Nerve Growth Factor (NGF) and Brain derived Neurotrophic Factor (BDNF) mature/precursor imbalance in tears and serum is suggested as a risk factor and symptomatology aggravation in ophthalmology and neuropsychiatric disturbances. Cognitive and mood alterations are reported by patients with Graves' Orbitopathy (GO), indicating neurotrophin alterations might be involved. To address this question, the expression levels of NGF and BDNF and their precursors in serum and tears of GO patients were analyzed and correlated with the ophthalmological and psycho-cognitive symptoms. Hamilton Rating Scale for Anxiety (HAM-A) and Depression (HAM-D), Temperament and Character Inventory (TCI), and Cambridge Neuropsychological Test Automated Battery (CANTAB) test were used as a score. NGF and BDNF levels were measured using ELISA and Western Blot and statistically analyzed for psychiatric/ocular variable trend association. GO patients show memorization time and level of distraction increase, together with high irritability and impulsiveness. HAM-A and CANTAB variables association, and some TCI dimensions are also found. NGF and BDNF expression correlates with ophthalmological symptoms only in tears, while mature/precursor NGF and BDNF correlate with the specific psycho-cognitive variables both in tears and serum. Our study is the first to show that changes in NGF and BDNF processing in tears and serum might profile ocular and cognitive alterations in patients.

Background/Objectives: Chronic migraine with medication-overuse headache (CM-MOH) is neurophysiologically characterized by increased cortical excitability with sensitization at both the thalamocortical and the cortical levels. It is unclear whether the increased cortical excitability could be reverted by medication withdrawal (i.e., brain state) or whether it is a brain trait of individuals predisposed to medication overuse. In this study, we aim to investigate whether withdrawal from overused drugs can influence and restore these neurophysiological abnormalities. Methods: Somatosensory evoked potentials (SSEPs) were elicited by electrical stimulation of the median nerve (M), the ulnar nerve (U), and the simultaneous stimulation of both nerves (MU) in 14 patients with CM-MOH before (T0) and after (T1) a three-week withdrawal protocol and, for comparison, in 14 healthy volunteers (HVs) of a comparable age distribution. We measured the level of thalamocortical (pre-HFO) and cortical activation (post-HFO) by analyzing the high-frequency oscillations (HFOs) embedded in parietal N20 median SSEPs. Furthermore, we calculated the habituation and the degree of cortical lateral inhibition (dLI) of N20-P25 low-frequency SSEPs. Results: After the three-week withdrawal protocol (T1), we observed a normalization of the baseline habituation deficit (T0: +0.10 ± 0.54; T1: −0.53 ± 0.8; p = 0.040) and a reduction in the amplitude for both pre-HFO (p < 0.009) and post-HFO (p = 0.042), with values comparable to those of the HVs. However, no effects were observed on the dLI (p = 0.141). Conclusions: Our findings showed that withdrawal from overused drugs could affect the increased excitability of the non-painful somatosensory system in patients with CM-MOH, reducing the level of sensitization at both the thalamocortical and the cortical levels. Background/Objectives: Habituation and sensitization are opposite phenomena that play a role in the pathophysiology of episodic migraine and its progression to chronic migraine (CM). There have been just a few studies that have investigated these phenomena in patients with medication overuse headache (MOH) in comparison to those with chronic migraine (CM) and healthy controls (HCs), and the findings have been inconsistent. Methods: We measured and examined visual evoked potentials (VEPs) in 81 patients with MOH and 24 patients with CM, as well as 24 HCs. The VEPs were used to assess sensitization by analysing the amplitude of the first block (100 sweeps) and to evaluate habituation by measuring the amplitude response decrement after six blocks. We further examined patients diagnosed with MOH based on their acute medication type and after a 3-week acute medication withdrawal program. Results: There were no significant differences between groups in terms of the first N1-P1 VEP amplitude block and its habituation. It was found that patients with MOH had a greater drop in the amplitude of the VEP P1-N2 complex after repeated stimulation than patients with CM or HC. The VEP parameters showed no significant differences based on the specific overused drug and after a 3-week acute medication withdrawal. Conclusions: We propose that the results obtained in patients with MOH indicate an abnormal activation of inhibitory circuits in the parieto-occipital region in response to repeated modulatory stimuli. Ancient stadia and circuses were considered by Greeks and Romans to be excellent places for live events. Back in ancient times, many people participated in public entertainment from athletic games, as typical of Greek traditions, to combats between gladiators and wild beasts. Among all of them, the most acclaimed were the horse races conducted with chariots, and this was the main sport of ancient Roman stadia. This paper deals with the digital reconstruction of three stadia belonging to the 2nd century B.C. (i.e., Panathenaic Stadium) and to the 1st century A.D. (i.e., Circus Maximum and Stadium of Domitian). The digital models have been rebuilt based on historical resources and archaeological discoveries to conduct the acoustic simulations and understand the acoustic behavior within these places. After the assessment of the main acoustic parameters, the noise levels from crowds have been predicted in different ways: based on information gathered from historic annals, and the comfort used for modern stadia to predict the ancient conditions with reference to the crowd noise levels measured in modern stadiums. The results indicate that the acoustic response of ancient stadia is very similar to the modern ones, in terms of both reverberation and noise level from crowds. [ABSTRACT FROM AUTHOR]

