2. Adipokinetic hormone enhances CarE‐mediated chlorpyrifos resistance in the brown planthopper, Nilaparvata lugens.

Adipokinetic hormone (AKH), the principal stress‐responsive neurohormone in insects, has been implicated in insect responses to insecticides. However, the functionality of AKH and its mode of signalling in insecticide resistance are unknown. Herein, we demonstrated that the enhanced activity of carboxylesterases (CarEs) is involved in the chlorpyrifos resistance in Nilaparvata lugens [brown planthopper (BPH)]. Chlorpyrifos exposure significantly induced the expression of AKH and its receptor AKHR in the susceptible BPH (Sus), and these two AKH signalling genes were over‐expressed in the chlorpyrifos‐resistant strain (Res) compared to Sus. RNA interference (RNAi) against AKH or AKHR decreased the CarE activity and suppressed the BPH's resistance to chlorpyrifos in Res. Conversely, AKH peptide injection elevated the CarE activity and enhanced the BPH's survival against chlorpyrifos in Sus. Furthermore, five CarE genes were identified to be positively affected by the AKH pathway using RNAi and AKH injection. Among these CarE genes, CarE and Esterase E4‐1 were found to be over‐expressed in Res compared to Sus, and knockdown of either gene decreased the BPH's resistance to chlorpyrifos. In conclusion, AKH plays a role in enhancing chlorpyrifos resistance in the BPH through positive influence on the expression of CarE genes and CarE enzyme activity. [ABSTRACT FROM AUTHOR]

An optimal control strategy for the random vibration reduction of nonlinear structures using piezoelectric stack inertial actuator is proposed. First, the dynamic model of the nonlinear structure considering the dynamics of a piezoelectric stack inertial actuator is established, and the motion equation of the coupled system is described by a quasi-non-integrable-Hamiltonian system. Then, using the stochastic averaging method, this quasi-non-integrable-Hamiltonian system is reduced to a one-dimensional averaged system for total energy. The optimal control law is determined by establishing and solving the dynamic programming equation. The proposed control law is analytical and can be fully executed by a piezoelectric stack inertial actuator. The responses of optimally controlled and uncontrolled systems are obtained by solving the Fokker–Planck–Kolmogorov (FPK) equation to evaluate the control effectiveness of the proposed strategy. Numerical results show that our proposed control strategy is effective for random vibration reduction of the nonlinear structures using piezoelectric stack inertial actuator, and the theoretical method is verified by comparing with the simulation results. Background: Vagus nerve stimulation (VNS) is an emerging electroceutical therapy for remedying gastric disorders that are poorly managed by pharmacological treatments and/or dietary changes. Such therapy seems promising as the vagovagal neurocircuitry modulates the enteric nervous system to influence gastric functions. Methods: Here, the modulatory effects of left cervical VNS on gastric emptying in rats were quantified using a (i) feeding protocol in which the animal voluntarily consumed a postfast, gadolinium‐labeled meal and (ii) a non‐invasive imaging method to measure antral motility, pyloric activity and gastric emptying based on contrast‐enhanced magnetic resonance imaging (MRI) and computer‐assisted image processing pipelines. Key Results: Vagus nerve stimulation significantly accelerated gastric emptying (sham vs VNS: 29.1% ± 1.5% vs 40.7% ± 3.9% of meal emptied per 4 hours), caused a greater relaxation of the pyloric sphincter (sham vs VNS: 1.5 ± 0.1 vs 2.6 ± 0.4 mm2 cross‐sectional area of lumen), and increased antral contraction amplitude (sham vs VNS: 23.3% ± 3.0% vs 32.5% ± 3.0% occlusion), peristaltic velocity (sham vs VNS: 0.50 ± 0.02 vs 0.67 ± 0.03 mm s−1), but not its contraction frequency (sham vs VNS: 6.1 ± 0.2 vs 6.4 ± 0.2 contractions per minute, P = .22). The degree to which VNS relaxed the pylorus was positively correlated with gastric emptying rate (r = .5887, P < .001). Conclusions & Inferences: The MRI protocol employed in this study is expected to enable advanced preclinical studies to understand stomach pathophysiology and its therapeutics. 5. Comparison of Different Perovskite Cathodes in Solid Oxide Fuel Cells.

