Stroke is the primary cause of disability without effective rehabilitation methods. Emerging brain-machine interfaces offer promise for regulating brain neural circuits and promoting the recovery of brain function disorders. Implantable probes play key roles in brain-machine interfaces, which are subject to two irreconcilable tradeoffs between conductivity and modulus match/transparency. In this work, mechanically interlocked polyrotaxane is incorporated into topological hydrogels to solve the two tradeoffs at the molecular level through the pulley effect of polyrotaxane. The unique performance of the topological hydrogels enables them to acquire brain neural information and conduct neuromodulation. The probe is capable of continuously recording local field potentials for eight weeks. Optogenetic neuromodulation in the primary motor cortex to regulate brain neural circuits and control limb behavior is realized using the probe. Most importantly, optogenetic neuromodulation is conducted using the probe, which effectively reduces the infarct regions of the brain tissue and promotes locomotor function recovery. The synthesis of chiral mechanically interlocked molecules has attracted a lot of attention in the last few years, with applications in different fields, such as asymmetric catalysis or sensing. Herein we describe the synthesis of orientational mechanostereoisomers, which include a benzylic amide macrocycle with a stereogenic center, and nonsymmetric N-(arylmethyl)fumaramides as the axis. The base-promoted cyclization of the initial fumaramide thread allows enantioenriched value-added compounds, such as lactams of different ring sizes and amino acids, to be obtained. The chiral information is effectively transmitted across the mechanical bond from the encircling ring to the interlocked lactam. A series of dynamic metalloporphyrin [2]rotaxane molecular shuttles comprising of bis-functionalised Zn(II) porphyrin axle and pyridyl functionalised macrocycle components are prepared in high yield via active metal template synthetic methodology. Extensive variable temperature 1 H NMR and quantitative UV-Vis spectroscopic titration studies demonstrate dynamic macrocycle translocation is governed by an inter-component co-ordination interaction between the macrocycle pyridyl and axle Zn(II) metalloporphyrin, which serves to bias a 'resting state' co-conformation. The dynamic shuttling behaviour of the interlocked structures is dramatically inhibited by the addition of a neutral Lewis base such as pyridine, but can also be tuned via post-synthetic rotaxane demetallation of the porphyrin axle core to give free-base, or upon subsequent metallation, Ni(II) [2]rotaxane analogues. Importantly, the Lewis acidic Zn(II) porphyrin axle component is also capable of coordinating anions which induces mechanical bond shuttling behaviour resulting in a novel optical sensing response., (© 2023 The Authors. Anion receptors employing two distinct sensory mechanisms are rare. Herein, we report the first examples of halogen-bonding porphyrin BODIPY [2]rotaxanes capable of both fluorescent and redox electrochemical sensing of anions. 1 H NMR, UV/visible and electrochemical studies revealed rotaxane axle triazole group coordination to the zinc(II) metalloporphyrin-containing macrocycle component, serves to preorganise the rotaxane binding cavity and dramatically enhances anion binding affinities. The efficacy of many drugs can be limited by undesirable properties, such as poor aqueous solubility, low bioavailability, and "off-target" interactions. To combat this, various drug carriers have been investigated to enhance the pharmacological profile of therapeutic agents. In this work, we demonstrate the use of mechanical protection to "cage" a DNA-targeting metallodrug within a photodegradable rotaxane. More specifically, we report the synthesis of rotaxanes incorporating as a stoppering unit a known G-quadruplex DNA binder, namely a Pt II -salphen complex. This compound cannot interact with DNA when it is part of the mechanically interlocked assembly. The second rotaxane stopper can be cleaved by either light or an esterase, releasing the Pt II -salphen complex. This system shows enhanced cell permeability and limited cytotoxicity within osteosarcoma cells compared to the free drug. Tough and self-healable substrates can enable stretchable electronics long service life. However, for substrates, it still remains a challenge to achieve both high toughness and autonomous self-healing ability at room temperature. Herein, a strategy by using the combined effects between quadruple H-bonding and slidable cross-links is proposed to solve the above issues in the elastomer. The elastomer exhibits high toughness (77.3 MJ m -3 ), fracture energy (≈127.2 kJ m -2 ), and good healing efficiency (91 %) at room temperature. The superior performance is ascribed to the inter and intra crosslinking structures of quadruple H-bonding and polyrotaxanes in the dual crosslinking system. Strain-induced crystallization of PEG in polyrotaxanes also contributes to the high fracture energy of the elastomers. Furthermore, based on the dual cross-linked supramolecular elastomer, a highly stretchable and self-healable electrode containing liquid metal is also fabricated, retaining resistance stability (0.16-0.26 Ω) even at the strain of 1600 %., (© 2023 Wiley-VCH GmbH.)

