Over the past decades, toxic concentrations of metals accumulated in soils by human activities, such as mining and industrial processes. In plants, metals interfere with essential physiological processes, like photosynthesis, transpiration and respiration, resulting in growth retardation and chlorosis. At the cellular level, metal exposure leads to an increased production of reactive oxygen species (ROS), thereby disturbing the cellular redox status and causing oxidative damage to cellular components. However, ROS can also act as signalling molecules in normal cell metabolism as well as in defence responses against biotic and abiotic stresses. In this way, ROS can interact with different signalling pathways in the cell, one of them being mitogen-activated protein kinase (MAPK) cascades. MAPK signalling pathways relay stress signals from the cell membrane to the nucleus by phosphorylation and activation of transcription factors, resulting in transcriptional control of stress-responsive genes. Although MAPK cascades are associated with copper (Cu) and cadmium (Cd) stress signalling, knowledge about their specific functioning in metal stress responses is rather scarce. To understand plant responses to toxic concentrations of metals, it is necessary to gain insight in the specific stress signalling pathways operating at the molecular level thereby regulating cellular responses to these metals. Therefore, the main objective of this work was to unravel MAPK cascades controlling metal stress responses. For the metal treatment, two metals (Cu and Cd) with distinct characteristics (essential versus non-essential, redox-active versus non-redoxactive) were used. In conclusion, the results obtained in this work are indicative of an essential role for MAPK cascades in metal-induced oxidative stress responses. Depending on the chemical properties of the applied metal, the same MAPK cascade is able to regulate different metal-specific transcription factors resulting in metal-specific stress responses. Knowledge about the molecular mechanisms controlling metal stress responses is essential to understand how plants cope with elevated metal concentrations at the physiological level. De afgelopen decennia hebben er in de bodem toxische concentraties aan metalen geaccumuleerd als gevolg van humane activiteiten, zoals mijnbouw en industrie. In planten interfereren metalen met essentiële fysiologische processen, zoals fotosynthese, transpiratie en respiratie, met dwerggroei en chlorose tot gevolg en aldus een verminderde biomassaproductie. Op cellulair niveau induceert blootstelling aan metalen een verhoogde productie van reactieve zuurstofvormen (ROS), waardoor de cellulaire redox status verstoord wordt en oxidatieve schade aan cellulaire componenten kan ontstaan. Daarnaast hebben ROS ook een belangrijke functie als signaalmolecule zowel in het normale celmetabolisme alsook in verdedigingsresponsen tegen biotische en abiotische stressors. Op deze manier kunnen ROS interageren met verschillende signaaltransductie pathways in de cel, waaronder de ‗mitogen-activated protein kinase‘ (MAPK) cascades. MAPK pathways geleiden stress signalen van het celmembraan naar de nucleus door fosforylatie en activatie van transcriptiefactoren, die de transcriptie van stressresponsieve genen controleren. Alhoewel MAPK-cascades worden geassocieerd met koper (Cu) en cadmium (Cd) geïnduceerde signaaltransductie, is er maar weinig geweten over hun specifieke functie in metaalgeïnduceerde responsen. Om te kunnen begrijpen hoe planten reageren op toxische metaalconcentraties in hun omgeving, is het noodzakelijk om meer inzicht te krijgen in de specifieke stress signaaltransductie pathways op moleculair niveau die aan de basis liggen van de fysiologische responsen op deze metalen. Daarom was het belangrijkste doel van dit werk om de MAPK-cascades, die de metaalstress responsen controleren, te ontrafelen. Hiervoor werden twee metalen (Cu en Cd) met verschillende eigenschappen (essentieel versus niet-essentieel, redox-actief versus niet redox-actief) gebruikt. Alles tesamen tonen de resultaten, bekomen in dit werk, aan dat MAPK-cascades een essentiële rol spelen in metaalgeïnduceerde oxidatieve stress responsen. Afhankelijk van de chemische eigenschappen van het toegediende metaal, is een bepaalde MAPK-cascade in staat verschillende metaalspecifieke transcriptiefactoren te reguleren, resulterend in metaalspecifieke stress responsen. Kennis over de moleculaire mechanismen, die metaalstress responsen controleren, is essentieel om te begrijpen hoe planten op fysiologisch niveau omgaan met verhoogde metaalconcentraties.

