Aims: Type 2 diabetes mellitus (T2DM) is associated with increased cardiovascular disease (CVD) risk, but whether T2DM directly causes adverse cardiac remodelling is uncertain. We performed a comprehensive Mendelian randomization (MR) analysis to investigate the causal relevance of T2DM to CVD outcomes and cardiac structure/function. Methods and results: Bidirectional two‐sample MR was conducted using summary‐level data from European‐ancestry genome‐wide association studies. The T2DM GWAS data included 80 154 cases and 853 816 controls from the DIAGRAM consortium. Outcomes included coronary artery disease (CAD), myocardial infarction (MI), stroke, heart failure, atrial fibrillation, and various quantitative cardiac imaging traits assessed by magnetic resonance imaging. MR analysis revealed causal associations between genetic predisposition to T2DM and increased risk of CAD (odds ratio [OR] 1.104, 95% confidence interval [CI] 1.078–1.130, P = 2.59e‐16), MI (OR 1.129, 95% CI 1.094–1.166, P = 6.02e‐14) and stroke (OR 1.086, 95% CI 1.064–1.109, P = 1.02e‐14). These associations were validated in the FinnGen cohort (CAD: OR 1.117, 95% CI 1.075–1.158, P = 1.56e‐9; MI: OR 1.132, 95% CI 1.083–1.184, P = 4.27e‐8; stroke: OR 1.138, 95% CI 1.107–1.170, P = 3.52e‐20). Multivariable MR show consistent findings (CAD: OR 1.063, 95% CI 1.031–1.097, P = 1.11e‐4; MI: OR 1.088, 95% CI 1.042–1.135, P = 1.12e‐4; stroke: OR 1.066, 95% CI 1.032–1.101, P = 1.18e‐4) after adjusting for cardiometabolic traits. T2DM was causally associated with higher left ventricular mass index (β = 0.473, 95% CI 0.193 to 0.752, P = 0.001), lower indexed right atrial minimum (β = −0.048, 95% CI −0.073 to −0.022, P = 2.1e‐5), and maximum (β = −0.042, 95% CI −0.065 to −0.019, P = 4.12e‐5) areas. The effects on right atrial size remained significant after adjusting for risk factors (minimum area: β = −0.041, 95% CI −0.072 to −0.010, P = 0.009; maximum area: β = −0.039, 95% CI −0.069 to −0.008, P = 0.012). Both apolipoprotein A1 and SBP are important mediators in the causal relationship between T2DM and left ventricular mass index. No reverse causal associations were identified. Conclusions: Our MR study demonstrates that genetic liability to T2DM plays causal roles in CAD, MI, stroke, and cardiac structure changes including left ventricular hypertrophy and reduced right atrial dimensions. The Kuroshio intrusion (KI) is a northwestward‐flowing branch of the Kuroshio Current, which enters the South China Sea (SCS) and regulates its temperature, salinity, and water mass exchanges. However, limited direct observations hinder our understanding of KI's mechanisms and responses to climate change. Here, we present a 60‐year bi‐monthly resolved coral oxygen isotope (δ18Oc) record from Dongsha Atoll, the northern SCS. The dry‐season (December–March) δ18Oc record reveals interannual to decadal variabilities of the KI. The impact of the East Asian winter monsoon (EAWM) on Dongsha δ18Oc was more pronounced during the 1970s and 1980s and after the early 2000s, while the influence of the El Niño‐Southern Oscillation (ENSO) on Dongsha δ18Oc was higher between the 1980s and 1990s. The Pacific Decadal Oscillation (PDO) may have a relatively minor effect on KI strength or may indirectly modulate KI strength through its influence on ENSO activities. Our Dongsha δ18Oc record highlight the importance of the EAWM, ENSO, and PDO in predicting future KI changes. Plain Language Summary: The Kuroshio intrusion is a branch of the Kuroshio Current. Here we present a study on the chemical composition of a coral core from Dongsha Atoll in the northern South China Sea. Our study shows that the intrusion varies markedly over the past 60 years and is connected to changes in sea surface salinity. Stronger intrusions align with stronger East Asian winter monsoon intensity and El Niño events. Studies have shown that antidiabetic drugs can alter the gut microbiota. The hypoglycemic effects of the drugs can be attributed in part to certain species in the gut microbiome that help the drugs work more effectively. In addition, increasing energy expenditure via the induction of adipose tissue browning has become an appealing strategy to treat obesity and associated metabolic complications. Currently, glucagon‐like peptide‐1 receptor agonist (GLP‐1 RA) treatment for metabolic disorders such as obesity and type 2 diabetes has been widely studied. To determine the mechanism of a long‐acting GLP‐1 RA affects adipose tissue browning and the gut microbiome, we treated high‐fat diet mice with GLP‐1 RA and demonstrated that the drug can regulate adipose tissue browning. 16S rRNA and untargeted metabolomics assays suggested that it increased the abundance of bacterium Lactobacillus reuteri and decreased serum ceramide levels in mice. L. reuteri was negatively correlated with ceramide. We found that the mechanism of ceramide decline was alkaline ceramidase 2 (Acer2) overexpression. Moreover, L. reuteri can play a therapeutic synergistic role with GLP‐1 RA, suggesting that gut microbiota can be used as a part of the treatment of diabetes. [ABSTRACT FROM AUTHOR]