Polyrotaxane is a supramolecular assembly consisting of multiple cyclic molecules threaded by a linear polymer. One of the unique properties of polyrotaxane is molecular mobility, cyclic molecules moving along the linear polymer. Molecular mobility of polyrotaxane surfaces affects cell spreading, differentiation, and other cell-related aspects through changing subcellular localization of yes-associated proteins (YAPs). Subcellular YAP localization is also related to cell senescence derived from oxidative stress, which is known to cause cancer, diabetes, and heart disease. Herein, the effects of polyrotaxane surface molecular mobility on subcellular YAP localization and cell senescence following H 2 O 2 -induced oxidative stress are evaluated in human mesenchymal stem cells (HMSCs) cultured on polyrotaxane surfaces with different molecular mobilities. Oxidative stress promotes cytoplasmic YAP localization in HMSCs on high-mobility polyrotaxane surfaces; however, low-mobility polyrotaxane surfaces more effectively maintain nuclear YAP localization, exhibiting lower senescence-associated β-galactosidase activity and senescence-related gene expression and DNA damage than that seen with the high-mobility surfaces. These results suggest that the molecular mobility of polyrotaxane surfaces regulates subcellular YAP localization, thereby protecting HMSCs from oxidative stress-induced cell senescence. The interlocking of ring and axle molecular components in rotaxanes provides a way to combine chromophoric, electron-donor and electron-acceptor moieties in the same molecular entity, in order to reproduce the features of photosynthetic reaction centers. To this aim, the photoinduced electron transfer processes involving a 1,8-naphthalimide chromophore, embedded in several rotaxane-based dyads, were investigated by steady-state and time-resolved absorption and luminescence spectroscopic experiments in the 300 fs-10 ns time window. Different rotaxanes built around the dialkylammonium/ dibenzo[24]crown-8 ether supramolecular motif were designed and synthesized to decipher the relevance of key structural factors, such as the chemical deactivation of the ammonium-crown ether recognition, the presence of a secondary site for the ring along the axle, and the covalent functionalization of the macrocycle with a phenothiazine electron donor. A [2]rotaxane built around a multi-responsive bis-acridinium macrocycle has been synthesized. Structural investigation has confirmed the interlocked nature of the molecule, and MD simulations illuminated its conformational dynamics with atomic resolution. Both halochromic and redox-switching properties were explored to shed light on the mechanical response and electronic changes that occur in the bis-acridinium [2]rotaxane. A [2]rotaxane-based molecular shuttle with an acid-responsive asymmetric macrocycle on a symmetric dumbbell axle is reported. Upon adding TFA, the macrocycle, namely the amine naphthotube, is protonated and translocates from the di(quaternary ammonium) station to the triazole stations because of electrostatic repulsion and weakened binding. The shuttling kinetics are slow due to the steric hindrance caused by the ethyl group on the quaternary ammonium center and can be followed by 1 H NMR spectroscopy. Interestingly, it was found that the shuttling kinetics depends on the concentration of TFA. A kinetic intermediate was detected and can even be captured in the presence of a high concentration of TFA. Surface properties of biomaterials affect the morphologies and inflammatory responses of macrophages. Recently, biomaterial design utilizing these properties has been explored to build a scaffold for balancing the immune system in vivo. In the present study, polyrotaxane surfaces with different functional groups including methyl, amino, and sulfo groups are utilized to clarify the effect of molecular mobility and zeta potential of these surfaces on RAW264.7 macrophage responses. At 24 h post-seeding, the majority of the cells adhere onto each surface, and the initial spreading is suppressed by more negatively-charged polyrotaxane surfaces. From 24 to 48 h of incubation, the spreading areas on the unmodified and methylated surfaces significantly increase, whereas those on the aminated and sulfonated surfaces remain unchanged. These results suggest that the initially cellular spreading process depends on the zeta potential, while the subsequent spreading process is governed by the molecular mobility. After lipopolysaccharide stimulation, the less mobile surfaces induce higher expression of inflammation-related genes than highly mobile surfaces, suggesting that molecular mobility is the main factor modulating the inflammatory activity in macrophages. Cyclodextrin (CD)-based polyrotaxanes (PR) are widely used to construct high-mechanical-performance materials because of the high degree of conformational freedom. However, strong hydrogen bonds between CDs greatly limit the application of CD-PR in the preparation of ductile neutral hydrogels. In this work, spiropyrane (SP) into α-CD-based PR is introduced to "visualize" the segment motion of the network in neutral water. The aggregation-induced cohesion and critical factors for the force transmission are disclosed. Mechanically interlocked molecules present opportunities to construct therapeutic drugs and diagnostic imaging agents but harnessing supramolecular