Abstract: This study compares the electrochemical performance of three perovskite cathode materials, La
0.6 Sr0.4 Co0.2 Fe0.8 O3 (LSCF), Ba0.5 Sr0.5 Co0.2 Fe0.8 O3 (BSCF), and Sm0.5 Sr0.5 Co0.2 Fe0.8 O3 (SSCF), at different operating temperatures, in order to provide optimal operating condition and performance for different solid oxide fuel cells. Among these three cathodes, BSCF has the highest power density of 39 mW cm−2 at 600 °C, 88 mW cm−2 at 650 °C, and 168 mW cm−2 at 700 °C; LSCF has the highest power density of 263 mW cm−2 at 750 °C and 456 mW cm−2 at 800 °C. The cathode overpotentials have the same trend as the power densities. Activation energies for the total cathode area specific resistance (ASR) are calculated to be 0.44 eV, 0.38 eV, and 0.52 eV for the LSCF, BSCF, and SSCF cathodes, respectively. Stability testing of 100 h shows that the open circuit voltages of the LSCF and BSCF cathodes drop 16.1% and 22.9% at 800 °C, respectively, as well as 15.3% and 11.3% at 600 °C, respectively, indicating that the LSCF cathode is more stable at 800 °C while the BSCF cathode is more stable at 600 °C. Background and purpose Physical activity is associated with a reduced incidence of first-time stroke. However, few studies have examined the effect of pre-stroke physical activity on post-stroke complications and clinical outcomes. Methods A total of 39 835 cases of stroke registered in the nationwide stroke registry system of Taiwan between 2006 and 2009 were analyzed according to five levels of severity as determined by National Institutes of Health Stroke Scale score upon hospital admission. Pre-stroke physical activity was defined in the Taiwan Stroke Registry as dedicated leisure-time physical activity for at least 30 min/day for 3 days/week for more than 6 months. A Cox model was used to compare complications and outcomes between active and inactive groups. Results The active and inactive groups were similar in age distribution and stroke type distribution, but the active group had better National Institutes of Health Stroke Scale scores upon admission. The active group also had significantly fewer post-stroke complications. Active patients had lower hospital mortality and better functional outcomes upon discharge as per the modified Rankin Scale. Improved functional status in the active group was significant at 1, 3 and 6 months post-stroke. Conclusion Dedicated leisure-time physical activity for at least 30 min/day, at least three times per week for more than 6 months was associated with decreased stroke severity, fewer post-stroke complications, lower mortality and better outcomes. Until now, there are no completely effective parasite-specific pharmaceuticals or immunotherapies for treatment against the zoonotic cryptosporidiosis. Sushi domain (CpSushi) is an important functional domain in Cryptosporidium parvum putative rhoptry protein-1 (Cp PRP1), which is the only reported C. parvum rhoptry protein and may play key role in the course of invasion. Here, a 708-bp fragment encoding the CpSushi domain was amplified and expressed in E. coli. Immunofluorescence detection showed that CpSushi was located on the surface of C. parvum oocysts and the apical pole to the sporozoites that belonged to the position of rhoptry. Three-week-old female ICR mice were used for detecting the immunoreactions and immunoprotection of recombinant CpSushi (rCpSushi) to artificial C. tyzzeri infection. The results indicated that a significant increase of anti-CpSushi antibody response was induced by the recombinant protein. Compared to blank, Tris- EDTA ( TE) buffer and adjuvant controls mice, rCpSushi-immunized mice produced specific spleen cell proliferation as well as enhanced IL4, IL5, IL12p70 and TNF-α production in vitro. The reduction rate of parasites shedding in stool in mice immunized with rCpSushi was 68.91% after challenging with C. tyzzeri. These results suggest that CpSushi could be a new promising cryptosporidiosis vaccine candidate antigen composition. [ABSTRACT FROM AUTHOR]

An 8-week feeding trial was performed to investigate effects of dietary selenium (sodium selenite, Na
2 SeO3 ; selenium nanoparticle, Nano-Se; selenium yeast, Se-yeast) on the growth, selenium status, antioxidant activities, muscle composition and meat quality of blunt snout bream. Na2 SeO3 and Nano-Se were supplemented at 0.2 mg Se kg−1 , and Se-yeast was supplemented at 0.1, 0.2, 0.4 and 0.8 mg Se kg−1 in basal diet; no Se was added as a control. The results indicated that groups of 0.2 and 0.4 mg kg−1 Se-yeast had significantly higher weight gain and Nano-Se, 0.2, 0.4 mg kg−1 Se-yeast had significantly lower feed conversation ratio compared with the control group. The Se concentrations of whole body, muscle and liver linearly increased with increasing dietary Se-yeast levels. Group of 0.4 mg kg−1 Se-yeast significantly increased activities of catalase and glutathione peroxidase. Muscle colour of Nano-Se, 0.2, 0.4 mg kg−1 Se-yeast groups, the water-holding capacity of 0.4, 0.8 mg kg−1 Se-yeast groups showed significant differences compared with the control group. The results suggest that Se-yeast and Nano-Se had a better growth performance than Na2 SeO3 at 0.2 mg Se kg−1 , and supplementing appropriate Se-yeast in diet can increase antioxidant activities and enhance meat quality of blunt snout bream. A droplet micro-reactor was developed to realize the internal sol-gel reaction of ZrO
2 inside the droplet. The reactants are only allowed to combine when the droplet forms, meanwhile, they are able to mix well and react to form gel particles with a proper designed structure of the micro-reactor. In this condition, the mixing efficiency was determined by the structure of the micro-reactor and the flow rate ratio of the two components. After sintering, ZrO2 ceramic microspheres are acquired with high sphericity and narrow size distribution. This droplet micro-reactor overcomes the gelation of the premixed sol and reactant in the channel, and it has been demonstrated to be more stable with a longer running time. A simple and cost-effective method was used for the fabrication of BaZrO