Diabetes manifests as chronic inflammation and leads to the development diabetic cardiomyopathy (DCM). Targeting key proteins in inflammatory signaling may provide new therapy for DCM. In this study, the authors explore the pharmacological effects and mechanisms of Schisandrin B (Sch B), a natural compound with anti‐inflammatory activity against DCM. It is shown that Sch B prevents high‐level glucose (HG)‐induced hypertrophic and fibrotic responses in cultured cardiomyocytes. RNA sequencing and inflammatory qPCR microarray show that Sch B mainly affects myeloid differentiation primary response 88 (MyD88)‐dependent inflammatory gene expression in HG‐challenged cardiomyocytes. Further studies indicate that Sch B directly binds to and inhibits MyD88 activation, but does not alter MyD88‐independent Toll‐like receptor signaling in vivo and in vitro. Inhibiting or silencing MyD88 is associated with reduced levels of HG‐induced inflammatory cytokines and myocardial injuries in vitro. Treatment of type 1 and type 2 diabetic mice with Sch B protects heart function, reduces myocardial injuries, and decreases secretion of inflammatory cytokines. Cardiomyocyte‐specific MyD88 knockout also protects mice against cardiac inflammation and injury in type 1 diabetic mice. The impacts of anthropogenic forcing on Indian summer monsoon (ISM) rainfall are obscure, partly due to limited availability of highly resolved hydroclimate proxy records as well as the highly regionalized nature of precipitation. Here, we report an annually‐resolved speleothem oxygen isotope record from Xianren Cave, southwestern China, which represents rainfall change over the broad ISM region. We find that the region has endured at least six decadal‐scale weak monsoon events in the past three hundred years. One of them, lasting from the early to mid 19th century, shares the similar gradual, persistent trend as the most recent decline in ISM rainfall and both have a magnitude substantially larger than the others dominated by natural variability. This early weak monsoon event occurred during a historical time of intensive deforestation in the region. We conclude that the ISM trend could have been altered by the changes in land‐use and land‐cover since the early 19th century. Here, we present a well‐resolved Indian summer monsoon (ISM) rainfall record, based on stable isotope studies on speleothems collected from a cave in southwestern China. The oxygen isotope record shows that ISM rainfall has six multi‐decadal decreases in the past three hundred years. One of the corresponding monsoon weakening, lasting from the early to mid 19th century, cannot be simply explained by natural variations in the climate system. Obesity is often associated with the risk of chronic inflammation and other metabolic diseases, such as diabetes, cardiovascular disease, and cancer. The composition and activity of the gut microbiota play an important role in this process, affecting a range of physiological processes, such as nutrient absorption and energy metabolism. The active gut microbiota can produce a large number of physiologically active substances during the process of intestinal metabolism and reproduction, including short‐chain/long‐chain fatty acids, secondary bile acids, and tryptophan metabolites with beneficial effects on metabolism, as well as negative metabolites, including trimethylamine N‐oxide, delta‐valerobetaine, and imidazole propionate. How gut microbiota specifically affect and participate in metabolic and immune activities, especially the metabolites directly produced by gut microbiota, has attracted extensive attention. So far, some animal and human studies have shown that gut microbiota metabolites are correlated with host obesity, energy metabolism, and inflammation. Some pathways and mechanisms are slowly being discovered. Here, we will focus on the important metabolites of gut microbiota (beneficial and negative), and review their roles and mechanisms in obesity and related metabolic diseases, hoping to provide a new perspective for the treatment and remission of obesity and other metabolic diseases from the perspective of metabolites. Objective: Analyze the clinical and genetic characteristics of a rare Chinese family with Multiple synostoses syndrome and identify the causative variant with the high‐throughput sequencing approach. Methods: The medical history investigation, physical examination, imaging examination, and audiological examination of the family members were performed. DNA samples were extracted from the family members. The candidate variant was identified by performing whole‐exome sequencing of the proband, then verified by Sanger sequencing in the family. Results: The family named HBSY‐018 from Hubei province had 18 subjects in three generations, and six subjects were diagnosed with conductive or mixed hearing loss. Meanwhile, characteristic features including short philtrum, hemicylindrical nose, and hypoplastic alae nasi were noticed among those patients. Symptoms of proximal interdigital joint adhesion and inflexibility were found. The family was diagnosed as Multiple synostoses syndrome type 1 (SYNS1).The inheritance pattern of this family was autosomal dominant. A novel mutation in the NOG gene c.533G>A was identified by performing whole‐exome sequencing of the proband. The substitution of cysteine encoding 178th