limited by training with images from one specific endoscopy platform. Methods In this study, we developed an AI system with a convolutional neural network (CNN) model using WLI images from Olympus and Fujifilm endoscopy platforms. The training dataset consisted of 5892 WLI images from 1283 patients, and the validation dataset included 4529 images from 1224 patients. We assessed the diagnostic performance of the AI system and compared it with that of endoscopists. We analyzed the system's ability to identify cancerous imaging characteristics and investigated the efficacy of the AI system as an assistant in diagnosis. Results In the internal validation set, the AI system's per-image analysis had a sensitivity, specificity, accuracy, positive predictive value (PPV), and negative predictive value (NPV) of 96.64%, 95.35%, 91.75%, 90.91%, and 98.33%, respectively. In patient-based analysis, these values were 90.17%, 94.34%, 88.38%, 89.50%, and 94.72%, respectively. The diagnostic results in the external validation set were also favorable. The CNN model’s diagnostic performance in recognizing cancerous imaging characteristics was comparable to that of expert endoscopists and significantly higher than that of mid-level and junior endoscopists. This model was competent in localizing SESCC lesions. Manual diagnostic performances were significantly improved with the assistance by AI system, especially in terms of accuracy (75.12% vs. 84.95%, p = 0.008), specificity (63.29% vs. 76.59%, p = 0.017) and PPV (64.95% vs. 75.23%, p = 0.006). Conclusions The results of this study demonstrate that the developed AI system is highly effective in automatically recognizing SESCC, displaying impressive diagnostic performance, and exhibiting strong generalizability. Furthermore, when used as an assistant in the diagnosis process, the system improved manual diagnostic performance.

Most theories of information security policy (ISP), except a few focused on the insider-centric view, are grounded in the control-centric perspective, and most ISP compliance models stem from Western countries. Regulatory focus theory (RFT) proposes two modes of motivational regulation, promotion and prevention focused that are supposed to motivate employee compliance in a trade-off. Culture is crucial to the study of ISP that puts control over human connections. Chinese guanxi, a specific dimension of Chinese culture, is better understood underlying the trust-distrust frame. To bridge the theoretical gap between the control-centric and the insider-centric perspectives, we develop an ISP behavioral model by taking an integrated approach from RFT and the trust-distrust frame. We employed scenario-based events about information security misconduct in the workplace to examine employees’ compliance intention and non-violation choice of ISP upon counterfactual thinking. Our empirical results improve the theoretical and practical implications of security practices.

This program evaluation examined integration of primary care nurse practitioners into assertive community treatment (ACT).From January to June 2019, primary care nurse practitioners in a postgraduate fellowship program were assigned to five ACT teams (N=305 participants). Focus groups explored staff members' and participants' experiences. Screening rates for hemoglobin A1c and cholesterol for ACT participants were compared over time.Staff and participants in ACT described improved engagement in primary care, citing benefit from colocation and consultation. Field visits were not found to be an efficient use of the primary care nurse practitioners' time to serve most ACT participants. A significant increase in screening was observed after 6 months for the ACT teams with integrated primary care.An integrated primary care nurse practitioner readily available for participant engagement and consultation with the ACT team, using a cardiometabolic registry to guide care, may offer a sustainable model of integration.

Silicone oil is a type of fluid with low viscosity, but it is not easy to form stable cone jet for electrohydrodynamic printing. In this paper, we proposed a new electrohydrodynamic printing method for patterns fabrication with this kind of low viscosity fluid. Dots array was first printed on the substrate at higher direct current voltage. Then by controlling the moving speed of the platform, the dots were connected into lines according to the fluidity of the silicone oil and its low surface tension. With the proposed method, the patterns with silicone oil can be successfully formed by electrohydrodynamic printing. In the experiment, the influence of main parameters including applied voltage, moving speed of substrate, distance from needle to substrate, and axial length of droplet on the quality of printed lines was studied. Finally, by optimizing the printing parameters, the silicone oil lines with width of 73 µm and low surface roughness were printed.