Neurodegenerative diseases constitute a global health issue and a major economic burden. They significantly impair both cognitive and motor functions, and their prevalence is expected to rise due to ageing societies and continuous population growth. Conventional therapies provide symptomatic relief, nevertheless, disease-modifying treatments that reduce or halt neuron death and malfunction are still largely unavailable. Amongst the common hallmarks of neurodegenerative diseases are protein aggregation, oxidative stress, neuroinflammation and mitochondrial dysfunction. Transcription factor nuclear factor-erythroid 2-related factor 2 (NRF2) constitutes a central regulator of cellular defense mechanisms, including the regulation of antioxidant, antiinflammatory and mitochondrial pathways, making it a highly attractive therapeutic target for disease modification in neurodegenerative disorders. Here, we describe the role of NRF2 in the common hallmarks of neurodegeneration, review the current pharmacological interventions and their challenges in activating the NRF2 pathway, and present alternative therapeutic approaches for disease modification. Ischemic stroke (IS) is a major cause of mortality and disability among adults. Recanalization of blood vessels to facilitate timely reperfusion is the primary clinical approach; however, reperfusion itself may trigger cerebral ischemiareperfusion injury. Emerging evidence strongly implicates the NLRP3 inflammasome as a potential therapeutic target, playing a key role in cerebral ischemia and reperfusion injury. The aberrant expression and function of NLRP3 inflammasome-mediated inflammation in cerebral ischemia have garnered considerable attention as a recent research focus. Accordingly, this review provides a comprehensive summary of the signaling pathways, pathological mechanisms, and intricate interactions involving NLRP3 inflammasomes in cerebral ischemiareperfusion injury. Moreover, notable progress has been made in investigating the impact of natural plant products (e.g., Proanthocyanidins, methylliensinine, salidroside, a-asarone, acacia, curcumin, morin, ginsenoside Rd, paeoniflorin, breviscapine, sulforaphane, etc.) on regulating cerebral ischemia and reperfusion bymodulating the NLRP3 inflammasome andmitigating the release of inflammatory cytokines. Neurodegenerative diseases are associated with oxidative stress, due to an imbalance in the oxidation-reduction reactions at the cellular level. Various treatments are available to treat these diseases, although they often do not cure them and have many adverse effects. Therefore, it is necessary to find complementary and/or alternative drugs that replace current treatments with fewer side effects. It has been demonstrated that natural products derived from plants, specifically phenolic compounds, have a great capacity to suppress oxidative stress and neutralize free radicals thus, they may be used as alternative alternative pharmacological treatments for pathological conditions associated with an increase in oxidative stress. The plant species that dominate the Mediterranean ecosystems are characterized by having a wide variety of phenolic compound content. Therefore, these species might be important sources of neuroprotective biomolecules. To evaluate this potential, 24 typical plant species of the Mediterranean ecosystems were selected, identifying the most important compounds present in them. This set of plant species provides a total of 403 different compounds. Of these compounds, 35.7% are phenolic acids and 55.6% are flavonoids. The most relevant of these compounds are gallic, vanillic, caffeic, chlorogenic, p-coumaric, and ferulic acids, apigenin, kaempferol, myricitrin, quercetin, isoquercetin, quercetrin, rutin, catechin and epicatechin, which are widely distributed among the analyzed plant species (in over 10 species) and which have been involved in the literature in the prevention of different neurodegenerative pathologies. It is also important to mention that three of these plant species, Pistacea lentiscus, Lavandula stoechas and Thymus vulgaris, have most of the described compounds with protective properties against neurodegenerative diseases. The present work shows that the plant species that dominate the studied geographic area can provide an important source of phenolic compounds for the pharmacological and biotechnological industry to prepare extracts or isolated compounds for therapy against neurodegenerative diseases. Conferences worldwide focus on a range of disciplines relating to the construction of the built environment. They tend to emphasize either the art or the science of building, the former focusing on architectural theory and design while the latter targets a range of topics from civil and/or building engineering to building physics. Vitruvius's De Architectura Libri Decem is a seminal treatise more than two millennia old which addresses these themes in a holistic manner. This text remains valid today for students and professionals engaged in architecture and building engineering. Translated as Ten Books on Architecture, it not only presents an overall view of the disciplines of town planning, architecture and civil engineering, along with the qualifications required to practice them, but also addresses building materials, civil-engineering structures and the science influencing buildings. Although grounded in the practice and technology of Ancient Rome, the principles put forward in this treatise are still valid nowadays for effective, sustainable architectural-engineering design based on rigorous education and good knowledge of building materials and construction. Vitruvius's