position with tyrosine (p.Cys178Tyr) was caused by this mutation, which was conserved across species. Co‐segregation of disease phenotypes was demonstrated by the family verification. Conclusion: The family diagnosed as SYNS1 was caused by the novel mutation (c.533G>A) of NOG. The combination of clinical diagnosis and molecular diagnosis had improved the understanding of this rare disease and provided a scientific basis for genetic counseling in the family. Marine biofouling is one of the most significant challenges hindering practical uranium extraction from seawater. Single atoms have been widely used in catalytic applications because of their remarkable redox property, implying that the single atom is highly capable of catalyzing the generation of reactive oxygen species (ROS) and acts as an anti‐biofouling substance for controlling biofouling. In this study, the Co single atom loaded polyacrylamidoxime (PAO) material, PAO‐Co, is fabricated based on the binding ability of the amidoxime group to uranyl and cobalt ions. Nitrogen and oxygen atoms from the amidoxime group stabilize the Co single atom. The fabricated PAO‐Co exhibits a broad range of antimicrobial activity against diverse marine microorganisms by producing ROS, with an inhibition rate up to 93.4%. The present study is the first to apply the single atom for controlling biofouling. The adsorbent achieves an ultrahigh uranium adsorption capacity of 9.7 mg g−1 in biofouling‐containing natural seawater, which decreased only by 11% compared with that in biofouling‐removed natural seawater. These findings indicate that applying single atoms would be a promising strategy for designing biofouling‐resistant adsorbents for uranium extraction from seawater. [ABSTRACT FROM AUTHOR]

Therioaphis trifolii (Monell) is a well‐known pest in alfalfa cultivation in China. Individuals of T. trifolii were allowed to feed on 10 alfalfa cultivars and their growth, reproduction, and population life table parameters were estimated. Nymphs that fed on Hunter River had the longest development time, whereas nymphs that fed on Defu had the shortest development time. Adults that fed on Debao, Zhongmu No. 2, and Defu had the shortest development time, whereas adults that fed on Golden Empress and Hunter River had the longest development times. Individuals that fed on Zhungeer produced the greatest number of offspring (13.950), and those that fed on Hunter River produced the fewest offspring (6.000). The survival rate of T. trifolii was the highest on Algonquin and lowest on Hunter River, whereas the survival time was the longest on Golden Empress and shortest on Debao. The population that fed on Algonquin had the highest intrinsic growth rate (0.560) and shortest population doubling time (1.237 days), whereas those that fed on Hunter River had the lowest intrinsic growth rate (0.217) and longest population doubling time (3.194 days). The life table parameters of the experimental T. trifolii population were used to quantify the resistance strengths of the cultivars. Results indicate that the Hunter River cultivar may effectively resist damage from T. trifolii. This study provides a theoretical basis for breeding aphid‐resistant alfalfa cultivars, enabling further research into the prevention and control of aphid infestation. [ABSTRACT FROM AUTHOR]

The detection of environmental uranyl is attracting increasing attention. However, the available detection strategies mainly depend on the selective recognition of uranyl, which is subject to severe interference by coexisting metal ions. Herein, based on the unique uranyl‐triggered photocleavage property, the protein BSA is labelled with fluorescent molecules that exhibit an aggregation‐induced emission effect for uranyl detection. Uranyl‐triggered photocleavage causes the separation of the fluorescent‐molecule‐labelled protein fragments, leading to attenuation of the emission fluorescence, which is used as a signal for uranyl detection. This detection strategy shows high selectivity for uranyl and an ultralow detection limit of 24 pM with a broad detection range covering five orders of magnitude. To investigate the relationship between red blood cell distribution width (RDW) and residual renal function (RRF) in patients undergoing continuous ambulatory peritoneal dialysis (CAPD). Seventy‐seven CAPD patients were enrolled in this study. According to receiver operator characteristic (ROC) curve analysis, patients were divided into high RDW (RDW > 14.95%) and low RDW (RDW ≤ 14.95%) groups. The data of baseline clinical, biochemical parameters, comorbidities, medication status, peritoneal function, and dialysis adequacy were compared. Survival curves were calculated using Kaplan–Meier method. Cox regression model was employed to analyze risk factors of decline in RRF. The overall median survival time was 24 months, the median survival time of high RDW group (46 patients) and low RDW group (31 patients) were 24 and 12 months, respectively. Compared with the low RDW group, patients in the high RDW group were older, higher rate of decline RRF and white blood cells count as well as lower total Kt/V (all p < 0.05). Kaplan–Meier survival curves showed that the low RDW group had higher survival of RRF compared with the high RDW group (p < 0.001). Multivariate Cox regression analysis showed that high RDW was independent risk factor for decline of RRF(hazard ratio = 1.441, 95% confidence interval: 1.089–1.905, p = 0.01). Increased baseline RDW is associated with decline of RRF in CAPD patients and RDW can be stratified as a valuable indicator for the risk of RRF decline. [ABSTRACT FROM AUTHOR]