chemistry to make biologically active probes in water is a challenge. Here, we describe a rotaxane-based approach to synthesise radiolabelled proteins and peptides for molecular imaging of cancer biomarkers in vivo. Host-guest chemistry using β-cyclodextrin- and cucurbit[6]uril-catalysed cooperative capture synthesis produced gallium-68 or zirconium-89 radiolabelled metallo[4]rotaxanes. Photochemical conjugation to trastuzumab led to a viable positron emission tomography (PET) radiotracer. The rotaxane architecture can be tuned to accommodate different radiometal ion complexes, other protein- or peptide-based drugs, and fluorophores for optical detection. Spatial distance is an important factor in controlling the functional interactions between molecular units in a conjugate; therefore, the bridging unit has been closely examined. Here, we examined the effect of the flexibility of bridging alkyl chains on the proximity of stacked porphyrin and phthalocyanine conjugated with a fourfold rotaxane linkage. We found that closely stacking two π systems requires bridging alkyl chains above a certain length, and the shorter bridges hinder stacking because of their lower flexibility. The stacking distance between porphyrin and phthalocyanine in the conjugate with decyl (C 10 ) chains was estimated to be 4.03 Å and showed a unique physical character arising from short-distance interactions. The longer alkyl chains minimized steric restriction inside the fourfold rotaxane and allowed efficient communication between the porphyrin and phthalocyanine units. We report the synthesis of two [2]rotaxanes containing an interlocked three dimensional binding cavity formed from a pyridinium bis(amide) axle component containing two phenol donors, and an isophthalamide based macrocycle. In the competitive solvent mixture 1 : 1 CDCl 3  : CD 3 OD, one of the receptors exhibits a much higher selectivity preference for chloride than an analogous rotaxane without the hydroxy groups. X-ray crystal structures reveal the chloride anion guest encapsulated within the interlocked binding cavity, though not all of the hydrogen bond donors are utilised. Computational semi-empirical simulations indicate that secondary intermolecular interactions occur between the axle hydroxy hydrogen bond donors and the [2]rotaxane macrocycle components, contributing to a more preorganised binding pocket, which may be responsible for the observed enhanced selectivity., (© 2022 The Authors. The demand for stretchable electronics with a broader working range is increasing for wide application in wearable sensors and e-skin. However, stretchable conductors based on soft elastomers always exhibit low working range due to the inhomogeneous breakage of the conductive network when stretched. Here, a highly stretchable and self-healable conductor is reported by adopting polyrotaxane and disulfide bonds into the binding layer. The binding layer (PR-SS) builds the bridge between polymer substrates (PU-SS) and silver nanowires (AgNWs). The incorporation of sliding molecules endows the stretchable conductor with a long sensing range (190%) due to the energy dissipation derived from the sliding nature of polyrotaxanes, which is two times higher than the working range (93%) of conductors based on AP-SS without polyrotaxanes. Furthermore, the mechanism of sliding effect for the polyrotaxanes in the elastomers is investigated by SEM for morphological change of AgNWs, in situ small-angle x-ray scattering, as well as stress relaxation experiments. This paper reports the synthesis and study of new pH-sensitive DB24C8-based [2]rotaxane molecular shuttles that contain within their axle four potential sites of interaction for the DB24C8: ammonium, amine, Weinreb amide, and ketone. In the protonated state, the DB24C8 lay around the best ammonium site. After either deprotonation or deprotonation-then-carbamoylation of the ammonium, different localizations of the DB24C8 were seen, depending on both the number and nature of the secondary stations and steric restriction. Unexpectedly, the results indicated that the Weinreb amide was not a proper secondary molecular station for the DB24C8. Nevertheless, through its methoxy side chain, it slowed down the shuttling of the macrocycle along the threaded axle, thereby partitioning the [2]rotaxane into two translational isomers on the NMR timescale. The restriction of local molecular motions is critical for improving the fluorescence quantum yields (FQYs) and the photostability of fluorescent dyes. Herein, we report a supramolecular approach to enhance the performance of fluorescent dyes by incorporating a linked rotaxane structure with permethylated α-cyclodextrins. Tetraphenylethylene (TPE) derivatives generally exhibit low FQYs in solution due to the molecular motions in the excited state. We show that TPE with linked rotaxane structures on two sides displays up to 15-fold higher FQYs. Detailed investigations with variable temperature 1 H NMR, UV-Vis, and photoluminescence spectroscopy revealed that the linked rotaxane structure rigidifies the TPE moiety and thus suppresses the local molecular motions and non-radiative decay. Moreover, the linked rotaxane structure enhances the FQY of the dye in various solvents, including aqueous solutions, and improves the photostability through the inhibition of local molecular motions in the excited TPE., (© 2022 Wiley-VCH GmbH.)