definition of architecture—the one still customarily used—is an inclusive philosophical statement on the essence of building for humanity to house humanity. In recent years, 3D technology has been widely used in various aspects of the entire workflow of conservation–restoration. However, in the majority of cases, researchers have mainly relied on 3D technologies to assist the conventional conservation–restoration process, and its advantages have not truly been exploited. In this paper, we applied a combination of digital acquisition, virtual anastylosis, virtual reconstruction, and 3D printing of a transparent reversible prosthesis with slots for the restoration of a gold foil decorated Black Ding bowl collected by the Chifeng Museum of the Inner Mongolia Autonomous Region. While completing the physical/aesthetic restoration of the artifact, the conservation–restoration principles of the integrity and authenticity of cultural heritage, minimal intervention and reversibility were followed to the utmost extent. At the same time, we also conducted preliminary performance tests on the 3D printing material of the prosthesis, and the results showed that the material has excellent mechanical properties and stain resistance, contributing to the long-term, stable preservation of the artifact. The Temple of Apollo Pythios in Gortyn (Crete, Greece) dates back to the mid-7th century BC. The temple underwent several transformations and an ultimate destruction over time that resulted in the current remains of a Roman colonnade composed of six fragmented stone columns lying on the ground within the naos. In addition, the region was struck by several earthquakes which contributed to the various changes. The analysis of the fragments composing the column portions and their geometric features provided a possible outline for their standing repositioning. Based on the current seismic hazard of the region, a predictive numerical model of the colonnade in the anastylosis conditions suggested the need for proper connections between the fragments and the bases to ensure their overall conservation as a compound structure. The comparison of various configurations of intervention and the simple superimposition of the fragments also provided details on the accomplishment of minimal and compatible solutions. The paper presents a practical approach to define a hypothetical virtual reconstruction of the ciborium and pergola of Monte Sorbo's Church. The high accuracy of archaeological fragments, digitally acquired, allowed to hypothesize the entire sequences of decorations of architectural elements and to define their geometrical rules. Starting from the acquisition to the 3d modelling phase, the proposed virtual reconstruction combines original fragments with their lost parts modelled in a digital environment.Digital technologies offer nowadays the great possibility to visualize and understand cultural heritage in a new and attractive way. Digital models and virtual reconstructions have a key role in encouraging and promoting the development of good practices for recording, documenting, and making accessible scientific processes beyond visual appearance. The term virtual reconstruction, within the project, is used to describe a procedure that consists of making a 3D model that reproduces a digital copy of the original, assembling digitally acquired fragments with elements philologically reconstructed following the evidence-based geometrical rules. This study aims to contribute to this growing area of research by exploring possibilities on digital integration of different 3d models to re-define a visual appearance of lost architectural elements. There has been significant progress in recent years aimed at the development of new analytical techniques for investigating structure-function relationships in hierarchically ordered materials. Inspired by these technological advances and the potential for applying these approaches to the study of construction materials from antiquity, we present a new set of high throughput characterization tools for investigating ancient Roman concrete, which like many ancient construction materials, exhibits compositional heterogeneity and structural complexity across multiple length scales. The detailed characterization of ancient Roman concrete at each of these scales is important for understanding its mechanics, resilience, degradation pathways, and for making informed decisions regarding its preservation. In this multi-scale characterization investigation of ancient Roman concrete samples collected from the ancient city of Privernum (Priverno, Italy), cm-scale maps with micron-scale features were collected using multi-detector energy dispersive spectroscopy (EDS) and confocal Raman microscopy on both polished cross-sections and topographically complex fracture surfaces to extract both bulk and surface information. Raman spectroscopy was used for chemical profiling and phase characterization, and data collected using EDS was used to construct ternary diagrams to supplement our understanding of the different phases. We also present a methodology for correlating data collected using different techniques on the same sample at different orientations, which shows remarkable potential in using complementary characterization approaches in the study of heterogeneous materials with complex surface topographies. Historic buildings are representations of cultural systems throughout time. Thus, it is important to shed light on any given historic building's chronological information by means of restitution, that is, the descriptions of different (re)compositions of a building for different historical states. However, the representation of information beyond the geometric, spatial, and physical characteristics of a historic building carries a crucial importance in the holistic understanding of historical changes. This requirement consists