The extraction of uranium from seawater, which is an abundant resource, has attracted considerable attention as a viable form of energy‐resource acquisition. The two critical factors for boosting the chemical thermodynamics of uranium extraction from seawater are the availability of sufficient amounts of uranyl ions for supply to adsorbents and increased interaction temperatures. However, current approaches only rely on the free diffusion of uranyl ions from seawater to the functional groups within adsorbents, which largely limits the uranium extraction capacity. Herein, inspired by the mechanism of plant transpiration, a plant‐mimetic directional‐channel poly(amidoxime) (DC‐PAO) hydrogel is designed to enhance the uranium extraction efficiency via the active pumping of uranyl ions into the adsorbent. Compared with the original PAO hydrogel without plant‐mimetic transpiration, the uranium extraction capacity of the DC‐PAO hydrogel increases by 79.33% in natural seawater and affords the fastest reported uranium extraction average rate of 0.917 mg g−1 d−1 among the most state‐of‐the‐art amidoxime group‐based adsorbents, along with a high adsorption capacity of 6.42 mg g−1 within 7 d. The results indicate that the proposed method can enhance the efficiency of solar‐transpiration‐based uranium extraction from seawater, particularly in terms of reducing costs and saving processing time. With the ability to activate certain signaling pathways, chemokines and their receptors may facilitate tumor progression at key steps, including proliferation, immunomodulation, and metastasis. Nevertheless, their prognostic value and regulatory mechanism warrant thorough studies in liver cancer. Here, by screening the expression profiles of all known chemokines in independent liver cancer cohorts, we found that CCL23 was frequently downregulated at mRNA and protein levels in liver cancer. Decreased CCL23 correlated with shortened patient survival, enrichment of signatures related to cancer stem cell property, and metastatic potential. In addition to serving as a tumor suppressor through recruiting CD8+ T cell infiltration in liver cancer, CCL23 could repress cancer cell proliferation, stemness, and mobility. Mechanistically, the expression of CCL23 was transcriptionally regulated by ESR1. On the other hand, CCL23 could suppress the activation of AKT signaling and thus promote the expression of ESR1, forming a feedback loop in liver cancer cells. Collectively, these findings reveal that loss of CCL23 drives liver cancer progression by coordinating immune evasion and metastasis initiation. Mixed‐halide perovskites (MHPs) have emerged as important semiconductor materials for optoelectronic devices due to the bandgap tunability by halide composition. However, their device applications are hampered by light‐induced ion migration and halide segregation, leading to the formation of lower bandgap domains with red‐shifted photoluminescence. Previous studies of phase segregation in MHPs have predominantly focused on their linear optical properties. Herein, nonlinear absorption (NLA) properties of MAPbBr2I films before and after femtosecond laser irradiation are investigated using the Micro‐I‐scan technique. An unusual NLA switching from reverse‐saturable absorption (RSA) to saturable absorption (SA) under intense laser illumination is found. It is unveiled that the initial RSA originates from two‐photon absorption in uniform bromide‐rich regions of as‐prepared film whereas the SA occurs due to state filling effect in the newly generated low bandgap I‐rich phases under laser irradiation. The switching threshold and effective NLA coefficient are highly dependent on the distribution of halide ions, which is controllable through the laser illumination time. Furthermore, an all‐optical logic gate scheme is demonstrated based on the unusual NLA switching. The results not only unveil that halide segregation results in NLA switching in MHPs but also pave the way for the realization of MHPs‐based nonlinear photonic devices. [ABSTRACT FROM AUTHOR]

Sex pheromones serve a critical role in Lepidopterans finding mates. Male moths perceive and react to sex pheromones emitted by conspecific females through a delicate pheromone communication system. Pheromone receptors (PRs) are the key sensory elements at the beginning of that process. The codling moth (Cydia pomnonella) is an important pome fruit pest globally and a serious invasive species in China. Pheromone‐based techniques have been used successfully in monitoring and controlling this species. We conducted ribonucleic acid sequencing analysis of the codling moth antennal transcriptome and identified 66 odorant receptors (ORs) in a population from Xinjiang province, China, of which 14 were PRs, including two novel PRs (CpomOR2e and CpomOR73). Four PRs that contain full‐length open reading