3 ( BZ) fiber. The crystallization of BZ fiber was a simultaneous process with the decomposition of Ba CO3 which was formed during the pyrolysis process of the precursor fiber. The surface cracks of the fiber induced by the organic decomposition converted to grain boundaries during the grain growth at high temperatures. A simple model was used to describe the evolution mechanism of the transformation from surface cracks to grain boundaries. This work reports the precise diameter control of electrospun yttria-stabilized zirconia ( YSZ) nanofibers from 200 to 900 nm after calcination. Fabricated YSZ nanofibers showed porous nanocrystalline structures with high aspect ratios of more than 500:1 and high surface-to-volume ratios with a specific surface area of 43.32 m
2 /g. The diameter of the YSZ nanofibers increased with the viscosity of the precursor solution, which was controlled by the concentrations of either polymers (polyacrylonitrile) or ceramic precursors ( YSZ). We present a modified correlation between the diameter of a nanofiber and the synthetic conditions, as the observed behavior for calcined ceramic nanofibers deviated from the expected behavior. Our results demonstrate a modified but simple approach to fabricate ceramic nanofibers with desired diameters, providing a new design guideline for many electrochemical applications. This study was conducted to investigate the effects of fructooligosaccharide ( FOS) and Bacillus licheniformis ( B. licheniformis) on growth performance, body composition, intestinal enzymes activities and gut histology of Megalobrama terminalis. Nine experimental diets were formulated to contain three FOS levels (0, 3 and 6 g kg
−1 ) and three B. licheniformis levels (0, 1 and 5 × 107 CFU g−1 ) following a 3 × 3 factorial design. Accordingly, diets were named as 0/0, 0/3, 0/6, 1/0, 1/3, 1/6, 5/0, 5/3 and 5/6 ( B. licheniformis/ FOS). At the end of the 8-week feeding trial, weight gain ( WG) and specific growth rate ( SGR) of fish fed 6 g kg−1 FOS were both significantly ( P < 0.01 and P < 0.05) higher than that of the other groups in terms of dietary FOS levels. Besides, WG and SGR of fish fed 1 × 107 CFU g−1 B. licheniformis were significantly ( P < 0.05) higher than that of the control group in terms of dietary B. licheniformis levels. In addition, a significant interaction ( P < 0.05) between dietary FOS and B. licheniformis was observed in finial weight, WG, SGR as well as the survival rate with the highest values all observed in fish fed diet 1/3. Hepatosomatic index, carcass lipid content, lipase activities as well as microvilli length increased significantly ( P < 0.05) from 0 to 1 × 107 CFU g−1 , but no significant difference ( P > 0.05) was observed in terms of dietary FOS levels. In addition, a significant ( P < 0.05) interaction of FOS and B. licheniformis was observed in both protease and Na+ , K+ - ATPase activities with the highest value obtained in fish fed diet 1/3. The results indicated that the dietary applications of dietary FOS and B. licheniformis alone or in combination can significantly improve the growth performance, survival rate, intestinal enzymes activities as well as microvilli length of triangular bream. In addition, there is a significant interaction between dietary FOS and B. licheniformis. The best combination for this species is 3 g kg−1 FOS with 1 × 107 CFU g−1 B. licheniformis. Neurocysticercosis, an infection of the central nervous system with the larval stage of the cestode Taenia solium, is common in developing countries but its occurrence and management in allogeneic hematopoietic stem cell transplantation ( HSCT) has not been reported previously, to our knowledge. We report the case of an immigrant female patient who underwent a matched-related allogeneic HSCT for acute lymphoblastic leukemia and was incidentally found to have a solitary viable neurocysticercosis lesion. However, despite severe immunosuppression, the size of the cyst did not increase. More importantly, restoration of the immune system did not induce significant inflammation or seizures. Subsequent follow-up demonstrated complete resolution of the neurocysticercosis lesion. Thus, in the setting of HSCT, an asymptomatic patient with a single neurocysticercosis lesion was successfully managed without the use of anthelmintics, steroids, or anti-epileptics. Contents This study was undertaken to investigate differences in protein expression between high- and low-motility sperm of swamp buffalo. The research used two-dimensional gel electrophoresis (2 DE) coupled to matrix-assisted laser desorption/ionization time-of-flight tandem mass spectrometry ( MALDI- TOF/ TOF- MS) to analyse the different proteins. The results showed 18 different expression protein spots between high- and low-motility buffalo sperm; eight of these proteins were up-regulated in low-motility sperm, five were down-regulated, one deleted and four proteins specifically expressed. Finally, four proteins were successfully identified by MS as belonging to three unique proteins; they are outer dense fibre of sperm tails protein 2 ( ODF2), ATP synthase subunit alpha ( ATP5A1) and succinyl-CoA synthetase subunit beta ( SUCLG2). In summary, these results help to develop an understanding of the molecular mechanisms associated with low-motility sperm and provide clues for finding molecular markers associated with sperm motility. 0.8 Sr0.2 MnO3 (LSM821) and La0.6 Sr0.4 Co0.2 Fe0.8 O3 (LSCF6428) cathodes at 800 °C by investigating the interfacial reaction and degradation through an AISI 441 interconnect/ LSM821 (LSCF6428) electrode/yttria-stabilized zirconia (YSZ) electrolyte tri-layer structure. Under the same processing condition, the grain size of the LSCF6428 cathode is smaller than that of the LSM821 cathode. Ohmic resistance and polarization resistance of the cathodes are analyzed by deconvoluting the electrochemical impedance spectroscopy (EIS) results. The LSCF6428 cathode has much smaller resistance than the LSM821 cathode. Moisture produces a larger effect on the ohmic resistance and polarization resistance of the LSM821 cathode than on those of the LSCF6428 cathode. More chromium diffuses from the interconnect to the cathode for both LSM821 and LSCF6428 electrodes thermally treated in moist air. Based on the structure, elemental distribution, and EIS analysis, the interaction mechanisms between the electrodes and the AISI 441 alloy interconnect are proposed. [ABSTRACT FROM AUTHOR]- Published