of representation, archival, assessment, and management aspects. Nevertheless, covering all these aspects is still quite challenging. Thus, three-dimensional (3D) visualization in digital platforms is considered, which indeed seems to be the most desirable way today. This article proposes a methodology through adopting an approach based on the Geographical Information System (GIS) and further develops a digital "container" as a 3D digital model for the archiving of a building's chronological information. This does not only cover the geometric and spatial aspects, but also the historical resources and their reliability. The paper concludes that the developed prototype may lessen the future investigation effort of heritage specialists by making the chronological information of a historical building more integrated and coherent, through increased readability, accessibility, and visibility. Psychological stress triggers headaches, but how this happens is unclear. To explore this, 38 migraine sufferers, 28 with tension-type headache (T-TH) and 20 controls rated nausea, negative affect, task-expectancies and headache at 5-min intervals during an aversive 20-min mental arithmetic task with a fixed failure rate. Blood pressure and pulse rate were measured every 3 min and salivary cortisol was sampled before and after the task. Multiple regression analysis indicated that irritation, anxiety and the absence of sluggishness (i.e., alertness) independently predicted increases in headache intensity during the task ( p < .001), but increases in headache were unrelated to changes in cardiovascular activity or cortisol. Changes that preceded headache onset were explored in repeated measures ANOVAs, comparing those who developed headache with those who did not. In general, nausea, negative affect and self-efficacy expectancies were higher in participants who went on to develop headache than in those who remained headache-free ( p < .05 to p < .001). Together, these findings suggest that headache developed when participants overextended themselves during a stressful task, adopting an information processing style which impeded emotional adjustment to changing situational demands. Learning to modify perceptions of threat, and adopting a more flexible, less outcome-dependent processing style, might help to prevent headache from spiralling upward. [ABSTRACT FROM AUTHOR]

Neurological disorders such as stroke, Alzheimer's and Parkinson's diseases are associated with high morbidity and mortality, and few or no effective options are available for their treatment. These disorders share common pathological characteristics like the induction of oxidative stress, abnormal protein aggregation, perturbed Ca2+ homeostasis, excitotoxicity, inflammation and apoptosis. A large body of evidence supports the beneficial effects of the Mediterranean diet in preventing neurodegeneration. As the Mediterranean diet is characterized by a high consumption of extra-virgin olive oil it has been hypothesized that olive oil, and in particular its phenols, could be responsible for the beneficial effect of the Mediterranean diet. This review provides an updated vision of the beneficial properties of olive oil and olive oil phenols in preventing/counteracting both acute and chronic neurodegenerative diseases. In March 2001, the world's largest depictions of standing Buddha figures located in Bamiyan (Afghanistan) were destroyed during a campaign of the Taliban leadership against all non-Muslim heritage in the country. Bamiyan and its archaeological remains have been nominated a World Heritage also to commemorate the events of destruction the site has experienced over the centuries. More than fifteen years after the Giant Buddha figures turned into dust, the UNESCO Safeguarding campaign for the preservation of the Bamiyan World Heritage property has made considerable progress. Upon continued request by the local population and the national government, the international community is asked to contribute proposals for the future presentation of the physical remains including the options of reassembling the original fragments. The achievements and backlashes of the UNESCO campaign are discussed and a proposal is given, how virtual technology can contribute to the ongoing discussion regarding the future of the site. At a time when deliberate raids during military conflicts against heritage places have become commonplace, the Bamiyan case has become emblematic and is considered now a key turning point in the heritage community on the question, which roles reconstruction can have especially in the aftermath of conflict situations. It is argued that immersive virtual reality technologies offer the chance to investigate how values attributed to cultural heritage are produced and experienced among different stakeholder groups. The Maritime Theatre is one of the iconic buildings of Hadrian's Villa, Tivoli. The state of conservation of the theatre is not only the result of weathering over time, but also due to restoration work carried out during the Fifties of the past century. Although this anastylosis process had the virtue of partially restoring a few of the fragments of the compound's original image, it now reveals diverse inconsistencies and genuine errors in the reassembling of the fragments. This study aims at carrying out a digital reinterpretation of the restoration of the architectural fragments in relation to the architectural order, with particular reference to the miscellaneous decoration of the frieze of the Teatro Marittimo (vestibule and atrium). Over the course of the last few years the Teatro Marittimo has been the target of numerous surveying campaigns using digital methodology (laser scanner and photogrammetry SfM/MVS). Starting with the study of the remains of the opus caementicium on the ground, it is possible to identify surfaces which are then used in the model for subsequent cross sections, so as to achieve the best fitting circumferences to use as reference points to put the fragments back into place. The development of close-range photogrammetry has produced a lot of new possibility to study cultural heritage. 