frames (CpomOR1, OR2a, OR5, OR7) and four PRs with ligands that have not been reported previously (CpomOR1, OR2a, OR5, OR7) were selected to deorphanize in the heterologous Xenopus oocyte expression system. Specifically, we found that CpomOR2a and CpomOR5 responded to (E,E)‐8, 10‐dodecadien‐1‐yl acetate (codlemone acetate). Furthermore, CpomOR5 (EC50 = 1.379 × 10−8 mol/L) was much more sensitive to codlemone acetate than CpomOR2a (EC50 = 1.663 × 10−6 mol/L). Since codlemone acetate is an important component of C. pomonella sex pheromone, our results improve the current understanding of pheromone communication in codling moths and will be helpful for the development of pest management strategies. Impaired wound healing is a common complication of diabetes. In diabetic wounds, macrophages present dysfunctional efferocytosis and abnormal phenotypes, which could result in excessive neutrophil accumulation and prolonged inflammation, thereby eventually hindering wound repair. ANXA1 N‐terminal peptide Ac2‐26 exhibits a high potential in mitigating inflammation and improving repair; however, its efficacy in diabetic wound repair remains unclear. In this study, a cutaneous excisional wound model was built in genetically diabetic mice. Ac2‐26 or a vehicle solution was employed locally in wound sites. Subsequently, wound zones were measured and sampled at different time intervals post‐wounding. Using hematoxylin‐eosin and Masson's trichrome staining, we observed the histopathological variations and collagen deposition in wound samples. Based on immunohistochemistry and immunofluorescence, the numbers of neutrophils, macrophages, and CD206‐positive macrophages in the wound samples were determined. Cytokine expression in wound samples was studied by immunoblot assay. Results showed that Ac2‐26 treatment could facilitate diabetic wound closure, down‐regulate the number of neutrophils, and improve angiogenesis and collagen deposition. In addition, Ac2‐26 application expedited macrophage recruitment and up‐regulated the percentage of macrophages expressing CD206, which is a marker for M2 macrophages. Moreover, Ac2‐26 inhibited the expressions of TNF‐α and IL‐6 and up‐regulated the expressions of IL‐10, TGF‐β, and VEGFA during diabetic wound healing. Hence, based on the aforementioned findings, Ac2‐26 application in diabetic wounds could exert anti‐inflammatory and pro‐repair effects by reducing neutrophil accumulation and facilitating M2 macrophage development. Constructed from rigid 2,5‐bis(4‐pyridyl)thiazolo[5,4‐d]thiazole (Py2TTz) and various dicarboxylic acids [2‐methyl‐1,4‐benzenedicarboxylic acid (2‐CH3‐H2BDC), 5‐methyl‐1,3‐benzenedicarboxylic acid (5‐CH3‐H2BDC), 2,5‐dimethyl‐1,4‐benzenedicarboxylic acid (2,5‐di‐CH3‐H2BDC), 2,5‐dihydroxyl‐1,4‐benzenedicarboxylic acid (2,5‐di‐OH‐H2BDC), 1,4‐naphthalenedicarboxylic acid (1,4‐H2NDC), and 2,6‐naphthalenedicarboxylic acid (2,6‐H2NDC)], nine novel Zn(II)/Cd(II) MOFs have been synthesized under solvothermal conditions. They possess diverse structures, which mainly result from the different shapes and sizes of the auxiliary dicarboxylate ligands. Linear dicarboxylate ligands (2‐CH3‐BDC, 2,5‐di‐CH3‐BDC, 1,4‐NDC, 2,6‐NDC) generate 3D multi‐fold interpenetrated structures, while the V‐shaped dicarboxylate ligand (5‐CH3‐BDC) affords a 2D layer structure. The well‐known Erzberg site represents the largest siderite (FeCO3) deposit in the world. It consists of various carbonates accounting for the formation of prominent CaCO3 (dominantly aragonite) precipitates filling vertical fractures of different width (centimetres to decimetres) and length (tens of metres). These commonly laminated precipitates are known as 'erzbergite'. This study focuses on the growth dynamics and environmental dependencies of these vein fillings. Samples recovered on‐site and from mineral collections were analyzed, and these analyses were further complemented by modern water analyses from different Erzberg sections. Isotopic signatures support meteoric water infiltration and sulphide oxidation as the principal hydrogeochemical mechanism of (Ca, Mg and Fe) carbonate host rock dissolution, mobilization and vein mineralization. Clumped isotope measurements revealed cool formation temperatures of ca 0 to 10°C for the aragonite, i.e. reflecting the elevated altitude Alpine setting, but unexpectedly low for aragonite nucleation. The 238U–234U–230Th dating yielded ages from 285·1 ± 3·9 to 1·03 ± 0·04 kyr bp and all samples collected on‐site formed after the Last Glacial Maximum. The observed CaCO3 polymorphism is primarily controlled by the high aqueous Mg/Ca ratios resulting from dissolution of Mg‐rich host rocks, with Mg/Ca further evolving during prior CaCO3 precipitation and CO2 outgassing in the fissured aquifer. Aragonite represents the 'normal' mode of erzbergite formation and most of the calcite is of diagenetic (replacing aragonite) origin. The characteristic lamination (millimetre‐scale) is an original growth feature and mostly associated with the deposition of stained (Fe‐rich) detrital particle layers. Broader zonations (centimetre‐scale) are commonly of diagenetic origin. Petrographic observations and radiometric dating support an irregular nature for most of the