A linear double pyridinium-terminated thread comprising a central chalcone moiety is shown to provide two independent binding sites with similar affinity for cucurbit[7]uril (CB7) macrocycles in water as judged from NMR, UV-Visible and fluorescence spectroscopies. Association results in [2] and [3]pseudorotaxanes, which are both pH and photosensitive. Switching from the neutral chalcone to the cationic flavylium form upon irradiation at 365 nm under acidic conditions provided an enhanced CB7 association (K 1:1 increases from 1.2×10 5  M -1 to 1.5×10 8  M -1 ), limiting spontaneous on-thread cucurbituril shuttling. This co-conformational change in the [2]pseudorotaxane is reversible in the dark with k obs =4.1×10 -4  s -1 . Threading the flavylium moiety into CB7 leads to a dramatic increase in the fluorescence quantum yield, from 0.29 in the free axle to 0.97 in the [2]pseudorotaxane and 1.0 in the [3]pseudorotaxane., (© 2021 Wiley-VCH GmbH.)

Solvent-mediated crystal-to-crystal transformations of [Au 6 Ag 3 Cu 3 (H 2 O) 3 (d-pen) 6 (tdme) 2 ] 3+ (d-[1(H 2 O) 3 ] 3+ ; pen 2- =penicillaminate, tdme=1,1,1-tris(diphenylphosphinomethyl)ethane) to form unique supramolecular species are reported. Soaking crystals of d-[1(H 2 O) 3 ] 3+ in aqueous Na 2 bdc (bdc 2- =1,4-benzenedicarboxylate) yielded crystals containing d-[1(bdc)(H 2 O) 2 ] + due to the replacement of a terminal aqua ligand in d-[1(H 2 O) 3 ] 3+ by a monodentate bdc 2- ligand. When γ-cyclodextrin (γ-CD) was added to aqueous Na 2 bdc, d-[1(H 2 O) 3 ] 3+ was transformed to d-[1(bdc@γ-CD)(H 2 O) 2 ] + , where a γ-CD ring was threaded by a bdc 2- molecule to construct a pseudorotaxane structure. While the use of dicarboxylates with an aliphatic carbon chain instead of bdc 2- afforded analogous pseudorotaxanes, such pseudorotaxane species were not formed when crystals of [Au 6 Ag 3 Cu 3 (H 2 O) 3 (l-pen) 6 (tdme) 2 ] 3+ (l-[1(H 2 O) 3 ] 3+ ) enantiomeric to d-[1(H 2 O) 3 ] 3+ were soaked in aqueous Na 2 bdc and γ-CD, affording only crystals containing l-[1(bdc)(H 2 O) 2 ] + ., (© 2021 Wiley-VCH GmbH.)