The chemical interaction study of AO-SiO
2 -B2 O3 -Y2 O3 (A = Ba, Sr) (BaY, SrY) glass with high-temperature electrolyte yttria-stabilized zirconia (YSZ 8 mol%) is reported as a function of different heat treatment durations. The as-prepared glass with 10 mol% of yttria shows limited amount of crystallization at 800 °C. Due to this yttria-based glasses BaY and SrY have been chosen to make diffusion couples with high-temperature electrolyte and interconnect material. These diffusion couples have been heat treated at 850 °C, for 100, 200, and 500 h. The heat-treated diffusion couples have been characterized using X-ray diffraction (XRD), scanning electron microscopy (SEM), and energy dispersive spectroscopy (EDS). Microstructural analysis of diffusion couples shows absence of any undesired oxides and detrimental reaction products at the interface. The glass has shown good bonding characteristics and absence of cracks, pores, or any kind of delamination from YSZ. Apart from this, SrY and BaY glass seals have also shown good adhesion characteristics with Crofer 22 APU, even after 500 h at 850 °C. Background The association between meat consumption and the risk of hepatocellular carcinoma ( HCC) is still inconclusive. Aim To conduct a systematic review and meta-analysis to quantitatively assess the relationship between meat intake and the risk of HCC. Methods We searched the PubMed, Web of Science and EMBASE databases for relevant studies published before July 2013. The summary relative risks were pooled using the fixed-effects model when no substantial heterogeneity was detected, otherwise, the random-effects model was used. Heterogeneity and publication bias were also analysed. Results Finally, seven cohort studies and 10 case-control studies were included. The pooled relative risks ( RRs) of HCC for the highest vs. lowest consumption levels were 1.10 (95% confidence interval, CI: 0.85-1.42) for red meat, 1.01 (95% CI: 0.79-1.28) for processed meat and 0.97 (95% CI: 0.85-1.11) for total meat. Moreover, white meat and fish consumption were found to be inversely associated with HCC risk, the summary RRs were 0.69 (95% CI: 0.58-0.81) and 0.78 (95% CI: 0.67-0.90) respectively, and the results remained quite stable after stratification by the confounding factors. Conclusions The present meta-analysis indicates that a high level of white meat or fish consumption can reduce the risk of HCC significantly, while intake of red meat, processed meat or total meat is not associated with HCC risk. Our findings suggest that dietary intervention may be a promising approach for prevention of HCC, which still need to be confirmed by further well-designed prospective studies and experimental research. We examined the pre-emptive analgesic effect of a cyclooxygenase (COX)-2 inhibitor in a rat surgical pain model and characterised the changes in cutaneous COX-2 around a surgical site. Thermal hyperalgesia and mechanical allodynia were tested in the rats for three days after incision and skin tissues were collected for analysis of COX-2. There was decreased expression of cutaneous COX-2 one day after surgical incision. Pre-incision injection of the COX-2 inhibitor significantly inhibited expression of COX-2 and also reduced thermal hyperalgesia (but not mechanical allodynia) compared with the post-incision COX-2-inhibitor injection group, one day after incision. The purpose of this study is to investigate the possible underlying pathogenesis of erectile dysfunction(ED) in young men with low risk of coronary heart disease and no well-known aetiology. To conduct this study, 122 patients with ED under the age of 40 were enrolled, along with 33 age-matched normal control subjects. The patients with ED had significantly higher levels of systolic blood pressure (SBP), total cholesterol and triglyceride, high sensitivity C-reactive protein (hs-CRP), greater carotid intima-media thickness (CIMT) and Framingham risk score (FRS) than the control group, though all of these values were within the respective normal range. Further, the brachial artery flow- mediated vasodilation (FMD) values were significantly lower in ED patients and correlated positively with the severity of ED ( r = 0.714, p < 0.001). When these significant factors were studied in the multivariate logistic regression model, FMD, SBP, hs-CRP and FRS remained the statistical significance. The receiver-operating characteristic (ROC) analysis demonstrated that FMD had a high ability to predict ED in young male with low FRS [area under the curve (AUC) 0.921, p < 0.001]. The cutoff value of FMD <10.25% had sensitivity of 82.8% and specificity of 100% for diagnosis of ED. FRS and hs- CRP were also proven to be predictors of ED (AUC 0.812, p < 0.001; AUC 0.645, p = 0.011, respectively). The results of this study validated that subclinical endothelial dysfunction and low-grade inflammation may be the underlying pathogenesis of ED with no well-known aetiology. Young patients complaining of ED should be screened for cardiovascular risk factors and possible subclinical atherosclerosis. Measurement of FMD, hs-CRP and FRS can improve our ability to predict and treat ED, as well as subclinical cardiovascular disease early for young male. [ABSTRACT FROM AUTHOR]