layering. Open fractures resulting from fault tectonics or gravitational mass movements provide water flow routes and fresh chemical reaction surfaces of the host rock carbonates and accessory sulphides. If these prerequisites are considered, including the hydrogeochemical mechanism, modern water compositions, young U‐Th ages and calculated precipitation rates, it seems unlikely that the fractures had stayed open over extended time intervals. In recent years, more and more nanomaterials‐based chemiluminescence (CL) systems have appeared to improve the sensitivity and expand the scope of the analytical applications with the explosive growth and development of nanomaterials and technology. As a fascinating class of luminescent carbon nanomaterials, carbon dots (CDs) are now substantially studied in fabricating CL based assays due to their unique optical and mechanical properties. Herein, we summarize and highlight the current developments of CDs‐involved weak or ultraweak CL systems, as well as the corresponding mechanisms and proper applications in some fields. CDs can take part in the CL reactions as oxidants, emitting species directly involved in redox oxidation, energy acceptors of CL energy transfer, or even catalysts involving other luminophores. In fact, they always have more than one role in many cases, owing to the formation of various excited species with short life in CL systems. Therefore, in this review article, the most recent progress of the different CDs‐assisted CL systems including the mechanisms and applications are presented. Finally, the conclusions and future prospects of this field are also discussed. Abstract: A GaN HEMT oscillator implemented with the WIN 0.25 μm GaN HEMT technology is designed. The oscillator consists of two HEMT amplifiers with cross‐coupled feedback topology. With the supply voltage of VDD = 0.4 V, the GaN VCO current and power consumption of the oscillator are 6.673 mA and 2.669 mW, respectively. The oscillator can generate differential signal at 4.7 GHz and it also supplies output power − 5.3 dBm. At 1 MHz frequency offset from the carrier the phase noise is −121.77 dBc/Hz. The C-H stretching vibration serves as an important probe for characterizing molecular structures and properties of hydrocarbons. In this work, we present a detailed study on gas-phase Raman spectrum of n-propanol in the C-H stretching region using stimulated photoacoustic Raman spectroscopy. A complete assignment was carried out with the aid of quantum chemistry calculations and depolarization ratio measurement as well as isotope substitutions, i.e. CH3CD2CD2OH, CD3CH2CD2OH and CD3CD2CH2OH. It is shown that the spectra of three C-H groups of n-propanol overlap each other because of Fermi resonance coupling and different molecular conformations, leading to complex features that were not determined previously. In addition, the comparisons between the spectra of three isotopologues reveal that the C-H vibrations at different sites of carbon chain exhibit different sensitivity to conformational change of n-propanol. The CH3 stretching vibration at terminated γ-carbon is not sensitive whereas the CH2 stretching vibrations at both α-carbon and β-carbon atoms are sensitive. The micro-structure of hydration shell of solute in water is significant for understanding the properties of aqueous solutions. However the spectra of hydration shell are difficult to be obtained. Herein, a novel Raman ratio spectra, which is obtained through dividing the Raman spectra of aqueous solutions from the spectrum of water, was applied to deduce the spectra of hydration shell of organic (acetone-D6) and inorganic compounds (NaNO3, NaSCN, NaClO4, Na2SO4, NaCl) in water. Those spectra of the hydration shell were employed to study the micro-structures of the first hydration shells of anions, the number of water molecules in the first hydration shell of free anions and acetone-D6, and the aggregation behavior of ions in the concentrated aqueous NaNO3. The number of water molecules in the hydration shell was supported by our molecular dynamic simulations. The Raman ratio spectra can be widely employed to obtain the spectra of the first hydration shell, and it is helpful to understand the micro-structure of aqueous solutions. A series of transition metal coordination complexes, {[Cu(3-Sptta)2] ·1/2H2O} n ( 1), {[Cd(3-Sptta)2] ·1/2H2O} n ( 2), [Zn(3-Sptta)2(H2O)4] ( 3), and [Co(3-Sptta)2(H2O)4] ( 4) were synthesized under solvothermal and solvent evaporation conditions using the newly designed heterocyclic N/NS carboxylate ligand [2-(3-pyridyl)-1,3,4-thiadiazole-5-]-thio-acetate (3-Sptta) as a main building block. Their structures were determined by single-crystal X-ray diffraction analyses and further characterized by elemental analyses and IR spectroscopy. The four complexes exhibit structural diversity: complexes 1 and 2 exhibit similar 2D 'wave-like' double-layer framework with two shares of mutually parallel left-handed and right-handed helical chains, whereas 3 and 4 present mononuclear structures. Despite advances in medical science, the causes of death can sometimes only be determined by pathologists after a complete autopsy. Few studies have investigated the importance of forensic autopsy in medically disputed cases among different levels of institutional settings. Our study aimed to analyze forensic autopsy in 120 cases of medical disputes among five levels of institutional settings between 2001 and 2012 in Wenzhou, China. The results showed an overall concordance rate of 55%. Of the 39% of clinically missed diagnosis, cardiovascular pathology comprises 55.32%, while respiratory pathology accounts for the remaining 44. 