The lasso peptide benenodin-1, a naturally occurring and bacterially produced [1]rotaxane, undergoes a reversible zip tie-like motion under heat activation, in which a peptidic wheel stepwise translates along a molecular thread in a cascade of "tail/loop pulling" equilibria. Conformational and structural analyses of four translational isomers, in solution and in the gas phase, reveal that the equilibrium distribution is controlled by mechanical and non-covalent forces within the lasso peptide. The development of artificial nanoscale motors that can use energy from a source to perform tasks requires systems capable of performing directionally controlled molecular movements and operating away from chemical equilibrium. Here, the design, synthesis and properties of pseudorotaxanes are described, in which a photon input triggers the unidirectional motion of a macrocyclic ring with respect to a non-symmetric molecular axle. The photoinduced energy ratcheting at the basis of the pumping mechanism is validated by measuring the relevant thermodynamic and kinetic parameters. Owing to the photochemical behavior of the azobenzene moiety embedded in the axle, the pump can repeat its operation cycle autonomously under continuous illumination. NMR spectroscopy was used to observe the dissipative non-equilibrium state generated in situ by light irradiation. We also show that fine changes in the axle structure lead to an improvement in the performance of the motor. Such results highlight the modularity and versatility of this minimalist pump design, which provides facile access to dynamic systems that operate under photoinduced non-equilibrium regimes., (© 2021 The Authors. Tensile forces influence a variety of important biological processes and force sensors are required to study these processes in vivo. Current force sensors are often tailor-made for a specific application, or activate at much higher forces than those observed at the cellular or tissue level. A versatile force sensor, with tunable mechanical and optical properties, activated at low pN forces will be ideal. The development of integrated systems that mimic the multi-stage stiffness change of marine animals such as the sea cucumber requires the design of molecularly tailored structures. Herein, we used an integrated biomimicry design to fabricate a sea cucumber mimic using sidechain polypseudorotaxanes with tunable nano-to-macroscale properties. A series of polyethylene glycol (PEG)-based sidechain copolymers were synthesized to form sidechain polypseudorotaxanes with α-cyclodextrins (α-CDs). By tailoring the copolymers' molecular weights and their PEG grafting densities, we rationally tuned the sizes of the formed polypseudorotaxanes crystalline domain and the physical crosslinking density of the hydrogels, which facilitated 3D printing and the mechanical adaptability to these hydrogels. After 3D printing and photo-crosslinking, the obtained hydrogels exhibited large tensile strain and broad elastic-to-plastic variations upon α-CD (de)threading. Incorporation of rotaxane scaffolds into the crosslinking points of polymer networks significantly affects their rheological and mechanical properties. The present study involves the synthesis of a new rotaxane-type crosslinker containing nitrile N-oxide functional groups on both the axle and wheel components. The prepared crosslinker is highly reactive; however, it can be isolated and applied in the crosslinking reaction of a commercially important polymer, namely ethylene-propylene-butadiene monomer rubber (EPDM), in the absence of additives and catalysts. Tensile tests reveal that compared to a network containing conventional crosslinking points, both breaking strength and strain of the network structure prepared herein are improved due to the incorporation of movable crosslinking points. The synthesized network structure also exhibits five times higher fracture energy. The fascinating pulley effect from moveable α-cyclodextrin (α-CD) based polyrotaxane favors toughening of hydrogels, but the strategy is rarely applied in bulk polymers because of the severe aggregation trend of α-CDs. Herein, the authors propose a simple approach to moderately modify the α-CDs of polyrotaxane by introducing large steric side groups and reactive CC so as to minimize the unwanted hydrogen bonds-induced aggregation of α-CDs and hydrophilicity of polyrotaxane. Accordingly, the proof-of-concept material, poly(methyl acrylate) crosslinked by the modified polyrotaxane, turns out to be rather homogeneous with optical transparency. The polyrotaxane crosslinks are movable under external force as disclosed by in situ small-angle X-ray scattering and other techniques, which is correlated to the relative amount of α-CDs. A few polyrotaxane crosslinkers prove to be sufficient to simultaneously improve strength and toughness of poly(methyl acrylate) owing to the stress equalization. A nanoring-rotaxane supramolecular assembly with a Cy7 cyanine dye (hexamethylindotricarbocyanine) threaded along the axis of the nanoring was synthesized as a model for the energy transfer between the light-harvesting complex LH1 and the reaction center in purple bacteria photosynthesis. The complex displays efficient energy transfer from the central cyanine dye to the surrounding zinc porphyrin nanoring. We present a theoretical model that