3D data acquired with conventional and low cost cameras can be used to document, investigate the full appearance, materials and conservation status, to help the restoration process and identify intervention priorities. At the same time, with 3D survey a lot of three-dimensional data are collected and analyzed by researchers, but there are a very few possibility of 3D output. The augmented reality is one of this possible output with a very low cost technology but a very interesting result. Using simple mobile technology (for iPad and Android Tablets) and shareware software (in the case presented "Augment") it is possible to share and visualize a large number of 3D models with your own device. The case study presented is a part of an architecture graduate thesis, made in Rome at Department of Architecture of Roma Tre University. We have developed a photogrammetric survey to study the Aurelian Wall at Castra Praetoria in Rome. The surveys of 8000 square meters of surface have allowed to identify stratigraphy and construction phases of a complex portion of Aurelian Wall, specially about the Northern door of Castra. During this study, the data coming out of 3D survey (photogrammetric and topographic), are stored and used to create a reverse 3D model, or virtual reconstruction, of the Northern door of Castra. This virtual reconstruction shows the door in the Tiberian period, nowadays it's totally hidden by a curtain wall but, little and significative architectural details allow to know its original feature. The 3D model of the ancient walls has been mapped with the exact type of bricks and mortar, oriented and scaled according to the existing one to use augmented reality. Finally, two kind of application have been developed, one on site, were you can see superimposed the virtual reconstruction on the existing walls using the image recognition. Research on architectural archaeology conducted by archaeologists, architects, engineers - and more recently by computer scientists - is characterized by different goals and methodologies. Only sometimes does this result in profitable contributions. Instead, attitudes very often remain deeply rooted in single-discipline vision unable to communicate or interact with each other in the best interest of the subject of the research in question. In this paper, by means of a series of case studies, we intend to outline the principles of "reverse designing," that is the integrated set of methods and skills enabling the retracings of the steps taken by the ancient builder. Multiple sclerosis (MS) is the most common multifocal inflammatory demyelinating disease of the central nervous system (CNS). Due to the progressive neurodegenerative nature of MS, developing treatments that exhibit direct neuroprotective effects are needed. Tecfidera™ (BG-12) is an oral formulation of the fumaric acid esters (FAE), containing the active metabolite dimethyl fumarate (DMF). Although BG-12 showed remarkable efficacy in lowering relapse rates in clinical trials, its mechanism of action in MS is not yet well understood. In this study, we reported the potential neuroprotective effects of dimethyl fumarate (DMF) on mouse and rat neural stem/progenitor cells (NPCs) and neurons. We found that DMF increased the frequency of the multipotent neurospheres and the survival of NPCs following oxidative stress with hydrogen peroxide (H2O2) treatment. In addition, utilizing the reactive oxygen species (ROS) assay, we showed that DMF reduced ROS production induced by H2O2. DMF also decreased oxidative stress-induced apoptosis. Using motor neuron survival assay, DMF significantly promoted survival of motor neurons under oxidative stress. We further analyzed the expression of oxidative stress-induced genes in the NPC cultures and showed that DMF increased the expression of transcription factor nuclear factor-erythroid 2-related factor 2 (Nrf2) at both levels of RNA and protein. Furthermore, we demonstrated the involvement of Nrf2-ERK1/2 MAPK pathway in DMF-mediated neuroprotection. Finally, we utilized SuperArray gene screen technology to identify additional anti-oxidative stress genes (Gstp1, Sod2, Nqo1, Srxn1, Fth1). 2014 is the hundredth anniversary of the outbreak of the First World War (WWI) - or Great War - in Europe and a number of initiatives have been planned to commemorate the tragic event. Until 1918, the Italian Trentino -- Alto Adige region was under the Austro -- Hungarian Empire and represented one of the most crucial and bloody war front between the Austrian and Italian territories. The region borders were constellated of military fortresses, theatre of battles between the two opposite troops. Unfortunately, most of these military buildings are now ruined and their architectures can be hardly appreciated. The paper presents the initial results of the VAST project (VAlorizzazione Storia e Territorio -- Valorization of History and Landscape), that aims to digitally reconstruct the forts located on the plateaus of Luserna, Lavarone and Folgaria. An integrated methodology has been adopted to collect and employ all possible source of information in order to derive precise and photo-realistic 3D digital representations of WWI forts. Alzheimer's Disease (AD) is a neurodegenerative disorder affecting up to one third of individuals reaching the age of 80. Different integrated responses exist in the brain to detect oxidative stress which is controlled by several genes termed Vitagenes. Vitagenes encode for cytoprotective heat shock proteins (Hsp), as well as thioredoxin, sirtuins and uncouple proteins (UCPs). In the present study we evaluate stress response mechanisms