68%. Factors that increase the likelihood of missed diagnoses were private clinics, community settings, and county hospitals. These results support that autopsy remains an important tool in establishing causes of death in medically disputed case, which may directly determine or exclude the fault of medical care and therefore in helping in resolving these cases. Despite recent advances in therapy, chronic lymphocytic leukaemia (CLL) remains incurable and new treatment strategies are therefore urgently required. Inhibitor of apoptosis proteins (IAPs) are over-expressed in CLL, suggesting both a role in disease pathogenesis and the potential for therapeutic targeting. To explore these questions, we evaluated the effects on primary CLL cells of AZD5582, a novel potent and selective inhibitor of IAPs. AZD5582 at nanomolar concentrations induced extensive degradation of cIAP-1 and cIAP-2, but minimally of X chromosome-linked IAP (XIAP). However, these effects of AZD5582 produced little or no direct cytotoxicity, nor did they sensitize CLL cells to p53-dependent killing by fludarabine or p53-independent killing by dexamethasone. In contrast, AZD5582 significantly enhanced apoptosis induced by the death receptor (DR) agonist tumour necrosis factor-related apoptosis-inducing ligand (TRAIL). Importantly, killing by TRAIL plus AZD5582 was independent of adverse prognostic features including TP53 deletion which is strongly associated with chemoresistance in CLL. Coculture experiments involving transfected mouse fibroblasts expressing human CD40L (CD154) to mimic the effect of T cells at sites of tissue involvement showed that CD40 stimulation almost completely prevented the killing of CLL cells by TRAIL plus AZD5582 despite up-regulating TRAIL receptors 1 and 2. In conclusion, our findings confirm the rate-limiting, upstream involvement of IAPs in the extrinsic but not intrinsic apoptotic pathway of CLL cells and suggest that drug combinations that simultaneously activate DRs and inhibit IAPs may have therapeutic potential in patients with CLL who have failed T-cell-depleting chemotherapy. This study was conducted to investigate the possibility that TP53 mRNA is variably expressed in chronic lymphocytic leukaemia (CLL) and that under-expression is associated with TP53 dysfunction and adverse outcome. Although TP53 mRNA levels did indeed vary among the 104 CLL samples examined, this variability resulted primarily from over-expression of TP53 mRNA in 18 samples, all of which lacked TP53 deletion/mutation. These patients had higher lymphocyte counts and shorter overall and treatment-free survival times compared to cases with low TP53 mRNA expression and no TP53 deletion/mutation. Furthermore, TP53 mRNA levels did not correlate with levels of TP53 protein or its transcriptional target CDKN1A. We speculated that the adverse outcome associated with TP53 mRNA over-expression might reflect variation in levels of MIR15A and MIR16-1, which are encoded on chromosome 13q14 and target TP53 and some oncogenes including BCL2. In keeping with our hypothesis, 13q14 copy number and levels of MIR15A/MIR16-1 correlated positively with one another but negatively with levels of TP53 mRNA and BCL2 mRNA. Our findings support a model in which loss of MIR15A/MIR16-1 at chromosome 13q14 results in adverse outcome due to de-repression of oncogenes such as BCL2, and up-regulation of TP53 mRNA as a bystander effect. We developed a dietary exposure assay for screening insecticidal compounds for their toxicity and for assessing the side effects of insecticidal proteins produced by genetically engineered (GE) plants on the planthopper Laodelphax striatellus Fallén. The fitness bioassay confirmed that the diet fulfills the requirements to be used in the dietary exposure system. To validate the efficacy of the dietary exposure system, nymphs of L. striatellus were fed diets treated with different concentrations of an inorganic stomach poison, potassium arsenate (PA), or a cysteine protease inhibitor, E-64. The results showed that with increasing concentrations of E-64, the larval development time was prolonged, the adult weight was reduced and the survival rate of L. striatellus was decreased. Similarly the survival rates of L. striatellus consistently decreased with increasing PA content in the diet. The data indicate that the dietary exposure assay is able to detect the effects of insecticidal compounds on L. striatellus. Subsequently, this assay was successfully used for assessing the potential toxicity of Cry2Aa. The results showed that L. striatellus larvae were not negatively affected when fed the artificial diet containing purified Cry2Aa at 300 μg/g diet. In the assay, the stability and bioactivity of crystal (Cry) proteins in the food sources were confirmed by enzyme-linked immunosorbent assay and sensitive-insect bioassays. These results show that L. striatellus is not sensitive to Cry2Aa. We conclude that the dietary exposure system is valid and useful for assessing the toxicity of insecticidal compounds produced by GE plants on planthoppers. [ABSTRACT FROM AUTHOR]