reproduces the absorption spectrum of the nanoring and quantifies the excitonic coupling between the nanoring and the central dye, thereby explaining the efficient energy transfer and demonstrating similarity with structurally related natural light-harvesting systems., (© 2020 The Authors. Stimuli-responsive materials have received considerable attention for their application in biomedical fields. Herein, the temperature-dependent phase transition behavior of acetylated polyrotaxanes (Ac-PRXs) consisting of acetylated cyclodextrins threaded onto a linear axle polymer in aqueous solution is investigated. The aqueous solutions of Ac-PRXs exhibit temperature-dependent transmittance changes when the degree of acetylation is 30-40%. Upon increasing the temperature, Ac-PRXs are dehydrated and acetyl groups are subsequently associated through hydrophobic interactions. Interestingly, the Ac-PRXs form coacervate droplets with a diameter of ≈2-3 µm above their cloud points. These temperature-responsivities of Ac-PRXs are observed for other PRXs with different axle polymer molecular weights and compositions. Diphenanthrioctaphyrin(1.1.1.0.1.1.1.0), an expanded carbaporphyrinoid incorporating two phenanthrenylene moieties, exists as two separate, yet interconvertible, locked stereoisomers. These species demonstrate complex dynamic behavior upon protonation, consisting in multiple conformational rearrangements and anion-binding events. The formation of one of the final dicationic forms is accompanied by the inclusion of a complex anion(s) within the macrocyclic cavity yielding a pseudorotaxane-like host-guest complex. Protonation with trifluoroacetic or dichloroacetic acids followed by neutralization afforded a conformation-switching cycle, which involves six structurally different species. Analogous acidification with chiral 10-camphorsulfonic acid and subsequent neutralization generated one of the free base stereoisomers with enantiomeric excess. Polyrotaxanes, consisting of poly(ethylene glycol) and α-cyclodextrins, are mechanically interlocked supermolecules. The structure allows α-cyclodextrins to move along the polymer, referred to as molecular mobility. Here, polyrotaxane-based triblock copolymers, composed of polyrotaxanes with different degrees of methylation and poly(benzyl methacrylate) at both terminals, are coated on culture surfaces to fabricate dynamic biointerfaces for myocyte differentiation. The molecular mobility increases with the degree of methylation and the contact angle hysteresis of water droplets and air bubbles. When the mouse myoblast cell line C2C12 is cultured on methylated polyrotaxane surfaces, the expression levels of myogenesis-related genes, myogenin (Myog) and myosin heavy chain (Myhc) are altered by the degree of methylation. Polyrotaxane surfaces with intermediate degrees of methylation promote the highest expression levels among all the surfaces. [3]Rotaxanes, which consist of one macrocyclic phenanthroline compound and two axle components, were prepared by the oxidative dimerization of an alkyne compound with bulky tris[4'-cyclohexyl-(1,1'-biphenyl)-4-yl]methyl blocking group. The catalytic activity of a macrocyclic phenanthroline-Cu complex was utilized to thread the two axle components inside the ring. The alkyne compound with chain of 15 or 20 methylene groups gave [3]rotaxanes in high yields, whereas the axle with a chain of six methylene groups afforded a [3]rotaxane in very poor yield. We also examined the effect of the ring size on the synthesis of [3]rotaxanes. The use of natural compounds to construct biomaterials, including delivery system, is an attractive strategy. In the present study, through threading functional α-cyclodextrins onto the conjugated macromolecules of poly(ethylene glycol) (PEG) and natural compound bile acid, glycopolymers of polyrotaxanes with the active targeting ability are obtained. These glycopolymers self-assemble into micelles as evidenced by dynamic light scattering and transmission electron microscopy, in which glucosamine, as an example of targeting groups, is introduced. These micelles after loading doxorubicin (DOX) exhibit the selective recognition with cancer cells 4T1. Meanwhile, the maximal half inhibitory concentration is determined to be ≈2.5 mg L -1 for the DOX-loaded micelles, close to the value of free DOX·HCl (1.9 mg L -1 ). The cumulative release of DOX at pH 5.5 is faster than at pH 7.4, which may be used as the controlled release system. This drug delivery system assembled by glycopolymers features high drug loading of DOX, superior biocompatibility. The strategy not only utilizes the micellization induced by bile acids, but also overcomes the major limitation of PEG such as the lack of targeting groups. Reversible covalent bonds yield polymeric materials with functional characteristics such as self-healing, shape memory, stress relaxation, and stimuli-responsiveness. Here, photo-reversibly cappable polyrotaxanes are designed and the on-off controlled dissociation of their supramolecular architectures is demonstrated. The polyrotaxanes are synthesized by capping dithiobenzoates at both terminals of polyethylene glycol threaded through multiple α-cyclodextrins. Since dethreading of the α-cyclodextrins is prevented by the dithiobenzoate stoppers, the supramolecular dissociation is induced by their