Contents Mitochondria are energy-supplying organelles, whose distribution and functional integrity are necessary for cell survival and development. In this study, the mitochondrial distribution pattern and activity during buffalo oocyte in vitro maturation, fertilization and preimplantation embryo development were revealed using a fluorescent dye and confocal laser scanning microscopy. Distribution of active mitochondria changed during buffalo oocyte in vitro maturation. Active mitochondria were transferred from the outer to inner and perinuclear cytoplasm as oocytes matured in vitro and aggregated around the pronuclei in the fertilized eggs. Active mitochondria were also observed in preimplantation embryos. In the two-cell stage, they were distributed throughout the cytoplasm. From four-cell to the spherical embryonic stages, active mitochondria translocated to the perinuclear and the periphery of the cytoplasm. These results confirm that mitochondria play an important role in oocyte and embryo. The distribution of active mitochondria might be a marked feature of buffalo oocyte maturation, fertilization and preimplantation embryo development in vitro. [ABSTRACT FROM AUTHOR]

Background: Platelet-rich fibrin (PRF) prepared by Choukroun's technique is derived from an autogenous preparation of concentrated platelets without any manipulation. PRF was found to increase osteoblast growth and proliferation. However, the underlying mechanisms are not yet completely understood. This study aimed to determine the effects of PRF on cell attachment, proliferation, phosphorylated Akt, heat shock protein 47 (HSP47) and lysyl oxidase (LOX) expression on human osteoblasts. Methods: Blood collection was carried out from 10 healthy volunteers. Cell attachment and proliferation were measured by colorimetric assay with WST-1 and alamar blue in human osteoblast cell line U2OS cells, respectively. Western blot was employed to evaluate the expression of p-Akt, HSP47 and LOX. Results: PRF alone was found to stimulate U2OS cell attachment compared with untreated controls (p < 0.05). PRF was found to increase osteoblast proliferation during a 5-day incubation period (p < 0.05). PRF was found to increase Akt phosphorylation in a time-dependent manner (p < 0.05). Collagen-related proteins HSP47 and LOX were significantly elevated by stimulation with PRF compared with untreated controls (p < 0.05). Conclusions: It is suggested that PRF is capable of increasing osteoblast attachment, proliferation and simultaneously upregulating collagen-related protein production. Nevertheless, 90.96% of these patients are managed on maintenance haemodialysis (HD), with only 9.03% enrolled in a peritoneal dialysis (PD) programme. Aim: The study aim was to identify the factors affecting Taiwanese patient's selection of PD in preference to HD for chronic kidney disease. Methods: A cross-sectional research design was utilized with 130 chronic renal failure (CRF) patients purposively selected from outpatient nephrology clinics at four separate Taiwan hospitals. Logistic regression was used to identify the main factors affecting the patient's choice of dialysis type. Results: Single-factor logistic regression found significant differences in opinion related to age, education level, occupation type, disease characteristics, lifestyle modifications, self-care ability, know-how of dialysis modality, security considerations and findings related to the decisions made by medical personnel ( P < 0.05). Moreover, multinomial logistic regression after adjustment for interfering variables found that self-care ability and dialysis modality know-how were the two main factors affecting the person's selection of dialysis type. Conclusions: Self-care ability and the person's knowledge of the different types of dialysis modality and how they function were the major determinants for selection of dialysis type in Taiwan based on the results from this study. The results indicate that the education of CRF patients about the types of dialysis available is essential to enable them to understand the benefits or limitations of both types of dialysis. Background Although asthma and allergic rhinitis (AR) are considered to be one syndrome, many questions remain unanswered. Why do some AR patients develop asthma but others do not, and which factors play a role in the development of asthma that have so far not been clearly elucidated. Objective We hypothesize that children with AR who have the Clara cell secretory protein (CC16, secretoglobin 1A1) 38A/38A genotype (rs3741240) have an increased likelihood of developing asthma. Methods The study sample included 117 children, with AR, but no asthma diagnosed within the following 5 years, as the control group. Cases group (n = 202) included age- and gendermatched children with AR first, and asthma developed 3-5 years later, as the study group. The CC16 genotype was determined by PCR and Sau96I restriction digestion of PCR products. The serum CC16 levels were measured by ELISA. Total serum IgE, allergen specific IgE, eosinophil count and pulmonary function were also measured. Results In children with rhinitis who develop asthma, the frequencies of the AA genotype were significantly higher than those who did not develop asthma [odds ratio (OR) = 2.527; 95% confidence interval (CI) = 1.571-4.065; Po0.01]. Serum CC16 levels in the children with rhinitis who develop asthma and carry the AA genotype were significantly lower than those who carry the non-AA genotype and those who did not develop asthma. Conclusions and Clinical Relevance Results of this study suggest that CC16 38A/38A genotype plays a role in the development of early asthma in children with AR. The vitellogenin receptor (VgR) belongs to the low-density lipoprotein receptor (LDLR) superfamily, and