Objective: The purpose of this paper is to investigate whether playing exergames can enhance quality of life among young adults and it examines the potential moderators. Methods: A 12-week randomized controlled trial was conducted. Quality of life was measured using the short-form 36-item version (SF-36) scale. All the participants were between 20 and 24 years old in Taiwan. Participants in the intervention group (n = 55) were asked to play exergames for 12 weeks, three times a week and 30 minutes at a time, while participants in the control group (n = 62) did not play exergames. The changes in the scores on quality of life between the beginning and the end of the 12-week trial were calculated. Independent t-tests were used to analyze the differences. Results: The intervention group participants experienced an enhanced quality of life in terms of physical functioning, role-physical (role limitations due to physical health), general health, and social functioning. Moreover, the intervention group participants who were not enthusiastic about exercisers experienced an enhanced quality of life in physical functioning, role-physical, and general health. The intervention group participants who attempted to control their weight experienced enhanced general health, vitality, and mental health. Conclusion: Playing exergaming could contribute to users’ quality of life in terms of both physical and mental health.

We present a unified approach to single-machine scheduling with position-based processing times, an availability constraint, and job rejection. The approach uses two general position-based processing time functions to model both the learning and aging effects in scheduling. In addition, taking machine availability and job rejection into consideration, the models are more realistic, which seek to minimize the sum of the makespan of the accepted jobs and the total penalty of the rejected jobs. We present fully polynomial-time approximation schemes to address the two NP-hard problems.

Aiming at the problem that the Beidou B1I navi¬gation receiver is susceptible to continuous-wave interfer¬ence (CWI), it is of great significance to investigate the interference effect and mechanism of the B1I navigation receiver. With the threshold of carrier-to-noise ratio (C/N0) as the criterion of satellite tracking missing, the CWI injection experiment is conducted on a kind of B1I navigation receiver. The results indicate that the power spectrum of the satellite signal affects the CWI's ability to interfere with each satellite. Then, a prediction model of satellite tracking loss under dual-frequency CWI is proposed, and the accuracy of the model is verified through experiments. The experiment results demonstrate that the power combination between the dual-frequency CWI is negatively correlated, and the error of this model is less than 2 dB. Finally, the sensitive frequency bands of the navigation receiver under out-of-band CWI are obtained through experiments. It is confirmed by theoretical and simulation analysis that the mixer is the main non-linear circuit generating these sensitive frequency bands. Furthermore, improving the selectivity of the radio frequency (RF) front-end circuit can enhance the performance of anti out-of-band interference.

Background Endoscopic screening is the widely accepted screening strategy for esophageal squmaous cell carcinoma (ESCC). However, massive endoscopic screening is expensive and not cost-efficient, and novel pre-endoscopy detection used as a preliminary screening method arouses new concerns. We are planning to launch an artificial intelligence (AI) assisted sponge cytology for detecting esophageal squmaous high-grade intraepithelial neoplasia (HGIN) and above lesions. The aim of this trail is to investigate the efficiency of AI-assisted sponge cytology in population-based screening of early esophageal squmaous epithelial lesions. Methods The study will be prospectively conducted in five regions with a high prevalence of ESCC. AI-assisted sponge cytology and endoscopic examination will be sequentially performed. Based on our previous data, at least 864 patients with esophageal HGIN and above lesions are needed to achieve enough statistical power. And, a calculated 112,500 individuals with high risks of ESCC will be recruited. In the first stage, each 24,000 participants who meet the inclusion criteria will be recruited on a voluntary basis. Setting pathological results as standard reference, diagnostic threshold and according performance of AI-assisted detection will be evaluated. A prediction model will be constructed by co-analyzing cytological results and relevant risk factors. Then, an external validation cohort will be used for validation of the model efficiency. Also, cost-efficiency analysis will be performed. This study protocol was registered on chineseclinicaltrial.gov (ChiCTR1900028524). Discussion Our study will determine whether this AI-assisted sponge cytology can be used as an effective pre-endoscopy detection tool for large-scale screening for ESCC in high-risk areas.