in plasma and lymphocytes of AD patients, as compared to controls, in order to provide evidence of an imbalance of oxidant/antioxidant mechanisms and oxidative damage in AD patients and the possible protective role of vitagenes. We found that the levels of Sirt-1 and Sirt-2 in AD lymphocytes were significantly higher than in control subjects. Interestingly, analysis of plasma showed in AD patients increased expression of Trx, a finding associated with reduced expression of UCP1, as compared to control group. This finding can open up new neuroprotective strategies, as molecules inducing this defense mechanisms can represent a therapeutic target to minimize the deleterious consequences associated to oxidative stress, such as in brain aging and neurodegenerative disorders. Neurodegenerative diseases (NDDs) and cardiovascular diseases (CVDs) are illnesses that affect the nervous system and heart, all of which are vital to the human body. To maintain health of the human body, vegetable diets serve as a preventive approach and particularly Brassica vegetables have been associated with lower risks of chronic diseases, especially NDDs and CVDs. Interestingly, glucosinolates (GLs) and isothiocyanates (ITCs) are phytochemicals that are mostly found in the Cruciferae family and they have been largely documented as antioxidants contributing to both cardio- and neuroprotective effects. The hydrolytic breakdown of GLs into ITCs such as sulforaphane (SFN), phenylethyl ITC (PEITC), moringin (MG), erucin (ER), and allyl ITC (AITC) has been recognized to exert significant effects with regards to cardio- and neuroprotection. From past in vivo and/or in vitro studies, those phytochemicals have displayed the ability to mitigate the adverse effects of reactive oxidation species (ROS), inflammation, and apoptosis, which are the primary causes of CVDs and NDDs. The Forma Urbis Romae (F.U.) was a 3rd-century-AD monumental map of ancient Rome consisting of 151 rectangular marble slabs. Several efforts have been made to reconstruct it from its current incomplete and fragmentary condition. In this paper, we explore the potential of an archaeometric approach to serve this purpose. Almost a hundred F.U. fragments have been characterized, particularly focusing on cathodoluminescence (CL) microscopy and stable isotopes (δ18O and δ13C). Different statistical methods have been used to quantify the similarity between samples. The central assumption is that samples from a given slab share similar CL colors and isotopic ratios. The assumption has been verified for samples from single fragments and then it has been used to check ten debated reconstruction hypotheses. The measured isotopic ratios confirm the Proconnesian nature of the F.U. marble, except for a fragment. Beyond provenance, the results cast doubts on four out of the ten checked reconstruction hypotheses and support the other six. The reconstruction of the F.U. remains a fascinating challenge, and both isotopic and CL analyses have demonstrated their potential to tackle it. Further research could extend the presented methodology to a higher number of samples. The innovative use of CL to reconstruct a fragmented artwork could be applied to other projects. An integrated research approach consisting of hydrogeologic and geochemical methods was applied to a coastal aquifer in the Ostia Antica archaeological park, Roma, Italy, to describe freshwater–saltwater interactions. The archaeological park of Ostia Antica is located on the left bank of the Tevere River delta which developed on a morphologically depressed area. The water monitoring program included the installation of multiparametric probes in some wells inside the archaeological area, with continuous measurement of temperature, electrical conductivity, and water table level. Field surveys, water sampling, and major elements and bromide analyses were carried out on a seasonal basis in 2016. In order to understand the detailed stratigraphic setting of the area, three surface boreholes were accomplished. Two distinct circulations were identified during the dry season, with local interaction in the rainy period: an upper one within the archaeological cover, less saline and with recharge inland; and a deeper one in the alluvial materials of Tevere River, affected by salinization. Oxygen and carbon isotopic signature of calcite in the sediments extracted from the boreholes, along with major elements and Br concentration, allowed us to recognize the sources of salinity (mainly, local interaction with Roman salt pans and agricultural practices) and the processes of gas–water–rock interaction occurring in the area. Neurodegenerative diseases are driven by several mechanisms such as inflammation, abnormal protein aggregation, excitotoxicity, mitochondrial dysfunction and oxidative stress. So far, no therapeutic strategies are available for neurodegenerative diseases and in recent years the research is focusing on bioactive molecules present in food. In particular, extra-virgin olive oil (EVOO) phenols have been associated to neuroprotection. In this study, we investigated the potential antioxidant and neuroprotective activity of two different EVOO extracts obtained from Quercetano cultivar trees grown in two different areas (plain and hill) of the Tuscany region (Italy). The different geographical origin of the orchards influenced phenol composition. Plain extract presented a higher content of phenyl ethyl alcohols, cinnammic acids, oleacein, oleocanthal and flavones; meanwhile, hill extract was richer in lignans. Hill extract was more effective in protecting differentiated SH-SY5Y cells from peroxide stress thanks to a marked upregulation of the antioxidant enzymes heme oxygenase 1, NADPH quinone oxidoreductase 1, thioredoxin Reductase 1 and glutathione reductase. Proteomic analysis revealed that hill extract plays a role in the regulation of proteins involved in neuronal plasticity and activation of neurotrophic factors such as BDNF. In conclusion, these data demonstrate that EVOOs can have important neuroprotective activities, but these effects are strictly related to their specific phenol composition. [ABSTRACT FROM AUTHOR]