photo-cleavage. Subsequently, the cleaved dithiobenzoates spontaneously re-cap the polyrotaxane terminals in darkness. Thus, the supramolecular dissociation can be modulated by photo-reversible capping of the dithiobenzoate stoppers. Mechanically interlocked supramolecular assemblies are appealing building blocks for creating functional nanodevices. Herein, we describe the multistep assembly of large DNA origami rotaxanes that are capable of programmable structural switching. We validated the topology and structural integrity of these rotaxanes by analyzing the intermediate and final products of various assembly routes by electrophoresis and electron microscopy. We further analyzed two structure-switching behaviors of our rotaxanes, which are both mediated by DNA hybridization. In the first mechanism, the translational motion of the macrocycle can be triggered or halted at either terminus. We report the stepwise assembly of supramolecular daisy chain rotaxanes (DCR) made of double-stranded DNA: Small dsDNA macrocycles bearing an axle assemble into a pseudo-DCR precursor that was connected to rigid DNA stoppers to form DCR with the macrocycles hybridized to the axles. In presence of release oligodeoxynucleotides (rODNs), the macrocycles are released from their respective hybridization sites on the axles, leading to stable mechanically interlocked DCRs. Besides the expected threaded DCRs, certain amounts of externally hybridized structures were observed, which dissociate into dumbbell structures in presence of rODNs. We show that the genuine DCRs have significantly higher degrees of freedom in their movement along the thread axle than the hybridized DCR precursors. Interlocking of DNA in DCRs might serve as a versatile principle for constructing functional DNA nanostructures where the movement of the subunits is restricted within precisely confined tolerance ranges., (© 2016 The Authors. Native α-cyclodextrin- (α-CD) and permethylated α-CD (PMeCD)-based rotaxanes with various short alkylene chains as axles can be synthesized through a urea end-capping method. Native α-CD tends to form [3]- or [5]pseudorotaxanes and not [2]- or [4]pseudorotaxanes, which indicates that the coupled CDs act as a single fragment. End-capping reactions of the pseudorotaxanes with C18 and C24 axle lengths do not occur because the axle termini are covered by the densely stacked CDs. The number of PMeCDs on the pseudorotaxane is flexible and mainly depends on the axle length. Peracetylated α-CD (PAcCD)-based rotaxanes are synthesized through O-acetylation of the α-CD-based rotaxanes without any decomposition of the rotaxanated structures. The structures of PMeCD-based [3]- and [4]rotaxanes, and the molecular dynamics calculations on [3]pseudorotaxanes, indicate that the tail face of PMeCDs is regularly directed toward the axle termini. A new, versatile chloride-anion-templating synthetic pathway is exploited for the preparation of a series of eight new [2]rotaxane host molecules, including the first sulfonamide interlocked system. (1)H NMR spectroscopic titration investigations demonstrate the rotaxanes' capability to selectively recognise the chloride anion in competitive aqueous solvent media. The interlocked host's halide binding affinity can be further enhanced and tuned through the attachment of electron-withdrawing substituents and by increasing its positive charge. A dicationic rotaxane selectively binds chloride in 35% water, wherein no evidence of oxoanion binding is observed. NMR spectroscopy, X-ray structural analysis and computational molecular dynamics simulations are used to account for rotaxane formation yields, anion binding strengths and selectivity trends.

Two Janus [2]rotaxanes, 5a and 5b, with α-cyclodextrin (α-CD) derivatives substituted on the 6-position with two recognition sites (azobenzene and heptamethylene (C7)) that were linked with linkers of different lengths (oligo(ethylene glycol) with a degree of polymerization equal to 2 or approximately 21) were synthesized and characterized. 2D ROESY NMR spectroscopy and circular dichroism (cd) spectra demonstrated that the recognition site of the α-CD moiety was switched by photoisomerization of the azobenzene moiety in 5a and 5b. The different size changes of 5a and 5b in hydrodynamic radius (R(H)) owing to the different length of linker between two recognition sites were observed by pulse-field-gradient spin-echo NMR spectroscopy. The kinetic results indicated that the different length of linker had no or a weak effect for the photoisomerization process of 5a and 5b.

We have developed a "threading-followed-by-swelling" protocol to synthesize [2]rotaxanes efficiently and atom economically. Our protocol employs cis-1-[(Z)-alk-1'-enyl]-2-vinylcyclopropane units as the termini of the threadlike components; these end groups are converted into more-sizable cycloheptadiene motifs, which function as stopper units, through Cope rearrangements at elevated temperature. We used this approach to synthesize [2]rotaxanes in good yield from [2]pseudorotaxanes featuring either one or two swellable termini to interlock three different types of macrocycle. The chiral centers created by the swelling process were "erased" by hydrogenating the cycloheptadiene termini into the corresponding cycloheptane units, affording achiral molecular [2]rotaxanes as the only final products.