Relative entropy identities yield basic decompositions of cat- egorical data log-likelihoods. These naturally lead to the development of information models in contrast to the hierarchical log-linear models. A recent study by the authors clarified the principal difference in the data likelihood analysis between the two model types. The proposed scheme of log-likelihood decomposition introduces a prototype of linear information models, with which a basic scheme of model selection can be formulated accordingly. Empirical studies with high-way contingency tables are exem- plified to illustrate the natural selections of information models in contrast to hierarchical log-linear models. Analysis of contingency tables with multi-way classifications has been a fun- damental area of research in the history of statistics. From testing hypothesis of independence in a 2 × 2 table (Pearson, 1904; Yule, 1911; Fisher, 1934; Yates, 1934) to testing interaction across a strata of 2 × 2 tables (Bartlett, 1935), many discussions had emerged in the literature to build up the foundation of statisti- cal inference of categorical data analysis. In this vital field of applied statistics, three closely related topics have gradually developed and are still theoretically incomplete even after half a century of investigation. The first and primary topic is born out of the initial hypothesis testing for independence in a 2 × 2 table. From 1960's utill the 1980's, Fisher's exact test re- peatedly received criticism for being conservative due to discrete nature (Berkson, 1978; Yates, 1984). Although the arguments in favor of the unconditional tests essentially focused on using the unconditional exact tests in the recent decades, the reasons for preferred p values and sensitivity of the unconditional tests have not been assured in theory. To assess the safety and biological activity of rozibafusp alfa, a first-in-class bispecific antibody-peptide conjugate targeting inducible costimulator ligand (ICOSL) and B cell activating factor (BAFF), in patients with rheumatoid arthritis (RA).This phase 1b, double-blind, placebo-controlled, multiple ascending dose study included 34 patients (18-75 years; 82.4% female) with active RA (Disease Activity Score of 28 joints-C-reactive protein [DAS28-CRP]2.6, on stable methotrexate) randomized 3:1 to receive rozibafusp alfa (n = 26, in four ascending dose cohorts of 70, 140, 210, and 420 mg) or a placebo (n = 8) subcutaneously once every 2 weeks for 10 weeks (six total doses), with 24 weeks of follow-up. The primary end point was the incidence of treatment-emergent adverse events (TEAEs). Additional assessments included serum pharmacokinetics (PK), pharmacodynamics (PD), immunogenicity, and RA disease activity measures (DAS28-CRP, Patient Global Assessment of Disease, and Physician Global Assessment of Disease).TEAEs occurred in 96.2% and 87.5% of patients receiving rozibafusp alfa and the placebo, respectively; most were mild or moderate in severity. Two (7.7%) patients treated with rozibafusp alfa reported serious TEAEs; none were considered treatment related. Multiple doses of rozibafusp alfa showed nonlinear PK (mean tMultiple ascending doses of rozibafusp alfa were well tolerated, with PK and PD reflecting dual ICOSL and BAFF blockade. Findings support further clinical evaluation of rozibafusp alfa in autoimmune disease.

Electrohydrodynamic (EHD) printing has great potential to be used in the field of electronic industry, precision optics and biological engineering. However, it is still a challenge to achieve a high scaling ratio for line patterns production by EHD jet printing technique, which can effectively prevent the needles to be blocked. In this paper, the influence of the key process parameters on the line width of EHD jet printing was investigated. The printing needle with the inner diameter of 890 μm was used, and the line with width of 3.02 μm was printed by optimizing the parameters. The scaling ratio between the needle inner diameter and the printed line width was up to 295 times. At the same time, the function model between the flow rate and printed line width of PVP/PEO composite was deduced. The error was found to be less than 1% by the verification of experiments. By utilizing this model, the printed line width can be predicted by controlling the flow rate, which will greatly improve the work efficiency. By controlling the flow rate, the goal of printing on demand can be realized. The proposed method and model can provide support for the fabrication of micro‐nano scale structure.

Nanofluidic devices with two-dimensional nanochannels have many applications in biology and chemistry, however, it is still a challenge to develop a low-cost and simple method for fabricating nano-masks that can be used to produce two-dimensional nanochannels. In this paper, a novel low-cost and simple method, based on UV lithography and oxygen plasma, was proposed to fabricate nano-mask. The influence of exposure time on the photoresist mesas was investigated in the ultraviolet lithography process. The parameters of RF power and treatment time on the width reduction of photoresist mesas were analyzed by the oxygen plasma. In our work, in order to increase the efficiency controllability of photoresist removal, a RF power of 90 W, a pressure of oxygen plasma 60 Pa, and the time division method were adopted to remove photoresist by oxygen plasma. Finally, nano-patterns of photoresist mesas with bottom width of 330 nm were successfully fabricated. The proposed method provides a low-cost way to produce high-throughput two-dimensional nanochannels.