is an important carrier for the uptake of vitellogenin (Vg) into developing oocytes of all oviparous species. The first full-length message for a VgR from a Lepidopteran insect was cloned and sequenced from the ovary of Spodoptera litura Fabricius (GenBank accession no. GU983858). The coding region consisted of 5370 bp flanked by a 49 bp 5′-untranslated region (UTR) and a 177 bp 3′-UTR, which encoded a 1798-residue protein with a predicted molecular weight (MW) of 201.69 kDa. S. litura VgR ( SlVgR)comprised two ligand binding sites with four LDLR class A repeats in the first domain and seven in the second domain, an epidermal growth factor-like domain containing an LDLR class B repeat and a YWXD motif, a transmembrane domain and a cytoplasmic domain. A phylogenetic relationship placed SlVgR as a separate group from the other insects. SlVgR messenger RNA (mRNA) was specifically expressed in the ovarian tissues. The developmental expression patterns showed that VgR mRNA was first transcribed in 6 day female pupae and the maximum level of VgR mRNA appeared in 36-h-old adults. Immunoblot analysis detected an ovary-specific VgR protein with a MW of ∼200 kDa, whose development profiles were consistent with VgR mRNA expression patterns. RNA inteference (RNAi) specifically disrupted the VgR gene by injection of 3 or 5 µg VgR double-stranded RNA per insect in 4 or 6 day pupae. RNAi of SlVgR led to a phenotype characterized by high Vg accumulation in the haemolymph, low Vg deposition in the ovary and the failure of insect spawning. These results mean that VgR is critical for binding Vg and transporting it into the oocytes of the insect ovary, thus playing an important role in insect reproduction. [ABSTRACT FROM AUTHOR]

Aim: To evaluate the efficacy and safety of taranabant in overweight and obese patients with type 2 diabetes mellitus (T2DM). Methods: This was a multicenter, double-blind, randomized, placebo-controlled study in overweight and obese patients with T2DM (ages ≥ 18 and ≤ 75 years) with a BMI ≥ 27 kg/m
2 and ≤ 43 kg/m2 and HbA1c ≥7.0 and ≤ 10.0%, who were either not on an antihyperglycaemic agent or on a stable dose of metformin (≥ 1500 mg/day). After a 2-week placebo run-in, patients were randomized to placebo (N = 156) or taranabant 0.5-mg (N = 155), 1-mg (N = 157), or 2-mg (N = 155) once daily for 52 weeks. Primary efficacy endpoints were changes from baseline in body weight (BW) and HbA1c at Week 36, with results at Week 52 being key secondary endpoints. Results: In the all-patients-treated population, using a last-observation-carried-forward analysis, reductions in BW were −2.5, −3.7, −4.5 and −5.1 kg at Week 36 and −2.4, −4.0, −4.6 and −5.3 kg at Week 52 in the placebo, 0.5-, 1- and 2-mg groups, respectively (all doses significant vs. placebo at both time points). The proportion of patients who lost ≥ 5 and ≥ 10% of their baseline BW was significantly greater in the 1- and 2-mg groups vs. placebo at Week 36 and all taranabant groups vs. placebo at Week 52. Reductions in HbA1c were −0.40, −0.47, −0.68 and −0.71% at Week 36 and −0.30, −0.43, −0.65 and −0.64% at Week 52, in the placebo, 0.5-, 1- and 2-mg groups, respectively (1- and 2-mg doses significant vs. placebo at both time points). After 52 weeks, the incidences of adverse experiences classified in the gastrointestinal (diarrhoea, nausea, vomiting), nervous system-related (dizziness, sensory-related), and psychiatric (irritability, depression-related) organ systems were numerically higher or statistically significantly higher in all taranabant groups compared with the placebo group. Conclusions: After 36 and 52 weeks, treatment with taranabant at the 1- and 2-mg doses led to clinically significant weight loss and improvement in glycaemic parameters in overweight and obese patients with T2DM that was associated with dose-related increases in adverse experiences. Based on these data and data from other Phase III clinical studies, it was determined that the overall safety and efficacy profile of taranabant did not support further development for the treatment of obesity. Eur J Clin Invest 2010; 40 (2): 164–171 Background The aim was to examine whether the inhibition of selective cyclooxygenase (COX) 2 activation could suppress the development of inflammatory reaction in visceral and subcutaneous abdominal fats of high-fat-induced obese rats. Materials and methods The rats were fed separately regular diet (CONT), high-fat diet ad libitum or energy-restrictedly (HFr) for 12 weeks. Rats fed high-fat diet ad libitum were further divided into those co-treated with vehicle (HFa), a selective COX2 inhibitor-celecoxib (HFa-Cel) or nimesulide (HFa-Nim). Oral glucose tolerance test (OGTT) was performed at the end of weeks 4, 8, 12. Another set of rats with similar grouping was divided into those with a 4-, 8- or 12-week intervention for tissue sampling. Results Body and epididymal fat weights were increased similarly in HFa, HFa-Cel and HFa-Nim. Time-dependent increases in plasma insulin, triglyceride, impaired OGTT shown in HFa were significantly reversed in HFa-Cel and HFa-Nim. The obese-linked increases in gene expressions of COX-2, monocyte chemoattractant protein-1 (MCP-1) and tumour necrosis factor-α (TNF-α) in epididymal and subcutaneous fats (especially in the former) were significantly suppressed in HFa-Cel and HFa-Nim. The protein contents of MCP-1 and TNF-α in epididymal fats were changed consistently with their gene expressions. Plasma MCP-1 was increased time-dependently in HFa and suppressed in HFa-Cel and HFa-Nim. The increased CD68 positive cells showed in both epididymal and subcutaneous fats of HFa were significantly attenuated in HFa-Cel and HFa-Nim. Grain-oriented polycrystalline alumina (PCA) ceramics with very high, nearly single-crystal transparency were successfully fabricated through colloidal processing in a high magnetic field followed by hot isostatic pressing. The c-axis of the produced material was oriented along the direction parallel to the magnetic field. The transmittance of the specimen with a thickness of 0.8 mm was 78% at a wavelength of 650 nm, close to the reported value of 84.5% for sapphire and more than 10% higher than the largest transmittance magnitude ever reported for PCA ceramics. In addition, the Na salt of 4,5-dihydroxy-1,3-benzenedisulfonic acid (named "Tiron") was found to be a highly effective dispersant for processing high-purity materials. [ABSTRACT FROM AUTHOR]