The vibrational spectra of gaseous and liquid 2-propanol in the C-H stretching region of 2800 ~ 3100 cm−1 were investigated by polarized photoacoustic Raman spectroscopy and conventional Raman spectroscopy, respectively. Using two deuterated samples, that is, CH3CDOHCH3 and CD3CHOHCD3, the overlapping spectral features between the CH and CH3 groups were identified. With the aid of depolarization ratio measurements and density functional theory calculations, a new spectral assignment was presented. In the gas phase, the band at 2884 cm−1 was assigned to the overlapping of one CH3 Fermi resonance mode and a CH stretching of gauche conformer. The bands at 2917 and 2933 cm−1 were assigned to another two CH3 Fermi resonance modes, but the latter includes weak contribution from CH stretching of trans conformer. The bands at 2950 and 2983 cm−1 were assigned to CH3 symmetric and antisymmetric stretching, respectively. The spectral features of liquid 2-propanol are similar to those in the gas phase except for the blue shift of CH and the red shift of CH3 band positions, which can be attributed to the intermolecular interaction in the liquid state. The new assignments not only clarify the confusions in previous studies from different spectral methods but also provide the reliable groundwork on spectral application of 2-propanol in the futures. In chronic lymphocytic leukaemia ( CLL), TP53 mutation and deletion are strongly associated with one another and with adverse clinical outcome. Mutant TP53 protein typically accumulates to high levels and has been reported to have transcriptional regulatory activity distinct from that of wild-type TP53. To investigate whether such an effect is relevant to CLL, carefully balanced primary CLL samples with or without TP53 mutation/deletion were compared for their gene expression profiles using high-density DNA microarrays. Ninety-six and eight differentially expressed genes were identified, respectively, using two alternative statistical approaches with different stringencies. None of the differentially expressed genes were known to be regulated by mutant TP53, and only four of the 67 under-expressed genes were known transcriptional targets of wild-type TP53. Significantly, both approaches showed that gene under-expression was the dominant feature of TP53-mutant CLL samples. Furthermore, a disproportionate number of the under-expressed genes were located on chromosome 17p, the most significant being TP53 itself. Together, these results indicate that any transcriptional regulatory effects of mutant TP53 in CLL cells are overshadowed by the under-expression of co-deleted TP53 and other genes on chromosome 17p. ABSTRACT PURPOSE Multimodal CT with CT angiography (CTA) and CT perfusion (CTP) are increasingly used in stroke triage. Our aim was to identify parameters most predictive of hemorrhagic transformation (HT), especially symptomatic intracerebral hemorrhage (SICH). METHODS This retrospective study included patients evaluated by baseline multimodal CT ≤ 9 hours from ictus with acute nonlacunar middle cerebral artery (MCA) territory infarction. Two readers independently evaluated CTP maps for ischemic severity and CTA source images (CTA-SI) for infarct extent (as measured by ASPECTS). Presence of proximal occlusion (ICA or M1) and degree of collateralization (collateral score) were also assessed on CTA. HT was defined as SICH if associated with deterioration ≥ 4-points on NIHSS. Multivariate logistic regression analysis identified independent predictors of SICH. ROC curves selected optimal thresholds. RESULTS Of 84 patients reviewed, HT occurred in 22 (26.2%) and SICH in 8 (9.5%). Univariate predictors for SICH were proximal occlusion (OR = 8.65, P= .049), collateral score (OR = .34, P= .017), ASPECTS (OR = .46, P= .001), and CBV (OR = .001, P= .005). Multivariate analysis revealed ASPECTS as the only independent predictor with optimal threshold ≤ 5 and sensitivity and specificity of 75.0% and 85.5%, respectively. It has been reported that low-pH-induced fusion of influenza virus with liposomes results in rapid and extensive release of both low- and high-molecular-weight substances from the liposomes [Gu¨nther-Ausborn et al., J. Biol. Chem. 270 (1995) 29279–29285; Shangguan et al., Biochemistry 35 (1996) 4956–4965]. Here, we demonstrate retention of encapsulated water-soluble compounds during fusion of Semliki Forest virus (SFV) or Sindbis virus with liposomes at low pH. Under conditions allowing complete fusion of the liposomes, a limited fluorescence dequenching of liposome-encapsulated calcein was observed, particularly for SFV. Also, radioactively labeled inulin or sucrose were largely retained. Freezing and thawing of the viruses in the absence of sucrose resulted in an enhanced leakiness of fusion. These results support the notion that the alphavirus fusion event per se is non-leaky and may well involve a discrete hemifusion intermediate. [Copyright &y& Elsevier]