The Western Desert is located in Egypt and it consists of a few extensional coastal rift-basins. It started as rifts and was formed during the Jurassic time in association with the opening of the Tethys Sea. There were three major tectonic events that occurred in Western Desert within Jurassic to Eocene time and resulted in NE-SW trend and NW-SE trend normal fault, and inversion of Western Desert basin due to rifting of Neo-Tethys followed by South America & Africa Atlantic rifting and Transpressional Syrian Arc event which had contributed to the formation of hydrocarbon trap. The generation, migration and accumulation of hydrocarbon started in the Late Cretaceous (95–90 Ma) and it continues to the present time. There is proven petroleum system named as Khatatba-Khatatba petroleum system within the Western Desert. The source of hydrocarbons is the Middle Jurassic Khatatba organicz-rich shales which contains type II- III and type III kerogen source migrated into Khatatba sandstones reservoir rock. Khatatba sandstones are mostly quartz arenite, which composed mainly of more than 95 % quartz. These sandstones have high porosity and high permeability with well sorted and are mostly subangular to subrounded grains. Masajid carbonate acts as regional seal within the basin. Hence, the Western Desert of Egypt has a significant hydrocarbon potential for exploration or development targeting on inversion structure.

Vaccination is a well-known method to protect the public against an epidemic outbreak, e.g., COVID-19. To this end, the government of a country or region would strive to achieve its target of vaccination coverage. Limited by the total vaccine capacity of public hospitals, the government may need to cooperate with private hospitals or clinics for more vaccination. Exploring in this paper government coordination of public and private resources for vaccination, we model a vaccine system consisting of a public hospital, a profit-maximizing private clinic, and self-interested individuals, under three scenarios: (1) without information sharing (concerning vaccine inventory and vaccine price), (2) with information sharing and subsidy, and (3) with information sharing and allocation. We find that, under scenario (1), the vaccine demand is fully satisfied by the public hospital and the private clinic cannot make any profit. Under scenario (2), the private clinic is willing to enter the vaccine market with a positive profit-maximizing vaccination coverage. Under scenario (3), the socially optimal vaccination coverage may be lower than that under scenario (1). Moreover, we conduct a sensitivity analysis to generate practical implications of the research findings for vaccination policy-making. Our results provide both theoretical and managerial insights on vaccine supply decision, government intervention, and vaccination coverage.

Background Creeping fat, a form of visceral adipose tissue (VAT) that wraps the intestinal wall, influences the formation of Crohn’s disease (CD) strictures. The degree of fat wrapping from intestinal stricture resections is correlated with the extent of chronic inflammation, fibrosis, stricture formation, and response to biologic therapy. VAT and subcutaneous adipose tissue (SAT) ratios from CTE (computed tomography) scans are elevated in CD strictures. However, the definition of strictures in these studies has been poorly defined and not included current well-recognized criteria: 1) bowel wall thickness (BWT), 2) narrowed luminal diameter, and 3) pre-stenotic dilation. (PSD). Purpose The objective of this pilot study was to assess the relationship of 2D and 3D VAT:SAT ratios with CT stricture parameters in patients with terminal ileal (TI) CD strictures. Method 2D VAT:SAT ratios from CT’s of CD patients with TI strictures defined as increased BWT, narrowed luminal diameter (< 50% relative to normal adjacent distended loop), and PSD greater than the stricture diameter were retrospectively obtained from a database and chart review. CT’s from fibrostenotic CD patients were sex and BMI matched to patients with only TI inflammatory behaviour. Patient demographics, medication, smoking, and surgical history were also obtained. Analyses were adjusted for age, sex, and BMI covariates. Unpaired t-tests and multi-variable logistic regression analyses were conducted. Result(s) Twenty-eight patients with stricturing CD had a significantly greater mean VAT:SAT volume ratio than 29 non-stricturing CD (41.5 cm3 vs 34.2 cm3, p=0.03). Thirty-six percent (10/28) of CD stricture patients had prior ileocolic resection with a mean disease duration of 13.5 years (range 0-48). The median ileal BWT (7.0 mm, range 4.0-13.0 mm) for the stricturing group was significantly greater than those with inflammatory behaviour (BWT 2.0 mm, p Image Conclusion(s) Fibrostenotic TI CD patients have increased VAT:SAT ratios in comparison to those with only inflammatory behaviour. Background Robots are introduced into health care contexts to assist health care professionals. However, we do not know how the benefits and maintenance of robots influence nurse–robot engagement. Objective This study aimed to examine how the benefits and maintenance of robots and nurses’ personal innovativeness impact nurses’ attitudes to robots and nurse–robot engagement. Methods Our study adopted a 2-wave follow-up design. We surveyed 358 registered nurses in operating rooms in a large-scale medical center in Taiwan. The first-wave data were collected from October to November 2019. The second-wave data were collected from December 2019 to February 2020. In total, 344 nurses participated in the first wave. We used telephone to follow up with them and successfully followed-up with 331 nurses in the second wave. Results Robot benefits are positively related to nurse–robot engagement (β=.13, P Conclusions Our study is the first to examine how the benefits and maintenance requirements of assistive robots influence nurses’ engagement with them. We found that the impact of robot benefits on nurse–robot engagement outweighs that of robot maintenance requirements. Hence, robot makers should consider emphasizing design and communication of robot benefits in the health care context.