- Abstract
Hypertrophic scarring remains a major problem for patients who have suffered from surgeries or burns. Vascularization plays an important role in the early phase of hypertrophic scarring. Therefore, the inhibition of angiogenesis might be used as a preventive strategy. In this study, we assessed the effect of anti-angiogenesis resulting from adenovirus-mediated METH1 (metalloprotease and thrombospondin1) gene expression on the hypertrophic scar formation in a rabbit ear model of hypertrophic scarring. We first investigated the number of microvessel and microcirculatory perfusion in untreated scars on days 10, 30, 60, and 90 after epithelialization. Then, we examined the effect of anti-angiogenesis by adenovirus-mediated METH1 expression on hypertrophic scar formation by calculating the scar elevation index, counting the microvessel and argyrophilic nucleolar organizer region particle, and detecting the amount of collagen on days 30 and 60 after treatment. We found that untreated scar tissues at the proliferative phase (days 10-60 after epithelialization) had a significantly higher density of microvessel and microcirculatory perfusion than those at the mature phase (day 90 after epithelization) (both p<0.05). On days 30 and 60 after treatment, the hypertrophic scar formation was significantly inhibited in the treatment group. There was significantly reduced scar elevation index, microvessel count, number of argyrophilic nucleolar organizer region, and total collagen content for treated scars. Our results demonstrate that METH1 has a markedly inhibitive effect on the formation of hypertrophic scar, and may thus have a promising application in the prevention of human hyperthropic scars. Objectives: Islet-like clusters (ILCs), differentiated from human embryonic stem cells (hESCs), were characterized both before and after transplantation under the kidney capsule of streptozotocin-induced diabetic immuno-incompetent mice. Materials and methods: Multiple independent ILC preparations ( n = 8) were characterized by immunohistochemistry, flow cytometry and cell insulin content, with six preparations transplanted into diabetic mice ( n = 42), compared to controls, which were transplanted with either a human fibroblast cell line or undifferentiated hESCs ( n = 28). Results: Prior to transplantation, ILCs were immunoreactive for the islet hormones insulin, C-peptide and glucagon, and for the ductal epithelial marker cytokeratin-19. ILCs also had cellular insulin contents similar to or higher than human foetal islets. Expression of islet and pancreas-specific cell markers was maintained for 70 days post-transplantation. The mean survival of recipients was increased by transplanted ILCs as compared to transplanted human fibroblast cells ( P < 0.0001), or undifferentiated hESCs ( P < 0.042). Graft function was confirmed by secretion of human C-peptide in response to an oral bolus of glucose. Conclusions: hESC-derived ILC grafts continued to contain cells that were positive for islet endocrine hormones and were shown to be functional by their ability to secrete human C-peptide. Further enrichment and maturation of ILCs could lead to generation of a sufficient source of insulin-producing cells for transplantation into patients with type 1 diabetes. [ABSTRACT FROM AUTHOR]

CONTEXT: Current studies have found limited evidence for an association between doctor emotional intelligence (EI) and the patient-doctor relationship (PDR). This study explored the associations among doctor EI, patient trust and the PDR using multi-source and multi-level approaches. METHODS: A total of 994 outpatients and 39 doctors representing 11 specialties were surveyed. RESULTS: Doctors' self-rated EI was not significantly correlated with any variables rated by the patients. The nurse-rated PDR and the EI score for the doctor were positively associated with patient trust at a significant level. CONCLUSIONS: Multi-sources for assessment of doctor EI may be more objective and predictive than doctor self-ratings in ascertaining the associations among patient trust, the PDR, and patient satisfaction. Emotional intelligence coaching for doctors and interdisciplinary collaboration among clinicians are needed to optimise the efficient and therapeutic function of the PDR for patients. Hepatitis B virus core (HBc) particles, self-assemble into capsid particles and are extremely immunogenic, hold promise as an immune-enhancing vaccine carrier for heterologous antigens. However, formation of virus-like particles (VLP) can be restricted by size and structure of heterlogous antigens. In the study, we investigated formation of VLP by modified HBc fused with specified foot-and-mouth disease virus (FMDV) multiepitopes and evaluated their immune effects. Firstly, three HBc display vectors (pHBc1, pHBc2 and pHBc3) were constructed by deletions of different lengths within the HBc c/e1 region: 75–78 amino acid (aa), 75–80 aa and 75–82 aa respectively. Secondly, we inserted different compositions of FMDV multiepitopes, BT [VP1(141–160)–VP4(21–40)] and BTB [VP1(141–160)–VP4(21–40)–VP1(141–160)], into modified regions. As a result, only plasmid pHBc3-BTB of six recombinant vectors was expressed as soluble protein, which resulted in the formation of complete VLP confirmed by electron microscopy. Recombinant VLP could be taken up by cells and presented in vitro and in vivo. Furthermore, the modified VLP displayed a significantly stronger immunogenicity than other five recombinant proteins and GST-BTB with a higher titer of peptide-specific and virus-specific antibody, elevated IFN- γ and interleukin-4 production, especially enhanced lymphocyte proliferation. The results encourage further work towards the development of FMDV vaccines using hepatitis B virus core particles fused with FMDV epitopes. [ABSTRACT FROM AUTHOR]