Sinds het einde van de 19e eeuw en het begin van de 20e eeuw werden zware metalen wijd verspreid in de lucht, de bodem en het water als gevolg van antropogene activiteiten. Door opwaaiend stof en accumulatie in landbouwproducten kunnen ze opgenomen worden door mens en dier. Zware metalen zijn zeer toxisch en verstoren verschillende fysiologische processen bij zowel mens en dier (long-, nier-, lever- en botschade) als bij planten (verstoring van de groei en fotosynthese). Op cellulair niveau is een algemeen effect waarneembaar, namelijk oxidatieve stress. Dit is een onevenwicht in de redoxbalans waarbij het aantal geoxideerde substraten in de cel groter is dan het aantal gereduceerde. Deze geoxideerde moleculen kunnen reageren met proteïnen en DNA in de cel. Ze kunnen ook aanleiding geven tot lipidenperoxidatie waardoor de celmembranen worden aangetast. Als bescherming tegen de reactie van deze geoxideerde moleculen, activeert de cel haar antioxidatief systeem. Dit bestaat uit enzymen (catalase, superoxidedismutase, …) en metabolieten (glutathion, ascorbaat). In deze studie werd er gekeken naar het effect van enkele zware metalen, Cu en Cd, op oxidatieve stress gerelateerde parameters in de cel. Als een testorganisme werd gebruik gemaakt van Arabidopsis thaliana omdat voor deze plant een grote beschikbaarheid bestaat van moleculaire informatie en technieken. Na behandeling werd een nutriëntenprofiel opgesteld en werd het gehalte aan lipidenperoxidatieproducten en H2O2 bepaald. De expressie van ROS-producerende enzymen zoals de lipoxygenasen en NADPH-oxidasen werd ook nagegaan. Verder werden de effecten van zware metalen op het antioxidatieve verdedigingsmechanisme onderzocht op zowel transcriptioneel als metabolisch niveau. Uit deze studie kan besloten worden dat blootstelling aan zware metalen, zoals Cu en Cd, leidt tot oxidatieve stress op cellulair niveau. Dit kan afgeleid worden uit de stijging in H2O2 en lipidenperoxidatie na toediening van Cu en Cd. De mechanismen die hiertoe leiden zijn echter verschillend voor beide metalen. Cu is een redoxactief metaal en kan via redoxcycling tussen zijn verschillende oxidatievormen hydroxylradicalen vormen, die vervolgens aanleiding kunnen geven tot oxidatieve stress. Bij Cd is het ontstaan van oxidatieve stress eerder een indirect mechanisme te wijten aan een verhoogde activiteit van NADPH-oxidasen en lipoxygenasen. Verder wordt het antioxidatief systeem aangetast op zowel transcriptioneel als metabolisch niveau.

The reactive oxygen species (ROS)-signaling pathway is very important in heavy metal toxicity. Induction of the antioxidative defense mechanism, comprising ROS-scavenging enzymes and metabolites, in plants after environmental uranium contamination has been insufficiently studied in the past. This study aimed to analyze oxidative stress related responses in Arabidopsis thaliana after uranium exposure. Seventeen-day-old seedlings were exposed to 0, 0.1, 1, 10 and 100 μM uranium for 3 days. After exposure to 100 μM uranium, a decrease in fresh weight for leaves and roots was observed, leaves colored anthocyanous and roots were stunted and yellow. To reveal the importance of oxidative stress in uranium toxicity, alterations in ROS-scavenging enzymes were studied at protein and transcriptional level. Superoxide dismutase (SOD) capacities increased in leaves and roots after exposure to 100 μM uranium but no differences were observed for catalase (CAT) capacities. Transcript levels of different SODs located at various cellular compartments were affected depending on the place of action. Gene expression of CAT in leaves and roots was also affected after uranium exposure. Results indicate that oxidative stress plays an important role in uranium toxicity but suggest that plant responses differ for leaves and roots.