AMG 966 is a bi-specific, heteroimmunoglobulin molecule that binds both tumor necrosis factor alpha (TNFα) and TNF-like ligand 1A (TL1A). In a first-in-human clinical study in healthy volunteers, AMG 966 elicited anti-drug antibodies (ADA) in 53 of 54 subjects (98.1%), despite a paucity of T cell epitopes observed in T cell assays. ADA were neutralizing and bound to all domains of AMG 966. Development of ADA correlated with loss of exposure.In vitrostudies demonstrated that at certain drug-to-target ratios, AMG 966 forms large immune complexes with TNFα and TL1A, partially restoring the ability of the aglycosylated Fc domain to bind FcγRIa and FcγRIIa, leading to the formation of ADA. In addition to ADA against AMG 966, antibodies to endogenous TNFα were also detected in the sera of subjects dosed with AMG 966. This suggests that the formation of immune complexes between a therapeutic and target can cause loss of tolerance and elicit an antibody response against the target.

Background Both the direct anterior approach (DAA) and posterior approach (PA) to THA have known advantages and disadvantages. The comparison between DAA and PA THA has been widely explored during the early postoperative period. However, few randomized trials have compared these approaches at a minimum follow-up of 5 years; doing so would be important to establish any differences in mid-term outcomes or complications. Questions/purposes We performed a randomized trial comparing DAA and PA in THA in terms of (1) patient-reported outcome scores, (2) quality of life and functional outcomes assessed by the EQ-5D and 10-meter walk test results, (3) radiographic analysis, and (4) survivorship and surgical complications at a minimum of 5 years follow-up. Methods Two hip specialist surgeons performed both DAA and PA THA using the same THA components at two hospital sites. One hundred twelve patients on the elective THA surgical waitlist were invited to participate in the study. Thirty-four patients did not meet the study's inclusion criteria and were excluded, and three patients declined to participate in the study. The remaining 75 patients who were eligible were randomized into DAA and PA groups. Thirty-seven patients were initially randomized to receive DAA THA, but two did not and were excluded, resulting in 48% (35 of 73) of patients who received DAA THA; 52% (38 of 73) of patients were randomized into and received PA THA. Over a minimum 5 years of follow-up, 3% (1 of 35) of DAA patients were lost to follow-up, and none of the patients undergoing PA THA were lost. A per-protocol analysis was adopted, resulting in further patients being excluded from analysis. Of the 73 study patients, 99% (72; DAA: 35, PA: 37) were analyzed at 1 year, 95% (69; DAA: 34, PA: 35) were analyzed at 2 years, and 72% (52; DAA: 23, PA: 29) were analyzed at 5 years. The primary outcome was the Oxford Hip Score (OHS) and WOMAC score. Secondary outcomes included the EQ-5D and EQ-5D VAS scores, 10-meter walk test results, radiographic evidence of loosening (femoral: lucency > 2 mm at the implant-bone interface, subsidence > 2 mm; acetabular: migration or change in inclination), 5-year survivorship analysis from all-cause revisions, and surgical complications. The study was powered to detect a 10-point difference in the WOMAC score, which is equivalent to the minimum clinically important difference (MCID). Results There were no differences in primary outcomes (OHS and WOMAC scores) or secondary outcomes (EQ-5D scores, EQ-5D VAS scores, and 10-meter walk test result) between the DAA and PA groups at the 5-year follow-up interval. The median (range) OHS at 5 years was 46 (16 to 48) for DAA and 47 (18 to 48) for PA groups (p = 0.93), and the median WOMAC score was 6 (0 to 81) for DAA and 7 (0 to 59) for PA groups (p = 0.96). The median EQ-5D score was 1 (0.1 to 1) for DAA and 1 (0.5 to 1) for PA groups (p = 0.45), and the median EQ-5D VAS score was 85 (60 to 100) for DAA and 95 (70 to 100) for PA groups (p = 0.29). There were no cases of component loosening on radiographs. There was no difference in component survival between the two approaches at 5 years (DAA: 97% [95% CI 85% to 100%] versus PA: 97% [95% CI 87% to 100%]). Eight of 23 patients in the DAA group reported decreased sensation in the lateral femoral cutaneous nerve distribution. Conclusion DAA and PA are both effective approaches in performing primary THA. Each approach has its associated risks and complications. The choice of THA should be based on individual patient factors, surgeon experience, and shared decision-making. Early registry data indicate DAA and PA THA are comparable, but longer-term data with larger numbers of patients will be required before one can safely conclude equal survivorship between both approaches. This study investigates the antecedents of environmentally responsible behaviour by creating a theoretical model of relationships between service quality, place attachment and environmentally responsible behaviour. A questionnaire survey was conducted among tourists who visited six famous tourist attractions in Shaanxi, China. The results of the research demonstrate that service quality exerts influence on environmentally responsible behaviour and place attachment in a positive manner. Additionally, the findings suggest that place attachment affects environmentally responsible behaviour in a direct and positive manner. Moreover, it is also found that service quality strengthens its effect on environmentally responsible behaviour through place attachment, indicating the mediating role of place attachment in the link between service quality and environmentally responsible behaviour. The current research contributes to tourism literature in that it helps to better understand the tourists’ environmentally responsible behaviour and provides management with evidence of the significance for improving the service quality at tourist attractions.