A novel receptor guanylyl cyclase (GC) has been identified from the oriental fruit fly Bactrocera dorsalis (Hendel) and has been designated BdmGC-1. Protein domain analysis revealed that BdmGC-1 possesses a characteristic domain organization similar to all known receptor GCs but with a unique carboxyl-terminal extension. When overexpressed in 293T cells, BdmGC-1 manifests as a cell-surface glycoprotein with a marked cGMP-generating activity but is unresponsive to all ligands known to activate mammalian receptor GCs. BdmGC-1 mRNAs were highly expressed during development but had low or no expression in adult tissues. On the basis of its unique sequence and distinct developmental expression pattern, BdmGC-1 represents a novel receptor GC that may play a critical role during the development of B. dorsalis. Abstract: Paclitaxel has been found to exhibit cytotoxic and antitumor activity. There is little information regarding the mechanisms of apoptotic-inducing effect of paclitaxel on human osteogenic sarcoma U-2 OS cells. Several key regulatory proteins are involved in the initiation of apoptosis. Caspase-3 plays a direct role in proteolytic cleavage of cellular proteins responsible for progression to apoptosis. We examined the effect of paclitaxel on the cell cycle arrest and apoptosis in U-2 OS cells using flow cytometric analysis and Western blotting. We also measured the inhibition of paclitaxel-induced apoptosis and the caspase-3 activity by the broad-spectrum caspase inhibitor z-VAD-fmk on U-2 OS cells. The increased levels of casapse-3 were also confirmed by cDNA microarray. Our observations were: (1) paclitaxel treatment resulted in G2/M-cycle arrest in U-2 OS cells; (2) time and dose dependent apoptosis of U-2 OS cells was induced by paclitaxel; (3) in U-2 OS cells, z-VAD-fmk blocked the paclitaxel-induced apoptosis and caspase-3 activation. These results suggest that paclitaxel-induced G2/M-cycle arrest of the G2/M phase and apoptosis via a caspase-3 pathway in U-2 OS cells. [Copyright &y& Elsevier]

This study was conducted to find out the group B streptococcus colonisation of pregnant women in Kocaeli, Turkey. A culture plus individualised high-risk-based antibiotic prophylaxis was compared with high-risk-based approach alone. The screening of women was performed via vaginal and anal cultures for group B streptococcus (GBS). The maternal GBS colonisation rate was found to be 6.5%. All colonised women or preterm labours with unavailable culture results until delivery received prophylactic antibiotics. Neonatal colonisation rate and early-onset neonatal sepsis due to GBS was 1/200. The unscreened 900 women received prophylactic antibiotics due to a risk factor-based approach. The neonatal colonisation rate was 17/900 (p = 0.1), and the rate of early-onset neonatal sepsis was 3/900 (p = 0.6). A culture plus individualised high-risk-based antibiotic prophylaxis provided an insignificant change in neonatal colonisation and early-onset neonatal sepsis with GBS when compared with high-risk-based approach alone. The aim of this study was to investigate the pharmacokinetics of desflurane uptake into the brain and body by comparing desflurane concentrations in internal jugular-bulb blood (Jdes), arterial blood (Ades) and pulmonary arterial blood (PAdes) at a fixed inspired desflurane concentration. Thirteen patients (aged 42–72 years) undergoing coronary artery bypass grafting surgery were enrolled in this study. They were anaesthetised using a constant 5% inspired desflurane concentration (CIdes) during the first hour of anaesthesia. Under constant volume-controlled ventilation, CIdes and end-tidal desflurane (CEdes) were measured with an infrared analyser. The desflurane concentration in the blood was analysed using gas chromatography, and cardiac output was measured using an Opti-Q pulmonary artery catheter. It took 24 min for the Jdes to equilibrate with Ades. Both CIdes–CEdes and Ades–PAdes gradients persisted during the study period. There was no further uptake of desflurane into the brain after 24 min but there was near-constant uptake into the body. [ABSTRACT FROM AUTHOR]

DNA tests in normal subjects and patients with ataxia and Parkinson's disease (PD) were carried out to assess the frequency of spinocerebellar ataxia (SCA) and to document the distribution of SCA mutations underlying ethnic Chinese in Taiwan. MJD/SCA3 (46%) was the most common autosomal dominant SCA in the Taiwanese cohort, followed by SCA6 (18%) and SCA1 (3%). No expansions of SCA types 2, 10, 12, or dentatorubropallidoluysian atrophy (DRPLA) were detected. The clinical phenotypes of these affected SCA patients were very heterogeneous. All of them showed clinical symptoms of cerebellar ataxia, with or without other associated features. The frequencies of large normal alleles are closely associated with the prevalence of SCA1, SCA2, MJD/SCA3, SCA6, and DRPLA among Taiwanese, Japanese, and Caucasians. Interestingly, abnormal expansions of SCA8 and SCA17 genes were detected in patients with PD. The clinical presentation for these patients is typical of idiopathic PD with the following characteristics: late onset of disease, resting tremor in the limbs, rigidity, bradykinesia, and a good response to levodopa. This study appears to be the first report describing the PD phenotype in association with an expanded allele in the TATA-binding protein gene and suggests that SCA8 may also be a cause of typical PD.