A broadly applicable chemical cleavage methodology to facilitate MS/MS sequencing was developed for macrocyclic and lasso peptides, which hold promise as exciting new therapeutics. Existing methods such as Edman degradation, CNBr cleavage, and enzymatic digestion are either limited in scope or completely fail in cleavage of constrained nonribosomal peptides. This paper describes a novel, highly efficient approach to the self-assembly of monomacrocycles and two-layered capsules by using dynamic covalent chemistry. Intramolecular hydrogen-bonding was used to preorganize aromatic amide-based monomers that contain aldehyde and tert-butoxycarbonylamino units. As a result, in the presence of an excess of trifluoroacetic acid (TFA), four monomers could self-couple to produce macrocycles selectively through the formation of three imine or hydrazone bonds. Three dipodal precursors were also prepared by connecting two hydrogen-bonded segments with a flexible linker. In the presence of TFA, these precursors could also self-couple, leading to the exclusive formation of two-layered capsules. As a result of intramolecular hydrogen-bonding, all the macrocycles and capsules were stable in solution and could be purified by simple recrystallization. The new capsules were able to form complexes with linear propylenediammonium derivatives to give unique two-layered pseudo[3]rotaxanes.

A series of water-soluble [3]rotaxanes-(n+2) and [3]pseudorotaxanes-(n+2) with short (propyl, n=1) and long (dodecyl, n=10) aliphatic spacers have been prepared in high yields by a 1,3-dipolar cycloaddition reaction catalyzed by cucurbit[6]uril (CB6). The pH-triggered dethreading and rethreading of CB6 on these pseudorotaxanes was monitored by 1H NMR spectroscopy. A previously reported [3]rotaxane-12 that is known to behave as a bistable molecular switch has two recognition sites for CB6, that is, the diaminotriazole moieties and the dodecyl spacer. By changing the pH of the system, it is possible to observe more than one state in the shuttling process. At low pH values both CB6 units are located on the diaminotriazole moieties owing to an ion-dipole interaction, whereas at high pH values both of the CB6 units are located on the hydrophobic dodecyl spacer. Surprisingly, the CB6 units shuttle back to their initial state very slowly after reprotonation of the axle. Even after eighteen days at room temperature, only about 50 % of the CB6 units had relocated back onto the diaminotriazole moieties. The rate constants for the shuttling processes were measured as a function of temperature over the range from 313 to 333 K and the activation parameters (enthalpy, entropy, and free energy) were calculated by using the Eyring equation. The results indicate that this [3]rotaxane behaves as a kinetically controlled molecular switch. The switching properties of [3]rotaxane-3 have also been studied. However, even under extreme pH conditions this rotaxane has not shown any switching action, which confirms that the propyl spacer is too short to accommodate CB6 units.

The anion-templated synthesis of three novel halogen-bonding 5-halo-1,2,3-triazolium axle containing [2]rotaxanes is described, and the effects of altering the nature of the halogen-bond donor atom together with the degree of inter-component preorganisation on the anion-recognition properties of the interlocked host investigated. The ability of the bromotriazolium motif to direct the halide-anion-templated assembly of interpenetrated [2]pseudorotaxanes was studied initially; bromide was found to be the most effective template. As a consequence, bromide anion templation was used to synthesise the first bromotriazolium axle containing [2]rotaxane, the anion-binding properties of which, determined by (1) H NMR spectroscopic titration experiments, revealed enhanced bromide and iodide recognition relative to a hydrogen-bonding protic triazolium rotaxane analogue. Two halogen-bonding [2]rotaxanes with bromo- and iodotriazolium motifs integrated into shortened axles designed to increase inter-component preorganisation were also synthesised. Anion (1) H NMR spectroscopic titration experiments demonstrated that these rotaxanes were able to bind halide anions even more strongly, with the iodotriazolium axle integrated rotaxane capable of recognising halides in aqueous solvent media. Synthetic receptors selective for target peptides or proteins have received attention because of their potential applications in the separation of biomolecules and biomedical diagnostics. Herein, a [2]rotaxane-based functional monomer containing monocarboxylated α-cyclodextrin (α-CD) was synthesized, and its crosslinked polymers were evaluated to determine their binding ability to a model peptide, angiotensin III (Arg-Val-Tyr-Ile-His-Pro-Phe), containing an arginine (Arg) residue. The binding ability of the resulting polymers toward angiotensin III, angiotensin IV (Val-Tyr-Ile-His-Pro-Phe), and FMRF-amide (Phe-Met-Arg-Phe) was examined by the batch-binding assay and compared with that of control polymers, in which maleic acid-introduced α-CD was chemically crosslinked. The results suggest that the [2]rotaxane-based functional monomer in the crosslinked polymer contributes to the high affinity toward angiotensin III.