Several research works have been carried out on Shoushan Basin, Western Desert including the petroleum system in the Shoushan Basin. However, there is no proper classification was done on Shoushan Basin, Western Desert, Egypt using Global Basin Classification. The objective of this study is to classify Shoushan Basin using Global Basin Classification as proposed by Kingston et al. (1983a, b). The study which was done by integrating different types of data including geochemical, well log, seismic data from literature review provided a better understanding of the evolution and the hydrocarbon potential of Western Desert. Shoushan Basin consists of two basin types which included Margin Sag (MS) Cycle Basin and Wrench Cycle Basin (LL). The combination of these cycles, stages and tectonic event results in the formula being written from the youngest to the oldest as follows: LL – 2/LD/MS – 321321321.

Six-day fieldwork was conducted in the north-west coast of Sarawak to examine the outcrops along Bintulu- Niah- Miri areas which cover southern part of Balingian Province and Baram Delta Province. The aim of this fieldwork is to synthesize the observations of structure and sedimentology of outcrops along Bintulu-Niah-Miri areas and discuss the implication of reservoir characteristics based on observation. The study was conducted by sketching the main structural elements of outcrops followed by detailed sedimentological analysis which include observation and facies description were conducted on different outcrops along Bintulu-Niah- Miri areas using sedimentary logs. The findings show that Bintulu- Niah- Miri areas outcrops consists of mixed-environment deposited succession with tidal and wave characteristic. This resulted in reservoir architect will be different and result in different in reservoir properties included horizontal and vertical permeability of fluids. Niah Cave is a good place to study the distribution of the types of breccia due to collapsed paleokarst at reservoir scale and also good analog for Central Luconia Platform where large resources of hydrocarbon have been discovered due to the its environment setting or forming process is same as Central Luconia Platform. In addition, Miri Airport Outcrop succession consist of Type 4- Fracture Create Flow Barriers which could lead to potential production problems.

This study aimed to clarify the mechanism of propofol on proliferation and apoptosis of colorectal cancer (CRC) cell. SW620 and HCT15 cells were exposed to different concentrations of propofol, the proliferation and apoptotic rate, were measured by MTT, colony formation and flow cytometry assays, respectively. The expressions of miR-1-3p and insulin-like growth factors 1 (IGF1) were examined by real-time polymerase chain reaction (RT-qPCR). Western bolt was employed to quantify the protein levels of IGF1 and apoptotic proteins. The molecular interaction between miR-1-3p and IGF1 was validated using dual-luciferase reporter assay. A xenograft tumor model was established to further assess the effects of propofol on CRC in vivo. Propofol dramatically decreased the proliferation and elevated apoptotic rate of CRC cells. RT-qPCR assay demonstrated that miR-1-3p was downregulated in CRC cells, and could be strikingly increased by propofol. Importantly, miR-1-3p inhibited IGF-1 expression through interacting with its 3'-UTR region, thus inactivating AKT/mTOR signals. Gain or loss of functional study revealed that miR-1-3p downregulation remarkedly diminished the anti-tumor roles of propofol by directly inhibiting IGF1. In vivo study showed that propofol inhibited tumor growth by regulating miR-1-3p/IGF1 axis. Our data eventually elucidated that propofol suppressed CRC progression by promoting miR-1-3p which targeted IGF1. These results might provide a scientific basis for the application of propofol on the clinical surgery and the prognosis of patients with CRC.