Sulforaphane (SFN) is a phytocompound belonging to the isothiocyanate family. Although it was also found in seeds and mature plants, SFN is mainly present in sprouts of many cruciferous vegetables, including cabbage, broccoli, cauliflower, and Brussels sprouts. SFN is produced by the conversion of glucoraphanin through the enzyme myrosinase, which leads to the formation of this isothiocyanate. SFN is especially characterized by antioxidant, anti-inflammatory, and anti-apoptotic properties, and for this reason, it aroused the interest of researchers. The aim of this review is to summarize the experimental studies present on Pubmed that report the efficacy of SFN in the treatment of neurodegenerative disease, including Alzheimer's disease (AD), Parkinson's disease (PD), and multiple sclerosis (MS). Glucosinolates (GSs) are common anionic plant secondary metabolites in the order Brassicales. Together with glucosinolate hydrolysis products (GSHPs), they have recently gained much attention due to their biological activities and mechanisms of action. We review herein the health benefits of GSs/GSHPs, approaches to improve the plant contents, their bioavailability and bioactivity. In this review, only literature published between 2010 and March 2020 was retrieved from various scientific databases. Findings indicate that these compounds (natural, pure, synthetic, and derivatives) play an important role in human/animal health (disease therapy and prevention), plant health (defense chemicals, biofumigants/biocides), and food industries (preservatives). Overall, much interest is focused on in vitro studies as anti-cancer and antimicrobial agents. GS/GSHP levels improvement in plants utilizes mostly biotic/abiotic stresses and short periods of phytohormone application. Their availability and bioactivity are directly proportional to their contents at the source, which is affected by methods of food preparation, processing, and extraction. This review concludes that, to a greater extent, there is a need to explore and improve GS-rich sources, which should be emphasized to obtain natural bioactive compounds/active ingredients that can be included among synthetic and commercial products for use in maintaining and promoting health. Furthermore, the development of advanced research on compounds pharmacokinetics, their molecular mode of action, genetics based on biosynthesis, their uses in promoting the health of living organisms is highlighted. Geological models are very useful tools for developing conceptual schemes owing to their capacity to optimize the management of stratigraphic information. This is particularly true in areas where archaeological heritage is exposed to hydrogeological hazards; 3D models can constitute the first step toward the construction of numerical models created to understand processes and plan mitigation actions to improve visitor safety and preserve archaeological heritage. This paper illustrates the results of a 3D hydrostratigraphic model of the site of the Colosseum in the Central Archaeological Area of Rome. In recent years, this area has experienced numerous floods caused by intense meteorological events. A new borehole survey provided the opportunity to update previous maps and cross sections and build a local scale 3D model. The resulting conceptual model was used to identify primary gaps in existing knowledge about the groundwater system and to optimize the planning of a piezometer monitoring network. Further studies can then focus on the development of groundwater numerical models to verify hypotheses regarding inflow-outflow dynamics and facilitate the optimization of water management. In this paper, crushed lava granulate was used as full silica sand replacement in composition of repair mortars based on hydrated lime, natural hydraulic lime, or cement-lime binder. Lava granules were analyzed by X-ray fluorescence analysis (XRF), X-ray diffraction (XRD), and scanning electron microscopy (SEM). Particle size distribution of both silica and lava aggregates was assessed using standard sieve analysis. Hygrothermal function of the developed lightweight materials was characterized by the measurement of complete set of hygric, thermal, and structural parameters of the hardened mortar samples that were tested for both 28 days and 90 days cured specimens. As the repair mortars must also meet requirements on mechanical performance, their compressive strength, flexural strength, and dynamic Young's modulus were tested. The newly developed mortars composed of lava aggregate and hydrated lime or natural hydraulic lime met technical, functional, compatibility, and performance criteria on masonry and rendering materials, and were found well applicable for repair of historically valuable buildings. Currently, the majority of cell-based studies on neurodegeneration are carried out on two-dimensional cultured cells that do not represent the cells residing in the complex microenvironment of the brain. Recent evidence has suggested that three-dimensional (3D) in vitro microenvironments may better model key features of brain tissues in order to study molecular mechanisms at the base of neurodegeneration. So far, no drugs have been discovered to prevent or halt the progression of neurodegenerative disorders. New therapeutic interventions can come from phytochemicals that have a broad spectrum of biological activities. On this basis, we evaluated the neuroprotective effect of three phytochemicals (sulforaphane, epigallocatechin gallate, and plumbagin) alone or in combination, focusing on their ability to counteract oxidative stress. The combined