Photosynthetic bacteria are beneficial to plants, but knowledge of photosynthetic bacterial community dynamics in field crops during different growth stages is scarce. The factors controlling the changes in the photosynthetic bacterial community during plant growth require further investigation. In this study, 35 microbial community samples were collected from the seedling, flowering, and mature stages of tomato, cucumber, and soybean plants. 35 microbial community samples were assessed using Illumina sequencing of the photosynthetic reaction center subunit M (pufM) gene. The results revealed significant alpha diversity and community structure differences among the three crops at the different growth stages. Proteobacteria was the dominant bacterial phylum, and Methylobacterium, Roseateles, and Thiorhodococcus were the dominant genera at all growth stages. PCoA revealed clear differences in the structure of the microbial populations isolated from leaf samples collected from different crops at different growth stages. In addition, a dissimilarity test revealed significant differences in the photosynthetic bacterial community among crops and growth stages (PMethylobacterium were present in varying abundances among different sample types, which we speculated was related to the function of different Methylobacterium species in promoting plant growth development and enhancing plant photosynthetic efficiency. In conclusion, the dynamics observed in this study provide new research ideas for the detailed assessments of the relationship between photosynthetic bacteria and different growth stages of plants.

Background The efficacy of risankizumab (RZB), an interleukin 23 p19 inhibitor, in patients with Crohn’s disease (CD) has been reported. Normalisation of high-sensitivity C-reactive protein (hs-CRP) and faecal calprotectin (FCP) are intermediate treatment targets in CD. Here, we evaluated changes in these objective inflammatory biomarkers and clinical outcomes with RZB treatment. Methods In 2 phase 3, randomised, double-blind studies (ADVANCE, NCT03105128; MOTIVATE, NCT03104413), patients with moderately to severely active CD received 12-week intravenous (IV) RZB induction therapy or placebo (PBO). Patients with clinical response to RZB IV induction were rerandomised in a 52-week maintenance study (FORTIFY, NCT03105102) to receive subcutaneous (SC) RZB or PBO (ie, RZB withdrawal). Induction analyses included patients who received either 600 mg RZB IV or PBO for 12 weeks. Maintenance analyses included patients who received 360 mg RZB SC every 8 weeks or withdrawal (PBO SC) for 52 weeks. Outcomes assessed were normalisation of hs-CRP and FCP concentrations at week 12 of induction and at week 52 of maintenance in patients with elevated biomarkers at baseline (hs-CRP > 5 mg/L and/or FCP > 250 μg/g), clinical biomarker response (defined as enhanced clinical response [≥ 60% average daily stool frequency (SF) decrease and/or ≥ 35% average daily abdominal pain score (APS) decrease] and ≥ 50% reduction in hs-CRP or FCP), and clinical biomarker remission (defined as clinical remission per CD Activity Index or SF/APS criteria and normal hs-CRP or FCP) during maintenance treatment. Nonresponder imputation was used for missing data. Results Greater proportions of patients receiving RZB vs PBO achieved normalisation of hs-CRP and FCP at the end of the 12-week induction period and the 52-week maintenance period (P < .0001 for all; Table). Among patients with clinical response to RZB IV induction and entered maintenance, rates of clinical biomarker response were maintained through week 52 in patients receiving 360 mg RZB SC and declined over time among patients in the withdrawal (PBO SC) arm (Figure A). Rates of clinical biomarker remission increased over time in patients receiving 360 mg RZB SC. At week 52, clinical remission and normalisation of hs-CRP or FCP was achieved by 41% of patients receiving RZB vs 28%–29% of patients in the withdrawal (PBO SC) arm (Figure B-C). The safety profile of RZB in CD was previously reported. Conclusion Normalisation of objective biomarkers of inflammation in CD was achieved with RZB induction and maintenance therapy. Improvements in both clinical outcomes and biomarker levels were sustained with continuous RZB maintenance therapy and decreased over time in patients discontinuing RZB.