treatment was found to be more effective than the single treatments. In particular, the combined treatment increased cell viability and reduced glutathione (GSH) levels, upregulated antioxidant enzymes and insulin-degrading enzymes, and downregulated nicotinamide adenine dinucleotide phosphate (NADPH) oxidase 1 and 2 in respect to peroxide-treated cells. Our data suggest that a combination of different phytochemicals could be more effective than a single compound in counteracting neurodegeneration, probably thanks to a pleiotropic mechanism of action. This study reviewed aspects of the biology of two members of the glucosinolate family, namely sinigrin and glucoraphanin and their anti-tumour and antimicrobial properties. Sinigrin and glucoraphanin are converted by the β-sulphoglucosidase myrosinase or the gut microbiota into their bioactive forms, allyl isothiocyanate (AITC) and sulphoraphanin (SFN) which constitute part of a sophisticated defence system plants developed over several hundred million years of evolution to protect them from parasitic attack from aphids, ticks, bacteria or nematodes. Delivery of these components from consumption of cruciferous vegetables rich in the glucosinolates also delivers many other members of the glucosinolate family so the dietary AITCs and SFN do not act in isolation. In vitro experiments with purified AITC and SFN have demonstrated their therapeutic utility as antimicrobials against a range of clinically important bacteria and fungi. AITC and SFN are as potent as Vancomycin in the treatment of bacteria listed by the World Health Organisation as antibiotic-resistant "priority pathogens" and also act as anti-cancer agents through the induction of phase II antioxidant enzymes which inactivate potential carcinogens. Glucosinolates may be useful in the treatment of biofilms formed on medical implants and catheters by problematic pathogenic bacteria such as Pseudomonas aeruginosa and Staphylococcus aureus and are potent antimicrobials against a range of clinically important bacteria and fungi. The glucosinolates have also been applied in the prevention of bacterial and fungal spoilage of food products in advanced atmospheric packaging technology which improves the shelf-life of these products. Crucifer vegetables, Brassicaceae and other species of the order Brassicales, e.g., Moringaceae that are commonly consumed as spice and food, have been reported to have potential benefits for the treatment and prevention of several health disorders. Though epidemiologically inconclusive, investigations have shown that consumption of those vegetables may result in reducing and preventing the risks associated with neurodegenerative disease development and may also exert other biological protections in humans. The neuroprotective effects of these vegetables have been ascribed to their secondary metabolites, glucosinolates (GLs), and their related hydrolytic products, isothiocyanates (ITCs) that are largely investigated for their various medicinal effects. Extensive pre-clinical studies have revealed more than a few molecular mechanisms of action elucidating multiple biological effects of GLs hydrolytic products. This review summarizes the most significant and up-to-date in vitro and in vivo neuroprotective actions of sulforaphane (SFN), moringin (MG), phenethyl isothiocyanate (PEITC), 6-(methylsulfinyl) hexyl isothiocyanate (6-MSITC) and erucin (ER) in neurodegenerative diseases. Oxidative stress is considered to be critically involved in the normal aging process but also in the development and progression of various human pathologies like cardiovascular and neurodegenerative diseases, as well as of infections and malignant tumors. These pathological conditions involve an overwhelming production of reactive oxygen species (ROS), which are released as part of an anti-proliferative strategy during pro-inflammatory immune responses. Moreover, ROS themselves are autocrine forward regulators of the immune response. Most of the beneficial effects of antioxidants are considered to derive from their influence on the immune system. Due to their antioxidant and/or radical scavenging nature, phytochemicals, botanicals and herbal preparations can be of great importance to prevent oxidation processes and to counteract the activation of redox-regulated signaling pathways. Antioxidants can antagonize the activation of T-cells and macrophages during the immune response and this anti-inflammatory activity could be of utmost importance for the treatment of above-mentioned disorders and for the development of immunotolerance. Herein, we provide an overview of in vitro assays for the measurement of antioxidant and antiinflammatory activities of plant-derived substances and extracts, by discussing possibilities and limitations of these methods. To determine the capacity of antioxidants, the oxygen radical absorbance capacity (ORAC) assay and the cell-based antioxidant activity (CAA) assay are widely applied. To examine the influence of compounds on the human immune response more closely, the model of mitogen stimulated human peripheral blood mononuclear (PBMC) cells can be applied, and the production of the inflammatory marker neopterin as well as the breakdown of the amino acid tryptophan in culture supernatants can be used as readout to indicate an immunomodulatory potential of the tested compound. These two biomarkers of immune system activation are robust and correlate with the course of cardiovascular, neurodegenerative and malignant tumor diseases, but also with the normal aging process, and they are strongly predictive. Thus, while the simpler ORAC and CAA assays provide insight into one peculiar chemical aspect, namely the neutralization of peroxyl radicals, the more complex PBMC assay is closer to the in vivo conditions as the assay comprehensively enlights several properties of immunomodulatory test compounds.
